We present one-dimensional, time-dependent models of the clumps generated by the linedeshadowing instability. In order to follow the clumps out to distances of more than 1000 R∗, we use an efficient moving-box technique. We show that, within the approximations, the wind can remain clumped well into the formation region of the radio continuum.
INTEGRAL tripled the number of super-giant high-mass X-ray binaries (sgHMXB) known in the Galaxy by revealing absorbed and fast transient (SFXT) systems. Quantitative constraints on the wind clumping of massive stars can be obtained from the study of the hard X-ray variability of SFXT. A large fraction of the hard X-ray emission is emitted in the form of flares with a typical duration of 3 ksec, frequency of 7 days and luminosity of $10^{36}$ erg/s. Such flares are most probably emitted by the interaction of a compact object orbiting at $\sim10~R_*$ with wind clumps ($10^{22 ... 23}$ g) representing a large fraction of the stellar mass-loss rate. The density ratio between the clumps and the inter-clump medium is $10^{2 ... 4}$. The parameters of the clumps and of the inter-clump medium, derived from the SFXT flaring behavior, are in good agreement with macro-clumping scenario and line-driven instability simulations. SFXT are likely to have larger orbital radius than classical sgHMXB.
Magnetic fields influence the dynamics of hot-star winds and create large scale structure. Based on numerical magnetohydrodynamic (MHD) simulations, we model the wind of θ¹ Ori C, and then use the SEI method to compute synthetic line profiles for a range of viewing angles as function of rotational phase. The resulting dynamic spectrum for a moderately strong line shows a distinct modulation, but with a phase that seems at odds with available observations.

Dynamical simulation of the “velocity-porosity” reduction in observed strength of stellar wind lines

I use dynamical simulations of the line-driven instability to examine the potential role of the resulting flow structure in reducing the observed strength of wind absorption lines. Instead of the porosity length formalism used to model effects on continuum absorption, I suggest reductions in line strength can be better characterized in terms of a velocity clumping factor that is insensitive to spatial scales. Examples of dynamic spectra computed directly from instability simulations do exhibit a net reduction in absorption, but only at a modest 10-20% level that is well short of the ca. factor 10 required by recent analyses of PV lines.
The James Webb Space Telescope (JWST) is a large, infrared-optimized space telescope scheduled for launch in 2013. JWST will find the first stars and galaxies that formed in the early universe, connecting the Big Bang to our own Milky Way galaxy. JWST will peer through dusty clouds to see stars forming planetary systems, connecting the MilkyWay to our own Solar System. JWST’s instruments are designed to work primarily in the infrared range of 1 - 28 μm, with some capability in the visible range. JWST will have a large mirror, 6.5 m in diameter, and will be diffraction-limited at 2 μm (0.1 arcsec resolution). JWST will be placed in an L2 orbit about 1.5 million km from the Earth. The instruments will provide imaging, coronography, and multi-object and integral-field spectroscopy across the 1 - 28 μm wavelength range. The breakthrough capabilities of JWST will enable new studies of massive star winds from the Milky Way to the early universe.
Clumps in hot star winds can originate from shock compression due to the line driven instability. One-dimensional hydrodynamic simulations reveal a radial wind structure consisting of highly compressed shells separated by voids, and colliding with fast clouds. Two-dimensional simulations are still largely missing, despite first attempts. Clumpiness dramatically affects the radiative transfer and thus all wind diagnostics in the UV, optical, and in X-rays. The microturbulence approximation applied hitherto is currently superseded by a more sophisticated radiative transfer in stochastic media. Besides clumps, i.e. jumps in the density stratification, so-called kinks in the velocity law, i.e. jumps in dv/dr, play an eminent role in hot star winds. Kinks are a new type of radiative-acoustic shock, and propagate at super-Abbottic speed.

We study the influence of clumping on the predicted wind structure of O-type stars. For this purpose we artificially include clumping into our stationary wind models. When the clumps are assumed to be optically thin, the radiative line force increases compared to corresponding unclumped models, with a similar effect on either the mass-loss rate or the terminal velocity (depending on the onset of clumping). Optically thick clumps, alternatively, might be able to decrease the radiative force.
We present the results of Monte Carlo mass-loss predictions for massive stars covering a wide range of stellar parameters. We critically test our predictions against a range of observed massloss rates – in light of the recent discussions on wind clumping. We also present a model to compute the clumping-induced polarimetric variability of hot stars and we compare this with observations of Luminous Blue Variables, for which polarimetric variability is larger than for O and Wolf-Rayet stars. Luminous Blue Variables comprise an ideal testbed for studies of wind clumping and wind geometry, as well as for wind strength calculations, and we propose they may be direct supernova progenitors.
Many hot stars exhibit stochastic polarimetric variability, thought to arise from clumping low in the wind. Here we investigate the wind properties required to reproduce this variability using analytic models, with particular emphasis on Luminous Blue Variables. We find that the winds must be highly structured, consisting of a large number of optically-thin clumps; while we find that the overall level of polarization should scale with mass-loss rate – consistent with observations of LBVs. The models also predict variability on very short timescales, which is supported by the results of a recent polarimetric monitoring campaign.
We investigate the effect of wind clumping on the dynamics of Wolf-Rayet winds, by means of the Potsdam Wolf-Rayet (PoWR) hydrodynamic atmosphere models. In the limit of microclumping the radiative acceleration is generally enhanced. We examine the reasons for this effect and show that the resulting wind structure depends critically on the assumed radial dependence of the clumping factor D(r). The observed terminal wind velocities for WR stars imply that D(r) increases to very large values in the outer part of the wind, in agreement with the assumption of detached expanding shells.
Overwhelming observational and theoretical evidence suggests that the winds of massive stars are highly clumped. We briefly discuss the influence of clumping on model diagnostics and the difficulties of allowing for the influence of clumping on model spectra. Because of its simplicity, and because of computational ease, most spectroscopic analyses incorporate clumping using the volume filling factor. The biases introduced by this approach are uncertain. To investigate alternative clumping models, and to help determine the validity of parameters derived using the volume filling factor method, we discuss results derived using an alternative model in which we assume that the wind is composed of optically thick shells.
We report FUSE observations in 2005–2006 of three O-type, double-lined spectroscopic binaries in the Magellanic Clouds. The systems have very short periods (1.4–2.25 d), represent rare, young evolutionary stages of massive stars and binaries, and provide a unique glimpse at some of the most massive systems that form in dense clusters of massive stars. Improved orbit parameters, including revised masses, for LH54-425 are derived from new ctio spectroscopy. The systems are: LH54-425 in the LMC (O3V + O5V, P=2.25d, 62+37M⊙), J053441-693139 in the LMC (O2-3If+O6V, P=1.4 d, 41+27M⊙), and Hodge 53-47 in the SMC (O6V + O4-5IIIf, P=2.2 d, 24+14M⊙, where the O4 star appears to be less massive than the O6 star). Their short periods indicates that wind interaction and mass transfer are likely important factors in their evolution. The spectra provide quantitative and systematic studies of phase-dependent stellar wind properties, wind collision effects in O+O binaries at lower metallicities, improved radial velocity curves, and FUV spectro-photometric changes as a function of orbital phase.
We present preliminary results of a tailored atmosphere analysis of six Galactic WC stars using UV, optical, and mid-infrared Spitzer IRS data. With these data, we are able to sample regions from 10 to 10³ stellar radii, thus to determine wind clumping in different parts of the wind. Ultimately, derived wind parameters will be used to accuratelymeasure neon abundances, and to so test predicted nuclear-reaction rates.
Mass accretion onto compact objects through accretion disks is a common phenomenon in the universe. It is seen in all energy domains from active galactic nuclei through cataclysmic variables (CVs) to young stellar objects. Because CVs are fairly easy to observe, they provide an ideal opportunity to study accretion disks in great detail and thus help us to understand accretion also in other energy ranges. Mass accretion in these objects is often accompanied by mass outflow from the disks. This accretion disk wind, at least in CVs, is thought to be radiatively driven, similar to O star winds. WOMPAT, a 3-D Monte Carlo radiative transfer code for accretion disk winds of CVs is presented.
We apply the 3-dimensional radiative transport codeWind3D to 3D hydrodynamic models of Corotating Interaction Regions to fit the detailed variability of Discrete Absorption Components observed in Si iv UV resonance lines of HD 64760 (B0.5 Ib). We discuss important effects of the hydrodynamic input parameters on these large-scale equatorial wind structures that determine the detailed morphology of the DACs computed with 3D transfer. The best fit model reveals that the CIR in HD 64760 is produced by a source at the base of the wind that lags behind the stellar surface rotation. The non-corotating coherent wind structure is an extended density wave produced by a local increase of only 0.6% in the smooth symmetric wind mass-loss rate.
Modeling expanding atmospheres is a difficult task because of the extreme non-LTE situation, the need to account for complex model atoms, especially for the iron-group elements with their millions of lines, and because of the supersonic expansion. Adequate codes have been developed e.g. by Hillier (CMFGEN), the Munich group (Puls, Pauldrach), and in Potsdam (PoWR code, Hamann et al.). While early work was based on the assumption of a smooth and homogeneous spherical stellar wind, the need to account for clumping became obvious about ten years ago. A relatively simple first-order clumping correction was readily implemented into the model codes. However, its simplifying assumptions are severe. Most importantly, the clumps are taken to be optically thin at all frequencies (”microclumping”). We discuss the consequences of this approximation and describe an approach to account for optically thick clumps (“macroclumping”). First results demonstrate that macroclumping can generally reduce the strength of spectral features, depending on their optical thickness. The recently reported discrepancy between the Hα diagnostic and the Pv resonance lines in O star spectra can be resolved without decreasing the mass-loss rates, when macroclumping is taken into account.
Clumping in Galactic WN stars : a comparison of mass loss rates from UV/optical & radio diagnostics

The mass loss rates and other parameters for a large sample of Galactic WN stars have been revised by Hamann et al. (2006), using the most up-to date Potsdam Wolf-Rayet (PoWR) model atmospheres. For a sub-sample of these stars exist measurements of their radio free-free emission. After harmonizing the adopted distance and terminal wind velocities, we compare the mass loss rates obtained from the two diagnostics. The differences are discussed as a possible consequence of different clumping contrast in the line-forming and radio-emitting regions.
Recent studies of massive O-type stars present clear evidences of inhomogeneous and clumped winds. O-type (H-rich) central stars of planetary nebulae (CSPNs) are in some ways the low mass–low luminosity analogous of those massive stars. In this contribution, we present preliminary results of our on-going multi-wavelength (FUV, UV and optical) study of the winds of Galactic CSPNs. Particular emphasis will be given to the clumping factors derived by means of optical lines (Hα and Heii 4686) and “classic” FUV (and UV) lines.
We exploit time-series $FUSE$ spectroscopy to {\it uniquely} probe spatial structure and clumping in the fast wind of the central star of the H-rich planetary nebula NGC~6543 (HD~164963). Episodic and recurrent optical depth enhancements are discovered in the P{\sc v} absorption troughs, with some evidence for a $\sim$ 0.17-day modulation time-scale. The characteristics of these features are essentially identical to the discrete absorption components' (DACs) commonly seen in the UV lines of massive OB stars, suggesting the temporal structures seen in NGC~6543 likely have a physical origin that is similar to that operating in massive, luminous stars. The mechanism for forming coherent perturbations in the outflows is therefore apparently operating equally in the radiation-pressure-driven winds of widely differing momenta ($\mdot$$v_\infty$$R_\star^{0.5}$) and flow times, as represented by OB stars and CSPN.
This paper outlines a newly-developed method to include the effects of time variability in the radiative transfer code CMFGEN. It is shown that the flow timescale is often large compared to the variability timescale of LBVs. Thus, time-dependent effects significantly change the velocity law and density structure of the wind, affecting the derivation of the mass-loss rate, volume filling factor, wind terminal velocity, and luminosity. The results of this work are directly applicable to all active LBVs in the Galaxy and in the LMC, such as AG Car, HR Car, S Dor and R 127, and could result in a revision of stellar and wind parameters. The massloss rate evolution of AG Car during the last 20 years is presented, highlighting the need for time-dependent models to correctly interpret the evolution of LBVs.
We discuss the results of time-resolved spectroscopy of three presumably single Population I Wolf-Rayet stars in the Small Magellanic Cloud, where the ambient metallicity is $\sim 1/5 Z_\odot$. We were able to detect and follow numerous small-scale wind-embedded inhomogeneities in all observed stars. The general properties of the moving features, such as their velocity dispersions, emissivities and average accelerations, closely match the corresponding characteristics of small-scale inhomogeneities in the winds of Galactic Wolf-Rayet stars.
The influence of the wind to the total continuum of OB supergiants is discussed. For wind velocity distributions with β > 1.0, the wind can have strong influence to the total continuum emission, even at optical wavelengths. Comparing the continuum emission of clumped and unclumped winds, especially for stars with high β values, delivers flux differences of up to 30% with maximum in the near-IR. Continuum observations at these wavelengths are therefore an ideal tool to discriminate between clumped and unclumped winds of OB supergiants.
Massive stars usually form groups such as OB associations. Their fast stellar winds sweep up collectively the surrounding insterstellar medium (ISM) to generate superbubbles. Observations suggest that superbubble evolution on the surrounding ISM can be very irregular. Numerical simulations considering these conditions could help to understand the evolution of these superbubbles and to clarify the dynamics of these objects as well as the difference between observed X-ray luminosities and the predicted ones by the standard model (Weaver et al. 1977).
We present the latest results on the observational dependence of the mass-loss rate in stellar winds of O and early-B stars on the metal content of their atmospheres, and compare these with predictions. Absolute empirical rates for the mass loss of stars brighter than 10$^{5.2} L_{\odot}$, based on H$\alpha$ and ultraviolet (UV) wind lines, are found to be about a factor of two higher than predictions. If this difference is attributed to inhomogeneities in the wind this would imply that luminous O and early-B stars have clumping factors in their H$\alpha$ and UV line forming regime of about a factor of 3--5. The investigated stars cover a metallicity range $Z$ from 0.2 to 1 $Z_{\odot}$. We find a hint towards smaller clumping factors for lower $Z$. The derived clumping factors, however, presuppose that clumping does not impact the predictions of the mass-loss rate. We discuss this assumption and explain how we intend to investigate its validity in more detail.
While there is strong evidence for clumping in the winds of massive hot stars, very little is known about clumping in the winds from Central Stars. We have checked [WC]-type CSPN winds for clumping by inspecting the electron-scattering line wings. At least for three stars we found indications for wind inhomogeneities.
We report on new mass-loss rate estimates for O stars in six massive binaries using the amplitude of orbital-phase dependent, linear-polarimetric variability caused by electron scattering off free electrons in the winds. Our estimated mass-loss rates for luminous O stars are independent of clumping. They suggest similar clumping corrections as for WR stars and do not support the recently proposed reduction in mass-loss rates of O stars by one or two orders of magnitude.
Clumping in O-star winds

