A catalog of genetic loci associated with kidney function from analyses of a million individuals

Chronic kidney disease (CKD) is responsible for a public health burden with multi-systemic complications. Through transancestry meta-analysis of genome-wide association studies of estimated glomerular filtration rate (eGFR) and independent replication (n = 1,046,070), we identified 264 associated loci (166 new). Of these,147 were likely to be relevant for kidney function on the basis of associations with the alternative kidney function marker blood urea nitrogen (n = 416,178). Pathway and enrichment analyses, including mouse models with renal phenotypes, support the kidney as the main target organ. A genetic risk score for lower eGFR was associated with clinically diagnosed CKD in 452,264 independent individuals. Colocalization analyses of associations with eGFR among 783,978 European-ancestry individuals and gene expression across 46 human tissues, including tubulo-interstitial and glomerular kidney compartments, identified 17 genes differentially expressed in kidney. Fine-mapping highlighted missense driver variants in 11 genes and kidney-specific regulatory variants. These results provide a comprehensive priority list of molecular targets for translational research.
JavaScript is the most popular programming language for web applications. Static analysis of JavaScript applications is highly challenging due to its dynamic language constructs and event-driven asynchronous executions, which also give rise to many security-related bugs. Several static analysis tools to detect such bugs exist, however, research has not yet reported much on the precision and scalability trade-off of these analyzers. As a further obstacle, JavaScript programs structured in Node. js modules need to be collected for analysis, but existing bundlers are either specific to their respective analysis tools or not particularly suitable for static analysis.
Evaluating creativity of verbal responses or texts is a challenging task due to psychometric issues associated with subjective ratings and the peculiarities of textual data. We explore an approach to objectively assess the creativity of responses in a sentence generation task to 1) better understand what language-related aspects are valued by human raters and 2) further advance the developments toward automating creativity evaluations. Over the course of two prior studies, participants generated 989 four-word sentences based on a four-letter prompt with the instruction to be creative. We developed an algorithm that scores each sentence on eight different metrics including 1) general word infrequency, 2) word combination infrequency, 3) context-specific word uniqueness, 4) syntax uniqueness, 5) rhyme, 6) phonetic similarity, and similarity of 7) sequence spelling and 8) semantic meaning to the cue. The text metrics were then used to explain the averaged creativity ratings of eight human raters. We found six metrics to be significantly correlated with the human ratings, explaining a total of 16% of their variance. We conclude that the creative impression of sentences is partly driven by different aspects of novelty in word choice and syntax, as well as rhythm and sound, which are amenable to objective assessment.
“Financial Analysis” is an online course designed for professionals consisting of three MOOCs, offering a professionally and institutionally recognized certificate in finance. The course is open but not free of charge and attracts mostly professionals from the banking industry. The primary objective of this study is to identify indicators that can predict learners at high risk of failure. To achieve this, we analyzed data from a previous course that had 875 enrolled learners and involve in the course during Fall 2021. We utilized correspondence analysis to examine demographic and behavioral variables.
The initial results indicate that demographic factors have a minor impact on the risk of failure in comparison to learners’ behaviors on the course platform. Two primary profiles were identified: (1) successful learners who utilized all the documents offered and spent between one to two hours per week, and (2) unsuccessful learners who used less than half of the proposed documents and spent less than one hour per week. Between these groups, at-risk students were identified as those who used more than half of the proposed documents and spent more than two hours per week. The goal is to identify those in group 1 who may be at risk of failing and those in group 2 who may succeed in the current MOOC, and to implement strategies to assist all learners in achieving success.
