Das 1817 erstmals schriftlich erwähnte Selen galt lange Zeit nur als toxisch und sogar als procancerogen, bis es 1957 von Schwarz und Foltz als essentielles Spurenelement erkannt wurde, dessen biologische Funktionen in Säugern durch Selenoproteine vermittelt werden. Die Familie der Glutathionperoxidasen nimmt hierbei eine wichtige Stellung ein. Für diese sind konkrete Funktionen und die dazugehörigen molekularen Mechanismen, welche über die von ihnen katalysierte Hydroperoxidreduktion und damit verbundene antioxidative Kapazität hinausgehen, bislang nur unzureichend beschrieben worden. Die Funktion der gastrointestinalen Glutathionperoxidase (GI-GPx) wird als Barriere gegen eine Hydroperoxidabsorption im Gastrointestinaltrakt definiert. Neuen Erkenntnissen zufolge wird die GI-GPx aber auch in verschiedenen Tumoren verstärkt exprimiert, was weitere, bis dato unbekannte, Funktionen dieses Enzymes wahrscheinlich macht. Um mögliche neue Funktionen der GI-GPx, vor allem während der Cancerogenese, abzuleiten, wurde hier die transkriptionale Regulation der GI-GPx detaillierter untersucht. Die Sequenzanalyse des humanen GI-GPx-Promotors ergab das Vorhandensein von zwei möglichen "antioxidant response elements" (ARE), bei welchen es sich um Erkennungssequenzen des Transkriptionsfaktors Nrf2 handelt. Die meisten der bekannten Nrf2-Zielgene gehören in die Gruppe der Phase-II-Enzyme und verfügen über antioxidative und/oder detoxifizierende Eigenschaften. Sowohl auf Promotorebene als auch auf mRNA- und Proteinebene konnte die Expression der GI-GPx durch typische, in der Nahrung enthaltene, Nrf2-Aktivatoren wie z.B. Sulforaphan oder Curcumin induziert werden. Eine direkte Beteiligung von Nrf2 wurde durch Cotransfektion von Nrf2 selbst bzw. von Keap1, das Nrf2 im Cytoplasma festhält, demonstriert. Somit konnte die GI-GPx eindeutig als Nrf2-Zielgen identifiziert werden. Ob sich die GI-GPx in die Gruppe der antiinflammatorischen und anticancerogenen Phase-II-Enzyme einordnen lässt, bleibt noch zu untersuchen. Die Phospholipidhydroperoxid Glutathionperoxidase (PHGPx) nimmt aufgrund ihres breiten Substratspektrums, ihrer hohen Lipophilie und ihrer Fähigkeit, Thiole zu modifizieren, eine Sonderstellung innerhalb der Familie der Glutathionperoxidasen ein. Mit Hilfe eines PHGPx-überexprimierenden Zellmodells wurden deshalb Beeinflussungen des zellulären Redoxstatus und daraus resultierende Veränderungen in der Aktivität redoxsensitiver Transkriptionsfaktorsysteme und in der Expression atheroskleroserelevanter Adhäsionsmoleküle untersucht. Als Transkriptionsfaktoren wurden NF-kB und Nrf2 ausgewählt. Die Bindung von NF-kB an sein entsprechendes responsives Element in der DNA erfordert das Vorhandensein freier Thiole, wohingegen Nrf2 durch Thiolmodifikation von Keap1 freigesetzt wird und in den Kern transloziert. Eine erhöhte Aktivität der PHGPx resultierte in einer Erhöhung des Verhältnisses von GSH zu GSSG, andererseits aber in einer verminderten Markierbarkeit freier Proteinthiole. PHGPx-Überexpression reduzierte die IL-1-induzierte NF-kB-Aktivität, die sich in einer verminderten NF-kB-DNA-Bindefähigkeit und Transaktivierungsaktivität ausdrückte. Auch war die Proliferationsrate der Zellen vermindert. Die Expression des NF-kB-regulierten vaskulären Zelladhäsionsmoleküls, VCAM-1, war ebenfalls deutlich verringert. Umgekehrt war in PHGPx-überexprimierenden Zellen eine erhöhte Nrf2-Aktivität und Expression der Nrf2-abhängigen Hämoxygenase-1 zu verzeichnen. Letzte kann für die meisten der beobachteten Effekte verantwortlich gemacht werden. Die hier dargestellten Ergebnisse verdeutlichen, dass eine Modifizierung von Proteinthiolen als wichtige Determinante für die Regulation der Expression und Funktion von Glutathionperoxidasen angesehen werden kann. Entgegen früheren Vermutungen, welche oxidative Vorgänge generell mit pathologischen Veränderungen assoziierten, scheint ein moderater oxidativer Stress, bedingt durch eine transiente Thiolmodifikation, durchaus günstige Auswirkungen zu haben, da, wie hier dargelegt, verschiedene, miteinander interagierende, cytoprotektive Mechanismen ausgelöst werden. Hieran wird deutlich, dass sich "antioxidative Wirkung" oder "oxidativer Stress" keineswegs nur auf "gute" oder "schlechte" Vorgänge beschränken lassen, sondern im Zusammenhang mit den beeinflussten (patho)physiologischen Prozessen und dem Ausmaß der "Störung" des physiologischen Redoxgleichgewichtes betrachtet werden müssen.
