This chapter investigates the trajectory of establishing the Forest Stewardship Council (FSC) in the early 1990s as the first private transnational certification organization with an antagonistic stakeholder body. Its main contribution is a micro-analysis of the founding assembly in 1993. By investigating the role of brokers within the negotiation as one institutional scope condition for ‘arguing’ having occurred, the chapter adopts a dramaturgical approach. It contends that the authority of brokers is not necessarily institutionally given, but needs to be gained: brokers have to prove situationally that their knowledge is relevant and that they are speaking impartially in the interest of progress rather than their own. The chapter stresses the importance of procedural knowledge which brokers provide in contrast to policy knowledge.
"Der unterirdische Kampf"

Die militärische Rüstungsbürokratie zwischen Niederlage des Deutschen Kaiserreiches und ›Totalem Krieg‹ der NS-Diktatur – in dieses Spannungsverhältnis begibt sich der Autor. Er untersucht, wie die Fachleute des Wehrwirtschafts- und Rüstungsamtes die Mobilisierung planten – und dabei scheiterten.
Bereits kurze Zeit nach der deutschen Niederlage von 1918 wurden die wirtschaftlichen Aspekte der Kriegführung wieder von einem kleinen, hochprofessionellen Offizierskreis diskutiert. Sie nahmen Gedanken aus der Zeit des Ersten Weltkrieges auf und forderten in der Reichswehr und noch stärker in der Wehrmacht die Steuerungshoheit des Militärs über die kriegsrelevante Wirtschaft. Ihre Planungen und Maßnahmen waren nicht nur im Offizierkorps selbst umstritten, sondern unterlagen letztlich auch den Widerständen der Privatwirtschaft und den Interessen von NS-Parteigrößen. Entlang dieser Fronten beschreibt der Autor eine militärische Organisation von ihrem Aufbau 1924 bis zu ihrer ›Zerschlagung‹ durch Albert Speer 1942. Dabei nimmt er auch das alles andere als eindeutige Verhalten des Führungspersonals im Amt in den Blick.
The aim of this study was to develop a one-step synthesis of gold nanotriangles (NTs) in the presence of mixed phospholipid vesicles followed by a separation process to isolate purified NTs. Negatively charged vesicles containing AOT and phospholipids, in the absence and presence of additional reducing agents (polyampholytes, polyanions or low molecular weight compounds), were used as a template phase to form anisotropic gold nanoparticles. Upon addition of the gold chloride solution, the nucleation process is initiated and both types of particles, i.e., isotropic spherical and anisotropic gold nanotriangles, are formed simultaneously. As it was not possible to produce monodisperse nanotriangles with such a one-step procedure, the anisotropic nanoparticles needed to be separated from the spherical ones. Therefore, a new type of separation procedure using combined polyelectrolyte/micelle depletion flocculation was successfully applied. As a result of the different purification steps, a green colored aqueous dispersion was obtained containing highly purified, well-defined negatively charged flat nanocrystals with a platelet thickness of 10 nm and an edge length of about 175 nm. The NTs produce promising results in surface-enhanced Raman scattering.
Study 1 targets grammatical restrictions, based on a corpus of peer group conversations among adolescents. We show that noncanonical variants have the form of bare NPs with or without preposition and appear in both multilingual and monolingual speech communities, following the same syntactic and semantic patterns. While there is a quantitative advantage for the multilingual group, noncanonical variants generally constitute only a minority compared to canonical full PP[DP]. Study 2 targets usage restrictions across communicative situations, based on a corpus of elicited productions by adolescents from a multilingual urban neighbourhood. Comparisons show significantly more noncanonical local expressions in informal, peer-group situations than in formal ones for both spoken and written modes. Taken together, results indicate a selective, grammatically restricted and register-bound choice of noncanonical local expressions.
"Seid mutig und aufrecht!"

Der 'Centralverein deutscher Staatsbürger jüdischen Glaubens' (C.V.) wurde 1893 in Berlin gegründet. Sein Tätigkeitsschwerpunkt lag ursprünglich auf dem Gebiet des Rechtsschutzes gegen antisemitische Diskriminierungen. Eine zweite Säule seiner Arbeit war die Aufklärung der Nichtjuden über das 'wahre Wesen' des Judentums. Johann Nicolai analysiert in diesem Band unter Auswertung der Moskauer Archivbestände die Entwicklung des Vereins von 1933 bis zu seiner Auflösung im Jahr 1938. Im Mittelpunkt der Studie steht die Frage, wie der Centralverein die Fortsetzung seiner Arbeit gegenüber seinen Mitgliedern begründete, obwohl im Nationalsozialismus die grundlegenden Bürgerrechte für die deutschen Juden, die er ursprünglich verteidigen wollte, nicht mehr gewährleistet waren. Ein besonderer Fokus liegt auf der Wandlung des Centralvereins nach den Nürnberger Gesetzen und der darauf erfolgten Umbenennung und Umstrukturierung. Wesentlicher Bestandteil ist auch die Auseinandersetzung mit der jüdischen Emigration nach Übersee. Nicolai zeigt auf, dass es bis tief in die 1930er-Jahre hinein starke Spannungen im deutschen Judentum gab.
