Light and gravity are two key determinants in orientating plant stems for proper growth and development. The organization and dynamics of the actin cytoskeleton are essential for cell biology and critically regulated by actin-binding proteins. However, the role of actin cytoskeleton in shoot negative gravitropism remains controversial. In this work, we report that the actin-binding protein Rice Morphology Determinant (RMD) promotes reorganization of the actin cytoskeleton in rice (Oryza sativa) shoots. The changes in actin organization are associated with the ability of the rice shoots to respond to negative gravitropism. Here, light-grown rmd mutant shoots exhibited agravitropic phenotypes. By contrast, etiolated rmd shoots displayed normal negative shoot gravitropism. Furthermore, we show that RMD maintains an actin configuration that promotes statolith mobility in gravisensing endodermal cells, and for proper auxin distribution in light-grown, but not dark-grown, shoots. RMD gene expression is diurnally controlled and directly repressed by the phytochrome-interacting factor-like protein OsPIL16. Consequently, overexpression of OsPIL16 led to gravisensing and actin patterning defects that phenocopied the rmd mutant. Our findings outline a mechanism that links light signaling and gravity perception for straight shoot growth in rice.
Magnetic reconnection is a fundamental process of topology change and energy release, taking place in plasmas on the Sun, in space, in astrophysical objects and in the laboratory. However, observational evidence has been relatively rare and typically only partial. Here we present evidence of fast reconnection in a solar filament eruption using high-resolution H-alpha images from the New Vacuum Solar Telescope, supplemented by extreme ultraviolet observations. The reconnection is seen to occur between a set of ambient chromospheric fibrils and the filament itself. This allows for the relaxation of magnetic tension in the filament by an untwisting motion, demonstrating a flux rope structure. The topology change and untwisting are also found through nonlinear force-free field modelling of the active region in combination with magnetohydrodynamic simulation. These results demonstrate a new role for reconnection in solar eruptions: the release of magnetic twist.
The Gongjue basin from the eastern Qiangtang terrane is located in the transition region where the regional structural lineation curves from east-west-oriented in Tibet to north-south-oriented in Yunnan. In this study, we sampled the red beds in the basin from the lower Gongjue to upper Ranmugou formations for the first time covering the entire stratigraphic profile. The stratigraphic ages are bracketed within 53-43Ma by new detrital zircon U-Pb ages constraining the maximum deposition age to 52.51.5Ma. Rock magnetic and petrographic studies indicate that detrital magnetite and hematite are the magnetic carriers. Positive reversals and fold tests demonstrate that the characteristic remanent magnetization has a primary origin. The Gongjue and Ranmugou formations yield mean characteristic remanent magnetization directions of D-s/I-s=31.0 degrees/21.3 degrees and D-s/I-s=15.9 degrees/22.0 degrees, respectively. The magnetic inclination of these characteristic remanent magnetizations is significantly shallowed compared to the expected inclination for the locality. However, the elongation/inclination correction method does not provide a meaningful correction, likely because of syn-depositional rotation. Rotations relative to the Eurasian apparent polar wander path occurred in three stages: Stage I, 33.33.4 degrees clockwise rotation during the deposition of the Gongjue and lower Ranmugou formations; Stage II, 26.93.7 degrees counterclockwise rotation during deposition of the lower and middle Ranmugou formation; and Stage III, 17.73.3 degrees clockwise rotation after 43Ma. The complex rotation history recorded in the basin is possibly linked to sinistral shear along the Qiangtang block during India indentation into Asia and the early stage of the extrusion of the northwestern Indochina blocks away from eastern Tibet.
