Composition and functions of microbial communities affect important traits in diverse hosts, from crops to humans. Yet, mechanistic understanding of how metabolism of individual microbes is affected by the community composition and metabolite leakage is lacking. Here, we first show that the consensus of automatically generated metabolic reconstructions improves the quality of the draft reconstructions, measured by comparison to reference models. We then devise an approach for gap filling, termed COMMIT, that considers metabolites for secretion based on their permeability and the composition of the community. By applying COMMIT with two soil communities from the Arabidopsis thaliana culture collection, we could significantly reduce the gap-filling solution in comparison to filling gaps in individual reconstructions without affecting the genomic support. Inspection of the metabolic interactions in the soil communities allows us to identify microbes with community roles of helpers and beneficiaries. Therefore, COMMIT offers a versatile fully automated solution for large-scale modelling of microbial communities for diverse biotechnological applications. <br /> Author summaryMicrobial communities are important in ecology, human health, and crop productivity. However, detailed information on the interactions within natural microbial communities is hampered by the community size, lack of detailed information on the biochemistry of single organisms, and the complexity of interactions between community members. Metabolic models are comprised of biochemical reaction networks based on the genome annotation, and can provide mechanistic insights into community functions. Previous analyses of microbial community models have been performed with high-quality reference models or models generated using a single reconstruction pipeline. However, these models do not contain information on the composition of the community that determines the metabolites exchanged between the community members. In addition, the quality of metabolic models is affected by the reconstruction approach used, with direct consequences on the inferred interactions between community members. Here, we use fully automated consensus reconstructions from four approaches to arrive at functional models with improved genomic support while considering the community composition. We applied our pipeline to two soil communities from the Arabidopsis thaliana culture collection, providing only genome sequences. Finally, we show that the obtained models have 90% genomic support and demonstrate that the derived interactions are corroborated by independent computational predictions.
Die Professionsorientierung der Lehramtsstudiengänge ist ein zentrales Anliegen der universitären Potsdamer Lehrkräftebildung. Seit 1999 finden Evaluationen zur Professionsorientierung statt, die Diskrepanzen zwischen der gewünschten und der erfahrenen Professionsorientierung durch die Studierenden aufzeigen. Im Wintersemester 2013/14 wurden neue Studiengänge an der Universität Potsdam eingeführt. Inwieweit damit auch eine stärkere Professionsorientierung und ein stärkerer Berufsbezug erfolgt ist, ist bislang ungeklärt. In einer Onlinebefragung im Dezember 2018 wurden Studierende der Lehramtsstudiengänge der Universität Potsdam gebeten, die inhaltliche Gestaltung der Lehramtsstudiengänge sowie die Professionsorientierung der Praxisphasen, die Betreuung und Beratung im Rahmen der Praktika, den Nutzen der Praktika für Studium und Beruf und ihre Lehrer:innenkompetenz einzuschätzen. Der Beitrag stellt erste empirische Analysen dar und diskutiert Anregungen zur Weiterentwicklung der Studiengänge mit Bezug auf die Praxisstudien.
Studien zu langen Praxisphasen konnten aufzeigen, dass das Kompetenzerleben der Studierenden von Beginn bis zum Ende der Praxisphase zunahm. Auf Basis einer langfristig angelegten Evaluationsstudie zu allen bildungswissenschaftlichen und fachdidaktischen Praktika im Verlauf des Ba-/Ma-Lehramtsstudiums wurde auch das selbst eingeschätzte Kompetenzerleben Studierender im Schulpraktikum (Praxissemester) analysiert. Die empirische Befundlage deutet darauf hin, dass die Ausprägung des Kompetenzerlebens hinsichtlich SuSorientiertem Handeln, Unterrichtlichem Handeln und Wertvermittelndem Handeln über den Praktikumszeitraum zunimmt. Innovierend-kooperatives Handeln zeigt keine signifikante Entwicklung. Spezielle Einflüsse konnten vor allem durch das wahrgenommene Kompetenzerleben zu Beginn des Schulpraktikums, aber auch durch die jeweilige Nachbereitung der Fachdidaktik und der Bildungswissenschaften ermittelt werden. Die Befunde werden mit Blick auf die bisherige Forschungslage und die Stichprobe der Untersuchung kritisch diskutiert.