Das seit 1957 als essentiell klassifizierte Spurenelement Selen vermittelt seine Funktion hauptsächlich durch seinen Einbau in Selenoproteine in Form der 21. proteinogenen Aminosäure Selenocystein. Insgesamt wurden 25 humane Gene für Selenoproteine identifiziert, deren genaue Funktion häufig noch nicht bekannt ist. Selen ist das einzige Mitglied aus der Gruppe der Mikronährstoffe, für das nach wie vor eine antikanzerogene Funktion vor allem in Bezug auf Darmkrebs postuliert wird. Die Grundlage dafür liefert eine Interventionsstudie, bei der 1.312 Probanden für 4,5 Jahre mit 200 μg Selen/Tag supplementiert wurden. Dies resultierte in einer Senkung der Gesamtkrebsmortalität um 50 %. Die Fragen einer optimalen Selenzufuhr, die nicht nur den Bedarf deckt, sondern auch die Entfaltung der antikanzerogenen Wirkung von Selen gewährleistet und die zugrunde liegenden molekularen Mechanismen sind noch ungeklärt. Zudem liegt die Selenzufuhr bei einem Großteil der europäischen Bevölkerung unter den Empfehlungen. Deshalb wurden in der vorliegenden Arbeit vier Wochen alte Mäuse für sechs Wochen marginal defizient (0,086 mg/kg Futter) bzw. selenadäquat (0,15 mg/kg Futter) gefüttert. Dieser geringe Unterschied im Selengehalt resultierte in einer Senkung des Plasmaselenspiegels der selenarmen Tiere auf 13 % und der GPx-Aktivität in der Leber auf 35 %. Zunächst wurde der Einfluss von Selen auf die globale Genexpression im murinen Colon mittels Microarray untersucht. Von den im Colon exprimierten Selenoproteinen reagierte die mRNA von SelW, SelH, GPx1 und SelM im Selenmangel besonders deutlich mit Expressionsverlust. Da diese Selenoproteine nicht nur im Colon, sondern auch in Leukozyten reguliert waren, sind sie auch als humane Biomarker für die in dieser Studie gewählte Schwankung des Selengehalts geeignet. Des Weiteren wurde auf Basis der Microarraydaten eine Signalweganalyse durchgeführt, die der Identifizierung krebsrelevanter Signalwege diente, um mögliche molekularbiologische Erklärungsansätze für die Rolle von Selen im Krebsgeschehen zu finden. Es zeigte sich, dass die mRNA von Schlüsselgenen des Wnt-Signalwegs wie β-Catenin, Gsk3β, Dvl2, Tle2, Lef1 und c-Myc auf Schwankungen des Selengehalts reagiert. Vor allem die Induktion von c-Myc, einem Zielgen des Wnt-Signalwegs, deutet darauf hin, dass dieser im Selenmangel tatsächlich aktiver ist als bei selenadäquater Versorgung. Ein weiterer möglicher Erklärungsansatz für die postulierte präventive Funktion von Selen gegenüber Darmkrebs ist die gastrointestinale Glutathionperoxidase (GPx2), die physiologisch in den proliferierenden Zellen des Kryptengrunds exprimiert wird. Die Regulation dieses Enzyms durch den Wnt-Signalweg, der ebenfalls in proliferierenden Zellen aktiv ist, konnte mittels Reportergenanalyse und endogen auf mRNA- und Proteinebene in Zellkultur gezeigt werden. Die Aktivierung verkürzter Promotorkonstrukte und die Mutation eines potentiellen Bindeelements identifizierten den für die Bindung von TCF und β-Catenin verantwortlichen Bereich. Als Zielgen des Wnt-Signalwegs scheint GPx2 zu den an Proliferationsprozessen beteiligten Genen zu gehören, was unter physiologischen Bedingungen die Aufrechterhaltung des intestinalen Epithels gewährleistet. Bei der Entstehung intestinaler Tumore, die in der Initiationsphase zu über 90 % mit einer konstitutiven Aktivierung des Wnt-Signalwegs einhergeht, wirkt GPx2 möglicherweise prokanzerogen. Die genaue Funktion von GPx2 während der Kanzerogenese bleibt weiter zu untersuchen.
