Context. For magnetically driven events, the magnetic energy of the system is the prime energy reservoir that fuels the dynamical evolution. In the solar context, the free energy (i.e., the energy in excess of the potential field energy) is one of the main indicators used in space weather forecasts to predict the eruptivity of active regions. A trustworthy estimation of the magnetic energy is therefore needed in three-dimensional (3D) models of the solar atmosphere, e. g., in coronal fields reconstructions or numerical simulations.
Aims. The expression of the energy of a system as the sum of its potential energy and its free energy (Thomson's theorem) is strictly valid when the magnetic field is exactly solenoidal. For numerical realizations on a discrete grid, this property may be only approximately fulfilled. We show that the imperfect solenoidality induces terms in the energy that can lead to misinterpreting the amount of free energy present in a magnetic configuration.
Methods. We consider a decomposition of the energy in solenoidal and nonsolenoidal parts which allows the unambiguous estimation of the nonsolenoidal contribution to the energy. We apply this decomposition to six typical cases broadly used in solar physics. We quantify to what extent the Thomson theorem is not satisfied when approximately solenoidal fields are used.
Results. The quantified errors on energy vary from negligible to significant errors, depending on the extent of the nonsolenoidal component of the field. We identify the main source of errors and analyze the implications of adding a variable amount of divergence to various solenoidal fields. Finally, we present pathological unphysical situations where the estimated free energy would appear to be negative, as found in some previous works, and we identify the source of this error to be the presence of a finite divergence.
Conclusions. We provide a method of quantifying the effect of a finite divergence in numerical fields, together with detailed diagnostics of its sources. We also compare the efficiency of two divergence-cleaning techniques. These results are applicable to a broad range of numerical realizations of magnetic fields.
We consider the area coverage of radial Levy flights in a finite square area with periodic boundary conditions. From simulations we show how the fractal path dimension d(f) and thus the degree of area coverage depends on the number of steps of the trajectory, the size of the area, and the resolution of the applied box counting algorithm. For sufficiently long trajectories and not too high resolution, the fractal dimension returned by the box counting method equals two, and in that sense the Levy flight fully covers the area. Otherwise, the determined fractal dimension equals the stable index of the distribution of jump lengths of the Levy flight. We provide mathematical expressions for the turnover between these two scaling regimes. As complementary methods to analyze confined Levy flights we investigate fractional order moments of the position for which we also provide scaling arguments. Finally, we study the time evolution of the probability density function and the first passage time density of Levy flights in a square area. Our findings are of interest for a general understanding of Levy flights as well as for the analysis of recorded trajectories of animals searching for food or for human motion patterns.
A new sulfonated aniline-modified poly(vinyl alcohol)/K-feldspar (SAPK) composite was prepared. The cation-exchange capacity of the composite was found to be S times that of neat feldspar. The specific surface area and point of zero charge also changed significantly upon modification, from 15.6 +/- 0.1 m(2)/g and 2.20 (K-feldspar) to 73.6 +/- 0.3 m(2)/g and 1.91 (SAPK). Zn2+, Cd2+, and Pb2+ adsorption was found to be largely independent of pH, and the metal adsorption rate on SAPK was higher than that on neat feldspar. This particularly applies to the initial adsorption rates. The adsorption process involves both film and pore diffusion; film diffusion initially controls the adsorption. The Freundlich and Langmuir models were found to fit metal-ion adsorption on SAPK most accurately. Adsorption on neat feldspar was best fitted with a Langmuir model, indicating the formation of adsorbate monolayers. Both pure feldspar and SAPK showed better selectivity for Pb2+ than for Cd2+ or Zn2+.
New hybrid clay adsorbent based on kaolinite clay and Carica papaya seeds with improved cation exchange capacity (CEC), rate of heavy metal ion uptake, and adsorption capacity for heavy metal ions were prepared. The CEC of the new material is ca. 75 meq/100 g in spite of the unexpectedly low surface area (approximate to 19 m(2)/g). Accordingly, the average particle size of the hybrid clay adsorbent decreased from over 200 to 100 pm. The hybrid clay adsorbent is a highly efficient adsorbent for heavy metals. With an initial metal concentration of 1 mg/L, the hybrid clay adsorbent reduces the Cd2+, Ni2+, and Pb2+ concentration in aqueous solution to <= 4, <= 7 and <= 20 mu g/L, respectively, from the first minute to over 300 min using a fixed bed containing 2 g of adsorbent and a flow rate of approximate to 7 mL/min. These values are (with the exception of Pb2+) in line with the WHO permissible limits for heavy metal ions. In a cocktail solution of Cd2+, and Ni2+, the hybrid clay shows a reduced rate of uptake but an increased adsorption capacity. The CEC data suggest that the adsorption of Pb2+, Cd2+, and Ni2+ on the hybrid clay adsorbent is essentially due to ion exchange. This hybrid clay adsorbent is prepared from materials that are abundant and by a simple means that is sustainable, easily recovered from aqueous solution, nonbiodegradable (unlike numerous biosorbent), and easily regenerated and is a highly efficient alternative to activated carbon for water treatment.

Current assessment of visual neglect involves paper-and-pencil tests or computer-based tasks. Both have been criticised because of their lack of ecological validity as target stimuli can only be presented in a restricted visual range. This study examined the user-friendliness and diagnostic strength of a new "Circle-Monitor" (CM), which enlarges the range of the peripersonal space, in comparison to a standard paper-and-pencil test (Neglect-Test, NET).
Methods: Ten stroke patients with neglect and ten age-matched healthy controls were examined by the NET and the CM test comprising of four subtests (Star Cancellation, Line Bisection, Dice Task, and Puzzle Test).
Results: The acceptance of the CM in elderly controls and neglect patients was high. Participants rated the examination by CM as clear, safe and more enjoyable than NET. Healthy controls performed at ceiling on all subtests, without any systematic differences between the visual fields. Both NET and CM revealed significant differences between controls and patients in Line Bisection, Star Cancellation and visuo-constructive tasks (NET: Figure Copying, CM: Puzzle Test). Discriminant analyses revealed cross-validated assignment of patients and controls to groups was more precise when based on the CM (hit rate 90%) as compared to the NET (hit rate 70%).
Conclusion: The CM proved to be a sensitive novel tool to diagnose visual neglect symptoms quickly and accurately with superior diagnostic validity compared to a standard neglect test while being well accepted by patients. Due to its upgradable functions the system may also be a valuable tool not only to test for non-visual neglect symptoms, but also to provide treatment and assess its outcome.
Supra-glacial deposition and flux of catastrophic rock-slope failure debris, south-central Alaska

The ongoing debate over the effects of global environmental change on Earth's cryosphere calls for detailed knowledge about process rates and their variability in cold environments. In this context, appraisals of the coupling between glacier dynamics and para-glacial erosion rates in tectonically active mountains remain rare. We contribute to filling this knowledge gap and present an unprecedented regional-scale inventory of supra-glacial sediment flux and hillslope erosion rates inferred from an analysis of 123 large (> 0 center dot 1km2) catastrophic bedrock landslides that fell onto glaciers in the Chugach Mountains, Alaska, as documented by satellite images obtained between 1972 to 2008. Assuming these supra-glacial landslide deposits to be passive strain markers we infer minimum decadal-scale sediment yields of 190 to 7400tkm-2yr-1 for a given glacier-surface cross-section impacted by episodic rock-slope failure. These rates compare to reported fluvial sediment yields in many mountain rivers, but are an order of magnitude below the extreme sediment yields measured at the snouts of Alaskan glaciers, indicating that the bulk of debris discharged derives from en-glacial, sub-glacial or ice-proximal sources. We estimate an average minimum para-glacial erosion rate by large, episodic rock-slope failures at 0 center dot 5-0 center dot 7mmyr-1 in the Chugach Mountains over a 50-yr period, with earthquakes likely being responsible for up to 73% of this rate. Though ranking amongst the highest decadal landslide erosion rates for this size of study area worldwide, our inferred rates of hillslope erosion in the Chugach Mountains remain an order of magnitude below the pace of extremely rapid glacial sediment export and glacio-isostatic surface uplift previously reported from the region.

Large Central European flood events of the past have demonstrated that flooding can affect several river basins at the same time leading to catastrophic economic and humanitarian losses that can stretch emergency resources beyond planned levels of service. For Germany, the spatial coherence of flooding, the contributing processes and the role of trans-basin floods for a national risk assessment is largely unknown and analysis is limited by a lack of systematic data, information and knowledge on past events. This study investigates the frequency and intensity of trans-basin flood events in Germany. It evaluates the data and information basis on which knowledge about trans-basin floods can be generated in order to improve any future flood risk assessment. In particu-lar, the study assesses whether flood documentations and related reports can provide a valuable data source for understanding trans-basin floods. An adaptive algorithm was developed that systematically captures trans-basin floods using series of mean daily discharge at a large number of sites of even time series length (1952-2002). It identifies the simultaneous occurrence of flood peaks based on the exceedance of an initial threshold of a 10 year flood at one location and consecutively pools all causally related, spatially and temporally lagged peak recordings at the other locations. A weighted cumulative index was developed that accounts for the spatial extent and the individual flood magnitudes within an event and allows quantifying the overall event severity. The parameters of the method were tested in a sensitivity analysis. An intensive study on sources and ways of information dissemination of flood-relevant publications in Germany was conducted. Based on the method of systematic reviews a strategic search approach was developed to identify relevant documentations for each of the 40 strongest trans-basin flood events. A novel framework for assessing the quality of event specific flood reports from a user’s perspective was developed and validated by independent peers. The framework was designed to be generally applicable for any natural hazard type and assesses the quality of a document addressing accessibility as well as representational, contextual, and intrinsic dimensions of quality. The analysis of time-series of mean daily discharge resulted in the identification of 80 trans-basin flood events within the period 1952-2002 in Germany. The set is dominated by events that were recorded in the hydrological winter (64%); 36% occurred during the summer months. The occurrence of floods is characterised by a distinct clustering in time. Dividing the study period into two sub-periods, we find an increase in the percentage of winter events from 58% in the first to 70.5% in the second sub-period. Accordingly, we find a significant increase in the number of extreme trans-basin floods in the second sub-period. A large body of 186 flood relevant documentations was identified. For 87.5% of the 40 strongest trans-basin floods in Germany at least one report has been found and for the most severe floods a substantial amount of documentation could be obtained. 80% of the material can be considered grey literature (i.e. literature not controlled by commercial publishers). The results of the quality assessment show that the majority of flood event specific reports are of a good quality, i.e. they are well enough drafted, largely accurate and objective, and contain a substantial amount of information on the sources, pathways and receptors/consequences of the floods. The inclusion of this information in the process of knowledge building for flood risk assessment is recommended. Both the results as well as the data produced in this study are openly accessible and can be used for further research. The results of this study contribute to an improved spatial risk assessment in Germany. The identified set of trans-basin floods provides the basis for an assessment of the chance that flooding occurs simultaneously at a number of sites. The information obtained from flood event documentation can usefully supplement the analysis of the processes that govern flood risk.
Sophisticated methods have been developed and become standard in analysing floods as well as for assessing flood risk. However, increasingly critique of the current standards and scientific practice can be found both in the flood hydrology community as well as in the risk community who argue that the considerable amount of information already available on natural disasters has not been adequately deployed and brought to effective use. We describe this phenomenon as a failure to synthesize knowledge that results from barriers and ignorance in awareness, use and management of the entire spectrum of relevant content, that is, data, information and knowledge. In this paper we argue that the scientific community in flood risk research ignores event-specific analysis and documentations as another source of data. We present results from a systematic search that includes an intensive study on sources and ways of information dissemination of flood-relevant publications. We obtain 186 documents that contain information on the sources, pathways, receptors and/or consequences for any of the 40 strongest trans-basin floods in Germany in the period 1952-2002. This study therefore provides the most comprehensive metadata collection of flood documentations for the considered geographical space and period. A total of 87.5% of all events have been documented, and especially the most severe floods have received extensive coverage. Only 30% of the material has been produced in the scientific/academic environment, and the majority of all documents (about 80%) can be considered grey literature (i.e. literature not controlled by commercial publishers). Therefore, ignoring grey sources in flood research also means ignoring the largest part of knowledge available on single flood events (in Germany). Further, the results of this study underpin the rapid changes in information dissemination of flood event literature over the last decade. We discuss the options and obstacles of incorporating this data into the knowledge-building process in light of the current technological developments and international, interdisciplinary debates for data curation.

