Preface
(2018)
UNC119 and PDE delta are lipid-binding proteins and are thought to form diffusible complexes with transducin-alpha and prenylated OS proteins, respectively, to mediate their trafficking to photoreceptor outer segments. Here, we investigate mechanisms of trafficking which are controlled by Arf-like protein 3 (Arl3), a small GTPase. The activity of ARL3 is regulated by a GEF (ARL13b) and a GAP (RP2). In a mouse germline knockout of RP2, ARL3-GTP is abundant as its intrinsic GTPase activity is extremely low. High levels of ARL3-GTP impair binding and trafficking of cargo to the outer segment. Germline knockout of ARL3 is embryonically lethal generating a syndromic ciliopathy-like phenotype. Retina-and rod-specific knockout of ARL3 allow to determine the precise mechanisms leading to photoreceptor degeneration. The knockouts reveal binary functions of ARL3-GTP as a key molecule in late-stage photoreceptor ciliogenesis and cargo displacement factor.
An Information System Supporting the Eliciting of Expert Knowledge for Successful IT Projects
(2018)
In order to guarantee the success of an IT project, it is necessary for a company to possess expert knowledge. The difficulty arises when experts no longer work for the company and it then becomes necessary to use their knowledge, in order to realise an IT project. In this paper, the ExKnowIT information system which supports the eliciting of expert knowledge for successful IT projects, is presented and consists of the following modules: (1) the identification of experts for successful IT projects, (2) the eliciting of expert knowledge on completed IT projects, (3) the expert knowledge base on completed IT projects, (4) the Group Method for Data Handling (GMDH) algorithm, (5) new knowledge in support of decisions regarding the selection of a manager for a new IT project. The added value of our system is that these three approaches, namely, the elicitation of expert knowledge, the success of an IT project and the discovery of new knowledge, gleaned from the expert knowledge base, otherwise known as the decision model, complement each other.
No other means of communication determines through its seemingly unrestricted possibilities our everyday life more than the internet. From the mid-90s onwards, more and more technical advancements in the field of communication appear on the market, which in turn call for new terminology. In the first place, it is the internet (essentially based on the interaction between users and experts), which requires effective nomenclature in order to mediate between lay users and their restricted knowledge on the one, and experts and their sophisticated terminology on the other hand. At the interface between the new and complex realities and the need for simple linguistic access, a huge quantity of metaphoric denominations is used, making abstract innovations more comprehensible. Metaphor in the internet discourse serves to "reduce verticality" (Stenschke 2006) between specialized terminology and common language. The paper deals with metaphors based on spatial concepts. Space and spatiality play a key role in cognitive theories of metaphor as these theories themselves (according to Lakoff/Johnson 1980) are often based on the application of spatial concepts to non-spatial relations. After describing spatial concepts in general (referring to the internet), the paper explores which kind of metaphor takes advantage of the complexity present in the internet and how the medial space is linguistically recaptured in terms of spatial perception.
The Aral Sea desiccation and related changes in hydroclimatic conditions on a regional level is a hot topic for past decades. The key problem of scientific research projects devoted to an investigation of modern Aral Sea basin hydrological regime is its discontinuous nature - the only limited amount of papers takes into account the complex runoff formation system entirely. Addressing this challenge we have developed a continuous prediction system for assessing freshwater inflow into the Small Aral Sea based on coupling stack of hydrological and data-driven models. Results show a good prediction skill and approve the possibility to develop a valuable water assessment tool which utilizes the power of classical physically based and modern machine learning models both for territories with complex water management system and strong water-related data scarcity. The source code and data of the proposed system is available on a Github page (https://github.com/SMASHIproject/IWRM2018).
