Active host-searching in bat ectoparasites. As a pilot study, this paper discribes the mobility of ectoparasites after removing from their hosts. Spinturnix acuminatus (Koch, 1836)(Acari, Spinturnicidae) a permanent stationary bat mite normally survives the removing only a few hours. This species moves easily on the patagium or on human skin. In contrast, on other surfaces the movements of this species seems to be less orientated and it is unlikely that the mite can reach a host in some distance. Nycteribia kolenatii Theodor & Moscona, 1954 (Diptera: Nycteribiidae) a bat fly, shows excellent movements in the fur of its host. The specimens survive the removing from the host several days. The ability to clamber up a wall was tested with a rough clay brick. The bat fly achieves only 9,6 cm on average. This result does not support good abilities for host searching in this species. Bat fleas (Siphonaptera: Ischnopsyllidae): Ischnopsyllus elongatus (Curtis, 1832), I. hexactenus (Kolenati, 1856), I. simplex (Rothschild, 1906)and other species survive the removing from their hosts several days. In case of a vertical barrier they showed a stereotype upside climbing, followed by a horizontal appetitive behavior. The fleas were climbing a distance in the upside of 6,45 m on average(1,2-21,68 m). As excellent climbers bat fleas are able to overcome edges and slopes and to crawl on the ceiling. Therefore, it seems to be no problem for a flea to parasite any bat in a resting room. Additionally, some bugs (Cimex lectularius Linnaeus, 1758) were examinated. They run with a speed of 63 cm per minute and were able to climb easily on the brick. On average their stretch of way achieved 2,85 m in ten minutes.

* 1. Large female insects usually have high potential fecundity. Therefore selection should favour an increase in body size given that these females get opportunities to realize their potential advantage by maturing and laying more eggs. However, ectotherm physiology is strongly temperature-dependent, and activities are carried out sufficiently only within certain temperature ranges. Thus it remains unclear if the fecundity advantage of a large size is fully realized in natural environments, where thermal conditions are limiting. * 2. Insect fecundity might be limited by temperature at two levels; first eggs need to mature, and then the female needs time for strategic ovipositing of the egg. Since a female cannot foresee the number of oviposition opportunities that she will encounter on a given day, the optimal rate of egg maturation will be governed by trade-offs associated with egg- and time-limited oviposition. As females of different sizes will have different amounts of body reserves, size-dependent allocation trade-offs between the mother"s condition and her egg production might be expected. * 3. In the temperate butterfly Pararge aegeria, the time and temperature dependence of oviposition and egg maturation, and the interrelatedness of these two processes were investigated in a series of laboratory experiments, allowing a decoupling of the time budgets for the respective processes. * 4. The results show that realized fecundity of this species can be limited by both the temperature dependence of egg maturation and oviposition under certain thermal regimes. Furthermore, rates of oviposition and egg maturation seemed to have regulatory effects upon each other. Early reproductive output was correlated with short life span, indicating a cost of reproduction. Finally, large females matured more eggs than small females when deprived of oviposition opportunities. Thus, the optimal allocation of resources to egg production seems dependent on female size. * 5. This study highlights the complexity of processes underlying rates of egg maturation and oviposition in ectotherms under natural conditions. We further discuss the importance of temperature variation for egg- vs. time-limited fecundity and the consequences for the evolution of female body size in insects.
Neben dem Habitatverlust gelten Konsequenzen der Habitatfragmentierung seit den 1990er Jahren als wesentliche Ursache der Gefaehrdung von Pflanzen und stehen damit nun auch im Fokus des botanischen Artenschutzes. Der vorliegende Beitrag gibt einen ueberblick ueber den Stand der populationsbiologischen und genetischen Forschung und versucht abzuschaetzen, welche Bedeutung Habitatfragmentierung und die dadurch entstehenden kleinen, isolierten Populationen auf heimische Pflanzenarten haben koennen. Als wesentliche und offenbar sehr weit verbreitete negative Effekte werden Zufallsereignisse, Randeffekte, Bestaeuberlimitierung, Gendrift und Inzuchtdepression identifiziert. Zusammen mit verringerter Habitatqualitaet durch Eutrophierung, Entwaesserung oder Nutzungsaenderung wirken sie zumeist negativ auf die Fitness der Individuen und Populationen und erhoehen so deren Aussterberisiko. Dieser negative Effekt kleiner Populationen auf die individuelle Fitness wird unabhaengig von der Ursache als Allee-Effekt bezeichnet. Eine durch einen Biotopverbund gefoerderte Metapopulationsdynamik kann das dauerhafte Aussterben von Pflanzenpopulationen verhindern und mindert die negativen genetischen Effekte der Habitatfragmentierung ueber einen erhoehten Genfluss durch Pollen und Samen. Die bisherigen wissenschaftlichen Studien in Mitteleuropa beruhen allerdings in ueberproportionaler Weise auf bestimmten Pflanzenfamilien (Gentianaceae, Primulaceae), Habitaten (Trocken- und Magerrasen, Wirtschaftsgruenland), insekten- und obligat fremdbestaeubten sowie weitgehend auf sexuelle Fortpflanzung angewiesenen Arten, waehrend etwa ueber Grasartige, Ruderalpflanzen, wind- und selbstbestaeubte sowie an vegetative Fortpflanzung angepasste Arten nur wenige Erkenntnisse vorliegen. Gerade diese und Pflanzenarten mit hohem Ausbreitungspotenzial muessen aber nach derzeitigem Wissensstand als weniger sensitiv gegenueber Habitatfragmentierung eingestuft werden. Auf diesen Befunden aufbauend werden fuer die Naturschutzpraxis Biotoptypen hinsichtlich ihrer Sensitivitaet gegenueber Habitatfragmentierung klassifiziert und ein auf biologischen Merkmalen basierender Kriterienkatalog zur Auswahl von Zielarten des Biotopverbunds vorgestellt. Schließlich wird eroertert, was bei Maßnahmen zur Regeneration kleiner bzw. bereits ausgestorbener Populationen zu beachten ist, und es werden allgemeine Folgerungen zur Ausgestaltung eines Biotopverbundskonzepts fuer Pflanzen gezogen.
Vegetation und Standort bodensaurer Buchenwälder am Arealrand : am Beispiel Mittelbrandenburgs

Different from NW Germany, the northern part of NE Germany and the "Hohe Flaeming" region, central Brandenburg is considered as being largely devoid of natural beech forests because of its subcontinental, dry climate. In the present study the vegetation ecology of beech forests of the region is comprehensively documented for the first time, and they are compared with NW German stands in Lower Saxony. In the study area beech forests are concentrated in the Berlin-Potsdam region along the Havel river lakes which is characterised by relatively high precipitation and a specific land use history. All belong to the Luzulo-Fagetum growing on acid soils. Four subtypes are distinguished according to nutrient availability and soil moisture. The central Brandenburg Luzulo-Fagetum does not markedly deviate from other beech forests in the northern German lowlands with respect to vegetation structure and edaphic subtypes. However, numerous indicator species for humid or moist conditions are less frequent than under atlantic climate conditions in the lowlands of Lower Saxony, a pattern occurring also in other forest communities. On the other hand, nitrogen and disturbance indicators are more frequent in central Brandenburg. As expected, podzolisation of the soils and humus accumulation is lower in beech forests under subcontinental climate, but surprisingly the soils are more sandy and thus drier. However, beech forests are lacking on south-exposed slopes, and they are notably occurring in northern exposition. A combined analysis of distribution patterns and climatic data, postglacial vegetation history and forest use history, and actual rejuvenation dynamics reveals that the present-day beech forests in central Brandenburg have to be considered as near-natural relics, which are currently spreading. The range of potentially natural beech forests is larger than assumed until now, but further on it is not clearly to define.
Die 11beta-HSD1 reguliert intrazellulär die Cortisolkonzentration durch Regeneration von Cortison z.B. aus dem Blutkreislauf, zu Cortisol. Daher stellt diese ein wichtiges Element in der Glucocorticoid-vermittelten Genregulation dar. Die 11beta-HSD1 wird ubiquitär exprimiert, auf hohem Niveau besonders in Leber, Fettgewebe und glatten Muskelzellen. Insbesondere die Bedeutung der 11beta-HSD1 in Leber und Fettgewebe konnte mehrfach nachgewiesen werden. In der Leber führte eine erhöhte Aktivität aufgrund einer Überexpression in Mäusen zu einer verstärkten Gluconeogeneserate. Des Weiteren konnte gezeigt werden, dass eine erhöhte Expression und erhöhte Enzymaktivität der 11beta-HSD1 im subkutanen und viszeralen Fettgewebe assoziiert ist mit Fettleibigkeit, Insulinresistenz und Dyslipidämie. Über die Regulation ist jedoch noch wenig bekannt. Zur Untersuchung der Promotoraktivität wurde der Promotorbereich von -3034 bis +188, vor und nach dem Translations- und Transkriptionsstart, der 11beta-HSD1 kloniert. 8 Promotorfragmente wurden mittels Dual-Luciferase-Assay in humanen HepG2-Zellen sowie undifferenzierten und differenzierten murinen 3T3-L1-Zellen untersucht. Anschließend wurde mittels nicht-radioaktiven EMSA die Bindung des TATA-Binding Proteins (TBP) sowie von CCAAT/Enhancer-Binding-Proteinen (C/EBP) an ausgewählte Promotorregionen analysiert. Nach der Charakterisierung des Promotors wurden spezifische endogene und exogene Regulatoren untersucht. Fettsäuren modifizieren die Entstehung von Adipositas und Insulinresistenz. Ihre Wirkung wird u.a. PPARgamma-abhängig vermittelt und kann durch das Inkretin (Glucose-dependent insulinotropic Peptide) GIP modifiziert werden. So wurden die Effekte von unterschiedlichen Fettsäuren, vom PPARgamma Agonisten Rosiglitazon sowie dem Inkretin GIP auf die Expression und Enzymaktivität der 11beta-HSD1 untersucht. Dies wurde in-vitro-, tierexperimentell und in humanen in-vivo-Studien realisiert. Zuletzt wurden 2 Single Nucleotide Polymorphismen (SNP) im Promotorbereich der 11beta-HSD1 in der Zellkultur im Hinblick auf potentielle Funktionalität analysiert sowie die Assoziation mit Diabetes mellitus Typ 2 und Körpergewicht in der MeSyBePo-Kohorte bei rund 1.800 Personen untersucht. Die Luciferase-Assays zeigten basal eine zell-spezifische Regulation der 11beta-HSD1, wobei in allen 3 untersuchten Zelltypen die Bindung eines Repressors nachgewiesen werden konnte. Zudem konnte eine mögliche Bindung des TBPs sowie von C/EBP-Proteinen an verschiedene Positionen gezeigt werden. Die Transaktivierungsassays mit den C/EBP-Proteinen -alpha, -beta und -delta zeigten eben-falls eine zellspezifische Regulation des 11beta-HSD1-Promotors. Die Aktivität und Expression der 11beta-HSD1 wurde durch die hier untersuchten endogenen und exogenen Faktoren spezifisch modifiziert, was sowohl in-vitro als auch in-vivo in unterschiedlichen Modellsystemen dargestellt werden konnte. Die Charakterisierung der MeSyBePo-Kohorte ergab keine direkten Assoziationen zwischen Polymorphismus und klinischem Phänotyp, jedoch Tendenzen für eine erhöhtes Körper-gewicht und Typ 2 Diabetes mellitus in Abhängigkeit des Genotyps. Der Promotor der 11beta-HSD1 konnte aufgrund der Daten aus den Luciferaseassays sowie den Daten aus den EMSA-Analysen näher charakterisiert werden. Dieser zeigt eine variable und zell-spezifische Regulation. Ein wichtiger Regulator stellen insbesondere in den HepG2-Zellen die C/EBP-Proteine -alpha, -beta und -delta dar. Aus den in-vivo-Studien ergab sich eine Regulation der 11beta-HSD1 durch endogene, exogene und pharmakologische Substanzen, die durch die Zellkulturversuche bestätigt und näher charakterisiert werden konnten.