Understanding metabolic acclimation of plants to challenging environmental conditions is essential for dissecting the role of metabolic pathways in growth and survival. As stresses involve simultaneous physiological alterations across all levels of cellular organization, a comprehensive characterization of the role of metabolic pathways in acclimation necessitates integration of genome-scale models with high-throughput data. Here, we present an integrative optimization-based approach, which, by coupling a plant metabolic network model and transcriptomics data, can predict the metabolic pathways affected in a single, carefully controlled experiment. Moreover, we propose three optimization-based indices that characterize different aspects of metabolic pathway behavior in the context of the entire metabolic network. We demonstrate that the proposed approach and indices facilitate quantitative comparisons and characterization of the plant metabolic response under eight different light and/or temperature conditions. The predictions of the metabolic functions involved in metabolic acclimation of Arabidopsis thaliana to the changing conditions are in line with experimental evidence and result in a hypothesis about the role of homocysteine-to-Cys interconversion and Asn biosynthesis. The approach can also be used to reveal the role of particular metabolic pathways in other scenarios, while taking into consideration the entirety of characterized plant metabolism.
The relaxation of a dissipative system to its equilibrium state often shows a multiexponential pattern with relaxation rates, which are typically considered to be independent of the initial condition. The rates follow from the spectrum of a Hermitian operator obtained by a similarity transformation of the initial Fokker-Planck operator. However, some initial conditions are mapped by this similarity transformation to functions which growat infinity. These cannot be expanded in terms of the eigenfunctions of a Hermitian operator, and show different relaxation patterns. Considering the exactly solvable examples of Gaussian and generalized Levy Ornstein-Uhlenbeck processes (OUPs) we show that the relaxation rates belong to the Hermitian spectrum only if the initial condition belongs to the domain of attraction of the stable distribution defining the noise. While for an ordinary OUP initial conditions leading to nonspectral relaxation can be considered exotic, for generalized OUPs driven by Levy noise, such initial conditions are the rule. Intensive Forschung hat in den vergangenen Jahrzehnten zu einer sehr detaillierten Charakterisierung des Geschmackssystems der Säugetiere geführt. Dennoch sind mit den bislang eingesetzten Methoden wichtige Fragestellungen unbeantwortet geblieben. Eine dieser Fragen gilt der Unterscheidung von Bitterstoffen. Die Zahl der Substanzen, die für den Menschen bitter schmecken und in Tieren angeborenes Aversionsverhalten auslösen, geht in die Tausende. Diese Substanzen sind sowohl von der chemischen Struktur als auch von ihrer Wirkung auf den Organismus sehr verschieden. Während viele Bitterstoffe potente Gifte darstellen, sind andere in den Mengen, die mit der Nahrung aufgenommen werden, harmlos oder haben sogar positive Effekte auf den Körper. Zwischen diesen Gruppen unterscheiden zu können, wäre für ein Tier von Vorteil. Ein solcher Mechanismus ist jedoch bei Säugetieren nicht bekannt. Das Ziel dieser Arbeit war die Untersuchung der Verarbeitung von Geschmacksinformation in der ersten Station der Geschmacksbahn im Mausgehirn, dem Nucleus tractus solitarii (NTS), mit besonderem Augenmerk auf der Frage nach der Diskriminierung verschiedener Bitterstoffe. Zu diesem Zweck wurde eine neue Untersuchungsmethode für das Geschmackssystem etabliert, die die Nachteile bereits verfügbarer Methoden umgeht und ihre Vorteile kombiniert. Die Arc-catFISH-Methode (cellular compartment analysis of temporal activity by fluorescent in situ hybridization), die die Charakterisierung der Antwort großer Neuronengruppen auf zwei Stimuli erlaubt, wurde zur Untersuchung geschmacksverarbeitender Zellen im NTS angewandt. Im Zuge dieses Projekts wurde erstmals eine stimulusinduzierte Arc-Expression im NTS gezeigt. Die ersten Ergebnisse offenbarten, dass die Arc-Expression im NTS spezifisch nach Stimulation mit Bitterstoffen auftritt und sich die Arc exprimierenden Neurone vornehmlich im gustatorischen Teil des NTS befinden. Dies weist darauf hin, dass Arc-Expression ein Marker für bitterverarbeitende gustatorische Neurone im NTS ist. Nach zweimaliger Stimulation mit Bittersubstanzen konnten überlappende, aber verschiedene Populationen von Neuronen beobachtet werden, die unterschiedlich auf die drei verwendeten Bittersubstanzen Cycloheximid, Chininhydrochlorid und Cucurbitacin I reagierten. Diese Neurone sind vermutlich an der Steuerung von Abwehrreflexen beteiligt und könnten so die Grundlage für divergentes Verhalten gegenüber verschiedenen Bitterstoffen bilden.
Die automatisierte Objektidentifikation stellt ein modernes Werkzeug in den Geoinformationswissenschaften dar (BLASCHKE et al., 2012). Um bei thematischen Kartierungen untereinander vergleichbare Ergebnisse zu erzielen, sollen aus Sicht der Geoinformatik Mittel für die Objektidentifikation eingesetzt werden. Anstelle von Feldarbeit werden deshalb in der vorliegenden Arbeit multispektrale Fernerkundungsdaten als Primärdaten verwendet. Konkrete natürliche Objekte werden GIS-gestützt und automatisiert über große Flächen und Objektdichten aus Primärdaten identifiziert und charakterisiert. Im Rahmen der vorliegenden Arbeit wird eine automatisierte Prozesskette zur Objektidentifikation konzipiert. Es werden neue Ansätze und Konzepte der objektbasierten Identifikation von natürlichen isolierten terrestrischen Oberflächenformen entwickelt und implementiert. Die Prozesskette basiert auf einem Konzept, das auf einem generischen Ansatz für automatisierte Objektidentifikation aufgebaut ist. Die Prozesskette kann anhand charakteristischer quantitativer Parameter angepasst und so umgesetzt werden, womit das Konzept der Objektidentifikation modular und skalierbar wird. Die modulbasierte Architektur ermöglicht den Einsatz sowohl einzelner Module als auch ihrer Kombination und möglicher Erweiterungen. Die eingesetzte Methodik der Objektidentifikation und die daran anschließende Charakteristik der (geo)morphometrischen und morphologischen Parameter wird durch statistische Verfahren gestützt. Diese ermöglichen die Vergleichbarkeit von Objektparametern aus unterschiedlichen Stichproben. Mit Hilfe der Regressionsund Varianzanalyse werden Verhältnisse zwischen Objektparametern untersucht. Es werden funktionale Abhängigkeiten der Parameter analysiert, um die Objekte qualitativ zu beschreiben. Damit ist es möglich, automatisiert berechnete Maße und Indizes der Objekte als quantitative Daten und Informationen zu erfassen und unterschiedliche Stichproben anzuwenden. Im Rahmen dieser Arbeit bilden Thermokarstseen die Grundlage für die Entwicklungen und als Beispiel sowie Datengrundlage für den Aufbau des Algorithmus und die Analyse. Die Geovisualisierung der multivariaten natürlichen Objekte wird für die Entwicklung eines besseren Verständnisses der räumlichen Relationen der Objekte eingesetzt. Kern der Geovisualisierung ist das Verknüpfen von Visualisierungsmethoden mit kartenähnlichen Darstellungen.
This paper proposes a method of real-time monitoring and modeling the ionospheric Total Electron Content (TEC) by Precise Point Positioning (PPP). Firstly, the ionospheric TEC and receiver's Differential Code Biases (DCB) are estimated with the undifferenced raw observation in real-time, then the ionospheric TEC model is established based on the Single Layer Model (SLM) assumption and the recovered ionospheric TEC. In this study, phase observations with high precision are directly used instead of phase smoothed code observations. In addition, the DCB estimation is separated from the establishment of the ionospheric model which will limit the impacts of the SLM assumption impacts. The ionospheric model is established at every epoch for real time application. The method is validated with three different GNSS networks on a local, regional, and global basis. The results show that the method is feasible and effective, the real-time ionosphere and DCB results are very consistent with the IGS final products, with a bias of 1-2 TECU and 0.4 ns respectively.
In order to speed up Precise Point Positioning (PPP)'s convergence, a combined PPP method with GPS and GLONASS which is based on using raw observations is proposed, and the positioning results and convergence time have been compared with that of single system. The ionospheric delays and receiver's Differential Code Bias (DCB) corrections are estimated as unknown parameters in this method. The numerical results show that the combined PPP has not caused significant impacts on the final solutions, but it greatly improved Position Dilution of Precision (PDOP) and convergence speed and enhanced the reliability of the solution. Meanwhile, the convergence speed is greatly influenced by the receiver's DCB, positioning results in horizontal which are better than 10 cm can be realized within 10 min. In addition, the ionosphere and DCB products can be provided with high precision.
In recent years, an increasing number of mutations in what would appear to be 'housekeeping genes' have been identified as having unexpectedly specific defects in multicellular organogenesis. This is also the case for organogenesis in seed plants. Although it is not surprising that loss-of-function mutations in 'housekeeping' genes result in lethality or growth retardation, it is surprising when (1) the mutant phenotype results from the loss of function of a 'housekeeping' gene and (2) the mutant phenotype is specific. In this review, by defining housekeeping genes as those encoding proteins that work in basic metabolic and cellular functions, we discuss unexpected links between housekeeping genes and specific developmental processes. In a surprising number of cases housekeeping genes coding for enzymes or proteins with functions in basic cellular processes such as transcription, post-transcriptional modification, and translation affect plant development.

