We previously showed that purified 1-methoxy-3-indolylmethyl (1-MIM) glucosinolate, a secondary plant metabolite in Brassica species, is mutagenic in various in vitro systems and forms DNA and protein adducts in mouse models. In the present study, we administered 1-MIM glucosinolate in a natural matrix to mice, by feeding a diet containing pak choi powder and extract. Groups of animals were killed after 1, 2, 4 and 8 days of pak choi diet, directly or, in the case of the 8-day treatment, after 0, 8 and 16 days of recovery with pak choi-free diet. DNA adducts [N-2-(1-MIM)-dG, N-6-(1-MIM)-dA] in six tissues, as well as protein adducts [tau N-(1-MIM)-His] in serum albumin (SA) and hemoglobin (Hb) were determined using UPLC-MS/MS with isotopically labeled internal standards. None of the samples from the 12 control animals under standard diet contained any 1-MIM adducts. All groups receiving pak choi diet showed DNA adducts in all six tissues (exception: lung of mice treated for a single day) as well as SA and Hb adducts. During the feeding period, all adduct levels continuously increased until day 8 (in the jejunum until day 4). During the 14-day recovery period, N-2-(1-MIM)-dG in liver, kidney, lung, jejunum, cecum and colon decreased to 52, 41, 59, 11, 7 and 2%, respectively, of the peak level. The time course of N-6-(1-MIM)-dA was similar. Immunohistochemical analyses indicated that cell turnover is a major mechanism of DNA adduct elimination in the intestine. In the same recovery period, protein adducts decreased more rapidly in SA than in Hb, to 0.7 and 37%, respectively, of the peak level, consistent with the differential turnover of these proteins. In conclusion, the pak choi diet lead to the formation of high levels of adducts in mice. Cell and protein turnover was a major mechanism of adduct elimination, at least in gut and blood.
Abdominal and general adiposity are independently associated with mortality, but there is no consensus on how best to assess abdominal adiposity. We compared the ability of alternative waist indices to complement body mass index (BMI) when assessing all-cause mortality. We used data from 352,985 participants in the European Prospective Investigation into Cancer and Nutrition (EPIC) and Cox proportional hazards models adjusted for other risk factors. During a mean follow-up of 16.1 years, 38,178 participants died. Combining in one model BMI and a strongly correlated waist index altered the association patterns with mortality, to a predominantly negative association for BMI and a stronger positive association for the waist index, while combining BMI with the uncorrelated A Body Shape Index (ABSI) preserved the association patterns. Sex-specific cohort-wide quartiles of waist indices correlated with BMI could not separate high-risk from low-risk individuals within underweight (BMI<18.5 kg/m(2)) or obese (BMI30 kg/m(2)) categories, while the highest quartile of ABSI separated 18-39% of the individuals within each BMI category, which had 22-55% higher risk of death. In conclusion, only a waist index independent of BMI by design, such as ABSI, complements BMI and enables efficient risk stratification, which could facilitate personalisation of screening, treatment and monitoring.

Among them, base excision repair (BER) plays a decisive role. BER is the major repair pathway for small oxidative base modifications and apurinic/apyrimidinic (AP) sites. We established a highly sensitive non-radioactive assay to measure BER incision activity in murine liver samples. Incision activity can be assessed towards the three DNA lesions 8-oxo-2’-deoxyguanosine (8-oxodG), 5-hydroxy-2’-deoxyuracil (5-OHdU), and an AP site analogue. We applied the established assay to murine livers of adult and old mice of both sexes. Furthermore, poly(ADP-ribosyl)ation (PARylation) was assessed, which is an important determinant in DDR and BER. Additionally, DNA damage levels were measured to examine the overall damage levels. No impact of ageing on the investigated endpoints in liver tissue were found. However, animal sex seems to be a significant impact factor, as evident by sex-dependent alterations in all endpoints investigated. Moreover, our results revealed interrelationships between the investigated endpoints indicative for the synergetic mode of action of the cellular DNA integrity maintaining machinery.

In order to assess the individual trace element status of humans for either medical or scientific purposes, amongst others, blood serum levels are determined. Furthermore, animal models are used to study interactions of trace elements. Most published methods require larger amounts (500-1000 mu L) of serum to achieve a reliable determination of multiple trace elements. However, oftentimes, these amounts of serum cannot be dedicated to a single analysis and the amount available for TE-determination is much lower. Therefore, a published ICP-MS/MS method for trace element determination in serum was miniaturized, optimized and validated for the measurement of Mn, Fe, Cu Zn, I and Se in as little as 50 mu L of human and murine serum and is presented in this work. For validation, recoveries of multiple LOTs and levels from commercially available human reference serum samples were determined, infra- and inter-day variations were assessed and limits of detection and quantification determined. It is shown, that the method is capable of giving accurate and reproducible results for all six elements within the relevant concentration ranges for samples from humans living in central Europe as well as from laboratory mice. As a highlight, the achieved limits of detection and quantification for Mn were found to be at 0.02 mu g/L serum and 0.05 mu g/L serum, respectively, while using an alkaline diluent for the parallel determination of iodine.
Zinc is an essential trace element, making it crucial to have a reliable biomarker for evaluating an individual’s zinc status. The total serum zinc concentration, which is presently the most commonly used biomarker, is not ideal for this purpose, but a superior alternative is still missing. The free zinc concentration, which describes the fraction of zinc that is only loosely bound and easily exchangeable, has been proposed for this purpose, as it reflects the highly bioavailable part of serum zinc. This report presents a fluorescence-based method for determining the free zinc concentration in human serum samples, using the fluorescent probe Zinpyr-1. The assay has been applied on 154 commercially obtained human serum samples. Measured free zinc concentrations ranged from 0.09 to 0.42 nM with a mean of 0.22 ± 0.05 nM. It did not correlate with age or the total serum concentrations of zinc, manganese, iron or selenium. A negative correlation between the concentration of free zinc and total copper has been seen for sera from females. In addition, the free zinc concentration in sera from females (0.21 ± 0.05 nM) was significantly lower than in males (0.23 ± 0.06 nM). The assay uses a sample volume of less than 10 µL, is rapid and cost-effective and allows us to address questions regarding factors influencing the free serum zinc concentration, its connection with the body’s zinc status, and its suitability as a future biomarker for an individual’s zinc status.
ABCB1/4 gallbladder cancer risk variants identified in India also show strong effects in Chileans

Background: The first large-scale genome-wide association study of gallbladder cancer (GBC) recently identified and validated three susceptibility variants in the ABCB1 and ABCB4 genes for individuals of Indian descent. We investigated whether these variants were also associated with GBC risk in Chileans, who show the highest incidence of GBC worldwide, and in Europeans with a low GBC incidence.
Methods: This population-based study analysed genotype data from retrospective Chilean case-control (255 cases, 2042 controls) and prospective European cohort (108 cases, 181 controls) samples consistently with the original publication.
Results: Our results confirmed the reported associations for Chileans with similar risk effects. Particularly strong associations (per-allele odds ratios close to 2) were observed for Chileans with high Native American (=Mapuche) ancestry. No associations were noticed for Europeans, but the statistical power was low.
Conclusion: Taking full advantage of genetic and ethnic differences in GBC risk may improve the efficiency of current prevention programs.

Macrophages have important protective functions during infection with herpes simplex virus type 1 (HSV-1). However, molecular mechanisms that restrict viral propagation and protect from severe disease are unclear. Here we show that macrophages take up HSV-1 via endocytosis and transport the virions into multivesicular bodies (MVBs). In MVBs, acid ceramidase (aCDase) converts ceramide into sphingosine and increases the formation of sphingosine-rich intraluminal vesicles (ILVs). Once HSV-1 particles reach MVBs, sphingosine-rich ILVs bind to HSV-1 particles, which restricts fusion with the limiting endosomal membrane and prevents cellular infection. Lack of aCDase in macrophage cultures or in Asah1(-/-) mice results in replication of HSV-1 and Asah1(-/-) mice die soon after systemic or intravaginal inoculation. The treatment of macrophages with sphingosine enhancing compounds blocks HSV-1 propagation, suggesting a therapeutic potential of this pathway. In conclusion, aCDase loads ILVs with sphingosine, which prevents HSV-1 capsids from penetrating into the cytosol.

Farber disease is a rare lysosomal storage disorder resulting from acid ceramidase deficiency and subsequent ceramide accumulation. No treatments for Farber disease are clinically available, and affected patients have a severely shortened lifespan. We have recently reported a novel acid ceramidase deficiency model that mirrors the human disease closely. Acid sphingomyelinase is the enzyme that generates ceramide upstream of acid ceramidase in the lysosomes. Using our acid ceramidase deficiency model, we tested if acid sphingomyelinase could be a potential novel therapeutic target for the treatment of Farber disease. A number of functional acid sphingomyelinase inhibitors are clinically available and have been used for decades to treat major depression. Using these as a therapeutic for Farber disease, thus, has the potential to improve central nervous symptoms of the disease as well, something all other treatment options for Farber disease can’t achieve so far. As a proof-of-concept study, we first cross-bred acid ceramidase deficient mice with acid sphingomyelinase deficient mice in order to prevent ceramide accumulation. Double-deficient mice had reduced ceramide accumulation, fewer disease manifestations, and prolonged survival. We next targeted acid sphingomyelinase pharmacologically, to test if these findings would translate to a setting with clinical applicability. Surprisingly, the treatment of acid ceramidase deficient mice with the acid sphingomyelinase inhibitor amitriptyline was toxic to acid ceramidase deficient mice and killed them within a few days of treatment. In conclusion, our study provides the first proof-of-concept that acid sphingomyelinase could be a potential new therapeutic target for Farber disease to reduce disease manifestations and prolong survival. However, we also identified previously unknown toxicity of the functional acid sphingomyelinase inhibitor amitriptyline in the context of Farber disease, strongly cautioning against the use of this substance class for Farber disease patients
Objective: We aimed to identify the role of the enzyme acid sphingomyelinase in the aging of stored units of packed red blood cells (pRBCs) and subsequent lung inflammation after transfusion.