Die vorliegende Untersuchung von Fledermaeusen in Sommer- und Winterquartieren liefert neue daten zur Verbreitung und Haeufigkeit von Ektoparasiten aus Brandenburg und anderen Bundeslaendern. An 11 Fledermausarten wurden insgesamt 7 Floharten (Ischnosyllidae), zwei Lausfliegenarten (Nycteribiidae) und zwei Wanzenarten (Cimicidae) nachgewiesen. Die Floharten Ischnopsyllus intermedius und die Artengruppe I. simplex/mysticus ergaenzen die Liste der aktuellen Funde fuer Brandenburg. Nach dem milden Winter 2006-7 wurden in einigen Winterquartieren Veraenderungen im Artenspektrum der Parasiten festgestellt. Nycteribia kolenatii an Myotis daubentoni und Nycteribia eusarca an Nyctalus noctula traten bei der vorliegenden Untersuchung relativ haeufig auf. Die anderen ektoparasitischen Floehe und Lausfliegen waren sehr selten zu finden. Die meisten Parasitenarten und die hoechste Befallsintensitaet wies die Wasserfledermaus (Myotis daubentoni) auf.
Auf dem Weg der genetischen Information stellt die Translation der RNA in eine Aminosäuresequenz den letzten Schritt dar. In Chloroplasten, den grünen Organellen der Pflanzenzellen, findet ein Großteil der Regulation der Genexpression auf Ebene der Initiation dieses Schrittes statt. Eine Vielzahl von Eigenschaften der RNA und von Faktoren, die an die RNA binden, entfalten einen Einfluss auf diesen Schritt. Bisher unvollständig aufgeklärt ist die Rolle einer konservierten Nukleotidsequenz in der untranslatierten Region der RNA -- der Shine-Dalgarno-Sequenz. Diese stellt in Bakterien, wie z.B. E. coli als Ribosomenbindestelle sicher, dass Ribosomen den Anfang der zu translatierenden Sequenz zuverlässig erkennen. Im Rahmen dieser Arbeit wurden diverse DNA-Konstrukte in Plastiden von Tabak eingebracht. Hierzu zählten Konstrukte, die sowohl eine erhöhte Anzahl von Ribosomenbindestellen enthielten als auch vermehrte Startpunkte der Translation. Zusätzlich wurden Konstrukte hergestellt, die die Situation von mehreren zu translatierenden Regionen in der RNA nachstellten. Es konnte festgestellt werden, dass plastidäre Ribosomen die strangaufwärts gelegenen Translationsstartpunkte bevorzugen -- im Gegensatz zu E. coli, wo alle Startpunkte gleichmäßig genutzt wurden. Hierdurch zeigten die prokaryotischen Ribosomen aus Chloroplasten, die sich aus bakteriellen Systemen ableiten, Eigenschaften von eukaryotischen Ribosomen. Ein zweites Teilprojekt dieser Arbeit beschäftigte sich mit der Inkompatibilität von Chloroplasten mit dem Kerngenom. In Kreuzungen von Arten der Gattung Pelargonium fielen Kombinationen auf, bei denen die Tochterpflanzen bleiche Blattbereiche bis hin zu vollständig weißen Pflanzen zeigten. Dieses Phänomen wird als Bastardbleichheit bezeichnet. In der Gattung Pelargonium werden Chloroplasten von beiden Elternteilen an die Tochterpflanzen vererbt. Da das Phänomen der Bastardbleichheit nur in einem der Plastiden vorkommt, nicht jedoch im anderen in der gleichen Pflanze, muss von einem Effekt ausgegangen werden, der von Plastiden ausgeht. Die Interaktionen zwischen Zellkern und Chloroplasten sind offensichtlich stark gestört. Zur detaillierten Untersuchung dieses Effekts wurde die Nukleotidsequenz von drei Chloroplastengenomen aufgeklärt. Es konnte eine Reihe von Sequenzunterschieden der Genome ermittelt werden. Aus diesen wurde eine Reihe von Unterschieden beobachtet, die einen solchen Effekt zur Folge haben können. Aus diesen Unterschieden wurde eine Reihe von potentiellen Kandidatengenen zusammengestellt, die in weiteren Arbeiten auf ihre Rolle in der Entstehung der Bastardbleichheit untersucht werden.

Plant population modelling has been around since the 1970s, providing a valuable approach to understanding plant ecology from a mechanistic standpoint. It is surprising then that this area of research has not grown in prominence with respect to other approaches employed in modelling plant systems. In this review, we provide an analysis of the development and role of modelling in the field of plant population biology through an exploration of where it has been, where it is now and, in our opinion, where it should be headed. We focus, in particular, on the role plant population modelling could play in ecological forecasting, an urgent need given current rates of regional and global environmental change. We suggest that a critical element limiting the current application of plant population modelling in environmental research is the trade-off between the necessary resolution and detail required to accurately characterize ecological dynamics pitted against the goal of generality, particularly at broad spatial scales. In addition to suggestions how to overcome the current shortcoming of data on the process-level we discuss two emerging strategies that may offer a way to overcome the described limitation: (1) application of a modern approach to spatial scaling from local processes to broader levels of interaction and (2) plant functional-type modelling. Finally we outline what we believe to be needed in developing these approaches towards a 'science of forecasting'.
Adenylates are metabolites with essential function in metabolism and signaling in all living organisms. As Cofactors, they enable thermodynamically unfavorable reactions to be catalyzed enzymatically within cells. Outside the cell, adenylates are involved in signalling processes in animals and emerging evidence suggests similar signaling mechanisms in the plants’ apoplast. Presumably, apoplastic apyrases are involved in this signaling by hydrolyzing the signal mediating molecules ATP and ADP to AMP. This PhD thesis focused on the role of adenylates on metabolism and development of potato (Solanum tuberosum) by using reverse genetics and biochemical approaches. To study the short and long term effect of cellular ATP and the adenylate energy charge on potato tuber metabolism, an apyrase from Escherichia coli targeted into the amyloplast was expressed inducibly and constitutively. Both approaches led to the identification of adaptations to reduced ATP/energy charge levels on the molecular and developmental level. These comprised a reduction of metabolites and pathway fluxes that require significant amounts of ATP, like amino acid or starch synthesis, and an activation of processes that produce ATP, like respiration and an immense increase in the surface-to-volume ratio. To identify extracellular enzymes involved in adenylate conversion, green fluorescent protein and activity localization studies in potato tissue were carried out. It was found that extracellular ATP is imported into the cell by an apoplastic enzyme complement consisting of apyrase, unspecific phosphatase, adenosine nucleosidase and an adenine transport system. By changing the expression of a potato specific apyrase via transgenic approaches, it was found that this enzyme has strong impact on plant and particular tuber development in potato. Whereas metabolite levels were hardly altered, transcript profiling of tubers with reduced apyrase activity revealed a significant upregulation of genes coding for extensins, which are associated with polar growth. The results are discussed in context of adaptive responses of plants to changes in the adenylate levels and the proposed role of apyrase in apoplastic purinergic signaling and ATP salvaging. In summary, this thesis provides insight into adenylate regulated processes within and outside non-photosynthetic plant cells.
KEPI is a protein kinase C-potentiated inhibitory protein for type 1 Ser/Thr protein phosphatases. We found no or reduced expression of KEPI in breast cancer cell lines, breast tumors and metastases in comparison to normal breast cell lines and tissues, respectively. KEPI protein expression and ubiquitous localization was detected with a newly generated antibody. Ectopic KEPI expression in MCF7 breast cancer cells induced differential expression of 95 genes, including the up-regulation of the tumor suppressors EGR1 (early growth response 1) and PTEN (phosphatase and tensin homolog), which is regulated by EGR1. We further show that the up-regulation of EGR1 in MCF7/KEPI cells is mediated by MEK-ERK signaling. The inhibition of this pathway by the MEK inhibitor UO126 led to a strong decrease in EGR1 expression in MCF7/KEPI cells. These results reveal a novel role for KEPI in the regulation of the tumor suppressor gene EGR1 via activation of the MEK-ERK MAPK pathway.
Oidium pedaliacearum sp. nov. (; O. sesami, nom. inval.) and Podosphaera macrospora comb. et stat. nov. (; Sphaerotheca alpina f. macrospora) are introduced, and the taxonomy and distribution of Erysiphe celosiae is discussed. New host species and new collections of Erysiphe cruciferarum (on Cleome hassleriana), E. flexuosa (on Aesculus hippocastanum), E. hedwigii (on Viburnum carlesii), E. heraclei (on Tinguarra montana), E. cf. macleayae (on Macleaya cordata), E. prunastri (on Prunus cerasifera), E. sedi (on Sedum aff. spectabilis), E. trifolii (on Trigonella caerulea), Golovinomyces cichoracearum (on Argyranthemum pinnatifidum subsp. succulentum), G. cf. hydrophyllacearum (on Nemophila menziesii), G. orontii (on Nolana spp.), G. cf. orontii (on Tiarella cordifolia), Neoerysiphe cumminsiana (on Bidens cf. ferulifolia), Oidium clitoriae (on Clitoria ternatea), O. cf. hortensiae (on Philadelphus coronarius), O. pedilanthi (on Pedilanthus tithymaloides), Oidium (Pseudoidium) sp. (on Utricularia alpina), Podosphaera sp. (on Bergia capensis), Sawadaea bicornis (on Acer platanoides) and S. tulasnei (on Acer ginnala and A. tatarica) are recorded from France, Germany, Greece and Mexico.