New forms of communication and greater accessibility of Islamic texts on-line allow Muslims to shape their own religiosity, to become less dependent on established sources of authority, and thereby to become more aware of their own cultural diversity as a community. New practices of transnational Islam, and the growth of new concepts of Muslim identities currently emerging in the on-line community, are relatively free from immediate constraints. This article provides the result of a sociological analysis of three Internet sites in Sydney which deliver on-line fatwas. Even if cyberspace has allowed the Muslim world to be de-territorialised and provides a way for people to distance themselves from traditional communities if they wish, this research points out a variety of approaches, including one case which is aiming at re-localising an Australian Muslim system of values. This case highlights ways in which first generation Muslims are re-territorialising Shari'a in a specific western country.
The article offers a rereading of Sibylla Schwarz’s prose eclogue, Faunus. By describing the circumstances and the development of a young love in detail, Schwarz directs the reader’s attention to the fact, that Christian moral and ethical standards are external, and, therefore, are to be rejected. Instead, she places the ‘anthropological’ dimension in the foreground, the interest in human beings and their emotional motivations. Affective control and adaptation to conventions are less important. In that regard, this short text can be seen as an integral part of a prehistory of ‘literary anthropology,’ which ultimately evolved only in the course of the 18th century, although, as will be shown, its precursors can already be found in the 17th century.
The fruit of Aristotelia chilensis is considered a "super fruit" due to its high concentration of polyphenols displaying exceptional antioxidant capacities ORAC. From maqui berries have been reported several anthocyanins and glycosylated flavonoids, those benefits increase the attention to restudy the plant. From the leaves of A. chilensis several indole alkaloids have been reported, we in addition to aristoteline, aristone, aristoquinoline and 3-fromylindole report the spectroscopic elucidation of 8-oxo-9-dehydromakomakine (1), hobartine (2) and a new alkaloid named 8-oxohobartine (3). Compound 1 to 3 did not show bactericidal activity against E. coli and S. aureus till 200 mu g.
We dissect the sources of error leading to inaccuracies in the description of the geometry and optical excitation energies of pi-conjugated polymers. While the ground-state bond length alternation is shown to be badly reproduced by standard functionals, the recently adapted functionals PBEh* and omega PBE* as well as the double hybrid functional XYGJ-OS manage to replicate results obtained at the CCSD(T) level. By analysis of the bond length alternation in the excited state, a sensitive dependence of the exciton localization on the long-range behavior of the functional and the amount of Hartree-Fock exchange present is shown. Introducing thermal disorder through molecular dynamics simulations allows the consideration of a range of thermally accessible configurations of each oligomer, including trans to cis rotations, which break the conjugation of the backbone. Thermal disorder has a considerable effect when combined with functionals that overestimate the delocalization of the excitation, such as B3LYP. For functionals with a larger amount of exact exchange such as our PBEh* and omega PBE*, however, the effect is small, as excitations are often localized enough to fit between twists in the chain.
We present an algorithm that performs sequentially one-dimensional inversion of subsurface magnetic permeability and electrical conductivity by using multi-configuration electromagnetic induction sensor data. The presented method is based on the conversion of the in-phase and out-of-phase data into effective magnetic permeability and electrical conductivity of the equivalent homogeneous half-space. In the case of small-offset systems, such as portable electromagnetic induction sensors, for which in-phase and out-of-phase data are moderately coupled, the effective half-space magnetic permeability and electrical conductivity can be inverted sequentially within an iterative scheme. We test and evaluate the proposed inversion strategy using synthetic and field examples. First, we apply it to synthetic data for some highly magnetic environments. Then, the method is tested on real field data acquired in a basaltic environment to image a formation of archaeological interest. These examples demonstrate that a joint interpretation of in-phase and out-of-phase data leads to a better characterisation of the subsurface in magnetic environments such as volcanic areas.
The Earth’s shallow subsurface with sedimentary cover acts as a waveguide to any incoming wavefield. Within the framework of my thesis, I focused on the characterization of this shallow subsurface within tens to few hundreds of meters of sediment cover. I imaged the seismic 1D shear wave velocity (and possibly the 1D compressional wave velocity). This information is not only required for any seismic risk assessment, geotechnical engineering or microzonation activities, but also for exploration and global seismology where site effects are often neglected in seismic waveform modeling.
