Öffentlich Private Partnerschaften (ÖPPs) haben in den letzten zehn bis fünfzehn Jahren in Deutschland einen beachtlichen Stellenwert erreicht. Auch zukünftig ist aufgrund der Finanzkrise der Kommunen mit einem weiteren Bedeutungszuwachs zu rechnen. Damit ÖPPs die von der öffentlichen Hand gewünschten Vorteile mit sich bringen können, wie zum Beispiel die Entlastung des öffentlichen Haushalts oder Effizienzsteigerungen, sollten sie im Vorfeld und im Tagesgeschäft aktiv und umsichtig begleitet werden. In diesem Zusammenhang ergibt sich der Ansatzpunkt für die Themenstellung der Dissertation sowie angesichts der Erkenntnis, dass bisher noch keine umfassend fundierten und systematischen Untersuchungen vorliegen, welche die bestehenden Praxiserfahrungen mit ÖPPs mit anwendbaren Theorien in Verbindung setzen und Entscheidungshilfen für öffentliche Akteure ableiten. Aufgrund der verschiedenen möglichen Ausprägungsformen wurde eine Eingrenzung des Themas auf institutionelle ÖPPs auf kommunaler Ebene vorgenommen. Die Untersuchung beginnt mit der Auseinandersetzung der Grundlagen zu ÖPPs, um ein generelles Verständnis für dieses Themengebiet zu schaffen. Nachdem der Begriff erläutert und Merkmale von ÖPPs herausgearbeitet wurden, erfolgt eine Abgrenzung zwischen vertraglichen und institutionellen ÖPPs. Daraufhin werden mögliche Motive der öffentlichen und privaten Seite beim Eingehen einer solchen Partnerschaft aufgeführt sowie erste mögliche Chancen und Risiken skizziert. Im Anschluss erfolgt mit Hilfe der wissenschaftlichen Theorie des Neuen Institutionalismus eine vertiefende Analyse zu institutionellen ÖPPs. Dabei schließt sich die Dissertation an die von Mayntz und Scharpf vorgenommene Einteilung in einen ökonomischen, (organisations-) soziologischen und politikwissenschaftlichen Neo-Institutionalismus an. Der Neue Ökonomische Institutionalismus wurde anhand der drei Teillehren Transaktionskostentheorie, Prinzipal-Agent-Theorie und Theorie der Verfügungsrechte untersucht. Zunächst werden theoretische Erkenntnisse zu den einzelnen Theorien herausgearbeitet und erste Schlussfolgerungen für institutionelle ÖPPs gezogen. Daraus werden nachfolgend Untersuchungskriterien in Form von Fragestellungen für den späteren Fallstudienvergleich entwickelt. Nach Abschluss des Theorieteils erfolgt eine Betrachtung institutioneller ÖPPs aus realer empirischer Sicht. Hierzu werden Fallstudien vorgestellt und an den Untersuchungskriterien, welche aus den einzelnen Theorien abgeleitet wurden, gespiegelt. Zuerst werden recherchierte Fallstudien analysiert, beginnend mit den Teilprivatisierungen der Stadtentwässerung Dresden GmbH und der Stadtwerke Görlitz AG, bei denen sich die Zusammenarbeit wohl positiv entwickelt. Als Negativbeispiel wird dann auf die Privatisierung der Wasserversorgung von Grenoble und ihre spätere Rekommunalisierung eingegangen. Im folgenden Schritt werden Fallstudien aus den realen Erfahrungen des Verfassers diskutiert. Hierbei bildet die Teilprivatisierung und anschließende Rekommunalisierung des Wasserbetriebes in Potsdam den Schwerpunkt. Ergänzt wird dies durch die Darstellung der positiven Zusammenarbeit mit dem privaten Gesellschafter bei der Energie und Wasser Potsdam GmbH. Abschließend werden die anfänglichen Probleme zwischen Kommune und Privat bei der teilprivatisierten STEP Stadtentsorgung Potsdam untersucht und aufgezeigt, wie die Partnerschaft zum Vorteil der öffentlichen Seite verändert wurde. Aus dem Vergleich von Theorie und Praxis konnten wissenschaftlich fundierte Schlussfolgerungen für institutionelle ÖPPs gezogen und Erfolgsfaktoren für das Gelingen einer solchen Kooperation abgeleitet werden. Die gewonnenen Erkenntnisse werden in Form von Thesen zusammengefasst und dienen als Basis für die Ableitung von Handlungsempfehlungen für kommunale Akteure beim Eingehen einer institutionellen ÖPP. Zu Beginn erfolgt eine Darstellung der Empfehlungen, die sich aus den Untersuchungskriterien der jeweiligen Theorien ergeben haben. Nachfolgend wird diese Betrachtung erweitert, indem näher auf die wesentlichen Phasen des Entscheidungsfindungsprozesses eingegangen und eine Untersetzung dieser Phasen mit den erarbeiteten Handlungsempfehlungen vorgenommen wird. Auf diese Weise kann den kommunalen Entscheidungsträgern eine sehr praxisnahe Hilfestellung gegeben werden. Insgesamt betrachtet, geht aus der Dissertation ein umfangreicher, fundierter und sehr praxisrelevanter Leitfaden hervor, der wichtige Anhaltspunkte für das Eingehen einer institutionellen ÖPP im kommunalen Bereich gibt. Aus der Spiegelung von Theorie und Praxis werden wertvolle Hinweise abgeleitet, wodurch insbesondere deutlich wird, an welchen Stellen sich die öffentliche Seite absichern sollte. Darüber hinaus können die kommunalen Entscheidungsträger durch die gewonnenen Erkenntnisse sensibilisiert und ihr Blick für den individuellen Fall geschärft werden. Letztendlich werden dadurch wichtige Voraussetzungen geschaffen, um ein solches Vorhaben zum Erfolg zu führen.
