In its Burmych and Others v. Ukraine judgment of October 2017 the European Court of Human Rights dismissed more than 12,000 applications due to the fact that they were not only repetitive in nature, but also mutatis mutandis identical to applications covered by a previous pilot judgment rendered against Ukraine. This raises fundamental issues as to the role of the Court within the human rights protection system established by the European Convention on Human Rights, as well as those concerning the interrelationship between the Court and the Committee of Ministers.
Bridging metabolomics with plant phenotypic responses is challenging. Multivariate analyses account for the existing dependencies among metabolites, and regression models in particular capture such dependencies in search for association with a given trait. However, special care should be undertaken with metabolomics data. Here we propose a modeling workflow that considers all caveats imposed by such large data sets.
The spatial control of DNA and of self-assembled DNA constructs is a prerequisite for the preparation of DNA-based nanostructures and microstructures and a useful tool for studies on single DNA molecules. Here we describe a protocol for the accumulation of dissolved lambda-DNA molecules between planar microelectrodes by the action of inhomogeneous radiofrequency electric fields. The resulting AC electrokinetic forces stretch the DNA molecules and align them parallel to the electric field. The electrode preparation from off-the-shelf electronic components is explained, and a detailed description of the electronic setup is given. The experimental procedure is controlled in real-time by fluorescence microscopy.
UNC119 and PDE delta are lipid-binding proteins and are thought to form diffusible complexes with transducin-alpha and prenylated OS proteins, respectively, to mediate their trafficking to photoreceptor outer segments. Here, we investigate mechanisms of trafficking which are controlled by Arf-like protein 3 (Arl3), a small GTPase. The activity of ARL3 is regulated by a GEF (ARL13b) and a GAP (RP2). In a mouse germline knockout of RP2, ARL3-GTP is abundant as its intrinsic GTPase activity is extremely low. High levels of ARL3-GTP impair binding and trafficking of cargo to the outer segment. Germline knockout of ARL3 is embryonically lethal generating a syndromic ciliopathy-like phenotype. Retina-and rod-specific knockout of ARL3 allow to determine the precise mechanisms leading to photoreceptor degeneration. The knockouts reveal binary functions of ARL3-GTP as a key molecule in late-stage photoreceptor ciliogenesis and cargo displacement factor.
Affect Disposition(ing)
(2018)
The “affective turn” has been primarily concerned not with what affect is, but what it does. This article focuses on yet another shift towards how affect gets organized, i.e., how it is produced, classified, and controlled. It proposes a genealogical as well as a critical approach to the organization of affect and distinguishes between several “affect disposition(ing) regimes”—meaning paradigms of how to interpret and manage affects, for e.g., encoding them as byproducts of demonic possession, judging them in reference to a moralistic framework, or subsuming them under an industrial regime. Bernard Stiegler’s concept of psychopower will be engaged at one point and expanded to include social media and affective technologies, especially Affective Computing. Finally, the industrialization and cybernetization of affect will be contrasted with poststructuralist interpretations of affects as events.
The Limits of Buyer Power
(2018)
This paper studies the behavior of buyers confronting an incumbent monopolist and a potential market entrant in a repeated trade situation. In the experiment, buyers have two possibilities to demand lower prices in future trade periods. First, they can withhold demand. Second, they can voluntarily pay a higher price to the entrant in order to encourage future re-entry. Both these forms of buyer behavior occur in the experiment. They are less frequent when the number of buyers is large as opposed to small. A control treatment tests to what extent such behavior can be attributed to strategic motives.
Background: Comprehension of non-canonical sentences is frequently characterised by chance level performance in people with aphasia (PWA). Chance level performance has been interpreted as guessing, but online data does not support this rendering. It is still not clear whether the incorrect sentence processing is guided by the compensatory strategies that PWA might employ to overcome linguistic difficulties.Aims: We aim to study to what extent people with non-fluent aphasia are aware of their sentence comprehension deficits. Methods & Procedures: This study combined offline and online data to investigate the effect of word order and error-awareness on sentence comprehension in a group of PWA and non-brain damaged (NBD) participants. The offline tasks involved auditory sentence picture-matching immediately followed by a confidence rating (CR). Participants were asked to judge the perceived correctness of their previous answer. Online data consisted of eye-tracking.Outcomes & Results: Replicating previous findings, PWA had significantly worse comprehension of Theme-Agent order compared to Agent-Theme order sentences. Controls showed ceiling level sentence comprehension. CR was a poor predictor of response accuracy in PWA, but moderate-good in NBD. A total of 6.8% of judgements were classified as guessing by PWA. Post hoc gaze data analysis indicated that CR was a predictor of the fixation pattern during the presentation of the linguistic stimuli. Conclusions: Results suggest that PWA were mostly unaware of their sentence comprehension errors and did not consciously employ strategies to compensate for their difficulties.