First, the conventional frequency-wavenumber (f - k) technique is used to derive the dispersion characteristic of the propagating surface waves recorded using distinct arrays of seismometers in 1D and 2D configurations. Further, the cross-correlation technique is applied to seismic array data to estimate the Green’s function between receivers pairs combination assuming one is the source and the other the receiver. With the consideration of a 1D media, the estimated cross-correlation Green’s functions are sorted with interstation distance in a virtual 1D active seismic experiment. The f - k technique is then used to estimate the dispersion curves. This integrated analysis is important for the interpretation of a large bandwidth of the phase velocity dispersion curves and therefore improving the resolution of the estimated 1D Vs profile.
Second, the new theoretical approach based on the Diffuse Field Assumption (DFA) is used for the interpretation of the observed microtremors H/V spectral ratio. The theory is further extended in this research work to include not only the interpretation of the H/V measured at the surface, but also the H/V measured at depths and in marine environments. A modeling and inversion of synthetic H/V spectral ratio curves on simple predefined geological structures shows an almost perfect recovery of the model parameters (mainly Vs and to a lesser extent Vp). These results are obtained after information from a receiver at depth has been considered in the inversion.
Finally, the Rayleigh wave phase velocity information, estimated from array data, and the H/V(z, f) spectral ratio, estimated from a single station data, are combined and inverted for the velocity profile information. Obtained results indicate an improved depth resolution in comparison to estimations using the phase velocity dispersion curves only. The overall estimated sediment thickness is comparable to estimations obtained by inverting the full micortremor H/V spectral ratio.
Here, we study the 3-D subduction initiation process induced by the interaction between a hot thermochemical mantle plume and oceanic lithosphere using thermo-mechanical viscoplastic finite difference marker-in-cell models. Our numerical modeling results show that self-sustaining subduction is induced by plume-lithosphere interaction when the plume is sufficiently buoyant, the oceanic lithosphere is sufficiently old and the plate is weak enough to allow the buoyant plume to. pass through it. Subduction initiation occurs following penetration of the lithosphere by the hot plume and the downward displacement of broken, nearly circular segments of lithosphere (proto-slabs) as a result of partially molten plume rocks overriding the proto-slabs. Our experiments show four different deformation regimes in response to plume-lithosphere interaction: a) self-sustaining subduction initiation, in which subduction becomes self-sustaining; b) frozen subduction initiation, in which subduction stops at shallow depths; c) slab break-off, in which the subducting circular slab breaks off soon after formation; and d) plume underplating, in which the plume does not pass through the lithosphere and instead spreads beneath it (i.e., failed subduction initiation). These regimes depend on several parameters, such as the size, composition, and temperature of the plume, the brittle/plastic strength and age of the oceanic lithosphere, and the presence/absence of lithospheric heterogeneities. The results show that subduction initiates and becomes self-sustaining when the lithosphere is older than 10 Myr and the non dimensional ratio of the plume buoyancy force and lithospheric strength above the plume is higher than approximately 2. The outcomes of our numerical experiments are applicable for subduction initiation in the modern and Precambrian Earth and for the origin of plume-related corona structures on Venus. Ice complex deposits are characteristic, ice-rich formations in northern East Siberia and represent an important part in the arctic carbon pool. Recently, these late Quaternary deposits are the objective of numerous investigations typically relying on outcrop and borehole data. Many of these studies can benefit from a 3D structural model of the subsurface for upscaling their observations or for constraining estimations of inventories, such as the local carbon stock. We have addressed this problem of structural imaging by 3D ground-penetrating radar (GPR), which, in permafrost studies, has been primarily used for 2D profiling. We have used a 3D kinematic GPR surveying strategy at a field site located in the New Siberian Archipelago on top of an ice complex. After applying a 3D GPR processing sequence, we were able to trace two horizons at depths below 20 m. Taking available borehole and outcrop data into account, we have interpreted these two features as interfaces of major lithologic units and derived a 3D cryostratigraphic model of the subsurface. Our data example demonstrated that a 3D surveying and processing strategy was crucial at our field site and showed the potential of 3D GPR to image geologic structures in complex ice-rich permafrost landscapes.
This study investigates the characteristics of narrative-speech production and the use of verbs in Turkish agrammatic speakers (n = 10) compared to non-brain-damaged controls (n = 10). To elicit narrative-speech samples, personal interviews and storytelling tasks were conducted. Turkish has a large and regular verb inflection paradigm where verbs are inflected for evidentiality (i.e. direct versus indirect evidence available to the speaker). Particularly, we explored the general characteristics of the speech samples (e.g. utterance length) and the uses of lexical, finite and non-finite verbs and direct and indirect evidentials. The results show that speech rate is slow, verbs per utterance are lower than normal and the verb diversity is reduced in the agrammatic speakers. Verb inflection is relatively intact; however, a trade-off pattern between inflection for direct evidentials and verb diversity is found. The implications of the data are discussed in connection with narrative-speech production studies on other languages.
