The major aim of this thesis was to study the effect of nitrate on primary metabolism and in development of the model plant Arabidopsis thaliana. The present work has two separate topics. First, to investigate the GDH family, a small gene family at the interface between nitrogen and carbon metabolisms. Second, to investigate the mechanisms whereby nitrogen is regulating the transition to flowering time in Arabidopsis thaliana. To gain more insights into the regulation of primary metabolism by the functional characterization of the glutamate dehydrogenase (GDH) family, an enzyme putatively involved in the metabolism of amino acids and thus suggested to play different and essential roles in carbon and nitrogen metabolism in plants, knock out mutants and transgenic plants carrying RNA interference construct were generated and characterized. The effect of silencing GDH on carbon and nitrogen metabolisms was investigated, especially the level of carbohydrates and the amino acid pool were further analysed. It has been shown that GDH expression is regulated by light and/or sugar status therefore, phenotypic and metabolic analysis were developed in plants grown at different points of the diurnal rhythm and in response to an extended night period. In addition, we are interested in the effect of nutrient availability in the transition from vegetative growth to flowering and especially in nitrate as a metabolite that triggers widespread and coordinated changes in metabolism and development. Nutrient availability has a dramatic effect on flowering time, with a marked delay of flowering when nitrate is supplied (Stitt, 1999). The use of different mutants and transgenic plants impaired in flowering signalling pathways was crucial to evaluate the impact of different nitrate concentrations on flowering time and to better understand the interaction of nitrate-dependent signals with other main flowering signalling pathways. Plants were grown on glutamine as a constitutive source of nitrogen, and the nitrate supply varied. Low nitrate led to earlier flowering. The response to nitrate is accentuated in short days and in the CONSTANS deficient co2 mutant, whereas long days or overexpression of CONSTANS overrides the nitrate response. These results indicate that nitrates acts downstream of the known flowering signalling pathways for photoperiod, autonomy, vernalization and gibberellic acid. Global analyses of gene expression of two independent flowering systems, a light impaired mutant (co2tt4) and a constitutive over-expresser of the potent repressor of flowering (35S::FLC), were to be investigated under two different concentrations of nitrate in order to identify candidate genes that may be involved in the regulation of flowering time by nitrate.
Each organ of a multicellular organism is unique at the level of its tissues and cells. Furthermore, responses to environmental stimuli or developmental signals occur differentially at the single cell or tissue level. This underlines the necessity of precise investigation of the “building block of life” -the individual cell. Although recently large amount of data concerning different aspects of single cell performance was accumulated, our knowledge about development and differentiation of individual cell within specialized tissue are still far from being complete. To get more insight into processes that occur in certain individual cell during its development and differentiation changes in gene expression during life cycle of A. thaliana leaf hair cell (trichome) were explored in this work. After onset of trichome development this cell changes its cell cycle: it starts endoreduplication (a modified cell cycle in which DNA replication continues in the absence of mitosis and cytokinesis). This makes trichomes a suitable model for studying cell cycle regulation, regulation of cell development and differentiation. Cells of interest were sampled by puncturing them with glass microcapillaries. Each sample contained as few as ten single cells. At first time trichomes in initial stage of trichome development were investigated. To allow their sampling they were specifically labelled by green fluorescent protein (GFP). In total three cell types were explored: pavement cells, trichome initials and mature trichomes. Comparison of gene expression profiles of these cells allowed identification of the genes differentially expressed in subsequent stages of trichome development. Bioinformatic analysis of genes preferentially expressed in trichome initials showed their involvement in hormonal, metal, sulphur response and cell-cycle regulation. Expression pattern of three selected candidate genes, involved in hormonal response and early developmental processes was confirmed by independent method. Effects of mutations in these genes on both trichome and plant development as well as on plant metabolism were analysed. As an outcome of this work novel components in the sophisticated machinery of trichome development and cell cycle progression were identified. These factors could integrate hormone stimuli and network interactions between characterized and as yet unknown members of this machinery. I expect findings presented in this work to enhance and complement our current knowledge about cell cycle progression and trichome development, as well as about performance of the individual cell in general.
Nature has always served as a model for mimicking and inspiration to humans in their efforts to improve their life. Researchers have been inspired by nature to produce biomimetic materials with molecular recognition properties by design rather than evolution. Molecular imprinting is one way to prepare such materials. Such smart materials with new functionalities are at the forefront of the development of a relevant number of ongoing and perspective applications ranging from consumer to space industry. Molecularly imprinted polymers were developed by mimicking the natural enzymes or antibodies that serve as host for binding target molecules. These imprints were used as a recognition element to substitute natural biomolecules in biosensors. The concept behind molecular imprinting is to mold a material (with the desired chemical properties) around individual molecules. Upon removal of the molecular templates, one is left with regions in the molded material that fit the shape of the template molecules. Thus, molecular imprinting results in materials that can selectively bind to molecules of interest. Imprinted materials resulted in applications ranging from chemical separation to bioanalytics. In this work attempts were made particularly in the development of molecularly imprinted polymer based thermometric sensors. The main effort was focused towards the development of an covalently imprinted polymer that would be able to selectively bind fructosyl valine (Fru-Val), the N-terminal constituent of hemoglobin A1c ß-chains. Taking into account the known advantages of imprinted polymers, e.g. robustness, thermal and chemical stability, imprinted materials were successfully used as a recognition element in the sensor. One of the serious problems associated with the development of MIP sensors and which lies in the absence of a generic procedure for the transformation of the polymer-template binding event into a detectable signal has been addressed by developing the "thermometric" approach. In general the developed approach gives a new insight on MIP/Analyte interactions.
Functional analysis of selected DOF transcription factors in the model plant Arabidopsis thaliana

Transcription factors (TFs) are global regulators of gene expression playing essential roles in almost all biological processes, and are therefore of great scientific and biotechnological interest. This project focused on functional characterisation of three DNA-binding-with-one-zinc-finger (DOF) TFs from the genetic model plant Arabidopsis thaliana, namely OBP1, OBP2 and AtDOF4;2. These genes were selected due to severe growth phenotypes conferred upon their constitutive over-expression. To identify biological processes regulated by OBP1, OBP2 and AtDOF4;2 in detail molecular and physiological characterization of transgenic plants with modified levels of OBP1, OBP2 and AtDOF4;2 expression (constitutive and inducible over-expression, RNAi) was performed using both targeted and profiling technologies. Additionally expression patterns of studied TFs and their target genes were analyzed using promoter-GUS lines and publicly available microarray data. Finally selected target genes were confirmed by chromatin immuno-precipitation and electrophoretic-mobility shift assays. This combinatorial approach revealed distinct biological functions of OBP1, OBP2 and AtDOF4;2. Specifically OBP2 controls indole glucosinolate / auxin homeostasis by directly regulating the enzyme at the branch of these pathways; CYP83B1 (Skirycz et al., 2006). Glucosinolates are secondary compounds important for defence against herbivores and pathogens in the plants order Caparales (e.g. Arabidopsis, canola and broccoli) whilst auxin is an essential plant hormone. Hence OBP2 is important for both response to biotic stress and plant growth. Similarly to OBP2 also AtDOF4;2 is involved in the regulation of plant secondary metabolism and affects production of various phenylpropanoid compounds in a tissue and environmental specific manner. It was found that under certain stress conditions AtDOF4;2 negatively regulates flavonoid biosynthetic genes whilst in certain tissues it activates hydroxycinnamic acid production. It was hypothesized that this dual function is most likely related to specific interactions with other proteins; perhaps other TFs (Skirycz et al., 2007). Finally OBP1 regulates both cell proliferation and cell expansion. It was shown that OBP1 controls cell cycle activity by directly targeting the expression of core cell cycle genes (CYCD3;3 and KRP7), other TFs and components of the replication machinery. Evidence for OBP1 mediated activation of cell cycle during embryogenesis and germination will be presented. Additionally and independently on its effects on cell proliferation OBP1 negatively affects cell expansion via reduced expression of cell wall loosening enzymes. Summing up this work provides an important input into our knowledge on DOF TFs function. Future work will concentrate on establishing exact regulatory networks of OBP1, OBP2 and AtDOF4;2 and their possible biotechnological applications.
