Der statistische Diskussionbeitrag untersucht, ob und wie sich Erwartungen und Stimmungen in der Wirtschaft bilden bzw. von welchen volkswirtschaftlichen Größen sie abhängen. Als Methodik werden Partial Least Squares (PLS) Modelle genutzt, eine Modellklasse der Pfadanalyse mit latenten Variablen. Die verwendeten Daten wurden vom Ifo-Institut und aus der amtlichen Statistik entnommen.
Dieser Beitrag reflektiert und ergänzt die aktuelle Diskussion über die Empfehlungen des Wissenschaftsrats zur Weiterentwicklung der Friedens- und Konfliktforschung. Wir richten dabei den Blick auf die vom Wissenschaftsrat attestierten Schwachstellen im Bereich empirisch-analytischer Methoden und erläutern ihre Auswirkungen auf Interdisziplinarität, Internationalität und Politikberatung der deutschen Friedens- und Konfliktforschung. Wir argumentieren, unter Verweis auf den Bericht des Wissenschaftsrats, dass eine breitere Methodenausbildung und -kenntnis von großer Bedeutung für interdisziplinäre und internationale Zusammenarbeit, aber auch für die Politikberatung ist. Zukünftige Initiativen innerhalb der Friedens- und Konfliktforschung sollten die Methodenvielfalt des Forschungsbereichs angemessen berücksichtigen und einen besonderen Fokus auf die Ausbildung im Bereich empirisch-analytischer Methoden legen, um das Forschungsfeld in diesem Bereich zu stärken. Unser Beitrag entspringt einer Diskussion innerhalb des Arbeitskreises „Empirische Methoden der Friedens- und Konfliktforschung“ der Arbeitsgemeinschaft Friedens- und Konfliktforschung.
The active metabolite of vitaminA, retinoic acid (RA), plays an important role in the female reproductive system. The synthesis of RA is tightly regulated by the activity of retinaldehyde dehydrogenases (Raldh). Among these, Raldh1 and Raldh2 exhibit specific temporal and spatial expression patterns in the mouse uterus, both during the oestrous cycle and early pregnancy. In the present study, we have assessed whether oestradiol and progesterone directly influence the uterine expression of Raldh1 and Raldh2 in ovariectomised mice. We investigated the effect of gestagen (promegestone 0.3 mg kg(-1) bodyweight), oestrogen (oestradiol 3 mu g kg(-1) bodyweight) and their combination on the uterine expression of Raldh2. Expression was analysed using in situ hybridisation and quantified using real-time detection reverse transcription-polymerase chain reaction. The results show that the expression of Raldh2 is rapidly (within 1-4 h) induced in stromal cells by oestrogen, but not by gestagen, treatment, whereas combined oestrogen + gestagen treatment leads to a more prolonged (48 h) response. In contrast, oestrogen, but not progesterone, treatment downregulates (within 4 - 24 h) Raldh1 expression in the uterine glandular epithelium. We conclude that the uterine RA concentrations are regulated by oestrogens via an effect on the expression of the Raldh synthesising enzymes. Such a regulation is consistent with the natural fluctuations of Raldh expression during the oestrous cycle, early pregnancy and blastocyst implantation

Collagen is the most abundant protein in mammals. In many tissues, collagen molecules assemble to form a hierarchical structure. In the smallest supramolecular unit, named fibril, each molecule is displaced in the axial direction with respect to its neighbors. This staggering creates a periodic gap and overlap regions, where the gap regions exhibit 20% less density. These fibril-forming collagens play an essential role in the strength of connective tissues. Despite much effort, directed at understanding collagen function and regulation, the influence of the chemical environment on the local structural and mechanical properties remains poorly understood. Recent studies, aimed at elucidating the effect of osmotic pressure, showed that collagen contracts upon water removal. This observation highlights the importance of water for the stabilization and mechanics of the collagen molecule.
Using collagen mimetic peptides (CMPs), which fold into triple helical structures reminiscent of natural collagen, the primary goal of this work was to investigate the effect of the osmotic pressure on specific collagen-mimetic sequences. CMPs were used as the model system as they provide sequence control, which is essential for discriminating local from global structural changes and for relating the observed effects to existing knowledge about the full-length collagen molecule. Of specific interest was the structure of individual collagen triple helices as well as their organization into self-assembled higher order structures. These key structural features were monitored with infrared spectroscopy (IR) and synchrotron X-ray scattering, while varying the osmotic pressure. For controlling the osmotic pressure, CMP powder samples were incubated in air of defined relative humidity, ranging from dry conditions to highly “humid”. In addition, to obtain more biologically relevant conditions, the CMPs were measured in ultrapure water and in solutions containing small molecule osmolytes.
Using the sequences (Pro-Pro-Gly)10, (Pro-Hyp-Gly)10 and (Hyp-Hyp-Gly)10, it was shown that CMPs with different degrees of proline hydroxylation (Hyp = hydroxyproline) exhibit a sequence-specific response to osmotic pressure. IR spectroscopy revealed that osmotic pressure changes affect the strength of the triple helix stabilizing, interchain hydrogen bond and that the extent of this change depends on the degree of hydroxylation. X-ray scattering experiments further showed that changes in osmotic pressure affect both the molecular length as well as the higher order organization of CMPs. Starting from a pseudo-hexagonal packing in the dry state, all three CMPs showed isotropic swelling when increasing the water content to approximately 1.2 water molecules per amino acid, again to different extents depending on the degree of hydroxylation. When increasing the water content further, this pseudo-hexagonal arrangement breaks down. In the fully hydrated state, each CMP is characterized by its own specific and more complex packing geometry.