The effect of moderate rates of nitrogen deposition on ground floor vegetation is poorly predicted by uncontrolled surveys or fertilization experiments using high rates of nitrogen (N) addition. We compared the temporal trends of ground floor vegetation in permanent plots with moderate (7-13 kg/ha/yr) and lower bulk N deposition (4-6 kg/ha/yr) in southern Sweden during 1982-1998. We examined whether trends differed between growth forms (vascular plants and bryophytes) and vegetation types (three types of coniferous forest, deciduous forest, and bog). Trends of site-standardized cover and richness varied among growth forms, vegetation types, and deposition regions. Cover in spruce forests decreased at the same rate with both moderate and low deposition. In pine forests cover decreased faster with moderate deposition and in bogs cover decreased faster with low deposition. Cover of bryophytes in spruce forests increased at the same rate with both moderate and low deposition. In pine forests cover decreased faster with moderate deposition and in bogs and deciduous forests there was a strong non-linear increase with moderate deposition. The trend of number of vascular plants was constant with moderate and decreased with low deposition. We found no trend in the number of bryophyte species. We propose that the decrease of cover and number with low deposition was related to normal ecosystem development (increased shading), suggesting that N deposition maintained or increased the competitiveness of some species in the moderate-deposition region. Deposition had no consistent negative effect on vegetation suggesting that it is less important than normal successional processes.

Die Speicheldrüsen der Schmeißfliege Calliphora vicina produzieren bei Stimulierung mit dem Neurohormon Serotonin (5-Hydroxytryptamine, 5-HT) einen KCl-reichen Primärspeichel. Der transepitheliale K+-Transport wird durch eine apikal lokalisierte vakuoläre H+-ATPase (V-ATPase) energetisiert. Stimulierung der Speicheldrüsen mit 5-HT aktiviert die apikale V-ATPase, die Protonen aus der Zelle in das Drüsenlumen transportiert. Trotz des auswärts gerichteten Protonentransportes führt die 5-HT-Stimulierung kurioserweise zu einer intrazellulären Ansäuerung. Die Ursachen dieser 5-HT-induzierten Ansäuerung waren unzureichend untersucht. Deshalb war das Ziel dieser Arbeit die Identifikation aller Transporter, die an der intrazellulären pH-(pHi)-Regulation in unstimulierten Speicheldrüsen von Calliphora vicina beteiligt sind und an der Entstehung und Regulation der 5-HT-induzierten pHi-Änderungen mitwirken. Von besonderem Interesse war hierbei die funktionelle Mitwirkung der V-ATPase, deren Beteiligung an der pHi-Regulation in tierischen Zellen bisher wenig untersucht war. Wesentliche Ergebnisse dieser Arbeit waren: • Messungen des pHi-Wertes in der unstimulierten Drüse zeigten, dass vor allem die V-ATPase und mindestens ein Na+-abhängiger HCO3--Transporter an der Aufrechterhaltung des Ruhe-pHi beteiligt sind. • Zur Wiederherstellung des Ruhe-pHi nach einer intrazellulären Ansäuerung (NH4Cl-Vorpuls) tragen ebenfalls im Wesentlichen die V-ATPase und mindestens ein Na+-abhängiger HCO3--Transporter bei. Der Na+/H+-Antiporter hat in der unstimulierten Drüse keinen messbaren Einfluss auf den Ruhe-pHi. • Die Wiederherstellung des Ruhe-pHi nach einer intrazellulären Alkalisierung (Na-acetat-Vorpuls) ist Cl--abhängig, aber auch unter extremen Bedingungen waren die Zellen noch in der Lage sich vollständig von einer intrazellullären Alkalisierung zu erholen. Einen entscheidenden Anteil daran hat offenbar die hohe intrazelluläre Pufferkapazität. • Ein Na+-abhängiger Glutamat-Transporter ist per se kein pHi-regulierender Transporter, seine Aktivität hat jedoch Einfluss auf den Ruhe-pHi in der unstimulierten Speicheldrüse von Calliphora vicina. • 10 nM 5-HT induzieren in den Calliphora Speicheldrüsen eine intrazelluläre Ansäuerung. An dieser Ansäuerung ist der Na+/H+-Antiporter entscheidend beteiligt. Auch eine klare Cl--Abhängigkeit der 5-HT-induzierten Ansäuerung konnte beobachtet werden. Wahrscheinlich ist eine gekoppelte Aktivität von Na+/H+-Antiporter und Cl-/HCO3--Antiporter. • Messungen mit einem O2-empfindlichen Fluoreszenzfarbstoff zeigten, dass Stimulierung der Speicheldrüsen mit 5-HT die Zellatmung aktivierte. Der cAMP- und der IP3/Ca2+-Weg tragen auf komplexe Weise zu der 5-HT-induzierten Aktivierung der Zellatmung und damit auch zu den 5-HT-induzierten pHi-Änderungen bei. • Mit molekularbiologischen Untersuchungen ist es gelungen den Na+-abhängigen Glutamat-Transporter, den Na+/H+-Antiporter, die Carboanhydrase und die Untereinheit C der V-ATPase in den Calliphora Speicheldrüsen direkt nachzuweisen. Zudem konnte erstmals der direkte Nachweis für die Expression eines nH+/K+-Antiporters in den Speicheldrüsen von Calliphora vicina erbracht werden. Diese Arbeit trug ganz wesentlich zum Verständnis der pHi-Regulation in der unstimulierten und stimulierten Speicheldrüse von Calliphora vicina bei. Mechanismen die zur Aufrechterhaltung und Wiederherstellung des Ruhe-pHi nach einer intrazellulären Ansäuerung bzw. Alkalisierung beitragen, konnten mit pHi-Messungen und auch molekularbiologisch nachgewiesen werden. Die Mechanismen, welche die 5-HT-induzierte intrazelluläre Ansäuerung verursachen, konnten ebenfalls aufgeklärt werden. Zudem wurde an den Calliphora Speicheldrüsen eine neue optische Methode zur Messung des O2-Verbrauchs in tierischen Geweben etabliert.

Die vakuoläre Protonen-ATPase, kurz V-ATPase, ist ein multimerer Enzymkomplex, der in fast jeder eukaryotischen Zelle zu finden ist und den aktiven elektrogenen Transport von Protonen über Membranen katalysiert. Die Aktivität der V-ATPase ist essentiell für eine Vielzahl physiologischer Prozesse. Ein grundlegender Mechanismus zur Regulation der V-ATPase-Aktivität ist die reversible Dissoziation des Holoenzyms in den integralen VO-Komplex, der als Protonenkanal dient, und den cytosolischen V1-Komplex, der ATP hydrolysiert und somit den Protonentransport energetisiert. Die Untereinheit C, die im dissoziierten Zustand der V-ATPase als einzige Untereinheit isoliert im Cytoplasma vorliegt, scheint bei der Bildung des aktiven Holoenzyms eine Schlüsselrolle zu übernehmen. In den Speicheldrüsen der Schmeißfliege Calliphora vicina ist die V-ATPase an der Speichelsekretion beteiligt. In den sekretorischen Zellen wird die Bildung des V-ATPase-Holoenzyms in der apikalen Plasmamembran durch das Neurohormon Serotonin (5-HT) stimuliert. Der Effekt von 5-HT auf die V-ATPase wird intrazellulär durch die Proteinkinase A (PKA) vermittelt und hält nur für die Dauer der Stimulierung an. In der vorliegenden Arbeit wurde mittels Phosphoproteinfärbungen und 2D-Elektrophorese nachgewiesen, dass infolge einer Stimulierung der Drüsenzellen mit 5-HT die Untereinheit C der V-ATPase durch die PKA reversibel phosphoryliert wird. Die Phosphorylierung geht einher mit einer Umverteilung der Untereinheit C aus dem Cytoplasma zur apikalen Plasmamembran und der Bildung des aktiven Holoenzyms. Immuncytochemische Untersuchungen zeigten, dass die katalytische Untereinheit der PKA ebenfalls umverteilt wird und in stimulierten Zellen im Bereich der apikalen Plasmamembran konzentriert vorliegt. Um herauszufinden welche Proteinphosphatase der PKA entgegenwirkt, wurden luminale pH-Messungen durchgeführt und der Effekt von spezifischen Proteinphosphatase-Inhibitoren und veresterten Komplexbildnern zweiwertiger Kationen auf die V-ATPase-Aktivität untersucht. Diese Messungen führten zu der Schlussfolgerung, dass eine Proteinphosphatase des Typs 2C an der Inaktivierung der V-ATPase beteiligt ist. Mit weiteren Phosphoproteinfärbungen konnte gezeigt werden, dass die Dephosphorylierung der Untereinheit C ebenfalls durch eine Proteinphosphatase 2C katalysiert wird und dies vermutlich die Dissoziation des VO- und V1-Komplexes begünstigt. Darüber hinaus konnte durch luminale pH-Messungen und ergänzende biochemische Untersuchungen eine Calcineurin-vermittelte Modulation des cAMP/PKA-Signalweges durch den parallel aktivierten IP3/Ca2+-Signalweg und damit einhergehend eine Beeinflussung der V-ATPase-Aktivität durch den [Ca2+]-Spiegel nachgewiesen werden.
