Contrastive focus

The article puts forward a discourse-pragmatic approach to the notoriously evasive phenomena of contrastivity and emphasis. It is argued that occurrences of focus that are treated in terms of ‘contrastive focus’, ‘kontrast’ (Vallduví & Vilkuna 1998) or ‘identificational focus’ (É. Kiss 1998) in the literature should not be analyzed in familiar semantic terms like introduction of alternatives or exhaustivity. Rather, an adequate analysis must take into account discourse-pragmatic notions like hearer expectation or discourse expectability of the focused content in a given discourse situation. The less expected a given content is judged to be for the hearer, relative to the Common Ground, the more likely a speaker is to mark this content by means of special grammatical devices, giving rise to emphasis.
We compute the local spectrum of the magnetic field near a metallic microstructure at finite temperature. Our main focus is on deviations from a plane-layered geometry for which we review the main properties. Arbitrary geometries are handled with the help of numerical calculations based on surface integral equations. The magnetic noise shows a significant polarization anisotropy above flat wires with finite lateral width, in stark contrast to an infinitely wide wire. Within the limits of a two-dimensional setting, our results provide accurate estimates for loss and dephasing rates in so-called `atom chip traps' based on metallic wires. A simple approximation based on the incoherent summation of local current elements gives qualitative agreement with the numerics, but fails to describe current correlations among neighboring objects.
[1] Observations of hydrological response often exhibit considerable scatter that is difficult to interpret. In this paper, we examine runoff production of 53 sprinkling experiments on the water-repellent soils in the southern Alps of Switzerland; simulated plot scale tracer transport in the macroporous soils at the Weiherbach site, Germany; and runoff generation data from the 2.3-km(2) Tannhausen catchment, Germany, that has cracking soils. The response at the three sites is highly dependent on the initial soil moisture state as a result of the threshold dynamics of the systems. A simple statistical model of threshold behavior is proposed to help interpret the scatter in the observations. Specifically, the model portrays how the inherent macrostate uncertainty of initial soil moisture translates into the scatter of the observed system response. The statistical model is then used to explore the asymptotic pattern of predictability when increasing the number of observations, which is normally not possible in a field study. Although the physical and chemical mechanisms of the processes at the three sites are different, the predictability patterns are remarkably similar. Predictability is smallest when the system state is close to the threshold and increases as the system state moves away from it. There is inherent uncertainty in the response data that is not measurement error but is related to the observability of the initial conditions.
CAMPHOR: A GOOD MODEL FOR ILLUSTRATING NMR TECHNIQUES. The use of Nuclear Magnetic Resonance spectroscopy to establish the three-dimensional structures of molecules is an important component of modern Chemistry courses. The combination of techniques that can be used for this purpose is conveniently illustrated by their application to the camphor molecule. This paper presents applications of several techniques used in NMR spectral interpretation in an increasing order of complexity. The result of individual experiments is illustrated in order to familiarize the user with the way connectivity through bonds and through space is established from 1D/2D-NMR spectra and molecular stereochemistry is determined from different NMR experiments
Electrically charged cellular polymer films can exhibit very high piezoelectric activity and are therefore more and more often employed in advanced electromechanical and electro-acoustical transducers. In this paper, we report an optimized sequence of steps for preparing such ferroelectrets from commercial nonvoided ploy(ethylene terephthalate) (PETP) films by means of foaming with CO2 biaxial mechanical stretching, controlled void inflation, and bipolar electric charging. The nonvoid PETP films foamed with supercritical CO2 at a suitably high pressure and subsequently annealed for stabilization. The cellular foam structure was further optimized by means of well controlled biaxial stretching in a commercial stretcher and sometimes subsequent inflamation in a pressure chamber. Bipolar electric charging of the internal voids was achieved through the application of high electric fields in an SF0 atmosphere. The new optimized PETP ferroelectric exhibit quite large piezoelectric coefficients up to almost 500 pCN(-1), for which unusually low elastic stiffness of only around 0.3 MPa are essential. The PETP foam ferroelectrics posses unclamped thickenss-extension resonance frequences between approximately 120 and 250 kHz, and are thus highly suitable for several established as well as novel ultrasonic-transductant applications.