While these changes in the lateral packing arrangement suggest swelling upon hydration, an overall decrease of the molecular length (i.e. contraction) was observed in the axial direction. Also for this structural feature, a strong dependency on the specific amino acid sequence was found. Interestingly, the observed contraction is the opposite of what has been reported for natural collagen. As (Pro-Pro-Gly)n, (Pro-Hyp-Gly)n and (Hyp-Hyp-Gly)n repeat units are found in collagen with a relatively high abundance, this suggests that other collagen sequence fragments need to respond to hydration in the opposite way to obtain a net elongation of the full-length collagen molecule.
To test this hypothesis, sequences predicted to be sensitive to osmotic pressure were considered. One such sequence, consisting of two repeat units (Ala-Arg-Gly-Ser-Asp-Gly), was inserted as a guest into a (Pro-Pro-Gly) host. When compared to the canonical CMP sequences investigated earlier, the lateral helix packing follows a similar trend with increasing hydration; however, the host-guest CMP axially elongates with increasing water content. This behavior is more similar to what has been found for natural collagen and suggests that different sequences do determine the molecular length of collagen sequences differently. Interestingly, the canonical sequences are more abundant in the overlap region while the guest sequence is found in the gap region. This allows to speculate that sequences in the gap and overlap regions possess a specifically fine-tuned local response to osmotic pressure changes. Clearly, more experiments with additional sequences are needed to confirm this.
In conclusion, the results obtained in this work indicate a highly sequence specific interaction between collagen and water. Osmotic pressure-induced conformational changes mostly originate from local geometries and bonding patterns and affect both the structure of individual triple helices as well as higher order assemblies. One key remaining question is how these conformational changes affect the local mechanical properties of the collagen molecule. As a first step, the stiffness (persistence length) of full-length collagen was determined using atomic force microscopy. In the future, experimental strategies need to be developed that allow for investigating the mechanical properties of specific collagen sequences, e.g. performing single-molecule force spectroscopy of CMPs.
Photon Density Wave (PDW) spectroscopy was applied for temperature dependent monitoring of melting and crystallization of milk fat within homogenized fresh milk. As an in-line process analytical technique, PDW spectroscopy quantifies continuously the optical properties of turbid material, providing an insight into its structural processes. Here, the measured absorption coefficients reflect temperature as well as fat content of milk and the reduced scattering coefficients probe physical changes of the light scattering fat droplets and casein micelles. Thermal processing reveals breakpoints within the temperature trend of the reduced scattering coefficient of fat containing milk. Found at 16 degrees C and 24 degrees C while cooling and heating, respectively, they are associated to the phase transitions of milk fat. Continuous isothermal measurement of the optical coefficients showed that the crystallization process requires several hours. The strongly changing reduced scattering coefficient implies that the thermal history of milk will have a major impact on any method based on light scattering as quantitative analytical technique.
This dissertation was carried out as part of the international and interdisciplinary graduate school StRATEGy. This group has set itself the goal of investigating geological processes that take place on different temporal and spatial scales and have shaped the southern central Andes. This study focuses on claystones and carbonates of the Yacoraite Fm. that were deposited between Maastricht and Dan in the Cretaceous Salta Rift Basin. The former rift basin is located in northwest Argentina and is divided into the sub-basins Tres Cruces, Metán-Alemanía and Lomas de Olmedo. The overall motivation for this study was to gain new knowledge about the evolution of marine and lacustrine conditions during the Yacoraite Fm. Deposit in the Tres Cruces and Metán-Alemanía sub-basins. Other important aspects that were examined within the scope of this dissertation are the conversion of organic matter from Yacoraite Fm. into oil and its genetic relationship to selected oils produced and natural oil spills. The results of my study show that the Yacoraite Fm. began to be deposited under marine conditions and that a lacustrine environment developed by the end of the deposition in the Tres Cruces and Metán-Alemanía Basins. In general, the kerogen of Yacoraite Fm. consists mainly of the kerogen types II, III and II / III mixtures. Kerogen type III is mainly found in samples from the Yacoraite Fm., whose TOC values are low. Due to the adsorption of hydrocarbons on the mineral surfaces (mineral matrix effect), the content of type III kerogen with Rock-Eval pyrolysis in these samples could be overestimated. Investigations using organic petrography show that the organic particles of Yacoraite Fm. mainly consist of alginites and some vitrinite-like particles. The pyrolysis GC of the rock samples showed that the Yacoraite Fm. generates low-sulfur oils with a predominantly low-wax, paraffinic-naphthenic-aromatic composition and paraffinic wax-rich oils. Small proportions of paraffinic, low-wax oils and a gas condensate-generating facies are also predicted. Here, too, mineral matrix effects were taken into account, which can lead to a quantitative overestimation of the gas-forming character.
The results of an additional 1D tank modeling carried out show that the beginning (10% TR) of the oil genesis took place between ≈10 Ma and ≈4 Ma. Most of the oil (from ≈50% to 65%) was generated prior to the development of structural traps formed during the Plio-Pleistocene Diaguita deformation phase. Only ≈10% of the total oil generated was formed and potentially trapped after the formation of structural traps. Important factors in the risk assessment of this petroleum system, which can determine the small amounts of generated and migrated oil, are the generally low TOC contents and the variable thickness of the Yacoraite Fm. Additional risks are associated with a low density of information about potentially existing reservoir structures and the quality of the overburden.
