Due to their ability to capture attention, emotional stimuli tend to benefit from enhanced perceptual processing, which can be helpful when such stimuli are task-relevant but hindering when they are task-irrelevant. Altered emotion-attention interactions have been associated with symptoms of affective disturbances, and emerging research focuses on improving emotion-attention interactions to prevent or treat affective disorders. In line with the Human Affectome Project's emphasis on linguistic components, we also analyzed the language used to describe attention-related aspects of emotion, and highlighted terms related to domains such as conscious awareness, motivational effects of attention, social attention, and emotion regulation. These terms were discussed within a broader review of available evidence regarding the neural correlates of (1) Emotion-Attention Interactions in Perception, (2) Emotion-Attention Interactions in Learning and Memory, (3) Individual Differences in Emotion-Attention Interactions, and (4) Training and Interventions to Optimize Emotion-Attention Interactions. This comprehensive approach enabled an integrative overview of the current knowledge regarding the mechanisms of emotion-attention interactions at multiple levels of analysis, and identification of emerging directions for future investigations.
Do stereotypes strike twice?

Stereotypes influence teachers' perception of and behaviour towards students, thus shaping students' learning opportunities. The present study investigated how 315 Australian pre-service teachers' stereotypes about giftedness and gender are related to their perception of students' intellectual ability, adjustment, and social-emotional ability, using an experimental vignette approach and controlling for social desirability in pre-service teachers' responses. Repeated-measures ANOVA showed that pre-service teachers associated giftedness with higher intellectual ability, but with less adjustment compared to average-ability students. Furthermore, pre-service teachers perceived male students as less socially and emotionally competent and less adjusted than female students. Additionally, pre-service teachers seemed to perceive female average-ability students' adjustment as most favourable compared to male average-ability students and gifted students. Findings point to discrepancies between actual characteristics of gifted female and male students and stereotypes in teachers' beliefs. Consequences of stereotyping and implications for teacher education are discussed.
Word forms such as walked or walker are decomposed into their morphological constituents (walk + -ed/-er) during language comprehension. Yet, the efficiency of morphological decomposition seems to vary for different languages and morphological types, as well as for first and second language speakers. The current study reports results from a visual masked priming experiment focusing on different types of derived word forms (specifically prefixed vs. suffixed) in first and second language speakers of German. We compared the present findings with results from previous studies on inflection and compounding and proposed an account of morphological decomposition that captures both the variability and the consistency of morphological decomposition for different morphological types and for first and second language speakers. This study addresses the question of whether and how growing up with more than one language shapes a child's language impairment. Our focus is on Specific Language Impairment (SLI) in bilingual (Turkish-German) children. We specifically investigated a range of phenomena related to the so-called CP (Complementizer Phrase) in German, the hierarchically highest layer of syntactic clause structure, which has been argued to be particularly affected in children with SLI. Spontaneous speech data were examined from bilingual children with SLI in comparison to two comparison groups: (i) typically-developing bilingual children, (ii) monolingual children with SLI. We found that despite persistent difficulty with subject-verb agreement, the two groups of children with SLI did not show any impairment of the CP-domain. We conclude that while subject-verb agreement is a suitable linguistic marker of SLI in German-speaking children, for both monolingual and bilingual ones, 'vulnerability of the CP-domain' is not.
Lake sediments are increasingly explored as reliable paleoflood archives. In addition to established flood proxies including detrital layer thickness, chemical composition, and grain size, we explore stable oxygen and carbon isotope data as paleoflood proxies for lakes in catchments with carbonate bedrock geology. In a case study from Lake Mondsee (Austria), we integrate high-resolution sediment trapping at a proximal and a distal location and stable isotope analyses of varved lake sediments to investigate flood-triggered detrital sediment flux. First, we demonstrate a relation between runoff, detrital sediment flux, and isotope values in the sediment trap record covering the period 2011-2013 CE including 22 events with daily (hourly) peak runoff ranging from 10 (24) m(3) s(-1) to 79 (110) m(3) s(-1). The three- to ten-fold lower flood-triggered detrital sediment deposition in the distal trap is well reflected by attenuated peaks in the stable isotope values of trapped sediments. Next, we show that all nine flood-triggered detrital layers deposited in a sediment record from 1988 to 2013 have elevated isotope values compared with endogenic calcite. In addition, even two runoff events that did not cause the deposition of visible detrital layers are distinguished by higher isotope values. Empirical thresholds in the isotope data allow estimation of magnitudes of the majority of floods, although in some cases flood magnitudes are overestimated because local effects can result in too-high isotope values. Hence we present a proof of concept for stable isotopes as reliable tool for reconstructing flood frequency and, although with some limitations, even for flood magnitudes.
Achromatium oxaliferum is a large sulfur bacterium easily recognized by large intracellular calcium carbonate bodies. Although these bodies often fill major parts of the cells' volume, their role and specific intracellular location are unclear. In this study, we used various microscopy and staining techniques to identify the cell compartment harboring the calcium carbonate bodies. We observed that Achromatium cells often lost their calcium carbonate bodies, either naturally or induced by treatments with diluted acids, ethanol, sodium bicarbonate and UV radiation which did not visibly affect the overall shape and motility of the cells (except for UV radiation). The water-soluble fluorescent dye fluorescein easily diffused into empty cavities remaining after calcium carbonate loss. Membranes (stained with Nile Red) formed a network stretching throughout the cell and surrounding empty or filled calcium carbonate cavities. The cytoplasm (stained with FITC and SYBR Green for nucleic acids) appeared highly condensed and showed spots of dissolved Ca2+ (stained with Fura-2). From our observations, we conclude that the calcium carbonate bodies are located in the periplasm, in extra-cytoplasmic pockets of the cytoplasmic membrane and are thus kept separate from the cell's cytoplasm. This periplasmic localization of the carbonate bodies might explain their dynamic formation and release upon environmental changes.
The knowledge of transformation pathways and identification of transformation products (TPs) of veterinary drugs is important for animal health, food, and environmental matters. The active agent Monensin (MON) belongs to the ionophore antibiotics and is widely used as a veterinary drug against coccidiosis in broiler farming. However, no electrochemically (EC) generated TPs of MON have been described so far. In this study, the online coupling of EC and mass spectrometry (MS) was used for the generation of oxidative TPs. EC-conditions were optimized with respect to working electrode material, solvent, modifier, and potential polarity. Subsequent LC/HRMS (liquid+ chromatography/high resolution mass spectrometry) and MS/MS experiments were performed to identify the structures of derived TPs by a suspected target analysis. The obtained EC-results were compared to TPs observed in metabolism tests with microsomes and hydrolysis experiments of MON. Five previously undescribed TPs of MON were identified in our EC/MS based study and one TP, which was already known from literature and found by a microsomal assay, could be confirmed. Two and three further TPs were found as products in microsomal tests and following hydrolysis, respectively. We found decarboxylation, O-demethylation and acid-catalyzed ring-opening reactions to be the major mechanisms of MON transformation
Dermal Delivery of the High-Molecular-Weight Drug Tacrolimus by Means of Polyglycerol-Based Nanogels

Polyglycerol-based thermoresponsive nanogels (tNGs) have been shown to have excellent skin hydration properties and to be valuable delivery systems for sustained release of drugs into skin. In this study, we compared the skin penetration of tacrolimus formulated in tNGs with a commercial 0.1% tacrolimus ointment. The penetration of the drug was investigated in ex vivo abdominal and breast skin, while different methods for skin barrier disruption were investigated to improve skin permeability or simulate inflammatory conditions with compromised skin barrier. The amount of penetrated tacrolimus was measured in skin extracts by liquid chromatography tandem-mass spectrometry (LC-MS/MS), whereas the inflammatory markers IL-6 and IL-8 were detected by enzyme-linked immunosorbent assay (ELISA). Higher amounts of tacrolimus penetrated in breast as compared to abdominal skin or in barrier-disrupted as compared to intact skin, confirming that the stratum corneum is the main barrier for tacrolimus skin penetration. The anti-proliferative effect of the penetrated drug was measured in skin tissue/Jurkat cells co-cultures. Interestingly, tNGs exhibited similar anti-proliferative effects as the 0.1% tacrolimus ointment. We conclude that polyglycerol-based nanogels represent an interesting alternative to paraffin-based formulations for the treatment of inflammatory skin conditions.

The development and deployment of new operational runoff forecasting systems are a strong focus of the scientific community due to the crucial importance of reliable and timely runoff predictions for early warnings of floods and flashfloods for local businesses and communities. OpenForecast, the first operational runoff forecasting system in Russia, open for public use, is presented in this study. We developed OpenForecast based only on open-source software and data-GR4J hydrological model, ERA-Interim meteorological reanalysis, and ICON deterministic short-range meteorological forecasts. Daily forecasts were generated for two basins in the European part of Russia. Simulation results showed a limited efficiency in reproducing the spring flood of 2019. Although the simulations managed to capture the timing of flood peaks, they failed in estimating flood volume. However, further implementation of the parsimonious data assimilation technique significantly alleviates simulation errors. The revealed limitations of the proposed operational runoff forecasting system provided a foundation to outline its further development and improvement.
Forest structure is a crucial component in the assessment of whether a forest is likely to act as a carbon sink under changing climate. Detailed 3D structural information about the tundra–taiga ecotone of Siberia is mostly missing and still underrepresented in current research due to the remoteness and restricted accessibility. Field based, high-resolution remote sensing can provide important knowledge for the understanding of vegetation properties and dynamics. In this study, we test the applicability of consumer-grade Unmanned Aerial Vehicles (UAVs) for rapid calculation of stand metrics in treeline forests. We reconstructed high-resolution photogrammetric point clouds and derived canopy height models for 10 study sites from NE Chukotka and SW Yakutia. Subsequently, we detected individual tree tops using a variable-window size local maximum filter and applied a marker-controlled watershed segmentation for the delineation of tree crowns. With this, we successfully detected 67.1% of the validation individuals. Simple linear regressions of observed and detected metrics show a better correlation (R2) and lower relative root mean square percentage error (RMSE%) for tree heights (mean R2 = 0.77, mean RMSE% = 18.46%) than for crown diameters (mean R2 = 0.46, mean RMSE% = 24.9%). The comparison between detected and observed tree height distributions revealed that our tree detection method was unable to representatively identify trees <2 m. Our results show that plot sizes for vegetation surveys in the tundra–taiga ecotone should be adapted to the forest structure and have a radius of >15–20 m to capture homogeneous and representative forest stands. Additionally, we identify sources of omission and commission errors and give recommendations for their mitigation. In summary, the efficiency of the used method depends on the complexity of the forest’s stand structure.
In this paper Lie group method in combination with Magnus expansion is utilized to develop a universal method applicable to solving a Sturm–Liouville problem (SLP) of any order with arbitrary boundary conditions. It is shown that the method has ability to solve direct regular (and some singular) SLPs of even orders (tested for up to eight), with a mix of (including non-separable and finite singular endpoints) boundary conditions, accurately and efficiently. The present technique is successfully applied to overcome the difficulties in finding suitable sets of eigenvalues so that the inverse SLP problem can be effectively solved. The inverse SLP algorithm proposed by Barcilon (1974) is utilized in combination with the Magnus method so that a direct SLP of any (even) order and an inverse SLP of order two can be solved effectively.
In this paper, we investigate the continuous version of modified iterative Runge–Kutta-type methods for nonlinear inverse ill-posed problems proposed in a previous work. The convergence analysis is proved under the tangential cone condition, a modified discrepancy principle, i.e., the stopping time T is a solution of ∥𝐹(𝑥𝛿(𝑇))−𝑦𝛿∥=𝜏𝛿+ for some 𝛿+>𝛿, and an appropriate source condition. We yield the optimal rate of convergence.
In animals and humans, behavior can be influenced by irrelevant stimuli, a phenomenon called Pavlovian-to-instrumental transfer (PIT). In subjects with substance use disorder, PIT is even enhanced with functional activation in the nucleus accumbens (NAcc) and amygdala. While we observed enhanced behavioral and neural PIT effects in alcohol-dependent subjects, we here aimed to determine whether behavioral PIT is enhanced in young men with high-risk compared to low-risk drinking and subsequently related functional activation in an a-priori region of interest encompassing the NAcc and amygdala and related to polygenic risk for alcohol consumption. A representative sample of 18-year old men (n = 1937) was contacted: 445 were screened, 209 assessed: resulting in 191 valid behavioral, 139 imaging and 157 genetic datasets. None of the subjects fulfilled criteria for alcohol dependence according to the Diagnostic and Statistical Manual of Mental Disorders-IV-TextRevision (DSM-IV-TR). We measured how instrumental responding for rewards was influenced by background Pavlovian conditioned stimuli predicting action-independent rewards and losses. Behavioral PIT was enhanced in high-compared to low-risk drinkers (b = 0.09, SE = 0.03, z = 2.7, p < 0.009). Across all subjects, we observed PIT-related neural blood oxygen level-dependent (BOLD) signal in the right amygdala (t = 3.25, p(SVC) = 0.04, x = 26, y = -6, z = -12), but not in NAcc. The strength of the behavioral PIT effect was positively correlated with polygenic risk for alcohol consumption (r(s) = 0.17, p = 0.032). We conclude that behavioral PIT and polygenic risk for alcohol consumption might be a biomarker for a subclinical phenotype of risky alcohol consumption, even if no drug-related stimulus is present. The association between behavioral PIT effects and the amygdala might point to habitual processes related to out PIT task. In non-dependent young social drinkers, the amygdala rather than the NAcc is activated during PIT; possible different involvement in association with disease trajectory should be investigated in future studies.
Die vorliegende Arbeit geht der Fragestellung nach, inwiefern der Beutelsbacher Konsens in einem ausgewählten Schulbuch für den Politikunterricht in der Sekundarstufe I in Brandenburg berücksichtigt wird. Um sich dieser Frage anzunähern, werden zunächst die drei Grundsätze des Konsenses wiedergegeben: das Überwältigungsverbot, das Kontroversitätsgebot und die Schülerorientierung. Da der Konsens, auch wenn er von einem Großteil der Fachdidaktikerinnen und Fachdidaktiker geteilt wird, immer wieder Gegenstand von Diskussionen ist, werden in einem ersten Schritt Ansätze zur Aktualisierung bzw. Erweiterung dargestellt und anschließend aktuelle Streitpunkte aufgezeigt. In einem kurzen Zwischenfazit wird dann ein für die Schulbuchanalyse unabdingliches, eindeutiges Verständnis des Konsenses entwickelt.
Im folgenden Schritt wird die Rolle von Schulbüchern als Lehr- und Lernmedien diskutiert. Dabei steht insbesondere die Frage im Zentrum, weshalb sich gerade Schulbücher für eine Analyse im Rahmen der vorliegenden Arbeit eignen. Vor diesem Hintergrund wird das Konzept der Schulbuchanalyse vorgestellt. In diesem Rahmen werden der Untersuchungsschwerpunkt (Kontroversitätsgebot) und der Untersuchungsgegenstand (Kontroverse um Migration und Integration) eingegrenzt. In der Folge wird das Schulbuch Politik und Co. 1 mithilfe des erarbeiteten Untersuchungsinstruments (Kodierleitfaden) analysiert. Zudem werden die Ergebnisse pointiert und die gewählte Vorgehensweise reflektiert.
Narratives are shaping our understanding of the world. They convey values and norms and point to desirable future developments. In this way, they justify and legitimize political actions and social practices. Once a narrative has emerged and this world view is supported by broad societal groups, narratives can provide powerful momentum to trigger innovation and changes in the course of action. Narratives, however, are not necessarily based on evidence and precise categories, but can instead be vague and ambiguous in order to be acceptable and attractive to different actors. However, the more open and inclusive a narrative is, the less impact can be expected. We investigate whether there is a shared narrative in research for the sustainable economy and how this can be evaluated in terms of its potential societal impact. The paper carves out the visions for the future that have been underlying the research projects conducted within the German Federal Ministry of Education and Research (BMBF) funding programme "The Sustainable Economy". It then analyzes whether these visions are compatible with narratives dominating societal discourse on the sustainable economy, and concludes how the use of visions and narratives in research can contribute to fostering societal transformations.
This dissertation is concerned with the relation between qualitative phonological organization in the form of syllabic structure and continuous phonetics, that is, the spatial and temporal dimensions of vocal tract action that express syllabic structure. The main claim of the dissertation is twofold. First, we argue that syllabic organization exerts multiple effects on the spatio-temporal properties of the segments that partake in that organization. That is, there is no unique or privileged exponent of syllabic organization. Rather, syllabic organization is expressed in a pleiotropy of phonetic indices. Second, we claim that a better understanding of the relation between qualitative phonological organization and continuous phonetics is reached when one considers how the string of segments (over which the nature of the phonological organization is assessed) responds to perturbations (scaling of phonetic variables) of localized properties (such as durations) within that string. Specifically, variation in phonetic variables and more specifically prosodic variation is a crucial key to understanding the nature of the link between (phonological) syllabic organization and the phonetic spatio-temporal manifestation of that organization. The effects of prosodic variation on segmental properties and on the overlap between the segments, we argue, offer the right pathway to discover patterns related to syllabic organization. In our approach, to uncover evidence for global organization, the sequence of segments partaking in that organization as well as properties of these segments or their relations with one another must be somehow locally varied. The consequences of such variation on the rest of the sequence can then be used to unveil the span of organization. When local perturbations to segments or relations between adjacent segments have effects that ripple through the rest of the sequence, this is evidence that organization is global. If instead local perturbations stay local with no consequences for the rest of the whole, this indicates that organization is local.
Das Preußische Erbrecht in der Judikatur des Berliner Obertribunals in den Jahren 1836 bis 1865