The paper presents a simulation and parameter-estimation approach for evaluating stochastic patterns of population growth and spread of an annual forest herb, Melampyrum pratense (Orobanchaceae). The survival of a species during large-scale changes in land use and climate will depend, to a considerable extent, on its dispersal and colonisation abilities. Predictions on species migration need a combination of field studies and modelling efforts. Our study on the ability of M. pratense to disperse into so far unoccupied areas is based on experiments in secondary woodland in NE Germany. Experiments started in 1997 at three sites where the species was not yet present, with 300 seeds sown within 1m2. Population development was then recorded until 2001 by mapping of individuals with a resolution of 5 cm. Additional observations considered density dependence of seed production. We designed a spatially explicit individual-based computer simulation model to explain the spatial patterns of population development and to predict future population spread. Besides primary drop of seeds (barochory) it assumed secondary seed transport by ants (myrmecochory) with an exponentially decreasing dispersal tail. An important feature of population-pattern explanation was the simultaneous estimation of both population-growth and dispersal parameters from consistent spatio-temporal data sets. As the simulation model produced stochastic time series and random spatially discrete distributions of individuals we estimated parameters by minimising the expectation of weighted sum of squares. These sums of squares criteria considered population sizes, radial population distributions around the area of origin and distributions of individuals within squares of 25cm×25 cm, the range of density action. Optimal parameter values, together with the precision of the estimates, were obtained from calculating sum of squares in regular grids of parameter values. Our modelling results showed that transport of fractions of seeds by ants over distances of 1-2m was indispensable for explaining the observed population spread that led to distances of at most 8mfrom population origin within 3 years. Projections of population development over four additional years gave a diffusion-like increase of population area without any "outposts". This prediction generated by the simulation model gave a hypothesis which should be revised by additional field observations. Some structural deviations between observations and model output already indicated that for full understanding of population spread the set of dispersal mechanisms assumed in the model may have to be extended by additional features of plant-animal mutualism.
A tight hormonal control of energy homeostasis is of pivotal relevance for animals. Recent evidence suggests an involvement of the nuclear receptor NR1i3 (CAR). Fasting induces CAR by largely unknown mechanisms and CAR-deficient mice are defective in fasting adaptation. In rat hepatocytes CAR was induced by WY14643, a PPARalpha-agonist. A DR1 motif in the CAR promoter was necessary and sufficient for this control. The PPARalpha-dependent increase in CAR potentiated the phenobarbital-induced transcription of the prototypical CAR-dependent gene CYP2B1. Since free fatty acids are natural ligands for PPARalpha, a fasting-induced increase in free fatty acids might induce CAR. In accordance with this hypothesis, CAR induction by fasting was abrogated in PPARalpha-deficient mice.
The mode of nutrition of mixotrophic flagellates determines the food quality for their consumers

Both the stoichiometry and complex stability constants of crown ether complexes with metal ions have been determined by examining gradual changes in their diffusional behavior in nonaqueous solution. Diffusion coefficients, D, were evaluated by pulsed field gradient (PFG) NMR titration experiments whilst complex stability constants were determined by nonlinear curve-fitting procedures, D versus csol., which also allow the treatment of multiple complexation equilibria (1:1 to 1:2 stoichiometries). Differences in the diffusion coefficients of the various free crown ethers with respect to their metal ion complexes indicate great sensitivity to both conformational changes and changes in molecular size upon complexation.
An investigation of the conformational landscape of 1,3-dithian-2-yl bearing porphyrins and the rotational behavior of the dithianyl substituents in meso position was carried out by variable-temperature (VT) NMR spectroscopy. Additionally, theoretical results for alternative conformations and energy barriers were obtained by molecular modeling. The study revealed different NH trans tautomers with regard to the orientation of the dithianyl ligands for the free base porphyrins 1-3. Relatively ruffled porphyrin core conformations were established for the transition states of the dithianyl rotation, resulting in a lower rotational energy barrier for the nickel(II) complex 4 compared to that of the free base systems. The data obtained and the first depiction of a rotational transition state for the rotation of bulky meso-alkyl substituents illustrate the close structural interplay between meso-alkyl substituents and the macrocycle conformation in porphyrins.
Phonology and intonation

Using a serial search paradigm, we observed several effects of within-object fixation position on spatial and temporal control of eye movements: the preferred viewing location, launch site effect, the optimal viewing position, and the inverted optimal viewing position of fixation duration. While these effects were first identified by eye-movement studies in reading, our approach permits an analysis of the functional relationships between the effects in a different paradigm. Our results demonstrate that the fixation position is an important predictor of the subsequent saccade by influencing both fixation duration and the selection of the next saccade target.
While the Information Structure (IS) is most naturally interpreted as 'structure of information', some may argue that it is structure of something else, and others may object to the use of the word 'structure'. This paper focuses on the question of whether the informational component can have structural properties such that it can be called 'structure'. The preliminary conclusion is that, although there are some vague indications of structurehood in it, it is perhaps better understood to be a representation that encodes a finite set of information-based partitions, rather than structure.
