Reproducibility is a defining feature of science, but the extent to which it characterizes current research is unknown. We conducted replications of 100 experimental and correlational studies published in three psychology journals using high-powered designs and original materials when available. Replication effects were half the magnitude of original effects, representing a substantial decline. Ninety-seven percent of original studies had statistically significant results. Thirty-six percent of replications had statistically significant results; 47% of original effect sizes were in the 95% confidence interval of the replication effect size; 39% of effects were subjectively rated to have replicated the original result; and if no bias in original results is assumed, combining original and replication results left 68% with statistically significant effects. Correlational tests suggest that replication success was better predicted by the strength of original evidence than by characteristics of the original and replication teams.

The RailChain project designed, implemented, and experimentally evaluated a juridical recorder that is based on a distributed consensus protocol. That juridical blockchain recorder has been realized as distributed ledger on board the advanced TrainLab (ICE-TD 605 017) of Deutsche Bahn.
For the project, a consortium consisting of DB Systel, Siemens, Siemens Mobility, the Hasso Plattner Institute for Digital Engineering, Technische Universität Braunschweig, TÜV Rheinland InterTraffic, and Spherity has been formed. These partners not only concentrated competencies in railway operation, computer science, regulation, and approval, but also combined experiences from industry, research from academia, and enthusiasm from startups.
Distributed ledger technologies (DLTs) define distributed databases and express a digital protocol for transactions between business partners without the need for a trusted intermediary. The implementation of a blockchain with real-time requirements for the local network of a railway system (e.g., interlocking or train) allows to log data in the distributed system verifiably in real-time. For this, railway-specific assumptions can be leveraged to make modifications to standard blockchains protocols.
EULYNX and OCORA (Open CCS On-board Reference Architecture) are parts of a future European reference architecture for control command and signalling (CCS, Reference CCS Architecture – RCA). Both architectural concepts outline heterogeneous IT systems with components from multiple manufacturers. Such systems introduce novel challenges for the approved and safety-relevant CCS of railways which were considered neither for road-side nor for on-board systems so far. Logging implementations, such as the common juridical recorder on vehicles, can no longer be realized as a central component of a single manufacturer. All centralized approaches are in question.
The research project RailChain is funded by the mFUND program and gives practical evidence that distributed consensus protocols are a proper means to immutably (for legal purposes) store state information of many system components from multiple manufacturers. The results of RailChain have been published, prototypically implemented, and experimentally evaluated in large-scale field tests on the advanced TrainLab. At the same time, the project showed how RailChain can be integrated into the road-side and on-board architecture given by OCORA and EULYNX.
Logged data can now be analysed sooner and also their trustworthiness is being increased. This enables, e.g., auditable predictive maintenance, because it is ensured that data is authentic and unmodified at any point in time.
Context
For a given body mass index (BMI), both impaired metabolic health (MH) and reduced cardiorespiratory fitness (CRF) associate with increased risk of cardiovascular disease (CVD).
Objective
It remains unknown whether both risk phenotypes relate to CVD independently of each other, and whether these relationships differ in normal weight, overweight, and obese subjects.
Methods
Data from 421 participants from the Tubingen Diabetes Family Study, who had measurements of anthropometrics, metabolic parameters, CRF (maximal aerobic capacity [VO2max]) and carotid intima-media thickness (cIMT), an early marker of atherosclerosis, were analyzed. Subjects were divided by BMI and MH status into 6 phenotypes.
Results
In univariate analyses, older age, increased BMI, and a metabolic risk profile correlated positively, while insulin sensitivity and VO2max negatively with cIMT. In multivariable analyses in obese subjects, older age, male sex, lower VO2max (std. ss -0.21, P = 0.002) and impaired MH (std. ss 0.13, P = 0.02) were independent determinants of increased cIMT. After adjustment for age and sex, subjects with metabolically healthy obesity (MHO) had higher cIMT than subjects with metabolically healthy normal weight (MHNW; 0.59 +/- 0.009 vs 0.52 +/- 0.01 mm; P < 0.05). When VO2max was additionally included in this model, the difference in cIMT between MHO and MHNW groups became statistically nonsignificant (0.58 +/- 0.009 vs 0.56 +/- 0.02 mm; P > 0.05).
Conclusion
These data suggest that impaired MH and low CRF independently determine increased cIMT in obese subjects and that low CRF may explain part of the increased CVD risk observed in MHO compared with MHNW.
The inner region of the Milky Way halo harbors a large amount of dark matter (DM). Given its proximity, it is one of the most promising targets to look for DM. We report on a search for the annihilations of DM particles using gamma-ray observations towards the inner 300 pc of the Milky Way, with the H.E.S.S. array of ground-based Cherenkov telescopes. The analysis is based on a 2D maximum likelihood method using Galactic Center (GC) data accumulated by H.E.S.S. over the last 10 years (2004-2014), and does not show any significant gamma-ray signal above background. Assuming Einasto and Navarro-Frenk-White DM density profiles at the GC, we derive upper limits on the annihilation cross section <sigma nu >. These constraints are the strongest obtained so far in the TeV DM mass range and improve upon previous limits by a factor 5. For the Einasto profile, the constraints reach <sigma nu > values of 6 x 10(-26) cm(3) s(-1) in the W+W- channel for a DM particle mass of 1.5 TeV, and 2 x 10(-26) cm(3) s(-1) in the tau(+)tau(-) channel for a 1 TeV mass. For the first time, ground-based gamma-ray observations have reached sufficient sensitivity to probe <sigma nu > values expected from the thermal relic density for TeV DM particles.

Die Schule ist Ort des Lebens und Lernens sowie der Tätigkeit von Kindern und Jugendlichen. Sie ist dem Schutz der seelischen und körperlichen Unversehrtheit, der geistigen Freiheit und der Entfaltungsmöglichkeiten aller Schülerinnen und Schüler verpflichtet. Dieser Auftrag ist tägliche Herausforderung, aber leider nicht tägliche Lebenswirklichkeit. Vor allem betrifft dies Jugendliche, die sich zum eigenen Geschlecht hingezogen fühlen oder aufgrund ihrer Identität als Inter- und Transsexuelle sowie Transgender in Konflikt mit den traditionellen Geschlechterrollen und den damit verbundenen gesellschaftlich akzeptierten Lebensweisen geraten. Mit der vorliegenden Handreichung soll dazu beigetragen werden, dass aus dem Brandenburger Verfassungsanspruch und dem Handlungsauftrag des Schulgesetzes für alle Schülerinnen und Schüler – auch lesbische, schwule, bisexuelle, intersexuelle, transsexuelle und transgender – eine Schulalltagsrealität wird. Die vorliegende Publikation richtet sich an Pädagoginnen und Pädagogen und enthält sowohl theoretische Einführungen als auch konkrete Unterrichtsvorschläge und Hinweise für weitere Informationsmöglichkeiten.
Reflexion ist eine Schlüsselkategorie für die professionelle Entwicklung von Lehrkräften, welche als Ausbildungsziel in den Bildungsstandards für die Lehrkräftebildung verankert ist. Eine Verstetigung universitär geprägter Forschung und Modellierung in der praxisnahen Anwendung im schulischen Kontext bietet Potentiale nachhaltiger Professionalisierung. Die Stärkung reflexionsbezogener Kompetenzen durch Empirie und Anwendung scheint eine phasenübergreifende Herausforderung der Lehrkräftebildung zu sein, die es zu bewältigen gilt. Ziele des Tagungsbandes Reflexion in der Lehrkräftebildung sind eine theoretische Schärfung des Konzeptes „Reflexive Professionalisierung“ und der Austausch über Fragen der Einbettung wirksamer reflexionsbezogener Lerngelegenheiten in die Lehrkräftebildung. Forschende und Lehrende der‚ drei Phasen (Studium, Referendariat sowie Fort- und Weiterbildung) der Lehrkräftebildung stellen Lehrkonzepte und Forschungsprojekte zum Thema Reflexion in der Lehrkräftebildung vor und diskutieren diese. Gemeinsam mit Teilnehmenden aller Phasen und von verschiedenen Standorten der Lehrkräftebildung werden zukünftige Herausforderungen identifiziert und Lösungsansätze herausgearbeitet.
Reduced expression of the Indy ("I am Not Dead, Yet") gene in lower organisms promotes longevity in a manner akin to caloric restriction. Deletion of the mammalian homolog of Indy (mIndy, Slc13a5) encoding for a plasma membrane-associated citrate transporter expressed highly in the liver, protects mice from high-fat diet-induced and aging-induced obesity and hepatic fat accumulation through a mechanism resembling caloric restriction. We studied a possible role of mIndy in human hepatic fat metabolism. In obese, insulin-resistant patients with nonalcoholic fatty liver disease, hepatic mIndy expression was increased and mIndy expression was also independently associated with hepatic steatosis. In nonhuman primates, a 2-year high-fat, high-sucrose diet increased hepatic mIndy expression. Liver microarray analysis showed that high mIndy expression was associated with pathways involved in hepatic lipid metabolism and immunological processes. Interleukin-6 (IL-6) was identified as a regulator of mIndy by binding to its cognate receptor. Studies in human primary hepatocytes confirmed that IL-6 markedly induced mIndy transcription through the IL-6 receptor and activation of the transcription factor signal transducer and activator of transcription 3, and a putative start site of the human mIndy promoter was determined. Activation of the IL-6-signal transducer and activator of transcription 3 pathway stimulated mIndy expression, enhanced cytoplasmic citrate influx, and augmented hepatic lipogenesis in vivo. In contrast, deletion of mIndy completely prevented the stimulating effect of IL-6 on citrate uptake and reduced hepatic lipogenesis. These data show that mIndy is increased in liver of obese humans and nonhuman primates with NALFD. Moreover, our data identify mIndy as a target gene of IL-6 and determine novel functions of IL-6 through mINDY. Conclusion: Targeting human mINDY may have therapeutic potential in obese patients with nonalcoholic fatty liver disease. German Clinical Trials Register: DRKS00005450.

Over the past years, NGS has become a crucial workhorse for open-view pathogen diagnostics.
Yet, long turnaround times result from using massively parallel high-throughput technologies as the analysis can only be performed after sequencing has finished. The interpretation of results can further be challenged by contaminations, clinically irrelevant sequences, and the sheer amount and complexity of the data.
We implemented PathoLive, a real-time diagnostics pipeline for the detection of pathogens from clinical samples hours before sequencing has finished.
Based on real-time alignment with HiLive2, mappings are scored with respect to common contaminations, low-entropy areas, and sequences of widespread, non-pathogenic organisms.
The results are visualized using an interactive taxonomic tree that provides an easily interpretable overview of the relevance of hits. For a human plasma sample that was spiked in vitro with six pathogenic viruses, all agents were clearly detected after only 40 of 200 sequencing cycles.
For a real-world sample from Sudan, the results correctly indicated the presence of Crimean-Congo hemorrhagic fever virus. In a second real-world dataset from the 2019 SARS-CoV-2 outbreak in Wuhan, we found the presence of a SARS coronavirus as the most relevant hit without the novel virus reference genome being included in the database.
For all samples, clinically irrelevant hits were correctly de-emphasized.
Our approach is valuable to obtain fast and accurate NGS-based pathogen identifications and correctly prioritize and visualize them based on their clinical significance: PathoLive is open source and available on GitLab and BioConda.