Understanding the interactions of predators and their prey and their responses to environmental changes is one of the striking features of ecological research. In this thesis, spring dynamics of phytoplankton and its consumers, zooplankton, were considered in dependence on the environmental conditions in a deep lake (Lake Constance) and a shallow marine water (mesocosms from Kiel Bight), using descriptive statistics, multiple regression models, and process-oriented dynamic simulation models. The development of the spring phytoplankton bloom, representing a dominant feature in the plankton dynamics in temperate and cold oceans and lakes, may depend on temperature, light, and mixing intensity, and the success of over-wintering phyto- and zooplankton. These factors are often correlated in the field. Unexpectedly, irradiance often dominated algal net growth rather than vertical mixing even in deep Lake Constance. Algal net losses from the euphotic layer to larger depth were induced by vertical mixing, but were compensated by the input from larger depth when algae were uniformly distributed over the water column. Dynamics of small, fast-growing algae were well predicted by abiotic variables, such as surface irradiance, vertical mixing intensity, and temperature. A simulation model additionally revealed that even in late winter, grazing may represent an important loss factor of phytoplankton during calm periods when losses due to mixing are small. The importance of losses by mixing and grazing changed rapidly as it depended on the variable mixing intensity. Higher temperature, lower global irradiance and enhanced mixing generated lower algal biomass and primary production in the dynamic simulation model. This suggests that potential consequences of climate change may partly counteract each other. The negative effect of higher temperatures on phytoplankton biomass was due to enhanced temperature-sensitive grazing losses. Comparing the results from deep Lake Constance to those of the shallow mesocosm experiments and simulations, confirmed the strong direct effect of light in contrast to temperature, and the importance of grazing already in early spring as soon as moderate algal biomasses developed. In Lake Constance, ciliates dominated the herbivorous zooplankton in spring. The start of ciliate net growth in spring was closely linked to that of edible algae, chlorophyll a and the vertical mixing intensity but independent of water temperature. The duration of ciliate dominance in spring was largely controlled by the highly variable onset of the phytoplankton bloom, and little by the less variable termination of the ciliate bloom by grazing of meta-zooplankton. During years with an extended spring bloom of algae and ciliates, they coexisted at relatively high biomasses over 15-30 generations, and internally forced species shifts were observed in both communities. Interception feeders alternated with filter feeders, and cryptomonads with non-cryptomonads in their relative importance. These dynamics were not captured by classical 1-predator-1-prey models which consistently predict pronounced predator-prey cycles or equilibria with either the predator or the prey dominating or suppressed. A multi-species predator-prey model with predator species differing in their food selectivity, and prey species in their edibility reproduced the observed patterns. Food-selectivity and edibility were related to the feeding and growth characteristics of the species, which represented ecological trade-offs. For example, the prey species with the highest edibility also had the highest maximum growth rate. Data and model revealed endogenous driven ongoing species alternations, which yielded a higher variability in species-specific biomasses than in total predator and prey biomass. This holds for a broad parameter space as long as the species differ functionally. A more sophisticated model approach enabled the simulation of a continuum of different functional types and adaptability of predator and prey communities to altered environmental conditions, and the maintenance of a rather low model complexity, i.e., low number of equations and free parameters. The community compositions were described by mean functional traits --- prey edibility and predator food-selectivity --- and their variances. The latter represent the functional diversity of the communities and thus, the potential for adaptation. Oscillations in the mean community trait values indicated species shifts. The community traits were related to growth and grazing characteristics representing similar trade-offs as in the multi-species model. The model reproduced the observed patterns, when nonlinear relationships between edibility and capacity, and edibility and food availability for the predator were chosen. A constant minimum amount of variance represented ongoing species invasions and thus, preserved a diversity which allows adaptation on a realistic time-span.
Nob1 (New Zealand obese 1) bezeichnet einen Adipositas-QTL auf Chr. 5 der Maus (LODBMI >3,3), der in einem Rückkreuzungsexperiment der Mausstämme NZO (adipös) und SJL (schlank) identifiziert wurde. Um Kandidatengene für Adipositas zu finden, wurden mehr als 300 Nob1-Transkripte mit Hilfe von Genexpressionsanalysen auf Unterschiede in stoffwechselrelevanten Geweben zwischen beiden Mausstämmen untersucht. Sieben Gene zeigten eine differentielle Expression: 2310045A20Rik, Tbc1d1, Ppp1cb, Mll5, Insig1, Abhd1 und Alox5ap. Die codierenden Bereiche dieser Gene wurden anschließend auf Sequenzunterschiede zwischen NZO und SJL untersucht. Nur im Gen Tbc1d1, das im Peak-Bereich des Nob1 lokalisiert ist, wurde eine SJL-spezifische Deletion von sieben Basen detektiert, die zu einer Leserasterverschiebung und einem vorzeitigen Abbruch des Proteins in der funktionellen Rab-GAP-Domäne führt (Loss-of-Function-Mutation). Interessanterweise wurde eine Variante von TBC1D1 (R125W) in Kopplungsanalysen mit Adipositas beim Menschen assoziiert (Stone et al., 2006). TBC1D1 zeigt eine hohe Homologie zu TBC1D4 (AS160), das im Insulinsignalweg eine wichtige Rolle spielt. In 17 weiteren Genen im Peak-Bereich des Nob1 wurde keine weitere SJL-spezifischen Mutation detektiert. Bei NZO-Tieren erfolgte die Tbc1d1-mRNA-Expression vorwiegend in glycolytischen Fasern des Skelettmuskels. Zudem wurden zwei gewebsspezifisch exprimierte Tbc1d1-Isoformen identifiziert, die sich durch alternatives Splicen der Exone 12 und 13 unterscheiden. Die im Rahmen dieser Arbeit gefundenen Ergebnisse machen Tbc1d1 zu einem plausiblen Kandidatengen für den Nob1-QTL. Welche Funktion Tbc1d1 im Glucose- und Fettstoffwechsel des Skelettmuskels hat, muss in weiteren Analysen untersucht werden.
The introduction of a new powdery mildew disease on Rhus hirta in various parts of Germany (Brandenburg, Rhine- Westphalia, Sachsen-Anhalt and Saxony) is reported. The anamorph found on this host agrees well with the North American Podosphaera pruinosa. Although the teleomorph has not yet been found in Germany and a molecular study has not yet been possible due to the lack of fresh North American material for a comparison, there is little doubt that the European outbreak of the Rhus powdery mildew disease may be referred to as Podosphaera pruinosa. Morphology, taxonomy and distribution of Podosphaera species on Rhus and other hosts of the Anacardiaceae are discussed in detail.
Large oligomeric proteins are usually thought to fold and assemble hierarchically: Domains fold and coalesce to form the subunits, and folded subunits can then associate to form the multimeric structure. We have investigated the refolding pathway of the ;-sheet protein pea seed lectin using spectroscopic and hydrodynamic techniques. In vivo, it is proteolytically processed post-translationally, so that the single-domain subunits of the initial homodimer themselves become heterodimers of intertwined fragment polypeptide chains. Despite this complex topology, mature pea seed lectin reassembles with considerable efficiency at low total protein concentration (10 ;g/mL) and low temperature (10 °C), albeit very slowly (t1/2 ; 2 days). Contrary to expectations, the primary assembly product is not the intact ;-sheet domain, but the larger fragment chains first dimerize to form the native-like subunit interface. The smaller fragment chains then associate with this preformed dimer.
Modeling the impacts of grazing land management on land-use change for the Jordan River region

In this article, we describe a simulation method for investigating the impacts of different grazing land management strategies on the productivity of (semi-)natural vegetation and the resulting feedback on land-use change. In a first application, we analyze the effects of sustainable and intensive grazing land management in the Jordan River region. For this purpose, we adapt and use the regional version of the spatially explicit modeling framework LandSHIFT. Our simulation experiments indicate that the modeled feedback mechanism has a strong effect on the spatial extent of grazing land. Consequently, the results of our study underline that the inclusion of such feedback mechanisms in land- use models can help to represent and analyze the complex interactions between humans and the environment in a more differentiated and realistic way, but they also identify the demand for more detailed empirical data on grazing land degradation in order to further improve the explanatory power of the model.
We have developed a microfluidic mixer optimized for rapid measurements of protein folding kinetics using synchrotron radiation circular dichroism (SRCD) spectroscopy. The combination of fabrication in fused silica and synchrotron radiation allows measurements at wavelengths below 220 nm, the typical limit of commercial instrumentation. At these wavelengths, the discrimination between the different types of protein secondary structure increases sharply. The device was optimized for rapid mixing at moderate sample consumption by employing a serpentine channel design, resulting in a dead time of less than 200 ;s. Here, we discuss the design and fabrication of the mixer and quantify the mixing efficiency using wide-field and confocal epi-fluorescence microscopy. We demonstrate the performance of the device in SRCD measurements of the folding kinetics of cytochrome c, a small, fast-folding protein. Our results show that the combination of SRCD with microfluidic mixing opens new possibilities for investigating rapid conformational changes in biological macromolecules that have previously been inaccessible.
Biosensoren werden oft für die Messung einzelner Substanzen in komplexen Medien verwendet, wie z.B. bei der Blutzuckerbestimmung. Sie bestehen aus einem physikochemischen Sensor, dem Transduktionselement, und einer darauf immobilisierten biologischen Komponente, dem Erkennungselement. In dieser Arbeit wurde als Transduktionselement eine Elektrode und als Biokomponente das Enzym „Meerrettich Peroxidase“ (engl. horseradish peroxidase, HRP) verwendet. Solche HRP-Elektroden werden für die Messung von Wasserstoffperoxid (H2O2) eingesetzt. H2O2 wird im Körper von weißen Blutkörperchen produziert, um Bakterien abzutöten, wird teilweise ausgeatmet und kann in kondensierter Atemluft nachgewiesen werden. Da viele weiße Blutkörperchen bei einer Chemotherapie abgetötet und dadurch die Patienten anfälliger für Infektionen werden, muss ihre Anzahl regelmäßig überwacht werden. Dazu wird zurzeit Blut abgenommen. Im ersten Teil dieser Arbeit wurde untersucht, ob eine Überwachung der Anzahl an weißen Blutkörperchen ohne Blutabnahme durch eine H2O2-Messung erfolgen kann. Ein direkter Zusammenhang zwischen der ausgeatmeten H2O2-Menge und der Zahl der weißen Blutkörperchen konnte dabei nicht festgestellt werden. Für empfindliche H2O2-Messungen mit einer HRP-Elektrode ist ein schneller Austausch von Elektronen zwischen der Elektrode und dem Enzym notwendig. Eine Vorraussetzung dafür ist eine kurze Distanz zwischen dem aktiven Zentrum des Enzyms und der Elektrodenoberfläche. Um einen kurzen Abstand zu erreichen wurden im zweiten Teil dieser Arbeit verschiedene poröse graphitähnliche Materialien aus pyrolysierten Kobalt-Porphyrinen für die Elektrodenherstellung verwendet. Dabei stellte sich heraus, dass eines der untersuchten Materialien, welches Poren von etwa der Größe eines Enzyms hat, Elektronen etwa 200mal schneller mit dem Enzym austauscht als festes Graphit. Die HRP selbst enthält in seinem aktiven Zentrum ein Eisen-Protoporphyrin, also ein aus vier Ringen bestehendes flaches Molekül mit einem Eisenatom im Zentrum. Reagiert die HRP mit H2O2, so entzieht es dem Peroxid zwei Elektronen. Eines dieser Elektronen wird am Eisen, das andere im Ringsystem zwischengespeichert, bevor sie an ein anderes Molekül oder an die Elektrode weitergegeben werden. Im letzten Teil dieser Arbeit wurde das Eisen durch Osmium ausgetauscht. Das so veränderte Enzym entzieht Peroxiden nur noch ein Elektron. Dadurch reagiert es zwar langsamer mit Wasserstoffperoxid, dafür aber schneller mit tert-Butylhydroperoxid, einem organischen Vertreter der Peroxid-Familie.