„Ein Buch für Alle und Keinen“ heißt es auf dem Titelblatt von Nietzsches Also sprach Zarathustra. Was wie ein Paradoxon anmutet, verweist jedoch auf den manifesten strukturellen Gehalt eines Werkes, welches der Forschung größtenteils nur mit seinem reichhaltigen narrativen Inhalt entgegentritt. An Nietzsches Also sprach Zarathustra soll in Bezug auf die Klärung des Untertitels untersucht werden, inwiefern die Struktur oder Form des Textes als philosophischer Gehalt verstanden werden kann. Gezeigt werden soll, dass der Zarathustra nicht nur selbstreferentiell den Schlüssel zu seinem Verständnis in sich birgt, sondern auch unabhängig von seinem Inhalt Informationen transportiert, die eigentlich unter einem vermeintlichen methodischen Ausschluss der Möglichkeit des Verstehens, einen philosophischen Sinnhorizont eröffnen.
Es soll gezeigt werden, dass der Zarathustra eine Eigendynamik aufweist, welche im Selbstvollzug ihren philosophischen Gehalt entfaltet. Dabei soll sich zeigen, dass der Text sich in einem Verweisungsgeflecht von in dieser Arbeit herauszuarbeitenden Textebenen in einem Akt der Selbsterfüllung auflöst. Darüber hinaus wird herausgestellt, wie dies geschieht, warum der herauszuarbeitende philosophische Gehalt dieser Dynamik die strukturelle Anforderung an den Text stellt und wie der Untertitel gleichsam als Chiffre, als Leseanweisung, sowie auch als ein den Text manipulierender Katalysator jener skizzierten Dynamik fungiert. Im Laufe dieser Untersuchung wird sich erhärten, was hier als Prämisse angeführt wird: In der Dynamik der Auflösung des Textes sowie in dem im Vollzug des Textes sich ergebenden Ausschluss kommunizierbarer einheitlicher Wahrheiten lösen sich ebenso die vermeintlichen philosophischen Schwergewichte des Zarathustra, wie der Topos des Übermenschen, der Willen zur Macht oder die Ewige Wiederkunft auf, sodass von Lehren oder gar Philosophemen im Also sprach Zarathustra nicht die Rede sein kann.
Diese Arbeit wird zeigen, dass der Text weniger inhaltlich als vielmehr im Vollzug seiner Struktur von philosophischer Bedeutung ist und gar als das folgerichtige Paradigma einer Philosophie angesehen werden kann, welche die Dichotomie des binären Diskurses von ‚wahr‘ und ‚falsch‘ der abendländischen Epistemologie kritisch überwunden zu haben scheint.
The near-infrared is an important part of the spectrum in astronomy, especially in cosmology because the light from objects in the early universe is redshifted to these wavelengths. However, deep near-infrared observations are extremely difficult to make from ground-based telescopes due to the bright background from the atmosphere. Nearly all of this background comes from the bright and narrow emission lines of atmospheric hydroxyl (OH) molecules. The atmospheric background cannot be easily removed from data because the brightness fluctuates unpredictably on short timescales. The sensitivity of ground-based optical astronomy far exceeds that of near-infrared astronomy because of this long-standing problem. GNOSIS is a prototype astrophotonic instrument that utilizes "OH suppression fibers" consisting of fiber Bragg gratings and photonic lanterns to suppress the 103 brightest atmospheric emission doublets between 1.47 and 1.7 mu m. GNOSIS was commissioned at the 3.9 m Anglo-Australian Telescope with the IRIS2 spectrograph to demonstrate the potential of OH suppression fibers, but may be potentially used with any telescope and spectrograph combination. Unlike previous atmospheric suppression techniques GNOSIS suppresses the lines before dispersion and in a manner that depends purely on wavelength. We present the instrument design and report the results of laboratory and on-sky tests from commissioning. While these tests demonstrated high throughput (approximate to 60%) and excellent suppression of the skylines by the OH suppression fibers, surprisingly GNOSIS produced no significant reduction in the interline background and the sensitivity of GNOSIS+IRIS2 is about the same as IRIS2. It is unclear whether the lack of reduction in the interline background is due to physical sources or systematic errors as the observations are detector noise dominated. OH suppression fibers could potentially impact ground-based astronomy at the level of adaptive optics or greater. However, until a clear reduction in the interline background and the corresponding increasing in sensitivity is demonstrated optimized OH suppression fibers paired with a fiber-fed spectrograph will at least provide a real benefit at low resolving powers.
TURBO2 - a MATLAB simulation to study the effects of bioturbation on paleoceanographic time series

Bioturbation (or benthic mixing) causes significant distortions in marine stable isotope signals and other palaeoceanographic records. Although the influence of bioturbation on these records is well known it has rarely been dealt systematically. The MATLAB program called TURBO2 can be used to simulate the effect of bioturbation on individual sediment particles. It can therefore be used to model the distortion of all physical, chemical, and biological signals in deep-sea sediments, such as Mg/Ca ratios and UK37-based sea-surface temperature (SST) variations. In particular, it can be used to study the distortions in paleoceanographic records that are based on individual sediment particles, such as SST records based on foraminifera assemblages. Furthermore. TURBO2 provides a tool to study the effect of benthic mixing of isotope signals such as C-14, delta O-18, and delta C-13, measured in a stratigraphic carrier such as foraminifera shells.
Interactive rendering techniques for focus+context visualization of 3D geovirtual environments

This thesis introduces a collection of new real-time rendering techniques and applications for focus+context visualization of interactive 3D geovirtual environments such as virtual 3D city and landscape models. These environments are generally characterized by a large number of objects and are of high complexity with respect to geometry and textures. For these reasons, their interactive 3D rendering represents a major challenge. Their 3D depiction implies a number of weaknesses such as occlusions, cluttered image contents, and partial screen-space usage. To overcome these limitations and, thus, to facilitate the effective communication of geo-information, principles of focus+context visualization can be used for the design of real-time 3D rendering techniques for 3D geovirtual environments (see Figure). In general, detailed views of a 3D geovirtual environment are combined seamlessly with abstracted views of the context within a single image. To perform the real-time image synthesis required for interactive visualization, dedicated parallel processors (GPUs) for rasterization of computer graphics primitives are used. For this purpose, the design and implementation of appropriate data structures and rendering pipelines are necessary. The contribution of this work comprises the following five real-time rendering methods: • The rendering technique for 3D generalization lenses enables the combination of different 3D city geometries (e.g., generalized versions of a 3D city model) in a single image in real time. The method is based on a generalized and fragment-precise clipping approach, which uses a compressible, raster-based data structure. It enables the combination of detailed views in the focus area with the representation of abstracted variants in the context area. • The rendering technique for the interactive visualization of dynamic raster data in 3D geovirtual environments facilitates the rendering of 2D surface lenses. It enables a flexible combination of different raster layers (e.g., aerial images or videos) using projective texturing for decoupling image and geometry data. Thus, various overlapping and nested 2D surface lenses of different contents can be visualized interactively. • The interactive rendering technique for image-based deformation of 3D geovirtual environments enables the real-time image synthesis of non-planar projections, such as cylindrical and spherical projections, as well as multi-focal 3D fisheye-lenses and the combination of planar and non-planar projections. • The rendering technique for view-dependent multi-perspective views of 3D geovirtual environments, based on the application of global deformations to the 3D scene geometry, can be used for synthesizing interactive panorama maps to combine detailed views close to the camera (focus) with abstract views in the background (context). This approach reduces occlusions, increases the usage the available screen space, and reduces the overload of image contents. • The object-based and image-based rendering techniques for highlighting objects and focus areas inside and outside the view frustum facilitate preattentive perception. The concepts and implementations of interactive image synthesis for focus+context visualization and their selected applications enable a more effective communication of spatial information, and provide building blocks for design and development of new applications and systems in the field of 3D geovirtual environments.
A new class of star-shaped, liquid crystalline, low-molecular weight compounds functionalized with photochromic azobenzene and mesogenic groups was investigated in terms of light-induced anisotropy. The behaviour of the materials under the action of light with simultaneous or subsequent thermal treatment was examined with respect to the induction of anisotropy. The unconventional UV light treatment prior to the irradiation with linearly polarized light allowed induction of very high values of anisotropy (D = 0.77) at room temperature. Moreover, the simultaneous action of light and temperature led to the induction of higher values of dichroism in comparison with anisotropy generated by the standard procedure. Subsequent thermal treatment led to dewetting and the formation of 3D macroscopic stripe- and dome-like structures for one of the investigated compounds. Despite photoinduction of anisotropy by a single beam, the formation of polarization and surface relief gratings by two-beam interference pattern was also investigated.
The occurrence of neritic microbial carbonates is often related to ecological refuges, where grazers and other competitors are reduced by environmental conditions, or to post-extinction events (e.g. in the Late Devonian, Early Triassic). Here, we present evidence for Middle Jurassic (Bajocian) microbial mounds formed in the normal marine, shallow neritic setting of an inner, ramp system from the High Atlas of Morocco. The microbial mounds are embedded in cross-bedded oolitic facies. Individual mounds show low relief domal geometries (up to 3 m high and 4.5 m across), but occasionally a second generation of mounds exhibits tabular geometries (<1 m high). The domes are circular in plan view and have intact tops, lacking evidence of current influence on mound preferred growth direction or distribution patterns, or truncation. The mound fades consists almost entirely of non-laminated, micritic thrombolites with branching morphologies and fine-grained, clotted and peloidal fabrics. Normal marine biota are present but infrequent. Several lines of evidence document that microbial mound growth alternates with time intervals of active ooid shoal deposition. This notion is of general significance when compared with modern Bahamian microbialites that co-exist with active sub-aquatic dunes. Furthermore, the lack of detailed studies of Middle Jurassic, normal marine shallow neritic microbial mounds adds a strong motivation for the present study. Specifically, Bajocian mounds formed on a firmground substratum during transgressive phases under condensed sedimentation. Furthermore, a transient increase in nutrient supply in the prevailing mesotrophic setting, as suggested by the heterotrophic-dominated biota, may have controlled microbial mound stages.
A considerable fraction of the central stars of planetary nebulae (CSPNe) are hydrogen-deficient. Almost all of these H-deficient central stars (CSs) display spectra with strong carbon and helium lines. Most of them exhibit emission-line spectra resembling those of massive WC stars. Therefore these stars are classed as CSPNe of spectral type [WC]. Recently, quantitative spectral analysis of two emission-line CSs, PB 8 and IC 4663, revealed that these stars do not belong to the [WC] class. Instead PB 8 has been classified as [WN/WC] type and IC 4663 as [WN] type. In this work we report the spectroscopic identification of another rare [WN] star, the CS of Abell 48. We performed a spectral analysis of Abell 48 with the Potsdam Wolf-Rayet (PoWR) models for expanding atmospheres. We find that the expanding atmosphere of Abell 48 is mainly composed of helium (85 per cent by mass), hydrogen (10 per cent) and nitrogen (5 per cent). The residual hydrogen and the enhanced nitrogen abundance make this object different from the other [WN] star IC 4663. We discuss the possible origin of this atmospheric composition.

This paper proposes a novel analysis of the Russian OVS construction and argues that the parametric variation in the availability of OVS cross-linguistically depends on the type of relative interpretative argument prominence that a language encodes via syntactic structure. When thematic and information-structural prominence relations do not coincide, only one of them can be structurally/linearly represented. The relation that is not structurally/linearly encoded must be made visible at the PF interface either via prosody or morphology.