Die Dissertation befasst sich mit dem Allgemeinen Preußischen Landrecht von 1794 und der hierzu ergangenen Rechtsprechung des Berliner Obertribunals. Im Fokus der Untersuchung stehen die erbrechtlichen Regelungen des Landrechts und deren Anwendung sowie Auslegung in der Judikatur des höchsten preußischen Gerichts. Der Forschungsgegenstand ergibt sich aus dem im Landrecht kodifizierten speziellen Gesetzesverständnisses. Nach diesem sollte die Gesetzesauslegung durch die Rechtsprechung auf ein Minimum, nämlich die Auslegung allein anhand des Wortlauts der Regelung reduziert werden, um dem absolutistischen Regierungsanspruch der preußischen Monarchen, namentlich Friedrich des Großen, hinreichend Rechnung zu tragen. In diesem Kontext wird der Frage nachgegangen, inwieweit das preußische Obertribunal das im Landrecht statuierte „Auslegungsverbot“ beachtet hat und in welchen Fällen sich das Gericht von der Vorgabe emanzipierte und weitere Auslegungsmethoden anwendete und sich so eine unabhängige Rechtsprechung entwickeln konnte.
Die Arbeit gliedert sich in drei Hauptabschnitte. Im Anschluss an die Einleitung, in der zunächst die rechtshistorische Bedeutung des Landrechts und des Erbrechts sowie der Untersuchungsgegenstand umrissen werden, folgt die Darstellung der Entstehungsgeschichte des Landrechts und des Berliner Obertribunals.
Hieran schließt sich in einem dritten Abschnitt eine Analyse der erbrechtlichen Vorschriften des Landrechts an. In dieser wird auf die Entstehungsgeschichte der verschiedenen erbrechtlichen Institute wie beispielsweise der gesetzlichen und gewillkürten Erbfolge, dem Pflichtteilsrecht etc., unter Berücksichtigung des zeitgenössischen wissenschaftlichen Diskurses eingegangen.
Im vierten Abschnitt geht es um die Judikate des Berliner Obertribunals aus den Jahren 1836-1865 in denen die zuvor dargestellten erbrechtlichen Regelungen entscheidungserheblich waren. Dabei wird der Forschungsfrage, inwieweit das Obertribunal das im Landrecht statuierte Auslegungsverbot beachtet hat und in welchen Fällen es von diesem abwich bzw. weitere Auslegungsmethoden anwendete, konkret nachgegangen wird. Insgesamt werden 26 Entscheidungen des Obertribunals unter dem Aspekt der Auslegungspraxis, der Kontinuität und der Beschleunigung der Rechtsprechung analysiert und ausgewertet.
Fünfundsiebzig Jahre nach dem Ende des Zweiten Weltkriegs steht die heutige Erinnerungskultur vor großen Herausforderungen. Immer weniger Überlebende können persönliches Zeugnis des Erlebten ablegen. Während sich seit den 1980er Jahren hat in der westlichen Welt eine gesellschaftliche und staatliche Erinnerungskultur etabliert hat, die sowohl in der Bildung, als auch im öffentlichen Raum ihren Platz gefunden hat. Jenseits des ehemaligen „Eisernen Vorhangs“ ist diese Erinnerungskultur keineswegs selbstverständlich, bis nicht erwünscht.
Diese Arbeit beschäftigt sich mit der besonderen Situation Belarus, welches heute immer noch stark postsowjetisch geprägt ist und als Land mit dem zweithöchsten Partisanenwiderstand gegen die deutsche Besatzung gilt. Aufgrund der besonderen geschichtlichen Situation geht der von der jüdischen Bevölkerung mitgetragene Widerstand im staatlich geprägten Heldennarrativ der siegreichen sowjetischen Armee unter. Gepaart wird der Sieg der Sowjetarmee mit einem Opfernarrativ, welches in der Folge in allgemeiner Ansprache fast ausschließlich nur verstorbenen Sowjetbürgern gedenkt, aber nicht explizit die jüdischen Mitbürger benennt. Somit geht sowohl die Erinnerung an den jüdischen Widerstand, als auch an die Verfolgung der jüdischen Bevölkerung in der Memorialkultur Belarus bis auf wenige Ausnahmen verloren. Tragischerweise war Widerstand jedoch oftmals die einzige Möglichkeit für Jüdinnen und Juden überhaupt zu überleben. Darüber hinaus muss ein staatlich getragener und geförderter Antisemitismus innerhalb der ehemaligen Sowjetunion mitgedacht werden, der die jüdische Bevölkerung weiterhin diskriminierte und verfolgte.
In einem geschichtlichen Abriss ab 1941 bis zur Gründung der Republik Belarus und ihrer aktuellen staatlichen Doktrin wird einleitend der jüdische Widerstand dargestellt, der in der Folge stattfindende offizielle Umgang mit dem Holocaust und heutige Orte des Erinnerns, sowohl staatlicherseits, als auch privater Initiativen. Einzelne konkrete Beispiele veranschaulichen die angesprochenen Themenbereiche.
Bis heute hat das Gedenken an die jüdischen Opfer, geschweige denn an deren Widerstand, einen sehr geringen Stellenwert in Belarus. Es bleibt zu befürchten, dass mit den letzten Zeitzeugen diese Erinnerung aus dem kollektiven Gedächtnis verschwindet.
Bisher ist die Ursache für die Entstehung der meisten Skoliosen noch ungeklärt und damit eine kausale Behandlung der Betroffenen unmöglich. Die vorliegende Arbeit geht davon aus, dass der Auslöser für die sogenannte idiopathische Skoliose eine funktionelle Störung von Muskeln ist, die sich in einer verminderten relativen Haltekraft äußert. Durch gezielte willkürliche Muskelanspannungen könnte es möglich sein, kompensatorisch auf die Deformität einzuwirken, um damit ein Fortschreiten zu verhindern bzw. sogar eine Regression hervorzurufen. Insbesondere Patientengruppen mit einem hohen Progressionsrisiko, wie Jugendliche im Wachstumsalter, könnten davon profitieren.
Ein Muskeltraining kann mit unterschiedlichsten Hilfsmitteln und Methoden erfolgen. Eine Möglichkeit bietet auch das Klettern. Im Kern wird daher ein Trainingskonzept zum Therapeutischen Klettern bei Jugendlichen mit Skoliose vorgestellt. Dabei beruft sich der Autor auf das Potsdamer Modell. Dieses Modell erlaubt es, gezielte Kraftübungen systematisiert an der Kletterwand in Absprunghöhe umzusetzen. Materielle Sicherungsmaßnahmen sind dadurch nicht erforderlich und eventuell notwendige Korrekturen bzw. Hilfestellungen können direkt erfolgen. Hauptinhalt eines Trainings nach dem vorgestellten Konzept sind spielerische Bewegungserfahrung innerhalb der Sportart Klettern und ein Systembouldertraining.
In einem beigefügten Übungskatalog werden für letzteres Möglichkeiten der praktischen Umsetzun-gen gegeben. Die Übungen fokussieren sich auf die Aktivierung und das Training wirbelkörperdero-tierender Muskeln. Im Hauptteil einer Trainingseinheit können sie dann in Kombination mit der Kor-rektur der Seitverbiegung und des sagittalen Profils (3D Autokorrektur) unter Aufsicht eines geschul-ten Therapeuten durchgeführt werden. Die Arbeit erhebt den Anspruch, einem Leser vom Fach, die Auswahl der Übungen und die darin enthaltene individuelle Anpassung an den Patienten aus funktionell-anatomischer Sicht zu begründen.
In naher Zukunft wird das Konzept in einer randomisiert kontrollierten Studie untersucht. Alle notwendigen Vorbereitungen wurden im Rahmen dieser Arbeit getroffen.
A digital filter is introduced which treats the problem of predictability versus time averaging in a continuous, seamless manner. This seamless filter (SF) is characterized by a unique smoothing rule that determines the strength of smoothing in dependence on lead time. The rule needs to be specified beforehand, either by expert knowledge or by user demand. As a result, skill curves are obtained that allow a predictability assessment across a whole range of time-scales, from daily to seasonal, in a uniform manner. The SF is applied to downscaled SEAS5 ensemble forecasts for two focus regions in or near the tropical belt, the river basins of the Karun in Iran and the Sao Francisco in Brazil. Both are characterized by strong seasonality and semi-aridity, so that predictability across various time-scales is in high demand. Among other things, it is found that from the start of the water year (autumn), areal precipitation is predictable with good skill for the Karun basin two and a half months ahead; for the Sao Francisco it is only one month, longer-term prediction skill is just above the critical level.

CH3NH3PbI3 als Archetyp dieser Materialklasse besitzt außergewöhnliche Eigenschaften wie eine sehr effiziente Umwandlung von Solarenergie, wobei besonders Ferroelektrizität als mögliche Erklärung in den Fokus gerückt ist. Diese erfordert allerdings eine nicht-zentrosymmetrische Kristallstruktur als notwendige Voraussetzung. Wir stellen hier eine Erklärung des Symmetriebruchs in diesem Material auf kristallographischem, d. h. fernordnungs-basiertem, Wege vor. Während das Molekülkation CH3NH3+ intrinsisch polar ist, ist es extrem fehlgeordnet und kann deshalb nicht die einzige Erklärung darstellen. Es verzerrt allerdings das umgebende Kristallgitter und ruft dadurch eine Verschiebung der Iod-Atome von den zentrosymmetrischen Positionen hervor.
Excellent conversion efficiencies of over 20% and facile cell production have placed hybrid perovskites at the forefront of novel solar cell materials, with CH3NH3PbI3 being an archetypal compound. The question why CH3NH3PbI3 has such extraordinary characteristics, particularly a very efficient power conversion from absorbed light to electrical power, is hotly debated, with ferroelectricity being a promising candidate. This does, however, require the crystal structure to be non-centrosymmetric and we herein present crystallographic evidence as to how the symmetry breaking occurs on a crystallographic and, therefore, long-range level. Although the molecular cation CH3NH3+ is intrinsically polar, it is heavily disordered and this cannot be the sole reason for the ferroelectricity. We show that it, nonetheless, plays an important role, as it distorts the neighboring iodide positions from their centrosymmetric positions.
Background: The distribution of pronouns varies cross-linguistically. This distribution has led to conflicting results in studies that investigated pronoun resolution in agrammatic indviduals. In the investigation of pronominal resolution, the linguistic phenomenon of "resumption" is understudied in agrammatism. The construction of pronominal resolution in Akan presents the opportunity to thoroughly examine resumption. Aims: To start, the present study examines the production of (pronominal) resumption in Akan focus constructions (who-questions and focused declaratives). Second, we explore the effect of grammatical tone on the processing of pronominal (resumption) since Akan is a tonal language. Methods & Procedures: First, we tested the ability to distinguish linguistic and non-linguistic tone in Akan agrammatic speakers. Then, we administered an elicitation task to five Akan agrammatic individuals, controlling for the structural variations in the realization of resumption: focused who-questions and declaratives with (i) only a resumptive pronoun, (ii) only a clause determiner, (iii) a resumptive pronoun and a clause determiner co-occurring, and (iv) neither a resumptive pronoun nor a clause determiner. Outcomes & Results: Tone discrimination .both for pitch and for lexical tone was unimpaired. The production task demonstrated that the production of resumptive pronouns and clause determiners was intact. However, the production of declarative sentences in derived word order was impaired; wh-object questions were relatively well-preserved. Conclusions: We argue that the problems with sentence production are highly selective: linguistic tones and resumption are intact but word order is impaired in non-canonical declarative sentences.
Open-circuit voltages of lead-halide perovskite solar cells are improving rapidly and are approaching the thermodynamic limit. Since many different perovskite compositions with different bandgap energies are actively being investigated, it is not straightforward to compare the open-circuit voltages between these devices as long as a consistent method of referencing is missing. For the purpose of comparing open-circuit voltages and identifying outstanding values, it is imperative to use a unique, generally accepted way of calculating the thermodynamic limit, which is currently not the case. Here a meta-analysis of methods to determine the bandgap and a radiative limit for open-circuit voltage is presented. The differences between the methods are analyzed and an easily applicable approach based on the solar cell quantum efficiency as a general reference is proposed.
The impact of the orientation of zwitterionic groups, with respect to the polymer backbone, on the antifouling performance of thin hydrogel films made of polyzwitterions is explored. In an extension of the recent discussion about differences in the behavior of polymeric phosphatidylcholines and choline phosphates, a quasi-isomeric set of three poly(sulfobetaine methacrylate)s is designed for this purpose. The design is based on the established monomer 3-[N-2-(methacryloyloxy)ethyl-N,N-dimethyl]ammonio-propane-1-sulfonate and two novel sulfobetaine methacrylates, in which the positions of the cationic and the ionic groups relative to the polymerizable group, and thus also to the polymer backbone, are altered. The effect of the varied segmental dipole orientation on their water solubility, wetting behavior by water, and fouling resistance is compared. As model systems, the adsorption of the model proteins bovine serum albumin (BSA), fibrinogen, and lysozyme onto films of the various polyzwitterion surfaces is studied, as well as the settlement of a diatom (Navicula perminuta) and barnacle cyprids (Balanus improvisus) as representatives of typical marine fouling communities. The results demonstrate the important role of the zwitterionic group's orientation on the polymer behavior and fouling resistance
Entrepreneurial persistence is demonstrated by an entrepreneur’s continued positive maintenance of entrepreneurial motivation and constantly renewed active engagement in a new business venture despite counterforces or enticing alternatives. It thus is a crucial factor for entrepreneurs when pursuing and exploiting their business opportunities and in realizing potential economic gains and benefits. Using rich data on a representative sample of German business founders, we investigated the determinants of entrepreneurial persistence. Next to observed survival, we also constructed a hybrid persistence measure capturing the motivational dimension of persistence. We analyzed the influence of individual-level (human capital and personality) and business-related characteristics on both measures as well as their relative importance. We found that the two indicators emphasize different aspects of persistence. For the survival indicator, the predictive power was concentrated in business characteristics and human capital, while for hybrid persistence the dominant factors were business characteristics and personality. Finally, we showed that results were heterogeneous across subgroups. In particular, formerly unemployed founders did not differ in survival chances, but they were more likely to lack a high psychological commitment to their business ventures.
We study travelling chimera states in a ring of nonlocally coupled heterogeneous (with Lorentzian distribution of natural frequencies) phase oscillators. These states are coherence-incoherence patterns moving in the lateral direction because of the broken reflection symmetry of the coupling topology. To explain the results of direct numerical simulations we consider the continuum limit of the system. In this case travelling chimera states correspond to smooth travelling wave solutions of some integro-differential equation, called the Ott–Antonsen equation, which describes the long time coarse-grained dynamics of the oscillators. Using the Lyapunov–Schmidt reduction technique we suggest a numerical approach for the continuation of these travelling waves. Moreover, we perform their linear stability analysis and show that travelling chimera states can lose their stability via fold and Hopf bifurcations. Some of the Hopf bifurcations turn out to be supercritical resulting in the observation of modulated travelling chimera states.

