Prostaglandin F(2alpha) (PGF(2alpha)), modulates hepatocyte functions via a heptahelical G(q)-coupled PGF(2alpha)-receptor (FP-R) which in liver is expressed exclusively in hepatocytes. The aim of the present study was to isolate the 5'-flanking region of the rat FP-R gene and to elucidate its basal and IL-6-modulated transcription control function in rat hepatocytes. The 5'-non-translated region of the rat hepatocyte FP-R mRNA differed from the corresponding region in rat fetal astrocyte or corpus luteum. It was encoded by exons 1a and 2 which were separated by a 1. 4 kb intron containing the exons 1b and 1c coding for the 5'-untranslated region of rat fetal astrocyte and corpus luteum FP-R mRNA, respectively. The transcription initiation site in hepatocytes was localized 263 bp upstream of the start ATG by 5'-RACE. A DNA-fragment covering the 5'-flanking region of the rFP-R gene from - 1 of the transcription initiation site to -2590 bp was cloned and sequenced. Its 3'-two thirds had a 65% sequence identity to the mouse FP-R promoter however no homology to the bovine FP-R promoter. In the overlapping sequence most of the putative transcription factor binding sites were conserved between mouse and rat. The rat promoter contained no classical TATA- or CAAT-boxes but putative binding sites for the transcription factors C/EBP, GATA-1, HNF-1, HNF-3beta, SP-1, and USF. Luciferase reporter gene constructs containing portions of the 5'-flanking region were transfected into rat hepatocytes. Luciferase expression ranked -181 >/= -608 < -1418 > -1821 >/= -2590. The strongest transcriptional activity was conferred by the region between -608 and -1418 containing a cluster of potential HNF-1 and HNF-3beta binding sites that might allow the exclusive expression of FP-R mRNA in hepatocytes. The amount of FP-R mRNA and the luciferase expression under control of the -2590 promoter fragment were reduced by IL-6 in hepatocytes. Copyright 2000 Academic Press.
Prostanoids, that are released from nonparenchymal liver cells in response to proinflammatory stimuli, are involved in the regulation of hepatic functions during inflammation. They exert their effects on their target cells via heptahelical receptors in the plasma membrane. For the 5 prostanoids prostaglandin E(2) (PGE(2)), prostaglandin F(2alpha), prostaglandin D(2) (PGD(2)), prostacyclin, and thromboxane A(2) there exist 8 receptors that are coupled to different heterotrimeric G proteins. These receptors are expressed differentially in the 4 principal liver cell types, i.e., hepatocytes, Kupffer cells, sinusoidal endothelial cells, and hepatic stellate cells. It was intriguing, that the messenger RNA (mRNA) of none of the G(s)-coupled prostanoid receptors (DP-R, EP2-R, EP4-R, and IP-R) that can attenuate the inflammatory reaction were present in hepatocytes. The current study shows that the expression of the G(s)-coupled prostanoid receptors EP2-R, EP4-R, and DP-R, but not the IP-R, was efficiently and rapidly up-regulated by treatment of hepatocytes in vitro or rats in vivo with the key acute phase cytokine interleukin 6 (IL-6). In IL-6-treated hepatocytes PGE(2) in turn attenuated the IL-6-induced alpha(2)-macroglobulin formation via a cyclic adenosine monophosphate (cAMP)- dependent signal chain. The data indicate that an IL-6-mediated induction of the previously not expressed EP2-R and EP4- R on hepatocytes might establish a prostanoid-mediated feedback inhibition loop for the attenuation of the acute phase response.
Eight heptahelical receptors have been characterized for prostaglandin (PG) D(2), PGE(2), PGF(2alpha), prostacyclin and thromboxane A(2). They share a sequence identity of 40%. All of them have potential N-glycosylation sites. The current study analysed the role of the two N-glycosylation sites in the rat EP3beta-subtype PGE(2) receptor for protein folding and sorting. The N-glycosylation consensus sequences were eliminated by site-directed mutagenesis and receptors expressed in HEK-293 cells. Both potential N-glycosylation sites were used. Their joint elimination resulted in the synthesis of a receptor protein with full binding competence, biological activity and no reduction of affinity; however, the half-life of the non-glycosylated receptor was slightly reduced. Ligand binding to intact stably transfected cells and confocal laser microscopic immunocytochemistry showed that the glycosylated receptor was correctly inserted into the plasma membrane to a much larger extent than the non-glycosylated receptor, which tended to accumulate in the perinuclear zone of the endoplasmic reticulum. Inhibition of N-glycosylation with tunicamycin resulted in a similar perinuclear distribution of the wild-type receptor. Therefore, glycosylation of the EP3beta receptor seems not to be necessary for correct folding of the receptor protein but for the efficient transport of the receptor protein to the plasma membrane. This contrasts with a previous finding which described a reduction of the affinity for PGE(2) of the EP3alpha receptor by elimination of the distal glycosylation site when the receptor protein was expressed in insect cells.
Der Autor diskutiert in seinem Aufsatz kritisch den Friedensvertrag von Lomé, der am 7. Juli 1999 offiziell den bewaffneten Konflikt in Sierra Leone beendete. Nach einer kurzen Zusammenfassung der allgemeinen Regelungen des Vertrags stellt der Autor die in Artikel 9 des Abkommens vorgesehene Generalamnestie den bindenden Grundsätzen des internationalen Rechts gegenüber. Internationale Verbrechen, wie Völkermord, Kriegsverbrechen oder Folterung sind als Verstoß gegen ius cogens-Normen von allen Staaten zu verfolgen. Nach der Erörterung der betreffenden Konventionen, internationalen Abkommen und Fallentscheidungen des IGH, die diesen Grundsatz festschreiben, beschreibt er den - Friedensprozessen inhärenten - Konflikt, ein Gleichgewicht zwischen notwendiger Versöhnung und strafrechtlicher Verfolgung zu finden. Bei der Betrachtung des Fallrechts schließt Phenyo neuere Entscheidungen ein, wie die des britischen House of Lords im Fall Pinochet, die sowohl nationalen wie internationalen Gerichten das Recht auf Strafverfolgung internationaler Verbrechen zugestand. Stellvertretend für die weite Kritik der Generalamnestie des Lomé-Abkommens zitiert der Autor den VN-Generalsekretär Kofi Annan, der die Generalamnestie als unvereinbar mit der Tätigkeit und Aufgabe der internationalen Straftribunale in Den Haag und Arusha sowie des zukünftigen Internationalen Strafgerichtshofes ansieht. Phenyo schließt sich mit seiner kurzen Analyse des Friedensabkommens der kritischen Haltung Annans an und sieht nur eine geringe Möglichkeit für die Durchsetzung der fraglichen Amnestie, deren Gültigkeit durch die wiederaufgeflammten Kämpfe in Sierra Leone auch faktisch in Frage gestellt worden sind. Reversible binding of the starch-related R1 protein to the surface of transitory starch granules