For linear inverse problems Y = A mu + zeta, it is classical to recover the unknown signal mu by iterative regularization methods ((mu) over cap,(m) = 0,1, . . .) and halt at a data-dependent iteration tau using some stopping rule, typically based on a discrepancy principle, so that the weak (or prediction) squared-error parallel to A((mu) over cap (()(tau)) - mu)parallel to(2) is controlled. In the context of statistical estimation with stochastic noise zeta, we study oracle adaptation (that is, compared to the best possible stopping iteration) in strong squared- error E[parallel to((mu) over cap (()(tau)) - mu)parallel to(2)]. For a residual-based stopping rule oracle adaptation bounds are established for general spectral regularization methods. The proofs use bias and variance transfer techniques from weak prediction error to strong L-2-error, as well as convexity arguments and concentration bounds for the stochastic part. Adaptive early stopping for the Landweber method is studied in further detail and illustrated numerically.
This study examined a theoretical model hypothesizing that reading strategies mediate the effects of intrinsic reading motivation, reading fluency, and vocabulary knowledge on reading comprehension. Using path analytic methods, we tested the direct and indirect effects specified in the hypothesized model in a sample of 1105 fifth-graders. In addition to standardized tests and questionnaires, we administered a performance test to assess students' proficiency in the application of three reading strategies. The overall fit of the model to the data was good. Both cognitive (fluency and vocabulary) and motivational (intrinsic reading motivation) variables had an indirect effect on reading comprehension through their influence on reading strategies. Reading strategies had a unique effect on reading comprehension and partially mediated the effects that cognitive and motivational variables had on fifth-graders' reading achievements.
Bryophytes constitute an important and permanent component of the grassland flora and diversity in Europe. As most bryophyte species are sensitive to habitat change, their diversity is likely to decline following land-use intensification. Most previous studies on bryophyte diversity focused on specific habitats of high bryophyte diversity, such as bogs, montane grasslands, or calcareous dry grasslands. In contrast, mesic grasslands are rarely studied, although they are the most common grassland habitat in Europe. They are secondary vegetation, maintained by agricultural use and thus, are influenced by different forms of land use. We studied bryophyte species richness in three regions in Germany, in 707 plots of 16 m(2) representing different land-use types and environmental conditions. Our study is one of the few to inspect the relationships between bryophyte richness and land use across contrasting regions and using a high number of replicates. Among the managed grasslands, pastures harboured 2.5 times more bryophyte species than meadows and mown pastures. Similarly, bryophyte cover was about twice as high in fallows and pastures than in meadows and mown pastures. Among the pastures, bryophyte species richness was about three times higher in sheep grazed plots than in the ones grazed by cattle or horses. In general, bryophyte species richness and cover was more than 50% lower in fertilized than in unfertilized plots. Moreover, the amount of suitable substrates was linked to bryophyte diversity. Species richness of bryophytes growing on stones increased with stone cover, and the one of bryophytes growing on bark and deadwood increased with larger values of woody plant species and deadwood cover. Our findings highlight the importance of low-intensity land use and high structural heterogeneity for bryophyte conservation. They also caution against an intensification of traditionally managed pastures. In the light of our results, we recommend to maintain low-intensity sheep grazing on sites with low productivity, such as slopes on shallow soils.
Swimming is of vital importance for aquatic organisms because it determines several aspects of fitness, such as encounter rates with food, predators, and mates. Generally, rotifer swimming speed is measured by manual tracking of the swimming paths filmed in videos. Recently, an open-source package has been developed that integrates different open-source software and allows direct processing and analysis of the swimming paths of moving organisms. Here, we filmed groups of females and males of Keratella cochlearis separately and in a mixed experimental setup. We extracted movement trajectories and swimming speeds and applied the classification method random forest to assign sex to individuals of the mixed setup. Finally, we used advanced statistical methods of movement ecology, namely a hidden Markov model, to investigate swimming states of females and males. When not discriminating swimming states, females swam faster than males, while when discriminating states males swam faster. Specifically, females and males showed two main states of movement with many individuals switching between states resulting in four modes of swimming. We suggest that switching between states is related to predator avoidance. Males of K. cochlearis especially exhibited switching between turning in a restricted area and swimming over longer distances. No mating or other male-female interactions were observed. Our study elucidates the steps necessary for automatic analysis of rotifer trajectories with open-source software. Application of sophisticated software and analytical models will broaden our understanding of zooplankton ecology from the individual to the population level.
Intensive rice-wheat cultivation cycle in Northern belt of India in general and in the State of Punjab in particular results in large volumes of straw and other post-harvest residue annually. The agricultural area, bordering the districts of Nawanshahr and Hoshiarpur, is popularly known as the seleniferous belt of India. The agri-residues, generated in seleniferous region of this state, are observed to contain significantly high concentration of selenium (Se). The present study was aimed to evaluate the Se uptake by different mushroom species: Pleurotus sajorcaju, Pleurotus ostreatus, Pleurotus citrinopileatus, Agaricus bisporus, and Volvariella volvacea, cultivated on Se-rich wheat and paddy straw from the seleniferous region. Wheat (Pleurotus species and A. bisporus) and paddy straw (V. volvacea) was inoculated with the mycelium spawn and left for 7-20 days, depending on the species, to grow. Control mushrooms were grown analogously using the agricultural residues from non-seleniferous area of the State of Punjab. All fruiting bodies were collected and analyzed in triplicate. Se was quantified using inductively coupled plasma sector field mass spectrometry. The Se accumulation was high in all species under study, being the highest in A. bisporus (1396 mu g/g vs. 46.8 mu g/g in controls - dry weight) and V. volvacea (231 mu g/g vs. 3.77 mu g/g - dry weight). The observed biological efficiency and total yield for all mushroom species showed good and unaltered productivity in Se-rich conditions, if compared to the controls. The Se-rich mushrooms can be prospective Se-supplements sourcing and biofortified foods, providing readily bioavailable and accessible Se for the diets deficient of this biologically essential element.
