Die vorliegende Arbeit thematisiert die Synthese und die Polymerisation von Monomeren auf der Basis nachwachsender Rohstoffe wie zum Beispiel in Gewürzen und ätherischen Ölen enthaltenen kommerziell verfügbaren Phenylpropanoiden (Eugenol, Isoeugenol, Zimtalkohol, Anethol und Estragol) und des Terpenoids Myrtenol sowie ausgehend von der Rinde einer Birke (Betula pendula) und der Korkeiche (Quercus suber). Ausgewählte Phenylpropanoide (Eugenol, Isoeugenol und Zimtalkohol) und das Terpenoid Myrtenol wurden zunächst in den jeweiligen Laurylester überführt und anschließend das olefinische Strukturelement epoxidiert, wobei 4 neue (2-Methoxy-4-(oxiran-2-ylmethyl)phenyldodecanoat, 2-Methoxy-4-(3-methyl-oxiran-2-yl)phenyldodecanoat, (3-Phenyloxiran-2-yl)methyldodecanoat, (7,7-Dimethyl-3-oxatricyclo[4.1.1.02,4]octan-2-yl)methyldodecanoat) und 2 bereits bekannte monofunktionelle Epoxide (2-(4-Methoxybenzyl)oxiran und 2-(4-Methoxyphenyl)-3-methyloxiran) erhalten wurden, die mittels 1H-NMR-, 13C-NMR- und FT-IR-Spektroskopie sowie mit DSC untersucht wurden. Die Photo-DSC Untersuchung der Epoxidmonomere in einer kationischen Photopolymerisation bei 40 °C ergab die maximale Polymerisationsgeschwindigkeit (Rpmax: 0,005 s-1 bis 0,038 s-1) sowie die Zeit (tmax: 13 s bis 26 s) bis zum Erreichen des Rpmax-Wertes und führte zu flüssigen Oligomeren, deren zahlenmittlerer Polymerisationsgrad mit 3 bis 6 mittels GPC bestimmt wurde. Die Umsetzung von 2-Methoxy-4-(oxiran-2-ylmethyl)phenyldodecanoat mit Methacrylsäure ergab ein Isomerengemisch (2-Methoxy-4-(2-hydroxy-3-(methacryloyloxy)propyl)phenyldodecanoat und 2-Methoxy-4-(2-(methacryl-oyloxy)-3-hydroxypropyl)phenyldodecanoat), das mittels Photo-DSC in einer freien radikalischen Photopolymerisation untersucht wurde (Rpmax: 0,105 s-1 und tmax: 5 s), die zu festen in Chloroform unlöslichen Polymeren führte.
Aus Korkpulver und gemahlener Birkenrinde wurden selektiv 2 kristalline ω-Hydroxyfettsäuren (9,10-Epoxy-18-hydroxyoctadecansäure und 22-Hydroxydocosansäure) isoliert. Die kationische Photopolymerisation der 9,10-Epoxy-18-hydroxyoctadecansäure ergab einen nahezu farblosen transparenten und bei Raumtemperatur elastischen Film, welcher ein Anwendungspotential für Oberflächenbeschichtungen hat. Aus der Reaktion von 9,10-Epoxy-18-hydroxyoctadecansäure mit Methacrylsäure wurde ein bei Raumtemperatur flüssiges Gemisch aus zwei Konstitutionsisomeren (9,18-Dihydroxy-10-(methacryloyloxy)octadecansäure und 9-(Methacryloyloxy)-10,18-dihydroxyoctadecansäure) erhalten (Tg: -60 °C). Die radikalische Photopolymerisation dieser Konstitutionsisomere wurde ebenfalls mittels Photo-DSC untersucht (Rpmax: 0,098 s-1 und tmax: 3,8 s). Die Reaktion von 22-Hydroxydocosansäure mit Methacryloylchlorid ergab die kristalline 22-(Methacryloyloxy)docosansäure, welche ebenfalls in einer radikalischen Photopolymerisation mittels Photo-DSC untersucht wurde (Rpmax: 0,023 s-1 und tmax: 9,6 s).