Growing recognition of the importance of long-distance dispersal (LDD) of plant seeds for various ecological and evolutionary processes has led to an upsurge of research into the mechanisms underlying LDD. We summarize these findings by formulating six generalizations stating that LDD is generally more common in open terrestrial landscapes, and is typically driven by large and migratory animals, extreme meteorological phenomena, ocean currents and human transportation, each transporting a variety of seed morphologies. LDD is often associated with unusual behavior of the standard vector inferred from plant dispersal morphology, or mediated by nonstandard vectors. To advance our understanding of LDD, we advocate a vector-based research approach that identifies the significant LDD vectors and quantifies how environmental conditions modify their actions.
Die vorliegende Dissertation behandelt die Ökologie von Cnidium dubium (Schkuhr) Thell. (Sumpf-Brenndolde), Gratiola officinalis L. (Gottes-Gnadenkraut) und Juncus atratus Krocker (Schwarze Binse), drei gefährdeten Arten, die als sogenannte Stromtalpflanzen in Mitteleuropa in ihrem Vorkommen eng an die Flussauen gebunden sind. Die Arbeit basiert auf verschiedenen Simulationsexperimenten und Feldstudien in der Unteren Havelniederung, einem „Feuchtgebiet von internationaler Bedeutung“. Sie behandelt Themenkomplexe wie das Samenbankverhalten, die Samenkeimung, die Stickstofflimitierung, die Konkurrenzkraft, das Verhalten der Pflanzen nach einer Sommertrockenheit und nach einer Winter/Frühjahrsüberflutung. Ferner widmet sie sich der Populationsbiologie der Arten und dem Verhalten der Pflanzen nach besonderen Störungsereignissen wie Mahd, Herbivorie und der Sommerflut 2002. Der Leser erfährt, wie die Pflanzen in verschiedenen Lebensphasen auf die auentypische Umwelt reagieren und erhält umfassende Einblicke in physiologische Mechanismen, die der Anpassung an die typischen Bedingungen einer mitteleuropäischen Flussaue dienen. Eine Interpretation der Ergebnisse zeigt auf, welche der spezifischen Eigenschaften zur Gefährdung der drei Stromtalarten beitragen. Die Arbeit ist für den Arten-, Biotop- und Landschaftsschutz interessant. Darüber hinaus bietet sie zahlreiche Anknüpfungspunkte zur ökophysiologischen Grundlagenforschung. Die verstärkte Nutzung physiologischer Methoden bei der Klärung ökologischer Fragestellungen wird angeregt.
Die ergonomische Anpassung von Produkten der körpernahen Umwelt an den menschlichen Körper in seiner gesamten Variabilität erfordert anthropometrische Grundlagen. Die vorliegende Arbeit beschreibt und analysiert die Körpermasse, 17 Längenmaße, 5 Skelettrobustizitätsmaße, 6 Korpulenzmaße, 3 Kopfmaße, 5 Handmaße, 3 Fußmaße, sowie 10 Beweglichkeitsmaße der Wirbelsäule, 8 Beweglichkeitsmaße der Hand, 2 Beweglichkeitsmaße des Beines und 7 Handkräfte von 295 Probanden der drei Altersgruppen 20 bis 29 Jahre, 50 bis 59 Jahre und 60 bis 69 Jahre. Die Untersuchungen wurden im Zeitraum von September 2006 bis April 2007 durchgeführt. Ziel der Arbeit ist es, für den überwiegenden Teil der untersuchten körperlichen Merkmale erstmals für die deutsche Bevölkerung geschlechts- und altersspezifische Mittelwerte und Variabilitätsbereiche bis zum vollendeten 70. Lebensjahr zur Verfügung zu stellen. Das gilt insbesondere für die untersuchten Beweglichkeitsmaße und Handkräfte. Erstmals werden Korrelationen zwischen der Körperform, wie sie sich im Maßzusammenhang der unterschiedlichen Körperbautypen darstellt, der Gelenkbeweglichkeit und den Handkräften vorgestellt. Darüber hinaus wird durch den Vergleich der Ergebnisse der jungen und der beiden älteren Erwachsenengruppen untersucht, welche Unterschiede zwischen den verschiedenen Altersgruppen bestehen. Im Hinblick auf die zeitliche Gültigkeit der aktuellen Untersuchungsergebnisse werden der Einfluss des säkularen Trends und der Einfluss der ontogenetischen Alternsprozesse auf Längenmaße und Korpulenzmaße diskutiert. Die Arbeit zeigt auf, dass innerhalb der untersuchten Probanden eine große Variationsbreite in den Körpermaßen auftritt. Es lassen sich typische Altersunterschiede erkennen. Die Älteren sind im Mittel kleiner, weisen jedoch größere Skelettrobustizitäts- und Korpulenzmaße auf. Die dynamischen Maße weisen auf eine geringere Beweglichkeit der Wirbelsäule, teilweise auch der Hand hin. Die Handkräfte der Frauen werden mit zunehmendem Alter geringer, bei den Männern sind die Älteren kräftiger als die jungen Erwachsenen. Die Ergebnisse deuten auf einen gegenüber früheren Generationen verzögerten Beginn von körperlichen Alterserscheinungen hin, der im Hinblick auf die steigende Lebenserwartung der Bevölkerung eingehender untersucht werden sollte.
Die Enzyme der Sulfotransferase-Gensuperfamilie (SULT) konjugieren nukleophile Gruppen von kleinen endogenen Verbindungen und Fremdstoffen mit der negativ geladenen Sulfo-Gruppe. Dadurch wird die Polarität dieser Verbindungen erhöht, ihre passive Permeation von Zellmembranen verhindert und somit ihre Ausscheidung erleichtert. Jedoch stellt die Sulfo-Gruppe in bestimmten chemischen Verbindungen eine gute Abgangsgruppen dar. Aus der Spaltung resultierende Carbenium- oder Nitreniumionen können mit DNA oder anderen zellulären Nukleophilen reagieren. In Testsystemen für Mutagenität wurden zahlreiche Verbindungen, darunter Nahrungsinhaltsstoffe und Umweltkontaminanten, durch SULT zu Mutagenen aktiviert. Dabei zeigten sich zum einen eine ausgeprägte Substratspezifität selbst orthologer SULT-Formen unterschiedlicher Spezies und zum anderen Interspezies-Unterschiede in der SULT-Gewebeverteilung. Daher könnten sich die Zielgewebe einer SULT-induzierten Krebsentstehung bei Mensch und Nager unterscheiden. Um die Beteiligung von humanen SULT an der Bioaktivierung von Fremdstoffen im Tiermodell untersuchen zu können, wurden transgene Mauslinien für den Cluster der humanen SULT1A1- und -1A2-Gene sowie für die humane SULT1B1 generiert. Zur Herstellung der transgenen Linien wurden große genomische Konstrukte verwendet, die die SULT-Gene sowie – zum Erreichen einer der Humansituation entsprechenden Gewebeverteilung der Proteinexpression – deren potentielle regulatorische Sequenzen enthielten. Es wurden je drei transgene Linien für hSULT1A1/hSULT1A2 und drei transgene Linien für hSULT1B1 etabliert. Die Expression der humanen Proteine konnte in allen Linien gezeigt werden und fünf der sechs Linien konnten zur Homozygotie bezüglich der Transgene gezüchtet werden. In der molekularbiologischen Charakterisierung der transgenen Linien wurde der chromosomale Integrationsort der Konstrukte bestimmt und die Kopienzahl pro Genom untersucht. Mit Ausnahme einer hSULT1A1/hSULT1A2-transgenen Linie, bei der Kopien des Konstrukts in zwei unterschiedliche Chromosomen integriert vorliegen, wiesen alle Linien nur einen Transgen-Integrationsort auf. Die Untersuchung der Transgen-Kopienzahl ergab, dass die Mauslinien zwischen einer und etwa 20 Kopien des Transgen-Konstrukts pro Genom trugen. In der proteinbiochemischen Charakterisierung wurde gezeigt, dass die transgenen Linien die humanen Proteine mit einer weitgehend der des Menschen entsprechenden Gewebeverteilung exprimieren. Die Intensität der im Immunblot nachgewiesenen Expression korrelierte mit der Kopienzahl der Transgene. Die zelluläre und subzelluläre Verteilung der Transgen-Expression wurden bei einer der hSULT1A1/hSULT1A2-transgenen Linien in Leber, Niere, Lunge, Pankreas, Dünndarm und Kolon und bei einer der hSULT1B1-transgenen Linien im Kolon untersucht. Sie stimmte ebenfalls mit der Verteilung der entsprechenden SULT-Formen im Menschen überein. Da sich die erzeugten transgenen Linien aufgrund ihrer mit dem Menschen vergleichbaren Gewebeverteilung der SULT-Expression als Modellsystem zur Untersuchung der menschlichen SULT-vermittelten metabolischen Aktivierung eigneten, wurde eine der hSULT1A1/hSULT1A2-transgenen Linien für zwei erste toxikologische Untersuchungen eingesetzt. Den Mäusen wurden chemische Verbindungen verabreicht, für die in in-vitro-Versuchen eine hSULT1A1/hSULT1A2-vermittelte Bioaktivierung zu Mutagenen gezeigt worden war. In beiden Untersuchungen wurde die Gewebeverteilung der entstandenen DNA-Addukte als Endpunkt einer gewebespezifischen genotoxischen Wirkung ermittelt. In der ersten Untersuchung wurden 90 mg/kg Körpergewicht 2-Amino-1-methyl-6-phenylimidazo[4,5-b]pyridin – ein in gebratenem Fleisch gebildetes heterozyklisches aromatisches Amin – transgenen sowie Wildtyp-Mäusen oral verabreicht. Acht Stunden nach Applikation wiesen die transgenen Mäuse signifikant höhere Adduktniveaus als die Wildtyp-Mäuse in Leber, Lunge, Niere, Milz und Kolon auf. In der Leber der transgen Mäuse war das Adduktniveau 17fach höher als in der Leber der Wildtyp-Mäuse. Die Leber war bei den transgenen Tieren das Organ mit dem höchsten, bei den Wildtyp-Tieren hingegen mit dem niedrigsten DNA-Adduktniveau. In der zweiten Untersuchung (Pilotstudie mit geringer Tierzahl) wurde transgenen und Wildtyp-Mäusen 19 mg/kg Körpergewicht des polyzyklischen aromatischen Kohlenwasserstoffs 1-Hydroxymethylpyren – ein Metabolit der Nahrungs- und Umweltkontaminante 1-Methylpyren – intraperitoneal verabreicht. Nach 30 Minuten wurden, verglichen mit den Wildtyp-Mäusen, bis zu 25fach erhöhte Adduktniveaus bei den transgenen Mäusen in Leber, Niere, Lunge und Jejunum nachgewiesen. Somit konnte anhand einer in dieser Arbeit generierten transgenen Mauslinie erstmals gezeigt werden, dass die Expression der humanen SULT1A1/hSULT1A2 tatsächlich sowohl auf die Stärke als auch die Zielgewebe der DNA-Adduktbildung in vivo eine Auswirkung hat.