Two sesquiterpenes, corymbolone and mustakone, isolated from the chloroform extract of the rhizomes of Cyperus articulatus, exhibited significant anti-plasmodial properties. Mustakone was approximately ten times more active than corymbolone against the sensitive strains of the Plasmodium falciparum.
A new precursor route for the preparation of bulk oxides and thin films of bismuth vanadates is proposed. The method involves the thermal treatment under air and mild conditions of hybrid organic-inorganic precursors, made from a zwitterionic salt-free polymer matrix and selected inorganic species. Monoclinic BiVO4 was obtained in the form of bulk oxide by calcination of the powdered homogeneous hybrid materials at 600 degrees C, from precursors containing Bi and V in stoichiometric amounts. In the same way, thermodiffractometry studies performed on a hybrid material exhibiting a Bi/ V molar ratio of 2 revealed that the ionic conductor gamma-Bi4V2O11 phase can be stabilized under very soft thermal conditions (300 degrees C). Additionally, thin films of yellow monoclinic BiVO4 were for the first time fabricated, by thermal treatment of the same hybrid polymeric precursors deposited on quartz substrates by spin coating, using a layer- by-layer technique. The presence of the target phase at the surface of the plates was confirmed by X-ray diffraction as well as UV-vis measurements
Homopolymers were prepared from diallylammonium monomers bearing 4-methylcoumarin and 4-cyanobiphenyl as fluorescent and mesogenic side groups, as well as their copolymers with diallyldimethylammonium chloride (DADMAC). Organic-inorganic hybrid films were electrostatically self-assembled via the layer-by-layer technique on silicon wafers and quartz plates from the chromophore-bearing polymers and an exfoliated synthetic hectorite. Photophysical studies performed in solution as well as in the self-assembled films demonstrated only a weak tendency for aggregation of the chromophores in the macromolecules. Moreover, assemblies made from the polymers carrying the cyanobiphenyl mesogen were found to exhibit a pronounced internal order
An unconventional but easily accessible precursor route involving the thermal treatment of hybrid precursors containing an ampholytic polymer matrix is developed to prepare multimetallic oxides of catalytic interest such as transition metal molybdates. A copolymer of diallyldimethylammonium chloride and a functionalized maleamic acid bearing an amine group suited for cation complexation was designed, synthesized and used as a matrix to stabilize inorganic species generated in solution from Ni(NO3)(2)center dot 6H(2)O, Co(NO3)(2)center dot 6H(2)O and/or Mn(NO3)(2)center dot 4H(2)O together with (NH4)(6)Mo(7)O(24)center dot 4H(2)O. UV-vis-NIR as well as C-13-NMR studies suggest that the interactions between the cations and the polymer in solution are mainly electrostatic. Only minor complexation interactions take place under certain conditions. Homogeneous hybrid blends were prepared from these solutions. The presence of a complexing amine group in addition to the charged betaine moieties in the polymer permits stabilization of more than stoichiometric amounts of the metal species in the blends. XRD measurements suggest that the homogeneity in the solid state can be kept up to about 1.5 mol of each metal that is incorporated ( anionic as well as cationic) per mol of repeat units of the copolymer. The blends were calcined under air at 600 degrees C to produce the simple as well as mixed nickel, cobalt and manganese molybdates. Characterization of the final phases by XRD and Raman spectroscopy indicates that the alpha- as well as the beta-molybdate phases can be prepared, and that the mixed structures are solid solutions of the simple NiMoO4, MnMoO4 and CoMoO4. If the precursors engaged are homogeneous, the pH of the precursor solution, the amount of metal that is incorporated in the matrix, and the nature of the polymer matrix seem to exert only a minor influence on the nature of the final phase, which demonstrates the versatility and facile applicability of the method
Hydrophobically substituted diallylamines bearing a hexyl, dodecyl, or octadecyl chain were synthesized and homopolymerized as hydrochlorides. Copolymerixation of the diallylamines with maleic acid produces alternating copolymers. The copolymers behave as amphiphilic polyampholytes and dissolve best in the acidic or in the basic form. Only the colpolymer with the hexyl chain could be dissolved in aqueous solvents and shows hydrophobic associaiton. The copolymers with the longer alkyl chains require polar protic organic solvents. All polymers are amorphous, but show a superstructure in bulk due to their amphiphilicity

Die Bedeutung der Bewegung im Computerspiel wird von vielen Seiten immer wieder betont. Dies ist keinesfalls verwunderlich, wenn man sich einmal die „Verben“ vor Augen führt, die den Handlungsraum klassischer Computerspiele konstituieren: „Laufen“, „Schießen“ und „Fangen“ sollen hier nur als Stellvertreter für ein Repertoire an möglichen Aktionen stehen, die allesamt auf der kinetischen Ebene stattfinden. Diese Handlungen erschöpfen sich aber nicht in sich selbst, sondern stellen auch Sinnzusammenhänge unterhalb der Spielelemente her. In den meisten klassischen Games sind es eben diese kinetischen Relationen, welche die singulären Elemente überhaupt erst zu einem diegetischen Spielraum zusammenfügen, und das Spiel so ermöglichen. Umso verwunderlicher mutet es da an, dass dieser Gestaltungsebene von analytischer Seite bislang so wenig Aufmerksamkeit zu Teil wurde. Mein Aufsatz soll an die Möglichkeiten einer kinetischen Perspektive bei der Betrachtung von Computerspielen heranführen und aufzeigen, welches Potential in einer Analyse wie auch der gestalterischen Nutzung dieser bislang weitgehend vernachlässigten Gestaltungsebene liegen könnte.