We have analyzed the spectra of seven Galactic O4 supergiants, with the NLTE wind code CMFGEN. For all stars, we have found that clumped wind models match well lines from different species spanning a wavelength range from FUV to optical, and remain consistent with Hα data. We have achieved an excellent match of the P V λλ1118, 1128 resonance doublet and N IV λ1718, as well as He II λ4686 suggesting that our physical description of clumping is adequate. We find very small volume filling factors and that clumping starts deep in the wind, near the sonic point. The most crucial consequence of our analysis is that the mass loss rates of O stars need to be revised downward significantly, by a factor of 3 and more compared to those obtained from smooth-wind models.
I discuss observational evidence – independent of the direct spectral diagnostics of stellar winds themselves – suggesting that mass-loss rates for O stars need to be revised downward by roughly a factor of three or more, in line with recent observed mass-loss rates for clumped winds. These independent constraints include the large observed mass-loss rates in LBV eruptions, the large masses of evolved massive stars like LBVs and WNH stars, WR stars in lower metallicity environments, observed rotation rates of massive stars at different metallicity, supernovae that seem to defy expectations of high mass-loss rates in stellar evolution, and other clues. I pay particular attention to the role of feedback that would result from higher mass-loss rates, driving the star to the Eddington limit too soon, and therefore making higher rates appear highly implausible. Some of these arguments by themselves may have more than one interpretation, but together they paint a consistent picture that steady line-driven winds of O-type stars have lower mass-loss rates and are significantly clumped.
The P v λλ1118, 1128 resonance doublet is an extraordinarily useful diagnostic of O-star winds, because it bypasses the traditional problems associated with determining mass-loss rates from UV resonance lines. We discuss critically the assumptions and uncertainties involved with using P v to diagnose mass-loss rates, and conclude that the large discrepancies between massloss rates determined from P v and the rates determined from “density squared” emission processes pose a significant challenge to the “standard model” of hot-star winds. The disparate measurements can be reconciled if the winds of O-type stars are strongly clumped on small spatial scales, which in turn implies that mass-loss rates based on Hα or radio emission are too large by up to an order of magnitude.
In the old days (pre ∼1990) hot stellar winds were assumed to be smooth, which made life fairly easy and bothered no one. Then after suspicious behaviour had been revealed, e.g. stochastic temporal variability in broadband polarimetry of single hot stars, it took the emerging CCD technology developed in the preceding decades (∼1970-80’s) to reveal that these winds were far from smooth. It was mainly high-S/N, time-dependent spectroscopy of strong optical recombination emission lines in WR, and also a few OB and other stars with strong hot winds, that indicated all hot stellar winds likely to be pervaded by thousands of multiscale (compressible supersonic turbulent?) structures, whose driver is probably some kind of radiative instability. Quantitative estimates of clumping-independent mass-loss rates came from various fronts, mainly dependent directly on density (e.g. electron-scattering wings of emission lines, UV spectroscopy of weak resonance lines, and binary-star properties including orbital-period changes, electron-scattering, and X-ray fluxes from colliding winds) rather than the more common, easier-to-obtain but clumping-dependent density-squared diagnostics (e.g. free-free emission in the IR/radio and recombination lines, of which the favourite has always been Hα). Many big questions still remain, such as: What do the clumps really look like? Do clumping properties change as one recedes from the mother star? Is clumping universal? Does the relative clumping correction depend on $\dot{M}$ itself?
Mass loss is a very important aspect of the life of massive stars. After briefly reviewing its importance, we discuss the impact of the recently proposed downward revision of mass loss rates due to clumping (difficulty to form Wolf-Rayet stars and production of critically rotating stars). Although a small reduction might be allowed, large reduction factors around ten are disfavoured. We then discuss the possibility of significant mass loss at very low metallicity due to stars reaching break-up velocities and especially due to the metal enrichment of the surface of the star via rotational and convective mixing. This significant mass loss may help the first very massive stars avoid the fate of pair-creation supernova, the chemical signature of which is not observed in extremely metal poor stars. The chemical composition of the very low metallicity winds is very similar to that of the most metal poor star known to date, HE1327-2326 and offer an interesting explanation for the origin of the metals in this star. We also discuss the importance of mass loss in the context of long and soft gamma-ray bursts and pair-creation supernovae. Finally, we would like to stress that mass loss in cooler parts of the HR-diagram (luminous blue variable and yellow and red supergiant stages) are much more uncertain than in the hot part. More work needs to be done in these areas to better constrain the evolution of the most massive stars.
Wasserdampf in der Stratosphäre und Troposphäre ist eines der wichtigsten atmosphärischen Treibhausgase. Neben seiner Bedeutung für das Klima hat es großen Einfluss auf die Bildung von polaren stratosphärischen Wolken sowie auf die atmosphärische Chemie. Weltweit erstmalig soll innerhalb eines Forscherverbundes in Deutschland ein leistungsstarkes, mobiles, abtastendes Wasserdampf-DIAL zur dreidimensional hochaufgelösten Messung des atmosphärischen Wasserdampfs entwickelt werden. Mit dem Wasserdampf-DIAL können Wasserdampfkonzentrationen in der Atmosphäre mit hoher zeitlicher und räumlicher Auflösung gemessen werden. Das DIAL basiert auf einem Titan-Saphir-Laser oder einem dazu alternativen OPO-Laser (optisch parametrischer Oszillator). Der für das optische Pumpen dieser Laser nötige Pumplaser wurde im Rahmen dieser Arbeit in der Arbeitsgruppe Nichtlineare Optik des Instituts für Physik der Universität Potsdam entwickelt. Ein hochauflösendes, mobiles DIAL erfordert einen Pumplaser mit großen Pulsenergien, guter Strahlqualität und einer hohen Effizienz. Um diese Ziele zu erreichen, wurde ein MOPA-System (Master Oscillator Power Amplifier) mit Frequenzstabilisierung auf der Basis von doppelbrechungskompensierten, transversal diodengepumpten Laserstäben entwickelt und untersucht. Auf dem Weg dahin wurden unterschiedliche Realisierungsmöglichkeiten des MOPA-Systems geprüft. Im Rahmen dessen wurden die Festkörperlasermaterialien Yb:YAG [1], kerndotierte Nd:YAG-Keramik [2] und herkömmliches Nd:YAG vorgestellt und hinsichtlich ihrer Eignung für dieses MOPA-System untersucht. Nachdem die Entscheidung für Nd:YAG als laseraktives Material gefallen war, konnte darauf aufbauend die Konzeptionierung des Lasersystems auf der Basis von Verstärkungsrechnungen vorgenommen werden. Die entwickelte Verstärkungsrechnung trägt den Tatbeständen von realen Systemen Rechnung, indem radiusabhängige Intensitäten und eine radiale, nicht homogene Inversionsdichte berücksichtigt werden. Die Frequenzstabilisierung des gepulsten Oszillators (Frequenzstabilität von 1 MHz) wurde mittels des Pound-Drever-Hall-Verfahrens vorgenommen. Mit der Heterodynmethode wird die Frequenzstabilität des Oszillators gemessen. Nach Untersuchungen über verschiedene Konfigurationen für lineare und ringförmige Oszillatoren, wurde ein Ringoszillator mit zwei Laserköpfen aufgebaut, in welchen von außen mit einem Laser fester Frequenz eingestrahlt wird. Dieser emittiert bei einer Wiederholrate von 400 Hz eine Pulsenergie von Eout = 21 mJ bei nahezu beugungsbegrenzter Strahlqualität (M2 < 1,2). Die Verstärkung dieser Laserpulse erfolgte zunächst durch eine Vorverstärkerstufe und anschließend durch zwei doppelbrechungskompensierte Hauptverstärker im Doppeldurchgang. Eine gute Strahlqualität (M2 = 1,75) konnte unter anderem erzielt werden, indem der Doppeldurchgang durch die Hauptverstärker mit einem phasenkonjugierenden Spiegel (SF6), auf der Basis der stimulierten Brillouin Streuung, realisiert wurde. Der entwickelte Laser emittiert Pulse mit einer Länge von 25 ns und einer Energie von 250 mJ. Insgesamt wurde ein bisher einmaliges Lasersystem entwickelt. In der Literatur sind die erreichte Frequenzstabilität, Strahlqualität und Leistung in dieser Kombination bisher nicht dokumentiert. In der Zukunft soll durch den Einsatz von kerndotierten, keramischen Lasermaterialien, höheren Pumpleistungen der Hauptverstärker und phasenkonjugierenden Spiegeln aus Quarz die Pulsenergie des Systems weiter erhöht werden. Each organ of a multicellular organism is unique at the level of its tissues and cells. Furthermore, responses to environmental stimuli or developmental signals occur differentially at the single cell or tissue level. This underlines the necessity of precise investigation of the “building block of life” -the individual cell. Although recently large amount of data concerning different aspects of single cell performance was accumulated, our knowledge about development and differentiation of individual cell within specialized tissue are still far from being complete. To get more insight into processes that occur in certain individual cell during its development and differentiation changes in gene expression during life cycle of A. thaliana leaf hair cell (trichome) were explored in this work. After onset of trichome development this cell changes its cell cycle: it starts endoreduplication (a modified cell cycle in which DNA replication continues in the absence of mitosis and cytokinesis). This makes trichomes a suitable model for studying cell cycle regulation, regulation of cell development and differentiation. Cells of interest were sampled by puncturing them with glass microcapillaries. Each sample contained as few as ten single cells. At first time trichomes in initial stage of trichome development were investigated. To allow their sampling they were specifically labelled by green fluorescent protein (GFP). In total three cell types were explored: pavement cells, trichome initials and mature trichomes. Comparison of gene expression profiles of these cells allowed identification of the genes differentially expressed in subsequent stages of trichome development. Bioinformatic analysis of genes preferentially expressed in trichome initials showed their involvement in hormonal, metal, sulphur response and cell-cycle regulation. Expression pattern of three selected candidate genes, involved in hormonal response and early developmental processes was confirmed by independent method. Effects of mutations in these genes on both trichome and plant development as well as on plant metabolism were analysed. As an outcome of this work novel components in the sophisticated machinery of trichome development and cell cycle progression were identified. These factors could integrate hormone stimuli and network interactions between characterized and as yet unknown members of this machinery. I expect findings presented in this work to enhance and complement our current knowledge about cell cycle progression and trichome development, as well as about performance of the individual cell in general.
The interaction between neuronal cells can be identified as the computing mechanism of the brain. Neurons are complex cells that do not operate in isolation, but they are organized in a highly connected network structure. There is experimental evidence that groups of neurons dynamically synchronize their activity and process brain functions at all levels of complexity. A fundamental step to prove this hypothesis is to analyze large sets of single neurons recorded in parallel. Techniques to obtain these data are meanwhile available, but advancements are needed in the pre-processing of the large volumes of acquired data and in data analysis techniques. Major issues include extracting the signal of single neurons from the noisy recordings (referred to as spike sorting) and assessing the significance of the synchrony. This dissertation addresses these issues with two complementary strategies, both founded on the manipulation of point processes under rigorous analytical control. On the one hand I modeled the effect of spike sorting errors on correlated spike trains by corrupting them with realistic failures, and studied the corresponding impact on correlation analysis. The results show that correlations between multiple parallel spike trains are severely affected by spike sorting, especially by erroneously missing spikes. When this happens sorting strategies characterized by classifying only good'' spikes (conservative strategies) lead to less accurate results than tolerant'' strategies. On the other hand, I investigated the effectiveness of methods for assessing significance that create surrogate data by displacing spikes around their original position (referred to as dithering). I provide analytical expressions of the probability of coincidence detection after dithering. The effectiveness of spike dithering in creating surrogate data strongly depends on the dithering method and on the method of counting coincidences. Closed-form expressions and bounds are derived for the case where the dither equals the allowed coincidence interval. This work provides new insights into the methodologies of identifying synchrony in large-scale neuronal recordings, and of assessing its significance.
Atmospheric circulation and the surface mass balance in a regional climate model of Antarctica

Understanding the Earth's climate system and particularly climate variability presents one of the most difficult and urgent challenges in science. The Antarctic plays a crucial role in the global climate system, since it is the principal region of radiative energy deficit and atmospheric cooling. An assessment of regional climate model HIRHAM is presented. The simulations are generated with the HIRHAM model, which is modified for Antarctic applications. With a horizontal resolution of 55km, the model has been run for the period 1958-1998 creating long-term simulations from initial and boundary conditions provided by the European Centre for Medium-Range Weather Forecasts (ECMWF) ERA40 re-analysis. The model output is compared with observations from observation stations, upper air data, global atmospheric analyses and satellite data. In comparison with the observations, the evaluation shows that the simulations with the HIRHAM model capture both the large and regional scale circulation features with generally small bias in the modeled variables. On the annual time scale the largest errors in the model simulations are the overestimation total cloud cover and the colder near-surface temperature over the interior of the Antarctic plateau. The low-level temperature inversion as well as low-level wind jet is well captured by the model. The decadal scale processes were studied based on trend calculations. The long-term run was divided into two 20 years parts. The 2m temperature, 500 hPa temperature, MSLP, precipitation and net mass balance trends were calculated for both periods and over 1958 - 1998. During the last two decades the strong surface cooling was observed over the Eastern Antarctica, this result is in good agreement with the result of Chapman and Walsh (2005) who calculated the temperature trend based on the observational data. The MSLP trend reveals a big disparity between the first and second parts of the 40 year run. The overall trend shows the strengthening of the circumpolar vortex and continental anticyclone. The net mass balance as well as precipitation show a positive trend over the Antarctic Peninsula region, along Wilkes Land and in Dronning Maud Land. The Antarctic ice sheet grows over the Eastern part of Antarctica with small exceptions in Dronning Maud Land and Wilkes Land and sinks in the Antarctic Peninsula; this result is in good agreement with the satellite-measured altitude presented in Davis (2005) . To better understand the horizontal structure of MSLP, temperature and net mass balance trends the influence of the Southern Annual Mode (SAM) on the Antarctic climate was investigated. The main meteorological parameters during the positive and negative Antarctic Oscillation (AAO) phases were compared to each other. A positive/negative AAO index means strengthening/weakening of the circumpolar vortex, poleward/northward storm tracks and prevailing/weakening westerly winds. For detailed investigation of global teleconnection, two positive and one negative periods of AAO phase were chosen. The differences in MSLP and 2m temperature between positive and negative AAO years during the winter months partly explain the surface cooling during the last decades.
Stellar winds play an important role for the evolution of massive stars and their cosmic environment. Multiple lines of evidence, coming from spectroscopy, polarimetry, variability, stellar ejecta, and hydrodynamic modeling, suggest that stellar winds are non-stationary and inhomogeneous. This is referred to as 'wind clumping'. The urgent need to understand this phenomenon is boosted by its far-reaching implications. Most importantly, all techniques to derive empirical mass-loss rates are more or less corrupted by wind clumping. Consequently, mass-loss rates are extremely uncertain. Within their range of uncertainty, completely different scenarios for the evolution of massive stars are obtained. Settling these questions for Galactic OB, LBV and Wolf-Rayet stars is prerequisite to understanding stellar clusters and galaxies, or predicting the properties of first-generation stars. In order to develop a consistent picture and understanding of clumped stellar winds, an international workshop on 'Clumping in Hot Star Winds' was held in Potsdam, Germany, from 18. - 22. June 2007. About 60 participants, comprising almost all leading experts in the field, gathered for one week of extensive exchange and discussion. The Scientific Organizing Committee (SOC) included John Brown (Glasgow), Joseph Cassinelli (Madison), Paul Crowther (Sheffield), Alex Fullerton (Baltimore), Wolf-Rainer Hamann (Potsdam, chair), Anthony Moffat (Montreal), Stan Owocki (Newark), and Joachim Puls (Munich). These proceedings contain the invited and contributed talks presented at the workshop, and document the extensive discussions.

Electron transfer phenomena in proteins represent one of the most common types of biochemical reactions. They play a central role in energy conversion pathways in living cells, and are crucial components in respiration and photosynthesis. These complex biochemical reaction cascades consist of a series of proteins and protein complexes that couple a charge transfer to different forms of chemical energy. The efficiency and sophisticated optimisation of signal transfer in these natural redox chains has inspired engineering of artificial architectures mimicking essential properties of their natural analogues. Implementation of direct electron transfer (DET) in protein assemblies was a breakthrough in bioelectronics, providing a simple and efficient way for coupling biological recognition events to a signal transducer. DET avoids the use of redox mediators, reducing potential interferences and side reactions, as well as being more compatible with in vivo conditions. However, only a few haem proteins, including the redox protein cytochrome c (cyt.c), and blue copper enzymes show efficient DET on different kinds of electrodes. Previous investigations with cyt.c have mainly focused on heterogeneous electron transfer of monolayers of this protein on gold. An important advance was the fabrication of cyt.c multilayers by electrostatic layer-by-layer self-assembly. The ease of fabrication, the stability, and the controllable permeability of polyelectrolyte multilayers have made them particularly attractive for electroanalytical applications. With cyt.c and sulfonated polyaniline it was for the first time possible that fully electro-active multilayers of the redox protein could be prepared. This approach was extended to design an analytical signal chain based on multilayers of cyt.c and xanthine oxidase (XOD). The system does not need an external mediator but relies on an in situ generation of a mediating radical and thus allows a signal transfer from hypoxanthine via the substrate converting enzyme and cyt.c to the electrode. Another kind of a signal chain is based on assembling proteins in complexes on electrodes in such a way that a direct protein-protein electron transfer becomes feasible. This design does not need a redox mediator in analogy to natural protein communication. For this purpose, cyt.c and the enzyme bilirubin oxidase (BOD, EC 1.3.3.5) are co-immobilized in a self-assembled polyelectrolyte multilayer on gold electrodes. Although these two proteins are not natural reaction partners, the protein architecture facilitates an electron transfer from the electrode via multiple protein layers to molecular oxygen resulting in a significant catalytic reduction current. Finally, we describe a novel strategy for multi-protein layer-by-layer self-assembly combining cyt.c with an enzyme sulfite oxidase (SOx) without use of any additional polymer. Electrostatic interactions between these two proteins with rather separated pI values during the assembly process from a low ionic strength buffer were found sufficient for the layer-by-layer deposition of the both biomolecules. It is anticipated that the concepts described in this work will stimulate further progress in multilayer design of even more complex biomimetic signal cascades taking advantage of direct communication between proteins.
The objective of this thesis is to improve the knowledge of control mechanisms of hydrological induced mass movements. To this end, detailed hydrological process studies and physically-based hydrological modelling were applied. The study site is a hillslope in the Dornbirn Ache valley near Bregenz, Austria. This so called Heumös slope features a deep-seated translational shear zone and surface near creep movements of up to 10 cm a year. The Cretaceous marlstones of the Austrian Helveticum have a high susceptibility for weathering and might form clay-rich cohesive sediments. In addition, glacial and post-glacial processes formed an unstable hillslope. High yearly precipitation depths of about 2100 mm and rainstorms with both high intensities and precipitation depths govern surface and subsurface hydrological processes. Pressure propagation induced in hydrological active areas influences laterally the groundwater reactions of the moving mass. A complex three-dimensional subsurface pressure system is the cause for fast groundwater reactions despite low hydraulic conductivities. To understand hillslope scale variability, hydrotopes representing specific dominating processes were mapped using vegetation association distribution and soil core analysis. Detailed small-scale soil investigations followed to refine the understanding of these hydrotopes. A perceptional model was developed from the hydrotope distribution and was corroborated by these detailed investigations. The moving hillslope is dominated by surface-runoff generation. Infiltration and deep percolation of water is inhibited through clay-rich gleysols; the yearly average soil moisture is close to saturation. Steep slopes adjacent to the moving hillslope are far more active concerning infiltration, preferential flow and groundwater fluctuations. Spring discharge observations at the toe of the steep slopes are in close relation to groundwater table observations on the moving hillslope body. Evidence of pressure propagation from the steep slopes towards the hillslope body is gathered by comparison of dominating structures and processes. The application of the physically-based hydrological model CATFLOW substantiates the idea of pressure propagation as a key process for groundwater reactions and as a possible trigger for movement in the hillslope.
Nature has always served as a model for mimicking and inspiration to humans in their efforts to improve their life. Researchers have been inspired by nature to produce biomimetic materials with molecular recognition properties by design rather than evolution. Molecular imprinting is one way to prepare such materials. Such smart materials with new functionalities are at the forefront of the development of a relevant number of ongoing and perspective applications ranging from consumer to space industry. Molecularly imprinted polymers were developed by mimicking the natural enzymes or antibodies that serve as host for binding target molecules. These imprints were used as a recognition element to substitute natural biomolecules in biosensors. The concept behind molecular imprinting is to mold a material (with the desired chemical properties) around individual molecules. Upon removal of the molecular templates, one is left with regions in the molded material that fit the shape of the template molecules. Thus, molecular imprinting results in materials that can selectively bind to molecules of interest. Imprinted materials resulted in applications ranging from chemical separation to bioanalytics. In this work attempts were made particularly in the development of molecularly imprinted polymer based thermometric sensors. The main effort was focused towards the development of an covalently imprinted polymer that would be able to selectively bind fructosyl valine (Fru-Val), the N-terminal constituent of hemoglobin A1c ß-chains. Taking into account the known advantages of imprinted polymers, e.g. robustness, thermal and chemical stability, imprinted materials were successfully used as a recognition element in the sensor. One of the serious problems associated with the development of MIP sensors and which lies in the absence of a generic procedure for the transformation of the polymer-template binding event into a detectable signal has been addressed by developing the "thermometric" approach. In general the developed approach gives a new insight on MIP/Analyte interactions.
Der Kampf um die parteipolitische Macht in der Russländischen Föderation : die KPRF 1991 - 1996