The most recent trend in the studies of LF intervention effects makes crucial reference to focusing effects on the interveners, and this paper critically examines the representative analyses of the focus-based approach. While each analysis has its own merits and shortcomings, I argue that a pragmatic analysis that does not make appeal to syntactic configurations is better equipped to deal with many of the complex and delicate facts surrounding intervention effects.
Spring algal development in deep temperate lakes is thought to be strongly influenced by surface irradiance, vertical mixing and temperature, all of which are expected to be altered by climate change. Based on long-term data from Lake Constance, we investigated the individual and combined effects of these variables on algal dynamics using descriptive statistics, multiple regression models and a processoriented dynamic simulation model. The latter considered edible and less-edible algae and was forced by observed or anticipated irradiance, temperature and vertical mixing intensity. Unexpectedly, irradiance often dominated algal net growth rather than vertical mixing for the following reason: algal dynamics depended on algal net losses from the euphotic layer to larger depth due to vertical mixing. These losses strongly depended on the vertical algal gradient which, in turn, was determined by the mixing intensity during the previous days, thereby introducing a memory effect. This observation implied that during intense mixing that had already reduced the vertical algal gradient, net losses due to mixing were small. Consequently, even in deep Lake Constance, the reduction in primary production due to low light was often more influential than the net losses due to mixing. In the regression model, the dynamics of small, fast-growing algae was best explained by vertical mixing intensity and global irradiance, whereas those of larger algae were best explained by their biomass 1 week earlier. The simulation model additionally revealed that even in late winter grazing may represent an important loss factor during calm periods when losses due to mixing are small. The importance of losses by mixing and grazing changed rapidly as it depended on the variable mixing intensity. Higher temperature, lower global irradiance and enhanced mixing generated lower algal biomass and primary production in the dynamic simulation model. This suggests that potential consequences of climate change may partly counteract each other.
Regulation of planktonic ciliate dynamics and functional composition during spring in Lake Constance

Protozoans are among the most important grazers of phytoplankton and remineralizers of nutrients in marine and freshwater ecosystems, but less is known about the regulation of their population dynamics. We analyzed a 12 yr data set of ciliate biomass and species composition in large, deep Lake Constance to understand the factors influencing ciliate spring development. The start of ciliate net growth in spring was closely linked to that of edible algae, chlorophyll a and the vertical mixing intensity, but independent of water temperature. During ciliate spring growth, the relative contribution of ciliated interception feeders was positively related to that of cryptomonads, whereas the relative contribution of filter feeders correlated positively with that of non-cryptomonads. The duration of ciliate dominance in spring was largely controlled by the highly variable onset of the phytoplankton bloom, as the termination of the ciliate bloom was less variable. During years with an extended spring bloom of algae and ciliates, internally forced species shifts were observed in both communities. Interception feeders alternated with filter feeders in their relative importance as did cryptomonads and non-cryptomonads. Extended spring blooms were observed when vertical mixing intensity was low at low temperatures during early spring, which will become less likely under the anticipated climate change scenarios. The termination of the ciliate spring bloom occurred prior to a reduction in food concentration and mostly also prior to the mass development of daphnids alone, but coincided with increased grazing by various predators together, such as rotifers, copepods and daphnids in late May/early June.
We present projects for future space missions using new quantum devices based on ultracold atoms. They will enable fundamental physics experiments testing quantum physics, physics beyond the standard model of fundamental particles and interactions, special relativity, gravitation and general relativity.
Chlamydomonas acidophila, a dominant phytoplankton species in the very acidic Lake 111 (pH 2.7) situated in Germany, faces low concentrations of inorganic phosphorus (P-i), inorganic carbon (C-i) and potassium (K+) in its environment, which may lead to a complex colimitation by these nutrients. We performed laboratory and field investigations to test for P-i limitation and its dependence on C-i and K+ concentrations. The minimum cell quota for phosphorus (Q(0)) and phosphatase enzyme activity were similar to those for neutrophilic algae, despite the low pH and high concentrations of iron and aluminium, indicating no extra metabolic costs or inhibition of enzymes by the extreme environment. The threshold concentration of soluble reactive phosphorus for growth (SRPt), the algal C:P ratio and the alkaline phosphatase enzyme activity all suggested a moderate P-i limitation of C. acidophila in Lake 111. SRPt and Q(0) were higher at low CO2 and K+ concentrations in culture, showing a relationship between C-i and P-i acquisition. Furthermore, SRPt and Q(0) were higher under K+/P-i-colimiting conditions than under P-i-limiting conditions alone, suggesting that K+ concentrations influence P-i limitation in C. acidophila as well. Our results show that a limitation by one macronutrient requires consideration of the availability of the others as their uptake mechanisms depend on each other. Notwithstanding these interactions, C-i or K+ concentrations had no clear influence on the P-i limitation of C. acidophila in Lake 111.