Das wirft die Frage auf, wie das notwendige Wissen erzeugt, bewahrt, verteilt und auffindbar gemacht werden kann. Ein solches Wissensmanagement kann die Arbeit der Behörden qualitativ verbessern und effizienter machen. Dennoch wird Wissen in der Verwaltungspraxis bisher nur unzureichend gemanagt.
Ein systematisches Wissensmanagement erfordert personelle, finanzielle und technische Ressourcen. Sind diese nicht vorhanden, können Verwaltungen zunächst auf einzelne Instrumente des Wissensmanagements zurückgreifen, um ihre Arbeit mit begrenztem Aufwand zu verbessern.
The public encounter

This thesis puts the citizen-state interaction at its center. Building on a comprehensive model incorporating various perspectives on this interaction, I derive selected research gaps. The three articles, comprising this thesis, tackle these gaps. A focal role plays the citizens’ administrative literacy, the relevant competences and knowledge necessary to successfully interact with public organizations. The first article elaborates on the different dimensions of administrative literacy and develops a survey instrument to assess these. The second study shows that public employees change their behavior according to the competences that citizens display during public encounters. They treat citizens preferentially that are well prepared and able to persuade them of their application’s potential. Thereby, they signal a higher success potential for bureaucratic success criteria which leads to the employees’ cream-skimming behavior. The third article examines the dynamics of employees’ communication strategies when recovering from a service failure. The study finds that different explanation strategies yield different effects on the client’s frustration. While accepting the responsibility and explaining the reasons for a failure alleviates the frustration and anger, refusing the responsibility leads to no or even reinforcing effects on the client’s frustration. The results emphasize the different dynamics that characterize the nature of citizen-state interactions and how they establish their short- and long-term outcomes.
Molecularly imprinted polymers (MIPs) mimic the binding sites of antibodies by substituting the amino acid-scaffold of proteins by synthetic polymers. In this work, the first MIP for the recognition of the diagnostically relevant enzyme butyrylcholinesterase (BuChE) is presented. The MIP was prepared using electropolymerization of the functional monomer o-phenylenediamine and was deposited as a thin film on a glassy carbon electrode by oxidative potentiodynamic polymerization. Rebinding and removal of the template were detected by cyclic voltammetry using ferricyanide as a redox marker. Furthermore, the enzymatic activity of BuChE rebound to the MIP was measured via the anodic oxidation of thiocholine, the reaction product of butyrylthiocholine. The response was linear between 50 pM and 2 nM concentrations of BuChE with a detection limit of 14.7 pM. In addition to the high sensitivity for BuChE, the sensor responded towards pseudo-irreversible inhibitors in the lower mM range.
Over the years, we developed highly selective fluorescent probes for K+ in water, which show K+-induced fluorescence intensity enhancements, lifetime changes, or a ratiometric behavior at two emission wavelengths (cf. Scheme 1, K1-K4). In this paper, we introduce selective fluorescent probes for Na+ in water, which also show Na+ induced signal changes, which are analyzed by diverse fluorescence techniques. Initially, we synthesized the fluorescent probes 2, 4, 5, 6 and 10 for a fluorescence analysis by intensity enhancements at one wavelength by varying the Na+ responsive ionophore unit and the fluorophore moiety to adjust different K-d values for an intra- or extracellular Na+ analysis. Thus, we found that 2, 4 and 5 are Na+ selective fluorescent tools, which are able to measure physiologically important Na+ levels at wavelengths higher than 500 nm. Secondly, we developed the fluorescent probes 7 and 8 to analyze precise Na+ levels by fluorescence lifetime changes. Herein, only 8 (K-d=106 mm) is a capable fluorescent tool to measure Na+ levels in blood samples by lifetime changes. Finally, the fluorescent probe 9 was designed to show a Na+ induced ratiometric fluorescence behavior at two emission wavelengths. As desired, 9 (K-d=78 mm) showed a ratiometric fluorescence response towards Na+ ions and is a suitable tool to measure physiologically relevant Na+ levels by the intensity change of two emission wavelengths at 404 nm and 492 nm.
The novel space-borne Global Navigation Satellite System Reflectometry (GNSS-R) technique has recently shown promise in monitoring the ocean state and surface wind speed with high spatial coverage and unprecedented sampling rate. The L-band signals of GNSS are structurally able to provide a higher quality of observations from areas covered by dense clouds and under intense precipitation, compared to those signals at higher frequencies from conventional ocean scatterometers. As a result, studying the inner core of cyclones and improvement of severe weather forecasting and cyclone tracking have turned into the main objectives of GNSS-R satellite missions such as Cyclone Global Navigation Satellite System (CYGNSS). Nevertheless, the rain attenuation impact on GNSS-R wind speed products is not yet well documented. Evaluating the rain attenuation effects on this technique is significant since a small change in the GNSS-R can potentially cause a considerable bias in the resultant wind products at intense wind speeds. Based on both empirical evidence and theory, wind speed is inversely proportional to derived bistatic radar cross section with a natural logarithmic relation, which introduces high condition numbers (similar to ill-posed conditions) at the inversions to high wind speeds. This paper presents an evaluation of the rain signal attenuation impact on the bistatic radar cross section and the derived wind speed. This study is conducted simulating GNSS-R delay-Doppler maps at different rain rates and reflection geometries, considering that an empirical data analysis at extreme wind intensities and rain rates is impossible due to the insufficient number of observations from these severe conditions. Finally, the study demonstrates that at a wind speed of 30 m/s and incidence angle of 30 degrees, rain at rates of 10, 15, and 20 mm/h might cause overestimation as large as approximate to 0.65 m/s (2%), 1.00 m/s (3%), and 1.3 m/s (4%), respectively, which are still smaller than the CYGNSS required uncertainty threshold. The simulations are conducted in a pessimistic condition (severe continuous rainfall below the freezing height and over the entire glistening zone) and the bias is expected to be smaller in size in real environments.
Humus forms are a distinctive morphological indicator of soil organic matter decomposition. The spatial distribution of humus forms depends on environmental factors such as topography, climate and vegetation. In montane and subalpine forests, environmental influences show a high spatial heterogeneity, which is reflected by a high spatial variability of humus forms. This study aims at examining spatial patterns of humus forms and their dependence on the spatial scale in a high mountain forest environment (Val di Sole/Val di Rabbi, Trentino, Italian Alps). On the basis of the distributions of environmental covariates across the study area, we described humus forms at the local scale (six sampling sites), slope scale (60 sampling sites) and landscape scale (30 additional sampling sites). The local variability of humus forms was analyzed with regard to the ground cover type. At the slope and landscape scale, spatial patterns of humus forms were modeled applying random forests and ordinary kriging of the model residuals. The results indicate that the occurrence of the humus form classes Mull, Mullmoder, Moder, Amphi and Eroded Moder generally depends on the topographical position. Local-scale patterns are mostly related to micro-topography (local accumulation and erosion sites) and ground cover, whereas slope-scale patterns are mainly connected with slope exposure and elevation. Patterns at the landscape scale show a rather irregular distribution, as spatial models at this scale do not account for local to slope-scale variations of humus forms. Moreover, models at the slope scale perform distinctly better than at the landscape scale. In conclusion, the results of this study highlight that landscape-scale predictions of humus forms should be accompanied by local- and slope-scale studies in order to enhance the general understanding of humus form patterns.
The possibility to manufacture perovskite solar cells (PSCs) at low temperatures paves the way to flexible and lightweight photovoltaic (PV) devices manufactured via high-throughput roll-to-roll processes. In order to achieve higher power conversion efficiencies, it is necessary to approach the radiative limit via suppression of non-radiative recombination losses. Herein, we performed a systematic voltage loss analysis for a typical low-temperature processed, flexible PSC in n-i-p configuration using vacuum deposited C-60 as electron transport layer (ETL) and two-step hybrid vacuum-solution deposition for CH3NH3PbI3 perovskite absorber. We identified the ETL/absorber interface as a bottleneck in relation to non-radiative recombination losses, the quasi-Fermi level splitting (QFLS) decreases from similar to 1.23 eV for the bare absorber, just similar to 90 meV below the radiative limit, to similar to 1.10 eV when C-60 is used as ETL. To effectively mitigate these voltage losses, we investigated different interfacial modifications via vacuum deposited interlayers (BCP, B4PyMPM, 3TPYMB, and LiF). An improvement in QFLS of similar to 30-40 meV is observed after interlayer deposition and confirmed by comparable improvements in the open-circuit voltage after implementation of these interfacial modifications in flexible PSCs. Further investigations on absorber/hole transport layer (HTL) interface point out the detrimental role of dopants in Spiro-OMeTAD film (widely employed HTL in the community) as recombination centers upon oxidation and light exposure. Role of GDF15 in active lifestyle induced metabolic adaptations and acute exercise response in mice

Physical activity is an important contributor to muscle adaptation and metabolic health. Growth differentiation factor 15 (GDF15) is established as cellular and nutritional stress-induced cytokine but its physiological role in response to active lifestyle or acute exercise is unknown. Here, we investigated the metabolic phenotype and circulating GDF15 levels in lean and obese male C57BI/6J mice with long-term voluntary wheel running (VWR) intervention. Additionally, treadmill running capacity and exercise-induced muscle gene expression was examined in GDF15-ablated mice. Active lifestyle mimic via VWR improved treadmill running performance and, in obese mice, also metabolic phenotype. The post-exercise induction of skeletal muscle transcriptional stress markers was reduced by VWR. Skeletal muscle GDF15 gene expression was very low and only transiently increased post-exercise in sedentary but not in active mice. Plasma GDF15 levels were only marginally affected by chronic or acute exercise. In obese mice, VWR reduced GDF15 gene expression in different tissues but did not reverse elevated plasma GDF15. Genetic ablation of GDF15 had no effect on exercise performance but augmented the post exercise expression of transcriptional exercise stress markers (Atf3, Atf6, and Xbp1s) in skeletal muscle. We conclude that skeletal muscle does not contribute to circulating GDF15 in mice, but muscle GDF15 might play a protective role in the exercise stress response.
Oscillatory systems under weak coupling can be described by the Kuramoto model of phase oscillators. Kuramoto phase oscillators have diverse applications ranging from phenomena such as communication between neurons and collective influences of political opinions, to engineered systems such as Josephson Junctions and synchronized electric power grids. This thesis includes the author's contribution to the theoretical framework of coupled Kuramoto oscillators and to the understanding of non-trivial N-body dynamical systems via their reduced mean-field dynamics.
The main content of this thesis is composed of four parts. First, a partially integrable theory of globally coupled identical Kuramoto oscillators is extended to include pure higher-mode coupling. The extended theory is then applied to a non-trivial higher-mode coupled model, which has been found to exhibit asymmetric clustering. Using the developed theory, we could predict a number of features of the asymmetric clustering with only information of the initial state provided.
The second part consists of an iterated discrete-map approach to simulate phase dynamics. The proposed map --- a Moebius map --- not only provides fast computation of phase synchronization, it also precisely reflects the underlying group structure of the dynamics. We then compare the iterated-map dynamics and various analogous continuous-time dynamics. We are able to replicate known phenomena such as the synchronization transition of the Kuramoto-Sakaguchi model of oscillators with distributed natural frequencies, and chimera states for identical oscillators under non-local coupling.
The third part entails a particular model of repulsively coupled identical Kuramoto-Sakaguchi oscillators under common random forcing, which can be shown to be partially integrable. Via both numerical simulations and theoretical analysis, we determine that such a model cannot exhibit stationary multi-cluster states, contrary to the numerical findings in previous literature. Through further investigation, we find that the multi-clustering states reported previously occur due to the accumulation of discretization errors inherent in the integration algorithms, which introduce higher-mode couplings into the model. As a result, the partial integrability condition is violated.
Lastly, we derive the microscopic cross-correlation of globally coupled non-identical Kuramoto oscillators under common fluctuating forcing. The effect of correlation arises naturally in finite populations, due to the non-trivial fluctuations of the meanfield. In an idealized model, we approximate the finite-sized fluctuation by a Gaussian white noise. The analytical approximation qualitatively matches the measurements in numerical experiments, however, due to other periodic components inherent in the fluctuations of the mean-field there still exist significant inconsistencies.
Interfacial properties of morpholine-2,5-dione-based oligodepsipeptides and multiblock copolymers