Wahlen sind wichtige Bestandteile demokratischer Strukturen und werfen schon im Vorfeld ihrer Durchführung ihre Schatten voraus. Die Russländische Föderation (RF) stellt hierbei keine Ausnahme dar, aber können in ihrem Falle nur selten westeuropäische Maßstäbe angewandt werden. Russischen Uhren ticken anders. Für die RF gelten keine Schablonen, keine allgemein übertragbaren Parameter. Fragen der Sozialisierung, Mentalitätsfragen und Fragen nach der politischen Kultur spielen hierbei in der Realität eine entscheidende Rolle. Es ist wichtig, die Akteure im System selbst zu untersuchen, nicht aber, wie es in der Literatur stattfindet, mit dem Blick auf die Wirkung der Einzelakteure auf das Gesamtsystem, sondern des Gesamtsystems auf einen Einzelakteur. Hierbei analysiert die Arbeit die spezifischen Eigenarten des russländischen politischen Systems, verdeutlicht die Entwicklung des politischen Lebens in der RF, besonders der politischen Vereinigungen und streicht die rechtliche Stellung sowie die politischen Lager heraus. Im vereinigungspolitischen Spektrum der RF haben sich nur wenige Konstanten herauskristallisieren können. Einer dieser Fixpunkte ist in der Kommunistischen Partei der Russländischen Föderation (KPRF) zu sehen. Die Vereinigung hatte es geschafft, wie ein Phönix aus der Asche aufzusteigen. Sich nicht nur über die Zeit des Putsches 1993 zu retten, sondern in der Folgezeit zur einzigen Vereinigung zu erwachsen, der es möglich war, sich gegen die rechtspopulistische Liberal-Demokratische Partei Russlands (LDPR), die Parteien der Macht´, aber auch gegen JABLOKO und die Jelzin-Administration zu behaupten. Keine Vereinigung dieser Zeit schaffte es, so viele Mitglieder und Wähler an sich zu binden, wie es die kommunistische Partei unter G. A. Sjuganow vermochte. Letztendlich ist es der KPRF gelungen, mit G. A. Sjuganow den aussichtsreichsten Gegenkandidaten zu B. N. Jelzin aufzubauen. Die Präsidentschaftswahl von 1996 ist, wie keine nach ihr, denkbar knapp ausgefallen und erst in der Stichwahl entschieden worden. Gleichsam stellt das Jahr 1996 für die Vereinigung eine tief greifende Zäsur dar. Für die Erfolge und Niederlagen der KPRF ist es von Vorteil, die sie beeinflussenden Faktoren in exogene, der Makroebene politische System der RF´ und in endogene, der Mikroebene KPRF´ entstammende Faktoren zu unterteilen. Auf die exogenen Faktoren, wie beispielsweise die exponierte Stellung des Präsidenten, eine fehlende Parteiengesetzgebung, die marginale Stellung der Staatsduma, die Politikverdrossenheit und Sozialisierung der Wahlbevölkerung oder die geografischen Dimensionen der Föderation, konnte die Sjuganow-Vereinigung nur geringfügig Einfluss nehmen. Die endogenen Faktoren, wie das politische Programm, das Statut oder eine fehlende innerparteiliche Homogenität, lagen in den Händen der Vereinigung und konnten von ihr aktiv, aus ihr heraus gestaltet werden. Für den angestrebten Sieg zur Präsidentschaftswahl 1996 oblag es der KPRF, geeignete Strategien zu finden. Als für das Ziel positiv kann der Schwenk von der unversöhnlichen zur konstruktiven Opposition, eine breite Koalitionsbereitschaft, das Nutzen dichter Netzwerke und ein breites Angebot programmatischer Schwerpunkte, gekoppelt an den Passus Partei leninistischen Typs´ gewertet werden. Dass G. A. Sjuganow in der Vereinigung umstritten war, der Vorwurf des fehlenden Charismas im Raum stand und es die Vereinigung nicht vermochte, ausreichend Mobilisierungspotential gegenüber Wählern außerhalb der KPRF-Stammwählerschaft zu erreichen, sind als dem Ziel abträglich zu betrachten. Ferner verfolgte die KPRF einen grauen, klassischen, aber z. T. unprofessionellen Wahlkampf, der der Kremladministration nicht gefährlich werden konnte und dem modernen, stark medialen Wahlkampf B. N. Jelzins nichts entgegenzusetzen hatte. Eine einseitige Ausrichtung auf russisch-nationale Themen kann ebenfalls als strategischer Fehler der KPRF gesehen werden, zeigten doch Umfragen, dass dem Großteil der Wahlbevölkerung mehr an der Verbesserung der sozial-ökonomischen Lage gelegen war, als an nationaler Rhetorik. Eine von exogener Seite geschürte Kommunismusangst und das Fehlen eines positiven Alternativprogramms von endogener Seite runden das Scheitern der KPRF zur Präsidentschaftswahl 1996 ab. Die Arbeit zeigt, dass die exogenen, wie auch endogenen Faktoren sowohl hemmend, als auch fördernd auf die KPRF einwirken konnten. Woran die KPRF beim Erreichen des Wahlsieges von 1996 letztendlich scheiterte, kann aus keiner der beiden Seiten eindeutig abgeleitet werden. Sowohl die endogenen, als auch die exogenen Faktoren leisteten ihren Betrag zum Scheitern der Vereinigung, doch muss festgehalten werden, dass die Einflussnahme der KPRF auf die hemmenden Faktoren im exogenen Bereich hätte stärker ausfallen können.
Functional analysis of selected DOF transcription factors in the model plant Arabidopsis thaliana

Transcription factors (TFs) are global regulators of gene expression playing essential roles in almost all biological processes, and are therefore of great scientific and biotechnological interest. This project focused on functional characterisation of three DNA-binding-with-one-zinc-finger (DOF) TFs from the genetic model plant Arabidopsis thaliana, namely OBP1, OBP2 and AtDOF4;2. These genes were selected due to severe growth phenotypes conferred upon their constitutive over-expression. To identify biological processes regulated by OBP1, OBP2 and AtDOF4;2 in detail molecular and physiological characterization of transgenic plants with modified levels of OBP1, OBP2 and AtDOF4;2 expression (constitutive and inducible over-expression, RNAi) was performed using both targeted and profiling technologies. Additionally expression patterns of studied TFs and their target genes were analyzed using promoter-GUS lines and publicly available microarray data. Finally selected target genes were confirmed by chromatin immuno-precipitation and electrophoretic-mobility shift assays. This combinatorial approach revealed distinct biological functions of OBP1, OBP2 and AtDOF4;2. Specifically OBP2 controls indole glucosinolate / auxin homeostasis by directly regulating the enzyme at the branch of these pathways; CYP83B1 (Skirycz et al., 2006). Glucosinolates are secondary compounds important for defence against herbivores and pathogens in the plants order Caparales (e.g. Arabidopsis, canola and broccoli) whilst auxin is an essential plant hormone. Hence OBP2 is important for both response to biotic stress and plant growth. Similarly to OBP2 also AtDOF4;2 is involved in the regulation of plant secondary metabolism and affects production of various phenylpropanoid compounds in a tissue and environmental specific manner. It was found that under certain stress conditions AtDOF4;2 negatively regulates flavonoid biosynthetic genes whilst in certain tissues it activates hydroxycinnamic acid production. It was hypothesized that this dual function is most likely related to specific interactions with other proteins; perhaps other TFs (Skirycz et al., 2007). Finally OBP1 regulates both cell proliferation and cell expansion. It was shown that OBP1 controls cell cycle activity by directly targeting the expression of core cell cycle genes (CYCD3;3 and KRP7), other TFs and components of the replication machinery. Evidence for OBP1 mediated activation of cell cycle during embryogenesis and germination will be presented. Additionally and independently on its effects on cell proliferation OBP1 negatively affects cell expansion via reduced expression of cell wall loosening enzymes. Summing up this work provides an important input into our knowledge on DOF TFs function. Future work will concentrate on establishing exact regulatory networks of OBP1, OBP2 and AtDOF4;2 and their possible biotechnological applications.
Giacconi et al. (1962) discovered a diffuse cosmic X-ray background with rocket experiments when they searched for lunar X-ray emission. Later satellite missions found a spectral peak in the cosmic X-ray background at ~30 keV. Imaging X-ray satellites such as ROSAT (1990-1999) were able to resolve up to 80% of the background below 2 keV into single point sources, mainly active galaxies. The cosmic X-ray background is the integration of all accreting super-massive (several million solar masses) black holes in the centre of active galaxies over cosmic time. Synthesis models need further populations of X-ray absorbed active galaxy nuclei (AGN) in order to explain the cosmic X-ray background peak at ~30 keV. Current X-ray missions such as XMM-Newton and Chandra offer the possibility of studying these additional populations. This Ph.D. thesis studies the populations that dominate the X-ray sky. For this purpose the 120 ksec XMM-Newton Marano field survey, named for an earlier optical quasar survey in the southern hemisphere, is analysed. Based on the optical follow-up observations the X-ray sources are spectroscopically classified. Optical and X-ray properties of the different X-ray source populations are studied and differences are derived. The amount of absorption in the X-ray spectra of type II AGN, which are considered as a main contributor to the X-ray background at ~30 keV, is determined. In order to extend the sample size of the rare type II AGN, this study also includes objects from another survey, the XMM-Newton Serendipitous Medium Sample. In addition, the dependence of the absorption in type II AGN with redshift and X-ray luminosity is analysed. We detected 328 X-ray sources in the Marano field. 140 sources were spectroscopically classified. We found 89 type I AGN, 36 type II AGN, 6 galaxies, and 9 stars. AGN, galaxies, and stars are clearly distinguishable by their optical and X-ray properties. Type I and II AGN do not separate clearly. They have a significant overlap in all studied properties. In a few cases the X-ray properties are in contradiction to the observed optical properties for type I and type II AGN. For example we find type II AGN that show evidence for optical absorption but are not absorbed in X-rays. Based on the additional use of near infra-red imaging (K-band), we were able to identify several of the rare type II AGN. The X-ray spectra of type II AGN from the XMM-Newton Marano field survey and the XMM-Newton Serendipitous Medium Sample were analysed. Since most of the sources have only ~40 X-ray counts in the XMM-Newton PN-detector, I carefully studied the fit results of simulated X-ray spectra as a function of fit statistic and binning method. The objects revealed only moderate absorption. In particular, I do not find any Compton-thick sources (absorbed by column densities of NH > 1.5 x 10^24 cm^−2). This gives evidence that type II AGN are not the main contributor of the X-ray background around 30 keV. Although bias effects may occur, type II AGN show no noticeable trend of the amount of absorption with redshift or X-ray luminosity.
Abenteuer Lesesprache : zum Aufbau interkomprehensiver Lesekompetenz in den romanischen Sprachen

