We have isolated a cDNA coding for a putative invertebrate-type dopamine receptor (Peadop2) from P. americana brain by using a PCR-based strategy. The mRNA is present in samples from brain and salivary glands. We analyzed the distribution of the PeaDOP2 receptor protein with specific affinity-purified polyclonal antibodies. On Western blots, PeaDOP2 was detected in protein samples from brain, subesophageal ganglion, thoracic ganglia, and salivary glands. In immunocytochemical experiments, we detected PeaDOP2 in neurons with their somata being located at the anterior edge of the medulla bilaterally innervating the optic lobes and projecting to the ventro-lateral protocerebrum. In order to determine the functional and pharmacological properties of the cloned receptor, we generated a cell line constitutively expressing PeaDOP2. Activation of PeaDOP2-expressing cells with dopamine induced an increase in intracellular cAMP. In contrast, a C-terminally truncated splice variant of this receptor did not exhibit any functional property by itself. The molecular and pharmacological characterization of the first dopamine receptor from P. americana provides the basis for forthcoming studies focusing on the significance of the dopaminergic system in cockroach behavior and physiology.
Die nichtproteinogene Aminosäure GABA (γ-Aminobuttersäure) gilt als der wichtigste inhibitorische Neurotransmitter im Zentralnervensystem von Vertebraten sowie Invertebraten und vermittelt ihre Wirkung u. a. über die metabotropen GABAB-Rezeptoren. Bisher sind diese Rezeptoren bei Insekten nur rudimentär untersucht. Für die Amerikanische Großschabe als etablierter Modellorganismus konnte pharmakologisch eine modulatorische Rolle der GABAB-Rezeptoren bei der Bildung von Primärspeichel nachgewiesen werden. Ziel dieser Arbeit war eine umfassende Charakterisierung der GABAB-Rezeptor-Subtypen 1 und 2 von Periplaneta americana. Unter Verwendung verschiedenster Klonierungsstrategien sowie der Kooperationsmöglichkeit mit der Arbeitsgruppe von Prof. Dr. T. Miura (Hokkaido, Japan) in Hinsicht auf eine dort etablierte P. americana EST-Datenbank gelang die Klonierung von zwei Rezeptor-cDNAs. Die Analyse der abgeleiteten Aminosäuresequenzen auf GB-spezifische Domänen und konservierte Aminosäure-Reste, sowie der Vergleich zu bekannten GB Sequenzen anderer Arten legen nahe, dass es sich bei den isolierten Sequenzen um die GABAB-Rezeptor-Subtypen 1 und 2 (PeaGB1 und PeaGB2) handelt. Für die funktionelle und pharmakologische Charakterisierung des Heteromers aus PeaGB1 und PeaGB2 wurden Expressionskonstrukte für die Transfektion in HEK-flpTM-Zellen hergestellt. Das Heteromer aus PeaGB1 und PeaGB2 hemmt bei steigenden GABA-Konzentrationen die cAMP-Produktion. Die Substanzen SKF97541 und 3-APPA konnten als Agonisten identifiziert werden. CGP55845 und CGP54626 wirken als vollwertige Antagonisten. Das in vitro ermittelte pharmakologische Profil im Vergleich zur Pharmakologie an der isolierten Drüse bestätigt, dass die GABA-Wirkung in der Speicheldrüse tatsächlich von GBs vermittelt wird. Für die immunhistochemische Charakterisierung konnte ein spezifischer polyklonaler Antikörper gegen die extrazelluläre Schleife 2 des PeaGB1 generiert werden. Ein weiterer Antikörper, welcher gegen den PeaGB2 gerichtet ist, erwies sich hingegen nicht als ausreichend spezifisch. Western-Blot-Analysen bestätigen das Vorkommen beider Subtypen im Zentralnervensystem von P. americana. Zudem wird der PeaGB1 in der Speicheldrüse und in den Geschlechtsdrüsen der Schabenmännchen exprimiert. Immunhistochemische Analysen zeigen eine PeaGB1-ähnliche Markierung in den GABAergen Fasern der Speicheldrüse auf. Demnach fungiert der PeaGB1 hier als Autorezeptor. Weiterhin konnte eine PeaGB1-ähnliche Markierung in nahezu allen Gehirnneuropilen festgestellt werden. Auch die akzessorischen Drüsen der Männchen, Pilzdrüse und Phallusdrüse, sind PeaGB1-immunreaktiv.