Die mittels AIBN in Dimethylsulfoxid initiierte Homopolymerisation der 22-(Methacryloyloxy)docosansäure und der Isomerengemische bestehend aus 2-Methoxy-4-(2-hydroxy-3-(methacryloyloxy)propyl)phenyldodecanoat und 2-Methoxy-4-(2-(methacryl-oyloxy)-3-hydroxypropyl)phenyldodecanoat sowie aus 9,18-Dihydroxy-10-(methacryloy-loxy)octadecansäure und 9-(Methacryloyloxy)-10,18-dihydroxyoctadecansäure ergab feste lösliche Polymere, die mittels 1H-NMR- und FT-IR-Spektroskopie, GPC (Poly(2-methoxy-4-(2-hydroxy-3-(methacryloyloxy)propyl)phenyldodecanoat / 2-methoxy-4-(2-(methacryloyloxy)-3-hydroxypropyl)phenyldodecanoat): Pn = 94) und DSC (Poly(2-methoxy-4-(2-hydroxy-3-(methacryloyloxy)propyl)phenyldodecanoat / 2-methoxy-4-(2-(methacryloyloxy)-3-hydroxypropyl)phenyldodecanoat): Tg: 52 °C; Poly(9,18-dihydroxy-10-(methacryloyloxy)-octadecansäure / 9-(methacryloyloxy)-10,18-dihydroxyoctadecansäure): Tg: 10 °C; Poly(22-(methacryloyloxy)docosansäure): Tm: 74,1 °C, wobei der Schmelzpunkt mit dem des Photopolymers (Tm = 76,8 °C) vergleichbar ist) charakterisiert wurden.
Das bereits bekannte Monomer 4-(4-Methacryloyloxyphenyl)butan-2-on wurde ausgehend von 4-(4-Hydroxyphenyl)butan-2-on hergestellt, welches aus Birkenrinde gewonnen werden kann, und unter identischen Bedingungen für einen Vergleich mit den neuen Monomeren polymerisiert. Die freie radikalische Polymerisation führte zu Poly(4-(4-methacryloyloxyphenyl)butan-2-on) (Pn: 214 und Tg: 83 °C). Neben der Homopolymerisation wurde eine statistische Copolymerisation des Isomerengemisches 2-Methoxy-4-(2-hydroxy-3-(methacryl-oyloxy)propyl)phenyldodecanoat / 2-Methoxy-4-(2-(methacryloyloxy)-3-hydroxypropyl)-phenyldodecanoat mit 4-(4-Methacryloyloxyphenyl)butan-2-on untersucht, wobei ein äquimolarer Einsatz der Ausgangsmonomere zu einem Anstieg der Ausbeute, der Molmassenverteilung und der Dispersität des Copolymers (Tg: 44 °C) führte. Die unter Verwendung von Diethylcarbonat als „grünes“ Lösungsmittel mittels AIBN initiierten freien radikalischen Homopolymerisationen von 4-(4-Methacryloyloxyphenyl)butan-2-on und von Laurylmethacrylat ergaben vergleichbare Polymerisationsgrade der Homopolymere (Pn: 150), welche jedoch aufgrund ihrer Strukturunterschiede deutlich unterschiedliche Glasübergangstemperaturen hatten (Poly(4-(4-methacryloyloxyphenyl)butan-2-on): Tg: 70 °C, Poly(laurylmethacrylat) Tg: -49 °C. Eine statistische Copolymerisation äquimolarer Stoffmengen der beiden Monomere in Diethylcarbonat führte bei einer Polymerisationszeit von 60 Minuten zu einem leicht bevorzugten Einbau des 4-(4-Methacryloyloxyphenyl)butan-2-on in das Copolymer (Tg: 17 °C). Copolymerisationsdiagramme für die freien radikalischen Copolymerisationen von 4-(4-Methacryloyloxyphenyl)butan-2-on mit n-Butylmethacrylat beziehungsweise 2-(Dimethylamino)ethylmethacrylat (t: 20 min bis 60 min; Molenbrüche (X) für 4-(4-Methacryloyloxyphenyl)butan-2-on: 0,2; 0,4; 0,6 und 0,8) zeigten ein nahezu ideales azeotropes Copolymerisationsverhalten, obwohl ein leicht bevorzugter Einbau von 4-(4-Methacryloyloxyphenyl)butan-2-on in das jeweilige Copolymer beobachtet wurde. Dabei korreliert ein Anstieg der Ausbeute und der Glasübergangstemperatur der erhaltenen Copolymere mit einem zunehmenden Gehalt an 4-(4-Methacryloyloxyphenyl)butan-2-on im Reaktionsgemisch. Die unter Einsatz der modifizierten Gibbs-DiMarzio-Gleichung berechneten Glasübergangstemperaturen der Copolymere stimmten mit den gemessenen Werten gut überein. Das ist eine gute Ausgangsbasis für die Bestimmung der Glasübergangstemperatur eines Copolymers mit einer beliebigen Zusammensetzung.