Design and Implementation of service-oriented architectures imposes a huge number of research questions from the fields of software engineering, system analysis and modeling, adaptability, and application integration. Component orientation and web services are two approaches for design and realization of complex web-based system. Both approaches allow for dynamic application adaptation as well as integration of enterprise application.
Commonly used technologies, such as J2EE and .NET, form de facto standards for the realization of complex distributed systems. Evolution of component systems has lead to web services and service-based architectures. This has been manifested in a multitude of industry standards and initiatives such as XML, WSDL UDDI, SOAP, etc. All these achievements lead to a new and promising paradigm in IT systems engineering which proposes to design complex software solutions as collaboration of contractually defined software services.
Service-Oriented Systems Engineering represents a symbiosis of best practices in object-orientation, component-based development, distributed computing, and business process management. It provides integration of business and IT concerns.
The annual Ph.D. Retreat of the Research School provides each member the opportunity to present his/her current state of their research and to give an outline of a prospective Ph.D. thesis. Due to the interdisciplinary structure of the research school, this technical report covers a wide range of topics. Proceedings of KogWis 2010 : 10th Biannual Meeting of the German Society for Cognitive Science

As the latest biannual meeting of the German Society for Cognitive Science (Gesellschaft für Kognitionswissenschaft, GK), KogWis 2010 at Potsdam University reflects the current trends in a fascinating domain of research concerned with human and artificial cognition and the interaction of mind and brain. The Plenary talks provide a venue for questions of the numerical capacities and human arithmetic (Brian Butterworth), of the theoretical development of cognitive architectures and intelligent virtual agents (Pat Langley), of categorizations induced by linguistic constructions (Claudia Maienborn), and of a cross-level account of the “Self as a complex system“ (Paul Thagard). KogWis 2010 integrates a wealth of experimental research, cognitive modelling, and conceptual analysis in 5 invited symposia, over 150 individual talks, 6 symposia, and more than 40 poster contributions. Some of the invited symposia reflect local and regional strenghts of research in the Berlin-Brandenburg area: the two largests research fields of the university Cognitive Sciences Area of Excellence in Potsdam are represented by an invited symposium on “Information Structure” by the Special Research Area 632 (“Sonderforschungsbereich”, SFB) of the same name, of Potsdam University and Humboldt-University Berlin, and by a satellite conference of the research group “Mind and Brain Dynamics”. The Berlin School of Mind and Brain at Humboldt-University Berlin takes part with an invited symposium on “Decision Making” from a perspective of cognitive neuroscience and philosophy and the DFG Cluster of Excellence “Languages of Emotion” of Free University presents interdisciplinary research results in an invited symposium on “Symbolising Emotions”.
(40)A/Ar-39 step-heating of mica and amphibole megacrysts from hauyne-bearing olivine melilitite scoria/tephra from the Zelezna hurka yielded a 435 +/- 108 ka isotope correlation age for phlogopite and a more imprecise 1.55 Ma total gas age of the kaersutite megacryst. The amphibole megacrysts may constitute the first, and the younger phlogopite megacrysts the later phase of mafic, hydrous melilitic magma crystallization. It cannot be ruled out that the amphibole megacrysts are petrogenetically unrelated to tephra and phlogopite megacrysts and were derived from mantle xenoliths or disaggregated older, deep crustal pegmatites. This is in line both with the rarity of amphibole at Zelezna hurka and with the observed signs of magmatic resorption at the edges of amphibole crystals.
We present density functional theory modeling of time-resolved optical pump/X-ray spectroscopic probe data of CO desorption from Ru(0001). The BEEF van der Waals functional predicts a weakly bound state as a precursor to desorption. The optical pump leads to a near-instantaneous (<100 fs) increase of the electronic temperature to nearly 7000 K. The temperature evolution and energy transfer between electrons, substrate phonons and adsorbate is described by the two-temperature model and found to equilibrate on a timescale of a few picoseconds to an elevated local temperature of similar to 2000K. Estimating the free energy based on the computed potential of mean force along the desorption path, we find an entropic barrier to desorption (and by time-reversal also to adsorption). This entropic barrier separates the chemisorbed and precursor states, and becomes significant at the elevated temperature of the experiment (similar to 1.4 eV at 2000 K). Experimental pump-probe X-ray absorption/X-ray emission spectroscopy indicates population of a precursor state to desorption upon laser-excitation of the system (Dell'Angela et al., 2013). Computing spectra along the desorption path confirms the picture of a weakly bound transient state arising from ultrafast heating of the metal substrate. (C) 2015 Elsevier B.V. All rights reserved.
Although temporal heterogeneity is a well-accepted driver of biodiversity, effects of interannual variation in land-use intensity (LUI) have not been addressed yet. Additionally, responses to land use can differ greatly among different organisms; therefore, overall effects of land-use on total local biodiversity are hardly known. To test for effects of LUI (quantified as the combined intensity of fertilization, grazing, and mowing) and interannual variation in LUI (SD in LUI across time), we introduce a unique measure of whole-ecosystem biodiversity, multidiversity. This synthesizes individual diversity measures across up to 49 taxonomic groups of plants, animals, fungi, and bacteria from 150 grasslands. Multidiversity declined with increasing LUI among grasslands, particularly for rarer species and aboveground organisms, whereas common species and belowground groups were less sensitive. However, a high level of interannual variation in LUI increased overall multidiversity at low LUI and was even more beneficial for rarer species because it slowed the rate at which the multidiversity of rare species declined with increasing LUI. In more intensively managed grasslands, the diversity of rarer species was, on average, 18% of the maximum diversity across all grasslands when LUI was static over time but increased to 31% of the maximum when LUI changed maximally over time. In addition to decreasing overall LUI, we suggest varying LUI across years as a complementary strategy to promote biodiversity conservation.
Early maternal care may counteract familial liability for psychopathology in the reward circuitry

Reward processing is altered in various psychopathologies and has been shown to be susceptible to genetic and environmental influences. Here, we examined whether maternal care may buffer familial risk for psychiatric disorders in terms of reward processing. Functional magnetic resonance imaging during a monetary incentive delay task was acquired in participants of an epidemiological cohort study followed since birth (N = 172, 25 years). Early maternal stimulation was assessed during a standardized nursing/playing setting at the age of 3 months. Parental psychiatric disorders (familial risk) during childhood and the participants’ previous psychopathology were assessed by diagnostic interview. With high familial risk, higher maternal stimulation was related to increasing activation in the caudate head, the supplementary motor area, the cingulum and the middle frontal gyrus during reward anticipation, with the opposite pattern found in individuals with no familial risk. In contrast, higher maternal stimulation was associated with decreasing caudate head activity during reward delivery and reduced levels of attention deficit hyperactivity disorder (ADHD) in the high-risk group. Decreased caudate head activity during reward anticipation and increased activity during delivery were linked to ADHD. These findings provide evidence of a long-term association of early maternal stimulation on both adult neurobiological systems of reward underlying externalizing behavior and ADHD during development.
Questions of identity and provenance of minerals that are parts of masterpieces in museums have become increasingly important in mineralogical and historical studies. Detailed investigations of valuable and unique objects require on-site, nondestructive and noninvasive methods because touching or removing them may cause irreparable damage. A mobile Raman-microprobe has been used to meet these demands for truly in situ mineralogical studies of the large collection of minerals and rocks of the Prussian kings in the Grotto Hall (Grottensaal) of the New Palace (Neues Palais), Park Sanssouci in Potsdam. Minerals on the walls of the Grotto Hall were analyzed to identify them and thereby to complete the data bank of the collection. Fluid and solid inclusions in the interior of a large quartz crystal have been studied to provide evidence of the provenance of the crystal. The fluid inclusions contain aqueous saline solutions, whereas the solid inclusions are needles of anhydrite with a length of about 1.5 mm. The quartz probably originated from an area in the eastern Alps, from the surroundings of Bad Gastein, Austria. This is the first on-site and in situ study of inclusions below the surface of a mineral with a mobile Raman-microprobe outside a laboratory.
Association between pubertal stage at first drink and neural reward processing in early adulthood

Puberty is a critical time period during human development. It is characterized by high levels of risk-taking behavior, such as increased alcohol consumption, and is accompanied by various neurobiological changes. Recent studies in animals and humans have revealed that the pubertal stage at first drink (PSFD) significantly impacts drinking behavior in adulthood. Moreover, neuronal alterations of the dopaminergic reward system have been associated with alcohol abuse or addiction. This study aimed to clarify the impact of PSFD on neuronal characteristics of reward processing linked to alcohol-related problems. One hundred sixty-eight healthy young adults from a prospective study covering 25 years participated in a monetary incentive delay task measured with simultaneous EEG-fMRI. PSFD was determined according to the age at menarche or Tanner stage of pubertal development, respectively. Alcohol-related problems in early adulthood were assessed with the Alcohol Use Disorder Identification Test (AUDIT). During reward anticipation, decreased fMRI activation of the frontal cortex and increased preparatory EEG activity (contingent negative variation) occurred with pubertal compared to postpubertal first alcohol intake. Moreover, alcohol-related problems during early adulthood were increased in pubertal compared to postpubertal beginners, which was mediated by neuronal activation of the right medial frontal gyrus. At reward delivery, increased fMRI activation of the left caudate and higher feedback-related EEG negativity were detected in pubertal compared to postpubertal beginners. Together with animal findings, these results implicate PSFD as a potential modulator of psychopathology, involving altered reward anticipation. Both PSFD timing and reward processing might thus be potential targets for early prevention and intervention.
Microorganisms are used for the production of foodstuffs since thousands of years. By now, these biotechnological processes are not restricted to some of the known microorganisms. The possibility to produce enzymes independently from their host organisms extended their range of application. Since then, exploration of new bioresources leads to novel enzymes and peptides applicable for a diverse set of food production processes: peptidases of grain pest beetles are able to hydrolyse gluten and antimicrobial active peptides of insects maybe of use for preservation of food. Examples of our own work depict strategies to identify novel biocatalysts for food biotechnology.
Integral-field spectrophotometry of the quadruple QSO HE 0435-1223 : Evidence for microlensing

We present the first spatially resolved spectroscopic observations of the recently discovered quadruple QSO and gravitational lens HE 0435-1223. Using the Potsdam Multi-Aperture Spectrophotometer (PMAS), we show that all four QSO components have very similar but not identical spectra. In particular, the spectral slopes of components A, B, and D are indistinguishable, implying that extinction due to dust plays no major role in the lensing galaxy. While also the emission line profiles are identical within the error bars, as expected from lensing, the equivalent widths show significant differences between components. Most likely, microlensing is responsible for this phenomenon. This is also consistent with the fact that component D, which shows the highest relative continuum level, has brightened by 0.07 mag since Dec. 2001. We find that the emission line flux ratios between the components are in better agreement with simple lens models than broad band or continuum measurements, but that the discrepancies between model and data are still unacceptably large. Finally, we present a detection of the lensing galaxy, although this is close to the limits of the data. Comparing with a model galaxy spectrum, we obtain a redshift estimate of zlens=0.44+/- 0.02.
We present spatially resolved spectrophotometric observations of multiply imaged QSOs, using the Potsdam Multi- Aperture Spectrophotometer (PMAS), with the intention to search for spectral differences between components indicative of either microlensing or dust extinction. For the quadruple QSO HE 0435-1223 we find that the continuum shapes are indistinguishable, therefore differential extinction is negligible. The equivalent widths of the broad emission lines are however significantly different, and we argue that this is most likely due to microlensing. Contrariwise, the two components of the well-known object UM 673 have virtually identical emission line properties, but the continuum slopes differ significantly and indicate different dust extinction along both lines of sight
The Chelyabinsk meteorite