To calibrate delta O-18 time-series from speleothems in the eastern Indian summer monsoon (ISM) region of India, and to understand the moisture regime over the northern Bay of Bengal (BoB) we analyze the delta O-18 and delta D of rainwater, collected in 2007 and 2008 near Cherrapunji, India. delta D values range from + 18.5 parts per thousand to 144.4 parts per thousand, while delta O-18 varies between +0.8 parts per thousand and 18.8 parts per thousand. The Local Meteoric Water Line (LMWL) is found to be indistinguishable from the Global Meteoric Water Line (GMWL). Late ISM (September-October) rainfall exhibits lowest delta O-18 and delta D values, with little relationship to the local precipitation amount. There is a trend to lighter isotope values over the course of the ISM, but it does not correlate with the patterns of temperature and rainfall amount delta O-18 and delta D time-series have to be interpreted with caution in terms of the 'amount effect' in this subtropical region. We find that the temporal trend in delta O-18 reflects increasing transport distance during the ISM, isotopic changes in the northern BoB surface waters during late ISM, and vapor re-equilibration with rain droplets. Using an isotope box model for surface ocean waters, we quantify the potential influence of river runoff on the isotopic composition of the seasonal freshwater plume in the northern BoB. Temporal variations in this source can contribute up to 25% of the observed changes in stable isotopes of precipitation in NE India. To delineate other moisture sources, we use backward trajectory computations and find a strong correlation between source region and isotopic composition. Palaeoclimatic stable isotope time-series from northeast Indian speleothems likely reflect changes in moisture source and transport pathway, as well as the isotopic composition of the BoB surface water, all of which in turn reflect ISM strength. Stalagmite records from the region can therefore be interpreted as integrated measures of the ISM strength.
During the last glacial period, climate records from the North Atlantic region exhibit a pronounced spectral component corresponding to a period of about 1470 years, which has attracted much attention. This spectral peak is closely related to the recurrence pattern of Dansgaard-Oeschger (DO) events. In previous studies a red noise random process, more precisely a first-order autoregressive (AR1) process, was used to evaluate the statistical significance of this peak, with a reported significance of more than 99%. Here we use a simple mechanistic two-state model of DO events, which itself was derived from a much more sophisticated ocean-atmosphere model of intermediate complexity, to numerically evaluate the spectral properties of random (i.e., solely noise-driven) events. This way we find that the power spectral density of random DO events differs fundamentally from a simple red noise random process. These results question the applicability of linear spectral analysis for estimating the statistical significance of highly non-linear processes such as DO events. More precisely, to enhance our scientific understanding about the trigger of DO events, we must not consider simple "straw men" as, for example, the AR1 random process, but rather test against realistic alternative descriptions.
To balance the steady decrease of conventional hydrocarbon resources, increased utilization of unconventional and new energy resources, such as shale gas and geothermal energy, is required. Also, the geological sequestration of carbon dioxide is being considered as a technology that may temporarily mitigate the effects of CO2 emission. Sites suitable for shale gas production, geothermal exploration, or CO2 sequestration are commonly characterized by electrical resistivities distinctly different from those of the surrounding rocks. Therefore, electromagnetic methods can be viable tools to help identify target sites suitable for exploration, and to monitor reservoirs during energy production or CO2 injection. Among the wide variety of electromagnetic methods available, controlled-source magnetotelluric (CSMT) may be particularly suitable because of (i) its ability to resolve both electrically resistive and conductive structures, (ii) controlled sources offering noise control and thus facilitating surveys in populated regions, and (iii) the potential of penetration throughout the depth range accessible by drilling. Nevertheless, CSMT has not yet been widely employed because of logistical challenges of field operations and the requirement of complex and highly computer-intensive data processing. With these difficulties gradually being mitigated by recent technological developments, CSMT may now be reconsidered as an exploration tool. Here, we investigate by 1D and 3D numerical simulations the feasibility of detecting gas shales and identifying sites eligible for geothermal exploration or CO2 sequestration from CSMT data. We consider surface-to-surface, borehole-to-surface, and cross-hole configurations of the sources and receivers. Results and conclusions on the detectability of the targets of interest are presented for various exploration and monitoring scenarios, which are roughly representative of the geological setting of the North German Basin.
We establish elements of a new approach to ellipticity and parametrices within operator algebras on manifolds with higher singularities, only based on some general axiomatic requirements on parameter-dependent operators in suitable scales of spaes. The idea is to model an iterative process with new generations of parameter-dependent operator theories, together with new scales of spaces that satisfy analogous requirements as the original ones, now on a corresponding higher level. The "full" calculus involves two separate theories, one near the tip of the corner and another one at the conical exit to infinity. However, concerning the conical exit to infinity, we establish here a new concrete calculus of edge-degenerate operators which can be iterated to higher singularities.
On a method for solution of the ordinary differential equations connected with Huygens' equations

Investigation of transient soil moisture profiles yields valuable information of near- surface processes. A recently developed reconstruction algorithm based on the telegraph equation allows the inverse estimation of soil moisture profiles along coated, three rod TDR probes. Laboratory experiments were carried out to prove the results of the inversion and to understand the influence of probe rod deformation and solid objects close to the probe in heterogonous media. Differences in rod geometry can lead to serious misinterpretations in the soil moisture profile but have small influence on the average soil moisture along the probe. Solids in the integration volume have almost no effect on average soil moisture but result in locally slightly decreased moisture values. Inverted profiles obtained in a loamy soil with a clay content of about 16% were in good agreement with independent measurements.
Shallowly situated evaporites in built-up areas are of relevance for urban and cultural development and hydrological regulation. The hazard of sinkholes, subrosion depressions and gypsum karst is often difficult to evaluate and may quickly change with anthropogenic influence. The geophysical exploration of evaporites in metropolitan areas is often not feasible with active industrial techniques. We collect and combine different passive geophysical data as microgravity, ambient vibrations, deformation and hydrological information to study the roof morphology of shallow evaporites beneath Hamburg, Northern Germany. The application of a novel gravity inversion technique leads to a 3-D depth model of the salt diapir under study. We compare the gravity-based depth model to pseudo-depths from H/V measurements and depth estimates from small-scale seismological array data. While the general range and trend of the diapir roof is consistent, a few anomalous regions are identified where H/V pseudo-depths indicate shallower structures not observed in gravity or array data. These are interpreted by shallow residual caprock floaters and zones of increased porosity. The shallow salt structure clearly correlates with a relative subsidence in the order of 2 mm yr(-1). The combined interpretation of roof morphology, yearly subsidence rates, chemical analyses of groundwater and of hydraulic head in aquifers indicates that the salt diapir beneath Hamburg is subject to significant ongoing dissolution that may possibly affect subrosion depressions, sinkhole distribution and land usage. The combined analysis of passive geophysical data may be exemplary for the study of shallow evaporites beneath other urban areas.