In semi-arid savannah ecosystems, the vegetation structure and composition, i.e. the architecture of trees, shrubs, grass tussocks and herbaceous plants, offer a great variety of habitats and niches to sustain animal diversity. In the last decades intensive human land use practises like livestock farming have altered the vegetation in savannah ecosystems worldwide. Extensive grazing leads to a reduction of the perennial and herbaceous vegetation cover, which results in an increased availability of bare soil. Both, the missing competition with perennial grasses and the increase of bare soils favour shrub on open ground and lead to area-wide shrub encroachment. As a consequence of the altered vegetation structure and composition, the structural diversity declines. It has been shown that with decreasing structural diversity animal diversity decline across a variety of taxa. Knowledge on the effects of overgrazing on reptiles, which are an important part of the ecosystem, are missing. Furthermore, the impact of habitat degradation on factors of a species population dynamic and life history, e.g., birth rate, survival rate, predation risk, space requirements or behavioural adaptations are poorly known. Therefore, I investigated the impact of overgrazing on the reptile community in the southern Kalahari. Secondly I analysed population dynamics and the behaviour of the Spotted Sand Lizard, Pedioplanis l. lineoocellata. All four chapters clearly demonstrate that habitat degradation caused by overgrazing had a severe negative impact upon (i) the reptile community as a whole and (ii) on population parameters of Pedioplanis l. lineoocellata. Chapter one showed a significant decline of regional reptile diversity and abundance in degraded habitats. In chapter two I demonstrated that P. lineoocellata moves more frequently, spends more time moving and covers larger distances in degraded than in non-degraded habitats. In addition, home range size of the lizard species increases in degraded habitats as shown by chapter three. Finally, chapter four showed the negative impacts of overgrazing on several population parameters of P. lineoocellata. Absolute population size of adult and juvenile lizards, survival rate and birth rate are significantly lower in degraded habitats. Furthermore, the predation risk was greatly increased in degraded habitats. A combination of a variety of aspects can explain the negative impact of habitat degradation on reptiles. First, reduced prey availability negatively affects survival rate, the birth rate and overall abundance. Second, the loss of perennial plant cover leads to a loss of niches and to a reduction of opportunities to thermoregulate. Furthermore, a loss of cover and is associated with increased predation risk. A major finding of my thesis is that the lizard P. lineoocellata can alter its foraging strategy. Species that are able to adapt and change behaviour, such as P. lineoocellata can effectively buffer against changes in their environment. Furthermore, perennial grass cover can be seen as a crucial ecological component of the vegetation in the semi-arid savannah system of the southern Kalahari. If perennial grass cover is reduced to a certain degree reptile diversity will decline and most other aspects of reptile life history will be negatively influenced. Savannah systems are characterised by a mixture of trees, shrubs and perennial grasses. These three vegetation components determine the composition and structure of the vegetation and accordingly influence the faunal diversity. Trees are viewed as keystone structures and focal points of animal activity for a variety of species. Trees supply animals with shelter, shade and food and act as safe sites, nesting sites, observation posts and foraging sites. Recent research demonstrates a positive influence of shrub patches on animal diversity. Moreover, it would seem that intermediate shrub cover can also sustain viable populations in savannah landscapes as has been demonstrated for small carnivores and rodent species. The influence of perennial grasses on faunal diversity did not receive the same attention as the influence of trees and shrubs. In my thesis I didn’t explicitly measure the direct effects of perennial grasses but my results strongly imply that it has an important role. If the perennial grass cover is significantly depleted my results suggest it will negatively influence reptile diversity and abundance and on several populations parameters of P. lineoocellata. Perennial grass cover is associated with the highest prey abundance, reptile diversity and reptile abundance. It provides reptiles both a refuge from predators and opportunities to optimise thermoregulation. The relevance of each of the three vegetation structural elements is different for each taxa and species. In conclusion, I can all three major vegetation structures in the savannah system are important for faunal diversity.
Homogene Immunoassays sind immunologische Testverfahren, bei deren Durchführung vollständig auf Separations- und Waschschritte verzichtet werden kann. Der Substrate Channeling Immunoassay beruht auf der Weitergabe eines Substrates in einem immunologischen Komplex aus zwei Enzymen. Das Produkt des ersten Enzyms dient dem zweiten Enzym als Substrat zur Generierung eines photometrisch nachweisbaren Produktes. Voraussetzung für diese Weitergabe ist die enge räumliche Nähe beider Enzyme. Diese Nähe wird durch eine Bindung zwischen Analyt und anti-Analyt Antikörper vermittelt. Ein solcher Substrate Channeling Immunoassay wurde unter Verwendung der Enzyme Glucoseoxidase und Peroxidase aufgebaut. Das so etablierte System war funktionstüchtig, jedoch blieb seine Sensitivität hinter der normaler, heterogener Immunoassays zurück. Die Grundlage eines Fluorescence Quenching Immunoassays ist der gegenseitige Ausschluß zweier Antikörper bei der Bindung eines Dihapten-Konjugates. Das Konjugat besteht dabei aus dem Analyten und einem Fluorophor. Die beiden um die Konjugatbindung konkurrierenden Antikörper sind ein anti-Analyt Antikörper und ein anti-Fluorophor Antikörper, der zudem über die Eigenschaft verfügt, bei Bindung des Fluorophors dessen Fluoreszenz zu löschen. Externe Gaben des freien Analyten verschieben das eingestellte Gleichgewicht in Richtung Fluorophor-Bindung und damit Fluoreszenz-Löschung. Die Änderung der Fluoreszenz ist direkt an die Konzentration des freien Analyten gekoppelt und dient zu deren Bestimmung. Ein solcher Fluorescence Quenching Immunoassays wurde für die Konzentrationsbestimmung des Herbizides Diuron etabliert. Die erreichten Sensitivitäten erlauben die praktische, immundiagnostische Anwendung des Systems. Ein Dihapten-Konjugat wurde ebenfalls zum Aufbau eines Verfahrens zur Selektion Antikörper produzierender Zellen eingesetzt. Die Selektion der Antikörper produzierenden Zellen erfolgt unter Verwendung eines Toxinkonjugates. Dieses Konjugat besteht aus einem Liganden und einem Toxin. Die Antikörperbindung des Liganden behindert sterisch die Wechselwirkung der Toxinkomponente im Konjugat mit deren Zielstruktur in oder auf der Zelle. Nur Zellen die einen geeigneten Antikörper sezernieren, überleben die Selektion und reichern sich in der Kultur an. Das Selektionsverfahren wurde erfolgreich für die Selektion von E.coli Zellen eingesetzt, die einen rekombinanten, Fluorescein bindenden Antikörper produzierten. Das hierfür synthetisierte Toxinkonjugat bestand aus Fluorescein (Ligand) und Ampicillin (Toxinkomponente). Eine Ablösung der bisher für diese Aufgabe gebräuchlichen, außerordentlich kostenintensiven, Screening Methoden wird damit möglich.
