Rehabilitationspädagogik

Die Rehabilitationspädagogik ist eine jüngere eigenständige Hybridwissenschaft im Feld der Humanwissenschaften. Sie setzt theoriebildend im Sinne des Neunten Buchs Sozialgesetzbuch (SGB IX) an den längerfristigen Folgen einer Krankheit oder eines biologischen Mangels an. Dabei orientiert sie sich konzeptionell zum Beispiel an der UN-Behindertenrechtskonvention (UN-BRK) und an der International Classification of Functioning, Disability and Health (ICF). Des Weiteren an den Konzepten der Humanontogenetik von K.-F. Wessel, insbesondere: dem ganzen Menschen, der Hierarchie der Kompetenzen, den sensiblen Phasen und der Souveränität.
Die Rehabilitationspädagogik ist Bestandteil der komplexen gesundheitlichen Rehabilitation und eine Tochterdisziplin der allgemeinen Pädagogik. Bei ihrem rehabilitationspädagogischen Prozess gilt das Richtziel, die umfassende Teilhabe des Menschen an individuellen Lebensbereichen durch rehabilitationspädagogische Mittel, Methoden und Organisationsformen zu unterstützen.
Die Dissertation setzt sich mittels Methoden der Hermeneutik mit der DDR-Rehabilitationspädagogik von K.- P. Becker und Autorenkollektiv kritisch-konstruktiv auseinander. Sie legt eine aktuelle fortführende Theorie der Rehabilitationspädagogik unter der Berücksichtigung der UN-BRK, der ICF und des SGB IX vor und liefert eine neue Sichtweise auf die Rehabilitationspädagogik aus historischer und aktueller Perspektive.
Participation has become an orthodoxy in the field of development, an essential element of projects and programmes. This book analyses participation in development interventions as an institutionalised expectation – a rationalized myth – and examines how organisations on different levels of government process it. At least two different objectives of participation are appropriate and legitimate for international organisations in the field: the empowerment of local beneficiaries and the achievement of programme goals. Both integrate participatory forums into the organisational logic of development interventions. Local administrations react to the institutionalised expectation with means-ends decoupling, where participatory forums are implemented superficially but de facto remain marginalised in local administrative processes and activities. The book furthermore provides a thick description of the organisationality of participation in development interventions. Participatory forums are shown to be a form of partial organisation. They establish an order in the relationship between administrations and citizens through the introduction of rules and the creation of a defined membership. At the same time, this order is found to be fragile and subject to criticism and negotiation.
Die Dissertation legt ihren Schwerpunkt auf die synchronische und diachronische Variation im Gebrauch der französischen Kausalkonjunktion parce que sowie auf die Interaktion mit den außersprachlichen Variablen Alter und sozioprofessionelle Kategorie. Basierend auf vorausgehenden makrodiachronischen Studien, die Anhaltspunkte dafür liefern, dass die Konjunktion einen Prozess der Pragmatikalisierung durchlaufen hat und weiterhin durchläuft, wurde ein Untersuchungskorpus von 56 Interviews aus den diachronisch distinkten Korpora ESLO1, ESLO2 und LangAge extrahiert. Dieses Untersuchungskorpus diente als Grundlage für Panelstudien und Trendstudien, die darauf ausgerichtet waren, die Pragmatikalisierung von parce que aus einem mikrodiachronischen Gesichtspunkt zu verifizieren. Zusätzlich zu der diachronischen Perspektive wurde eine synchronische Perspektive eingenommen, um die Variation im Gebrauch der Konjunktion so einem diachronischen Phänomen wie dem age grading oder der apparent time zuordnen zu können. Ausgehend von der Theorie der Konstruktionsgrammatik wurden parce que enthaltende Konstruktionen bottom-up annotiert und in fünf Pragmatikalitätsgrade kategorisiert (pra0–pra4). Diese wurden anschließend quantifiziert und in Abhängigkeit des Geburtsjahres und der sozioprofessionellen Kategorie der (männlichen) Sprecher mithilfe mehrerer R-Modelle wie ctrees, trees, lm, hclust und kmeans analysiert.
Die Frequenzentwicklung der Pragmatikalitätsgrade bestätigte die Pragmatikalisierungshypothese in einem mikrodiachronischen Rahmen. Zudem konnte ein quantitativer Rückgang im Gebrauch der Konstruktionen am nicht- oder weniger pragmatikalisierten (pra0, pra1) Pol festgestellt werden, während Verwendungsweisen höherer Pragmatikalisierungsgrade (pra2–pra4) über 40 Jahre vergleichsweise stabil blieben.
