The G protein-coupled estrogen receptor (GPER1) is acknowledged as an important mediator of estrogen signaling. Given the ubiquitous expression of GPER1, it is likely that the receptor plays a role in a variety of malignancies, not only in the classic hormonally regulated tissues (e.g., breast, ovary, and prostate), but also in the colon. As colorectal cancer (CRC) is the third most common cancer in both men and women worldwide and environmental factors and dietary habits are important risk factors, it is increasingly recognized that natural and synthetic hormones and their associated receptors might play a role in CRC. Through oral consumption, environmental contaminants with endocrine activity are in contact with the gastrointestinal mucosa, where they might exert their toxic effects. Although GPER1 has been shown to be engaged in physiological and pathophysiological processes, its role in CRC remains poorly understood. Thus, pro- as well as anti-tumorigenic effects are described in the literature. This thesis has uncovered novel roles of GPER1 in mediating major CRC-associated phenotypes in transformed and non-transformed colon cell lines. Exposure to the estrogens 17β-estradiol (E2), bisphenol-A (BPA) and diethylstilbestrol (DES) but also the androgen dihydrotestosterone (DHT) resulted in GPER1-dependent induction of supernumerary centrosomes, whole chromosomal instability (w-CIN) and aneuploidy. Indeed, both knockdown and inhibition of GPER1 attenuated the generation of (xeno)hormone-driven supernumerary centrosomes and karyotype instability. Mechanistically, (xeno)hormone-induced centrosome amplification was associated with transient multipolar mitosis and the generation of so called anaphase “lagging” chromosomes. The results of this thesis propose a GPER1/PKA/AKAP9-pathway in regulating centrosome numbers in colorectal cancer cells and the involvement of the centriolar protein centrin. Remarkably, exposure to (xeno)hormones resulted in atypical enlargement and unexpected phosphorylation of the centriole marker centrin in interphase. These findings provide a novel role for GPER1 in key CRC-prone lesions and shed light on underlying mechanisms that involve GPER1 function in the colon. Elucidating to what extent centrosomal proteins are involved in the GPER1-mediated aneugenic effect will be an important task for future studies. The present study was intended to lay a first foundation to understand the molecular basis and potential risk factors of CRC which might help to reduce the use of laboratory animals. Since numerous animal experiments are conducted in biomedical research, the development of alternative methods is indispensable. The Federal Institute for Risk Assessment (BfR) as the German Center for the Protection of Laboratory Animals (Bf3R) addresses this issue by uncovering underlying mechanisms leading to colorectal cancer as necessary prerequisite in order to develop alternative methods.
Krafttraining ist ein etabliertes Mittel für die Entwicklung körperlicher Fitness und sport-artspezifischer Leistungen in verschiedenen Sportarten. Dabei scheint dem Krafttraining auch im Hinblick auf den langfristigen Leistungsaufbau im Nachwuchsleistungssport Rudern eine besondere Rolle zuzukommen. Die vorliegende Arbeit widmet sich der Optimie-rung der Leistungsdiagnostik sowie der Bewertung der Effektivität von Krafttrainingsmethoden zur Verbesserung der körperlichen Fitness und der sportartspezifischen Leistung im Nachwuchsleistungsrudern.
Die erste Studie im Rahmen dieser kumulativen Arbeit beinhaltet die Analyse eines sport-artspezifischen Anforderungsprofils der Sportart Rudern. Die Studienergebnisse zeigen, dass die Energiebereitstellung im Ruderwettkampf zu einem großen Anteil aus aeroben und zu einem geringen Anteil aus anaeroben Stoffwechselprozessen erfolgt. Während der vier Phasen des rudertechnischen Leitbildes werden nahezu alle großen Muskelgruppen beansprucht. Aufgrund der hohen Trainingsumfänge auf internationalem Leistungsniveau besteht bei Ruderern ein erhöhtes Risiko, Verletzungen im lumbalen Rückensegment, den Knien sowie den Rippen zu erleiden.
Im Rahmen der zweiten Studie wurden die Effekte von Krafttraining auf die körperliche Fitness und die sportartspezifische Leistung von Ruderern unterschiedlichen Expertisen-niveaus untersucht. Die Studienergebnisse zeigen, dass signifikante kleine Effekte des Krafttrainings auf die körperliche Fitness (Maximalkraft der unteren Gliedmaßen) und auf die sportartspezifische Leistung existieren. Subgruppenanalysen für Krafttrainingstyp und Expertisenniveau zeigten nicht signifikante Unterschiede zwischen den jeweiligen Subgruppen der Ruderer.
Die dritte Studie analysierte die Zusammenhänge zwischen der biologischen Reife, der Körperkonstitution und der körperlichen Fitness mit der Leistung auf dem Ruderergometer über 700-m bei weiblichen Nachwuchseliteruderern. Die Analyse zeigte signifikante mittle-re bis hohe Zusammenhänge zwischen der biologischen Reife, Kennwerten der Körperkonstitution, der körperlichen Fitness (z. B. Maximalkraft, Kraftausdauer) und der Leistung auf dem Ruderergometer. Zusätzlich konnte gezeigt werden, dass die Prädiktoren Körperkonstitution (Magermasse) und Kraftausdauer (Bourban-Test) die Leistung des 700-m Ruderergometertest am besten abbilden.
Die vierte Studie untersuchte die Effekte von zwei unterschiedlichen Krafttrainingsmetho-den, die jeweils über neun Wochen durchgeführt wurden, auf die körperliche Fitness und die Leistung auf dem Ruderergometer bei weiblichen Elite-Nachwuchsruderern. Die Stu-dienergebnisse zeigen, dass beide Gruppen signifikante Steigerungen auf ausgewählte Komponenten der körperlichen Fitness (z. B. Maximalkraft, Schnellkraft, anaerobe Aus-dauer und Richtungswechselgeschwindigkeit) und die sportartspezifische Leistung erzielten. Signifikant größere Zuwächse resultierten nach Krafttraining mit höheren Intensitäten [HI] bei gleichem Volumen im Vergleich zu Kraftausdauertraining [KA] in ausgewählten Komponenten der körperlichen Fitness (z. B. Maximalkraft). Im Gegensatz dazu, führte KA bei gleichem Volumen im Vergleich zu HI zu größeren Steigerungen der sportartspezifischen Leistung.
Zusammenfassend zeigen die Ergebnisse, dass Krafttraining ein effektives Mittel zur Steigerung ausgewählter Komponenten der körperlichen Fitness und der sportartspezifischen Leistung in der Sportart Rudern für RuderInnen und im Speziellen für den Nachwuchssport ist. Insbesondere im Nachwuchsleistungsrudern zeigte sich, dass HI zusätzlich zum regulären Rudertraining effektiver zur Verbesserung der körperlichen Fitness (z. B. Maximalkraft) ist als KA. Zur Verbesserung der sportartspezifischen Leistung jedoch, stellte KA im Vergleich zu HI bei weiblichen Elite-Nachwuchsruderern die effektivere Krafttrainings-methode dar. Weiterhin wird Trainern im Nachwuchsrudersport empfohlen, ausgewählte Merkmale der biologischen Reife, Körperkonstitution sowie der körperlichen Fitness (Maximalkraft, Kraftausdauer) für die Talententwicklung zu berücksichtigen.
In nature, plants often encounter biotic and abiotic stresses, which can cause reduced crop yield and quality, and threaten the nutrition of a growing human population. As heat stress (HS) is one of the main abiotic stresses, and is projected to increase due to global warming, it is necessary to better understand how plants respond and survive under HS. In Arabidopsis thaliana, plants can survive under severe HS if primed by a non-lethal HS, a process called acquisition of thermotolerance. This primed stated can be maintained for several days, and the ability of plants to maintain the primed state is called maintenance of acquired thermotolerance (mATT) or HS memory. According to current research, two Heat shock factors (HSFs) HSFA2 and HSFA3 are known to account for the majority of mATT capability, and there are other HSFs e.g. HSFA1b and HSFA6b in HSF complexes containing HSFA2 and/or HSFA3, however, the roles of these HSFs in HS memory is not clearly understood. Moreover, the mechanism of these HSFs in regulating HS memory is unclear, whether transcriptional machinery e.g. the Mediator complex contributes to transcriptional memory. This work investigates the role of HSFs and Mediator subunits in HS memory in A. thaliana. For the role of HSFs, the interaction between HSFA1b and HSFA2 during HS memory phase was confirmed by in vivo co- immunoprecipitation (Co-IP). HSFA1b, HSFA2, HSFA3 and HSFA6b targeted HS memory-related genes according to DNA affinity purification sequencing (DAP-seq) data, and targets of HSFA1b were confirmed in vivo by chromatin immunoprecipitation qPCR (ChIP-qPCR). The mutant of hsfa6b showed an HS memory deficiency phenotype in mATT survival assay. These data confirmed the role for HSFA2 and HSFA3 in HS memory, and suggest that HSFA1b and HSFA6b also function in HS memory. The Mediator complex functions as an RNA Polymerase II (RNA Pol II) co-regulator, and includes Head, Middle, Tail and Kinase modules. Both MED23 and MED32 belong to the Tail module, and they have a positive role in HS memory. MED23 interacted with HSFA3, as determined by yeast two hybrid (Y2H) and in vivo Co-IP assays. The med23 mutant showed a decreased HS memory phenotype, reduced expression of Type I (sustained expression) memory genes following HS, and reduced accumulation of the memory-associated Tri-methylation of histone H3 lysine 4 (H3K4me3)histone modification at HS memory-related gene loci after HS. MED23 was recruited to HS-inducible memory and non-memory genes after HS, as determined by ChIP-qPCR. The med32
mutant showed a reduced HS memory phenotype, decreased expression of Type I and Type II (hyper-induction) memory genes, and lower accumulation of H3K4me3 at memory gene lociafter HS. However, MED32 did not show interaction with any tested HSF in Y2H or in vivo Co-IP. MED32 regulated the recruitment of RNA Pol II at HS-inducible genes after HS, but was not itself recruited to HS memory genes after HS. These results provided more evidence
that the Mediator subunits MED23 and MED32 regulate HS memory on transcriptional and epigenetic levels. In general, this work provides a better insight into the molecular mechanism of how HSFs and Mediator subunits regulate HS memory in plants and will provide new perspectives to breed crops with improved thermotolerance.
Functional materials, also called "Smart Materials", are described by their ability to fulfill a desired task through targeted interaction with its environment. Due to this functional integration, such materials are of increased interest, especially in areas where the increasing micronization of components is required. Modern manufacturing processes (e.g. microfluidics) and the availability of a wide variety of functional materials (e.g. shape memory materials) now enable the production of particle-based switching components. This category includes micropumps and microvalves, whose basic function is the active control of liquid flows. One approach in realizing those microcomponents as pursued by this work, enables variable size-switching of water-filled microballoons by implementing a stimulus-sensitive switching motif in the capsule's membrane shell, while being under the influence of a constant driving force. The switching motif with its gatekeeper function has a critical influence on one or more material parameters, which modulate the capsule's resistance against the driving force in microballoon expansion process. The advantage of this concept is that even non-variable analyte conditions, such as concentration levels of ions, can be capitalized to generate external force fields that, under the control of the membrane, cause an inflation of the microballoon by an osmotically driven water influx. In case of osmotic pressure gradients as the driving force for the capsule expansion, material parameters associated with the gatekeeper function are specifically the permeability and the mechanical stiffness of the shell material. While a modulation of the shell permeability could be utilized to kinetically impede the water influx on large time scales, a modulation of the shell's mechanical stiffness even might be utilized to completely prevent the capsule inflation due to a possible non-deformability beneath a certain threshold pressure. In polymer networks, which are a suitable material class for the demanded capsule shell because of their excellent elasticity, both the permeability and the mechanical properties are strongly influenced by the crystallinity of the material. Since the permeability is effectively reduced with increasing crystallinity, while the mechanical stiffness is simultaneously greatly increased, both effects point in the same direction in terms of their functional relationship. For this reason and due to a reversible and contactless modulation of the membrane crystallinity by heat input, crystallites may be suitable switching motifs for controlling the capsule expansion. As second design element of reversible expandable microballoons, the capsule geometry, defined by an aqueous core enveloped by the temperature-sensitive polymer network membrane, should allow an osmotic pressure gradient across the membrane layer. The strength of the inflation pressure and the associated inflation velocity upon membrane melting should be controlled by the salt concentration within the aqueous core, while a turn in the osmotic gradient should furthermore allow the reversible process of capsule deflation. Therefore, it should be possible to build either microvalves and micropumps, while their intended action of either pumping or valving is determined by their state of expansion and the direction of the osmotic pressure gradient.. Microballoons of approximately 300 µm in diameter were formed via droplet-based microfluidics from double-emulsion templates (w/o/w). The elastomeric capsule membrane was formed by photo-crosslinking of methacrylate (MA) functionalized oligo(ε-caprolactone) precursors (≈ 3.8 MA-arms, Mn ≈ 12000 g mol-1) within the organic medium layer (o) via UV-exposure after droplet-formation. After removal of the toluene/chloroform mixture by slow extraction via the continuous aqueous phase, the capsules solidified under the development of a characteristic "mushroom"-like shape at specific experimental conditions (e.g. λ = 308 nm, 57 mJ·s-1·cm-2, 16 min). It could be furthermore shown that in dependency to the process parameters: oligomer concentration and curing-time also spherical capsules were accessible. Long curing-times and high oligomer concentrations at a fixed light-intensity favored the formation of "mushroom"-like capsules, whereas the contrary led to spherical shaped capsules. A comparative study on thin polymer network films of same composition and equal treatment proved a correlation between the film's crosslink density and their contraction capability, while stronger crosslinked polymer networks showed a stronger contraction after solvent removal. In combination with observations during capsule solidification via light-microscopy, where a continuous shaping from almost spherical crosslinked templates to "mushroom"-shaped and solidified capsules was stated, the following mechanism was proposed. In case of low oligomer contents and short curing-times, the contraction of the capsule shell during solvent removal is strongly diminished due to a low degree of crosslinking. Therefore, the solidifying shell could freely collapse onto the aqueous core. In the other case, high oligomer concentrations and long curing-times will favor the formation of highly crosslinked capsule membranes with a strong contraction capability. Due to an observed decentered location of the aqueous core within the swollen polymer network, an uneven radial stress along the capsule's circumference is exerted to the incompressible core. This lead to an uneven contraction during solvent removal and a directed flow of the core fluid into the direction of the minimal stress vector. In consequence, the initially thicker spherical cap contracts, whereas the opposing thinner spherical cap get stretched. The "mushroom"-shape over some advantages over their spherical shaped counterparts, why they were selected for the further experiments. Besides the necessity of a high density of crosslinking for the purpose of extraordinary elasticity and toughness, the form-anisotropy promotes a faster microballoon expandability due to a partial reduction of the membrane thickness. Additionally, pre-stretched regions of thin thickness might provide a better resistance against inflation pressure than spherical but non-stretched capsules of equal membrane thickness. The resulting "mushroom"-shaped microcapsules exhibited a melting point of Tm ≈ 50 - 60 °C and a degree of crystallinity of Xc ≈ 29 - 38 % depending on the membrane thickness and internal salt content, which is slightly lower than for the non-crosslinked oligomer and reasoned by a limited chain mobility upon crosslinking. Nonetheless, the melting transition of the polymer network was associated with a strong drop in its mechanical stiffness, which was shown to have a strong influence on the osmotic driven expansion of the microcapsules. Capsules that were subjected to osmotic pressures between 1.5 and 4.7 MPa did not expand if the temperature was well below the melting point of the capsule's membrane, i.e. at room temperature. In contrast, a continuous expansion, while approaching asymptotically to a final capsule size, was observed if the temperature exceeded the melting point, i.e. 60 °C. Microballoons, which were kept for 56 days at ∆Π = 1.5 MPa and room temperature, did not change significantly in diameter, why the impact of the mechanical stiffness on the expansion behavior is considered to be the greater than the influence of the shell permeability. The time-resolved expansion behavior of the microballoons above their Tm was subsequently modeled, using difusion equations that were corrected for shape anisotropy and elastic restoring forces. A shape-related and expansion dependent pre-factor was used to dynamically address the influence of the shell thickness differences along the circumference on the inflation velocity, whereas the microballoon's elastic contraction upon inflation was rendered by the inclusion of a hyperelastic constitutive model. An important finding resulting from this model was the pronounced increase in inflation velocity compared to hypothetical capsules with a homogeneous shell thickness, which stresses the benefit of employing shape anisotropic balloon-like capsules in this study. Furthermore, the model was able to predict the finite expandability on basis of entropy-elastic recovery forces and strain-hardening effects. A comparison of six different microballoons with different shell thicknesses and internal salt contents showed the linear relationship between the volumetric expansion, the shell thickness and the applied osmotic pressure, as represented by the model. As the proposed model facilitates the prediction of the expansion kinetics depending on the membranes mechanical and diffusional characteristics, it might be a screening tool for future material selections. In course of the microballoon expansion process, capsules of intermediate diameters could be isolated by recrystallization of the membrane, which is mainly caused by a restoration of the membrane's mechanical stiffness and is otherwise difficult to achieve with other stimuli-sensitive systems. The capsule's crystallinity of intermediate expansion states was nearly unchanged, whereas the lamellar crystal size tends to decreased with the expansion ratio. Therefore, it was assumed that the elastic modulus was only minimally altered and might increased due to the networks segment-chain extension. In addition to the volume increase achieved by inflation, a turn in the osmotic gradient also facilitated the reversible deflation, which was shown in inflation/deflation cycles. These both characteristics of the introduced microballoons are important parameter regarding the realization of micropumps and microvalves. The fixation of expanded microcapsules via recrystallization enabled the storage of entropy-elastic strain-energy, which could be utilized for pumping actions in non-aqueous media. Here, the pumping velocity depended on both, the type of surrounding medium and the applied temperature. Surrounding media that supported the fast transport of pumped liquid showed an accelerated deflation, while high temperatures further accelerate the pumping velocity. Very fast rejection of the incorporated payload was furthermore realized with pierced expanded microballoons, which were subjected to temperatures above their Tm. The possible fixation of intermediate particle sizes provide opportunities for vent constructions that allowed the precise adjustment of specific flow-rates and multiple valve openings and closings. A valve construction was realized by the insertion of a single or multiple microballoons in a microfluidic channel. A complete and a partial closing of the microballoon-valves was demonstrated as a function of the heating period. In this context, a difference between the inflation and deflation velocity was stated, summarizing slower expansion kinetics. Overall, microballoons, which presented both on-demand pumping and reversible valving by a temperature-triggered change in the capsule's volume, might be suitable components that help to design fully integrated LOC devices, due to the implementation of the control switch and controllable inflation/deflation kinetics. In comparison to other state of the art stimuli-sensitive materials, one has to highlight the microballoons capability of stabilizing almost continuously intermediate capsule sizes by simple recrystallization of the microballoon's membrane.
In times of ongoing biodiversity loss, understanding how communities are structured and what mechanisms and local adaptations underlie the patterns we observe in nature is crucial for predicting how future ecological and anthropogenic changes might affect local and regional biodiversity. Aquatic zooplankton are a group of primary consumers that represent a critical link in the food chain, providing nutrients for the entire food web. Thus, understanding the adaptability and structure of zooplankton communities is essential. In this work, the genetic basis for the different temperature adaptations of two seasonally shifted (i.e., temperature-dependent) occurring freshwater rotifers of a formerly cryptic species complex (Brachionus calyciflorus) was investigated to understand the overall genetic diversity and evolutionary scenario for putative adaptations to different temperature regimes. Furthermore, this work aimed to clarify to what extent the different temperature adaptations may represent a niche partitioning process thus enabling co-existence. The findings were then embedded in a metacommunity context to understand how zooplankton communities assemble in a kettle hole metacommunity located in the northeastern German "Uckermark" and which underlying processes contribute to the biodiversity patterns we observe. Using a combined approach of newly generated mitochondrial resources (genomes/cds) and the analysis of a candidate gene (Heat Shock Protein 40kDa) for temperature adaptation, I showed that the global representatives of B. calyciflorus s.s.. are genetically more similar than B. fernandoi (average pairwise nucleotide diversity: 0.079 intraspecific vs. 0.257 interspecific) indicating that both species carry different standing genetic variation. In addition to differential expression in the thermotolerant B. calyciflorus s.s. and thermosensitive B. fernandoi, the HSP 40kDa also showed structural variation with eleven fixed and six positively selected sites, some of which are located in functional areas of the protein. The estimated divergence time of ~ 25-29 Myr combined with the fixed sites and a prevalence of ancestral amino acids in B. calyciflorus s.s. indicate that B. calyciflorus s.s. remained in the ancestral niche, while B. fernandoi partitioned into a new niche. The comparison of mitochondrial and nuclear markers (HPS 40kDa, ITS1, COI) revealed a hybridisation event between the two species. However, as hybridisation between the two species is rare, it can be concluded that the temporally isolated niches (i.e., seasonal-shifted occurrence) they inhabit based on their different temperature preferences most likely represent a pre-zygotic isolation mechanism that allows sympatric occurrence while maintaining species boundaries. To determine the processes underlying zooplankton community assembly, a zooplankton metacommunity comprising 24 kettle holes was sampled over a two-year period. Active (i.e., water samples) and dormant communities (i.e., dormant eggs hatched from sediment) were identified using a two-fragment DNA metabarcoding approach (COI and 18S). Species richness and diversity as well as community composition were analysed considering spatial, temporal and environmental parameters. The analysis revealed that environmental filtering based on parameters such as pH, size and location of the habitat patch (i.e., kettle hole) and surrounding field crops largely determined zooplankton community composition (explained variance: Bray-Curtis dissimilarities: 10.5%; Jaccard dissimilarities: 12.9%), indicating that adaptation to a particular habitat is a key feature of zooplankton species in this system. While the spatial configuration of the kettle holes played a minor role (explained variance: Bray-Curtis dissimilarities: 2.8% and Jaccard dissimilarities: 5.5%), the individual kettle hole sites had a significant influence on the community composition. This suggests monopolisation/priority effects (i.e., dormant communities) of certain species in individual kettle holes. As environmental filtering is the dominating process structuring zooplankton communities, this system could be significantly influenced by future land-use change, pollution and climate change.
Das Eigene und das Fremde