Phytol aus dem Chlorophyllabbau ist limitierend für die Tocopherol (Vitamin E)-Synthese Als Bestandteil von Chlorophyll ist Phytol das am häufigsten vorkommende Isoprenoid in der Biosphäre. Große Mengen an Chlorophyll werden jährlich degradiert und dabei wird Phytol freigesetzt, über dessen Verbleib jedoch wenig bekannt ist. Es sollte der Nachweis erbracht werden, dass im Zuge des Chlorophyllabbaus hydrolysiertes Phytol Eingang in die Synthese anderer Phytylderivate findet. Während der Gehalt an Tocopherol, Chlorophyll und Fettsäurephytylester entwicklungs- bzw. seneszenzabhängig ist, bleibt der Gehalt an Phyllochinon etwa gleich. Auch in Samen ist der Gehalt von Tocopherol, Chlorophyll und Fettsäurephytylester entwicklungsabhängig. Es wurde gefolgert, dass nur die Synthesen von Tocopherol und Fettsäurephytylester während des Chlorophyllabbaus stimuliert werden. Daher sollten Mutanten analysiert werden, welche im Chlorophyllabbau inhibiert sind. Da Chlorophyllase den ersten Schritt des Chlorophyllabbaus katalysiert, wurden zwei unabhängige T-DNA-Insertionsmutanten für Chlorophyllase1 (CHL1) und eine T-DNA-Insertionsmutante für Chlorophyllase2 (CHL2) identifiziert und eine chl1-1chl2-Doppelmutante erzeugt. Die Analyse der Chlorophyllidanteile ergab eine im Vergleich zum Wildtyp starke Reduktion in den chl1-Mutanten, während der Chlorophyllidanteil von chl2 ähnlich hoch dem Wildtyp ist. Der Chlorophyllidanteil sich entwickelnder chl1-1chl2-Pflanzen nahm in der Seneszenz zu. Die Chlorophyllasemutanten zeigten kein verändertes Seneszenzverhalten im Vergleich zu den Wildtypen. Ferner konnte in den chl1-Linien nur geringfügig weniger Tocopherol und Fettsäurephytylester als in den Wildtypen nachgewiesen werden. Auch der Tocopherolgehalt der Samen war in den Chlorophyllasemutanten unverändert zu den Wildtypen. Aufgrund dessen wurde gefolgert, dass neben den Chorophyllasen CHL1 und CHL2 weitere Chlorophyllhydrolasen in Samen und Blättern von Arabidopsis existieren. Daher wurde auf andere Mutanten zurückgegriffen, in denen der Chlorophyllabbau stark inhibiert ist und die Seneszenz nach Dunkelinkubation im Vergleich zum Wildtyp deutlich verzögert ist. Eine deutliche Korrelation zwischen vermindertem Chlorophyllabbau und Gehalt an Tocopherol und Fettsäurephytylester konnte in den staygreen-Mutanten pao1 und zwei unabhängigen SGR (staygreen)-RNAi-Linien nachgewiesen werden. Damit konnte eindeutig gezeigt werden, dass die Synthese von Tocopherol und der Fettsäurephytylester durch die Chlorophyllhydrolyse induziert wird. Es wurde gefolgert, dass vor allem unter Seneszenz- bzw. Stressbedingungen dieser alternative Syntheseweg von Phytol eine Rolle spielt. Dennoch kommt der Phytylsynthese durch die de novo-Isoprenoidsynthese auch eine Bedeutung zu. Nach Behandlung von stickstoffmangelgestressten Wildtyppflanzen mit dem Inhibitor Fosmidomycin, welcher die plastidäre de novo-Isoprenoidsynthese hemmt, war der Tocopherolgehalt gegenüber stickstoffmangelgestressten Kontrollpflanzen stark reduziert. Ferner konnte eine T-DNA-Insertionsmutante der Geranylgeranylreduktase (GGR) identifiziert werden. Diese Mutante kann nur auf Nährmedium überleben, hat nur wenige grüne Blätter und bildet keine Samen. Es konnte kein Phyllochinon, Chlorophyll und keine Fettsäurephytylester, jedoch geringe Mengen Tocopherol nachgewiesen werden. Der Resttocopherolgehalt wird auf die Nebenaktivität einer anderen Reduktase zurückgeführt. Weiterhin wurde nur das Geranylgeranylderivat des Chlorophylls identifiziert. Diese Ergebnisse erlauben den Schluss, dass die phytylgruppenübertragenen Enzyme der Tocopherol-, Phyllochinon- und Fettsäurephytylestersynthese eine hohe Substratspezifität für die Phytylgruppe aufweisen. Nach Fütterung von Phytol konnte in ggr Tocopherol und Chlorophyll bestimmt werden. Aufgrund dessen kann gefolgert werden, dass Chlorophyllsynthetase aus Arabidopsis sowohl Geranylgeranyl-, als auch Phytylpyrophosphat als Substrat nutzen kann und damit ein breiteres Substratspektrum aufweist.
The cytoskeletal motor protein kinesin-1 (conventional kinesin) is the fast carrier for intracellular cargo transport along microtubules. So far most studies aimed at investigating the transport properties of individual motor molecules. However, the transport in cells usually involves the collective work of more than one motor. In the present work, we have studied the movement of beads as artificial loads/organelles pulled by several kinesin-1 motors in vitro. For a wide range of motor coverage of the beads and different bead (cargo) sizes the transport parameters walking distance or run length, velocity and force generation are measured. The results indicate that the transport parameters are influenced by the number of motors carrying the bead. While the transport velocity slightly decreases, an increase in the run length was measured and higher forces are determined, when more motors are involved. The effective number of motors pulling a bead is estimated by measuring the change in the hydrodynamic diameter of kinesin-coated beads using dynamic light scattering. The geometrical constraints imposed by the transport system have been taken into account. Thus, results for beads of different size and motor-surface coverage could be compared. In addition, run length-distributions obtained for the smallest bead size were matched to theoretically calculated distributions. The latter yielded an average number of pulling motors, which is in agreement with the effective motor numbers determined experimentally.
Effekte einer reduzierten Dosis von Pflanzenschutzmitteln auf tritrophische Systeme im Ackerbau

Chemische Pflanzenschutzmittel (PSM) bekämpfen nicht nur Schadorganismen, sondern haben aufgrund ihrer hohen Toxizität auch negative Auswirkungen auf Nicht-Ziel-Organismen. Die Fragestellung der Arbeit war es, ob mit reduzierten Anwendungen von PSM ihr Gefährdungspotenzial für Prädatoren von Schädlingen verringert und dadurch das Potenzial der natürlichen Schädlingsregulation erhöht wird. In dreijährigen Freilanduntersuchungen wurden die Effekte einer dauerhaft reduzierten Dosis von chemischen PSM auf die ökologische Situation im Ackerbau anhand von drei Fallbeispielen in einem konventionell bewirtschafteten Betrieb in der Magdeburger Börde untersucht. Drei über 15 ha große Felder wurden dauerhaft in zwei Teilflächen geteilt, wobei eine Teilfläche mit der vom Landwirt gewünschten Dosis (100 %-Variante) und die andere mit jeweils genau der halben Dosis (50 %-Variante) behandelt wurde. Mittels dieser Halbfelder-Vergleiche wurden die ökologischen Situationen bezüglich des Auftretens von Blattläusen und ihren Prädatoren sowie Unkräutern vor und nach der jeweiligen PSM-Behandlung aufgenommen und ökonomische Parameter ermittelt. Ergänzend wurden im Labor Modellgefäßversuche mit abgestuften Dosierungen von Insektiziden und Herbiziden durchgeführt. Die Insektizidbehandlung übte einen großen Einfluss auf die Blattläuse und ihre Prädatoren aus, während alle vorherigen Herbizid- und Fungizidbehandlungen zu keinen Unterschieden in der Abundanz der Blattläuse und ihrer Prädatoren zwischen beiden Varianten hervorriefen. Die reduzierte Insektiziddosis führte zu keiner guten Blattlauskontrolle, während die Abundanz der blattlausspezifischen Prädatoren positiv beeinflusst wurde. Die Araneae reagierten auf die reduzierte Dosis mit einer teilweise erhöhten Aktivitätsdichte und Artendiversität. Dagegen waren diesbezüglich keine eindeutigen Effekte auf die Carabidae festzustellen. Es traten keine strukturellen Veränderungen in Form einer erhöhten Unkrautdichte durch die reduzierte Herbiziddosis auf. Erste Hinweise auf mögliche langfristige Auswirkungen einer dauerhaft reduzierten PSM-Anwendung konnten nur bei der Verunkrautung und der Aktivitätsdichte der Araneae beobachtet werden. Blattläuse profitierten demnach mehr von der reduzierten Anwendung der PSM als ihre Prädatoren, so dass zwar das Potenzial der natürlichen Blattlausregulation erhöht, die Selbstregulation aber nicht verbessert wurde. Die geschonten Prädatoren schafften es nicht, die vorhandene Restpopulation der Blattläuse zu reduzieren. Dagegen konnte in den Laborversuchen gezeigt werden, das schon bei deutlich reduzierten Insektiziddosen eine ausreichende Blattlausbekämpfung möglich ist und eine weitere Einsparung durch Ausnutzung der natürlichen Regulation durch Prädatoren erreicht werden kann. Allerdings ist eine Übertragung der Ergebnisse von Laboruntersuchungen auf Freilandbedingungen schwierig. Es kann zu einer Überschätzung der Prädatorleistung führen.