Assessing the human and economic threat introduced by sliding or creeping masses is of major importance in landslide hazard assessment and mitigation. Especially, in the densely populated alpine region unstable hillslopes represent a major hazard to men and infrastructure. Detailed knowledge, especially, of the dominant site-specific controlling factors such as subsurface architecture and geology is thereby key in assessing slope vulnerability. In order to quantify the geological variations at a creeping hillslope in the Austrian Alps, we have collected six 2D refraction seismic profiles. We propose using a layer-based inversion strategy to reconstruct P-wave velocity models from first arrival times. Considering the geological complexity at such sites, the selected inversion approach eases the interpretability of geological structures given intrinsic optimization for only a discrete, user-defined, number of layers. As the applied layer-based inversion approach fits our travel time data equally well as traditional smooth inversion approaches, it represents a feasible mean to summarize the structural complexity often present at such sites. Analysis of the inversion results illustrates that bedrock topography clearly deviates from a previously assumed planar surface and exhibits distinct variations across the slope extension. Bedrock topography additionally impacts the intermediate geological units and, thus, this information is critical for further analyses such as geomechanical modeling. (C) 2012 Elsevier B.V. For a detailed characterization of near-surface environments, geophysical techniques are increasingly used to support more conventional point-based techniques such as borehole and direct-push logging. Because the underlying parameter relations are often complex, site-specific, or even poorly understood, a remaining challenging task is to link the geophysical parameter models to the actual geotechnical target parameters measured only at selected points. We propose a workflow based on nonparametric regression to establish functional relationships between jointly inverted geophysical parameters and selected geotechnical parameters as measured, for example, by different borehole and direct-push tools. To illustrate our workflow, we present field data collected to characterize a near-surface sedimentary environment Our field data base includes crosshole ground penetrating radar (GPR), seismic P-, and S-wave data sets collected between 25 m deep boreholes penetrating sand- and gravel dominated sediments. Furthermore, different typical borehole and direct-push logs are available. We perform a global joint inversion of traveltimes extracted from the crosshole geophysical data using a recently proposed approach based on particle swarm optimization. Our inversion strategy allows for generating consistent models of GPR, P-wave, and S-wave velocities including an appraisal of uncertainties. We analyze the observed complex relationships between geophysical velocities and target parameter logs using the alternating conditional expectation (ACE) algorithm. This nonparametric statistical tool allows us to perform multivariate regression analysis without assuming a specific functional relation between the variables. We are able to explain selected target parameters such as characteristic grain size values or natural gamma activity by our inverted geophysical data and to extrapolate these parameters to the inter-borehole plane covered by our crosshole experiments. We conclude that the ACE algorithm is a powerful tool to analyze a multivariate petrophysical data base and to develop an understanding of how a multi-parameter geophysical model can be linked and translated to selected geotechnical parameters.
Assessing uncertainty in refraction seismic traveltime inversion using a global inversion strategy

To analyse and invert refraction seismic travel time data, different approaches and techniques have been proposed. One common approach is to invert first-break travel times employing local optimization approaches. However, these approaches result in a single velocity model, and it is difficult to assess the quality and to quantify uncertainties and non-uniqueness of the found solution. To address these problems, we propose an inversion strategy relying on a global optimization approach known as particle swarm optimization. With this approach we generate an ensemble of acceptable velocity models, i.e., models explaining our data equally well. We test and evaluate our approach using synthetic seismic travel times and field data collected across a creeping hillslope in the Austrian Alps. Our synthetic study mimics a layered near-surface environment, including a sharp velocity increase with depth and complex refractor topography. Analysing the generated ensemble of acceptable solutions using different statistical measures demonstrates that our inversion strategy is able to reconstruct the input velocity model, including reasonable, quantitative estimates of uncertainty. Our field data set is inverted, employing the same strategy, and we further compare our results with the velocity model obtained by a standard local optimization approach and the information from a nearby borehole. This comparison shows that both inversion strategies result in geologically reasonable models (in agreement with the borehole information). However, analysing the model variability of the ensemble generated using our global approach indicates that the result of the local optimization approach is part of this model ensemble. Our results show the benefit of employing a global inversion strategy to generate near-surface velocity models from refraction seismic data sets, especially in cases where no detailed a priori information regarding subsurface structures and velocity variations is available.