The presented work describes new concepts of fast switching elements based on principles of photonics. The waveguides working in visible and infra-red ranges are put in a basis of these elements. And as materials for manufacturing of waveguides the transparent polymers, dopped by molecules of the dyes possessing second order nonlinear-optical properties are proposed. The work shows how nonlinear-optical processes in such structures can be implemented by electro-optical and opto-optical control circuit signals. In this paper we consider the complete cycle of fabrication of several types of integral photonic elements. The theoretical analysis of high-intensity beam propagation in media with second-order optical nonlinearity is performed. Quantitative estimations of necessary conditions of occurrence of the nonlinear-optical phenomena of the second order taking into account properties of used materials are made. The paper describes the various stages of manufacture of the basic structure of the integrated photonics: a planar waveguide. Using the finite element method the structure of the electromagnetic field inside the waveguide in different modes was analysed. A separate part of the work deals with the creation of composite organic materials with high optical nonlinearity. Using the methods of quantum chemistry, the dependence of nonlinear properties of dye molecules from its structure were investigated in details. In addition, the paper discusses various methods of inducing of an optical nonlinearity in dye-doping of polymer films. In the work, for the first time is proposed the use of spatial modulation of nonlinear properties of waveguide according Fibonacci law. This allows involving several different nonlinear optical processes simultaneously. The final part of the work describes various designs of integrated optical modulators and switches constructed of organic nonlinear optical waveguides. A practical design of the optical modulator based on Mach-Zehnder interferometer made by a photolithography on polymer film is presented.
Das Ziel dieser Arbeit ist die Untersuchung der aktiven Komponenten und ihrer Wechselwirkungen in teilorganischen Hybrid-Solarzellen. Diese bestehen aus einer dünnen Titandioxidschicht, kombiniert mit einer dünnen Polymerschicht. Die Effizienz der Hybrid-Solarzellen wird durch die Lichtabsorption im Polymer, die Dissoziation der gebildeten Exzitonen an der aktiven Grenzfläche zwischen TiO2 und Polymer, sowie durch Generation und Extraktion freier Ladungsträger bestimmt. Zur Optimierung der Solarzellen wurden grundlegende physikalische Wechselwirkungen zwischen den verwendeten Materialen sowie der Einfluss verschiedener Herstellungsparameter untersucht. Unter anderem wurden Fragen zum optimalen Materialeinsatz und Präparationsbedingungen beantwortet sowie grundlegende Einflüsse wie Schichtmorphologie und Polymerinfiltration näher betrachtet. Zunächst wurde aus unterschiedlich hergestelltem Titandioxid (Akzeptor-Schicht) eine Auswahl für den Einsatz in Hybrid-Solarzellen getroffen. Kriterium war hierbei die unterschiedliche Morphologie aufgrund der Oberflächenbeschaffenheit, der Film-Struktur, der Kristallinität und die daraus resultierenden Solarzelleneigenschaften. Für die anschließenden Untersuchungen wurden mesoporöse TiO2–Filme aus einer neuen Nanopartikel-Synthese, welche es erlaubt, kristalline Partikel schon während der Synthese herzustellen, als Elektronenakzeptor und konjugierte Polymere auf Poly(p-Phenylen-Vinylen) (PPV)- bzw. Thiophenbasis als Donatormaterial verwendet. Bei der thermischen Behandlung der TiO2-Schichten erfolgt eine temperaturabhängige Änderung der Morphologie, jedoch nicht der Kristallstruktur. Die Auswirkungen auf die Solarzelleneigenschaften wurden dokumentiert und diskutiert. Um die Vorteile der Nanopartikel-Synthese, die Bildung kristalliner TiO2-Partikel bei tiefen Temperaturen, nutzen zu können, wurden erste Versuche zur UV-Vernetzung durchgeführt. Neben der Beschaffenheit der Oxidschicht wurde auch der Einfluss der Polymermorphologie, bedingt durch Lösungsmittelvariation und Tempertemperatur, untersucht. Hierbei konnte gezeigt werden, dass u.a. die Viskosität der Polymerlösung die Infiltration in die TiO2-Schicht und dadurch die Effizienz der Solarzelle beeinflusst. Ein weiterer Ansatz zur Erhöhung der Effizienz ist die Entwicklung neuer lochleitender Polymere, welche möglichst über einen weiten spektralen Bereich Licht absorbieren und an die Bandlücke des TiO2 angepasst sind. Hierzu wurden einige neuartige Konzepte, z.B. die Kombination von Thiophen- und Phenyl-Einheiten näher untersucht. Auch wurde die Sensibilisierung der Titandioxidschicht in Anlehnung an die höheren Effizienzen der Farbstoffzellen in Betracht gezogen. Zusammenfassend konnten im Rahmen dieser Arbeit wichtige Einflussparameter auf die Funktion hybrider Solarzellen identifiziert und z.T. näher diskutiert werden. Für einige limitierende Faktoren wurden Konzepte zur Verbesserung bzw. Vermeidung vorgestellt.