Die vorliegende Arbeit stellt eine Untersuchung des Fremdverstehens von Lehrkräften im Mathematikunterricht dar. Mit ‚Fremdverstehen‘ soll dabei – in Anlehnung an den Soziologen Alfred Schütz – der Prozess bezeichnet werden, in welchem eine Lehrkraft versucht, das Verhalten einer Schülerin oder eines Schülers zu verstehen, indem sie dieses Verhalten auf ein Erleben zurückführt, das ihm zugrunde gelegen haben könnte. Als ein wesentliches Merkmal des Prozesses stellt Schütz in seiner Theorie des Fremdverstehens heraus, dass das Fremdverstehen eines Menschen immer auch auf seinen eigenen Erlebnissen basiert. Aus diesem Grund wird in der Arbeit ein methodischer Zweischritt vorgenommen: Es werden zunächst die mathematikbezogenen Erlebnisse zweier Lehrkräfte nachgezeichnet, bevor dann ihr Fremdverstehen in konkreten Situationen im Mathematikunterricht rekonstruiert wird. In der ersten Teiluntersuchung (= der Rekonstruktion eigener Erlebnisse der untersuchten Lehrkräfte) erfolgt die Datenerhebung mit Hilfe biographisch-narrativer Interviews, in denen die untersuchten Lehrkräfte angeregt werden, ihre mathematikbezogene Lebensgeschichte zu erzählen. Die Analyse dieser Interviews wird im Sinne der rekonstruktiven Fallanalyse vorgenommen. Insgesamt führt die erste Teiluntersuchung zu textlichen Darstellungen der rekonstruierten mathematikbezogenen Lebensgeschichte der untersuchten Mathematiklehrkräfte. In der zweiten Teiluntersuchung (= der Rekonstruktion des Fremdverstehens der untersuchten Lehrkräfte) werden dann narrative Interviews geführt, in denen die untersuchten Lehrkräfte von ihrem Fremdverstehen in konkreten Situationen im Mathematikunterricht erzählen. Die Analyse dieser Interviews erfolgt mit Hilfe eines dreischrittigen Analyseverfahrens, welches die Autorin eigens zum Zweck der Rekonstruktion von Fremdverstehen entwickelte. Am Ende dieser zweiten Teiluntersuchung werden sowohl das rekonstruierte Fremdverstehen der Lehrkräfte in verschiedenen Unterrichtssituationen dargestellt als auch Strukturen, die sich in ihrem Fremdverstehen abzeichnen. Mit Hilfe einer theoretischen Verallgemeinerung werden schließlich – auf Basis der Ergebnisse der zweiten Teiluntersuchung – Aussagen über fünf Merkmale des Fremdverstehens von Lehrkräften im Mathematikunterricht im Allgemeinen gewonnen. Mit diesen Aussagen vermag die Arbeit eine erste Beschreibung davon hervorzubringen, wie sich das Phänomen des Fremdverstehens von Lehrkräften im Mathematikunterricht ausgestalten kann.
Von Ulfila bis Rekkared

Für etwa 200 Jahre, vom Ende des 4. Jahrhunderts bis 589 n. Chr., gehören die Westgoten einem von der Orthodoxie der Reichskirche abweichenden christlichen Bekenntnis an. Auf dem 3. Konzil von Toledo beendet König Rekkared diesen Zustand religiöser Alterität durch die Konversion zum Katholizismus. Die antiken Berichte zeichnen nur vordergründig ein kohärentes Bild des Christentums der Goten. Dagegen weist Eike Faber Widersprüche und Fehler in der Überlieferung nach und bietet eine Reihe neuer Interpretationen an. Die genauere Betrachtung vermag dabei sowohl den dogmatischen Gehalt des Christentums der Goten schärfer zu konturieren als auch die Beweggründe für die Annahme des neuen Glaubens herauszustellen und die traditionelle Datierung dieses Vorgangs zu widerlegen. Schließlich wird deutlich, welche Funktion das fortgesetzte Festhalten an einer demonstrativ anderen religiösen Überzeugung für die Goten hatte. Prägnante Wegmarken der gotischen Geschichte, wie die Bibelübersetzung Ulfilas, die Eroberung und Plünderung Roms 410 n. Chr. oder die demonstrative Aufgabe der religiösen Differenz durch König Rekkared, werden erst durch diesen neuen, umfassenden Kontext verständlich.
Mitochondria and plastids are organelles with an endosymbiotic origin. During evolution, many genes are lost from the organellar genomes and get integrated in the nuclear genome, in what is known as intracellular/endosymbiotic gene transfer (IGT/EGT). IGT has been reproduced experimentally in Nicotiana tabacum at a gene transfer rate (GTR) of 1 event in 5 million cells, but, despite its centrality to eukaryotic evolution, there are no genetic factors known to influence the frequency of IGT in higher eukaryotes. The focus of this work was to determine the role of different DNA repair pathways of double strand break repair (DSBR) in the integration step of organellar DNA in the nuclear genome during IGT. Here, a CRISPR/Cas9 mutagenesis strategy was implemented in N. tabacum, with the aim of generating mutants in nuclear genes without expected visible phenotypes. This strategy led to the generation of a collection of independent mutants in the LIG4 (necessary for non-homologous end joining, NHEJ) and POLQ genes (necessary for microhomology mediated end joining, MMEJ). Targeting of other DSBR genes (KU70, KU80, RPA1C) generated mutants with unexpectedly strong developmental phenotypes.. These factors have telomeric roles, hinting towards a possible relationship between telomere length, and strength of developmental disruption upon loss of telomere structure in plants. The mutants were made in a genetic background encoding a plastid-encoded IGT reporter, that confers kanamycin resistance upon transfer to the nucleus. Through large scale independent experiments, increased IGT from the chloroplast to the nucleus was observed in lig4 mutants, as well as lines encoding a POLQ gene with a defective polymerase domain (polqΔPol). This shows that NHEJ or MMEJ have a double-sided relationship with IGT: while transferred genes may integrate using either pathway, the presence of both pathways suppresses IGT in wild-type somatic cells, thus demonstrating for the first time the extent on which nuclear genes control IGT frequency in plants. The IGT frequency increases in the mutants are likely mediated by increased availability of double strand breaks for integration. Additionally, kinetic analysis reveals that gene transfer (GT) events accumulate linearly as a function of time spent under antibiotic selection in the experiment, demonstrating that, contrary to what was previously thought, there is no such thing as a single GTR in somatic IGT experiments. Furthermore, IGT in tissue culture experiments appears to be the result of a "race against the clock" for integration in the nuclear genome, that starts when the organellar DNA arrives to the nucleus granting transient antibiotic resistance. GT events and escapes of kanamycin selection may be two possible outcomes from this race: those instances where the organellar DNA gets to integrate are recovered as GT events, and in those cases where timely integration fails, antibiotic resistance cannot be sustained, and end up considered as escapes. In the mutants, increased opportunities for integration in the nuclear genome change the overall ratio between IGT and escape events. The resources generated here are promising starting points for future research: (1) the mutant collection, for the further study of processes that depend on DNA repair in plants (2) the collection of GT lines obtained from these experiments, for the study of the effect of DSBR pathways over integration patterns and stability of transferred genes and (3) the developed CRISPR/Cas9 workflow for mutant generation, to make N. tabacum meet its potential as an attractive model for answering complex biological questions.

È qui analizzata la corsa ad ostacoli del romanzo tra congruenze ed eccezioni che assume i tratti di un giallo letterario. L'opera di un principe defunto scavalca il Muro grazie ad Alfred Kurella, potente funzionario della SED. Nella sua postfazione Il Gattopardo non è canto della decadenza, ma preannuncio di una nuova epoca, manifesto dello Stato in procinto di entrare nell'era socialista. La chiave di lettura è un unicum con una marcata impronta ideologica che ne determina prima la fortuna e poi l'isolamento. Al Gattopardo nella Germania Est Bernardina Rago dà per la prima volta voce.

The aim of this work is the study of silica Arrayed Waveguide Gratings (AWGs) in the context of applications in astronomy. The specific focus lies on the investigation of the feasibility and technology limits of customized silica AWG devices for high resolution near-infrared spectroscopy. In a series of theoretical and experimental studies, AWG devices of varying geometry, foot-print and spectral resolution are constructed, simulated using a combination of a numerical beam propagation method and Fraunhofer diffraction and fabricated devices are characterized with respect to transmission efficiency, spectral resolution and polarization sensitivity. The impact of effective index non-uniformities on the performance of high-resolution AWG devices is studied numerically. Characterization results of fabricated devices are used to extrapolate the technology limits of the silica platform. The important issues of waveguide birefringence and defocus aberration are discussed theoretically and addressed experimentally by selection of an appropriate aberration-minimizing anastigmatic AWG layout structure. The drawbacks of the anastigmatic AWG geometry are discussed theoretically. From the results of the experimental studies, it is concluded that fabrication-related phase errors and waveguide birefringence are the primary limiting factors for the growth of AWG spectral resolution. It is shown that, without post-processing, the spectral resolving power is phase-error-limited to R < 40, 000 and, in the case of unpolarized light, birefringence-limited to R < 30, 000 in the AWG devices presented in this work. Necessary measures, such as special waveguide geometries and post-fabrication phase error correction are proposed for future designs. The elimination of defocus aberration using an anastigmatic AWG geometry is successfully demonstrated in experiment. Finally, a novel, non-planar dispersive in-fibre waveguide structure is proposed, discussed and studied theoretically.
Das menschenrechtliche Prinzip des Non-Refoulement vor den Vertragsorganen der Vereinten Nationen

Die Vertragsorgane der Vereinten Nationen können durch die Abstimmung ihrer Praxis Rechtssicherheit schaffen, und zwar sowohl für Betroffene als auch für die Vertragsstaaten. Durch den ständigen Dialog mit den Vertragsstaaten, die Beeinflussung der Vertragsorgane untereinander und das Aufgreifen der Praxis durch andere internationale Akteure lässt sich Gewohnheitsrecht identifizieren. -- Dabei ist das menschenrechtliche Prinzip des Non-Refoulement besonders geeignet, dieses Potenzial der Vertragsorgane zu veranschaulichen. Hierbei handelt es sich um ein Rechtsprinzip, das zwar dem Grunde nach allgemein anerkannt ist, dessen Reichweite im Detail jedoch kontinuierlich umstritten ist. Erstmals wird ein umfassender Überblick über die gesamte Praxis der Vertragsorgane zum Prinzip des Non-Refoulement gegeben. Es wird gezeigt, wie sich die Vertragsorgane sowohl auf prozessualer Ebene als auch bei der Bestimmung des materiellen Schutzbereichs von Refoulementverboten einander annähern.
In jüngerer Vergangenheit häuften sich Fälle, in denen die Verwaltung Entscheidungen von Verwaltungsgerichten bewusst nicht umsetzte. Der Autor nimmt diese Entwicklung zum Anlass und befasst sich mit der Frage, welchen Anteil die Vollstreckungsregeln der Verwaltungsgerichtsordnung an dieser Entwicklung haben. Dabei erfasst er sowohl die staatsrechtliche als auch die grundrechtliche Perspektive. Er überprüft insbesondere, ob die Vorschriften, die die Vollstreckung aus verwaltungsgerichtlichen Entscheidungen gegen den Staat regeln, verfassungsgemäß sind. Nach ausführlicher Diskussion denkbarer Lösungsansätze kommt der Autor zu dem Ergebnis, dass eine Änderung der Vollstreckungsvorschriften dringend geboten ist.
The African weakly electric fish genus Campylomormyrus includes 15 described species mostly native to the Congo River and its tributaries. They are considered sympatric species, because their distribution area overlaps. These species generate species-specific electric organ discharges (EODs) varying in waveform characteristics, including duration, polarity, and phase number. They exhibit also pronounced divergence in their snout, i.e. the length, thickness, and curvature. The diversifications in these two phenotypical traits (EOD and snout) have been proposed as key factors promoting adaptive radiation in Campylomormyrus. The role of EODs as a pre-zygotic isolation mechanism driving sympatric speciation by promoting assortative mating has been examined using behavioral, genetical, and histological approaches. However, the evolutionary effects of the snout morphology and its link to species divergence have not been closely examined. Hence, the main objective of this study is to investigate the effect of snout morphology diversification and its correlated EOD to better understand their sympatric speciation and evolutionary drivers. Moreover, I aim to utilize the intragenus and intergenus hybrids of Campylomormyrus to better understand trait divergence as well as underlying molecular/genetic mechanisms involved in the radiation scenario. To this end, I utilized three different approaches: feeding behavior analysis, diet assessment, and geometric morphometrics analysis. I performed feeding behavior experiments to evaluate the concept of the phenotype-environment correlation by testing whether Campylomormyrus species show substrate preferences. The behavioral experiments showed that the short snout species exhibits preference to sandy substrate, the long snout species prefers a stone substrate, and the species with intermediate snout size does not exhibit any substrate preference. The experiments suggest that the diverse feeding apparatus in the genus Campylomormyrus may have evolved in adaptation to their microhabitats. I also performed diet assessments of sympatric Campylomormyrus species and a sister genus species (Gnathonemus petersii) with markedly different snout morphologies and EOD using NGS-based DNA metabarcoding of their stomach contents. The diet of each species was documented showing that aquatic insects such as dipterans, coleopterans and trichopterans represent the major diet component. The results showed also that all species are able to exploit diverse food niches in their habitats. However, comparing the diet overlap indices showed that different snout morphologies and the associated divergence in the EOD translated into different prey spectra. These results further support the idea that the EOD could be a ‘magic trait’ triggering both adaptation and reproductive isolation. Geometric morphometrics method was also used to compare the phenotypical shape traits of the F1 intragenus (Campylomormyrus) and intergenus (Campylomormyrus species and Gnathonemus petersii) hybrids relative to their parents. The hybrids of these species were well separated based on the morphological traits, however the hybrid phenotypic traits were closer to the short-snouted species. In addition, the likelihood that the short snout expressed in the hybrids increases with increasing the genetic distance of the parental species. The results confirmed that additive effects produce intermediate phenotypes in F1-hybrids. It seems, therefore, that morphological shape traits in hybrids, unlike the physiological traits, were not expressed straightforward.
Beinahe 30 Jahre lang glänzte die »Völkerfreundschaft« zwischen Kuba und der DDR im öffentlichen Diskurs der SED als Musterbeispiel ihres proletarischen Internationalismus. Doch die Rhetorik täuscht: Besonders in den Anfangsjahren der bilateralen Beziehungen nahmen die deutschen Kader ihre kubanischen »Genossen« in der Karibik als notorische Querschläger wahr, die mit ihrem Aufbegehren gegen den ideologischen Suprematieanspruch des Kremls die Stabilität des Ostblocks gefährdeten.
Anhand bislang unveröffentlichten Quellenmaterials aus deutschen und kubanischen Archiven veranschaulicht Antonia Bihlmayer die Bemühungen der Regierung Walter Ulbrichts, die widerspenstigen Sozialisten in der Karibik auf Moskau auszurichten. Auf politisch-ideologischer, wirtschaftlicher und kulturpolitischer Ebene analysiert ihre Studie zum einen die Charakteristika dieser sozialistischen Zivilisierungsmission. Zum anderen nimmt sie diejenigen Faktoren in den Blick, die dafür ausschlaggebend waren, dass sich diese beiden sozialistischen Enklaven ab Mitte der 1970er Jahre schließlich zu gleichwertigen »Juniorpartnern« der Sowjetunion entwickelten.
Answer Set Programming (ASP) allows us to address knowledge-intensive search and optimization problems in a declarative way due to its integrated modeling, grounding, and solving workflow. A problem is modeled using a rule based language and then grounded and solved. Solving results in a set of stable models that correspond to solutions of the modeled problem. In this thesis, we present the design and implementation of the clingo system---perhaps, the most
widely used ASP system. It features a rich modeling language originating from the field of knowledge representation and reasoning, efficient grounding algorithms based on database evaluation techniques, and high performance solving algorithms based on Boolean satisfiability (SAT) solving technology.
The contributions of this thesis lie in the design of the modeling language, the design and implementation of the grounding algorithms, and the design and implementation of an Application Programmable Interface (API) facilitating the use of ASP in real world applications and the implementation of complex forms of reasoning beyond the traditional ASP workflow.
Für die Sondernutzung des gemeindlichen Verkehrsraumes durch die Verlegung und den Betrieb von Leitungen kann die Gemeinde vom Wasserversorger eine Konzessionsabgabe erheben. Gleichwohl kleine Regie- und Eigenbetriebe den größten Teil der Wasserversorger ausmachen, ein Großteil der Abnehmerbeziehungen über Gebühren ausgestaltet ist, und Konzessionsabgaben einen beachtlichen Faktor in den kommunalen Haushalten darstellen, ist die Frage, ob von einem Eigenbetrieb gezahlte Konzessionsabgaben in die Gebührenkalkulation eingestellt werden können, umstritten. Mehrere Oberverwaltungsgerichte verneinen dies. Die Verfasserin untersucht die zugrundeliegende Thematik aus zahlreichen rechtlichen Perspektiven und widerspricht der Rechtsprechung.
The Media Privilege under Data Protection Law: Journalism and data protection are fundamentally at odds with each other. In view of current developments, the need for a functioning regulatory concept for both legal positions is probably more important than ever. This conceptual balance is provided by the journalistic exemption in data protection law. The thesis focuses on the scope of the exception. It also identifies existing coherence problems between European and national law.
Investment control has experienced a considerable increase in importance in M&A transactions, as it has been constantly adapted to real economic conditions and successively tightened. The result is a partially inconsistent review regime that contains value contradictions and creates legal uncertainties. In a comprehensive approach, this doctoral thesis analyzes the problems of the current investment control regime and states concrete reform proposals.
In der vorliegenden Arbeit liegt der Fokus auf jungen russischsprachigen Jüdinnen und Juden, deren Eltern in den 90er Jahren des 20 Jahrhunderts nach Deutschland eingewandert sind. Im Rahmen dieser Studie wird der Bildungsweg dieser MigrantInnengruppe und deren Erfahrungen in Deutschland aus der biographischen Perspektive nachvollzogen. Der Fokus wird insbesondere auf die biographischen Lebenserfahrungen gelegt, d.h. die allgemeinen Lebensumstände, Hürden und Schwierigkeiten, die die jungen russischsprachigen Jüdinnen und Juden in Deutschland überwinden mussten, um auf ihrem Bildungsweg an ihr Ziel zu kommen. Des Weiteren werden die Rolle des sozialen Umfelds auf die Auswahl ihres Bildungsweges sowie ihre Zugehörigkeit und ihr Beitrag zur deutschen Gesellschaft beleuchtet. Ein weiteres Hauptaugenmerk dieser Arbeit liegt auf den gesellschaftlichen, politischen und familiären Rahmenbedingungen, die den BiographInnen den Zugang zum Bildungsweg ermöglichten.
Die in der Arbeit formulierten Forschungsfragen wurden mithilfe der interpretativen Sozialforschung, genauer, der fallrekonstruktiven Auswertung nach Gabriele Rosenthal beantwortet.
Über den biographischen Verlauf der Lebensgeschichte der jungen russischsprachigen Jüdinnen und Juden wurde deutlich, dass die BiographInnen in die säkulare Gesellschaft gehen müssen, um ihren Bildungsweg erfolgreich zu gestalten. Dort erfahren sie einen sehr starken Antisemitismus und sind diesem schutzlos ausgeliefert. Bei allen drei Interviewten wurde diese Erfahrung in der Schule gemacht, an einem Ort, an dem sie Schutz erfahren sollten. Diesen Anfeindungen begegneten sie auf unterschiedliche Weise und sie entwickelten verschiedene Handlungsstrategien. Einige BiographInnen setzen sich auf der intellektuellen Ebene bewusst damit auseinander, andere wiederum versuchen, nicht hinzuschauen und es zu ignorieren.
Des Weiteren wurde als Resultat der Untersuchung in einigen Fällen ein übereinstimmendes, in anderen ein nicht übereinstimmendes Passungsverhältnis zwischen elterlichen Vorstellungen und den Bildungswegen der jungen russischsprachigen Jüdinnen und Juden gefunden.