Polyplexes, composed of Salmon DNA and very small gold nanoparticles embedded into a dendritic glycopolymer architecture of sugar-modified poly(ethyleneimine) (PEI-Mal) with a molar mass of about 25,000 g/mol, were characterized by dynamic light scattering (DLS), zeta potential measurements, micro differential scanning calorimetry (mu-DSC) and transmission electron microscopy (TEM). The PEI-Mal-entrapped gold nanoparticles of about 2 nm in diameter influence the polyplex formation of the hyperbranched PEI containing bulky maltose, and in consequence the DNA is more compactized in the inner part of spherical polyplex particles of about 150 nm in diameter. The resulting more compact core shell polyplex particles with embedded gold nanoparticles in the outer polymer shell will be used as components in forthcoming gene delivery experiments. Polyplexes between a double-stranded Salmon DNA and hyperbranched poly(ethyleneimine) (PEI) as well as a maltosylated PEI-Mal were incorporated into a gelatin/chitosan hydrogel scaffold. Calorimetric experiments of the polyplexes show a decrease of the melting temperature in presence of PEI and a peak splitting in presence of PEI-Mal, which can be interpreted to a partial compaction of the DNA strands in presence of PEI-Mal. When the polyplexes are incorporated into a gelatin/chitosan scaffold in the swollen state, the DNA melting peaks at 90 and 93 degrees C, respectively, indicate in both cases the release of the DNA at the surface of the hydrogel scaffold in a more compact form. Specific interactions between the PEI-Mal shell and gelatin are responsible for the tuning of the release properties in presence of the maltose units in the hyperbranched PEI.
Polyplexes between Salmon DNA and non-modified hyperbranched poly(ethyleneimines) of varying molar mass, i.e., PEI(5 k) with 5000 g/mol and PEI(25 k) with 25,000 g/mol, and modified PEI(5 k) with maltose units (PEI-Mal) were investigated in dependence on the molar N/P ratio by using dynamic light scattering (DLS), zeta potential measurements, micro differential scanning calorimetry (mu-DSC), scanning-transmission electron microscopy (STEM), and cryo-scanning electron microscopy (cryo-SEM). A reloading of the polyplexes can be observed by adding the unmodified PEI samples of different molar mass. In excess of PEI a morphological transition from core-shell particles (at N/P 8) to loosely packed onion-like polyplexes (at N/P 40) is observed. The shift of the DSC melting peak from 88 degrees C to 76 degrees C indicates a destabilization of the DNA double helix due to the complexation with the unmodified PEI. Experiments with the maltose-modified PEI show a reloading already at a lower N/P ratio. Due to the presence of the sugar units in the periphery of the polycation electrostatic interactions between DNA become weaker, but cooperative H-bonding forces are reinforced. The resulting less-toxic, more compact polyplexes in excess of the PEI-Mal with two melting points and well distributed DNA segments are of special interest for extended gene delivery experiments. We report on the theoretical study of interaction of ionic surfactants with oppositely charged microgel particles in dilute solutions. Two approaches are proposed. Within the first approach, the micellization of the surfactants inside the microgel is taken into account while the second model focuses on the hydrophobic interactions of the surfactant tails with the hydrophobic parts of microgel subchains. It has been shown that microgels effectively absorb surfactant ions. At low surfactant concentration this absorption is realized due to an ion exchange between microgel counterions and surfactant ions. The ion exchange is significantly affected by the amount of the microgel counterions initially trapped within the microgel particles which depends on the size of the microgel, its ionization degree, cross-linking density as well as polymer concentration in the solution. Increase of the surfactant concentration causes contraction of the microgels, which can be realized as either a continuous shrinking or a jump-like collapse transition depending on the system parameters. In the collapsed state additional absorption of surfactants by microgels takes place due to an energy gain from micellization or hydrophobic interactions. This leads to microgel precipitation and successive microgel overcharging at an excess of the surfactant in the solution. The theoretical results are compared with the existing experimental data, in particular, on photosensitive surfactant/microgel complexes.
We present one-dimensional, time-dependent models of the clumps generated by the linedeshadowing instability. In order to follow the clumps out to distances of more than 1000 R∗, we use an efficient moving-box technique. We show that, within the approximations, the wind can remain clumped well into the formation region of the radio continuum.
In mammals, epoxy-polyunsaturated fatty acids (epoxy-PUFA) are enzymatically formed from naturally occurring all-cis PUFA by cytochrome P450 monooxygenases leading to the generation of cis-epoxy-PUFA (mixture of R,S- and S,R-enantiomers). In addition, also non-enzymatic chemical peroxidation gives rise to epoxy-PUFA leading to both, cis- and trans-epoxy-PUFA (mixture of R,R- and S,S-enantiomers). Here, we investigated for the first time trans-epoxy-PUFA and the trans/cis-epoxy-PUFA ratio as potential new biomarker of lipid peroxidation. Their formation was analyzed in correlation with the formation of isoprostanes (IsoP), which are commonly used as biomarkers of oxidative stress. Five oxidative stress models were investigated including incubations of three human cell lines as well as the in vivo model Caenorhabditis elegans with tert-butyl hydroperoxide (t-BOOH) and analysis of murine kidney tissue after renal ischemia reperfusion injury (IRI). A comprehensive set of IsoP and epoxy-PUFA derived from biologically relevant PUFA (ARA, EPA and DHA) was simultaneously quantified by LC-ESI(-)-MS/MS. Following renal IRI only a moderate increase in the kidney levels of IsoP and no relevant change in the trans/cis-epoxy-PUFA ratio was observed. In all investigated cell lines (HCT-116, HepG2 and Caki-2) as well as C. elegans a dose dependent increase of both, IsoP and the trans/cis-epoxy-PUFA ratio in response to the applied t-BOOH was observed. The different cell lines showed a distinct time dependent pattern consistent for both classes of autoxidatively formed oxylipins. Clear and highly significant correlations of the trans/cisepoxy-PUFA ratios with the IsoP levels were found in all investigated cell lines and C. elegans. Based on this, we suggest the trans/cis-epoxy-PUFA ratio as potential new biomarker of oxidative stress, which warrants further investigation.