Summary Background Data: Large volume pRBC transfusions are associated with multiple adverse clinical sequelae, including lung inflammation. Microparticles are formed in stored pRBCs over time and have been shown to contribute to lung inflammation after transfusion.
Methods: Human and murine pRBCs were stored with or without amitriptyline, a functional inhibitor of acid sphingomyelinase, or obtained from acid sphingomyelinase-deficient mice, and lung inflammation was studied in mice receiving transfusions of pRBCs and microparticles isolated from these units.
Results: Acid sphingomyelinase activity in pRBCs was associated with the formation of ceramide and the release of microparticles. Treatment of pRBCs with amitriptyline inhibited acid sphingomyelinase activity, ceramide accumulation, and microparticle production during pRBC storage. Transfusion of aged pRBCs or microparticles isolated from aged blood into mice caused lung inflammation. This was attenuated after transfusion of pRBCs treated with amitriptyline or from acid sphingomyelinase-deficient mice.
Conclusions: Acid sphingomyelinase inhibition in stored pRBCs offers a novel mechanism for improving the quality of stored blood.
Acid sphingomyelinase mediates murine acute lung injury following transfusion of aged platelets

Pulmonary complications from stored blood products are the leading cause of mortality related to transfusion. Transfusion-related acute lung injury is mediated by antibodies or bioactive mediators, yet underlying mechanisms are incompletely understood. Sphingolipids such as ceramide regulate lung injury, and their composition changes as a function of time in stored blood. Here, we tested the hypothesis that aged platelets may induce lung injury via a sphingolipid-mediated mechanism. To assess this hypothesis, a two-hit mouse model was devised. Recipient mice were treated with 2 mg/kg intraperitoneal lipopolysaccharide (priming) 2 h before transfusion of 10 ml/kg stored (1-5 days) platelets treated with or without addition of acid sphingomyelinase inhibitor ARC39 or platelets from acid sphingomyelinase-deficient mice, which both reduce ceramide formation. Transfused mice were examined for signs of pulmonary neutrophil accumulation, endothelial barrier dysfunction, and histological evidence of lung injury. Sphingolipid profiles in stored platelets were analyzed by mass spectrophotometry. Transfusion of aged platelets into primed mice induced characteristic features of lung injury, which increased in severity as a function of storage time. Ceramide accumulated in platelets during storage, but this was attenuated by ARC39 or in acid sphingomyelinase-deficient platelets. Compared with wild-type platelets, transfusion of ARC39-treated or acid sphingomyelinase-deficient aged platelets alleviated lung injury. Aged platelets elicit lung injury in primed recipient mice, which can be alleviated by pharmacological inhibition or genetic deletion of acid sphingomyelinase. Interventions targeting sphingolipid formation represent a promising strategy to increase the safety and longevity of stored blood products.
In the isolated rat liver perfused in situ, stimulation of the nerve bundles around the hepatic artery and portal vein caused an increase of glucose and lactate output and a reduction of perfusion flow. These changes could be inhibited completely by α-receptor blockers. The possible involvement of inositol phosphates in the intracellular signal transmission was studied. 1. In cell-suspension experiments, which were performed as a positive control, noradrenaline caused an increase in glucose output and, in the presence of 10 mM LiCl, a dose-dependent and time-dependent increase of inositol mono, bis and trisphosphate. 2. In the perfused rat liver 1 μM noradrenaline caused an increase of glucose and lactate output and in the presence of 10 mM LiCl a time-dependent increase of inositol mono, bis and trisphosphate that was comparable to that observed in cell suspensions. 3. In the perfused rat liver stimulation of the nerve bundles around the portal vein and hepatic artery caused a similar increase in glucose and lactate output to that produced by noradrenaline, but in the presence of 10 mM LiCl there was a smaller increase of inositol monophosphate and no increase of inositol bis and trisphosphate. These findings are in line with the proposal that circulating noradrenaline reaches every hepatocyte, causing a clear overall increase of inositol phosphate formation and thus calcium release from the endoplasmic reticulum, while the hepatic nerves reach only a few cells causing there a small local change of inositol phosphate metabolism and thence a propagation of the signal via gap junctions.
Asymmetric dimethylarginine (ADMA) is a competitive inhibitor of the nitric oxide (NO)-synthase and a biomarker of endothelial dysfunction (ED). ED plays an important role in the pathogenesis of contrast-induced nephropathy (CIN). The aim of our study was to evaluate serum ADMA concentration as a biomarker of an acute renal damage during the follow-up of 90 days after contrast medium (CM) application. Blood samples were obtained from 330 consecutive patients with diabetes mellitus or mild renal impairment immediately before, 24 and 48 hours after the CM application for coronary angiography. The patients were followed for 90 days. The composite endpoints were major adverse renal events (MARE) defined as occurrence of death, initiation of dialysis, or a doubling of serum creatinine concentration. Overall, ADMA concentration in plasma increased after CM application, although, there was no differences between ADMA levels in patients with and without CIN. ADMA concentration 24 hours after the CM application was predictive for dialysis with a specificity of 0.889 and sensitivity of 0.653 at values higher than 0.71 mu mol/L (area under the curve: 0.854, 95% confidential interval: 0.767-0.941, P<0.001). This association remained significant in multivariate Cox regression models adjusted for relevant factors of long-term renal outcome. 24 hours after the CM application, ADMA concentration in plasma was predictive for MARE with a specificity of 0.833 and sensitivity of 0.636 at a value of more than 0.70 mu mol/L (area under the curve: 0.750, 95% confidence interval: 0.602-0.897, P=0.004). Multivariate logistic regression analysis confirmed that ADMA and anemia were significant predictors of MARE. Further analysis revealed that increased ADMA concentration in plasma was highly significant predictor of MARE in patients with CIN. Moreover, patients with CIN and MARE had the highest plasma ADMA levels 24 hours after CM exposure in our study cohort. The impact of ADMA on MARE was independent of such known CIN risk factors as anemia, pre-existing renal failure, pre-existing heart failure, and diabetes. ADMA concentration in plasma is a promising novel biomarker of major contrast-induced nephropathy-associated events 90 days after contrast media exposure.
Zellbasierte heterologe Expressionssysteme bieten ein einfaches und schnelles Verfahren, um neue Süßstoffe oder Süßverstärker zu finden. Unter Verwendung eines solchen Testsystems, konnte ich in Zusammenarbeit mit der Symrise AG, Holzminden und dem Institut für Pflanzenbiochemie in Halle/Saale die vietnamesische Pflanze Mycetia balansae als Quelle eines neuen Süßstoffs identifizieren. Deren Hauptkomponenten, genannt Balansine, aktivieren spezifisch den humanen Süßrezeptor. Chimäre Rezeptoren zeigten, dass die amino-terminalen Domänen der Süßrezeptoruntereinheiten, welche ein Großteil der Liganden des Süßrezeptors binden, für dessen Aktivierung durch Balansin A nicht notwendig sind.
Voraussetzung für die Anwendung zellbasierter Testsysteme zum Auffinden neuer Süßstoffe ist jedoch, dass süße Substanzen gesichert identifiziert werden, während nicht süße Substanzen zuverlässig keine Rezeptoraktivierung aufweisen. Während in HEK293 TAS1R2 TAS1R3To Galpha15i3-Zellen Süßrezeptoraktivierung gegenüber nicht süß schmeckenden Substanzen beobachtet wurde, konnte mit den HEK293PEAKrapid Galpha15-Zellen ein zuverlässiges Testsystem identifiziert, welches den Süßgeschmack der untersuchten Substanzen widerspiegelte.
Es fanden sich keine Hinweise, dass akzessorische Proteine oder verwandte Rezeptoren des Süßrezeptors das unterschiedliche Verhalten der Zellen verursachen. Es konnte gezeigt werden, dass die Verwendung unterschiedlicher G-Proteine die Signalamplituden des Süßrezeptors beeinflusst, die Unterschiede zwischen den Zellsystemen jedoch nicht vollständig erklärt. Keine der untersuchten Galpha-Proteinchimären spiegelte die intrinsische Süße der Substanzen wider.
Wenn auch nicht ursächlich für die Diskrepanz zwischen Süßrezeptoraktivierung in vitro und Süßgeschmack in vivo, so weisen die Ergebnisse dieser Arbeit auf eine Interaktion der Süßrezeptoruntereinheiten mit dem humanen Calcium-sensing Rezeptor hin. Vanillin und Ethylvanillin konnten als neue Agonisten des Calcium-sensing Rezeptors identifiziert werden.
Wie die vorliegende Arbeit zeigt, können sich kleine Unterschiede im Zellhintergrund deutlich auf die Funktionsweise heterolog exprimierter Rezeptoren auswirken. Dies zeigt wie wichtig die Wahl der Zellen für solche Screeningsysteme ist.