Struggling over crisis
(2018)
If you put two economists in a room, you get two opinions, unless one of them is Lord Keynes, in which case you get three opinions.” Following the premise of this quotation attributed to Winston Churchill, varying perceptions of the European crisis by academic economists and their structural homology to economists’ positions in the field of economics are examined. The dataset analysed using specific multiple correspondence analysis (MCA) and hierarchical agglomerative clustering (HAC) comprises information on the careers of 480 German-speaking economists and on statements they made concerning crisis-related issues. It can be shown that the main structural differences in the composition and amount of scientific and academic capital held by economists as well as their age and degree of transnationalisation are linked to how they see the crisis: as a national sovereign debt crisis, as a European banking crisis, or as a crisis of European integration and institutions.
A geological transect across the suture separating northwestern South America from the Panama Arc helps document the provenance and thermal history of both crustal domains and the suture zone. During middle Miocene, strata were being accumulated over the suture zone between the Panama Arc and the continental margin. Integrated provenance analyses of those middle Miocene strata show the presence of mixed sources that includes material derived from the two major crustal domains: the old northwestern South American orogens and the younger Panama Arc. Coeval moderately rapid exhumation of Upper Cretaceous to Paleogene sediments forming the reference continental margin is suggested from our inverse thermal modeling. Strata within the suture zone are intruded by similar to 12 Ma magmatic arc-related plutons, marking the transition from a collisional orogen to a subduction-related one. Renewed late Miocene to Pliocene acceleration of the exhumation rates is the consequence of a second tectonic pulse, which is likely to be triggered by the onset of a flat-slab subduction of the Nazca plate underneath the northernmost Andes of Colombia, suggesting that late Miocene to Pliocene orogeny in the Northern Andes is controlled by at least two different tectonic mechanisms.
Breaking the Barrier
(2018)
Topical administration permits targeted, sustained delivery of therapeutics to human skin. Delivery to the skin, however, is typically limited to lipophilic molecules with molecular weight of < 500 Da, capable of crossing the stratum corneum. Nevertheless, there are indications protein delivery may be possible in barrier deficient skin, a condition found in several inflammatory skin diseases such as psoriasis, using novel nanocarrier systems. Methods: Water in water thermo-nanoprecipitation; dynamic light scattering; zeta potential measurement; nanoparticle tracking analysis; atomic force microscopy; cryogenic transmission electron microscopy; UV absorption; centrifugal separation membranes; bicinchoninic acid assay; circular dichroism; TNF alpha binding ELISA; inflammatory skin equivalent construction; human skin biopsies; immunohistochemistry; fluorescence microscopy; western blot; monocyte derived Langerhans cells; ELISA Results: Here, we report the novel synthesis of thermoresponsive nanogels (tNG) and the stable encapsulation of the anti-TNFa fusion protein etanercept (ETR) (similar to 150 kDa) without alteration to its structure, as well as temperature triggered release from the tNGs. Novel tNG synthesis without the use of organic solvents was conducted, permitting in situ encapsulation of protein during assembly, something that holds great promise for easy manufacture and storage. Topical application of ETR loaded tNGs to inflammatory skin equivalents or tape striped human skin resulted in efficient ETR delivery throughout the SC and into the viable epidermis that correlated with clear anti-inflammatory effects. Notably, effective ETR delivery depended on temperature triggered release following topical application. Conclusion: Together these results indicate tNGs hold promise as a biocompatible and easy to manufacture vehicle for stable protein encapsulation and topical delivery into barrier-deficient skin.
That’s not quite it

We present a novel empirical study on German directly comparing the exhaustivity inference in es-clefts to exhaustivity inferences in definite pseudoclefts, exclusives, and plain intonational focus constructions. We employ mouse-driven verification/falsification tasks in an incremental information-retrieval paradigm across two experiments in order to assess the strength of exhaustivity in the four sentence types. The results are compatible with a parallel analysis of clefts and definite pseudoclefts, in line with previous claims in the literature (Percus 1997, Buring & Kriz 2013). In striking contrast with such proposals, in which the exhaustivity inference is conventionally coded in the cleft-structure in terms of maximality/homogeneity, our study found that the exhaustivity inference is not systematic or robust in es-clefts nor in definite pseudoclefts: Whereas some speakers treat both constructions as exhaustive, others treat both constructions as non-exhaustive. In order to account for this unexpected finding, we argue that the exhaustivity inference in both clefts and definite pseudoclefts-specifically those with the compound definite derjenige - is pragmatically derived from the anaphoric existence presupposition that is common to both constructions.