We consider a distributed learning approach in supervised learning for a large class of spectral regularization methods in an reproducing kernel Hilbert space (RKHS) framework. The data set of size n is partitioned into m = O (n(alpha)), alpha < 1/2, disjoint subsamples. On each subsample, some spectral regularization method (belonging to a large class, including in particular Kernel Ridge Regression, L-2-boosting and spectral cut-off) is applied. The regression function f is then estimated via simple averaging, leading to a substantial reduction in computation time. We show that minimax optimal rates of convergence are preserved if m grows sufficiently slowly (corresponding to an upper bound for alpha) as n -> infinity, depending on the smoothness assumptions on f and the intrinsic dimensionality. In spirit, the analysis relies on a classical bias/stochastic error analysis.
For linear inverse problems Y = A mu + zeta, it is classical to recover the unknown signal mu by iterative regularization methods ((mu) over cap,(m) = 0,1, . . .) and halt at a data-dependent iteration tau using some stopping rule, typically based on a discrepancy principle, so that the weak (or prediction) squared-error parallel to A((mu) over cap (()(tau)) - mu)parallel to(2) is controlled. In the context of statistical estimation with stochastic noise zeta, we study oracle adaptation (that is, compared to the best possible stopping iteration) in strong squared- error E[parallel to((mu) over cap (()(tau)) - mu)parallel to(2)]. For a residual-based stopping rule oracle adaptation bounds are established for general spectral regularization methods. The proofs use bias and variance transfer techniques from weak prediction error to strong L-2-error, as well as convexity arguments and concentration bounds for the stochastic part. Adaptive early stopping for the Landweber method is studied in further detail and illustrated numerically.
Background: Comprehension of non-canonical sentences is frequently characterised by chance level performance in people with aphasia (PWA). Chance level performance has been interpreted as guessing, but online data does not support this rendering. It is still not clear whether the incorrect sentence processing is guided by the compensatory strategies that PWA might employ to overcome linguistic difficulties.Aims: We aim to study to what extent people with non-fluent aphasia are aware of their sentence comprehension deficits. Methods & Procedures: This study combined offline and online data to investigate the effect of word order and error-awareness on sentence comprehension in a group of PWA and non-brain damaged (NBD) participants. The offline tasks involved auditory sentence picture-matching immediately followed by a confidence rating (CR). Participants were asked to judge the perceived correctness of their previous answer. Online data consisted of eye-tracking.Outcomes & Results: Replicating previous findings, PWA had significantly worse comprehension of Theme-Agent order compared to Agent-Theme order sentences. Controls showed ceiling level sentence comprehension. CR was a poor predictor of response accuracy in PWA, but moderate-good in NBD. A total of 6.8% of judgements were classified as guessing by PWA. Post hoc gaze data analysis indicated that CR was a predictor of the fixation pattern during the presentation of the linguistic stimuli. Conclusions: Results suggest that PWA were mostly unaware of their sentence comprehension errors and did not consciously employ strategies to compensate for their difficulties.
That’s not quite it

We present a novel empirical study on German directly comparing the exhaustivity inference in es-clefts to exhaustivity inferences in definite pseudoclefts, exclusives, and plain intonational focus constructions. We employ mouse-driven verification/falsification tasks in an incremental information-retrieval paradigm across two experiments in order to assess the strength of exhaustivity in the four sentence types. The results are compatible with a parallel analysis of clefts and definite pseudoclefts, in line with previous claims in the literature (Percus 1997, Buring & Kriz 2013). In striking contrast with such proposals, in which the exhaustivity inference is conventionally coded in the cleft-structure in terms of maximality/homogeneity, our study found that the exhaustivity inference is not systematic or robust in es-clefts nor in definite pseudoclefts: Whereas some speakers treat both constructions as exhaustive, others treat both constructions as non-exhaustive. In order to account for this unexpected finding, we argue that the exhaustivity inference in both clefts and definite pseudoclefts-specifically those with the compound definite derjenige - is pragmatically derived from the anaphoric existence presupposition that is common to both constructions.
