Die Toxizität und Kanzerogenität von rein aromatischen polyzyklischen aromatischen Kohlenwasserstoffen (PAK) ist seit Jahrzehnten bekannt und umfassend erforscht. Die alkylierten PAK (alkPAK) besitzen jedoch aufgrund ihrer Alkylgruppe eine weitere Möglichkeit zur Bioaktivierung und müssen daher gesondert betrachtet werden. Die Alkylgruppe wird zunächst hydroxyliert, anschließend zur Säure oxidiert oder direkt konjugiert. Entstehen hierbei instabile benzylische Sulfokonjugate, so können diese DNA-Addukte bilden und zu Mutationen führen. In Hinblick auf die Bioaktivierung von alkPAK galt es daher zu klären welchen Einfluss die Struktur auf die benzylische Hydroxylierung hat und welche humanen Formen der löslichen Sulfotransferasen besonders an der Umsetzung der alkPAK-Derivate beteiligt sind. Die Untersuchung der Albuminbindung von Schwefelsäureestern sowie ihre Aufnahme in Nierenzellen sollten Aufschluss hinsichtlich möglicher Transportvorgänge geben. Für die in-vivo-Situation wurde weiterhin die Modulation des Metabolismus ausgewählter benzylischer Alkohole durch verschiedene Nahrungsmittelbestandteile, Arzneimittel und Fremdstoffe an Ratten untersucht. Als Biomarker wurden benzylische Carbonsäuren im Urin und die entsprechenden Mercaptursäuren in Urin und Fäzes betrachtet. Zunächst wurde anhand von Inkubationen mit Rattenlebermikrosomen festgestellt, dass insbesondere größere Ringsystemen wie etwa alkylierte Benzo[a]pyrene im Gegensatz zu Methylpyrenen in wesentlich geringerem Umfang zum benzylischen Alkohol umgesetzt werden. Dies wurde auch in Untersuchungen mit humanen Lebermikrosomen bestätigt. Untersuchungen an einzelnen humanen Cytochromen P450 zeigten, dass insbesondere die durch PAK induzierbaren Formen hCYP1A1 und 1B1 hohe Umsatzraten aufwiesen. Die hepatisch exprimierten Formen hCYP1A2 und 3A4 waren jedoch auch zur Bildung der benzylischen Alkohole in der Lage. Für die anschließende Sulfonierung der benzylischen Alkohole wurden besonders hohe Aktivitäten mit den humanen Sulfotransferasen hSULT1A1, 1A2, 1C2 und 1E1 festgestellt. Aufgrund der Enzymexpression und der guten Durchblutung, die eine gute Substratversorgung ermöglicht, ist die Leber als Hauptort der benzylischen Hydroxylierung und Sulfonierung anzusehen. Ergebnisse unserer Arbeitsgruppe zeigen jedoch, dass nach 1-Hydroxymethylpyren-Applikation bei Ratten die Niere die höchste Zahl an DNA-Addukten aufweist. Wegen der Fokussierung der Sulfonierung auf die Leber ist die systemische Verteilung der Schwefelsäureester die einzig plausible Erklärung. So wurde im Rahmen dieser Arbeit eine hochaffine Bindungsstelle für 2-Sulfoxymethylpyren an Albumin beschrieben und die Aufnahme von benzylischen Sulfaten durch die humanen organischen Anionentransporter hOAT1, 3 und 4 in Nierenzellen in vitro gezeigt. Für die in-vivo-Situation wurde der Einfluss von Ethanol, 4-Methylpyrazol, Pentachlorphenol, Quercetin und Disulfiram untersucht. Neben der durch die Detoxifizierung mittels Alkoholdehydrogenase und Aldehyddehydrogenase entstandenen benzylischen Carbonsäure kann als Biomarker die entsprechende Mercaptursäure herangezogen werden. Sie ist ein indirekter Nachweis für die reaktiven und toxischen benzylischen Sulfate der alkPAK. Für die beiden im Tierversuch eingesetzten benzylischen Alkohole (1-Hydroxymethylpyren und 1-Hydroxymethyl-8-methylpyren) konnte sie in Urin und