Active host-searching in bat ectoparasites. As a pilot study, this paper discribes the mobility of ectoparasites after removing from their hosts. Spinturnix acuminatus (Koch, 1836)(Acari, Spinturnicidae) a permanent stationary bat mite normally survives the removing only a few hours. This species moves easily on the patagium or on human skin. In contrast, on other surfaces the movements of this species seems to be less orientated and it is unlikely that the mite can reach a host in some distance. Nycteribia kolenatii Theodor & Moscona, 1954 (Diptera: Nycteribiidae) a bat fly, shows excellent movements in the fur of its host. The specimens survive the removing from the host several days. The ability to clamber up a wall was tested with a rough clay brick. The bat fly achieves only 9,6 cm on average. This result does not support good abilities for host searching in this species. Bat fleas (Siphonaptera: Ischnopsyllidae): Ischnopsyllus elongatus (Curtis, 1832), I. hexactenus (Kolenati, 1856), I. simplex (Rothschild, 1906)and other species survive the removing from their hosts several days. In case of a vertical barrier they showed a stereotype upside climbing, followed by a horizontal appetitive behavior. The fleas were climbing a distance in the upside of 6,45 m on average(1,2-21,68 m). As excellent climbers bat fleas are able to overcome edges and slopes and to crawl on the ceiling. Therefore, it seems to be no problem for a flea to parasite any bat in a resting room. Additionally, some bugs (Cimex lectularius Linnaeus, 1758) were examinated. They run with a speed of 63 cm per minute and were able to climb easily on the brick. On average their stretch of way achieved 2,85 m in ten minutes.

* 1. Large female insects usually have high potential fecundity. Therefore selection should favour an increase in body size given that these females get opportunities to realize their potential advantage by maturing and laying more eggs. However, ectotherm physiology is strongly temperature-dependent, and activities are carried out sufficiently only within certain temperature ranges. Thus it remains unclear if the fecundity advantage of a large size is fully realized in natural environments, where thermal conditions are limiting. * 2. Insect fecundity might be limited by temperature at two levels; first eggs need to mature, and then the female needs time for strategic ovipositing of the egg. Since a female cannot foresee the number of oviposition opportunities that she will encounter on a given day, the optimal rate of egg maturation will be governed by trade-offs associated with egg- and time-limited oviposition. As females of different sizes will have different amounts of body reserves, size-dependent allocation trade-offs between the mother"s condition and her egg production might be expected. * 3. In the temperate butterfly Pararge aegeria, the time and temperature dependence of oviposition and egg maturation, and the interrelatedness of these two processes were investigated in a series of laboratory experiments, allowing a decoupling of the time budgets for the respective processes. * 4. The results show that realized fecundity of this species can be limited by both the temperature dependence of egg maturation and oviposition under certain thermal regimes. Furthermore, rates of oviposition and egg maturation seemed to have regulatory effects upon each other. Early reproductive output was correlated with short life span, indicating a cost of reproduction. Finally, large females matured more eggs than small females when deprived of oviposition opportunities. Thus, the optimal allocation of resources to egg production seems dependent on female size. * 5. This study highlights the complexity of processes underlying rates of egg maturation and oviposition in ectotherms under natural conditions. We further discuss the importance of temperature variation for egg- vs. time-limited fecundity and the consequences for the evolution of female body size in insects.