For centuries, Amaranthus sp. were used as food, ornamentals, and medication. Molecular mechanisms, explaining the health beneficial properties of amaranth, are not yet understood, but have been attributed to secondary metabolites, such as phenolic compounds. One of the most abundant phenolic compounds in amaranth leaves is 2-caffeoylisocitric acid (C-IA) and regarding food occurrence, C-IA is exclusively found in various amaranth species. In the present study, the anti-inflammatory activity of C-IA, chlorogenic acid, and caffeic acid in LPS-challenged macrophages (RAW 264.7) has been investigated and cellular contents of the caffeic acid derivatives (CADs) were quantified in the cells and media. The CADs were quantified in the cell lysates in nanomolar concentrations, indicating a cellular uptake. Treatment of LPS-challenged RAW 264.7 cells with 10 µM of CADs counteracted the LPS effects and led to significantly lower mRNA and protein levels of inducible nitric oxide synthase, tumor necrosis factor alpha, and interleukin 6, by directly decreasing the translocation of the nuclear factor κB/Rel-like containing protein 65 into the nucleus. This work provides new insights into the molecular mechanisms that attribute to amaranth’s anti-inflammatory properties and highlights C-IA’s potential as a health-beneficial compound for future research.
IMPORTANCE Inflammatory processes have been suggested to have an important role in colorectal cancer (CRC) etiology. Chemerin is a recently discovered inflammatory biomarker thought to exert chemotactic, adipogenic, and angiogenic functions. However, its potential link with CRC has not been sufficiently explored. OBJECTIVE To evaluate the prospective association of circulating plasma chemerin concentrations with incident CRC. DESIGN, SETTING, AND PARTICIPANTS Prospective case-cohort study based on 27 548 initially healthy participants from the European Prospective Investigation Into Cancer and Nutrition (EPIC)-Potsdam cohort who were followed for up to 16 years. Baseline study information and samples were collected between August 23, 1994, and September 25, 1998. Recruitment was according to random registry sampling from the geographical area of Potsdam, Germany, and surrounding municipalities. The last date of study follow-up was May 10, 2010. Statistical analysis was conducted in 2018. MAIN OUTCOMES AND MEASURES Incident CRC, colon cancer, and rectal cancer. Baseline chemerin plasma concentrations were measured by enzyme-linked immunosorbent assay. CONCLUSIONS AND RELEVANCE This study found that the association between chemerin concentration and the risk of incident CRC was linear and independent of established CRC risk factors. Further studies are warranted to evaluate chemerin as a novel immune-inflammatory agent in colorectal carcinogenesis.
Aims/hypothesis This study aimed to evaluate associations of height as well as components of height (sitting height and leg length) with risk of type 2 diabetes and to explore to what extent associations are explainable by liver fat and cardiometabolic risk markers. Methods A case-cohort study within the European Prospective Investigation into Cancer and Nutrition (EPIC)-Potsdam study comprising 26,437 participants who provided blood samples was designed. We randomly selected a subcohort of 2500 individuals (2029 diabetes-free at baseline and with anamnestic, anthropometrical and metabolic data for analysis). Of the 820 incident diabetes cases identified in the full cohort during 7 years of follow-up, 698 remained for analyses after similar exclusions. Results After adjustment for age, potential lifestyle confounders, education and waist circumference, greater height was related to lower diabetes risk (HR per 10 cm, men 0.59 [95% CI 0.47, 0.75] and women 0.67 [0.51, 0.88], respectively). Leg length was related to lower risk among men and women, but only among men if adjusted for total height. Adjustment for liver fat and triacylglycerols, adiponectin and C-reactive protein substantially attenuated associations between height and diabetes risk, particularly among women. Conclusions/interpretation We observed inverse associations between height and risk of type 2 diabetes, which was largely related to leg length among men. The inverse associations may be partly driven by lower liver fat content and a more favourable cardiometabolic profile.
Die allergische Kontaktdermatitis ist eine immunologisch bedingte Hauterkrankung mit insbesondere in den westlichen Industrienationen hoher und weiter ansteigender Prävalenz. Es handelt sich hierbei um eine Hypersensitivitätsreaktion vom Typ IV, die sich nach Allergenkontakt durch Juckreiz, Rötung, Bläschenbildung und Abschälung der Haut äußert. Zahlreiche Xenobiotika besitzen das Potenzial, Kontaktallergien auszulösen, darunter Konservierungsstoffe, Medikamente, Duftstoffe und Chemikalien. Die wirksamste Maßnahme zur Eindämmung der Erkrankung ist die Expositionsprophylaxe, also die Vermeidung des Kontakts mit den entsprechenden Substanzen. Dies wiederum setzt die Kenntnis des jeweiligen sensibilisierenden Potenzials einer Substanz voraus, dessen Bestimmung aus diesem Grund eine hohe toxikologische Relevanz besitzt. Zu diesem Zweck existieren von der OECD veröffentlichte Testleitlinien, welche auf entsprechend validierten Testmethoden basieren. Goldstandard bei der Prüfung auf hautsensibilisierendes Potenzial war über lange Zeit der murine Lokale Lymphknotentest. Seit der 7. Änderung der EU-Kosmetikrichtlinie, welche Tierversuche für Kosmetika und deren Inhaltsstoffe untersagt, wurden vermehrt Alternativmethoden in die OECD-Testleitlinien implementiert.. Die bestehenden in vitro Methoden sind jedoch alleinstehend nur begrenzt aussagekräftig, da sie lediglich singuläre Mechanismen bei der Entstehung einer Kontaktallergie abbilden. Die Entwicklung von Testmethoden, welche mehrere dieser Schlüsselereignisse berücksichtigen, erscheint daher richtungsweisend. Einen vielversprechenden Ansatz liefert hierbei der Loose-fit coculture-based sensitisation assay (LCSA), welcher eine Kokultur aus primären Keratinozyten und PBMC darstellt. Bei der Kokultivierung von Immunzellen mit anderen Zelltypen stellt sich allerdings die Frage, inwiefern die Nutzung von Zellen derselben Spender*innen (autologe Kokultur) bzw. verschiedener Spender*innen (allogene Kokultur) einen Einfluss nimmt. Zu diesem Zweck wurden im Rahmen dieser Arbeit Hautzellen spenderspezifisch aus gezupften Haarfollikeln isoliert und der LCSA mit den generierten HFDK in autologen und allogenen Ansätzen verglichen. Zusätzlich wurde auch ein Vergleich zwischen der Nutzung von HFDK und NHK, welche aus humaner Vorhaut isoliert wurden, im LCSA durchgeführt. Dabei ergaben sich keine signifikanten Unterschiede zwischen autologen und allogenen Kokulturen bzw. zwischen der Verwendung von HFDK und NHK. Die Verwendung allogener Zellen aus anonymem Spendermaterial sowie die Nutzung von Keratinozyten aus unterschiedlichen Quellen scheint im Rahmen des LCSA problemlos möglich. Einige der getesteten Kontaktallergene, darunter DNCB und NiCl2, erwiesen sich im LCSA jedoch als problematisch und konnten nicht zufriedenstellend als sensibilisierend detektiert werden. Daher wurde eine Optimierung der Kokultur durch Verwendung ex vivo differenzierter Langerhans Zellen (MoLC) angestrebt, welche ein besseres Modell primärer epidermaler Langerhans Zellen darstellen als die dendritischen Zellen aus dem LCSA. Zusätzlich wurden weitere, den Erfolg der Kokultur beeinflussende Faktoren, wie die Art und Zusammensetzung des Mediums und die Kokultivierungsdauer, untersucht und angepasst. Das schlussendlich etablierte Kokultivierungsprotokoll führte zu einer maßgeblich verstärkten Expression von CD207 (Langerin) auf den MoLC, was auf eine wirkungsvolle Interaktion zwischen Haut- und Immunzellen in der Kokultur hindeutete. Des Weiteren konnten DNCB und NiCl2 im Gegensatz zum LCSA durch Verwendung des kostimulatorischen Moleküls CD86 sowie des Reifungsmarkers CD83 als Ausleseparameter eindeutig als Kontaktallergene identifiziert werden. Die Untersuchungen zur Kokultur von MoLC und HFDK wurden jeweils vergleichend in autologen und allogenen Ansätzen durchgeführt. Ähnlich wie beim LCSA kam es aber auch hier zu keinen signifikanten Unterschieden, weder hinsichtlich der Expression von Charakterisierungs- und Aktivierungsmarkern auf MoLC noch hinsichtlich der Zytokinsekretion in den Zellkulturüberstand. Die Hinweise aus zahlreichen Studien im Mausmodell, dass Zellen des angeborenen Immunsystems zur Erkennung von und Aktivierung durch allogene Zellen bzw. Gewebe in der Lage sind, bestätigten sich im Rahmen dieser Arbeit dementsprechend nicht. Aus diesem Grund wurden abschließend CD4+ T-Lymphozyten, die Effektorzellen des adaptiven Immunsystems, in die Kokultur aus MoLC und autologen bzw. allogenen HFDK integriert. Überraschenderweise traten auch hier keine verstärkten Aktivierungen in allogener Kokultur im Vergleich zur autologen Kokultur auf. Die Nutzung autologer Primärzellen scheint im Rahmen der hier getesteten Methoden nicht notwendig zu sein, was die Validierung von Kokulturen und deren Implementierung in die OECD-Testleitlinien erleichtern dürfte. Zuletzt wurde eine Kokultivierung primärer Haut- und Immunzellen auch im 3D-Vollhautmodell durchgeführt, wobei autologe MoLC in die Epidermisäquivalente entsprechender Modelle integriert werden sollten. Obwohl die erstellten Hautmodelle unter Verwendung autologer Haarfollikel-generierter Keratinozyten und Fibroblasten eine zufriedenstellende Differenzierung und Stratifizierung aufwiesen, gestaltete sich die Inkorporation der MoLC als problematisch und konnte im Rahmen dieser Arbeit nicht erreicht werden.