Breaking the Barrier
(2018)
Topical administration permits targeted, sustained delivery of therapeutics to human skin. Delivery to the skin, however, is typically limited to lipophilic molecules with molecular weight of < 500 Da, capable of crossing the stratum corneum. Nevertheless, there are indications protein delivery may be possible in barrier deficient skin, a condition found in several inflammatory skin diseases such as psoriasis, using novel nanocarrier systems. Methods: Water in water thermo-nanoprecipitation; dynamic light scattering; zeta potential measurement; nanoparticle tracking analysis; atomic force microscopy; cryogenic transmission electron microscopy; UV absorption; centrifugal separation membranes; bicinchoninic acid assay; circular dichroism; TNF alpha binding ELISA; inflammatory skin equivalent construction; human skin biopsies; immunohistochemistry; fluorescence microscopy; western blot; monocyte derived Langerhans cells; ELISA Results: Here, we report the novel synthesis of thermoresponsive nanogels (tNG) and the stable encapsulation of the anti-TNFa fusion protein etanercept (ETR) (similar to 150 kDa) without alteration to its structure, as well as temperature triggered release from the tNGs. Novel tNG synthesis without the use of organic solvents was conducted, permitting in situ encapsulation of protein during assembly, something that holds great promise for easy manufacture and storage. Topical application of ETR loaded tNGs to inflammatory skin equivalents or tape striped human skin resulted in efficient ETR delivery throughout the SC and into the viable epidermis that correlated with clear anti-inflammatory effects. Notably, effective ETR delivery depended on temperature triggered release following topical application. Conclusion: Together these results indicate tNGs hold promise as a biocompatible and easy to manufacture vehicle for stable protein encapsulation and topical delivery into barrier-deficient skin.
Bryophytes constitute an important and permanent component of the grassland flora and diversity in Europe. As most bryophyte species are sensitive to habitat change, their diversity is likely to decline following land-use intensification. Most previous studies on bryophyte diversity focused on specific habitats of high bryophyte diversity, such as bogs, montane grasslands, or calcareous dry grasslands. In contrast, mesic grasslands are rarely studied, although they are the most common grassland habitat in Europe. They are secondary vegetation, maintained by agricultural use and thus, are influenced by different forms of land use. We studied bryophyte species richness in three regions in Germany, in 707 plots of 16 m(2) representing different land-use types and environmental conditions. Our study is one of the few to inspect the relationships between bryophyte richness and land use across contrasting regions and using a high number of replicates. Among the managed grasslands, pastures harboured 2.5 times more bryophyte species than meadows and mown pastures. Similarly, bryophyte cover was about twice as high in fallows and pastures than in meadows and mown pastures. Among the pastures, bryophyte species richness was about three times higher in sheep grazed plots than in the ones grazed by cattle or horses. In general, bryophyte species richness and cover was more than 50% lower in fertilized than in unfertilized plots. Moreover, the amount of suitable substrates was linked to bryophyte diversity. Species richness of bryophytes growing on stones increased with stone cover, and the one of bryophytes growing on bark and deadwood increased with larger values of woody plant species and deadwood cover. Our findings highlight the importance of low-intensity land use and high structural heterogeneity for bryophyte conservation. They also caution against an intensification of traditionally managed pastures. In the light of our results, we recommend to maintain low-intensity sheep grazing on sites with low productivity, such as slopes on shallow soils.
This study examined a theoretical model hypothesizing that reading strategies mediate the effects of intrinsic reading motivation, reading fluency, and vocabulary knowledge on reading comprehension. Using path analytic methods, we tested the direct and indirect effects specified in the hypothesized model in a sample of 1105 fifth-graders. In addition to standardized tests and questionnaires, we administered a performance test to assess students' proficiency in the application of three reading strategies. The overall fit of the model to the data was good. Both cognitive (fluency and vocabulary) and motivational (intrinsic reading motivation) variables had an indirect effect on reading comprehension through their influence on reading strategies. Reading strategies had a unique effect on reading comprehension and partially mediated the effects that cognitive and motivational variables had on fifth-graders' reading achievements.
The electret state stability in nonpolar semicrystalline polymers is largely determined by the traps located at crystalline/ amorphous phase interfaces. Thus, the thermal history of such polymers should considerably influence their electret properties. In the present work, we investigate how recrystallization influences charge stability in low-density polyethylene corona electrets. It has been found that electret charge stability in quenched samples is higher than in slowly-crystallized ones. Phenomenologicaly, this can be explained by the increased number of deeper traps in samples with smaller crystallite size.