Fäzes nachgewiesen werden. Es wurde jedoch ein deutlicher Unterschied in der gebildeten Menge sowie der Verteilung zwischen Urin und Fäzes für die beiden Mercaptursäuren festgestellt. Hierfür sind wahrscheinlich Unterschiede im Transport der benzylischen Schwefelsäureester sowie der Spezifität der an der Mercaptursäurebildung beteiligten Enzyme verantwortlich. In diesem Zusammenhang konnte gezeigt werden, dass der humane organische Anionentransporter hOAT1 1,8-Dimethylpyrenmercaptursäure nicht und der hOAT3 nur mit niedrigen Umsatzraten transportiert. Bei den Modulatoren zeigte die Gabe der kompetitiven Alkoholdehydrogenase-Hemmstoffe Ethanol und 4-Methylpyrazol die Bedeutung der Alkoholdehydrogenasen für die Entgiftung der benzylischen Alkohole: Die Oxidation zur entsprechenden Carbonsäure war reduziert und die Bildung der Mercaptursäure erhöht. Eine Hemmung der Toxifizierung vermittelt durch Sulfotransferase-Inhibitoren konnte nur für Pentachlorphenol beim Metabolismus des 1-Hydroxymethylpyrens beobachtet werden. Gleichzeitig erwies sich Pentachlorphenol als kompetitiver Alkoholdehydrogenase-Inhibitor, da eine signifikant geminderte Carbonsäureausscheidung zu beobachten war. Bei 1-Hydroxymethyl-8-methylpyren traten diese Effekte nicht auf. Die unterschiedlichen bzw. unterschiedlich starken Effekte der Modulatoren beim Metabolismus der verschiedenen benzylischen Alkohole bestätigen die Beobachtungen aus den in-vitro-Untersuchungen, dass unterschiedliche Enzym- und Transporteraffinitäten und –aktivitäten vorliegen.
Alkylierte polyzyklische aromatische Kohlenwasserstoffe werden in vielen Matrizes wie Fahrzeugabgasen und Tabakrauch und auch als Kontaminanten in Nahrungsmitteln neben rein aromatischen Kongeneren gefunden. Alkylierte PAK können über die Alkylseitenkette über benzylische Hydroxylierung und nachfolgende Sulfonierung katalysiert über Sulfotransferasen (SULT) zu reaktiven Schwefelsäureestern umgesetzt werden. Die SULT-vermittelte Bioaktivierung zu einem genotoxischen Schwefelsäureester wurde für den benzylischen Alkohol 1-Hydroxymethylpyren des Hepatokanzerogens 1-Methylpyren in früheren Arbeiten gezeigt. In der vorliegenden Arbeit wurde überprüft, ob die benzylischen Alkohole weiterer alkylierter PAK über Sulfonierung zu genotoxischen Schwefelsäureestern umgesetzt werden. Hierzu wurde eine Gruppe von 17 Modellsubstanzen ausgewählt, um die Ableitung von Struktur-Aktivitäts-Beziehungen zu ermöglichen. Das genotoxische Potenzial authentischer benzylischer Schwefelsäureester der Modellsubstanzen wurde zunächst in vitro über DNA-Adduktbildung im zellfreien System und Mutagenität im Salmonella-Rückmutationstest untersucht. Die Sulfate zeigten große Reaktivitätsunterschiede in Abhängigkeit von der Struktur des aromatischen Systems und der Position der Alkylseitenkette, wobei die Endpunkte DNA-Adduktbildung und Mutagenität gut korrelierten. Des Weiteren wurde der Salmonella-Mutagenitätstest mit den benzylischen Alkoholen der untersuchten alkylierten PAK und gentechnisch veränderten S. typhimurium-Stämmen, die SULT-Formen des Menschen heterolog exprimieren, durchgeführt. Bis auf die Alkohole 2- und 4-HMP zeigten alle untersuchten benzylischen Alkohole deutliche mutagene Effekte in einem oder mehreren humane SULT exprimierenden Stämmen. Die durchgeführten in vitro-Versuche zeigten das Potenzial der benzylischen Metabolite