Das ITC SFN und der Mikronährstoff Se sind bekannt als chemopräventive Inhaltsstoffe von Gemüse der Brassica-Familie, welcher auch Brokkoli angehört. Die Wirkungen von sowohl SFN als auch Se beruhen auf zahlreichen verschiedenen Mechanismen. Es existieren jedoch Schnittstellen, an welchen Interaktionen beider Substanzen möglich sind. Basierend auf diesem Wissen wurden in dieser Arbeit Wechselwirkungen zwischen SFN und Se auf die Aktivität sowie Expression von Phase II Enzymen und Selenoproteinen untersucht. Der Einfluss der Kombination von SFN und Se auf die unter physiologischen Bedingungen stattfindende Proliferation und Apoptose war ebenso Gegenstand der Arbeit wie die Modulation von Entzündungsprozessen sowie der Tumorentstehung während der entzündungsverstärkten Colonkanzerogenese im Mausmodell. Das hinsichtlich seiner Wirksamkeit mit aus GRA hydrolysiertem SFN zunächst als vergleichbar befundene synthetische SFN wurde für die Untersuchung im AOM/DSS-induzierten Colontumormodell gewählt und in Kombination mit 3 verschiedenen Selendiäten verabreicht. Der Einfluss von SFN und Se auf Phase II Enzyme und Selenoproteine entlang des GIT war organabhängig und nach 4 Wochen geringer als nach 7 Tagen. Die schwächere Induktion deutet auf eine Anpassung des Organismus hin. Ein SFN-vermittelter Effekt auf NQO1 war im Selenmangel am deutlichsten. Die Aktivität des Selenoproteins TrxR wurde hingegen erst bei ausreichender Selenversorgung durch SFN beeinflusst. Die als Nrf2-Zielgen bekannte und in der Hierarchie der Selenoproteine einen hohen Rang einnehmende GPx2 konnte in bestimmten Organen bereits unter selenarmen Bedingungen durch SFN induziert werden. Eine Überexpression des Enzyms war jedoch nicht möglich. SFN steigerte, unabhängig vom Selenstatus, im oberen Abschnitt des GIT und im Colon die Aktivität der GST. Eine Induktion des eigenen Metabolismus wäre somit denkbar. Im Falle eines Mangels an GPx2 wurde GPx1 bei hinreichender Selenversorgung stärker exprimiert, allerdings konnte sie die Funktion von GPx2 nicht völlig erset-zen. Im Selenmangel kann die Aktivitätssteigerung der TrxR im Dünndarm, dem Ab-schnitt der Selenabsorption, als ein Versuch der GPx2-Kompensation angesehen werden. SFN war nicht in der Lage, über eine Aktivierung des Nrf2/ARE-Signalweges kompensatorische Effekte zu induzieren. Apoptotische Prozesse wurden unter physiologischen Bedingungen nur marginal durch SFN und Se moduliert. Das elektrophile ITC konnte lediglich im Selenmangel Apoptose im luminalen Bereich der Colonkrypten induzieren. Die durch supranutritive Selenkonzentration induzierte Apoptose im Kryptengrund wurde nicht durch SFN beeinflusst. Einer bei Abwesenheit der GPx2 erhöhten Apoptoserate im Kryptengrund wirkte SFN bei adäquater Selenversorgung entgegen, war indessen proapoptotisch unter selendefizienten Konditionen. Der Einfluss von SFN auf die Entzündung war deutlich abhängig vom Selenstatus. Während SFN im Selenmangel anscheinend prooxidative Prozesse induzierte und die Entzündungssymptome verschlimmerte, wirkte es unter adäquatem Selenstatus an-tiinflammatorisch. Den vergleichsweise milden Grad der Entzündung im selensupplementierten Status konnte SFN nicht zusätzlich beeinflussen. SFN veränderte die Inzi-denz colorektaler Tumore nicht. Ein, die Tumorinitiation blockierender SFN-Effekt durch direkte Hemmung der metabolischen Aktivierung des Prokanzerogens im selenadäquaten Zustand scheint offensichtlich. Eine Überversorgung mit Se kann protektiv im Hinblick auf Entzündung oder Colonkanzerogenese sein, jedoch bewirkt SFN keinen zusätzlichen Schutz. Kombinationseffekte von SFN und Se in Bezug auf Phase II Enzyme, Selenoproteine und Apoptose sowie die entzündungsverstärkte Colonkanzerogenese sind nicht eindeutiger Natur und können, abhängig vom Endpunkt, synergistische oder antagonistische Züge aufweisen. Eine bei Selendefizienz deutlichere Wirkung von SFN kann mit Hilfe der gesteigerten Aktivierung von Nrf2 erklärt werden, dies muss jedoch nicht von Vorteil sein. Bei adäquater Selenversorgung kann SFN kurzfristig antiinflammatorische und antikanzerogene Prozesse induzieren. Von einer längerfristigen ständigen SFN-Aufnahme in Form von GRA-reichen Brassicacea ist jedoch abzuraten, da von einer Adaptation auszugehen ist. Die Wirkung von SFN innerhalb der komplexen Pflanzenmatrix bleibt Gegenstand zukünftiger Untersuchungen.
