Point-of-care and in-vivo bio-diagnostic tools are the current need for the present critical scenarios in the healthcare industry. The past few decades have seen a surge in research activities related to solving the challenges associated with precise on-site bio-sensing. Cutting-edge fiber optic technology enables the interaction of light with functionalized fiber surfaces at remote locations to develop a novel, miniaturized and cost-effective lab on fiber technology for bio-sensing applications. The recent remarkable developments in the field of nanotechnology provide innumerable functionalization methodologies to develop selective bio-recognition elements for label free biosensors. These exceptional methods may be easily integrated with fiber surfaces to provide highly selective light-matter interaction depending on various transduction mechanisms. In the present review, an overview of optical fiber-based biosensors has been provided with focus on physical principles used, along with the functionalization protocols for the detection of various biological analytes to diagnose the disease. The design and performance of these biosensors in terms of operating range, selectivity, response time and limit of detection have been discussed. In the concluding remarks, the challenges associated with these biosensors and the improvement required to develop handheld devices to enable direct target detection have been highlighted.
We report a comparison of two photonic techniques for single-molecule sensing: fluorescence nanoscopy and optoplasmonic sensing. As the test system, oligonucleotides with and without fluorescent labels are transiently hybridized to complementary "docking" strands attached to gold nanorods. Comparing the measured single-molecule kinetics helps to examine the influence of the fluorescent labels as well as factors arising from different sensing geometries. Our results demonstrate that DNA dissociation is not significantly altered by the fluorescent labels and that DNA association is affected by geometric factors in the two techniques. These findings open the door to exploiting plasmonic sensing and fluorescence nanoscopy in a complementary fashion, which will aid in building more powerful sensors and uncovering the intricate effects that influence the behavior of single molecules.
Es wird ein Überblick über verschiedene spektroskopische Techniken, mit denen dünne organische Schichten, wie sie in der Fotovoltaik, bei Leuchtdioden oder organischen Halbleitern Anwendung finden, gegeben. Mit einfachen Zusatzgeräten lassen sich Schichtdicke, Schichtaufbau und Zusammensetzung untersuchen. Die Schichtdicke kann monomolekular sein. Unter bestimmten Voraussetzungen sind einzelne Moleküle in einer Schicht charakterisierbar.
LCST-type synthetic thermoresponsive polymers can reversibly respond to certain stimuli in aqueous media with a massive change of their physical state. When fluorophores, that are sensitive to such changes, are incorporated into the polymeric structure, the response can be translated into a fluorescence signal. Based on this idea, this thesis presents sensing schemes which transduce the stimuli-induced variations in the solubility of polymer chains with covalently-bound fluorophores into a well-detectable fluorescence output. Benefiting from the principles of different photophysical phenomena, i.e. of fluorescence resonance energy transfer and solvatochromism, such fluorescent copolymers enabled monitoring of stimuli such as the solution temperature and ionic strength, but also of association/disassociation mechanisms with other macromolecules or of biochemical binding events through remarkable changes in their fluorescence properties. For instance, an aqueous ratiometric dual sensor for temperature and salts was developed, relying on the delicate supramolecular assembly of a thermoresponsive copolymer with a thiophene-based conjugated polyelectrolyte. Alternatively, by taking advantage of the sensitivity of solvatochromic fluorophores, an increase in solution temperature or the presence of analytes was signaled as an enhancement of the fluorescence intensity. A simultaneous use of the sensitivity of chains towards the temperature and a specific antibody allowed monitoring of more complex phenomena such as competitive binding of analytes. The use of different thermoresponsive polymers, namely poly(N-isopropylacrylamide) and poly(meth)acrylates bearing oligo(ethylene glycol) side chains, revealed that the responsive polymers differed widely in their ability to perform a particular sensing function. In order to address questions regarding the impact of the chemical structure of the host polymer on the sensing performance, the macromolecular assembly behavior below and above the phase transition temperature was evaluated by a combination of fluorescence and light scattering methods. It was found that although the temperature-triggered changes in the macroscopic absorption characteristics were similar for these polymers, properties such as the degree of hydration or the extent of interchain aggregations differed substantially. Therefore, in addition to the demonstration of strategies for fluorescence-based sensing with thermoresponsive polymers, this work highlights the role of the chemical structure of the two popular thermoresponsive polymers on the fluorescence response. The results are fundamentally important for the rational choice of polymeric materials for a specific sensing strategy.
Eine Nutzung der optischen Anisotropie dünner Schichten ist vor allem für die Displaytechnologie, die optische Datenspeicherung und für optische Sicherheitselemente von hoher Bedeutung. Diese Doktorarbeit befasst sich mit theoretischen und experimentellen Untersuchung von dreidimensionaler Anisotropie und dabei insbesondere mit der Untersuchung von lichtinduzierter dreidimensionaler Anisotropie in organischen dünnen Polymer-Schichten. Die gewonnenen Erkentnisse und entwickelten Methoden können wertvolle Beiträge für Optimierungsprozesse, wie bei der Kompensation der Blickwinkelabhängigkeit von Flüssigkristall-Displays, liefern. Die neue Methode der Immersions-Transmissions-Ellipsometrie (ITE) zur Untersuchung von dünneren Schichten wurde im Rahmen dieser Dissertation entwickelt. Diese Methode gestattet es, in Kombination mit konventioneller Reflexions- und Transmissionsellipsometrie, die absoluten dreidimensionalen Brechungsindices einer biaxialen Schicht zu bestimmen. Erstmals gelang es damit, das dreidimensionale Brechungsindexellipsoid von transparenten, dünneren (150 nm) Filmen hochgenau (drei Stellen hinter dem Komma) zu bestimmen. Die ITE-Methode hat demzufolge das Potential, auch bei noch dünneren Schichten mit Gewinn eingesetzt werden zu können. Die lichtinduzierte Generierung von dreidimensionaler Anisotropie wurde in dünnen Schichten von azobenzenhaltigen und zimtsäurehaltigen, amorphen und flüssig-kristallinen Homo- und Copolymeren untersucht. Erstmals wurden quantitative Untersuchungen zur Änderung von lichtinduzierten, dreidimensionalen Anisotropien in dünnen Schichten von azobenzenhaltigen und zimtsäurehaltigen Polymeren bei Tempern oberhalb der Glastemperatur durchgeführt. Bei vielen der untersuchten Polymere war die dreidimensionale Ordnung nach dem Bestrahlen mit polarisiertem Licht und anschließendem Tempern oberhalb der Glastemperatur scheinbar von der Schichtdicke abhängig. Die Ursache liegt wohl in der, mit der neuentwickelten ITE-Methode detektierten, planaren Ausgangsorientierung der aufgeschleuderten dünneren Schichten. Um Verkippungs-Gradienten in dickeren Polymerschichten in ihrem Verlauf zu bestimmen, wurde eine spezielle Methode unter Benutzung der Wellenleitermoden-Spektroskopie entwickelt. Quantenchemisch bestimmte, maximal induzierbare Doppelbrechungen in flüssig-kristallinen Polymeren wurden mit den experimentell gefundenen Ordnungen verglichen.
