The aromatic peroxygenase (APO; EC 1.11.2.1) from the agraric basidomycete Marasmius rotula (MroAPO) immobilized at the chitosan-capped gold-nanoparticle-modified glassy carbon electrode displayed a pair of redox peaks with a midpoint potential of -278.5 mV vs. AgCl/AgCl (1 M KCl) for the Fe(2+)/Fe(3+) redox couple of the heme-thiolate-containing protein. MroAPO oxidizes aromatic substrates such as aniline, p-aminophenol, hydroquinone, resorcinol, catechol, and paracetamol by means of hydrogen peroxide. The substrate spectrum overlaps with those of cytochrome P450s and plant peroxidases which are relevant in environmental analysis and drug monitoring. In M. rotula peroxygenase-based enzyme electrodes, the signal is generated by the reduction of electrode-active reaction products (e.g., p-benzoquinone and p-quinoneimine) with electro-enzymatic recycling of the analyte. In these enzyme electrodes, the signal reflects the conversion of all substrates thus representing an overall parameter in complex media. The performance of these sensors and their further development are discussed.
Die „Potsdamer Beiträge zur Sorabistik – Podstupimske psinoski k Sorabistice“ Nr. 9 sind wiederum ein Sammelband mit verschiedenen Aufsätzen. Die Thematik ist in diesem Band „Domownja/Heimat. Sorbische/wendische Perspektiven auf die Lausitz“. Die Schrift ist in drei Themenbereiche unterteilt, einen Tagungsbericht von einem internationalen Kolloquium, das im Februar 2009 an der Brandenburgischen Technischen Universität in Cottbus (BTU) veranstaltet wurde und o.g. Titel trug. Darüber hinaus sind im Heft vier Referate eines zweiten Kolloquiums zu Ehren des niedersorbischen Pfarrers Kito Pank (1808-1895) abgedruckt. Im weiteren finden sich in der Ausgabe unter der Rubrik: Neues aus dem sorbischen/wendischen Leben zwei Aufsätze zu einer Schrifttumsausstellung des Wendischen Museums in Cottbus, ein Umfrageresultat zur Wahrnehmung der Sorben/Wenden in der Stadt Cottbus und ein umfassender Artikel zum sorbischen/wendischen Siedlungsgebiet im Rahmen der Minderheitenpolitik in Brandenburg.

Die Schrift ist dazu in drei Themenbereiche unterteilt, einen Tagungsbericht von einem internationalen Kolloquium, das im November 2007 an der Brandenburgischen Technischen Universität in Cottbus veranstaltet wurde und den Titel „Sind die sorbische/wendische Sprache und Identität noch zu retten“ trug. Darüber hinaus sind im Heft drei Referate des Kolloquiums abgedruckt. Im weiteren finden sich im Heft zwei Aufsätze zur sorbischen/wendischen Musikgeschichte und zwei Artikel zur sorbischen/wendischen Kirchengeschichte. Diesen Artikeln ist eine umfangreiche Statistik der evangelischen Gottesdienste in niedersorbischer Sprache beigefügt, die vorrangig nach der politischen Wende 1989 in der Niederlausitz gehalten wurden.
Low thermal conductivity boulder with high porosity identified on C-type asteroid (162173) Ryugu

C-type asteroids are among the most pristine objects in the Solar System, but little is known about their interior structure and surface properties. Telescopic thermal infrared observations have so far been interpreted in terms of a regolith-covered surface with low thermal conductivity and particle sizes in the centimetre range. This includes observations of C-type asteroid (162173) Ryugu1,2,3. However, on arrival of the Hayabusa2 spacecraft at Ryugu, a regolith cover of sand- to pebble-sized particles was found to be absent4,5 (R.J. et al., manuscript in preparation). Rather, the surface is largely covered by cobbles and boulders, seemingly incompatible with the remote-sensing infrared observations. Here we report on in situ thermal infrared observations of a boulder on the C-type asteroid Ryugu. We found that the boulder’s thermal inertia was much lower than anticipated based on laboratory measurements of meteorites, and that a surface covered by such low-conductivity boulders would be consistent with remote-sensing observations. Our results furthermore indicate high boulder porosities as well as a low tensile strength in the few hundred kilopascal range. The predicted low tensile strength confirms the suspected observational bias6 in our meteorite collections, as such asteroidal material would be too frail to survive atmospheric entry7
Rigorous runtime analysis is a major approach towards understanding evolutionary computing techniques, and in this area linear pseudo-Boolean objective functions play a central role. Having an additional linear constraint is then equivalent to the NP-hard Knapsack problem, certain classes thereof have been studied in recent works. In this article, we present a dynamic model of optimizing linear functions under uniform constraints. Starting from an optimal solution with respect to a given constraint bound, we investigate the runtimes that different evolutionary algorithms need to recompute an optimal solution when the constraint bound changes by a certain amount. The classical (1+1) EA and several population-based algorithms are designed for that purpose, and are shown to recompute efficiently. Furthermore, a variant of the (1+(λ,λ))GA for the dynamic optimization problem is studied, whose performance is better when the change of the constraint bound is small.
Congenital adrenal hyperplasia (CAH) is the most common form of adrenal insufficiency in childhood; it requires cortisol replacement therapy with hydrocortisone (HC, synthetic cortisol) from birth and therapy monitoring for successful treatment. In children, the less invasive dried blood spot (DBS) sampling with whole blood including red blood cells (RBCs) provides an advantageous alternative to plasma sampling.
Potential differences in binding/association processes between plasma and DBS however need to be considered to correctly interpret DBS measurements for therapy monitoring. While capillary DBS samples would be used in clinical practice, venous cortisol DBS samples from children with adrenal insufficiency were analyzed due to data availability and to directly compare and thus understand potential differences between venous DBS and plasma. A previously published HC plasma pharmacokinetic (PK) model was extended by leveraging these DBS concentrations.
In addition to previously characterized binding of cortisol to albumin (linear process) and corticosteroid-binding globulin (CBG; saturable process), DBS data enabled the characterization of a linear cortisol association with RBCs, and thereby providing a quantitative link between DBS and plasma cortisol concentrations. The ratio between the observed cortisol plasma and DBS concentrations varies highly from 2 to 8. Deterministic simulations of the different cortisol binding/association fractions demonstrated that with higher blood cortisol concentrations, saturation of cortisol binding to CBG was observed, leading to an increase in all other cortisol binding fractions.
In conclusion, a mathematical PK model was developed which links DBS measurements to plasma exposure and thus allows for quantitative interpretation of measurements of DBS samples.