Chlamydomonas acidophila faces high heavy-metal concentrations in acidic mining lakes, where it is a dominant phytoplankton species. To investigate the importance of metals to C. acidophila in these lakes, we examined the response of growth, photosynthesis, cell structure, heat-shock protein (Hsp) accumulation, and metal adsorption after incubation in metal-rich lake water and artificial growth medium enriched with metals (Fe, Zn). Incubation in both metal-rich lake water and medium caused large decreases in photosystem II function (though no differences among lakes), but no decrease in growth rate (except for medium + Fe). Concentrations of small Hsps were higher in algae incubated in metal-rich lake- water than in metal-enriched medium, whereas Hsp60 and Hsp70A were either less or equally expressed. Cellular Zn and Fe contents were lower, and metals adsorbed to the cell surface were higher, in lake-water-incubated algae than in medium- grown cells. The results indicate that high Zn or Fe levels are likely not the main or only contributor to the low primary production in mining lakes, and multiple adaptations of C. acidophila (e.g., high Hsp levels, decreased metal accumulation) increase its tolerance to metals and permit survival under such adverse environmental conditions. Supposedly, the main stress factor present in the lake water is an interaction between low P and high Fe concentrations.

Multinuclear dynamic NMR spectroscopy of 3,5-bis(trifluoromethylsulfonyl)-1,3,5-oxadiazinane (3) revealed the existence of two conformers with differently oriented CF3 groups with respect to the ring, and two dynamic processes: ring inversion and restricted rotation about the N-S bond. Two transition states connecting the two conformers and corresponding to clockwise and counterclockwise rotations about the N-S bond were found; the calculated activation barriers of about 12 kcal/mol are in excellent agreement with those measured experimentally for the related molecule 1,3,5-tris(trifluoromethylsulfonyl)-1,3,5-triazinane (1). X-ray analysis proved the existence of the symmetric isomer of 3, which is the minor isomer in solutions but the only one in the crystal due to packing effects. The normal Perlin effect (JCHax < JCHeq)observed for 2(6)-CH2 in 3, whereas the reversed Perlin effect was found for the 4-CH2 group in 3 as well as for all CH2 groups in 1 both experimentally and theoretically. The latter effect in compounds 1, 3, and 1- (methylsulfonyl)-3,5-bis(trifluoromethylsulfonyl)-1,3,5-triazinane (2) can be considered as a genuine reverse Perlin effect since larger values of 1JCH are observed for longer C-H bonds.
Lists as embedded structures and the prosody of list construction as an interactional resource

After giving an overview of the treatment of lists in the literature, I describe lists in German talk-in- interaction. I show that, apart from the preference for three-part lists described by Jefferson (1990), lists are embedded in a larger three-component structure that the list is the middle part of. For lists proper, I suggest to differentiate between closed and open lists that are produced with different kinds of practices. It is the prosody that is used to suggest the list as made up of a closed or an open number of list items, irrespective of its syntactic embedding. I then concentrate on open lists, in particular their intonation. Open lists may be produced with different kinds of, albeit similar, intonation contours. But it is not so much the particular intonation contour that is constitutive of lists, but a variety of similar contours plus the repetition of the chosen contour for at least some or even all of the list items. Furthermore, intonation is deployed to suggest the interpretation of a potential final list item as either a designed list completer or as another designed item of the list. The design of this final list item as a completer or as another list item is used as a practice to signal the non-completion or completion of the list proper. But even after completing the list proper, the larger three-component structure also has to be closed in order to embed and accommodate the list into the surrounding sequential interaction. For the analysis of the practices of list construction I am concentrating on the role of prosody, especially intonation, giving evidence to show that intonation is indeed one of the methodically used constitutive cues that makes the production and structuring of lists recognizable for recipients.