Biogene Amine sind eine Substanzklasse, die bei Vertebraten und Invertebraten eine wichtige Komponente des endokrinen Systems darstellen. Sie binden an spezifische Rezeptoren der Gruppe der G-Protein gekoppelten Rezeptoren. In dieser Arbeit wurden zwei neue Rezeptoren aus der Schabe Periplaneta americana kloniert. Durch verschiedene Ansätze konnten zwei vollständige cDNA-Sequenzen isoliert werden. Die Aminosäuresequenzen weisen die größte Ähnlichkeit zu bereits bekannten Tyraminrezeptoren aus Locusta/Bombyx bzw. zu Dopaminrezeptoren aus Apis/Drosophila auf. Entsprechend wurden diese Rezeptoren Pea (P. americana) TYR1 und PeaDOP2 genannt. Deutliche Hinweise auf ihre Funktion lassen sich an den abgeleiteten Aminosäuresequenzen ablesen. Aminosäuren, die wichtig bei der Bildung der Bindungstasche, der Rezeptoraktivierung und der Kopplung eines G-Proteins sind, treten bei beiden Rezeptoren auf. Sequenzalignments stellen die Rezeptoren in die Gruppe anderer Tyraminrezeptoren bzw. der Invertebraten-Typ Dopaminrezeptoren. Das Transkript der beiden Rezeptoren konnte durch RT-PCR in verschiedenen Geweben nachgewiesen werden. Ein Ziel der Arbeit war die Gewinnung eines polyklonalen Antiserums gegen PeaTYR1. Dieses Serum detektiert im Homogenat von Gehirnen mehrere Banden, darunter auch eine mit der kalkulierten Masse von PeaTYR1. Präabsorption des Serums mit dem Peptid, welches zur Reinigung verwendet wurde, zeigt dessen Spezifität. An Gehirnschnitten markiert das Serum große Teile des Protocerebrums aber auch feinere Strukturen der Antennalloben, der optischen Loben und des Zentralkomplexes. Ein weiteres Serum gegen Tyramin führte zu einer Markierung von mehreren Neuronengruppen, welche sich in die optischen Loben und den Zentralkomplex verzweigen. Der αPeaTYR1-CPL3 Antikörper markierte die Plasmamembran von transfizierten HEK293-Zellen. Die Lokalisierung von Rezeptor und Ligand deuten darauf hin, dass Tyramin die optische und olfaktorische Wahrnehmung beeinflussen könnte.
Being living systems unable to adjust their location to changing environmental conditions, plants display homeostatic networks that have evolved to maintain transition metal levels in a very narrow concentration range in order to avoid either deficiency or toxicity. Hence, plants possess a broad repertoire of mechanisms for the cellular uptake, compartmentation and efflux, as well as for the chelation of transition metal ions. A small number of plants are hypertolerant to one or a few specific transition metals. Some metal tolerant plants are also able to hyperaccumulate metal ions. The Brassicaceae family member Arabidopis halleri ssp. halleri (L.) O´KANE and AL´SHEHBAZ is a hyperaccumulator of zinc (Zn), and it is closely related to the non-hypertolerant and non-hyperaccumulating model plant Arabidopsis thaliana (L.) HEYNHOLD. The close relationship renders A. halleri a promising emerging model plant for the comparative investigation of the molecular mechanisms behind hypertolerance and hyperaccumulation. Among several potential candidate genes that are probably involved in mediating the zinc-hypertolerant and zinc-hyperaccumulating trait is AhHMA3. The AhHMA3 gene is highly similar to AtHMA3 (AGI number: At4g30120) in A. thaliana, and its encoded protein belongs to the P-type IB ATPase family of integral membrane transporter proteins that transport transition metals. In contrast to the low AtHMA3 transcript levels in A. thaliana, the gene was found to be constitutively highly expressed across different Zn treatments in A. halleri, especially in shoots. In this study, the cloning and characterisation of the HMA3 gene and its promoter from Arabidopsis halleri (L.) O´KANE and AL´SHEHBAZ and Arabidopsis thaliana (L.) HEYNHOLD is described. Heterologously expressed AhHMA3 mediated enhanced tolerance to Zn and to a much lesser degree to cadmium (Cd) but not to cobalt (Co) in metal-sensitive mutant strains of budding yeast. It is demonstrated that the genome of A. halleri contains at least four copies of AhHMA3, AhHMA3-1 to AhHMA3-4. A copy-specific real-time RT-PCR indicated that an AhHMA3-1 related gene copy is the source of the constitutively high transcript level in A. halleri and not a gene copy similar to AhHMA3-2 or AhHMA3-4. In accordance with the enhanced AtHMA3mRNA transcript level in A. thaliana roots, an AtHMA3 promoter-GUS gene construct mediated GUS activity predominantly in the vascular tissues of roots and not in shoots. However, the observed AhHMA3-1 and AhHMA3-2 promoter-mediated GUS activity in A. thaliana or A. halleri plants did not reflect the constitutively high expression of AhHMA3 in shoots of A. halleri. It is suggested that other factors e. g. characteristic sequence inserts within the first intron of AhHMA3-1 might enable a constitutively high expression. Moreover, the unknown promoter of the AhHMA3-3 gene copy could be the source of the constitutively high AhHMA3 transcript levels in A. halleri. In that case, the AhHMA3-3 sequence is predicted to be highly homologous to AhHMA3-1. The lack of solid localisation data for the AhHMA3 protein prevents a clear functional assignment. The provided data suggest several possible functions of the AhHMA3 protein: Like AtHMA2 and AtHMA4 it might be localised to the plasma membrane and could contribute to the efficient translocation of Zn from root to shoot and/or to the cell-to-cell distribution of Zn in the shoot. If localised to the vacuolar membrane, then a role in maintaining a low cytoplasmic zinc concentration by vacuolar zinc sequestration is possible. In addition, AhHMA3 might be involved in the delivery of zinc ions to trichomes and mesophyll leaf cells that are major zinc storage sites in A. halleri.
