For a long time, there were things on this planet that only humans could do, but this time might be coming to an end. By using the universal tool that makes us unique – our intelligence – we have worked to eliminate our uniqueness, at least when it comes to solving cognitive tasks. Artificial intelligence is now able to play chess, understand language, and drive a car – and often better than we.
How did we get here? The philosopher Aristotle formulated the first “laws of thought” in his syllogisms, and the mathematicians Blaise Pascal and Wilhelm Leibniz built some of the earliest calculating machines. The mathematician George Boole was the first to introduce a formal language to represent logic. The natural scientist Alan Turing created his deciphering machine “Colossus,” the first programmable computer. Philosophers, mathematicians, psychologists, and linguists – for centuries, scientists have been developing formulas, machines, and theories that were supposed to enable us to reproduce and possibly even enhance our most valuable ability.
But what exactly is “artificial intelligence”? Even the name calls for comparison. Is artificial intelligence like human intelligence? Alan Turing came up with a test in 1950 to provide a satisfying operational definition of intelligence: According to him, a machine is intelligent if its thinking abilities equal those of humans. It has to reach human levels for any cognitive task. The machine has to prove this by convincing a human interrogator that it is human. Not an easy task: After all, it has to process natural language, store knowledge, draw conclusions, and learn something new. In fact, over the past ten years, a number of AI systems have emerged that have passed the test one way or another in chat conversations with automatically generated texts or images. Nowadays, the discussion usually centers on other questions: Does AI still need its creators? Will it not only outperform humans but someday replace them – be it in the world of work or even beyond? Will AI solve our problems in the age of all-encompassing digital networking – or will it become a part of the problem?
Artificial intelligence, its nature, its limitations, its potential, and its relationship to humans were being discussed even before it existed. Literature and film have created scenarios with very different endings. But what is the view of the scientists who are actually researching with or about artificial intelligence? For the current issue of our research magazine, a cognitive scientist, an education researcher, and a computer scientist shared their views. We also searched the University for projects whose professional environment reveals the numerous opportunities that AI offers for various disciplines. We cover the geosciences and computer science as well as economics, health, and literature studies.
At the same time, we have not lost sight of the broad research spectrum at the University: a legal expert introduces us to the not-so-distant sphere of space law while astrophysicists work on ensuring that state-of-the-art telescopes observe those regions in space where something “is happening” at the right time. A chemist explains why the battery of the future will come from a printer, and molecular biologists explain how they will breed stress-resistant plants. You will read about all this in this issue as well as about current studies on restless legs syndrome in children and the situation of Muslims in Brandenburg. Last but not least, we will introduce you to the sheep currently grazing in Sanssouci Park – all on behalf of science. Quite clever!

Introduction: Cystic fibrosis (CF) is a genetic disease which disrupts the function of an epithelial surface anion channel, CFTR (cystic fibrosis transmembrane conductance regulator). Impairment to this channel leads to inflammation and infection in the lung causing the majority of morbidity and mortality. However, CF is a multiorgan disease affecting many tissues, including vascular smooth muscle. Studies have revealed young people with cystic fibrosis lacking inflammation and infection still demonstrate vascular endothelial dysfunction, measured per flow-mediated dilation (FMD). In other disease cohorts, i.e. diabetic and obese, endurance exercise interventions have been shown improve or taper this impairment. However, long-term exercise interventions are risky, as well as costly in terms of time and resources. Nevertheless, emerging research has correlated the acute effects of exercise with its long-term benefits and advocates the study of acute exercise effects on FMD prior to longitudinal studies. The acute effects of exercise on FMD have previously not been examined in young people with CF, but could yield insights on the potential benefits of long-term exercise interventions.
The aims of these studies were to 1) develop and test the reliability of the FMD method and its applicability to study acute exercise effects; 2) compare baseline FMD and the acute exercise effect on FMD between young people with and without CF; and 3) explore associations between the acute effects of exercise on FMD and demographic characteristics, physical activity levels, lung function, maximal exercise capacity or inflammatory hsCRP levels.
Methods: Thirty young volunteers (10 people with CF, 10 non-CF and 10 non-CF active matched controls) between the ages of 10 and 30 years old completed blood draws, pulmonary function tests, maximal exercise capacity tests and baseline FMD measurements, before returning approximately 1 week later and performing a 30-min constant load training at 75% HRmax. FMD measurements were taken prior, immediately after, 30 minutes after and 1 hour after constant load training. ANOVAs and repeated measures ANOVAs were employed to explore differences between groups and timepoints, respectively. Linear regression was implemented and evaluated to assess correlations between FMD and demographic characteristics, physical activity levels, lung function, maximal exercise capacity or inflammatory hsCRP levels. For all comparisons, statistical significance was set at a p-value of α < 0.05.