Proteasomes are key proteases in regulating protein homeostasis. Their holo-enzymes are composed of 40 different subunits which are arranged in a proteolytic core (CP) flanked by one to two regulatory particles (RP). Proteasomal proteolysis is essential for the degradation of proteins which control time-sensitive processes like cell cycle progression and stress response. In dividing yeast and human cells, proteasomes are primarily nuclear suggesting that proteasomal proteolysis is mainly required in the nucleus during cell proliferation. In yeast, which have a closed mitosis, proteasomes are imported into the nucleus as immature precursors via the classical import pathway. During quiescence, the reversible absence of proliferation induced by nutrient depletion or growth factor deprivation, proteasomes move from the nucleus into the cytoplasm. In the cytoplasm of quiescent yeast, proteasomes are dissociated into CP and RP and stored in membrane-less cytoplasmic foci, named proteasome storage granules (PSGs). With the resumption of growth, PSGs clear and mature proteasomes are transported into the nucleus by Blm10, a conserved 240 kDa protein and proteasome-intrinsic import receptor. How proteasomes are exported from the nucleus into the cytoplasm is unknown.

Several morphological and mitochondrial lineages of the alpine ringlet butterfly species Erebia pronoe have been described, indicating a complex phylogenetic structure. However, the existing data were insufficient and allow neither a reconstruction of the biogeographic history, nor an assessment of the genetic lineages. Therefore, we analysed mitochondrial (COI, NDI) and nuclear (EF1 alpha, RPS5) gene sequences and compared them with sequences from the sister species Erebia melas. Additionally, we combined this information with morphometric data of the male genitalia and the infection patterns with Wolbachia strains, based on a WSP analysis. We obtained a distinct phylogeographic structure within the E. pronoe-melas complex with eight well-distinguishable geographic groups, but also a remarkable mito-nuclear discordance. The mito-nuclear discordance in E. melas and E. pronoe glottis can be explained by different ages of Wolbachia infections with different Wolbachia strains, associated selective sweeps, and hybridisation inhibition. Additionally, we found indications for incipient speciation of E. pronoe glottis in the Pyrenees and a pronounced range dynamic within and among the other high mountain systems of Europe. Our results emphasize the importance of combined approaches in reconstructing biogeographic patterns and evaluating phylogeographic splits.
Simple Summary:& nbsp;High alpine meadows are home to numerous endemic butterfly species. A combination of climate change and changes in agricultural practices has led to a severe decline in many species. A seemingly unaffected representative of this habitat is Erebia pronoe. We studied the behaviour, resource use and population structure of this species to explain its resilience and estimate its future survival potential. This species shows pronounced protandry in combination with serial eclosion. Males were significantly more active and mobile and were also caught significantly more often than females, resulting in a pronounced shift in sex ratio in the predicted population structure. The adults use a wide range of nectar plants and establish homeranges in areas of high habitat quality. Thus, Erebia pronoe adults use a wide array of resources combined with a slight specialisation to avoid niche overlap with closely related species. The resulting ecological flexibility seems to be an adaptation to unpredictable environmental conditions, which should be the result of a long-lasting adaptation process. Moreover, the combination of opportunism and modest specialisation should also be a good basis for coping with future changes caused by climate and land-use change.</p> <br /> <br></p> <br /> A mark-recapture study of the nominotypical Erebia pronoe in the Alps was conducted to survey its ecological demands and characteristics. Population structure analysis revealed a combination of protandry (one-week earlier eclosion of males) and serial eclosion. Significant differences between both sexes were found in population density (males: 580/ha & PLUSMN; 37 SE; females: 241/ha & PLUSMN; 66 SE), sex-ratio (2.4) and behaviour (57.7 vs. 11.9% flying). Both sexes used a wide range of nectar plants (Asteraceae, 77.3%; Dipsacaceae, 12.3%; Gentianaceae, 9.7%). The use of nectar plants shows a non-specific spectrum, which, however, completely avoids overlap with the locally co-occurring species Erebia nivalis. Movement patterns show the establishment of homeranges, which significantly limits the migration potential. Due to its broad ecological niche, E. pronoe will probably be able to react plastically to the consequences of climate change. The formation of high population densities, the unconcerned endangerment status, the unspecific resource spectrum and the sedentary character of the species make E. pronoe a potential indicator of the quality and general resource occurrence of alpine rupicolous grasslands.