The tailspike protein P22 recognizes an octasaccharide derived from the O-antigen polysaccharide of Salmonella enteritidis in a shallow groove and molecular docking successfully identifies this binding region on the protein surface. Analysis by 2D 1H,1H-T-ROESY and transferred NOESY NMR experiments indicate that the bound octasaccharide ligand has a conformation similar to that observed in solution. The results from a saturation transfer difference NMR experiment show that a large number of protons in the octasaccharide are in close contact with the protein as a result of binding. A comparison of the crystal structure of the complex and a molecular dynamics simulation of the octasaccharide with explicit water molecules suggest that only minor conformational changes are needed upon binding to the tailspike protein.
Monitoring animal populations in changing environments is crucial to wildlife conservation and management, but restrictions in resources are a recurring problem for wildlife managers and researchers throughout Africa. Land-use- induced shrub encroachment in Kalahari savannah rangelands has led to fragmentation of the landscape. Mammalian carnivores are particularly vulnerable to local extinction in fragmented landscapes, but their low numbers and their often nocturnal and secretive habits make them difficult to monitor. In this study, we tested the applicability of a passive tracking method and compared two measurement methods and index calculations for monitoring small carnivores across a grazing gradient in the southern Kalahari. During the four years of monitoring in a five-year period, we used the knowledge of indigenous Khoisan Bushmen for the identification of carnivore tracks on 640 sand transects (5 m x 250 m). Our results showed that this simple and inexpensive observation method enabled detailed monitoring of 10 small carnivore species across the grazing gradient. A binary index calculated an index based on presence/absence of a species' tracks on each transect, whereas the track intrusion index used the number of track intrusions to each transect for each species in its calculations. For less common species, the two indices were similar in trend and magnitude, because the number of intrusions to each transect was typically 1 or 0. Usually, the two indices showed relatively strong correlations. However, species with patchy distributions of higher numbers presented difficulties for the binary index to monitor trends, but not for the track intrusion index.

Zur Detektion neuer IgE- reaktiver Proteine wurde in dieser Arbeit ein zweidimensionales Proteintrennverfahren verwendet. Resultierende Proteinfraktionen wurden mithilfe von 18 tomatensensibiliesierten Patientenseren im Immunoblot getestet. Detektierte Proteine in der SDS-PAGE wurden mittels LC-MS/MS identifiziert. Dadurch konnten 2 Tomatensamenproteine, die im Immunoblot ein IgE- reaktives Signal zeigten eindeutig mittels Massenspektrometrie identifiziert werden. Diese Proteine sind Legumin und Vicilin. Durch Sequenzabgleich und Proteinstrukturmodellierung im Vergleich zu bereits bekannten Allergenen (Erdnuss und Cashewnuss), konnte eine hohe Homologie gezeigt werden.
In order to function properly, organisms have a complex control mechanism, in which a given gene is expressed at a particular time and place. One way to achieve this control is to regulate the initiation of transcription. This step requires the assembly of several components, i.e., a basal/general machinery common to all expressed genes, and a specific/regulatory machinery, which differs among genes and is the responsible for proper gene expression in response to environmental or developmental signals. This specific machinery is composed of transcription factors (TFs), which can be grouped into evolutionarily related gene families that possess characteristic protein domains. In this work we have exploited the presence of protein domains to create rules that serve for the identification and classification of TFs. We have modelled such rules as a bipartite graph, where families and protein domains are represented as nodes. Connections between nodes represent that a protein domain should (required rule) or should not (forbidden rule) be present in a protein to be assigned into a TF family. Following this approach we have identified putative complete sets of TFs in plant species, whose genome is completely sequenced: Cyanidioschyzon merolae (red algae), Chlamydomonas reinhardtii (green alga), Ostreococcus tauri (green alga), Physcomitrella patens (moss), Arabidopsis thaliana (thale cress), Populus trichocarpa (black cottonwood) and Oryza sativa (rice). The identification of the complete sets of TFs in the above-mentioned species, as well as additional information and reference literature are available at http://plntfdb.bio.uni-potsdam.de/. The availability of such sets allowed us performing detailed evolutionary studies at different levels, from a single family to all TF families in different organisms in a comparative genomics context. Notably, we uncovered preferential expansions in different lineages, paving the way to discover the specific biological roles of these proteins under different conditions. For the basic leucine zipper (bZIP) family of TFs we were able to infer that in the most recent common ancestor (MRCA) of all green plants there were at least four bZIP genes functionally involved in oxidative stress and unfolded protein responses that are bZIP-mediated processes in all eukaryotes, but also in light-dependent regulations. The four founder genes amplified and diverged significantly, generating traits that benefited the colonization of new environments. Currently, following the approach described above, up to 57 TF and 11 TR families can be identified, which are among the most numerous transcription regulatory families in plants. Three families of putative TFs predate the split between rhodophyta (red algae) and chlorophyta (green algae), i.e., G2-like, PLATZ, and RWPRK, and may have been of particular importance for the evolution of eukaryotic photosynthetic organisms. Nine additional families, i.e., ABI3/VP1, AP2-EREBP, ARR-B, C2C2-CO-like, C2C2-Dof, PBF-2-like/Whirly, Pseudo ARR-B, SBP, and WRKY, predate the split between green algae and streptophytes. The identification of putative complete list of TFs has also allowed the delineation of lineage-specific regulatory families. The families SBP, bHLH, SNF2, MADS, WRKY, HMG, AP2-EREBP and FHA significantly differ in size between algae and land plants. The SBP family of TFs is significantly larger in C. reinhardtii, compared to land plants, and appears to have been lost in the prasinophyte O. tauri. The families bHLH, SNF2, MADS, WRKY, HMG, AP2-EREBP and FHA preferentially expanded with the colonisation of land, and might have played an important role in this great moment in evolution. Later, after the split of bryophytes and tracheophytes, the families MADS, AP2-EREBP, NAC, AUX/IAA, PHD and HRT have significantly larger numbers in the lineage leading to seed plants. We identified 23 families that are restricted to land plants and that might have played an important role in the colonization of this new habitat. Based on the list of TFs in different species we have started to develop high-throughput experimental platforms (in rice and C. reinhardtii) to monitor gene expression changes of TF genes under different genetic, developmental or environmental conditions. In this work we present the monitoring of Arabidopsis thaliana TFs during the onset of senescence, a process that leads to cell and tissue disintegration in order to redistribute nutrients (e.g. nitrogen) from leaves to reproductive organs. We show that the expression of 185 TF genes changes when leaves develop from half to fully expanded leaves and finally enter partial senescence. 76% of these TFs are down-regulated during senescence, the remaining are up-regulated. The identification of TFs in plants in a comparative genomics setup has proven fruitful for the understanding of evolutionary processes and contributes to the elucidation of complex developmental programs.
Plants are the primary producers of biomass and thereby the basis of all life. Many varieties are cultivated, mainly to produce food, but to an increasing amount as a source of renewable energy. Because of the limited acreage available, further improvements of cultivated species both with respect to yield and composition are inevitable. One approach to further progress in developing improved plant cultivars is a systems biology oriented approach. This work aimed to investigate the primary metabolism of the model plant A.thaliana and its relation to plant growth using quantitative genetics methods. A special focus was set on the characterization of heterosis, the deviation of hybrids from their parental means for certain traits, on a metabolic level. More than 2000 samples of recombinant inbred lines (RILs) and introgression lines (ILs) developed from the two accessions Col-0 and C24 were analyzed for 181 metabolic traces using gas-chromatography/ mass-spectrometry (GC-MS). The observed variance allowed the detection of 157 metabolic quantitative trait loci (mQTL), genetic regions carrying genes, which are relevant for metabolite abundance. By analyzing several hundred test crosses of RILs and ILs it was further possible to identify 385 heterotic metabolic QTL (hmQTL). Within the scope of this work a robust method for large scale GC-MS analyses was developed. A highly significant canonical correlation between biomass and metabolic profiles (r = 0.73) was found. A comparable analysis of the results of the two independent experiments using RILs and ILs showed a large agreement. The confirmation rate for RIL QTL in ILs was 56 % and 23 % for mQTL and hmQTL respectively. Candidate genes from available databases could be identified for 67 % of the mQTL. To validate some of these candidates, eight genes were re-sequenced and in total 23 polymorphisms could be found. In the hybrids, heterosis is small for most metabolites (< 20%). Heterotic QTL gave rise to less candidate genes and a lower overlap between both populations than was determined for mQTL. This hints that regulatory loci and epistatic effects contribute to metabolite heterosis. The data described in this thesis present a rich source for further investigation and annotation of relevant genes and may pave the way towards a better understanding of plant biology on a system level.
The vacuolar H+-ATPase (V-ATPase) in the apical membrane of blowfly (Calliphora vicina) salivary gland cells energizes the secretion of a KCl-rich saliva in response to the neurohormone serotonin (5-HT). We have shown previously that exposure to 5-HT induces a cAMP-mediated reversible assembly of V-0 and V-1 subcomplexes to V-ATPase holoenzymes and increases V-ATPase-driven proton transport. Here, we analyze whether the effect of cAMP on V-ATPase is mediated by protein kinase A (PKA) or exchange protein directly activated by cAMP (Epac), the cAMP target proteins that are present within the salivary glands. Immunofluorescence microscopy shows that PKA activators, but not Epac activators, induce the translocation of V1 components from the cytoplasm to the apical membrane, indicative of an assembly of V-ATPase holoenzymes. Measurements of transepithelial voltage changes and microfluorometric pH measurements at the luminal surface of cells in isolated glands demonstrate further that PKA-activating cAMP analogs increase cation transport to the gland lumen and induce a V-ATPase-dependent luminal acidification, whereas activators of Epac do not. Inhibitors of PKA block the 5-HT-induced V-1 translocation to the apical membrane and the increase in proton transport. We conclude that cAMP exerts its effects on V-ATPase via PKA.