The world’s appetite for energy is producing growing quantities of CO2, a pollutant that contributes to the warming of the planet and which currently cannot be removed or stored in any significant way. Other natural reserves are also being devoured at alarming rates and current assessments suggest that we will need to identify alternative sources in the near future. With the aid of materials chemistry it should be possible to create a world in which energy use needs not be limited and where usable energy can be produced and stored wherever it is needed, where we can minimize and remediate emissions as new consumer products are created, whilst healing the planet and preventing further disruptive and harmful depletion of valuable mineral assets. In achieving these aims, the creation of new and very importantly greener industries and new sustainable pathways are crucial. In all of the aforementioned applications, new materials based on carbon, ideally produced via inexpensive, low energy consumption methods, using renewable resources as precursors, with flexible morphologies, pore structures and functionalities, are increasingly viewed as ideal candidates to fulfill these goals. The resulting materials should be a feasible solution for the efficient storage of energy and gases. At the end of life, such materials ideally must act to improve soil quality and to act as potential CO2 storage sinks. This is exactly the subject of this habilitation thesis: an alternative technology to produce carbon materials from biomass in water using low carbonisation temperatures and self-generated pressures. This technology is called hydrothermal carbonisation. It has been developed during the past five years by a group of young and talented researchers working under the supervision of Dr. Titirici at the Max-Planck Institute of Colloids and Interfaces and it is now a well-recognised methodology to produce carbon materials with important application in our daily lives. These applications include electrodes for portable electronic devices, filters for water purification, catalysts for the production of important chemicals as well as drug delivery systems and sensors.
We report observations of dusty clouds in Saturn's rings, which we interpret as resulting from impacts onto the rings that occurred between 1 and 50 hours before the clouds were observed. The largest of these clouds was observed twice; its brightness and cant angle evolved in a manner consistent with this hypothesis. Several arguments suggest that these clouds cannot be due to the primary impact of one solid meteoroid onto the rings, but rather are due to the impact of a compact stream of Saturn-orbiting material derived from previous breakup of a meteoroid. The responsible interplanetary meteoroids were initially between 1 centimeter and several meters in size, and their influx rate is consistent with the sparse prior knowledge of smaller meteoroids in the outer solar system.
The M-type kinesin isoform, Kif9, has recently been implicated in maintaining a physical connection between the centrosome and nucleus in Dictyostelium discoideum. However, the mechanism by which Kif9 functions to link these two organelles remains obscure. Here we demonstrate that the Kif9 protein is localized to the nuclear envelope and is concentrated in the region underlying the centrosome point of attachment. Nuclear anchorage appears mediated through a specialized transmembrane domain located in the carboxyl terminus. Kif9 interacts with microtubules in in vitro binding assays and effects an endwise depolymerization of the polymer. These results suggest a model whereby Kif9 is anchored to the nucleus and generates a pulling force that reels the centrosome up against the nucleus. This is a novel activity for a kinesin motor, one important for progression of cells into mitosis and to ensure centrosome-nuclear parity in a multinuclear environment.
A high resolution multi proxy (pollen, grain size, total organic carbon) record from a small mountain lake (Lake Khuisiin; 46.6 degrees N, 101.8 degrees E; 2270 m a.s.l.) in the south eastern Khangai Mountains of central Mongolia has been used to explore changes in vegetation and climate over the last 1200 years. The pollen data indicates that the vegetation changed from dry steppe dominated by Poaceae and Artemisia (ca AD 760-950), to Larix forest steppe (ca AD 950-1170), Larix Betula forest steppe (ca AD 1170-1380), meadow dominated by Cyperaceae and Poaceae (ca AD 1380-1830), and Larix Betula forest steppe (after similar to AD 1830). The cold-wet period between AD 1380 and 1830 may relate to the Little Ice Age. Environmental changes were generally subtle and climate change seems to have been the major driver of variations in vegetation until at least the early part of the 20th century, suggesting that either the level of human activity was generally low, or the relationship between human activity and vegetation did not alter substantially between AD 760 and 1830. A review of centennial scale moisture records from China and Mongolia revealed that most areas experienced major changes at ca AD 1500 and AD 1900. However, the moisture availability since AD 1500 varied between sites, with no clear regional pattern or relationship to present day conditions. Both the reconstructions and the moisture levels simulation on a millennium scale performed in the MPI Earth System Model indicate that the monsoon-westerlies transition area shows a greater climate variability than those areas influenced by the westerlies, or by the summer monsoon only.

These predictions are exact for reaction networks composed exclusively of first order reactions or for networks involving bimolecular reactions and large numbers of molecules. It is however well known that gene regulation involves bimolecular interactions with molecule numbers as small as a single copy of a particular gene. It is therefore questionable how reliable are the LNA predictions for these systems.
Results: We implement in the software package intrinsic Noise Analyzer (iNA), a system size expansion based method which calculates the mean concentrations and the variances of the fluctuations to an order of accuracy higher than the LNA. We then use iNA to explore the parametric dependence of the Fano factors and of the coefficients of variation of the mRNA and protein fluctuations in models of genetic networks involving nonlinear protein degradation, post-transcriptional, post-translational and negative feedback regulation. We find that the LNA can significantly underestimate the amplitude and period of noise-induced oscillations in genetic oscillators. We also identify cases where the LNA predicts that noise levels can be optimized by tuning a bimolecular rate constant whereas our method shows that no such regulation is possible. All our results are confirmed by stochastic simulations.
Conclusion: The software iNA allows the investigation of parameter regimes where the LNA fares well and where it does not. We have shown that the parametric dependence of the coefficients of variation and Fano factors for common gene regulatory networks is better described by including terms of higher order than LNA in the system size expansion. This analysis is considerably faster than stochastic simulations due to the extensive ensemble averaging needed to obtain statistically meaningful results. Hence iNA is well suited for performing computationally efficient and quantitative studies of intrinsic noise in gene regulatory networks.

Water management and environmental protection is vulnerable to extreme low flows during streamflow droughts. During the last decades, in most rivers of Central Europe summer runoff and low flows have decreased. Discharge projections agree that future decrease in runoff is likely for catchments in Brandenburg, Germany. Depending on the first-order controls on low flows, different adaption measures are expected to be appropriate. Small catchments were analyzed because they are expected to be more vulnerable to a changing climate than larger rivers. They are mainly headwater catchments with smaller ground water storage. Local characteristics are more important at this scale and can increase vulnerability. This thesis mutually evaluates potential adaption measures to sustain minimum runoff in small catchments of Brandenburg, Germany, and similarities of these catchments regarding low flows. The following guiding questions are addressed: (i) Which first-order controls on low flows and related time scales exist? (ii) Which are the differences between small catchments regarding low flow vulnerability? (iii) Which adaption measures to sustain minimum runoff in small catchments of Brandenburg are appropriate considering regional low flow patterns? Potential adaption measures to sustain minimum runoff during periods of low flows can be classified into three categories: (i) increase of groundwater recharge and subsequent baseflow by land use change, land management and artificial ground water recharge, (ii) increase of water storage with regulated outflow by reservoirs, lakes and wetland water management and (iii) regional low flow patterns have to be considered during planning of measures with multiple purposes (urban water management, waste water recycling and inter-basin water transfer). The question remained whether water management of areas with shallow groundwater tables can efficiently sustain minimum runoff. Exemplary, water management scenarios of a ditch irrigated area were evaluated using the model Hydrus-2D. Increasing antecedent water levels and stopping ditch irrigation during periods of low flows increased fluxes from the pasture to the stream, but storage was depleted faster during the summer months due to higher evapotranspiration. Fluxes from this approx. 1 km long pasture with an area of approx. 13 ha ranged from 0.3 to 0.7 l\s depending on scenario. This demonstrates that numerous of such small decentralized measures are necessary to sustain minimum runoff in meso-scale catchments. Differences in the low flow risk of catchments and meteorological low flow predictors were analyzed. A principal component analysis was applied on daily discharge of 37 catchments between 1991 and 2006. Flows decreased more in Southeast Brandenburg according to meteorological forcing. Low flow risk was highest in a region east of Berlin because of intersection of a more continental climate and the specific geohydrology. In these catchments, flows decreased faster during summer and the low flow period was prolonged. A non-linear support vector machine regression was applied to iteratively select meteorological predictors for annual 30-day minimum runoff in 16 catchments between 1965 and 2006. The potential evapotranspiration sum of the previous 48 months was the most important predictor (r²=0.28). The potential evapotranspiration of the previous 3 months and the precipitation of the previous 3 months and last year increased model performance (r²=0.49, including all four predictors). Model performance was higher for catchments with low yield and more damped runoff. In catchments with high low flow risk, explanatory power of long term potential evapotranspiration was high. Catchments with a high low flow risk as well as catchments with a considerable decrease in flows in southeast Brandenburg have the highest demand for adaption. Measures increasing groundwater recharge are to be preferred. Catchments with high low flow risk showed relatively deep and decreasing groundwater heads allowing increased groundwater recharge at recharge areas with higher altitude away from the streams. Low flows are expected to stay low or decrease even further because long term potential evapotranspiration was the most important low flow predictor and is projected to increase during climate change. Differences in low flow risk and runoff dynamics between catchments have to be considered for management and planning of measures which do not only have the task to sustain minimum runoff.
Floricolous downy mildews are a monophyletic group of members of the genus Peronospora (Oomycota, Peronosporales). These downy mildews can be found on a variety of families of the Asteridae, including Asteraceae, Campanulaceae, Dipsacaceae, Lamiaceae, and Orobanchaceae. With the exception of Peronospora radii, which can also cause economically relevant losses, sporulation usually takes place only on floral parts of their hosts. However, only very few specimens of these mostly inconspicuous downy mildews have so far been included in molecular phylogenies. Focusing on Lamiaceae, we have investigated multiple specimens of floricolous downy mildews for elucidating species boundaries and host specificity in this group. Based on both mitochondrial and nuclear loci, it became apparent that phylogenetic lineages in the Lamiaceae seem to be host genus specific and significant sequence diversity could be found between lineages. Based on distinctiveness in both phylogenetic reconstructions and morphology, the downy mildew on flowers of Stachys palustris is introduced as a new species, Peronospora jagei sp. nov., which can be morphologically distinguished from Peronospora stigmaticola by broader and shorter conidiospores. The diversity of the floricolous down mildews might be higher than previously assumed, although specimens from a much broader set of samples will be needed to confirm this view.

Im vorspanischen Zentralkolumbien, vor allem in der Muisca-Kultur, wurden Hochreliefs auf Steinpatrizen zur Abformung von Wachsmatrizen für den Guss in verlorener Form benutzt, die Alexander von Humboldt irrtümlich für „Kalendersteine“ der Muisca hielt. A polymer analogous reaction was carried out to generate a porous polymeric network with N-heterocyclic carbenes (NHC) in the polymer backbone. Using a stepwise approach, first a polyimine network is formed by polymerization of the tetrafunctional amine tetrakis(4-aminophenyl)methane. This polyimine network is converted in the second step into polyimidazolium chloride and finally to a polyNHC network. Furthermore a porous Cu(II)-coordinated polyNHC network can be generated. Supercritical drying generates polymer networks with high permanent surface areas and porosities which can be applied for different catalytic reactions. The catalytic properties were demonstrated for example in the activation of CO2 or in the deoxygenation of sulfoxides to the corresponding sulfides.