Plurilinguismus oder „English only“? Als politische Institution, in der es nur sprachliche und kulturelle Minderheiten gibt und geben wird, folgt die Europäische Union einer plurilinguistischen Orientierung, womit ein erheblicher Bedarf an Sprach- und Kulturkenntnissen (und deren Erwerb) verbunden ist. Grosso modo umfasst die EU drei große indoeuropäische Sprachfamilien: die romanische, slawische und germanische. Innerhalb jeder dieser Sprachgruppen gibt es etymologisch bedingte Gemeinsamkeiten, die v.a. die Phonologie, Morphologie, Lexik und Syntax betreffen. Die Kenntnis dieser synchron erkennbaren gemeinsamen Elemente bzw. ihrer einzelsprachlich äquivalenten Varianten ermöglicht transferhaftes Lernen im Sinne der Interkomprehension in Sprachfamilien. Grundlage der romanischen Interkomprehension ist das „Vulgärlatein“, dessen Spezifik v.a. gegenüber den heutigen Sprachen der Romania herausgestellt wird. Den lerntheoretischen Hintergrund der Interkomprehension bilden die Interlanguage-Hypothese, die Annahme mentaler Netzwerke sowie die verschiedenen Verarbeitungsstadien des interkomprehensiven Spracherwerbs: Spontangrammatik, Mehrsprachenspeicher und didaktischer Monitor. Zudem wird die Rolle mutter- und fremdsprachlicher Transferbasen sowie die der einzelnen Transferdomänen (Form, Inhalt, Funktion, Pragmatik, Didaktik) erläutert. Schwerpunkte der vorliegenden Arbeit bilden die Motivation und die einzelnen Verarbeitungsstadien beim interkomprehensiven Lesen, wobei dem sprachlichen Vorwissen der Lesenden besondere Bedeutung zukommt. Der vorgeschlagene Leitfaden für das erschließende Lesen umfasst drei Gesamtlektüren und berücksichtigt dabei sowohl sprachsystematische als auch textlinguistische Kriterien. Zur Illustration dient ein aktueller Text in Nissart, der auch verschiedenen Probanden vorgelegt wurde. Die Analyse ergab, dass ein globales Textverständnis durch die Kenntnis einer romanischen Sprache spontan gegeben ist, dieses Globalverständnis im Einzelnen jedoch deutlich differieren kann. Am Ende der Arbeit werden mit der Darstellung der strukturellen Besonderheiten des Rumänischen die Grenzen interkomprehensiven Leseverständnisses gezeigt, aber auch der erleichterte Einstieg in die Sprachproduktion dargelegt.
Diese Arbeit beschäftigt sich mit der Rolle von Konkordanzmarkierungen innerhalb einer Phrase für die Segmentierung eines fremden Sprachstroms. Das Merkmal der Konkordanz tritt auf, wenn alle Bestandteile einer Phrase gleichermaßen durch eine identische Markierung gekennzeichnet sind (z. B. los muchachos ricosSpanisch = die reichen Männer). Da diese wiederkehrenden Markierungen zumeist aus Affixen bestehen, kann Konkordanz als ein Sonderfall der Flexionsmorphologie betrachtet werden. Es wurde untersucht, ob die formale Korrespondenz zwischen den Bestandteilen konkordanter Phrasen als Hinweis auf die Grenzen der linguistisch relevanten Einheit Phrase im Spracherwerb fungieren kann. Zusätzlich wird auf das Zusammenspiel einzelner Hinweisreize untereinander eingegangen. Mit Kindern im Alter von zehn Monaten wurden vier Experimente mit dem Headturn Preference Paradigma (Jusczyk & Aslin, 1995) durchgeführt. Es wurde zunächst bei deutschen und englischen Kleinkindern untersucht, ob sie sensibel für gleich bleibende Suffixe innerhalb einer Phrase sind und diese für die Segmentierung nutzen können. Außerdem wurde das Zusammenspiel der Hinweise Konkordanz und Prosodie bei der Auffindung von Phrasengrenzen betrachtet. Es zeigte sich, dass deutsche Kinder in besonderer Weise auf konkordante Markierungen reagieren. Neben einer Sensitivität für Konkordanzmarkierungen zeigte das Ergebnis der deutschen Kinder auch, dass sie Flexionssuffixe im Deutschen bereits im Sprachstrom bemerken können. Ein solches Ergebnismuster ließ sich bei den Englisch lernenden Kindern nicht beobachten. Verschiedene Erklärungsmöglichkeiten für diesen Unterschied werden erläutert. Insgesamt weisen die Daten aus den Kindersprachexperimenten darauf hin, dass bereits im Alter von zehn Monaten bei Kindern eine Sensibilität für wiederholt in ähnlicher / gleicher Form auftretende sprachliche Elemente innerhalb der Domäne der Phrase vorhanden ist. Außerdem lassen die Resultate darauf schließen, dass Konkordanzmarkierungen bereits früh zur Segmentierung von kontinuierlicher Sprache verwendet werden. Diese Leistung steht in Zusammenhang mit der Beachtung von statistischen Regularitäten im Sprachstrom. Untersuchungen dazu zeigen, dass m. H. statistischer Lernmechanismen wiederkehrende Elemente im Sprachstrom erkannt werden können (Bonatti, Peña, Nespor, & Mehler, 2005; Newport & Aslin, 2004; Saffran, 2001; Saffran, Aslin & Newport, 1996). Anscheinend ist das Auftreten identischer Segmente innerhalb einer relativ kleinen Domäne im Sprachstrom für Lerner ein hervorstechendes Merkmal, das dazu beiträgt, diese Domäne aus dem Signal hervorzuheben und somit die Segmentierung des Sprachstroms in kleinere Anteile zu unterstützen. Neben den Untersuchungen mit den Kleinkindern wurden zusätzlich drei Reaktionszeitexperimente mit deutschen und englischen Erwachsenen zur Rolle von Konkordanzmarkierungen bei der Verarbeitung der Fremdsprachen Spanisch, Suaheli und (für die englischen Probanden) Deutsch durchgeführt. Das erste Experiment befasste sich mit der Stimulussprache Spanisch, in der es bei Konkordanz zum mehrfachen Auftreten von identischen Suffixen mit Vollvokalen kommt. Dabei war zu beobachten, dass deutsche und englische Muttersprachler die zu erinnernden Phrasen besser in einem kontinuierlichen spanischen Sprachstrom wieder erkannten, wenn die kritischen Phrasen konkordant waren, als wenn sie nicht konkordant waren. Das zweite Experiment verwendete die Stimulussprache Suaheli (konkordante vs. nicht konkordante Präfixe). Dabei zeigte sich ein solches Muster ausschließlich bei den englischen Muttersprachlern. Das dritte Experiment untersuchte englische Muttersprachler mit deutschem Stimulusmaterial, wobei Konkordanz durch Suffixe markiert wird, die aus einer Schwa-Silbe bestehen. Hier ergab sich kein Hinweis für eine Nutzung konkordanter Markierungen bei der Erkennung von Phrasen. Als Grund dafür wird die reduzierte Vokalqualität angenommen, die Schwa-Silben u.U. schwerer wahrnehmbar macht als Vollvokalsilben (z.B. Widera & Portele, 1999; Goméz Lacabex, García Lecumberri, & Cooke, 2005). Es werden weitere Erklärungshypothesen bzgl. der Ergebnisunterschiede bei deutschen und englischen Muttersprachlern beschrieben, die auch auf den Unterschied zwischen der Verarbeitung von konkordanten Suffixen vs. Präfixen eingehen. Zusätzlich erfolgt eine Diskussion der Ergebnisse vor dem Hintergrund von Annahmen über Arten von (nicht-)sprachlichen Ähnlichkeiten und ihren Einfluss auf die Wahrnehmung von ähnlichen Elementen. Die vorliegenden Daten stützen die Annahme von Morgan (1986), dass der Input für einen Sprachlerner bereits zahlreiche Hinweise über die Struktur der jeweiligen Sprache enthält. Sowohl Kleinkinder als auch erwachsene Sprachlerner scheinen für einen beachtlichen Teil dieser Hinweisreize sensibel zu sein. Die bislang kaum beachteten konkordante Markierungen innerhalb von Phrasen scheinen zumindest einen Teil dieser Hinweisreize auszumachen.
In dieser Arbeit wurde die Variabilität der Atmosphäre in einem neuen gekoppelten Klimamodell (ECHO-GiSP) untersucht, welches eine vereinfachte Stratosphärenchemie (bis 80 km Höhe) enthält. Es wurden 2 Simulationen über 150 Jahre durchgeführt. In einer der Simulationen wurde die atmosphärische Chemie modelliert, hatte aber keinen Einfluß auf die Dynamik des Klimamodelles. In der zweiten Simulation wurde hingegen die Wirkung der Chemie auf die Klimadynamik explizit berücksichtigt, die über die Strahlungsbilanz des Modelles erfolgt. Dies ist die erste Langzeitsimulation mit einem voll gekoppelten globalen Klimamodell mit interaktiver Chemie. Die Simulation mit rückgekoppelter Chemie zeigt eine Abschwächung des atmosphärischen Variabilitätsmusters der Arktischen Oszillation (AO). Zudem kommt es in der Troposphäre zu einer Reduzierung der mittleren Windgeschwindigkeiten der gemäßigten Breiten aufgrund verringerter Temperaturgegensätze zwischen den Tropen und den Polargebieten. Auch in der Stratosphäre ergibt sich eine Abschwächung und Erwärmung des Polarwirbels. Diese Auswirkungen der Kopplung zwischen der atmosphärischen Chemie und der Dynamik des Klimamodelles sind eine wichtige Erkenntnis, da in früheren Klimasimulationen die Variabilität der AO oft zu stark ausgeprägt war. In der Stratosphäre reduziert sich infolge des abgeschwächten Polarwirbels auch die großräumige Zirkulation zwischen den beiden Hemisphären der Erde. Zudem kommt es in den Tropen zu Temperaturänderungen durch stratosphärische Ozonschwankungen in Abhängigkeit von der AO. Allgemein verändert sich die Kopplung zwischen Troposphäre und Stratosphäre, einschließlich des durch die Anregung von langen atmosphärischen Wellen erfolgenden vertikalen Energieübertrages aus der Troposphäre in die Stratosphäre.
"Considerons une particule mobile se mouvant aleatoirement sur la droite (ou sur un segment de droite). Supposons qu'il existe une probabilite F(x,y;s,t) bien definie pour que la particule se trouvant a l'instant s dans la position x se trouve a l'instant t (> s) a gauche de y, probabilite independante du mouvement anterieur de la particule...." Mit diesen Worten beginnt eines der berühmtesten mathematischen Manuskripte des letzten Jahrhunderts. Es stammt vom Soldaten Wolfgang Döblin, Sohn des deutschen Schriftstellers Alfred Döblin, und trägt den Titel "Sur l'equation de Kolmogoroff". Seine Veröffentlichung verbindet sich mit einer unglaublichen Geschichte. Wolfgang Döblin, stationiert mit seiner Einheit in den Ardennen im Winter 1939/1940, arbeitete an diesem Manuskript. Er entschloss sich, es als versiegeltes Manuskript an die Academie des Sciences in Paris zu schicken. Aber er kehrte nie aus diesem Krieg zurück. Sein Manuskript blieb 60 Jahre unter Verschluss im Archiv, und wurde erst im Jahre 2000 geöffnet. Wie weit Döblin damit seiner Zeit voraus war, wurde erkannt, nachdem es von Bernard Bru und Marc Yor ausgewertet worden war. Im ersten Satz umschreibt W. Döblin gleichzeitig das Programm des Manuskripts: "Wir betrachten ein bewegliches Teilchen, das sich zufällig auf der Geraden (oder einem Teil davon) bewegt." Er widmet sich damit der Aufgabe, die Fundamente eines Gebiets zu legen, das wir heute als stochastische Analysis bezeichnen.
Die vorliegende Arbeit befasst sich mit der wissensbasierten Modellierung von Audio-Signal-Klassifikatoren (ASK) für die Bioakustik. Sie behandelt ein interdisziplinäres Problem, das viele Facetten umfasst. Zu diesen gehören artspezifische bioakustische Fragen, mathematisch-algorithmische Details und Probleme der Repräsentation von Expertenwissen. Es wird eine universelle praktisch anwendbare Methode zur wissensbasierten Modellierung bioakustischer ASK dargestellt und evaluiert. Das Problem der Modellierung von ASK wird dabei durchgängig aus KDD-Perspektive (Knowledge Discovery in Databases) betrachtet. Der grundlegende Ansatz besteht darin, mit Hilfe von modifizierten KDD-Methoden und Data-Mining-Verfahren die Modellierung von ASK wesentlich zu erleichtern. Das etablierte KDD-Paradigma wird mit Hilfe eines detaillierten formalen Modells auf den Bereich der Modellierung von ASK übertragen. Neunzehn elementare KDD-Verfahren bilden die Grundlage eines umfassenden Systems zur wissensbasierten Modellierung von ASK. Methode und Algorithmen werden evaluiert, indem eine sehr umfangreiche Sammlung akustischer Signale des Großen Tümmlers mit ihrer Hilfe untersucht wird. Die Sammlung wurde speziell für diese Arbeit in Eilat (Israel) angefertigt. Insgesamt werden auf Grundlage dieses Audiomaterials vier empirische Einzelstudien durchgeführt: - Auf der Basis von oszillographischen und spektrographischen Darstellungen wird ein phänomenologisches Klassifikationssystem für die vielfältigen Laute des Großen Tümmlers dargestellt. - Mit Hilfe eines Korpus halbsynthetischer Audiodaten werden verschiedene grundlegende Verfahren zur Modellierung und Anwendung von ASK in Hinblick auf ihre Genauigkeit und Robustheit untersucht. - Mit einem speziell entwickelten Clustering-Verfahren werden mehrere Tausend natürliche Pfifflaute des Großen Tümmlers untersucht. Die Ergebnisse werden visualisiert und diskutiert. - Durch maschinelles mustererkennungsbasiertes akustisches Monitoring wird die Emissionsdynamik verschiedener Lauttypen im Verlaufe von vier Wochen untersucht. Etwa 2.5 Millionen Klicklaute werden im Anschluss auf ihre spektralen Charakteristika hin untersucht. Die beschriebene Methode und die dargestellten Algorithmen sind in vielfältiger Hinsicht erweiterbar, ohne dass an ihrer grundlegenden Architektur etwas geändert werden muss. Sie lassen sich leicht in dem gesamten Gebiet der Bioakustik einsetzen. Hiermit besitzen sie auch für angrenzende Disziplinen ein hohes Potential, denn exaktes Wissen über die akustischen Kommunikations- und Sonarsysteme der Tiere wird in der theoretischen Biologie, in den Kognitionswissenschaften, aber auch im praktischen Naturschutz, in Zukunft eine wichtige Rolle spielen.
In semi-arid savannah ecosystems, the vegetation structure and composition, i.e. the architecture of trees, shrubs, grass tussocks and herbaceous plants, offer a great variety of habitats and niches to sustain animal diversity. In the last decades intensive human land use practises like livestock farming have altered the vegetation in savannah ecosystems worldwide. Extensive grazing leads to a reduction of the perennial and herbaceous vegetation cover, which results in an increased availability of bare soil. Both, the missing competition with perennial grasses and the increase of bare soils favour shrub on open ground and lead to area-wide shrub encroachment. As a consequence of the altered vegetation structure and composition, the structural diversity declines. It has been shown that with decreasing structural diversity animal diversity decline across a variety of taxa. Knowledge on the effects of overgrazing on reptiles, which are an important part of the ecosystem, are missing. Furthermore, the impact of habitat degradation on factors of a species population dynamic and life history, e.g., birth rate, survival rate, predation risk, space requirements or behavioural adaptations are poorly known. Therefore, I investigated the impact of overgrazing on the reptile community in the southern Kalahari. Secondly I analysed population dynamics and the behaviour of the Spotted Sand Lizard, Pedioplanis l. lineoocellata. All four chapters clearly demonstrate that habitat degradation caused by overgrazing had a severe negative impact upon (i) the reptile community as a whole and (ii) on population parameters of Pedioplanis l. lineoocellata. Chapter one showed a significant decline of regional reptile diversity and abundance in degraded habitats. In chapter two I demonstrated that P. lineoocellata moves more frequently, spends more time moving and covers larger distances in degraded than in non-degraded habitats. In addition, home range size of the lizard species increases in degraded habitats as shown by chapter three. Finally, chapter four showed the negative impacts of overgrazing on several population parameters of P. lineoocellata. Absolute population size of adult and juvenile lizards, survival rate and birth rate are significantly lower in degraded habitats. Furthermore, the predation risk was greatly increased in degraded habitats. A combination of a variety of aspects can explain the negative impact of habitat degradation on reptiles. First, reduced prey availability negatively affects survival rate, the birth rate and overall abundance. Second, the loss of perennial plant cover leads to a loss of niches and to a reduction of opportunities to thermoregulate. Furthermore, a loss of cover and is associated with increased predation risk. A major finding of my thesis is that the lizard P. lineoocellata can alter its foraging strategy. Species that are able to adapt and change behaviour, such as P. lineoocellata can effectively buffer against changes in their environment. Furthermore, perennial grass cover can be seen as a crucial ecological component of the vegetation in the semi-arid savannah system of the southern Kalahari. If perennial grass cover is reduced to a certain degree reptile diversity will decline and most other aspects of reptile life history will be negatively influenced. Savannah systems are characterised by a mixture of trees, shrubs and perennial grasses. These three vegetation components determine the composition and structure of the vegetation and accordingly influence the faunal diversity. Trees are viewed as keystone structures and focal points of animal activity for a variety of species. Trees supply animals with shelter, shade and food and act as safe sites, nesting sites, observation posts and foraging sites. Recent research demonstrates a positive influence of shrub patches on animal diversity. Moreover, it would seem that intermediate shrub cover can also sustain viable populations in savannah landscapes as has been demonstrated for small carnivores and rodent species. The influence of perennial grasses on faunal diversity did not receive the same attention as the influence of trees and shrubs. In my thesis I didn’t explicitly measure the direct effects of perennial grasses but my results strongly imply that it has an important role. If the perennial grass cover is significantly depleted my results suggest it will negatively influence reptile diversity and abundance and on several populations parameters of P. lineoocellata. Perennial grass cover is associated with the highest prey abundance, reptile diversity and reptile abundance. It provides reptiles both a refuge from predators and opportunities to optimise thermoregulation. The relevance of each of the three vegetation structural elements is different for each taxa and species. In conclusion, I can all three major vegetation structures in the savannah system are important for faunal diversity.
Die sozialen Sicherungs- und Fürsorgesysteme in Deutschland befinden sich in einem radikalen Umbau im Sinne eines neoliberalen Gesellschaftsmodells mit der Folge, dass sich die gesellschaftlichen Widersprüche und Konflikte zuspitzen. Gleichzeitig weisen die aktuellen Debatten in der Sozialen Arbeit darauf hin, dass die gesellschaftlichen, und damit strukturellen, Widersprüche in deren Mitte angekommen sind. Sie manifestieren sich u.a. in Handlungsdilemmata, mit denen die Sozialarbeiter/innen in ihrem Berufsalltag und in ihrer pädagogischen Praxis konfrontiert sind und auf die sie reagieren (müssen). Hier liegt auch der Ausgangspunkt der vorliegenden Arbeit. Es wird der Frage nachgegangen, welche Strategien die in der Jugendberufshilfe tätigen Sozialarbeiter/innen bei der Konfrontation mit sich widersprechenden Handlungsanforderungen entwickeln, die sich aus den institutionellen und gesellschaftlichen Rahmenbedingungen einerseits und der sozialpädagogischen Berufspraxis andererseits ergeben. Von besonderem Interesse ist die Frage, welche Probleme und Handlungsdilemmata überhaupt von den Sozialarbeiter/innen wahrgenommen werden. Der Fokus dieser Arbeit liegt auf der Jugendsozialarbeit, und insbesondere auf der Jugendberufshilfe, als demjenigen Teil des sozialen Sicherungssystems, der sich speziell an individuell beeinträchtigte und sozial benachteiligte Jugendliche richtet und an ihrer Übergangsproblematik ansetzt. Dem qualitativen Forschungsansatz der grounded theory folgend werden zwei Fallanalysen von Sozialarbeiterinnen durchgeführt, die als Beraterinnen am Übergang Schule – Beruf tätig sind. Die Ergebnisse weisen darauf hin, dass die Umgangsformen mit Handlungsdilemmata in einem engen individuellen, institutionellen sowie gesellschaftlichen Kontext stehen, da sie stets gleichsam eine individuelle, institutionelle und gesellschaftliche Funktion erfüllen.
Writing an alternative Australia : women and national discourse in nineteenth-century literature

In this thesis, I want to outline the emergence of the Australian national identity in colonial Australia. National identity is not a politically determined construct but culturally produced through discourse on literary works by female and male writers. The emergence of the dominant bushman myth exhibited enormous strength and influence on subsequent generations and infused the notion of “Australianness” with exclusively male characteristics. It provided a unique geographical space, the bush, on and against which the colonial subject could model his identity. Its dominance rendered non-male and non-bush experiences of Australia as “un-Australian.” I will present a variety of contemporary voices – postcolonial, Aboriginal, feminist, cultural critics – which see the Australian identity as a prominent topic, not only in the academia but also in everyday culture and politics. Although positioned in different disciplines and influenced by varying histories, these voices share a similar view on Australian society: Australia is a plural society, it is home to millions of different people – women, men, and children, Aboriginal Australians and immigrants, newly arrived and descendents of the first settlers – with millions of different identities which make up one nation. One version of national identity does not account for the multitude of experiences; one version, if applied strictly, renders some voices unheard and oppressed. After exemplifying how the literature of the 1890s and its subsequent criticism constructed the itinerant worker as “the” Australian, literary productions by women will be singled out to counteract the dominant version by presenting different opinions on the state of colonial Australia. The writers Louisa Lawson, Barbara Baynton, and Tasma are discussed with regard to their assessment of their mother country. These women did not only present a different picture, they were also gifted writers and lived the ideal of the “New Women:” they obtained divorces, remarried, were politically active, worked for their living and led independent lives. They paved the way for many Australian women to come. In their literary works they allowed for a dual approach to the bush and the Australian nation. Louisa Lawson credited the bushwoman with heroic traits and described the bush as both cruel and full of opportunities not known to women in England. She understood women’s position in Australian society as oppressed and tried to change politics and culture through the writings in her feminist magazine the Dawn and her courageous campaign for women suffrage. Barbara Baynton painted a gloomy picture of the Australian bush and its inhabitants and offered one of the fiercest critiques of bush society. Although the woman is presented as the able and resourceful bushperson, she does not manage to survive in an environment which functions on male rules and only values the economic potential of the individual. Finally, Tasma does not present as outright a critique as Barbara Baynton, however, she also attests the colonies a fascination with wealth which she renders questionable. She offers an informed judgement on colonial developments in the urban surrounds of the city of Melbourne through the comparison of colonial society with the mother country England. These three women, as writers, commentators, and political activists, faced exclusion from the dominant literary discourses. Their assessment of colonial society remained unheard for a long time. Now, after much academic excavation, these voices speak to us from the past and remind us that people are diverse, thus nation is diverse. Dominant power structures, the institutions and individuals who decide who can contribute to the discourse on nation, have to be questioned and reassessed, for they mute voices which contribute to a wider, to the “full”, and maybe “real” picture of society.
In this work, some new results to exploit the recurrence properties of quasiperiodic dynamical systems are presented by means of a two dimensional visualization technique, Recurrence Plots(RPs). Quasiperiodicity is the simplest form of dynamics exhibiting nontrivial recurrences, which are common in many nonlinear systems. The concept of recurrence was introduced to study the restricted three body problem and it is very useful for the characterization of nonlinear systems. I have analyzed in detail the recurrence patterns of systems with quasiperiodic dynamics both analytically and numerically. Based on a theoretical analysis, I have proposed a new procedure to distinguish quasiperiodic dynamics from chaos. This algorithm is particular useful in the analysis of short time series. Furthermore, this approach demonstrates to be efficient in recognizing regular and chaotic trajectories of dynamical systems with mixed phase space. Regarding the application to real situations, I have shown the capability and validity of this method by analyzing time series from fluid experiments.