Während der Zeit des historischen Kolonialismus wurden in Völkerkundemuseen komplexe Formen rassistischer und religiöser Diskriminierung institutionalisiert, z.B. in den dort gültigen Ästhetik- und Kunstbegriffen. Viele der heutigen Museumsangestellten erklären sich deswegen zu Reformen bereit. Doch können sie sich tatsächlich vom Kolonialismus trennen? Ist eine Dekolonisation ethnologischer Museen mit kolonialer Beute je abschließend möglich? Am Beispiel umstrittener Heiligtümer lebender Kulturen untersucht Christoph Balzar das Verfahren der Musealisierung durch die Linse der Diskriminierungskritik. Im Fokus stehen dabei die Sammlungen der »Staatlichen Museen zu Berlin«.
Zu Seiner Majestät allerhöchstem Interesse als titelgebendem Zitat bekannten sich die zeitgenössischen Amtsträger in ihrer Beeidigung als grundlegende Maxime des durch den König verkörperten werdenden preussischen Staates.Die Domänenpolitik Friedrich Wilhelms I., dem zweiten preussischen König, war die wesentliche Voraussetzung für die folgende Entwicklung Preussens von einer Regionalmacht im Heiligen Römischen Reich zu einer europäischen Grossmacht. Ohne nennenswerte Rohstoffe und handwerkliche Traditionen, agrarisch geprägt und ganz dem merkantilistischen Wirtschaftsmodell und seiner kameralistischen Methodik verhaftet, war der Ausbau und die Intensivierung der landesherrlichen Gutswirtschaft der für Preussen verheissungsvollste Weg zur Erwirtschaftung der Mittel, die es zum Schuldenabbau, dem Aufbau einer starken Militärmacht und Anhäufung eines Staatsschatzes befähigte. Erreicht wurde dies durch konsequente Einnahmen- und Ausgabenkontrolle und die Schaffung eines effizienten Verwaltungsapparates mit detaillierten Vorschriften, was knapp einhundert Jahre Bestand behalten sollte und bis zu nahezu 50 Prozent der Staatseinnahmen hervorbrachte. Das friderizianische Preussen wäre ohne die Leistungen Friedrich Wilhelms I. nicht möglich gewesen.Der vorliegende Band stellt diese Entwicklung mit einer Fülle von Quellenmaterial und dessen Auswertungen sowie einem umfangreichen Anhang unter Beweis. Der allein 22seitige Personenindex mit über 1.300 Verweisen und 103 Kurzbiogra-fien der kurmärkischen Kammerräte machen deutlich, wer hinter den Leistungen dieser Epoche stand.

Mit mehr als 500 historischen Grabanlagen, seinem Ensemble aus Friedhofsbauten sowie aufgrund seiner Landschaftsarchitektur gehört dieser Ort heute zum UNESCO-Welterbe.

Nach einem Blick auf die Rolle der Jüdischen Gemeinde Potsdams als Eigentümerin des Friedhofs erfolgt unter Auswertung von umfangreichem Archivmaterial eine detaillierte Darstellung der Geschichte des „Guten Ortes“. Eine Untersuchung sämtlicher Grabmale auf unterschiedliche Gestaltungsmerkmale visualisiert und rekonstruiert die Veränderungen in der erhaltenen Sepulkralkultur. Abschließend richtet sich der Fokus auf den Umgang mit dem Friedhof als Ort der Erinnerung.
Teil 2 bietet die Dokumentation von 370 Grabanlagen. Um den Friedhof in seiner Gesamtheit abzubilden, wurde die 1992 erfolgte Teildokumentation der 158 Grabsteine des gesamten ältesten Begräbnisfeldes ergänzend hinzugenommen. Mit mehr als 1.000 Fotos wird hier ein einmaliges Zeugnis der Brandenburger Juden dokumentiert.

Eine Untersuchung sämtlicher Grabmale auf unterschiedliche Gestaltungsmerkmale visualisiert und rekonstruiert die Veränderungen in der erhaltenen Sepulkralkultur. Abschließend richtet sich der Fokus auf den Umgang mit dem Friedhof als Ort der Erinnerung.
Teil 2 bietet die Dokumentation von 370 Grabanlagen. Um den Friedhof in seiner Gesamtheit abzubilden, wurde die 1992 erfolgte Teildokumentation der 158 Grabsteine des gesamten ältesten Begräbnisfeldes ergänzend hinzugenommen. Mit mehr als 1.000 Fotos wird hier ein einmaliges Zeugnis der Brandenburger Juden dokumentiert.
Die betriebliche oder „arbeitsplatzorientierte Grundbildung (AoG)“ ist ein noch junges Forschungsgebiet der wissenschaftlichen Erwachsenenbildung. Neuerdings wenden sich auch betriebliche Kursangebote an die „Bildungsverlierer“ unseres Schulsystems, die mit z.T. massiven Problemen die Haupt- oder Sonderschule verlassen. Diese qualitative Studie ist dem Bereich der subjektwissenschaftlichen Lernforschung zuzuordnen. Sie rekonstruiert die subjektiven Lernbegründungen der formal gering literalisierten Beschäftigten, wenn sie an Grundbildungskursen am Arbeitsplatz teilnehmen. Die Studie zeigt auf, dass erhebliche lernbiografisch begründete Widerstände überwunden werden müssen, um – eingebettet in Anerkennungsverhältnisse im Kurs – zu persönlich bedeutsamem produktivem Lernen vorzudringen. Zunächst sind Freiwilligkeit und Teilnehmendenorientierung in Kursen häufig in Frage gestellt. Mehr oder weniger reflektierte Lernwiderstände mit defensiven Lerngründen werden insofern vom Autor als subjektiv sinnvolle Handlungsstrategien verstanden. Als lebenslang tätiger Praktiker erläutert der Autor, wie die Resultate der Studie für eine gute Praxis betrieblicher Grundbildung genutzt werden können.

Um sich der gesellschaftlichen Verantwortung im Rahmen des Ausstellens von Kunst zu stellen, erprobt sie queer-feministische, kulturwissenschaftliche und selbstreflektierende Methoden in der Praxis und Theorie des Kuratierens. Momente kuratorischer Störung werden zu produktiv-schöpferischen Momenten der Unterbrechung, mit denen sie reflektierte, offene, engagierte und vulnerable Perspektiven auf das Ausstellen eröffnet und tradierte Strukturen durchbricht.

Das 15. Jahrhundert ist das Zeitalter der letzten Massenkonversionen zum Christentum auf der Iberischen Halbinsel. Unter starkem Druck und zum Teil mit Gewalt wurde die zuvor große jüdische Bevölkerung gedrängt, sich taufen zu lassen. Gleichwohl akzeptierten große Teile der christlichen Mehrheit die Neubekehrten nicht als gleichwertig und bezweifelten ihre Rechtgläubigkeit. Gegen diese Diskriminierung wandten sich immer wieder Geistliche und Gelehrte mit Predigten, Briefen und Denkschriften. Die vorliegende Studie zeichnet erstmals detailliert die wesentlichen Inhalte ihrer Theologie nach, ihre spezielle Auslegung der Bibel und deren Wurzeln in Recht und Tradition der lateinischen Kirche.
Die Arbeit ist ein Beitrag zu einer grundlegenden Diskussion der Kapitalmarktforschung, dem messbaren Erfolg „aktiver vs. passiver“ Investmentstrategien. Der Autor setzt sich kritisch mit den wesentlichen Anlagestrategien und Modellen für Indexprodukte auseinander und beleuchtet zugleich Closet Indexing bei aktiv gemanagten Investmentfonds. Das Ergebnis zeigt, dass Closet Indexing nicht nur sporadisch auftritt, sondern eine weit verbreitete Anlagestrategie in vielen vermeintlich aktiv gemanagten Aktieninvestmentfonds ist.
Social networking site use and well-being - a nuanced understanding of a complex relationship

Social Networking Sites (SNSs) are ubiquitous and attract an enormous chair of the digital population. Their functionalities allow users to connect and interact with others and weave complex social networks in which social information is continuously disseminated between users. Besides the social value SNSs are generating, they likewise attract companies and allow for new forms of marketing, thereby creating considerable economic value alike. However, as SNSs grew in popularity, so did concerns about the impact of their use on social interactions in general and the well-being of individual users in particular. While existing scientific evidence points to both risk as well as benefits of SNS use, research still lacks a profound understanding of which aspects of SNSs enable an impact on well-being and which psychological processes on the part of the users underly and explain this relationship. Therefore, this thesis is dedicated to an in-depth exploration of the relationship between SNS use and well-being and aims to answer how SNS use can impact well-being. Primarily, it focuses on the unique technological features that characterize SNSs and enable potential well- being alterations and on specific psychological processes on the part of the users, underlying and explaining the relationship. For this purpose, the thesis first introduces the concept of well- being. It continues by presenting SNSs’ unique technological features, divided into specifics of the content disseminated on SNSs and the network structure of SNSs. Further, the thesis introduces three classes of psychological processes assumed most relevant for the relationship between SNSs and well-being: other-focused, self-focused, and contrastive processes.. It is assumed that the course and quality of these common processes change in the SNS context and that a complex interplay between the unique features of SNSs and these processes determines how SNSs may ultimately affect users' well-being - both in positive and negative ways. The dissertation comprises seven research articles, each of which focusses on a particular set of SNS characteristics, their interplay with one or more of the proposed psychological processes, and ultimately the resulting effects on user well-being or its key resilience and risk factors. The seven articles investigate this relationship using different methodological approaches. Three articles are based on either systematic or narrative literature reviews, one applies an empirical cross-sectional research design, and three articles present an experimental investigation. Thematically, two articles revolve around SNS use’s effect on self-esteem. Three articles examine the specific role of the emotion of envy and its potential to establish and perpetuate a well-being-damaging social climate on SNSs. The two last articles of this thesis revolve around the established assumption that active and passive SNS use, as different modalities of SNS use, cause differential effects on users’ well-being due to the involvement of different psychological processes. The results of this thesis illustrate different ways how SNSs can affect users’ well-being. The results suggest that especially contrastive processes play a decisive role in explaining potential well-being risks for SNS users. Their interplay with certain SNS features seems to foster upward social comparisons and feelings of envy, potentially leading to a complex set of deleterious effects on users’ well-being. At the same time, the findings illuminate ways in which SNSs can benefit users and their self-esteem – especially when SNS use promotes self- focused and social-feedback-based other-focused processes. The thesis and their findings illustrate that the relationship between SNSs and well-being is complex. Therefore, a nuanced perspective, taking into consideration both the technological uniqueness of SNSs and the psychological processes they are enabling, is crucial to understand how these technologies affect their users in good and potentially harmful ways. On the one hand, the gathered insights contribute to research, providing novel insights into the complex relationship between SNS use and well-being. On the other hand, the results enable a focused and action-oriented derivation of recommendations for stakeholders such as individual users, policymakers, and platform providers. The findings of this thesis can help them to better combat SNS-related risks and ultimately ensure a healthy and sustainable environment for users - and thus also the economic values of SNSs - in the long term.
Seismology, like many scientific fields, e.g., music information retrieval and speech signal pro- cessing, is experiencing exponential growth in the amount of data acquired by modern seismo- logical networks. In this thesis, I take advantage of the opportunities offered by "big data" and by the methods developed in the areas of music information retrieval and machine learning to predict better the ground motion generated by earthquakes and to study the properties of the surface layers of the Earth. In order to better predict seismic ground motions, I propose two approaches based on unsupervised deep learning methods, an autoencoder network and Generative Adversarial Networks. The autoencoder technique explores a massive amount of ground motion data, evaluates the required parameters, and generates synthetic ground motion data in the Fourier amplitude spectra (FAS) domain. This method is tested on two synthetic datasets and one real dataset. The application on the real dataset shows that the substantial information contained within the FAS data can be encoded to a four to the five-dimensional manifold. Consequently, only a few independent parameters are required for efficient ground motion prediction. I also propose a method based on Conditional Generative Adversarial Networks (CGAN) for simulating ground motion records in the time-frequency and time domains. CGAN generates the time-frequency domains based on the parameters: magnitude, distance, and shear wave velocities to 30 m depth (VS30). After generating the amplitude of the time-frequency domains using the CGAN model, instead of classical conventional methods that assume the amplitude spectra with a random phase spectrum, the phase of the time-frequency domains is recovered by minimizing the observed and reconstructed spectrograms. In the second part of this dissertation, I propose two methods for the monitoring and characterization of near-surface materials and site effect analyses. I implement an autocorrelation function and an interferometry method to monitor the velocity changes of near-surface materials resulting from the Kumamoto earthquake sequence (Japan, 2016). The observed seismic velocity changes during the strong shaking are due to the non-linear response of the near-surface materials. The results show that the velocity changes lasted for about two months after the Kumamoto mainshock. Furthermore, I used the velocity changes to evaluate the in-situ strain-stress relationship. I also propose a method for assessing the site proxy "VS30" using non-invasive analysis. In the proposed method, a dispersion curve of surface waves is inverted to estimate the shear wave velocity of the subsurface. This method is based on the Dix-like linear operators, which relate the shear wave velocity to the phase velocity. The proposed method is fast, efficient, and stable. All of the methods presented in this work can be used for processing "big data" in seismology and for the analysis of weak and strong ground motion data, to predict ground shaking, and to analyze site responses by considering potential time dependencies and nonlinearities.
Starch is an insoluble polyglucan, comprises of two polymers, namely, the branched α-1,4: α-1,6-D-glucan amylopectin and the almost unbranched α-1,4-D-glucan amylose. The growth of all plants is directly dependent on the accumulation of transitory starch during the daytime when photosynthesis takes place and subsequently starch degradation during the night. Starch phosphorylation takes place by starch-related dikinases called α-glucan, water dikinase (GWD), and phosphoglucan, water dikinase (PWD), and is a very important step in starch degradation. The biochemical mechanisms of phosphorylation of starch are not properly understood. Recent studies have found that there are two starch binding proteins namely, Early Starvation1 (ESV1) and Like Early Starvation1 (LESV), which play an important role in starch metabolism. It has been shown that ESV1 and LESV proteins affect the starch phosphorylation activity of GWD and PWD enzymes, which control the rate of degradation of starch granules. In this thesis, various in vitro assays were performed to identify and understand the mechanism of recombinant proteins; ESV1 and LESV on the starch degradation. The starch degradation was performed by phosphorylation enzymes, GWD and PWD separately. In various enzymatic assays, the influence of the ESV1 and LESV on the actions of GWD and PWD on the surfaces of different native starch granules were analysed. Furthermore, ESV1 and LESV have specifically shown influences on the phosphorylation activities of GWD and PWD on the starch granule surfaces in an antagonistic pattern in such a way that, the GWD mediated phosphorylation were significantly reduced while PWD mediated phosphorylation were significantly increased respectively. In another set of experiments, ISA and BAM hydrolyzing enzymes were used to alter the structure of starch, and then determine the effect of both dikinases mediated phosphorylation in the presence of ESV1 and LESV on the altered starch granules surfaces. In these results, significant decreases in both GWD and PWD mediated phosphorylation were observed in all the treatments containing either ESV1 or LESV proteins only or both ESV1 and LESV. It was also found that LESV preferentially binds to both amylose and amylopectin, while ESV1 binds to highly ordered glucans such as maltodextrins and amylopectin, which are crystalline in structure. Both ESV1 or LESV proteins either individually or in combination have shown influence on the activity of GWD and PWD phosphate incorporation into the starch granules via reduction even though at different percentages depending on the sources of starch, therefore it is difficult to distinguish the specific function between them. The biochemical studies have shown that protein-glucan interaction specifically between ESV1 or LESV or in combination with different species of starch granules has very strong surface binding, or it might be possible that both the proteins not only bind to the surface of the starch granules but also have entered deep inside the glucan structure of the starch granules. However, the results also revealed that ESV1 and LESV did not alter the autophosphorylation of the dikinases. Also, the chain length distribution pattern of the released glucan chains after treatment of starch with ISA enzyme was evaluated with respect to the degree of polymerization (DP) of the different starch granules. Capillary electrophoresis was employed to study the effect of LESV and ESV1 on the chain length distribution. In summary, this study confirms that ESV1 and LESV play an important role in organizing and regulating the starch metabolism process. In the later half, studies were performed to monitor whether the metabolism of carbohydrates and partitioning, contribute to the higher salt tolerance of the facultative halophyte Hordeum marinum when compared to glycophyte Hordeum vulgare. Seedlings with the same size from both species were hydroponically grown at 0, 150, and 300 mM of NaCl for 3 weeks. H. marinum maintained a high relative growth rate, which was found concomitant in higher aptitude plants to maintain efficient shoot tissue hydration and integrity of membrane under salt conditions when compared to H. vulgare. Hence, our data suggested that the change in the starch storage, distribution of soluble sugar concentrations between source and sink organs, and also changes in the level of enzymes involved in the starch metabolism was significant to give insights into the importance of carbohydrate metabolism in barley species with regards to the salt tolerance. Although these results are still in their nascent state, it could be vital for other researchers to formulate future studies. The preliminary results which were studies about the carbohydrate metabolism and partitioning in salt responses in the halophyte H. marinum and the glycophyte H. vulgare revealed that salt tolerance in barley species is not due to osmotic adjustments, but due to other reasons that were not explored in the past studies. However, the activity of DPE2 in H. vulgare was not hampered by the presence of NaCl as observed. While Pho1 and Pho2, activities were highly increased in cultivated barley. These findings could be suggestive of a possible role of these enzymes in the responses of carbohydrate metabolism to salinity. When sea and cultivated barley species were compared, it was discovered that the former had more versatility in carbohydrate metabolism and distribution.
Does youth matter?