The present study approaches the Spanish postposed constructions creo Ø and creo yo ‘[p], [I] think’ from a cognitive-constructionist perspective. It is argued that both constructions are to be distinguished from one another because creo Ø has a subjective function, while in creo yo, it is the intersubjective dimension that is particularly prominent. The present investigation takes both a qualitative and a quantitative perspective. With regard to the latter, the problem of quantitative representativity is addressed. The discussion posed the question of how empirical research can feed back into theory, more precisely, into the framework of Cognitive Construction Grammar. The data to be analyzed here are retrieved from the corpora Corpus de Referencia del Español Actual and Corpus del Español.
The importance of cultural ecosystem services in agricultural landscapes is increasingly recognized as agricultural scale enlargement and abandonment affect aesthetic and recreational values of agricultural landscapes. Landscape preference studies addressing these type of values often yield context-specific outcomes, limiting the applicability of their outcomes in landscape policy. Our approach measures the relative importance of landscape features across agricultural landscapes. This approach was applied in the agricultural landscapes of Winterswijk, The Netherlands (n=191) and the Markische Schweiz, Germany (n=113) among visitors in the agricultural landscape. We set up a parallel designed choice experiment, using regionally specific, photorealistic visualizations of four comparable landscape attributes. In the Dutch landscape visitors highly value hedgerows and tree lines, whereas groups of trees and crop diversity are highly valued in the German landscape. Furthermore, we find that differences in relative preference for landscape attributes are, to some extent, explained by socio-cultural background variables such as education level and affinity with agriculture of the visitors. This approach contributes to a better understanding of the cross-regional variation of aesthetic and recreational values and how these values relate to characteristics of the agricultural landscape, which could support the integration of cultural services in landscape policy. In this paper two groups supporting different views on the mechanism of light induced polymer deformation argue about the respective underlying theoretical conceptions, in order to bring this interesting debate to the attention of the scientific community. The group of Prof. Nicolae Hurduc supports the model claiming that the cyclic isomerization of azobenzenes may cause an athermal transition of the glassy azobenzene containing polymer into a fluid state, the so-called photo-fluidization concept. This concept is quite convenient for an intuitive understanding of the deformation process as an anisotropic flow of the polymer material. The group of Prof. Svetlana Santer supports the re-orientational model where the mass-transport of the polymer material accomplished during polymer deformation is stated to be generated by the light-induced re-orientation of the azobenzene side chains and as a consequence of the polymer backbone that in turn results in local mechanical stress, which is enough to irreversibly deform an azobenzene containing material even in the glassy state. For the debate we chose three polymers differing in the glass transition temperature, 32 degrees C, 87 degrees C and 95 degrees C, representing extreme cases of flexible and rigid materials. Polymer film deformation occurring during irradiation with different interference patterns is recorded using a homemade set-up combining an optical part for the generation of interference patterns and an atomic force microscope for acquiring the kinetics of film deformation. We also demonstrated the unique behaviour of azobenzene containing polymeric films to switch the topography in situ and reversibly by changing the irradiation conditions. We discuss the results of reversible deformation of three polymers induced by irradiation with intensity (IIP) and polarization (PIP) interference patterns, and the light of homogeneous intensity in terms of two approaches: the re-orientational and the photo-fluidization concepts. Both agree in that the formation of opto-mechanically induced stresses is a necessary prerequisite for the process of deformation. Using this argument, the deformation process can be characterized either as a flow or mass transport.
Individuals with agrammatic Broca's aphasia experience difficulty when processing reversible non-canonical sentences. Different accounts have been proposed to explain this phenomenon. The Trace Deletion account (Grodzinsky, 1995, 2000, 2006) attributes this deficit to an impairment in syntactic representations, whereas others (e.g., Caplan, Waters, Dede, Michaud, & Reddy, 2007; Haarmann, Just, & Carpenter, 1997) propose that the underlying structural representations are unimpaired, but sentence comprehension is affected by processing deficits, such as slow lexical activation, reduction in memory resources, slowed processing and/or intermittent deficiency, among others. We test the claims of two processing accounts, slowed processing and intermittent deficiency, and two versions of the Trace Deletion Hypothesis (TDH), in a computational framework for sentence processing (Lewis & Vasishth, 2005) implemented in ACT-R (Anderson, Byrne, Douglass, Lebiere, & Qin, 2004). The assumption of slowed processing is operationalized as slow procedural memory, so that each processing action is performed slower than normal, and intermittent deficiency as extra noise in the procedural memory, so that the parsing steps are more noisy than normal. We operationalize the TDH as an absence of trace information in the parse tree. To test the predictions of the models implementing these theories, we use the data from a German sentence—picture matching study reported in Hanne, Sekerina, Vasishth, Burchert, and De Bleser (2011). The data consist of offline (sentence-picture matching accuracies and response times) and online (eye fixation proportions) measures. From among the models considered, the model assuming that both slowed processing and intermittent deficiency are present emerges as the best model of sentence processing difficulty in aphasia. The modeling of individual differences suggests that, if we assume that patients have both slowed processing and intermittent deficiency, they have them in differing degrees.