Im Rahmen der Untersuchung zur Entwicklung der pelagischen Gemeinschaft des ehemals sauren Tagebauseenkomplexes Goitsche (pH~3) während dessen Flutung und Neutralisierung wurden Wechselwirkungen zwischen der Zusammensetzung von Organismengemeinschaften und der Variabilität des abiotischen Umfeldes untersucht.Im Mittelpunkt standen zwei von ihrer Kausalität her unterschiedliche Aspekte: • Der erste Aspekt betraf die Reifung von Ökosystemen: War der Reifungsprozess von pelagischen Gemeinschaften anhand der von Odum (1969) formulierten Kriterien zur Energetik der Gemeinschaft, zu den Nährstoffkreisläufen sowie zu strukturellen Merkmalen auf Ökosystem- und Individuenebene zu erfassen? Führten der physiologische Stress durch den niedrigen pH und physikalische Störungen der Schichtung durch das einströmende Flutungswasser zu einer Umkehr des Reifungsprozesses? Auf welchen Organisationsebenen der Lebensgemeinschaften waren die Auswirkungen dieser Stressoren erkennbar? • Der zweite Aspekt behandelte die Entwicklung der Artenzahl, die Gleichverteilung der Dominanz von Arten (Eveness) und die Diversität von Planktongemeinschaften entlang des Produktivitätsgradienten. Speziell wurde untersucht, ob die Artenzahl und die Diversität eine monoton positive oder eine unimodale Funktion der Produktivität waren und ob die Eveness eine monoton abnehmende Funktion der Produktivität war. Zur besseren Vorhersagbarkeit der Entwicklung dieser Indizes wurden in einem nächsten Schritt zusätzliche biotische und abiotische Faktoren (z.B. Konsumenteneffekte, physikalische Störung, Immigration) berücksichtigt. Zuletzt wurde die Hypothese getestet, dass unter dem Einfluss von extremem physiologischem Stress keine Abhängigkeit zwischen den betrachteten Indizes und der Produktivität von Ökosystemen besteht. Die Untersuchungen der vorliegenden Arbeit führten zu folgenden Ergebnissen und Schlussfolgerungen: 1) Der Reifungsprozess der Planktongemeinschaft war unter neutralen Bedingungen nicht eindeutig an einzelnen Kriterien festzumachen. Vielmehr schienen idiosynkratische Effekte einzelner Arten auf die Zusammensetzung und Funktion der Organismengemeinschaft von Bedeutung. Coenobiumbildende Kieselalgen sowie größere Cladoceren und Copepoden dominierten sehr rasch die Planktongemeinschaft und vermittelten den Eindruck eines reifen Ökosystems fast unmittelbar nach der Neutralisierung des Tagebausees. 2) Der Einfluss von physiologischem Säurestress und physikalischer Störung der Schichtung durch das eintretende Flutungswasser war gegenüber einem neutralen, ungestörten Teilbecken des Tagebausees (Referenzzustand) eindeutig zu erkennen. Die isolierte Betrachtung der Wirkung der Stressoren lieferte hinsichtlich fast aller Kriterien Anzeichen einer Verjüngung des Systems sensu Odum (1969, 1985). 3) Im betrachteten Ökosystem existierte eine Hierarchie innerhalb der Stressoren. Der Einfluss des Säurestresses dominierte gegenüber dem Einfluss der physikalischen Störung, wahrscheinlich indem er die Reaktionsmöglichkeiten der Planktongemeinschaft einschränkte. 4) Für Primärproduzenten war die Artenzahl eine monoton positive Funktion der realisierten Biomasse (einem Surrogatparameter für die Produktivität des Systems). Die Eveness war eine monoton negative Funktion der Produktivität. Die beobachtete unimodale Beziehung zwischen der Diversität und der Produktivität der Primärproduzenten muss als eine Folge der mathematischen Formulierung dieser Indizes betrachtet werden. 5) Die Ergebnisse multivariater Modelle zur Vorhersage der Artenzahl und der Eveness der Primärproduzenten in Abhängigkeit zusätzlicher erfassbarer biotischer und abiotischer Faktoren ermöglichten eine differenziertere Betrachtung der Ergebnisse: • Im Tagebausee Goitsche war die Eveness hauptsächlich von dichteabhängigen Prozessen gesteuert (negative Abhängigkeit zum Quadrat der Biomassen und zu den Lichtverhältnissen). • Die Entwicklung der Artenzahl war neben dem primären Einfluss der zunehmenden Biomasse auch durch qualitative und quantitative Aspekte der Konsumentengemeinschaft (Diversität und Biomasse des Zooplanktons) beeinflusst. Der Einfluss einer erhöhten Immigration auf die Artenzahl wurde nur zu Beginn der Flutung des Tagebausees beobachtet. 6) Auf Ebene der Konsumenten war die einzige eindeutig feststellbare Abhängigkeit ein Anstieg der Artenzahl mit steigender Biomasse. Das Fehlen von weiteren Beziehungen zwischen Diversitätsindizes und dem Produktivitätsgradienten wird darauf zurückgeführt, dass auf den unteren trophischen Ebenen der Primärproduzenten Ressourceneffekte (Bottom-Up) stärker ausgeprägt sind, wohingegen auf höheren trophischen Ebenen Konsumenteneffekte (Top-Down) dominieren. 7) In durch physiologischen Stress beeinflussten Systemen bestand keine Abhängigkeit zwischen den Diversitätsindizes (Artenzahl, Eveness und Diversität) und der Produktivität.
The need to develop sustainable resource management strategies for semi-arid and arid rangelands is acute as non-adapted grazing strategies lead to irreversible environmental problems such as desertification and associated loss of economic support to society. In such vulnerable ecosystems, successful implementation of sustainable management strategies depends on well-founded under-standing of processes at different scales that underlay the complex system dynamic. There is ample evidence that, in contrast to traditional sectoral approaches, only interdisciplinary research does work for resolving problems in conservation and natural resource management. In this thesis I combined a range of modeling approaches that integrate different disciplines and spatial scales in order to contribute to basic guidelines for sustainable management of semi-arid and arid range-lands. Since water availability and livestock management are seen as most potent determinants for the dynamics of semi-arid and arid ecosystems I focused on (i) the interaction of ecological and hydro-logical processes and (ii) the effect of farming strategies. First, I developed a grid-based and small-scaled model simulating vegetation dynamics and inter-linked hydrological processes. The simulation results suggest that ecohydrological interactions gain importance in rangelands with ascending slope where vegetation cover serves to obstruct run-off and decreases evaporation from the soil. Disturbances like overgrazing influence these positive feedback mechanisms by affecting vegetation cover and composition. In the second part, I present a modeling approach that has the power to transfer and integrate ecological information from the small scale vegetation model to the landscape scale, most relevant for the conservation of biodiversity and sustainable management of natural resources. I combined techniques of stochastic modeling with remotely sensed data and GIS to investigate to which ex-tent spatial interactions, like the movement of surface water by run-off in water limited environments, affect ecosystem functioning at the landscape scale. My simulation experiments show that overgrazing decreases the number of vegetation patches that act as hydrological sinks and run-off increases. The results of both simulation models implicate that different vegetation types should not only be regarded as provider of forage production but also as regulator of ecosystem functioning. Vegetation patches with good cover of perennial vegetation are capable to catch and conserve surface run-off from degraded surrounding areas. Therefore, downstream out of the simulated system is prevented and efficient use of water resources is guaranteed at all times. This consequence also applies to commercial rotational grazing strategies for semi-arid and arid rangelands with ascending slope where non-degraded paddocks act as hydrological sinks. Finally, by the help of an integrated ecological-economic modeling approach, I analyzed the relevance of farmers’ ecological knowledge for longterm functioning of semi-arid and arid grazing systems under current and future climatic conditions. The modeling approach consists of an ecological and an economic module and combines relevant processes on either level. Again, vegetation dynamics and forage productivity is derived by the small-scaled vegetation model. I showed that sustainable management of semi-arid and arid rangelands relies strongly on the farmers’ knowledge on how the ecosystem works. Furthermore, my simulation results indicate that the projected lower annual rainfall due to climate change in combination with non-adapted grazing strategies adds an additional layer of risk to these ecosystems that are already prone to land degradation. All simulation models focus on the most essential factors and ignore specific details. Therefore, even though all simulation models are parameterized for a specific dwarf shrub savanna in arid southern Namibia, the conclusions drawn are applicable for semi-arid and arid rangelands in general.
