In der vorliegenden Dissertation wird die Bedeutung von Brachen für Artenvielfalt und Stabilität von Blüte-Bestäuber-Nahrungsnetzen in agrarisch genutzten Landschaften anhand ausgewählter blütenbesuchender Insektengruppen (Syrphidae, Lepidoptera) untersucht. Die Freilandarbeiten fanden von 1998-2000 im Raum der Feldberger Seenlandschaft, Mecklenburg-Vorpommern, statt. Es werden die beiden Hauptnahrungsquellen Nektar und Pollen betrachtet, dabei fanden Untersuchungen zur Intensität der Blüte-Bestäuber-Interaktion auf Stilllegungsflächen, zum flächenbezogenen quantitativen Nektarangebot im Jahresverlauf, zur individuellen Pollennutzung bei Syrphiden und zur Breite und Überlappung der Nahrungsnischen bei den dominanten Arten Episyrphus balteatus, Metasyrphus corollae, Syritta pipiens und Sphaerophoria scripta statt. Im Ergebnis zeigt sich eine hohe Bedeutung der Brachflächen für die Stabilität des Blüte-Bestäuber-Netzes, während die Diversität von anderen, eher landschaftsbezogenen Faktoren abhängig ist.
Mononuclear molybdoenzymes catalyze a broad range of redox reactions and are highly conserved in all kingdoms of life. This study addresses the question of how the Mo cofactor (Moco) is incorporated into the apo form of human sulfite oxidase (hSO) by using site-directed spin labeling to determine intramolecular distances in the nanometer range. Comparative measurements of the holo and apo forms of hSO enabled the localization of the corresponding structural changes, which are localized to a short loop (residues 263-273) of the Moco-containing domain. A flap-like movement of the loop provides access to the Moco binding-pocket in the apo form of the protein and explains the earlier studies on the in vitro reconstitution of apo-hSO with Moco. Remarkably, the loop motif can be found in a variety of structurally similar molybdoenzymes among various organisms, thus suggesting a common mechanism of Moco incorporation.
Microorganisms accumulate molar concentrations of compatible solutes like ectoine to prevent proteins from denaturation. Direct structural or spectroscopic information on the mechanism and about the hydration shell around ectoine are scarce. We combined surface plasmon resonance (SPR), confocal Raman spectroscopy, molecular dynamics simulations, and density functional theory (DFT) calculations to study the local hydration shell around ectoine and its influence on the binding of a gene-S-protein (G5P) to a single-stranded DNA (dT(25)). Due to the very high hygroscopicity of ectoine, it was possible to analyze the highly stable hydration shell by confocal Raman spectroscopy. Corresponding molecular dynamics simulation results revealed a significant change of the water dielectric constant in the presence of a high molar ectoine concentration as compared to pure water. The SPR data showed that the amount of protein bound to DNA decreases in the presence of ectoine, and hence, the protein-DNA dissociation constant increases in a concentration-dependent manner. Concomitantly, the Raman spectra in terms of the amide I region revealed large changes in the protein secondary structure. Our results indicate that ectoine strongly affects the molecular recognition between the protein and the oligonudeotide, which has important consequences for osmotic regulation mechanisms.
Stillegungsflächen nehmen einen bedeutenden Anteil der landwirtschaftlich nutzbaren Fläche Deutschlands ein. Kompensiert durch Ausgleichszahlungen entfällt in diesen Bereichen die Produktionsfunktion der Agrarlandschaft, ohne dass andere Landschaftsfunktionen explizit gefördert werden. Dabei können Stillegungsflächen eine wichtige Rolle im ökosystemaren Gefüge einer intensiv genutzten Agrarlandschaft spielen. Die vorgestellte Arbeit untersucht das Potential stillgelegter Flächen als Nahrungsquelle für blütenbesuchende Insekten. Dabei wird von der These ausgegangen, dass Stillegungsflächen besonders attraktiv für Blütenbesucher sind, da sie die einzigen Bereiche in einer intensiv genutzten Agrarlandschaft darstellen, deren Vegetation im Jahresverlauf nicht durch Nutzung beeinträchtigt wird. Es werden erste Ergebnisse einer nahrungsökologischen Studie vorgestellt, bei der das Fraßverhalten dominanter Arten der Syrphidae, einer der individuenreichsten Blütenbesuchergruppen Mitteleuropas, auf verschiedenen Stillegungsflächen untersucht wird. Durch wöchentliche Begehungen von Dauerbeobachtungsflächen wurde die Stärke und Dynamik des Blütendargebotes erfasst. Nektarproben ergaben ein Bild von der Quantität und Verfügbarkeit einzelner Nahrungsressourcen. Über die Untersuchungsperiode hinweg wurden Syrphiden mit einem Kescher gefangen und konserviert. Individuen dominanter Arten wurde mit einer Abdominalpräparation der Verdauungstrakt entnommen und pollenanalytisch aufgearbeitet. Die enthaltenen Pollen wurden determiniert und ausgezählt.
Aims. We aim to characterize the multiwavelength emission from Markarian 501 (Mrk 501), quantify the energy-dependent variability, study the potential multiband correlations, and describe the temporal evolution of the broadband emission within leptonic theoretical scenarios. Methods. We organized a multiwavelength campaign to take place between March and July of 2012. Excellent temporal coverage was obtained with more than 25 instruments, including the MAGIC, FACT and VERITAS Cherenkov telescopes, the instruments on board the Swift and Fermi spacecraft, and the telescopes operated by the GASP-WEBT collaboration. Results. Mrk 501 showed a very high energy (VHE) gamma-ray flux above 0.2 TeV of similar to 0.5 times the Crab Nebula flux (CU) for most of the campaign. The highest activity occurred on 2012 June 9, when the VHE flux was similar to 3 CU, and the peak of the high-energy spectral component was found to be at similar to 2 TeV. Both the X-ray and VHE gamma-ray spectral slopes were measured to be extremely hard, with spectral indices <2 during most of the observing campaign, regardless of the X-ray and VHE flux. This study reports the hardest Mrk 501 VHE spectra measured to date. The fractional variability was found to increase with energy, with the highest variability occurring at VHE. Using the complete data set, we found correlation between the X-ray and VHE bands; however, if the June 9 flare is excluded, the correlation disappears (significance <3 sigma) despite the existence of substantial variability in the X-ray and VHE bands throughout the campaign. Conclusions. The unprecedentedly hard X-ray and VHE spectra measured imply that their low- and high-energy components peaked above 5 keV and 0.5 TeV, respectively, during a large fraction of the observing campaign, and hence that Mrk 501 behaved like an extreme high-frequency-peaked blazar (EHBL) throughout the 2012 observing season. This suggests that being an EHBL may not be a permanent characteristic of a blazar, but rather a state which may change over time. The data set acquired shows that the broadband spectral energy distribution (SED) of Mrk 501, and its transient evolution, is very complex, requiring, within the framework of synchrotron self-Compton (SSC) models, various emission regions for a satisfactory description. Nevertheless the one-zone SSC scenario can successfully describe the segments of the SED where most energy is emitted, with a significant correlation between the electron energy density and the VHE gamma-ray activity, suggesting that most of the variability may be explained by the injection of high-energy electrons. The one-zone SSC scenario used reproduces the behavior seen between the measured X-ray and VHE gamma-ray fluxes, and predicts that the correlation becomes stronger with increasing energy of the X-rays.