As land-cover conversion continues to expand into ever more remote areas in the humid tropics, montane rainforests are increasingly threatened. In the south Ecuadorian Andes, they are not only subject to man-made disturbances but also to naturally occurring landslides. I was interested in the impact of this ecosystem dynamics on a key parameter of the hydrologic cycle, the soil saturated hydraulic conductivity (synonym: permeability; Ks from here on), because it is a sensitive indicator for soil disturbances. My general objective was to quantify the effects of the regional natural and human disturbances on the saturated hydraulic conductivity and to describe the resulting spatial-temporal patterns. The main hypotheses were: 1) disturbances cause an apparent displacement of the less permeable soil layer towards the surface, either due to a loss of the permeable surface soil after land-sliding, or as a consequence of the surface soil compaction under cattle pastures; 2) ‘recovery’ from disturbance, either because of landslide re-vegetation or because of secondary succession after pasture abandonment, involves an apparent displacement of the less permeable layer back towards the original depth an 3) disturbances cause a simplification of the Ks spatial structure, i.e. the spatially dependent random variation diminishes; the subsequent recovery entails the re-establishment of the original structure. In my first study, I developed a synthesis of recent geostatistical research regarding its applicability to soil hydraulic data, including exploratory data analysis and variogram estimation techniques; I subsequently evaluated the results in terms of spatial prediction uncertainty. Concerning the exploratory data analysis, my main results were: 1) Gaussian uni- and bivariate distributions of the log-transformed data; 2) the existence of significant local trends; 3) no need for robust estimation; 4) no anisotropic variation. I found partly considerable differences in covariance parameters resulting from different variogram estimation techniques, which, in the framework of spatial prediction, were mainly reflected in the spatial connectivity of the Ks-field. Ignoring the trend component and an arbitrary use of robust estimators, however, would have the most severe consequences in this respect. Regarding variogram modeling, I encouraged restricted maximum likelihood estimation because of its accuracy and independence on the selected lags needed for experimental variograms. The second study dealt with the Ks spatial-temporal pattern in the sequences of natural and man-made disturbances characteristic for the montane rainforest study area. To investigate the disturbance effects both on global means and the spatial structure of Ks, a combined design-and model-based sampling approach was used for field-measurements at soil depths of 12.5, 20, and 50 cm (n=30-150/depth) under landslides of different ages (2 and 8 years), under actively grazed pasture, fallows following pasture abandonment (2 to 25 years of age), and under natural forest. Concerning global means, our main findings were 1) global means of the soil permeability generally decrease with increasing soil depth; 2) no significant Ks differences can be observed among landslides and compared to the natural forest; 3) a distinct permeability decrease of two orders of magnitude occurs after forest conversion to pasture at shallow soil depths, and 4) the slow regeneration process after pasture abandonment requires at least one decade. Regarding the Ks spatial structure, we found that 1) disturbances affect the Ks spatial structure in the topsoil, and 2) the largest differences in spatial patterns are associated with the subsoil permeability. In summary, the regional landslide activity seems to affect soil hydrology to a marginal extend only, which is in contrast to the pronounced drop of Ks after forest conversion. We used this spatial-temporal information combined with local rain intensities to assess the partitioning of rainfall into vertical and lateral flowpaths under undisturbed, disturbed, and regenerating land-cover types in the third study. It turned out that 1) the montane rainforest is characterized by prevailing vertical flowpaths in the topsoil, which can switch to lateral directions below 20 cm depth for a small number of rain events, which may, however, transport a high portion of the annual runoff; 2) similar hydrological flowpaths occur under the landslides except for a somewhat higher probability of impermeable layer formation in the topsoil of a young landslide, and 3) pronounced differences in runoff components can be observed for the human disturbance sequence involving the development of near-surface impeding layers for 24, 44, and 8 % of rain events for pasture, a two-year-old fallow, and a ten-year-old fallow, respectively.
In der vorliegenden Arbeit werden Methoden der Erdsystemanalyse auf die Untersuchung der Habitabilität terrestrischer Exoplaneten angewandt. Mit Hilfe eines parametrisierten Konvektionsmodells für die Erde wird die thermische Evolution von terrestrischen Planeten berechnet. Bei zunehmender Leuchtkraft des Zentralsterns wird über den globalen Karbonat-Silikat-Kreislauf das planetare Klima stabilisiert. Für eine photosynthetisch-aktive Biosphäre, die in einem bestimmten Temperaturbereich bei hinreichender CO2-Konzentration existieren kann, wird eine Überlebenspanne abgeschätzt. Der Abstandsbereich um einen Stern, in dem eine solche Biosphäre produktiv ist, wird als photosynthetisch-aktive habitable Zone (pHZ) definiert und berechnet. Der Zeitpunkt, zu dem die pHZ in einem extrasolaren Planetensystem endgültig verschwindet, ist die maximale Lebenspanne der Biosphäre. Für Supererden, massereiche terrestrische Planeten, ist sie umso länger, je massereicher der Planet ist und umso kürzer, je mehr er mit Kontinenten bedeckt ist. Für Supererden, die keine ausgeprägten Wasser- oder Landwelten sind, skaliert die maximale Lebenspanne mit der Planetenmasse mit einem Exponenten von 0,14. Um K- und M-Sterne ist die Überlebensspanne einer Biosphäre auf einem Planeten immer durch die maximale Lebensspanne bestimmt und nicht durch das Ende der Hauptreihenentwicklung des Zentralsterns limitiert. Das pHZ-Konzept wird auf das extrasolare Planetensystem Gliese 581 angewandt. Danach könnte die 8-Erdmassen-Supererde Gliese 581d habitabel sein. Basierend auf dem vorgestellten pHZ-Konzept wird erstmals die von Ward und Brownlee 1999 aufgestellte Rare-Earth-Hypothese für die Milchstraße quantifiziert. Diese Hypothese besagt, dass komplexes Leben im Universum vermutlich sehr selten ist, wohingegen primitives Leben weit verbreitet sein könnte. Unterschiedliche Temperatur- und CO2-Toleranzen sowie ein unterschiedlicher Einfluss auf die Verwitterung für komplexe und primitive Lebensformen führt zu unterschiedlichen Grenzen der pHZ und zu einer unterschiedlichen Abschätzung für die Anzahl der Planeten, die mit den entsprechenden Lebensformen besiedelt sein könnten. Dabei ergibt sich, dass komplex besiedelte Planeten heute etwa 100-mal seltener sein müssten als primitiv besiedelte.
Today about 24 Million people worldwide suffer from dementia, Alzheimer’s Disease accounts for approximately 50-60% of all dementia cases. As the prevalence of dementia grows with increasing age Alzheimer’s Disease becomes more and more of an issue for society as the proportion of elderly people increases from year to year. It is well established, that the amino acid glutamate - quantitatively being the most important neurotransmitter in the central nervous system (CNS) - may reach toxic concentrations if not cleared from the synaptic cleft into which it is released during transmittance of action potentials. In Alzheimer’s Disease there is strong evidence for a generally impaired glutamate uptake system which in turn is thought to result in toxic levels of the amino acid with the potential to kill off neurons. The excitatory amino acid transporter 1 (EAAT1) belongs to the family of Na+-dependent glutamate transporter and accounts together with EAAT2 for most of the glutamate uptake in the CNS. In this project a new splice variant of EAAT1, skipping exon 3 was detected in human brain samples and subsequently called EAAT1Δ3, this being the second splice variant found after the recent detection of EAAT1Δ9. A method was developed to quantify the transcript of EAAT1 wt, EAAT1Δ3 and EAAT1Δ9 by means of real-time PCR. Samples were taken from different brain areas of a set of control and AD cases. The areas chosen for examination are affected differently in Alzheimer’s Disease, this was used an internal control for the experiments done in this project as to determine whether any effect observed is specific for AD, i.e. AD affected areas or is generally seen in all areas examined. The results of this project show that EAAT1Δ3 is transcribed in very low copy numbers making up a proportion of 0.15% of EAAT1 wt whereas EAAT1Δ9 is transcribed in a considerably large proportion of EAAT1 wt of 26.6%. It was moreover found that all EAAT1 variants are transcribed at significantly lower rates (P<0.0001) in AD cases, supporting the theory that EAAT1 protein expression is reduced to a point where glutamate uptake normally mediated by this transporter is impaired. This in turn is thought to result in toxic levels glutamate accounting for neuronal loss in the disease. No area-dependent effects were found, suggesting that the reduction of EAAT1 transcription is rather a result of an underlying general mechanism present in AD. Further research will have to be done to assess the degree of EAAT1 expression in AD and whether those future findings match with the result of this project.
In dieser Arbeit wird die Interaktion der Protagonisten in den Romanen von Horacio Castellanos Moya untersucht. Betrachtet werden die Bücher: La diabla en el espejo (2000), El arma en el hombre (2001), Donde no estén ustedes (2003), Insensatez (2004) und Desmoronamiento (2006). In der Reihenfolge abnehmender sozialer Distanz geht es nacheinander um Konzepte von professionellen Beziehungen, Freundschaft, familiären Beziehungen und Intimität. Geprüft wird hierbei die These, dass all diese Beziehungen als von Gewaltverhältnissen unterwandert dargestellt werden. Zum anderen soll eine detaillierte Analyse Aufschluss über Inszenierungsweisen, Implikationen und Folgen der dargestellten Gewalt ermöglichen. Als zentral erweisen sich hierbei einerseits der Begriff des „Epitaphs“, wie ihn Jacques Derrida in seiner Politiques de l’ amitié entwickelt und andererseits die „Theorie des kommunikativen Rabens“, welche Ottmar Ette in seinem Band ÜberLebenswissen: die Aufgabe der Philologie darstellt. Die Arbeit hofft außerdem dazu beizutragen, den Bekanntheitsgrad Castellanos Moyas, als einem herausragenden Vertreter zentralamerikanischer Belletristik, zu erhöhen.
Im Rahmen der fünften Erweiterungswelle der EU um zwölf neue Mitgliedsstaaten setzte die EU-Kommission erstmals auf das Instrument der Verwaltungspartnerschaften. Das sogenannte Twinning umfasst Projekte, bei denen Behördenmitarbeiter aus Verwaltungen der EU-Mitgliedsstaaten in Partnerbehörden der Beitrittsländer entsandt werden, um diese bei der Umsetzung des EU-Rechtsbestandes zu beraten. Im Gegensatz zur bisherigen Forschungsliteratur, die sich vor allem auf die Europäisierung der Beitrittsländer konzentriert, geht die Autorin dieser Forschungsarbeit davon aus, dass Twinning-Projekte auch zur Europäisierung der beteiligten deutschen Verwaltungen beitragen. Mithilfe neo-institutionalistischer Ansätze entwickelt die Autorin die These, dass Twinning-Projekte den Handlungsspielraum der involvierten Akteure innerhalb der europäischen Dimension erheblich erweitern. Inwieweit diese Akteure die von der EU-Kommission im Rahmen des PHARE-Programms zur Verfügung gestellten materiellen und immateriellen Ressourcen tatsächlich genutzt haben, wurde für vier deutsche Bundesressorts (BMWi, BMU, BMI, BMG) vergleichend untersucht. Als Ergebnis kann festgehalten werden, dass Effekte vertikaler Europäisierung stark von der Unterstützung nationaler Entscheidungsträger abhängen. Effekte horizontaler Europäisierung in den deutschen Ministerialverwaltungen werden hingegen vom Engagement der einzelnen Akteure bestimmt. Hier entstehen neue Kontakte und Netzwerke, die auf Formen neuartigen Regierens in der EU hinweisen (European Governance).
Das Instrument der direkten Demokratie, die Abwahl von Bürgermeistern, wird in der vorliegenden Arbeit spezifisch untersucht. Die Fallanalyse konzentriert sich auf die kommunalpolitische Ebene und lässt die Bundes- und Landesebene weitgehend außen vor. Dabei wird zunächst darauf eingegangen, warum es sich bei der Abwahl des Bürgermeisters um ein direktdemokratisches Mittel handelt. Ausgehend von der Unterteilung der lokalen Demokratie in eine repräsentative und direkte Komponente wird danach die Bürgermeisterabwahl nach politischen, juristischen und persönlichen Motiven eingeordnet. Zwar ist die Möglichkeit der Abwahl des Amtsinhabers bei vielen Bundesländern in der Gemeindeordnung bzw. den einzelnen Kommunalwahlgesetzen verankert, aber die Anwendung wird nur selten vom Stadt-/ Gemeinderat oder der Stadtverordnetenversammlung sowie den Bürgern praktiziert. Aus diesem Grund wird versucht, die bisherige Anwendung der Abwahl eines Bürgermeisters, in den betreffenden Bundesländern einerseits statistisch zu erfassen und anderseits akteurszentriert zu beschreiben.
The biological function and the technological applications of semiflexible polymers, such as DNA, actin filaments and carbon nanotubes, strongly depend on their rigidity. Semiflexible polymers are characterized by their persistence length, the definition of which is the subject of the first part of this thesis. Attractive interactions, that arise e.g.~in the adsorption, the condensation and the bundling of filaments, can change the conformation of a semiflexible polymer. The conformation depends on the relative magnitude of the material parameters and can be influenced by them in a systematic manner. In particular, the morphologies of semiflexible polymer rings, such as circular nanotubes or DNA, which are adsorbed onto substrates with three types of structures, are studied: (i) A topographical channel, (ii) a chemically modified stripe and (iii) a periodic pattern of topographical steps. The results are compared with the condensation of rings by attractive interactions. Furthermore, the bundling of two individual actin filaments, whose ends are anchored, is analyzed. This system geometry is shown to provide a systematic and quantitative method to extract the magnitude of the attraction between the filaments from experimentally observable conformations of the filaments.
Übergewicht, Diabetes oder Fettstoffwechselstörungen sind mit erniedrigten Adiponectinspiegeln assoziiert. Eine Modulation des Adiponectins kann durch genetische und metabolische Gegebenheiten erfolgen. Das Ziel dieser Arbeit war die Analyse von Faktoren, welche die Adiponectinspiegel beeinflussen können, sowie eine Charakterisierung der Oligomerverteilung unter verschiedenen metabolischen Bedingungen. In der MeSyBePo-Kohorte waren die zirkulierenden Adiponectinspiegel mit den Promotorpolymorphismen ADIPOQ -11377 C/G und ADIPOQ -11391 G/A im Adiponectingen assoziiert. Im Hinblick auf die metabolischen Faktoren korrelierte Adiponectin eng mit Parametern des Glukose- und Fettstoffwechsels sowie dem Übergewicht. Innerhalb von hyperinsulinämischen euglykämischen Clamps führte eine akute Hyperinsulinämie zu einer Abnahme der Adiponectinspiegel. Adiponectin zirkuliert im Serum als hochmolekulare (HMW), mittelmolekulare (MMW) und niedrigmolekulare (LMW) Spezies. Mit zunehmendem Körpergewicht konnte eine Verlagerung von HMW-Spezies hin zu den LMW-Spezies beobachtet werden. Durch eine moderate Gewichtsabnahme erhöhten sich die Anteile an HMW- und MMW-Adiponectin wieder. Während sich in Abhängigkeit vom Glukosemetabolismus keine Unterschiede in den Gesamtspiegeln ergaben, wurden bei Personen mit normaler Glukosetoleranz signifikant höhere Anteile an MMW-Adiponectin detektiert als bei Personen mit einem gestörten Glukosestoffwechsel. Insgesamt scheinen die HMW- und MMW-Spezies gegensätzlich zur LMW-Spezies reguliert zu werden. Die Arbeit unterstreicht die wichtige Rolle des Adiponectins im Glukose- und Fettstoffwechsel sowie bei einer Adipositas in vivo. Dabei waren Änderungen der Adiponectinspiegel bei Vorliegen von Insulinresistenz und Adipositas stets mit einer Umverteilung der Oligomerfraktionen verbunden. Vor allem die HMW- und MMW-Spezies des Adiponectins scheinen von entscheidender Bedeutung zu sein.
Intuitive Modelle der Informatik sind gedankliche Vorstellungen über informatische Konzepte, die mit subjektiver Gewissheit verbunden sind. Menschen verwenden sie, wenn sie die Arbeitsweise von Computerprogrammen nachvollziehen oder anderen erklären, die logische Korrektheit eines Programms prüfen oder in einem kreativen Prozess selbst Programme entwickeln. Intuitive Modelle können auf verschiedene Weise repräsentiert und kommuniziert werden, etwa verbal-abstrakt, durch ablauf- oder strukturorientierte Abbildungen und Filme oder konkrete Beispiele. Diskutiert werden in dieser Arbeit grundlegende intuitive Modelle für folgende inhaltliche Aspekte einer Programmausführung: Allokation von Aktivität bei einer Programmausführung, Benennung von Entitäten, Daten, Funktionen, Verarbeitung, Kontrollstrukturen zur Steuerung von Programmläufen, Rekursion, Klassen und Objekte. Mit Hilfe eines Systems von Online-Spielen, der Python Visual Sandbox, werden die psychische Realität verschiedener intuitiver Modelle bei Programmieranfängern nachgewiesen und fehlerhafte Anwendungen (Fehlvorstellungen) identifiziert.
Homogene Immunoassays sind immunologische Testverfahren, bei deren Durchführung vollständig auf Separations- und Waschschritte verzichtet werden kann. Der Substrate Channeling Immunoassay beruht auf der Weitergabe eines Substrates in einem immunologischen Komplex aus zwei Enzymen. Das Produkt des ersten Enzyms dient dem zweiten Enzym als Substrat zur Generierung eines photometrisch nachweisbaren Produktes. Voraussetzung für diese Weitergabe ist die enge räumliche Nähe beider Enzyme. Diese Nähe wird durch eine Bindung zwischen Analyt und anti-Analyt Antikörper vermittelt. Ein solcher Substrate Channeling Immunoassay wurde unter Verwendung der Enzyme Glucoseoxidase und Peroxidase aufgebaut. Das so etablierte System war funktionstüchtig, jedoch blieb seine Sensitivität hinter der normaler, heterogener Immunoassays zurück. Die Grundlage eines Fluorescence Quenching Immunoassays ist der gegenseitige Ausschluß zweier Antikörper bei der Bindung eines Dihapten-Konjugates. Das Konjugat besteht dabei aus dem Analyten und einem Fluorophor. Die beiden um die Konjugatbindung konkurrierenden Antikörper sind ein anti-Analyt Antikörper und ein anti-Fluorophor Antikörper, der zudem über die Eigenschaft verfügt, bei Bindung des Fluorophors dessen Fluoreszenz zu löschen. Externe Gaben des freien Analyten verschieben das eingestellte Gleichgewicht in Richtung Fluorophor-Bindung und damit Fluoreszenz-Löschung. Die Änderung der Fluoreszenz ist direkt an die Konzentration des freien Analyten gekoppelt und dient zu deren Bestimmung. Ein solcher Fluorescence Quenching Immunoassays wurde für die Konzentrationsbestimmung des Herbizides Diuron etabliert. Die erreichten Sensitivitäten erlauben die praktische, immundiagnostische Anwendung des Systems. Ein Dihapten-Konjugat wurde ebenfalls zum Aufbau eines Verfahrens zur Selektion Antikörper produzierender Zellen eingesetzt. Die Selektion der Antikörper produzierenden Zellen erfolgt unter Verwendung eines Toxinkonjugates. Dieses Konjugat besteht aus einem Liganden und einem Toxin. Die Antikörperbindung des Liganden behindert sterisch die Wechselwirkung der Toxinkomponente im Konjugat mit deren Zielstruktur in oder auf der Zelle. Nur Zellen die einen geeigneten Antikörper sezernieren, überleben die Selektion und reichern sich in der Kultur an. Das Selektionsverfahren wurde erfolgreich für die Selektion von E.coli Zellen eingesetzt, die einen rekombinanten, Fluorescein bindenden Antikörper produzierten. Das hierfür synthetisierte Toxinkonjugat bestand aus Fluorescein (Ligand) und Ampicillin (Toxinkomponente). Eine Ablösung der bisher für diese Aufgabe gebräuchlichen, außerordentlich kostenintensiven, Screening Methoden wird damit möglich.
Real costs of freight transportation have strong increased in Russia particularly during the period of price liberalization 1992–93. This paper investigates possible connections between rising transport costs and the evolution of the size structure of the system of cities in the Russian Federation and its federal subjects. Empirical findings suggest that under conditions of a closed system agglomeration processes according to the predictions of the model of Tabuchi et al. (2005) would have taken place especially in the periphere regions of the North and Far East.