Sulphur, a macronutrient essential for plant growth, is among the most versatile elements in living organisms. Unfortunately, little is known about regulation of sulphate uptake and assimilation by plants. Identification of sulphate signalling processes will allow to control sulphate acquisition and assimilation and may prove useful in the future to improve sulphur-use efficiency in agriculture. Many of genes involved in sulphate metabolism are regulated on transcriptional level by products of other genes called transcription factors (TF). Several published experiments revealed TF genes that respond to sulphate deprivation, but none of these have been so far been characterized functionally. Thus, we aimed at identifying and characterising transcription factors that control sulphate metabolism in the model plant Arabidopsis thaliana. To achieve that goal we postulated that factors regulating Arabidopsis responses to inorganic sulphate deficiency change their transcriptional levels under sulphur-limited conditions. By comparing TF transcript profiles from plants grown on different sulphate regimes, we identified TF genes that may specifically induce or repress changes in expression of genes that allow plants to adapt to changes in sulphate availability. Candidate genes obtained from this screening were tested by reverse genetics approaches. Transgenic plants constitutively overproducing selected TF genes and mutant plants, lacking functional selected TF genes (knock out), were used. By comparing metabolite and transcript profiles from transgenic and wild type plants we aimed at confirming the role of selected AP2 TF candidate genes in plant adaptation to sulphur unavailability. After preliminary characterisation of WRKY24 and MYB93 TF genes, we postulate that these factors are involved in a complex multifactorial regulatory network, in which WRKY24 and MYB93 would act as superior factors regulating other transcription factors directly involved in the regulation of S-metabolism genes. Results obtained for plants overproducing TOE1 and TOE2 TF genes suggests that these factors may be involved in a mechanism, which is promoting synthesis of an essential amino acid, methionine, over synthesis of another amino acid, cysteine. Thus, TOE1 and TOE2 genes might be a part of transcriptional regulation of methionine synthesis. Approaches creating genetically manipulated plants may produce plant phenotypes of immediate biotechnological interest, such as plants with increased sulphate or sulphate-containing amino acid content, or better adapted to the sulphate unavailability.
Biomakromoleküle sind in der Natur für viele Abläufe in lebenden Organismen verantwortlich. Dies reicht vom Aufbau der extrazellulären Matrix und dem Cytoskelett über die Erkennung von Botenstoffen durch Rezeptoren bis hin zur Katalyse der verschiedensten Reaktionen in den Zellen selbst. Diese Aufgaben werden zum größten Teil von Proteinen übernommen, und besonders das spezifische Erkennen der Interaktionspartner ist für alle diese Moleküle äußerst wichtig, um eine fehlerfreie Funktion zu gewährleisten. Als Alternative zur evolutiven Erzeugung von optimalen Bindern und Katalysatoren auf der Basis von Aminosäuren und Nukleotiden wurden von Wulff, Shea und Mosbach synthetische molekular geprägte Polymere (molecularly imprinted polymers, MIPs) konzipiert. Das Prinzip dieser künstlichen Erkennungselemente beruht auf der Tatsache, dass sich funktionelle Monomere spezifisch um eine Schablone (Templat) anordnen. Werden diese Monomere dann vernetzend polymerisiert, entsteht ein Polymer mit molekularen Kavitäten, in denen die Funktionalitäten komplementär zum Templat fixiert sind. Dadurch ist die selektive Bindung des Templats in diese Kavitäten möglich. Aufgrund ihrer hohen chemischen und thermischen Stabilität und ihrer geringen Kosten haben “bio-inspirierte” molekular geprägte Polymere das Potential, biologische Erkennungselemente in der Affinitätschromatographie sowie in Biosensoren und Biochips zu ersetzen. Trotz einiger publizierter Sensorkonfigurationen steht der große Durchbruch noch aus. Ein Hindernis für Routineanwendungen ist die Signalgenerierung bei Bindung des Analyten an das Polymer. Eine Möglichkeit für die markerfreie Detektion ist die Benutzung von Kalorimetern, die Bindungs- oder Reaktionswärmen direkt messen können. In der Enzymtechnologie wird der Enzym-Thermistor für diesen Zweck eingesetzt, da enzymatische Reaktionen eine Enthalpie in einer Größenordnung von 5 – 100 kJ/mol besitzen. In dieser Arbeit wird die Herstellung von katalytisch geprägten Polymeren nach dem Verfahren des Oberflächenprägens erstmalig beschrieben. Die Methode zur Immobilisierung des Templats auf der Oberfläche von porösem Kieselgel sowie die Polymerzusammensetzung wurden optimiert. Weiter wird die Evaluation der katalytischen Eigenschaften über einen optischen Test, sowie das erste Mal die Kombination eines kalorimetrischen Transduktors – des Thermistors – mit der Analyterkennung durch ein katalytisch aktives MIP gezeigt. Bei diesen Messungen konnte zum ersten Mal gleichzeitig die Bindung/Desorption, sowie die katalytische Umwandlung des Substrats durch konzentrationsabhängige Wärmesignale nachgewiesen werden.
There is already strong evidence that temperate lakes have been highly vulnerable to human induced climate warming during the last century. Hitherto climate impact studies have mainly focussed on the impacts of the recent long-term warming in winter and spring and little is known on the influence of climate warming on temperate lakes in summer. In the present thesis, I studied some aspects, which may have been strongly involved in determining the response of a lake to climate warming in summer. Thereby I have focussed on climate induced impacts on the thermal characteristics and the phenology and abundance of summer plankton in a shallow polymictic lake (Müggelsee, Germany). First, the influence of climate warming on the phenology and abundance of the lake plankton was investigated across seasons. Fast-growing spring phytoplankton and zooplankton (Daphnia) advanced largely synchronously, whereas long-term changes in the phenology of slow-growing summer zooplankton were clearly species-specific and not synchronised. The phenology and/or abundance of several summer copepod species changed according to their individual thermal requirements at decisive developmental stages such as emergence from diapause in spring. The study emphasises that not only the degree of warming, but also its timing within the annual cycle is of great ecological importance. To analyse the impact of climate change on the thermal characteristics of the lake, I examined the long-term development of the daily epilimnetic temperature extrema during summer. The study demonstrated for the first time for lakes that the daily epilimnetic minima (during nighttime) have increased more rapidly than the daily epilimnetic maxima (during daytime), resulting in a distinct decrease in the daily epilimnetic temperature range. This day-night asymmetry in epilimnetic temperature was likely caused by an increased nighttime emission of long-wave radiation from the atmosphere. This underlines that not only increases in air temperature, but also changes in other meteorological variables such as wind speed, relative humidity and cloud cover may play an important role in determining the lake temperature with respect to further climate change. Furthermore, a short-term analysis on the mixing regime of the polymictic lake was conducted to examine the frequency and duration of stratification events and their impacts on dissolved oxygen, dissolved nutrients and summer phytoplankton. Even during the longest stratification events (heatwaves in 2003 and 2006) the thermal characteristics of the lake differed from those typically found in shallow dimictic lakes, which exhibit a continuous stratification during summer. Particularly, hypolimnetic temperatures were higher, favouring the depletion of oxygen and the accumulation of dissolved nutrient in the hypolimnion. Thermal stratification will be very likely amplified in the future, thus, I conclude that polymictic lakes will be very vulnerable to alterations in the thermal regime with respect to projections of further climate change during summer. Finally, a long-term case study on the long and short-term changes in the development of the planktonic larvae of the freshwater mussel Dreissena polymorpha was performed to analyse the impacts of simultaneous changes in the thermal and in the trophic regime of the lake. Both the climate warming and the decrease in external nutrient load were important in determining the abundance of the pelagic larvae by affecting different features of the life-history of this species throughout the warm season. The long-term increase in the abundance and length of larvae was related to the decrease in external nutrient loading and the change in phytoplankton composition. However, the recent heatwaves in 2003 and 2006 have offset this positive effect on larval abundance, due to unfavourable low oxygen concentrations that had resulted from extremely long stratification events, mimicking the effects of nutrient enrichment. Climate warming may thus induce counteracting effects in productive shallow lakes that underwent lake restoration through a decrease in external nutrient loading. I conclude that not only the nature of climate change and thus the timing of climate warming throughout the seasons and the occurrence of climatic extremes as heatwaves, but also site-specific lake conditions as the thermal mixing regime and the trophic state are crucial factors governing the impacts of climate warming on internal lake processes during summer. Consequently, further climate impact research on lake functioning should focus on how the different lake types respond to the complex environmental forcing in summer, to allow for a comprehensive understanding of human induced environmental changes in lakes.