Subdividing space through interfaces leads to many space partitions that are relevant to soft matter self-assembly. Prominent examples include cellular media, e.g. soap froths, which are bubbles of air separated by interfaces of soap and water, but also more complex partitions such as bicontinuous minimal surfaces.
Using computer simulations, this thesis analyses soft matter systems in terms of the relationship between the physical forces between the system's constituents and the structure of the resulting interfaces or partitions. The focus is on two systems, copolymeric self-assembly and the so-called Quantizer problem, where the driving force of structure formation, the minimisation of the free-energy, is an interplay of surface area minimisation and stretching contributions, favouring cells of uniform thickness.
In the first part of the thesis we address copolymeric phase formation with sharp interfaces. We analyse a columnar copolymer system "forced" to assemble on a spherical surface, where the perfect solution, the hexagonal tiling, is topologically prohibited. For a system of three-armed copolymers, the resulting structure is described by solutions of the so-called Thomson problem, the search of minimal energy configurations of repelling charges on a sphere. We find three intertwined Thomson problem solutions on a single sphere, occurring at a probability depending on the radius of the substrate.
We then investigate the formation of amorphous and crystalline structures in the Quantizer system, a particulate model with an energy functional without surface tension that favours spherical cells of equal size. We find that quasi-static equilibrium cooling allows the Quantizer system to crystallise into a BCC ground state, whereas quenching and non-equilibrium cooling, i.e. cooling at slower rates then quenching, leads to an approximately hyperuniform, amorphous state. The assumed universality of the latter, i.e. independence of energy minimisation method or initial configuration, is strengthened by our results. We expand the Quantizer system by introducing interface tension, creating a model that we find to mimic polymeric micelle systems: An order-disorder phase transition is observed with a stable Frank-Caspar phase.
The second part considers bicontinuous partitions of space into two network-like domains, and introduces an open-source tool for the identification of structures in electron microscopy images. We expand a method of matching experimentally accessible projections with computed projections of potential structures, introduced by Deng and Mieczkowski (1998). The computed structures are modelled using nodal representations of constant-mean-curvature surfaces. A case study conducted on etioplast cell membranes in chloroplast precursors establishes the double Diamond surface structure to be dominant in these plant cells. We automate the matching process employing deep-learning methods, which manage to identify structures with excellent accuracy.
Die vorliegende Arbeit thematisiert die Synthese und Charakterisierung von neuen funktionalisierten ionischen Flüssigkeiten und deren Polymerisation. Die ionischen Flüssigkeiten wurden dabei sowohl mit polymerisierbaren Kationen als auch Anionen hergestellt. Zum einen wurden bei thermisch initiierten Polymerisationen Azobis(isobutyronitril) (AIBN) verwendet und zum anderen dienten bei photochemisch initiierten Polymerisationen Bis-4-(methoxybenzoyl)diethylgermanium (Ivocerin®) als Radikalstarter.
Mittels Gelpermeationschromatographie konnte das Homopolymer Polydimethylaminoethylmethacrylat untersucht werden, welches erst im Anschluss an die GPC-Messungen polymeranalog modifiziert wurde. Dabei wurden nach einer Quaternisierung und anschließender Anionenmetathese bei diesen Polymeren die Grenzviskositäten bestimmt und mit den Grenzviskositäten der direkt polymerisierten ionischen Flüssigkeiten verglichen. Bei der direkten Polymerisation von Poly(N-[2-(Methacryloyloxy)ethyl]-N-butyl-N,N-dimethyl-ammoniumbis(trifluormethylsulfonyl)imid) lag [η_Huggins] bei 100 mL/g und bei dem polymeranalog hergestellten Polymer betrug [η_Huggins] = 40 mL/g.