We present results of petrographic, mineralogical, and chemical investigations of three Chelyabinsk meteorite fragments. Three distinct lithologies were identified: light S3LL5, dark S4-S5LL5 material, and opaque fine-grained former impact melt. Olivine-spinel thermometry revealed an equilibration temperature of 703 +/- 23 degrees C for the light lithology. All plagioclase seems to be secondary, showing neither shock-induced fractures nor sulfide-metal veinlets. Feldspathic glass can be observed showing features of extensive melting and, in the dark lithology, as maskelynite, lacking melt features and retaining grain boundaries of former plagioclase. Olivine of the dark lithology shows planar deformation features. Impact melt is dominated by Mg-rich olivine and resembles whole-rock melt. Melt veins (<2mm) are connected to narrower veinlets. Melt vein textures are similar to pegmatite textures showing chilled margins, a zone of inward-grown elongated crystals and central vugs, suggesting crystallization from supercooled melt. Sulfide-metal droplets indicate liquid immiscibility of both silicate and sulfide as well as sulfide and metal melts. Impact melting may have been an important factor for differentiation of primitive planetary bodies. Graphite associated with micrometer-sized melt inclusions in primary olivine was detected by Raman mapping. Carbon isotopic studies of graphite could be applied to test a possible presolar origin.
The pressures required for diamond and coesite formation far exceed conditions reached by even the deepest present-day orogenic crustal roots. Therefore the occurrence of metamorphosed continental crust containing these minerals requires processes other than crustal thickening to have operated in the past. Here we report the first in situ finding of diamond and coesite, characterized by micro-Raman spectroscopy, in high-pressure granulites otherwise indistinguishable from granulites found associated with garnet peridotite throughout the European Variscides. Our discovery confirms the provenance of Europe's first reliable diamond, the "Bohemian diamond," found in A.D. 1870, and also represents the first robust evidence for ultrahigh-pressure conditions in a major Variscan crustal rock type. A process of deep continental subduction is required to explain the metamorphic pressures and the granulite-garnet peridotite association, and thus tectonometamorphic models for these rocks involving a deep orogenic crustal root need to be significantly modified.
The 10th edition of the International Congress on the Application of Raman Spectroscopy in Art and Archaeology (RAA2019) was held in Potsdam (Germany) from 3 to 7 September 2019, with eight keynote lectures, 35 oral presentations and 18 Poster Presentations. The number of active participants was 68 delegates from 20 countries among the 236 authors that presented at least one work.
Three diatomite beds exposed in the Ol Njorowa Gorge south of Lake Naivasha, Central Kenya Rift, document three major lake-level highstands between 175 and 60 kyr BP. Diatom transfer-function estimates of hydrological and hydrochemical parameters suggest that a deep and large freshwater lake existed during the highstands at 135 and 80 kyr BP. In contrast, a shallower but more expanded freshwater lake existed at 110 kyr BP. The best analog for the most extreme highstand at 135 kyr BP is the highstand during the Early Holocene humid period from 10 to 6 kyr BP. The environmental conditions as reconstructed from diatom assemblages suggest long-lasting episodes of increased humidity during the high lake periods. This contrasts to the modern situation with a relatively shallow Lake Naivasha characterized by rapid water level fluctuations within a few decades. The most likely cause for the variable hydrological conditions since 175 kyr BP is orbitally driven insolation changes on the equator and increased lateral moisture transport from the ocean.
The structure and alterations of subducted oceanic lithosphere ( e. g., thickness and seismic velocity of oceanic crust) can be obtained by analyzing guided seismic waves generated by earthquakes within the slab (Wadati- Benioff zone). In northern Chile prominent secondary phases from intermediate-depth seismicity, observed in the forearc region can be interpreted as guided waves. For the observation of guided waves it is usually required to have stations close to the wave guide, a fact which is not directly given for forearc stations in subduction zone environments. With the help of finite difference simulations we model the decoupling mechanism of guided waves at the contact between the descending oceanic plate and the upper plate crust where the wave guide is opened due to the equalization of seismic velocities. Provided that suited stations are available, this mechanism allows for the use of intermediate depth seismicity to study the shallow subduction zone structure ( <= 100 km depth)
Thermobarometrical and mineral-chemical investigations by electron microprobe and LA-ICP-MS on a sillimanite- bearing pegmatoid from the Reinbolt Hills provide important constraints on the P-T-X-age relations of part of East Antarctica during Pan-African tectonism. U-Th-total Pb ages of monazite imply that the pegmatoid of originally Grenvillan age (zircon U-Pb age of ca. 900 Ma) underwent a major, late Pan-African (Cambrian) regional, granulite-facies metamorphism between 500 and 550 Ma. Most of the monazite formed during this event, as result of apatite metasomatism owing to infiltration of high-grade metamorphic fluids. Apatite-biotite and other mineral thermobarometers define the peak metamorphic temperatures and pressures with 850-950 degrees C and 0.8-1.0 GPa. The F-Cl-OH relations in apatite, and biotite, the chemistry of fluid inclusions and the presence of K-feldspar microveins suggest that the metasomatising fluid was a CO2-bearing, diluted KCl brine. The pegmatoid is the first record of monazite-(Ce) formed from fluorapatite that is rich in U (up to 2.6 Wt% UO2) and possesses Th/U ratios <1 (0.09 on average). These chemical signatures are direct reflection of the U and Th concentration patterns in the parental fluorapatite

Die vorliegende Publikation fällt ein wenig aus dem Rahmen der Reihe „Sorben (Wenden) - Eine Brandenburger Minderheit und ihre Thematisierung im Unterricht“. Im Gegensatz zu den anderen Teilen widmet sie sich nur einem Gegenstand - dem Stoff der Krabat-Sagen. Damit richtet sie sich hauptsächlich an Lehrkräfte im Fach Deutsch, wobei auch fachübergreifende und Fächer verbindende Aspekte berücksichtigt werden. Die Krabat-Sage zählt vor allem in der Bearbeitung von Preußler zu den bekanntesten sorbischen Stoffen. Diese sorbischen Wurzeln werden allerdings nur selten thematisiert. Daher sind viele der vorliegenden Betrachtungen zu ausgewählten Aspekten auch als Anregungen zu verstehen, unter welchen Gesichtspunkten Krabat behandelt oder gar neu interpretiert und weiterentwickelt werden könnte. Diese Handreichung ist so konzipiert, dass sie je nach Interesse ausschnittweise gelesen werden kann: Auf einen Überblick über verschiedene Krabat-Bearbeitungen folgen Betrachtungen sorbischer Aspekte als auch Möglichkeiten einer Thematisierung über den Deutsch-Unterricht hinaus. Dabei wird auf Möglichkeiten einer Exkursion in die historische Krabat-Region in der Lausitz eingegangen. Es folgen Texte zu ausgewählten Einzelaspekten. Zudem enthält diese Publikation eine Zusammenstellung von verschiedenen Krabat-Materialien und Hinweise auf Unterrichtsprojekte zur Anregung, weiteren Vertiefung bzw. für den eigenen Unterrichtseinsatz.
The rigorous development, application and validation of distributed hydrological models obligates to evaluate data in a spatially distributed way. In particular, spatial model predictions such as the distribution of soil moisture, runoff generating areas or nutrient-contributing areas or erosion rates, are to be assessed against spatially distributed observations. Also model inputs, such as the distribution of modelling units derived by GIS and remote sensing analyses, should be evaluated against groundbased observations of landscape characteristics. So far, however, quantitative methods of spatial field comparison have rarely been used in hydrology. In this paper, we present algorithms that allow to compare observed and simulated spatial hydrological data. The methods can be applied for binary and categorical data on regular grids. They comprise cell-by-cell algorithms, cell-neighbourhood approaches that account for fuzziness of location, and multi-scale algorithms that evaluate the similarity of spatial fields with changing resolution. All methods provide a quantitative measure of the similarity of two maps. The comparison methods are applied in two mountainous catchments in southern Germany (Brugga, 40 km<sup>2) and Austria (Löhnersbach, 16 km<sup>2). As an example of binary hydrological data, the distribution of saturated areas is analyzed in both catchments. For categorical data, vegetation zones that are associated with different runoff generation mechanisms are analyzed in the Löhnersbach. Mapped spatial patterns are compared to simulated patterns from terrain index calculations and from satellite image analysis. It is discussed how particular features of visual similarity between the spatial fields are captured by the quantitative measures, leading to recommendations on suitable algorithms in the context of evaluating distributed hydrological models.

From the acetone extract of the roots of Derris trifoliata an isollavonoid derivative, named 7a-O- methyldeguelol, a modified rotenoid with an open ring-C, representing a new sub-class of isollavonoids (the sub-class is here named as rotenoloid), was isolated and characterised. In addition, the known rotenoids, rotenone, deguelin and alpha-toxicarol, were identified. The structures were determined on the basis of spectroscopic evidence. Gilbert et al. conclude that evidence from the Open Science Collaboration’s Reproducibility Project: Psychology indicates high reproducibility, given the study methodology. Their very optimistic assessment is limited by statistical misconceptions and by causal inferences from selectively interpreted, correlational data. Using the Reproducibility Project: Psychology data, both optimistic and pessimistic conclusions about reproducibility are possible, and neither are yet warranted.
Lithium is an important component of some tourmalines, especially in chemically evolved granites and pegmatites. All attempts at synthesizing Li-rich tourmaline have so far been unsuccessful. Here we describe the first synthesis of rossmanitic tourmaline at 4 GPa and 700 degrees C in the system Li2OAl2O3SiO2B2O3H2O (LASBH) from seed-free solid starting materials consisting of a homogenous mixture of Li2O, gamma-Al2O3, quartz and H3BO3. The solid run products after 12-day run duration comprise rossmanitic tourmaline (68 wt%), dumortierite (28 wt%) and traces of spodumene (3 wt%) and coesite (1 wt%). Tourmaline forms idiomorphic, large prismatic crystals (30 X 100 mu m), which are inclusion free and chemically unzoned. The refined cell dimensions of the tourmaline are: a = 15.7396(9) angstrom, c = 7.0575(5) angstrom, V = 1514.1(2) angstrom 3. Conventionally, the Li+ ion is assumed to exclusively occupy the octahedral Y site in the tourmaline structure to a maximum of 2 Li per formula unit (pfu). However, the chemical composition of our synthetic tourmaline determined by electron microprobe and secondary ion mass spectroscopy results in the formula: (X)(square Li-0.67(11)(0.33(11)))(Y)(Al2.53(10)Li0.47(10))(Z)(Al-6)T(Si5.42(15)B0.58(15))O-18(B)(BO3)(3)(V+W)[(OH)(2.40(3))O-1.60(3)], wherein a significant amount of Li occupies the X site for charge balance requirements. Reliable assignment of the OH-stretching vibrations in a polarized single-crystal Raman spectrum such as a single-crystal XRD structure refinement, confirms the incorporation of Li at the X site [0.24(9) and 0.15(5) Li-X pfu, respectively]. The SREF data show that the LiO1 distances are shortened significantly in order to compensate for the smaller ionic radius of Li+ compared to Na+, K+ or Ca2+ at the X site, i.e., Li is closer to the Si6O18 ring and to a sevenfold coordination with oxygen.

