Speleothems, secondary cave carbonates, are important tools for climate reconstruction, especially as they often can be very precisely dated with the UTh method. If the U-Th method fails, dating becomes difficult, and often results in abandonment of samples and study sites. Radiocarbon dating is the only other radiometric dating technique applicable to the last similar to 50 ka, but presents complexities related to temporal variability of the reservoir effect in speleothems. Thus, radiocarbon dating of speleothems is not straightforward, and there are currently no publicly available tools to define proper age-depth relationships with this method. Here, we present an improved version of a previously published radiocarbon based age-depth modelling approach (star, Lechleitner et al., 2016b), which is now made freely available. The software is easy to use and provides the possibility to obtain reliable age-depth relationships, without prior knowledge of reservoir effects and their variability. In addition, star is able to detect and handle growth stops and phases with different growth rates. We test star on artificially constructed data sets and illustrate steps to improve the model performance. Furthermore, we apply the new approach to published radiocarbon data of U-Th dated stalagmites. This offers the possibility to investigate the strengths and weaknesses of the new approach with respect to potentially significant long term trends in the radiocarbon reservoir effect, which might otherwise remain undetected. In summary, we have produced a valuable software, which easily enables to construct age-depth relationships on the basis of reservoir effect disturbed radiocarbon measurements.
Light quality-induced changes of carotenoid composition in pak choi Brassica rapa ssp. chinensis

Carotenoids as part of the photosystems are crucial for their assembly, light-harvesting, and photoprotection. Light of different wavelengths impacts the composition and structure of photosystems, thus offering the possibility to influence the carotenoid concentrations and composition in photosystems by illumination with specific narrow-banded light spectra. Key components involved in the regulation of gene transcription are still poorly characterized, particularly in leafy vegetables as compared to model plants. In particular, the effect of different light qualities and its connection to redox control mechanisms, which also determine the photosystem composition and structure, is not yet well understood. Furthermore, light quality effects are species-dependent, and thus, increase the need to perform research on individual vegetable species such as pak choi Brassica rapa ssp. chinensis. Here, we investigated the carotenoid concentrations and composition of pak choi sprouts grown for 6 days under blue, red, or white light emitting diodes (LEDs) as light source. After 6 days, the total carotenoid content was the highest under white and slightly reduced under blue or red LEDs. Blue, red, and white light differently affected the carotenoid composition mainly due to variations of the beta-carotene content which could be correlated to changes in the transcript levels of beta-carotene hydroxylase 1 (beta-OHASE1). Further investigations implied a redox controlled gene expression of beta-OHASE1. In addition, transcription factors related to light signaling and the circadian clock differed in their transcriptional abundance after exposure to blue and red light. RNA-Seq analysis also revealed increased transcript levels of genes encoding the outer antenna complex of photosystem II under red compared to blue light, indicating an adjustment of the photosystems to the different light qualities which possibly contributed to the alternations in the carotenoid content and composition.
Enterprises reach out for collaborations with other organizations in order to offer complex products and services to the market. Such collaboration and coordination between different organizations, for a good share, is facilitated by information technology. The BPMN process choreography is a modeling language for specifying the exchange of information and services between different organizations at the business level. Recently, there is a surging use of the REST architectural style for the provisioning of services on the web, but few systematic engineering approach to design their collaboration. In this paper, we address this gap in a comprehensive way by defining a semi-automatic method for the derivation of RESTful choreographies from process choreographies. The method is based on natural language analysis techniques to derive interactions from the textual information in process choreographies. The proposed method is evaluated in terms of effectiveness resulting in the intervention of a web engineer in only about 10% of all generated RESTful interactions.