Salivary gland cells of the blowfly Calliphora vicina have a vacuolar-type H+-ATPase (V-ATPase) that lies in their apical membrane and energizes the secretion of a KCl-rich primary saliva upon stimulation with serotonin (5-hydroxytryptamine). Whether and to what extent V-ATPase contributes to intracellular pH (pH(i)) regulation in unstimulated gland cells is unknown. We used the fluorescent dye BCECF to study intracellular pH(i) regulation microfluorometrically and show that: (1) under resting conditions, the application of Na+-free physiological saline induces an intracellular alkalinization attributable to the inhibition of the activity of a Na+-dependent glutamate transporter; (2) the maintenance of resting pHi is Na+, Cl-, concanamycin A and DIDS sensitive; (3) recovery from an intracellular acid load is Na+ sensitive and requires V-ATPase activity; (4) the Na+/H+ antiporter is not involved in pHi recovery after a NH4Cl prepulse; and (5) at least one Na+-dependent transporter and the V-ATPase maintain recovery from an intracellular acid load. Thus, under resting conditions, the V-ATPase and at least one Na+-dependent transporter maintain normal pH(i) values of pH.7.5. We have also detected the presence of a Na+-dependent glutamate transporter, which seems to act as an acid loader. Despite this not being a common pH(i)-regulating transporter, its activity affects steady-state pH(i) in C. vicina salivary gland cells.
Die acinösen Speicheldrüsen der Schabe Periplaneta americana sind reich durch serotonerge, dopaminerge und GABAerge Fasern innerviert. Die biogenen Amine Serotonin (5-HT) und Dopamin (DA) induzieren die Sekretion eines NaCl-haltigen Primärspeichels. Die physiologische Rolle der GABAergen Innervation des Drüsenkomplexes war bislang unbekannt. Weiterhin wurde vermutet, dass Tyramin (TA) und Octopamin (OA) an der Speichelbildung beteiligt sind. Mittels intrazellulärer Ableitungen von sekretorischen Acinuszellen mit und ohne Stimulierung des Speicheldrüsennervs (SDN) sollte daher die Wirkung von GABA, TA und OA im Speicheldrüsenkomplex untersucht werden. Intrazelluläre Ableitungen aus Acinuszellen zeigten, dass sowohl DA als auch 5 HT biphasische Änderungen des Membranpotentials induzierten. Diese bestanden aus einer initialen Hyperpolarisation und einer darauf folgenden transienten Depolarisation. Stimulierung des SDN mittels einer Saugelektrode verursachte ebenfalls biphasische Änderungen des Membranpotentials der Acinuszellen, die mit den DA- bzw. 5-HT-induzierten Änderungen kinetisch identisch waren. Dieses Ergebnis zeigte, dass die elektrische Stimulierung des SDN im Nerv-Speicheldrüsenpräparat eine verlässliche Methode zur Untersuchung der Wirkungen von Neuromodulatoren auf die dopaminerge und/oder sertotonerge Neurotransmission ist. Die Hyperpolarisation der DA-induzierten Potentialänderungen wurde durch eine intrazelluläre Ca2+-Freisetzung und die Öffnung basolateral lokalisierter Ca2+-gesteuerter K+-Kanäle verur-sacht. Die DA- und 5-HT-induzierte Depolarisation hing kritisch von der Aktivität eines basolateral lokalisierten Na+-K+-2Cl--Symporters ab. GABA, TA und OA potenzierten die elektrischen Antworten der Acinuszellen, wenn diese durch SDN-Stimulierung hervorgerufen wurden. Dabei war OA wirksamer als TA. Dieses Ergebnis zeigte, dass diese Substanzen als im Drüsenkomplex präsynaptisch und erregend als Neuromodulatoren wirken. Pharmakologische Untersuchungen ergaben, dass die erregende Wirkung von GABA durch einen G-Protein-gekoppelten GABAB-Rezeptor vermittelt wurde. Messungen der durch SDN-Stimulierung induzierten Flüssigkeits- und Proteinsekretionsraten zeigten, dass beide Parameter in Anwesenheit von GABA verstärkt waren. Dies ließ auf eine verstärkte serotonerge Neurotransmission schließen, da nur 5-HT die Bildung eines Protein-haltigen Speichels verursacht. Immuncytochemische Untersuchungen zeigten, dass die Drüsen tyraminerge und octopaminerge Innervation empfangen. Weiterhin wurde der erste charakterisierte TA-Rezeptor (PeaTYR1) der Schabe auf einem paarigen, lateral zur Drüse ziehenden Nerv markiert, der auch tyraminerge Fasern enthielt. Die vorliegende Arbeit trug zum Verständnis der komplexen Funktionsweise der Speicheldrüse der Schabe bei und erweiterte das lückenhafte Wissen über die neuronale Kontrolle exokriner Drüsen in Insekten.