Results: Young people with CF presented with decreased lung function and maximal exercise capacity compared to matched controls. Baseline FMD was also significantly decreased in the CF group (CF: 5.23% v non-CF: 8.27% v non-CF active: 9.12%). Immediately post-training, FMD was significantly attenuated (approximately 40%) in all groups with CF still demonstrating the most minimal FMD. Follow-up measurements of FMD revealed a slow recovery towards baseline values 30 min post-training and improvements in the CF and non-CF active groups 60 min post-training. Linear regression exposed significant correlations between maximal exercise capacity (VO2 peak), BMI and FMD immediately post-training.
Conclusion: These new findings confirm that CF vascular endothelial dysfunction can be acutely modified by exercise and will aid in underlining the importance of exercise in CF populations. The potential benefits of long-term exercise interventions on vascular endothelial dysfunction in young people with CF warrants further investigation.
Die vorliegende Forschungsarbeit untersucht den Umgang mit Dilemmata von Topmanagern. Dilemmata sind ein alltägliches Geschäft im Topmanagement. Die entsprechenden Akteure sind daher immer wieder mit diesen konfrontiert und mit ihnen umzugehen, gehört gewissermaßen zu ihrer Berufsbeschreibung. Hinzu kommen Dilemmata im nicht direkt geschäftlichen Bereich, wie zum Beispiel jene zwischen Familien- und Arbeitszeit. Doch stellt dieses Feld ein kaum untersuchtes Forschungsgebiet dar. Während Dilemmata in anderen Bereichen eine zunehmende Aufmerksamkeit erfuhren, wurden deren Besonderheiten im Topmanagement genauso wenig differenziert betrachtet wie zugehörige Umgangsweisen. Theorie und Praxis stellen bezüglich Dilemmata von Topmanagern vor allem einen Gegensatz dar, beziehungsweise fehlt es an einer theoretischen Fundierung der Empirie. Diesem Umstand wird mittels dieser Studie begegnet. Auf der Grundlage einer differenzierten und breiten Erfassung von Theorien zu Dilemmata, so diese auch noch nicht auf Topmanager bezogen wurden, und einer empirischen Erhebung, die im Mittelpunkt dieser Arbeit stehen, soll das Feld Dilemmata von Topmanagern der Forschung geöffnet werden. Empirische Grundlage sind vor allem narrative Interviews mit Topmanagern über ihre Dilemmata-Wahrnehmung, ausgemachte Ursachen, Umgangsweisen und Resultate. Dies erlaubt es, Topmanagertypen sowie Dilemmata-Arten, mit denen sie konfrontiert sind oder waren, analytisch herauszuarbeiten. Angesichts der Praxisrelevanz von Dilemmata von Topmanagern wird jedoch nicht nur ein theoretisches Modell zu dieser Thematik erarbeitet, es werden auch Reflexionen auf die Praxis in Form von Handlungsempfehlungen vorgenommen. Schließlich gilt es, die allgemeine Theorie zu Dilemmata, ohne konkreten Bezug zu Topmanagern, mit den theoretischen Erkenntnissen dieser Studie auf empirischer Basis zu kontrastieren. Dabei wird im Rahmen der empirischen Erfassung und Auswertung dem Ansatz der Grounded-Theory-Methodologie gefolgt.
The hydration process of Portland cement in a cementitious system is crucial for development of the high‐quality cement‐based construction material. Complementary experiments of X‐ray diffraction analysis (XRD), scanning electron microscopy (SEM) and time‐resolved laser fluorescence spectroscopy (TRLFS) using europium (Eu(III)) as an optical probe are used to analyse the hydration process of two cement systems in the absence and presence of different organic admixtures. We show that different analysed admixtures and the used sulphate carriers in each cement system have a significant influence on the hydration process, namely on the time‐dependence in the formation of different hydrate phases of cement. Moreover, the effect of a particular admixture is related to the type of sulphate carrier used. The quantitative information on the amounts of the crystalline cement paste components is accessible via XRD analysis. Distinctly different morphologies of ettringite and calcium−silicate−hydrates (C−S−H) determined by SEM allow visual conclusions about formation of these phases at particular ageing times. The TRLFS data provides information about the admixture influence on the course of the silicate reaction. The dip in the dependence of the luminescence decay times on the hydration time indicates the change in the structure of C−S−H in the early hydration period. Complementary information from XRD, SEM and TRLFS provides detailed information on distinct periods of the cement hydration process.
