Die Honigbiene Apis mellifera zeigt innerhalb einer Kolonie eine an das Alter gekoppelte Arbeitsteilung. Junge Honigbienen versorgen die Brut (Ammenbienen), während ältere Honigbienen (Sammlerinnen) außerhalb des Stocks Pollen und Nektar eintragen. Die biogenen Amine Octopamin und Tyramin sind an der Steuerung der Arbeitsteilung maßgeblich beteiligt. Sie interagieren mit Zielzellen über die Bindung an G Protein gekoppelte Rezeptoren. A. mellifera besitzt fünf charakterisierte Octopaminrezeptoren (AmOctαR1, AmOctβR1-4), einen charakterisierten Tyraminrezeptor (AmTyr1) sowie einen weiteren putativen Tyraminrezeptor.
In der vorliegenden Arbeit wurde dieser putative Aminrezeptor als zweiter Tyraminrezeptor (AmTyr2) identifiziert, lokalisiert und pharmakologisch charakterisiert.
Die von der cDNA abgeleitete Aminosäuresequenz weist strukturelle Eigenschaften und konservierte Motive von G Protein gekoppelten Rezeptoren auf. Phylogenetisch ordnet sich der AmTyr2 Rezeptor bei den Tyramin 2 Rezeptoren anderer Insekten ein. Die funktionelle und pharmakologische Charakterisierung des putativen Tyraminrezeptors erfolgte in modifizierten HEK293 Zellen, die mit der Rezeptor cDNA transfiziert wurden. Die Applikation von Tyramin aktiviert Adenylylcyclasen in diesen Zellen und resultiert in einem Anstieg des intrazellulären cAMP Gehalts. Der AmTyr2 Rezeptor kann durch Tyramin in nanomolaren Konzentrationen halbmaximal aktiviert werden. Während es sich bei Octopamin um einen wirkungsvollen Agonisten des Rezeptors handelt, sind Mianserin und Yohimbin effektive Antagonisten. Für die Lokalisierung des Rezeptorproteins wurde ein polyklonaler Antikörper generiert. Eine AmTyr2-ähnliche Immunreaktivität zeigt sich im Gehirn in den optischen Loben, den Antennalloben, dem Zentralkomplex und in den Kenyon Zellen der Pilzkörper.
Des Weiteren wurde die Rolle der Octopamin- und Tyraminrezeptoren bei der Steuerung der altersabhängigen Arbeitsteilung analysiert.
Die Genexpression des AmOctαR1 in verschiedenen Gehirnteilen korreliert unabhängig vom Alter mit der sozialen Rolle, während sich die Genexpression von AmOctβR3/4 und den Tyraminrezeptoren AmTyr1 und AmTyr2 maximal mit dem Alter aber nicht der sozialen Rolle ändert. Sammlerinnen weisen einen höheren Octopamingehalt im Gesamtgehirn auf als Ammenbienen; bei Tyramin zeigen sich keine Unterschiede. Während Tyramin offensichtlich keine direkte Rolle spielt, werden durch Octopamin gesteuerte Prozesse der altersabhängigen Arbeitsteilung bei der Honigbiene vermutlich über den AmOctαR1 vermittelt.
Die Ergebnisse der vorliegenden Arbeit zeigen die wichtige Rolle von biogenen Aminen, insbesondere Octopamin bei der sozialen Organisation von Insektenstaaten.
Die acinösen Speicheldrüsen der Schabe Periplaneta americana sind reich durch serotonerge, dopaminerge und GABAerge Fasern innerviert. Die biogenen Amine Serotonin (5-HT) und Dopamin (DA) induzieren die Sekretion eines NaCl-haltigen Primärspeichels. Die physiologische Rolle der GABAergen Innervation des Drüsenkomplexes war bislang unbekannt. Weiterhin wurde vermutet, dass Tyramin (TA) und Octopamin (OA) an der Speichelbildung beteiligt sind. Mittels intrazellulärer Ableitungen von sekretorischen Acinuszellen mit und ohne Stimulierung des Speicheldrüsennervs (SDN) sollte daher die Wirkung von GABA, TA und OA im Speicheldrüsenkomplex untersucht werden. Intrazelluläre Ableitungen aus Acinuszellen zeigten, dass sowohl DA als auch 5 HT biphasische Änderungen des Membranpotentials induzierten. Diese bestanden aus einer initialen Hyperpolarisation und einer darauf folgenden transienten Depolarisation. Stimulierung des SDN mittels einer Saugelektrode verursachte ebenfalls biphasische Änderungen des Membranpotentials der Acinuszellen, die mit den DA- bzw. 5-HT-induzierten Änderungen kinetisch identisch waren. Dieses Ergebnis zeigte, dass die elektrische Stimulierung des SDN im Nerv-Speicheldrüsenpräparat eine verlässliche Methode zur Untersuchung der Wirkungen von Neuromodulatoren auf die dopaminerge und/oder sertotonerge Neurotransmission ist. Die Hyperpolarisation