We investigate numerically the appearance of heteroclinic behavior in a three-dimensional, buoyancy-driven fluid layer with stress-free top and bottom boundaries, a square horizontal periodicity with a small aspect ratio, and rotation at low to moderate rates about a vertical axis. The Prandtl number is 6.8. If the rotation is not too slow, the skewed-varicose instability leads from stationary rolls to a stationary mixed-mode solution, which in turn loses stability to a heteroclinic cycle formed by unstable roll states and connections between them. The unstable eigenvectors of these roll states are also of the skewed-varicose or mixed-mode type and in some parameter regions skewed-varicose like shearing oscillations as well as square patterns are involved in the cycle. Always present weak noise leads to irregular horizontal translations of the convection pattern and makes the dynamics chaotic, which is verified by calculating Lyapunov exponents. In the nonrotating case the primary rolls lose, depending on the aspect ratio, stability to traveling waves or a stationary square pattern. We also study the symmetries of the solutions at the intermittent fixed points in the heteroclinic cycle.
A numerical bifurcation analysis of the electrically driven plane sheet pinch is presented. The electrical conductivity varies across the sheet such as to allow instability of the quiescent basic state at some critical Hartmann number. The most unstable perturbation is the two-dimensional tearing mode. Restricting the whole problem to two spatial dimensions, this mode is followed up to a time-asymptotic steady state, which proves to be sensitive to three- dimensional perturbations even close to the point where the primary instability sets in. A comprehensive three- dimensional stability analysis of the two-dimensional steady tearing-mode state is performed by varying parameters of the sheet pinch. The instability with respect to three-dimensional perturbations is suppressed by a sufficiently strong magnetic field in the invariant direction of the equilibrium. For a special choice of the system parameters, the unstably perturbed state is followed up in its nonlinear evolution and is found to approach a three-dimensional steady state.