Obwohl für pra2 kein signifikanter Wandel hervortrat, wies dessen Entwicklung bei den Sprechern im mittleren Lebensalter sowie das synchronische Muster in Abhängigkeit von Alter (oder Geburtsjahr) und von sozioprofessioneller Kategorie dennoch in Richtung einer zugrundeliegenden diachronischen Variation. Diese könnte als ein durch die sozialen Transformationen der 1960er und 1970er Jahre katalysiertes Phänomen des age grading interpretiert werden. Für die näher am pragmatischen Pol situierten Gebrauchsweisen (pra3 und pra4) konnte keine klare Tendenz ermittelt werden.
Die Ergebnisse fordern diachronische Konzepte wie age grading und apparent time heraus, indem sie die Simplizität der zugrundeliegenden Mechanismen sowie die gängigen Methoden, diese zu identifizieren, infrage stellen.
Selfsustained oscillations are some of the most commonly observed phenomena in biological systems. They emanate from non-linear systems in a heterogeneous environment and can be described by the theory of dynamical systems. Part of this theory considers reduced models of the oscillator dynamics by means of amplitudes and a phase variable. Such variables are highly attractive for theoretical and experimental studies. Theoretically these variables correspond to an integrable linearization of the generally non-linear system. Experimentally, there exist well established approaches to extract phases from oscillator signals. Notably, one can define phase models also for networks of oscillators. One highly active field examines effects of non-local coupling among oscillators, which is thought to play a key role in networks with strong coupling. The dissertation introduces and expands the knowledge about high-order phase coupling in networks of oscillators. Mathematical calculations consider the Stuart-Landau oscillator. A novel phase estimation scheme for direct observations of an oscillator dynamics is introduced based on numerics. A numerical study of high-order phase coupling applies a Fourier fit for the Stuart-Landau and for the van-der-Pol oscillator. The numerical approach is finally tested on observation-based phase estimates of the Morris-Lecar neuron. A popular approach for the construction of phases from signals is based on phase demodulation by means of the Hilbert transform. Generally, observations of oscillations contain a small and generic variation of their amplitude. The work presents a way to quantify how much the variations of signal amplitude spoil a phase demodulation procedure. For the ideal case of phase modulated signals, amplitude modulations vanish. However, the Hilbert transform produces artificial variations of the reconstructed amplitude even in this case. The work proposes a novel procedure called Iterative Hilbert Transform Embedding to obtain an optimal demodulation of signals. The text presents numerous examples and tests of application for the method, covering multicomponent signals, observables of highly stable limit cycle oscillations and noisy phase dynamics. The numerical results are supported by a spectral theory of convergence for weak phase modulations.
Die vorliegende Arbeit beschäftigt sich mit der Synthese von Disulfiden, der Thiol-Disulfid Metathesereaktion als Möglichkeit, Polymere zu funktionalisieren, und der Synthese von Polydisulfiden. Im ersten Teil der Arbeit wird die Aminolyse von RAFT-Polymeren und die Abhängigkeit der Polymer-Polymer Disulfidbildung von der Molmasse untersucht. Dabei wurde durch die Aufnahme von Reaktionskinetiken mittels Gel-Permeations-Chromatographie (GPC) festgestellt, dass je länger die Polymerketten sind, desto weniger Disulfid Polymerkopplung tritt auf. RAFT-Polymere werden oft genutzt, um die RAFT-Polymer Endgruppe nach der Polymerisation zu modifizieren oder in einer chemischen Reaktion zu funktionalisieren. Hier kann die Aminolyse in Anwesenheit von kurzkettigen Disulfiden, wie zum Beispiel Cystin, durchgeführt werden, um die Bildung von Polymer-Polymer Disulfiden vollständig zu unterdrücken und ein endgruppenfunktionalisiertes Polymer zu erhalten. Bei dieser Reaktion greift das bei der Aminolyse entstehende Polymerthiolat die kurzkettigen Disulfide an, und es kommt zur Bildung von funktionalisierten Polymeren. Es wurde ein Polyethylenglykoldisulfid eingesetzt, um ein amphiphiles Blockcopolymer zu erhalten. Als RAFT-Polymer wurde Polystyrol (PS) verwendet, und es konnte die Bildung von Polystyrol-Polyethylenglykol Copolymeren nachgewiesen werden. Das amphiphile Polymer bildet im wässrigen Medium Vesikel. Die Oberfläche der Vesikel konnte mittels der Thiol-Disulfid Metathese umfunktionalisiert werden. Die Aminolyse von PS RAFT-Polymeren mit einem Polylaktiddisulfid oder einem Polybenzylglutamatdisulfid ergab Polystyrol-block-Polyester und Polystyrol-block-Polyaminosäuren Copolymere. Im zweiten Teil der Arbeit liegt der Fokus auf der Synthese von Polydisulfiden und ihren thermischen Eigenschaften. Es wurden verschiedene Alkyldithiole synthetisiert und mittels Wasserstoffperoxid und Triethylamin polymerisiert. Dabei konnte gezeigt werden, dass die Polymere teilkristallin sind und dass der Schmelzpunkt und die Kristallinität der Polymere mit steigender Alkylkettenlänge zwischen den Disulfidbindungen zunehmen. Die Möglichkeit einer Polymerkettenerweiterung nach der Polymerisation ist mit diesem System gegeben. Die Abbaubarkeit der Polydisulfide konnte durch den Einsatz von Thiolen im basischen Milieu gezeigt werden.