Tailspike interactions with lipopolysaccharide effect DNA ejection from phage P22 particles in vitro

Initial attachment of bacteriophage P22 to the Salmonella host cell is known to be mediated by interactions between lipopolysaccharide (LPS) and the phage tailspike proteins (TSP), but the events that subsequently lead to DNA injection into the bacterium are unknown. We used the binding of a fluorescent dye and DNA accessibility to DNase and restriction enzymes to analyze DNA ejection from phage particles in vitro. Ejection was specifically triggered by aggregates of purified Salmonella LPS but not by LPS with different O-antigen structure, by lipid A, phospholipids, or soluble O-antigen polysaccharide. This suggests that P22 does not use a secondary receptor at the bacterial outer membrane surface. Using phage particles reconstituted with purified mutant TSP in vitro, we found that the endorhamnosidase activity of TSP degrading the O-antigen polysaccharide was required prior to DNA ejection in vitro and DNA replication in vivo. If, however, LPS was pre-digested with soluble TSP, it was no longer able to trigger DNA ejection, even though it still contained five O-antigen oligosaccharide repeats. Together with known data on the structure of LPS and phage P22, our results suggest a molecular model. In this model, tail-spikes position the phage particles on the outer membrane surface for DNA ejection. They force gp26, the central needle and plug protein of the phage tail machine, through the core oligosaccharide layer and into the hydrophobic portion of the outer membrane, leading to refolding of the gp26 lazo-domain, release of the plug, and ejection of DNA and pilot proteins.
Pektatlyase (Pel-15) aus dem alkalophilen Bodenbakterium Bacillus spec. KSM-P15 ist mit 197 Aminosäuren eines der kleinsten, bekannten β-3-Solenoidproteine. Sie spaltet Polygalakturonsäurederivate in einem Ca2+-abhängigen β-Eliminierungsprozess. Wie bei allen Proteinen dieser Enzymfamilie ist auch die Polypeptidkette von Pel-15 zu einer einsträngigen, rechtsgängigen, parallelen β-Helix aufgewunden. In diesem Strukturmotiv enthält jede Windung drei β-Stränge, die jeweils durch flexible Schleifenbereiche miteinander verbunden sind. Insgesamt acht Windungen stapeln sich in Pel-15 übereinander und bilden entlang der Helixachse flächige, parallele β-Faltblätter aus. Im Bereich dieser β-Faltblätter existiert ein ausgedehntes Netzwerk von Wasserstoffbrückenbindungen, durch das der hydrophobe Kern, der sich im Inneren der β-Helix befindet, vom umgebenden Lösungsmittel abgeschirmt wird. Besondere Abschlussstrukturen an beiden Enden der β-Helix, wie sie typischerweise bei anderen Ver-tretern dieser Strukturklasse ausgeprägt werden, sind in Pel-15 nicht zu beobachten. Stattdessen sind die terminalen Bereiche der β-Helix über Salzbrücken und hydrophobe Seitenkettenkontakte stabilisiert. In der vorliegenden Dissertation wurde die Pektatlyase Pel-15 hinsichtlich ihres Faltungsgleichgewichtes, ihrer enzymatischen Aktivität und der Kinetik ihrer Strukturbildung charakterisiert. In eine evolutionär konservierte Helixwindung wurden destabilisierende Mutationen eingeführt, und deren Auswirkungen mittels spektroskopischer Methoden analysiert. Die Ergebnisse zeigen, dass Pel-15 in Gegenwart des Denaturierungsmittels Guanidiniumhydrochlorid einen hyperfluoreszenten Gleichgewichtsustand (HF) populiert, der nach Messungen von Faltungs- und Entfaltungskinetiken ein konformationelles Ensemble aus den Zuständen HFslow und HFfast darstellt. Diese HF-Zustände sind durch eine hohe Aktivierungsbarriere voneinander getrennt. In Rückfaltungsexperimenten populieren nur etwa 80 % der faltenden Moleküle den Zwischenzustand HFslow, der mit einer Zeitkonstante von ca. 100 s zu HFfast weiterreagiert. Die Denaturierungsmittelabhängigkeit dieser Reaktion ist sehr gering, was eine trans-/cis-Prolylisomerisierung als geschwindigkeitslimitierenden Schritt nahelegt. Die Existenz eines cis-Peptides in der nativen Struktur macht es erforderlich, den denaturierten Zustand als ein Ensemble kinetisch separierter Konformationen, kurz: DSE, zu betrachten, das durch die Spezies Ufast und Uslow populiert wird. Nach dem in dieser Arbeit aufgestellten „Minimalmodell der Pel-15 Faltung“ stehen die HF-Spezies (HFslow, HFfast) mit den Konformationen des DSE in einem thermodynamischen Kreisprozess. Das Modell positioniert HFfast und die native Konformation N auf die „native Seite“ der Aktivierungsbarriere und trägt damit der Tatsache Rechnung, dass die Gleichgewichtseinstellung zwischen diesen Spezies zu schnell ist, um mit manuellen Techniken erfasst zu werden. Die hochaffine Bindung von Ca2+ (Kd = 10 μM) verschiebt sich das Faltungsgleichgewicht bereits in Gegenwart von 1 mM CaCl2 soweit auf die Seite des nativen Zustandes, das HFfast nicht länger nachweisbar ist. Entgegen anfänglicher Vermutungen kommt einer lokalen, evolutionär konservierten Disulfidbrücke im Zentrum der β-Helix eine wichtige Stabilisierungsfunktion zu. Die Disulfidbrücke befindet sich in einem kurzen Schleifenbereich der β-Helix nahe dem aktiven Zentrum. Obwohl ihr Austausch gegen die Reste Val und Ala die freie Stabilisierungsenthalpie des Proteins um ca. 10 kJ/mol reduziert, lässt die Struktur im Bereich der Mutationsstelle keine gravierende Veränderung erkennen. Auch die katalytisch relevante Ca2+-Bindungsaffinität bleibt unbeeinflusst; dennoch zeigen Enzymaktivitätstests für VA-Mutanten eine Reduktion der enzymatischen Aktivität um fast 50 % an. Die evolutionär konservierte Helixwindung im Allgemeinen und die in ihr enthaltene Disulfidbrücke im Besonderen müssen nach den vorliegenden Ergebnissen also eine zentrale Funktion sowohl für die Struktur des katalytischen Zentrums als auch für die Strukturbildung der β-Helix während der Faltungsreaktion besitzen. Die Ergebnisse dieser Arbeit finden in mehreren Punkten Anklang an Faltungseigenschaften, die für andere β -Helixproteine beschrieben wurden. Vor allem aber prädestinieren sie Pel-15 als ein neues, β-helikales Modellprotein. Aufgrund seiner einfachen Topologie, seiner niedrigen Windungszahl und seiner hohen thermodynamischen Stabilität ist Pel-15 sehr gut geeignet, die Determinanten von Stabilität und Strukturbildung des parallelen β-Helix-Motivs in einer Auflösung zu studieren, die aufgrund der Komplexität bestehender β-helikaler Modellsysteme bislang nicht zur Verfügung stand.