The process of leaf senescence consists of the final stage of leaf development. It has evolved as a mechanism to degrade macromolecules and micronutrients and remobilize them to other developing parts of the plant; hence it plays a central role for the survival of plants and crop production. During senescence, a range of physiological, morphological, cellular, and molecular events occur, which are generally referred to as the senescence syndrome that includes several hallmarks such as visible yellowing, loss of chlorophyll and water content, increase of ion leakage and cell death, deformation of chloroplast and cell structure, as well as the upregulation of thousands of so-called senescence-associated genes (SAGs) and downregulation of photosynthesis-associated genes (PAGs). This chapter is devoted to methods characterizing the onset and progression of leaf senescence at the morphological, physiological, cellular, and molecular levels. Leaf senescence normally progresses in an age-dependent manner but is also induced prematurely by a variety of environmental stresses in plants. Focused on the hallmarks of the senescence syndrome, a series of protocols is described to asses quantitatively the senescence process caused by developmental cues or environmental perturbations. We first briefly describe the senescence process, the events associated with the senescence syndrome, and the theories and methods to phenotype senescence. Detailed protocols for monitoring senescence in planta and in vitro, using the whole plant and the detached leaf, respectively, are presented. For convenience, most of the protocols use the model plant species Arabidopsis and rice, but they can be easily extended to other plants.
Why choice matters
(2018)
Measures of democracy are in high demand. Scientific and public audiences use them to describe political realities and to substantiate causal claims about those realities. This introduction to the thematic issue reviews the history of democracy measurement since the 1950s. It identifies four development phases of the field, which are characterized by three recurrent topics of debate: (1) what is democracy, (2) what is a good measure of democracy, and (3) do our measurements of democracy register real-world developments? As the answers to those questions have been changing over time, the field of democracy measurement has adapted and reached higher levels of theoretical and methodological sophistication. In effect, the challenges facing contemporary social scientists are not only limited to the challenge of constructing a sound index of democracy. Today, they also need a profound understanding of the differences between various measures of democracy and their implications for empirical applications. The introduction outlines how the contributions to this thematic issue help scholars cope with the recurrent issues of conceptualization, measurement, and application, and concludes by identifying avenues for future research.
An area of increasing interest amongst teachers and researchers is the availability of tools for the design and implementation of literacy interventions with Spanish speaking children. The present systematic literature review contributes to this need by summarizing available findings on evidence-based literacy interventions (EBI) for children from first to third year of primary school. Our results are based on 20 EBI that aimed at improving at least one of the critical components mentioned by the NRP (2000): phonological awareness, phonics, fluency, vocabulary and comprehension. As 90% of the studies were completed with English-speaking children, we critically discussed the applicability of this evidence to the specific context of Spanish-speaking countries. Although many of the general characteristics of the EBI completed with English speaking children could also guide interventions in Spanish, it remains crucial to take into account structural differences between the orthographies of both languages. Moreover, we identified transversal strategies and implementation techniques that due to their universal character could also be useful for early literacy interventions in Spanish. La música como rizoma
(2018)
This introductory essay to the HSR Special Issue “Economists, Politics, and Society” argues for a strong field-theoretical programme inspired by Pierre Bourdieu to research economic life as an integral part of different social forms. Its main aim is threefold. First, we spell out the very distinct Durkheimian legacy in Bourdieu’s thinking and the way he applies it in researching economic phenomena. Without this background, much of what is actually part of how Bourdieu analysed economic aspects of social life would be overlooked or reduced to mere economic sociology. Second, we sketch the main theoretical concepts and heuristics used to analyse economic life from a field perspective. Third, we focus on practical methodological issues of field-analytical research into economic phenomena. We conclude with a short summary of the basic characteristics of this approach and discuss the main insights provided by the contributions to this special issue.