Oligodepsipeptides (ODPs) with alternating amide and ester bonds prepared by ring-opening polymerization of morpholine-2,5-dione derivatives are promising matrices for drug delivery systems and building blocks for multifunctional biomaterials. Here, we elucidate the behavior of three telechelic ODPs and one multiblock copolymer containing ODP blocks at the air-water interface. Surprisingly, whereas the oligomers and multiblock copolymers crystallize in bulk, no crystallization is observed at the air-water interface. Furthermore, polarization modulation infrared reflection absorption spectroscopy is used to elucidate hydrogen bonding and secondary structures in ODP monolayers. The results will direct the development of the next ODP-based biomaterial generation with tailored properties for highly sophisticated applications.
A common feature in Answer Set Programming is the use of a second negation, stronger than default negation and sometimes called explicit, strong or classical negation. This explicit negation is normally used in front of atoms, rather than allowing its use as a regular operator. In this paper we consider the arbitrary combination of explicit negation with nested expressions, as those defined by Lifschitz, Tang and Turner. We extend the concept of reduct for this new syntax and then prove that it can be captured by an extension of Equilibrium Logic with this second negation. We study some properties of this variant and compare to the already known combination of Equilibrium Logic with Nelson's strong negation.
Temperature-memory technology was utilized to generate flat substrates with a programmable stiffness pattern from cross-linked poly(ethylene-co-vinyl acetate) substrates with cylindrical microstructures. Programmed substrates were obtained by vertical compression at temperatures in the range from 60 to 100 degrees C and subsequent cooling, whereby a flat substrate was achieved by compression at 72 degrees C, as documented by scanning electron microscopy and atomic force microscopy (AFM). AFM nanoindentation experiments revealed that all programmed substrates exhibited the targeted stiffness pattern. The presented technology for generating polymeric substrates with programmable stiffness pattern should be attractive for applications such as touchpads. optical storage, or cell instructive substrates.
Cold-regulated (COR) 15A is an intrinsically disordered protein (IDP) from Arabidopsis thaliana important for freezing tolerance. During freezing-induced cellular dehydration, COR15A transitions from a disordered to mostly alpha-helical structure. We tested whether mutations that increase the helicity of COR15A also increase its protective function. Conserved glycine residues were identified and mutated to alanine. Nuclear magnetic resonance (NMR) spectroscopy was used to identify residue-specific changes in helicity for wildtype (WT) COR15A and the mutants. Circular dichroism (CD) spectroscopy was used to monitor the coil-helix transition in response to increasing concentrations of trifluoroethanol (TFE) and ethylene glycol. The impact of the COR15A mutants on the stability of model membranes during a freeze-thaw cycle was investigated by fluorescence spectroscopy. The results of these experiments showed the mutants had a higher content of alpha-helical structure and the increased alpha-helicity improved membrane stabilization during freezing. Comparison of the TFE- and ethylene glycol-induced coil-helix transitions support our conclusion that increasing the transient helicity of COR15A in aqueous solution increases its ability to stabilize membranes during freezing. Altogether, our results suggest the conserved glycine residues are important for maintaining the disordered structure of COR15A but are also compatible with the formation of alpha-helical structure during freezing induced dehydration.
Farber disease is a rare lysosomal storage disorder resulting from acid ceramidase deficiency and subsequent ceramide accumulation. No treatments for Farber disease are clinically available, and affected patients have a severely shortened lifespan. We have recently reported a novel acid ceramidase deficiency model that mirrors the human disease closely. Acid sphingomyelinase is the enzyme that generates ceramide upstream of acid ceramidase in the lysosomes. Using our acid ceramidase deficiency model, we tested if acid sphingomyelinase could be a potential novel therapeutic target for the treatment of Farber disease. A number of functional acid sphingomyelinase inhibitors are clinically available and have been used for decades to treat major depression. Using these as a therapeutic for Farber disease, thus, has the potential to improve central nervous symptoms of the disease as well, something all other treatment options for Farber disease can’t achieve so far. As a proof-of-concept study, we first cross-bred acid ceramidase deficient mice with acid sphingomyelinase deficient mice in order to prevent ceramide accumulation. Double-deficient mice had reduced ceramide accumulation, fewer disease manifestations, and prolonged survival. We next targeted acid sphingomyelinase pharmacologically, to test if these findings would translate to a setting with clinical applicability. Surprisingly, the treatment of acid ceramidase deficient mice with the acid sphingomyelinase inhibitor amitriptyline was toxic to acid ceramidase deficient mice and killed them within a few days of treatment. In conclusion, our study provides the first proof-of-concept that acid sphingomyelinase could be a potential new therapeutic target for Farber disease to reduce disease manifestations and prolong survival. However, we also identified previously unknown toxicity of the functional acid sphingomyelinase inhibitor amitriptyline in the context of Farber disease, strongly cautioning against the use of this substance class for Farber disease patients
Zinc is an essential trace element, making it crucial to have a reliable biomarker for evaluating an individual’s zinc status. The total serum zinc concentration, which is presently the most commonly used biomarker, is not ideal for this purpose, but a superior alternative is still missing. The free zinc concentration, which describes the fraction of zinc that is only loosely bound and easily exchangeable, has been proposed for this purpose, as it reflects the highly bioavailable part of serum zinc. This report presents a fluorescence-based method for determining the free zinc concentration in human serum samples, using the fluorescent probe Zinpyr-1. The assay has been applied on 154 commercially obtained human serum samples. Measured free zinc concentrations ranged from 0.09 to 0.42 nM with a mean of 0.22 ± 0.05 nM. It did not correlate with age or the total serum concentrations of zinc, manganese, iron or selenium. A negative correlation between the concentration of free zinc and total copper has been seen for sera from females. In addition, the free zinc concentration in sera from females (0.21 ± 0.05 nM) was significantly lower than in males (0.23 ± 0.06 nM). The assay uses a sample volume of less than 10 µL, is rapid and cost-effective and allows us to address questions regarding factors influencing the free serum zinc concentration, its connection with the body’s zinc status, and its suitability as a future biomarker for an individual’s zinc status.
Matching participants (as suggested by Hope, 2015) may be one promising option for research on a potential bilingual advantage in executive functions (EF). In this study we first compared performances in three EF-tasks of a naturally heterogeneous sample of monolingual (n = 69, age = 9.0 y) and multilingual children (n = 57, age = 9.3 y). Secondly, we meticulously matched participants pairwise to obtain two highly homogeneous groups to rerun our analysis and investigate a potential bilingual advantage. The initally disadvantaged multilinguals (regarding socioeconomic status and German lexicon size) performed worse in updating and response inhibition, but similarly in interference inhibition. This indicates that superior EF compensate for the detrimental effects of the background variables. After matching children pairwise on age, gender, intelligence, socioeconomic status and German lexicon size, performances became similar except for interference inhibition. Here, an advantage for multilinguals in the form of globally reduced reaction times emerged, indicating a bilingual executive processing advantage.
Trace elements, like Cu, Zn, Fe, or Se, are important for the proper functioning of antioxidant enzymes. However, in excessive amounts, they can also act as pro-oxidants. Accordingly, trace elements influence redox-modulated signaling pathways, such as the Nrf2 pathway. Vice versa, Nrf2 target genes belong to the group of transport and metal binding proteins. In order to investigate whether Nrf2 directly regulates the systemic trace element status, we used mice to study the effect of a constitutive, whole-body Nrf2 knockout on the systemic status of Cu, Zn, Fe, and Se. As the loss of selenoproteins under Se-deprived conditions has been described to further enhance Nrf2 activity, we additionally analyzed the combination of Nrf2 knockout with feeding diets that provide either suboptimal, adequate, or supplemented amounts of Se. Experiments revealed that the Nrf2 knockout partially affected the trace element concentrations of Cu, Zn, Fe, or Se in the intestine, liver, and/or plasma. However, aside from Fe, the other three trace elements were only marginally modulated in an Nrf2-dependent manner. Selenium deficiency mainly resulted in increased plasma Zn levels. One putative mediator could be the metal regulatory transcription factor 1, which was up-regulated with an increasing Se supply and downregulated in Se-supplemented Nrf2 knockout mice.
Using two crystals for spontaneous parametric down-conversion in a parallel setup, we observe two-photon interference with high visibility. The high visibility is consistent with complementarity and the absence of which-path information. The observations are explained as the effects of entanglement or equivalently in terms of interfering probability amplitudes and also by the calculation of a second-order field correlation function in the Heisenberg picture. The latter approach brings out explicitly the role of the vacuum fields in the down-conversion at the crystals and in the photon coincidence counting. For comparison, we show that the Hong–Ou–Mandel dip can be explained by the same approach in which the role of the vacuum signal and idler fields, as opposed to entanglement involving vacuum states, is emphasized. We discuss the fundamental limitations of a theory in which these vacuum fields are treated as classical, stochastic fields.
Peroxisome biogenesis disorders (PBDs) are nontreatable hereditary diseases with a broad range of severity. Approximately 65% of patients are affected by mutations in the peroxins Pex1 and Pex6. The proteins form the heteromeric Pex1/Pex6 complex, which is important for protein import into peroxisomes. To date, no structural data are available for this AAA+ ATPase complex. However, a wealth of information can be transferred from low-resolution structures of the yeast scPex1/scPex6 complex and homologous, well-characterized AAA+ ATPases. We review the abundant records of missense mutations described in PBD patients with the aim to classify and rationalize them by mapping them onto a homology model of the human Pex1/Pex6 complex. Several mutations concern functionally conserved residues that are implied in ATP hydrolysis and substrate processing. Contrary to fold destabilizing mutations, patients suffering from function-impairing mutations may not benefit from stabilizing agents, which have been reported as potential therapeutics for PBD patients.
During the last few decades, the rapid separation of the Small Aral Sea from the isolated basin has changed its hydrological and ecological conditions tremendously. In the present study, we developed and validated the hybrid model for the Syr Darya River basin based on a combination of state-of-the-art hydrological and machine learning models. Climate change impact on freshwater inflow into the Small Aral Sea for the projection period 2007–2099 has been quantified based on the developed hybrid model and bias corrected and downscaled meteorological projections simulated by four General Circulation Models (GCM) for each of three Representative Concentration Pathway scenarios (RCP). The developed hybrid model reliably simulates freshwater inflow for the historical period with a Nash–Sutcliffe efficiency of 0.72 and a Kling–Gupta efficiency of 0.77. Results of the climate change impact assessment showed that the freshwater inflow projections produced by different GCMs are misleading by providing contradictory results for the projection period. However, we identified that the relative runoff changes are expected to be more pronounced in the case of more aggressive RCP scenarios. The simulated projections of freshwater inflow provide a basis for further assessment of climate change impacts on hydrological and ecological conditions of the Small Aral Sea in the 21st Century.
The aim of this study is to monitor short-term seasonal development of young Olympic weightlifters’ anthropometry, body composition, physical fitness, and sport-specific performance. Fifteen male weightlifters aged 13.2 ± 1.3 years participated in this study. Tests for the assessment of anthropometry (e.g., body-height, body-mass), body-composition (e.g., lean-body-mass, relative fat-mass), muscle strength (grip-strength), jump performance (drop-jump (DJ) height, countermovement-jump (CMJ) height, DJ contact time, DJ reactive-strength-index (RSI)), dynamic balance (Y-balance-test), and sport-specific performance (i.e., snatch and clean-and-jerk) were conducted at different time-points (i.e., T1 (baseline), T2 (9 weeks), T3 (20 weeks)). Strength tests (i.e., grip strength, clean-and-jerk and snatch) and training volume were normalized to body mass. Results showed small-to-large increases in body-height, body-mass, lean-body-mass, and lower-limbs lean-mass from T1-to-T2 and T2-to-T3 (∆0.7–6.7%; 0.1 ≤ d ≤ 1.2). For fat-mass, a significant small-sized decrease was found from T1-to-T2 (∆13.1%; d = 0.4) and a significant increase from T2-to-T3 (∆9.1%; d = 0.3). A significant main effect of time was observed for DJ contact time (d = 1.3) with a trend toward a significant decrease from T1-to-T2 (∆–15.3%; d = 0.66; p = 0.06). For RSI, significant small increases from T1-to-T2 (∆9.9%, d = 0.5) were noted. Additionally, a significant main effect of time was found for snatch (d = 2.7) and clean-and-jerk (d = 3.1) with significant small-to-moderate increases for both tests from T1-to-T2 and T2-to-T3 (∆4.6–11.3%, d = 0.33 to 0.64). The other tests did not change significantly over time (0.1 ≤ d ≤ 0.8). Results showed significantly higher training volume for sport-specific training during the second period compared with the first period (d = 2.2). Five months of Olympic weightlifting contributed to significant changes in anthropometry, body-composition, and sport-specific performance. However, hardly any significant gains were observed for measures of physical fitness. Coaches are advised to design training programs that target a variety of fitness components to lay an appropriate foundation for later performance as an elite athlete.

Im Nachvollzug der Konstellationen des Berührens zeigt sich, dass dem Berühren, als Berühren, nicht begrifflich beizukommen ist – es entzieht sich dem aneignenden Zugriff. Berühren ist eben nicht Begriff. Deshalb muss sich das Gastmahl der Berührung auf andere Weise nähern, nämlich berührend – wofür die narratologische Konstruktion des Textes von entscheidender Wichtigkeit ist. Many human infants grow up learning more than one language simultaneously but only recently has research started to study early language acquisition in this population more systematically. The paper gives an overview on findings on early language acquisition in bilingual infants during the first two years of life and compares these findings to current knowledge on early language acquisition in monolingual infants. Given the state of the research, the overview focuses on research on phonological and early lexical development in the first two years of life. We will show that the developmental trajectory of early language acquisition in these areas is very similar in mono- and bilingual infants suggesting that these early steps into language are guided by mechanisms that are rather robust against the differences in the conditions of language exposure that mono- and bilingual infants typically experience.
In a recent line of research, two familiar concepts from logic programming semantics (unfounded sets and splitting) were extrapolated to the case of epistemic logic programs. The property of epistemic splitting provides a natural and modular way to understand programs without epistemic cycles but, surprisingly, was only fulfilled by Gelfond's original semantics (G91), among the many proposals in the literature. On the other hand, G91 may suffer from a kind of self-supported, unfounded derivations when epistemic cycles come into play. Recently, the absence of these derivations was also formalised as a property of epistemic semantics called foundedness. Moreover, a first semantics proved to satisfy foundedness was also proposed, the so-called Founded Autoepistemic Equilibrium Logic (FAEEL). In this paper, we prove that FAEEL also satisfies the epistemic splitting property something that, together with foundedness, was not fulfilled by any other approach up to date. To prove this result, we provide an alternative characterisation of FAEEL as a combination of G91 with a simpler logic we called Founded Epistemic Equilibrium Logic (FEEL), which is somehow an extrapolation of the stable model semantics to the modal logic S5.
The current study investigates how bilingual children encode and produce morphologically complex words. We employed a silent-production-plus-delayed-vocalization paradigm in which event-related brain potentials (ERPs) were recorded during silent encoding of inflected words which were subsequently cued to be overtly produced. The bilingual children's spoken responses and their ERPs were compared to previous datasets from monolingual children on the same task. We found an enhanced negativity for regular relative to irregular forms during silent production in both bilingual children's languages, replicating the ERP effect previously obtained from monolingual children. Nevertheless, the bilingual children produced more morphological errors (viz. over-regularizations) than monolingual children. We conclude that mechanisms of morphological encoding (as measured by ERPs) are parallel for bilingual and monolingual children, and that the increased over-regularization rates are due to their reduced exposure to each of the two languages (relative to monolingual children).
The development of new and better optimization and approximation methods for Job Shop Scheduling Problems (JSP) uses simulations to compare their performance. The test data required for this has an uncertain influence on the simulation results, because the feasable search space can be changed drastically by small variations of the initial problem model. Methods could benefit from this to varying degrees. This speaks in favor of defining standardized and reusable test data for JSP problem classes, which in turn requires a systematic describability of the test data in order to be able to compile problem adequate data sets. This article looks at the test data used for comparing methods by literature review. It also shows how and why the differences in test data have to be taken into account. From this, corresponding challenges are derived which the management of test data must face in the context of JSP research.

The innovative dual-purpose chicken approach aims at contributing to the transition towards sustainable poultry production by avoiding the culling of male chickens. To successfully integrate sustainability aspects into innovation, goal congruency among actors and clearly communicating the added value within the actor network and to consumers is needed. The challenge of identifying common sustainability goals calls for decision support tools. The objectives of our research were to investigate whether the tool could assist in improving communication and marketing with respect to sustainability and optimizing the value chain organization. Three actor groups participated in the tool application, in which quantitative and qualitative data were collected. The results showed that there were manifold sustainability goals within the innovation network, but only some goals overlapped, and the perception of their implementation also diverged. While easily marketable goals such as ‘animal welfare’ were perceived as being largely implemented, economic goals were prioritized less often, and the implementation was perceived as being rather low. By visualizing congruencies and differences in the goals, the tool helped identify fields of action, such as improved information flows and prompted thinking processes. We conclude that the tool is useful for managing complex decision processes with several actors involved.
Children's online use of word order and morphosyntactic markers in Tagalog thematic role assignment

We investigated whether Tagalog-speaking children incrementally interpret the first noun as the agent, even if verbal and nominal markers for assigning thematic roles are given early in Tagalog sentences. We asked five- and seven-year-old children and adult controls to select which of two pictures of reversible actions matched the sentence they heard, while their looks to the pictures were tracked. Accuracy and eye-tracking data showed that agent-initial sentences were easier to comprehend than patient-initial sentences, but the effect of word order was modulated by voice. Moreover, our eye-tracking data provided evidence that, by the first noun phrase, seven-year-old children looked more to the target in the agent-initial compared to the patient-initial conditions, but this word order advantage was no longer observed by the second noun phrase. The findings support language processing and acquisition models which emphasize the role of frequency in developing heuristic strategies (e.g., Chang, Dell, & Bock, 2006).
Most of the matter in the universe consists of hydrogen. The hydrogen in the intergalactic medium (IGM), the matter between the galaxies, underwent a change of its ionisation state at the epoch of reionisation, at a redshift roughly between 6>z>10, or ~10^8 years after the Big Bang. At this time, the mostly neutral hydrogen in the IGM was ionised but the source of the responsible hydrogen ionising emission remains unclear. In this thesis I discuss the most likely candidates for the emission of this ionising radiation, which are a type of galaxy called Lyman alpha emitters (LAEs). As implied by their name, they emit Lyman alpha radiation, produced after a hydrogen atom has been ionised and recombines with a free electron. The ionising radiation itself (also called Lyman continuum emission) which is needed for this process inside the LAEs could also be responsible for ionising the IGM around those galaxies at the epoch of reionisation, given that enough Lyman continuum escapes. Through this mechanism, Lyman alpha and Lyman continuum radiation are closely linked and are both studied to better understand the properties of high redshift galaxies and the reionisation state of the universe.
Before I can analyse their Lyman alpha emission lines and the escape of Lyman continuum emission from them, the first step is the detection and correct classification of LAEs in integral field spectroscopic data, specifically taken with the Multi-Unit Spectroscopic Explorer (MUSE). After detecting emission line objects in the MUSE data, the task of classifying them and determining their redshift is performed with the graphical user interface QtClassify, which I developed during the work on this thesis. It uses the strength of the combination of spectroscopic and photometric information that integral field spectroscopy offers to enable the user to quickly identify the nature of the detected emission lines. The reliable classification of LAEs and determination of their redshifts is a crucial first step towards an analysis of their properties.
Through radiative transfer processes, the properties of the neutral hydrogen clouds in and around LAEs are imprinted on the shape of the Lyman alpha line. Thus after identifying the LAEs in the MUSE data, I analyse the properties of the Lyman alpha emission line, such as the equivalent width (EW) distribution, the asymmetry and width of the line as well as the double peak fraction. I challenge the common method of displaying EW distributions as histograms without taking the limits of the survey into account and construct a more independent EW distribution function that better reflects the properties of the underlying population of galaxies. I illustrate this by comparing the fraction of high EW objects between the two surveys MUSE-Wide and MUSE-Deep, both consisting of MUSE pointings (each with the size of one square arcminute) of different depths. In the 60 MUSE-Wide fields of one hour exposure time I find a fraction of objects with extreme EWs above EW_0>240A of ~20%, while in the MUSE-Deep fields (9 fields with an exposure time of 10 hours and one with an exposure time of 31 hours) I find a fraction of only ~1%, which is due to the differences in the limiting line flux of the surveys. The highest EW I measure is EW_0 = 600.63 +- 110A, which hints at an unusual underlying stellar population, possibly with a very low metallicity.
With the knowledge of the redshifts and positions of the LAEs detected in the MUSE-Wide survey, I also look for Lyman continuum emission coming from these galaxies and analyse the connection between Lyman continuum emission and Lyman alpha emission. I use ancillary Hubble Space Telescope (HST) broadband photometry in the bands that contain the Lyman continuum and find six Lyman continuum leaker candidates. To test whether the Lyman continuum emission of LAEs is coming only from those individual objects or the whole population, I select LAEs that are most promising for the detection of Lyman continuum emission, based on their rest-frame UV continuum and Lyman alpha line shape properties. After this selection, I stack the broadband data of the resulting sample and detect a signal in Lyman continuum with a significance of S/N = 5.5, pointing towards a Lyman continuum escape fraction of ~80%. If the signal is reliable, it strongly favours LAEs as the providers of the hydrogen ionising emission at the epoch of reionisation and beyond.