Bei der Entdeckung der Glutathionperoxidase-2 (GPx2) wurde zunächst davon ausgegangen, dass die Funktion dieses Enzyms im Kryptengrund des Colons einzig in der Reduktion von H2O2 besteht. Im Laufe der weiteren Erforschung zeigte sich, dass GPx2 auch in verschiedenen Tumorgeweben vermehrt exprimiert wird. Dabei wird diskutiert, ob die Wirkung von GPx2 im Tumor eher als pro- oder als antikarzinogen einzustufen ist. Mehrere Experimente in vitro und in vivo zeigten antiinflammatorische Eigenschaften der GPx2. Aufgrund dieser Befunde wird derzeit über weitere Funktionen der GPx2 spekuliert. In dieser Arbeit wurde die physiologische Funktion von GPx2 näher erforscht, dazu wurden Wildtyp- und GPx2-Knockout-Mäuse in Hinblick auf Veränderungen der Enzymexpression und der Colonmorphologie untersucht. Es wurden drei verschiedene Selendiäten verfüttert: selenarmes, selenadäquates und selensupplementiertes Futter. Unter physiologischen Bedingungen ist am Kryptengrund des Colons, innerhalb der proliferierenden Zone, die Mitoserate am höchsten. Der Großteil der apoptotischen Zellen ist hingegen an der Kryptenspitze vorzufinden. Durch den Knockout von GPx2 kam es zu einer signifikanten Erhöhung der Apoptoserate am Kryptengrund. Dabei war der größte Effekt auf selenarmem Futter zu verzeichnen. Hierbei wurde sogar eine Veränderung der Colonmorphologie dokumentiert, da die Verschiebung der Proliferationszone in Richtung Kryptenspitze eine Verlängerung der Krypten nach sich zog. Im Wildtyp wurden keine Apoptosen im Kryptengrund detektiert. GPx1 wird unter physiologischen Bedingungen im Gegensatz zur GPx2 in der Kryptenspitze exprimiert und ist im Selenmangel nicht mehr detektierbar. Der Knockout von GPx2 erhöhte die GPx1-Expression im Kryptengrund auf allen drei Selendiäten. Diese Überexpression von GPx1 am Kryptengrund soll vermutlich den Verlust von GPx2 an dieser Stelle kompensieren. Da jedoch dort die massive Apoptoserate detektiert wurde, kann die GPx1 nicht die komplette Funktion von GPx2 kompensieren. Diese Ergebnisse deuten darauf hin, dass die Funktion von GPx2 nicht nur in der Reduktion von H2O2 liegt. Vielmehr kann eine Rolle bei der Aufrechterhaltung der Homöostase von Zellen postuliert werden. Ein weiterer Bestandteil dieser Arbeit war die Klärung der Frage, welchen Einfluss GPx2 auf die entzündungsassoziierte Colonkarzinogenese ausübt. In dem hierfür verwendeten AOM/DSS-Model wird der karzinogene Prozess durch Entzündung vorangetrieben. Es erfolgte sowohl im Wildtyp als auch im GPx2-Knockout zum einen die Bewertung des Entzündungsstatus des Colons und zum anderen wurde die Anzahl von ACF und Tumoren verglichen. Das Colon im GPx2-Knockout war wesentlich stärker entzündet als im Wildtyp. Diese Ergebnisse bestätigen die für die GPx2 postulierte antiinflammatorische Funktion. Normalerweise führt eine Erhöhung der Mitoseanzahl zur Regeneration des entzündeten Gewebes. Jedoch beeinflusst der Verlust von GPx2 vermutlich den Ablauf der Entzündung, indem beispielsweise die Regeneration des Gewebes durch die enorm hohe Apoptoserate am Kryptengrund verlangsamt wird. Des Weiteren hatten sich im GPx2-Knockout tendenziell mehr Tumore entwickelt. Somit korrelierte die Entzündung des Colons mit der Entwicklung von Tumoren. Der Verlust von GPx2 begünstigte vermutlich sowohl die Tumorinitiation als auch die Tumorprogression. Allerdings stimulierte die Expression von GPx2 ebenfalls das Tumorwachstum. Es kann geschlussfolgert werden, dass eine adäquate GPx2-Expression vor Entzündung schützt und somit das Risiko für Colonkrebs senkt. Ob GPx2 aber insgesamt pro- oder antikarzinogen wirkt, hängt vermutlich vom Stadium des Colonkarzinogenese ab.