Granitic melt inclusions were found in layers of garnet clinopyroxenites from orogenic peridotites hosted in high-pressure felsic granulites of the Granulitgebirge, central Europe. The inclusions are both glassy and crystallized, and occur as clusters in the garnet. Microstructural features suggest that the inclusions formed while garnet was growing as a peritectic phase, likely alongside clinopyroxene. The chemistry of the melt, in particular its trace element signature, shows a crustal contribution, probably due to the involvement of phengite in the melt-producing reaction, most likely in the presence of a fluid. The presence of a granitoid melt in mantle rocks may be the result of localized melting of a phengite-bearing protolith either already present in the peridotites or, more likely, within the local deeply subducted crustal units. In the latter case, the melt would have infiltrated the peridotites and generated pyroxenite via metasomatism. In either case, the presence of granitoid inclusions in orogenic peridotite provides direct evidence for a genetic connection between a high-pressure crustal melt and garnet pyroxenites. The in situ characterization of these remnants of natural melt provides direct quantitative constraints on (one of) the agents responsible for the interaction between crust and mantle.
Raman spectra of gas hydrates : differences and analogies to ice 1h and (gas saturated) water

It is generally accepted that Raman spectroscopic investigations of gas hydrates provide vital information regarding the structure of the hydrate, hydrate composition and cage occupancies, but most research is focused on the vibrational spectra of the guest molecules. We show that the shape and position of the Raman signals of the host molecules (H2O) also contain useful additional information. In this study, Raman spectra (200-4000cm(-1)) of (mixed) gas hydrates with variable compositions and different structures are presented. The bands in the O-H stretching region (3000- 3800cm(-1)), the O-H bending region (1600-1700cm(-1)) and the O-O hydrogen bonded stretching region (100-400cm(-1)) are compared with the corresponding bands in Raman spectra of ice Ih and liquid water. The interpretation of the differences and similarities with respect to the crystal structure and possible interactions between guest and host molecules are presented. (c) 2005 Elsevier B.V. Cell-to-cell diversity in a synchronized chlamydomonas culture as revealed by single-cell analyses