Aus der Einführung: Betrachtet man Größe und Funktionsfähigkeit von Städten weltweit, so scheint der Eindruck unabwendbar, daß es einerseits Städte gibt, die über das Maß einer organisierbaren urbanen Einheit hinausgewachsen sind (Megacities) und andererseits kleine Städte existieren, die nicht in der Lage sind, ihren Einwohnern in einem notwendigen Umfang Dienste zur Verfügung zu stellen. Diese Überlegung wird vielfach unterstützt durch die Tatsache, daß die Megacity größer ist, als sie aufgrund ihres Bedeutungsüberschusses in einem hierarchischen Städtesystem im Sinne Christallers sein müßte. Für kleine Städte in einem dünn besiedelten Umland ergibt sich das Problem, daß es für bestimmte private Dienste und öffentliche Leistungen technisch bedingte Mindestmengen der Produktion gibt (z. B. Feuerwehr, Medizintechnik, Ausbildungsstätten etc.), die Nachfrage aus Stadt und Umland aber kleiner ist und aus diesem Grund das Angebot fehlt. Die betriebswirtschaftliche Entscheidung, die entsprechenden Dienste nicht anzubieten, kann zu gesamtwirtschaftlichen Wohlfahrtsverlusten führen, die aus den weiträumigen Beschaffungsfahrten der Haushalte resultieren. Wenn es nun einerseits Städte gibt, die zu groß sind und andererseits Städte existieren, die offenbar zu klein sind, so stellt sich die Frage, ob es eine optimale Stadtgröße gibt. Diese Frage könnte in eine weitere übertragen werden: Gibt es eine - möglicherweise von Christaller abweichende - optimale Städtehierarchie? Die Antworten auf die Fragen können von verschiedenen Disziplinen gegeben werden, sowohl die Stadtsoziologie als auch die Stadtplanung kann Beiträge liefern; in den nachfolgenden Überlegungen sollen wirtschaftliche Gesichtspunkte im Zentrum einer raumwirtschaftlichen und stadtökonomischen Analyse stehen.
This paper gives a review on the theoretical foundation for fiscal decentralisation and a status quo analysis of the intergovernmental relations in Mongolia. It consists of two parts. Part I briefly reviews the theories of fiscal decentralisation and its impact on the nations’ welfare considering the major challenges for a transition economy. Part II of the paper describes the general structure and scope of the government and examines the current fiscal autonomy in Mongolia focusing on the four main areas of intergovernmental relations. This paper concludes that local governments in Mongolia are still far away from having the political, administrative and fiscal autonomy. New approaches for the assignments of expenditures and revenues in Mongolia are urgently needed.
The paper describes the exchange program in between the University of Wisconsin/ Milwaukee and the University of Potsdam in the field of economics. It discusses in detail the development of the program, including the problems and challenges. Additionally a brief description of the curriculum is presented. Then the future possibilities of the Transatlantic Degree Program (TDP) are discussed and the influences and problems of the Bologna process analysed.
The role played by azobenzene polymers in the modern photonic, electronic and opto-mechanical applications cannot be underestimated. These polymers are successfully used to produce alignment layers for liquid crystalline fluorescent polymers in the display and semiconductor technology, to build waveguides and waveguide couplers, as data storage media and as labels in quality product protection. A very hot topic in modern research are light-driven artificial muscles based on azobenzene elastomers. The incorporation of azobenzene chromophores into polymer systems via covalent bonding or even by blending gives rise to a number of unusual effects under visible (VIS) and ultraviolet light irradiation. The most amazing effect is the inscription of surface relief gratings (SRGs) onto thin azobenzene polymer films. At least seven models have been proposed to explain the origin of the inscribing force but none of them describes satisfactorily the light induced material transport on the molecular level. In most models, to explain the mass transport over micrometer distances during irradiation at room temperature, it is necessary to assume a considerable degree of photoinduced softening, at least comparable with that at the glass transition. Contrary to this assumption, we have gathered a convincing evidence that there is no considerable softening of the azobenzene layers under illumination. Presently we can surely say that light induced softening is a very weak accompanying effect rather than a necessary condition for the formation of SRGs. This means that the inscribing force should be above the yield point of the azobenzene polymer. Hence, an appropriate approach to describe the formation and relaxation of SRGs is a viscoplastic theory. It was used to reproduce pulse-like inscription of SRGs as measured by VIS light scattering. At longer inscription times the VIS scattering pattern exhibits some peculiarities which can be explained by the appearance of a density grating that will be shown to arise due to the final compressibility of the polymer film. As a logical consequence of the aforementioned research, a thermodynamic theory explaining the light-induced deformation of free standing films and the formation of SRGs is proposed. The basic idea of this theory is that under homogeneous illumination an initially isotropic sample should stretch itself along the polarization direction to compensate the entropy decrease produced by the photoinduced reorientation of azobenzene chromophores. Finally, some ideas about further development of this controversial topic will be discussed.
Vitamin E ist der Überbegriff für 4 Tocopherole (α, β, γ und δ) sowie 4 Tocotrienole (α, β, γ und δ), die als gemeinsames Merkmal ein Chromanolringsystem sowie eine gesättigte (Tocopherole) bzw. ungesättigte (Tocotrienole) Seitenkette aufweisen. Neben ihrer antioxidativen Wirkung (Schutz von Membranen vor Lipidperoxidaton) konnten für einige Vitamin E - Formen auch eine Reihe von hochspezifischen, nicht-antioxidativen Wirkungen in vitro nachgewiesen werden. Meist bleibt jedoch unklar, ob ein solcher Effekt auch in vivo, also im Tiermodel oder direkt im Menschen, gefunden werden kann. In erster Linie müsste hierbei geklärt werden, ob die jeweilige Vitamin E - Form auch bioverfügbar, also in für eine Wirkung ausreichender Konzentration im Organismus vorhanden ist, oder aber vorher eliminiert und ausgeschieden wird. In dieser Doktorarbeit wurden deshalb wichtige Grundlagen zum Abbau der Tocopherole und Tocotrienole erarbeitet. • In HepG2-Zellen konnte der Abbau der Tocotrienole mit Hilfe flüssig- sowie gaschromatographischer Analysemethoden vollständig aufgeklärt werden. Wie sich hierbei ergab, verläuft der Abbau weitgehend in Analogie zum Abbau der Tocopherole über eine durch Cytochrom P450 katalysierte initiale ω-Hydroxylierung mit 5 nachfolgenden β-Oxidationsschritten. • In vitro konnten in HepG2 – Zellen die Abbauraten der verschiedenen Vitamin E - Formen bestimmt werden. Dies nahmen in folgender Reihenfolge zu: α-Tocopherol < γ-Tocopherol < α-Tocotrienol < γ-Tocotrienol. • Wie sich mit Hilfe eines mit Cytochrom P450 hochangereicherten Homogenats aus Rattenlebern ergab, stellt die initiale ω-Hydroxylierung einen geschwindigkeitsbestimmenden Schritt des Abbaus dar: α-Tocopherol wurde weit langsamer hydroxyliert als alle anderen Vitamin E – Formen. • Der unterschiedliche Abbau von α-Tocopherol und γ-Tocotrienol konnte auch im Mäuseversuch in vivo bestätigt werden. Nach Fütterung von Mäusen mit α-Tocopherol wurden nur geringe Mengen von α-Tocopherolmetaboliten im Urin der Mäuse gefunden, während nach Applikation von γ-Tocotrienol hohe Konzentrationen der γ-Tocotrienolmetabolite nachgewiesen wurden. In Plasma und Leber wiederum wurden (dem Futtergehalt entsprechende) hohe α-Tocopherolkonzentrationen entdeckt, während γ-Tocotrienol selbst nach hoher Gabe nicht oder nur in Spuren nachweisbar war. In HepG2 – Zellen konnte gezeigt werden, dass γ-Tocotrienol eine cytotoxische Wirkung auf die Hepatocarcinoma-Zelllinie HepG2 entfalten kann, indem durch die Aktivierung der proteolytischen Caspase 3 die Induktion des programmierten Zelltodes (Apoptose) ausgelöst wird. Abschliessend lässt sich festhalten, dass der Körper lediglich das natürliche α-Tocopherol vor dem Abbau bewahrt, die anderen Vitamin E – Formen jedoch als Fremdstoffe behandelt und rapide ausscheidet. Als doppelter Schutz vor Verlust des “wertvollen” α-Tocopherol dienen hierbei das α-Tocopherol Transfer Protein sowie die in dieser Arbeit gefundenen Unterschiede im ersten Schritt des Abbaus, der Cytochrom P450 - katalysierten ω-Hydroxylierung. Beides erklärt die bevorzugte Retention von α-Tocopherol im Organsimus und seine hohe Bioaktivität. Will man deshalb in vitro Ergebnisse anderer Vitamin E – Formen auf die in vivo Situation übertragen, muss man die geringe Bioverfügbarkeit dieser Substanzen berücksichtigen.
Magnetorotational instability (MRI) is one of the most important and most common instabilities in astrophysics. Today it is widely accepted that it serves as a major source of turbulent viscosity in accretion disks, the most energy efficient objects in the universe. The importance of the MRI for astrophysics has been realized only in recent fifteen years. However, originally it was discovered much earlier, in 1959, in a very different context. Theoretical flow of a conducting liquid confined between differentially rotating cylinders in the presence of an external magnetic field was analyzed. The central conclusion is that the additional magnetic field parallel to the axis of rotation can destabilize otherwise stable flow. Theory of non-magnetized fluid motion between rotating cylinders has much longer history, though. It has been studied already in 1888 and today such setup is usually referred as a Taylor-Couette flow. To prove experimentally the existence of MRI in a magnetized Taylor-Couette flow is a demanding task and different MHD groups around the world try to achieve it. The main problem lies in the fact that laboratory liquid metals which are used in such experiments are characterized by small magnetic Prandtl number. Consequently rotation rates of the cylinders must be extremely large and vast amount of technical problems emerge. One of the most important difficulties is an influence of plates enclosing the cylinders in any experiment. For fast rotation the plates tend to dominate the whole flow and the MRI can not be observed. In this thesis we discuss a special helical configuration of the applied magnetic field which allows the critical rotation rates to be much smaller. If only the axial magnetic field is present, the cylinders must rotate with angular velocities corresponding to Reynolds numbers of order Re ≈ 10^6. With the helical field this number is dramatically reduced to Re ≈ 10^3. The azimuthal component of the magnetic field can be easily generated by letting an electric current through the axis of rotation, In a Taylor-Couette flow the (primary) instability manifests itself as Taylor vortices. The specific geometry of the helical magnetic field leads to a traveling wave solution and the vortices are drifting in a direction determined by rotation and the magnetic field. In an idealized study for infinitely long cylinders this is not a problem. However, if the cylinders have finite length and are bounded vertically by the plates the situation is different. In this dissertation it is shown, with use of numerical methods, that the traveling wave solution also exists for MHD Taylor-Couette flow at finite aspect ratio H/D, H being height of the cylinders, D width of the gap between them. The nonlinear simulations provide amplitudes of fluid velocity which are helpful in designing an experiment. Although the plates disturb the flow, parameters like the drift velocity indicate that the helical MRI operates in this case. The idea of the helical MRI was implemented in a very recent experiment PROMISE. The results provided, for the first time, an evidence that the (helical) MRI indeed exists. Nevertheless, the influence of the vertical endplates was evident and the experiment can be, in principle, improved. Exemplary methods of reduction of the end-effect are here proposed. Near the vertical boundaries develops an Ekman-Hartmann layer. Study of this layer for the MHD Taylor-Couette system as well as its impact on the global flow properties is presented. It is shown that the plates, especially if they are conducting, can disturb the flow far more then previously thought also for relatively slow rotation rates.