Das Selenoprotein Glutathionperoxidase 2 (GPx2) ist ein epithelzellspezifisches, Hydroperoxide-reduzierendes Enzym, welches im Darmepithel, vor allem in den proliferierenden Zellen des Kryptengrundes, exprimiert wird. Die Aufrechterhaltung der GPx2-Expression im Kryptengrund auch bei subadäquatem Selenstatus könnte darauf hinweisen, dass sie hier besonders wichtige Funktionen wahrnimmt. Tatsächlich weisen GPx2 knockout (KO)-Mäuse eine erhöhte Apoptoserate im Kryptengrund auf. Ein Ziel dieser Arbeit war es deshalb, die physiologische Funktion der GPx2 näher zu untersuchen. In Kryptengrundepithelzellen aus dem Colon selenarmer GPx2 KO-Mäuse wurde eine erhöhte Caspase 3/7-Aktivität im Vergleich zum Wildtyp (WT) festgestellt. Zudem wiesen diese Zellen eine erhöhte Suszeptibilität für oxidativen Stress auf. Die GPx2 gewährleistet also den Schutz der proliferierenden Zellen des Kryptengrundes auch bei subadäquater Selenversorgung. Des Weiteren wurde im Colon selenarmer (-Se) und -adäquater (+Se) GPx2 KO-Mäuse im Vergleich zum WT eine erhöhte Tumornekrosefaktor α-Expression und eine erhöhte Infiltration von Makrophagen festgestellt. Durch Fütterung einer selensupplementierten Diät (++Se) konnte dies verhindert werden. In GPx2 KO-Mäusen liegt demnach bereits basal eine niedriggradige Entzündung vor. Dies unterstreicht, dass GPx2 vor allem eine wichtige antiinflammatorische Funktion im Darmepithel besitzt. Dem Mikronährstoff Selen werden protektive Funktionen in der Colonkanzerogenese zugeschrieben. In einem Mausmodell der Colitis-assoziierten Colonkanzerogenese wirkte GPx2 antiinflammatorisch und hemmte so die Tumorentstehung. Auf der anderen Seite wurden jedoch auch prokanzerogene Eigenschaften der GPx2 aufgedeckt. Deshalb sollte in dieser Arbeit untersucht werden, welchen Effekt ein GPx2 knockout in einem Modell der sporadischen, durch Azoxymethan (AOM) induzierten, Colonkanzerogenese hat. Im WT kam es in präneoplastischen Läsionen häufig zu einer erhöhten GPx2-Expression im Vergleich zur normalen Darmmucosa. Eine derartige Steigerung der GPx2-Expression wurde auch in der humanen Colonkanzerogenese beschrieben. Das Fehlen der GPx2 resultierte in einer verminderten Entstehung von Tumoren (-Se und ++Se) und präneoplastischen Läsionen (-Se und +Se). Somit förderte GPx2 die Tumorentstehung im AOM-Modell. Acht Stunden nach AOM-Gabe war im GPx2 KO-Colon im Vergleich zum WT eine erhöhte Apoptoserate in der Kryptenmitte (-Se, +Se), nicht jedoch im Kryptengrund oder in der ++Se-Gruppe zu beobachten. Möglicherweise wirkte GPx2 prokanzerogen, indem sie die effiziente Elimination geschädigter Zellen in der Tumorinitiationsphase verhinderte. Eine ähnliche Wirkung wäre auch durch die erhöhte GPx2-Expression in der Promotionsphase denkbar. So könnte GPx2 proliferierende präneoplastische Zellen vor oxidativem Stress, Apoptosen, oder auch der Antitumorimmunität schützen. Dies könnte durch ein Zusammenwirken mit anderen Selenoproteinen wie GPx1 und Thioredoxinreduktasen, für die ebenfalls auch prokanzerogene Funktionen beschrieben wurden, verstärkt werden. Eine wichtige Rolle könnte hier die Modulation des Redoxstatus in Tumorzellen spielen. Die Variation des Selengehalts