Mechanistische und kinetische Untersuchungen von komplexen zellulären Prozessen in situ sind in den vergangenen Jahren durch den Einsatz photoaktivierbarer Biomoleküle, sogenannter caged Verbindungen, möglich geworden. Bei den caged Verbindungen handelt es sich um photolabile inaktive Derivate von biologisch aktiven Molekülen, aus denen durch ultraviolettes Licht mit Hilfe einer photochemischen Reaktion die natürliche, biologisch aktive Substanz schnell freigesetzt werden kann. Im Rahmen der vorliegenden Arbeit wurden caged Verbindungen von den Neurotransmittern Octopamin und Dopamin, dem Octopamin-Antagonist Epinastin, den Proteinsyntheseinhibitoren Emetin und Anisomycin, dem Protonophor CCCP und dem Riechstoff Bourgeonal hergestellt. Zur Synthese dieser caged Verbindungen wurden sowohl bekannte als auch verschiedene im Rahmen dieser Arbeit neu entwickelte photolabile Schutzgruppen mit einem (Cumarin-4-yl)methyl- bzw. einem 2-Nitrobenzyl-Gerüst eingesetzt. Entsprechende Syntheseverfahren wurden erarbeitet. Anschließend erfolgte eine umfassende physikalisch-chemische sowie photochemische Charakterisierung der erhaltenen caged Verbindungen. Dabei wurde besonders auf gute Löslichkeit in Wasser bei physiologischer Ionenstärke, schnelle und effiziente Photoreaktivität, hohe Extinktion bei Wellenlängen von 350-430 nm und gute solvolytische Stabilität bei geringer Toxizität der freigesetzten Schutzgruppe geachtet. Ein Schwerpunkt bei der photochemischen Charakterisierung bildeten die Untersuchungen zur Quantifizierung der 2-Photonen-Anregung, uncaging action cross-sections, der Cumarinylmethyl-caged Verbindungen, aufgrund ihrer Bedeutung für die Photofreisetzung von Biomolekülen, da die gleichzeitige Absorption von 2 IR-Photonen eine höhere dreidimensionale Auflösung und eine wesentlich tiefere Gewebepenetration erlaubt. Mit Hilfe von Kooperationspartnern wurden zeitaufgelösten Fluoreszenz- und IR-Messungen an verschiedenen (Cumarin-4-yl)methoxycarbonyl-caged Modellverbindungen durchgeführt, mit denen die Geschwindigkeitskonstanten k1 und kdecarb des Photolysemechanismus ermittelt wurde. Am Ende folgten die Anwendungserprobungen ausgewählter caged Verbindungen in einem Translationsassay bzw. in Zelluntersuchungen.
Z,E-Diene sind ein häufig auftretendes Strukturmerkmal in Naturstoffen. Aus diesem Grund ist die einfache Darstellung dieser Struktureinheit von großen Interesse in der organischen Chemie.
Das erste Ziel der vorliegenden Arbeit war daher die Weiterentwicklung der Ringschlussmetathese-/ baseninduzierten Ringöffnungs-/ Veresterungssequenz (RBRV-Sequenz) zur Synthese von (2Z,4E)-Diencarbonsäureethylestern ausgehend von Butenoaten. Dazu wurde zunächst die RBRV-Sequenz optimiert. Diese aus drei Schritten bestehende Sequenz konnte in einem Eintopf-Verfahren angewendet werden. Die Ringschlussmetathese gelang mit einer Katalysatorbeladung von 1 mol% des GRUBBS-Katalysators der zweiten Generation in Dichlormethan. Für die baseninduzierte Ringöffnung des β,γ-ungesättigten δ Valerolactons wurde NaHMDS verwendet. Die Alkylierung der Carboxylatspezies gelang mit dem MEERWEIN-Reagenz. Die Anwendbarkeit der Sequenz wurde für verschiedene Substrate demonstriert.
Die Erweiterung der Methode auf α-substituierte Butenoate unterlag starken Einschränkungen. So konnte der Zugang für α Hydroxyderivate realisiert werden. Bei der Anwendung der RBRV-Sequenz auf die α-substituierten Butenoate wurde festgestellt, dass diese sich nur in moderaten Ausbeuten umsetzen ließen und zudem nicht selektiv zu den (2E,4E)-konfigurierten α-substituierten-Dienestern reagierten.
Der Einsatz von Eninen unter den Standardbedingungen der RBRV-Sequenz gelang nicht. Erst nach Modifizierung der Sequenz (höhere Katalysatorbeladung, Wechsel des Lösungsmittels) konnten die [3]Dendralen-Produkte in geringen Ausbeuten erhalten werden.