Die Arbeit beschreibt die Synthese, Charakterisierung und Anwendung von meso- und mikroporösen Hochleistungspolymeren. Im ersten Teil wird die Synthese von mesoporösen Polybenzimidazol (PBI) auf der Basis einer Templatierungsmethode vorgestellt. Auf der Grundlage kommerzieller Monomere und Silikatnanopartikel sowie eines neuen Vernetzers wurde ein Polymer-Silikat-Hybridmaterial aufgebaut. Das Herauslösen des Silikats mit Ammoniumhydrogendifluorid führt zu mesoporösen Polybenzimidazolen mit spherischen Poren von 9 bis 11 nm Durchmesser. Die Abhängigkeit der beobachteten Porosität vom Massenverhältnis Silikat zu Polymer wurde ebenso untersucht wie die Abhängigkeit der Porosität vom Vernetzergehalt. Die Porosität vollvernetzter Proben zeigt eine lineare Abhängigkeit vom Verhältnis Silikat zu Polymer bis zu einem Grenzwert von 1. Wird der Grenzwert überschritten, ist teilweiser Porenkollaps zu beobachten. Die Abhängigkeit der Porosität vom Vernetzergehalt bei festem Silikatgehalt ist nichtlinear. Oberhalb einer kritischen Vernetzerkonzentration wird eine komplette Replikation der Nanopartikel gefunden. Ist die Vernetzerkonzentration dagegen kleiner als der kritische Wert, so ist der völlige Kollaps einiger Poren bei Stabilität der verbleibenden Poren zu beobachten. Ein komplett unporöses PBI resultiert bei Abwesenheit des Vernetzers. Die mesoporösen PBI-Netzwerke konnten kontrolliert mit Phosphorsäure beladen werden. Die erhaltenen Addukte wurden auf ihre Protonenleitfähigkeit untersucht. Es kann gezeigt werden, dass die Nutzung der vordefinierten Morphologie im Vergleich zu einem unstrukturierten PBI in höheren Leitfähigkeiten resultiert. Durch die vernetzte Struktur war des Weiteren genügend mechanische Stabilität gegeben, um die Addukte reversibel und bei sehr guten Leitfähigkeiten bis zu Temperaturen von 190°C bei 0% relativer Feuchtigkeit zu untersuchen. Dies ist für unstrukturierte Phosphorsäure/PBI - Addukte aus linearem PBI nicht möglich. Im zweiten Teil der Arbeit wird die Synthese intrinsisch mikroporöser Polyamide und Polyimide vorgestellt. Das Konzept intrinsisch mikroporöser Polymere konnte damit auf weitere Polymerklassen ausgeweitet werden. Als zentrales, strukturinduzierendes Motiv wurde 9,9'-Spirobifluoren gewählt. Dieses Molekül ist leicht und vielfältig zu di- bzw. tetrafunktionellen Monomeren modifizierbar. Dabei wurden bestehende Synthesevorschriften modifiziert bzw. neue Vorschriften entwickelt. Ein erster Schwerpunkt innerhalb des Kapitels lag in der Synthese und Charakterisierung von löslichen, intrinsisch mikroporösen, aromatischen Polyamid und Polyimid. Es konnte gezeigt werden, dass das Beobachten von Mikroporosität stark von der molekularen Architektur und der Verarbeitung der Polymere abhängig ist. Die Charakterisierung der Porosität erfolgte unter Nutzung von Stickstoffsorption, Kleinwinkelröntgenstreuung und Molecular Modeling. Es konnte gezeigt werden, dass die Proben stark vom Umgebungsdruck abhängigen Deformationen unterliegen. Die starke Quellung der Proben während des Sorptionsvorgangs konnte durch Anwendung des "dual sorption" Modells, also dem Auftreten von Porenfüllung und dadurch induzierter Henry-Sorption, erklärt werden. Der zweite Schwerpunkt des Kapitels beschreibt die Synthese und Charakterisierung mikroporöser Polyamid- und Polyimidnetzwerke. Während Polyimidnetzwerke auf Spirobifluorenbasis ausgeprägte Mikroporosität und spezifische Oberflächen von ca. 1100 m²/g aufwiesen, war die Situation für entsprechende Polyamidnetzwerke abweichend. Mittels Stickstoffsorption konnte keine Mikroporosität nachgewiesen werden, jedoch konnte mittels SAXS eine innere Grenzfläche von ca. 300 m²/g nachgewiesen werden. Durch die in dieser Arbeit gezeigten Experimente kann die Grenze zwischen Polymeren mit hohem freien Volumen und mikroporösen Polymeren somit etwas genauer gezogen werden. ausgeprägte Mikroporosität kann nur in extrem steifen Strukturen nachgewiesen werden. Die Kombination der Konzepte "Mesoporosität durch Templatierung" und "Mikroporosität durch strukturierte Monomere" hatte ein hierarchisch strukturiertes Polybenzimidazol zum Ergebnis. Die Präsenz einer Strukturierung im molekularen Maßstab konnte SAXS bewiesen werden. Das so strukturierte Polybenzimidazol zeichnete sich durch eine höhere Protonenleitfähigkeit im Vergleich zu einem rein mesoporösen PBI aus. Der letzte Teil der Arbeit beschäftigte sich mit der Entwicklung einer neuen Synthesemethode zur Herstellung von Polybenzimidazol. Es konnte gezeigt werden, dass lineares PBI in einer eutektischen Salzschmelze aus Lithium- und Kaliumchlorid synthetisiert werden kann. Die Umsetzung der spirobifluorenbasierten Monomere zu löslichem oder vernetztem PBI ist in der Salzschmelze möglich.

This work is concerned with the spatio-temporal structures that emerge when non-identical, diffusively coupled oscillators synchronize. It contains analytical results and their confirmation through extensive computer simulations. We use the Kuramoto model which reduces general oscillatory systems to phase dynamics. The symmetry of the coupling plays an important role for the formation of patterns. We have studied the ordering influence of an asymmetry (non-isochronicity) in the phase coupling function on the phase profile in synchronization and the intricate interplay between this asymmetry and the frequency heterogeneity in the system. The thesis is divided into three main parts. Chapter 2 and 3 introduce the basic model of Kuramoto and conditions for stable synchronization. In Chapter 4 we characterize the phase profiles in synchronization for various special cases and in an exponential approximation of the phase coupling function, which allows for an analytical treatment. Finally, in the third part (Chapter 5) we study the influence of non-isochronicity on the synchronization frequency in continuous, reaction diffusion systems and discrete networks of oscillators.
The styles of deformation of the fore-arc wedges along the Chilean convergent margin are observed to vary significantly, despite similar plate kinematic conditions. Here, I focus on the analysis of fore-arc deformation on two regions along the Chilean convergent margin at 20°-24°S and 37°-42°S. Although both regions are subjected to the oblique subduction of the oceanic Nazca plate and backstopped by the Andes mountain chain; they display different patterns of deformation. The northern Chilean study area (20° - 24°S) is characterized by an exceptionally thick crust of about 60 km beneath the Altiplano – Puna plateau, lack of an accretionary wedge in the fore-arc due to hyperarid climate, and consequently a sediment starved trench. Two major margin parallel strike slip faults are observed in this area, the Atacama Fault Zone (AFZ) and the Precordilleran Fault System (PFS). Both strike-slip faults do not exhibit significant recent displacement. The southern study area (37° - 42°S), compared to the northern study area, is characterized by lower topography, high precipitation rates (~2000 mm/yr), and a younger subducted oceanic plate. An active strike-slip fault, the Liquiñe-Ofqui-Fault-Zone (LOFZ), shows ~1 cm/yr recent dextral movement and shapes the surface of this area. Thus, the southern Chilean study area exhibits localized strike-slip motion. Within this area the largest earthquake ever recorded, the 1960 Valdivia earthquake, occurred with a moment magnitude of MW=9.5. I have constructed 2D thermal models and 3D mechanical models for both Chilean study areas to study processes related to active subduction. The applied numerical method is the finite element technique by means of the commercial software package ABAQUS. The thermal models are focused on the thermal conditions along the plate interface. The thermal structure along the plate interface reveals the limits of coupling but also the type of transition from coupled to uncoupled and vice versa. The model results show that shear heating at the plate interface is an important mechanism that should be taken into account. The models also show that the thermal condition at the downdip limit of the coupling zone leads to a sharp decrease of friction along the interface. Due to the different geometries of the two Chilean study areas, such as the slab dip and the thickness of the continental crust, the downdip limit of the southern study area is slightly shallower than that of the northern study area. The results of the 2D thermal models are used to constrain the spatial extent of the coupling zone in the 3D mechanical models. 3D numerical simulations are used to investigate how geometry, rheology and mechanical parameters influence strain partitioning and styles of deformation in the Chilean fore-arc. The general outline of the models is based on the fore-arc geometry and boundary conditions as derived from geophysical and geological field data. I examined the influence of different rheological approaches and varying physical properties of the fore-arc to identify and constrain the parameters controlling the difference in surface deformation between the northern and southern study area. The results of numerical studies demonstrate that a small slab dip, a high coefficient of basal friction, a high obliquity of convergence, and a high Young’s modulus favour localisation of deformation in the fore-arc wedge. This parameter study helped me to constrain preferred models for the two Chilean study areas that fit to first order observations. These preferred models explain the difference in styles of deformation as controlled by the angle of obliquity, the dip of subducting slab, and the strength of wedge material. The difference in styles can be even larger if I apply stronger coupling between plates within the southern area; however, several independent observations indicate opposite tendency showing southward decrease of intensity of coupling. The weaker wedge material of the preferred model for the northern study area is associated with advanced development of the adjacent orogen, the Central Andes. Analysis of world-wide examples of oblique subduction zones supports the conclusion that more mature subduction zones demonstrate less pronounced localization of strike-slip motion.
Phytol aus dem Chlorophyllabbau ist limitierend für die Tocopherol (Vitamin E)-Synthese Als Bestandteil von Chlorophyll ist Phytol das am häufigsten vorkommende Isoprenoid in der Biosphäre. Große Mengen an Chlorophyll werden jährlich degradiert und dabei wird Phytol freigesetzt, über dessen Verbleib jedoch wenig bekannt ist. Es sollte der Nachweis erbracht werden, dass im Zuge des Chlorophyllabbaus hydrolysiertes Phytol Eingang in die Synthese anderer Phytylderivate findet. Während der Gehalt an Tocopherol, Chlorophyll und Fettsäurephytylester entwicklungs- bzw. seneszenzabhängig ist, bleibt der Gehalt an Phyllochinon etwa gleich. Auch in Samen ist der Gehalt von Tocopherol, Chlorophyll und Fettsäurephytylester entwicklungsabhängig. Es wurde gefolgert, dass nur die Synthesen von Tocopherol und Fettsäurephytylester während des Chlorophyllabbaus stimuliert werden. Daher sollten Mutanten analysiert werden, welche im Chlorophyllabbau inhibiert sind. Da Chlorophyllase den ersten Schritt des Chlorophyllabbaus katalysiert, wurden zwei unabhängige T-DNA-Insertionsmutanten für Chlorophyllase1 (CHL1) und eine T-DNA-Insertionsmutante für Chlorophyllase2 (CHL2) identifiziert und eine chl1-1chl2-Doppelmutante erzeugt. Die Analyse der Chlorophyllidanteile ergab eine im Vergleich zum Wildtyp starke Reduktion in den chl1-Mutanten, während der Chlorophyllidanteil von chl2 ähnlich hoch dem Wildtyp ist. Der Chlorophyllidanteil sich entwickelnder chl1-1chl2-Pflanzen nahm in der Seneszenz zu. Die Chlorophyllasemutanten zeigten kein verändertes Seneszenzverhalten im Vergleich zu den Wildtypen. Ferner konnte in den chl1-Linien nur geringfügig weniger Tocopherol und Fettsäurephytylester als in den Wildtypen nachgewiesen werden. Auch der Tocopherolgehalt der Samen war in den Chlorophyllasemutanten unverändert zu den Wildtypen. Aufgrund dessen wurde gefolgert, dass neben den Chorophyllasen CHL1 und CHL2 weitere Chlorophyllhydrolasen in Samen und Blättern von Arabidopsis existieren. Daher wurde auf andere Mutanten zurückgegriffen, in denen der Chlorophyllabbau stark inhibiert ist und die Seneszenz nach Dunkelinkubation im Vergleich zum Wildtyp deutlich verzögert ist. Eine deutliche Korrelation zwischen vermindertem Chlorophyllabbau und Gehalt an Tocopherol und Fettsäurephytylester konnte in den staygreen-Mutanten pao1 und zwei unabhängigen SGR (staygreen)-RNAi-Linien nachgewiesen werden. Damit konnte eindeutig gezeigt werden, dass die Synthese von Tocopherol und der Fettsäurephytylester durch die Chlorophyllhydrolyse induziert wird. Es wurde gefolgert, dass vor allem unter Seneszenz- bzw. Stressbedingungen dieser alternative Syntheseweg von Phytol eine Rolle spielt. Dennoch kommt der Phytylsynthese durch die de novo-Isoprenoidsynthese auch eine Bedeutung zu. Nach Behandlung von stickstoffmangelgestressten Wildtyppflanzen mit dem Inhibitor Fosmidomycin, welcher die plastidäre de novo-Isoprenoidsynthese hemmt, war der Tocopherolgehalt gegenüber stickstoffmangelgestressten Kontrollpflanzen stark reduziert. Ferner konnte eine T-DNA-Insertionsmutante der Geranylgeranylreduktase (GGR) identifiziert werden. Diese Mutante kann nur auf Nährmedium überleben, hat nur wenige grüne Blätter und bildet keine Samen. Es konnte kein Phyllochinon, Chlorophyll und keine Fettsäurephytylester, jedoch geringe Mengen Tocopherol nachgewiesen werden. Der Resttocopherolgehalt wird auf die Nebenaktivität einer anderen Reduktase zurückgeführt. Weiterhin wurde nur das Geranylgeranylderivat des Chlorophylls identifiziert. Diese Ergebnisse erlauben den Schluss, dass die phytylgruppenübertragenen Enzyme der Tocopherol-, Phyllochinon- und Fettsäurephytylestersynthese eine hohe Substratspezifität für die Phytylgruppe aufweisen. Nach Fütterung von Phytol konnte in ggr Tocopherol und Chlorophyll bestimmt werden. Aufgrund dessen kann gefolgert werden, dass Chlorophyllsynthetase aus Arabidopsis sowohl Geranylgeranyl-, als auch Phytylpyrophosphat als Substrat nutzen kann und damit ein breiteres Substratspektrum aufweist.
Characterisation of silica in Equisetum hyemale and its transformation into biomorphous ceramics

Equisetum spp. (horsetail / “Schachtelhalm”) is the only surviving genus of the primitive Sphenopsids vascular plants which reached their zenith during the Carboniferous era. It is an herbaceous plant and is distinguished by jointed stems with fused whorl of nodal leaves. The plant has been used for scouring kitchen utensils and polishing wood during the past time due to its high silica encrustations in the epidermis. Equisetum hyemale (scouring rush) can accumulate silica up to 16% dry weight in its tissue, which makes this plant an interesting candidate as a renewable resource of silica for the synthesis of biomorphous ceramics. The thesis comprises a comprehensive experimental study of silica accumulations in E.hyemale using different characterisation techniques at all hierarchical levels. The obtained results shed light on the local distribution, chemical form, crystallinity, and nanostructure of biogenic silica in E.hyemale which were quite unclear until now. Furthermore, isolation of biogenic silica from E.hyemale to obtain high grade mesoporous silica with high purity is investigated. Finally, syntheses of silicon carbide (b-SiC) by a direct thermoconversion process of E.hyemale is attempted, which is a promising material for high performance ceramics. It is found that silica is deposited continuously on the entire epidermal layer with the highest concentration on the knobs. The highest silicon content is at the knob tips (≈ 33%), followed by epidermal flank (≈ 17%), and inner lower knob (≈ 6%), whereas there is almost no silicon found in the interior parts. Raman spectroscopy reveals the presence of at least two silica modifications in E.hyemale. The first type is pure hydrated amorphous silica restricted to the knob tips. The second type is accumulated on the entire continuous outer layer adjacent to the epidermis cell walls. It is lacking silanol groups and is intimately associated with polysaccharides (cellulose, hemicellulose, pectin) and inorganic compounds. Silica deposited in E.hyemale is found to be mostly amorphous with almost negligible amounts of crystalline silica in the form of a-quartz (< 7%). The silica primary particles have a plate-like shape with a thickness of about 2 nm. Pure mesoporous amorphous silica with an open surface area up to 400 m2/g can be obtained from E.hyemale after leaching the plant with HCl to remove the inorganic impurities followed by a calcination treatment. The optimum calcination temperature appears to be around 500°C. Calcination of untreated E.hyemale causes a collapse of the biogenic silica structure which is mainly attributed to the detrimental action of alkali ions present in the native plant. Finally, pure b-SiC with a surface area of about 12 m2/g is obtained upon direct pyrolysis of HCl-treated E.hyemale samples in argon atmosphere. The original structure of native E.hyemale is substantially retained in the biomorphous b-SiC. The results of this thesis lead to a better understanding of the silicification process and allow to draw conclusions about the role of silica in E.hyemale. In particular, a templating role of the plant biopolymers for the synthesis of the nanostructured silica within the plant body can be deduced. Moreover, the high grade ultrafine amorphous silica isolated from E.hyemale promises applications as adsorbent and catalyst support and as silica source for the fabrication of silica-based composites. The synthesis of biomorphous b-SiC from sustainable and low-cost E.hyemale is still in its initial stage. The present thesis demonstrates the principal possibility of carbothermal synthesis of SiC from E.hyemale with the prospect of potential applications, for instance as refractory materials, catalyst supports, or high performance advanced ceramics.
Exploring elections features from a geographical perspective is the focus of this study. Its primary objective is to develop a scientific approach based on geoinformation technology (GIT) that promotes deeper understanding how geographical settings affect the spatial and temporal variations of voting behaviour and election outcomes. For this purpose, the five parliamentary elections (1991-2005) following the political turnaround in 1990 in the South East European reform country Albania have been selected as a case study. Elections, like other social phenomena that do not develop uniformly over a territory, inherit a spatial dimension. Despite of fact that elections have been researched by various scientific disciplines ranging from political science to geography, studies that incorporate their spatial dimension are still limited in number and approaches. Consequently, the methodologies needed to generate an integrated knowledge on many facets that constitute election features are lacking. This study addresses characteristics and interactions of the essential elements involved in an election process. Thus, the baseline of the approach presented here is the exploration of relations between three entities: electorate (political and sociodemographic features), election process (electoral system and code) and place (environment where voters reside). To express this interaction the concept of electoral pattern is introduced. Electoral patterns are defined by the study as the final view of election results, chiefly in tabular and/or map form, generated by the complex interaction of social, economic, juridical, and spatial features of the electorate, which has occurred at a specific time and in a particular geographical location. GIT methods of geoanalysis and geovisualization are used to investigate the characteristics of electoral patterns in their spatial and temporal distribution. Aggregate-level data modelled in map form were used to analyse and visualize the spatial distribution of election patterns components and relations. The spatial dimension of the study is addressed in the following three main relations: One, the relation between place and electorate and its expression through the social, demographic and economic features of the electorate resulting in the profile of the electorate’s context; second, the electorate-election interaction which forms the baseline to explore the perspective of local contextual effects in voting behaviour and election results; third, the relation between geographical location and election outcomes reflecting the implication of determining constituency boundaries on election results. To address the above relations, three types of variables: geo, independent and dependent, have been elaborated and two models have been created. The Data Model, developed in a GIS environment, facilitates structuring of election data in order to perform spatial analysis. The peculiarity of electoral patterns – a multidimensional array that contains information on three variables, stored in data layers of dissimilar spatial units of reference and scales of value measurement – prohibit spatial analysis based on the original source data. To perform a joint spatial analysis it is therefore mandatory to restructure the spatial units of reference while preserving their semantic content. In this operation, all relevant electoral as well as socio-demographic data referenced to different administrative spatial entities are re-referenced to uniform grid cells as virtual spatial units of reference. Depending on the scale of data acquisition and map presentation, a cell width of 0.5 km has been determined. The resulting fine grid forms the basis of subsequent data analyses and correlations. Conversion of the original vector data layers into target raster layers allows for unification of spatial units, at the same time retaining the existing level of detail of the data (variables, uniform distribution over space). This in turn facilitates the integration of the variables studied and the performance of GIS-based spatial analysis. In addition, conversion to raster format makes it possible to assign new values to the original data, which are based on a common scale eliminating existing differences in scale of measurement. Raster format operations of the type described are well-established data analysis techniques in GIT, yet they have rarely been employed to process and analyse electoral data. The Geovisualization Model, developed in a cartographic environment, complements the Data Model. As an analog graphic model it facilitates efficient communication and exploration of geographical information through cartographic visualization. Based on this model, 52 choropleth maps have been generated. They represent the outcome of the GIS-based electoral data analysis. The analog map form allows for in-depth visual analysis and interpretation of the distribution and correlation of the electoral data studied. For researchers, decision makers and a wider public the maps provide easy-to-access information on and promote easy-to-understand insight into the spatial dimension, regional variation and resulting structures of the electoral patterns defined.
This collection contains 13 papers presented in the workshop on the "The Celtic Languages in Contact" organised by Hildegard L. C. Tristram at the XIII International Celtic Congress in Bonn (Germany), July 23rd - 27th, 2007. The authors of two papers from another section also contributed their papers to this volume, as they deal with closely related issues. The time-span covered ranges from potential pre-historic contacts of Celtic with Altaic languages or Nostratic cognates in Celtic, through the hypothesis of Afro-Asiatic as a possible substrate for Celtic, Latin and Gaulish contacts in Gaul, the impact of Vulgar Latin on the formation of the Insular Celtic Languages as a linguistic area (Sprachbund), to various contact scenarios involving the modern Insular Celtic languages as well as English and French. The final paper reflects on the political status of the modern Insular Celtic languages in the Europe of the 27 EU countries.
Entwicklung, Implementierung und Erprobung eines planetaren Informationssystems auf Basis von ArcGIS

