For the first time a molecularly imprinted polymer (MIP) with direct electron transfer (DET) and bioelectrocatalytic activity of the target protein is presented. Thin films of MIPs for the recognition of a hexameric tyrosine-coordinated heme protein (HTHP) have been prepared by electropolymerization of scopoletin after oriented assembly of HTHP on a self-assembled monolayer (SAM) of mercaptoundecanoic acid (MUA) on gold electrodes. Cavities which should resemble the shape and size of HTHP were formed by template removal. Rebinding of the target protein sums up the recognition by non-covalent interactions between the protein and the MIP with the electrostatic attraction of the protein by the SAM. HTHP bound to the MIP exhibits quasi-reversible DET which is reflected by a pair of well pronounced redox peaks in the cyclic voltammograms (CVs) with a formal potential of −184.4 ± 13.7 mV vs. Ag/AgCl (1 M KCl) at pH 8.0 and it was able to catalyze the cathodic reduction of peroxide. At saturation the MIP films show a 12-fold higher electroactive surface concentration of HTHP than the non-imprinted polymer (NIP).
This study aimed to determine the relative and absolute reliability of ultrasound (US) measurements of the thickness and echogenicity of the plantar fascia (PF) at different measurement stations along its length using a standardized protocol. Twelve healthy subjects (24 feet) were enrolled. The PF was imaged in the longitudinal plane. Subjects were assessed twice to evaluate the intra-rater reliability. A quantitative evaluation of the thickness and echogenicity of the plantar fascia was performed using Image J, a digital image analysis and viewer software. A sonography evaluation of the thickness and echogenicity of the PF showed a high relative reliability with an Intra class correlation coefficient of 0.88 at all measurement stations. However, the measurement stations for both the PF thickness and echogenicity which showed the highest intraclass correlation coefficient (ICCs) did not have the highest absolute reliability. Compared to other measurement stations, measuring the PF thickness at 3 cm distal and the echogenicity at a region of interest 1 cm to 2 cm distal from its insertion at the medial calcaneal tubercle showed the highest absolute reliability with the least systematic bias and random error. Also, the reliability was higher using a mean of three measurements compared to one measurement. To reduce discrepancies in the interpretation of the thickness and echogenicity measurements of the PF, the absolute reliability of the different measurement stations should be considered in clinical practice and research rather than the relative reliability with the ICC.
Ionogels (IGs) based on poly(methyl methacrylate) (PMMA) and the metal-containing ionic liquids (ILs) bis-1-butyl-3-methlimidazolium tetrachloridocuprate(II), tetrachloride cobaltate(II), and tetrachlorido manganate(II) have been synthesized and their mechanical and electrical properties have been correlated with their microstructure. Unlike many previous examples, the current IGs show a decreasing stability in stress-strain experiments on increasing IL fractions. The conductivities of the current IGs are lower than those observed in similar examples in the literature. Both effects are caused by a two-phase structure with micrometer-sized IL-rich domains homogeneously dispersed an IL-deficient continuous PMMA phase. This study demonstrates that the IL-polymer miscibility and the morphology of the IGs are key parameters to control the (macroscopic) properties of IGs.
Planar bis(1,2-dithiooxalato)nickelate(II), [Ni(dto)]2− reacts in aqueous solutions with lanthanide ions (Ln3+) to form pentanuclear, hetero-bimetallic complexes of the general composition [{Ln(H2O)n}2{Ni(dto)2}3]·xH2O. (n = 4 or 5; x = 9–12). The complex [{Ho(H2O)5}2{Ni(dto)2}3]·10H2O, Ho2Ni3, was synthesized and characterized by single crystal X-ray structure analysis and powder diffraction. The Ho2Ni3 complex crystallizes as monoclinic crystals in the space group P21/c. The channels and cavities, appearing in the crystal packing of the complex molecules, are occupied by a varying amount of non-coordinated water molecules.
Materials based on biodegradable polyesters, such as poly(butylene terephthalate) (PBT) or poly(butylene terephthalate-co-poly(alkylene glycol) terephthalate) (PBTAT), have potential application as pro-regenerative scaffolds for bone tissue engineering. Herein, the preparation of films composed of PBT or PBTAT and an engineered spider silk protein, (eADF4(C16)), that displays multiple carboxylic acid moieties capable of binding calcium ions and facilitating their biomineralization with calcium carbonate or calcium phosphate is reported. Human mesenchymal stem cells cultured on films mineralized with calcium phosphate show enhanced levels of alkaline phosphatase activity suggesting that such composites have potential use for bone tissue engineering.