Im zweiten Teil der Arbeit wurde der Einsatz von (2Z,4E)-Diencarbonsäureethylestern in der Totalsynthese von Naturstoffen untersucht. Dazu wurden zunächst die Transformationsmöglichkeiten der Ester geprüft. Es konnte gezeigt werden, dass sich (2Z,4E)-Diencarbonsäureethylester insbesondere zur Synthese von (2Z,4E)-Aldehyden sowie zum Aufbau der (3Z,5E)-Dien-1-in-Struktur eignen.
Anhand dieser Ergebnisse wurde im Anschluss die RBRV-Sequenz in der Totalsynthese eingesetzt. Dazu wurde zunächst der (2Z,4E)-Dienester Microsphaerodiolin in seiner ersten Totalsynthese auf drei verschiedene Routen hergestellt. Im Anschluss wurden sechs verschiedene Polyacetylene mit einer (3Z,5E)-Dien-1-in-Einheit hergestellt. Schlüsselschritte in ihrer Synthese waren immer die RBRV-Sequenz zum Aufbau der Z,E-Dien-Einheit, die Transformation des Esters in ein terminales Alkin sowie die CADIOT-CHODKIEWICZ-Kupplung zum Aufbau unsymmetrischer Polyine. Alle sechs Polyacetylene wurden zum ersten Mal in einer Totalsynthese synthetisiert. Drei Polyacetylene wurden ausgehend von (S)-Butantriol enantiomerenrein dargestellt. Anhand ihrer Drehwerte konnte eine Revision der von YAO und Mitarbeitern vorgenommen Zuordnung der Absolutkonfiguration der Naturstoffe vorgenommen werden.

The students first used both scaffolds (stepped supporting tools and a task navigator) on two tasks and could then choose for one other task the scaffold that suited them more. The scaffolds were evaluated in a mixedmethods study by the use of questionnaires and the conducting of a focus group interview. Both scaffolds were assessed as being helpful. However, students who thought they didn’t need different sorts of tips, as provided by the task navigator, chose the stepped supporting tools. All students reflected on their use of the scaffolds; their choices for one of both are therefore well-founded. As the reasons for choosing the scaffold are very individual, in future seminars both types of scaffolds will be provided.

Abstract. Catalysis is one of the most effective tools for the highly efficient assembly of complex molecular structures. Nevertheless, it is mainly represented by transition metal-based catalysts and typically is an energy consuming process. Therefore, photocatalysis utilizing solar energy is one of the appealing approaches to overcome these problems. A great alternative to classic transition metal-based photocatalysts, carbon nitrides, a group of organic polymeric semiconductors, have already shown their efficiency in water splitting, CO2 reduction, and organic pollutants degradation. However, these materials have also a great potential for the use in functionalization of complex organic molecules for synthetic needs as it was shown in recent years.
This work addresses the challenge to develop efficient system for heterogeneous organic photocatalysis, employing cheap and environmentally benign photocatalysts – carbon nitrides. Herein, fundamental properties of semiconductors are studied from the organic chemistry standpoint; the inherent properties of carbon nitrides, such as ability to accumulate electrons, are deeply investigated and their effect on the reaction outcome is established. Thus, understanding of the electron charging processes allowed for the synthesis of otherwise hardly-achieved diazetidines-1,3 by tetramerization of benzylamines. Furthermore, the high electron capacity of Potassium Poly(heptazine imide)s (K-PHI) made possible a multi-electron reduction of aromatic nitro compounds to bare or formylated anilines. Additionally, two deep eutectic solvents (DES) were designed as a sustainable reaction media and reducing reagent for this reaction. Eventually, the high oxidation ability of carbon nitride K-PHI is employed in a challenging reaction of halide anion oxidation (Cl―, Br―) to accomplish electrophilic substitution in aromatic ring. The possibility to utilize NaCl solution (seawater mimetic) for the chlorination of electron rich arenes was shown. Eventually, light itself is used as a tool in a chromoselective photocatalytic oxidation of aromatic thiols and thioacetatas to three different compounds, using UV, blue, and red LEDs.
All in all, the work enhances understanding the mechanism of heterogeneous photocatalysis in synthetic organic reactions and therefore, is a step forward to the sustainable methods of synthesis in organic chemistry.