Die ionischen Flüssigkeiten mit polymerisierbaren funktionellen Gruppen wurden mittels Photo-DSC hinsichtlich der maximalen Polymerisationsgeschwindigkeit (Rpmax), der Zeit, in der dieses Maximum erreicht wurde, tmax, ihrer Glasüberganstemperatur (Tg) und des Umsatzes an Vinylprotonen untersucht. Bei diesen Messungen wurde zum einen der Einfluss der unterschiedlichen Alkylkettenlänge am Ammoniumion und der Einfluss von verschiedenen Anionen bei gleichbleibender Kationenstruktur analysiert. So polymerisierte das ethylsubstituierte Kation mit einer tmax von 21 Sekunden am langsamsten. Die maximale Polymerisationsgeschwindigkeit (Rpmax) betrug 3.3∙10-2 s-1. Die tmax Werte der übrigen alkylsubstituierten ionischen Flüssigkeiten mit einer polymerisierbaren funktionellen Gruppe hingegen lagen zwischen 10 und 15 Sekunden. Die Glasübergangstemperaturen der mittels photoinduzierter Polymerisation hergestellten Polymere lagen mit 44 bis 55 °C nahe beieinander. Alle Monomere zeigten einen hohen Umsatz der Vinylprotonen; er betrug zwischen 93 und 100%.
Mithilfe einer Bandanlage, ausgerüstet mit einer LED (λ = 395 nm), konnten Polymerfilme hergestellt werden. Der Umsatz an Doppelbindungsäquivalenten dieser Filme wurde anhand der 1H-NMR Spektroskopie bestimmt. Bei der dynamisch-mechanischen Analyse wurden die Polymerfilme mit einer konstanten Heizrate und Frequenz periodisch wechselnden Beanspruchungen ausgesetzt, um die Glasübergangstemperaturen zu bestimmen. Die niedrigste Tg mit 26 °C besaß das butylsubstituierte N-[2-(Methacryloyloxy)ethyl]-N-butyl-N,N-dimethyl-ammoniumbis(trifluormethylsulfonyl)imid, welches als Polymerfilm mit Ivocerin® als Initiator hergestellt wurde, wohingegen die höchste Tg bei dem gleichen Polymer, welches direkt durch freie radikalische Polymerisation der ionischen Flüssigkeit in Masse mit AIBN hergestellt wurde, 51 °C betrug. Zusätzlich wurden die Filme unter dem Aspekt der Topographie mit einem Rasterkraftmikroskop untersucht, welches eine Domänenstruktur des Polymers N-[2-(methacryloyloxy)ethyl]-N-butyl-N,N-dimethyl-ammonium tris(pentafluorethyl)trifluorphosphat offenbarte.
In recent years people have realised non-renewability of our modern society which relays on spending huge amounts of energy mostly produced from fosil fuels, such as oil and coal, and the shift towards more sustainable energy sources has started. However, sustainable sources of energy, such as wind-, solar- and hydro-energy, produce primarily electrical energy and can not just be poured in canister like many fosil fuels, creating necessity for rechragable batteries. However, modern Li-ion batteries are made from toxic heavy metals and sustainable alternatives are needed. Here we show that naturally abundant catecholic and guaiacyl groups can be utilised to replace heavy metals in Li-ion batteries.
Foremost vanillin, a naturally occurring food additive that can be sustainably synthesised from industrial biowaste, lignin, was utilised to synthesise materials that showed extraordinary performance as cathodes in Li-ion batteries. Furthermore, behaviour of catecholic and guiacyl groups in Li-ion system was compared, confirming usability of guiacayl containing biopolymers as cathodes in Li-ion batteries. Lastly, naturally occurring polyphenol, tannic acid, was incorporated in fully bioderived hybrid material that shows performance comparable to commercial Li-ion batteries and good stability.
This thesis presents an important advancement in understanding of biowaste derived cathode materials for Li-ion batteries. Further research should be conducted to better understand behaviour of guaiacyl groups during Li-ion battery cycling. Lastly, challenges of incorporation of lignin, an industrial biowaste, have to be addressed and lignin should be incorporated as a cathode material in Li-ion batteries.
New bio-based polymers

Redox-responsive polymers, such as poly(disulfide)s, are a versatile class of polymers with potential applications including gene- and drug-carrier systems. Their degradability under reductive conditions allows for a controlled response to the different redox states that are present throughout the body. Poly(disulfide)s are typically synthesized by step growth polymerizations. Step growth polymerizations, however, may suffer from low conversions and therefore low molar masses, limiting potential applications. The purpose of this thesis was therefore to find and investigate new synthetic routes towards the synthesis of amino acid-based poly(disulfide)s.