Permafrost is warming globally, which leads to widespread permafrost thaw and impacts the surrounding landscapes, ecosystems and infrastructure. Especially ice-rich permafrost is vulnerable to rapid and abrupt thaw, resulting from the melting of excess ground ice. Local remote sensing studies have detected increasing rates of abrupt permafrost disturbances, such as thermokarst lake change and drainage, coastal erosion and RTS in the last two decades. All of which indicate an acceleration of permafrost degradation.
In particular retrogressive thaw slumps (RTS) are abrupt disturbances that expand by up to several meters each year and impact local and regional topographic gradients, hydrological pathways, sediment and nutrient mobilisation into aquatic systems, and increased permafrost carbon mobilisation. The feedback between abrupt permafrost thaw and the carbon cycle is a crucial component of the Earth system and a relevant driver in global climate models. However, an assessment of RTS at high temporal resolution to determine the dynamic thaw processes and identify the main thaw drivers as well as a continental-scale assessment across diverse permafrost regions are still lacking.
In northern high latitudes optical remote sensing is restricted by environmental factors and frequent cloud coverage. This decreases image availability and thus constrains the application of automated algorithms for time series disturbance detection for large-scale abrupt permafrost disturbances at high temporal resolution. Since models and observations suggest that abrupt permafrost disturbances will intensify, we require disturbance products at continental-scale, which allow for meaningful integration into Earth system models.
The main aim of this dissertation therefore, is to enhance our knowledge on the spatial extent and temporal dynamics of abrupt permafrost disturbances in a large-scale assessment. To address this, three research objectives were posed:
1. Assess the comparability and compatibility of Landsat-8 and Sentinel-2 data for a combined use in multi-spectral analysis in northern high latitudes.
2. Adapt an image mosaicking method for Landsat and Sentinel-2 data to create combined mosaics of high quality as input for high temporal disturbance assessments in northern high latitudes.
3. Automatically map retrogressive thaw slumps on the landscape-scale and assess their high temporal thaw dynamics.
We assessed the comparability of Landsat-8 and Sentinel-2 imagery by spectral comparison of corresponding bands. Based on overlapping same-day acquisitions of Landsat-8 and Sentinel-2 we derived spectral bandpass adjustment coefficients for North Siberia to adjust Sentinel-2 reflectance values to resemble Landsat-8 and harmonise the two data sets. Furthermore, we adapted a workflow to combine Landsat and Sentinel-2 images to create homogeneous and gap-free annual mosaics. We determined the number of images and cloud-free pixels, the spatial coverage and the quality of the mosaic with spectral comparisons to demonstrate the relevance of the Landsat+Sentinel-2 mosaics. Lastly, we adapted the automatic disturbance detection algorithm LandTrendr for large-scale RTS identification and mapping at high temporal resolution. For this, we modified the temporal segmentation algorithm for annual gradual and abrupt disturbance detection to incorporate the annual Landsat+Sentinel-2 mosaics. We further parametrised the temporal segmentation and spectral filtering for optimised RTS detection, conducted further spatial masking and filtering, and implemented a binary object classification algorithm with machine-learning to derive RTS from the LandTrendr disturbance output. We applied the algorithm to North Siberia, covering an area of 8.1 x 106 km2.
The spectral band comparison between same-day Landsat-8 and Sentinel-2 acquisitions already showed an overall good fit between both satellite products. However, applying the acquired spectral bandpass coefficients for adjustment of Sentinel-2 reflectance values, resulted in a near-perfect alignment between the same-day images. It can therefore be concluded that the spectral band adjustment succeeds in adjusting Sentinel-2 spectral values to those of Landsat-8 in North Siberia.
The number of available cloud-free images increased steadily between 1999 and 2019, especially intensified after 2016 with the addition of Sentinel-2 images. This signifies a highly improved input database for the mosaicking workflow. In a comparison of annual mosaics, the Landsat+Sentinel-2 mosaics always fully covered the study areas, while Landsat-only mosaics contained data-gaps for the same years. The spectral comparison of input images and Landsat+Sentinel-2 mosaic showed a high correlation between the input images and the mosaic bands, testifying mosaicking results of high quality. Our results show that especially the mosaic coverage for northern, coastal areas was substantially improved with the Landsat+Sentinel-2 mosaics. By combining data from both Landsat and Sentinel-2 sensors we reliably created input mosaics at high spatial resolution for comprehensive time series analyses.
This research presents the first automatically derived assessment of RTS distribution and temporal dynamics at continental-scale. In total, we identified 50,895 RTS, primarily located in ice-rich permafrost regions, as well as a steady increase in RTS-affected areas between 2001 and 2019 across North Siberia. From 2016 onward the RTS area increased more abruptly, indicating heightened thaw slump dynamics in this period. Overall, the RTS-affected area increased by 331 % within the observation period. Contrary to this, five focus sites show spatiotemporal variability in their annual RTS dynamics, alternating between periods of increased and decreased RTS development. This suggests a close relationship to varying thaw drivers. The majority of identified RTS was active from 2000 onward and only a small proportion initiated during the assessment period. This highlights that the increase in RTS-affected area was mainly caused by enlarging existing RTS and not by newly initiated RTS.