Zusammenfassung zur Dissertation „Neuartige DBD-Fluoreszenzfarbstoffe: Synthese, Untersuchungen und Anwendungen“ von Leonard John
In dieser Arbeit konnten auf Basis der etablierten [1,3]-Dioxolo[4,5-f][1,3]benzodioxol (DBD) Fluoreszenzfarbstoffe zwei neue Konzepte zur Darstellung unsymmetrisch funktionalisierter DBD-Fluorophore entwickelt werden. Die Variation der elektronenziehenden Reste führte zu einer Erweiterung des Farbspektrums an DBD-Fluorophoren, wobei alle weiteren spektroskopischen Parameter (Fluoreszenzlebenszeit, -quantenausbeute und STOKES-Verschiebung) unverändert hohe Werte aufweisen. Neben der Variation der elektronenziehenden Reste wurde das "pi"-System des DBD-Farbstoffs mit der Einführung von Stilben-, und Tolan-Derivaten vergrößert. Stilben-Derivate zeigten ähnlich gute spektroskopische Eigenschaften wie die bereits etablierten DBD-Farbstoffe.
Fluorophore mit langwelliger Emission sind auf Grund der großen Gewebe-Eindringtiefe besonders interessant für biologische Anwendungen. Da der langwelligste Vertreter der O4-DBD-Farbstoffe in polaren Medien nur schwer löslich ist, wurde ein Weg zur Einführung löslichkeitsvermittelnder Gruppen gesucht. Hierbei fiel die Wahl auf eine Carbonsäure-Gruppe zur Steigerung der Hydrophilie. Eine von vier untersuchten Methoden erwies sich als zielführend, sodass das gewünschte Molekül isoliert werden konnte. Eine erhöhte Wasserlöslichkeit wurde allerdings nicht beobachtet.
Zur Erforschung von Fettstoffwechselkrankheiten wie der ALZHEIMER-Krankheit werden fluoreszenzmarkierte Lipide benötigt. Um unterschiedliche Bereiche einer Membran zu untersuchen, war das Ziel, den Fluorophor an unterschiedlichen Stellen innerhalb der Fettsäure zu lokalisieren. Hierbei sollte die Gesamtkettenlänge des DBD-Lipids einer C18-Kette, analog der Stearinsäure, entsprechen. Durch die stufenweise Einführung der Reste gelang es, drei DBD-Lipide herzustellen, wobei sich der Fluorophor an unterschiedlichen Positionen innerhalb der Kette befindet. Die photophysikalischen Eigenschaften der Lipide weichen nur marginal von denen der reinen Fluorophore ab. Eine Einlagerung in giant unilamellar vesicles (GUVs) konnte für zwei Derivate beobachtet werden, wobei keine domänenspezifisch war.
Ein weiteres Ziel dieser Arbeit war es, die vier Sauerstoffatome im DBD-Grundkörper stufenweise durch Schwefelatome zu ersetzen und die Ringgrößen des DBD-Fluorophors zu variieren. Für die Ringgröße zeigte der 1,2-S2-DBD mit jeweils zwei Fünfringen die besten spektroskopischen Eigenschaften. Durch die Synthese von zwei weiteren schwefelhaltigen DBD-Grundkörpern (S1- und 1,4-S2-DBD) konnten insgesamt drei neue Farbstoffklassen zugänglich gemacht werden. Für alle neuen Chromophore wurden elektronenziehende Reste (Aldehyd, Acyl, Ester, Carboxy) eingeführt und die jeweiligen Derivate spektroskopisch untersucht. Mit steigender Anzahl an Schwefel-Atomen im Grundkörper zeigt sich eine bathochrome Verschiebung der Emission,
wobei die Werte für die Fluoreszenzlebenszeit- und -quantenausbeute abnehmen. Die optimalen spektroskopischen Eigenschaften aus langwelliger Emission, hoher Fluoreszenzlebenszeit und -quantenausbeute zeigt das 1,4-S2-Dialdehyd-Derivat. Für die S1- und 1,2-S2-Dialdehyd-
Derivate wurden Konzepte entwickelt, um bioreaktive Reste (Alkin, HOSu, Maleimid) einzuführen und die Fluorophore in biologischen Systemen anwenden zu können.
Natural products have proved to be a major resource in the discovery and development of many pharmaceuticals that are in use today. There is a wide variety of biologically active natural products that contain conjugated polyenes or benzofuran structures. Therefore, new synthetic methods for the construction of such building blocks are of great interest to synthetic chemists. The recently developed one-pot tethered ring-closing metathesis approach allows for the formation of Z,E-dienoates in high stereoselectivity. The extension of this method with a Julia-Kocienski olefination protocol would allow for the formation of conjugated trienes in a stereoselective manner. This strategy was applied in the total synthesis of conjugated triene containing (+)-bretonin B. Additionally, investigations of cross metathesis using methyl substituted olefins were pursued. This methodology was applied, as a one-pot cross metathesis/ring-closing metathesis sequence, in the total synthesis of benzofuran containing 7-methoxywutaifuranal. Finally, the design and synthesis of a catalyst for stereoretentive metathesis in aqueous media was investigated.