Pollen influx (number of pollen grains cm−2 year−1) can objectively reflect the dispersal and deposition features of pollen within a certain time and space, and is often used as a basis for the quantitative reconstruction of palaeovegetation; however, little is known about the features and mechanisms of vertical dispersal of pollen. Here we present the results from a 5 year (2006–2010) monitoring program using pollen traps placed at different heights from ground level up to 60 m and surface soil samples in a mixed coniferous and deciduous broad-leaved woodland in the Changbai mountains, northeastern China. The pollen percentages and pollen influx from the traps have very similar characteristics to the highest values for Betula, Fraxinus, Quercus and Pinus, among the tree taxa and Artemisia, Chenopodiaceae and Asteraceae among the herb taxa. Pollen influx values vary significantly with height and show major differences between three distinct layers, above-canopy (≥32 m), within the trunk layer (8 ≤ 32 m) and on the ground (0 m). These differences in pollen influx are explained by differences in (i) the air flows in each of these layers and (ii) the fall speed of pollen of the various taxa. We found that the pollen recorded on the ground surface is a good representation of the major part of the pollen transported in the trunk space of the woodland. Comparison of the pollen influx values with the theoretical, calculated “characteristic pollen source area” (CPSA) of 12 selected taxa indicates that the pollen deposited on the ground surface of the woodland is a fair representation with 85–90 % of the total pollen deposited at a wind speed of 2.4 m s−1 coming from within ca. 1–5 km for Pinus and Quercus, ca. 5–10 km for Ulmus, Tilia, Oleaceae and Betula, ca. 20–40 km for Fraxinus, Poaceae, Chenopodiaceae, Populus and Salix, and ca. 30–60 km for Artemisia; it is also a good representation with 90–98 % of the total pollen deposited coming from within 60 km at a wind speed of 2.4 m s−1, or 100 km at a wind speed: 6 m s−1, for the 12 selected taxa used in the CPSA calculation. Furthermore, comparison with the vegetation map of the area around the sampling site shows that the pollen deposited on the ground represents all plant communities which grow in the study area within 70 km radius of the sampling site. In this study, the pollen percentages obtained from the soil surface samples are significantly biased towards pollen taxa with good preservation due to thick and robust pollen walls. Therefore, if mosses are available instead, soil samples should be avoided for pollen studies, in particular for the study of pollen-vegetation relationships, the estimation of pollen productivities and quantitative reconstruction of past vegetation. The results also indicate that the existing model of pollen dispersal and deposition, Prentice’s model, provides a fair description of the actual pollen dispersal and deposition in this kind of woodland, which suggests that the application of the landscape reconstruction algorithm would be relevant for reconstruction of this type of woodland in the past.
In plants, transcripts move to distant body parts to potentially act as systemic signals regulating development and growth. Thousands of messenger RNAs (mRNAs) are transported across graft junctions via the phloem to distinct plant parts. Little is known regarding features, structural motifs, and potential base modifications of transported transcripts and how these may affect their mobility. We identified Arabidopsis thalianam RNAs harboring the modified base 5-methylcytosine (m(5)C) and found that these are significantly enriched in mRNAs previously described as mobile, moving over graft junctions to distinct plant parts. We confirm this finding with graft-mobile methylated mRNAs TRANSLATIONALLY CONTROLLED TUMOR PROTEIN 1 (TCTP1) and HEAT SHOCK COGNATE PROTEIN 70.1 (HSC70.1), whose mRNA transport is diminished in mutants deficient in m(5)C mRNA methylation. Together, our results point toward an essential role of cytosine methylation in systemic mRNA mobility in plants and that TCTP1 mRNA mobility is required for its signaling function.
Camellia sinensis synthesizes and emits a large variety of volatile phenylpropanoids and benzenoids (VPB). To investigate the enzymes involved in the formation of these VPB compounds, a new C. sinensis short-chain dehydrogenase/reductase (CsSDR) was isolated, cloned, sequenced, and functionally characterized. The complete open reading frame of CsSDR contains 996 nucleotides with a calculated protein molecular mass of 34.5 kDa. The CsSDR recombinant protein produced in Escherichia coli exhibited dehydrogenase-reductase activity towards several major VPB compounds in C. sinensis flowers with a strong preference for NADP/NADPH co-factors, and showed affinity for (R)/(S)-1-phenylethanol (1PE), phenylacetaldehyde, benzaldehyde, and benzyl alcohol, and no affinity for acetophenone (AP) and 2-phenylethanol. CsSDR showed the highest catalytic efficiency towards (R)/(S)-1PE. Furthermore, the transient expression analysis in Nicotiana benthamiana plants validated that CsSDR could convert 1PE to AP in plants. CsSDR transcript level was not significantly affected by floral development and some jasmonic acid-related environmental stress, and CsSDR transcript accumulation was detected in most floral tissues such as receptacle and anther, which were main storage locations of VPB compounds. Our results indicate that CsSDR is expressed in C. sinensis flowers and is likely to contribute to a number of floral VPB compounds including the 1PE derivative AP.
Electromagnetic ion cyclotron waves have long been recognized to play a crucial role in the dynamic loss of ring current protons. While the field-aligned propagation approximation of electromagnetic ion cyclotron waves was widely used to quantify the scattering loss of ring current protons, in this study, we find that the wave normal distribution strongly affects the pitch angle scattering efficiency of protons. Increase of peak normal angle or angular width can considerably reduce the scattering rates of <= 10 keV protons. For >10 keV protons, the field-aligned propagation approximation results in a pronounced underestimate of the scattering of intermediate equatorial pitch angle protons and overestimates the scattering of high equatorial pitch angle protons by orders of magnitude. Our results suggest that the wave normal distribution of electromagnetic ion cyclotron waves plays an important role in the pitch angle evolution and scattering loss of ring current protons and should be incorporated in future global modeling of ring current dynamics.