Meteorological extreme events have great potential for damaging railway infrastructure and posing risks to the safety of train passengers. In the future, climate change will presumably have serious implications on meteorological hazards in the Alpine region. Hence, attaining insights on future frequencies of meteorological extremes with relevance for the railway operation in Austria is required in the context of a comprehensive and sustainable natural hazard management plan of the railway operator. In this study, possible impacts of climate change on the frequencies of so-called critical meteorological conditions (CMCs) between the periods 1961-1990 and 2011-2040 are analyzed. Thresholds for such CMCs have been defined by the railway operator and used in its weather monitoring and early warning system. First, the seasonal climate change signals for air temperature and precipitation in Austria are described on the basis of an ensemble of high-resolution Regional Climate Model (RCM) simulations for Europe. Subsequently, the RCM-ensemble was used to investigate changes in the frequency of CMCs. Finally, the sensitivity of results is analyzed with varying threshold values for the CMCs. Results give robust indications for an all-season air temperature rise, but show no clear tendency in average precipitation. The frequency analyses reveal an increase in intense rainfall events and heat waves, whereas heavy snowfall and cold days are likely to decrease. Furthermore, results indicate that frequencies of CMCs are rather sensitive to changes of thresholds. It thus emphasizes the importance to carefully define, validate, andif neededto adapt the thresholds that are used in the weather monitoring and warning system of the railway operator. For this, continuous and standardized documentation of damaging events and near-misses is a pre-requisite.
Biomimetic binders and catalysts have been generated in order to substitute the biological pendants in separation techniques and bioanalysis. The two major approaches use either "evolution in the test tube" of nucleotides for the preparation of aptamers or total chemical synthesis for molecularly imprinted polymers (MIPs). The reproducible production of aptamers is a clear advantage, whilst the preparation of MIPs typically leads to a population of polymers with different binding sites. The realization of binding sites in the total bulk of the MIPs results in a higher binding capacity, however, on the expense of the accessibility and exchange rate. Furthermore, the readout of the bound analyte is easier for aptamers since the integration of signal generating labels is well established. On the other hand, the overall negative charge of the nucleotides makes aptamers prone to non-specific adsorption of positively charged constituents of the sample and the "biological" degradation of non-modified aptamers and ionic strength-dependent changes of conformation may be challenging in some application.
Human development has far-reaching impacts on the surface of the globe. The transformation of natural land cover occurs in different forms, and urban growth is one of the most eminent transformative processes. We analyze global land cover data and extract cities as defined by maximally connected urban clusters. The analysis of the city size distribution for all cities on the globe confirms Zipf’s law. Moreover, by investigating the percolation properties of the clustering of urban areas we assess the closeness to criticality for various countries. At the critical thresholds, the urban land cover of the countries undergoes a transition from separated clusters to a gigantic component on the country scale. We study the Zipf-exponents as a function of the closeness to percolation and find a systematic dependence, which could be the reason for deviating exponents reported in the literature. Moreover, we investigate the average size of the clusters as a function of the proximity to percolation and find country specific behavior. By relating the standard deviation and the average of cluster sizes—analogous to Taylor’s law—we suggest an alternative way to identify the percolation transition. We calculate spatial correlations of the urban land cover and find long-range correlations. Finally, by relating the areas of cities with population figures we address the global aspect of the allometry of cities, finding an exponent δ ≈ 0.85, i.e., large cities have lower densities.
Continuous exercise (CON) and high-intensity interval exercise (HIIE) can be safely performed with type 1 diabetes mellitus (T1DM). Additionally, continuous glucose monitoring (CGM) systems may serve as a tool to reduce the risk of exercise-induced hypoglycemia. It is unclear if CGM is accurate during CON and HIIE at different mean workloads. Seven T1DM patients performed CON and HIIE at 5% below (L) and above (M) the first lactate turn point (LTP1), and 5% below the second lactate turn point (LTP2) (H) on a cycle ergometer. Glucose was measured via CGM and in capillary blood (BG). Differences were found in comparison of CGM vs. BG in three out of the six tests (p < 0.05). In CON, bias and levels of agreement for L, M, and H were found at: 0.85 (−3.44, 5.15) mmol·L−1, −0.45 (−3.95, 3.05) mmol·L−1, −0.31 (−8.83, 8.20) mmol·L−1 and at 1.17 (−2.06, 4.40) mmol·L−1, 0.11 (−5.79, 6.01) mmol·L−1, 1.48 (−2.60, 5.57) mmol·L−1 in HIIE for the same intensities. Clinically-acceptable results (except for CON H) were found. CGM estimated BG to be clinically acceptable, except for CON H. Additionally, using CGM may increase avoidance of exercise-induced hypoglycemia, but usual BG control should be performed during intense exercise.
Pluvial floods have caused severe damage to urban areas in recent years. With a projected increase in extreme precipitation as well as an ongoing urbanization, pluvial flood damage is expected to increase in the future. Therefore, further insights, especially on the adverse consequences of pluvial floods and their mitigation, are needed. To gain more knowledge, empirical damage data from three different pluvial flood events in Germany were collected through computer-aided telephone interviews. Pluvial flood awareness as well as flood experience were found to be low before the respective flood events. The level of private precaution increased considerably after all events, but is mainly focused on measures that are easy to implement. Lower inundation depths, smaller potential losses as compared with fluvial floods, as well as the fact that pluvial flooding may occur everywhere, are expected to cause a shift in damage mitigation from precaution to emergency response. However, an effective implementation of emergency measures was constrained by a low dissemination of early warnings in the study areas. Further improvements of early warning systems including dissemination as well as a rise in pluvial flood preparedness are important to reduce future pluvial flood damage.