The abundant evapotranspiration provided by the Amazon forests is an important component of the hydrological cycle, both regionally and globally. Since the last century, deforestation and expanding agricultural activities have been changing the ecosystem and its provision of moisture to the atmosphere. However, it remains uncertain how the ongoing land use change will influence rainfall, runoff, and water availability as findings from previous studies differ. Using moisture tracking experiments based on observational data, we provide a spatially detailed analysis recognizing potential teleconnection between source and sink regions of atmospheric moisture. We apply land use scenarios in upwind moisture sources and quantify the corresponding rainfall and runoff changes in downwind moisture sinks. We find spatially varying responses of water regimes to land use changes, which may explain the diverse results from previous studies. Parts of the Peruvian Amazon and western Bolivia are identified as the sink areas most sensitive to land use change in the Amazon and we highlight the current water stress by Amazonian land use change on these areas in terms of the water availability. Furthermore, we also identify the influential source areas where land use change may considerably reduce a given target sink's water reception (from our example of the Ucayali River basin outlet, rainfall by 5–12 % and runoff by 19–50 % according to scenarios). Sensitive sinks and influential sources are therefore suggested as hotspots for achieving sustainable land–water management.
Many phenomena of high relevance for economic development such as human capital, geography and climate vary considerably within countries as well as between them. Yet, global data sets of economic output are typically available at the national level only, thereby limiting the accuracy and precision of insights gained through empirical analyses. Recent work has used interpolation and downscaling to yield estimates of sub-national economic output at a global scale, but respective data sets based on official, reported values only are lacking. We here present DOSE — the MCC-PIK Database Of Sub-national Economic Output. DOSE contains harmonised data on reported economic output from 1,661 sub-national regions across 83 countries from 1960 to 2020. To avoid interpolation, values are assembled from numerous statistical agencies, yearbooks and the literature and harmonised for both aggregate and sectoral output. Moreover, we provide temporally- and spatially-consistent data for regional boundaries, enabling matching with geo-spatial data such as climate observations. DOSE provides the opportunity for detailed analyses of economic development at the subnational level, consistent with reported values.
After the United Kingdom has left the European Union it remains unclear whether the two parties can successfully negotiate and sign a trade agreement within the transition period. Ongoing negotiations, practical obstacles and resulting uncertainties make it highly unlikely that economic actors would be fully prepared to a “no-trade-deal” situation. Here we provide an economic shock simulation of the immediate aftermath of such a post-Brexit no-trade-deal scenario by computing the time evolution of more than 1.8 million interactions between more than 6,600 economic actors in the global trade network. We find an abrupt decline in the number of goods produced in the UK and the EU. This sudden output reduction is caused by drops in demand as customers on the respective other side of the Channel incorporate the new trade restriction into their decision-making. As a response, producers reduce prices in order to stimulate demand elsewhere. In the short term consumers benefit from lower prices but production value decreases with potentially severe socio-economic consequences in the longer term.
Background: High-intensity muscle actions have the potential to temporarily improve the performance which has been denoted as postactivation performance enhancement.
Objectives: This study determined the acute effects of different stretch-shortening (fast vs. low) and strength (dynamic vs. isometric) exercises executed during one training session on subsequent balance performance in youth weightlifters.