Carbonate rocks, widely used for paleomagnetically quantifying the drift history of the Gondwana derived continental blocks of the Tibetan Plateau and evolution of the Paleo/Meso/Neo-Tethys Oceans, are prone to pervasive remagnetization. Identifying remagnetization is difficult because it is commonly undetectable through the classic paleomagnetic field tests. Here we apply comprehensive paleomagnetic, rock magnetic, and petrographic studies to upper Triassic limestones in the eastern Qiangtang block. Our results reveal that detrital/biogenic magnetite, which may carry the primary natural remanent magnetization (NRM), is rarely preserved in these rocks. In contrast, authigenic magnetite and hematite pseudomorphs after pyrite, and monoclinic pyrrhotite record three episodes of remagnetization. The earliest remagnetization was induced by oxidation of early diagenetic pyrite to magnetite, probably related to the collision between the northeastern Tibetan Plateau and the Qiangtang block after closure of the Paleo-Tethys Ocean in the Late Triassic. The second remagnetization, residing in hematite and minor goethite, which is the further subsurface oxidation product of pyrite/magnetite, is possibly related to the development of the localized Cenozoic basins soon after India-Asia collision in the Paleocene. The youngest remagnetization is a combination of thermoviscous and chemical remanent magnetization carried by authigenic magnetite and pyrrhotite, respectively. Our analyses suggest that a high supply of organic carbon during carbonate deposition, prevailing sulfate reducing conditions during early diagenesis, and widespread orogenic fluid migration related to crustal shortening during later diagenesis, have altered the primary remanence of the shallow-water Tethyan carbonate rocks of the Tibetan Plateau. We emphasize that all paleomagnetic results from these rocks must be carefully examined for remagnetization before being used for paleogeographic reconstructions. Future paleomagnetic investigations of the carbonate rocks in orogenic belts should be accompanied by thorough rock magnetic and petrographic studies to determine the origin of the NRM. Paleogeographic reconstructions of terranes can greatly benefit from the provenance analysis of sediments. A series of Cenozoic basins provide key sedimentary archives for investigating the growth of the Tibetan Plateau, yet the provenance of the sediments in these basins has never been constrained robustly. Here we report sedimentary petrological and detrital zircon geochronological data from the Paleocene-Eocene Nangqian-Xialaxiu and Gongjue basins. Sandstone detrital modes and zircon morphology suggest that the samples collected in these two basins were sourced from recycled orogen. Detrital zircon geochronology indicates that sediments in the Nangqian-Xialaxiu Basin are characterized by two distinct age populations at 220-280 Ma and 405-445 Ma. In contrast, three predominant age populations of 207-256 Ma, 423-445 Ma, and 1851-1868 Ma, and two subordinate age populations of similar to 50 Ma and similar to 2500 Ma, are recognized in the Gongjue Basin. Comparison with detrital zircon ages from the surrounding terranes suggests that sediments in the Nangqian-Xialaxiu Basin come from the neighboring thrust belts, whereas sediments from the Gongjue Basin are predominantly derived from the distant Songpan-Ganzi Terrane with minor contribution from the surrounding areas. A three-stage Cenozoic evolution of the eastern Tibetan Plateau is proposed. During the Paleocene, the Nangqian-Xialaxiu Basin appeared as a set of small intermontane sub-basins and received plentiful sediments from the neighboring mountain belts; during the Eocene, the Gongjue Basin kept a relatively low altitude and was a depression at the edge of a proto-Plateau; since the Oligocene, the Tibetan Plateau further uplifted and the marginal Gongjue Basin was involved in the Tibetan interior orogeny, indicating the eastward propagation of the Tibetan Plateau.
An association has been proved between high salt consumption and cardiovascular mortality. In vertebrates, the heart is the first functional organ to be formed. However, it is not clear whether high-salt exposure has an adverse impact on cardiogenesis. Here we report high-salt exposure inhibited basement membrane breakdown by affecting RhoA, thus disturbing the expression of Slug/E-cadherin/N-cadherin/Laminin and interfering with mesoderm formation during the epithelial-mesenchymal transition(EMT). Furthermore, the DiI(+) cell migration trajectory in vivo and scratch wound assays in vitro indicated that high-salt exposure restricted cell migration of cardiac progenitors, which was caused by the weaker cytoskeleton structure and unaltered corresponding adhesion junctions at HH7. Besides, down-regulation of GATA4/5/6, Nkx2.5, TBX5, and Mef2c and up-regulation of Wnt3a/-catenin caused aberrant cardiomyocyte differentiation at HH7 and HH10. High-salt exposure also inhibited cell proliferation and promoted apoptosis. Most importantly, our study revealed that excessive reactive oxygen species(ROS)generated by high salt disturbed the expression of cardiac-related genes, detrimentally affecting the above process including EMT, cell migration, differentiation, cell proliferation and apoptosis, which is the major cause of malformation of heart tubes.
