Die nachfolgende Länderstudie ist während eines längeren Arbeitsaufenthaltes im Rahmen der internationalen Zusammenarbeit in Kamerun entstanden. Mit ihr versuchen wir, unsere persönlichen Eindrücke und täglichen Beobachtungen in einem Land zu verarbeiten, in dem offenbar alle Hoffnung darauf beruht, dass der alternde Staatspräsident Paul Biya seinen Abschied von der politischen Bühne nimmt und damit ein autokratisches, korruptes Regime sein Ende findet. Diese Hoffnung scheint mit der Erwartung von Francis Fukuyama verbunden zu werden, der 1992 nach dem Zusammenbruch des Sowjet-Imperiums das „Ende der Geschichte“ erklärte, in der Überzeugung, dass das demokratische Gesellschaftsmodell bald überall Einzug halten würde. Bekanntlich hat sich diese Erwartung als zu optimistisch erwiesen. Mit unserer Untersuchung versuchen wir aufzuzeigen, warum sich die Hoffnung auf eine gerechtere Gesellschaft trotz langjähriger Bemühungen westlicher Geber um die Stärkung der Zivilgesellschaft und die Dezentralisierung staatlicher Aufgaben auch in Kamerun kaum erfüllen wird. Ein „Ende der Geschichte“ lässt sich auch für die Zeit nach Paul Biya nicht prognostizieren.

Die vorliegende Arbeit befasst sich mit der Untersuchung von Wanderungsüberlegungen der Einwohner der Stadt Visaginas vor dem Hintergrund steigender ökonomischer Unsicherheit, bedingt durch die bevorstehende Schließung des Atomkraftwerkes 'Ignalina'. Visaginas befindet sich im Nordosten Litauens, in einer der strukturschwächsten Regionen des Landes. Von Mitte der 70er bis Ende der 80er Jahre entstand die Stadt als Wohnstätte für die Arbeiter des sich zu dieser Zeit ebenfalls im Bau befindlichen Atomkraftwerkes 'Ignalina' sowie für deren Angehörige. Noch heute ist die Stadt wirtschaftlich vom Kraftwerk abhängig: Nicht nur ist das staatliche Unternehmen der größte Arbeitgeber für die ca. 30.000 Einwohner, sondern darüber hinaus stellt auch die Mehrzahl der unabhängigen Firmen ihre Leistungen und Produktion in dessen Dienst. Auf Druck der Europäischen Union, welche bereits Ende der 90er Jahre aus sicherheitstechnischen Gründen die Schließung des Atomkraftwerkes forderte, beschloss die Regierung Litauens, das Atomkraftwerk stillzulegen. Am 1. Januar 2005 wird der Betrieb des ersten Reaktors eingestellt. Die endgültige Abschaltung ist bis zum Januar 2009 vorgesehen. Auf diese Weise wird der Stadt Visaginas, und damit verbunden vielen ihrer Einwohner, die wirtschaftliche Existenzgrundlage entzogen. Bedingt durch den Wegfall ihres Arbeitsplatzes wird ein Großteil der Einwohner gezwungen sein, sich nach Beschäftigungsalternativen umzuschauen. Da jedoch auch im Umland der Stadt keine ausreichenden Möglichkeiten bestehen, die freigesetzten Arbeitskräfte aufzufangen, wird für viele Betroffene die einzige Alternative darin liegen, Visaginas auf der Suche nach einem neuen Arbeitsplatz zu verlassen. Die Tatsache, dass der Großteil der heutigen Einwohner aus anderen Unionsrepubliken der damaligen Sowjetunion nach Visaginas kam und nur selten über gute Kenntnisse in der Landessprache Litauisch verfügt, stellt dabei ein besonderes Problem dar. Der Arbeit lag folgende zentrale Fragestellung zugrunde: Warum wandern unter gleichen strukturellen (vor allem ökonomischen) Rahmenbedingungen Menschen aus Visaginas ab, während andere dies nicht tun bzw. eine Abwanderung gar nicht in Erwägung ziehen? This article deals with the explanation of failed democratisation as caused by political culture. Against the background of the Belarus’ autocracy, the author questions that political culture can be considered a reason for failed democratisation. The Belarus’ paternalistic political culture does not essentially differ from that of successfully democratising neighbouring states. A weak national conscience is the only specific characteristic of the Belarus autocracy, but it lacks a convincing theoretical link with democratisation. Nevertheless, in paternalistic political cultures, successful democratisation seems to need more incentives for people, due to higher adaptation costs.