alkylierter PAK für genotoxische Wirkungen. Nachfolgend musste geklärt werden, welche Relevanz die beobachteten Effekte für die komplexere in vivo-Situation haben. Nach Verabreichung verschiedener benzylischer Schwefelsäureester und Alkohole an männliche Ratten konnten DNA-Addukte in den untersuchten Organen detektiert werden, was im Fall der Schwefelsäureester deren systemische Bioverfügbarkeit und im Fall der benzylischen Alkohole deren Umsatz durch SULT der männlichen Ratte zeigte. Da im Gegensatz zum Menschen die SULT-Expression in der Ratte auf die Leber fokussiert ist, musste ein Großteil des Umsatzes zu genotoxischen Sulfaten in der Leber stattgefunden haben. DNA-Addukte wurden jedoch auch in extrahepatischen Organen gefunden, was über einen hepatischen Export der gebildeten reaktiven Sulfate und deren Transport über den Blutkreislauf zu diesen Geweben erklärt werden kann. Für die weiterführenden in vivo-Studien wurden die benzylischen Alkohole 1-HMP und 1-HM-8-MP ausgewählt, die trotz großer struktureller Ähnlichkeit toxikodynamische Unterschiede zeigten. Zur Untersuchung der Bedeutung des SULT-vermittelten Toxifizierungsweges als auch konkurrierender detoxifizierender oxidativer Stoffwechselprozesse, wurden für 1-HMP und 1-HM-8-MP in vivo-Inhibitionsstudien mit SULT-Inhibitoren und für 1-HM-8-MP auch mit ADH/ALDH-Inhibitoren durchgeführt. Eine Vorbehandlung mit dem SULT-Hemmstoff Pentachlorphenol führte zu einer Reduktion der DNA-Adduktniveaus in Organen 1-HMP- und 1-HM-8-MP-behandelter Tiere. Die Verabreichung von Quercetin hatte keine Auswirkung auf die DNA-Adduktniveaus. Die Hemmung der DNA-Adduktbildung bei Verabreichung von Pentachlorphenol verdeutlichte jedoch, dass benzylische Alkohole alkylierter PAK in vivo über Sulfonierung bioaktiviert werden. Eine Vorbehandlung mit dem ADH-Inhibitor 4-Methylpyrazol und dem ADH-Substrat Ethanol führte zu erhöhten DNA-Adduktniveaus in Organen 1-HM-8-MP-behandelter Tiere. Den gleichen Effekt, jedoch in geringerem Ausmaß, hatte auch die Vorbehandlung mit dem ALDH-Inhibitor Disulfiram. Dies deutet darauf hin, dass oxidative Modifikationen an der Seitenkette des 1-HM-8-MP einen Detoxifizierungsmechanismus darstellen. Nach Verabreichung benzylischer Metabolite alkylierter PAK wurden oftmals hohe Adduktniveaus in der Niere detektiert. Als mögliche Ursache hierfür wurde eine Transporter-vermittelte renale Sekretion reaktiver Sulfate postuliert, die über Vorbehandlung mit Probenecid vor Verabreichung von 1-HMP und 1-HM-8-MP überprüft wurde. Der Haupteffekt der Probenecid-Behandlung wurde jedoch nicht in der Niere, sondern in der Leber beobachtet, die stark erhöhte Adduktniveaus zeigte. Eine mögliche Erklärung hierfür ist die Hemmung des Exportes in der Leber gebildeter reaktiver Sulfate über Inhibition hepatischer organischer Anionentransporter.
Das seit 1957 als essentiell klassifizierte Spurenelement Selen vermittelt seine Funktion hauptsächlich durch seinen Einbau in Selenoproteine in Form der 21. proteinogenen Aminosäure Selenocystein. Insgesamt wurden 25 humane Gene für Selenoproteine identifiziert, deren genaue Funktion häufig noch nicht bekannt ist. Selen ist das einzige Mitglied aus der Gruppe der Mikronährstoffe, für das nach wie vor eine antikanzerogene Funktion vor allem in Bezug auf Darmkrebs postuliert wird. Die Grundlage dafür liefert eine Interventionsstudie, bei