Multimodal representation learning has gained increasing importance in various real-world multimedia applications. Most previous approaches focused on exploring inter-modal correlation by learning a common or intermediate space in a conventional way, e.g. Canonical Correlation Analysis (CCA). These works neglected the exploration of fusing multiple modalities at higher semantic level. In this paper, inspired by the success of deep networks in multimedia computing, we propose a novel unified deep neural framework for multimodal representation learning. To capture the high-level semantic correlations across modalities, we adopted deep learning feature as image representation and topic feature as text representation respectively. In joint model learning, a 5-layer neural network is designed and enforced with a supervised pre-training in the first 3 layers for intra-modal regularization. The extensive experiments on benchmark Wikipedia and MIR Flickr 25K datasets show that our approach achieves state-of-the-art results compare to both shallow and deep models in multimodal and cross-modal retrieval.
The molecular biomarker composition of two sediment cores from Sanabria Lake (NW Iberian Peninsula) and a survey of modern plants in the watershed provide a reconstruction of past vegetation and landscape dynamics since deglaciation. During a proglacial stage in Lake Sanabria (prior to 14.7 cal ka BP), very low biomarker concentration and carbon preference index (CPI) values similar to 1 suggest that the n-alkanes could have derived from eroded ancient sediment sources or older organic matter with high degree of maturity. During the Late glacial (14.7-11.7 cal ka BP) and the Holocene (last 11.7 cal ka BP) intervals with higher biomarker and triterpenoid concentrations (high %nC(29) , nC(31) alkanes), higher CPI and average carbon length (ACL), and lower P-aq (proportion of aquatic plants) are indicative of major contribution of vascular land plants from a more forested watershed (e.g. Mid Holocene period 7.0-4.0 cal ka BP). Lower biomarker concentrations (high %nC(27) alkanes), CPI and ACL values responded to short phases with decreased allochthonous contribution into the lake that correspond to centennial-scale periods of regional forest decline (e.g. 4-3 ka BP, Roman deforestation after 2.0 ka, and some phases of the LIA, seventeenth-nineteenth centuries). Human activities in the watershed were significant during early medieval times (1.3-1.0 cal ka BP) and since 1960 CE, in both cases associated with relatively higher productivity stages in the lake (lower biomarker and triterpenoid concentrations, high %nC(23) and %nC(31) respectively, lower ACL and CPI values and higher P-aq). The lipid composition of Sanabria Lake sediments indicates a major allochthonous (watershed-derived) contribution to the organic matter budget since deglaciation, and a dominant oligotrophic status during the lake history. The study constrains the climate and anthropogenic forcings and watershed versus lake sources in organic matter accumulation processes and helps to design conservation and management policies in mountain, oligotrophic lakes.

When sampling animal movement paths, the frequency at which location measurements are attempted is a critical feature for data analysis. Important quantities derived from raw data, e.g. travel distance or sinuosity, can differ largely based on the temporal resolution of the data. Likewise, when movement models are fitted to data, parameter estimates have been demonstrated to vary with sampling rate. Thus, biological statements derived from such analyses can only be made with respect to the resolution of the underlying data, limiting extrapolation of results and comparison between studies. To address this problem, we investigate whether there are models that are robust against changes in temporal resolution. First, we propose a mathematically rigorous framework, in which we formally define robustness as a model property. We then use the framework for a thorough assessment of a range of basic random walk models, in which we also show how robustness relates to other probabilistic concepts. While we found robustness to be a strong condition met by few models only, we suggest a new method to extend models so as to make them robust. Our framework provides a new systematic, mathematically founded approach to the question if, and how, sampling rate of movement paths affects statistical inference.