To meet the demands of a growing world population while reducing carbon dioxide (CO2) emissions, it is necessary to capture CO2 and convert it into value-added compounds. In recent years, metabolic engineering of microbes has gained strong momentum as a strategy for the production of valuable chemicals. As common microbial feedstocks like glucose directly compete with human consumption, the one carbon (C1) compound formate was suggested as an alternative feedstock. Formate can be easily produced by various means including electrochemical reduction of CO2 and could serve as a feedstock for microbial production, hence presenting a novel entry point for CO2 to the biosphere and a storage option for excess electricity. Compared to the gaseous molecule CO2, formate is a highly soluble compound that can be easily handled and stored. It can serve as a carbon and energy source for natural formatotrophs, but these microbes are difficult to cultivate and engineer. In this work, I present the results of several projects that aim to establish efficient formatotrophic growth of E. coli – which cannot naturally grow on formate – via synthetic formate assimilation pathways. In the first study, I establish a workflow for growth-coupled metabolic engineering of E. coli. I demonstrate this approach by presenting an engineering scheme for the PFL-threonine cycle, a synthetic pathway for anaerobic formate assimilation in E. coli. The described methods are intended to create a standardized toolbox for engineers that aim to establish novel metabolic routes in E. coli and related organisms. The second chapter presents a study on the catalytic efficiency of C1-oxidizing enzymes in vivo. As formatotrophic growth requires generation of both energy and biomass from formate, the engineered E. coli strains need to be equipped with a highly efficient formate dehydrogenase, which provides reduction equivalents and ATP for formate assimilation. I engineered a strain that cannot generate reducing power and energy for cellular growth, when fed on acetate. Under this condition, the strain depends on the introduction of an enzymatic system for NADH regeneration, which could further produce ATP via oxidative phosphorylation. I show that the strain presents a valuable testing platform for C1-oxidizing enzymes by testing different NAD-dependent formate and methanol dehydrogenases in the energy auxotroph strain. Using this platform, several candidate enzymes with high in vivo activity, were identified and characterized as potential energy-generating systems for synthetic formatotrophic or methylotrophic growth in E. coli. In the third chapter, I present the establishment of the serine threonine cycle (STC) – a synthetic formate assimilation pathway – in E. coli. In this pathway, formate is assimilated via formate tetrahydrofolate ligase (FtfL) from Methylobacterium extorquens (M. extorquens). The carbon from formate is attached to glycine to produce serine, which is converted into pyruvate entering central metabolism. Via the natural threonine synthesis and cleavage route, glycine is regenerated and acetyl-CoA is produced as the pathway product. I engineered several selection strains that depend on different STC modules for growth and determined key enzymes that enable high flux through threonine synthesis and cleavage. I could show that expression of an auxiliary formate dehydrogenase was required to achieve growth via threonine synthesis and cleavage on pyruvate. By overexpressing most of the pathway enzymes from the genome, and applying adaptive laboratory evolution, growth on glycine and formate was achieved, indicating the activity of the complete cycle. The fourth chapter shows the establishment of the reductive glycine pathway (rGP) – a short, linear formate assimilation route – in E. coli. As in the STC, formate is assimilated via M. extorquens FtfL. The C1 from formate is condensed with CO2 via the reverse reaction of the glycine cleavage system to produce glycine. Another carbon from formate is attached to glycine to form serine, which is assimilated into central metabolism via pyruvate. The engineered E. coli strain, expressing most of the pathway genes from the genome, can grow via the rGP with formate or methanol as a sole carbon and energy source.