This dissertation is a compilation of publications and submitted publication manuscripts that seek to improve the understanding of modern partnership trajectories. Romantic relationships constitute one of the most important dimensions in a person’s life. They serve to satisfy social and emotional needs (Arránz Becker, 2008) and have an impact on various other dimensions of life. Since the 1970s, partnership formation has been characterized by increased heterogeneity, has become less ordered and much more diverse in terms of living arrangements and the number of unions across the life course (Helske et al, 2015; Ross et al, 2009). This dissertation argues that while partnerships have become more unstable, the need for attachment and the importance of relationship have remained high, if not increased, as evidenced by the prevalence of couple relationships that have remained quite stable (Eckhardt, 2015). The life course perspective (Elder, 1994; Elder et al., 2004; Mayer, 2009) offers an appropriate framework for the understanding of partnership formations throughout the life course. This perspective stresses the path dependency of the life course as well as the interdependencies of life domains (Bernardi et al., 2019). Thus, it can be argued that conditions, resources, and experiences in youth have a substantial influence on later life course outcomes. Given the increasing heterogeneity of partnership trajectories, research to understand partnership processes cannot be based only on single events (e.g., marriage or divorce) or life stages, but must be explored in a dynamic context and over a longer period of time. In sum, this thesis argues that partnership trajectories have to be considered from a holistic perspective. Not only single transitions or events are useful to describe modern partnership histories adequately, but rather the whole process. Additionally, as partnership trajectories are linked to various outcomes (e.g., economics, health, effects on children), it is therefore highly relevant to improve our understanding of partnership dynamics and their determinants and consequences. Findings in this field of research contribute to a better understanding of how childhood and youth are of prospective importance for the later partnership trajectories and whether there are any long-term effects of the conditions and resources formed and stabilized in youth, which then help to understand and explain partnership dynamics. Thus, the interest of this thesis lies in the longitudinal description and prediction of the dynamics of partnership trajectories in light of the individual resources formed and stabilized in youth, as well as in the investigation of the consequences of different partnership trajectory patterns on individual well-being. For these objectives, a high demand on the data is required, as prospective data at the beginning of the partnership biography are needed, as well as data on current life dimensions and the detailed partnership history. The German LifE Study provides this particular data structure as it examines life courses of more than 1,300 individuals from adolescence to middle adulthood. With regard to the overall aim of this dissertation, the main conclusion is that early life conditions, experiences, and resources influence the dynamics of individual partnership trajectories. The results illustrate that youth matters and that characteristics and resources anchored in youth influence the timing of early status passages, which sets individuals on specific life paths. However, in addition to personal and social resources, partnership trajectories were also significantly influenced by individuals’ sociodemographic placement. Additionally, individual resources are also linked to the overall turbulence or stability of partnership trajectories. This overall dynamic, which is reflected in different partnership patterns, influences individual well-being, with stability being associated with greater satisfaction, and instability (women), or permanent singlehood (men), having a negative impact on well-being. My analyses contribute to life course research by examining path dependency against the background of various individual factors (socio-structural and psychological characteristics) to model decision-making processes in partnerships in more detail. They do so by including also non-cohabitational union types in the analyses, by accounting for pre-trajectory life conditions and resources, and, most importantly, by modeling the partnership trajectory in a holistic and dynamic perspective, applying this perspective to appropriate and modern statistical methods on a unique dataset.

Die vorliegende Untersuchung befasst sich mit den allgegenwärtigen Fragen, welches rechtliche Schicksal der menschliche Körper nach dem Tod nimmt, ob der Leichnam vererbt wird und wer in welchem Umfang über ihn bestimmen darf. Die Autorin gelangt zu dem Ergebnis, dass das Totenfürsorgerecht als wesentlicher Bestandteil des - im Grundgesetz als Staatszielbestimmung zu verankernden - sog. postmortalen Persönlichkeitsschutzes auf die Wahrung der Pietät ziele und zuvörderst dem Willen des Verstorbenen verpflichtet sei. Bei unbekanntem Verstorbenenwillen dürfe der Totenfürsorgeberechtigte aber in einigen wenigen Bereichen auch eigene Entscheidungen über den ihm anvertrauten Leichnam treffen. Der Umgang mit dem Leichnam lasse sich bislang keinem bekannten Rechtsinstitut zuordnen und stelle Gewohnheitsrecht dar. Gegenwärtig herrsche Rechtsunsicherheit. Zur Behebung des gesetzgeberischen Defizits schlägt die Autorin ein Bundesgesetz vor und unterbreitet hierfür einen Gesetzesvorschlag.
Die allergische Kontaktdermatitis ist eine immunologisch bedingte Hauterkrankung mit insbesondere in den westlichen Industrienationen hoher und weiter ansteigender Prävalenz. Es handelt sich hierbei um eine Hypersensitivitätsreaktion vom Typ IV, die sich nach Allergenkontakt durch Juckreiz, Rötung, Bläschenbildung und Abschälung der Haut äußert. Zahlreiche Xenobiotika besitzen das Potenzial, Kontaktallergien auszulösen, darunter Konservierungsstoffe, Medikamente, Duftstoffe und Chemikalien. Die wirksamste Maßnahme zur Eindämmung der Erkrankung ist die Expositionsprophylaxe, also die Vermeidung des Kontakts mit den entsprechenden Substanzen. Dies wiederum setzt die Kenntnis des jeweiligen sensibilisierenden Potenzials einer Substanz voraus, dessen Bestimmung aus diesem Grund eine hohe toxikologische Relevanz besitzt. Zu diesem Zweck existieren von der OECD veröffentlichte Testleitlinien, welche auf entsprechend validierten Testmethoden basieren. Goldstandard bei der Prüfung auf hautsensibilisierendes Potenzial war über lange Zeit der murine Lokale Lymphknotentest. Seit der 7. Änderung der EU-Kosmetikrichtlinie, welche Tierversuche für Kosmetika und deren Inhaltsstoffe untersagt, wurden vermehrt Alternativmethoden in die OECD-Testleitlinien implementiert.. Die bestehenden in vitro Methoden sind jedoch alleinstehend nur begrenzt aussagekräftig, da sie lediglich singuläre Mechanismen bei der Entstehung einer Kontaktallergie abbilden. Die Entwicklung von Testmethoden, welche mehrere dieser Schlüsselereignisse berücksichtigen, erscheint daher richtungsweisend. Einen vielversprechenden Ansatz liefert hierbei der Loose-fit coculture-based sensitisation assay (LCSA), welcher eine Kokultur aus primären Keratinozyten und PBMC darstellt. Bei der Kokultivierung von Immunzellen mit anderen Zelltypen stellt sich allerdings die Frage, inwiefern die Nutzung von Zellen derselben Spender*innen (autologe Kokultur) bzw. verschiedener Spender*innen (allogene Kokultur) einen Einfluss nimmt. Zu diesem Zweck wurden im Rahmen dieser Arbeit Hautzellen spenderspezifisch aus gezupften Haarfollikeln isoliert und der LCSA mit den generierten HFDK in autologen und allogenen Ansätzen verglichen. Zusätzlich wurde auch ein Vergleich zwischen der Nutzung von HFDK und NHK, welche aus humaner Vorhaut isoliert wurden, im LCSA durchgeführt. Dabei ergaben sich keine signifikanten Unterschiede zwischen autologen und allogenen Kokulturen bzw. zwischen der Verwendung von HFDK und NHK. Die Verwendung allogener Zellen aus anonymem Spendermaterial sowie die Nutzung von Keratinozyten aus unterschiedlichen Quellen scheint im Rahmen des LCSA problemlos möglich. Einige der getesteten Kontaktallergene, darunter DNCB und NiCl2, erwiesen sich im LCSA jedoch als problematisch und konnten nicht zufriedenstellend als sensibilisierend detektiert werden. Daher wurde eine Optimierung der Kokultur durch Verwendung ex vivo differenzierter Langerhans Zellen (MoLC) angestrebt, welche ein besseres Modell primärer epidermaler Langerhans Zellen darstellen als die dendritischen Zellen aus dem LCSA. Zusätzlich wurden weitere, den Erfolg der Kokultur beeinflussende Faktoren, wie die Art und Zusammensetzung des Mediums und die Kokultivierungsdauer, untersucht und angepasst. Das schlussendlich etablierte Kokultivierungsprotokoll führte zu einer maßgeblich verstärkten Expression von CD207 (Langerin) auf den MoLC, was auf eine wirkungsvolle Interaktion zwischen Haut- und Immunzellen in der Kokultur hindeutete. Des Weiteren konnten DNCB und NiCl2 im Gegensatz zum LCSA durch Verwendung des kostimulatorischen Moleküls CD86 sowie des Reifungsmarkers CD83 als Ausleseparameter eindeutig als Kontaktallergene identifiziert werden. Die Untersuchungen zur Kokultur von MoLC und HFDK wurden jeweils vergleichend in autologen und allogenen Ansätzen durchgeführt. Ähnlich wie beim LCSA kam es aber auch hier zu keinen signifikanten Unterschieden, weder hinsichtlich der Expression von Charakterisierungs- und Aktivierungsmarkern auf MoLC noch hinsichtlich der Zytokinsekretion in den Zellkulturüberstand. Die Hinweise aus zahlreichen Studien im Mausmodell, dass Zellen des angeborenen Immunsystems zur Erkennung von und Aktivierung durch allogene Zellen bzw. Gewebe in der Lage sind, bestätigten sich im Rahmen dieser Arbeit dementsprechend nicht. Aus diesem Grund wurden abschließend CD4+ T-Lymphozyten, die Effektorzellen des adaptiven Immunsystems, in die Kokultur aus MoLC und autologen bzw. allogenen HFDK integriert. Überraschenderweise traten auch hier keine verstärkten Aktivierungen in allogener Kokultur im Vergleich zur autologen Kokultur auf. Die Nutzung autologer Primärzellen scheint im Rahmen der hier getesteten Methoden nicht notwendig zu sein, was die Validierung von Kokulturen und deren Implementierung in die OECD-Testleitlinien erleichtern dürfte. Zuletzt wurde eine Kokultivierung primärer Haut- und Immunzellen auch im 3D-Vollhautmodell durchgeführt, wobei autologe MoLC in die Epidermisäquivalente entsprechender Modelle integriert werden sollten. Obwohl die erstellten Hautmodelle unter Verwendung autologer Haarfollikel-generierter Keratinozyten und Fibroblasten eine zufriedenstellende Differenzierung und Stratifizierung aufwiesen, gestaltete sich die Inkorporation der MoLC als problematisch und konnte im Rahmen dieser Arbeit nicht erreicht werden.
Cardiovascular diseases are the main cause of death worldwide, and their prevalence is expected to rise in the coming years. Polymer-based artificial replacements have been widely used for the treatment of cardiovascular diseases. Coagulation and thrombus formation on the interfaces between the materials and the human physiological environment are key issues leading to the failure of the medical device in clinical implantation. The surface properties of the materials have a strong influence on the protein adsorption and can direct the blood cell adhesion behavior on the interfaces. Furthermore, implant-associated infections will be induced by bacterial adhesion and subsequent biofilm formation at the implantation site. Thus, it is important to improve the hemocompatibility of an implant by altering the surface properties. One of the effective strategies is surface passivation to achieve protein/cell repelling ability to reduce the risk of thrombosis.
This thesis consists of synthesis, functionalization, sterilization, and biological evaluation of bulk poly(glycerol glycidyl ether) (polyGGE), which is a highly crosslinked polyether-based polymer synthesized by cationic ring-opening polymerization. PolyGGE is hypothesized to be able to resist plasma protein adsorption and bacterial adhesion due to analogous chemical structure as polyethylene glycol and hyperbranched polyglycerol. Hydroxyl end groups of polyGGE provide possibilities to be functionalized with sulfates to mimic the anti-thrombogenic function of the endothelial glycocalyx.
PolyGGE was synthesized by polymerization of the commercially available monomer glycerol glycidyl ether, which was characterized as a mixture of mono-, di- and tri-glycidyl ether. Cationic ring opening-polymerization of this monomer was carried out by ultraviolet (UV) initiation of the photo-initiator diphenyliodonium hexafluorophosphate. With the increased UV curing time, more epoxides in the side chains of the monomers participated in chemical crosslinking, resulting in an increase of Young’s modulus, while the value of elongation at break of polyGGE first increased due to the propagation of the polymer chains then decreased with the increase of crosslinking density. Eventually, the chain propagation can be effectively terminated by potassium hydroxide aqueous solution. PolyGGE exhibited different tensile properties in hydrated conditions at body temperature compared to the values in the dry state at room temperature. Both Young’s modulus and values of elongation at break were remarkably reduced when tested in water at 37 °C, which was above the glass transition temperature of polyGGE. At physiological conditions, entanglements of the ployGGE networks unfolded and the free volume of networks were replaced by water molecules as softener, which increased the mobility of the polymer chains, resulting in a lower Young’s modulus.
Protein adsorption analysis was performed on polyGGE films with 30 min UV curing using an enzyme-linked immunosorbent assay. PolyGGE could effectively prevent the adsorption of human plasma fibrinogen, albumin, and fibronectin at the interface of human plasma and polyGGE films. The protein resistance of polyGGE was comparable to the negative controls: the hemocompatible polydimethylsiloxane (PDMS), showing its potential as a coating material for cardiovascular implants. Moreover, antimicrobial tests of bacterial activity using isothermal microcalorimetry and the microscopic image of direct bacteria culturing demonstrated that polyGGE could directly interfere biofilm formation and growth of both Gram-negative and antibiotic-resistant Gram-positive bacteria, indicating the potential application of polyGGE for combating the risk of hospital-acquired infections and preventing drug-resistant superbug spreading.
To investigate its cell compatibility, polyGGE films were extracted by different solvents (ethanol, chloroform, acetone) and cell culture medium. Indirect cytotoxicity tests showed extracted polyGGE films still had toxic effects on L929 fibroblast cells. High-performance liquid chromatography/electrospray ionization mass spectrometry revealed the occurrence of organochlorine-containing compounds released during the polymer-cell culture medium interaction. A constant level of those organochlorine-containing compounds was confirmed from GGE monomer by a specific peak of C-Cl stretching in infrared spectra of GGE. This is assumed to be the main reason causing the increased cell membrane permeability and decreased metabolic activity, leading to cell death. Attempts as changing solvents were made to remove toxic substances, however, the release of these small molecules seems to be sluggish. The densely crosslinked polyGGE networks can possibly contribute to the trapping of organochlorine-containing compounds. These results provide valuable information for exploring the potentially toxic substances, leaching from polyGGE networks, and propose a feasible strategy for minimizing the cytotoxicity via reducing their crosslinking density.
Sulfamic acid/ N-Methyl-2-pyrrolidone (NMP) were selected as the reagents for the sulfation of polyGGE surfaces. Fourier transform attenuated total reflection infrared spectroscopy (ATR-FT-IR) was used to monitor the functionalization kinetics and the results confirmed the successful sulfate grafting on the surface of polyGGE with the covalent bond -C-O-S-. X-ray photoelectron spectroscopy was used to determine the element composition on the surface and the cross-section of the functionalized polyGGE and sulfation within 15 min guarantees the sulfation only takes place on the surface while not occurring in the bulk of the polymer. The concentration of grafted sulfates increased with the increasing reaction time. The hydrophilicity of the surface of polyGGE was highly increased due to the increase of negatively charged end groups. Three sterilization techniques including autoclaving, gamma irradiation, and ethylene oxide (EtO) sterilization were used for polyGGE sulfates. Results from ATR-FT-IR and Toluidine Blue O quantitative assay demonstrated the total loss of the sulfates after autoclave sterilization, which was also confirmed by the increased water contact angle. Little influence on the concentration of sulfates was found for gamma-irradiated and autoclaving sterilized polyGGE sulfates. To investigate the thermal influence on polyGGE sulfates, one strategy was to use poly(hydroxyethyl acrylate) sulfates (PHEAS) for modeling. The thermogravimetric analysis profile of PHEAS demonstrated that sulfates are not thermally stable independent of the substrate materials and decomposition of sulfates occurs at around 100 °C. Although gamma irradiation also showed little negative effect on the sulfate content, the color change in the polyGGE sulfates indicates chemical or physical change might occur in the polymer. EtO sterilization was validated as the most suitable sterilization technique to maintain the chemical structure of polyGGE sulfates.
In conclusion, the conducted work proved that bulk polyGGE can be used as an antifouling coating material and shows its antimicrobial potential. Sulfates functionalization can be effectively realized using sulfamic acid/NMP. EtO sterilization is the most suitable sterilization technique for grafted sulfates. Besides, this thesis also offers a good strategy for the analysis of toxic leachable substances using suitable physicochemical characterization techniques. Future work will focus on minimizing/eliminating the release of toxic substances via reducing the crosslinking density. Another interesting aspect is to study whether grafted sulfates can meet the need for anti-thrombogenicity.
Despite the importance of negotiations in companies and their contribution to strategic corporate planning, researchers have not yet focused on assessing the development of negotiations in the future. To broaden the field of futures research in negotiations and to provide empirical guidance about strategic business decisions to negotiators and managers, this work exploratively investigates the future of negotiations. The impact of trends on negotiations and negotiation behavior, as well as the development of future negotiation scenarios are therefore examined. Moreover, the preparation of negotiators for the future is analyzed and how effective negotiation teaching can be designed to improve negotiation performance.
Am 1.5.2014 ist das Gesetz zur Regelung der vertraulichen Geburt in Kraft getreten. Damit sollte eine rechtssichere Alternative zu Babyklappen oder anonymen Geburten geschaffen werden. Die Regelungen ermöglichen schwangeren Personen, die bei der Geburt ihre Identität geheim halten wollen, eine medizinisch begleitete Entbindung und sichern gleichzeitig das Recht des Kindes auf Kenntnis seiner Herkunft sowie dessen Unterbringung. Neben juristischen Fragestellungen überprüft die Autorin die Anwendung der Regelungen in der Praxis anhand eigener Umfragen bei Familiengerichten und Jugendämtern. Sie analysiert die Frage, ob sich die vertrauliche Geburt als rechtssicheres Verfahren gegenüber vollständig anonymen Kindesabgaben durchsetzen konnte.
Die Zeit um 1600 markiert eine Zäsur in der Ausbildung junger Adliger, da neben der etablierten humanistischen Ausbildung zunehmend moderne Fremdsprachen und die institutionalisierte Ausbildung standesspezifischer Inhalte wie Fechten, Tanzen und Reiten an Bedeutung gewannen. Im vorliegenden Band werden die Ausbildungsgänge der Söhne dreier südwestdeutscher Freiherren- und Grafenfamilien in der Zeit des Späthumanismus untersucht. Dabei werden standesspezifische Bildungsstrategien herausgearbeitet und familiäre sowie konfessionelle Unterschiede aufgezeigt. Die Familien der Reichsgrafen orientierten sich – soweit es ihr Budget zuließ – einerseits am Fürstenstand, andererseits waren sie bestrebt, eigene Akzente zu setzen, sei es aus standespolitischen Erwägungen oder, weil sie eigenen Traditionen verpflichtet waren. Die systematische Auswertung breiter Quellenbestände förderte dabei auch eine Fülle an Informationen zu weiteren Wissensgebieten wie etwa der Reise-, Medizin-, Musikgeschichte oder der Alltagskultur zutage.
Epigenetische Mechanismen spielen eine entscheidende Rolle bei der Pathogenese von Colitis ulcerosa (CU). Ihr Einfluss auf das beobachtete Ungleichgewicht zwischen pro- und anti-inflammatorischen Cytokinen ist hingegen weitgehend unerforscht. Einige der wichtigsten immunmodulatorischen Cytokine sind die Mitglieder der heterodimeren Interleukin- (IL-) 12-Familie, die durch das Kombinieren einer der drei α-Ketten (IL-12p35, IL-27p28, IL-23p19) mit den ß-Untereinheiten IL-12p40 oder EBI3 (Epstein-Barr Virus-induziertes Gen 3) charakterisiert sind. IL-35 (IL-12p35/EBI3) spielt eine bedeutende anti-inflammatorische Rolle bei verschiedenen Erkrankungen, wohingegen seine Level bei chronischen Entzündungen erniedrigt sind. Eine mögliche Ursache könnte eine transkriptionelle Stilllegung über epigenetische Modifikationen sein. Tatsächlich konnte durch die Stimulation mit dem DNA-Methyltransferase-Inhibitor (DNMTi) Decitabin (DAC; Dacogen®) eine Induktion von EBI3 in humanen Epithelzellen aus gesundem Colon (HCEC) erreicht werden, die als Modell für ein lokales Entzündungsgeschehen dienten. Diese Regulation über DNA-Methylierung konnte in weiteren humanen Zellen unterschiedlichen Ursprungs sowie durch Stimulation von HCEC-Zellen mit zwei weiteren DNMTi, dem Cytosin-Analogon Azacytidin (AZA; Vidaza®) und dem natürlich vorkommenden, epigenetisch wirksamen Polyphenol Epigallocatechingallat (EGCG), verifiziert werden. Die kombinierte Inkubation mit Tumor-Nekrose-Faktor α (TNFα) resultierte jeweils in einer über-additiven Induktion von EBI3.
Weiterführende Untersuchungen zeigten, dass TNFα trotz Beeinflussung der epigenetischen DNMT- und Ten-eleven Translocation- (TET-) Enzyme keinen Einfluss auf die globalen Methylierungs- oder Hydroxymethylierungslevel hatte, jedoch eine genspezifische DNA-Hypomethylierung im EBI3-Promotor induzierte. Durch Nutzung verschiedener Inhibitoren konnte darüber hinaus nachgewiesen werden, dass der beobachtete synergistische Effekt der gemeinsamen DAC und TNFα-Stimulation hauptsächlich über NFκB (Nuclear factor “kappa-light-chain-enhancer” of activated B-cells) vermittelt wird. Ein Teil verläuft dabei über p38 MAPK (mitogen-activated protein kinases), während die JNK- (c-Jun N-terminale Kinasen-) und ERK- (extracellular-signal-regulated kinases) Signalwege keine Rolle spielen.
In der vorliegenden Arbeit wurde zudem gezeigt, dass die DNA-Hypomethylierung während eines entzündlichen Zustandes auch in einer erhöhten EBI3-Proteinexpression resultiert. Die Höhe der immunologisch detektierten Banden wies auf eine Dimerbildung sowohl im Zelllysat als auch im Überstand hin. Humane Colonepithelzellen sind demnach in der Lage, Cytokine zu bilden und zu sezernieren, was die Bedeutung von Nicht-Immunzellen bei der lokalen Immunantwort unterstreicht. Mittels Genexpressionsanalysen wurden IL-12p35 und IL-23p19 als mögliche Bindungspartner identifiziert. Aufgrund kreuzreaktiver Antikörper ist ein direkter Nachweis der EBI3-Dimere derzeit nicht möglich. Die stattdessen genutzte Kombination verschiedener Methoden dient als geeigneter Ersatz für die problematischen Antikörper-basierten Analysen wie Immunpräzipitation oder ELISA. Durch molekularbiologische, immunologische und massenspektrometrische Methoden konnte IL-35 identifiziert werden, während IL-39 (IL-23p19/EBI3) nicht detektiert wurde. Dies ist in Einklang mit den Erkenntnissen mehrerer Forschungsgruppen, die eine Bildung des nativen humanen Dimers aus IL-23p19 und EBI3 bezweifeln. Des Weiteren wurde die biologische Aktivität des behandlungsinduzierten IL 35-Proteins durch einen Funktionsassay nachgewiesen.
Neben einer DNMTi-bedingten transkriptionellen Aktivierung konnte eine Regulation von EBI3 über Histonacetylierungen gezeigt werden. Der EBI3-induzierende Effekt des Histondeacetylasen-Inhibitors (HDACi) Trichostatin A (TSA) wurde durch SAHA (suberoylanilide hydroxamic acid (Vorinostat; Zolinza®)) verifiziert. Ähnlich zu der Stimulation mit den hypomethylierenden Substanzen wurde ein synergistischer Effekt bei paralleler Inkubation mit TNFα beobachtet, der in einer gesteigerten Bildung des EBI3-Proteins resultierte.
Um die Befunde in einem komplexeren in vivo-Modell zu untersuchen, wurde eine chronische Colitis in Ebi3-defizienten Mäusen und dem dazugehörigen Wildtypstamm C57BL/6 durch zyklische Applikation von Natriumdextransulfat (Dextran sodium sulfate (DSS)) induziert. Der Vergleich klinischer Parameter wie Mortalitätsrate und Körper- sowie Milzgewicht wies bei Abwesenheit von Ebi3 signifikant stärkere colitische Symptome auf. Dies bestätigte die zentrale Rolle von Ebi3 in der Colitisentwicklung und deutete auf eine bevorzugte Bildung des anti-inflammatorisch wirkenden IL-35 statt des pro-inflammatorischen IL-39 in den Wildtyptieren hin. Durch zusätzliche therapeutische Behandlung der C57BL/6-Mäuse nach der DSS-Gabe konnte die in der Literatur beschriebene positive Wirkung von SAHA auf die Colitismanifestation bestätigt werden. Im Gegensatz dazu war der HDACi in den Ebi3-defizienten Tieren nicht in der Lage, die colitischen Parameter zu verbessern beziehungsweise verschlimmerte den Krankheitsphänotyp. Expressionsanalysen von Up- und Downstream-Target-Genen lieferten weitere Hinweise darauf, dass bei Anwesenheit von Ebi3 IL-35 statt IL-39 gebildet wird, was in Einklang mit den in vitro-Untersuchungen steht.
Die vorliegende Arbeit konnte durch den Vergleich der C57BL/6-Mäuse mit den Ebi3-defizienten Tieren neue Erkenntnisse über die Wirkungsweise von SAHA erbringen. Histonacetylierende Bedingungen verbessern colitische Symptome über einen Mechanismus, der die epigenetische Induktion von Ebi3 mit nachfolgender IL-35-Bildung involviert. Durch Kooperation der epigenetischen Mechanismen Hypomethylierung und Histonacetylierung wurde der stärkste Effekt auf die EBI3-Induktion bewirkt.
Insgesamt konnte in der vorliegenden Arbeit durch in vitro- und in vivo-Analysen die epigenetische und NFκB-vermittelte Induktion von EBI3 über DNA-Demethylierung und Histonacetylierung mit nachfolgender IL-35-Bildung und –Sezernierung nachgewiesen werden. Da IL-35 in der Lage ist, colitische Symptome zu mildern, stellt die epigenetische Reaktivierbarkeit von EBI3 durch DNMTi und HDACi eine vielversprechende Alternative für die derzeit genutzten, oft nicht oder nur kurzfristig wirksamen Therapien bei der Behandlung einer CU dar. Einer übermäßigen Immunantwort während schubweiser entzündlicher Phasen könnte entgegengewirkt und Komplikationen wie die Bildung Colitis-assoziierter Karzinome verhindert werden.