A comprehensive molecular analysis of a simple aqueous complexing system. U(VI) acetate. selected to be independently investigated by various spectroscopic (vibrational, luminescence, X-ray absorption, and nuclear magnetic resonance spectroscopy) and quantum chemical methods was achieved by an international round-robin test (RRT). Twenty laboratories from six different countries with a focus on actinide or geochemical research participated and contributed to this scientific endeavor. The outcomes of this RRT were considered on two levels of complexity: first, within each technical discipline, conformities as well as discrepancies of the results and their sources were evaluated. The raw data from the different experimental approaches were found to be generally consistent. In particular, for complex setups such as accelerator-based X-ray absorption spectroscopy, the agreement between the raw data was high. By contrast, luminescence spectroscopic data turned out to be strongly related to the chosen acquisition parameters. Second, the potentials and limitations of coupling various spectroscopic and theoretical approaches for the comprehensive study of actinide molecular complexes were assessed. Previous spectroscopic data from the literature were revised and the benchmark data on the U(VI) acetate system provided an unambiguous molecular interpretation based on the correlation of spectroscopic and theoretical results. The multimethodologic approach and the conclusions drawn address not only important aspects of actinide spectroscopy but particularly general aspects of modern molecular analytical chemistry.
Inositol phosphates (IPs) and their turnover products have been implicated to play important roles in stress signaling in eukaryotic cells. In higher plants genes encoding inositol polyphosphate kinases have been identified previously, but their physiological functions have not been fully resolved. Here we expressed Arabidopsis inositol polyphosphate 6-/3-kinase (AtIpk2 beta) in two heterologous systems, i.e. the yeast Saccharomyces cerevisiae and in tobacco (Nicotiana tabacum), and tested the effect on abiotic stress tolerance. Expression of AtIpk2 beta rescued the salt-, osmotic- and temperature-sensitive growth defects of a yeast mutant strain (arg82 Delta) that lacks inositol polyphosphate multikinase activity encoded by the ARG82/IPK2 gene. Transgenic tobacco plants constitutively expressing AtIpk2 beta under the control of the Cauliflower Mosaic Virus 35S promoter were generated and found to exhibit improved tolerance to diverse abiotic stresses when compared to wild type plants. Expression patterns of various stress responsive genes were enhanced, and the activities of anti-oxidative enzymes were elevated in transgenic plants, suggesting a possible involvement of AtIpk2 beta in plant stress responses.
Morphological and genetic evidence for early Holocene cattle management in northeastern China

The domestication of cattle is generally accepted to have taken place in two independent centres: around 10,500 years ago in the Near East, giving rise to modern taurine cattle, and two millennia later in southern Asia, giving rise to zebu cattle. Here we provide firmly dated morphological and genetic evidence for early Holocene management of taurine cattle in northeastern China. We describe conjoining mandibles from this region that show evidence of oral stereotypy, dated to the early Holocene by two independent C-14 dates. Using Illumina high-throughput sequencing coupled with DNA hybridization capture, we characterize 15,406 bp of the mitogenome with on average 16.7-fold coverage. Phylogenetic analyses reveal a hitherto unknown mitochondrial haplogroup that falls outside the known taurine diversity. Our data suggest that the first attempts to manage cattle in northern China predate the introduction of domestic cattle that gave rise to the current stock by several thousand years.
Aims:This study aims to investigate the relationship between plasma endothelin-1 (ET-1) concentrations, ET-1 gene polymorphisms in loci rs5370, rs1630736, 3A/4A and clinical features of primary nephrotic syndrome (NS) in children.
Materials and methods: Thirty-six children with primary NS were selected as case group, and 94 healthy children were selected as control group. All subjects were genotyped for three single nucleotide polymorphisms (SNPs) (rs5370, rs10478694 [3A4A) and rs 1630736) in the ET-1 gene by gene sequencing. The plasma ET-1 concentrations were measured using a radio-immunoassay.
Key findings: Plasma ET-1 concentrations were higher in NS patients (P = 0.007) as compared to healthy children. The allele frequencies between control and NS patients were significantly different only with respect to the rs10478694 SNP of the ET-1 gene. The allele frequencies between control and NS patients for the rs5370 SNP showed a trend towards difference (P = 0.057). Plasma cholesterol in NS patients is associated with both: the Cl genotype in locus rs5370 and the 3A4A genotype in locus rs10478694 (P < 0.05 in both cases).
Significance: The ET systems might play a disease modifying role in pediatric NS. Plasma cholesterol, a hallmark of NS. seems to be associated with genetic variations within the human ET-1 gene. (C) 2014 Elsevier Inc. All rights reserved.
This article addresses the problem of estimating the Quality of Service (QoS) of a composite service given the QoS of the services participating in the composition. Previous solutions to this problem impose restrictions on the topology of the orchestration models, limiting their applicability to well-structured orchestration models for example. This article lifts these restrictions by proposing a method for aggregate QoS computation that deals with more general types of unstructured orchestration models. The applicability and scalability of the proposed method are validated using a collection of models from industrial practice.
