Objective: Hormone secretion from metabolically active tissues, such as pancreatic islets, is governed by specific and highly regulated signaling pathways. Defects in insulin secretion are among the major causes of diabetes. The molecular mechanisms underlying regulated insulin secretion are, however, not yet completely understood. In this work, we studied the role of the GTPase ARFRP1 on insulin secretion from pancreatic 13-cells. <br /> Methods: A 13-cell-specific Arfrp1 knockout mouse was phenotypically characterized. Pulldown experiments and mass spectrometry analysis were employed to screen for new ARFRP1-interacting proteins. Co-immunoprecipitation assays as well as super-resolution microscopy were applied for validation. <br /> Results: The GTPase ARFRP1 interacts with the Golgi-associated PDZ and coiled-coil motif-containing protein (GOPC). Both proteins are co localized at the trans-Golgi network and regulate the first and second phase of insulin secretion by controlling the plasma membrane localization of the SNARE protein SNAP25. Downregulation of both GOPC and ARFRP1 in Min6 cells interferes with the plasma membrane localization of SNAP25 and enhances its degradation, thereby impairing glucose-stimulated insulin release from 13-cells. In turn, overexpression of SNAP25 as well as GOPC restores insulin secretion in islets from 13-cell-specific Arfrp1 knockout mice. <br /> Conclusion: Our results identify a hitherto unrecognized pathway required for insulin secretion at the level of trans-Golgi sorting. Understanding the key factors influencing the water quality of large river systems forms an important basis for the assessment and protection of cross-regional ecosystems and the implementation of adapted water management concepts. However, identifying these factors requires in-depth comprehension of the unique environmental systems, which can only be achieved by detailed water quality monitoring.
Within the scope of the joint science and sports event "Elbschwimmstaffel" (swimming relay on the river Elbe) in June/July 2017 organized by the German Ministry of Education and Research, water quality data were acquired along a 550 km long stretch of the Elbe River in Germany. During the survey, eight physiochemical water quality parameters were recorded in high spatial and temporal resolution with the BIOFISH multisensor system. Multivariate statistical methods were applied to identify and delineate processes influencing the water quality.
The BIOFISH dataset revealed that phytoplankton activity has a major impact on the water quality of the Elbe River in the summer months. The results suggest that phytoplankton biomass constitutes a substantial proportion of the suspended particles and that photosynthetic activity of phytoplankton is closely related to significant temporal changes in pH and oxygen saturation.
An evaluation of the BIOFISH data based on the combination of statistical analysis with weather and discharge data shows that the hydrological and meteorological history of the sampled water body was the main driver of phytoplankton dynamics. This study demonstrates the capacity of longitudinal river surveys with the BIOFISH or similar systems for water quality assessment, the identification of pollution sources and their utilization for online in situ monitoring of rivers.
Background: In the animal kingdom body size is often linked to dominance and subsequently the standing in social hierarchy. Similarly, human growth has been associated and linked to socioeconomic factors, including one’s social status. This has already been proposed in the early 1900s where data on young German school girls from different social strata have been compared.
Objectives: This paper aims to summarize and analyze these results and make them accessible for non-German speakers. The full English translation of the historic work of Dikanski (Dikanski, 1914) is available as a supplement. Further, this work aims to compare the historical data with modern references, to test three hypotheses: (1) higher social class is positively associated with body height and weight, (2) affluent people from the used historical data match modern references in weight and height and (3) weight distributions are skewed in both modern and historical populations.
Methods: Comparison of historical data from 1914 with WHO and 1980s German data. The data sets, for both body weight and height for 6.0- and 7.0-year-old girls, were fitted onto centile curves and quantile correlation coefficients were calculated.
Results: In historical data social status is positively associated with body height and weight while both are also normally distributed, which marks a significant difference to modern references.
Conclusion: Social status is positively associated with height, signaling social dominance, making children of affluent classes taller. Children from the historical data do not reach the average height of modern children, even under the best environmental conditions. The children of the upper social class were not skewed in weight distribution, although they had the means to become as obese as modern children.