Being born small (SGA) or large for gestational age (LGA) is associated with adverse birth outcomes and metabolic diseases in later life of the offspring. It is known that aberrations in growth during gestation are related to altered placental function. Placental function is regulated by epigenetic mechanisms such as DNA methylation. Several studies in recent years have demonstrated associations between altered patterns of DNA methylation and adverse birth outcomes. However, larger studies that reliably investigated global DNA methylation are lacking. The aim of this study was to characterize global placental DNA methylation in relationship to size for gestational age. Global DNA methylation was assessed in 1023 placental samples by LC-MS/MS. LGA offspring displayed significantly higher global placental DNA methylation compared to appropriate for gestational age (AGA; p<0.001). ANCOVA analyses adjusted for known factors impacting on DNA methylation demonstrated an independent association between placental global DNA methylation and LGA births (p<0.001). Tertile stratification according to global placental DNA methylation levels revealed a significantly higher frequency of LGA births in the third tertile. Furthermore, a multiple logistic regression analysis corrected for known factors influencing birth weight highlighted an independent positive association between global placental DNA methylation and the frequency of LGA births (p=0.001).

Durch die Zunahme metabolischer Stoffwechselstörungen und Erkrankungen in der Weltbevölkerung wird in der Medizin und den Lebenswissenschaften vermehrt nach Präventionsstrategien und Ansatzpunkten gesucht, die die Gesundheit fördern, Erkrankungen verhindern helfen und damit auch die Gesamtlast auf die Gesundheitssysteme erleichtern. Ein Ansatzpunkt wird dabei in der Ernährung gesehen, da insbesondere der Konsum von gesättigten Fetten die Gesundheit nachträglich zu beeinflussen scheint. Dabei wird übersehen, dass in vielen Studien Hochfettdiäten nicht ausreichend von den Einflüssen einer zum Bedarf hyperkalorischen Energiezufuhr getrennt werden, sodass die Datenlage zu dem Einfluss von (gesättigten) Fetten auf den Metabolismus bei gleichbleibender Energieaufnahme noch immer unzureichend ist.
In der NUtriGenomic Analysis in Twins-Studie wurden 46 Zwillingspaare (34 monozygot, 12 dizygot) über einen Zeitraum von sechs Wochen mittels einer kohlenhydratreichen, fettarmen Diät nach Richtlinien der Deutschen Gesellschaft für Ernährung für ihr Ernährungsverhalten standardisiert, ehe sie zu einer kohlenhydratarmen, fettreichen Diät, die insbesondere gesättigte Fette enthielt, für weitere sechs Wochen wechselten. Beide Diäten waren dem individuellen Energiebedarf der Probanden angepasst, um so sowohl akut nach einerWoche als auch längerfristig nach sechs Wochen Änderungen des Metabolismus beobachten zu können, die sich in der vermehrten Aufnahme von (gesättigten) Fetten begründeten.
Die über die detaillierte Charakterisierung der Probanden an den klinischen Untersuchungstagen generierten Datensätze wurden mit statistischen und mathematischen Methoden (z.B. lineare gemischte Modellierung) analysiert, die der Größe der Datensätze und damit ihrem Informationsvolumen angepasst waren.
Es konnte gezeigt werden, dass die metabolisch gesunden und relativ jungen Probanden, die eine gute Compliance zeigten, im Hinblick auf ihren Glukosestoffwechsel adaptieren konnten, indem die Akutantwort nach einer Woche im Nüchterninsulin und dem Index für Insulinresistenz in den weiteren fünf Wochen ausgeglichen wurde.
Der Lipidstoffwechsel in Form der klassischen Marker wie Gesamtcholesterin, LDL und HDL war dagegen stärker beeinflusst und auch nach insgesamt sechs Wochen deutlich erhöht.
Letzteres unterstützt die Beobachtung im Transkriptom des weißen, subkutanen Fettgewebes, bei der eine Aktivierung der über die Toll-like receptors und das Inflammasom vermittelten subklinischen Inflammation beobachtet werden konnte.
Die auftretenden Veränderungen in Konzentration und Komposition des Plasmalipidoms zeigte ebenfalls nur eine teilweise und auf bestimmte Spezies begrenzte Gegenregulation.
Diesbezüglich kann also geschlussfolgert werden, dass auch die isokalorische Aufnahme von (gesättigten) Fetten zu Veränderungen im Metabolismus führt, wobei die Auswirkungen in weiteren (Langzeit-)Studien und Experimenten noch genauer untersucht werden müssen. Insbesondere wäre dabei ein längerer Zeitraum unter isokalorischen Bedingungen von Interesse und die Untersuchung von Probanden mit metabolischer Vorbelastung (z.B. Insulinresistenz).
Darüber hinaus konnte in NUGAT aber ebenfalls gezeigt werden, dass die Nutrigenetik und Nutrigenomik zwei nicht zu vernachlässigende Faktoren darstellen. So zeigten unter anderem die Konzentrationen einiger Lipidspezies eine starke Erblichkeit und Abhängigkeit der Diät.
Zudem legen die Ergebnisse nahe, dass laufende wie geplante Präventionsstrategien und medizinische Behandlungen deutlich stärker den Patienten als Individuum mit einbeziehen müssen, da die Datenanalyse interindividuelle Unterschiede identifizierte und Hinweise lieferte, dass einige Probanden die nachteiligen, metabolischen Auswirkungen einer Hochfettdiät besser ausgleichen konnten als andere.
Der Bittergeschmack warnt den Organismus vor potentiell verdorbener oder giftiger Nahrung und ist somit ein wichtiger Kontrollmechanismus. Die initiale Detektion der zahlreich vorkommenden Bitterstoffe erfolgt bei der Maus durch 35 Bitterrezeptoren (Tas2rs), die sich im Zungengewebe befinden. Die Geschmacksinformation wird anschließend von der Zunge über das periphere (PNS) ins zentrale Nervensystem (ZNS) geleitet, wo deren Verarbeitung stattfindet. Die Verarbeitung der Geschmacksinformation konnte bislang nicht gänzlich aufgeklärt werden. Neue Studien deuten auf eine Expression von Tas2rs auch im PNS und ZNS entlang der Geschmacksbahn hin. Über Vorkommen und Aufgaben dieser Rezeptoren bzw. Rezeptorzellen im Nervensystem ist bislang wenig bekannt.
Im Rahmen dieser Arbeit wurde die Tas2r-Expression in verschiedenen Mausmodellen untersucht, Tas2r-exprimierende Zellen identifiziert und deren Funktionen bei der Übertragung der Geschmacksinformationen analysiert. Im Zuge der Expressionsanalysen mittels qRT-PCR konnte die Expression von 25 der 35 bekannten Bittergeschmacksrezeptoren im zentralen Nervensystem der Maus nachgewiesen werden. Die Expressionsmuster im PNS sowie im ZNS lassen darüber hinaus Vermutungen zu Funktionen in verschiedenen Bereichen des Nervensystems zu. Basierend auf den Ergebnissen der Expressionsanalysen war es möglich, stark exprimierte Tas2rs mittels In-situ-Hybridisierung in verschiedenen Zelltypen zu visualisieren. Des Weiteren konnten immunhistochemische Färbungen unter Verwendung eines genetisch modifizierten Mausmodells die Ergebnisse der Expressionsanalysen bestätigen. Sie zeigten eine Expression von Tas2rs, am Beispiel des Tas2r131-Rezeptors, in cholinergen, dopaminergen, GABAergen, noradrenergen und glycinerg-angesteuerten Projektionsneuronen sowie in Interneuronen. Die Ergebnisse der vorliegenden Arbeit zeigen daher erstmals das Vorkommen von Tas2rs in verschiedenen neuronalen Zelltypen in weiten Teilen des ZNS. Dies lässt den Schluss zu, dass Tas2r-exprimierende Zellen potentiell multiple Funktionen innehaben. Anhand von Verhaltensexperimenten in genetisch modifizierten Mäusen wurde die mögliche Funktion von Tas2r131-exprimierenden Neuronen (Tas2r131-Neurone) bei der Geschmackswahrnehmung untersucht. Die Ergebnisse weisen auf eine Beteiligung von Tas2r131-Neuronen an der Signalweiterleitung bzw. -verarbeitung der Geschmacksinformation für eine Auswahl von Bittersubstanzen hin. Die Analysen zeigen darüber hinaus, dass Tas2r131-Neuronen nicht an der Geschmackswahrnehmung anderer Bitterstoffe sowie Geschmacksstimuli anderer Qualitäten (süß, umami, sauer, salzig), beteiligt sind. Eine spezifische „Tas2r131-Bittergeschmacksbahn“, die mit anderen potentiellen „Bitterbahnen“ teils unabhängige, teils überlappende Signalwege bzw. Verarbeitungsbereiche besitzt, bildet eine mögliche zelluläre Grundlage zur Unterscheidung von Bitterstoffen. Die im Rahmen dieser Arbeit entstandene Hypothese einer potentiellen Diskriminierung von Bitterstoffen soll daher in weiterführenden Studien durch die Etablierung eines Verhaltenstest mit Mäusen geprüft werden.