Materials and Methods: Sixteen male and female young weightlifters, aged 11.3±0.6years, performed four strength exercise conditions in randomized order, including dynamic strength (DYN; 3 sets of 3 repetitions of 10 RM) and isometric strength exercises (ISOM; 3 sets of maintaining 3s of 10 RM of back-squat), as well as fast (FSSC; 3 sets of 3 repetitions of 20-cm drop-jumps) and slow (SSSC; 3 sets of 3 hurdle jumps over a 20-cm obstacle) stretch-shortening cycle protocols. Balance performance was tested before and after each of the four exercise conditions in bipedal stance on an unstable surface (i.e., BOSU ball with flat side facing up) using two dependent variables, i.e., center of pressure surface area (CoP SA) and velocity (CoP V).
Results: There was a significant effect of time on CoP SA and CoP V [F(1,60)=54.37, d=1.88, p<0.0001; F(1,60)=9.07, d=0.77, p=0.003]. In addition, a statistically significant effect of condition on CoP SA and CoP V [F(3,60)=11.81, d=1.53, p<0.0001; F(3,60)=7.36, d=1.21, p=0.0003] was observed. Statistically significant condition-by-time interactions were found for the balance parameters CoP SA (p<0.003, d=0.54) and CoP V (p<0.002, d=0.70). Specific to contrast analysis, all specified hypotheses were tested and demonstrated that FSSC yielded significantly greater improvements than all other conditions in CoP SA and CoP V [p<0.0001 (d=1.55); p=0.0004 (d=1.19), respectively]. In addition, FSSC yielded significantly greater improvements compared with the two conditions for both balance parameters [p<0.0001 (d=2.03); p<0.0001 (d=1.45)].
Conclusion: Fast stretch-shortening cycle exercises appear to be more effective to improve short-term balance performance in young weightlifters. Due to the importance of balance for overall competitive achievement in weightlifting, it is recommended that young weightlifters implement dynamic plyometric exercises in the fast stretch-shortening cycle during the warm-up to improve their balance performance.
Wind influences the development, architecture and morphology of plant roots and may modify subsequent interactions between plants and soil (plant–soil feedbacks—PSFs). However, information on wind effects on fine root morphology is scarce and the extent to which wind changes plant–soil interactions remains unclear. Therefore, we investigated the effects of two wind intensity levels by manipulating surrounding vegetation height in a grassland PSF field experiment. We grew four common plant species (two grasses and two non-leguminous forbs) with soil biota either previously conditioned by these or other species and tested the effect of wind on root:shoot ratio, fine root morphological traits as well as the outcome for PSFs. Wind intensity did not affect biomass allocation (i.e. root:shoot ratio) in any species. However, fine-root morphology of all species changed under high wind intensity. High wind intensity increased specific root length and surface area and decreased root tissue density, especially in the two grasses. Similarly, the direction of PSFs changed under high wind intensity in all four species, but differences in biomass production on the different soils between high and low wind intensity were marginal and most pronounced when comparing grasses with forbs. Because soils did not differ in plant-available nor total nutrient content, the results suggest that wind-induced changes in root morphology have the potential to influence plant–soil interactions. Linking wind-induced changes in fine-root morphology to effects on PSF improves our understanding of plant–soil interactions under changing environmental conditions.
When it comes to footnotes, Alexander von Humboldt was ahead of his times even though his references leave much to be desired by today’s academic standards. This article examines the footnotes of Humboldt’s Essai politique sur l‘île de Cuba (1826). While it is not always easy to decipher his sometimes cryptic references, the undertaking is worthwhile: Humboldt’s footnotes do not only reveal his vast networks of knowledge. They also provide glimpses of ongoing, contemporary disputes among different scholars that involve Humboldt’s writings. They also present Humboldt’s reactions to such disputes. Exploring Humboldt’s footnotes consequently allows the reader to access both Humboldt the scholar and Humboldt the human being.