der 1.312 Probanden für 4,5 Jahre mit 200 μg Selen/Tag supplementiert wurden. Dies resultierte in einer Senkung der Gesamtkrebsmortalität um 50 %. Die Fragen einer optimalen Selenzufuhr, die nicht nur den Bedarf deckt, sondern auch die Entfaltung der antikanzerogenen Wirkung von Selen gewährleistet und die zugrunde liegenden molekularen Mechanismen sind noch ungeklärt. Zudem liegt die Selenzufuhr bei einem Großteil der europäischen Bevölkerung unter den Empfehlungen. Deshalb wurden in der vorliegenden Arbeit vier Wochen alte Mäuse für sechs Wochen marginal defizient (0,086 mg/kg Futter) bzw. selenadäquat (0,15 mg/kg Futter) gefüttert. Dieser geringe Unterschied im Selengehalt resultierte in einer Senkung des Plasmaselenspiegels der selenarmen Tiere auf 13 % und der GPx-Aktivität in der Leber auf 35 %. Zunächst wurde der Einfluss von Selen auf die globale Genexpression im murinen Colon mittels Microarray untersucht. Von den im Colon exprimierten Selenoproteinen reagierte die mRNA von SelW, SelH, GPx1 und SelM im Selenmangel besonders deutlich mit Expressionsverlust. Da diese Selenoproteine nicht nur im Colon, sondern auch in Leukozyten reguliert waren, sind sie auch als humane Biomarker für die in dieser Studie gewählte Schwankung des Selengehalts geeignet. Des Weiteren wurde auf Basis der Microarraydaten eine Signalweganalyse durchgeführt, die der Identifizierung krebsrelevanter Signalwege diente, um mögliche molekularbiologische Erklärungsansätze für die Rolle von Selen im Krebsgeschehen zu finden. Es zeigte sich, dass die mRNA von Schlüsselgenen des Wnt-Signalwegs wie β-Catenin, Gsk3β, Dvl2, Tle2, Lef1 und c-Myc auf Schwankungen des Selengehalts reagiert. Vor allem die Induktion von c-Myc, einem Zielgen des Wnt-Signalwegs, deutet darauf hin, dass dieser im Selenmangel tatsächlich aktiver ist als bei selenadäquater Versorgung. Ein weiterer möglicher Erklärungsansatz für die postulierte präventive Funktion von Selen gegenüber Darmkrebs ist die gastrointestinale Glutathionperoxidase (GPx2), die physiologisch in den proliferierenden Zellen des Kryptengrunds exprimiert wird. Die Regulation dieses Enzyms durch den Wnt-Signalweg, der ebenfalls in proliferierenden Zellen aktiv ist, konnte mittels Reportergenanalyse und endogen auf mRNA- und Proteinebene in Zellkultur gezeigt werden. Die Aktivierung verkürzter Promotorkonstrukte und die Mutation eines potentiellen Bindeelements identifizierten den für die Bindung von TCF und β-Catenin verantwortlichen Bereich. Als Zielgen des Wnt-Signalwegs scheint GPx2 zu den an Proliferationsprozessen beteiligten Genen zu gehören, was unter physiologischen Bedingungen die Aufrechterhaltung des intestinalen Epithels gewährleistet. Bei der Entstehung intestinaler Tumore, die in der Initiationsphase zu über 90 % mit einer konstitutiven Aktivierung des Wnt-Signalwegs einhergeht, wirkt GPx2 möglicherweise prokanzerogen. Die genaue Funktion von GPx2 während der Kanzerogenese bleibt weiter zu untersuchen.