The method of current extraction under linear increasing voltages (CELIV) allows for the simultaneous determination of charge mobilities and charge densities directly in thin-film geometries as used in organic photovoltaic (OPV) cells. It has been specifically applied to investigate the interrelation of microstructure and charge-transport properties in such systems. Numerical and analytical calculations presented in this work show that the evaluation of CELIV transients with the commonly used analysis scheme is error prone once charge recombination and, possibly, field- dependent charge mobilities are taken into account. The most important effects are an apparent time dependence of charge mobilities and errors in the determined field dependencies. Our results implicate that reports on time-dependent mobility relaxation in OPV materials obtained by the CELIV technique should be carefully revisited and confirmed by other measurement methods.
The long-term persistence of populations and species depends on the successful recruitment of individuals. The generative recruitment of plants may be limited by a lack of suitable germination and establishment conditions. Establishment limitation may especially be caused by the competitive effect of surrounding dense vegetation, which is believed to restrict the recruitment success of many plant species to small open patches ('safe sites'). We conducted experiments to clarify the roles of germination and seedling establishment as limiting processes in the recruitment of Juncus atratus Krock., a rare and threatened herbaceous perennial river corridor plant in Central Europe. Light intensity had a positive effect on germination. However, some seedlings emerged even in total darkness and the germination rate at 1% light intensity was more than half of that at 60% light intensity. Seedling establishment in the field after 10 weeks was 30% on bare ground, but it was close to zero in grassland. Establishment in the growth chamber after 8 weeks was close to 75% for seedlings that germinated underwater, but only about 35% for seedlings that germinated afloat. Furthermore, establishment decreased with flooding duration on bare ground, but increased with flooding duration in grassland. These data indicate that establishment, rather than germination, is a critical life stage in Central European populations off. atratus. They furthermore indicate that the competition of surrounding vegetation for water limits seedling establishment under field conditions without flooding, largely restricting establishment success to bare ground habitats. In contrast, grassland is more suitable for the recruitment off. atratus than bare ground under prolonged flooding. Grassland may facilitate the establishment off. atratus seedlings during long- lasting floods by supplying oxygen to the soil through aerenchyma. The shift from competition to facilitation in grassland occurred after 30 days of flooding, i.e. within the ontogeny of individual plants. The specific recruitment requirements off. arrows may be a main cause of its rarity in modern Central Europe. In order to prevent regional extinction off. atratus, we suggest maintaining or re-establishing natural hydrodynamics in the species' habitats.
Aims: Factors limiting distributions of species are fundamental to ecology and evolution but have rarely been addressed experimentally for multiple species. The conspicuous linear distribution patterns of plant species confined to river corridors in the Central European lowlands constitute an especially long-standing distribution puzzle. We experimentally tested our novel hypothesis that the tolerance of species to river corridor conditions is independent of the degree of confinement to river corridor habitats, but that species not confined to river corridors are better able to take advantage of the more benign non-river corridor conditions. Methods: We grew 42 herbaceous species differing in their confinement to river corridors in a common garden experiment on loamy soil typical for river corridor areas and sandy soil typical for non-river corridor areas, and with and without a flooding period. For a subset of species, we grew plants of both river corridor and non-river corridor origin to test for adaptation to river corridor conditions. Important findings: Species more confined to river corridor areas benefited less from the more benign non-flooded and non-river corridor soil conditions than species of wider distributional range did. For subsets of 7 and 12 widespread species, the response to flooding and soil origin, respectively, did not differ between plants from river corridor sites and plants from other sites, suggesting that the habitat tolerance of widespread species is clue to phenotypic plasticity rather than to local adaptation. Overall, we found clear support for our novel hypothesis that species not confined to river corridors are more able to take advantage of the more benign non-river corridor conditions. Our study provides a general hypothesis on differences between species confined to stressful habitats and widespread species out for test in further multispecies comparative experiments.

Investigation of transient soil moisture profiles yields valuable information of near- surface processes. A recently developed reconstruction algorithm based on the telegraph equation allows the inverse estimation of soil moisture profiles along coated, three rod TDR probes. Laboratory experiments were carried out to prove the results of the inversion and to understand the influence of probe rod deformation and solid objects close to the probe in heterogeneous media. Differences in rod geometry can lead to serious misinterpretations in the soil moisture profile, but have small influence on the average soil moisture along the probe. Solids in the integration volume have almost no effect on average soil moisture, but result in locally slightly decreased moisture values. Inverted profiles obtained in a loamy soil with a clay content of about 16% were in good agreement with independent measurements.
This study presents an application of an innovative sampling strategy to assess soil moisture dynamics in a headwater of the Weisseritz in the German eastern Ore Mountains. A grassland site and a forested site were instrumented with two Spatial TDR clusters (STDR) that consist of 39 and 32 coated TDR probes of 60 cm length. Distributed time series of vertically averaged soil moisture data from both sites/ensembles were analyzed by statistical and geostatistical methods. Spatial variability and the spatial mean at the forested site were larger than at the grassland site. Furthermore, clustering of TDR probes in combination with long-term monitoring allowed identification of average spatial covariance structures at the small field scale for different wetness states. The correlation length of soil water content as well as the sill to nugget ratio at the grassland site increased with increasing average wetness and but, in contrast, were constant at the forested site. As soil properties at both the forested and grassland sites are extremely variable, this suggests that the correlation structure at the forested site is dominated by the pattern of throughfall and interception. We also found a very strong correlation between antecedent soil moisture at the forested site and runoff coefficients of rainfall-runoff events observed at gauge Rehefeld. Antecedent soil moisture at the forest site explains 92% of the variability in the runoff coefficients. By combining these results with a recession analysis we derived a first conceptual model of the dominant runoff mechanisms operating in this catchment. Finally, we employed a physically based hydrological model to shed light on the controls of soil- and plant morphological parameters on soil average soil moisture at the forested site and the grassland site, respectively. A homogeneous soil setup allowed, after fine tuning of plant morphological parameters, most of the time unbiased predictions of the observed average soil conditions observed at both field sites. We conclude that the proposed sampling strategy of clustering TDR probes is suitable to assess unbiased average soil moisture dynamics in critical functional units, in this case the forested site, which is a much better predictor for event scale runoff formation than pre-event discharge. Long term monitoring of such critical landscape elements could maybe yield valuable information for flood warning in headwaters. We thus think that STDR provides a good intersect of the advantages of permanent sampling and spatially highly resolved soil moisture sampling using mobile rods.