Cloning and characterization of a single chain antibody to glucose oxidase from a murine hybridoma

Glucose oxidase (GOD) is an oxidoreductase catalyzing the reaction of glucose and oxygen to peroxide and gluconolacton (EC 1.1.3.4.). GOD is a widely used enzyme in biotechnology. Therefore the production of monoclonal antibodies and antibody fragments to GOD are of interest in bioanalytics and even tumor therapy. We describe here the generation of a panel of monoclonal antibodies to native and heat inactivated GOD. One of the hybridomas, E13BC8, was used for cloning of a single chain antibody (scFv). This scFv was expressed in Escherichia coli XL1-blue with the help of the vector system pOPE101. The scFv was isolated from the periplasmic fraction and detected by western blotting. It reacts specifically with soluble active GOD but does not recognize denatured GOD adsorbed to the solid phase. The same binding properties were also found for the monoclonal antibody E13BC8.
The paper investigates focus marking devices in the scarcely documented North-Ghanaian Gur language Konkomba. The two particles lé and lá occur under specific focus conditions and are therefore regarded as focus markers in the sparse literature. Comparing the distribution and obligatoriness of both alleged focus markers however, I show that one of the particles, lé, is better analyzed as a connective particle, i.e. as a syntactic rather than as a genuine pragmatic marker, and that comparable syntactic focus marking strategies for sentence-initial constituents are also known from related languages.
Design Issues in the Implementation of MPI2 One Sided Communication in Ethernet based Networks

In current research, one sided communication of the MPI2 standard is pushed as a promising technique [6, 7, 10, 18]. But measurements of applications and MPI2 primitives show a different picture [17]. In this paper we analyze de sign issues of MPI2 one sided communication and its im plementations. We focus on asynchronous communication for parallel applications in Ethernet cluster environments. Further, one sided communication is compared to two sided communication. This paper will prove that the key problem to performance is not only the implementation of MPI2 one sided communication - it is the design.
The field equations following from a Lagrangian L(R) will be deduced and solved for special cases. If L is a non-linear function of the curvature scalar, then these equations are of fourth order in the metric. In the introduction we present the history of these equations beginning with the paper of H. Weyl from 1918, who first discussed them as alternative to Einstein's theory. In the third part, we give details about the cosmic no hair theorem, i.e., the details how within fourth order gravity with L= R + R^2 the inflationary phase of cosmic evolution turns out to be a transient attractor. Finally, the Bicknell theorem, i.e. the conformal relation from fourth order gravity to scalar- tensor theory, will be shortly presented.
Aims: To investigate whether concurrent alcohol and tobacco use during early adolescence characterizes a subgroup that differs from users of one substance only regarding several risk factors for later substance use problems. Methods: Participants were from a prospective longitudinal cohort study of 384 children at risk for later psychopathology, with the majority being born with obstetric complications and psychosocial adversities. Assessments of adolescent drug consumption and related intrapersonal characteristics were obtained at age 15. Results: Compared to consumers of alcohol only, 15-year-olds drinking and smoking during the same time period (past 4 weeks) had significantly higher levels of consumption and more excessive use of alcohol, started drinking at an earlier age, had higher scores on the Fagerstrom Test for Nicotine Dependence, and more cannabis use. This group could be distinguished from users of alcohol only by higher novelty seeking and more positive alcohol effect expectancies. Compared to consumers of tobacco only, concurrent users reported higher nicotine dependence and more cannabis use. No significant differences were observed regarding frequency and age at initiation of tobacco use, tobacco-related sensitivity, self- efficacy and instrumentality as well as novelty seeking. Conclusions: Concurrent alcohol and tobacco use during early adolescence is associated with characteristics that are well known as risk factors for later alcohol use problems and dependence and that should be targeted by prevention programs.
A recombinant single chain antibody fragment (designated scDE1) of the murine monoclonal anti-fluorescein antibody B13-DE1 was generated using the original hybridoma cells as source for the variable antibody heavy and light chain (VH and VL) genes. After cloning the variable genes into a phage vector a functional antibody fragment was selected by phage display panning. Recombinant antibody could be expressed as phage antibody and as soluble single chain antibody in Escherichia coli. High yield of scDE1 could also be detected in bacterial culture supernatant. The scDE1 showed the same binding specificity as the parental monoclonal antibody, i.e. it bound fluorescein, fluorescein derivatives and a fluorescein peptide mimotope. Surface plasmon resonance revealed a K(D) of 19 nM for the scDE1 compared to 0.7 nM for the monoclonal antibody. The isolated soluble scDE1 could easily be conjugated to horseradish peroxidase which allowed the use of the conjugate as universal indicator for the detection of fluorescein-labelled proteins in different immunoassays. Detection of hCG in urine was performed as a model system using scDE1. In addition to E. coli the scFv genes could also be transferred and expressed in eukaryotic cells. Finally, we generated HEK293 cells expressing the scDE1 at the cell surface.