Development and application of novel genetic transformation technologies in maize (Zea mays L.)

Plant genetic engineering approaches are of pivotal importance to both basic and applied research. However, rapid commercialization of genetically engineered crops, especially maize, raises several ecological and environmental concerns largely related to transgene flow via pollination. In most crops, the plastid genome is inherited uniparentally in a maternal manner. Consequently, a trait introduced into the plastid genome would not be transferred to the sexually compatible relatives of the crops via pollination. Thus, beside its several other advantages, plastid transformation provides transgene containment, and therefore, is an environmentally friendly approach for genetic engineering of crop plants. Reliable in vitro regeneration systems allowing repeated rounds of regeneration are of utmost importance to development of plastid transformation technologies in higher plants. While being the world’s major food crops, cereals are among the most difficult-to-handle plants in tissue culture which severely limits genetic engineering approaches. In maize, immature zygotic embryos provide the predominantly used material for establishing regeneration-competent cell or callus cultures for genetic transformation experiments. The procedures involved are demanding, laborious and time consuming and depend on greenhouse facilities. In one part of this work, a novel tissue culture and plant regeneration system was developed that uses maize leaf tissue and thus is independent of zygotic embryos and greenhouse facilities. Also, protocols were established for (i) the efficient induction of regeneration-competent callus from maize leaves in the dark, (ii) inducing highly regenerable callus in the light, and (iii) the use of leaf-derived callus for the generation of stably transformed maize plants. Furthermore, several selection methods were tested for developing a plastid transformation system in maize. However, stable plastid transformed maize plants could not be yet recovered. Possible explanations as well as suggestions for future attempts towards developing plastid transformation in maize are discussed. Nevertheless, these results represent a first essential step towards developing chloroplast transformation technology for maize, a method that requires multiple rounds of plant regeneration and selection to obtain genetically stable transgenic plants. In order to apply the newly developed transformation system towards metabolic engineering of carotenoid biosynthesis, the daffodil phytoene synthase (PSY) gene was integrated into the maize genome. The results illustrate that expression of a recombinant PSY significantly increases carotenoid levels in leaves. The beta-carotene (pro-vitamin A) amounts in leaves of transgenic plants were increased by ~21% in comparison to the wild-type. These results represent evidence for maize to have significant potential to accumulate higher amounts of carotenoids, especially beta-carotene, through transgenic expression of phytoene synthases. Finally, progresses were made towards developing transformation technologies in Peperomia (Piperaceae) by establishing an efficient leaf-based regeneration system. Also, factors determining plastid size and number in Peperomia, whose species display great interspecific variation in chloroplast size and number per cell, were investigated. The results suggest that organelle size and number are regulated in a tissue-specific manner rather than in dependency on the plastid type. Investigating plastid morphology in Peperomia species with giant chloroplasts, plasmatic connections between chloroplasts (stromules) were observed under the light microscope and in the absence of tissue fixation or GFP overexpression demonstrating the relevance of these structures in vivo. Furthermore, bacteria-like microorganisms were discovered within Peperomia cells, suggesting that this genus provides an interesting model not only for studying plastid biology but also for investigating plant-microbe interactions.
Alle Organismen sind für ihr Überleben auf Metalle angewiesen. Hierbei gibt es für jedes Metall einen Konzentrationsbereich, der das Optimum zwischen Metallmangel, -bedarf und -toxizität darstellt. Es gilt mittlerweile als erwiesen, dass alle Organismen zur Aufrechterhaltung des Metallgleichgewichts ein komplexes Netzwerk von Proteinen und niedermolekularen Verbindungen entwickelt haben. Die molekularen Komponenten dieses Netzwerks sind nur zu einem Teil bekannt und charakterisiert: In den letzten Jahren wurden einige Proteinfamilien identifiziert, deren Mitglieder Metalle durch Lipidmembranen transportieren. Eine dieser Metalltransporterfamilien ist die Cation Diffusion Facilitator (CDF)-Familie: Alle charakterisierten Mitglieder exportieren Metalle aus dem Zytoplasma – entweder in zelluläre Kompartimente oder aus der Zelle heraus. Von den zwölf Mitgliedern dieser Familie in Arabidopsis thaliana (A. thaliana) – Metall Toleranz Protein (MTP)-1 bis -12 – wurden bisher AtMTP1 und AtMTP3 charakterisiert. In dieser Arbeit wird die Charakterisierung von AtMTP2 beschrieben. Wie die homologen Proteine AtMTP1 und AtMTP3 führt AtMTP2 zu Zn-Toleranz, wenn es heterolog in Zn-sensitiven Hefemutanten exprimiert wird. Mit AtMTP2 transformierte Hefemutanten zeigten darüber hinaus erhöhte Co-Toleranz. Expression von chimären AtMTP2/GFP Fusionsproteinen in Hefe, A.thaliana protoplasten und in stabil transformierten A.thalinana Planzenlinien deutet auf Lokalisation of AtMTP2 in Membranen des Endoplasmatischen Retikulums (ER) hin, wenn GFP an den C-Terminus von MTP2 fusioniert wird. Fusion of GFP an den N-Terminus von AtMTP2 führte zu Lokalisation in der vakuolären Membran, was wahrscheinlichsten auf Fehllokalisierung durch Maskierung eines ER-Retentionsmotivs (XXRR) am N-Terminus von AtMTP2 zurückgeht. Dies legt nahe, dass AtMTP2 die erwähnten Metalle in das Endomembransystem der Zelle transportieren kann. Eine gewebespezifische Lokalisierung wurde mit Pflanzen durchgeführt, die das β-Glucuronidase (GUS)-Reporterprotein bzw. chimäre Fusionsproteine aus EGFP und AtMTP2 unter Kontrolle des nativen pMTP2-Promotors exprimierten. Diese Experimente bestätigten zum einen, dass der pMTP2-Promotor nur unter Zn-Defizienz aktiv ist. GUS-Aktivität wurde unter diesen Bedingungen in zwei Zonen der Wurzelspitze beobachtet: in den isodiametrischen Zellen der meristematischen Zone und in der beginnenden Wurzelhaarzone. Darüber hinaus konnte gezeigt werden, dass die EGFP-Fusionsproteine unter Kontrolle des nativen pMTP2-Promotors nur in epidermalen Zellen exprimiert werden. Für eine homozygote Knockout- Linie, mtp2-S3, konnte bisher kein eindeutiger Phänotyp identifiziert werden. Auf Grundlage der bisher durchgeführten Charakterisierung von AtMTP2 erscheinen zwei Modelle der Funktion von AtMTP2 in der Pflanze möglich: AtMTP2 könnte essentiell für die Versorgung des ER mit Zn unter Zn-Mangelbedingungen sein. Hierfür spricht, dass AtMTP2 in jungen, teilungsaktiven und damit Zn-benötigenden Wurzelzonen exprimiert wird. Die auf die Epidermis beschränkte Lokalisation könnte bei diesem Modell auf die Möglichkeit der zwischenzellulären Zn-Verteilung innerhalb des ER über Desmotubules hindeuten. Alternativ könnte