The statistical distribution of energies among particles responsible for long gamma-ray burst (GRB) emission is analyzed in light of recent results of the Fermi Observatory. The all-sky flux, F., recorded by the Gamma-Ray Burst Monitor (GBM) is shown, despite its larger energy range, to be not significantly larger than that reported by the Burst and Transient Explorer, suggesting a relatively small flux in the 3-30MeV energy range. The present-day energy input rate in gamma-rays recorded by the GBM from long GRBs is found, assuming star formation rates in the literature, to be W(0) = 0.5F gamma H/c = 5x10(42) erg Mpc(-3) yr(-1). The Large Area Telescope fluence, when observed, is about 5%-10% per decade of the total, in good agreement with the predictions of saturated, nonlinear shock acceleration. The high- energy component of long GRBs, as measured by Fermi, is found to contain only similar to 10-2.5 of the energy needed to produce ultrahigh-energy cosmic rays (UHECRs) above 4 EeV, assuming the latter to be extragalactic, when various numerical factors are carefully included, if the cosmic-ray source spectrum has a spectral index of -2. The observed. - ray fraction of the required UHECR energy is even smaller if the source spectrum is softer than E-2. The AMANDA II limits rule out such a GRB origin for UHECRs if much more than 10(-2) of the cosmic-ray energy goes into neutrinos that are within, and simultaneous with, the gamma-ray beam. It is suggested that "orphan" neutrinos out of the gamma-ray beam might be identifiable via orphan afterglow or other wide angle signatures of GRBs in lieu of coincidence with prompt gamma-rays, and it is recommended that feasible single neutrino trigger criteria be established to search for such coincidences.
There is growing evidence that aging and muscle fatigue result in impaired postural reflexes in humans. Therefore, the objective of this study was to examine the effects of ankle fatigue on functional reflex activity (ERA) during gait perturbations in young and elderly men. Twenty-eight young (27.0 +/- 3.1 years, n = 14) and old (67.2 +/- 3.7 years, n = 14) healthy active men participated in this study. Fatigue of the plantarflexors and dorsiflexors was induced by isokinetic contractions. Pre and post-fatigue, subjects were tested for their ability to compensate for decelerating gait perturbations while walking on a treadmill. Latency, ERA of lower extremity muscles and angular velocity of the ankle joint complex were analysed by means of surface electromyography and goniometry. After the fatigue protocol, no significant main and interaction effects were detected for the parameter latency in m. tibialis anterior (TA). For both groups, a significant pre to post-test decrease in ERA in TA (P<.001) was observed coming along with increases in antagonist coactivity (P=.013) and maximal angular velocity of the ankle joint (p=.007). However, no significant group x test interactions were found for the three parameters. Ankle fatigue has an impact on the ability to compensate for gait perturbations in young and elderly adults. However, no significant differences in all analysed parameters were detected between young and elderly subjects. These results may imply that age-related deteriorations in the postural control system do not specifically affect the ability to compensate for gait perturbations under fatigued condition.
Dass Universitäten über Jahrhunderte hinweg Gegenstand von Literatur waren, verwundert kaum. Gehen doch Lesen und Bildung Hand in Hand. Und doch verzeichnen Literaturwissenschaftler für die Jahre zwischen 1690 und 1720 ungewöhnlich viele Romane, die sich der Beschreibung des zeitgenössischen Studentenlebens oder der Gelehrtensatire widmen. In ihrer Antrittsvorlesung am 23. Juni geht Prof. Dr. Stefanie Stockhorst der Frage nach, warum die Universität für den Roman um 1700 zu einem so wichtigen Thema wurde. Dabei entwickelt sie die These, dass sich in den viel gelesen Texten der Zeit wie dem "Academischen Roman" (1690) von Werner Happel bereits jener Bildungsoptimismus Bahn bricht, der, an der Schwelle zur Aufklärung stehend, mehr und mehr an Bedeutung gewinnt.
Bulk electron transport and charge injection in a high mobility n-type semiconducting polymer

Bulk electron transport in a high mobility n-type polymer is studied by time-of-flight photocurrent measurements and electron-only devices. Bulk electron mobilities of similar to 5 x 10(-3) cm(2)/Vs are obtained. The analysis of the electron currents suggests the presence of an injection barrier for all conventionally used low workfunction cathodes.
In dieser Arbeit wurden die Möglichkeiten und Grenzen für Zirkulardichroismus-Messungen mit Synchrotronstrahlung untersucht. Dazu wurde ein Messaufbau für Zirkulardichroismus-Messungen an zwei Strahlrohren am Berliner Elektronenspeicherring für Synchrotronstrahlung eingesetzt, die für Messungen im Bereich des ultravioletten Lichts geeignet sind. Eigenschaften der Strahlrohre und des Messaufbau wurden in einigen wichtigen Punkten mit kommerziellen Zirkulardichroismus-Spektrometern verglichen. Der Schwerpunkt lag auf der Ausdehnung des zugänglichen Wellenlängenbereichs unterhalb von 180 nm zur Untersuchung des Zirkulardichroismus von Proteinen in diesem Bereich. In diesem Bereich ist es nicht nur die Lichtquelle sondern vor allem die Absorption des Lichts durch Wasser, die den Messbereich bei der Messung biologischer Proben in wässriger Lösung einschränkt. Es wurden Bedingungen gefunden, unter denen der Messbereich auf etwa 160 nm, in einigen Fällen bis auf 130 nm ausgedehnt werden konnte. Dazu musste die Pfadlänge deutlich reduziert werden und verschieden Probenküvetten wurden getestet. Der Einfluss der dabei auftretenden Spannungsdoppelbrechung in den Probenküvetten auf das Messsignal konnte mit einem alternativen Messaufbau deutlich reduziert werden. Systematische Fehler im Messsignal und auftretende Strahlenschäden begrenzen jedoch die Zuverlässigkeit der gemessenen Spektren. Bei Proteinfilmen schränkt die Absorption von Wasser den Messbereich kaum ein. Es wurden jedoch meist deutliche Unterschiede zwischen den Spektren von Proteinfilmen und den Spektren von Proteinen in wässriger Lösung festgestellt. Solange diese Unterschiede nicht minimiert werden können, stellen Proteinfilme keine praktikable Alternative zu Messungen in wässriger Lösung dar.