The present work introduces a metronome-driven speech elicitation paradigm in which participants were asked to utter repetitive sequences of elementary consonant-vowel syllables. This paradigm, explicitly designed to cover speech rates from a substantially wider range than has been explored so far in previous work, is demonstrated to satisfy the important prerequisites for assessing so far difficult to access aspects of speech. Specifically, the paradigm's extensive speech rate manipulation enabled elicitation of a great range of movement speeds as well as movement durations and excursions of the relevant effectors. The presence of such variation is a prerequisite to assessing whether invariant relations between these and other parameters exist and thus provides the foundation for a rigorous evaluation of the two laws examined in the first two contributions of this work.
In the data resulting from this paradigm, it is shown that speech movements obey the same fundamental laws as movements from other domains of motor control do. In particular, it is demonstrated that speech strongly adheres to the power law relation between speed and curvature of movement with a clear speech rate dependency of the power law's exponent. The often-sought or reported exponent of one third in the statement of the law is unique to a subclass of movements which corresponds to the range of faster rates under which a particular utterance is produced. For slower rates, significantly larger values than one third are observed. Furthermore, for the first time in speech this work uncovers evidence for the presence of Fitts' law. It is shown that, beyond a speaker-specific speech rate, speech movements of the tongue clearly obey Fitts' law by emergence of its characteristic linear relation between movement time and index of difficulty. For slower speech rates (when temporal pressure is small), no such relation is observed. The methods and datasets obtained in the two assessment above provide a rigorous foundation both for addressing implications for theories and models of speech as well as for better understanding the status of speech movements in the context of human movements in general.
All modern theories of language rely on a fundamental segmental hypothesis according to which the phonological message of an utterance is represented by a sequence of segments or phonemes. It is commonly assumed that each of these phonemes can be mapped to some unit of speech motor action, a so-called speech gesture.
For the first time here, it is demonstrated that the relation between the phonological description of simple utterances and the corresponding speech motor action is non-unique. Specifically, by the extensive speech rate manipulation in the herein used experimental paradigm it is demonstrated that speech exhibits clearly distinct dynamical organizations underlying the production of simple utterances. At slower speech rates, the dynamical organization underlying the repetitive production of elementary /CV/ syllables can be described by successive concatenations of closing and opening gestures, each with its own equilibrium point. As speech rate increases, the equilibria of opening and closing gestures are not equally stable yielding qualitatively different modes of organization with either a single equilibrium point of a combined opening-closing gesture or a periodic attractor unleashed by the disappearance of both equilibria. This observation, the non-uniqueness of the dynamical organization underlying what on the surface appear to be identical phonemic sequences, is an entirely new result in the domain of speech. Beyond that, the demonstration of periodic attractors in speech reveals that dynamical equilibrium point models do not account for all possible modes of speech motor behavior.
While some pronouncements of expert treaty bodies have been considered ‘key catalysts’ for the development of international human rights law, others are only selectively referred to in legal practice. This article argues that the varying normative impact is due to the informal character of pronouncements. In the absence of treaty provisions specifying their legal effect, practitioners tend to rely on different factors and arguments when either drawing on or rejecting certain pronouncements. Scholars in turn face difficulties when trying to identify explanatory patterns within this diverging practice as the informal character confronts both international lawyers and international relations scholars with their respective methodological ‘blind spots’. In light of these intradisciplinary challenges, this article explores the extent as to which an interdisciplinary approach helps to assess the reasons for the varying impact of pronouncements. After analysing the factors determining their legal significance on the basis of State practice and the academic debate, this article identifies the drafting process as a factor which promises to be particularly insightful when explored from an interdisciplinary perspective and sketches out a framework for future research.
Solar wind observations show that geomagnetic storms are mainly driven by interplanetary coronal mass ejections (ICMEs) and corotating or stream interaction regions (C/SIRs). We present a binary classifier that assigns one of these drivers to 7,546 storms between 1930 and 2015 using ground‐based geomagnetic field observations only. The input data consists of the long‐term stable Hourly Magnetospheric Currents index alongside the corresponding midlatitude geomagnetic observatory time series. This data set provides comprehensive information on the global storm time magnetic disturbance field, particularly its spatial variability, over eight solar cycles. For the first time, we use this information statistically with regard to an automated storm driver identification. Our supervised classification model significantly outperforms unskilled baseline models (78% accuracy with 26[19]% misidentified interplanetary coronal mass ejections [corotating or stream interaction regions]) and delivers plausible driver occurrences with regard to storm intensity and solar cycle phase. Our results can readily be used to advance related studies fundamental to space weather research, for example, studies connecting galactic cosmic ray modulation and geomagnetic disturbances. They are fully reproducible by means of the underlying open‐source software (Pick, 2019, http://doi.org/10.5880/GFZ.2.3.2019.003)
Garnet of eclogite (formerly termed garnet clinopyroxenite) hosted in lenses of orogenic garnet peridotite from the Granulitgebirge, NW Bohemian Massif, contains unique inclusions of granitic melt, now either glassy or crystallized. Analysed glasses and re‐homogenized inclusions are hydrous, peraluminous, and enriched in highly incompatible elements characteristic of the continental crust such as Cs, Li, B, Pb, Rb, Th, and U. The original melt thus represents a pristine, chemically evolved metasomatic agent, which infiltrated the mantle via deep continental subduction during the Variscan orogeny. The bulk chemical composition of the studied eclogites is similar to that of Fe‐rich basalt and the enrichment in LILE and U suggest a subduction‐related component. All these geochemical features confirm metasomatism. In comparison with many other garnet+clinopyroxene‐bearing lenses in peridotites of the Bohemian Massif, the studied samples from Rubinberg and Klatschmühle are more akin to eclogite than pyroxenites, as reflected in high jadeite content in clinopyroxene, relatively low Mg, Cr, and Ni but relatively high Ti. However, trace elements of both bulk rock and individual mineral phases show also important differences making these samples rather unique. Metasomatism involving a melt requiring a trace element pattern very similar to the composition reported here has been suggested for the source region of rocks of the so‐called durbachite suite, that is, ultrapotassic melanosyenites, which are found throughout the high‐grade Variscan basement. Moreover, the Th, U, Pb, Nb, Ta, and Ti patterns of these newly studied melt inclusions (MI) strongly resemble those observed for peridotite and its enclosed pyroxenite from the T‐7 borehole (Staré, České Středhoři Mountains) in N Bohemia. This suggests that a similar kind of crustal‐derived melt also occurred here. This study of granitic MI in eclogites from peridotites has provided the first direct characterization of a preserved metasomatic melt, possibly responsible for the metasomatism of several parts of the mantle in the Variscides.

Eine achtköpfige Delegation der Universität Potsdam (UP) besuchte vom 18. bis 21. November 2018 die Tel Aviv University (TAU) in Israel. Die Kooperation zwischen beiden Einrichtungen wurde durch einen »staff exchange« intensiviert. So konnten Mitarbeiter*innen der UP ihren Gegenpart an der TAU kennenlernen und sich inhaltlich austauschen. Der vorliegende Bericht basiert auf Gesprächen mit drei der insgesamt fünf Bibliotheksdirektorinnen sowie zwei weiteren Kolleginnen aus dem Bereich Erwerbung und Katalogisierung. Dabei wurden unterschiedliche Themenbereiche aus dem Bibliothekswesen und die Sicht beziehungsweise der Stand dazu an der TAU und der UP angesprochen. Der vorliegende Bericht geht auf die Themenfelder Bibliothekssystem, Open Access und Bibliothek als Raum ein.
Increasing concerns regarding the environmental impact of our chemical production have shifted attention towards possibilities for sustainable biotechnology. One-carbon (C1) compounds, including methane, methanol, formate and CO, are promising feedstocks for future bioindustry. CO2 is another interesting feedstock, as it can also be transformed using renewable energy to other C1 feedstocks for use. While formaldehyde is not suitable as a feedstock due to its high toxicity, it is a central intermediate in the process of C1 assimilation. This thesis explores formaldehyde metabolism and aims to engineer formaldehyde assimilation in the model organism Escherichia coli for the future C1-based bioindustry.
The first chapter of the thesis aims to establish growth of E. coli on formaldehyde via the most efficient naturally occurring route, the ribulose monophosphate pathway. Linear variants of the pathway were constructed in multiple-gene knockouts strains, coupling E. coli growth to the activities of the key enzymes of the pathway. Formaldehyde-dependent growth was achieved in rationally designed strains. In the final strain, the synthetic pathway provides the cell with almost all biomass and energy requirements.
In the second chapter, taking advantage of the unique feature of its reactivity, formaldehyde assimilation via condensation with glycine and pyruvate by two promiscuous aldolases was explored. Facilitated by these two reactions, the newly designed homoserine cycle is expected to support higher yields of a wide array of products than its counterparts. By dividing the pathway into segments and coupling them to the growth of dedicated strains, all pathway reactions were demonstrated to be sufficiently active. The work paves a way for future implementation of a highly efficient route for C1 feedstocks into commodity chemicals.
In the third chapter, the in vivo rate of the spontaneous formaldehyde tetrahydrofolate condensation to methylene-tetrahydrofolate was assessed in order to evaluate its applicability as a biotechnological process. Tested within an E. coli strain deleted in essential genes for native methylene-tetrahydrofolate biosynthesis, the reaction was shown to support the production of this essential intermediate. However, only low growth rates were observed and only at high formaldehyde concentrations. Computational analysis dependent on in vivo evidence from this strain deduced the slow rate of this spontaneous reaction, thus ruling out its substantial contribution to growth on C1 feedstocks.
The reactivity of formaldehyde makes it highly toxic. In the last chapter, the formation of thioproline, the condensation product of cysteine and formaldehyde, was confirmed to contribute this toxicity effect. Xaa-Pro aminopeptidase (PepP), which genetically links with folate metabolism, was shown to hydrolyze thioproline-containing peptides. Deleting pepP increased strain sensitivity to formaldehyde, pointing towards the toxicity of thioproline-containing peptides and the importance of their removal. The characterization in this study could be useful in handling this toxic intermediate.
Overall, this thesis identified challenges related to formaldehyde metabolism and provided novel solutions towards a future bioindustry based on sustainable C1 feedstocks in which formaldehyde serves as a key intermediate.

Design and Implementation of service-oriented architectures imposes a huge number of research questions from the fields of software engineering, system analysis and modeling, adaptability, and application integration. Component orientation and web services are two approaches for design and realization of complex web-based system. Both approaches allow for dynamic application adaptation as well as integration of enterprise application.
Commonly used technologies, such as J2EE and .NET, form de facto standards for the realization of complex distributed systems. Evolution of component systems has lead to web services and service-based architectures. This has been manifested in a multitude of industry standards and initiatives such as XML, WSDL UDDI, SOAP, etc. All these achievements lead to a new and promising paradigm in IT systems engineering which proposes to design complex software solutions as collaboration of contractually defined software services.
Service-Oriented Systems Engineering represents a symbiosis of best practices in object-orientation, component-based development, distributed computing, and business process management. It provides integration of business and IT concerns.
The annual Ph.D. Retreat of the Research School provides each member the opportunity to present his/her current state of their research and to give an outline of a prospective Ph.D. thesis. Dynamic earthquake rupture modeling provides information on the rupture physics as the rupture velocity, frictions or tractions acting during the rupture process. Nevertheless, as often based on spatial gridded preset geometries, dynamic modeling is depending on many free parameters leading to both a high non-uniqueness of the results and large computation times. That decreases the possibilities of full Bayesian error analysis.
To assess the named problems we developed the quasi-dynamic rupture model which is presented in this work. It combines the kinematic Eikonal rupture model with a boundary element method for quasi-static slip calculation.
The orientation of the modeled rupture plane is defined by a previously performed moment tensor inversion. The simultanously inverted scalar seismic moment allows an estimation of the extension of the rupture. The modeled rupture plane is discretized by a set of rectangular boundary elements. For each boundary element an applied traction vector is defined as the boundary value.
For insights in the dynamic rupture behaviour the rupture front propagation is calculated for incremental time steps based on the 2D Eikonal equation. The needed location-dependent rupture velocity field is assumed to scale linearly with a layered shear wave velocity field.
At each time all boundary elements enclosed within the rupture front are used to calculate the quasi-static slip distribution. Neither friction nor stress propagation are considered. Therefore the algorithm is assumed to be “quasi-static”. A series of the resulting quasi-static slip snapshots can be used as a quasi-dynamic model of the rupture process.
As many a priori information is used from the earth model (shear wave velocity and elastic parameters) and the moment tensor inversion (rupture extension and orientation) our model is depending on few free parameters as the traction field, the linear factor between rupture and shear wave velocity and the nucleation point and time. Hence stable and fast modeling results are obtained as proven from the comparison to different infinite and finite static crack solutions.
First dynamic applications show promissing results. The location-dependent rise time is automatically derived by the model. Different simple kinematic models as the slip-pulse or the penny-shaped crack model can be reproduced as well as their corresponding slip rate functions. A source time function (STF) approximation calculated from the cumulative sum of moment rates of each boundary element gives results similar to theoretical and empirical known STFs.
The model was also applied to the 2015 Illapel earthquake. Using a simple rectangular rupture geometry and a 2-layered traction regime yields good estimates of both the rupture front propagation and the slip patterns which are comparable to literature results. The STF approximation shows a good fit with previously published STFs.
The quasi-dynamic rupture model is hence able to fastly calculate reproducable slip results. That allows to test full Bayesian error analysis in the future. Further work on a full seismic source inversion or even a traction field inversion can also extend the scope of our model.
Die Universität Potsdam hat 2018 zum zweiten Mal erfolgreich am Times Higher Education-Ranking (THE-Ranking) teilgenommen. Hierfür wurde erneut das Publikationsaufkommen inklusive der Verbreitung der IDs von Autorinnen und Autoren, wie bspw. ORCiD, Researcher-ID oder Google Scholar-ID, ermittelt. Die aus den Erkenntnissen der ersten bibliometrischen Output-Analyse abgeleiteten Maßnahmen wurden nun hinsichtlich ihrer Wirksamkeit beurteilt. Der vorliegende Artikel vergleicht die Ergebnisse beider Analysen und thematisiert die Entwicklungen und Implikationen seit dem Ergreifen der ersten Maßnahmen.
The Postmasburg Manganese Field (PMF), Northern Cape Province, South Africa, once represented one of the largest sources of manganese ore worldwide. Two belts of manganese ore deposits have been distinguished in the PMF, namely the Western Belt of ferruginous manganese ores and the Eastern Belt of siliceous manganese ores. Prevailing models of ore formation in these two belts invoke karstification of manganese-rich dolomites and residual accumulation of manganese wad which later underwent diagenetic and low-grade metamorphic processes. For the most part, the role of hydrothermal processes and metasomatic alteration towards ore formation has not been adequately discussed. Here we report an abundance of common and some rare Al-, Na-, K- and Ba-bearing minerals, particularly aegirine, albite, microcline, banalsite, sérandite-pectolite, paragonite and natrolite in Mn ores of the PMF, indicative of hydrothermal influence. Enrichments in Na, K and/or Ba in the ores are generally on a percentage level for most samples analysed through bulk-rock techniques. The presence of As-rich tokyoite also suggests the presence of As and V in the hydrothermal fluid. The fluid was likely oxidized and alkaline in nature, akin to a mature basinal brine. Various replacement textures, particularly of Na- and K- rich minerals by Ba-bearing phases, suggest sequential deposition of gangue as well as ore-minerals from the hydrothermal fluid, with Ba phases being deposited at a later stage. The stratigraphic variability of the studied ores and their deviation from the strict classification of ferruginous and siliceous ores in the literature, suggests that a re-evaluation of genetic models is warranted. New Ar-Ar ages for K-feldspars suggest a late Neoproterozoic timing for hydrothermal activity. This corroborates previous geochronological evidence for regional hydrothermal activity that affected Mn ores at the PMF but also, possibly, the high-grade Mn ores of the Kalahari Manganese Field to the north. A revised, all-encompassing model for the development of the manganese deposits of the PMF is then proposed, whereby the source of metals is attributed to underlying carbonate rocks beyond the Reivilo Formation of the Campbellrand Subgroup. The main process by which metals are primarily accumulated is attributed to karstification of the dolomitic substrate. The overlying Asbestos Hills Subgroup banded iron formation (BIF) is suggested as a potential source of alkali metals, which also provides a mechanism for leaching of these BIFs to form high-grade residual iron ore deposits.
Hochschulen und deren Zentraleinrichtungen beschäftigen sich zunehmend mit dem Thema Forschungsdatenmanagement (FDM), um ihre Forschenden adäquat zu unterstützen. Nicht zuletzt aufgrund neuer Verlags- und Förderanforderungen wünschen sich Forschende Beratung und Orientierung, wie sie mit ihren Forschungsdaten umgehen sollen. Damit Hochschulen schnell und nachhaltig Lösungen zum institutionellen FDM etablieren können, haben fünf Berliner und Brandenburger Universitäten im gemeinsamen Verbundvorhaben FDMentor mit Förderung des Bundesministeriums für Bildung und Forschung (BMBF) entsprechende Leitfäden und Werkzeuge erarbeitet. Die innerhalb von zwei Jahren (2017–2019) entstandenen Ergebnisse in den Bereichen Strategieentwicklung, Forschungsdaten-Policy, rechtliche Aspekte und Kompetenzausbau finden über das Verbundprojekt hinaus ihre Anwendung.
Lanthanide-doped upconverting nanoparticles (UCNP) are being extensively studied for bioapplications due to their unique photoluminescence properties and low toxicity. Interest in RET applications involving UCNP is also increasing, but due to factors such as large sizes, ion emission distributions within the particles, and complicated energy transfer processes within the UCNP, there are still many questions to be answered. In this study, four types of core and core-shell NaYF4-based UCNP co-doped with Yb3+ and Tm3+ as sensitizer and activator, respectively, were investigated as donors for the Methyl 5-(8-decanoylbenzo[1,2-d:4,5-d ']bis([1,3]dioxole)-4-yl)-5-oxopentanoate (DBD-6) dye. The possibility of resonance energy transfer (RET) between UCNP and the DBD-6 attached to their surface was demonstrated based on the comparison of luminescence intensities, band ratios, and decay kinetics. The architecture of UCNP influenced both the luminescence properties and the energy transfer to the dye: UCNP with an inert shell were the brightest, but their RET efficiency was the lowest (17%). Nanoparticles with Tm3+ only in the shell have revealed the highest RET efficiencies (up to 51%) despite the compromised luminescence due to surface quenching.
Hasidic myth-activism