If high temperatures prohibit solid walls, a conducting fluid can be held together by the action of an electric current passing through it with the pressure gradients being balanced by the Lorentz force. The resultant configuration is known as a pinch. In this paper we report on studies of the pinch in the geometry of a plane sheet.

We compute the shift of the critical temperature Tc with respect to the ideal case for a weakly interacting uniform Bose gas. We work in the framework of the canonical ensemble, extending the criterion of condensation provided by the canonical particle counting statistics for the zero-momentum state of the uniform ideal gas. The perturbative solution of the crossover equation to lowest order in power of the scattering length yields (Tc - Tc0)/Tc0=-0,93ap 1/3, where Tc0 is the transition temperature of the corresponding ideal Bose gas , a is the scattering length, and p is the particle number density. This is at vaiance with the standard grand canonical prediction of a null shift of the critical temperature in the lowest perturbative order. The non-equevalence of statistical ensemble for the ideal Bose gas is thus confirm (at the lowestperturbative level) also in the presence of interactions.
We consider entanglement-assisted remote quantum state manipulation of bipartite mixed states. Several aspects are addressed: we present a class of mixed states of rank two that can be transformed into another class of mixed states under entanglement-assisted local operations with classical communication, but for which such a transformation is impossible without assistance. Furthermore, we demonstrate enhancement of the efficiency of purification protocols with the help of entanglement-assisted operations. Finally, transformations from one mixed state to mixed target states which are sufficiently close to the source state are contrasted with similar transformations in the pure-state case.
We analyze the multi-time correlations of a laser-induced Bose Einstein condensate. We use quantum stochastic methods to obtain under certain circumstances a Fokker-Planck equation which describes the phase-difussion process, and obtain an analytical expression of the two-time correlations. We perform also quantum Monte Carlo numerical simulations of the correlations, which are in good agreement with the predicted analytical results.
Quantum games

In these lecture notes we investigate the implications of the identification of strategies with quantum operations in game theory beyond the results presented in [J. Eisert, M. Wilkens, and M. Lewenstein, Phys. Rev. Lett. 83, 3077 (1999)]. After introducing a general framework, we study quantum games with a classical analogue in order to flesh out the peculiarities of game theoretical settings in the quantum domain. Special emphasis is given to a detailed investigation of different sets of quantum strategies.
We establish a quantitative connection between the amount of lost classical information about a quantum state and the concomitant loss of entanglement. Using menthods that have been developed for the optimal purification of miced states, we find a class of miced states with known distillable entanglement. These results can be used to determine the quantum capacity of a quantum channel which randomizes the order of transmitted signals.
We discuss heating and decoherencw in traps fpr ions and neutral paricles close to metallic surfaces. We focus on simple trap geometries and compute noise spectra of thermally excited electromagnetic fields. If the trap is located in the near field of the substrate, the field fluctuations are largely increased compared to the level of the blackbody field, leading to much shorter coherence and life times of the trapped atoms. The correspinding time constants are computed for ion traps and magnetic traps. Analytical estimates for the size dependence of the noise spectrum are given. We finally discuss prospects for the coherent transport of matter waves in integrated surface waveguides.
We analyze the spatial coherence of the electromagnetic field emitted by a half-space at temperature T close to the interface. An asymptotic analysis allows to identify three different contributions to the cross-spectral density tensor in the near-field regime. It is shown that the coherence length can be either much larger or much shorter than the wavelength depending on the dominant contribution.