Non-alcoholic fatty liver diseases (NAFLD) including the severe form with steatohepatitis (NASH) are highly prevalent ailments to which no approved pharmacological treatment exists. Dietary intervention aiming at 10% weight reduction is efficient but fails due to low compliance. Increase in physical activity is an alternative that improved NAFLD even in the absence of weight reduction. The underlying mechanisms are unclear and cannot be studied in humans. Here, a rat NAFLD model was developed that reproduces many facets of the diet-induced NAFLD in humans. The impact of endurance exercise was studied in this model. Male Wistar rats received control chow or a NASH-inducing diet rich in fat, cholesterol, and fructose. Both diet groups were subdivided into a sedentary and an endurance exercise group. Animals receiving the NASH-inducing diet gained more body weight, got glucose intolerant and developed a liver pathology with steatosis, hepatocyte hypertrophy, inflammation and fibrosis typical of NAFLD or NASH. Contrary to expectations, endurance exercise did not improve the NASH activity score and even enhanced hepatic inflammation. However, endurance exercise attenuated the hepatic cholesterol overload and the ensuing severe oxidative stress. In addition, exercise improved glucose tolerance possibly in part by induction of hepatic FGF21 production.
Online hate is a topic that has received considerable interest lately, as online hate represents a risk to self-determination and peaceful coexistence in societies around the globe. However, not much is known about the explanations for adolescents posting or forwarding hateful online material or how adolescents cope with this newly emerging online risk. Thus, we sought to better understand the relationship between a bystander to and perpetrator of online hate, and the moderating effects of problem-focused coping strategies (e.g., assertive, technical coping) within this relationship. Self-report questionnaires on witnessing and committing online hate and assertive and technical coping were completed by 6829 adolescents between 12 and 18 years of age from eight countries. The results showed that increases in witnessing online hate were positively related to being a perpetrator of online hate. Assertive and technical coping strategies were negatively related with perpetrating online hate. Bystanders of online hate reported fewer instances of perpetrating online hate when they reported higher levels of assertive and technical coping strategies, and more frequent instances of perpetrating online hate when they reported lower levels of assertive and technical coping strategies. In conclusion, our findings suggest that, if effective, prevention and intervention programs that target online hate should consider educating young people about problem-focused coping strategies, self-assertiveness, and media skills. Implications for future research are discussed.
In natural heterogeneous environments, the fitness of animals is strongly influenced by the availability and composition of food. Food quantity and biochemical quality constraints may affect individual traits of consumers differently, mediating fitness response variation within and among species. Using a multifactorial experimental approach, we assessed population growth rate, fecundity, and survival of six strains of the two closely related freshwater rotifer species Brachionus calyciflorus sensu stricto and Brachionus fernandoi. Therefore, rotifers fed low and high concentrations of three algal species differing in their biochemical food quality. Additionally, we explored the potential of a single limiting biochemical nutrient to mediate variations in population growth response. Therefore, rotifers fed a sterol-free alga, which we supplemented with cholesterol-containing liposomes. Co-limitation by food quantity and biochemical food quality resulted in differences in population growth rates among strains, but not between species, although effects on fecundity and survival differed between species. The effect of cholesterol supplementation on population growth was strain-specific but not species-specific. We show that fitness response variations within and among species can be mediated by biochemical food quality. Dietary constraints thus may act as evolutionary drivers on physiological traits of consumers, which may have strong implications for various ecological interactions.