Neben dem Habitatverlust gelten Konsequenzen der Habitatfragmentierung seit den 1990er Jahren als wesentliche Ursache der Gefaehrdung von Pflanzen und stehen damit nun auch im Fokus des botanischen Artenschutzes. Der vorliegende Beitrag gibt einen ueberblick ueber den Stand der populationsbiologischen und genetischen Forschung und versucht abzuschaetzen, welche Bedeutung Habitatfragmentierung und die dadurch entstehenden kleinen, isolierten Populationen auf heimische Pflanzenarten haben koennen. Als wesentliche und offenbar sehr weit verbreitete negative Effekte werden Zufallsereignisse, Randeffekte, Bestaeuberlimitierung, Gendrift und Inzuchtdepression identifiziert. Zusammen mit verringerter Habitatqualitaet durch Eutrophierung, Entwaesserung oder Nutzungsaenderung wirken sie zumeist negativ auf die Fitness der Individuen und Populationen und erhoehen so deren Aussterberisiko. Dieser negative Effekt kleiner Populationen auf die individuelle Fitness wird unabhaengig von der Ursache als Allee-Effekt bezeichnet. Eine durch einen Biotopverbund gefoerderte Metapopulationsdynamik kann das dauerhafte Aussterben von Pflanzenpopulationen verhindern und mindert die negativen genetischen Effekte der Habitatfragmentierung ueber einen erhoehten Genfluss durch Pollen und Samen. Die bisherigen wissenschaftlichen Studien in Mitteleuropa beruhen allerdings in ueberproportionaler Weise auf bestimmten Pflanzenfamilien (Gentianaceae, Primulaceae), Habitaten (Trocken- und Magerrasen, Wirtschaftsgruenland), insekten- und obligat fremdbestaeubten sowie weitgehend auf sexuelle Fortpflanzung angewiesenen Arten, waehrend etwa ueber Grasartige, Ruderalpflanzen, wind- und selbstbestaeubte sowie an vegetative Fortpflanzung angepasste Arten nur wenige Erkenntnisse vorliegen. Gerade diese und Pflanzenarten mit hohem Ausbreitungspotenzial muessen aber nach derzeitigem Wissensstand als weniger sensitiv gegenueber Habitatfragmentierung eingestuft werden. Auf diesen Befunden aufbauend werden fuer die Naturschutzpraxis Biotoptypen hinsichtlich ihrer Sensitivitaet gegenueber Habitatfragmentierung klassifiziert und ein auf biologischen Merkmalen basierender Kriterienkatalog zur Auswahl von Zielarten des Biotopverbunds vorgestellt. Schließlich wird eroertert, was bei Maßnahmen zur Regeneration kleiner bzw. bereits ausgestorbener Populationen zu beachten ist, und es werden allgemeine Folgerungen zur Ausgestaltung eines Biotopverbundskonzepts fuer Pflanzen gezogen.
Vegetation und Standort bodensaurer Buchenwälder am Arealrand : am Beispiel Mittelbrandenburgs

Different from NW Germany, the northern part of NE Germany and the "Hohe Flaeming" region, central Brandenburg is considered as being largely devoid of natural beech forests because of its subcontinental, dry climate. In the present study the vegetation ecology of beech forests of the region is comprehensively documented for the first time, and they are compared with NW German stands in Lower Saxony. In the study area beech forests are concentrated in the Berlin-Potsdam region along the Havel river lakes which is characterised by relatively high precipitation and a specific land use history. All belong to the Luzulo-Fagetum growing on acid soils. Four subtypes are distinguished according to nutrient availability and soil moisture. The central Brandenburg Luzulo-Fagetum does not markedly deviate from other beech forests in the northern German lowlands with respect to vegetation structure and edaphic subtypes. However, numerous indicator species for humid or moist conditions are less frequent than under atlantic climate conditions in the lowlands of Lower Saxony, a pattern occurring also in other forest communities. On the other hand, nitrogen and disturbance indicators are more frequent in central Brandenburg. As expected, podzolisation of the soils and humus accumulation is lower in beech forests under subcontinental climate, but surprisingly the soils are more sandy and thus drier. However, beech forests are lacking on south-exposed slopes, and they are notably occurring in northern exposition. A combined analysis of distribution patterns and climatic data, postglacial vegetation history and forest use history, and actual rejuvenation dynamics reveals that the present-day beech forests in central Brandenburg have to be considered as near-natural relics, which are currently spreading. The range of potentially natural beech forests is larger than assumed until now, but further on it is not clearly to define.