This paper describes the principle of a homogeneous indirect fluorescence quenching immunoassay that uses monoclonal antibodies. It is a carrier-free assay system that is performed completely in solution. The assay system was established for the determination of a low molecular weight substance (hapten), the herbicide diuron, used as a model analyte. A fluorescein-monuron conjugate together with a fluorescence-quenching monoclonal anti-fluorescein antibody and an anti-analyte antibody (here an anti-diuron/monuron monoclonal antibody) were used as central components of the assay. The fluorescein-monuron conjugate can be bound either by the anti-fluorescein monoclonal antibody or by the anti-diuron/ monuron monoclonal antibody. Due to steric hindrance, binding of both antibodies to the conjugate was not possible at the same time. By selecting the antibody concentrations appropriately, a dynamic equilibrium can be established that permits the preferential binding of the anti-diuron/monuron antibody to the conjugate, which allows the fluorescein in the conjugate to fluoresce. This equilibrium can be easily altered by adding free analyte (diuron), which competes with the conjugate to bind to the anti-diuron/monuron antibody. A reduction of anti-diuron/monuron antibody binding to the conjugate results in an increase in the binding of the anti-fluorescein antibody, which leads to a decrease in the fluorescence of the conjugate. The fluorescence is therefore a direct indicator of the state of equilibrium of the system and thus also the presence of free unconjugated analyte. The determination of an analyte based on this test principle does not require any washing steps. After the test components are mixed, the dynamic equilibrium is rapidly reached and the results can be obtained in less than 5 min by measuring the fluorescence of the fluorescein. We used this test principle for the determination of diuron, which was demonstrated for concentrations of approximately 5 nM.
Die Xanthin-Dehydrogenase aus Rhodobacter capsulatus ist ein cytoplasmatisches Enzym, welches ein (αβ)₂ Heterotetramer mit einer Größe von 275 kDa bildet. Die drei Kofaktoren (Moco, 2[2Fe2S], FAD) sind auf zwei unterschiedlichen Polypeptidketten gebunden. So sind die beiden spektroskopisch unterscheidbaren Eisen-Schwefel-Zentren und das FAD in der XdhA-Untereinheit und der Moco in der XdhB-Untereinheit gebunden. Im ersten Teil dieser Arbeit sollte untersucht werden, warum die R. capsulatus XDH ein Dimer bildet und ob ein intramolekularer Elektronentransfer existiert. Dafür wurde eine chimäre XDH-Variante [(α)₂(β₁wt/β₂E730A)] erzeugt, welche eine aktive und eine inaktive XdhB-Untereinheit trägt. Mit Hilfe von Reduktionsspektren sowie mit der Bestimmung der kinetischen Parameter für die Substrate Xanthin und NAD+ konnte gezeigt werden, dass die chimäre XDH-Variante katalytisch halb so aktiv war, wie der auf gleiche Weise gereinigte XDH-Wildtyp. Dies verdeutlicht, dass die noch aktive Untereinheit der Chimären selbstständig und unabhängig Substrat binden und hydroxylieren kann und ein intramolekularer Elektronentransfer zwischen den beiden XdhB-Untereinheiten nicht stattfindet. Ein weiteres Ziel war die funktionelle Charakterisierung der Mus musculus AOX1 sowie der humanen AOX1 hinsichtlich ihrer Substratspezifitäten und ihrer biophysikalischen Eigenschaften sowie der Charakterisierung der konservierten Aminosäuren im aktiven Zentrum der mAOX1. Da bislang noch kein heterologes Expressionssystem für ein aktives und stabiles rekombinantes AO-Protein existierte, wurde ein E. coli Expressionssystem mit der gleichzeitigen Expression der entsprechenden Mocosulfurase für mAOX1 und hAOX1 in dieser Arbeit etabliert. Mit Hilfe dieser Koexpression konnte die Aktivität der rekombinanten mAOX1 um 50 % gesteigert werden, wenn gleich auch der sulfurierte Moco-Anteil nur 20 % betrug. Um die konservierten Aminosäuren im aktiven Zentrum hinsichtlich ihrer Funktion der Substratbindung zu charakterisieren, wurden folgende Varianten erzeugt: V806E, M884R, V806/M884R sowie E1265Q. Mit Hilfe von kinetischen Substratuntersuchungen konnte gezeigt werden, dass die beiden Aminosäuren Val806 und Met884 für die Erkennung und die Stabilisierung von Aldehyden und N-Heterozyklen essentiell sind. Ein Austausch dieser beiden gegen Glutamat bzw. Arginin (wie bei R. capsulatus XDH) zeigte jedoch keine Xanthin- oder Hypoxanthinumsetzung. Für das Glu1265 wurde ebenfalls die Rolle als die Katalyse initiierende Aminosäure belegt.
In dieser Arbeit wird die Entwicklung eines bifunktionellen Biosensors nach dem Vorbild eines Baukastensystems beschrieben. Das Ziel wird durch die Kombination verschiedenster molekularer Erkennungselemente erreicht. Solche molekularen Erkennungselemente im verwendeten System sind: • Propidium und die periphere anionische Bindungsstelle der Acetylcholinesterase (AChE) • Organophosphate und das aktive Zentrum der AChE • ein an die AChE gekoppeltes Hapten und das Epitop eines Antikörpers • ein an die AChE gekoppeltes Hapten, das als Ligand ein weiteres Enzym bindet Neben dem molekularen Erkennungselement wird ein Biosensor ebenso durch die Art des Transducers charakterisiert. Hier werden Quarzplättchen mit Goldelektroden zur Signalumwandlung eingesetzt. Die Verwendung solcher Sensoren mit einem EQCM-Gerät (electrochemical quartz crystal microbalance) ermöglicht es zwei Messsignale gleichzeitig aufzunehmen: die piezoelektrische Bestimmung einer Massebeladung und die amperometrische Detektion von Enzymaktivität auf der Sensoroberfläche. Für die Analytik stehen somit zwei verschiedene Assay-Varianten zur Verfügung: die Bestimmung der Inhibition der ACHE-Aktivität und ein Bindungstest über das Hapten. Die Basis beider Tests ist die Modifizierung der piezoelektrischen Kristalle mit Propidium – einem reversiblen Inhibitor der Acetylcholinesterase. Dies ermöglicht die Beladung des Sensors mit AChE über die Wechselwirkung mit der peripheren anionischen Bindungsstelle des Enzyms. Die Aktivität der so immobilisierten AChE und die Inhibition durch Organophosphate (Pestizide) werden amperometrisch bestimmt. Durch die chemische Kopplung eines Hapten an die Cholinesterase wird ein weiteres Erkennungselement eingeführt. Das eröffnet die Möglichkeit, an die auf dem Propidium-modifizierten Sensor immobilisierte, haptenisierte Cholinesterase einen Antikörper zu binden. Als Voraussetzung für elektrochemische Bestimmung der AChE-Aktivität wurde zunächst die Optimierung der amperometrischen Messmethode vorgenommen. Die Oxidatationspotentiale für die Detektion von Thiocholin wurden im Bereich von 150 mV bis 300 mV variiert. Dabei wurde für die nachfolgenden Untersuchungen eine Arbeitspotential von 200 mV (vs. Ag/AgCl) festgelegt, da hier das beste Verhältnis von gemessenem Oxidationsstrom und Langzeitstabilität der Propidium-modifizierten Sensoren erzielt wurde. Dieses Potential war deutlich geringer als die bisher publizierten Mediator-freien AChE-Biosensoren. Es wurde ein Vergleich verschiedener Organophosphate über ihre Inhibitionskonstanten durchgeführt, um diejenigen herauszufinden, die möglichst schnell mit dem aktiven Zentrum der Acetylcholinesterase reagieren. Das verwendete Messsystem beruht nicht auf der Vorinkubation der AChE und damit einer Einstellung des Inhibitionsgleichgewichts. Stattdessen wurde die Inhibition der AChE direkt im Fließsystem verfolgt. Daher war eine schnelle Inhibitionskinetik für einen empfindlichen Organophosphat-Nachweis erforderlich. Da einige Inhibitoren nur als Phosphothionat vorlagen, wurde die Überführung dieser Substanzen in die entsprechenden Oxo-Formen mittels N-Bromsuccinimid untersucht. Die NBS-Aktivierung wurde erfolgreich durchgeführt, die erwartete Inhibitionsstärke konnte jedoch aufgrund hydrolytischer Vorgänge nicht erreicht werden. Untersuchungen mit Diisopropylfluorophosphat (DFP) und Chlorpyriphos-oxon (CPO) konnten die Voruntersuchungen über die Inhibitionskinetik in Bezug auf die erreichten Nachweisgrenzen von 2E-06 M für DFP und 5E-08 M für CPO bestätigen. Für die chemische Modifizierung der Acetylcholinesterase wurde zunächst 2,4-Dichlorphenoxyessigsäure (2,4-D) als Hapten ausgewählt. 2,4-D wird als Herbizid eingesetzt und in der EU über die Gewässerschutzrichtlinie reguliert. 2,4-D konnte in unterschiedlichen molaren Verhältnissen von 2,6 : 1 bis 260 : 1 (2,4-D : AChE) nach Aktivierung mit einem Norbornendicarboximido-Derivat an die AChE gekoppelt werden. Dabei konnte die spezifische Aktivität der Acetylcholinesterase erhalten und die Bindung eines anti-2,4-D-Antikörpers ermöglicht werden. Zur Verstärkung des piezolelektrischen Signals der Antikörperbindung wurden die Immunoglobuline zunächst an Goldnanopartikel gekoppelt. Damit konnte eine Verstärkung um den Faktor 10 erreicht werden. Allerdings waren die Antikörper-modifizierten Goldnanopartikel nicht langzeitstabil. Daher wurden auch Silica-Nanopartikel als Matrix für die Antikörperkopplung getestet. Mit diesem System konnte eine Verstärkung um den Faktor von 5 bis 13 je nach Grad der Beladung den Nanopartikel mit Antikörper bestimmt werden. Die hohe unspezifische Bindung der Antikörper-Nanopartikel-Konjugate an den Propidium-modifizierten QCM-Sensor konnte keinen empfindlichen 2,4-D-Nachweis ermöglichen. Als Alternative wurde Kokain (Benzoylecgonin, BZE) als Hapten an die Aceytlcholinesterase gekoppelt. Da Kokain selbst auch als Inhibitor im aktiven Zentrum der AChE binden kann, wurden zwei verschiedene Strategien zur Konjugatsynthese verfolgt. Durch Zugabe von Kokain während der Kopplung sollte die kovalente Fixierung des Kokain-Derivats BZE-DADOO im aktiven Zentrum verhindert werden (Konjugat B). In der Tat konnten mit dieser Synthesestrategie 67% der spezifischen Cholinesterase-Aktivität erhalten werden, während im Kokain-freien Ansatz (Konjugat A) nur 2% der Ausgangsaktivität wiedergefunden wurden. Das BZE-AChE-Konjugat ermöglichte auch die Untersuchung der Bindungskinetik der anti-BZE-Antikörper. Dabei konnte eine Assoziationsgeschwindigkeitskonstante