Since the 1970s, Buber has often been suspected of being a Volkish thinker. This essay reconsiders the affinity of Buber's late writings with Volkish ideology. It examines the allegations against Buber's Volkish thought in light of his later biblical and Hasidic writings. By illuminating the ideological affinity between these two modes of thought, the essay explains how Buber aims to depart from the dangers of myth without rejecting myth as such. I argue that Buber's relationship to myth can help us to explain his critique of nationalism. My basic argument is that in his struggle with hyper-nationalism, Buber follows the Baal Shem Tov and his struggle against Sabbateanism. Like the Besht, Buber does not reject myth, but seeks instead to repair it from within. Whereas hyper-nationalism uses myth to advance its political goals, Buber seeks to reposition ethics within a mythic framework. I view Buber's exegesis and commentaries on biblical and Hasidic myths as myth-activism.
Aluminum oxide is an Earth-abundant geological material, and its interaction with water is of crucial importance for geochemical and environmental processes. Some aluminum oxide surfaces are also known to be useful in heterogeneous catalysis, while the surface chemistry of aqueous oxide interfaces determines the corrosion, growth and dissolution of such materials. In this doctoral work, we looked mainly at the (0001) surface of α-Al 2 O 3 and its reactivity towards water. In particular, a great focus of this work is dedicated to simulate and address the vibrational spectra of water adsorbed on the α-alumina(0001) surface in various conditions and at different coverages. In fact, the main source of comparison and inspiration for this work comes from the collaboration with the “Interfacial Molecular Spectroscopy” group led by Dr. R. Kramer Campen at the Fritz-Haber Institute of the MPG in Berlin. The expertise of our project partners in surface-sensitive Vibrational Sum Frequency (VSF) generation spectroscopy was crucial to develop and adapt specific simulation schemes used in this work. Methodologically, the main approach employed in this thesis is Ab Initio Molecular Dynamics (AIMD) based on periodic Density Functional Theory (DFT) using the PBE functional with D2 dispersion correction. The analysis of vibrational frequencies from both a static and a dynamic, finite-temperature perspective offers the ability to investigate the water / aluminum oxide interface in close connection to experiment.
The first project presented in this work considers the characterization of dissociatively adsorbed deuterated water on the Al-terminated (0001) surface. This particular structure is known from both experiment and theory to be the thermodynamically most stable surface termination of α-alumina in Ultra-High Vacuum (UHV) conditions. Based on experiments performed by our colleagues at FHI, different adsorption sites and products have been proposed and identified for D 2 O. While previous theoretical investigations only looked at vibrational frequencies of dissociated OD groups by staticNormal Modes Analysis (NMA), we rather employed a more sophisticated approach to directly assess vibrational spectra (like IR and VSF) at finite temperature from AIMD. In this work, we have employed a recent implementation which makes use of velocity-velocity autocorrelation functions to simulate such spectral responses of O-H(D) bonds. This approach allows for an efficient and qualitatively accurate estimation of Vibrational Densities of States (VDOS) as well as IR and VSF spectra, which are then tested against experimental spectra from our collaborators.
In order to extend previous work on unimolecularly dissociated water on α-Al 2 O 3 , we then considered a different system, namely, a fully hydroxylated (0001) surface, which results from the reconstruction of the UHV-stable Al-terminated surface at high water contents. This model is then further extended by considering a hydroxylated surface with additional water molecules, forming a two-dimensional layer which serves as a potential template to simulate an aqueous interface in environmental conditions. Again, employing finite-temperature AIMD trajectories at the PBE+D2 level, we investigated the behaviour of both hydroxylated surface (HS) and the water-covered structure derived from it (known as HS+2ML). A full range of spectra, from VDOS to IR and VSF, is then calculated using the same methodology, as described above. This is the main focus of the second project, reported in Chapter 5. In this case, comparison between theoretical spectra and experimental data is definitely good. In particular, we underline the nature of high-frequency resonances observed above 3700 cm −1 in VSF experiments to be associated with surface OH-groups, known as “aluminols” which are a key fingerprint of the fully hydroxylated surface.
In the third and last project, which is presented in Chapter 6, the extension of VSF spectroscopy experiments to the time-resolved regime offered us the opportunity to investigate vibrational energy relaxation at the α-alumina / water interface. Specifically, using again DFT-based AIMD simulations, we simulated vibrational lifetimes for surface aluminols as experimentally detected via pump-probe VSF. We considered the water-covered HS model as a potential candidate to address this problem. The vibrational (IR) excitation and subsequent relaxation is performed by means of a non-equilibrium molecular dynamics scheme. In such a scheme, we specifically looked at the O-H stretching mode of surface aluminols. Afterwards, the analysis of non-equilibrium trajectories allows for an estimation of relaxation times in the order of 2-4 ps which are in overall agreement with measured ones.
The aim of this work has been to provide, within a consistent theoretical framework, a better understanding of vibrational spectroscopy and dynamics for water on the α-alumina(0001) surface,ranging from very low water coverage (similar to the UHV case) up to medium-high coverages, resembling the hydroxylated oxide in environmental moist conditions.
Modern health care systems are characterized by pronounced prevention and cost-optimized treatments. This dissertation offers novel empirical evidence on how useful such measures can be. The first chapter analyzes how radiation, a main pollutant in health care, can negatively affect cognitive health. The second chapter focuses on the effect of Low Emission Zones on public heath, as air quality is the major external source of health problems. Both chapters point out potentials for preventive measures. Finally, chapter three studies how changes in treatment prices affect the reallocation of hospital resources. In the following, I briefly summarize each chapter and discuss implications for health care systems as well as other policy areas. Based on the National Educational Panel Study that is linked to data on radiation, chapter one shows that radiation can have negative long-term effects on cognitive skills, even at subclinical doses. Exploiting arguably exogenous variation in soil contamination in Germany due to the Chernobyl disaster in 1986, the findings show that people exposed to higher radiation perform significantly worse in cognitive tests 25 years later. Identification is ensured by abnormal rainfall within a critical period of ten days. The results show that the effect is stronger among older cohorts than younger cohorts, which is consistent with radiation accelerating cognitive decline as people get older. On average, a one-standarddeviation increase in the initial level of CS137 (around 30 chest x-rays) is associated with a decrease in the cognitive skills by 4.1 percent of a standard deviation (around 0.05 school years). Chapter one shows that sub-clinical levels of radiation can have negative consequences even after early childhood. This is of particular importance because most of the literature focuses on exposure very early in life, often during pregnancy. However, population exposed after birth is over 100 times larger. These results point to substantial external human capital costs of radiation which can be reduced by choices of medical procedures. There is a large potential for reductions because about one-third of all CT scans are assumed to be not medically justified (Brenner and Hall, 2007). If people receive unnecessary CT scans because of economic incentives, this chapter points to additional external costs of health care policies. Furthermore, the results can inform the cost-benefit trade-off for medically indicated procedures. Chapter two provides evidence about the effectiveness of Low Emission Zones. Low Emission Zones are typically justified by improvements in population health. However, there is little evidence about the potential health benefits from policy interventions aiming at improving air quality in inner-cities. The chapter ask how the coverage of Low Emission Zones air pollution and hospitalization, by exploiting variation in the roll out of Low Emission Zones in Germany. It combines information on the geographic coverage of Low Emission Zones with rich panel data on the universe of German hospitals over the period from 2006 to 2016 with precise information on hospital locations and the annual frequency of detailed diagnoses. In order to establish that our estimates of Low Emission Zones’ health impacts can indeed be attributed to improvements in local air quality, we use data from Germany’s official air pollution monitoring system and assign monitor locations to Low Emission Zones and test whether measures of air pollution are affected by the coverage of a Low Emission Zone. Results in chapter two confirm former results showing that the introduction of Low Emission Zones improved air quality significantly by reducing NO2 and PM10 concentrations. Furthermore, the chapter shows that hospitals which catchment areas are covered by a Low Emission Zone, diagnose significantly less air pollution related diseases, in particular by reducing the incidents of chronic diseases of the circulatory and the respiratory system. The effect is stronger before 2012, which is consistent with a general improvement in the vehicle fleet’s emission standards. Depending on the disease, a one-standard-deviation increase in the coverage of a hospitals catchment area covered by a Low Emission Zone reduces the yearly number of diagnoses up to 5 percent. These findings have strong implications for policy makers. In 2015, overall costs for health care in Germany were around 340 billion euros, of which 46 billion euros for diseases of the circulatory system, making it the most expensive type of disease caused by 2.9 million cases (Statistisches Bundesamt, 2017b). Hence, reductions in the incidence of diseases of the circulatory system may directly reduce society’s health care costs. Whereas chapter one and two study the demand-side in health care markets and thus preventive potential, chapter three analyzes the supply-side. By exploiting the same hospital panel data set as in chapter two, chapter three studies the effect of treatment price shocks on the reallocation of hospital resources in Germany. Starting in 2005, the implementation of the German-DRG-System led to general idiosyncratic treatment price shocks for individual hospitals. Thus far there is little evidence of the impact of general price shocks on the reallocation of hospital resources. Additionally, I add to the exiting literature by showing that price shocks can have persistent effects on hospital resources even when these shocks vanish. However, simple OLS regressions would underestimate the true effect, due to endogenous treatment price shocks. I implement a novel instrument variable strategy that exploits the exogenous variation in the number of days of snow in hospital catchment areas. A peculiarity of the reform allowed variation in days of snow to have a persistent impact on treatment prices. I find that treatment price increases lead to increases in input factors such as nursing staff, physicians and the range of treatments offered but to decreases in the treatment volume. This indicates supplier-induced demand. Furthermore, the probability of hospital mergers and privatization decreases. Structural differences in pre-treatment characteristics between hospitals enhance these effects. For instance, private and larger hospitals are more affected. IV estimates reveal that OLS results are biased towards zero in almost all dimensions because structural hospital differences are correlated with the reallocation of hospital resources. These results are important for several reasons. The G-DRG-Reform led to a persistent polarization of hospital resources, as some hospitals were exposed to treatment price increases, while others experienced reductions. If hospitals increase the treatment volume as a response to price reductions by offering unnecessary therapies, it has a negative impact on population wellbeing and public spending. However, results show a decrease in the range of treatments if prices decrease. Hospitals might specialize more, thus attracting more patients. From a policy perspective it is important to evaluate if such changes in the range of treatments jeopardize an adequate nationwide provision of treatments. Furthermore, the results show a decrease in the number of nurses and physicians if prices decrease. This could partly explain the nursing crisis in German hospitals. However, since hospitals specialize more they might be able to realize efficiency gains which justify reductions in input factors without loses in quality. Further research is necessary to provide evidence for the impact of the G-DRG-Reform on health care quality. Another important aspect are changes in the organizational structure. Many public hospitals have been privatized or merged. The findings show that this is at least partly driven by the G-DRG-Reform. This can again lead to a lack in services offered in some regions if merged hospitals specialize more or if hospitals are taken over by ecclesiastical organizations which do not provide all treatments due to moral conviction. Overall, this dissertation reveals large potential for preventive health care measures and helps to explain reallocation processes in the hospital sector if treatment prices change. Furthermore, its findings have potentially relevant implications for other areas of public policy. Chapter one identifies an effect of low dose radiation on cognitive health. As mankind is searching for new energy sources, nuclear power is becoming popular again. However, results of chapter one point to substantial costs of nuclear energy which have not been accounted yet. Chapter two finds strong evidence that air quality improvements by Low Emission Zones translate into health improvements, even at relatively low levels of air pollution. These findings may, for instance, be of relevance to design further policies targeted at air pollution such as diesel bans. As pointed out in chapter three, the implementation of DRG-Systems may have unintended side-effects on the reallocation of hospital resources. This may also apply to other providers in the health care sector such as resident doctors.
La carte de Tupaia constitue l’un des artéfacts les plus célèbres et les plus énigmatiques à émerger des toutes premières rencontres entre Européens et îliens du Pacifique. Elle a été élaborée entre août 1769 et février 1770 par Tupaia, prêtre ’arioi, conseiller royal et maître de navigation originaire de Ra’iātea, aux Îles Sous-le-Vent de la Société. En collaboration avec divers membres d’équipage de l’Endeavour de James Cook, en deux temps distincts de cartographie et trois ébauches. L’identité de bien des îles qui y figurent et la logique de leur agencement demeuraient jusqu’à présent des énigmes. En se fiant en partie à des pièces d’archives restées ignorées, nous proposons, dans ce long essai, une nouvelle compréhension de sa logique cartographique, une reconstitution détaillée de sa genèse et donc, pour la toute première fois, une lecture exhaustive. La carte de Tupaia n’illustre pas seulement la magnitude et la maîtrise de la navigation polynésienne, elle réalise aussi une remarquable synthèse représentationnelle de deux systèmes d’orientation très différents.
This paper addresses issues of translating both words and rituals as Muslim cemetery keepers care for Jewish graves and recite traditional prayers for the dead in Morocco. Several issues of translation must be dealt with while considering these rare and disappearing practices. The first issue to be discussed is the translation of Hebrew inscriptions into French by cemetery keepers. One cemetery keeper in Meknes has tried to compile an exhaustive index of the names and dates represented on the gravestones under her care. The Muslim guard of the Jewish cemetery in Sefrou, on the other hand, has somewhat famously told visitors differing stories about his ability and willingness to pray the Kaddish over the graves of emigrated relatives who cannot return to mark an anniversary death. These practices provide the context for considering how the act of Muslims caring for Jewish graves creates linguistic and ritual translations of traditional Jewish ancestor care.
Rabbi Jacob ben Isaac of Yanova (d. 1623) is best known as the author of the Ze’enah U-Re’enah; the Melits Yosher (“Intercessor before God”) is one of his lesser known works. It was first published in Lublin in 1622 and reprinted once in Amsterdam in 1688. Like the Ze’enah U-Re’enah, it was a Torah commentary, but composed for men who had some yeshivah education, but who could not continue their studies. The commentary on the Song of Songs by Isaac Sulkes is another Yiddish work that addresses the same audience as the Melits Yosher. The purpose of this article is to bring to scholarly attention an audience that has not been noticed or studied in the previous scholarship on early modern Yiddish literature.
Carbonfasern haben sich in der Luft- und Raumfahrt etabliert und gewinnen in Alltagsanwendungen wie dem Automobilbereich, Windkraft- und Sportbereich durch ihre hohen Zugfestigkeiten, insbesondere ihrer hohen E-Moduli, und ihrer geringen Dichte immer mehr an Bedeutung. Auf Grund ihrer hohen Kosten, welche sich zur Hälfte aus der Precursorherstellung, inklusive seiner Synthese und seinem Verspinnprozess, dem Lösungsspinnverfahren, ergeben, erhalten zunehmend alternative und schmelzspinnbare Precursoren Interesse. Für die Carbonfaserherstellung wird fast ausschließlich Polyacrylnitril (PAN) verwendet, das vor dem Schmelzen irreversible exotherme Zyklisierungsreaktionen aufweist, welchen sich seine Zersetzung anschließt. Eine Möglichkeit der Reduzierung der Schmelztemperatur von Polymeren ist die Einbringung von Comonomeren zur Erhöhung des freien Volumens und die Reduzierung der intermolekularen Wechselwirkungen als interne Weichmacher. Wie am Fraunhofer IAP gezeigt wurde, kann mittels 2-Methoxyethylacrylat (MEA) die Schmelztemperatur zu neuartigen PAN-basierten Precursoren verringert werden. Um den PAN-co-MEA-Precursor für die nachfolgenden Prozessschritte der Carbonfaserherstellung zu verwenden, müssen die thermoplastischen Fasern in thermisch stabile Fasern ohne thermoplastisches Verhalten überführt werden. Es wurde ein neuer Prozessschritt (Prästabilisierung) eingeführt, welcher unter alkalischen Bedingungen zur Abspaltung der Comonomerseitenkette führt. Neben der Esterhydrolyse finden Reaktionen statt, welche an diesem Material noch nicht hinreichend untersucht wurden. Weiterhin stellt sich die Frage nach der Kinetik der Prästabilisierung und der Ermittlung einer geeigneten Prozessführung.
Hierzu wurde die Prästabilisierung in den Labormaßstab überführt und die möglichen Zusammensetzungen des aus DMSO und einer KOH-Lösung bestehenden Reaktionsmediums evaluiert. Weiterhin wurde die Behandlung bei verschiedenen Prästabilisierungszeiten von maximal 30 min und Temperaturen von 40, 50 und 60 °C durchgeführt, um primär mittels NMR-Spektroskopie die chemischen Strukturänderungen aufzuklären. Die Esterhydrolyse des Comonomers, welche zur Abspaltung des 2-Methoxyethanols führt, wurde mittels 1H-NMR-spektroskopischer Untersuchungen detektiert.
Es wurde ein Modell aufgestellt, das die chemisch-physikalischen Strukturänderungen während der Prästabilisierung aufzeigt. Die zuerst ablaufende Reaktion ist die Esterhydrolyse am Comonomer, welche vom Faserrand nach innen verläuft und durch die Präsenz des DMSO in Kombination mit der KOH-Lösung (Superbase) initiiert wird. Der zeitliche Reaktionsverlauf der Esterhydrolyse kann in drei Bereiche eingeteilt werden. Der erste Bereich ab dem Prästabilisierungsbeginn wird durch die Diffusion der basischen Anionen in die Faser, der zweite Bereich durch die Reaktion an der Estergruppe des Comonomers und der dritte Bereich durch letzte Reaktionen im Faserinneren und diffusiven Prozessen der Produkte und Edukte charakterisiert. Der zweite Bereich kann mit einer Reaktion pseudo 1. Ordnung abgebildet werden, da in diesem Bereich bereits eine ausreichende Diffusion der Edukte in die Faser stattgefunden hat. Bei 50 °C spielt die Diffusion im ersten Bereich im Vergleich zur Reaktion eine untergeordnete Rolle. Mit Erhöhung der Temperatur auf 60 °C kann eine im Verhältnis geringere Diffusions- als Reaktionsgeschwindigkeit beobachtet werden. Die Nebenreaktionen wurden mittels 13C-CP/MAS-NMR-spektroskopischen, elementaranlaytischen Untersuchungen sowie Doppelbrechungsmessungen charakterisiert. Während der alkalischen Esterhydrolyse beginnt die Reduzierung der Nitrilgruppen unter der Bildung von primären Carbonsäureamiden und Carbonsäuren. Zur Beschreibung dieser Umsetzung wurde eine Methode entwickelt, welche die Addition von 13C-CP/MAS-NMR-Spektren der Modellsubstanzen PAN, PAM und PAA beinhaltet. Weitere stattfindende Reaktionen sind die Bildung von konjugierten Doppelbindungen, welche insbesondere auf eine Zyklisierung der Nitrile hinweisen. Die nasschemisch initiierte Zyklisierung der Nitrilgruppen kann zu kürzeren Stabilisierungszeiten und einem besser kontrollierbaren Stabilisierungsprozess durch geringere Wärmefreisetzung und schlussendlich zu einer Kostenersparnis des gesamten Verfahrens führen. Die Umsetzung der Nitrilgruppen konnte mit einer Reaktion pseudo 1. Ordnung gut abgebildet werden. DMSO initiiert die Esterhydrolyse, wobei die KOH-Konzentration einen höheren Einfluss auf die Reaktionsgeschwindigkeit der Ester- und Nitrilhydrolyse als die DMSO-Konzentration besitzt. Beide Reaktionen zeigen eine vergleichbare Abhängigkeit von der Temperatur. Die Erhöhung der Prästabilisierungszeit und der KOH- bzw. DMSO-Konzentration führt zur Migration niedermolekularer Bestandteile des Fasermaterials an die Oberfläche und der Bildung punktueller Ablagerungen bis hin zu miteinander verbundenen Einzelfasern. Eine weitere Erhöhung der Prästabilisierungszeit bzw. der Konzentration führt zu einem steigenden Carbonsäureanteil und zur Quellung des Fasermaterials, wodurch die Ablagerungen in das Reaktionsmedium diffundieren. Die Ablagerungen enthalten Chlor, welches durch den Waschvorgang mit HCl in das Materialsystem gelangt ist und durch Parameteranpassungen reduziert wurde. Die schmelzbaren Fasern konnten durch die Prästabilisierung erfolgreich über eine Kern-Mantel-Struktur in nicht-thermoplastische Fasern überführt werden.
Zur Ermittlung eines geeigneten Prozessfensters für nachfolgende thermische Beanspruchungen der prästabilisierten Fasern wurden drei Kriterien identifiziert, anhand welcher die Evaluation erfolgte. Das erste Kriterium beinhaltet die Notwendigkeit der vollständigen Aufhebung der thermoplastischen Eigenschaft der Fasern. Als zweites Kriterium diente die Fasermorphologie. Anhand von REM-Aufnahmen wurden Faserbündel mit separierten Einzelfasern ohne Ablagerungen für die nachfolgende Stabilisierung ausgewählt. Das dritte Kriterium bezieht sich auf eine möglichst geringe Umsetzung der Nitrilgruppen, um Prästabilisierungsbedingungen mit Nebenreaktionen zu vermeiden.
Aus den Untersuchungen konnte eine Prästabilisierungstemperatur von 60 °C als geeignet identifiziert werden. Weiterhin führen hoch alkalische Zusammensetzungen des Reaktionsmediums mit KOH-Konzentrationen von 1, 1,5 und 2 M, vorzugsweise 1,5 M und 50 vol% DMSO mit Reaktionszeiten von unter 10 min zu geeigneten Fasern. Ein MEA-Anteil unterhalb von 2 mol% bewirkt eine Überführung in die Unschmelzbarkeit. Thermisch stabile und für die nachfolgende Stabilisierung geeignete Fasern besitzen weiterhin 68 – 80 mol% Nitrilgruppen, 20 – 25 mol% Carbonsäuren, bis zu 15 mol% primäre Carbonsäureamide und zyklisierte Strukturen.
Additive Manufacturing (AM) in terms of laser powder-bed fusion (L-PBF) offers new prospects regarding the design of parts and enables therefore the production of lattice structures. These lattice structures shall be implemented in various industrial applications (e.g. gas turbines) for reasons of material savings or cooling channels. However, internal defects, residual stress, and structural deviations from the nominal geometry are unavoidable.
In this work, the structural integrity of lattice structures manufactured by means of L-PBF was non-destructively investigated on a multiscale approach.
A workflow for quantitative 3D powder analysis in terms of particle size, particle shape, particle porosity, inter-particle distance and packing density was established. Synchrotron computed tomography (CT) was used to correlate the packing density with the particle size and particle shape. It was also observed that at least about 50% of the powder porosity was released during production of the struts.
Struts are the component of lattice structures and were investigated by means of laboratory CT. The focus was on the influence of the build angle on part porosity and surface quality. The surface topography analysis was advanced by the quantitative characterisation of re-entrant surface features. This characterisation was compared with conventional surface parameters showing their complementary information, but also the need for AM specific surface parameters.
The mechanical behaviour of the lattice structure was investigated with in-situ CT under compression and successive digital volume correlation (DVC). The deformation was found to be knot-dominated, and therefore the lattice folds unit cell layer wise.
The residual stress was determined experimentally for the first time in such lattice structures. Neutron diffraction was used for the non-destructive 3D stress investigation. The principal stress directions and values were determined in dependence of the number of measured directions. While a significant uni-axial stress state was found in the strut, a more hydrostatic stress state was found in the knot. In both cases, strut and knot, seven directions were at least needed to find reliable principal stress directions.