Die Induktion antioxidativer Enzyme gilt als eine Möglichkeit, die antioxidative Kapazität von Zellen zu steigern und dadurch mit oxidativem Stress assoziierten Erkrankungen (z. B. Herz-Kreislauf-Erkrankungen, Neurodegeneration, Atherosklerose) vorzubeugen. Ausgehend davon wurde in der vorliegenden Arbeit der Dual-Luciferase-Reportergen-(DLR)-Assay zum Nachweis der Induktion der antioxidativen Enzyme Katalase (CAT), zytosolische Glutathion-Peroxidase (GPX1) und Kupfer-Zink-Superoxid-Dismutase (SOD1) entwickelt. Im Zuge dessen wurden drei Säugetierzelllinien (CaCo2, IEC-18, V79) auf ihre Eignung zur Modellzelllinie untersucht. Aufgrund der Transfektionseffizienz wurde die Fibroblastenzelllinie V79 ausgewählt. Zur Gewährleistung eines hohen Substanzdurchsatzes des DLR-Assays wurden bei der Etablierung Parameter wie Kulturplattenformat, DNA-Menge, Luciferasen-Kinetik berücksichtigt. Nach erfolgreicher Etablierung des Versuchs im 96-Well-Format wurden L-Carnitin, Catechin, Epigallocatechingallat, Genistein, Wasserstoffperoxid (H2O2), Natrium-Ascorbat, Paraquat, Quercetin, 12-O-Tetradecanoylphorbol-13-Acetat (TPA) und Trolox in nicht-zytotoxischen Konzentrationen hinsichtlich der Aktivierung des Ratten-CAT-, des humanen GPX1- und des humanen SOD1-Promotors untersucht. Die Bestimmung der maximal tolerierbaren Behandlungskonzentration erfolgte im Vorfeld mittels Resazurintest. Von den zehn Verbindungen zeichneten sich drei Substanzen als potente Induktoren für die SOD1 und die GPX1 aus. Die 24-stündige Behandlung von mit Reportergenkonstrukten transient transfizierten V79-Zellen mit 100 µM Paraquat resultierte in einer Verdopplung der relativen SOD1-Promotor-Aktivität und einer Erhöhung der relativen GPX1-Promotor-Aktivität auf 1,6 bzw. 1,7. Die Stimulation mit 20 µM Genistein oder 10 µM Quercetin führte wiederum zu einer Verdopplung bis Verdreifachung der relativen SOD1- und GPX1-Promotor-Aktivität. Der Promotor der Rattenkatalase konnte demgegenüber nur durch 50 µM H2O2 aktiviert werden (1,5fach). Für diesen DLR-Assays bieten sich folglich Genistein, Quercetin wie auch H2O2 als Referenzsubstanzen an. Um aber eine qualitative Charakterisierung der einzelnen Verbindungen hinsichtlich ihres Induktionspotentials zu gewährleisten, sollten von allen getesteten Substanzen Dosis-Wirkungskurven aufgenommen werden. Zudem wird für den routinemäßigen Einsatz die Verwendung stabil transfizierter Zellen zur Vermeidung von mit der Transfektion verbundenen experimentellen Schwankungen empfohlen.
Der Fettsäurestoffwechsel unterliegt vielfältigen Kontrollmechanismen. So wird der Fettsäureabbau über die Induktion und Aktivität spezifischer Enzyme reguliert. Ein zentraler Regulator ist dabei der nukleäre Rezeptor Peroxisomenproliferator-aktivierter-Rezeptor-α (PPARα). PPARα wird durch freie Fettsäuren in der Zelle aktiviert und fördert über die Induktion von Zielgenen den Fettsäuretransport und -abbau sowie die Gluconeogenese und Ketogenese. Der Anstieg an freien Fettsäuren beim Fasten, aber auch im Diabetes aktiviert PPARα. Unabhängig davon wurde in beiden Stoffwechsellagen auch eine erhöhte Expression des nukleären Rezeptors Constitutiver-Androstan-Rezeptor (CAR) und einiger CAR-Zielgene, vorrangig Enzyme des Fremdstoffmetabolismus wie Cytochrom P450 2B (CYP2B), festgestellt. Bei der Adaption an eine Fastensituation scheinen PPARα- und CAR-Signalwege über einen bisher unbekannten Mechanismus miteinander verschaltet zu sein. In der vorliegenden Arbeit sollte der der Verschaltung zugrunde liegende Mechanismus anhand eines Modelsystems, der PPARα-Agonisten-vermittelten Verstärkung der Phenobarbital (PB)-abhängigen Induktion des CAR-Zielgens CYP2B, in vitro untersucht werden. Zudem sollte die physiologische Relevanz einer durch PPARα-Agonisten