Boron-associated shifts in sex ratios at birth were suggested earlier and attributed to a decrease in Y- vs. X-bearing sperm cells. As the matter is pivotal in the discussion of reproductive toxicity of boron/borates, re-investigation in a highly borate-exposed population was required. In the present study, 304 male workers in Bandirma and Bigadic (Turkey) with different degrees of occupational and environmental exposure to boron were investigated. Boron was quantified in blood, urine and semen, and the persons were allocated to exposure groups along B blood levels. In the highest ("extreme") exposure group (n = 69), calculated mean daily boron exposures, semen boron and blood boron concentrations were 44.91 +/- 18.32 mg B/day, 1643.23 +/- 965.44 ng B/g semen and 553.83 +/- 149.52 ng B/g blood, respectively. Overall, an association between boron exposure and Y:X sperm ratios in semen was not statistically significant (p > 0.05). Also, the mean Y:X sperm ratios in semen samples of workers allocated to the different exposure groups were statistically not different in pairwise comparisons (p > 0.05). Additionally, a boron-associated shift in sex ratio at birth towards female offspring was not visible. In essence, the present results do not support an association between boron exposure and decreased Y:X sperm ratio in males, even under extreme boron exposure conditions.
Consumption of Brassica vegetables is linked to health benefits, as they contain high concentrations of the following secondary plant metabolites (SPMs): glucosinolate breakdown products, carotenoids, chlorophylls, and phenolic compounds. Especially Brassica vegetables are consumed as microgreens (developed cotyledons). It was investigated how different ontogenetic stages (microgreens or leaves) of pak choi (Brassica rapa subsp. chinensis) and kale (Brassica oleracea var. sabellica) differ in their SPM concentration. The impact of breadmaking on SPMs in microgreens (7 days) and leaves (14 days) in pak choi and kale as a supplement in mixed wheat bread was assessed. In leaves, carotenoids, chlorophylls, and phenolic compounds were higher compared to those of microgreens. Breadmaking caused a decrease of SPMs. Chlorophyll degradation was observed, leading to pheophytin and pyropheophytin formation. In kale, sinapoylgentiobiose, a hydroxycinnamic acid derivative, concentration increased. Thus, leaves of Brassica species are suitable as natural ingredients for enhancing bioactive SPM concentrations in bread.
Mitochondria are critical for hypothalamic function and regulators of metabolism. Hypothalamic mitochondrial dysfunction with decreased mitochondrial chaperone expression is present in type 2 diabetes (T2D). Recently, we demonstrated that a dysregulated mitochondrial stress response (MSR) with reduced chaperone expression in the hypothalamus is an early event in obesity development due to insufficient insulin signaling. Although insulin activates this response and improves metabolism, the metabolic impact of one of its members, the mitochondrial chaperone heat shock protein 10 (Hsp10), is unknown. Thus, we hypothesized that a reduction of Hsp10 in hypothalamic neurons will impair mitochondrial function and impact brain insulin action. Therefore, we investigated the role of chaperone Hsp10 by introducing a lentiviral-mediated Hsp10 knockdown (KD) in the hypothalamic cell line CLU-183 and in the arcuate nucleus (ARC) of C57BL/6N male mice. We analyzed mitochondrial function and insulin signaling utilizing qPCR, Western blot, XF96 Analyzer, immunohistochemistry, and microscopy techniques. We show that Hsp10 expression is reduced in T2D mice brains and regulated by leptin in vitro. Hsp10 KD in hypothalamic cells induced mitochondrial dysfunction with altered fatty acid metabolism and increased mitochondria-specific oxidative stress resulting in neuronal insulin resistance. Consequently, the reduction of Hsp10 in the ARC of C57BL/6N mice caused hypothalamic insulin resistance with acute liver insulin resistance.

Prostaglandin (PG)F₂α has previously been shown to increase glucose output from perfused livers and isolated hepatocytes, where it stimulated glycogen phosphorylase via an inositol-trisphosphatedependent signal pathway. In this study, PGF₂α binding sites on hepatocyte plasma membranes, that might represent the putative receptor, were characterized. Binding studies could not be performed with intact hepatocytes, because PGF₂α accumulated within the cells even at 4°C. The intracellular accumulation was an order of magnitude higher than binding to plasma membranes. Purified hepatocyte plasma membranes had a high-affinity/low-capacity and a low-affinity/highcapacity binding'site for PGF₂α. The respective binding constants for the high-affinity site were Kd = 3 nM and Bmax = 6 fmol/mg membrane protein, and for the low-affinity site Kd = 426 nM and Bmax = 245 fmol/mg membrane protein. Specific PGF₂α binding to the low-affinity site, but not to the high-affinity site, could be enhanced most potently by GTP[γS] followed by GDP[ϐS] and GTP, but not by ATP[γS] or GMP. PGF₂α competed most potently with [³H]PGF₂α for specific binding to hepatocyte plasma membranes, followed by PGD₂ and PGE₂. Since the low-affinity PGF₂α-binding site had a Kd in the concentration range in which PG had previously been shown to be half-maximally active, and since this binding site showed a sensitivity to GTP, it is concluded that it might represent the receptor involved in the PGF₂α signal chain in hepatocytes. A biological function of the high-affinity site is currently not known.
Inhibition of acid sphingomyelinase (ASM), a lysosomal enzyme that catalyzes the hydrolysis of sphingomyelin into ceramide and phosphorylcholine, may serve as an investigational tool or a therapeutic intervention to control many diseases. Specific ASM inhibitors are currently not sufficiently characterized. Here, we found that 1-aminodecylidene bis-phosphonic acid (ARC39) specifically and efficiently (>90%) inhibits both lysosomal and secretory ASM in vitro. Results from investigating sphingomyelin phosphodiesterase 1 (SMPD1/Smpd1) mRNA and ASM protein levels suggested that ARC39 directly inhibits ASM's catalytic activity in cultured cells, a mechanism that differs from that of functional inhibitors of ASM. We further provide evidence that ARC39 dose- and time-dependently inhibits lysosomal ASM in intact cells, and we show that ARC39 also reduces platelet- and ASM-promoted adhesion of tumor cells. The observed toxicity of ARC39 is low at concentrations relevant for ASM inhibition in vitro, and it does not strongly alter the lysosomal compartment or induce phospholipidosis in vitro. When applied intraperitoneally in vivo, even subtoxic high doses administered short-term induced sphingomyelin accumulation only locally in the peritoneal lavage without significant accumulation in plasma, liver, spleen, or brain. These findings require further investigation with other possible chemical modifications. In conclusion, our results indicate that ARC39 potently and selectively inhibits ASM in vitro and highlight the need for developing compounds that can reach tissue concentrations sufficient for ASM inhibition in vivo.

Background: Being an essential trace element, copper is involved in diverse physiological processes. However, excess levels might lead to adverse effects. Disrupted copper homeostasis, particularly in the brain, has been associated with human diseases including the neurodegenerative disorders Wilson and Alzheimer?s disease. In this context, astrocytes play an important role in the regulation of the copper homeostasis in the brain and likely in the prevention against neuronal toxicity, consequently pointing them out as a potential target for the neurotoxicity of copper. Major toxic mechanisms are discussed to be directed against mitochondria probably via oxidative stress. However, the toxic potential and mode of action of copper in astrocytes is poorly understood, so far. Methods: In this study, excess copper levels affecting human astrocytic cell model and their involvement in the neurotoxic mode of action of copper, as well as, effects on the homeostasis of other trace elements (Mn, Fe, Ca and Mg) were investigated. Results: Copper induced substantial cytotoxic effects in the human astrocytic cell line following 48 h incubation (EC30: 250 ?M) and affected mitochondrial function, as observed via reduction of mitochondrial membrane potential and increased ROS production, likely originating from mitochondria. Moreover, cellular GSH metabolism was altered as well. Interestingly, not only cellular copper levels were affected, but also the homeostasis of other elements (Ca, Fe and Mn) were disrupted. Conclusion: One potential toxic mode of action of copper seems to be effects on the mitochondria along with induction of oxidative stress in the human astrocytic cell model. Moreover, excess copper levels seem to interact with the homeostasis of other essential elements such as Ca, Fe and Mn. Disrupted element homeostasis might also contribute to the induction of oxidative stress, likely involved in the onset and progression of neurodegenerative disorders. These insights in the toxic mechanisms will help to develop ideas and approaches for therapeutic strategies against copper-mediated diseases.
Im Sinne des Refinements von Tierversuchen sollen alle Bedingungen während der Zucht, der Haltung und des Transports von zu Versuchszwecken gehaltenen Tieren und alle Methoden während des Versuchs so verbessert werden, dass die verwendeten Tiere ein minimales Maß an potentiellem Distress, Schmerzen oder Leiden erfahren. Zudem soll ihr Wohlbefinden durch die Möglichkeit des Auslebens speziesspezifischer Verhaltensweisen und die Anwendung tierschonender Verfahren maximal gefördert werden. Zur Etablierung von Grundsätzen des Refinements sind grundlegende Kenntnisse über die physiologischen Bedürfnisse und Verhaltensansprüche der jeweiligen Spezies unabdingbar. Die Experimentatoren sollten das Normalverhalten der Tiere kennen, um potentielle Verhaltensabweichungen, wie Stereotypien, zu verstehen und interpretieren zu können. Standardisierte Haltungsbedingungen von zu Versuchszwecken gehaltenen Mäusen weichen in diversen Aspekten von der natürlichen Umgebung ab und erfordern eine gewisse Adaptation. Ist ein Tier über einen längeren Zeitraum unfähig, sich an die gegebenen Umstände anzupassen, können abnormale Verhaltensweisen, wie Stereotypien auftreten. Stereotypien werden definiert als Abweichungen vom Normalverhalten, die repetitiv und ohne Abweichungen im Ablauf ausgeführt werden, scheinbar keiner Funktion dienen und der konkreten Umweltsituation nicht immer entsprechen.
Bisher war unklar, in welchem Ausmaß stereotypes Verhalten den metabolischen Phänotyp eines Individuums beeinflusst. Ziel dieser Arbeit war es daher, das stereotype Verhalten der FVB/NJ-Maus erstmals detailliert zu charakterisieren, systematisch zusammenzutragen, welche metabolischen Konsequenzen dieses Verhalten bedingt und wie sich diese auf das Wohlbefinden der Tiere und die Verwendung stereotyper Tiere in Studien mit tierexperimentellem Schwerpunkt auswirken.