In a synchronized photoautotrophic culture of Chlamydomonas reinhardtii, cell size, cell number, and the averaged starch content were determined throughout the light-dark cycle. For single-cell analyses, the relative cellular starch was quantified by measuring the second harmonic generation (SHG). In destained cells, amylopectin essentially represents the only biophotonic structure. As revealed by various validation procedures, SHG signal intensities are a reliable relative measure of the cellular starch content. During photosynthesis-driven starch biosynthesis, synchronized Chlamydomonas cells possess an unexpected cell-to-cell diversity both in size and starch content, but the starch-related heterogeneity largely exceeds that of size. The cellular volume, starch content, and amount of starch/cell volume obey lognormal distributions. Starch degradation was initiated by inhibiting the photosynthetic electron transport in illuminated cells or by darkening. Under both conditions, the averaged rate of starch degradation is almost constant, but it is higher in illuminated than in darkened cells. At the single-cell level, rates of starch degradation largely differ but are unrelated to the initial cellular starch content. A rate equation describing the cellular starch degradation
Second harmonic generation (SHG) microscopy is employed to study changes in crystalline organization due to altered gene expression and hydration in barley starch granules. SHG intensity and susceptibility ratio values (R'(SHG)) are obtained using reduced Stokes-Mueller polarimetric microscopy. The maximum R'(SHG) values occur at moderate moisture indicating the narrowest orientation distribution of nonlinear dipoles from the cylindrical axis of glucan helices. The maximum SHG intensity occurs at the highest moisture and amylopectin content. These results support the hypothesis that SHG is caused by ordered hydrogen and hydroxyl bond networks which increase with hydration of starch granules. (C) 2015 Optical Society of America
Research collaboration promises a useful approach to bridging the gap between research and practice and thus promoting evidence-informed education. This study examines whether information on research collaboration can influence the reception of research knowledge. We assume that the composition of experts from the field and scientists in a research team sends out signals that influence trust in as well as the relevance and applicability of the finding. In a survey experiment with practitioners from the field of adult education the influence of different research team compositions around an identical finding is tested. The results show overall high trust, relevance and applicability ratings with regard to the finding, regardless of the composition of the research team. We discuss the potential importance of additional information about research collaborations for effective knowledge translation and point out the need for more empirical research.
Mit dem vorliegenden Band „Nach Bologna: Praktika im Studium – Pflicht oder Kür? Empirische Analysen und Empfehlungen für die Hochschulpraxis“ von Wilfried Schubarth, Karsten Speck und Andreas Seidel wird die Reihe „Potsdamer Beiträge zur Lehrevaluation“ unter neuem Titel und veränderter inhaltlicher Schwerpunktsetzung fortgeführt. Die Umbenennung in „Potsdamer Beiträge zur Hochschulforschung“ versteht sich als ein Schritt hin zu einer thematischen Öffnung der Reihe für die verschiedensten Felder der Hochschulforschung. Der vorliegende Band widmet sich einem der zentralen Reformziele von Bologna: der Frage des Praxis- und Berufsbezugs und dabei insbesondere den Praxisphasen im Studium. Mit der Bologna-Reform werden im bildungspolitischen Bereich sehr vielfältige strukturelle und inhaltliche Ziele verfolgt. Das Ziel dieses Bandes besteht deshalb darin, empirische Forschungen zu Praxisbezügen und Praxisphasen im Studium vorzustellen, diese in den Kontext aktueller Debatten um Studienqualität und Studienreform zu stellen sowie Folgerungen für die Gestaltung von Praxisphasen abzuleiten. Inhaltliche Schwerpunkte bilden das BMBF-Forschungsprojekt ProPrax und die Praxisphasen im Lehramtsstudium. Die Beiträge dieses Bandes gehen aus einem gleichnamigen Workshop hervor, der am 1. Oktober 2010 in Potsdam stattfand.
There are a plethora of ways to guide and support people to learn about MOOC (massive open online course) development, from their first interest, sourcing supportive resources, methods and tools to better aid their understanding of the concepts and pedagogical approaches of MOOC design, to becoming a MOOC developer. This contribution highlights tools and methods that are openly available and re-usable under Creative Commons licenses. Our collection builds upon the experiences from three MOOC development and hosting teams with joint experiences of several hundred MOOCs (University of Applied Sciences in Lübeck, Graz University of Technology, University of Glasgow) in three European countries, which are Germany, Austria and the UK. The contribution recommends and shares experiences with short articles and poster for first information sharing a Monster MOOC assignment for beginners, a MOOC canvas for first sketches, the MOOC design kit for details of instructional design and a MOOC for MOOC makers and a MOOC map as introduction into a certain MOOC platform.
Silk protein-based materials show promise for application as biomaterials for tissue engineering. The simple and rapid photochemical modification of silk protein-based materials composed of either Bombyx mori silkworm silk or engineered spider silk proteins (eADF4(C16)) is reported. Radicals formed on the silk-based materials initiate the polymerization of monomers (acrylic acid, methacrylic acid, or allylamine) which functionalize the surface of the silk materials with poly(acrylic acid) (PAA), poly(methacrylic acid) (PMAA), or poly(allylamine) (PAAm). To demonstrate potential applications of this type of modification, the polymer-modified silks are mineralized. The PAA- and PMAA-functionalized silks are mineralized with calcium carbonate, whereas the PAAm-functionalized silks are mineralized with silica, both of which provide a coating on the materials that may be useful for bone tissue engineering, which will be the subject of future investigations.
Background:
Given the rising number of strokes worldwide, and the large number of individuals left with disabilities after stroke, novel strategies to reduce disability, increase functions in the motor and the cognitive domains, and improve quality of life are of major importance. Physical activity is a promising intervention to address these challenges but, as yet, there is no study demonstrating definite outcomes. Our objective is to assess whether additional treatment in the form of physical fitness-based training for patients early after stroke will provide benefits in terms of functional outcomes, in particular gait speed and the Barthel Index (co-primary outcome measures) reflecting activities of daily living (ADL). We will gather secondary functional outcomes as well as mechanistic parameters in an exploratory approach.
Methods/Design: Our phase III randomised controlled trial will recruit 215 adults with moderate to severe limitations of walking and ADL 5 to 45 days after stroke onset. Participants will be stratified for the prognostic variables of "centre", "age", and "stroke severity", and randomly assigned to one of two groups. The interventional group receives physical fitness training delivered as supported or unsupported treadmill training (cardiovascular active aerobic training; five times per week, over 4 weeks; each session 50 minutes; total of 20 additional physical fitness training sessions) in addition to standard rehabilitation treatment. The control intervention consists of relaxation sessions (non-cardiovascular active; five times per week week, over 4 weeks; each session 50 minutes) in addition to standard rehabilitation treatment. Co-primary efficacy endpoints will be gait speed (in m/s, 10 m walk) and the Barthel Index (100 points total) at 3 months post-stroke, compared to baseline measurements. Secondary outcomes include standard measures of quality of life, sleep and mood, cognition, arm function, maximal oxygen uptake, and cardiovascular risk factors including blood pressure, pulse, waist-to-hip ratio, markers of inflammation, immunity and the insulin-glucose pathway, lipid profile, and others.
Discussion: The goal of this endpoint-blinded, phase III randomised controlled trial is to provide evidence to guide post-stroke physical fitness-based rehabilitation programmes, and to elucidate the mechanisms underlying this intervention.
Was wird unter „nachhaltiger Prävention“ in der Präventionsforschung verstanden? Welche guten Beispiele für nachhaltige Prävention gibt es in der Praxis? Und v. a.: Wie lässt sich Prävention in den verschiedenen Bereichen wie Kriminalität, Gewalt und Rechtsextremismus nachhaltig gestalten? Diesen Fragen will der vorliegende Sammelband nachgehen und damit der Präventionsdebatte neue Impulse verleihen. Der Band will insbesondere die nationale sowie internationale Fachdebatte konstruktiv aufgreifen, Theorie und Praxis verbinden, „good practice“ Beispiele darstellen sowie Perspektiven nachhaltiger Prävention aufzeigen. Mit diesem Themenspektrum richtet er sich sowohl an die Wissenschaft als auch an die Praxis sowie insgesamt an eine interessierte Öffentlichkeit.
Aims/hypothesis Polycystic ovary syndrome (PCOS) is a risk factor of type 2 diabetes. Screening for impaired glucose metabolism (IGM) with an OGTT has been recommended, but this is relatively time-consuming and inconvenient. Thus, a strategy that could minimise the need for an OGTT would be beneficial. Materials and methods Consecutive PCOS patients (n=118) with fasting glucose < 6.1 mmol/l were included in the study. Parameters derived from medical history, clinical examination and fasting blood samples were assessed by decision tree modelling for their ability to discriminate women with IGM (2-h OGTT value >= 7.8 mmol/l) from those with NGT. Results According to the OGTT results, 93 PCOS women had NGT and 25 had IGM. The best decision tree consisted of HOMA-IR, the proinsulin:insulin ratio, proinsulin, 17-OH progesterone and the ratio of luteinising hormone:follicle-stimulating hormone. This tree identified 69 women with NGT. The remaining 49 women included all women with IGM (100% sensitivity, 74% specificity to detect IGM). Pruning this tree to three levels still identified 53 women with NGT (100% sensitivity, 57% specificity to detect IGM). Restricting the data matrix used for tree modelling to medical history and clinical parameters produced a tree using BMI, waist circumference and WHR. Pruning this tree to two levels separated 27 women with NGT (100% sensitivity, 29% specificity to detect IGM). The validity of both trees was tested by a leave-10%-out cross-validation. Conclusions/interpretation Decision trees are useful tools for separating PCOS women with NGT from those with IGM. They can be used for stratifying the metabolic screening of PCOS women, whereby the number of OGTTs can be markedly reduced.
High-density polyethylene becomes optically transparent during tensile drawing when previously saturated with diesel fuel. This unusual phenomenon is investigated as it might allow conclusions with respect to the material behavior. Microscopy, differential scanning calorimetry, density measurements are applied together with two scanning X-ray scattering techniques: wide angle X-ray scattering (WAXS) and X-ray refraction, able to extract the spatially resolved crystal orientation and internal surface, respectively. The sorbed diesel softens the material and significantly alters the yielding characteristics. Although the crystallinity among stretched regions is similar, a virgin reference sample exhibits strain whitening during stretching, while the diesel-saturated sample becomes transparent. The WAXS results reveal a pronounced fiber texture in the tensile direction in the stretched region and an isotropic orientation in the unstretched region. This texture implies the formation of fibrils in the stretched region, while spherulites remain intact in the unstretched parts of the specimens. X-ray refraction reveals a preferred orientation of internal surfaces along the tensile direction in the stretched region of virgin samples, while the sample stretched in the diesel-saturated state shows no internal surfaces at all. Besides from stretching saturated samples, optical transparency is also obtained from sorbing samples in diesel after stretching.
Defensive behaviors in animals and humans vary dynamically with increasing proximity of a threat and depending upon the behavioral repertoire at hand. The current study investigated physiological and behavioral adjustments and associated brain activation when participants were exposed to dynamically approaching threat that was either inevitable or could be avoided by motor action. When the approaching threat was inevitable, attentive freezing was observed as indicated by fear bradycardia, startle potentiation, and a dynamic increase in activation of the anterior insula and the periaqueductal grey. In preparation for active avoidance a switch in defensive behavior was observed characterized by startle inhibition and heart rate acceleration along with potentiated activation of the amygdala and the periaqueductal grey. Importantly, the modulation of defensive behavior according to threat imminence and the behavioral option at hand was associated with activity changes in the ventromedial prefrontal cortex. These findings improve our understanding of brain mechanisms guiding human behavior during approaching threat depending on available resources.
We present an excerpt of the document "Quantum Information Processing and Communication: Strategic report on current status, visions and goals for research in Europe", which has been recently published in electronic form at the website of FET (the Future and Emerging Technologies Unit of the Directorate General Information Society of the European Commission, http://www.cordis.lu/ist/fet/qipc-sr.htm). Besides being a document addressed to policy makers and funding agencies (both at the European and national level), the document contains a detailed scientific assessment of the state-of-the-art, main research goals, challenges, strengths, weaknesses, visions and perspectives of all the most relevant QIPC sub-fields, that we report here
Surface self-assembly of spherical nanoparticles of sizes below 10 nm into hierarchical heterostructures is under arising development despite the inherent difficulties of obtaining complex ordering patterns on a larger scale. Due to template-mediated interactions between oil-dispersible superparamagnetic nanoparticles (MNPs) and polyethylenimine- stabilized gold nanoparticles (Au(PEI)NPs) at the water-oil interface of microemulsions, complex nanostructured films can be formed. Characterization of the reverse microemulsion phase by UV-vis absorption revealed the formation of heteroclusters from Winsor type II phases (WPII) using Aerosol-OT (AOT) as the surfactant. SAXS measurements verify the mechanism of initial nanoparticle clustering in defined dimensions. XPS suggested an influence of AOT at the MNP surface. Further, cryo-SEM and TEM visualization demonstrated the elongation of the reverse microemulsions into cylindrical, wormlike structures, which subsequently build up larger nanoparticle superstructure arrangements. Such WPII phases are thus proven to be a new form of soft template, mediating the self-assembly of different nanoparticles in hierarchical network-like filaments over a substrate during solvent evaporation.
Raman microspectroscopy on carbonaceous material (RSCM) from the eastern Tauern Window indicates contrasting peak-temperature patterns in three different fabric domains, each of which underwent a poly-metamorphic orogenic evolution: Domain 1 in the northeastern Tauern Window preserves oceanic units (Glockner Nappe System, Matrei Zone) that attained peak temperatures (T-p) of 350-480 degrees C following Late Cretaceous to Palaeogene nappe stacking in an accretionary wedge. Domain 2 in the central Tauern Window experienced T-p of 500-535 degrees C that was attained either within an exhumed Palaeogene subduction channel or during Oligocene Barrovian-type thermal overprinting within the Alpine collisional orogen. Domain 3 in the Eastern Tauern Subdome has a peak-temperature pattern that resulted from Eo-Oligocene nappe stacking of continental units derived from the distal European margin. This pattern acquired its presently concentric pattern in Miocene time due to post-nappe doming and extensional shearing along the Katschberg Shear Zone System (KSZS). T-p values in the largest (Hochalm) dome range from 612 degrees C in its core to 440 degrees C at its rim. The maximum peak-temperature gradient (70 degrees Ckm(-1)) occurs along the eastern margin of this dome where mylonitic shearing of the Katschberg Normal Fault (KNF) significantly thinned the Subpenninic- and Penninic nappe pile, including the pre-existing peak-temperature gradient.
Organic management is one of the most popular strategies to reduce negative environmental impacts of intensive agriculture. However, little is known about benefits for biodiversity and potential worsening of yield under organic grasslands management across different grassland types, i.e. meadow, pasture and mown pasture. Therefore, we studied the diversity of vascular plants and foliage-living arthropods (Coleoptera, Araneae, Heteroptera, Auchenorrhyncha), yield, fodder quality, soil phosphorus concentrations and land-use intensity of organic and conventional grasslands across three study regions in Germany. Furthermore, all variables were related to the time since conversion to organic management in order to assess temporal developments reaching up to 18 years. Arthropod diversity was significantly higher under organic than conventional management, although this was not the case for Araneae, Heteroptera and Auchenorrhyncha when analyzed separately. On the contrary, arthropod abundance, vascular plant diversity and also yield and fodder quality did not considerably differ between organic and conventional grasslands. Analyses did not reveal differences in the effect of organic management among grassland types. None of the recorded abiotic and biotic parameters showed a significant trend with time since transition to organic management, except soil organic phosphorus concentrations which decreased with time. This implies that permanent grasslands respond slower and probably weaker to organic management than crop fields do. However, as land-use intensity and inorganic soil phosphorus concentrations were significantly lower in organic grasslands, overcoming seed and dispersal limitation by re-introducing plant species might be needed to exploit the full ecological potential of organic grassland management. We conclude that although organic management did not automatically increase the diversity of all studied taxa, it is a reasonable and useful way to support agro-biodiversity.
Remnants of hydrous melt formed at mantle depths have been identified and characterized within high-pressure leucogranulites of the Orlica-Snieznik Dome (Bohemian Massif, central Europe). They occur as nanogranites in garnet formed via partial melting of granitoids during the Variscan orogeny. Melt composition and H2O content have been investigated in situ after experimental re-homogenization of the nanogranites, and are consistent with melts produced experimentally from crustal lithologies at mantle depths. This is the first geochemical study of melt inclusions from natural crustal rocks equilibrated close to the stability field of coesite, shedding light on how continental crust melts during deep subduction. Whereas decompressional melting is commonly invoked for deeply subducted crustal lithologies, melting occurred near or at the metamorphic peak pressure in the Orlica-Snieznik granulites. Melting of deeply subducted crustal rocks significantly modifies the rheology and thus promotes fast exhumation: this process has a critical influence on the geodynamic evolution of subduction-collision orogens as well as crustal differentiation at depth.
Systems of parallel finite automata communicating by states are investigated. We consider deterministic and nondeterministic devices and distinguish four working modes. It is known that systems in the most general mode are as powerful as one-way multi-head finite automata. Here we solve some open problems on the computational capacity of systems working in the remaining modes. In particular, it is shown that deterministic returning and non-returning devices are equivalent, and that there are languages which are accepted by deterministic returning and centralized systems but cannot be accepted by deterministic non-returning centralized systems. Furthermore, we show that nondeterministic systems are strictly more powerful than their deterministic variants in all the four working modes. Finally, incomparability with the classes of (deterministic) (linear) context-free languages as well as the Church-Rosser languages is derived.
This study monitors regional changes in the crystallinity of carbonaceous matter (CM) by applying Micro-Raman spectroscopy to a total of 214 metasediment samples (largely so-called Bundnerschiefer) dominantly metamorphosed under blueschist- to amphibolite-facies conditions. They were collected within the northeastern margin of the Lepontine dome and easterly adjacent areas of the Swiss Central Alps. Three-dimensional mapping of isotemperature contours in map and profile views shows that the isotemperature contours associated with the Miocene Barrow-type Lepontine metamorphic event cut across refolded nappe contacts, both along and across strike within the northeastern margin of the Lepontine dome and adjacent areas. Further to the northeast, the isotemperature contours reflect temperatures reached during the Late Eocene subduction-related blueschist-facies event and/or during subsequent near-isothermal decompression; these contours appear folded by younger, large-scale post-nappe-stacking folds. A substantial jump in the recorded maximum temperatures across the tectonic contact between the frontal Adula nappe complex and surrounding metasediments indicates that this contact accommodated differential tectonic movement of the Adula nappe with respect to the enveloping Bundnerschiefer after maximum temperatures were reached within the northern Adula nappe, i.e. after Late Eocene time.
We present a new integral field spectroscopic dataset of the central part of the Orion Nebula (M 42), observed with the MUSE instrument at the ESO VLT. We reduced the data with the public MUSE pipeline. The output products are two FITS cubes with a spatial size of similar to 5'9 x 4'9 (corresponding to similar to 0.76 x 0.63 pc(2)) and a contiguous wavelength coverage of 4595 ... 9366 angstrom, spatially sampled at 0 ''.2. We provide two versions with a sampling of 1.25 angstrom and 0.85 angstrom in dispersion direction. Together with variance cubes these files have a size of 75 and 110 GiB on disk. They are the largest integral field mosaics to date in terms of information content. We make them available for use in the community. To validate this dataset, we compare world coordinates, reconstructed magnitudes, velocities, and absolute and relative emission line fluxes to the literature values and find excellent agreement. We derive a 2D map of extinction and present de-reddened flux maps of several individual emission lines and of diagnostic line ratios. We estimate physical properties of the Orion Nebula, using the emission line ratios [N II] and [S III] (for the electron temperature T-e) and [S II] and [Cl III] (for the electron density N-e), and show 2D images of the velocity measured from several bright emission lines.
Ultrahigh-pressure (UHP), coesite-bearing edogites in the Himalaya have been documented from the Kaghan Valley in Pakistan and the Tso Morani area in northwest India. These complexes are part of the northern edge of the Indian plate that has been subducted to, and metamorphosed at, mantle depths of more than 100 km before being exhumed. Both UHP complexes are located today directly adjacent to the Indus-Tsangpo suture zone and are not separated by non-metamorphosed sequences of Tethyan sediments from the Asian margin. Herein, we present new data for one fresh coesite-bearing eclogite from the Tso Moran massif. Therein, garnets are zoned reflecting their growth during prograde and peak metamorphism and showing a thin retrograde overgrowth. Inclusions can be directly correlated to the compositional zoning and are seen as either relicts of the protolith mineral paragenesis and as "snap shots" of the mineral paragenesis during subduction and under peak conditions. Rare earth element concentrations (REE) were obtained for garnet, mineral inclusions in garnet and matrix minerals. The REE pattern in garnet reflects a sequential change in matrix minerals and their proportions due to net transfer reactions during subduction and peak metamorphism. Using conventional geothermobarometry, a peak pressure of ca. 44-48 kbar at 560-760 degrees C followed by an S-shaped exhumation curve has been deduced. Extreme droughts, heat waves, frosts, precipitation, wind storms and other climate extremes may impact the structure, composition and functioning of terrestrial ecosystems, and thus carbon cycling and its feedbacks to the climate system. Yet, the interconnected avenues through which climate extremes drive ecological and physiological processes and alter the carbon balance are poorly understood. Here, we review the literature on carbon cycle relevant responses of ecosystems to extreme climatic events. Given that impacts of climate extremes are considered disturbances, we assume the respective general disturbance-induced mechanisms and processes to also operate in an extreme context. The paucity of well-defined studies currently renders a quantitative meta-analysis impossible, but permits us to develop a deductive framework for identifying the main mechanisms (and coupling thereof) through which climate extremes may act on the carbon cycle. We find that ecosystem responses can exceed the duration of the climate impacts via lagged effects on the carbon cycle. The expected regional impacts of future climate extremes will depend on changes in the probability and severity of their occurrence, on the compound effects and timing of different climate extremes, and on the vulnerability of each land-cover type modulated by management. Although processes and sensitivities differ among biomes, based on expert opinion, we expect forests to exhibit the largest net effect of extremes due to their large carbon pools and fluxes, potentially large indirect and lagged impacts, and long recovery time to regain previous stocks. At the global scale, we presume that droughts have the strongest and most widespread effects on terrestrial carbon cycling. Comparing impacts of climate extremes identified via remote sensing vs. ground-based observational case studies reveals that many regions in the (sub-)tropics are understudied. Hence, regional investigations are needed to allow a global upscaling of the impacts of climate extremes on global carbon-climate feedbacks.
Parallel communicating finite automata (PCFAs) are systems of several finite state automata which process a common input string in a parallel way and are able to communicate by sending their states upon request. We consider deterministic and nondeterministic variants and distinguish four working modes. It is known that these systems in the most general mode are as powerful as one-way multi-head finite automata. It is additionally known that the number of heads corresponds to the number of automata in PCFAs in a constructive way. Thus, undecidability results as well as results on the hierarchies induced by the number of heads carry over from multi-head finite automata to PCFAs in the most general mode. Here, we complement these undecidability and hierarchy results also for the remaining working modes. In particular, we show that classical decidability questions are not semi-decidable for any type of PCFAs under consideration. Moreover, it is proven that the number of automata in the system induces infinite hierarchies for deterministic and nondeterministic PCFAs in three working modes.
Are gastropods, rather than ants, important dispersers of seeds of myrmecochorous forest herbs?