The phenolamines octopamine and tyramine control, regulate, and modulate many physiological and behavioral processes in invertebrates. Vertebrates possess only small amounts of both substances, and thus, octopamine and tyramine, together with other biogenic amines, are referred to as “trace amines.” Biogenic amines evoke cellular responses by activating G-protein-coupled receptors. We have isolated a complementary DNA (cDNA) that encodes a biogenic amine receptor from the American cockroach Periplaneta americana, viz., Peatyr1, which shares high sequence similarity to members of the invertebrate tyramine-receptor family. The PeaTYR1 receptor was stably expressed in human embryonic kidney (HEK) 293 cells, and its ligand response has been examined. Receptor activation with tyramine reduces adenylyl cyclase activity in a dose-dependent manner (EC50 350 nM). The inhibitory effect of tyramine is abolished by co-incubation with either yohimbine or chlorpromazine. Receptor expression has been investigated by reverse transcription polymerase chain reaction and immunocytochemistry. The mRNA is present in various tissues including brain, salivary glands, midgut, Malpighian tubules, and leg muscles. The effect of tyramine on salivary gland acinar cells has been investigated by intracellular recordings, which have revealed excitatory presynaptic actions of tyramine. This study marks the first comprehensive molecular, pharmacological, and functional characterization of a tyramine receptor in the cockroach.
Source, topography and excitatory effects of GABAergic innervation in cockroach salivary glands

Cockroach salivary glands are innervated by dopaminergic and serotonergic neurons. Both transmitters elicit saliva secretion. We studied the distribution pattern of neurons containing gamma-aminobutyric acid ( GABA) and their physiological role. Immunofluorescence revealed a GABA-immunoreactive axon that originates within the subesophageal ganglion at the salivary neuron 2 (SN2) and this extends within the salivary duct nerve towards the salivary gland. GABA-positive fibers form a network on most acinar lobules and a dense plexus in the interior of a minor fraction of acinar lobules. Co-staining with anti-synapsin revealed that some putative GABAergic terminals seem to make pre-synaptic contacts with GABA-negative release sites. Many putative GABAergic release sites are at some distance from other synapses and at distance from the acinar tissue. Intracellular recordings from isolated salivary glands have revealed that GABA does not affect the basolateral membrane potential of the acinar cells directly. When applied during salivary duct nerve stimulation, GABA enhances the electrical response of the acinar cells and increases the rates of fluid and protein secretion. The effect on electrical cell responses is mimicked by the GABA(B) receptor agonists baclofen and SKF97541, and blocked by the GABAB receptor antagonists CGP52432 and CGP54626. These findings indicate that GABA has a modulatory role in the control of salivation, acting presynaptically on serotonergic and/or dopaminergic neurotransmission.