Der Versuch begann mit der Charakterisierung der mütterlichen Fürsorge in der Parentalgeneration. Insgesamt wurden 35 Jungtiere der F1-Generation vom Absatz an, über einen Zeitraum von 11 Wochen einzeln gehalten, kontinuierlich beobachtet, bis zum Versuchsende wöchentlich Kotproben gesammelt und das Körpergewicht bestimmt. Zusätzlich erfolgten begleitende Untersuchungen wie Verhaltenstests und die Erfassung der physischen Aktivität und metabolischer Parameter. Anschließend wurden u.a. die zerebralen Serotonin- und Dopamingehalte, fäkale Glucocorticoidlevels, hepatisches Glykogen und muskuläre Glykogen- und Triglyceridlevels bestimmt.
Nahezu unabhängig von der mütterlichen Herkunft entwickelte sich bei mehr als der Hälfte der 35 Jungtiere in der F1-Generation stereotypes Verhalten. Diese Daten deuten darauf hin, dass es keine Anzeichen für das Erlernen oder eine direkte genetische Transmission stereotypen Verhaltens bei der FVB/NJ-Maus gibt. Über den gesamten Beobachtungszeitraum zeichneten sich die stereotypen FVB/NJ-Mäuse durch ein eingeschränktes Verhaltensrepertoire aus. Zu Gunsten der erhöhten Aktivität und des Ausübens stereotypen Verhaltens lebten sie insgesamt weniger andere Verhaltensweisen (Klettern, Graben, Nagen) aus. Darüber hinaus waren Stereotypien sowohl im 24-Stunden Open Field Test als auch in der Messeinrichtung der indirekten Tierkalorimetrie mit einer erhöhten Aktivität und Motilität assoziiert, während die circadiane Rhythmik nicht divergierte. Diese erhöhte körperliche Betätigung spiegelte sich in den niedrigeren Körpergewichtsentwicklungen der stereotypen Tiere wieder. Außerdem unterschieden sich die Körperfett- und Körpermuskelanteile.
Zusammenfassend lässt sich sagen, dass das Ausüben stereotypen Verhaltens zu Differenzen im metabolischen Phänotyp nicht-stereotyper und stereotyper FVB/NJ-Mäuse führt. Im Sinne der „Guten Wissenschaftlichen Praxis“ sollte das zentrale Ziel jedes Wissenschaftlers sein, aussagekräftige und reproduzierbare Daten hervorzubringen. Jedoch können keine validen Resultate von Tieren erzeugt werden, die in Aspekten variieren, die für den vorgesehenen Zweck der Studie nicht berücksichtigt wurden. Deshalb sollten nicht-stereotype und stereotype Individuen nicht innerhalb einer Versuchsgruppe randomisiert werden. Stereotype Tiere demzufolge von geplanten Studien auszuschließen, würde allerdings dem Gebot des zweiten R’s – der Reduction – widersprechen. Um Refinement zu garantieren, sollte der Fokus auf der maximal erreichbaren Prävention stereotypen Verhaltens liegen. Diverse Studien haben bereits gezeigt, dass die Anreicherung der Haltungsumwelt (environmental enrichment) zu einer Senkung der Prävalenz von Stereotypien bei Mäusen führt, dennoch kommen sie weiterhin vor. Daher sollte environmental enrichment zukünftig weniger ein „Kann“, sondern ein „Muss“ sein – oder vielmehr: der Goldstandard. Zudem würde eine profunde phänotypische Charakterisierung dazu beitragen, Mausstämme zu erkennen, die zu Stereotypien neigen und den für den spezifischen Zweck am besten geeigneten Mausstamm zu identifizieren, bevor ein Experiment geplant wird.
Die Ausstattung der gastrointestinalen Mukosa des Menschen und der Ratte mit Sulfotransferasen wurde mit Hilfe von Immunodetektion und Enzymaktivitätsmessungen untersucht. In Proben aus Colon und Rektum von 39 Personen wurden die Formen h1A1, h1A3 und h1B1 identifiziert, wobei in einer weiteren Probe, die als einzige von einem an Colitis Ulcerosa erkrankten Patienten stammte, keine Sulfotransferasen nachgewiesen werden konnten. Bei der Immunblot-Analyse war das Expressionsmuster der einzelnen Formen in allen Proben ähnlich. In wenigen Proben waren die relativen Signalintensitäten der h1A1 und der h1B1 um die Hälfte erniedrigt. Der Gehalt von SULT an zytosolischem Protein zeigte einen bis zu 8 - 10fachen Unterschied, er betrug jedoch bei zwei Dritteln der Proben zwischen 0,15 und 0,3 (h1A1 und h1A3) bzw. 0,6 und 0,8 Promille (h1B1). Die Variation konnte nicht auf Alter, Geschlecht oder Krankheitsbild der Patienten zurückgeführt werden. Auch der für die allelischen Varianten der h1A1 beschriebene Effekt auf die Enzymaktiviät bzw. Stabilität konnte in der Menge an immunreaktivem Protein nicht in diesem Ausmaß detektiert werden. Die Allelhäufigkeit von h1A1*R und h1A1*H war gegenüber der gesunden Bevölkerung nicht verändert. In den sieben Proben aus dem Dünndarm (Coecum, viermal Ileum, Jejunum) konnten zusätzlich die Formen h1E1 und h2A1 identifiziert werden. Ein möglicherweise der Form h1C1 entsprechendes Protein wurde im Magen detektiert. Im Vergleich zum Menschen war die Expression in der Ratte stärker auf die Leber konzentriert. Während beim Menschen in allen untersuchten Abschnitten Sulfotransferasen in Mengen detektiert wurden, die in zwei Fällen (h1B1 und h1A3) sogar den Gehalt in der Leber überstiegen, beschränkte sich die Expression in der Ratte auf im Vergleich zur Leber geringe Mengen im Magen und Dickdarm. Nachgewiesen wurden die r1B1, r1A1 sowie eine nicht identifizierte Form von 35kD, bei der es sich vermutlich um die r1C2 handelt. Im Vergleich zur Leber enthielt der Dickdarm der Ratte 20 - 30 % an r1B1 und 3 % an r1A1, während im Dickdarm des Menschen die 3 - 5fache Menge an h1B1 und 25 - 50 % an h1A1 gefunden wurden. Die nicht identifizierte Form verhielt sich wie die r1B1. Die für die Leber der Ratte bekannte geschlechtsabhängige Expression wurde im Gastrointestinaltrakt nicht beobachtet. Die Verteilung der Sulfotransferasen im Colon und Ileum des Menschen wurde immunhistochemisch untersucht; für die Gewebe der Ratte war die Spezifität der zur Verfügung stehenden Antiseren nicht ausreichend. Im Colon traten h1B1-spezifische Färbungen in den differenzierten Enterozyten am oberen Ende der Krypten auf, im Dünndarm wurden die Epithelzellen der Zotten gefärbt. Die Färbung konzentrierte sich auf das Zytoplasma. Eine ähnliche Verteilung zeigte sich für h1A1 und h1A3, außer daß zusätzlich eine intensive Färbung der Endothelzellen der Kapillaren in der Submukosa des Ileums auftrat. Im Dickdarm war dies nur bei den Kapillaren in den Lymphfollikeln zu erkennen. Die h2A1 war lediglich im Zytoplasma der Epithelzellen der Zotten des Ileums nachzuweisen, während im Colon keine Farbreaktion auftrat. Durch die Verwendung der rekombinanten Indikatorstämme TA1538-h1A1, -h1A3 und -h1B1 und des Ausgangsstammes Salmonella typhimurium TA1538 im Ames-Test wurde gezeigt, daß verschiedene benzylische und allylische Alkohole durch im humanen Colon exprimierte Sulfotransferasen zu Mutagenen aktiviert werden. In den meisten Fällen erwies sich eine der drei Sulfotransferasen als besonders effizient in der Bioaktivierung, während durch die anderen Formen kein oder nur ein schwacher Effekt verursacht wurde. Die Bioaktivierung von Promutagenen durch Sulfotransferasen im Colon muß im Zusammenhang mit der Lokalisation diskutiert werden. Die Zellen im Darm, in denen immunhistochemisch Sulfotransferasen detektiert wurden, haben mit Ausnahme des Endothels je nach Abschnitt eine Lebensdauer von maximal fünf Tagen und machen keine weiteren Zellteilungen mehr durch. Daher sind DNA-Schäden in diesen Zellen ein sehr geringes Risiko für den Organismus. Soweit die reaktiven Metabolite in diesen Zellen gefangen bleiben, kann die Bioaktivierung in diesen Zellen und die Bildung von Addukten als protektiv betrachten werden, da letztere nach wenigen Tagen mit den toten Zellen in das Darmlumen abgegeben werden. Für den Vergleich der Bioaktiverung von Promutagenen durch die Form 1B1 des Menschen und der Ratte wurden aus V79 Lungenfibroblasten des Chinesischen Hamsters abgeleitete Zellinien hergestellt, die je eine der beiden Formen stabil exprimieren. Damit standen 1B1-profiziente Indikatorzellen für den HPRT-Genmutationstest zur Verfügung, und die 1B1-abhängige Bioaktivierung konnte in einem System untersucht werden, die dem eukaryontischen Organismus näher steht als die für die Ames-Tests verwendeten Bakterien. So war z.B. die Sulfotransferase wie im Gewebe im Zytoplasma lokalisiert. Als Modellsubstanzen wurden hierbei die bereits in TA1538-h1B1 mutagen wirkenden benzylischen Alkohole 6-Hydroxymethylbenzo[a]pyren und 4-Hydroxycyclopenta-[def]chrysen getestet. Da die Sensitivität einer Sulfotransferase-exprimierenden V79-Zellinie sowohl