Seed dispersal by ants (myrmecochory) is widespread, and seed adaptations to myrmecochory are common, especially in the form of fatty appendices (elaiosomes). In a recent study, slugs were identified as seed dispersers of myrmecochores in a central European beech forest. Here we used 105 beech forest sites to test whether myrmecochore presence and abundance is related to ant or gastropod abundance and whether experimentally exposed seeds are removed by gastropods. Myrmecochorous plant cover was positively related to gastropod abundance but was negatively related to ant abundance. Gastropods were responsible for most seed removal and elaiosome damage, whereas insects (and rodents) played minor roles. These gastropod effects on seeds were independent of region or forest management. We suggest that terrestrial gastropods can generally act as seed dispersers of myrmecochorous plants and even substitute myrmecochory, especially where ants are absent or uncommon.
Characterization of freeze-fractured epithelial plasma membranes on nanometer scale with ToF-SIMS

Time-of-flight secondary ion mass spectrometry (ToF-SIMS) was used to characterize the freeze-fracturing process of human epithelial PANC-1 and UROtsa cells. For this purpose, phosphatidylcholine, sphingomyelin, phosphatidylethanolamine, and phosphatidylserine standard samples were investigated to find specific signals with both high specificity and signal intensity. The results were used to investigate single cells of subconfluent cell layers prepared with a special silicon wafer sandwich preparation technique. This freeze-fracturing technique strips cell membranes off the cells, isolating them on opposing silicon wafer substrates. Criteria were found for defining regions with stripped off cell membranes and, on the opposing wafer, complementary regions with the remaining cells. Measured ethanolamine/choline and serine/choline ratios in these regions clearly showed that in the freeze-fracturing process, the lipid bilayer of the plasma membrane is split along its central zone. Accordingly, only the outer lipid monolayer is stripped off the cell, while the inner lipid monolayer remains attached to the cell on the opposing wafer, thus allowing detailed analysis of a single lipid monolayer. Furthermore, it could be shown that using different washing procedures did not influence the transmembrane lipid distribution. Under optimized preparation conditions, it became feasible to detect lipids with a lateral resolution of approximately 100 nm. The data indicate that ToF-SIMS would be a very useful technique to study with very high lateral resolution changes in lipid composition caused, for example, by lipid storage diseases or pharmaceuticals that interfere with the lipid metabolism.
Positive coping styles and perigenual ACC volume: two related mechanisms for conferring resilience?