Background/Aims: Angiogenesis plays a key role during embryonic development. The vascular endothelin (ET) system is involved in the regulation of angiogenesis. Lipopolysaccharides (LPS) could induce angiogenesis. The effects of ET blockers on baseline and LPS-stimulated angiogenesis during embryonic development remain unknown so far. Methods: The blood vessel density (BVD) of chorioallantoic membranes (CAMs), which were treated with saline (control), LPS, and/or BQ123 and the ETB blocker BQ788, were quantified and analyzed using an IPP 6.0 image analysis program. Moreover, the expressions of ET-1, ET-2, ET3, ET receptor A (ETRA), ET receptor B (ETRB) and VEGFR2 mRNA during embryogenesis were analyzed by semi-quantitative RT-PCR. Results: All components of the ET system are detectable during chicken embryogenesis. LPS increased angiogenesis substantially. This process was completely blocked by the treatment of a combination of the ETA receptor blockers-BQ123 and the ETB receptor blocker BQ788. This effect was accompanied by a decrease in ETRA, ETRB, and VEGFR2 gene expression. However, the baseline angiogenesis was not affected by combined ETA/ETB receptor blockade. Increasingly available high-frequency data during storm events, when hydrological dynamics most likely activate nitrate storage-flux exchanges, reveal insights into catchment nitrate dynamics. In this study, we explored impacts of seasonality and landscape gradients on nitrate concentration-discharge (C-Q) hysteresis patterns in the Selke catchment, central Germany, which has heterogeneous combinations of meteorological, hydrogeological and land use conditions. Three nested gauging stations established along the main Selke River captured flow and nitrate export dynamics from the uppermost subcatchment (mixed forest and arable land), middle subcatchment (pure steep forest) and lowermost subcatchment (arable and urban land). We collected continuous high-frequency (15-min) discharge and nitrate concentration data from 2012 to 2017 and analyzed the 223 events detected at all three stations. A dominant hysteresis pattern in the uppermost and middle subcatchments was counter-clockwise and combined with an accretion effect, indicating many proximal and mobilized distal nitrate sources. However, 66% of all events at the catchment outlet experienced a dilution effect, possibly due to mechanisms that vary seasonally. During wetting/wet periods (October-March), it was combined mainly with a counter-clockwise pattern due to the dominance of event runoff volume from the uppermost and middle subcatchments. During drying/dry periods (April-September), however, it was combined mainly with a clockwise pattern due to occasional quick surface flows from lowland near-stream urban areas. In addition, the clockwise hysteresis occurred mainly from May-October during mostly drying/dry periods at all three sites, indicating little distal nitrate transport in response to the low terrestrial hydrological connectivity, especially in the lowermost dry and flat sub-catchment. This comprehensive analysis (i.e., clockwise vs. counter-clockwise, accretion vs. dilution) enables in-depth analysis of nitrate export mechanisms during certain periods under different landscape conditions. Specific combination of C-Q relationships could identify target locations for agricultural management actions that decrease nitrate output. Therefore, we strongly encourage long-term multisite and high-frequency monitoring strategies in heterogeneous nested catchment(s), which can help understand process mechanisms, generate data for physical-based water-quality modeling and provide guidance for water and agricultural management.
Young Genes out of the Male: An Insight from Evolutionary Age Analysis of the Pollen Transcriptome

The birth of new genes in genomes is an important evolutionary event. Several studies reveal that new genes in animals tend to be preferentially expressed in male reproductive tissues such as testis (Betran et al., 2002; Begun et al., 2007; Dubruille et al., 2012), and thus an "out of testis' hypothesis for the emergence of new genes has been proposed (Vinckenbosch et al., 2006; Kaessmann, 2010). However, such phenomena have not been examined in plant species. Here, by employing a phylostratigraphic method, we dated the origin of protein-coding genes in rice and Arabidopsis thaliana and observed a number of young genes in both species. These young genes tend to encode short extracellular proteins, which may be involved in rapid evolving processes, such as reproductive barriers, species specification, and antimicrobial processes. Further analysis of transcriptome age indexes across different tissues revealed that male reproductive cells express a phylogenetically younger transcriptome than other plant tissues. Compared with sporophytic tissues, the young transcriptomes of the male gametophyte displayed greater complexity and diversity, which included a higher ratio of anti-sense and inter-genic transcripts, reflecting a pervasive transcription state that facilitated the emergence of new genes. Here, we propose that pollen may act as an "innovation incubator' for the birth of de novo genes. With cases of male-biased expression of young genes reported in animals, the "new genes out of the male' model revealed a common evolutionary force that drives reproductive barriers, species specification, and the upgrading of defensive mechanisms against pathogens.