vermittelten Modulierung der CYP2B-Aktivität in einer Ganztierstudie in vivo belegt werden. Die verwendeten synthetischen PPARα-Agonisten steigerten in primären Hepatozyten der Ratte signifikant die Phenobarbital (PB)-abhängige mRNA- und Protein-Expression sowie die Aktivität von CYP2B. Ohne vorherige PB-Behandlung induzierten PPARα-Agonisten CYP2B nicht. In Gegenwart von PB war die Steigerung der CYP2B-Aktivität durch PPARα-Agonisten dosisabhängig. In einem Luciferase-Reportergenassay wurde gezeigt, dass die Induktion durch PB unter der Kontrolle des CYP2B1-Promotors von einem distalen PBREM (PB-responsive-enhancer-module), an welches CAR binden kann, abhängig war. PPARα-Agonisten steigerten diese PB- und PBREM-abhängige Reportergentranskription und induzierten die CAR-mRNA und CAR-Proteinexpression. Sie aktivierten die Transkription eines Reportergens unter der Kontrolle eines Promotorfragments von bis zu 4,4 kb oberhalb des mutmaßlichen CAR-Transkriptionsstarts. Mit Hilfe von Deletionskonstrukten konnte ein potentielles Peroxisomenproliferator-aktivierter-Rezeptor-responsives Element (PPRE) im CAR-Promotorbereich von -942 bp bis -930 bp identifiziert werden, welches essentiell für die Initiation der Transkription durch PPARα-Agonisten ist. In band shift Experimenten akkumulierte verstärkt Kernprotein mit diesem PPRE. Ein Überschuss an unmarkiertem Wildtyp-CAR-Reportergenvektor, nicht aber an CAR-Reportergenvektor mit PPRE-Deletion, konnte mit dem markierten PPRE um die Bindung von Kernprotein konkurrieren. Nach Chromatin-Immunpräzipitation mit einem PPARα-Antikörper wiederum wurde das betreffende PPRE amplifiziert. Bei in vivo Experimenten an männlichen Ratten resultierte die Behandlung mit PPARα-Agonisten in einer signifikanten Induktion der CAR-mRNA-Expression und signifikant erhöhter PB-abhängiger CYP2B-Aktivität. Die physiologisch Relevanz wurde durch weiterführenden Experimente unterstrichen, in denen gezeigt wurde, dass die Fasten-abhängige Induktion von CAR in PPARα-defizienten Mäusen unterdrückt war. Diese Experimente legen nahe, dass durch PPARα-Agonisten aktiviertes PPARα an das PPRE im CAR-Promotorbereich von -942 bp bis -930 bp bindet und dadurch die CAR-Transkription induziert. Somit kann CAR als PPARα-Zielgen betrachtet werden, was die Schlussfolgerung zulässt, dass die PPARα- und CAR-Signalwege über die direkte Bindung von PPARα an den CAR-Promotor unmittelbar miteinander verknüpft sind. Allerdings ist davon unabhängig eine Aktivierung von CAR, etwa durch PB, für die vermehrte Induktion von CAR-Zielgenen notwendig . Die physiologische Relevanz der PPARα-abhängige CAR-Expression zeigt sich in den Ganztierexperimenten, bei denen die Wirksamkeit der PPARα-Agonisten bestätigt werden konnte. CAR-abhängig induzierte Enzyme sind nicht nur in großem Umfang am Fremdstoffmetabolismus beteiligt, sondern auch am Abbau von Schilddrüsenhormonen und Glucocorticoiden. Sie können damit direkt Einfluss auf den Kohlenhydrat- und Energiestoffwechsel sowie die Regulation der Nahrungsaufnahme nehmen. Über eine PPARα-abhängige Induktion von CAR im Rahmen der Fastenadaption könnten die CAR-Zielgene UDP-Glucuronyltransferase 1A1 und Sulfotransferase N beispielsweise verstärkt Schilddrüsenhormone abbauen und in der Folge den Grundumsatz senken. Der in dieser Arbeit erstmals beschriebene Mechanismus ist dafür von zentraler Bedeutung.