“Jewish, Gay and Proud”

This publication examines the foundation and institutional integration of the first gay-lesbian synagogue Beth Chayim Chadashim, which was founded in Los Angeles in 1972. As early as June 1974, the synagogue was admitted to the Union of American Hebrew Congregations, the umbrella organization of the Reform congregations in the United States. Previously, the potential acceptance of a congregation by and for homosexual Jews triggered an intense and broad debate within Reform Judaism. The work asks how it was possible to successfully establish a gay-lesbian synagogue at a time when homosexual acts were considered unnatural and contrary to tradition by almost the entire Jewish community. The starting point of the argumentation is, in addition to general changes in American synagogues after World War II, the assumption that Los Angeles was the most suitable place for this foundation. Los Angeles has an impressive queer history and the Jewish community was more open, tolerant and innovative here than its counterpart on the East Coast. The Metropolitan Community Church was also founded in the city, and as the largest religious institution for homosexual Christians, it also served as the birthplace of queer synagogues.
Reform Judaism was chosen as the place of institutional integration of the community because a relative openness for such an endeavor was only seen here. Responsa written in response to a potential admission of Beth Chayim Chadashim can be used to understand the arguments and positions of rabbis and psychologists regarding homosexuality and communities for homosexual Jews in the early 1970s.
Ultimately, the commitment and dedication of the congregation and its heterosexual supporters convinced the decision-makers in Reform Judaism. The decisive impulse to question the situation of homosexual Jews in Judaism came from Los Angeles. With its analysis, the publication contributes to the understanding of Queer Jewish History in general and queer synagogues in particular.

Responding to the poor performance of generic automated sentiment analysis solutions on domain-specific texts, we collect a dataset of 10,000 tweets discussing the topics of finance and investing. We manually assign each tweet its market sentiment, i.e., the investor’s anticipation of a stock’s future return. Using this data, we show that all existing sentiment models trained on adjacent domains struggle with accurate market sentiment analysis due to the task’s specialized vocabulary. Consequently, we design, train, and deploy our own sentiment model. It outperforms all previous models (VADER, NTUSD-Fin, FinBERT, TwitterRoBERTa) when evaluated on Twitter posts. On posts from a different platform, our model performs on par with BERT-based large language models. We achieve this result at a fraction of the training and inference costs due to the model’s simple design. We publish the artifact as a python library to facilitate its use by future researchers and practitioners.
Increasing greenhouse gas emissions are likely to impact not only natural systems but economies worldwide. If these impacts alter future economic development, the financial losses will be significantly higher than the mere direct damages. So far, potentially aggravating investment responses were considered negligible. Here we consistently incorporate an empirically derived temperature-growth relation into the simple integrated assessment model DICE. In this framework we show that, if in the next eight decades varying temperatures impact economic growth as has been observed in the past three decades, income is reduced by similar to 20% compared to an economy unaffected by climate change. Hereof similar to 40% are losses due to growth effects of which similar to 50% result from reduced incentive to invest. This additional income loss arises from a reduced incentive for future investment in anticipation of a reduced return and not from an explicit climate protection policy. Under economically optimal climate-change mitigation, however, optimal investment would only be reduced marginally as mitigation efforts keep returns high.
Earth’s surface temperature will continue to rise for another 20 to 30 years even with the strongest carbon emission reduction currently considered. The associated changes in rainfall patterns can result in an increased flood risk worldwide. We compute the required increase in flood protection to keep high-end fluvial flood risk at present levels. The analysis is carried out worldwide for subnational administrative units. Most of the United States, Central Europe, and Northeast and West Africa, as well as large parts of India and Indonesia, require the strongest adaptation effort. More than half of the United States needs to at least double their protection within the next two decades. Thus, the need for adaptation to increased river flood is a global problem affecting industrialized regions as much as developing countries.