Postcolonial Surveillance investigates the long history of the European border regime, focusing on the colonial forerunners of today’s border technologies. The book takes a longue durée perspective to uncover how Europe’s colonial history continues to shape the high-tech political present and has morphed into EU border migration policies, border security, and surveillance apparatuses. It exposes the racial hierarchies and power relations that form these systems and highlights key moments when the past and present interact and collide, such as in panoptic surveillance, biopolitical registers, biometric sorting, and deterrent media infrastructure. The technological genealogies assembled in this book reveal the unacknowledged histories that had to be rejected for the seemingly clean, unbiased, and neutral technologies to emerge as such.

Vom Weltuntergang mit Richard Wagners "Tristan und Isolde" über die Gesangsperformance einer zum Tode verurteilten Björk am Galgen bis hin zu Johann Sebastian Bachs Orgelmusik als Erklärungsmodell für Hypersexualität: So seltsam das Kino des Lars von Trier erscheinen mag, so vielfältige Möglichkeiten bietet es, über ein Musikphänomen nachzudenken, das einen Großteil der heutigen Kunst- und Unterhaltungswelt prägt.
In Lars von Triers Filmen erklingt hauptsächlich Musik, die es bereits vor den Filmen gab. Einerseits besitzt solche präexistente Musik ein ausgeprägtes Eigenleben, andererseits entsteht aus der filmischen Aneignung etwas Neues. Am Beispiel eines der einflussreichsten Regisseure der Gegenwart untersucht Pascal Rudolph, wie Filmschaffende Musik adaptieren und wie dadurch Bedeutungen und Wirkungen entstehen. Erstmals bietet das Buch auf Grundlage von unveröffentlichtem Produktionsmaterial und Insider-Interviews detaillierte Einblicke in die Arbeit bei Lars von Triers Filmprojekten im Besonderen, aber auch in die Arbeitsprozesse der filmmusikalischen Gestaltung im Allgemeinen. Der musikalischen Vielfalt in den Filmen wird die Studie durch ihren multiperspektivischen und transdisziplinären Ansatz gerecht. Die zehn Kapitel beleuchten das Zusammenwirken von Musik und Film auf diese Weise aus verschiedenen Blickwinkeln.
The knowledge of transformation pathways and transformation products of veterinary drugs is important for health, food and environmental matters. Residues, consisting of original veterinary drug and transformation products, are found in food products of animal origin as well as the environment (e.g., soil or surface water). Several transformation processes can alter the original veterinary drug, ranging from biotransformation in living organism to environmental degradation processes like photolysis, hydrolysis, or microbial processes. In this thesis, four veterinary drugs were investigated, three ionophore antibiotics Monensin, Salinomycin and Lasalocid and the macrocyclic lactone Moxidectin. Ionophore antibiotics are mainly used to cure and prevent coccidiosis in poultry especially prophylactic in broiler farming. Moxidectin is an antiparasitic drug that is used for the treatment of internal and external parasites in food-producing and companion animals. The main objective of this work is to employ different laboratory approaches to generate and identify transformation products. The identification was conducted using high-resolution mass spectrometry (HRMS). A major focus was placed on the application of electrochemistry for simulation of transformation processes. The electrochemical reactor – equipped with a three-electrode flow-through cell – enabled the oxidation or reduction by applying a potential. The transformation products derived were analyzed by online coupling of the electrochemical reactor and a HRMS and offline by liquid chromatography (LC) combined with HRMS. The main modification reaction of the identified transformation products differed for each investigated veterinary drug. Monensin showed decarboxylation and demethylation as the main modification reactions, for Salinomycin mostly decarbonylation occurred and for Lasalocid methylation was prevalent. For Moxidectin, I observed an oxidation (hydroxylation) reaction and adduct formation with solvent. In general, for Salinomycin and Lasalocid, more transient transformation products (online measurement) than stable transformation products (offline measurements) were detected. By contrast, the number of transformation products using online and offline measurements were identical for Monensin and Moxidectin. As a complementary approach, metabolism tests with rat or human liver microsomes were conducted for the ionophore antibiotics. Monensin was investigated by using rat liver microsomes and the transformation products identified were based on decarboxylation and demethylation. Salinomycin and Lasalocid were converted by human and rat liver microsomes. For both substances, more transformation products were found by using human liver microsomes. The transformation products of the rat liver microsome conversion were redundant, and the transformation products were also found at the human liver microsome assay. Oxidation (hydroxylation) was found to be the main modification reaction for both. In addition, a frequent ion exchange between sodium and potassium was identified. The final two experiments were performed for one substance each, whereby the hydrolysis of Monensin and the photolysis of Moxidectin was investigated. The transformation products of the pH-dependent hydrolysis were based on ring-opening and dehydration. Moxidectin formed several transformation products by irradiation with UV-C light and the main modification reactions were isomeric changes, (de-)hydration and changes of the methoxime moiety. In summary, transformation products of the four investigated veterinary drugs were generated by the different laboratory approaches. Most of the transformation products were identified for the first time. The resulting findings provide an improved understanding of clarifying the transformation behavior.
Das Promotionsvorhaben verfolgt das Ziel, die Zuverlässigkeit der Datenspeicherung und die Speicherdichte von neu entwickelten Speichern (Emerging Memories) mit Multi-Level-Speicherzellen zu verbessern bzw. zu erhöhen. Hierfür werden Codes zur Erkennung von unidirektionalen Fehlern analysiert, modifiziert und neu entwickelt, um sie innerhalb der neuen Speicher anwenden zu können. Der Fokus liegt dabei auf sog. Berger-Codes und m-aus-n-Codes. Da Multi-Level-Speicherzellen nicht mehr binär, sondern mit mehreren Leveln arbeiten, können bisher verwendete Codes nicht mehr verwendet werden, bzw. müssen entsprechend angepasst werden. Auf Basis der Berger-Codes und m-aus-n-Codes werden in dieser Arbeit neue Codes abgeleitet, welche in der Lage sind, Daten auch in mehrwertigen Systemen zu schützen.
Die Arbeit untersucht bewaffnete Konfliktszenarien, in denen an multinationalen Militäroperationen beteiligte Staaten während einer Gewahrsamsoperation gegnerische Kräfte oder andere Personen in Gewahrsam nehmen und diese dann an die Kräfte eines anderen Staates, oftmals der Hostnation mit zweifelhafter Menschenrechtsreputation, überstellen. Gewahrsamspersonen laufen dann Gefahr, Opfer erheblicher Rechtsverletzungen zu werden
Die vorliegende Untersuchung zum internationalen Kryptowerterecht schafft einen umfassenden Überblick für die Praxis, indem eine systematische Analyse der Sachverhalte unter Berücksichtigung der technischen Besonderheiten, insbesondere der Blockchain, erfolgt. Kern der Arbeit stellen die vier untersuchten Sachverhalte dar, angefangen beim Mining, der Ausgabe neuer Kryptowerte im Rahmen eines ICO, Transaktionen auf dem Sekundärmarkt und der Prospekthaftung bei ICOs. Aus rechtlicher Sicht wird vor allem die Frage der internationalen Zuständigkeit nach der Brüssel Ia-VO und des anwendbaren Rechts nach der Rom I und II-VO behandelt. Zusätzlich werden Aspekte des Kapitalmarkt-IPR erörtert.
The prevalence of depression and anxiety is increased in obese patients compared to healthy humans, which is partially due to a shared pathogenesis, including insulin resistance and inflammation. These factors are also linked to intestinal dysbiosis. Additionally, the chronic consumption of diets rich in saturated fats results in body weight gain, hormonal resistances and unfavorable changes in the microbiome composition. The intake of Lactobacilli has already been shown to improve dysbiosis along with metabolism and mood. Yet, the beneficial role and the underlying mechanism of Lactobacillus rhamnosus GG (LGG) to improve emotional behavior in established diet-induced obese conditions are, so far, unknown. To characterize the role of LGG in diet-induced obesity, female and male C57BL/6N mice were fed a semi-synthetic low-fat diet (LFD, 10 % kcal from fat) or a conventional high-fat diet (HFD, 45 % kcal from fat) for initial 6 weeks, which was followed by daily oral gavage of vehicle or 1x10^8 CFU of LGG until the end of the experiment. Mice were subjected to basic metabolic and extensive behavioral phenotyping, with a focus on emotional behavior. Moreover, composition of cecal gut microbiome, metabolomic profile in plasma and cerebrospinal fluid was investigated and followed by molecular analyses. Both HFD-feeding and LGG application resulted in sex-specific differences. While LGG prevented the increase of plasma insulin, adrenal gland weight and hyperactivity in diet-induced obese female mice, there was no regulation of anxiodepressive-like behavior. In contrast, metabolism of male mice did not benefit from LGG application, but strikingly, LGG decreased specifically depressive-like behavior in the Mousetail Suspension Test which was confirmed by the Splash Test characterizing motivation for ’self-care’. The microbiome analysis in male mice revealed that HFD-feeding, but not LGG application, altered cecal microbiome composition, indicating a direct effect of LGG on behavioral regulation. However, in female mice, both HFD-feeding and LGG application resulted in changes of microbiome composition, which presumably affected metabolism. Moreover, as diet-induced obese female mice unexpectedly did not exhibit anxiodepressive-like behavior, follow-up analyses were conducted in male mice. Here, HFD-feeding significantly altered abundance of plasma lipids whereas LGG decreased branched chain amino acids which associated with improved emotional behavior. In nucleus accumbens (NAcc) and VTA/SN, which belong to the dopaminergic system, LGG restored HFD-induced decrease of tyrosine hydroxylase, the rate-limiting enzyme in dopamine synthesis, on gene expression level. Lastly, transcriptome analysis in the NAcc identified gene expression of cholecystokinin as a potential mediator of the effect of LGG on HFD-induced emotional alterations. In summary, this thesis revealed the beneficial effects of LGG application on emotional alterations in established diet-induced obesity. Furthermore, both HFD-feeding and LGG treatment exhibited sex-specific effects, resulting in metabolic improvements in female mice while LGG application mitigated depressive-like behavior in obese male mice along with a molecular signature of restored dopamine synthesis and neuropeptide signaling.
The morphogenesis of sessile plants is mainly driven by directional cell growth and cell division. The organization of their cytoskeleton and the mechanical properties of the cell wall greatly influence morphogenetic events in plants. It is well known that cortical microtubules (CMTs) contribute to directional growth by regulating the deposition of the cellulose microfibrils, as major cell wall fortifying elements. More recent findings demonstrate that mechanical stresses existing in cells and tissues influence microtubule organization. Also, in dividing cells, mechanical stress directions contribute to the orientation of the new cell wall. In comparison to the microtubule cytoskeleton, the role of the actin cytoskeleton in regulating shoot meristem morphogenesis has not been extensively studied.
This thesis focuses on the functional relevance of the actin cytoskeleton during cell and tissue scale morphogenesis in the shoot apical meristem (SAM) of Arabidopsis thaliana. Visualization of transcriptional reporters indicates that ACTIN2 and ACTIN7 are two highly expressed actin genes in the SAM. A link between the actin cytoskeleton and SAM development derives from the observation that the act2-1 act7-1 double mutant has abnormal cell shape and perturbed phyllotactic patterns. Live-cell imaging of the actin cytoskeleton further shows that its organization correlates with cell shape, which indicates a potential role of actin in influencing cellular morphogenesis.
In this thesis, a detailed characterization of the act2-1 act7-1 mutant reveals that perturbation of actin leads to more rectangular cellular geometries with more 90° cell internal angles, and higher incidences of four-way junctions (four cell boundaries intersecting together). This observation deviates from the conventional tricellular junctions found in epidermal cells. Quantitative cellular-level growth data indicates that such differences in the act2-1 act7-1 mutant arise due to the reduced accuracy in the placement of the new cell wall, as well as its mechanical maturation. Changes in cellular morphology observed in the act2-1 act7-1 mutant result in cell packing defects that subsequently compromise the flow of information among cells in the SAM.