Proxy-based reconstructions and modeling of Holocene spatiotemporal precipitation patterns for China and Mongolia have hitherto yielded contradictory results indicating that the basic mechanisms behind the East Asian Summer Monsoon and its interaction with the westerly jet stream remain poorly understood. We present quantitative reconstructions of Holocene precipitation derived from 101 fossil pollen records and analyse them with the help of a minimal empirical model. We show that the westerly jet-stream axis shifted gradually southward and became less tilted since the middle Holocene. This was tracked by the summer monsoon rain band resulting in an early-Holocene precipitation maximum over most of western China, a mid-Holocene maximum in north-central and northeastern China, and a late-Holocene maximum in southeastern China. Our results suggest that a correct simulation of the orientation and position of the westerly jet stream is crucial to the reliable prediction of precipitation patterns in China and Mongolia.
Aggregation of chromophores in the solid state commonly causes undesirable red shifts in the emission spectra and/or emission quenching. To overcome this problem, we have prepared soluble perylenetetracarboxidiimide dyes in which the chromophores are effectively shielded by polyphenylene dendrimers attached in the bay positions. Models show that attachment of the shielding units in the bay position should provide more efficient shielding than attaching them via the imide moieties. The dendrimers possess excellent film-forming properties due to alkyl substituents on their peripheries. The lack of a red shift in emission upon going from solution to the solid state indicates the dendrons suppress interaction of the emissive cores, leading to pure red-orange emission. Single-layer LEDs produce red-orange emission with relatively low efficiency especially for the higher generation dendrons, which is attributed to poor charge conduction. LEDs using blends of the dendrimers and the undendronized dye as a model compound in PVK have been investigated, and a model to extract relative charge injection rates through the dendritic scaffold from the spectral contributions in the EL spectra is developed
Alpine ecosystems on the Tibetan Plateau are being threatened by ongoing climate warming and intensified human activities. Ecological time-series obtained from sedimentary ancient DNA (sedaDNA) are essential for understanding past ecosystem and biodiversity dynamics on the Tibetan Plateau and their responses to climate change at a high taxonomic resolution. Hitherto only few but promising studies have been published on this topic. The potential and limitations of using sedaDNA on the Tibetan Plateau are not fully understood. Here, we (i) provide updated knowledge of and a brief introduction to the suitable archives, region-specific taphonomy, state-of-the-art methodologies, and research questions of sedaDNA on the Tibetan Plateau; (ii) review published and ongoing sedaDNA studies from the Tibetan Plateau; and (iii) give some recommendations for future sedaDNA study designs. Based on the current knowledge of taphonomy, we infer that deep glacial lakes with freshwater and high clay sediment input, such as those from the southern and southeastern Tibetan Plateau, may have a high potential for sedaDNA studies. Metabarcoding (for microorganisms and plants), metagenomics (for ecosystems), and hybridization capture (for prehistoric humans) are three primary sedaDNA approaches which have been successfully applied on the Tibetan Plateau, but their power is still limited by several technical issues, such as PCR bias and incompleteness of taxonomic reference databases. Setting up high-quality and open-access regional taxonomic reference databases for the Tibetan Plateau should be given priority in the future. To conclude, the archival, taphonomic, and methodological conditions of the Tibetan Plateau are favorable for performing sedaDNA studies. More research should be encouraged to address questions about long-term ecological dynamics at ecosystem scale and to bring the paleoecology of the Tibetan Plateau into a new era.
Temporally changing drivers for late-Holocene vegetation changes on the northern Tibetan Plateau

Fossil pollen records have been widely used as indicators of past changes in vegetation and variations in climate. The driving mechanisms behind these vegetation changes have, however, remained unclear. In order to evaluate vegetation changes that have occurred in the northern part of the Tibetan Plateau and the possible drivers behind these changes, we have applied a moving-window Redundancy Analysis (RDA) to high resolution (10-15 years) pollen and sedimentary data from Lake Kusai covering the last 3770 years. Our analyses reveal frequent fluctuations in the relative abundances of alpine steppe and alpine desert components. The sedimentary proxies (including total organic carbon content, total inorganic carbon content, and "end-member" indices from grain-size analyses) that explain statistically some of the changes in the pollen assemblage vary significantly with time, most probably reflecting multiple underlying driving processes. Climate appears to have had an important influence on vegetation changes when conditions were relatively wet and stable. However, a gradual decrease in vegetation cover was identified after 1500 cal a BP, after which the vegetation appears to have been affected more by extreme events such as dust-storms or fluvial erosion than by general climatic trends. Furthermore, pollen spectra over the last 600 years are shown by Procrustes analysis to be statistically different from those recovered from older samples, which we attribute to increased human impact that resulted in unprecedented changes to the vegetation composition. Overall, changes in vegetation and climate on the northern part of the Tibetan Plateau appear to have roughly followed the evolution of the Asian Summer Monsoon. After taking into account the highly significant millennial (1512 years) periodicity revealed by time-series analysis, the regional vegetation and climate changes also show variations that appear to match variations in the mid-latitude westerlies.