Drosophila melanogaster photoreceptors are highly polarized cells and their plasma membrane is organized into distinct domains. Zonula adherens junctions separate a smooth peripheral surface, the equivalent of the basolateral surface in other epithelial cells, from the central surface (cong apical surface). The latter consists of the microvillar rhabdomere and the juxtarhabdomeric domain, a nonmicrovillar area between the rhabdomere and the zonulae adherens. The distribution of Na/K-ATPase over these domains was examined by immunocytochemical, developmental, and genetic approaches. Immunofluorescence and immunogold labeling of adult compound eyes reveal that the distribution of Na/ K-ATPase is concentrated at the peripheral surface in the photoreceptors R1-R6, but extends over the juxtarhabdomeric domain to the rhabdomere in the photoreceptors R7/R8. Developmental analysis demonstrates further that Na/K-ATPase is localized over the entire plasma membrane in all photoreceptors in early pupal eyes. Redistribution of Na/K-ATPase in R1- R6 occurs at about 78% of pupal life, coinciding with the onset of Rh1-rhodopsin expression on the central surface of these cells. Despite the essential role of Rh1 in structural development and intracellular trafficking, Rh1 mutations do not affect the distribution of Na/K-ATPase. These results suggest that Na/K-ATPase and rhodopsin are involved in distinct intracellular localization mechanisms, which are maintained independent of each other.
Selenides of the type R1Se-EMe3 (E = Si, Ge, Sn, Pb) react with xenon difluoride by cleavage of the Se-E bond to yield the R1Se-F intermediate and the fluorides Me3E-F, whereas the Se-C bond in PhSe-tBu (E = C) is stable against XeF2. The presence of R1Se-F intermediates is confirmed by addition to acetylenes (4-octyne, 3-hexyne). Thus, the fluoroselenenylation of acetylenes gives fluoro(organylseleno)olefins in preparative yields. In the cases of E = Si, Ge, Sn, and Pb, aryl and n-alkyl groups are suitable as the substituent R1. The X-ray crystal structural analysis of (E)-3- (p-carboxyphenylseleno)-4-fluorohex-3-ene - the first example of an uncharged fluoroselenoolefin synthesized from p- EtO2C-C6H4-Se-SnMe3, XeF2, and 3-hexyne followed by an ester hydrolysis - shows that the addition of the selenenylfluoride intermediate to the acetylene proceeds via a trans-addition, as is known for the R2Se2-XeF2 reagents.
Long-term impacts of livestock herbivory on herbaceous and woody vegetation in semiarid savannas

Water column mixing is known to have a decisive impact on plankton communities. The underlying mechanisms depend on the size and depth of the water body, the nutrient status, and the plankton community structure and they are well understood for shallow polymictic and deep stratified lakes. Two consecutive mixing events of similar intensity under different levels of herbivory were performed in enclosures in a shallow, but periodically stratified, eutrophic lake, in order to investigate the effects of water column mixing on bacteria abundance, phytoplankton abundance and diversity, and rotifer abundance and fecundity. When herbivory by filter-feeding zooplankton was low, water column mixing provoking a substantial nutrient input into the euphotic zone, led to an intense net increase of bacteria and phytoplankton biomass. Phytoplankton diversity was lower in the mixed enclosures than in the undisturbed ones owing to the larger contribution of a few fast-growing species. After the second mixing event at high biomass of filter-feeding crustaceans, the increase of phytoplankton biomass was lower than after the first mixing, and diversity remained unchanged as the enhanced growth of small fast-growing was prevented by zooplankton grazing. Bacteria abundance did not increase after the second mixing, when cladoceran biomass was high. Rotifer fecundity indicated a transmission of the phytoplankton response to the next trophic level. Our results suggest that water column mixing in shallow eutrophic lakes with periodic stratification has a strong effect on the plankton community by enhanced nutrient availability rather than resuspension or reduced light availability. This fuels the basis of the classic and microbial food chain via enhanced phytoplankton and bacteria growth, but the effects on biomass may be dampened by high levels of herbivory.

Ökobilanzen können angewendet werden, um die Umweltwirkungen verschiedener Abfallbehandlungsvarianten zu vergleichen. Es zeigt sich dabei, dass die ökologischen Vorteile bzw. Nachteile einzelner Verfahren stark von den vorliegenden Rahmenbedingungen abhängen. Beispielhaft werden Berechnungsergebnisse vorgestellt, bei denen die mechanisch-biologische Abfallbehandlung mit der direkten Deponierung der Abfälle und der Verbrennung verglichen werden.