der DA-induzierten Potentialänderungen wurde durch eine intrazelluläre Ca2+-Freisetzung und die Öffnung basolateral lokalisierter Ca2+-gesteuerter K+-Kanäle verur-sacht. Die DA- und 5-HT-induzierte Depolarisation hing kritisch von der Aktivität eines basolateral lokalisierten Na+-K+-2Cl--Symporters ab. GABA, TA und OA potenzierten die elektrischen Antworten der Acinuszellen, wenn diese durch SDN-Stimulierung hervorgerufen wurden. Dabei war OA wirksamer als TA. Dieses Ergebnis zeigte, dass diese Substanzen als im Drüsenkomplex präsynaptisch und erregend als Neuromodulatoren wirken. Pharmakologische Untersuchungen ergaben, dass die erregende Wirkung von GABA durch einen G-Protein-gekoppelten GABAB-Rezeptor vermittelt wurde. Messungen der durch SDN-Stimulierung induzierten Flüssigkeits- und Proteinsekretionsraten zeigten, dass beide Parameter in Anwesenheit von GABA verstärkt waren. Dies ließ auf eine verstärkte serotonerge Neurotransmission schließen, da nur 5-HT die Bildung eines Protein-haltigen Speichels verursacht. Immuncytochemische Untersuchungen zeigten, dass die Drüsen tyraminerge und octopaminerge Innervation empfangen. Weiterhin wurde der erste charakterisierte TA-Rezeptor (PeaTYR1) der Schabe auf einem paarigen, lateral zur Drüse ziehenden Nerv markiert, der auch tyraminerge Fasern enthielt. Die vorliegende Arbeit trug zum Verständnis der komplexen Funktionsweise der Speicheldrüse der Schabe bei und erweiterte das lückenhafte Wissen über die neuronale Kontrolle exokriner Drüsen in Insekten.
Biogene Amine sind kleine organische Verbindungen, die sowohl bei Wirbeltieren als auch bei Wirbellosen als Neurotransmitter, Neuromodulatoren und/oder Neurohormone wirken können. Sie bilden eine bedeutende Gruppe von Botenstoffen und entfalten ihre Wirkungen über die Bindung an eine bestimmte Klasse von Rezeptorproteinen, die als G-Protein-gekoppelte Rezeptoren bezeichnet werden. Bei Insekten gehören zur Substanzklasse der biogenen Amine die Botenstoffe Dopamin, Tyramin, Octopamin, Serotonin und Histamin. Neben vielen anderen Wirkung ist z.B. gezeigt worden, daß einige dieser biogenen Amine bei der Honigbiene (Apis mellifera) die Geschmacksempfindlichkeit für Zuckerwasser-Reize modulieren können. Ich habe verschiedene Aspekte der aminergen Signaltransduktion an den „Modellorganismen“ Honigbiene und Amerikanische Großschabe (Periplaneta americana) untersucht. Aus der Honigbiene, einem „Modellorganismus“ für das Studium von Lern- und Gedächtnisvorgängen, wurden zwei Dopamin-Rezeptoren, ein Tyramin-Rezeptor, ein Octopamin-Rezeptor und ein Serotonin-Rezeptor charakterisiert. Die Rezeptoren wurden in kultivierten Säugerzellen exprimiert, um ihre pharmakologischen und funktionellen Eigenschaften (Kopplung an intrazelluläre Botenstoffwege) zu analysieren. Weiterhin wurde mit Hilfe verschiedener Techniken (RT-PCR, Northern-Blotting, in situ-Hybridisierung) untersucht, wo und wann während der Entwicklung die entsprechenden Rezeptor-mRNAs im Gehirn der Honigbiene exprimiert werden. Als Modellobjekt zur Untersuchung der zellulären Wirkungen biogener Amine wurden die Speicheldrüsen der Amerikanischen Großschabe genutzt. An isolierten Speicheldrüsen läßt sich sowohl mit Dopamin als auch mit Serotonin Speichelproduktion auslösen, wobei Speichelarten unterschiedlicher Zusammensetzung gebildet werden. Dopamin induziert die Bildung eines völlig proteinfreien, wäßrigen Speichels. Serotonin bewirkt die Sekretion eines proteinhaltigen Speichels. Die Serotonin-induzierte Proteinsekretion wird durch eine Erhöhung der Konzentration des intrazellulären Botenstoffs cAMP vermittelt. Es wurden die pharmakologischen Eigenschaften der Dopamin-Rezeptoren der Schaben-Speicheldrüsen untersucht sowie mit der molekularen Charakterisierung putativer aminerger Rezeptoren der Schabe begonnen. Weiterhin habe ich das ebony-Gen der Schabe charakterisiert. Dieses Gen kodiert für ein Enzym, das wahrscheinlich bei der Schabe (wie bei anderen Insekten) an der Inaktivierung biogener Amine beteiligt ist und im Gehirn und in den Speicheldrüsen der Schabe exprimiert wird.