Hitze ist eine bedeutende klimatische Bedingung, die das Wachstum und das Überleben von Pflanzen bedroht. Extreme Temperaturereignisse in der Natur werden gravierender, häufiger, länger anhaltend, was sich nachteilig auf die landwirtschaftliche Produktion auswirkt. Daher ist es wichtig, mehr über die Mechanismen zu erfahren, die zu einer erhöhten Hitzetoleranz bei Pflanzen führen. Um auszuhalten und zu überleben, haben höhere Pflanzen komplexe Mechanismen entwickelt, um auf verschiedene Intensitäten von Hitzestress zu reagieren. Pflanzen haben eine thermische Toleranz, die es ihnen ermöglicht, schnelle und dramatische Temperaturanstiege für eine begrenzte Zeit zu überleben. Pflanzen können auch darauf vorbereitet werden, Hitzestress (HS) zu widerstehen, der ansonsten tödlich wäre, indem man sie kurzen, moderaten und nicht-tödlichen HS (als Priming-Stimulus bezeichnet) aussetzt, bevor sie hohem HS ausgesetzt werden. Eine erworbene Thermotoleranz kann bei Pflanzen unter optimalen Bedingungen lange aufrechterhalten werden, was bedeutet, dass Pflanzen während dieser Zeit Informationen speichern können. Mehrere Studien haben gezeigt, dass sich erworbene Thermotoleranz (Thermopriming) auf die erhöhte Widerstandsfähigkeit von Zellen, Geweben und Organismen gegenüber erhöhten Temperaturen nach vorheriger Hitzeeinwirkung bezieht. Die Aufrechterhaltung der erworbenen Thermotoleranz (Thermomemory) ist mit der Synthese von speziellen Stressproteinen verbunden, die am Zellschutz und der beschleunigten Gewebereparatur beteiligt sind, wie z. B. Hitzeschockproteine (HSPs). Neuere Studien haben eine Beteiligung von Hitzeschockproteinen, z.B. HSP21, in Chloroplasten an der Regulation des Thermogedächtnisses belegt. Als wichtiges Organell ist die mitochondriale Funktion entscheidend für die Reaktion von Pflanzenzellen auf Hitze. Es ist jedoch noch unbekannt, wie die molekulare und physiologische Beteiligung von HSPs an der mitochondrialen Funktion im Thermogedächtnis erfolgt. In unserer Studie haben wir gezeigt, dass Thermopriming Transkript- und Proteinspiegel von zwei mitochondrialen kleinen Hitzeschockproteinen, HSP23.56 (AT5G51440) und HSP23.6 (AT4G25200), induziert, die während der Thermogedächtnisphase 2-3 Tage andauern. Die morphologische Analyse von HSP23.5/6-transgenen Pflanzen zeigte eine HSP23.5/6-Funktionsredundanz bei Hitzestress. Wir zeigten, dass hsp23.5/6-Doppel-Knockout-Pflanzen Anomalien im Thermogedächtnis im Keimlingsstadium aufwiesen und dass reife hsp23.5/6-Pflanzen sowohl mit basaler Thermotoleranz als auch mit Thermogedächtnis empfindlicher sind. Die Wärmebehandlung beeinflusste die Atmungsrate von hsp23.5/6-Keimlingen im Vergleich zu WT signifikant, was auf eine mitochondriale Dysfunktion in Abhängigkeit von HSP23.5 und HSP23.6 hinweist. Darüber hinaus haben wir die Chaperon-Aktivität von HSP23.6 gegenüber dem Modellsubstratprotein Malatdehydrogenase (MDH) in vitro getestet und bestätigt, was darauf hindeutet, dass HSP23.6 möglicherweise zur Aufrechterhaltung der zellulären Lebensfähigkeit beiträgt. Darüber hinaus entdeckten wir ein neues HSP23.6-Clientprotein, CIB22, ein mitochondriales Komplex-I-Untereinheitsprotein. Nach experimentellen Daten (BiFC und Co-IP) interagieren HSP23.6 und CIB22 in Pflanzenzellen. Wir identifizierten auch einen Hitzereaktionsphänotyp in der cib22-Mutante im Vergleich zu WT sowie einen CIB22-Proteinabbau in der hsp23.5/6-Mutante, wenn sie Hitze ausgesetzt wurde. Unsere Ergebnisse legen nahe, dass die beiden mitochondrial lokalisierten
Hitzeschockproteine eine Rolle bei der Thermotoleranz spielen, vermutlich indem sie die mitochondriale Funktion und Struktur beeinflussen. Um neue genetische Komponenten zu identifizieren, die mit dem Thermogedächtnis in Pflanzen verbunden sind, haben wir weiterhin ein Proteom-Profiling von Arabidopsis WT (Col-0) -Keimlingen während des Thermogedächtnisses durchgeführt. Mehrere Zeitpunkte von Priming und Triggerung mit Kontrollen wurden gesammelt und analysiert, um dynamische Proteomänderungen während der Gedächtnisphase in
Arabidopsis-Zellen aufzudecken. Unter den Top-gedächtnis-assoziierten Proteinen entdeckten wir, dass HSP70-4 nach dem Priming signifikant hochreguliert wurde und für die nächsten vier Tage auf hohem Niveau bleibt (mindestens 2-fach erhöht). Durch Analyse ihres Hitzestressverhaltens konnten wir verifizieren, dass HSP70-4 an der 7 Reaktion von Pflanzen auf Hitzestress beteiligt ist. Interessant ist, dass HSP70-4-GFP nach dem Priming zytosolische Foci erzeugt, die für einige Tage während der Erholungsphase bestehen bleiben. Wir schlagen vor, dass der Fokus mit SGs verbunden ist, da Cycloheximid (CHX) GFP-Foci-Signale unterdrückt, wenn sie der Hitze ausgesetzt werden. Diese Ergebnisse weisen auf eine HSP70-4-vermittelte Transkriptions- und Translationssteuerungsverbindung (Modul) während der basalen Thermotoleranz und des Thermogedächtnisses sowie auf ihre potenzielle(n) Rolle(n) bei der Reaktion auf Hitzestress hin.
Zusammenfassend bietet unsere Forschung neue Einblicke in die Rolle von Hitzeschockproteinen bei der Kontrolle der Hitzestresstoleranz und des Gedächtnisses.

The Letting of Real Estate as a Problem of the Fulfillment of Statutory Requirements in the System of Income Taxes. A Value-Oriented Approach to the Specification of Statutory Commercial Elements Using the Example of the Differentiation of Special and Supplementary Services«: The thesis addresses the income tax classification of the letting of real estate as either asset management or a commercial activity. The process of decision-making in practice, which often appears to be very casuistic, is contrasted with a methodical approach, which - by revealing the principles and valuations of income tax law - is intended to lead to consistent results. In doing so, the fundamental findings of legal methodology are elaborated and made fruitful.
Heat stress (HS) is one of the most common abiotic stresses, frequently affecting plant growth and crop production. With its fluctuating nature, HS episodes are frequently interspersed by stress-free intervals. Plants can be primed by HS, allowing them to survive better a recurrent stress episode. A memory of this priming can be maintained during stress-free intervals and this memory is closely correlated with transcriptional memory at several HS-inducible loci. This transcriptional memory is evident from hyper-induction of a locus upon a recurrent HS. ASCORBATE PEROXIDASE 2 (APX2) shows such hyper-induction upon recurring HS, however, the molecular basis of this transcriptional memory is not understood. Previous research showed that the HSinduced transcriptional memory at APX2 can last for up to seven days, and that it is controlled by cis-regulatory elements within the APX2 promoter.
To identify regulators involved in HS transcriptional memory, an unbiased forward genetic screening using EMS mutated seeds of pAPX2::LUC was performed from this screen. Two EMS mutants with affected transcriptional memory of LUC were identified. I confirmed that both two EMS mutants resulted from the gene mutations of HISTONE ACETYLTRANSFERASE 1 (HAC1). Besides pAPX2::LUC, the HS-induced transcription of other HS memory genes were also affected in hac1 mutants. Moreover, HAC1 may promote HS transcriptional memory by acetylating promoters of HS memory genes.
On the other hand, to identify cis-regulatory elements that are required for transcriptional memory of APX2, I performed promoter analysis of the four conserved HSEs identified within a functional APX2 promoter. I found out that one of the HSEs (HSE1) is necessary for both HS-induced APX2 transcription and transcriptional memory, while another one of HSEs (HSE2) is important for HS-induced APX2 transcriptional memory. I also found out that the HSE1 itself (with 10 bp of flanking sequence) is sufficient to confer HS-induced APX2 transcriptional memory, and HSE1 is also necessary for HSFA2 to bind on APX2 promoter and activate APX2 transcription. The findings will provide important clues for the molecular mechanism of transcriptional memory and will enable engineering of enhanced stress tolerance in crops.

Maria Weilandt betrachtet sie unter einem literatur- und kunstkomparatistischen Blickwinkel nicht als starre und unveränderliche Entitäten, sondern als Ensemble verflochtener Erzählungen. Anhand des Stereotyps der Pariserin im 19. und beginnenden 20. Jahrhundert entwickelt sie ein eigenes Analysekonzept. Neben literarischen Texten und Kunstwerken beleuchtet sie dabei auch die sogenannte Parisienne als Werbefigur der Pariser Warenhauskultur sowie als Repräsentation eines spezifischen intersektionalen Konzepts von Nation, etwa im Rahmen der Weltausstellung 1900.
Der Verteidigungsausschuss des Deutschen Bundestags steht seit seiner Gründung in rationaler und emotionaler Auseinandersetzung mit Parlament und Öffentlichkeit. Wolfgang Geist untersucht in seiner Langzeitanalyse die wechselnde Stellung des Ausschusses im Bundestag und gegenüber dessen Fraktionen unter den sich wandelnden politischen und gesellschaftlichen Gegebenheiten. Uncovering the interplay between nutrient availability and cellulose biosynthesis inhibitor activity

All plant cells are surrounded by a dynamic, carbohydrate-rich extracellular matrix known as the cell wall. Nutrient availability affects cell wall composition via uncharacterized regulatory mechanisms, and cellulose deficient mutants develop a hypersensitive root response to growth on high concentrations of nitrate. Since cell walls account for the bulk of plant biomass, it is important to understand how nutrients regulate cell walls. This could provide important knowledge for directing fertilizer treatments and engineering plants with higher nutrient use efficiency. The direct effect of nitrate on cell wall synthesis was investigated through growth assays on varying concentrations of nitrate, measuring cellulose content of roots and shoots, and assessing cellulose synthase activity (CESA) using live cell imaging with spinning disk confocal microscopy. A forward genetic screen was developed to isolate mutants impaired in nutrient-mediated cell wall regulation, revealing that cellulose biosynthesis inhibitor (CBI) activity is modulated by nutrient availability. Various non-CESA mutants were isolated that displayed CBI resistance, with the majority of mutations causing perturbation of mitochondria-localized proteins. To investigate mitochondrial involvement, the CBI mechanism of action was investigated using a reverse genetic screen, a targeted pharmacological screen, and -omics approaches. The results generated suggest that CBI-induced cellulose inhibition is due to off-target effects. This provides the groundwork to investigate uncharacterized processes of CESA regulation and adds valuable knowledge to the understanding of CBI activity, which could be harnessed to develop new and improved herbicides.
Natural hazards pose a threat to human health and life. In Germany, where the research for this thesis was conducted, numerous weather extremes occurred in the recent past that caused high numbers of fatalities and huge financial losses. The focus of this research is centred around two relevant natural hazards: heat stress and flooding. Preventing negative health impacts and deaths, as well as structural and monetary damage is the purpose of risk management and this requires citizens to adapt as well. Risk communication is implemented to foster people’s risk perception and motivate individual adaptation. However, methods of risk and crisis communication are often not evaluated in a structured manner. Much interdisciplinary research exists on both risk perception and adaptation, however, not much is known on the connection between the two. Furthermore, the existing research on risk communication is often not theory-driven and its impact on individual adaptation and risk perception is not thoroughly documented. This dissertation follows three research aims: (1) Compare psychological theories that contribute to natural hazard research. (2) Explore risk perception and adaptive behaviour by applying multiple methods. And (3) evaluate one risk communication method and one crisis communication method in a theory-driven manner to determine their impact on risk perception and adaptive behaviour. First, a literature review is provided on existing psychological theories which aim to explain the behaviour of individuals with regards to natural hazards. The three key theories included are the Protection Motivation Theory (PMT), the Protective Action Decision Model (PADM), and the Risk Information Seeking and Processing Model (RISP). Each of these are described and compared to each other with a focus on their explanatory power and practical significance in interdisciplinary research. Theoretical adaptations and possible extensions for future research are proposed for the presented approaches. Second, a multimethod field study on heat stress at an open-air event is presented. Face-to-face surveys (n = 306) and behavioural observations (n = 2750) were carried out at a horticultural show in Würzburg in summer 2018. The visitors’ risk perception, adaptive behaviour, and activity level were analysed and compared between hot days, summer days, and rainy days, applying correlation analyses, ANOVA, and multiple regression analyses. Heat risk perception was generally high, but most respondents were unaware of heat warnings on the day of their visit. During hot days the highest level of adaptation and lower activity levels were observed. Discrepancies between reported and observed adaptation emerged for different age groups.. Third, a telephone and web-based household survey on heat stress was conducted in the cities of Würzburg, Potsdam, and Remscheid in 2019 (n = 1417). The PADM served as the study’s theoretical framework. In multiple regression analyses the PADM factors of environmental and demographic context, risk communication, and psychological processes explained a substantial share of variance of protection motivation, protective response, and emotion-focused coping. Elements of crisis communication of a heat warning were evaluated experimentally. Results showed that understanding and adaptation intention was significantly higher in individuals that had received action recommendations alongside the heat warning. Fourth, the focus is set on a risk communication method of the flood context. A series of workshops on individual flood protection was carried out in six different settings. The participants (n = 115) answered a pretest-posttest questionnaire. Mixed-model analyses revealed significant increases in self-efficacy, subjective knowledge, and protection motivation. Stronger effects were observed in younger participants and those with lower levels of previous knowledge on flood adaptation as well as no flood experience. The findings of this thesis help to understand individual adaptation, as well as possible impacts of risk and crisis communication on risk perception and adaptation. The scientific background of this work is rooted in the disciplines of psychology and geosciences. The two theories PMT and PADM proved to be useful theoretical frameworks for the presented studies to suggest improvements in risk communication methods. A broad picture of individual adaptation is captured through a variety of methods of self-reports (face-to-face, telephone-based, web-based, and paper-pencil surveys) and behavioural observations, which recorded past and intended behaviour. Alongside with further methodological recommendations, the theory-driven evaluations of risk and crisis communication methods can serve as best-practice examples for future evaluation studies in natural hazard research but also other sciences dealing with risk behaviour to identify and improve effective risk communication pathways.

Das Konzept des Gefährdungsschadens gehört seit mindestens 130 Jahren zum festen strafrechtlichen Repertoire. Allerdings ist bis heute nicht eindeutig geklärt, wo die Grenze zwischen vermögensrelevanten und -irrelevanten Gefährdungen verläuft. Im Gegensatz zu den bisherigen sachverhalts- und deliktsbezogenen Fallgruppen bildet Melanie Epe daher zehn induktiv-dogmatische Fallgruppen auf Basis aller in der Standardliteratur publizierten 382 Entscheidungen des Reichsgerichts, des BGH und der Obergerichte. Die Leistungsfähigkeit der Fallgruppen ist, dass diese die Figur des Gefährdungsschadens in der Praxis greifbarer machen. Denn es wird an dogmatische, wirtschaftliche Besonderheiten für die Begründung des Gefährdungsschadens angeknüpft.