COR15A and COR15B form a tandem repeat of highly homologous genes in Arabidopsis thaliana. Both genes are highly cold induced and the encoded proteins belong to the Pfam LEA_4 group (group 3) of the late embryogenesis abundant (LEA) proteins. Both proteins were predicted to be intrinsically disordered in solution. Only COR15A has previously been characterized and it was shown to be localized in the soluble stroma fraction of chloroplasts. Ectopic expression of COR15A in Arabidopsis resulted in increased freezing tolerance of both chloroplasts after freezing and thawing of intact leaves and of isolated protoplasts frozen and thawed in vitro. In the present study we have generated recombinant mature COR15A and COR15B for a comparative study of their structure and possible function as membrane protectants. CD spectroscopy showed that both proteins are predominantly unstructured in solution and mainly a-helical after drying. Both proteins showed similar effects on the thermotropic phase behavior of dry liposomes. A decrease in the gel to liquid-crystalline phase transition temperature depended on both the unsaturation of the fatty acyl chains and lipid headgroup structure. FTIR spectroscopy indicated no strong interactions between the proteins and the lipid phosphate and carbonyl groups, but significant interactions with the galactose headgroup of the chloroplast lipid monogalactosyldiacylglycerol. These findings were rationalized by modeling the secondary structure of COR15A and COR15B. Helical wheel projection indicated the presence of amphipathic a-helices in both proteins. The helices lacked a clear separation of positive and negative charges on the hydrophilic face, but contained several hydroxylated amino acids.
TSPs (tailspike proteins) are essential infection organelles of bacteriophage P22. Upon infection, P22TSP binds to and cleaves the O-antigen moiety of the LPS (lipopolysaccharide) of its Salmonella host To elucidate the role of TSP during infection, we have studied binding to oligosaccharides and polysaccharides of Salmonella enteric Typhimurium and Enteritidis in vitro. P22TSP is a trimeric beta-helical protein with a carbohydrate-binding site on each subunit. Octasaccharide O-antigen fragments bind to P22TSP with micromolar dissociation constants. Moreover, P22TSP is an endorhamnosidase and cleaves the host O-antigen. Catalytic residues lie at the periphery of the high-affinity binding site, which enables unproductive binding modes, resulting in slow hydrolysis. However, the role of this hydrolysis function during infection remains unclear. Binding of polysaccharide to P22TSP is of high avidity with slow dissociation rates when compared with oligosaccharides. In vivo, the infection of Salmonella with phage P22 can be completely inhibited by the addition of LPS, indicating that binding of phage to its host via TSP is an essential step for infection.
This paper aims to highlight the potential of using elemental and stable isotope analyses of aquatic macrophytes in palaeolimnological studies. Potamogeton pectinatus material was collected from modem plants (n=68) and from late glacial and Holocene-aged sediments from Koucha Lake (northeastern Tibetan Plateau; 34.0 degrees N; 97.2 degrees E; 4540 m a.s.l.). It was analyzed for delta C-13(Potamogeton) (modern: -23 to 0 parts per thousand, fossil: -19 to -4 parts per thousand) and delta N-15(Potamogeton) (modern: -11.0 to +13.8 parts per thousand, fossil: -9.5 to +6.7 parts per thousand) in addition to elemental carbon and nitrogen (modem C/N-Potamogeton: 7 to 29; fossil: 13 to 68) and sulfur (fossil: 188-899 mu mol/g dry weight). Fossil data were interpreted in terms of palaeo-nutrient availability and palaeo-productivity based on the modem relationships between various proxies and certain environmental data. Productivity of Potamogeton pectinatus mats at Koucha Lake as indicated by palaeo-epsilon(Potamogeton-TIC) (i.e. the enrichment of delta C-13(Potamogeton) relative to the delta(CTIC)-C-13) was reduced during periods of high conductivity, especially between 10.3 and 7.4 cal kyr BP. Potamogeton pectinatus material from these periods was also characterized by high S-Potamogeton indicating high sulfide concentrations and anoxic conditions within the sediments. However, C/N- Potamogeton ratios and delta N-15(Potamogeton) from the lower core section were found to have been altered by decompositional processes. A pronounced shift in the aquatic productivity of Lake Koucha occurred at similar to 7.4 cal kyr BP when the hydrological conditions shifted towards an open lake system and water depth increased. At this time a strong increase in productivity led to a strong decrease in the water HCO3- concentration as inferred from the application of a epsilon-(Potamogeton-TIC)-InHCO3- transfer function. A comparison of reconstructed productivity changes from Koucha Lake with further environmental proxies suggests that primary productivity changes are probably a function of internal lake dynamics and were only indirectly triggered by climate change.