Die 11beta-HSD1 reguliert intrazellulär die Cortisolkonzentration durch Regeneration von Cortison z.B. aus dem Blutkreislauf, zu Cortisol. Daher stellt diese ein wichtiges Element in der Glucocorticoid-vermittelten Genregulation dar. Die 11beta-HSD1 wird ubiquitär exprimiert, auf hohem Niveau besonders in Leber, Fettgewebe und glatten Muskelzellen. Insbesondere die Bedeutung der 11beta-HSD1 in Leber und Fettgewebe konnte mehrfach nachgewiesen werden. In der Leber führte eine erhöhte Aktivität aufgrund einer Überexpression in Mäusen zu einer verstärkten Gluconeogeneserate. Des Weiteren konnte gezeigt werden, dass eine erhöhte Expression und erhöhte Enzymaktivität der 11beta-HSD1 im subkutanen und viszeralen Fettgewebe assoziiert ist mit Fettleibigkeit, Insulinresistenz und Dyslipidämie. Über die Regulation ist jedoch noch wenig bekannt. Zur Untersuchung der Promotoraktivität wurde der Promotorbereich von -3034 bis +188, vor und nach dem Translations- und Transkriptionsstart, der 11beta-HSD1 kloniert. 8 Promotorfragmente wurden mittels Dual-Luciferase-Assay in humanen HepG2-Zellen sowie undifferenzierten und differenzierten murinen 3T3-L1-Zellen untersucht. Anschließend wurde mittels nicht-radioaktiven EMSA die Bindung des TATA-Binding Proteins (TBP) sowie von CCAAT/Enhancer-Binding-Proteinen (C/EBP) an ausgewählte Promotorregionen analysiert. Nach der Charakterisierung des Promotors wurden spezifische endogene und exogene Regulatoren untersucht. Fettsäuren modifizieren die Entstehung von Adipositas und Insulinresistenz. Ihre Wirkung wird u.a. PPARgamma-abhängig vermittelt und kann durch das Inkretin (Glucose-dependent insulinotropic Peptide) GIP modifiziert werden. So wurden die Effekte von unterschiedlichen Fettsäuren, vom PPARgamma Agonisten Rosiglitazon sowie dem Inkretin GIP auf die Expression und Enzymaktivität der 11beta-HSD1 untersucht. Dies wurde in-vitro-, tierexperimentell und in humanen in-vivo-Studien realisiert. Zuletzt wurden 2 Single Nucleotide Polymorphismen (SNP) im Promotorbereich der 11beta-HSD1 in der Zellkultur im Hinblick auf potentielle Funktionalität analysiert sowie die Assoziation mit Diabetes mellitus Typ 2 und Körpergewicht in der MeSyBePo-Kohorte bei rund 1.800 Personen untersucht. Die Luciferase-Assays zeigten basal eine zell-spezifische Regulation der 11beta-HSD1, wobei in allen 3 untersuchten Zelltypen die Bindung eines Repressors nachgewiesen werden konnte. Zudem konnte eine mögliche Bindung des TBPs sowie von C/EBP-Proteinen an verschiedene Positionen gezeigt werden. Die Transaktivierungsassays mit den C/EBP-Proteinen -alpha, -beta und -delta zeigten eben-falls eine zellspezifische Regulation des 11beta-HSD1-Promotors. Die Aktivität und Expression der 11beta-HSD1 wurde durch die hier untersuchten endogenen und exogenen Faktoren spezifisch modifiziert, was sowohl in-vitro als auch in-vivo in unterschiedlichen Modellsystemen dargestellt werden konnte. Die Charakterisierung der MeSyBePo-Kohorte ergab keine direkten Assoziationen zwischen Polymorphismus und klinischem Phänotyp, jedoch Tendenzen für eine erhöhtes Körper-gewicht und Typ 2 Diabetes mellitus in Abhängigkeit des Genotyps. Der Promotor der 11beta-HSD1 konnte aufgrund der Daten aus den Luciferaseassays sowie den Daten aus den EMSA-Analysen näher charakterisiert werden. Dieser zeigt eine variable und zell-spezifische Regulation. Ein wichtiger Regulator stellen insbesondere in den HepG2-Zellen die C/EBP-Proteine -alpha, -beta und -delta dar. Aus den in-vivo-Studien ergab sich eine Regulation der 11beta-HSD1 durch endogene, exogene und pharmakologische Substanzen, die durch die Zellkulturversuche bestätigt und näher charakterisiert werden konnten.
Die vorliegende Untersuchung von Fledermaeusen in Sommer- und Winterquartieren liefert neue daten zur Verbreitung und Haeufigkeit von Ektoparasiten aus Brandenburg und anderen Bundeslaendern. An 11 Fledermausarten wurden insgesamt 7 Floharten (Ischnosyllidae), zwei Lausfliegenarten (Nycteribiidae) und zwei Wanzenarten (Cimicidae) nachgewiesen. Die Floharten Ischnopsyllus intermedius und die Artengruppe I. simplex/mysticus ergaenzen die Liste der aktuellen Funde fuer Brandenburg. Nach dem milden Winter 2006-7 wurden in einigen Winterquartieren Veraenderungen im Artenspektrum der Parasiten festgestellt. Nycteribia kolenatii an Myotis daubentoni und Nycteribia eusarca an Nyctalus noctula traten bei der vorliegenden Untersuchung relativ haeufig auf. Die anderen ektoparasitischen Floehe und Lausfliegen waren sehr selten zu finden. Die meisten Parasitenarten und die hoechste Befallsintensitaet wies die Wasserfledermaus (Myotis daubentoni) auf.