der Diät hatte im WT einen eher U-förmigen Effekt. So traten in der –Se und ++Se-Gruppe tendenziell mehr und größere Tumore auf, als in der +Se Gruppe. Zusammenfassend schützt GPx2 also die proliferierenden Zellen des Kryptengrundes. Sie könnte jedoch auch proliferierende transformierte Zellen schützen und so die sporadische, AOM-induzierte Colonkanzerogenese fördern. In einem Modell der Colitis-assoziierten Colonkanzerogenese hatte GPx2 auf Grund ihrer antiinflammatorischen Wirkung einen gegenteiligen Effekt und hemmte die Tumorentstehung. Die Rolle der GPx2 in der Colonkanzerogenese ist also abhängig vom zugrunde liegenden Mechanismus und wird maßgeblich von der Beteiligung einer Entzündung bestimmt.
Kolorektalkrebs (CRC) ist die dritthäufigste Tumorerkrankung weltweit. Neben dem Alter spielt auch die Ernährung eine wichtige Rolle bei der Entstehung der Krankheit. Eine vermutlich krebspräventive Wirkung wird dabei dem Spurenelement Selen zugeschrieben, das fast ausschließlich über Lebensmittel aufgenommen wird. So hängt beispielsweise ein niedriger Selenstatus mit dem Risiko, im Laufe des Lebens an CRC zu erkranken, zusammen. Seine Funktionen vermittelt Selen dabei überwiegend durch Selenoproteine, in denen es in Form von Selenocystein eingebaut wird. Zu den bisher am besten untersuchten Selenoproteinen mit möglicher Funktion während CRC zählen die Glutathionperoxidasen (GPXen). Die Mitglieder dieser Familie tragen aufgrund ihrer Hydroperoxid-reduzierenden Eigenschaften entscheidend zum Schutz der Zellen vor oxidativem Stress bei. Dies kann je nach Art und Stadium des Tumors entweder krebshemmend oder -fördernd wirken, da auch transformierte Zellen von dieser Schutzfunktion profitieren.
In dieser Arbeit wurde die GPX2 in HT29-Darmkrebszellen mithilfe stabil-transfizierter shRNA herunterreguliert, um die Funktion des Enzyms vor allem in Hinblick auf regulierte Signalwege zu untersuchen. Ein Knockdowns (KD) der strukturell ähnlichen GPX1 kam ebenfalls zum Einsatz, um gezielt Isoform-spezifische Funktionen unterscheiden zu können. Anhand eines PCR-Arrays wurden Signalwege identifiziert, die auf einen Einfluss der beiden Proteine im Zellwachstum hindeuteten. Anschließende Untersuchungen ließen auf einen verminderten Differenzierungsstatus in den GPX1- und GPX2-KDs aufgrund einer geringeren Aktivität der Alkalischen Phosphatase schließen. Zudem war die Zellviabilität im Neutralrot-Assay (NRU) bei Fehlen der GPX1 bzw. GPX2 im Vergleich zur Kontrolle reduziert. Die Ergebnisse des PCR-Arrays, und speziell für die GPX2 frühere Untersuchungen der Arbeitsgruppe, wiesen weiterhin auf eine Rolle der beiden Proteine in der entzündungsgetriebenen Karzinogenese hin. Daher wurden auch mögliche Interaktionen mit dem NFκB-Signalweg analysiert. Eine Stimulation der Zellen mit dem proinflammatorischen Zytokin IL1β ging mit einer verstärkten Aktivierung der MAP-Kinasen ERK1/2 in den Zellen mit GPX1- bzw. GPX2-KD einher. Die gleichzeitige Behandlung mit dem Antioxidans NAC führte nicht zur Rücknahme der Effekte in den KDs, sodass möglicherweise nicht nur die antioxidativen Eigenschaften der Enzyme bei der Interaktion mit diesen Signalwegsproteinen relevant sind.
