Parkinson's disease (PD) shows high heterogeneity with regard to the underlying molecular pathogenesis involving multiple pathways and mechanisms. Diagnosis is still challenging and rests entirely on clinical features. Thus, there is an urgent need for robust diagnostic biofluid markers. Untargeted metabolomics allows establishing low-molecular compound biomarkers in a wide range of complex diseases by the measurement of various molecular classes in biofluids such as blood plasma, serum, and cerebrospinal fluid (CSF). Here, we applied untargeted high-resolution mass spectrometry to determine plasma and CSF metabolite profiles. We semiquantitatively determined small-molecule levels (<= 1.5 kDa) in the plasma and CSF from early PD patients (disease duration 0-4 years; n = 80 and 40, respectively), and sex-and age-matched controls (n = 76 and 38, respectively). We performed statistical analyses utilizing partial least square and random forest analysis with a 70/30 training and testing split approach, leading to the identification of 20 promising plasma and 14 CSF metabolites. The semetabolites differentiated the test set with an AUC of 0.8 (plasma) and 0.9 (CSF). Characteristics of the metabolites indicate perturbations in the glycerophospholipid, sphingolipid, and amino acid metabolism in PD, which underscores the high power of metabolomic approaches. Further studies will enable to develop a potential metabolite-based biomarker panel specific for PD
Two experiments examined how individuals respond to a restriction presented within an approach versus an avoidance frame. In Study 1, working on a problem-solving task, participants were initially free to choose their strategy, but for a second task were told to change their strategy. The message to change was embedded in either an approach or avoidance frame. When confronted with an avoidance compared to an approach frame, the participants’ reactance toward the request was greater and, in turn, led to impaired performance. The role of reactance as a response to threat to freedom was explicitly examined in Study 2, in which participants evaluated a potential change in policy affecting their program of study herein explicitly varying whether a restriction was present or absent and whether the message was embedded in an approach versus avoidance frame. When communicated with an avoidance frame and as a restriction, participants showed the highest resistance in terms of reactance, message agreement and evaluation of the communicator. The difference in agreement with the change was mediated by reactance only when a restriction was present. Overall, avoidance goal frames were associated with more resistance to change on different levels of experience (reactance, performance, and person perception). Reactance mediated the effect of goal frame on other outcomes only when a restriction was present.

Phosphorus availability is commonly assumed to limit productivity in lowland tropical rainforests, yet there is relatively little information on the chemical forms of soil phosphorus in such ecosystems. We used the Hedley sequential fractionation scheme to assess phosphorus chemistry in five soils supporting tropical rainforest on Barro Colorado Island, Republic of Panama. The soils represented a range of orders (Inceptisols, Alfisols, and Oxisols) formed on contrasting geological substrates and topography, but under uniform climate and vegetation. Total phosphorus in surface horizons ranged between 315 and 1114 mg P kg(-1), being lowest on a soil derived from marine sediments and highest on soils derived from andesite. The majority of the phosphorus occurred in recalcitrant forms, although between 14% and 39% occurred as organic phosphorus. Readily-available phosphate, as extracted by anion-exchange membranes, occurred in small concentrations (4-13 mg P kg(-1)), although labile phosphorus, defined as phosphate extracted by anion-exchange membrane plus inorganic and organic phosphorus extracted by 0.5 M NaHCO3, accounted for between 4.7% and 11.4% of the total soil phosphorus. Our results indicate a strong control of geology and topography on soil phosphorus in tropical rainforests, which may have important implications for understanding the diversity and distribution of plant species in such ecosystems. Further, some of the most common soils on Barro Colorado Island, including those on the 50 ha forest dynamics plot, are rich in phosphorus despite their relatively advanced stage of pedogenesis.
Shallowly situated evaporites in built-up areas are of relevance for urban and cultural development and hydrological regulation. The hazard of sinkholes, subrosion depressions and gypsum karst is often difficult to evaluate and may quickly change with anthropogenic influence. The geophysical exploration of evaporites in metropolitan areas is often not feasible with active industrial techniques. We collect and combine different passive geophysical data as microgravity, ambient vibrations, deformation and hydrological information to study the roof morphology of shallow evaporites beneath Hamburg, Northern Germany. The application of a novel gravity inversion technique leads to a 3-D depth model of the salt diapir under study. We compare the gravity-based depth model to pseudo-depths from H/V measurements and depth estimates from small-scale seismological array data. While the general range and trend of the diapir roof is consistent, a few anomalous regions are identified where H/V pseudo-depths indicate shallower structures not observed in gravity or array data. These are interpreted by shallow residual caprock floaters and zones of increased porosity. The shallow salt structure clearly correlates with a relative subsidence in the order of 2 mm yr(-1). The combined interpretation of roof morphology, yearly subsidence rates, chemical analyses of groundwater and of hydraulic head in aquifers indicates that the salt diapir beneath Hamburg is subject to significant ongoing dissolution that may possibly affect subrosion depressions, sinkhole distribution and land usage. The combined analysis of passive geophysical data may be exemplary for the study of shallow evaporites beneath other urban areas.
Myriapods (e. g., centipedes and millipedes) display a simple homonomous body plan relative to other arthropods. All members of the class are terrestrial, but they attained terrestriality independently of insects. Myriapoda is the only arthropod class not represented by a sequenced genome. We present an analysis of the genome of the centipede Strigamia maritima. It retains a compact genome that has undergone less gene loss and shuffling than previously sequenced arthropods, and many orthologues of genes conserved from the bilaterian ancestor that have been lost in insects. Our analysis locates many genes in conserved macro-synteny contexts, and many small-scale examples of gene clustering. We describe several examples where S. maritima shows different solutions from insects to similar problems. The insect olfactory receptor gene family is absent from S. maritima, and olfaction in air is likely effected by expansion of other receptor gene families. For some genes S. maritima has evolved paralogues to generate coding sequence diversity, where insects use alternate splicing. This is most striking for the Dscam gene, which in Drosophila generates more than 100,000 alternate splice forms, but in S. maritima is encoded by over 100 paralogues. We see an intriguing linkage between the absence of any known photosensory proteins in a blind organism and the additional absence of canonical circadian clock genes. The phylogenetic position of myriapods allows us to identify where in arthropod phylogeny several particular molecular mechanisms and traits emerged. For example, we conclude that juvenile hormone signalling evolved with the emergence of the exoskeleton in the arthropods and that RR-1 containing cuticle proteins evolved in the lineage leading to Mandibulata. We also identify when various gene expansions and losses occurred. The genome of S. maritima offers us a unique glimpse into the ancestral arthropod genome, while also displaying many adaptations to its specific life history.

The electrophoretic deposition process was used to produce multi-layered ceramics consisting of alternating layers of fully stabilized cubic zirconia and partially stabilized tetragonal zirconia to make use of their different mechanical behaviour, investigating the possibility to deflect advancing cracks at the interfaces of the different layers. This crack deflection is apparently impacted by a toughening mechanism only found in the tetragonal stabilized zirconia polymorph and is characterized by the stress induced transformation of the metastable tetragonal phase into the monoclinic one, which is accompanied by a volume increase resulting in a closing mechanism for advancing cracks. While improving the electrophoretic deposition process, we investigated the transformation toughening mechanism at the layer interfaces and their effect on crack propagation. Investigations involved a combination of different imaging methods, including light microscopy, white light interferometry, atomic force microscopy, scanning electron microscopy, energy dispersive X-ray spectroscopy and Raman spectroscopy.