Im Rahmen dieser Dissertation wurde der Sauerstoff im Grundgerüst der [1,3]-Dioxolo[4.5-f]benzodioxol-Fluoreszenzfarbstoffe (DBD-Fluoreszenzfarbstoffe) vollständig mit Schwefel ausgetauscht und daraus eine neue Klasse von Fluoreszenzfarbstoffen entwickelt, die Benzo[1,2-d:4,5-d']bis([1,3]dithiol)-Fluorophore (S4-DBD-Fluorophore). Insgesamt neun der besonders interessanten, difunktionalisierten Vertreter konnten synthetisiert werden, die sich in ihren elektronenziehenden Gruppen und in ihrer Anordnung unterschieden.
Durch den Austausch von Sauerstoff mit Schwefel kam es zu teilweise auffälligen Veränderungen in den Fluoreszenzparametern, wie eine Abnahme der Fluoreszenzquantenausbeuten und -lebenszeiten aber auch eine deutliche Rotverschiebung in den Absorptions- und Emissionswellenlängen mit großen STOKES-Verschiebungen. Damit sind die S4-DBD-Fluorophore eine wertvolle Ergänzung für die DBD-Farbstoffe.
Die Ursachen für die Abnahme der Lebenszeiten und Quantenausbeuten konnte auf eine hohe Besetzung des Triplett-Zustandes zurückgeführt werden, welcher durch die verstärkten Spin-Bahn-Kopplungen des Schwefels hervorgerufen wird. Zusammen mit dem Arbeitskreis physikalische Chemie der Universität Potsdam konnten auch die photophysikalischen Prozesse über die Transienten-Absorptionsspektroskopie (TAS) aufgeklärt werden.
Eine Strategie zur Funktionalisierung der S4-DBD-Farbstoffe am Thioacetalgerüst konnte entwickelt werden. So gelang es Alkohol-, Propargyl-, Azid-, NHS-Ester-, Carbonsäure-, Maleimid- und Tosyl-Gruppen an S4-DBD-Dialdehyden anzubringen.
Erweiternd wurden molekulare Stäbe auf Basis von Schwefel-Oligo-Spiro-Ketalen (SOSKs) untersucht, bei denen Sauerstoff durch Schwefel ersetzt wurde. Hier konnten die Synthesen der löslichkeitsvermittelnden TER-Muffe und auch des Tetrathiapentaerythritols als Grundbaustein deutlich verbessert werden. Aus diesen konnte ein einfaches SOSK-Polymer hergestellt werden. Weitere Versuche zum Aufbau eines Stabes müssen aber noch untersucht werden. Um einen S-OSK-Stab aufzubauen hat sich dabei die Dithiocarbonat-Gruppe in ersten Versuchen als potenzielle geeignete Schutzgruppe für das Tetrathiapentaerythritol herausgestellt.
Mechanism comics as a task in a written exam in organic chemistry for pre-service chemistry teachers

In this paper, we describe and evaluate a study on the use of mechanism comics for writing solutions to a task in a written exam for the course "Organic Chemistry I for Pre-Service Chemistry Teachers."
The students had to design a reaction mechanism for a reaction that was unknown to them and write captions explaining every step of their reaction mechanism.
The students' work was evaluated using the method of qualitative content analysis in four rounds by both authors. The majority of the captions were coded as "descriptive" and only a minority as "causal."
This means that the students mostly described "what" happened, but seldom "why" this happened. Implicit electron movement was also described more often than explicit electron movement. The majority of the captions were technically correct. In summary, the students were capable of designing and describing a reaction mechanism for a previously unknown reaction.
The quality of their reasoning could be improved, however. In the new course, the quality of students' mechanistic reasoning and then especially their explanations of "why" mechanistic steps occur will be given much clearer emphasis.
Das Konstrukt des erweiterten Fachwissens für den schulischen Kontext, welches in der ersten Förderphase von PSI-Potsdam entwickelt wurde, diente als Grundlage für die Entwicklung von Aufgaben, Lehrformate sowie einem Laborpraktikum im Bereich organischer Chemie. Eine Delphi-Studie wurde zur Validierung der Anwendung des Konstruktes auf die organische Chemie durchgeführt. Alle Aufgaben und Lehrformate wurden in Mixed-Methods-Studien evaluiert. Es zeigte sich, dass die Studierenden sowohl die Aufgaben als auch die Lehrformate und das Praktikum als relevant für ihren späteren Beruf als Chemielehrkräfte bewerteten.