The acinar salivary gland of the cockroach, Periplaneta americana, is innervated by dopaminergic and serotonergic nerve fibers. Stimulation of the glands by serotonin (5-hydroxytryptamine, 5-HT) results in the production of a protein-rich saliva, whereas stimulation by dopamine results in saliva that is protein-free. Thus, dopamine acts selectively on ion-transporting peripheral cells within the acini, and 5-HT acts on protein-producing central cells. We have investigated the pharmacology of the 5-HT-induced secretory activity of isolated salivary glands of P. americana by testing several 5-HT receptor agonists and antagonists. The effects of 5-HT can be mimicked by the non-selective 5-HT receptor agonist 5-methoxytryptamine. All tested agonists that display at least some receptor subtype specificity in mammals, i.e., 5-carboxamidotryptamine, (+/-)-8-OH-DPAT, (+/-)-DOI, and AS 19, were ineffective in stimulating salivary secretion. 5-HT-induced secretion can be blocked by the vertebrate 5-HT receptor antagonists methiothepin, cyproheptadine, and mianserin. Flüssigkeitssekretion und Proteinsekretion werden in Speicheldrüsen von Insekten über Hormone und Neurotransmitter gesteuert. Diese entfalten ihre physiologische Wirkung in den sekretorischen Drüsenzellen hauptsächlich über den zyklischen Adenosinmonophosphat (cAMP)-Signalweg und den Inositoltrisphosphat (IP<SUB>3</SUB>) / Ca<sup>2+-Signalweg. Die Mechanismen möglicher Wechselwirkungen zwischen diesen Signalwegen und ihre physiologischen Auswirkungen sind unzureichend bekannt. Im Mittelpunkt dieser Arbeit stand die Frage, ob und wie sich der Ca<sup>2+-Signalweg und der cAMP-Signalweg in der Speicheldrüse der Diptere Calliphora vicina beeinflussen. Substanzen wie 5-Fluoro-α-Methyltryptamin und Histamin wurden in früheren Arbei-ten als Agonisten genutzt, um in den Speicheldrüsen von C. vicina selektiv den cAMP-Signalweg (getrennt vom IP<SUB>3</SUB>/Ca<sup>2+-Signalweg) zu aktivieren. Es zeigte sich in transepithelialen Potentialmessungen und mikrofluorometrischen Ca<sup>2+-Untersuchungen, dass beide Substanzen sowohl den cAMP-Weg als auch den Ca<sup>2+-Signalweg aktivierten. Die physiologischen Ursachen der Histamin-induzierten Ca<sup>2+-Erhöhung wurden genauer untersucht. Zusammengefasst zeigten diese Untersuchungen, dass Histamin wie 5-HT den cAMP-Weg und die Phosphoinositidkaskade aktivierte. Im Gegensatz zu den 5-HT-induzierten Ca<sup>2+-Oszillationen, welche durch interzelluläre Ca<sup>2+-Wellen synchronisiert werden, verursachte Histamin bei niedrigen Konzentrationen lokale Ca<sup>2+-Oszillationen in einzelnen Zellen (keine Wellen). Bei höheren Histamin-Konzentrationen war eine anhaltende Ca<sup>2+-Erhöhung oder ein synchrones <quote>Ca<sup>2+-beating</quote> in der gesamten Drüse zu beobachten. Des Weiteren wurde die Frage untersucht, ob eine Erhöhung der intrazellulären cAMP-Konzentration den IP<SUB>3</SUB> Ca<sup>2+-Signalweg in den Epithelzellen der Speicheldrüse beeinflussen kann. Es zeigte sich, dass cAMP den durch schwellennahe 5-HT-Konzentrationen induzierten Ca<sup>2+-Anstieg verstärkte. Diese Verstärkung wurde durch eine PKA-vermittelte Sensitivierung des IP<SUB>3</SUB>-Rezeptor/Ca<sup>2+-Kanals für IP<SUB>3</SUB> verursacht. Immunzytochemische Untersuchungen deuten dar-auf hin, dass die Proteinkinase A eng mit dem IP<SUB>3</SUB>-Rezeptor/Ca<sup>2+-Kanal assoziiert ist. Diese Messungen zeigen erstmals, dass auch bei Invertebraten der Botenstoff cAMP, PKA-vermittelt, den IP<SUB>3</SUB>-Rezeptor/Ca<sup>2+-Kanal des ER für IP<SUB>3</SUB> sensitiviert.