Wheat is one of the most consumed foods in the world and unfortunately causes allergic reactions which have important health effects. The α-amylase/trypsin inhibitors (ATIs) have been identified as potentially allergen components of wheat. Due to a lack of data on optimization of ATI extraction, a new wheat ATIs extraction approach combining solvent extraction and selective precipitation is proposed in this work. Two types of wheat cultivars (Triticum aestivum L.), Julius and Ponticus were used and parameters such as solvent type, extraction time, temperature, stirring speed, salt type, salt concentration, buffer pH and centrifugation speed were analyzed using the Plackett-Burman design. Salt concentration, extraction time and pH appeared to have significant effects on the recovery of ATIs (p < 0.01). In both wheat cultivars, Julius and Ponticus, ammonium sulfate substantially reduced protein concentration and inhibition of amylase activity (IAA) compared to sodium chloride. The optimal conditions with desirability levels of 0.94 and 0.91 according to the Doehlert design were: salt concentrations of 1.67 and 1.22 M, extraction times of 53 and 118 min, and pHs of 7.1 and 7.9 for Julius and Ponticus, respectively. The corresponding responses were: protein concentrations of 0.31 and 0.35 mg and IAAs of 91.6 and 83.3%. Electrophoresis and MALDI-TOF/MS analysis showed that the extracted ATIs masses were between 10 and 20 kDa. Based on the initial LC-MS/MS analysis, up to 10 individual ATIs were identified in the extracted proteins under the optimal conditions. The positive implication of the present study lies in the quick assessment of their content in different varieties especially while considering their allergenic potential.
Im Rahmen dieser Dissertation konnten neue Kalium- und Natrium-Ionen Fluoreszenzfarbstoffe von der Klasse der Fluoroionophore synthetisiert und charakterisiert werden. Sie bestehen aus einem N Phenylazakronenether als Ionophor und unterschiedlichen Fluorophoren und sind über einen π-konjugierten 1,2,3-Triazol-1,4-diyl Spacer verbunden. Dabei lag der Fokus während ihrer Entwicklung darauf, diese in ihrer Sensitivität, Selektivität und in ihren photophysikalischen Eigenschaften so zu funktionalisieren, dass sie für intra- bzw. extrazelluläre Konzentrationsbestimmungen geeignet sind.
Durch Variation der in ortho Position der N-Phenylazakronenether befindlichen Alkoxy-Gruppen und der fluorophoren Gruppe der Fluoroionophore konnte festgestellt werden, dass die Sensitivität und Selektivität für Kalium- bzw. Natrium-Ionen jeweils durch eine bestimmte Isomerie der 1,2,3-Triazol-1,4-diyl-Einheit erhöht wird. Des Weiteren wurde gezeigt, dass durch eine erhöhte Einschränkung der N,N-Diethylamino-Gruppe des Fluorophors eine Steigerung der Fluoreszenzquantenausbeute und eine Verschiebung des Emissionsmaximums auf über 500 nm erreicht werden konnte. Die Einführung einer Isopropoxy-Gruppe an einem N-Phenylaza-[18]krone-6-ethers resultierte dabei in einem hoch selektiven Kalium-Ionen Fluoroionophor und ermöglichte eine in vitro Überwachung von 10 – 80 mM Kalium-Ionen. Die Substitution einer Methoxy-Gruppe an einem N-Phenylaza-[15]krone-5-ether kombiniert mit unterschiedlich N,N-Diethylamino-Coumarinen lieferte hingegen zwei Natrium-Ionen Fluoroionophore, die für die Überwachung von intra- bzw. extrazellulären Natrium-Ionen Konzentrationen geeignet sind.
In einem weiteren Schritt wurden N-Phenylaza-[18]krone-6-ether mit einem Fluorophor, basierend auf einem [1,3]-Dioxolo[4,5-f][1,3]benzodioxol-(DBD)-Grundgerüst, funktionalisiert. Die im Anschluss durchgeführten spektroskopischen Untersuchungen ergaben, dass die Isopropoxy-Gruppe in ortho Position des N-Phenylaza-[18]krone-6-ether in einen für extrazelluläre Kalium-Ionen Konzentrationen selektiven Fluoroionophor resultierte, der die Konzentrationsbestimmungen über die Fluoreszenzintensität und -lebensdauer ermöglicht.
In einem abschließenden Schritt konnte unter Verwendung eines Pyrens als fluorophore Gruppe ein weiterer für extrazelluläre Kalium-Ionen Konzentrationen geeigneter Fluoroionophor entwickelt werden. Die Bestimmung der Kalium-Ionen Konzentration erfolgte hierbei anhand der Fluoreszenzintensitätsverhältnisse bei zwei Emissionswellenlängen.
Insgesamt konnten 17 verschiedene neue Fluoroionophore für die Bestimmung von Kalium- bzw. Natrium-Ionen synthetisiert und charakterisiert werden. Sechs dieser neuen Moleküle ermöglichen in vitro Messungen der intra- oder extrazellulären Kalium- und Natrium-Ionen Konzentrationen und könnten zukünftig für in vivo Konzentrationsmessungen verwendet werden.