Transient receptor potential (TRP) channels have emerged as potential sensors and transducers of inflammatory pain. The aims of this study were to investigate (1) the expression of TRP channels in intervertebral disc (IVD) cells in normal and inflammatory conditions and (2) the function of Transient receptor potential ankyrin 1 (TRPA1) and Transient receptor potential vanilloid 1 (TRPV1) in IVD inflammation and matrix homeostasis. RT-qPCR was used to analyze human fetal, healthy, and degenerated IVD tissues for the gene expression of TRPA1 and TRPV1. The primary IVD cell cultures were stimulated with either interleukin-1 beta (IL-1) or tumor necrosis factor alpha (TNF-) alone or in combination with TRPA1/V1 agonist allyl isothiocyanate (AITC, 3 and 10 mu M), followed by analysis of calcium flux and the expression of inflammation mediators (RT-qPCR/ELISA) and matrix constituents (RT-qPCR). The matrix structure and composition in caudal motion segments from TRPA1 and TRPV1 wild-type (WT) and knock-out (KO) mice was visualized by FAST staining. Gene expression of other TRP channels (A1, C1, C3, C6, V1, V2, V4, V6, M2, M7, M8) was also tested in cytokine-treated cells. TRPA1 was expressed in fetal IVD cells, 20% of degenerated IVDs, but not in healthy mature IVDs. TRPA1 expression was not detectable in untreated cells and it increased upon cytokine treatment, while TRPV1 was expressed and concomitantly reduced. In inflamed IVD cells, 10 mu M AITC activated calcium flux, induced gene expression of IL-8, and reduced disintegrin and metalloproteinase with thrombospondin motifs 5 (ADAMTS5) and collagen 1A1, possibly via upregulated TRPA1. TRPA1 KO in mice was associated with signs of degeneration in the nucleus pulposus and the vertebral growth plate, whereas TRPV1 KO did not show profound changes. Cytokine treatment also affected the gene expression of TRPV2 (increase), TRPV4 (increase), and TRPC6 (decrease). TRPA1 might be expressed in developing IVD, downregulated during its maturation, and upregulated again in degenerative disc disease, participating in matrix homeostasis. However, follow-up studies with larger sample sizes are needed to fully elucidate the role of TRPA1 and other TRP channels in degenerative disc disease.
French participants learned English pseudowords either with the orthographic form displayed under the corresponding picture (Audio-Ortho) or without (Audio). In a naming task, pseudowords learned in the Audio-Ortho condition were produced faster and with fewer errors, providing a first piece of evidence that orthographic information facilitates the learning and on-line retrieval of productive vocabulary in a second language. Formant analyses, however, showed that productions from the Audio-Ortho condition were more French-like (i.e., less target-like), a result confirmed by a vowel categorization task performed by native speakers of English. It is argued that novel word learning and pronunciation accuracy should be considered together. (C) 2019 Acoustical Society of America
Models of ring current electron dynamics unavoidably contain uncertainties in boundary conditions, electric and magnetic fields, electron scattering rates, and plasmapause location. Model errors can accumulate with time and result in significant deviations of model predictions from observations. Data assimilation offers useful tools which can combine physics-based models and measurements to improve model predictions. In this study, we systematically analyze performance of the Kalman filter applied to a log-transformed convection model of ring current electrons and Van Allen Probe data. We consider long-term dynamics of mu = 2.3 MeV/G and K = 0.3 G(1/2) R-E electrons from 1 February 2013 to 16 June 2013. By using synthetic data, we show that the Kalman filter is capable of correcting errors in model predictions associated with uncertainties in electron lifetimes, boundary conditions, and convection electric fields. We demonstrate that reanalysis retains features which cannot be fully reproduced by the convection model such as storm-time earthward propagation of the electrons down to 2.5 R-E. The Kalman filter can adjust model predictions to satellite measurements even in regions where data are not available. We show that the Kalman filter can adjust model predictions in accordance with observations for mu = 0.1, 2.3, and 9.9 MeV/G and constant K = 0.3 G(1/2) R-E electrons. The results of this study demonstrate that data assimilation can improve performance of ring current models, better quantify model uncertainties, and help deeper understand the physics of the ring current particles.
Cognitive representations and beliefs are what comprise an individual’s self-concept. A positive self-concept is related to and influences academic achievement, and the relationship between a domain-specific self-concept and achievement in the same domain is positive and strong. However, insufficient attention has been paid to these issues among multilingual children. More importantly, since instruction strongly contributes to the development of metacognition and executive functions (EFs), and since the bilingual advantage hypothesis holds that the constant management of multiple languages entails benefits for EF, we bring together these important issues in the present study. We examine the relationship between domain-specific self-concepts and standardized assessment of reading and spelling competences against the background of potential differences in self-concept between monolingual and multilingual German children. While between-group comparisons revealed no significant differences for self-concept nor reading competency, monolinguals outperformed multilinguals in spelling. Correlations between domain-specific self-concepts and academic achievement in reading comprehension, reading fluency, and spelling were positive and significant for both groups. Regardless of language background, children’s evaluations of their academic achievement (reading and spelling) were realistic. We argue, on a theoretical basis, that metacognition and EFs could facilitate a bilingual advantage and improve educational outcomes.