Wie verändert sich die Beziehung von Gesellschaften zu ihrer natürlichen Umgebung über die Zeit? Wie werden natürliche Systeme »in Wert« gesetzt? Und welchen Einfluss hat das auf die von uns so bezeichnete »Natur«? Am Beispiel eines Korkeichenwaldes in Marokko geht Juliane Schumacher diesen Fragen nach. Unter Bezugnahme auf Ansätze der Politischen Ökologie, der Science and Technology Studies und Foucaults Gouvernementalitätsanalyse zeigt sie, wie sich seit der Kolonialzeit die Bewirtschaftung des Waldes verändert hat. Dabei wird deutlich, wie Programme zur Integration der Wälder in globale Finanz- und Kohlenstoffmärkte zu neuen, experimentellen Formen der »Regierung des Waldes« führen.

Die Kapitalanlage von Pensionskassen in Investmentfonds wird dadurch erschwert, dass sich die sie betreffenden aufsichts- und steuerrechtlichen Normen widersprechen. Die den Pensionskassen aufsichtsrechtlich zugestandene Kaitalanlage, wird durch die aktuelle Lesart ihrer Steuerbefreiung beschränkt. Die Steuerbefreiung soll demnach entfallen, soweit die Pensionskasse durch die Kapitalanlage gewerbliche Einkünfte erzielt. Dann sei die dauernde Einkünfte- und Vermögensbindung für die Zwecke der Kasse nach § 5 Abs. 1 Nr. 3 lit. c KStG nicht gesichert. Das benachteiligt die Pensionskassen bei der Erwirtschaftung von Rentenleistungen. Zugleich findet diese Lesart bei der Auslegung der entsprechenden steuerrechtlichen Normen keinen Zuspruch.
Illegale Preisabsprachen und andere Kartellverstöße verursachen jährlich Schäden in Millionenhöhe. The Turkish language in diaspora is in process of change due to different language constellations of immigrants and the dominance of majority languages. This led to a great interest in various research areas, particularly in linguistics. Against this background, this study focuses on developmental change in the use of adverbial clause-combining constructions in Turkish-German bilingual students’ oral and written text production. It illustrates the use of non-finite constructions and some unique alternative strategies to express adverbial relations with authentic examples in Turkish and German. The findings contribute to a better understanding of how bilingual competencies vary in expressing adverbial relations depending on language contact and extra-linguistic factors.
The Annamites mountain range of Southeast Asia which runs along the border of Viet Nam and Laos is an important biodiversity hotspot with high levels of endemism. However, that biodiversity is threatened by unsustainable hunting, and many protected areas across the region have been emptied of their wildlife. To better protect the unique species in the Annamites, it is crucial to have a better understanding of their ecology and distribution. Additionally, basic genetic information is needed to provide conservation stakeholders with essential information to facilitate conservation breeding and counteract the illegal wildlife trade. To date, this baseline information is lacking for many Annamites species.
This thesis aims to assess the effectiveness of using non-invasive collection methods, i.e. camera-trap surveys and leech-derived wildlife host DNA, in order to improve and enhance our understanding of ecology, distribution, and genetic diversity of the Annamites terrestrial mammals.
In chapter 1, we analysed data from a systematic landscape camera-trap survey using single-species occupancy models to assess the ecology and distribution of two little-known Annamite endemics, the Annamite dark muntjac (Muntiacus rooseveltorum / truongsonensis) and Annamite striped rabbit (Nesolagus timminsi), in multiple protected areas across the Annamites. This chapter provided the first in-depth information on their ecology, as well as distribution patterns at large spatial scales. Most notably, we found that the Annamite dark muntjac was predominantly found at higher elevations, while responses to elevation varied among study areas for the Annamite striped rabbit. We estimated occupancy probabilities for both endemics by using their responses to environmental and anthropogenic influences and used this information to make recommendations for targeted conservation actions. We discuss how the approach we used for these two Annamites endemics can be expanded for other little-known and threatened species in other tropical regions.
As is the case with ecology and distribution, very little is known about the genetic diversity of the Annamite striped rabbit and other mammals of the Annamites. This poor understanding is mainly attributed to the lack of a comprehensive DNA sample collection that covers the species’ entire distribution range, which is believed to be a consequence of the low density of mammals or the remoteness of species’ habitat. In order to overcome the difficulties when trying to collect DNA samples from elusive mammals, we applied invertebrate-derived DNA (iDNA) sampling via hematophagous leeches to indirectly obtain genetic materials of their terrestrial host mammals.
In chapter 2, leech-derived DNA was used to study the genetic diversity of the Annamite striped rabbit population. By analysing the DNA extracted from leech samples collected at multiple study areas of the central Annamites, we found a genetic variation with five haplotypes among nine obtained sequences. Despite this diversity, we found no clear phylogeographic pattern among the lagomorph’s populations in central Annamites. The findings have direct conservation implications for the species, as local stakeholders are currently establishing a conservation rescue and breeding facility for Annamite endemic species. Thus our results suggested that Annamite striped rabbits from multiple protected areas in central Annamites can be used as founders for the breeding program.
In chapter 3, the genetic material of six mammals, which are frequently found in Indochina's illegal wildlife trade, was extracted from leeches collected at six study sites across the Anamites. Species-specific genetic markers were used to obtain DNA fragments that were analysed together with Genbank reference sequences from other parts of the species’ distribution range. Our results showed that invertebrate-derived DNA can be used to fill the sampling gaps and provide genetic reference data that is needed for conservation breeding programmes or to counteract the illegal wildlife trade.
Overal, this dissertation provides the first insights in the ecology, distribution, and genetics of rare and threatened species of the Annamites by utilising camera traps and leech-derived DNA as two non-invasive collection methods. This information is essential for improving conservation efforts of local stakeholders and managers, especially for the Annamite endemics. Results in this dissertation also show the effectiveness of both non-invasive methods for studying terrestrial mammals at a landscape level. By expanding the application of these methods to other protected areas across the Annamites, we will further our understanding of ecology, distribution, and genetics of Annamite endemics. With such landscape-scale surveys, we are able to provide stakeholders with an overview of the current status of wildlife in the Annamites which supports efforts to protect these secretive species from illegal hunting and thus their extinction.
Throughout their lifetime plants need to adapt to temperature changes. Plants adapt to nonfreezing cold temperatures in a process called cold priming (cold acclimation) and lose the acquired freezing tolerance during warmer temperatures through deacclimation. The alternation of both processes is essential for plants to achieve optimal fitness in response to different temperature conditions. Cold acclimation has been extensively studied, however, little is known about the regulation of deacclimation. This thesis elucidates the process of deacclimation on a physiological and molecular level in Arabidopsis thaliana. Electrolyte leakage measurements during cold acclimation and up to four days of deacclimation enabled the identification of four knockout mutants (hra1, lbd41, mbf1c and jub1) with a slower rate of deacclimation compared to the wild type. A transcriptomic study using RNA-Sequencing in A. thaliana Col-0, jub1 and mbf1c identified the importance of the inhibition of stress responsive and Jasmonate-ZIM-domain genes as well as the regulation of cell wall modifications during deacclimation. Moreover, measurements of alcohol dehydrogenase activity and gene expression changes of hypoxia markers during the first four days of deacclimation evidently showed that a hypoxia response is activated during deacclimation. Epigenetic regulation was observed to be extensively involved during cold acclimation and 24 h of deacclimation in A. thaliana. Further, both deacclimation studies showed that the previous hypothesis that heat stress might play a role in early deacclimation, is not likely. A number of DNA- and histone demethylases as well as histone variants were upregulated during deacclimation suggesting a role in plant memory. Recently, multiple studies have shown that plants are able to retain memory of a previous cold stress even after a week of deacclimation. In this work, transcriptomic and metabolomic analyses of Arabidopsis during 24 h of priming (cold acclimation) and triggering (recurring cold stress after deacclimation) revealed a uniquely significant and transient induction of DREB1D, DREB1E and DREB1F transcription factors during triggering contributing to fine-tuning of the second cold stress response. Furthermore, genes encoding Late Embryogenesis Abundant (LEA) and antifreeze proteins and proteins detoxifying reactive oxygen species were higher induced during late triggering (24 h) compared to primed samples, while cell wall remodelers of the class xyloglucan endotransglucosylase/hydrolase were early responders of triggering. The high induction of cell wall remodelers during deacclimation as well as triggering proposes that these proteins play an essential role in the stabilization of the cells during growth as well as the response to recurring stresses. Collectively this work gives new insights on the regulation of deacclimation and cold stress memory in A. thaliana and opens the door to future targeted studies of essential genes in this process.

Wo ist Deutschlands Raketenabwehr? Diese Frage rückte nach der völkerrechtswidrigen Annexion der Krim durch Russland 2014 in den Fokus der Presseberichterstattung. Für die Abwehr von ballistischen Raketen ist die Flugabwehrraketentruppe der Luftwaffe zuständig. Im Ost-West-Konflikt schützten rund 18.600 deutsche Soldaten im Rahmen der Integrierten NATO-Luftverteidigung die westliche Allianz vor Luftangriffen durch den Warschauer Pakt. Nach der Wiedervereinigung befand sich der Luftverteidigungsgürtel des Bündnisses nicht nur in einer geografisch wirkungslosen Position, sondern ihm fehlte auch die Daseinsberechtigung. Mit seiner Auflösung ging ein erheblicher Abbau von Personal und Material der Flugabwehrraketenverbände einher. Nach der Neuausrichtung der Bundeswehr 2012 blieb diesem Dienstbereich der Luftwaffe nur noch ein Geschwader mit rund 2.300 Dienstposten. Der alte Feind war weg – und Deutschland nach 1989/90 umgeben von Freunden und Verbündeten. Warum also sollte die Regierung in eine Fähigkeit investieren, die Deutschland für sich selbst nicht brauchte?

In everyday life, we take there to be ordinary objects such as persons, tables, and stones bearing certain properties such as color and shape and standing in various causal relationships to each other. Basic convictions such as these form our everyday picture of the world: the manifest image.
The scientific image, on the other hand, is a system of beliefs that is only based on scientific results. It contains many beliefs that are not contained in the manifest image. At first glance, this may not seem to be a problem. But Mulamustafi? shows convincingly that this is a mistake: The world as it is in itself cannot be both the way the manifest image depicts it and the way the scientific image describes it to be.
Adem Mulamustafic studied and completed his PhD in philosophy at the University of Potsdam. His areas of specialization are metaphysics, philosophy of science, and critical thinking.
The energy required to drive photochemical reactions is derived from charge separation across the thylakoid membrane. As the consequence of difference in proton concentration between chloroplasts stroma and thylakoid lumen, a proton motive force (pmf) is generated. The pmf is composed out of the proton gradient (ΔpH) and membrane potential (ΔΨ), and together they drive the ATP synthesis. In nature, the amount of energy fueling photosynthesis varies due to frequent changes in the light intensity. Thylakoid ion transport can adapt the energy flow through a photosynthetic apparatus to the light availability by adjusting the pmf composition. Dissipation of ΔΨ reduces the charge recombination at the photosystem II, allowing for an increase in ΔpH component to trigger a feedback downregulation of photosynthesis. K+ Exchange Antiporter 3 (KEA3) driven K+/H+ antiport reduces the ΔpH fraction of pmf, thereby dampening a non-photochemical quenching (NPQ). As a result, it increases the photosynthesis efficiency during the transition to lower light intensity. This thesis aimed to find the answers for questions concerning KEA3 activity regulation and its role in plant development. Presented data shows that in plants lacking chloroplast ATP synthase assembly factor CGL160 with decreased ATP synthase activity, KEA3 has a pivotal role in photosynthesis regulation and plant growth during steady-state conditions. Lack of KEA3 in cgl160 mutant results in a strong growth impairment, as photosynthesis is limited due to increased pH-dependent NPQ and decreased electron flow through cytochrome b6f complex. Overexpression of KEA3 in cgl160 mutant increases charge recombination at photosystem II, promoting photosynthesis. Thus, during periods of low ATP synthase activity, plants benefit from KEA3 activity. The KEA3 undergoes dimerization via its regulatory C-terminus (RCT). The RCT responds to changes in light intensity as the plants expressing KEA3 without this domain show reduced photo-protective mechanism in light intensity transients. However, those plants fix more carbon during the photosynthesis induction phase as a trade-off for a long-term photoprotection, showing KEA3 regulatory role in plant development. The KEA3 RCT is facing thylakoid stroma, thus its regulation depends on light-induced changes in the stromal environment. KEA3 activity regulation overlaps with the stromal pH changes occurring during light fluctuations. The ATP and ADP has shown to have an affinity towards heterologously expressed KEA3 RCT. Such interaction causes conformational changes in RCT structure. The fold change of RCT-ligand interaction depends on the environmental pH value. With a combination of bioinformatics and in vitro approach, the ATP binding site at RCT was located. Introduction of binding site point mutation in planta KEA3 RCT resulted in antiporter activity deregulation during transition to low light. Together, the data presented in this thesis allowed us to assess more broadly a KEA3 role in photosynthesis adjustment and propose the models of KEA3 activity regulation throughout transition in light intensity.
Während das nationale und das europäische Wettbewerbsrecht seit vielen Jahrzehnten eine differenzierte Regelung und wissenschaftliche Durchdringung erfahren haben, ist ein vergleichbarer wettbewerbsrechtlicher Normenbestand auf internationaler Ebene nicht zu verzeichnen. Diese Dissertation greift diese Forschungslücke auf und plädiert für die Schaffung eines internationalen, multilateralen Wettbewerbsrechts. Dabei wird der Bestand an hard-law und soft-law untersucht und als Ergebnis gefordert, neue multilaterale Wettbewerbsregelungen zu entwerfen. In institutioneller Hinsicht ist zu fragen, innerhalb welcher internationaler Organisation dies sinnvoll erfolgen kann. Insgesamt unternimmt die Dissertation den Versuch, mögliche Konturen einer globalen Wettbewerbsrechtsordnung aufzuzeigen und vertieft zu begründen.

Seit Aufstellung der Bundeswehr 1955 verloren über 3300 Soldaten und Soldatinnen ihr Leben im Dienst. Nur eine kleine Minderheit von ihnen fiel während des Kampfeinsatzes in Afghanistan. Die weitaus meisten Soldaten starben bei Unfällen. Ihnen hat die Bundeswehr lange das öffentliche Gedenken verweigert.
Erst mit Beginn des Afghanistan-Einsatzes 2002 setzte letztlich ein Umdenken ein, das in der Einweihung des Berliner Ehrenmales der Bundeswehr 2009 seinen vorläufigen Höhepunkt findet. Seitdem gedenkt die Bundeswehr offiziell und öffentlich ihrer toten Soldaten.
Aber warum verweigerte die Bundeswehr ihren Toten so lange ein öffentlich sichtbares und dauerhaftes Gedenken?

Fanatismus, Krieg und Terror – öffentliche Deutungen und Stereotype über Muslim*innen in der Bundesrepublik.
Ein großer Teil der deutschen Bevölkerung hat heutzutage eine negative Wahrnehmung von Muslim*innen. Ihnen wird pauschal ein Hang zu Gewalt, religiösem Fanatismus, Extremismus und Unterdrückung von Frauen unterstellt. Diese Zuschreibungen bestehen nicht erst seit den Terroranschlägen vom 11. September 2001, sondern haben sich bereits in den drei Jahrzehnten zuvor etabliert.
Alexander Konrad untersucht den Wandel der bundesdeutschen Wahrnehmungen von Muslim*innen von den Siebzigerjahren bis zur Jahrtausendwende. Dabei nimmt er öffentliche Aussagen und Handlungen von Akteur*innen aus Politik, Medien, Wissenschaft, Religionsgemeinschaften und Zivilgesellschaft kritisch in den Blick. Hintergründe, argumentative Überschneidungen und Agenden stehen im Zentrum seiner Analyse. Auch den damaligen Bemühungen um reflektierte Sichtweisen zu Muslim*innen spürt der Autor nach. Mit seiner Studie leistet Alexander Konrad einen fundamentalen Beitrag zur zeithistorischen Dekonstruktion von Denkweisen über Islam und Muslim*innen.
In der fortwährenden Diskussion zur Familienbesteuerung bewertet der Autor vor dem Hintergrund des deutschen Verfassungsrechts die geltenden Regelungen und mögliche Reformmodelle rechtlich, steuerpolitisch und ökonomisch. Unter dem besonderen Blickwinkel eines seit mehr als 20 Jahren tätigen Steuerberaters kann der Autor die Wirkung des Splittingtarifs und deren Alternativen ökonomisch gut durchdringen.
Die Ehe und Familie stehen unter dem besonderen Schutz der deutschen staatlichen Ordnung. Diese Schutzpflicht aus Art. 6 Abs. 1 GG geht deutlich über ein Abwehrrecht hinaus. Sie gebietet es, die Ehe zu schützen, ihr einen grundsätzlich unantastbaren Kernbereich privater Lebensführung zu gewähren und innerfamiliär staatlicherseits keine bestimmte innereheliche Arbeits- und Aufgabenverteilung vorzuschreiben.
Die Ehe wird häufig als „Keimzelle“ jeglicher menschlichen Gesellschaft bezeichnet. Das erscheint folgerichtig und historisch bedeutsam. Im heutigen Zeitalter jedoch ist sie mehr oder weniger eine Erwerbs-, Konsum- und Verantwortungsgemeinschaft. Insofern stellt sich vornehmlich die Frage nach einer genügenden Rechtfertigung für die Gewährung eines Splittingtarifs.
Ausgangspunkt der umfassenden systematischen Darstellung ist die historischen Entwicklung in Deutschland und den angrenzenden Staaten; insbesondere der heutigen Europäischen Union. Denn in anderen Staaten werden bereits unterschiedliche Modelle wie beispielsweise die Einzel- oder Familienbesteuerung angewendet.
Verfassungsrechtlich hat der Gesetzgeber Veränderungen der gesellschaftlichen Rahmenbedingungen in und für die Familie in seine gesetzgeberischen Entscheidungen zwingend einzubeziehen. Es stellt sich demnach die Frage, ob beispielsweise seit der Entscheidung des Bundesverfassungsgerichts zum Ehegattensplitting im Jahr 1957 relevante Veränderungen eingetreten sind, die eine Änderung der Familienbesteuerung rechtfertigen oder gar erfordern. Die zwischenzeitliche Anerkennung von Lebenspartnerschaften und die Ehe für alle sind Ausdruck einer solchen Veränderung. Sie implizieren die bereits eingetretenen Veränderungen der Lebensverhältnisse. Daher ist die politische Diskussion für eine Veränderung nachvollziehbar. Ob jedoch Alternativen vorzugswürdig sind, wird ausführlich dargestellt.
Der Autor untersucht das System des gerichtlichen Rechtsschutzes gegen beamtenrechtliche Personalauswahlentscheidungen im Hinblick auf dessen tatsächliche Wirksamkeit zur Durchsetzung des Grundrechts auf gleichen Zugang zu öffentlichen Ämtern aus Art. 33 Abs. 2 GG. Maßstab der Wirksamkeitsprüfung ist die Rechtsschutzgarantie aus Art. 19 Abs. 4 S. 1 GG. Berücksichtigt werden insbesondere die Modifikationen der etablierten Rechtsschutzdogmatik durch das Urteil des BVerwG vom 04.11.2010. Der Autor konstatiert, dass Bewerbern um ein öffentliches Amt nun zwar ein formell lückenloser Primärrechtsschutz eingeräumt wird. Dessen praktische Wirksamkeit ist jedoch durch zahlreiche prozessuale Besonderheiten und die Handhabung des dem Dienstherrn bei der Auswahlentscheidung zugebilligten weiten Beurteilungs- und Ermessensspielraums erheblich eingeschränkt. Der Autor folgert, dass der geforderte effektive gerichtliche Rechtsschutz nur durch eine rechtsschutzfreundliche Gestaltung des behördlichen Auswahlverfahrens gewährleistet werden kann, und leitet bestimmte organisatorische Mindestanforderungen an das Auswahlverfahren her.