durch die Menge an Sulfotransferase als auch durch die Verfügbarkeit des Sulfodonors limitiert sein könnte, wurden die Mutagenitätsexperimente mit V79-r1B1-Zellinien durchgeführt, die sich in ihrer Enzymaktivität um das Zwanzigfache unterschieden: V79-r1B1/A und -/B. Eine starke Erhöhung der Mutantenfrequenz wurde nur in der hoch exprimierenden Zellinie V79-r1B1/A (1019 ± 224 pmol/mg/min) beobachtet, so daß eine gravierende Beeinträchtigung der Sensitivität durch einen Mangel an Kosubstrat ausgeschlossen wurde. In der niedriger exprimierenden Zellinie V79-r1B1/B (57 ± 9 pmol/mg/min) war nur mit 6-Hydroxymethylbenzo[a]pyren ein schwacher Anstieg der Mutantenfrequenz zu erkennen, der mit 0,3 µM bei einer in etwa 100fach höheren Konzentration begann als bei V79-r1B1/A. Die zytosolische Fraktion aus V79-r1B1/B-Zellen enthielt in etwa die dreifache Menge an r1B1-Protein wie die aus Colonmucosa der Ratte. Da zumindest für die humane Mukosa gezeigt wurde, daß die 1B1 nur im einschichtigen Epithel, nicht aber in allen Zellen der Mukosa exprimiert wird, repräsentiert die zytosolische Fraktion aus der Mukosa nur bedingt die Expression in den Epithelzellen und der Vergleich mit den V79-1B1-Zellen ist grob. Im Gegensatz zu V79-r1B1/B war die Zellinie V79-h1B1, die ebenfalls nur mit Darm und Leber vergleichbare Mengen an h1B1 exprimierte, in der Lage, beide benzylischen Alkohole zu aktivieren. Der Erhöhung der Mutantenfrequenz im Vergleich zur KontrollZellinie war ähnlich wie bei der stark exprimierenden Zellinie V79-r1B1/A, erforderte aber 10fach höhere Konzentrationen. Somit unterscheiden sich Mensch und Ratte nicht nur insgesamt in ihrer Ausstattung des Gastrointestinaltrakts mit Sulfotransferasen, auch bei Betrachtung einer einzelnen Form zeigten sich deutliche Unterschiede in der Aktivierung von zwei Promutagenen. Die Ratte ist daher ein ungeeignetes Modell, um die Rolle von Sulfotransferasen bei tumorinitiierenden Prozessen im Darm zu untersuchen. Dies unterstreicht die Bedeutung von rekombinanten in-vitro-Systemen für die Erfassung des humanen Metabolismus von Fremdstoffen. Insgesamt kennt man nur eine geringe Anzahl von Substanzen, die im Tierexperiment Colontumore erzeugen, und mit Ausnahme der heterozyklischen aromatischen Amine sind diese lediglich von experimenteller Bedeutung. Dies spricht für effiziente Schutzmechanismen der Darmmukosa gegenüber Mutagenen und läßt die Frage nach der hohen Inzidenz des Kolorektalkarzinoms offen.
Housing in metabolic cages can induce a pronounced stress response. Metabolic cage systems imply housing mice on metal wire mesh for the collection of urine and feces in addition to monitoring food and water intake. Moreover, mice are single-housed, and no nesting, bedding, or enrichment material is provided, which is often argued to have a not negligible impact on animal welfare due to cold stress. We therefore attempted to reduce stress during metabolic cage housing for mice by comparing an innovative metabolic cage (IMC) with a commercially available metabolic cage from Tecniplast GmbH (TMC) and a control cage. Substantial refinement measures were incorporated into the IMC cage design. In the frame of a multifactorial approach for severity assessment, parameters such as body weight, body composition, food intake, cage and body surface temperature (thermal imaging), mRNA expression of uncoupling protein 1 (Ucp1) in brown adipose tissue (BAT), fur score, and fecal corticosterone metabolites (CMs) were included. Female and male C57BL/6J mice were single-housed for 24 h in either conventional Macrolon cages (control), IMC, or TMC for two sessions. Body weight decreased less in the IMC (females—1st restraint: 6.94%; 2nd restraint: 6.89%; males—1st restraint: 8.08%; 2nd restraint: 5.82%) compared to the TMC (females—1st restraint: 13.2%; 2nd restraint: 15.0%; males—1st restraint: 13.1%; 2nd restraint: 14.9%) and the IMC possessed a higher cage temperature (females—1st restraint: 23.7°C; 2nd restraint: 23.5 °C; males—1st restraint: 23.3 °C; 2nd restraint: 23.5 °C) compared with the TMC (females—1st restraint: 22.4 °C; 2nd restraint: 22.5 °C; males—1st restraint: 22.6 °C; 2nd restraint: 22.4 °C). The concentration of fecal corticosterone metabolites in the TMC (females—1st restraint: 1376 ng/g dry weight (DW); 2nd restraint: 2098 ng/g DW; males—1st restraint: 1030 ng/g DW; 2nd restraint: 1163 ng/g DW) was higher compared to control cage housing (females—1st restraint:
640 ng/g DW; 2nd restraint: 941 ng/g DW; males—1st restraint: 504 ng/g DW; 2nd restraint: 537 ng/g DW). Our results show the stress potential induced by metabolic cage restraint that is markedly influenced by the lower housing temperature. The IMC represents a first attempt to target cold stress reduction during metabolic cage application thereby producing more animal welfare friendly data.
In reconstructed skin and diffusion cell studies, core-multishell nanocarriers (CMS-NC) showed great potential for drug delivery across the skin barrier. Herein, we investigated penetration, release of dexamethasone (DXM), in excised full-thickness human skin with special focus on hair follicles (HF). Four hours and 16 h after topical application of clinically relevant dosages of 10 mu g DXM/cm(2) skin encapsulated in CMS-NC (12 nm diameter, 5.8% loading), presence of DXM in the tissue as assessed by fluorescence microscopy of anti-DXM-stained tissue sections as well as ELISA and HPLC-MS/MS in tissue extracts was enhanced compared to standard LAW-creme but lower compared to DXM aqueous/alcoholic solution. Such enhanced penetration compared to conventional cremes offers high potential for topical therapies, as recurrent applications of corticosteroid solutions face limitations with regard to tolerability and fast drainage. The findings encourage more detailed investigations on where and how the nanocarrier and drug dissociate within the skin and what other factors, e.g. thermodynamic activity, influence the penetration of this formulations. Microscopic studies on the spatial distribution within the skin revealed accumulation in HF and furrows accompanied by limited cellular uptake assessed by flow cytometry (up to 9% of total epidermal cells). FLIM clearly visualized the presence of CMS-NC in the viable epidermis and dermis. When exposed in situ a fraction of up to 25% CD1a(+) cells were found within the epidermal CMS-NC+ population compared to approximately 3% CD1a(+)/CMS-NC+ cells after in vitro exposure in short-term cultures of epidermal cell suspensions. The latter reflects the natural percentage of Langerhans cells (LC) in epidermis suspensions and indicated that CMS-NC were not preferentially internalized by one cell type. The increased CMS-NC+ LC proportion after exposure within the tissue is in accordance with the strategic suprabasal LC-localization. More specifically we postulate that the extensive dendrite meshwork, their position around HF orifices and their capacity to modulate tight junctions facilitated a preferential uptake of CMS-NC by LC within the skin. This newly identified aspect of CMS-NC penetration underlines the potential of CMS-NC for dermatotherapy and encourages further investigations of CMS-NC for the delivery of other molecule classes for which intracellular delivery is even more crucial.
An insufficient adaptive beta-cell compensation is a hallmark of type 2 diabetes (T2D). Primary cilia function as versatile sensory antennae regulating various cellular processes, but their role on compensatory beta-cell replication has not been examined. Here, we identify a significant enrichment of downregulated, cilia-annotated genes in pancreatic islets of diabetes-prone NZO mice as compared with diabetes-resistant B6-ob/ob mice. Among 327 differentially expressed mouse cilia genes, 81 human orthologs are also affected in islets of diabetic donors. Islets of nondiabetic mice and humans show a substantial overlap of upregulated cilia genes that are linked to cell-cycle progression. The shRNA-mediated suppression of KIF3A, essential for ciliogenesis, impairs division of MINE beta cells as well as in dispersed primary mouse and human islet cells, as shown by decreased BrdU incorporation. These findings demonstrate the substantial role of cilia-gene regulation on islet function and T2D risk.