For many years, psycholinguistic evidence has been predominantly based on findings from native speakers of Indo-European languages, primarily English, thus providing a rather limited perspective into the human language system. In recent years a growing body of experimental research has been devoted to broadening this picture, testing a wide range of speakers and languages, aiming to understanding the factors that lead to variability in linguistic performance. The present dissertation investigates sources of variability within the morphological domain, examining how and to what extent morphological processes and representations are shaped by specific properties of languages and speakers. Firstly, the present work focuses on a less explored language, Hebrew, to investigate how the unique non-concatenative morphological structure of Hebrew, namely a non-linear combination of consonantal roots and vowel patterns to form lexical entries (L-M-D + CiCeC = limed ‘teach’), affects morphological processes and representations in the Hebrew lexicon. Secondly, a less investigated population was tested: late learners of a second language. We directly compare native (L1) and non-native (L2) speakers, specifically highly proficient and immersed late learners of Hebrew. Throughout all publications, we have focused on a morphological phenomenon of inflectional classes (called binyanim; singular: binyan), comparing productive (class Piel, e.g., limed ‘teach’) and unproductive (class Paal, e.g., lamad ‘learn’) verbal inflectional classes. By using this test case, two psycholinguistic aspects of morphology were examined: (i) how morphological structure affects online recognition of complex words, using masked priming (Publications I and II) and cross-modal priming (Publication III) techniques, and (ii) what type of cues are used when extending morpho-phonological patterns to novel complex forms, a process referred to as morphological generalization, using an elicited production task (Publication IV).
The findings obtained in the four manuscripts, either published or under review, provide significant insights into the role of productivity in Hebrew morphological processing and generalization in L1 and L2 speakers. Firstly, the present L1 data revealed a close relationship between productivity of Hebrew verbal classes and recognition process, as revealed in both priming techniques. The consonantal root was accessed only in the productive class (Piel) but not the unproductive class (Paal). Another dissociation between the two classes was revealed in the cross-modal priming, yielding a semantic relatedness effect only for Paal but not Piel primes. These findings are taken to reflect that the Hebrew mental representations display a balance between stored undecomposable unstructured stems (Paal) and decomposed structured stems (Piel), in a similar manner to a typical dual-route architecture, showing that the Hebrew mental lexicon is less unique than previously claimed in psycholinguistic research. The results of the generalization study, however, indicate that there are still substantial differences between inflectional classes of Hebrew and other Indo-European classes, particularly in the type of information they rely on in generalization to novel forms. Hebrew binyan generalization relies more on cues of argument structure and less on phonological cues.
Secondly, clear L1/L2 differences were observed in the sensitivity to abstract morphological and morpho-syntactic information during complex word recognition and generalization. While L1 Hebrew speakers were sensitive to the binyan information during recognition, expressed by the contrast in root priming, L2 speakers showed similar root priming effects for both classes, but only when the primes were presented in an infinitive form. A root priming effect was not obtained for primes in a finite form. These patterns are interpreted as evidence for a reduced sensitivity of L2 speakers to morphological information, such as information about inflectional classes, and evidence for processing costs in recognition of forms carrying complex morpho-syntactic information. Reduced reliance on structural information cues was found in production of novel verbal forms, when the L2 group displayed a weaker effect of argument structure for Piel responses, in comparison to the L1 group. Given the L2 results, we suggest that morphological and morphosyntactic information remains challenging for late bilinguals, even at high proficiency levels.
This thesis investigates whether multilingual speakers’ use of grammatical constraints in an additional language (La) is affected by the native (L1) and non-native grammars (L2) of their linguistic repertoire.
Previous studies have used untimed measures of grammatical performance to show that L1 and L2 grammars affect the initial stages of La acquisition. This thesis extends this work by examining whether speakers at intermediate levels of La proficiency, who demonstrate mature untimed/offline knowledge of the target La constraints, are differentially affected by their L1 and L2 knowledge when they comprehend sentences under processing pressure. With this purpose, several groups of La German speakers were tested on word order and agreement phenomena using online/timed measures of grammatical knowledge. Participants had mirror distributions of their prior languages and they were either L1English/L2Spanish speakers or L1Spanish/L2English speakers. Crucially, in half of the phenomena the target La constraint aligned with English but not with Spanish, while in the other half it aligned with Spanish but not with English. Results show that the L1 grammar plays a major role in the use of La constraints under processing pressure, as participants displayed increased sensitivity to La constraints when they aligned with their L1, and reduced sensitivity when they did not. Further, in specific phenomena in which the L2 and La constraints aligned, increased L2 proficiency resulted in an enhanced sensitivity to the La constraint. These findings suggest that both native and non-native grammars affect how speakers use La grammatical constraints under processing pressure. However, L1 and L2 grammars differentially influence on participants’ performance: While L1 constraints seem to be reliably recruited to cope with the processing demands of real-time La use, proficiency in an L2 can enhance sensitivity to La constraints only in specific circumstances, namely when L2 and La constraints align.
Phytoplankton growth depends not only on the mean intensity but also on the dynamics of the light supply. The nonlinear light-dependency of growth is characterized by a small number of basic parameters: the compensation light intensity PARcompμ, where production and losses are balanced, the growth efficiency at sub-saturating light αµ, and the maximum growth rate at saturating light µmax. In surface mixed layers, phytoplankton may rapidly move between high light intensities and almost darkness. Because of the different frequency distribution of light and/or acclimation processes, the light-dependency of growth may differ between constant and fluctuating light. Very few studies measured growth under fluctuating light at a sufficient number of mean light intensities to estimate the parameters of the growth-irradiance relationship. Hence, the influence of light dynamics on µmax, αµ and PARcompμ are still largely unknown. By extension, accurate modelling predictions of phytoplankton development under fluctuating light exposure remain difficult to make. This PhD thesis does not intend to directly extrapolate few experimental results to aquatic systems – but rather improving the mechanistic understanding of the variation of the light-dependency of growth under light fluctuations and effects on phytoplankton development.
In Lake TaiHu and at the Three Gorges Reservoir (China), we incubated phytoplankton communities in bottles placed either at fixed depths or moved vertically through the water column to mimic vertical mixing. Phytoplankton at fixed depths received only the diurnal changes in light (defined as constant light regime), while phytoplankton received rapidly fluctuating light by superimposing the vertical light gradient on the natural sinusoidal diurnal sunlight. The vertically moved samples followed a circular movement with 20 min per revolution, replicating to some extent the full overturn of typical Langmuir cells. Growth, photosynthesis, oxygen production and respiration of communities (at Lake TaiHu) were
measured. To complete these investigations, a physiological experiment was performed in the laboratory on a toxic strain of Microcystis aeruginosa (FACBH 1322) incubated under 20 min period fluctuating light. Here, we measured electron transport rates and net oxygen production at a much higher time resolution (single minute timescale).
The present PhD thesis provides evidence for substantial effects of fluctuating light on the eco-physiology of phytoplankton. Both experiments performed under semi-natural conditions in Lake TaiHu and at the Three Gorges Reservoir gave similar results. The significant decline in community growth efficiencies αµ under fluctuating light was caused for a great share by different frequency distribution of light intensities that shortened the effective daylength for production. The remaining gap in community αµ was attributed to species-specific photoacclimation mechanisms and to light-dependent respiratory losses. In contrast, community maximal growth rates µmax were similar between incubations at constant and fluctuating light. At daily growth saturating light supply, differences in losses for biosynthesis between the two light regimes were observed. Phytoplankton experiencing constant light suffered photo-inhibition - leading to photosynthesis foregone and additional respiratory costs for photosystems repair. On the contrary, intermittent exposure to low and high light intensities prevented photo-inhibition of mixed algae but forced them to develop alternative light strategy. They better harvested and exploited surface irradiance by enhancing their photosynthesis. In the laboratory, we showed that Microcystis aeruginosa increased its oxygen consumption by dark respiration in the light few minutes only after exposure to increasing light intensities. More, we proved that within a simulated Langmuir cell, the net production at saturating light and the compensation light intensity for production at limiting light are positively related. These results are best explained by an accumulation of photosynthetic products at increasing irradiance and mobilization of these fresh resources by rapid enhancement of dark respiration for maintenance and biosynthesis at decreasing irradiance. At the daily timescale, we showed that the enhancement of photosynthesis at high irradiance for biosynthesis of species increased their maintenance respiratory costs at limiting light. Species-specific growth at saturating light µmax and compensation light intensity for growth PARcompμ of species incubated in Lake TaiHu were positively related. Because of this species-specific physiological tradeoff, species displayed different light affinities to limiting and saturating light - thereby exhibiting a gleaner-opportunist tradeoff. In Lake TaiHu, we showed that inter-specific differences in light acquisition traits (µmax and PARcompμ) allowed coexis¬tence of species on a gradient of constant
light while avoiding competitive exclusion. More interestingly we demonstrated for the first time that vertical mixing (inducing fluctuating light supply for phytoplankton) may alter or even reverse the light utilization strategies of species within couple of days. The intra-specific variation in traits under fluctuating light increased the niche space for acclimated species, precluding competitive exclusion.
Overall, this PhD thesis contributes to a better understanding of phytoplankton eco-physiology under fluctuating light supply. This work could enhance the quality of predictions of phytoplankton development under certain weather conditions or climate change scenarios.
L’extériorisation de toute communication est assujettie à un mode d’accès du locuteur aux informations véhiculées. Les constatations faites de nos données prouvent que tous les huit verbes étudiés traduisent des mécanismes d’acquisition des connaissances que nous avons appelés en emprunt à (Vogeleer, 1995 :92) « l’accès cognitif au savoir ». C’est cette valeur intrinsèque qui vaut à ces termes la dénomination de verbes médiatifs. En d’autres mots, ce sont des éléments qui explicitent des processus d’accès du locuteur au savoir. Une source du savoir qui peut être directe (la vue, le touché, l’ouïe, l’odorat…) ou indirecte (ouï-dire) et surtout inférée. Nous entendons par inférence un processus d’analyse et de mise en relation d’éléments (prémisses), lesquelles permettent de tirer une conclusion par déduction, induction ou par abduction. Et selon que lesdites prémisses tendent à être plus ou moins fiables, ces processus inférentiels impliqueront des valeurs épistémiques à des degrés divers.
Sur le plan rhétorico-syntaxique, nos analyses ont montré tous les verbes cognitifs (VC) de cette étude exigent l’occurrence d’autres constituants (actants) phrastiques qu’ils régissent. C’est grâce à cette valence verbale qu’ils gardent un pouvoir rectionnel dans les constructions asyndétiques. Ce sont donc les matrices des éléments sur lesquels ils se rapportent. Quant au cinétisme de ces verbes, il possède une fonction rhétorique et syntaxique. En effet, cet agencement particulier et souvent perturbant permet de traduire l’expression d’une figure de syntaxe à effet rhétorique : l’hyperbate. Une construction atypique qui, à travers les agencements anticonformistes, donne un sens de regressivité à l’énoncé et confère une saillance à des termes mis ce fait en exergue.
Accurate weather observations are the keystone to many quantitative applications, such as precipitation monitoring and nowcasting, hydrological modelling and forecasting, climate studies, as well as understanding precipitation-driven natural hazards (i.e. floods, landslides, debris flow). Weather radars have been an increasingly popular tool since the 1940s to provide high spatial and temporal resolution precipitation data at the mesoscale, bridging the gap between synoptic and point scale observations. Yet, many institutions still struggle to tap the potential of the large archives of reflectivity, as there is still much to understand about factors that contribute to measurement errors, one of which is calibration. Calibration represents a substantial source of uncertainty in quantitative precipitation estimation (QPE). A miscalibration of a few dBZ can easily deteriorate the accuracy of precipitation estimates by an order of magnitude. Instances where rain cells carrying torrential rains are misidentified by the radar as moderate rain could mean the difference between a timely warning and a devastating flood.
Since 2012, the Philippine Atmospheric, Geophysical, and Astronomical Services Administration (PAGASA) has been expanding the country’s ground radar network. We had a first look into the dataset from one of the longest running radars (the Subic radar) after devastating week-long torrential rains and thunderstorms in August 2012 caused by the annual southwestmonsoon and enhanced by the north-passing Typhoon Haikui. The analysis of the rainfall spatial distribution revealed the added value of radar-based QPE in comparison to interpolated rain gauge observations. However, when compared with local gauge measurements, severe miscalibration of the Subic radar was found. As a consequence, the radar-based QPE would have underestimated the rainfall amount by up to 60% if they had not been adjusted by rain gauge observations—a technique that is not only affected by other uncertainties, but which is also not feasible in other regions of the country with very sparse rain gauge coverage.
Relative calibration techniques, or the assessment of bias from the reflectivity of two radars, has been steadily gaining popularity. Previous studies have demonstrated that reflectivity observations from the Tropical Rainfall Measuring Mission (TRMM) and its successor, the Global Precipitation Measurement (GPM), are accurate enough to serve as a calibration reference for ground radars over low-to-mid-latitudes (± 35 deg for TRMM; ± 65 deg for GPM). Comparing spaceborne radars (SR) and ground radars (GR) requires cautious consideration of differences in measurement geometry and instrument specifications, as well as temporal coincidence. For this purpose, we implement a 3-D volume matching method developed by Schwaller and Morris (2011) and extended by Warren et al. (2018) to 5 years worth of observations from the Subic radar. In this method, only the volumetric intersections of the SR and GR beams are considered.
Calibration bias affects reflectivity observations homogeneously across the entire radar domain. Yet, other sources of systematic measurement errors are highly heterogeneous in space, and can either enhance or balance the bias introduced by miscalibration. In order to account for such heterogeneous errors, and thus isolate the calibration bias, we assign a quality index to each matching SR–GR volume, and thus compute the GR calibration bias as a qualityweighted average of reflectivity differences in any sample of matching SR–GR volumes. We exemplify the idea of quality-weighted averaging by using beam blockage fraction (BBF) as a quality variable. Quality-weighted averaging is able to increase the consistency of SR and GR observations by decreasing the standard deviation of the SR–GR differences, and thus increasing the precision of the bias estimates.
To extend this framework further, the SR–GR quality-weighted bias estimation is applied to the neighboring Tagaytay radar, but this time focusing on path-integrated attenuation (PIA) as the source of uncertainty. Tagaytay is a C-band radar operating at a lower wavelength and is therefore more affected by attenuation. Applying the same method used for the Subic radar, a time series of calibration bias is also established for the Tagaytay radar.
Tagaytay radar sits at a higher altitude than the Subic radar and is surrounded by a gentler terrain, so beam blockage is negligible, especially in the overlapping region. Conversely, Subic radar is largely affected by beam blockage in the overlapping region, but being an SBand radar, attenuation is considered negligible. This coincidentally independent uncertainty contributions of each radar in the region of overlap provides an ideal environment to experiment with the different scenarios of quality filtering when comparing reflectivities from the two ground radars. The standard deviation of the GR–GR differences already decreases if we consider either BBF or PIA to compute the quality index and thus the weights. However, combining them multiplicatively resulted in the largest decrease in standard deviation, suggesting that taking both factors into account increases the consistency between the matched samples.
The overlap between the two radars and the instances of the SR passing over the two radars at the same time allows for verification of the SR–GR quality-weighted bias estimation method. In this regard, the consistency between the two ground radars is analyzed before and after bias correction is applied. For cases when all three radars are coincident during a significant rainfall event, the correction of GR reflectivities with calibration bias estimates from SR overpasses dramatically improves the consistency between the two ground radars which have shown incoherent observations before correction. We also show that for cases where adequate SR coverage is unavailable, interpolating the calibration biases using a moving average can be used to correct the GR observations for any point in time to some extent. By using the interpolated biases to correct GR observations, we demonstrate that bias correction reduces the absolute value of the mean difference in most cases, and therefore improves the consistency between the two ground radars.
This thesis demonstrates that in general, taking into account systematic sources of uncertainty that are heterogeneous in space (e.g. BBF) and time (e.g. PIA) allows for a more consistent estimation of calibration bias, a homogeneous quantity. The bias still exhibits an unexpected variability in time, which hints that there are still other sources of errors that remain unexplored. Nevertheless, the increase in consistency between SR and GR as well as between the two ground radars, suggests that considering BBF and PIA in a weighted-averaging approach is a step in the right direction.
Despite the ample room for improvement, the approach that combines volume matching between radars (either SR–GR or GR–GR) and quality-weighted comparison is readily available for application or further scrutiny. As a step towards reproducibility and transparency in atmospheric science, the 3D matching procedure and the analysis workflows as well as sample data are made available in public repositories. Open-source software such as Python and wradlib are used for all radar data processing in this thesis. This approach towards open science provides both research institutions and weather services with a valuable tool that can be applied to radar calibration, from monitoring to a posteriori correction of archived data.
Hepcidin-25 (Hep-25) plays a crucial role in the control of iron homeostasis. Since the dysfunction of the hepcidin pathway leads to multiple diseases as a result of iron imbalance, hepcidin represents a potential target for the diagnosis and treatment of disorders of iron metabolism. Despite intense research in the last decade targeted at developing a selective immunoassay for iron disorder diagnosis and treatment and better understanding the ferroportin-hepcidin interaction, questions remain. The key to resolving these underlying questions is acquiring exact knowledge of the 3D structure of native Hep-25. Since it was determined that the N-terminus, which is responsible for the bioactivity of Hep-25, contains a small Cu(II)-binding site known as the ATCUN motif, it was assumed that the Hep-25-Cu(II) complex is the native, bioactive form of the hepcidin. This structure has thus far not been elucidated in detail. Owing to the lack of structural information on metal-bound Hep-25, little is known about its possible biological role in iron metabolism. Therefore, this work is focused on structurally characterizing the metal-bound Hep-25 by NMR spectroscopy and molecular dynamics simulations. For the present work, a protocol was developed to prepare and purify properly folded Hep-25 in high quantities. In order to overcome the low solubility of Hep-25 at neutral pH, we introduced the C-terminal DEDEDE solubility tag. The metal binding was investigated through a series of NMR spectroscopic experiments to identify the most affected amino acids that mediate metal coordination. Based on the obtained NMR data, a structural calculation was performed in order to generate a model structure of the Hep-25-Ni(II) complex. The DEDEDE tag was excluded from the structural calculation due to a lack of NMR restraints. The dynamic nature and fast exchange of some of the amide protons with solvent reduced the overall number of NMR restraints needed for a high-quality structure. The NMR data revealed that the 20 Cterminal Hep-25 amino acids experienced no significant conformational changes, compared to published results, as a result of a pH change from pH 3 to pH 7 and metal binding. A 3D model of the Hep-25-Ni(II) complex was constructed from NMR data recorded for the hexapeptideNi(II) complex and Hep-25-DEDEDE-Ni(II) complex in combination with the fixed conformation of 19 C-terminal amino acids. The NMR data of the Hep-25-DEDEDE-Ni(II) complex indicates that the ATCUN motif moves independently from the rest of the structure. The 3D model structure of the metal-bound Hep-25 allows for future works to elucidate hepcidin’s interaction with its receptor ferroportin and should serve as a starting point for the development of antibodies with improved selectivity.
Arboreal epiphytes (plants residing in forest canopies) are present across all major climate zones and play important roles in forest biogeochemistry. The substantial water storage capacity per unit area of the epiphyte “bucket” is a key attribute underlying their capability to influence forest hydrological processes and their related mass and energy flows. It is commonly assumed that the epiphyte bucket remains saturated, or near-saturated, most of the time; thus, epiphytes (particularly vascular epiphytes) can store little precipitation, limiting their impact on the forest canopy water budget. We present evidence that contradicts this common assumption from (i) an examination of past research; (ii) new datasets on vascular epiphyte and epi-soil water relations at a tropical montane cloud forest (Monteverde, Costa Rica); and (iii) a global evaluation of non-vascular epiphyte saturation state using a process-based vegetation model, LiBry. All analyses found that the external and internal water storage capacity of epiphyte communities is highly dynamic and frequently available to intercept precipitation. Globally, non-vascular epiphytes spend <20% of their time near saturation and regionally, including the humid tropics, model results found that non-vascular epiphytes spend ~1/3 of their time in the dry state (0–10% of water storage capacity). Even data from Costa Rican cloud forest sites found the epiphyte community was saturated only 1/3 of the time and that internal leaf water storage was temporally dynamic enough to aid in precipitation interception. Analysis of the epi-soils associated with epiphytes further revealed the extent to which the epiphyte bucket emptied—as even the canopy soils were often <50% saturated (29–53% of all days observed). Results clearly show that the epiphyte bucket is more dynamic than currently assumed, meriting further research on epiphyte roles in precipitation interception, redistribution to the surface and chemical composition of “net” precipitation waters reaching the surface.
Fractures serve as highly conductive preferential flow paths for fluids in rocks, which are difficult to exactly reconstruct in numerical models. Especially, in low-conductive rocks, fractures are often the only pathways for advection of solutes and heat. The presented study compares the results from hydraulic and tracer tomography applied to invert a theoretical discrete fracture network (DFN) that is based on data from synthetic cross-well testing. For hydraulic tomography, pressure pulses in various injection intervals are induced and the pressure responses in the monitoring intervals of a nearby observation well are recorded. For tracer tomography, a conservative tracer is injected in different well levels and the depth-dependent breakthrough of the tracer is monitored. A recently introduced transdimensional Bayesian inversion procedure is applied for both tomographical methods, which adjusts the fracture positions, orientations, and numbers based on given geometrical fracture statistics. The used Metropolis-Hastings-Green algorithm is refined by the simultaneous estimation of the measurement error’s variance, that is, the measurement noise. Based on the presented application to invert the two-dimensional cross-section between source and the receiver well, the hydraulic tomography reveals itself to be more suitable for reconstructing the original DFN. This is based on a probabilistic representation of the inverted results by means of fracture probabilities.
Surface modification by polyzwitterions of the sulfabetaine-type, and their resistance to biofouling