Biogene Amine sind kleine organische Verbindungen, die sowohl bei Wirbeltieren als auch bei Wirbellosen als Neurotransmitter, Neuromodulatoren und/oder Neurohormone wirken können. Sie bilden eine bedeutende Gruppe von Botenstoffen und entfalten ihre Wirkungen über die Bindung an eine bestimmte Klasse von Rezeptorproteinen, die als G-Protein-gekoppelte Rezeptoren bezeichnet werden. Bei Insekten gehören zur Substanzklasse der biogenen Amine die Botenstoffe Dopamin, Tyramin, Octopamin, Serotonin und Histamin. Neben vielen anderen Wirkung ist z.B. gezeigt worden, daß einige dieser biogenen Amine bei der Honigbiene (Apis mellifera) die Geschmacksempfindlichkeit für Zuckerwasser-Reize modulieren können. Ich habe verschiedene Aspekte der aminergen Signaltransduktion an den „Modellorganismen“ Honigbiene und Amerikanische Großschabe (Periplaneta americana) untersucht. Aus der Honigbiene, einem „Modellorganismus“ für das Studium von Lern- und Gedächtnisvorgängen, wurden zwei Dopamin-Rezeptoren, ein Tyramin-Rezeptor, ein Octopamin-Rezeptor und ein Serotonin-Rezeptor charakterisiert. Die Rezeptoren wurden in kultivierten Säugerzellen exprimiert, um ihre pharmakologischen und funktionellen Eigenschaften (Kopplung an intrazelluläre Botenstoffwege) zu analysieren. Weiterhin wurde mit Hilfe verschiedener Techniken (RT-PCR, Northern-Blotting, in situ-Hybridisierung) untersucht, wo und wann während der Entwicklung die entsprechenden Rezeptor-mRNAs im Gehirn der Honigbiene exprimiert werden. Als Modellobjekt zur Untersuchung der zellulären Wirkungen biogener Amine wurden die Speicheldrüsen der Amerikanischen Großschabe genutzt. An isolierten Speicheldrüsen läßt sich sowohl mit Dopamin als auch mit Serotonin Speichelproduktion auslösen, wobei Speichelarten unterschiedlicher Zusammensetzung gebildet werden. Dopamin induziert die Bildung eines völlig proteinfreien, wäßrigen Speichels. Serotonin bewirkt die Sekretion eines proteinhaltigen Speichels. Die Serotonin-induzierte Proteinsekretion wird durch eine Erhöhung der Konzentration des intrazellulären Botenstoffs cAMP vermittelt. Es wurden die pharmakologischen Eigenschaften der Dopamin-Rezeptoren der Schaben-Speicheldrüsen untersucht sowie mit der molekularen Charakterisierung putativer aminerger Rezeptoren der Schabe begonnen. Weiterhin habe ich das ebony-Gen der Schabe charakterisiert. Dieses Gen kodiert für ein Enzym, das wahrscheinlich bei der Schabe (wie bei anderen Insekten) an der Inaktivierung biogener Amine beteiligt ist und im Gehirn und in den Speicheldrüsen der Schabe exprimiert wird.
In der vorliegenden Arbeit habe ich wichtige Teilmechanismen der Erregungs-Sekretionskopplung in der Speicheldrüse der Schabe Periplaneta americana (L.) untersucht. Die Speicheldrüse ist von dopaminergen und serotonergen Fasern innerviert (Baumann et al., 2002). Beide Transmitter stimulieren eine unterschiedliche Reaktion der Drüse: Dopamin (DA) stimuliert die P-Zellen der Acini und die Ausführgangzellen, während Serotonin (5-HT) die P- und C-Zellen der Acini stimuliert, nicht jedoch die Ausführgangzellen. Der Endspeichel ist nach einer DA-Stimulierung proteinfrei. Dagegen enthält er nach einer 5-HT-Stimulierung Proteine, die von den C-Zellen sezerniert werden (Just & Walz, 1996). Im ersten Teil meiner Arbeit habe ich mittels Kapillarelektrophoretischer Analyse (CE-Analyse) die Elektrolytkonzentrationen im Endspeichel untersucht sowie die Raten der Flüssigkeitssekretion gemessen. Damit wollte ich klären, welche Transporter an der Sekretion des Primärspeichels und an dessen Modifikation beteiligt sind. Ausserdem wollte ich die Rolle der transportaktiven