A polymer analogous reaction for the formation of imidazolium and NHC based porous polymer networks

A polymer analogous reaction was carried out to generate a porous polymeric network with N-heterocyclic carbenes (NHC) in the polymer backbone. Using a stepwise approach, first a polyimine network is formed by polymerization of the tetrafunctional amine tetrakis(4-aminophenyl)methane. This polyimine network is converted in the second step into polyimidazolium chloride and finally to a polyNHC network. Furthermore a porous Cu(II)-coordinated polyNHC network can be generated. Supercritical drying generates polymer networks with high permanent surface areas and porosities which can be applied for different catalytic reactions. The catalytic properties were demonstrated for example in the activation of CO2 or in the deoxygenation of sulfoxides to the corresponding sulfides.
The northward motion of the Pamir indenter with respect to Eurasia has resulted in coeval thrusting, strike-slip faulting, and normal faulting. The eastern Pamir is currently deformed by east-west oriented extension, accompanied by uplift and exhumation of the Kongur Shan (7719m) and Muztagh Ata (7546m) gneiss domes. Both domes are an integral part of the footwall of the Kongur Shan extensional fault system (KES), a 250 km long, north-south oriented graben. Why active normal faulting within the Pamir is primarily localized along the KES and not distributed more widely throughout the orogen has remained unclear. In addition, relatively little is known about how deformation has evolved throughout the Cenozoic, despite refined estimates on present-day crustal deformation rates and microseismicity, which indicate where crustal deformation is presently being accommodated. To better constrain the spatiotemporal evolution of faulting along the KES, we present 39 new apatite fission track, zircon U-Th-Sm/He, and Ar-40/Ar-39 cooling ages from a series of footwall transects along the KES graben shoulder. Combining these data with present-day topographic relief, 1-D thermokinematic and exhumational modeling documents successive stages, rather than synchronous deformation and gneiss dome exhumation. While the exhumation of the Kongur Shan commenced during the late Miocene, extensional processes in the Muztagh Ata massif began earlier and have slowed down since the late Miocene. We present a new model of synorogenic extension suggesting that thermal and density effects associated with a lithospheric tear fault along the eastern margin of the subducting Alai slab localize extensional upper plate deformation along the KES and decouple crustal motion between the central/western Pamir and eastern Pamir/Tarim basin.
In the last decade growing interest has emerged in quantifying the spatial and temporal variations in mountain building. Until recently, insufficient data have been available to attempt such a task at the scale of large orogens such as the Himalaya. The Himalaya accommodates ongoing convergence between India and Eurasia and is a focal point for studying orogen evolution and hypothesized interactions between tectonics and climate. Here we integrate 1126 published bedrock mineral cooling ages with a transient 1D Monte-Carlo thermal-kinematic erosion model to quantify the denudation histories along similar to 2700 km of the Himalaya. The model free parameter is a temporally variable denudation rate from 50 Ma to present. Thermophysical material properties and boundary conditions were tuned to individual study areas. Monte-Carlo simulations were conducted to identify the range of denudation histories that can reproduce the observed cooling ages. Results indicate large temporal and spatial variations in denudation and these are resolvable across different tectonic units of the Himalaya. More specifically, across > 1000 km of the southern Greater Himalaya denudation rates were highest (similar to 1.5-3 mm/yr) between similar to 10 and 2 Ma and lower (0.5-2.6 mm/yr) over the last 2 My. These differences are best determined in the NW-Himalaya. In contrast to this, across the similar to 2500 km length of the northern Greater Himalaya denudation rates vary over length scales of similar to 300-1700 km. Slower denudation (<1 mm/yr) occurred between 10 and 4 Ma followed by a large increase (1.2-2.6 mm/yr) in the last similar to 4 Ma. We find that only the southern Greater Himalayan Sequence clearly supports a continuous co-evolution of tectonics, climate and denudation. Results from the higher elevation northern Greater Himalaya suggest either tectonic driven variations in denudation due to a ramp-flat geometry in the main decollement and/or recent glacially enhanced denudation.
We recorded large data sets of swimming trajectories of the soil bacterium Pseudomonas putida. Like other prokaryotic swimmers, P. putida exhibits a motion pattern dominated by persistent runs that are interrupted by turning events. An in-depth analysis of their swimming trajectories revealed that the majority of the turning events is characterized by an angle of phi(1) = 180 degrees (reversals). To a lesser extent, turning angles of phi(2 Sigma Sigma Sigma Sigma) = 00 are also found. Remarkably, we observed that, upon a reversal, the swimming speed changes by a factor of two on average a prominent feature of the motion pattern that, to our knowledge, has not been reported before. A theoretical model, based on the experimental values for the average run time and the rotational diffusion, recovers the mean-square displacement of P. putida if the two distinct swimming speeds are taken into account. Compared to a swimmer that moves with a constant intermediate speed, the mean-square displacement is strongly enhanced. We furthermore observed a negative dip in the directional autocorrelation at intermediate times, a feature that is only recovered in an extended model, where the nonexponential shape of the run-time distribution is taken into account.
In the presence of a solid-liquid or liquid-air interface, bacteria can choose between a planktonic and a sessile lifestyle. Depending on environmental conditions, cells swimming in close proximity to the interface can irreversibly attach to the surface and grow into three-dimensional aggregates where the majority of cells is sessile and embedded in an extracellular polymer matrix (biofilm). We used microfluidic tools and time lapse microscopy to perform experiments with the polarly flagellated soil bacterium Pseudomonas putida (P. putida), a bacterial species that is able to form biofilms. We analyzed individual trajectories of swimming cells, both in the bulk fluid and in close proximity to a glass-liquid interface. Additionally, surface related growth during the early phase of biofilm formation was investigated. In the bulk fluid, P.putida shows a typical bacterial swimming pattern of alternating periods of persistent displacement along a line (runs) and fast reorientation events (turns) and cells swim with an average speed around 24 micrometer per second. We found that the distribution of turning angles is bimodal with a dominating peak around 180 degrees. In approximately six out of ten turning events, the cell reverses its swimming direction. In addition, our analysis revealed that upon a reversal, the cell systematically changes its swimming speed by a factor of two on average. Based on the experimentally observed values of mean runtime and rotational diffusion, we presented a model to describe the spreading of a population of cells by a run-reverse random walker with alternating speeds. We successfully recover the mean square displacement and, by an extended version of the model, also the negative dip in the directional autocorrelation function as observed in the experiments. The analytical solution of the model demonstrates that alternating speeds enhance a cells ability to explore its environment as compared to a bacterium moving at a constant intermediate speed. As compared to the bulk fluid, for cells swimming near a solid boundary we observed an increase in swimming speed at distances below d= 5 micrometer and an increase in average angular velocity at distances below d= 4 micrometer. While the average speed was maximal with an increase around 15% at a distance of d= 3 micrometer, the angular velocity was highest in closest proximity to the boundary at d=1 micrometer with an increase around 90% as compared to the bulk fluid. To investigate the swimming behavior in a confinement between two solid boundaries, we developed an experimental setup to acquire three-dimensional trajectories using a piezo driven objective mount coupled to a high speed camera. Results on speed and angular velocity were consistent with motility statistics in the presence of a single boundary. Additionally, an analysis of the probability density revealed that a majority of cells accumulated near the upper and lower boundaries of the microchannel. The increase in angular velocity is consistent with previous studies, where bacteria near a solid boundary were shown to swim on circular trajectories, an effect which can be attributed to a wall induced torque. The increase in speed at a distance of several times the size of the cell body, however, cannot be explained by existing theories which either consider the drag increase on cell body and flagellum near a boundary (resistive force theory) or model the swimming microorganism by a multipole expansion to account for the flow field interaction between cell and boundary. An accumulation of swimming bacteria near solid boundaries has been observed in similar experiments. Our results confirm that collisions with the surface play an important role and hydrodynamic interactions alone cannot explain the steady-state accumulation of cells near the channel walls. Furthermore, we monitored the number growth of cells in the microchannel under medium rich conditions. We observed that, after a lag time, initially isolated cells at the surface started to grow by division into colonies of increasing size, while coexisting with a comparable smaller number of swimming cells. After 5:50 hours, we observed a sudden jump in the number of swimming cells, which was accompanied by a breakup of bigger clusters on the surface. After approximately 30 minutes where planktonic cells dominated in the microchannel, individual swimming cells reattached to the surface. We interpret this process as an emigration and recolonization event. A number of complementary experiments were performed to investigate the influence of collective effects or a depletion of the growth medium on the transition. Similar to earlier observations on another bacterium from the same family we found that the release of cells to the swimming phase is most likely the result of an individual adaption process, where syntheses of proteins for flagellar motility are upregulated after a number of division cycles at the surface.
Background: Serotonin plays a pivotal role in regulating and modulating physiological and behavioral processes in both vertebrates and invertebrates. In the honeybee (Apis mellifera), serotonin has been implicated in division of labor, visual processing, and learning processes. Here, we present the cloning, heterologous expression, and detailed functional and pharmacological characterization of two honeybee 5-HT2 receptors.
Methods: Honeybee 5-HT2 receptor cDNAs were amplified from brain cDNA. Recombinant cell lines were established constitutively expressing receptor variants. Pharmacological properties of the receptors were investigated by Ca2+ imaging experiments. Quantitative PCR was applied to explore the expression patterns of receptor mRNAs.
Results: The honeybee 5-HT2 receptor class consists of two subtypes, Am5-HT2 alpha and Am5-HT2 beta. Each receptor gene also gives rise to alternatively spliced mRNAs that possibly code for truncated receptors. Only activation of the full-length receptors with serotonin caused an increase in the intracellular Ca2+ concentration. The effect was mimicked by the agonists 5-methoxytryptamine and 8-OH-DPAT at low micromolar concentrations. Receptor activities were blocked by established 5-HT receptor antagonists such as clozapine, methiothepin, or mianserin. High transcript numbers were detected in exocrine glands suggesting that 5-HT2 receptors participate in secretory processes in the honeybee.
Conclusions: This study marks the first molecular and pharmacological characterization of two 5-HT2 receptor subtypes in the same insect species. The results presented should facilitate further attempts to unravel central and peripheral effects of serotonin mediated by these receptors.
Korrelation zwischen der genetischen und der funktionellen Diversität humaner Bitterrezeptoren

Der Mensch besitzt ~25 funktionelle Bitterrezeptoren (TAS2R), die für die Wahrnehmung potenziell toxischer Substanzen in der Nahrung verantwortlich sind. Aufgrund der großen genetischen Variabilität der TAS2R-Gene könnte es eine Vielzahl funktionell unterschiedlicher TAS2R-Haplotypen geben, die zu Unterschieden der Bitterwahrnehmung führen. Dies konnte bereits in funktionellen Analysen und sensorischen Studien für einzelne Bitterrezeptoren gezeigt werden. In dieser Arbeit wurden die häufigsten Haplotypen aller 25 Bitterrezeptoren verschiedener Ethnien funktionell charakterisiert. Das Ziel war eine umfassende Aussage über die funktionelle Diversität der TAS2Rs, die die molekulare Grundlage für individuelle Bitterwahrnehmung bildet, treffen zu können. Fehlende Varianten wurden aus genomischer DNA kloniert oder durch gezielte Mutagenese bereits vorhandener TAS2R-Konstrukte generiert. Die funktionelle Analyse erfolgte mittels Expression der TAS2R-Haplotypen in HEK293TG16gust44 Zellen und anschließenden Calcium-Imaging-Experimenten mit zwei bekannten Agonisten. Die Haplotypen der fünf orphanen TAS2Rs wurden mit über hundert Bitterstoffen stimuliert. Durch die gelungene Deorphanisierung des TAS2R41 in dieser Arbeit, wurden für die 21 aktivierbaren TAS2Rs 36 funktionell-unterschiedliche Haplotypen identifiziert. Die tatsächliche funktionelle Vielfalt blieb jedoch deutlich hinter der genetischen Variabilität der TAS2Rs zurück. Neun Bitterrezeptoren wiesen funktionell homogene Haplotypen auf oder besaßen nur eine weltweit vorherrschende Variante. Funktionell heterogene Haplotypen wurden für zwölf TAS2Rs identifiziert. Inaktive Varianten der Rezeptoren TAS2R9, TAS2R38 und TAS2R46 sollten die Wahrnehmung von Bitterstoffen wie Ofloxacin, Cnicin, Hydrocortison, Limonin, Parthenolid oder Strychnin beeinflussen. Unterschiedlich sensitive Varianten, besonders der Rezeptoren TAS2R47 und TAS2R49, sollten für Agonisten wie Absinthin, Amarogentin oder Cromolyn ebenfalls zu phänotypischen Unterschieden führen. Wie für den TAS2R16 bereits gezeigt, traten Haplotypen des funktionell heterogenen TAS2R7 und TAS2R41 ethnien-spezifisch auf, was auf lokale Anpassung und verschiedene Phänotypen hinweisen könnte. Weiterführend muss nun eine Analyse der funktionell-variablen TAS2Rs in sensorischen Tests erfolgen, um ihre phänotypische Relevanz zu prüfen. Die Analyse der funktionsmodulierenden Aminosäurepositionen, z.Bsp. des TAS2R44, TAS2R47 oder TAS2R49, könnte weiterführend zum besseren Verständnis der Rezeptor-Ligand- und Rezeptor-G-Protein-Interaktion beitragen.