The different routes in this thesis include entropy-driven ring opening polymerizations of novel macrocyclic monomers, derived from cystine derivatives. These monomers were obtained with overall yields of up to 77% and were analyzed by mass spectrometry as well as by 1D and 2D NMR spectroscopy. The kinetics of the entropy-driven ring-opening metathesis polymerization (ED-ROMP) were thoroughly investigated in dependence of temperature, monomer concentration, and catalyst concentration. The polymerization was optimized to yield poly(disulfide)s with weight average molar masses of up to 80 kDa and conversions of ~80%, at the thermodynamic equilibrium. Additionally, an alternative metal free polymerization, namely the entropy-driven ring-opening disulfide metathesis polymerization (ED-RODiMP) was established for the polymerization of the macrocyclic monomers. The effect of different solvents, concentrations and catalyst loadings on the polymerization process and its kinetics were studied. Polymers with very high weight average molar masses of up to 177 kDa were obtained. Moreover, various post-polymerization reactions were successfully performed.
This work provides the first example of the homopolymerization of endo-cyclic disulfides by ED-ROMP and the first substantial study into the kinetics of the ED-RODiMP process.
In the present work side-chain polystyrenes were synthesized and characterized, in order to be applied in multilayer OLEDs fabricated by solution process techniques. Manufacture of optoelectronic devices by solution process techniques is meant to decrease significantly fabrication cost and allow large scale production of such devices.
This dissertation focusses in three series, enveloped in two material classes. The two classes differ to each other in the type of charge transport exhibited, either ambipolar transport or electron transport. All materials were applied in all-organic solution processed green Ir-based devices.
In the first part, a series of ambipolar host materials were developed to transport both charge types, holes and electrons, and be applied especially as matrix for green Ir-based emitters. It was possible to increase devices efficacy by modulating the predominant charge transport type. This was achieved by modification of molecules electron transport part with more electron-deficient heterocycles or by extending the delocalization of the LUMO. Efficiencies up to 28.9 cd/A were observed for all-organic solution-process three layer devices.
In the second part, suitability of triarylboranes and tetraphenylsilanes as electron transport materials was studied. High triplet energies were obtained, up to 2.95 eV, by rational combination of both molecular structures. Although the combination of both elements had a low effect in materials electron transport properties, high efficiencies around 24 cd/A were obtained for the series in all-organic solution-processed two layer devices.
In the last part, benzene and pyridine were chosen as the series electron-transport motif. By controlling the relative pyridine content (RPC) solubility into methanol was induced for polystyrenes with bulky side-chains. Materials with RPC ≥ 0.5 could be deposited orthogonally from solution without harming underlying layers. From the best of our knowledge, this is the first time such materials are applied in this architecture showing moderate efficiencies around 10 cd/A in all-organic solution processed OLEDs.
Overall, the outcome of these studies will actively contribute to the current research on materials for all-solution processed OLEDs.
Organische Halbleiter besitzen neue, bemerkenswerte Materialeigenschaften, die sie für die grundlegende Forschung wie auch aktuelle technologische Entwicklung (bsw. org. Leuchtdioden, org. Solarzellen) interessant werden lassen. Aufgrund der starken konformative Freiheit der konjugierten Polymerketten führt die Vielzahl der möglichen Anordnungen und die schwache intermolekulare Wechselwirkung für gewöhnlich zu geringer struktureller Ordnung im Festkörper. Die Morphologie hat gleichzeitig direkten Einfluss auf die elektronische Struktur der organischen Halbleiter, welches sich meistens in einer deutlichen Reduktion der Ladungsträgerbeweglichkeit gegenüber den anorganischen Verwandten zeigt. So stellt die Beweglichkeit der Ladungen im Halbleiter einen der limitierenden Faktoren für die Leistungsfähigkeit bzw. den Wirkungsgrad von funktionellen organischen Bauteilen dar. Im Jahr 2009 wurde ein neues auf Naphthalindiimid und Bithiophen basierendes Dornor/Akzeptor Copolymer vorgestellt [P(NDI2OD‑T2)], welches sich durch seine außergewöhnlich hohe Ladungsträgermobilität auszeichnet. In dieser Arbeit wird die Ladungsträgermobilität in P(NDI2OD‑T2) bestimmt, und der Transport durch eine geringe energetischer Unordnung charakterisiert. Obwohl dieses Material zunächst als amorph beschrieben wurde zeigt eine detaillierte Analyse der optischen Eigenschaften von P(NDI2OD‑T2), dass bereits in Lösung geordnete Vorstufen supramolekularer Strukturen (Aggregate) existieren. Quantenchemische Berechnungen belegen die beobachteten spektralen Änderungen. Mithilfe