Plant X-tender

Cloning multiple DNA fragments for delivery of several genes of interest into the plant genome is one of the main technological challenges in plant synthetic biology. Despite several modular assembly methods developed in recent years, the plant biotechnology community has not widely adopted them yet, probably due to the lack of appropriate vectors and software tools. Here we present Plant X-tender, an extension of the highly efficient, scarfree and sequence-independent multigene assembly strategy AssemblX,based on overlapdepended cloning methods and rare-cutting restriction enzymes. Plant X-tender consists of a set of plant expression vectors and the protocols for most efficient cloning into the novel vector set needed for plant expression and thus introduces advantages of AssemblX into plant synthetic biology. The novel vector set covers different backbones and selection markers to allow full design flexibility. We have included ccdB counterselection, thereby allowing the transfer of multigene constructs into the novel vector set in a straightforward and highly efficient way. Vectors are available as empty backbones and are fully flexible regarding the orientation of expression cassettes and addition of linkers between them, if required. We optimised the assembly and subcloning protocol by testing different scar-less assembly approaches: the noncommercial SLiCE and TAR methods and the commercial Gibson assembly and NEBuilder HiFi DNA assembly kits. Plant X-tender was applicable even in combination with low efficient homemade chemically competent or electrocompetent Escherichia coli. We have further validated the developed procedure for plant protein expression by cloning two cassettes into the newly developed vectors and subsequently transferred them to Nicotiana benthamiana in a transient expression setup. Thereby we show that multigene constructs can be delivered into plant cells in a streamlined and highly efficient way. Our results will support faster introduction of synthetic biology into plant science.
Polar nuclear migration is crucial during the development of diverse eukaryotes. In plants, root hair growth requires polar nuclear migration into the outgrowing hair. However, knowledge about the dynamics and the regulatory mechanisms underlying nuclear movements in root epidermal cells remains limited. Here, we show that both auxin and Rho-of-Plant (ROP) signaling modulate polar nuclear position at the inner epidermal plasma membrane domain oriented to the cortical cells during cell elongation as well as subsequent polar nuclear movement to the outer domain into the emerging hair bulge in Arabidopsis (Arabidopsis thaliana). Auxin signaling via the nuclear AUXIN RESPONSE FACTOR7 (ARF7)/ARF19 and INDOLE ACETIC ACID7 pathway ensures correct nuclear placement toward the inner membrane domain. Moreover, precise inner nuclear placement relies on SPIKE1 Rho-GEF, SUPERCENTIPEDE1 Rho-GDI, and ACTIN7 (ACT7) function and to a lesser extent on VTI11 vacuolar SNARE activity. Strikingly, the directionality and/or velocity of outer polar nuclear migration into the hair outgrowth along actin strands also are ACT7 dependent, auxin sensitive, and regulated by ROP signaling. Thus, our findings provide a founding framework revealing auxin and ROP signaling of inner polar nuclear position with some contribution by vacuolar morphology and of actin-dependent outer polar nuclear migration in root epidermal hair cells.
The paper describes the content of the project “Tax-/Transfer-Reform and Fiscal Responsibility: Pathways to a Sustainable Fiscal Policy in Georgia”, which is supported for the period Fall 2005 to Spring 2008 by the VolkswagenStiftung. Within the project a knowledgetransfer is organized, concentrating on state theory, tax and transfer reform, revenue sharing as well as fiscal equalization. A short overview on the current situation in Georgia is presented. Then the main project components – tax reform, tax revenue estimation and microsimulation – are briefly discussed.
Background: Children’s spontaneous focusing on numerosity (SFON) is related to numerical skills. This study aimed to examine (1) the developmental trajectory of SFON and (2) the interrelations between SFON and early numerical skills at pre-school as well as their influence on arithmetical skills at school. Method: Overall, 1868 German pre-school children were repeatedly assessed until second grade. Nonverbal intelligence, visual attention, visuospatial working memory, SFON and numerical skills were assessed at age five (M = 63 months, Time 1) and age six (M = 72 months, Time 2), and arithmetic was assessed at second grade (M = 95 months, Time 3). Results: SFON increased significantly during pre-school. Path analyses revealed interrelations between SFON and several numerical skills, except number knowledge. Magnitude estimation and basic calculation skills (Time 1 and Time 2), and to a small degree number knowledge (Time 2), contributed directly to arithmetic in second grade. The connection between SFON and arithmetic was fully mediated by magnitude estimation and calculation skills at pre-school. Conclusion: Our results indicate that SFON first and foremost influences deeper understanding of numerical concepts at pre-school and—in contrast to previous findings –affects only indirectly children’s arithmetical development at school.