The outermost cell layer of plants, the epidermis, and its outer (lateral) membrane domain facing the environment are continuously challenged by biotic and abiotic stresses. Therefore, the epidermis and the outer membrane domain provide important selective and protective barriers. However, only a small number of specifically outer membrane-localized proteins are known. Similarly, molecular mechanisms underlying the trafficking and the polar placement of outer membrane domain proteins require further exploration. Here, we demonstrate that ACTIN7 (ACT7) mediates trafficking of the PENETRATION3 (PEN3) outer membrane protein from the trans-Golgi network (TGN) to the plasma membrane in the root epidermis of Arabidopsis (Arabidopsis thaliana) and that actin function contributes to PEN3 endocytic recycling. In contrast to such generic ACT7-dependent trafficking from the TGN, the EXOCYST84b (EXO84b) tethering factor mediates PEN3 outer-membrane polarity. Moreover, precise EXO84b placement at the outer membrane domain itself requires ACT7 function. Hence, our results uncover spatially and mechanistically distinct requirements for ACT7 function during outer lateral membrane cargo trafficking and polarity establishment. They further identify an exocyst tethering complex mediator of outer lateral membrane cargo polarity.

Sphingolipids are major components of the plasma membrane. In particular, ceramide serves as an essential building hub for complex sphingolipids, but also as an organizer of membrane domains segregating receptors and signalosomes. Sphingomyelin breakdown as a result of sphingomyelinase activation after ligation of a variety of receptors is the predominant source of ceramides released at the plasma membrane. This especially applies to T lymphocytes where formation of ceramide-enriched membrane microdomains modulates TCR signaling. Because ceramide release and redistribution occur very rapidly in response to receptor ligation, novel tools to further study these processes in living T cells are urgently needed. To meet this demand, we synthesized nontoxic, azido-functionalized ceramides allowing for bio-orthogonal click-reactions to fluorescently label incorporated ceramides, and thus investigate formation of ceramide-enriched domains. Azido-functionalized C-6-ceramides were incorporated into and localized within plasma membrane microdomains and proximal vesicles in T cells. They segregated into clusters after TCR, and especially CD28 ligation, indicating efficient sorting into plasma membrane domains associated with T cell activation; this was abolished upon sphingomyelinase inhibition. Importantly, T cell activation was not abrogated upon incorporation of the compound, which was efficiently excluded from the immune synapse center as has previously been seen in Ab-based studies using fixed cells. Therefore, the functionalized ceramides are novel, highly potent tools to study the subcellular redistribution of ceramides in the course of T cell activation. Moreover, they will certainly also be generally applicable to studies addressing rapid stimulation-mediated ceramide release in living cells.
Liverwort Blasia pusilla L. recruits soil nitrogen-fixing cyanobacteria of genus Nostoc as symbiotic partners. In this work we compared Nostoc community composition inside the plants and in the soil around them from two distant locations in Northern Norway. STRR fingerprinting and 16S rDNA phylogeny reconstruction showed a remarkable local diversity among isolates assigned to several Nostoc clades. An extensive web of negative allelopathic interactions was recorded at an agricultural site, but not at the undisturbed natural site. The cell extracts of the cyanobacteria did not show antimicrobial activities, but four isolates were shown to be cytotoxic to human cells. The secondary metabolite profiles of the isolates were mapped by MALDI-TOF MS, and the most prominent ions were further analyzed by Q-TOF for MS/MS aided identification. Symbiotic isolates produced a great variety of small peptide-like substances, most of which lack any record in the databases. Among identified compounds we found microcystin and nodularin variants toxic to eukaryotic cells. Microcystin producing chemotypes were dominating as symbiotic recruits but not in the free-living community. In addition, we were able to identify several novel aeruginosins and banyaside-like compounds, as well as nostocyclopeptides and nosperin.

A holistic design and verification environment to investigate driving assistance systems is presented, with an emphasis on system-on-chip architectures for video applications. Starting with an executable specification of a driving assistance application, subsequent transformations are performed across different levels of abstraction until the final implementation is achieved. The hardware/software partitioning is facilitated through the integration of OpenCV and SystemC in the same design environment, as well as OpenCV and Linux in the run-time system. We built a rapid prototyping, FPGA-based camera system, which allows designs to be explored and evaluated in realistic conditions. Using lane departure and the corresponding performance speedup, we show that our platform reduces the design time, while improving the verification efforts.
A desirable goal is to synthesize easily accessible and highly K+/Na+-selective fluoroionophores to monitor physiological K+ levels in vitro and in vivo. Therefore, highly K+/Na+-selective ionophores have to be developed. Herein, we obtained in a sequence of only four synthetic steps a set of K+-responsive fluorescent probes 4, 5 and 6. In a systematic study, we investigated the influence of the alkoxy substitution in ortho position of the aniline moiety in -conjugated aniline-1,2,3-triazole-coumarin-fluoroionophores 4, 5 and 6 [R=MeO (4), EtO (5) and iPrO (6)] towards the K+-complex stability and K+/Na+ selectivity. The highest K+-complex stability showed fluoroionophore 4 with a dissociation constant K-d of 19mm, but the K-d value increases to 31mm in combined K+/Na+ solutions, indicating a poor K+/Na+ selectivity. By contrast, 6 showed even in the presence of competitive Na+ ions equal K-d values (K-d(K+)=45mm and K-d(K+/Na+)=45mm) and equal K+-induced fluorescence enhancement factors (FEFs=2.3). Thus, the fluorescent probe 6 showed an outstanding K+/Na+ selectivity and is a suitable fluorescent tool to measure physiological K+ levels in the range of 10-80mm in vitro. Further, the isopropoxy-substituted N-phenylaza[18]crown-6 ionophore in 6 is a highly K+-selective building block with a feasible synthetic route.