der NMR-Spektroskopie kann die Bildung der Aggregate unabhängig von optischer Spektroskopie bestätigt werden. Die Analytische Ultrazentrifugation an P(NDI2OD‑T2) Lösungen legt nahe, dass sich die Aggregation innerhalb der einzelnen Ketten unter Reduktion des hydrodynamischen Radius vollzieht. Die Ausbildung supramolekularen Strukturen nimmt auch eine signifikante Rolle bei der Filmbildung ein und verhindert gleichzeitig die Herstellung amorpher P(NDI2OD‑T2) Filme. Durch chemische Modifikation der P(NDI2OD‑T2)-Kette und verschiedener Prozessierungs-Methoden wurde eine Änderung des Kristallinitätsgrades und gleichzeitig der Orientierung der kristallinen Domänen erreicht und mittels Röntgenbeugung quantifiziert. In hochauflösenden Elektronenmikroskopie-Messungen werden die Netzebenen und deren Einbettung in die semikristallinen Strukturen direkt abgebildet. Aus der Kombination der verschiedenen Methoden erschließt sich ein Gesamtbild der Nah- und Fernordnung in P(NDI2OD‑T2). Über die Messung der Elektronenmobilität dieser Schichten wird die Anisotropie des Ladungstransports in den kristallographischen Raumrichtungen von P(NDI2OD‑T2) charakterisiert und die Bedeutung der intramolekularen Wechselwirkung für effizienten Ladungstransport herausgearbeitet. Gleichzeitig wird deutlich, wie die Verwendung von größeren und planaren funktionellen Gruppen zu höheren Ladungsträgermobilitäten führt, welche im Vergleich zu klassischen semikristallinen Polymeren weniger sensitiv auf die strukturelle Unordnung im Film sind.
We study buckling instabilities of filaments in biological systems. Filaments in a cell are the building blocks of the cytoskeleton. They are responsible for the mechanical stability of cells and play an important role in intracellular transport by molecular motors, which transport cargo such as organelles along cytoskeletal filaments. Filaments of the cytoskeleton are semiflexible polymers, i.e., their bending energy is comparable to the thermal energy such that they can be viewed as elastic rods on the nanometer scale, which exhibit pronounced thermal fluctuations. Like macroscopic elastic rods, filaments can undergo a mechanical buckling instability under a compressive load. In the first part of the thesis, we study how this buckling instability is affected by the pronounced thermal fluctuations of the filaments. In cells, compressive loads on filaments can be generated by molecular motors. This happens, for example, during cell division in the mitotic spindle. In the second part of the thesis, we investigate how the stochastic nature of such motor-generated forces influences the buckling behavior of filaments. In chapter 2 we review briefly the buckling instability problem of rods on the macroscopic scale and introduce an analytical model for buckling of filaments or elastic rods in two spatial dimensions in the presence of thermal fluctuations. We present an analytical treatment of the buckling instability in the presence of thermal fluctuations based on a renormalization-like procedure in terms of the non-linear sigma model where we integrate out short-wavelength fluctuations in order to obtain an effective theory for the mode of the longest wavelength governing the buckling instability. We calculate the resulting shift of the critical force by fluctuation effects and find that, in two spatial dimensions, thermal fluctuations increase this force. Furthermore, in the buckled state, thermal fluctuations lead to an increase in the mean projected length of the filament in the force direction. As a function of the contour length, the mean projected length exhibits a cusp at the buckling instability, which becomes rounded by thermal fluctuations. Our main result is the observation that a buckled filament is stretched by thermal fluctuations, i.e., its mean projected length in the direction of the applied force increases by thermal fluctuations. Our analytical results are confirmed by Monte Carlo simulations for buckling of semiflexible filaments in two spatial dimensions. We also perform Monte Carlo simulations in higher spatial dimensions and show that the increase in projected length by thermal fluctuations is less pronounced than in two dimensions and strongly depends on the choice of the boundary conditions. In the second part of this work, we present a model for buckling of semiflexible filaments under the action of molecular motors. We investigate a system in which a group of motors moves along a clamped filament carrying a second filament as a cargo. The cargo-filament is pushed against the wall and eventually buckles. The force-generating motors can stochastically unbind and rebind to the filament during the buckling process. We formulate a stochastic model of this system and calculate the mean first passage time for the unbinding of all linking motors which corresponds to the transition back to the unbuckled state of the cargo filament in a mean-field model. Our results show that for sufficiently short microtubules the movement of kinesin-I-motors is affected by the load force generated by the cargo filament. Our predictions could be tested in future experiments.