Generating a novel and descriptive caption of an image is drawing increasing interests in computer vision, natural language processing, and multimedia communities. In this work, we propose an end-to-end trainable deep bidirectional LSTM (Bi-LSTM (Long Short-Term Memory)) model to address the problem. By combining a deep convolutional neural network (CNN) and two separate LSTM networks, our model is capable of learning long-term visual-language interactions by making use of history and future context information at high-level semantic space. We also explore deep multimodal bidirectional models, in which we increase the depth of nonlinearity transition in different ways to learn hierarchical visual-language embeddings. Data augmentation techniques such as multi-crop, multi-scale, and vertical mirror are proposed to prevent over-fitting in training deep models. To understand how our models "translate" image to sentence, we visualize and qualitatively analyze the evolution of Bi-LSTM internal states over time. The effectiveness and generality of proposed models are evaluated on four benchmark datasets: Flickr8K, Flickr30K, MSCOCO, and Pascal1K datasets. We demonstrate that Bi-LSTM models achieve highly competitive performance on both caption generation and image-sentence retrieval even without integrating an additional mechanism (e.g., object detection, attention model). Our experiments also prove that multi-task learning is beneficial to increase model generality and gain performance. We also demonstrate the performance of transfer learning of the Bi-LSTM model significantly outperforms previous methods on the Pascal1K dataset.
Thematic maps are a common tool to visualize semantic data with a spatial reference. Combining thematic data with a geometric representation of their natural reference frame aids the viewer’s ability in gaining an overview, as well as perceiving patterns with respect to location; however, as the amount of data for visualization continues to increase, problems such as information overload and visual clutter impede perception, requiring data aggregation and level-of-detail visualization techniques. While existing aggregation techniques for thematic data operate in a 2D reference frame (i.e., map), we present two aggregation techniques for 3D spatial and spatiotemporal data mapped onto virtual city models that hierarchically aggregate thematic data in real time during rendering to support on-the-fly and on-demand level-of-detail generation. An object-based technique performs aggregation based on scene-specific objects and their hierarchy to facilitate per-object analysis, while the scene-based technique aggregates data solely based on spatial locations, thus supporting visual analysis of data with arbitrary reference geometry. Both techniques can apply different aggregation functions (mean, minimum, and maximum) for ordinal, interval, and ratio-scaled data and can be easily extended with additional functions. Our implementation utilizes the programmable graphics pipeline and requires suitably encoded data, i.e., textures or vertex attributes. We demonstrate the application of both techniques using real-world datasets, including solar potential analyses and the propagation of pressure waves in a virtual city model.
This paper aims to contribute to exploring the design possibilities of robots for use in human-robot interaction. In an experiment, we investigate the influence of the human's personality and the robot's design, especially its humanization, on its acceptance. We use the Almere model, the Big 5 personality traits, and the anthropomorphic gestalt variants to build the foundation for our investigation. The assumption that an anthropomorphized robot variant would, in principle, be preferred to the standard variant when a natural choice is enforced could not be evidenced in our experiment. This allows for the interpretation that anthropomorphism does not necessarily lead to intentional perception and, consequently, does not guarantee that it can automatically generate acceptance.
Sustained glacier melt in the Himalayas has gradually spawned more than 5,000 glacier lakes that are dammed by potentially unstable moraines. When such dams break, glacier lake outburst floods (GLOFs) can cause catastrophic societal and geomorphic impacts. We present a robust probabilistic estimate of average GLOFs return periods in the Himalayan region, drawing on 5.4 billion simulations. We find that the 100-y outburst flood has an average volume of 33.5(+3.7)/(-3.7) x 10(6) m(3) (posterior mean and 95% highest density interval [HDI]) with a peak discharge of 15,600(+2.000)/(-1,800) m(3).S-1. Our estimated GLOF hazard is tied to the rate of historic lake outbursts and the number of present lakes, which both are highest in the Eastern Himalayas. There, the estimated 100-y GLOF discharge (similar to 14,500 m(3).s(-1)) is more than 3 times that of the adjacent Nyainqentanglha Mountains, and at least an order of magnitude higher than in the Hindu Kush, Karakoram, and Western Himalayas. The GLOF hazard may increase in these regions that currently have large glaciers, but few lakes, if future projected ice loss generates more unstable moraine-dammed lakes than we recognize today. Flood peaks from GLOFs mostly attenuate within Himalayan headwaters, but can rival monsoon-fed discharges in major rivers hundreds to thousands of kilometers downstream. Projections of future hazard from meteorological floods need to account for the extreme runoffs during lake outbursts, given the increasing trends in population, infrastructure, and hydropower projects in Himalayan headwaters.