Albert Berg (1825–1884) was the first director of the Silesian Art Museum in Breslau. He was supported by a network of sponsors, who advised him on planning his trips and bought paintings (or arranged purchases). A particularly important mentor beginning in 1849 was Alexander von Humboldt, also the singer Amalie Joachim. Berg made a number of important contacts with fellow students during his law studies in Bonn. He formed a particularly close relationship with Friedrich Franz II von Mecklenburg-Schwerin, who, in 1844, invited Berg to a journey. This expedition marked the beginning of a lifelong friendship. Friedrich Franz II bought numerous paintings from Berg and worked behind the scenes to organize Berg's career.
Der Erwerb der südamerikanischen Reisetagebücher Alexander von Humboldts durch die Staatsbibliothek Preußischer Kulturbesitz war 2014 ein bedeutendes Ereignis. In einem ersten Schritt wertet der Artikel unveröffentlichte Teile der Tagebücher und Dokumente aus dem Nachlass aus, um den „Kosmos Humboldt´scher Interessen“ zum Thema Pflanzengeographie zu umreißen, darunter auch seine Überlegungen zum Einfluss von Mensch und Tier auf die Ausbreitung von Pflanzen und die Veränderung von Vegetation. Die Einsichten des Gelehrten zur Abhängigkeit Europas von der sogenannten „Neuen Welt“ hinsichtlich der Nahrungsmittel werden ebenso behandelt. So vertrat Humboldt in seinen Ausführungen zum Thema „Was sind Barbaren?“ die These, dass Isolierung von Menschen aus der Gemeinschaft, Elend, Nahrungsmangel, klimatische Ursachen usw. „die Spuren von Zivilisation vertilgen“. Er betonte, dass es nötig sein, in einem weltweiten Wettstreit intellektueller Kräfte die Ressourcenknappheit zu überwinden.
Franz Ferdinand Julius Meyen (1804–1840) veröffentlichte auf sehr vielen Gebieten der Biologie wichtige Arbeiten, vor allem in Botanik und Zoologie. Mit Unterstützung Alexander von Humboldts gelang es ihm, auf dem Forschungsschiff „Prinzess Louise“ anzuheuern und als Schiffsarzt an einer mehrjährigen Weltreise teilzunehmen. Humboldt, der einige Regionen ebenfalls besucht und erforscht hatte, unterstützte seinen jungen Kollegen bei der Vorbereitung seiner Reise. Meyen veröffentlichte seine Reiseergebnisse bereits kurz nach seiner Rückkehr, gefördert von Christian Gottfried Nees von Esenbeck (1776–1858), sowohl als zweiteiligen historischen Reisebericht als auch in sieben Einzelabhandlungen. Einige Ergebnisse, die er, schwerkrank, nicht selbst verwerten konnte, übergab er noch bei Lebzeiten zur Bearbeitung an Spezialisten, andere wurden posthum von Mitgliedern der Leopoldina publiziert. Meyen brachte u. a. eine umfangreiche Sammlung an Vogelpräparaten mit, zu denen auch das Erstbeschreibungsexemplar des von ihm so genannten Spheniscus humboldti (Trivialname: Humboldt-Pinguin) gehört.

We present the first systematic literature review on stress and burnout in K−12 teachers during the COVID-19 pandemic. Based on a systematic literature search, we identified 17 studies that included 9,874 K−12 teachers from around the world. These studies showed some indication that burnout did increase during the COVID-19 pandemic. There were, however, almost no differences in the levels of stress and burnout experienced by K−12 teachers compared to individuals employed in other occupational fields. School principals' leadership styles emerged as an organizational characteristic that is highly relevant for K−12 teachers' levels of stress and burnout. Individual teacher characteristics associated with burnout were K−12 teachers' personality, self-efficacy in online teaching, and perceived vulnerability to COVID-19. In order to reduce stress, there was an indication that stress-management training in combination with training in technology use for teaching may be superior to stress-management training alone. Future research needs to adopt more longitudinal designs and examine the interplay between individual and organizational characteristics in the development of teacher stress and burnout during the COVID-19 pandemic and beyond.