AtMTP2 eine Funktion bei der Detoxifizierung von Zn unter Zn-Schock Bedingungen haben: Es ist bekannt, dass unter Zn- Mangelbedingungen die Expression der zellulären Zn-Aufnahmesysteme hochreguliert wird. Wenn nun die Zn-Verfügbarkeit im Boden z. B durch eine pH-Änderung innerhalb kurzer Zeit stark ansteigt, besteht die Notwendigkeit der Entgiftung von Zn innerhalb der Zelle, bis der starke Einstrom von Zn ins Zytoplasma durch die Deaktivierung der Zn-Aufnahmesysteme und einer geringeren Expression in der Pflanze gedrosselt ist. Ein ähnlicher Mechanismus wurde in der Bäckerhefe S. cerevisae beschrieben, in der darüber hinaus ein Zn-Transporter verstärkt exprimiert wird, der Zn durch Transport in die Vakuole entgiften kann. Es ist durchaus möglich, dass in Arabidopsis AtMTP2 die Zn-Detoxifizierung unter diesen speziellen Bedingungen durch Zn-Transport in das ER oder die Vakuole vermittelt. Zur Identifikation weiterer Komponenten des Metallhomöostasenetzwerks sind verschiedene Ansätze denkbar. In dieser Arbeit wurde in Hefe ein heterologer Screen durchgeführt, um Interaktoren für vier Mitglieder der Arabidopsis-CDF-Familie zu identifizieren. Unter den 11 im Hefesystem bestätigten Kandidaten befindet sich mit AtSPL1 ein AtMTP1-Interaktionskandidat, der möglicherweise eine Rolle bei der Cu-,Zn-Homöostase spielt. Als wahrscheinliche AtMTP3-Interaktionskandidaten wurde die c”-Untereinheit der vakuolären H+-ATPase AtVHA identifiziert sowie mit AtNPSN13 ein Protein, das vermutlich eine Rolle bei Fusionen von Vesikeln mit Zielmembranen spielt. Ein anderer Ansatz zur Identifikation neuer Metallhomöostasegene ist die vergleichende Elementanalyse von natürlichen oder mutagenisierten Pflanzenpopulationen. Voraussetzung für diesen Ansatz ist die schnelle und genaue Analyse des Elementgehalts von Pflanzen. Eine etablierte Methode zur simultanen Bestimmung von bis zu 65 Elementen in einer Probe ist die Inductively Coupled Plasma Optical Emission Spectrometry (ICP OES). Der limitierende Faktor für einen hohen Probendurchsatz ist die Notwendigkeit, Proben für die Analyse zu verflüssigen. Eine alternative Methode der Probenzuführung zum Analysegerät ist die elektrothermale Verdampfung (ETV) der Probe. Zur weitgehend automatisierten Analyse von Pflanzenmaterial mit minimiertem Arbeitsaufwand wurde eine Methode entwickelt, die auf der Kopplung der ETV mit der ICP OES basiert.
During the last decades, the global change of the environment has caused a dramatic loss of habitats and species. In Central Europe, open habitats are particularly affected. The main objective of this thesis was to experimentally test the suitability of wild megaherbivore grazing as a conservation tool to manage open habitats. We studied the effect of wild ungulates in a 160 ha game preserve in NE Germany in three successional stages (i) Corynephorus canescens-dominated grassland, (ii) ruderal tall forb vegetation dominated by Tanacetum vulgare and (iii) Pinus sylvestris-pioneer forest over three years. Our results demonstrate that wild megaherbivores considerably affected species composition and delayed successional pathways in open habitats. Grazing effects differed considerably between successional stages: species richness was higher in grazed ruderal and pioneer forest plots, but not in the Corynephorus sites. Species composition changed significantly in the Corynephorus and ruderal sites. Grazed ruderal sites had turned into sites with very short vegetation dominated by Agrostis spp. and the moss Brachythecium albicans, most species did not flower. Woody plant cover was significantly affected only in the pioneer forest sites. Young pine trees were severely damaged and tree height was considerably reduced, leading to a “Pinus-macchie”-appearance. Ecological patterns and processes are known to vary with spatial scale. Since grazing by megaherbivores has a strong spatial component, the scale of monitoring success of grazing may largely differ among and within different systems. Thus, the second aim of this thesis was to test whether grazing effects are consistent over different spatial scales, and to give recommendations for appropriate monitoring scales. For this purpose, we studied grazing effects on plant community structure using multi-scale plots that included three nested spatial scales (0.25 m2, 4 m2, and 40 m2). Over all vegetation types, the scale of observation directly affected grazing effects on woody plant cover and on floristic similarity, but not on the proportion of open soil and species richness. Grazing effects manifested at small scales regarding floristic similarity in pioneer forest and ruderal sites and regarding species richness in ruderal sites. The direction of scale-effects on similarity differed between vegetation types: Grazing effects on floristic similarity in the Corynephorus sites were significantly higher at the medium and large scale, while in the pioneer forest sites they were significantly higher at the smallest scale. Disturbances initiate vegetation changes by creating gaps and affecting colonization and extinction rates. The third intention of the thesis was to investigate the effect of small-scale disturbances on the species-level. In a sowing experiment, we studied early establishment probabilities of Corynephorus canescens, a key species of open sandy habitats. Applying two different regimes of mechanical ground disturbance (disturbed and undisturbed) in the three successional stages mentioned above, we focused on the interactive effects of small-scale disturbances, successional stage and year-to-year variation. Disturbance led to higher emergence in a humid and to lower emergence in a very dry year. Apparently, when soil moisture was sufficient, the main factor limiting C. canescens establishment was competition, while in the dry year water became the limiting factor. Survival rates were not affected by disturbance. In humid years, C. canescens emerged in higher numbers in open successional stages while in the dry year, emergence rates were higher in late stages, suggesting an important role of late successional stages for the persistence of C. canescens. We conclude that wild ungulate grazing is a useful tool to slow down succession and to preserve a species-rich, open landscape, because it does not only create disturbances, thereby supporting early successional stages, but at the same time efficiently controls woody plant cover. However, wild ungulate grazing considerably changed the overall appearance of the landscape. Additional measures like shifting exclosures might be necessary to allow vulnerable species to flower and reproduce. We further conclude that studying grazing impacts on a range of scales is crucial, since different parameters are affected at different spatial scales. Larger scales are suitable for assessing grazing impact on structural parameters like the proportion of open soil or woody plant cover, whereas species richness and floristic similarity are affected at smaller scales. Our results further indicate that the optimal strategy for promoting C. canescens is to apply disturbances just before seed dispersal and not during dry years. Further, at the landscape scale, facilitation by late successional species may be an important mechanism for the persistence of protected pioneer species.