Wie jede öffentlichkeitssuchende Institution oder Person legen auch die katholische Kirche und ihre Päpste großen Wert auf das eigene Image. Dabei weicht die intendierte Selbstdarstellung oftmals von der medialen Wahrnehmung ab oder steht ihr sogar diametral entgegen. Ganz gleich ob Pius XII. als „Hitlers Papst“, Johannes XXIII. als „Revoluzzer“ oder Paul VI. als „Antipillenpapst“ – es existieren eine Fülle von außen zugetragener printmedialer Papst-Images, deren Entstehung, Verlauf und Brüche es in diesem Buch mithilfe einer mediensemiotischen, textbasierten Methodik auszumachen und nachzuzeichnen gilt.
Leonhard Frank (1882-1961) hat bereits ein an Erfahrungen reiches Leben hinter sich, als er 1950 nach Deutschland zurückkehrt. Das langjährige Exil lässt der Schriftsteller nicht zuletzt aufgrund mangelnder Veröffentlichungs- und Verdienstmöglichkeiten sowie einer rigiden Immigrationsgesetzgebung in den USA hinter sich. Vor allem jedoch zieht es ihn in seine Heimat als das Land seiner Sprache zurück. Und nur hier weiß er sein Publikum, dessen Resonanz er als existenziell empfindet.
In dieser Studie werden die Bemühungen nachvollzogen, die ein während der Weimarer Republik renommierter und ökonomisch erfolgreicher Autor nach dem erzwungenen Exil während der Zeit des Nationalsozialismus unternimmt, um als linker Schriftsteller seine Position innerhalb der besonderen literarischen und kulturpolitischen Bedingungen in der BRD und der DDR im Jahrzehnt der 1950er Jahre zu finden. Ausgehend von umfangreichen Archivmaterialien werden die Lebens- und Arbeitsumstände Franks in seinem letzten Lebensjahrzehnt in die jeweiligen biographischen, sozialen, politischen und kulturellen Kontexte gesetzt.
Besondere Aufmerksamkeit erfährt dabei die Besonderheit, dass Frank als Bürger der Bundesrepublik fortdauernde Beziehungen zu Institutionen und Personen in der DDR unterhielt. Aus dieser Konstellation resultiert eingedenk der politisch-historischen Prozesse, in die sie sich einbettete, ein Spannungsfeld, dessen Spezifika transparent gemacht werden.
This thesis focuses on the synthesis of novel functional materials based on plasmonic nanoparticles. Three systems with targeted surface modification and functionalization have been designed and synthesized, involving modified perylenediimide doped silica-coated silver nanowires, polydopamine or TiO2 coated gold-palladium nanorods and thiolated poly(ethylene glycol) (PEG-SH)/dodecanethiol (DDT) modified silver nanospheres. Their possible applications as plasmonic resonators, chiral sensors as well as photo-catalysts have been studied. In addition, the interaction between silver nanospheres and 2,3,5,6-Tetrafluoro-7,7,8,8-tetracyanoquinodimethane (F4TCNQ) molecules has also been investigated in detail.
In the first part of the thesis, surface modification on Ag nanowires (NWs) with optimized silica coating through a modified Stöber method has been firstly conducted, employing sodium hydroxide (NaOH) to replace ammonia solution (NH4OH). The coated silver nanowires with a smooth silica shell have been investigated by single-particle dark-field scattering spectroscopy, transmission electron microscopy and electron-energy loss spectroscopy to characterize the morphologies and structural components. The silica-coated silver nanowires can be further functionalized with fluorescent molecules in the silica shell via a facile one-step coating method. The as-synthesized nanowire is further coupled with a gold nanosphere by spin-coating for the application of the sub-diffractional chiral sensor for the first time. The exciton-plasmon-photon interconversion in the system eases the signal detection in the perfectly matched 1D nanostructure and contributes to the high contrast of the subwavelength chiral sensing for the polarized light.
In the second part of the thesis, dumbbell-shaped Au-Pd nanorods coated with a layer of polydopamine (PDA) or titanium dioxide (TiO2) have been constructed. The PDA- and TiO2- coated Au-Pd nanorods show a strong photothermal conversion performance under NIR illumination. Moreover, the catalytic performance of the particles has been investigated using the reduction of 4-nitrophenol (4-NP) as the model reaction. Under light irradiation, the PDA-coated Au-Pd nanorods exhibit a superior catalytic activity by increasing the reaction rate constant of 3 times. The Arrhenius-like behavior of the reaction with similar activation energies in the presence and absence of light irradiation indicates the photoheating effect to be the dominant mechanism of the reaction acceleration. Thus, we attribute the enhanced performance of the catalysis to the strong photothermal effect that is driven by the optical excitation of the gold surface plasmon as well as the synergy with the PDA layer.
In the third part, the kinetic study on the adsorption of 2,3,5,6-Tetrafluoro-7,7,8,8-tetracyanoquino-dimethane (F4TCNQ) on the surface of Ag nanoparticles (Ag NPs) in chloroform has been reported in detail. Based on the results obtained from the UV-vis-NIR absorption spectroscopy, cryogenic transmission electron microscopy (cryo-TEM), scanning nano-beam electron diffraction (NBED) and electron energy loss spectroscopy (EELS), a two-step interaction kinetics has been proposed for the Ag NPs and F4TCNQ molecules. It includes the first step of electron transfer from Ag NPs to F4TCNQ indicated by the ionization of F4TCNQ, and the second step of the formation of Ag-F4TCNQ complex. The whole process has been followed via UV-vis-NIR absorption spectroscopy, which reveals distinct kinetics at two stages: the instantaneous ionization and the long-term complex formation. The kinetics and the influence of the molar ratio of Ag NPs/F4TCNQ molecules on the interaction between Ag NPs and F4TCNQ molecules in the organic solution are reported herein for the first time. Furthermore, the control experiment with silica-coated Ag NPs indicates that the charge transfer at the surface between Ag NPs and F4TCNQ molecules has been prohibited by a silica layer of 18 nm.
Schulleitungen nehmen eine zentrale Rolle bei der Weiterentwicklung der schulischen Qualität ein. Hierfür existieren Hinweise empirischer Untersuchungen, die Schulleitungen attestieren, dass ihr berufliches Handeln im direkten Zusammenhang mit Strukturen und Prozessen der schulischen Arbeit und mit Merkmalen auf Ebene der Lehrkräfte steht (z.B. Berufszufriedenheit, professionelle Weiterentwicklung; Kim & Lee, 2020; Liu, Keeley, Sui & Sang, 2021). Über diese Zusammenhänge steht das Führungshandeln von Schulleitungen in einer indirekten Beziehung mit der Qualität des Unterrichts sowie mit den Leistungen von Schüler*innen (z.B. Grissom, Egalite & Lindsay, 2021; Özdemir, Gün & Yirmibes, 2021; Pietsch, Lücken, Thonke, Klitsche & Musekamp, 2016). Im Rahmen der schulischen Entwicklung tragen Schulleitungen wesentlich dazu bei, die Entwicklungsbedarfe der Schule zu identifizieren, konkrete Zielstellungen zu definieren sowie entsprechende Maßnahmen zur Erreichung dieser Ziele zu planen, zu implementieren und zu evaluieren (vgl. Dalin, Rolff & Buchen, 1998). In all diese Prozesse können Schulleitungen weitere Mitglieder ihres Leitungsteams sowie Lehrkräfte involvieren und ihnen somit indirekt Führungsverantwortung übertragen (Heck & Hallinger, 2010). Auch aus normativer Perspektive nehmen Schulleitungen eine wichtige Funktion für die schulische Qualität ein. So verfügen sie neben Aufgaben der Administration und Verwaltung der Schule auch zunehmend über die Verantwortung, die schulische Qualität weiterzuentwickeln (Schratz, Wiesner, Kemethofer, George, Rauscher, Krenn & Huber, 2016). In Anbetracht der hohen Relevanz schulischen Führungshandelns für die Schulentwicklung, müssen Schulleitungen über die Kompetenzen zur Ausübung entsprechender Tätigkeiten verfügen. Diese können sie im Zuge einer vorbereitenden Ausbildung sowie durch Angebote der berufsbegleitenden Fort- und Weiterbildung erwerben. Die vorliegende Dissertation schließt an bestehende Forschungsarbeiten an und untersucht mithilfe von insgesamt vier Teilstudien, (1) über welche Aufgaben Schulleitungen hinsichtlich der Schulentwicklung verfügen, (2) wie ihr Handeln mit der Kooperation von Lehrkräften zusammenhängt, (3) wie sie die Zusammenarbeit von Lehrkräften im Rahmen der Schulentwicklung fördern können und (4) wie Schulleitungen auf Tätigkeiten der Schulentwicklung im Rahmen einer Fortbildung vorbereitet werden können. Insgesamt unterstreichen die vier Teilstudien in vielfältiger Weise die Relevanz von Schulleitungen und ihrem beruflichen Handeln für Strukturen und Prozesse der Schulentwicklung. Die Befunde können zeigen, inwiefern Schulleitungen über Aufgaben im Bereich der Schulentwicklung verfügen, wir ihr Handeln mit der Partizipation und Zusammenarbeit und von Lehrkräften im Rahmen der Schulentwicklung zusammenhängt und wie Schulleitungen auf Tätigkeiten der Schulentwicklung im Kontext von Fortbildungen vorbereitet werden können. Diesen Befunden sollte in Zukunft Rechnung getragen werden, indem Schulleitungen einerseits mehr Zeiten und Möglichkeiten für Tätigkeiten der Schulentwicklung eingeräumt werden. Anderseits betonen die Befunde die hohe Relevanz grundständig ausgebildeter Schulleitungen, um die Qualität der schulischen Arbeit längerfristig und eigenverantwortlich weiterentwickeln zu können. Dies sollte im Rahmen der Aus- und Fortbildung von Schulleitungen berücksichtigt werden.

Fibroblast growth differentiation factor 21 (FGF21) is known as a pivotal regulator of the glucose and lipid metabolism. As such, it is considered beneficial and has even been labelled a longevity hormone. Nevertheless, recent observational studies have shown that FGF21 is increased in higher age with possible negative effects such as loss of lean and bone mass as well as decreased survival. Hepatic FGF21 secretion can be induced by various nutritional stimuli such as starvation, high carbohydrate and fat intake as well as protein deficiency.. So far it is still unclear whether the FGF21 response to different macronutrients is altered in older age. An altered response would potentially contribute to explain the higher FGF21 concentrations found in older age. In this publication-based doctoral dissertation, a cross-sectional study as well as a dietary challenge were conducted to investigate the influence of nutrition on FGF21 concentrations and response in older age. In a cross-sectional study, FGF21 concentrations were assessed in older patients with and without cachexia anorexia syndrome anorexia syndrome compared to an older community-dwelling control group. Cachexia anorexia syndrome is a multifactorial syndrome frequently occurring in old age or in the context of an underlying disease. It is characterized by a severe involuntary weight loss, loss of appetite (anorexia) and reduced food intake, therefore representing a state of severe nutrient deficiency, in some aspects similar to starvation. The highest FGF21 concentrations were found in patients with cachexia anorexia syndrome. Moreover, FGF21 was positively correlated with weight loss and loss of appetite. In addition, cachexia anorexia syndrome itself was associated with FGF21 independent of sex, age and body mass index. As cachectic patients presumably exhibit protein malnutrition and FGF21 has been proposed a marker for protein insufficiency, the higher levels of FGF21 in patients with cachexia anorexia syndrome might be partly explained by insufficient protein intake. In order to investigate the acute response of FGF21 to different nutritional stimuli, a dietary challenge with a parallel group design was conducted. Here, healthy older (65-85 years) and younger (18-35 years) adults were randomized to one of four test meals: a dextrose drink, a high carbohydrate, high fat or high protein meal. Over the course of four hours, postprandial FGF21 concentrations (dynamics) were assessed and the FGF21 response (incremental area under the curve) to each test meal was examined.. In a sub-group of older and younger women, also the adiponectin response was investigated, as adiponectin is a known mediator of FGF21 effects on glucose and lipid metabolism. The dietary meal challenge revealed that dextrose and high carbohydrate intake result in higher FGF21 concentrations after four hours in older adults. This was partly explained by higher postprandial glucose concentrations in the old. For high fat ingestion no age differences were found. For the first time, acute FGF21 response to high protein intake was shown. Here, protein ingestion resulted in lower FGF21 concentrations in younger compared to older adults. Furthermore, sufficient protein intake, according to age-dependent recommendations, of the previous day, was associated with lower FGF21 concentrations in both age groups. The higher FGF21 response to dextrose ingestion resulted in a higher adiponectin response in older women, independent of fat mass, insulin resistance, triglyceride concentrations, inflammation and oxidative stress. Following the high fat meal, adiponectin concentrations declined in older women. Adiponectin response was not affected by meal composition in younger women. In summary, this thesis showed a positive association of FGF21 and cachexia anorexia syndrome with concomitant anorexia in older patients. Regarding the acute FGF21 response, a higher response following dextrose and carbohydrate ingestion was found in older compared with younger subjects. This might be attributed to a higher glucose response in older age. Furthermore, it was shown that the higher FGF21 response after dextrose ingestion possibly contributes to a higher adiponectin response in older women, independent of potential metabolic and inflammatory confounders. Acute protein ingestion resulted in a significant decrease in FGF21 concentrations. Moreover, protein intake of the previous day was inversely associated with fasting FGF21 concentrations. This might explain why FGF21 concentrations are higher in cachexia anorexia syndrome. These results therefore support the role of FGF21 as a sensor of protein restriction.

Hypoxic pulmonary vasoconstriction is an active alveolar hypoxia-caused physiological response redirecting pulmonary blood flow from poorly ventilated areas to better oxygenated lung regions in order to optimize oxygen supply. However, the signaling pathways underlying this pulmonary vascular response remain an area under investigation. In the present study I investigated the functional relevance of Pannexin 1 (Panx1)-mediated ATP release in hypoxic pulmonary vasoconstriction and chronic hypoxic pulmonary hypertension using murine isolated perfused lungs, chronic hypoxic mice, and pulmonary artery smooth muscle cell culture. In isolated mouse lungs, switch to hypoxic gas induced a marked increase in pulmonary artery pressure. Pharmacological inhibition of Panx1 using probenecid, Panx1 specific inhibitory peptide (10Panx1) or spironolactone as well as genetic deletion of Panx1 in smooth muscle cells diminished hypoxic pulmonary vasoconstriction in isolated perfused mouse lungs. Fura-2 imaging revealed a reduced Ca2+ response to hypoxia in pulmonary artery smooth muscle cells treated with spironolactone or 10Panx1. Although these findings suggested an important role of Panx1 in HPV, neither smooth muscle cell nor endothelial cell specific genetic deletion of Panx1 prevented the development of pulmonary hypertension in chronic hypoxic mice. Surprisingly, hypoxia did not induce ATP release and inhibition of purinergic receptors or ATP degradation by ATPase failed to decrease the pulmonary vasoconstriction response to hypoxia in isolated perfused mouse lungs. However, Panx1 antagonism as well as TRPV4 inhibition prevented the hypoxia-induced increase in intracellular Ca2+ concentration in pulmonary artery smooth muscle cells in an additive manner suggesting that Panx1 might modulate intracellular Ca2+ signaling independently of the ATP-P2-TRPV4 signaling axis. In line with this assumption, overexpression of Panx1 in HeLa cells increased intracellular Ca2+ concentrations in response to acute hypoxia. Conclusion: In this study I identifiy Panx1 as novel regulator of HPV.. Yet, the role of Panx1 was not attributable to the release of ATP and downstream P2 signaling pathways or activation of TRPV4 but rathter relates to a role of Panx1 as indirect or direct modulator of the Ca2+ response to hypoxia in PASMCs. Genetic deletion of Panx1 did not influence the development of chronic hypoxic pulmonary hypertension in mice.
Receptors predominantly expressed on tumor cells represent one of the key prerequisites of targeted radiotherapy. The gastric inhibitory polypeptide receptor (GIPR) has emerged as a promising target due to its substantial overexpression in neuroendocrine neoplasms (NENs) and virtual absence in healthy tissues (Waser 2012). So far, only radiolabeled peptides targeting the somatostatin receptor 2 (SSTR2) are approved for targeted radiotherapy of inoperable, metastatic NENs.
The aim of this thesis was to develop highly affine GIPR tracers for targeted radiotherapy by continuous in vitro and in vivo characterization of peptide sequence modifications. It was hypothesized that a GIPR antagonist might increase the sensitivity to detect GIPR-positive tumors relative to the agonist GIP(1-30), as shown for SSTR2 tracers (Reubi 2017). Further comparison to the SSTR2 agonist and antagonist (DOTATATE, JR11) should allow compound ranking regarding their ability to detect NENs.
The novel GIPR-targeting antagonists were conjugated to DOTA, enabling complexation of diagnostic (e. g. 111In) and therapeutic radionuclides (e. g. 177Lu). Among the high number of compounds screened, 3BP-3775 proved to be the most promising candidate for further preclinical and clinical development. High GIPR affinity and long receptor residence time in vitro were reflected in strong tumor uptake and retention in vivo. Low initial kidney accumulation and fast subsequent clearance represented a major advantage relative to previously described GIPR-targeting molecules (Gourni 2014). Furthermore, administration of 177Lu-3BP-3775 demonstrated for the first time strong therapeutic efficacy of a GIPR-targeting compound. In vitro receptor autoradiography with 111In-3BP-3626 (GIPR antagonist) demonstrated up to 6-fold higher signals in gastroenteropancreatic and bronchial NENs, relative to the clinically utilized SSTR2 agonist 111In-DOTATATE. Both receptor antagonists, however, revealed similar signal strength. In contrast to 111In-JR11, 111In-3BP-3626 showed no binding to non-target tissues, which led to higher tumor-to-background ratios for 111In-3BP-3626. Signal strength of the GIPR agonist 111In-GIP(1-30) was close to background in all investigated tumor samples. The ranking of compounds according to their ability to detect NENs based on autoradiographic signal intensities was determined to be: 111In-3BP-3626 ~ 111In-JR11> 111In-DOTATATE > 111In GIP(1-30).
In summary, this thesis proposes the application of the GIPR antagonist 3BP-3775 for a targeted radiotherapy in GEP- and bronchial NENs.

So entsteht das Bild einer Technik, welche die in sie gesetzten Erwartungen nicht erfüllte, einer fehlerhaften und statischen Doktrin, die für zahlreiche Rückschläge sorgte, und von individualistisch agierenden Piloten, die sich dem militärhierarchischen System entfremdeten.