ka von 12911 l/(mol•s) berechnet werden. Dieser Wert ist trotz der vergleichsweise geringen Oberflächenbeladung vergleichbar mit den in der Literatur angegebenen Werten. Die Dissoziationsgeschwindigkeitskonstante ist mit 2,89E−3 1/s um den Faktor 30 höher als der Literaturwert. Diese Abweichung ist auf Unterschiede im Bindungsmodell zurückzuführen. Mit beiden BZE-AChE-Konjugaten konnte ein kompetetiver Immunoassay mit Kokain im Fließsystem durchgeführt werden. Dabei zeigte sich für beide Konjugate ein ähnlicher Testmittelpunkt: IC50 = 4,40E−8 mol/l für Konjugat A bzw. IC50 = 1,77E−8 mol/l für Konjugat B. Diese Werte sind vergleichbar zu bereits publizierten Kokainassays im Fließsystem. Wie vorstehend beschrieben, bindet Kokain als Inhibitor auch im aktiven Zentrum von Cholinesterasen. Diese Eigenschaft wurde genutzt, um ein zweites Enzym – Butyrylcholinesterase (BChE) – an die BZE-AChE zu binden. Die Spezifität dieser Bindung konnte durch die Abwesenheit einer Affinität der BChE zum Propidium und durch die Blockierbarkeit der Bindung von BChE und BZE-AChE durch Kokain nachgewiesen werden. Damit konnte erfolgreich die Kombination mehrere molekularer Erkennungselemente demonstriert werden. Die Propidium-Plattform ermöglicht den Aufbau einer Architektur aus verschiedenen Cholinesterasen, die über unterschiedliche Bindungsstellen wechselwirken. Sowohl freie als auch BZE-modifizierte AChE können über die Affinität zum Propidium auf dem EQCM-Sensor immobilisiert werden. Mit Kokain als Substrat der Butyrylcholinesterase kann Benzoylecgonin nicht nur als Epitop für die Bindung eines Antikörpers, sondern auch als Erkennungselement für die BChE genutzt werden. Auf der anderen Seite erschwert die geringe Affinität der BChE im Gegensatz zum anti-BZE-Antikörper den Einsatz dieses Systems für analytische Zwecke. Durch die Verwendung anderer Ligand-Enzym-Kombinationen läßt sich das in dieser Arbeit vorgestellte Konzept noch weiter ausbauen und ermöglicht damit eine Entwicklung ausgehend von „einfachen“ molekularen Erkennungselementen (MRE) hin zu „multifunktionellen“ Erkennungselementsystemen. In dieser Arbeit konnte demonstriert werden, dass der Aufbau solch komplexe Systeme möglich ist, ohne Abstriche in Bezug auf die Empfindlichkeit der einzelnen Assays hinzunehmen.
Background To improve the understanding of consequences of climate change for annual plant communities, I used a detailed, grid-based model that simulates the effect of daily rainfall variability on individual plants in five climatic regions on a gradient from 100 to 800 mm mean annual precipitation (MAP). The model explicitly considers moisture storage in the soil. I manipulated daily rainfall variability by changing the daily mean rain (DMR, rain volume on rainy days averaged across years for each day of the year) by ± 20%. At the same time I adjusted intervals appropriately between rainy days for keeping the mean annual volume constant. In factorial combination with changing DMR I also changed MAP by ± 20%. Results Increasing MAP generally increased water availability, establishment, and peak shoot biomass. Increasing DMR increased the time that water was continuously available to plants in the upper 15 to 30 cm of the soil (longest wet period, LWP). The effect of DMR diminished with increasing humidity of the climate. An interaction between water availability and density-dependent germination increased the establishment of seedlings in the arid region, but in the more humid regions the establishment of seedlings decreased with increasing DMR. As plants matured, competition among individuals and their productivity increased, but the size of these effects decreased with the humidity of the regions. Therefore, peak shoot biomass generally increased with increasing DMR but the effect size diminished from the semiarid to the mesic Mediterranean region. Increasing DMR reduced via LWP the annual variability of biomass in the semiarid and dry Mediterranean regions. Conclusion More rainstorms (greater DMR) increased the recharge of soil water reservoirs in more arid sites with consequences for germination, establishment, productivity, and population persistence. The order of magnitudes of DMR and MAP overlapped partially so that their combined effect is important for projections of climate change effects on annual vegetation.

Conformational changes associated with the assembly of recombinant ;2-microglobulin in vitro under acidic conditions were investigated using infrared spectroscopy and static and dynamic light scattering. In parallel, the morphology of the different aggregated species obtained under defined conditions was characterized by electron microscopy. The initial salt-induced aggregate form of ;2-microglobulin, composed of small oligomers (dimers to tetramers), revealed the presence of ;-strands organized in an intramolecular-like fashion. Further particle growth was accompanied by the formation of intermolecular ;-sheet structure and led to short curved forms. An increase in temperature by only 25 °C was able to disaggregate these assemblies, followed by the formation of longer filamentous structures. In contrast, a rise in temperature up to 100 °C was associated with a reorganization of the short curved forms at the level of secondary structure and the state of assembly, leading to a species with a characteristic infrared spectrum different from those of all the other aggregates observed before, suggesting a unique overall structure. The infrared spectral features of this species were nearly identical to those of ;2-microglobulin assemblies formed at low ionic strength with agitation, indicating the presence of fibrils, which was confirmed by electron microscopy. The observed spectroscopic changes suggest that the heat-triggered conversion of the short curved assemblies into fibrils involves a reorganization of the ;-strands from an antiparallel arrangement to a parallel arrangement, with the latter being characteristic of amyloid fibrils of ;2-microglobulin.
Natürliche Standorte der Waldkiefer gibt es in Deutschland nur kleinflächig. Während Kiefernforste anstelle natürlicher Laubwälder heute oft landschaftsprägend sind, bildet die konkurrenzschwache und lichtbedürftige Kiefer ausschließlich auf extrem trockenen oder nassen, nährstoffarmen Standorten naturnahe Schlusswaldgesellschaften. Regionale Schwerpunkte liegen in subkontinentalen Regionen wie dem nordostdeutschen Tiefland und Bayern, ein „natürliches Kiefernareal" lässt sich aber kaum abgrenzen. An der Trockengrenze des Waldes finden sich auf Kalk- und Dolomitgesteinen artenreiche Karbonat-Trockenkiefernwälder mit Elementen der alpinen Rasen und Kalkmagerrasen in der Bodenvegetation. Diese Wälder besiedeln steile, südexponierte Felsen und morphodynamisch aktive Bereiche wie Rutschhänge und FlussSchotterböden im Umkreis der Alpen, kommen aber auch in den Mittelgebirgen vor. Ihr Gegenstück auf sauren Standorten sind die Sand- und Silikat-Kiefernwälder der Quarzsande und Sandstein-Verwitterungsböden, deren Bodenvegetation durch Zwergsträucher, Moose und Strauchflechten geprägt ist. Hier siedelt die Kiefer in den Tieflagen besonders auf Binnendünen und Sandern, aber auch auf Küstendünen der Ostsee, in den Mittelgebirgen z. B. auf den Sandsteinriffen der Sächsischen Schweiz. Der dritte Wuchsbereich natürlicher Kiefernwälder sind saure, nährstoffarme Moore, die ganz überwiegend von Regenwasser gespeist werden. Auch die Kiefern-Moorwälder sind in Nordostdeutschland und Bayern am häufigsten. Von diesen Standorten ausgehend, wo ihr Platz kaum von anderen Baumarten streitig gemacht wird, tritt die Waldkiefer immer wieder als Pionier auf weniger extremen Standorten auf. In der Naturlandschaft kam dies etwa nach Waldbränden oder Stürmen vor, doch der Mensch förderte die Kiefer durch Auflichtung der Wälder, Waldweide und Streunutzung stark. Auch die damit verbundene Nährstoffverarmung macht eine exakte Abgrenzung natürlicher Kiefernstandorte unmöglich. Die schlechtwüchsigen und forstwirtschaftlich nicht interessanten, ästhetisch aber sehr ansprechenden natürlichen Kiefernbestände sind heute vor allem durch Stickstoff-Immissionen gefährdet. Trotz ihrer oft kargen Erscheinung besitzen sie einen hohen Wert für die Biodiversität und den Artenschutz. Neben bodenbewohnenden Flechten und regionalen Relikt-Endemiten ist vor allem die in den letzten Jahrzehnten zunehmend gefährdete Vielfalt an Mykorrhiza-Pilzen hervorzuheben, die der Kiefer das Leben auf extrem nährstoffarmen Standorten überhaupt ermöglichen. Abschließend werden mögliche Schutz- bzw. Regenerationsmaßnahmen wie das Abplaggen flechtenreicher Kiefernstandorte vorgestellt.
Bacteriophage HK620 infects Escherichia coli H and is closely related to Shigella phage Sf6 and Salmonella phage P22. All three Podoviridae recognize and cleave their respective host cell receptor polysaccharide by homotrimeric tailspike proteins. The three proteins exhibit high sequence identity in the 110 residues of their N-terminal particle- binding domains, but no apparent sequence similarity in their major, receptor-binding parts. We have biochemically characterized the receptor-binding part of HK620 tailspike and determined its crystal structure to 1.38 Å resolution. Its major domain is a right-handed parallel ;-helix, as in Sf6 and P22 tailspikes. HK620 tailspike has endo-N- acetylglucosaminidase activity and produces hexasaccharides of an O18A1-type O-antigen. As indicated by the structure of a hexasaccharide complex determined at 1.6 Å resolution, the endoglycosidase-active sites are located intramolecularly, as in P22, and not between subunits, as in Sf6 tailspike. In contrast, the extreme C-terminal domain of HK620 tailspike forms a ;-sandwich, as in Sf6 and unlike P22 tailspike. Despite the different folds, structure-based sequence alignments of the C-termini reveal motifs conserved between the three proteins. We propose that the tailspike genes of P22, Sf6 and HK620 have a common precursor and are not mosaics of unrelated gene fragments.