Overall, this research showed the advantages of combining Landsat and Sentinel-2 data in northern high latitudes and the improvements in spatial and temporal coverage of combined annual mosaics. The mosaics build the database for automated disturbance detection to reliably map RTS and other abrupt permafrost disturbances at continental-scale. The assessment at high temporal resolution further testifies the increasing impact of abrupt permafrost disturbances and likewise emphasises the spatio-temporal variability of thaw dynamics across landscapes. Obtaining such consistent disturbance products is necessary to parametrise regional and global climate change models, for enabling an improved representation of the permafrost thaw feedback.

Remote sensing provides the means to detect, map, and quantify these changes homogeneously across large regions and time scales. Existing Landsat-based algorithms assess different types of disturbances with similar spatiotemporal requirements. However, Landsat-based analyses are restricted in northern high latitudes due to the long repeat interval and frequent clouds, in particular at Arctic coastal sites. We therefore propose to combine Landsat and Sentinel-2 data for enhanced data coverage and present a combined annual mosaic workflow, expanding currently available algorithms, such as LandTrendr, to achieve more reliable time series analysis. We exemplary test the workflow for twelve sites across the northern high latitudes in Siberia. We assessed the number of images and cloud-free pixels, the spatial mosaic coverage and the mosaic quality with spectral comparisons. The number of available images increased steadily from 1999 to 2019 but especially from 2016 onward with the addition of Sentinel-2 images. Consequently, we have an increased number of cloud-free pixels even under challenging environmental conditions, which then serve as the input to the mosaicking process. In a comparison of annual mosaics, the Landsat+Sentinel-2 mosaics always fully covered the study areas (99.9–100 %), while Landsat-only mosaics contained data-gaps in the same years, only reaching coverage percentages of 27.2 %, 58.1 %, and 69.7 % for Sobo Sise, East Taymyr, and Kurungnakh in 2017, respectively. The spectral comparison of Landsat image, Sentinel-2 image, and Landsat+Sentinel-2 mosaic showed high correlation between the input images and mosaic bands (e.g., for Kurungnakh 0.91–0.97 between Landsat and Landsat+Sentinel-2 mosaic and 0.92–0.98 between Sentinel-2 and Landsat+Sentinel-2 mosaic) across all twelve study sites, testifying good quality mosaic results. Our results show that especially the results for northern, coastal areas was substantially improved with the Landsat+Sentinel-2 mosaics. By combining Landsat and Sentinel-2 data we accomplished to create reliably high spatial resolution input mosaics for time series analyses. Our approach allows to apply a high temporal continuous time series analysis to northern high latitude permafrost regions for the first time, overcoming substantial data gaps, and assess permafrost disturbance dynamics on an annual scale across large regions with algorithms such as LandTrendr by deriving the location, timing and progression of permafrost thaw disturbances
The Arctic-Boreal regions experience strong changes of air temperature and precipitation regimes, which affect the thermal state of the permafrost. This results in widespread permafrost-thaw disturbances, some unfolding slowly and over long periods, others occurring rapidly and abruptly. Despite optical remote sensing offering a variety of techniques to assess and monitor landscape changes, a persistent cloud cover decreases the amount of usable images considerably. However, combining data from multiple platforms promises to increase the number of images drastically. We therefore assess the comparability of Landsat-8 and Sentinel-2 imagery and the possibility to use both Landsat and Sentinel-2 images together in time series analyses, achieving a temporally-dense data coverage in Arctic-Boreal regions. We determined overlapping same-day acquisitions of Landsat-8 and Sentinel-2 images for three representative study sites in Eastern Siberia. We then compared the Landsat-8 and Sentinel-2 pixel-pairs, downscaled to 60 m, of corresponding bands and derived the ordinary least squares regression for every band combination. The acquired coefficients were used for spectral bandpass adjustment between the two sensors. The spectral band comparisons showed an overall good fit between Landsat-8 and Sentinel-2 images already. The ordinary least squares regression analyses underline the generally good spectral fit with intercept values between 0.0031 and 0.056 and slope values between 0.531 and 0.877. A spectral comparison after spectral bandpass adjustment of Sentinel-2 values to Landsat-8 shows a nearly perfect alignment between the same-day images. The spectral band adjustment succeeds in adjusting Sentinel-2 spectral values to Landsat-8 very well in Eastern Siberian Arctic-Boreal landscapes. After spectral adjustment, Landsat and Sentinel-2 data can be used to create temporally-dense time series and be applied to assess permafrost landscape changes in Eastern Siberia. Permafrost is warming globally which leads to widespread permafrost thaw. Particularly ice-rich permafrost is vulnerable to rapid thaw and erosion, impacting whole landscapes and ecosystems. Retrogressive thaw slumps (RTS) are abrupt permafrost disturbances that expand by several meters each year and lead to an increased soil organic carbon release. Local Remote Sensing studies identified increasing RTS activity in the last two decades by increasing number of RTS or heightened RTS growth rates. However, a large-scale assessment across diverse permafrost regions and at high temporal resolution allowing to further determine RTS thaw dynamics and its main drivers is still lacking. In this study we apply the disturbance detection algorithm LandTrendr for automated large-scale RTS mapping and high temporal thaw dynamic assessment to North Siberia (8.1 x 106km2). We adapted and parametrised the temporal segmentation algorithm for abrupt disturbance detection to incorporate Landsat+Sentinel-2 mosaics, conducted spectral filtering, spatial masking and filtering, and a binary machine-learning object classification of the disturbance output to separate between RTS and false positives (F1 score: 0.609). Ground truth data for calibration and validation of the workflow was collected from 9 known RTS cluster sites using very highresolution RapidEye and PlanetScope imagery. Our study presents the first automated detection and assessment of RTS and their temporal dynamics at largescale for 2001-2019. We identified 50,895 RTS and a steady increase in RTS-affected area from 2001 to 2019 across North Siberia, with a more abrupt increase from 2016 onward. Overall the RTS-affected area increased by 331% compared to 2000 (2000: 20,158 ha, 2001-2019: 66,699 ha). Contrary to this, 5 focus sites show spatiotemporal variability in their annual RTS dynamics, with alternating periods of increased and decreased RTS development, indicating a close relationship to thaw drivers. The majority of identified RTS was active from 2000 onward and only a small proportion initiated during the assessment period, indicating that the increase in RTS-affected area was mainly caused by enlarging existing RTS and not by new RTS. The detected increase in RTS dynamics suggests advancing permafrost thaw and underlines the importance of assessing abrupt permafrost disturbances with high spatial and temporal resolution at large-scales. Obtaining such consistent disturbance products will help to parametrise regional and global climate change models.