The retinitis pigmentosa 2 polypeptide (RP2) functions as a GTPase-activating protein (GAP) for ARL3 (Arf-like protein 3), a small GTPase. ARL3 is an effector of phosphodiesterase 6 Delta (PDE6D), a prenyl-binding protein and chaperone of prenylated protein in photoreceptors. Mutations in the human RP2 gene cause X-linked retinitis pigmentosa (XLRP) and cone-rod dystrophy (XL-CORD). To study mechanisms causing XLRP, we generated an RP2 knockout mouse. The RP2h(-/-) mice exhibited a slowly progressing rod-cone dystrophy simulating the human disease. RP2h(-/-) scotopic a-wave and photopic b-wave amplitudes declined at 1 mo of age and continued to decline over the next 6 mo. Prenylated PDE6 subunits and G-protein coupled receptor kinase 1 (GRK1) were unable to traffic effectively to the RP2h(-/-) outer segments. Mechanistically, absence of RP2 GAP activity increases ARL3-GTP levels, forcing PDE6D to assume a predominantly "closed" conformation that impedes binding of lipids. Lack of interaction disrupts trafficking of PDE6 and GRK1 to their destination, the photoreceptor outer segments. We propose that hyperactivity of ARL3-GTP in RP2 knockout mice and human patients with RP2 null alleles leads to XLRP resembling recessive rod-cone dystrophy.
The Tian Shan range is an inherited intracontinental structure reactivated by the far-field effects of the India-Asia collision. A growing body of thermochronology and magnetostratigraphy datasets shows that the range grew through several tectonic pulses since similar to 25 Ma, however the early Cenozoic history remains poorly constrained. The time-lag between the Eocene India-Asia collision and the Miocene onset of Tian Shan exhumation is particularly enigmatic. This peculiar period is potentially recorded along the southwestern Tian Shan piedmont. There, late Eocene marine deposits of the proto-Paratethys epicontinental sea transition to continental foreland basin sediments of unknown age were recently dated. We provide magnetostratigraphic dating of these continental sediments from the 1700-m-thick Mine section integrated with previously published detrital apatite fission track and U/Pb zircon ages. The most likely correlation to the geomagnetic polarity time scale indicates an age span from 20.8 to 13.3 Ma with a marked increase in accumulation rates at 19-18 Ma. This implies that the entire Oligocene period is missing between the last marine and first continental sediments, as suggested by previous southwestern Tian Shan results. This differs from the southwestern Tarim basin where Eocene marine deposits are continuously overlain by late Eocene-Oligocene continental sediments. This supports a simple evolution model of the western Tarim basin with Eocene-Oligocene foreland basin activation to the south related to northward thrusting of the Kunlun Shan, followed by early Miocene activation of northern foreland basin related to overthrusting of the south Tian Shan. Our data also support southward propagation of the Tian Shan piedmont from 20 to 18 Ma that may relate to motion on the Talas Fergana Fault. The coeval activation of a major right-lateral strike-slip system allowing indentation of the Pamir Salient into the Tarim basin, suggests far-field deformation from the India-Asia collision zone affected the Tian Shan and the Talas Fergana fault by early Miocene. Rapid decline of glomerular filtration rate estimated from creatinine (eGFRcrea) is associated with severe clinical endpoints. In contrast to cross-sectionally assessed eGFRcrea, the genetic basis for rapid eGFRcrea decline is largely unknown. To help define this, we meta-analyzed 42 genome-wide association studies from the Chronic Kidney Diseases Genetics Consortium and United Kingdom Biobank to identify genetic loci for rapid eGFRcrea decline. Two definitions of eGFRcrea decline were used: 3 mL/min/1.73m(2)/year or more ("Rapid3"; encompassing 34,874 cases, 107,090 controls) and eGFRcrea decline 25% or more and eGFRcrea under 60 mL/min/1.73m(2) at follow-up among those with eGFRcrea 60 mL/min/1.73m(2) or more at baseline ("CKDi25"; encompassing 19,901 cases, 175,244 controls). Seven independent variants were identified across six loci for Rapid3 and/or CKDi25: consisting of five variants at four loci with genome-wide significance (near UMOD-PDILT (2), PRKAG2, WDR72, OR2S2) and two variants among 265 known eGFRcrea variants (near GATM, LARP4B). All these loci were novel for Rapid3 and/or CKDi25 and our bioinformatic follow-up prioritized variants and genes underneath these loci. The OR2S2 locus is novel for any eGFRcrea trait including interesting candidates. For the five genome-wide significant lead variants, we found supporting effects for annual change in blood urea nitrogen or cystatin-based eGFR, but not for GATM or (LARP4B). Individuals at high compared to those at low genetic risk (8-14 vs. 0-5 adverse alleles) had a 1.20-fold increased risk of acute kidney injury (95% confidence interval 1.08-1.33). Thus, our identified loci for rapid kidney function decline may help prioritize therapeutic targets and identify mechanisms and individuals at risk for sustained deterioration of kidney function.
