Biosensors representing the technological counterpart of living senses have found routine application in amperometric enzyme electrodes for decentralized blood glucose measurement, interaction analysis by surface plasmon resonance in drug development, and to some extent DNA chips for expression analysis and enzyme polymorphisms. These technologies have already reached a highly advanced level and need minor improvement at most. The dream of the "100-dollar' personal genome may come true in the next few years provided that the technological hurdles of nanopore technology or of polymerase-based single molecule sequencing can be overcome. Tailor-made recognition elements for biosensors including membrane-bound enzymes and receptors will be prepared by cell-free protein synthesis. As alternatives for biological recognition elements, molecularly imprinted polymers (MIPs) have been created. They have the potential to substitute antibodies in biosensors and biochips for the measurement of low-molecular-weight substances, proteins, viruses, and living cells. They are more stable than proteins and can be produced in large amounts by chemical synthesis. Integration of nanomaterials, especially of graphene, could lead to new miniaturized biosensors with high sensitivity and ultrafast response. In the future individual therapy will include genetic profiling of isoenzymes and polymorphic forms of drug-metabolizing enzymes especially of the cytochrome P450 family. For defining the pharmacokinetics including the clearance of a given genotype enzyme electrodes will be a useful tool. For decentralized online patient control or the integration into everyday "consumables' such as drinking water, foods, hygienic articles, clothing, or for control of air conditioners in buildings and cars and swimming pools, a new generation of "autonomous' biosensors will emerge.
Was wird unter „nachhaltiger Prävention“ in der Präventionsforschung verstanden? Welche guten Beispiele für nachhaltige Prävention gibt es in der Praxis? Und v. a.: Wie lässt sich Prävention in den verschiedenen Bereichen wie Kriminalität, Gewalt und Rechtsextremismus nachhaltig gestalten? Diesen Fragen will der vorliegende Sammelband nachgehen und damit der Präventionsdebatte neue Impulse verleihen. Der Band will insbesondere die nationale sowie internationale Fachdebatte konstruktiv aufgreifen, Theorie und Praxis verbinden, „good practice“ Beispiele darstellen sowie Perspektiven nachhaltiger Prävention aufzeigen. Mit diesem Themenspektrum richtet er sich sowohl an die Wissenschaft als auch an die Praxis sowie insgesamt an eine interessierte Öffentlichkeit.
The concept of bonding and antibonding orbitals is fundamental in chemistry. The population of those orbitals and the energetic difference between the two reflect the strength of the bonding interaction. Weakening the bond is expected to reduce this energetic splitting, but the transient character of bond-activation has so far prohibited direct experimental access. Here we apply time-resolved soft X-ray spectroscopy at a free electron laser to directly observe the decreased bonding antibonding splitting following bond-activation using an ultrashort optical laser pulse.
An English double-embedded relative clause from which the middle verb is omitted can often be processed more easily than its grammatical counterpart, a phenomenon known as the grammaticality illusion. This effect has been found to be reversed in German, suggesting that the illusion is language specific rather than a consequence of universal working memory constraints. We present results from three self-paced reading experiments which show that Dutch native speakers also do not show the grammaticality illusion in Dutch, whereas both German and Dutch native speakers do show the illusion when reading English sentences. These findings provide evidence against working memory constraints as an explanation for the observed effect in English. We propose an alternative account based on the statistical patterns of the languages involved. In support of this alternative, a single recurrent neural network model that is trained on both Dutch and English sentences is shown to predict the cross-linguistic difference in the grammaticality effect.
Cell-free protein synthesis (CFPS) is a valuable method for the fast expression of difficult-to-express proteins as well as posttranslationally modified proteins. Since cell-free systems circumvent possible cytotoxic effects caused by protein overexpression in living cells, they significantly enlarge the scale and variety of proteins that can be characterized. We demonstrate the high potential of eukaryotic CFPS to express various types of membrane proteins covering a broad range of structurally and functionally diverse proteins. Our eukaryotic cell-free translation systems are capable to provide high molecular weight membrane proteins, fluorescent-labeled membrane proteins, as well as posttranslationally modified proteins for further downstream analysis.
Hazelnuts are rarely cultivated in Germany, although they are a valuable source for macro- and micronutrients and can thus contribute to a healthy diet. Near the present, 15 varieties were cultivated in Thuringia, Germany, as a pilot study for further research. The aim of our study was to evaluate the micro- and macronutrient composition of representative, randomly mixed samples of the 15 different hazelnut cultivars. Protein, fat, and fiber contents were determined using established methods. Fatty acids, tocopherols, minerals, trace elements, and ultra-trace elements were analyzed using gas chromatography, high-performance liquid chromatography, and inductively coupled plasma triple quadrupole mass-spectrometry, respectively. We found that the different hazelnut varieties contained valuable amounts of fat, protein, dietary fiber, minerals, trace elements, and alpha-tocopherol, however, in different quantities. The variations in nutrient composition were independent of growth conditions, which were identical for all hazelnut varieties. Therefore, each hazelnut cultivar has its specific nutrient profile.