Aim: Fossil pollen spectra from lake sediments on the Tibetan Plateau have been used for qualitative climate reconstruction, but no modern pollen-climate calibration set based on lake sediments is available to infer past climate quantitatively. This study aims to develop such a dataset and apply it to fossil data. Location: The Tibetan Plateau, between 30 and 40 degrees N and 87 and 103 degrees E. Methods: We collected surface sediments from 112 lakes and analysed them palynologically. The lakes span a wide range of mean annual precipitation (P-ann; 31-1022 mm), mean annual temperature (T-ann; -6.5 to 1 degrees C), and mean July temperature (T-July; 2.6-19.7 degrees C). Redundancy analysis showed that the modern pollen spectra are characteristic of their respective vegetation types and local climate. Transfer functions for P-ann, T-ann and T-July were developed with weighted averaging partial least squares. Model performance was assessed by leave-one-out cross-validation. Results: The root mean square errors of prediction (RMSEP) were 104 mm (P-ann), 1.18 degrees C (T-ann) and 1.17 degrees C (T-July). The RMSEPs, when expressed as percentages of the gradient sampled, were 10.6% (P-ann), 15.7% (T-ann) and 11.9% (T-July). These low values indicate the good performance of our models. An application of the models to fossil pollen spectra covering the last c. 50 kyr yielded realistic results for Luanhaizi Lake in the Qilian Mountains on the north-eastern Tibetan Plateau (modern P-ann 480 mm; T-ann-1 degrees C). T-ann and P-ann values similar to present ones were reconstructed for late Marine Isotope Stage 3, with minimum values for the Last Glacial Maximum (c. 300 mm and 2 degrees C below present), and maximum values for the early Holocene (c. 70 mm and 0.5 degrees C greater than present). Main conclusions: The modern pollen-climate calibration set will potentially be useful for quantitative climate reconstructions from lake-sediment pollen spectra from the Tibetan Plateau, an area of considerable climatic and biogeographical importance.
The apparent isotope enrichment factor epsilon(macrophyte) of submerged plants (epsilon(macrophyte-DIC) = delta C-13(macrophyte) - delta C-13(DIC)) is indicative of dissolved inorganic carbon (DIC) supply in neutral to alkaline waters and is related to variations in aquatic productivity (Papadimitriou et al. in Limnol Oceanogr 50:1084-1095, 2005). This paper aims to evaluate the usage of epsilon(macrophyte) inferred from isotopic analyses of submerged plant fossils in addition to analyses of lake carbonate as a palaeolimnological proxy for former HCO3 (-) concentrations. Stable carbon isotopic analysis of modern Potamogeton pectinatus leaves and its host water DIC from the Tibetan Plateau and Central Yakutia (Russia) yielded values between -23.3 and +0.4aEuro degrees and between +14.0 and +6.5aEuro degrees, respectively. Values of epsilon (Potamogeton-DIC) (range -15.4 to +1.1aEuro degrees) from these lakes are significantly correlated with host water HCO3 (-) concentration (range 78-2,200 mg/l) (r = -0.86; P < 0.001), thus allowing for the development of a transfer function. Palaeo-epsilon (Potamogeton-ostracods) values from Luanhaizi Lake on the NE Tibetan Plateau, as inferred from the stable carbon isotope measurement of fossil Potamogeton pectinatus seeds (range -24 to +2.8aEuro degrees) and ostracods (range -7.8 to +7.5%) range between -14.8 and 1.6aEuro degrees. Phases of assumed disequilibrium between delta C-13(DIC) and delta C-13(ostracods) known to occur in charophyte swards (as indicated by the deposition of charophyte fossils) were excluded from the analysis of palaeo-epsilon. The application of the epsilon (Potamogeton-DIC)-HCO3 (-) transfer function yielded a median palaeo-HCO3 (-) -concentration of 290 mg/l. Variations in the dissolved organic carbon supply compare well with aquatic plant productivity changes and lake level variability as inferred from a multiproxy study of the same record including analyses of plant macrofossils, ostracods, carbonate and organic content.
Aim: Atmospheric CO2 concentrations depend, in part, on the amount of biomass locked up in terrestrial vegetation. Information on the causes of a broad-scale vegetation transition and associated loss of biomass is thus of critical interest for understanding global palaeoclimatic changes. Pollen records from the north-eastern Tibet-Qinghai Plateau reveal a dramatic and extensive forest decline beginning c. 6000 cal. yr bp. The aim of this study is to elucidate the causes of this regional-scale change from high-biomass forest to low-biomass steppe on the Tibet-Qinghai Plateau during the second half of the Holocene. Location: Our study focuses on the north-eastern Tibet-Qinghai Plateau. Stratigraphical data used are from Qinghai Lake (3200 m a.s.l., 36 degrees 32'-37 degrees 15' N, 99 degrees 36'-100 degrees 47' E). Methods: We apply a modern pollen-precipitation transfer function from the eastern and north-eastern Tibet-Qinghai Plateau to fossil pollen spectra from Qinghai Lake to reconstruct annual precipitation changes during the Holocene. The reconstructions are compared to a stable oxygen-isotope record from the same sediment core and to results from two transient climate model simulations. Results: The pollen-based precipitation reconstruction covering the Holocene parallels moisture changes inferred from the stable oxygen-isotope record. Furthermore, these results are in close agreement with simulated model-based past annual precipitation changes. Main conclusions: In the light of these data and the model results, we conclude that it is not necessary to attribute the broad-scale forest decline to human activity. Climate change as a result of changes in the intensity of the East Asian Summer Monsoon in the mid-Holocene is the most parsimonious explanation for the widespread forest decline on the Tibet-Qinghai Plateau. Moreover, climate feedback from a reduced forest cover accentuates increasingly drier conditions in the area, indicating complex vegetation-climate interactions during this major ecological change.
Pollen taxa of known indicator value are of great potential in the qualitative interpretation of pollen diagrams. Here we apply several numerical approaches to a lake-sediment based pollen data-set from the eastern and central Tibetan Plateau (112 samples) to assess the indicator value of Tibetan pollen taxa for modem vegetation types and for modern climate. Results from Multi-Response Permutation Procedures indicate that the differences between groups of pollen spectra originating from the same vegetation type (temperate desert, temperate steppe, alpine desert, alpine steppe, high-alpine meadow, subalpine shrub, and patchy forest) are statistically significant. Indicator Species Analyses identify several indicator taxa for most vegetation types. Multivariate regression tree analysis indicates that about 390 mm of annual precipitation is the most critical threshold for the modern pollen spectra. This roughly separates desert and steppe vegetation from high-alpine meadow, subalpine shrub, and patchy forest vegetation. A strong pollen-climate relationship on the Tibetan Plateau is confirmed by the large number of statistically significant pollen taxa-climate (annual precipitation or/and annual temperature) relationships as evaluated by statistical response- modelling, involving generalised linear models.