We elaborate a boundary Fourier method for studying an analogue of the Hilbert problem for analytic functions within the framework of generalised Cauchy-Riemann equations. The boundary value problem need not satisfy the Shapiro-Lopatinskij condition and so it fails to be Fredholm in Sobolev spaces. We show a solvability condition of the Hilbert problem, which looks like those for ill-posed
problems, and construct an explicit formula for approximate solutions.
In nature, plants often encounter chronic or recurring stressful conditions. Recent results indicate that plants can remember a past exposure to stress to be better prepared for a future stress incident. However, the molecular basis of this is poorly understood. Here, we report the involvement of chromatin modifications in the maintenance of acquired thermotolerance (heat stress [HS] memory). HS memory is associated with the accumulation of histone H3 lysine 4 di- and trimethylation at memory-related loci. This accumulation outlasts their transcriptional activity and marks them as recently transcriptionally active. High accumulation of H3K4 methylation is associated with hyper-induction of gene expression upon a recurring HS. This transcriptional memory and the sustained accumulation of H3K4 methylation depend on HSFA2, a transcription factor that is required for HS memory, but not initial heat responses. Interestingly, HSFA2 associates with memory-related loci transiently during the early stages following HS. In summary, we show that transcriptional memory after HS is associated with sustained H3K4 hyper-methylation and depends on a hit-and-run transcription factor, thus providing a molecular framework for HS memory.
Transposable elements (TEs) make up a large proportion of eukaryotic genomes. As their mobilization creates genetic variation that threatens genome integrity, TEs are epigenetically silenced through several pathways, and this may spread to neighboring sequences. JUMONJI (JMJ) proteins can function as antisilencing factors and prevent silencing of genes next to TEs. Whether TE silencing is counterbalanced by the activity of antisilencing factors is still unclear. Here, we characterize JMJ24 as a regulator of TE silencing. We show that loss of JMJ24 results in increased silencing of the DNA transposon AtMu1c, while overexpression of JMJ24 reduces silencing. JMJ24 has a JumonjiC (JmjC) domain and two RING domains. JMJ24 autoubiquitinates in vitro, demonstrating E3 ligase activity of the RING domain(s). JMJ24-JmjC binds the N-terminal tail of histone H3, and full-length JMJ24 binds histone H3 in vivo. JMJ24 activity is anticorrelated with histone H3 Lys 9 dimethylation (H3K9me2) levels at AtMu1c. Double mutant analyses with epigenetic silencing mutants suggest that JMJ24 antagonizes histone H3K9me2 and requires H3K9 methyltransferases for its activity on AtMu1c. Genome-wide transcriptome analysis indicates that JMJ24 affects silencing at additional TEs. Our results suggest that the JmjC domain of JMJ24 has lost demethylase activity but has been retained as a binding domain for histone H3. This is in line with phylogenetic analyses indicating that JMJ24 (with the mutated JmjC domain) is widely conserved in angiosperms. Taken together, this study assigns a role in TE silencing to a conserved JmjC-domain protein with E3 ligase activity, but no demethylase activity.
This manuscript proposes a method to assess hydrological drought in semi-arid environments under high impoundment rate and applies it to the semi-arid Jaguaribe River basin in Brazil. It analyzes droughts (1) in the largest reservoir systems; (2) in the Upper Basin, considering 4744 reservoirs, 800 wells and almost 18,000 cisterns; and (3) in reservoirs of different sizes during multiyear droughts. Results show that the water demand is constrained in the basin; hydrological and meteorological droughts are often out of phase; there is a negative correlation between storage level and drought severity; and the small systems cannot cope with long-term droughts.
Cells contain a finite set of resources that must be distributed across many processes to ensure survival. Among them, the largest proportion of cellular resources is dedicated to protein translation. Synthetic biology often exploits these resources in executing orthogonal genetic circuits, yet the burden this places on the cell is rarely considered. Here, we develop a minimal model of ribosome allocation dynamics capturing the demands on translation when expressing a synthetic construct together with endogenous genes required for the maintenance of cell physiology. Critically, it contains three key variables related to design parameters of the synthetic construct covering transcript abundance, translation initiation rate, and elongation time. We show that model-predicted changes in ribosome allocation closely match experimental shifts in synthetic protein expression rate and cellular growth. Intriguingly, the model is also able to accurately infer transcript levels and translation times after further exposure to additional ambient stress. Our results demonstrate that a simple model of resource allocation faithfully captures the redistribution of protein synthesis resources when faced with the burden of synthetic gene expression and environmental stress. The tractable nature of the model makes it a versatile tool for exploring the guiding principles of efficient heterologous expression and the indirect interactions that can arise between synthetic circuits and their host chassis because of competition for shared translational resources.