Plant-derived Transcription Factors for Orthologous Regulation of Gene Expression in the Yeast Saccharomyces cerevisiae
Control of gene expression by transcription factors (TFs) is central in many synthetic biology projects where tailored expression of one or multiple genes is often needed. As TFs from evolutionary distant organisms are unlikely to affect gene expression in a host of choice, they represent excellent candidates for establishing orthogonal control systems. To establish orthogonal regulators for use in yeast (Saccharomyces cerevisiae), we chose TFs from the plant Arabidopsis thaliana. We established a library of 106 different combinations of chromosomally integrated TFs, activation domains (yeast GAL4 AD, herpes simplex virus VP64, and plant EDLL) and synthetic promoters harbouring cognate cis-regulatory motifs driving a yEGFP reporter. Transcriptional output of the different driver / reporter combinations varied over a wide spectrum, with EDLL being a considerably stronger transcription activation domain in yeast, than the GAL4 activation domain, in particular when fused to Arabidopsis NAC TFs. Notably, the strength of several NAC - EDLL fusions exceeded that of the strong yeast TDH3 promoter by 6- to 10-fold. We furthermore show that plant TFs can be used to build regulatory systems encoded by centromeric or episomal plasmids. Our library of TF – DNA-binding site combinations offers an excellent tool for diverse synthetic biology applications in yeast.
COMPASS: Rapid combinatorial optimization of biochemical pathways based on artificial transcription factors
We established a high-throughput cloning method, called COMPASS for COMbinatorial Pathway ASSembly, for the balanced expression of multiple genes in Saccharomyces cerevisiae. COMPASS employs orthogonal, plant-derived artificial transcription factors (ATFs) for controlling the expression of pathway genes, and homologous recombination-based cloning for the generation of thousands of individual DNA constructs in parallel. The method relies on a positive selection of correctly assembled pathway variants from both, in vivo and in vitro cloning procedures. To decrease the turnaround time in genomic engineering, we equipped COMPASS with multi-locus CRISPR/Cas9-mediated modification capacity. In its current realization, COMPASS allows combinatorial optimization of up to ten pathway genes, each transcriptionally controlled by nine different ATFs spanning a 10-fold difference in expression strength. The application of COMPASS was demonstrated by generating cell libraries producing beta-carotene and co-producing beta-ionone and biosensor-responsive naringenin. COMPASS will have many applications in other synthetic biology projects that require gene expression balancing.
CaPRedit: Genome editing using CRISPR-Cas9 and plant-derived transcriptional regulators for the redirection of flux through the FPP branch-point in yeast. Technologies developed over the past decade have made Saccharomyces cerevisiae a promising platform for production of different natural products. We developed CRISPR/Ca9- and plant derived regulator-mediated genome editing approach (CaPRedit) to greatly accelerate strain modification and to facilitate very low to very high expression of key enzymes using inducible regulators. CaPRedit can be implemented to enhance the production of yeast endogenous or heterologous metabolites in the yeast S. cerevisiae. The CaPRedit system aims to faciltiate modification of multiple targets within a complex metabolic pathway through providing new tools for increased expression of genes encoding rate-limiting enzymes, decreased expression of essential genes, and removed expression of competing pathways. This approach is based on CRISPR/Cas9-mediated one-step double-strand breaks to integrate modules containing IPTG-inducible plant-derived artificial transcription factor and promoter pair(s) in a desired locus or loci. Here, we used CaPRedit to redirect the yeast endogenous metabolic flux toward production of farnesyl diphosphate (FPP), a central precursor of nearly all yeast isoprenoid products, by overexpression of the enzymes lead to produce FPP from glutamate. We found significantly higher beta-carotene accumulation in the CaPRedit-mediated modified strain than in the wild type (WT) strain. More specifically, CaPRedit_FPP 1.0 strain was generated, in which three genes involved in FPP synthesis, tHMG1, ERG20, and GDH2, were inducibly overexpressed under the control of strong plant-derived ATFPs. The beta–carotene accumulated in CaPRedit_FPP 1.0 strain to a level 1.3-fold higher than the previously reported optimized strain that carries the same overexpressed genes (as well as additional genetic modifications to redirect yeast endogenous metabolism toward FPP production). Furthermore, the genetic modifications implemented in CaPRedit_FPP 1.0 strain resulted in only a very small growth defect (growth rate relative to the WT is ~ -0.03).