At z < 1 a large fraction of the baryons is thought to reside in diffuse gas that has been shock-heated to high temperatures (10 (5)-10 (6) K). Absorption by the 770.41, 780.32 A doublet of Ne viii in quasar spectra represents a unique tool to study this elusive warm-hot phase. We have developed an analytic model for the properties of Ne viii absorbers that allows for an inhomogeneous metal distribution. Our model agrees with the predictions of a simulation from the OverWhelmingly Large Simulations project indicating that the average line-of-sight metal-filling fraction within the absorbing gas is low (c(L) similar to 0.1). Most of the Ne viii in our model is produced in low-density, collisionally ionized gas (n(H) = 10(-6)-10(-4) cm(-3), T = 10 (5)-10 (6) K). Strong Ne viii absorbers (log(10)(N-NeVIII/cm(-2))14), like those recently detected by Hubble Space Telescope/Cosmic Origins Spectrograph, are found to arise in higher density gas (n(H) greater than or similar to 10(-4) cm(-3), T approximate to 5 x 10 (5) K). Ne viii cloudlets harbour only 1 per cent of the cosmic baryon budget. The baryon content of the surrounding gas (which has similar densities and temperatures as the Ne viii cloudlets) is a factor c(-1)L higher. We conclude that Ne viii absorbers are robust probes of shock-heated diffuse gas, but that spectra with signal-to-noise ratios S/N > 100 would be required to detect the bulk of the baryons in warm-hot gas.
Nonoxidized, biologically active parathyroid hormone determines mortality in hemodialysis patients

Background: It was shown that nonoxidized PTH (n-oxPTH) is bioactive, whereas the oxidation of PTH results in a loss of biological activity.
Methods: In this study we analyzed the association of n-oxPTH on mortality in hemodialysis patients using a recently developed assay system.
Results: Hemodialysis patients (224 men, 116 women) had a median age of 66 years. One hundred seventy patients (50%) died during the follow-up period of 5 years. Median n-oxPTH levels were higher in survivors (7.2 ng/L) compared with deceased patients (5.0 ng/L; P = .002). Survival analysis showed an increased survival in the highest n-oxPTH tertile compared with the lowest n-oxPTH tertile (chi(2), 14.3; P = 0008). Median survival was 1702 days in the highest n-oxPTH tertile, whereas it was only 453 days in the lowest n-oxPTH tertile. Multivariable-adjusted Cox regression showed that higher age increased odds for death, whereas higher n-oxPTH reduced the odds for death. Another model analyzing a subgroup of patients with intact PTH (iPTH) concentrations at baseline above the upper normal range of the iPTH assay (70 ng/L) revealed that mortality in this subgroup was associated with oxidized PTH but not with n-oxPTH levels.
Conclusions: The predictive power of n-oxPTH and iPTH on the mortality of hemodialysis patients differs substantially. Measurements of n-oxPTH may reflect the hormone status more precisely. The iPTH-associated mortality is most likely describing oxidative stress-related mortality.
Silver nanoparticles (SNP) are among the most commercialized nanoparticles. Here, we show that peptide-coated SNP cause functional impairment of human macrophages. A dose-dependent inhibition of phagocytosis is observed after nanoparticle treatment, and pretreatment of cells with N-acetyl cysteine (NAC) can counteract the phagocytosis disturbances caused by SNP.
Using the surface-sensitive mode of time-of-flight secondary ion mass spectrometry, in combination with multivariate statistical methods, we studied the composition of cell membranes in human macrophages upon exposure to SNP with and without NAC preconditioning. This method revealed characteristic changes in the lipid pattern of the cellular membrane outer leaflet in those cells challenged by SNP. Statistical analyses resulted in 19 characteristic ions, which can be used to distinguish between NAC pretreated and untreated macrophages. The present study discusses the assignments of surface cell membrane phospholipids for the identified ions and the resulting changes in the phospholipid pattern of treated cells. We conclude that the adverse effects in human macrophages caused by SNP can be partially reversed through NAC administration. Some alterations, however, remained.
We perform experiments with 72 electronic limit-cycle oscillators, globally coupled via a linear or nonlinear feedback loop. While in the linear case we observe a standard Kuramoto-like synchronization transition, in the nonlinear case, with increase of the coupling strength, we first observe a transition to full synchrony and then a desynchronization transition to a quasiperiodic state. However, in this state the ensemble remains coherent so that the amplitude of the mean field is nonzero, but the frequency of the mean field is larger than frequencies of all oscillators. Next, we analyze effects of common periodic forcing of the linearly or nonlinearly coupled ensemble and demonstrate regimes when the mean field is entrained by the force whereas the oscillators are not.
The coastal stretch of north-eastern Mediterranean Morocco holds vitally important ecological, social, and economic functions. The implementation of large-scale luxury tourism resorts shall push socio-economic development and facilitate the shift from a mainly agrarian to a service economy. Sufficient water availability and intact beaches are among the key requirements for the successful realization of regional development plans. The water situation is already critical, additional water-intense sectors could overstrain the capacity of water resources. Further, coastal erosion caused by sea-level rise is projected. Regional climate change is observable, and must be included in regional water management. Long-term climate trends are assessed for the larger region (Moulouya basin) and for the near-coastal zone at Saidia. The amount of additional water demand is assessed for the large-dimensioned Saidia resort; including the monthly, seasonal and annual tourist per capita water need under inclusion of irrigated golf courses and garden areas. A shift of climate patterns is observed, a lengthening of the dry summer season, and as well a significant decline of annual precipitation. Thus, current water scarcity is mainly human-induced; however, climate change will aggravate the situation. As a consequence, severe environmental damage due to water scarcity is likely and could impinge on the quality of local tourism. The re-adjustment of current management routines is therefore essential. Possible adjustments are discussed and the analysis concludes with management recommendations for innovative regional water management of tourism facilities.
Fluorescence probes consisting of well-established fluorophores in combination with rigid molecular rods based on spirane-type structures were investigated with respect to their fluorescence properties under different solvent conditions. The attachment of the dyes was accomplished by 1,3-dipolar cycloaddition between alkynes and azides (click' reaction) and is a prime example for a novel class of sensor constructs. Especially, the attachment of two (different) fluorophores on opposite sides of the molecular rods paves the way to new sensor systems with less bulky (compared to the conventional DNA- or protein-based concepts), nevertheless rigid spacer constructs, e.g., for FRET-based sensing applications. A detailed photophysical characterization was performed in MeOH (and in basic H2O/MeOH mixtures) for i) rod constructs containing carboxyfluorescein, ii) rod constructs containing carboxyrhodamine, iii) rod constructs containing both carboxyfluorescein and carboxyrhodamine, and iv) rod constructs containing both pyrene and perylene parts. For each dye (pair), two rod lengths with different numbers of spirane units were synthesized and investigated. The rod constructs were characterized in ensemble as well as single-molecule fluorescence experiments with respect to i) specific roddye and ii) dyedye interactions. In addition to MeOH and MeOH/NaOH, the rod constructs were also investigated in micellar systems, which were chosen as a simplified model for membranes.
This study focuses on the evolutionary relationships among Turkish species of the cave cricket genus Troglophilus. Fifteen populations were studied for sequence variation in a fragment (543 base pairs) of the mitochondrial DNA (mtDNA) 16S rDNA gene (16S) to reconstruct their phylogenetic relationships and biogeographic history. Genetic data retrieved three main clades and at least three divergent lineages that could not be attributed to any of the taxa known for the area. Molecular time estimates suggest that the diversification of the group took place between the Messinian and the Plio-Pleistocene.