We propose simple and fast methods based on nearest neighbors that order objects from high-dimensional data sets from typical points to untypical points. On the one hand, we show that these easy-to-compute orderings allow us to detect outliers (i.e. very untypical points) with a performance comparable to or better than other often much more sophisticated methods. On the other hand, we show how to use these orderings to detect prototypes (very typical points) which facilitate exploratory data analysis algorithms such as noisy nonlinear dimensionality reduction and clustering. Comprehensive experiments demonstrate the validity of our approach.
We used single-molecule FRET in combination with other biophysical methods and molecular simulations to investigate the effect of temperature on the dimensions of unfolded proteins. With singlemolecule FRET, this question can be addressed even under nearnative conditions, where most molecules are folded, allowing us to probe a wide range of denaturant concentrations and temperatures. We find a compaction of the unfolded state of a small cold shock protein with increasing temperature in both the presence and the absence of denaturant, with good agreement between the results from single-molecule FRET and dynamic light scattering. Although dissociation of denaturant from the polypeptide chain with increasing temperature accounts for part of the compaction, the results indicate an important role for additional temperaturedependent interactions within the unfolded chain. The observation of a collapse of a similar extent in the extremely hydrophilic, intrinsically disordered protein prothymosin suggests that the hydrophobic effect is not the sole source of the underlying interactions. Circular dichroism spectroscopy and replica exchange molecular dynamics simulations in explicit water show changes in secondary structure content with increasing temperature and suggest a contribution of intramolecular hydrogen bonding to unfolded state collapse.
Effects of isoflavones on estrogen sensitive tissues are discussed controversially This study was designed to investigate tissue specific effects of an isoflavone exposure through different periods of life in female Wistar rats and to compare the effects of genistein (GEN) to those of mixed dietary isoflavones, GEN and daidzein (DAI). One group received an isoflavone-free diet (IDD). another was fed an isoflavone-rich diet (IRD) and the third group an IDD supplemented with GEN (GEN(d)) prior to mating. throughout pregnancy and up to weaning The offspring were kept on the respective diets during growth. puberty and adulthood The weight of the uterus, the height of the uterine and vaginal epithelium, the bone mineral density of the tibia, and the expression of the estrogen sensitive gene CaBP9K in the liver were determined. At d21, the uterine weight, the uterine epithelium and the expression of CaBP9K in the liver were significantly stimulated in GEN(d) animals compared to IDD and IRD Interestingly, bone mineral density was increased in GEN(d) and in IRD animals Around puberty (d50) neither uterine wet weights nor trabecular bone density differed significantly among the isoflavone groups and the IDD control. At d80 nosignificant differences in uterine weight were observed among IDD. GEN(d) and IRD animals. However, bone mineral density was increased in GEN(d) and IRD animals In summary, Our results demonstrate that lifelong dietary exposure to isoflavones can affect estrogen sensitive tissues, apparently in a tissue selective manner With respect to health risk and benefit our data indicate that an increased bone mineral density can be achieved by lifelong exposure to an IRD. which, in contrast to GEN supplementation, does not seem to stimulate the proliferation of the uterine epithelium
The most recent intense earthquake swarm in West Bohemia lasted from 6 October 2008 to January 2009. Starting 12 days after the onset, the University of Potsdam monitored the swarm by a temporary small-aperture seismic array at 10 km epicentral distance. The purpose of the installation was a complete monitoring of the swarm including micro-earthquakes (M (L) < 0). We identify earthquakes using a conventional short-term average/long-term average trigger combined with sliding-window frequency-wavenumber and polarisation analyses. The resulting earthquake catalogue consists of 14,530 earthquakes between 19 October 2008 and 18 March 2009 with magnitudes in the range of -aEuro parts per thousand 1.2 a parts per thousand currency signaEuro parts per thousand M (L) a parts per thousand currency signaEuro parts per thousand 2.7. The small-aperture seismic array substantially lowers the detection threshold to about M (c) = -aEuro parts per thousand 0.4, when compared to the regional networks operating in West Bohemia (M (c) > 0.0). In the course of this work, the main temporal features (frequency-magnitude distribution, propagation of back azimuth and horizontal slowness, occurrence rate of aftershock sequences and interevent-time distribution) of the recent 2008/2009 earthquake swarm are presented and discussed. Temporal changes of the coefficient of variation (based on interevent times) suggest that the swarm earthquake activity of the 2008/2009 swarm terminates by 12 January 2009. During the main phase in our studied swarm period after 19 October, the b value of the Gutenberg-Richter relation decreases from 1.2 to 0.8. This trend is also reflected in the power-law behavior of the seismic moment release. The corresponding total seismic moment release of 1.02x10(17) Nm is equivalent to M (L,max) = 5.4.