Ziel dieser Arbeit ist die phänomenologische Untersuchung der Feuchteempfindlichkeit der elektrischen Eigenschaften dünner Polymerschichten. Diese Untersuchungen stellen gleichzeitig Vorarbeiten zur Entwicklung von Prototypen von zwei polymeren Dünnschicht-Feuchtesensoren dar, die sich durch die spezielle Auswahl der feuchtesensitiven Materialien jeweils durch eine besondere Eigenschaft gegenüber kommerziellen Massenprodukten auszeichnen. Ziel der Entwicklungsarbeiten für den ersten Prototypen war die Konstruktion eines schnellen Feuchtesensors, der plötzliche und sprunghafte Feuchteänderungen in der umgebenden Atmosphäre möglichst rasch detektieren kann. Dafür wurden dünne Schichten von Poly-DADMAC auf Interdigitalstrukturen aufgebracht, die einen möglichst direkten Kontakt zwischen feuchtesensitiver Schicht und umgebender, feuchter Atmosphäre gewährleisten. Als Messgrößen dienten die Wechselstromgrößen Widerstand und Kapazität der Schichten. Die Feuchtekennlinien der Schichten zeigen gute Konstanz und hohe Reproduzierbarkeit. Der Widerstand der Schichten ändert sich durch den Einfluss von Feuchte je nach Schichtdicke um 3 bis 5 Größenordnungen und eignet sich als Messgröße für die Feuchtigkeit im gesamten Feuchtebereich. Die Hysterese der Filme konnte auf kleiner als 2,5% r.F. bestimmt werden, die Reproduzierbarkeit auf besser als 1% r.F. Die Ansprechzeit der Schichten lässt sich schichtdickenabhängig zu 1 bis 10 Sekunden bestimmen. Hierbei zeigen besonders die dünnen Schichten kurze Ansprechzeiten. Zielstellung für den zweiten Feuchtesensor war die Entwicklung eines Prototypen, dessen sensitive Schicht sich biostatisch und biozid verhält, so dass er in biotischen Umgebungen eingesetzt werden kann. Es wurden fünf Polysulfobetaine synthetisiert, deren Biozidität und Biostatik mit dem Kontakttest nach Rönnpagel, dem ISO846-Test und Abbautests bestimmt wurde. Zwei Polymere – Poly-DMMAAPS (BT2) und Poly-[MSA-Styren-Sulfobetain] (BT5) – erwiesen sich als ausreichend biozid und biostatisch. Schichten dieser Polymere wurden auf Interdigitalstrukturen aufgezogen, anschließend wurden die Kennlinien dieser Proben aufgenommen. Die Messwerte zeigen für beide Polymere gute Konstanz und eine hohe Reproduzierbarkeit. BT2-Proben sind zwischen 20% und 80% r.F. besonders empfindlich und zeigen über einen Monat keine Langzeitdrift. Vernetzte Proben zeigen bis 50°C keinen temperaturbedingten Abfall der Feuchteempfindlichkeit. Der Einsatz vernetzter BT5-Schichten als kapazitiver Feuchtesensor ist bis etwa 70°C möglich, die Schichten sind selbst nach Lagerung im Hochvakuum und mehrfacher Betauung stabil. Damit liegen zwei funktionsfähige Prototypen von Feuchtesensoren vor, für die die meisten Kennwerte denen von vergleichbaren kommerziellen Feuchtesensoren entsprechen. Gleichzeitig zeichnen sie sich aber durch eine sehr niedrige Ansprechzeit bzw. eine ausreichende Lebensdauer unter biotischen Bedingungen aus.