Investigation of processes that contribute to the maintenance of genomic stability is one crucial factor in the attempt to understand mechanisms that facilitate ageing. The DNA damage response (DDR) and DNA repair mechanisms are crucial to safeguard the integrity of DNA and to prevent accumulation of persistent DNA damage. Among them, base excision repair (BER) plays a decisive role. BER is the major repair pathway for small oxidative base modifications and apurinic/apyrimidinic (AP) sites. We established a highly sensitive non-radioactive assay to measure BER incision activity in murine liver samples. Incision activity can be assessed towards the three DNA lesions 8-oxo-2’-deoxyguanosine (8-oxodG), 5-hydroxy-2’-deoxyuracil (5-OHdU), and an AP site analogue. We applied the established assay to murine livers of adult and old mice of both sexes. Furthermore, poly(ADP-ribosyl)ation (PARylation) was assessed, which is an important determinant in DDR and BER. Additionally, DNA damage levels were measured to examine the overall damage levels. No impact of ageing on the investigated endpoints in liver tissue were found. However, animal sex seems to be a significant impact factor, as evident by sex-dependent alterations in all endpoints investigated. Moreover, our results revealed interrelationships between the investigated endpoints indicative for the synergetic mode of action of the cellular DNA integrity maintaining machinery.
Enterprise solutions, specifically enterprise systems, have allowed companies to integrate enterprises’ operations throughout. The integration scope of enterprise solutions has increasingly widened, now often covering customer activities, activities along supply chains, and platform ecosystems. IS research has contributed a wide range of explanatory and design knowledge dealing with this class of IS. During the last two decades, many technological as well as managerial/organizational innovations extended the affordances of enterprise solutions—but this broader scope also challenges traditional approaches to their analysis and design. This position paper presents an enterprise-level (i.e., cross-solution) perspective on IS, discusses the challenges of complexity and coordination for IS design and management, presents selected enterprise-level insights for IS coordination and governance, and explores avenues towards a more comprehensive body of knowledge on this important level of analysis.
The effects of exercise interventions on unspecific chronic low back pain (CLBP) have been investigated in many studies, but the results are inconclusive regarding exercise types, efficiency, and sustainability. This may be because the influence of psychosocial factors on exercise induced adaptation regarding CLBP is neglected. Therefore, this study assessed psychosocial characteristics, which moderate and mediate the effects of sensorimotor exercise on LBP. A single-blind 3-arm multicenter randomized controlled trial was conducted for 12-weeks. Three exercise groups, sensorimotor exercise (SMT), sensorimotor and behavioral training (SMT-BT), and regular routines (CG) were randomly assigned to 662 volunteers. Primary outcomes (pain intensity and disability) and psychosocial characteristics were assessed at baseline (M1) and follow-up (3/6/12/24 weeks, M2-M5). Multiple regression models were used to analyze whether psychosocial characteristics are moderators of the relationship between exercise and pain, meaning that psychosocial factors and exercise interact. Causal mediation analysis were conducted to analyze, whether psychosocial characteristics mediate the exercise effect on pain. A total of 453 participants with intermittent pain (mean age = 39.5 ± 12.2 years, f = 62%) completed the training. It was shown, that depressive symptomatology (at M4, M5), vital exhaustion (at M4), and perceived social support (at M5) are significant moderators of the relationship between exercise and the reduction of pain intensity. Further depressive mood (at M4), social-satisfaction (at M4), and anxiety (at M5 SMT) significantly moderate the exercise effect on pain disability. The amount of moderation was of clinical relevance. In contrast, there were no psychosocial variables which mediated exercise effects on pain. In conclusion it was shown, that psychosocial variables can be moderators in the relationship between sensorimotor exercise induced adaptation on CLBP which may explain conflicting results in the past regarding the merit of exercise interventions in CLBP. Results suggest further an early identification of psychosocial risk factors by diagnostic tools, which may essential support the planning of personalized exercise therapy.