Weiterhin wurden Analysen zum Substratspektrum der GPX2 in HCT116-Zellen mit einer Überexpression des Proteins durchgeführt. Dabei zeigte sich mittels NRU-Assay und DNA-Laddering, dass die GPX2 besonders vor den proapoptotischen Effekten einer Behandlung mit den Lipidhydroperoxiden HPODE und HPETE schützt.
Im Gegensatz zur GPX2 lässt sich Selenoprotein H (SELENOH) stärker durch die alimentäre Selenzufuhr beeinflussen. Einer möglichen Nutzung als Biomarker oder gar als Ansatzpunkt bei der Prävention bzw. Behandlung von CRC steht allerdings unvollständiges Wissen über die Funktion des Proteins gegenüber. Zur genaueren Charakterisierung von SELENOH wurden daher stabil-transfizierte KD-Klone in HT29- und Caco2-Zellen hergestellt und zunächst auf ihre Tumorigenität untersucht.
Zellen mit SELENOH-KD bildeten mehr und größere Kolonien im Soft Agar und zeigten ein erhöhtes Proliferations- und Migrationspotenzial im Vergleich zur Kontrolle.
Ein Xenograft in Nacktmäusen resultierte zudem in einer stärkeren Tumorbildung nach Injektion von KD-Zellen. Untersuchungen zur Beteiligung von SELENOH an der Zellzyklusregulation deuten auf eine hemmende Rolle des Proteins in der G1/S-Phase hin.
Die weiterhin beobachtete Hochregulation von SELENOH in humanen Adenokarzinomen und präkanzerösem Mausgewebe lässt sich möglicherweise mit der postulierten Schutzfunktion vor oxidativen Zell- und DNA-Schäden erklären. In gesunden Darmepithelzellen war das Protein vorrangig am Kryptengrund lokalisiert, was zu einer potenziellen Rolle während der gastrointestinalen Differenzierung passt.
Selen und Jod sind essenzielle Spurenelemente, die gemeinsam für eine optimale Funktionstüchtigkeit der Schilddrüse erforderlich sind. Der Mangel eines oder beider Elemente führt zu Verschiebungen auf Ebene der Schilddrüsenhormonproduktion mit weitreichenden Konsequenzen für Stoffwechselprozesse, neurologische Entwicklung und Erkrankungen. Auch bei Autoimmunerkrankungen der Schilddrüse spielt die Versorgung mit Jod und Selen eine wichtige Rolle. Als Biomarker für den Selenstatus eignet sich der Gehalt des Gesamtselens oder der des Selenoproteins P im Serum. Zur Bestimmung des Jodstatus wird in der Regel der Jodgehalt im Urin herangezogen. Um den Versorgungszustand an diesen und vier weiteren essenziellen Spurenelementen besser zu erfassen, charakterisiert die Forschungsgruppe TraceAge alters- und geschlechtsspezifische Spurenelementprofile und neue funktionelle Biomarker der einzelnen Spurenelemente. Außerdem sollen Interaktionen weiterer Spurenelemente genauer untersucht werden.
Selenium (Se) is an essential trace element that is ubiquitously present in the environment in small concentrations. Essential functions of Se in the human body are manifested through the wide range of proteins, containing selenocysteine as their active center. Such proteins are called selenoproteins which are found in multiple physiological processes like antioxidative defense and the regulation of thyroid hormone functions. Therefore, Se deficiency is known to cause a broad spectrum of physiological impairments, especially in endemic regions with low Se content. Nevertheless, being an essential trace element, Se could exhibit toxic effects, if its intake exceeds tolerable levels. Accordingly, this range between deficiency and overexposure represents optimal Se supply. However, this range was found to be narrower than for any other essential trace element. Together with significantly varying Se concentrations in soil and the presence of specific bioaccumulation factors, this represents a noticeable difficulty in the assessment of Se
epidemiological status. While Se is acting in the body through multiple selenoproteins, its intake occurs mainly in form of small organic or inorganic molecular mass species. Thus, Se exposure not only depends on daily intake but also on the respective chemical form, in which it is present.
The essential functions of selenium have been known for a long time and its primary forms in different food sources have been described. Nevertheless, analytical capabilities for a comprehensive investigation of Se species and their derivatives have been introduced only in the last decades. A new Se compound was identified in 2010 in the blood and tissues of bluefin tuna. It was called selenoneine (SeN) since it is an isologue of naturally occurring antioxidant ergothioneine (ET), where Se replaces sulfur. In the following years, SeN was identified in a number of edible fish species and attracted attention as a new dietary Se source and potentially strong antioxidant. Studies in populations whose diet largely relies on fish revealed that SeN
represents the main non-protein bound Se pool in their blood. First studies, conducted with enriched fish extracts, already demonstrated the high antioxidative potential of SeN and its possible function in the detoxification of methylmercury in fish. Cell culture studies demonstrated, that SeN can utilize the same transporter as ergothioneine, and SeN metabolite was found in human urine.