Stress exposure has been linked to increased rates of depression and anxiety in adults, particularly in females, and has been associated with maladaptive changes in the anterior cingulate cortex (ACC), which is an important brain structure involved in internalizing disorders. Coping styles are important mediators of the stress reaction by establishing homeostasis, and may thus confer resilience to stress-related psychopathology. Anatomical scans were acquired in 181 healthy participants at age 25 years. Positive coping styles were determined using a self-report questionnaire (German Stress Coping Questionnaire, SVF78) at age 22 years. Adult anxiety and depression symptoms were assessed at ages 22, 23 and 25 years with the Young Adult Self-Report. Information on previous internalizing diagnoses was obtained by diagnostic interview (2-19 years). Positive coping styles were associated with increased ACC volume. ACC volume and positive coping styles predicted anxiety and depression in a sex-dependent manner with increased positive coping and ACC volume being related to lower levels of psychopathology in females, but not in males. These results remained significant when controlled for previous internalizing diagnoses. These findings indicate that positive coping styles and ACC volume are two linked mechanisms, which may serve as protective factors against internalizing disorders.
Converging evidence emphasizes the role of an interaction between monoamine oxidase A (MAOA) genotype, environmental adversity, and sex in the pathophysiology of aggression. The present study aimed to clarify the impact of this interaction on neural activity in aggression-related brain systems. Functional magnetic resonance imaging was performed in 125 healthy adults from a high-risk community sample followed since birth. DNA was genotyped for the MAOA-VNTR (variable number of tandem repeats). Exposure to childhood life stress (CLS) between the ages of 4 and 11 years was assessed using a standardized parent interview, aggression by the Youth/Young Adult Self-Report between the ages of 15 and 25 years, and the VIRA-R (Vragenlijst Instrumentele En Reactieve Agressie) at the age of 15 years. Significant interactions were obtained between MAOA genotype, CLS, and sex relating to amygdala, hippocampus, and anterior cingulate cortex (ACC) response, respectively. Activity in the amygdala and hippocampus during emotional face-matching increased with the level of CLS in male MAOA-L, while decreasing in male MAOA-H, with the reverse pattern present in females. Findings in the opposite direction in the ACC during a flanker NoGo task suggested that increased emotional activity coincided with decreased inhibitory control. Moreover, increasing amygdala activity was associated with higher Y(A)SR aggression in male MAOA-L and female MAOA-H carriers. Likewise, a significant association between amygdala activity and reactive aggression was detected in female MAOA-H carriers. The results point to a moderating role of sex in the MAOAx CLS interaction for intermediate phenotypes of emotional and inhibitory processing, suggesting a possible mechanism in conferring susceptibility to violence-related disorders.
Carbonatites are peculiar magmatic rocks with mantle-related genesis, commonly interpreted as the products of melting of CO2-bearing peridotites, or resulting from the chemical evolution of mantle derived magmas, either through extreme "differentiation or secondary immiscibility. Here we report the first finding of anatectic carbonatites of crustal origin, preserved as calcite-rich polycrystalline inclusions in garnet from low-to-medium pressure migmatites of the Oberpfalz area, SW Bohemian Massif (Central Europe). These inclusions originally trapped a melt of calciocarbonatitic composition with a characteristic enrichment in Ba, Sr and LREE. This interpretation is supported by the results of a detailed microstructural and microchemical investigation, as well as re-melting experiments using a piston cylinder apparatus. Carbonatitic inclusions coexist in the same cluster with crystallized silicate melt inclusions (nanogranites) and COH fluid inclusions, suggesting conditions of primary immiscibility between two melts and a fluid during anatexis. The production of both carbonatitic and granitic melts during the same anatectic event requires a suitable heterogeneous protolith. This may be represented by a sedimentary sequence containing marble lenses of limited extension, similar to the one still visible in the adjacent central Moldanubian Zone. The presence of CO2-rich fluid inclusions suggests furthermore that high CO2 activity during anatexis may be required to stabilize a carbonate-rich melt in a silica-dominated, system. This natural occurrence displays a remarkable similarity with experiments on carbonate-silicate melt immiscibility, where CO2 saturation is a condition commonly imposed. In conclusion, this study shows how the investigation of partial melting through melt inclusion studies may unveil unexpected processes whose evidence, while preserved in stiff minerals such as garnet, is completely obliterated in the rest of the rock due to metamorphic re-equilibration. Our results thus provide invaluable new insights into the processes which shape the geochemical evolution of our planet, such as the redistribution of carbon and strategic metals during orogenesis. Findings on the etiology of aggressive behavior have provided evidence for an effect both of genetic factors, such as variation in the monoamine oxidase A (MAOA) gene, and adverse environmental factors. Recent studies have supported the existence of gene × environment interactions, with early experiences playing a key role. In the present study, the effects of prenatal nicotine exposure, MAOA genotype and their interaction on aggressive behavior during young adulthood were examined. In a sample of 272 young adults (129 males, 143 females) from an epidemiological cohort study, smoking during pregnancy was measured with a standardized parent interview at the offspring’s age of 3 months. Aggressive behavior was assessed between the ages of 19 and 25 years using the Young Adult Self-Report. DNA was genotyped for the MAOA 5′ untranslated region variable number of tandem repeats polymorphism (VNTR). Results revealed a significant interaction between MAOA and smoking during pregnancy, indicating higher levels of aggressive behavior in young adults carrying the MAOA low-expressing genotype who had experienced prenatal nicotine exposure (n = 8, p = .025). In contrast, in carriers of the MAOA high-expressing genotype, maternal smoking during pregnancy had no effect on aggressive behavior during young adulthood (n = 20, p = .145). This study extends earlier findings demonstrating an interaction between MAOA genotype and prenatal nicotine exposure on aggressive behavior into young adulthood. The results point to the long-term adverse effects of smoking during pregnancy on the offspring’s mental health, possibly underlining the importance of smoking cessation during pregnancy. According to the nature of the study (particularly sample size and power), analyses are exploratory and results need to be interpreted cautiously.
A great number of Central Asian wall paintings, archeological materials, architectural fragments, and textiles, as well as painting fragments on silk and paper, make up the so called Turfan Collection at the Asian Art Museum in Berlin. The largest part of the collection comes from the Kucha region, a very important cultural center in the third to ninth centuries. Between 1902 and 1914, four German expeditions traveled along the northern Silk Road. During these expeditions, wall paintings were detached from their original settings in Buddhist cave complexes. This paper reports a technical study of a wall painting, existing in eight fragments, from the Buddhist cave no. 40 (Ritterhohle). Its original painted surface is soot blackened and largely illegible. Gruwedel, leader of the first and third expeditions, described the almost complete destruction of the rediscovered temple complex and evidence of fire damage. The aim of this case study is to identify the materials used for the wall paintings. Furthermore, soot deposits as well as materials from conservation interventions were of interest. Non-invasive analyses were preferred but a limited number of samples were taken to provide more precise information on the painting technique. By employing optical and scanning electron microscopy, energy dispersive X-ray spectroscopy, micro X-ray fluorescence spectroscopy, X-ray diffraction analysis, and Raman spectroscopy, a layer sequence of earthen render, a ground layer made of gypsum, and a paint layer containing a variety of inorganic pigments were identified.
A unique assemblage including kumdykolite and kokchetavite, polymorphs of albite and K-feldspar, respectively, together with cristobalite, micas, and calcite has been identified in high-pressure granulites of the Orlica-Snieznik dome (Bohemian Massif) as the product of partial melt crystallization in preserved nanogranites. Previous reports of both kumdykolite and kokchetavite in natural rocks are mainly from samples that passed through the diamond stability field. However, because the maximum pressure recorded in these host rocks is <3 GPa, our observations indicate that high pressure is not required for the formation of kumdykolite and kokchetavite, and their presence is not therefore an indicator of ultrahigh-pressure conditions. Detailed microstructural and microchemical investigation of these inclusions indicates that such phases should instead be regarded as (1) a direct mineralogical criteria to identify former melt inclusions with preserved original compositions, including H2O and CO2 contents and (2) indicators of rapid cooling of the host rocks. Thus, the present study provides novel criteria for the interpretation of melt inclusions in natural rocks and allows a more rigorous characterization of partial melts during deep subduction to mantle depth as well as their behavior on exhumation.
Converging evidence has highlighted the association between poverty and conduct disorder (CD) without specifying neurobiological pathways. Neuroimaging research has emphasized structural and functional alterations in the orbitofrontal cortex (OFC) as one key mechanism underlying this disorder. The present study aimed to clarify the long-term influence of early poverty on OFC volume and its association with CD symptoms in healthy participants of an epidemiological cohort study followed since birth. At age 25 years, voxel-based morphometry was applied to study brain volume differences. Poverty (0 = non-exposed (N = 134), I = exposed (N = 33)) and smoking during pregnancy were determined using a standardized parent interview, and information on maternal responsiveness was derived from videotaped mother infant interactions at the age of 3 months. CD symptoms were assessed by diagnostic interview from 8 to 19 years of age. Information on life stress was acquired at each assessment and childhood maltreatment was measured using retrospective self-report at the age of 23 years. Analyses were adjusted for sex, parental psychopathology and delinquency, obstetric adversity, parental education, and current poverty. Individuals exposed to early life poverty exhibited a lower OFC volume. Moreover, we replicated previous findings of increased CD symptoms as a consequence of childhood poverty. This effect proved statistically mediated by OFC volume and exposure to life stress and smoking during pregnancy, but not by childhood maltreatment and maternal responsiveness. These findings underline the importance of studying the impact of early life adversity on brain alterations and highlight the need for programs to decrease income-related disparities.
Albian-Turonian subduction-accretionary complexes are exposed widely in the Central Pontides. A major portion of the accretionary complexes is made up of a metaflysch sequence consisting of slate/phyllite and metasandstone intercalation with blocks of marble, Na-amphibole bearing metabasite, and serpentinite. The metaflysch sequence represents distal parts of a large Lower Cretaceous submarine turbidite fan deposited on the Laurasian active continental margin that was subsequently accreted and metamorphosed during the Albian. Raman spectra of carbonaceous material of the metapelitic rocks revealed that the metaflysch consists of metamorphic packets with distinct peak metamorphic temperatures. The majority of the metapelites are low-temperature (ca. 330 degrees C) slates characterized by lack of differentiation of the graphite (G) and D2 defect bands. They possibly represent offscraped distal turbidites along the toe of the Albian accretionary wedge. Other phyllites are characterized by a slightly pronounced G band with a D2 defect band occurring on its shoulder. Peak metamorphic temperatures of these phyllites are constrained to 370-385 degrees C. The phyllites are associated with a strip of incipient blueschist facies metabasites and are found as a sliver within the offscraped distal turbidites. We interpret the phyllites as underplated continental sediments together with oceanic crustal basalt along the basal decollement. Tectonic emplacement of the underplated rocks into the offscraped distal turbidites was possibly achieved by out-of-sequence thrusting causing tectonic thickening and uplift of the wedge.
Over the past years, family 18 chitinases have been validated as potential targets for the design of drugs against human pathogens that contain or interact with chitin during their normal life cycles. Thus far, only one potent chitinase inhibitor has been described in detail, the pseudotrisaccharide allosamidin. Recently, however, two potent natural-product cyclopentapeptide chitinase inhibitors, argifin and argadin, were reported. Here, we describe high- resoln. crystal structures that reveal the details of the interactions of these cyclopeptides with a family 18 chitinase. The structures are examples of complexes of a carbohydrate-processing enzyme with high-affinity peptide-based inhibitors and show in detail how the peptide backbone and side chains mimic the interactions of the enzyme with chitooligosaccharides. Together with enzymol. characterization, the structures explain why argadin shows an order of magnitude stronger inhibition than allosamidin, whereas argifin shows weaker inhibition. The peptides bind to the chitinase in remarkably different ways, which may explain the differences in inhibition consts. The two complexes provide a basis for structure-based design of potent chitinase inhibitors, accessible by std. peptide chem.