Mit der Entwicklung der modernen Raumfahrt Mitte der 60er-Jahre des zwanzigsten Jahrhunderts und der Eroberung des Weltraums brach eine neue Epoche der bis dato auf Beobachtungen mit dem Teleskop gestützten planetaren Forschung an. Während des Wettrennens um die technologische Führerschaft im All zur Zeit des Kalten Krieges war das erste Ziel die Entsendung von Satelliten zur Erdbeobachtung, denen aber schon bald Sonden zum Mond und den benachbarten Planeten folgten. Diese Missionen lieferten eine enorme Fülle von Informationen in Form von Bildern und Messergebnissen in unterschiedlichen Datenformaten. Diese galt und gilt es zu strukturieren, zu verwalten, zu aktualisieren und zu interpretieren. Für die Interpretation terrestrischer Daten werden geographische Informationssysteme (GIS) hinzugezogen, die jedoch für planetare Anwendungen aufgrund unterschiedlicher Voraussetzungen nicht ohne weiteres eingesetzt werden können. Daher wurde im Rahmen dieser Arbeit die für die Verwaltung von geographischen Daten der Erdfernerkundung kommerziell erhältliche Software ArcGIS Desktop 9.0 / 9.1 (ESRI) mit eigenen Programmen und Modulen für die Planetenforschung angepasst. Diese ermöglichen die Aufbereitung und den Import planetarer Bild- und Textinformation in die kommerzielle Software. Zusätzlich wurde eine planetare Datenbank zur Speicherung und zentralen Verwaltung der Informationen aufgebaut. Die im Rahmen dieser Arbeit entwickelten Softwarekomponenten ermöglichen die schnelle und benutzerfreundliche Aufbereitung der in der Datenbank gehaltenen Informationen und das Auslesen in Dateiformate, die für geographische Informationssysteme geeignet sind. Des Weiteren wurde eine „Werkzeugleiste“ für ArcGIS entwickelt, die das Arbeiten mit planetaren Datensätzen beträchtlich beschleunigt und vereinfacht. Sie beinhaltet auch Module zur wissenschaftlichen Interpretation der planetaren Informationen, wie beispielsweise der Berechnung der Oberflächenrauigkeit der Marsoberfläche inklusive der flächendeckenden Kalibrierung der Eingangs-Basisdaten. Exemplarisch konnte gezeigt werden, dass das Verfahren eine verbesserte Berechnung der Oberflächenrauigkeit ermöglicht, als bisher angewandte Ansätze. Zudem wurde eine auf ArcGIS basierende Prozesskette zur Berechnung von hierarchischen Flussnetzen entwickelt und erprobt. Das terrestrische Beispiel, die Analyse eines Abflusssystems auf Island, zeigte eine sehr große Übereinstimmung der errechneten Gewässernetze mit den morphologischen Gegebenheiten vor Ort. Daraus ließ sich eine hohe Genauigkeit der mit demselben Ansatz errechneten Gewässernetze auf dem Mars ableiten. Auf der Grundlage der in dieser Arbeit entwickelten Programme und Module lassen sich auch Daten zukünftiger Missionen aufbereiten und in ein solches System einbinden, um diese mit eigenen Ansätzen zu verwalten, zu aktualisieren und für neue wissenschaftliche Fragestellungen perfekt anzupassen, einzusetzen und zu präsentieren, um so neue wissenschaftliche Erkenntnisse in der Planetenforschung zu gewinnen.
Fragestellung (Auszug): Die polnische Landwirtschaft wird im Hinblick auf ihre Rolle in der EU als nicht wettbewerbsfähig einge-schätzt und es scheint unumstritten, dass strukturelle Reformen unabdingbar sind. Ratschläge für die Ent-wicklung der polnischen Landwirtschaft gibt es viele. Unter anderem wird Polen die Besetzung von potentiellen Marktnischen empfohlen. Als Hoffnungsträger gilt dabei der ökologische Landbau, für den die extensive, traditionelle polnische Landwirtschaft eine optimale Voraussetzung darstellt. Neben einer EU-weit steigenden Nachfrage nach Bio-Lebensmitteln spricht für diese verstärkte Nischenbesetzung die Tatsache, dass die arbeitsintensive ökologische Wirtschaftsweise zumindest teilweise der zu erwartenden hohen Arbeitslosigkeit im Agrarsektor entgegen wirken kann. Damit würde der Biolandbau einen entscheidenden Beitrag für eine nachhaltige Regionalentwicklung leisten. Trotz positiver Entwicklungen in den letzten Jahren, verlief die Entwicklung des ökologischen Landbaus in Polen allerdings schleppend, und das Potential Polens scheint in diesem Bereich noch längst nicht ausgeschöpft. Diese Arbeit widmet sich der Frage, welche institutionellen Erklärungen es für diese Entwicklung gibt, verbunden mit der Hoffnung, institutionelle Faktoren ausfindig zu machen, welche die Entwicklung besonders positiv befördern oder andererseits abbremsen. Dabei werden ausgehend von einem institutionentheoretischen Hintergrund relevante formale und informelle Institutionen auf polnischer und EU-Ebene untersucht, sowie ver-schiedene Akteursgruppen, sowie deren Interaktion mit den institutionellen Entwicklungen beleuchtet.
Aus der Einleitung: Vor 15 Jahren hat Polen mit dem Transformationsprozess begonnen, der das Land von einer sozialistischen Planwirtschaft mit autoritärem Regime in einen modernen demokratischen Staat marktwirtschaftlicher Ausrichtung verwandelt hat. Seither genießt das polnische Volk nach vielen Jahren der Fremdherrschaft wieder seine eigene Souveränität. Mit dem Ende der sozialistischen Ära begann eine Phase umfangreicher und weit reichender institutioneller, wirtschaftlicher und sozialer Veränderungen. Diese gehen einher mit hoher Arbeitslosigkeit und starker Verunsicherung im Umgang mit den sich neu bildenden Strukturen, was eine große Belastung für die Bevölkerung darstellt. Den Herausforderungen zum Trotz haben es die Polen geschafft, in nur wenigen Jahren ein institutionelles und wirtschaftliches Niveau zu erreichen, das sich an dem Niveau westeuropäischer Staaten orientiert. Diese Leistung wurde u. a. durch die Europäische Kommission anerkannt, indem 1998 zunächst die Beitrittsverhandlungen mit Polen eröffnet wurden und das Land im Mai 2004 zur Europäischen Union (EU) beitrat. Während die alten Mitgliedsstaaten der Europäischen Union eine lang andauernde Phase wirtschaftlichen Abschwungs erleiden, findet im angrenzenden Polen ein rasanter wirtschaftlicher Aufholungsprozess statt. Wachstumsraten von 5 % und mehr sind hierbei keine Seltenheit. Diese Entwicklung wird von vielen Bürgern der alten EU Mitgliedssaaten in Zeiten steigender Arbeitslosigkeit und sinkender Löhne wahrgenommen und teilweise als Ursache der eigenen Misere identifiziert. Tatsache aber ist, dass sogar bei anhaltender Dynamik der polnischen Wirtschaft ein Erreichen des westeuropäischen Standards, wenn überhaupt, erst in mehreren Jahrzehnten als realistisch erscheint. Ebenso wie im wirtschaftlichen Bereich stellt sich die institutionelle Diskrepanz zwischen Polen und den Ländern der EU der 15 dar. Zu groß sind die Unterschiede, als dass sie in nur einem Jahrzehnt hätten nachgeholt und ausgeglichen werden könnten. Als zusätzlich Hindernis ist zu werten, dass sich in den letzten Jahren die Berichte der Europäischen Kommission, der Weltbank oder Transparency International mehren, Korruption in Polen bedrohe sämtliche Sphären des öffentlichen und privaten Lebens und die junge „alte“ Demokratie somit durch hausgemachte Probleme ins Strudeln geraten könnte. Innerhalb der Europäischen Kommission scheint man sich des Problems der Korruption in Polen bewusst zu sein und befürchtet diese in Form eines trojanischen Pferdes importiert zu bekommen (Vgl. Dehley 2002: 4). Andere Organisationen wie GRECO1 zeichnen ein düstereres Szenario, da sie der Korruption in Polen attestieren, die demokratischen Institutionen auszuhöhlen.
Problemstellung: Schuleingangsuntersuchungen belegen zunehmend Defizite in der motorischen und kognitiven Entwicklung von Einschülern. Im Hinblick auf Präventivmaßnahmen gilt dem motorischen und kognitiven Entwicklungsstand von Vorschulkindern ein verstärktes Forschungsinteresse. Die bisherige Wissensbasis soll mit aktuellen Untersuchungsergebnissen zum Zusammenhang von Motorik und Kognition bei Vorschulkindern erweitert werden. Methodik: Untersucht wurden 101 Kinder in vier Potsdamer Kitas im Alter von 3,5-4,6 Jahren. Zur Überprüfung der koordinativen und konditionellen Fähigkeiten wurden Standweitsprung, Sechs-Meter-Lauf, Balancieren, Einbeinstand und Seitliches Umsetzen sowie Handkoordination eingesetzt (KTK; Kiphard & Schilling 1972; Vogt 1978). Zusätzlich wurden in Anlehnung an das Brain-Gym-Konzept (Dennison 1984) körpermitteüberschreitende Armbewegungen qualitativ bewertet und auf Validität und Reliabilität geprüft. Die kognitiven Fähigkeiten wurden mit dem Bildbasierten Intelligenztest getestet (Schaarschmidt, Ricken, Kieschke & Preuß 2004). Ergebnisse: Jungen sind deutlich schneller und kräftiger als Mädchen, können aber geringfügig schlechter balancieren und feinkoordinieren. Ein Entwicklungssprung in den motorischen Leistungen ab dem vierten Lebensjahr konnte nachgewiesen werden. Weiterhin zeigte sich ein positiver signifikanter Zusammenhang zwischen der Gleichgewichtsfähigkeit und kognitiven Fähigkeiten bei den männlichen 3,5-Jährigen (r=.28-r=.32; p=0,01). Dies bestätigt: Je jünger die Kinder, desto stärker der Zusammenhang von Motorik und Intelligenz. Bei einseitiger Testung zeigten sich schwache signifikante Korrelationen von kognitiven Leistungen der Kinder und Qualifikation der Eltern (r=.18 - r=.21; p=0,01). Die Qualitative Bewertung der Arm- und Handbewegungen eignet sich wegen unbestätigter Validität und Reliabilität nicht als Untersuchungsmethode im Sinne einer frühfunktionellen Diagnostik.
Der vorliegende Text beschäftigt sich mit der religiös, kulturell und rassisch begründeten Ausgrenzung von Minderheiten durch die Mehrheitsgesellschaft im Zeitalter der europäischen Nationalstaaten, die fast immer im Rückgriff auf konstruierte historische Belege geschieht. Am Beispiel von Deutschland und Österreich wird der Zusammenhang aufgezeigt zwischen der staatsbürgerlichen Gleichstellung der Juden im 19. und frühen 20. Jahrhundert einerseits, und Krieg und Nationsbildung, bürgerlichem Selbstverständnis und Geschichtsdeutung in legitimatorischer Absicht andererseits.
The Arctic plays a key role in Earth’s climate system as global warming is predicted to be most pronounced at high latitudes and because one third of the global carbon pool is stored in ecosystems of the northern latitudes. In order to improve our understanding of the present and future carbon dynamics in climate sensitive permafrost ecosystems, the present study concentrates on investigations of microbial controls of methane fluxes, on the activity and structure of the involved microbial communities, and on their response to changing environmental conditions. For this purpose an integrated research strategy was applied, which connects trace gas flux measurements to soil ecological characterisation of permafrost habitats and molecular ecological analyses of microbial populations. Furthermore, methanogenic archaea isolated from Siberian permafrost have been used as potential keystone organisms for studying and assessing life under extreme living conditions. Long-term studies on methane fluxes were carried out since 1998. These studies revealed considerable seasonal and spatial variations of methane emissions for the different landscape units ranging from 0 to 362 mg m-2 d-1. For the overall balance of methane emissions from the entire delta, the first land cover classification based on Landsat images was performed and applied for an upscaling of the methane flux data sets. The regionally weighted mean daily methane emissions of the Lena Delta (10 mg m-2 d-1) are only one fifth of the values calculated for other Arctic tundra environments. The calculated annual methane emission of the Lena Delta amounts to about 0.03 Tg. The low methane emission rates obtained in this study are the result of the used remotely sensed high-resolution data basis, which provides a more realistic estimation of the real methane emissions on a regional scale. Soil temperature and near soil surface atmospheric turbulence were identified as the driving parameters of methane emissions. A flux model based on these variables explained variations of the methane budget corresponding to continuous processes of microbial methane production and oxidation, and gas diffusion through soil and plants reasonably well. The results show that the Lena Delta contributes significantly to the global methane balance because of its extensive wetland areas. The microbiological investigations showed that permafrost soils are colonized by high numbers of microorganisms. The total biomass is comparable to temperate soil ecosystems. Activities of methanogens and methanotrophs differed significantly in their rates and distribution patterns along both the vertical profiles and the different investigated soils. The methane production rates varied between 0.3 and 38.9 nmol h-1 g-1, while the methane oxidation ranged from 0.2 to 7.0 nmol h-1 g-1. Phylogenetic analyses of methanogenic communities revealed a distinct diversity of methanogens affiliated to Methanomicrobiaceae, Methanosarcinaceae and Methanosaetaceae, which partly form four specific permafrost clusters. The results demonstrate the close relationship between methane fluxes and the fundamental microbiological processes in permafrost soils. The microorganisms do not only survive in their extreme habitat but also can be metabolic active under in situ conditions. It was shown that a slight increase of the temperature can lead to a substantial increase in methanogenic activity within perennially frozen deposits. In case of degradation, this would lead to an extensive expansion of the methane deposits with their subsequent impacts on total methane budget. Further studies on the stress response of methanogenic archaea, especially Methanosarcina SMA-21, isolated from Siberian permafrost, revealed an unexpected resistance of the microorganisms against unfavourable living conditions. A better adaptation to environmental stress was observed at 4 °C compared to 28 °C. For the first time it could be demonstrated that methanogenic archaea from terrestrial permafrost even survived simulated Martian conditions. The results show that permafrost methanogens are more resistant than methanogens from non-permafrost environments under Mars-like climate conditions. Microorganisms comparable to methanogens from terrestrial permafrost can be seen as one of the most likely candidates for life on Mars due to their physiological potential and metabolic specificity.
The cytoskeletal motor protein kinesin-1 (conventional kinesin) is the fast carrier for intracellular cargo transport along microtubules. So far most studies aimed at investigating the transport properties of individual motor molecules. However, the transport in cells usually involves the collective work of more than one motor. In the present work, we have studied the movement of beads as artificial loads/organelles pulled by several kinesin-1 motors in vitro. For a wide range of motor coverage of the beads and different bead (cargo) sizes the transport parameters walking distance or run length, velocity and force generation are measured. The results indicate that the transport parameters are influenced by the number of motors carrying the bead. While the transport velocity slightly decreases, an increase in the run length was measured and higher forces are determined, when more motors are involved. The effective number of motors pulling a bead is estimated by measuring the change in the hydrodynamic diameter of kinesin-coated beads using dynamic light scattering. The geometrical constraints imposed by the transport system have been taken into account. Thus, results for beads of different size and motor-surface coverage could be compared. In addition, run length-distributions obtained for the smallest bead size were matched to theoretically calculated distributions. The latter yielded an average number of pulling motors, which is in agreement with the effective motor numbers determined experimentally.