In probabilistic seismic-hazard analysis, epistemic uncertainties are commonly treated within a logic-tree framework in which the branch weights express the degree of belief of an expert in a set of models. For the calculation of the distribution of hazard curves, these branch weights represent subjective probabilities. A major challenge for experts is to provide logically consistent weight estimates (in the sense of Kolmogorovs axioms), to be aware of the multitude of heuristics, and to minimize the biases which affect human judgment under uncertainty. We introduce a platform-independent, interactive program enabling us to quantify, elicit, and transfer expert knowledge into a set of subjective probabilities by applying experimental design theory, following the approach of Curtis and Wood (2004). Instead of determining the set of probabilities for all models in a single step, the computer-driven elicitation process is performed as a sequence of evaluations of relative weights for small subsets of models. From these, the probabilities for the whole model set are determined as a solution of an optimization problem. The result of this process is a set of logically consistent probabilities together with a measure of confidence determined from the amount of conflicting information which is provided by the expert during the relative weighting process. We experiment with different scenarios simulating likely expert behaviors in the context of knowledge elicitation and show the impact this has on the results. The overall aim is to provide a smart elicitation technique, and our findings serve as a guide for practical applications.
The dynamics of tail-like current sheets under the influence of small-scale plasma turbulence

A 2D-magnetohydrodynamic model of current-sheet dynamics caused by anomalous electrical resistivity as result of small-scale plasma turbulence is proposed. The anomalous resistivity is assumed to be proportional to the square of the gradient of the magnetic pressure as may be valid for instance in the case of lower-hybrid-drift turbulence. The initial resistivity pulse is given. Then the temporal and spatial evolution of the magnetic and electric fields, plasma density, pressure, convection and resistivity are considered. The motion of the induced electric field is discussed as indicator of the plasma disturbances. The obtained results found using much improved numerical methods show a magnetic field evolution with x-line formation and plasma acceleration. Besides, in the current sheet, three types of magnetohydrodynamic waves occur, fast magnetoacoustic waves of compression and rarefaction as well as slow magnetoacoustic waves.
The author's recently published monograph on Alexander von Humboldt[1] describes the multiple images of this great cultural icon. The book is a metabiographical study that shows how from the middle of the nineteenth century to the present day Humboldt has served as a nucleus of crystallisation for a variety of successive socio-political ideologies, each producing its own distinctive representation of him. The historiographical implications of this biographical diversity are profound and support current attempts to understand historical scholarship in terms of memory cultures.

Zeitgleich erfuhr die britische Gesellschaft eine nie dagewesene Demokratisierung. Die Arbeit versucht darzulegen, wie auch vermeintlich arkane Bereiche staatlichen Handelns in öffentliche Aushandlungsprozesse eingebettet sind und rekonstruiert deshalb erstmals systematisch öffentliche und fachöffentliche Diskurse über Nachrichtendienste Großbritanniens im Zeitalter der Weltkriege.

The peace processes in Liberia and Sierra Leone share similar contexts and have an interrelated history. They are also often portrayed as successful cases of peacebuilding. This conclusion seems valid, as war has not returned, and political power was handed over peacefully; however, both cases differ with regard to the inclusiveness of the peace processes and the role of local leaders. This article aims to add to the critical peacebuilding debate by focusing on local perceptions about the position of local leaders in these two peace processes. We conducted a public opinion survey in five regions in Sierra Leone and Liberia and expert interviews with peacebuilding actors to examine changing perceptions about the roles of local leaders in both countries. This article speaks to the broader peacebuilding debate by highlighting the importance of including local voices in the peace process and by discussing challenges of inclusive peacebuilding.
Der Beitrag beleuchtet jüngere Entwicklungen der unbefugten Verwendung von Daten im Sinne des § 263 a StGB und versucht, die unterschiedlichen Strömungen beispielsnah einzufangen, um die Schwierigkeiten des Computerbetrugs in der Fallbehandlung aufzuzeigen sowie davon ausgehend darzulegen, wie die eigene Meinung in der Klausur argumentativ zu stützen ist.