A catalog of genetic loci associated with kidney function from analyses of a million individuals

Chronic kidney disease (CKD) is responsible for a public health burden with multi-systemic complications. Through transancestry meta-analysis of genome-wide association studies of estimated glomerular filtration rate (eGFR) and independent replication (n = 1,046,070), we identified 264 associated loci (166 new). Of these,147 were likely to be relevant for kidney function on the basis of associations with the alternative kidney function marker blood urea nitrogen (n = 416,178). Pathway and enrichment analyses, including mouse models with renal phenotypes, support the kidney as the main target organ. A genetic risk score for lower eGFR was associated with clinically diagnosed CKD in 452,264 independent individuals. Colocalization analyses of associations with eGFR among 783,978 European-ancestry individuals and gene expression across 46 human tissues, including tubulo-interstitial and glomerular kidney compartments, identified 17 genes differentially expressed in kidney. Fine-mapping highlighted missense driver variants in 11 genes and kidney-specific regulatory variants. These results provide a comprehensive priority list of molecular targets for translational research.

The establishment and evolution of the Asian monsoons and arid interior have been linked to uplift of the Tibetan Plateau, retreat of the inland proto-Paratethys Sea and global cooling during the Cenozoic. However, the respective role of these driving mechanisms remains poorly constrained. This is partly due to a lack of continental records covering the key Eocene epoch marked by the onset of Tibetan Plateau uplift, proto-Paratethys Sea incursions and long-term global cooling. In this study, we reconstruct paleoenvironments in the Xining Basin, NE Tibet, to show a long-term drying of the Asian continental interior from the early Eocene to the Oligocene. Superimposed on this trend are three alternations between arid mudflat and wetter saline lake intervals, which are interpreted to reflect atmospheric moisture fluctuations in the basin. We date these fluctuations using magnetostratigraphy and the radiometric age of an intercalated tuff layer. The first saline lake interval is tentatively constrained to the late Paleocene-early Eocene. The other two are firmly dated between similar to 46 Ma (top magnetochron C21n) and similar to 41 Ma (base C18r) and between similar to 40 Ma (base C18n) and similar to 37 Ma (top C17n). Remarkably, these phases correlate in time with highstands of the proto-Paratethys Sea. This strongly suggests that these sea incursions enhanced westerly moisture supply as far inland as the Xining Basin. We conclude that the proto-Paratethys Sea constituted a key driver of Asian climate and should be considered in model and proxy interpretations. (C) 2019 Elsevier B.V. All rights reserved.
Biodegradation of polyester polyurethane by the marine fungus Cladosporium halotolerans 6UPA1

Lack of degradability and the accumulation of polymeric wastes increase the risk for the health of the environment. Recently, recycling of polymeric waste materials becomes increasingly important as raw materials for polymer synthesis are in short supply due to the rise in price and supply chain disruptions. As an important polymer, polyurethane (PU) is widely used in modern life, therefore, PU biodegradation is desirable to avoid its accumulation in the environment. In this study, we isolated a fungal strain Cladosporium halotolerans from the deep sea which can grow in mineral medium with a polyester PU (Impranil DLN) as a sole carbon source. Further, we demonstrate that it can degrade up to 80% of Impranil PU after 3 days of incubation at 28 celcius by breaking the carbonyl groups (1732 cm(-1)) and C-N-H bonds (1532 cm(-1) and 1247 cm(-1)) as confirmed by Fourier-transform infrared (FTIR) spectroscopy analysis. Gas chromatography-mass spectrometry (GC-MS) analysis revealed polyols and alkanes as PU degradation intermediates, indicating the hydrolysis of ester and urethane bonds. Esterase and urease activities were detected in 7 days-old cultures with PU as a carbon source. Transcriptome analysis showed a number of extracellular protein genes coding for enzymes such as cutinase, lipase, peroxidase and hydrophobic surface binding proteins A (HsbA) were expressed when cultivated on Impranil PU. The yeast two-hybrid assay revealed that the hydrophobic surface binding protein ChHsbA1 directly interacts with inducible esterases, ChLip1 (lipase) and ChCut1 (cutinase). Further, the KEGG pathway for "fatty acid degradation " was significantly enriched in Impranil PU inducible genes, indicating that the fungus may use the degradation intermediates to generate energy via this pathway. Taken together, our data indicates secretion of both esterase and hydrophobic surface binding proteins by C. halotolerans plays an important role in Impranil PU absorption and subsequent degradation. Our study provides a mechanistic insight into Impranil PU biodegradation by deep sea fungi and provides the basis for future development of biotechnological PU recycling.