Films of zwitterionic polymers are increasingly explored for conferring fouling resistance to materials. Yet, the structural diversity of polyzwitterions is rather limited so far, and clear structure-property relationships are missing. Therefore, we synthesized a series of new polyzwitterions combining ammonium and sulfate groups in their betaine moieties, so-called poly(sulfabetaine)s. Their chemical structures were varied systematically, the monomers carrying methacrylate, methacrylamide, or styrene moieties as polymerizable groups. High molar mass homopolymers were obtained by free radical polymerization. Although their solubilities in most solvents were very low, brine and lower fluorinated alcohols were effective solvents in most cases. A set of sulfabetaine copolymers containing about 1 mol % (based on the repeat units) of reactive benzophenone methacrylate was prepared, spin-coated onto solid substrates, and photo-cured. The resistance of these films against the nonspecific adsorption by two model proteins (bovine serum albumin—BSA, fibrinogen) was explored, and directly compared with a set of references. The various polyzwitterions reduced protein adsorption strongly compared to films of poly(n-butyl methacrylate) that were used as a negative control. The poly(sulfabetaine)s showed generally even somewhat higher anti-fouling activity than their poly(sulfobetaine) analogues, though detailed efficacies depended on the individual polymer–protein pairs. Best samples approach the excellent performance of a poly(oligo(ethylene oxide) methacrylate) reference.