Epithelzellen der Ausführgänge für die Modifikation des Primärspeichels untersuchen. Dafür habe ich einen Vergleich der Elektrolytkonzentrationen im DA- und 5-HT-stimulierten Endspeichel durchgeführt. Der Elektrolytgehalt des DA- und 5-HT-stimulierten Endspeichels unterscheidet sich nicht signifikant voneinander. Er ist nach beiden Stimulierungen hypoosmotisch zum verwendeten Ringer. Die Ausführgangzellen werden durch DA stimuliert und modifizieren den Primärspeichel durch eine netto-Ionenreabsorption. Meine Versuche zeigen jedoch, dass auch die während einer 5-HT-Stimulierung der Drüse unstimulierten Ausführgangzellen den Primärspeichel modifizieren. In einer nachfolgenden Versuchsreihe habe ich den Einfluss von Ouabain, einem Hemmstoff der Na+-K+-ATPase, und Bumetanid, einem Hemmstoff des NKCC, auf die Raten der Flüssigkeitssekretion sowie den Elektrolytgehalt des Endspeichels untersucht. Ich habe gefunden, dass die Aktivität der Na+-K+-ATPase wichtig für die Modifikation des DA-stimulierten Primärspeichels ist. Im Gegensatz dazu ist sie für die Modifikation des 5-HT-stimulierten Primärspeichels nicht von Bedeutung. Bezüglich der Flüssigkeitssekretion habe ich keinen Einfluss der Na+-K+-ATPase-Aktivität auf die DA-stimulierten Sekretionsraten gefunden, dagegen ist die 5-HT-stimulierte Sekretionsrate in Anwesenheit von Ouabain gesteigert. Die Aktivität des NKCC ist für beide sekretorische Prozesse, die Ionen- und die Flüssigkeitssekretion, wichtig. Eine Hemmung des NKCC bewirkt eine signifikante Verringerung der Raten der Flüssigkeitssekretion nach DA- und 5-HT-Stimulierung sowie in beiden Fällen einen signifikanten Abfall der Ionenkonzentrationen im Endspeichel. Im zweiten Teil meiner Arbeit habe ich versucht, Änderungen der intrazellulären Ionenkonzentrationen in den Acinuszellen während einer DA- oder 5-HT-Stimulierung zu messen. Diese Experimente sollten mit der Methode des "ratiometric imaging" durchgeführt werden. Messungen mit dem Ca2+-sensitiven Fluoreszenzfarbstoff Fura-2 zeigten keinen globalen Anstieg in der intrazellulären Ca2+-Konzentration der P-Zellen. Aufgrund von Problemen mit einer schlechten Beladung der Zellen, einer starken und sich während der Stimulierung ändernden Autofluoreszenz der Zellen sowie Änderungen im Zellvolumen wurden keine Messungen mit Na+- und K+-sensitiven Fluoreszenzfarbstoffen durchgeführt. Im dritten Teil dieser Arbeit habe ich die intrazellulären Signalwege untersucht, die zwischen einer 5-HT-Stimulierung der Drüse und der Proteinsekretion vermitteln. Dazu wurde der Proteingehalt im Endspeichel biochemisch mittels eines modifizierten Bradford Assay gemessen. Eine erstellte Dosis-Wirkungskurve zeigt, dass die Rate der Proteinsekretion von der zur Stimulierung verwendeten 5-HT-Konzentration abhängt. In einer Serie von Experimenten habe ich die intrazellulären Konzentrationen von Ca2+, cAMP und / oder cGMP erhöht und anschließend den Proteingehalt im Endspeichel gemessen. Ein Anstieg der intrazellulären Ca2+-Konzentration aktiviert nur eine geringe Rate der Proteinsekretion. Dagegen kann die Steigerung der intrazellulären cAMP-Konzentration eine stärkere Proteinsekretion aktivieren, die sich nicht signifikant von der nach 5-HT-Stimulierung unterscheidet. Die cAMP-stimulierte Proteinsekretion kann durch gleichzeitige Erhöhung der intrazellulären Ca2+-Konzentration weiter gesteigert werden. Dagegen aktivierte eine Erhöhung der intrazellulären cGMP-Konzentration die Proteinsekretion nicht. Aufgrund dieser Ergebnisse postuliere ich die Existenz eines die Adenylatcyclase aktivierenden 5-HT-Rezeptors in der Basolateralmembran der C-Zellen.