We analyze in detail appearance and stability properties of this state in possibly the simplest setup of a biharmonic Kuramoto-Daido phase model as well as demonstrate the effect in limit-cycle relaxational Rayleigh oscillators. Such a regime extends the notion of splay state from a uniform distribution of phases to an oscillating one. Suitable collective observables such as the Kuramoto order parameter allow detecting the presence of an inhomogeneous distribution. The characteristic and most peculiar property of self-consistent partial synchrony is the difference between the frequency of single units and that of the macroscopic field.

The ground-state classical path approximation is utilized to compute molecular absorption spectra in a mixed quantum-classical frame. To improve the description for high-frequency vibrational satellites, related quantum correction factors are introduced. A model analysis of mechanisms for radial microtubular patterns at root hair initiation sites

Diffuse growth where whole cell walls extend in specific directions, guided by anisotropically positioned cellulose fibers, and tip growth, with inhomogeneous addition of new cell wall material at the tip of the structure. Cells are known to regulate these processes via molecular signals and the cytoskeleton. Mechanical stress has been proposed to provide an input to the positioning of the cellulose fibers via cortical microtubules in diffuse growth. In particular, a stress feedback model predicts a circumferential pattern of fibers surrounding apical tissues and growing primordia, guided by the anisotropic curvature in such tissues. In contrast, during the initiation of tip growing root hairs, a star-like radial pattern has recently been observed. Here, we use detailed finite element models to analyze how a change in mechanical properties at the root hair initiation site can lead to star-like stress patterns in order to understand whether a stress-based feedback model can also explain the microtubule patterns seen during root hair initiation. We show that two independent mechanisms, individually or combined, can be sufficient to generate radial patterns. In the first, new material is added locally at the position of the root hair. In the second, increased tension in the initiation area provides a mechanism. Finally, we describe how a molecular model of Rho-of-plant (ROP) GTPases activation driven by auxin can position a patch of activated ROP protein basally along a 2D root epidermal cell plasma membrane, paving the way for models where mechanical and molecular mechanisms cooperate in the initial placement and outgrowth of root hairs.

We have synthesized mesoporous silica (monoliths) with defined surface chemistry by means of a number of addition reactions: (i) coupling of an isocyanate to a surface-immobilized thiol, (ii) addition of an epoxide to a surface-immobilized thiol, (iii) cross-metathesis between two olefins, and (iv) Huisgen [2+3] cycloaddition of an alkyne-functionalized silica monolith with an azide. Functionalization of the mesopores was observed, but there are significant differences between individual approaches. Isocyanate and epoxide additions lead to high degrees of functionalization, whereas olefin metathesis and [2+3] cycloaddition are less effective. We further show that the efficiency of the modification is about twice as high in mesoporous silica particles than in macroscopic silica monoliths.
The knowledge of the contemporary in situ stress state is a key issue for safe and sustainable subsurface engineering. However, information on the orientation and magnitudes of the stress state is limited and often not available for the areas of interest. Therefore 3-D geomechanical-numerical modelling is used to estimate the in situ stress state and the distance of faults from failure for application in subsurface engineering. The main challenge in this approach is to bridge the gap in scale between the widely scattered data used for calibration of the model and the high resolution in the target area required for the application. We present a multi-stage 3-D geomechanical-numerical approach which provides a state-of-the-art model of the stress field for a reservoir-scale area from widely scattered data records. Therefore, we first use a large-scale regional model which is calibrated by available stress data and provides the full 3-D stress tensor at discrete points in the entire model volume. The modelled stress state is used subsequently for the calibration of a smaller-scale model located within the large-scale model in an area without any observed stress data records. We exemplify this approach with two-stages for the area around Munich in the German Molasse Basin. As an example of application, we estimate the scalar values for slip tendency and fracture potential from the model results as measures for the criticality of fault reactivation in the reservoir-scale model. The modelling results show that variations due to uncertainties in the input data are mainly introduced by the uncertain material properties and missing S-Hmax magnitude estimates needed for a more reliable model calibration. This leads to the conclusion that at this stage the model's reliability depends only on the amount and quality of available stress information rather than on the modelling technique itself or on local details of the model geometry. Any improvements in modelling and increases in model reliability can only be achieved using more high-quality data for calibration.