Hilft Beten? Christen gehen davon aus, dass sie Gott nicht umsonst anrufen. Wirkt Gott in uns oder ist das Gebet als ein selbstreflexiver Vollzug des inneren Menschen anzusehen? Darf das Gebet von seiner Erhörung abhängig gemacht werden, muss es davon unabhängig gemacht werden? Diese Anfragen an das Gebet betreffen nicht nur die Glaubwürdigkeit einer der wesentlichsten christlichen Praktiken, sondern den Kern des Glaubens und des Gottesbegriffs. Soll die Logik des Gebets verstanden werden, ist somit die innere Systematik der Theologie zu befragen, liturgie- und frömmigkeitsgeschichtliche Zugänge sind zu bedenken und die äußere Begründbarkeit zu erörtern.
Dieser Band zum Gebet umfasst Beiträge aus unterschiedlichen Konfessionen. Die große Mehrzahl ist im Rahmen der Jahrestagung des Interkonfessionellen Theologischen Arbeitskreises im Januar 2014 zu diesem Thema entstanden. Einige weitere wurden gezielt als Ergänzung aufgenommen. Der Schwerpunkt des Bandes ist systematisch-theologischer Art, da die Reflexionen auf die Denkmöglichkeiten des Betens zugeschnitten sind.
The limited capacity of cartilage to heal large lesions through endogenous mechanisms has led to extensive effort to develop materials to facilitate chondrogenesis. Although physical-chemical properties of biomaterials have been shown to impact in vitro chondrogenesis, whether these findings are translatable in vivo is subject of debate. Herein, architectured 3D hydrogel scaffolds (ArcGel) (produced by crosslinking gelatin with ethyl lysine diisocyanate (LDI)) were used as a model system to investigate the interplay between scaffold mechanical properties and degradation on matrix deposition by human articular chondrocytes (HAC) from healthy donors in vitro and in vivo. Using ArcGel scaffolds of different tensile and shear modulus, and degradation behavior; in this study, we compared the fate of ex vivo engineeredArcGels-chondrocytes constructs, i.e. the traditional tissue engineering approach, with the de novo formation of cartilaginous tissue in HAC laden ArcGels in an ectopic nude mouse model. While the softer and fast degrading ArcGel (LNCO3) was more efficient at promoting chondrogenic differentiation in vitro, upon ectopic implantation, the stiffer and slow degrading ArcGel (LNCO8) was superior in maintaining chondrogenic phenotype in HAC and retention of cartilaginous matrix. Furthermore, surprisingly the de novo formation of cartilage tissue was promoted only in LNCO8. Since HAC cultured for only three days in the LNCO8 environment showed upregulation of hypoxia-associated genes, this suggests a potential role for hypoxia in the observed in vivo outcomes. In summary, this study sheds light on how immediate environment (in vivo versus in vitro) can significantly impact the outcomes of cell-laden biomaterials. Statement of Significance In this study, 3D architectured hydrogels (ArcGels) with different mechanical and biodegradation properties were investigated for their potential to promote formation of cartilaginous matrix by human articular chondrocytes in vitro and in vivo. Two paradigms were explored (i) ex vivo engineering followed by in vivo implantation in ectopic site of nude mice and (ii) short in vitro culture (3 days) followed by implantation to induce de novo cartilage formation. Softer and fast degrading ArcGel were better at promoting chondrogenesis in vitro, while stiffer and slow degrading ArcGel were strikingly superior in both maintaining chondrogenesis in vivo and inducing de novo formation of cartilage. Our findings highlight the importance of the interplay between scaffold mechanics and degradation in chondrogenesis.
We modeled the two most extreme highstands of Lake Naivasha during the last 175 k.y. to estimate potential precipitation/ evaporation changes in this basin. In a first step, the bathymetry of the paleolakes at f135 and 9 k.y. BP was reconstructed from sediment cores and surface outcrops. Second, we modeled the paleohydrologic budget during the highstands using a simplified coupled energy mass-balance model. Our results show that the hydrologic and hence the climate conditions at f135 and 9 k.y. BP were similar, but significantly different from today. The main difference is a f15% higher value in precipitation compared to the present. An adaptation and migration of vegetation in the cause of climate changes would result in a f30% increase in precipitation. The most likely cause for such a wetter climate at f135 and 9 k.y. BP is a more intense intertropical convergence and increased precipitation in East Africa.
Orbitolinids are larger foraminifera widespread in Lower Cretaceous shallow-water carbonates of the Tethyan realm. They are among the most important fossil groups used for Biostratigraphy. Despite this and although the structural features of the group have been described in detail, very little is known about the composition of their agglutinated test and the process by which they selected foreign grains. In this study, the test of Orbitolina d'Orbigny, 1850 (subgenus Mesorbitolina Schroeder, 1962) from Aptian shallow-water carbonate deposits of southern Italy has been studied in detail. We combine petrographic techniques (optical microscope and SEM) with energy-dispersive x-ray spectrometry (EDS), electron probe microanalyzer (EPMA), X-ray diffraction and Raman spectroscopy analyses.
The results show that the test of Mesorbitolina is composed of carbonate and non-carbonate agglutinated grains with the latter distributed across the test with a specific pattern, moving from the marginal to the central zone. In the marginal zone, non-carbonate grains are found only in the epidermis and along the septa which are composed of quartz, with smaller amounts of illite/muscovite and K-feldspar grains. In the central zone of the test, non-carbonate grains are distributed in two ways. Coarse grains of quartz and K-feldspar are abundant and randomly placed in the endoskeleton embedded in a mosaic of minute carbonate grains. Flat grains, mainly of illite/muscovite constitute the external part of the septa. Our observations indicate that Mesorbitolina did select and place agglutinated grains across its test, mainly according to their shape, whereas it did not select particles according to grain size. The distribution of agglutinated particles according to their mineralogical composition shows some contradictory evidence and therefore, at the moment, grain selection in function of mineralogy cannot be completely confirmed or ruled out. Analogies in the test composition of Mesorbitolina specimens from coeval deposits from different areas of southern Italy indicate that the features of their agglutinated test are typical characters of the genus Mesorbitolina. However, it is still unclear what advantage was obtained by the foraminifer by the described test features.
The potential of ecological models for supporting environmental decision making is increasingly acknowledged. However, it often remains unclear whether a model is realistic and reliable enough. Good practice for developing and testing ecological models has not yet been established. Therefore, TRACE, a general framework for documenting a model's rationale, design, and testing was recently suggested. Originally TRACE was aimed at documenting good modelling practice. However, the word 'documentation' does not convey TRACE's urgency. Therefore, we re-define TRACE as a tool for planning, performing, and documenting good modelling practice. TRACE documents should provide convincing evidence that a model was thoughtfully designed, correctly implemented, thoroughly tested, well understood, and appropriately used for its intended purpose. TRACE documents link the science underlying a model to its application, thereby also linking modellers and model users, for example stakeholders, decision makers, and developers of policies. We report on first experiences in producing TRACE documents. We found that the original idea underlying TRACE was valid, but to make its use more coherent and efficient, an update of its structure and more specific guidance for its use are needed. The updated TRACE format follows the recently developed framework of model 'evaludation': the entire process of establishing model quality and credibility throughout all stages of model development, analysis, and application. TRACE thus becomes a tool for planning, documenting, and assessing model evaludation, which includes understanding the rationale behind a model and its envisaged use. Incorporating photochromic molecules into organic/inorganic hybrid materials may lead to photoresponsive systems. In such systems, the second-order nonlinear properties can be controlled via external stimulation with light at an appropriate wavelength. By creating photochromic molecular switches containing self-assembled monolayers on Si(111), we can demonstrate efficient reversible switching, which is accompanied by a pronounced modulation of the nonlinear optical (NLO) response of the system. The concept of utilizing functionalized photoswitchable Si surfaces could be a way for the generation of two-dimensional NLO switching materials, which are promising for applications in photonic and optoelectronic devices.
The central European Bohemian Massif has undergone over two centuries of scientific investigation which has made it a pivotal area for the development and testing of modern geological theories. The discovery of melt inclusions in high-grade rocks, either crystallized as nanogranitoids or as glassy inclusions, prompted the re-evaluation of the area with an ‘inclusionist’ eye. Melt inclusions have been identified in a wide range of rocks, including felsic/perpotassic granulites, migmatites, eclogites and garnet clinopyroxenites, all the result of melting events albeit over a wide range of pressure/temperature conditions (800–1000°C/0.5–5 GPa). This contribution provides an overview of such inclusions and discusses the qualitative and quantitative constraints they provide for melting processes, and the nature of melts and fluids involved in these processes. In particular, data on trace-element signatures of melt inclusions trapped at mantle depths are presented and discussed. Moreover, experimental re-homogenization of nanogranitoids provided microstructural criteria allowing assessment of the conditions at which melt and host are mutually stable during melting. Overall this work aims to provide guidelines and suggestions for petrologists wishing to explore the fascinating field of melt inclusions in metamorphic terranes worldwide, based on the newest discoveries from the still-enigmatic Bohemian Massif.
Garnet of eclogite (formerly termed garnet clinopyroxenite) hosted in lenses of orogenic garnet peridotite from the Granulitgebirge, NW Bohemian Massif, contains unique inclusions of granitic melt, now either glassy or crystallized. Analysed glasses and re‐homogenized inclusions are hydrous, peraluminous, and enriched in highly incompatible elements characteristic of the continental crust such as Cs, Li, B, Pb, Rb, Th, and U. The original melt thus represents a pristine, chemically evolved metasomatic agent, which infiltrated the mantle via deep continental subduction during the Variscan orogeny. The bulk chemical composition of the studied eclogites is similar to that of Fe‐rich basalt and the enrichment in LILE and U suggest a subduction‐related component. All these geochemical features confirm metasomatism. In comparison with many other garnet+clinopyroxene‐bearing lenses in peridotites of the Bohemian Massif, the studied samples from Rubinberg and Klatschmühle are more akin to eclogite than pyroxenites, as reflected in high jadeite content in clinopyroxene, relatively low Mg, Cr, and Ni but relatively high Ti. However, trace elements of both bulk rock and individual mineral phases show also important differences making these samples rather unique. Metasomatism involving a melt requiring a trace element pattern very similar to the composition reported here has been suggested for the source region of rocks of the so‐called durbachite suite, that is, ultrapotassic melanosyenites, which are found throughout the high‐grade Variscan basement. Moreover, the Th, U, Pb, Nb, Ta, and Ti patterns of these newly studied melt inclusions (MI) strongly resemble those observed for peridotite and its enclosed pyroxenite from the T‐7 borehole (Staré, České Středhoři Mountains) in N Bohemia. This suggests that a similar kind of crustal‐derived melt also occurred here. This study of granitic MI in eclogites from peridotites has provided the first direct characterization of a preserved metasomatic melt, possibly responsible for the metasomatism of several parts of the mantle in the Variscides.

An der Universität Potsdam wird seit 2015 im Rahmen der „Qualitätsoffensive Lehrerbildung“ das Projekt „Professionalisierung – Schulpraktische Studien – Inklusion“ (PSI-Potsdam) durchgeführt und am Zentrum für Lehrerbildung und Bildungsforschung (ZeLB) koordiniert. Zur ersten Projektförderphase (2015-2018) erschien der Band „PSI-Potsdam – Ergebnisbericht zu den Aktivitäten im Rahmen der Qualitätsoffensive Lehrerbildung (2015-2018)“ zum Auftakt der Reihe „Potsdamer Beiträge zur Lehrerbildung und Bildungsforschung“.
Der vorliegende Band aus der gleichen Reihe gibt in den Kapiteln „Erhebungen“, „Lehrkonzepte“ und „Vernetzungen“ einen Überblick über alle Teilprojekte der zweiten Projektförderphase (2019-2023). Wissenschaftler:innen aus verschiedenen Fachdidaktiken, Fachwissenschaften sowie aus den Bildungswissenschaften und der Inklusionspädagogik haben im Rahmen des Projektes kooperiert. Sowohl praxisnahe Forschung als auch die Entwicklung neuer Lehrkonzepte sowie Strategien zur Vernetzung innerhalb der Lehrkräftebildung stehen im Fokus dieses Bandes. Die Praxisphasen, die im Rahmen des „Potsdamer Modells der Lehrerbildung“ eine zentrale Rolle spielen, wurden in einer großen Studie über alle Praxisphasen untersucht.
Der Band gibt interessante Einblicke in die Ergebnisse der Teilprojekte und Anregungen sowohl für die eigene Forschung als auch für Entwicklungsarbeit wie zum Beispiel die Entwicklung neuer Lehrkonzepte. Herausgegeben wird dieser Band von PD Dr. Jolanda Hermanns (Gesamtkoordinatorin PSI-Potsdam und Chemiedidaktikerin).
Objectives: Severe pneumonia may evoke acute lung injury, and sphingosine-1-phosphate is involved in the regulation of vascular permeability and immune responses. However, the role of sphingosine-1-phosphate and the sphingosine-1-phosphate producing sphingosine kinase 1 in pneumonia remains elusive. We examined the role of the sphingosine-1-phosphate system in regulating pulmonary vascular barrier function in bacterial pneumonia. Design: Controlled, in vitro, ex vivo, and in vivo laboratory study. Subjects: Female wild-type and SphK1-deficient mice, 8-10 weeks old. Human postmortem lung tissue, human blood-derived macrophages, and pulmonary microvascular endothelial cells. Interventions: Wild-type and SphK1-deficient mice were infected with Streptococcus pneumoniae. Pulmonary sphingosine-1-phosphate levels, messenger RNA expression, and permeability as well as lung morphology were analyzed. Human blood-derived macrophages and human pulmonary microvascular endothelial cells were infected with S. pneumoniae. Transcellular electrical resistance of human pulmonary microvascular endothelial cell monolayers was examined. Further, permeability of murine isolated perfused lungs was determined following exposition to sphingosine-1-phosphate and pneumolysin. Measurements and Main Results: Following S. pneumoniae infection, murine pulmonary sphingosine-1-phosphate levels and sphingosine kinase 1 and sphingosine-1-phosphate receptor 2 expression were increased. Pneumonia-induced lung hyperpermeability was reduced in SphK1(-/-) mice compared with wild-type mice. Expression of sphingosine kinase 1 in macrophages recruited to inflamed lung areas in pneumonia was observed in murine and human lungs. S. pneumoniae induced the sphingosine kinase 1/sphingosine-1-phosphate system in blood-derived macrophages and enhanced sphingosine-1-phosphate receptor 2 expression in human pulmonary microvascular endothelial cell in vitro. In isolated mouse lungs, pneumolysin-induced hyperpermeability was dose dependently and synergistically increased by sphingosine-1-phosphate. This sphingosine-1-phosphate-induced increase was reduced by inhibition of sphingosine-1-phosphate receptor 2 or its downstream effector Rho-kinase. Conclusions: Our data suggest that targeting the sphingosine kinase 1-/sphingosine-1-phosphate-/sphingosine-1-phosphate receptor 2-signaling pathway in the lung may provide a novel therapeutic perspective in pneumococcal pneumonia for prevention of acute lung injury.