Dieser Tagungsband beinhaltet die auf dem E-Learning Symposium 2012 an der Universität Potsdam vorgestellten Beiträge zu aktuellen Anwendungen, innovativen Prozesse und neuesten Ergebnissen im Themenbereich E-Learning. Lehrende, E-Learning-Praktiker und -Entscheider tauschten ihr Wissen über etablierte und geplante Konzepte im Zusammenhang mit dem Student-Life-Cycle aus. Der Schwerpunkt lag hierbei auf der unmittelbaren Unterstützung von Lehr- und Lernprozessen, auf Präsentation, Aktivierung und Kooperation durch Verwendung von neuen und etablierten Technologien.
A unified understanding of the relationship between disturbance and biodiversity is needed to predict biotic responses to global change. Recent advances have identified the need to deconstruct traditional models of disturbance into intensity and frequency to reconcile empirical studies that appear to generate contradictory associations between species diversity and disturbance. We integrate results from theoretical simulation modelling, field-based surveys of 5176 vegetation plots from 48 transects across 6 sites, and experimental pot-based manipulations of flooding to identify how disturbance drives species diversity within ephemeral wetlands in South Island, New Zealand. We find empirical, hump-shaped and positive relationships between species diversity and both disturbance intensity and frequency, mirroring patterns from a simulation model in which species differed in their demographic responses to disturbance. More generally, our simulations show that the relationships between diversity and disturbance shift from positive to hump-shaped to negative as species that are favored at low disturbance because of their resistance strategies, defined by low mortality and recruitment, decline within communities relative to resilient species. Resilient species with higher mortality and recruitment rates are instead favored as disturbance intensity and frequency intensify. Our theoretical findings suggest that sites must also have a third group of unique species with intermediate resilience and resistance. Analyses of community composition along our disturbance gradients support this prediction, emphasizing that shifts in community-level resistance and resilience drive empirical associations between diversity and disturbance. Overall, terrestrial plants may be unable to resist intense and frequent flooding, even with specialized traits. Only fast-growing species with high regeneration from seed may respond once flooding subsides and dominate community composition in these situations, especially on nutrient-rich soils. However, different strategies can co-occur at intermediate disturbance, ultimately increasing species richness. As disturbances become more pervasive globally, our results suggest that differences in the niches of species, rather than demographic stochasticity, drive biodiversity patterns. These niche-based processes may especially prevail, without accompanying losses in species richness, where sites are initially dominated by resistant taxa or life history strategies that balance resistance and resilience.
We investigate the potential of numerical algorithms to decipher the kinetic parameters involved in multi-step chemical reactions. To this end, we study dimerization kinetics of protein as a model system. We follow the dimerization kinetics using a stochastic simulation algorithm and combine it with three different optimization techniques (genetic algorithm, simulated annealing and parallel tempering) to obtain the rate constants involved in each reaction step. We find good convergence of the numerical scheme to the rate constants of the process. We also perform a sensitivity test on the reaction kinetic parameters to see the relative effects of the parameters for the associated profile of the monomer/dimer distribution.
We report the discovery of an unidentified, extended source of very-high-energy gamma-ray emission, VER J2019+407, within the radio shell of the supernova remnant SNR G78.2+2.1, using 21.4 hr of data taken by the VERITAS gamma-ray observatory in 2009. These data confirm the preliminary indications of gamma-ray emission previously seen in a two-year (2007-2009) blind survey of the Cygnus region by VERITAS. VER J2019+407, which is detected at a post-trials significance of 7.5 standard deviations in the 2009 data, is localized to the northwestern rim of the remnant in a region of enhanced radio and X-ray emission. It has an intrinsic extent of 0 degrees.23 +/- 0 degrees.03(stat-0 degrees.02sys)(+0 degrees.04) and its spectrum is well-characterized by a differential power law (dN/dE = N-0 x (E/TeV)-Gamma) with a photon index of Gamma = 2.37 +/- 0.14(stat) +/- 0.20(sys) and a flux normalization of N-0 = 1.5 +/- 0.2(stat) +/- 0.4(sys) x 10(-12) photon TeV-1 cm(-2) s(-1). This yields an integral flux of 5.2 +/- 0.8(stat) +/- 1.4(sys) x 10(-12) photon cm(-2) s(-1) above 320 GeV, corresponding to 3.7% of the Crab Nebula flux. We consider the relationship of the TeV gamma-ray emission with the GeV gamma-ray emission seen from SNR G78.2+2.1 as well as that seen from a nearby cocoon of freshly accelerated cosmic rays. Multiple scenarios are considered as possible origins for the TeV gamma-ray emission, including hadronic particle acceleration at the SNR shock.
Galaxy clusters are the largest known gravitationally bound objects, their study is important for both an intrinsic understanding of their systems and an investigation of the large scale structure of the universe. The multi- component nature of galaxy clusters offers multiple observable signals across the electromagnetic spectrum. At X-ray wavelengths, galaxy clusters are simply identified as X-ray luminous, spatially extended, and extragalactic sources. X-ray observations offer the most powerful technique for constructing cluster catalogues. The main advantages of the X-ray cluster surveys are their excellent purity and completeness and the X-ray observables are tightly correlated with mass, which is indeed the most fundamental parameter of clusters. In my thesis I have conducted the 2XMMi/SDSS galaxy cluster survey, which is a serendipitous search for galaxy clusters based on the X-ray extended sources in the XMM-Newton Serendipitous Source Catalogue (2XMMi-DR3). The main aims of the survey are to identify new X-ray galaxy clusters, investigate their X-ray scaling relations, identify distant cluster candidates, and study the correlation of the X-ray and optical properties. The survey is constrained to those extended sources that are in the footprint of the Sloan Digital Sky Survey (SDSS) in order to be able to identify the optical counterparts as well as to measure their redshifts that are mandatory to measure their physical properties. The overlap area be- tween the XMM-Newton fields and the SDSS-DR7 imaging, the latest SDSS data release at the starting of the survey, is 210 deg^2. The survey comprises 1180 X-ray cluster candidates with at least 80 background-subtracted photon counts, which passed the quality control process. To measure the optical redshifts of the X-ray cluster candidates, I used three procedures; (i) cross-matching these candidates with the recent and largest optically selected cluster catalogues in the literature, which yielded the photometric redshifts of about a quarter of the X-ray cluster candidates. (ii) I developed a finding algorithm to search for overdensities of galaxies at the positions of the X-ray cluster candidates in the photometric redshift space and to measure their redshifts from the SDSS-DR8 data, which provided the photometric redshifts of 530 groups/clusters. (iii) I developed an algorithm to identify the cluster candidates associated with spectroscopically targeted Luminous Red Galaxies (LRGs) in the SDSS-DR9 and to measure the cluster spectroscopic redshift, which provided 324 groups and clusters with spectroscopic confirmation based on spectroscopic redshift of at least one LRG. In total, the optically confirmed cluster sample comprises 574 groups and clusters with redshifts (0.03 ≤ z ≤ 0.77), which is the largest X-ray selected cluster catalogue to date based on observations from the current X-ray observatories (XMM-Newton, Chandra, Suzaku, and Swift/XRT). Among the cluster sample, about 75 percent are newly X-ray discovered groups/clusters and 40 percent are new systems to the literature. To determine the X-ray properties of the optically confirmed cluster sample, I reduced and analysed their X-ray data in an automated way following the standard pipelines of processing the XMM-Newton data. In this analysis, I extracted the cluster spectra from EPIC(PN, MOS1, MOS2) images within an optimal aperture chosen to maximise the signal-to-noise ratio. The spectral fitting procedure provided the X-ray temperatures kT (0.5 - 7.5 keV) for 345 systems that have good quality X-ray data. For all the optically confirmed cluster sample, I measured the physical properties L500 (0.5 x 10^42 – 1.2 x 10^45 erg s-1 ) and M500 (1.1 x 10^13 – 4.9 x 10^14 M⊙) from an iterative procedure using published scaling relations. The present X-ray detected groups and clusters are in the low and intermediate luminosity regimes apart from few luminous systems, thanks to the XMM-Newton sensitivity and the available XMM-Newton deep fields The optically confirmed cluster sample with measurements of redshift and X-ray properties can be used for various astrophysical applications. As a first application, I investigated the LX - T relation for the first time based on a large cluster sample of 345 systems with X-ray spectroscopic parameters drawn from a single survey. The current sample includes groups and clusters with wide ranges of redshifts, temperatures, and luminosities. The slope of the relation is consistent with the published ones of nearby clusters with higher temperatures and luminosities. The derived relation is still much steeper than that predicted by self-similar evolution. I also investigated the evolution of the slope and the scatter of the LX - T relation with the cluster redshift. After excluding the low luminosity groups, I found no significant changes of the slope and the intrinsic scatter of the relation with redshift when dividing the sample into three redshift bins. When including the low luminosity groups in the low redshift subsample, I found its LX - T relation becomes after than the relation of the intermediate and high redshift subsamples. As a second application of the optically confirmed cluster sample from our ongoing survey, I investigated the correlation between the cluster X-ray and the optical parameters that have been determined in a homogenous way. Firstly, I investigated the correlations between the BCG properties (absolute magnitude and optical luminosity) and the cluster global proper- ties (redshift and mass). Secondly, I computed the richness and the optical luminosity within R500 of a nearby subsample (z ≤ 0.42, with a complete membership detection from the SDSS data) with measured X-ray temperatures from our survey. The relation between the estimated optical luminosity and richness is also presented. Finally, the correlation between the cluster optical properties (richness and luminosity) and the cluster global properties (X-ray luminosity, temperature, mass) are investigated.
The Tibetan Plateau (TP), including its surrounding mountain ranges, represents the largest store of ice outside the polar regions. It hosts numerous lakes as well as the head waters of major Asian rivers, on which billions of people depend, and it is particularly sensitive to climate change. The moisture transport to the TP is controlled by the Indian and Pacific monsoon and the Westerlies. Understanding the evolution of the interaction of these circulation systems requires studies on climate archives in different spatial and temporal contexts. The objective of this study is to learn more about the interannual variability of precipitation patterns across the TP and how different hydrologic systems react to different climatic factors.
Aragonite shells of the aquatic gastropod Radix, which is widely distributed in the region, may represent suitable archives for inferring hydrologic and climatic signals in particularly high resolution. Therefore, sclerochronological studies of delta O-18 and delta C-13 ratios in Radix shells from seven lakes were conducted, each representing a different hydrologic and climatic setting, on a transect from the Pamirs across the TP.
The shell patterns exhibit an increasing influence of precipitation and a decreasing influence of evaporation on the isotope compositions from west to east. delta O-18 values of shells from lakes on the eastern and central TP (Donggi Cona, Yamdrok Yumco, Tarab Co) mirror monsoon signals, indicated by more negative values and higher variabilities compared to the more western lakes (Karakul, Bangong/Nyak, Manasarovar). In Yadang Co, located on the central southern TP, the monsoon rains did not reach the lake in the sampling year, although it is located in a region which is usually affected by monsoon circulation. The delta O-18 values are used to differentiate the annual hydrological cycle into ice cover period, melt water period, precipitation period and evaporation period. delta C-13 compositions in the shells particularly depend on specific habitats, which vary in biological productivity and in carbon sources. delta O-18 and delta C-13 patterns show a positive covariance in shells originating from large closed basins. The results show that Radix shells mirror general climatic differences between the seven lake regions. These differences reflect both regional and local climate signals in sub-seasonal resolution, without noticeable dependence on the particular lake system.

Die neue Ausgabe des Potsdamer Forschungsmagazins widmet sich dem Thema „Grenzen“ aus unterschiedlichsten Perspektiven.
Als Sprachwissenschaftlerin denke ich bei diesem Stichwort an sprachliche Grenzen und die Wirkungen, die sich aus dem Kontakt von zwei Sprachen an einer Sprachgrenze ergeben können. Vielfältig sind die Belege für das sogenannte Code-Switching, dem Wechsel von einer Sprache in die andere in einer bestimmten Äußerungssituation. Die Motive für einen solchen Sprachwechsel können ganz unterschiedlicher Natur sein: So lässt sich Code-Switching einerseits auf eine eingeschränkte sprachliche Kompetenz zurückführen, wenn beispielsweise einer Sprecherin ein bestimmtes Wort in der Zweitsprache fehlt, andererseits kann das Code-Switching prestigebedingt sein, wenn ein Sprecher durch einen sprachlichen Wechsel seine Zugehörigkeit zu einer bestimmten gesellschaftlichen Gruppe demonstrieren möchte.
Wenn Code-Switching nicht nur punktuell stattfindet, sondern ganze Sprach- gemeinschaften über einen längeren Zeitraum erfasst, kann es zu weitreichenden Veränderungen der betroffenen Sprachen kommen. Welche Sprache „gibt“ und welche Sprache „nimmt“, hängt von sprachexternen Faktoren ab. So ist es ein Leichtes vorherzusagen, dass die deutschen Varietäten der Sprachinseln in Süd- und Osteuropa wie auch in Nord- und Südamerika wohl zunehmend Material und Muster aus den sie umgebenden Sprachen aufnehmen und letztendlich in ihnen aufgehen werden, wenn sie nicht durch politischen Willen konserviert werden. Das Ausmaß von Sprachkontakt ist mit der zunehmenden räumlichen Mobilität moderner Gesellschaften stark angestiegen, und lässt sich sicher nicht auf den aktuell immer wieder thematisierten Sprachkontakt des Deutschen mit dem Englischen reduzieren. Historisch gesehen ist das Deutsche vor allem durch die romanischen Sprachen stark geprägt worden – in Potsdam denkt man hier unwillkürlich an den starken Einfluss des Französischen im 18. Jahrhundert.
Die Überwindung sprachlicher Grenzen zeigt sich auch im Alltag einer international ausgerichteten Forschungsuniversität: So hat im März dieses Jahres in Potsdam die Jahrestagung der deutschen Gesellschaft für Sprachwissenschaft mit über 500 Teilnehmern stattgefunden. Lingua Franca der Tagung war Englisch, was den Anteil der internationalen Teilnehmer gegenüber früheren Jahrestagungen nochmals vergrößert hat.
Zahlreiche andere Zugänge zum Thema Grenzen bieten die Beiträger dieser Ausgabe des Forschungsmagazins: Auf den Spuren von „Grenzvermessungen“ bewegen sich die Texte zum Australienforscher Ludwig Leichhardt oder zur Energiebilanz im Spitzensport. „Grenzgänger“ stehen im Fokus der Beiträge über eine Forschergruppe zur Literatur der kolonialen Karibik oder die tief in die Erde reichenden Forschungen eines italienischen Geologen. Auf der Suche nach dem „Grenzenlosen“ folgen die Autoren den Wissenschaftlern etwa zur Frage „Why love hurts?“ oder hinein in eine Geschichte des Musikhörens. Den umgekehrten Weg, nämlich „Grenzziehungen“, beobachtet „Portal Wissen“ in der Arbeit des Potsdamer MenschenRechtsZentrums oder den Auswertungen des Nationalen Dopingpräventionsplans. Belege für erfolgreiche „Grenzüberschreitungen“ liefern schließlich Blicke ins „Taschentuchlabor“ oder die digitale Edition mittelalterlicher Prosaepen, um nur einige Beiträge aus diesem Heft herauszugreifen.