The biological function and the technological applications of semiflexible polymers, such as DNA, actin filaments and carbon nanotubes, strongly depend on their rigidity. Semiflexible polymers are characterized by their persistence length, the definition of which is the subject of the first part of this thesis. Attractive interactions, that arise e.g.~in the adsorption, the condensation and the bundling of filaments, can change the conformation of a semiflexible polymer. The conformation depends on the relative magnitude of the material parameters and can be influenced by them in a systematic manner. In particular, the morphologies of semiflexible polymer rings, such as circular nanotubes or DNA, which are adsorbed onto substrates with three types of structures, are studied: (i) A topographical channel, (ii) a chemically modified stripe and (iii) a periodic pattern of topographical steps. The results are compared with the condensation of rings by attractive interactions. Furthermore, the bundling of two individual actin filaments, whose ends are anchored, is analyzed. This system geometry is shown to provide a systematic and quantitative method to extract the magnitude of the attraction between the filaments from experimentally observable conformations of the filaments.
In the present work, we discuss two subjects related to the nonequilibrium dynamics of polymers or biological filaments adsorbed to two-dimensional substrates. The first part is dedicated to thermally activated dynamics of polymers on structured substrates in the presence or absence of a driving force. The structured substrate is represented by double-well or periodic potentials. We consider both homogeneous and point driving forces. Point-like driving forces can be realized in single molecule manipulation by atomic force microscopy tips. Uniform driving forces can be generated by hydrodynamic flow or by electric fields for charged polymers. In the second part, we consider collective filament motion in motility assays for motor proteins, where filaments glide over a motor-coated substrate. The model for the simulation of the filament dynamics contains interactive deformable filaments that move under the influence of forces from molecular motors and thermal noise. Motor tails are attached to the substrate and modeled as flexible polymers (entropic springs), motor heads perform a directed walk with a given force-velocity relation. We study the collective filament dynamics and pattern formation as a function of the motor and filament density, the force-velocity characteristics, the detachment rate of motor proteins and the filament interaction. In particular, the formation and statistics of filament patterns such as nematic ordering due to motor activity or clusters due to blocking effects are investigated. Our results are experimentally accessible and possible experimental realizations are discussed.
Es ist schon seit längerer Zeit bekannt, dass nach Kontakt des Biomaterials mit der biologischen Umgebung bei Implantation oder extrakorporaler Wechselwirkung zunächst Proteine aus dem umgebenden Milieu adsorbiert werden, wobei die Oberflächeneigenschaften des Materials die Zusammensetzung der Proteinschicht und die Konformation der darin enthaltenden Proteine determinieren. Die nachfolgende Wechselwirkung von Zellen mit dem Material wird deshalb i.d.R. von der Adsorbatschicht vermittelt. Der Einfluss der Oberflächen auf die Zusammensetzung und Konformation der Proteine und die nachfolgende Wechselwirkung mit Zellen ist von besonderem Interesse, da einerseits eine Aussage über die Anwendbarkeit ermöglicht wird, andererseits Erkenntnisse über diese Zusammenhänge für die Entwicklung neuer Materialien mit verbesserter Biokompatibilität genutzt werden können. In der vorliegenden Habilitationsschrift wurde deshalb der Einfluss der Zusammensetzung von Polymeren bzw. von deren Oberflächeneigenschaften auf die Adsorption von Proteinen, den Aktivitätszustand der plasmatischen Gerinnung und die Adhäsion von Zellen untersucht. Dabei wurden auch Möglichkeiten zur Beeinflussung dieser Vorgänge über eine Veränderung der Volumenzusammensetzung oder durch Oberflächenmodifikationen von Biomaterialien vorgestellt. Erkenntnisse aus diesen Arbeiten konnten für die Entwicklung von Membranen für Biohybrid-Organe genutzt werden.

The surface of biological cells consists of a lipid membrane and a large amount of various proteins and polymers, which are embedded in the membrane or attached to it. We investigate how membranes are influenced by polymers, which are anchored to the membrane by one end. The entropic pressure exerted by the polymer induces a curvature, which bends the membrane away from the polymer. The resulting membrane shape profile is a cone in the vicinity of the anchor segment and a catenoid far away from it. The perturbative calculations are confirmed by Monte-Carlo simulations. An additional attractive interaction between polymer and membrane reduces the entropically induced curvature. In the limit of strong adsorption, the polymer is localized directly on the membrane surface and does not induce any pressure, i.e. the membrane curvature vanishes. If the polymer is not anchored directly on the membrane surface, but in a non-vanishing anchoring distance, the membrane bends towards the polymer for strong adsorption. In the last part of the thesis, we study membranes under the influence of non-anchored polymers in solution. In the limit of pure steric interactions between the membrane and free polymers, the membrane curves towards the polymers (in contrast to the case of anchored polymers). In the limit of strong adsorption the membrane bends away from the polymers.