Until recently, studies on SeN properties were severely limited due to the lack of ways to obtain the pure compound. As a predisposition to this work was firstly a successful approach to SeN synthesis in the University of Graz, utilizing genetically modified yeasts. In the current study, by use of HepG2 liver carcinoma cells, it was demonstrated, that SeN does not cause toxic effectsup to 100 μM concentration in hepatocytes. Uptake experiments showed that SeN is not bioavailable to the used liver cells.
In the next part a blood-brain barrier (BBB) model, based on capillary endothelial cells from the porcine brain, was used to describe the possible transfer of SeN into the central nervous system (CNS). The assessment of toxicity markers in these endothelial cells and monitoring of barrier conditions during transfer experiments demonstrated the absence of toxic effects from SeN on the BBB endothelium up to 100 μM concentration. Transfer data for SeN showed slow but substantial transfer. A statistically significant increase was observed after 48 hours following SeN incubation from the blood-facing side of the barrier. However, an increase in Se content was clearly visible already after 6 hours of incubation with 1 μM of SeN. While the transfer rate of SeN after application of 0.1 μM dose was very close to that for 1 μM, incubation with 10 μM of SeN resulted in a significantly decreased transfer rate. Double-sided application of SeN caused no side-specific transfer of SeN, thus suggesting a passive diffusion mechanism of SeN across the BBB. This data is in accordance with animal studies, where ET accumulation was observed in the rat brain, even though rat BBB does not have the primary ET transporter – OCTN1. Investigation of capillary endothelial cell monolayers after incubation with SeN and reference selenium compounds showed no significant increase of intracellular selenium concentration. Speciesspecific Se measurements in medium samples from apical and basolateral compartments, as good as in cell lysates, showed no SeN metabolization. Therefore, it can be concluded that SeN may reach the brain without significant transformation.
As the third part of this work, the assessment of SeN antioxidant properties was performed in Caco-2 human colorectal adenocarcinoma cells. Previous studies demonstrated that the intestinal epithelium is able to actively transport SeN from the intestinal lumen to the blood side and accumulate SeN. Further investigation within current work showed a much higher antioxidant potential of SeN compared to ET. The radical scavenging activity after incubation with SeN was close to the one observed for selenite and selenomethionine. However, the SeN effect on the viability of intestinal cells under oxidative conditions was close to the one caused by ET. To answer the question if SeN is able to be used as a dietary Se source and induce the activity of selenoproteins, the activity of glutathione peroxidase (GPx) and the secretion of selenoprotein P (SelenoP) were measured in Caco-2 cells, additionally. As expected, reference selenium compounds selenite and selenomethionine caused efficient induction of GPx activity. In contrast to those SeN had no effect on GPx activity. To examine the possibility of SeN being embedded into the selenoproteome, SelenoP was measured in a culture medium. Even though Caco-2 cells effectively take up SeN in quantities much higher than selenite or selenomethionine, no secretion of SelenoP was observed after SeN incubation.
Summarizing, we can conclude that SeN can hardly serve as a Se source for selenoprotein synthesis. However, SeN exhibit strong antioxidative properties, which appear when sulfur in ET is exchanged by Se. Therefore, SeN is of particular interest for research not as part of Se metabolism, but important endemic dietary antioxidant.
The trace elements, selenium (Se) and copper (Cu) play an important role in maintaining normal brain function. Since they have essential functions as cofactors of enzymes or structural components of proteins, an optimal supply as well as a well-defined homeostatic regulation are crucial. Disturbances in trace element homeostasis affect the health status and contribute to the incidence and severity of various diseases. The brain in particular is vulnerable to oxidative stress due to its extensive oxygen consumption and high energy turnover, among other factors. As components of a number of antioxidant enzymes, both elements are involved in redox homeostasis. However, high concentrations are also associated with the occurrence of oxidative stress, which can induce cellular damage. Especially high Cu concentrations in some brain areas are associated with the development and progression of neurodegenerative diseases such as Alzheimer's disease (AD). In contrast, reduced Se levels were measured in brains of AD patients. The opposing behavior of Cu and Se renders the study of these two trace elements as well as the interactions between them being particularly relevant and addressed in this work.