Homocystein (tHcy) gilt als unabhängiger kardiovaskulärer Risikofaktor und korreliert eng mit einer endothelialen Dysfunktion, welche nichtinvasiv mittels der flussinduzierten Vasodilatation (FMD) messbar ist. Experimentelle Hyperhomocysteinämie ist mit einer reduzierten Bioverfügbarkeit von endothelialen Stickstoffmonoxid (NO) bei gleichzeitig erhöhten Spiegeln des kompetetiven Inhibitors der NO-Biosynthese asymmetrisches Dimethylarginin (ADMA) assoziiert. In-vivo senkt eine Östrogenbehandlung neben tHcy auch die ADMA-Spiegel und verbessert signifikant die Endothelfunktion. Hinsichtlich ihrer Wirkung als selektive Östrogenrezeptormodulatoren wird angenommen, dass Phytoöstrogene, speziell Sojaisoflavone, ähnliche Effekte hervorrufen. Innerhalb einer europäischen, multizentrischen, doppelblinden Interventionsstudie an 89 gesunden, postmenopausalen Frauen wurde der Einfluss von Sojaisoflavonen auf den Homocysteinmetabolismus, den Blutdruck und die in-vivo Endothelfunktion untersucht. Die cross-over Studie umfasste zwei achtwöchige Interventionsperioden, die von einer gleichlangen Wash-out-Phase unterbrochen waren. Die Zuteilung zum Isoflavon- (50 mg/d) oder Plazeboregime für die erste Interventionsphase erfolgte randomisiert. Endpunkterhebungen fanden jeweils in den Wochen 0 und 8 der Interventionsperioden statt. Die renale Ausscheidung von Genistein, Daidzein und Equol war während der Isoflavonintervention signifikant erhöht (P>0,001). Die Phyoöstrogene hatten weder einen Effekt auf die tHcy-Konzentration (P=0,286), noch auf ADMA, Erythrozytenfolat und Vitamin B-12 (P>0,05) im Plasma. Während die Summe aus Nitrat und Nitrit (NOx), welche die NO-Bioverfügbarkeit reflektiert, im Verlaufe der Plazebobehandlung abfiel, wurde ein leichter Anstieg bei der Isoflavonsupplementation beobachtet (Delta Wo8-Wo0: -2,60 [-8,75; 2,25] vs. 1,00 [-6,65; 7,85] µmol/L P<0,001), was zu einem signifikanten Behandlungseffekt führte. Weiterhin wurde eine positive Korrelation zwischen ADMA und Vitamin B-12 gefunden (R=0,252; P=0,018). Die flussinduzierte Vasodilatation (P=0,716), ein Maß für die Endothelfunktion, blieb durch die Isoflavonbehandlung unbeeinflusst, obwohl sich diese über die Zeit insgesamt verbesserte (P>0,001). Bis auf einen marginalen Anstieg des systolischen Wertes (P=0,032) im Vergleich zur Plazebobehandlung blieb der Blutdruck während der Isoflavonintervention unverändert. Im Gegensatz zu Östrogen übten Sojaisoflavone weder einen Einfluss auf die in-vivo Endothelfunktion noch auf die traditionellen und neuen kardiovaskulären Risikofaktoren den Blutdruck, tHcy und ADMA aus. Demzufolge ist der gesundheitliche Nutzen isolierter Isoflavone hinsichtlich einer Prävention hormonmangelbedingter Erkrankungen in gesunden postmenopausalen Frauen fraglich.
Vitamin E ist der Überbegriff für 4 Tocopherole (α, β, γ und δ) sowie 4 Tocotrienole (α, β, γ und δ), die als gemeinsames Merkmal ein Chromanolringsystem sowie eine gesättigte (Tocopherole) bzw. ungesättigte (Tocotrienole) Seitenkette aufweisen. Neben ihrer antioxidativen Wirkung (Schutz von Membranen vor Lipidperoxidaton) konnten für einige Vitamin E - Formen auch eine Reihe von hochspezifischen, nicht-antioxidativen Wirkungen in vitro nachgewiesen werden. Meist bleibt jedoch unklar, ob ein solcher Effekt auch in vivo, also im Tiermodel oder direkt im Menschen, gefunden werden kann. In erster Linie müsste hierbei geklärt werden, ob die jeweilige Vitamin E - Form auch bioverfügbar, also in für eine Wirkung ausreichender Konzentration im Organismus vorhanden ist, oder aber vorher eliminiert und ausgeschieden wird. In dieser Doktorarbeit wurden deshalb wichtige Grundlagen zum Abbau der Tocopherole und Tocotrienole erarbeitet. • In HepG2-Zellen konnte der Abbau der Tocotrienole mit Hilfe flüssig- sowie gaschromatographischer Analysemethoden vollständig aufgeklärt werden. Wie sich hierbei ergab, verläuft der Abbau weitgehend in Analogie zum Abbau der Tocopherole über eine durch Cytochrom P450 katalysierte initiale ω-Hydroxylierung mit 5 nachfolgenden β-Oxidationsschritten. • In vitro konnten in HepG2 – Zellen die Abbauraten der verschiedenen Vitamin E - Formen bestimmt werden. Dies nahmen in folgender Reihenfolge zu: α-Tocopherol < γ-Tocopherol < α-Tocotrienol < γ-Tocotrienol. • Wie sich mit Hilfe eines mit Cytochrom P450 hochangereicherten Homogenats aus Rattenlebern ergab, stellt die initiale ω-Hydroxylierung einen geschwindigkeitsbestimmenden Schritt des Abbaus dar: α-Tocopherol wurde weit langsamer hydroxyliert als alle anderen Vitamin E – Formen. • Der unterschiedliche Abbau von α-Tocopherol und γ-Tocotrienol konnte auch im Mäuseversuch in vivo bestätigt werden. Nach Fütterung von Mäusen mit α-Tocopherol wurden nur geringe Mengen von α-Tocopherolmetaboliten im Urin der Mäuse gefunden, während nach Applikation von γ-Tocotrienol hohe Konzentrationen der γ-Tocotrienolmetabolite nachgewiesen wurden. In Plasma und Leber wiederum wurden (dem Futtergehalt entsprechende) hohe α-Tocopherolkonzentrationen entdeckt, während γ-Tocotrienol selbst nach hoher Gabe nicht oder nur in Spuren nachweisbar war. In HepG2 – Zellen konnte gezeigt werden, dass γ-Tocotrienol eine cytotoxische Wirkung auf die Hepatocarcinoma-Zelllinie HepG2 entfalten kann, indem durch die Aktivierung der proteolytischen Caspase 3 die Induktion des programmierten Zelltodes (Apoptose) ausgelöst wird. Abschliessend lässt sich festhalten, dass der Körper lediglich das natürliche α-Tocopherol vor dem Abbau bewahrt, die anderen Vitamin E – Formen jedoch als Fremdstoffe behandelt und rapide ausscheidet. Als doppelter Schutz vor Verlust des “wertvollen” α-Tocopherol dienen hierbei das α-Tocopherol Transfer Protein sowie die in dieser Arbeit gefundenen Unterschiede im ersten Schritt des Abbaus, der Cytochrom P450 - katalysierten ω-Hydroxylierung. Beides erklärt die bevorzugte Retention von α-Tocopherol im Organsimus und seine hohe Bioaktivität. Will man deshalb in vitro Ergebnisse anderer Vitamin E – Formen auf die in vivo Situation übertragen, muss man die geringe Bioverfügbarkeit dieser Substanzen berücksichtigen.
Dermal Delivery of the High-Molecular-Weight Drug Tacrolimus by Means of Polyglycerol-Based Nanogels

Polyglycerol-based thermoresponsive nanogels (tNGs) have been shown to have excellent skin hydration properties and to be valuable delivery systems for sustained release of drugs into skin. In this study, we compared the skin penetration of tacrolimus formulated in tNGs with a commercial 0.1% tacrolimus ointment. The penetration of the drug was investigated in ex vivo abdominal and breast skin, while different methods for skin barrier disruption were investigated to improve skin permeability or simulate inflammatory conditions with compromised skin barrier. The amount of penetrated tacrolimus was measured in skin extracts by liquid chromatography tandem-mass spectrometry (LC-MS/MS), whereas the inflammatory markers IL-6 and IL-8 were detected by enzyme-linked immunosorbent assay (ELISA). Higher amounts of tacrolimus penetrated in breast as compared to abdominal skin or in barrier-disrupted as compared to intact skin, confirming that the stratum corneum is the main barrier for tacrolimus skin penetration. The anti-proliferative effect of the penetrated drug was measured in skin tissue/Jurkat cells co-cultures. Interestingly, tNGs exhibited similar anti-proliferative effects as the 0.1% tacrolimus ointment. We conclude that polyglycerol-based nanogels represent an interesting alternative to paraffin-based formulations for the treatment of inflammatory skin conditions.

Wheat alpha-amylase/trypsin inhibitors remain a subject of interest considering the latest findings showing their implication in wheat-related non-celiac sensitivity (NCWS). Understanding their functions in such a disorder is still unclear and for further study, the need for pure ATI molecules is one of the limiting problems. In this work, a simplified approach based on the successive fractionation of ATI extracts by reverse phase and ion exchange chromatography was developed. ATIs were first extracted from wheat flour using a combination of Tris buffer and chloroform/methanol methods. The separation of the extracts on a C18 column generated two main fractions of interest F1 and F2. The response surface methodology with the Doehlert design allowed optimizing the operating parameters of the strong anion exchange chromatography. Finally, the seven major wheat ATIs namely P01083, P17314, P16850, P01085, P16851, P16159, and P83207 were recovered with purity levels (according to the targeted LC-MS/MS analysis) of 98.2 ± 0.7; 98.1 ± 0.8; 97.9 ± 0.5; 95.1 ± 0.8; 98.3 ± 0.4; 96.9 ± 0.5, and 96.2 ± 0.4%, respectively. MALDI-TOF-MS analysis revealed single peaks in each of the pure fractions and the mass analysis yielded deviations of 0.4, 1.9, 0.1, 0.2, 0.2, 0.9, and 0.1% between the theoretical and the determined masses of P01083, P17314, P16850, P01085, P16851, P16159, and P83207, respectively. Overall, the study allowed establishing an efficient purification process of the most important wheat ATIs. This paves the way for further in-depth investigation of the ATIs to gain more knowledge related to their involvement in NCWS disease and to allow the absolute quantification in wheat samples.