In this work, the development of temperature- and protein-responsive sensor materials based on biocompatible, inverse hydrogel opals (IHOs) is presented. With these materials, large biomolecules can be specifically recognised and the binding event visualised. The preparation of the IHOs was performed with a template process, for which monodisperse silica particles were vertically deposited onto glass slides as the first step. The obtained colloidal crystals with a thickness of 5 μm displayed opalescent reflections because of the uniform alignment of the colloids. As a second step, the template was embedded in a matrix consisting of biocompatible, thermoresponsive hydrogels. The comonomers were selected from the family of oligo(ethylene glycol)methacrylates. The monomer solution was injected into a polymerisation mould, which contained the colloidal crystals as a template. The space in-between the template particles was filled with the monomer solution and the hydrogel was cured via UV-polymerisation. The particles were chemically etched, which resulted in a porous inner structure. The uniform alignment of the pores and therefore the opalescent reflection were maintained, so these system were denoted as inverse hydrogel opals. A pore diameter of several hundred nanometres as well as interconnections between the pores should facilitate a diffusion of bigger (bio)molecules, which was always a challenge in the presented systems until now. The copolymer composition was chosen to result in a hydrogel collapse over 35 °C. All hydrogels showed pronounced swelling in water below the critical temperature. The incorporation of a reactive monomer with hydroxyl groups ensured a potential coupling group for the introduction of recognition units for analytes, e.g. proteins. As a test system, biotin as a recognition unit for avidin was coupled to the IHO via polymer-analogous Steglich esterification. The amount of accessible biotin was quantified with a colorimetric binding assay. When avidin was added to the biotinylated IHO, the wavelength of the opalescent reflection was significantly shifted and therefore the binding event was visualised. This effect is based on the change in swelling behaviour of the hydrogel after binding of the hydrophilic avidin, which is amplified by the thermoresponsive nature of the hydrogel. A swelling or shrinking of the pores induces a change in distance of the crystal planes, which are responsible for the colour of the reflection. With these findings, the possibility of creating sensor materials or additional biomolecules in the size range of avidin is given.
Through the reactions of 1-aminomethyl-2-naphthol and substituted 1-aminobenzyl-2-naphthols with 3,4-dihydroisoquinoline or 6,7-dimethoxy-3,4-dihydroisoquinoline under microwave conditions, naphth[1,2-e][1,3]oxazino[2,3-a]-isoquinoline derivatives were prepared in good yields. The latter reaction was extended by using 2-aminoarylmethyl-1-naphthols, leading to isomeric naphth-[2,1-e][1,3]oxazino[2,3-a] isoquinolines. Beside the detailed NMR spectroscopic and theoretical study of both stereochemistry and dynamic behaviour of these new conformational flexible heterocyclic ring systems an unexpected dynamic process between two diastereomers was observed in solution, studied by variable temperature H-1 NMR spectroscopy and the mechanism proved by theoretical DFT computations.
Annually laminated (varved) lake sediments with intercalated detrital layers resulting from sedimentary input by runoff events are ideal archives to establish precisely dated records of past extreme runoff events. In this study, the mid- to late Holocene varved sediments of Lake Mondsee (Upper Austria) were analysed by combining sedimentological, geophysical and geochemical methods. This approach allows to distinguish two types of detrital layers related to different types of extreme runoff events (floods and debris flows) and to detect changes in flood activity during the last 7100 years. In total, 271 flood and 47 debris flow layers, deposited during spring and summer, were identified, which cluster in 18 main flood episodes (FE 1-18) with durations of 30-50 years each. These main flood periods occurred during the Neolithic (7100-7050 vyr BP and 6470-4450 vyr BP), the late Bronze Age and the early Iron Age (3300-3250 and 2800-2750 vyr BP), the late Iron Age (2050-2000 vyr BP), throughout the Dark Ages Cold Period (1500-1200 vyr BP), and at the end of the Medieval Warm Period and the Little Ice Age (810-430 vyr BP).
Summer flood episodes in Lake Mondsee are generally more abundant during the last 1500 years, often coinciding with major advances of Alpine glaciers. Prior to 1500 vyr BP, spring/summer floods and debris flows are generally less frequent, indicating a lower number of intense rainfall events that triggered erosion. In comparison with the increase of late Holocene flood activity in western and northwestern (NW) Europe, commencing already as early as 2800 yr BP, the hydro-meteorological shift in the Lake Mondsee region occurred much later. These time lags in the onset of increased hydrological activity might be either due to regional differences in atmospheric circulation pattern or to the sensitivity of the individual flood archives. The Lake Mondsee sediments represent the first precisely dated and several millennia long summer flood record for the northeastern (NE) Alps, a key region at the climatic boundary of Atlantic, Mediterranean and East European air masses, aiding a better understanding of regional and seasonal peculiarities of flood occurrence under changing climate conditions. Context. Recent studies of O-type stars have demonstrated that discrepant mass-loss rates are obtained when different diagnostic methods are employed. Fitting the unsaturated UV resonance lines (e.g., P v) gives drastically lower values than obtained from the Ha emission. Wind inhomogeneity (so-called "clumping") may be the main cause of this discrepancy. Aims. In a previous paper, we presented 3D Monte-Carlo calculations for the formation of scattering lines in a clumped stellar wind. In the present paper we select five O-type supergiants (from 04 to 07) and test whether the reported discrepancies can be resolved this way. Methods. In the first step, the analyses started with simulating the observed spectra with Potsdam Wolf-Rayet (PoWR) non-LTE model atmospheres. The mass-loss rates are adjusted to fit to the observed Ha emission lines best. For the unsaturated UV resonance lines (i.e., P v) we then applied our 3D Monte-Carlo code, which can account for wind clumps of any optical depths ("macroclumping"), a non-void interclump medium, and a velocity dispersion inside the clumps. The ionization stratifications and underlying photospheric spectra were adopted from the PoWR models. The properties of the wind clumps were constrained by fitting the observed resonance line profiles. Results. Our results show that with the mass-loss rates that fit Ha (and other Balmer and He II lines), the UV resonance lines (especially the unsaturated doublet of P v) can also be reproduced with no problem when macroclumping is taken into account. There is no need to artificially reduce the mass-loss rates or to assume a subsolar phosphorus abundance or an extremely high clumping factor, unlike what was claimed by other authors. These consistent mass-loss rates are lower by a factor of 1.3 to 2.6, compared to the mass-loss rate recipe from Vink et al. Conclusions. Macroclumping resolves the previously reported discrepancy between Ha and P v mass-loss diagnostics.
A detailed study of inelastic x-ray scattering from the ground state to the (3)Sigma(g)(3 sigma(-1)(g)3s(g)(1)) state of the O-2 molecule is presented. The observed angular anisotropy shows that the vibrational excitations within this final state are strongly dependent on the polarization of the incident radiation. The analysis demonstrates that this is a manifestation of interference between resonant and direct nonresonant inelastic x-ray scattering. This interference provides a new tool to monitor nuclear dynamics by relative rotation of the polarization vectors of the incident and scattered photons.
Natural genetic diversity provides a powerful tool to study the complex interrelationship between metabolism and growth. Profiling of metabolic traits combined with network-based and statistical analyses allow the comparison of conditions and identification of sets of traits that predict biomass. However, it often remains unclear why a particular set of metabolites is linked with biomass and to what extent the predictive model is applicable beyond a particular growth condition. A panel of 97 genetically diverse Arabidopsis (Arabidopsis thaliana) accessions was grown in near-optimal carbon and nitrogen supply, restricted carbon supply, and restricted nitrogen supply and analyzed for biomass and 54 metabolic traits. Correlation-based metabolic networks were generated from the genotype-dependent variation in each condition to reveal sets of metabolites that show coordinated changes across accessions. The networks were largely specific for a single growth condition. Partial least squares regression from metabolic traits allowed prediction of biomass within and, slightly more weakly, across conditions (cross-validated Pearson correlations in the range of 0.27-0.58 and 0.21-0.51 and P values in the range of <0.001-<0.13 and <0.001-<0.023, respectively). Metabolic traits that correlate with growth or have a high weighting in the partial least squares regression were mainly condition specific and often related to the resource that restricts growth under that condition. Linear mixed-model analysis using the combined metabolic traits from all growth conditions as an input indicated that inclusion of random effects for the conditions improves predictions of biomass. Thus, robust prediction of biomass across a range of conditions requires condition-specific measurement of metabolic traits to take account of environment-dependent changes of the underlying networks.

Context. The Be/X-ray binary SXP 1062 is of especial interest owing to the large spin period of the neutron star, its large spin-down rate, and the association with a supernova remnant constraining its age. This makes the source an important probe for accretion physics.
Aims. To investigate the long-term evolution of the spin period and associated spectral variations, we performed an XMM-Newton target-of-opportunity observation of SXP 1062 during X-ray outburst.
Methods. Spectral and timing analysis of the XMM-Newton data was compared with previous studies, as well as complementary Swift/XRT monitoring and optical spectroscopy with the SALT telescope were obtained.
Results. The spin period was measured to be P-s = (1071.01 +/- 0.16) s on 2012 Oct. 14. The X-ray spectrum is similar to that of previous observations. No convincing cyclotron absorption features, which could be indicative for a high magnetic field strength, are found. The high-resolution RGS spectra indicate the presence of emission lines, which may not completely be accounted for by the SNR emission. The comparison of multi-epoch optical spectra suggest an increasing size or density of the decretion disc around the Be star.
Conclusions. SXP 1062 showed a net spin-down with an average of P-s = ( 2.27 +/- 0.44) s yr(-1) over a baseline of 915 days.
Facies bodies in geostatistical models of deep-water depositional environments generally represent channel-levee-overbank-lobe morphologies. Such models adequately capture one set of the erosional and depositional processes resulting from turbidity currents traveling downslope to the ocean basin floor. However, depositional morphologies diverge from the straight forward channel-levee-overbank-lobe paradigm when the topography of the slope or the shape of the basin impacts the timing and magnitude of turbidity current deposition. Subaqueous mass-transport-deposits (MTDs) present the need for an exception to the channel-levee-overbank-lobe archetype. Irregular surface topography of subaqueous MTDs can play a primary role in controlling sand deposition from turbidity currents. MTD topography creates mini-basins in which sand accumulates in irregularly-shaped deposits. These accumulations are difficult to laterally correlate using well-log data due to their variable and unpredictable shape and size. Prediction is further complicated because sandstone bodies typical of this setting are difficult to resolve in seismic-reflection data. An event-based model is presented, called DFTopoSim, which simulates debris flows and turbidity currents. The accommodation space on top of and between debris flow lobes is filled in by sand from turbidity currents. When applied to a subsurface case in the Molasse Basin of Upper Austria, DFTopoSim predicts sand packages consistent with observations from core, well, and seismic data and the interpretation of the sedimentologic processes. DFTopoSim expands the set of available geostatistical deep-water depositional models beyond the standard channel-levee-overbank-lobe model.
