Background/Aims: The tumor suppressor p53 is rarely mutated in gastroenteropancreatic neuroendocrine neoplasms (GEP-NEN) but they frequently show a strong expression of negative regulators of p53, rendering these tumors excellent targets for a p53 recovery therapy. Therefore, we analyzed the mechanisms of a p53 recovery therapy on intestinal neuroendocrine tumors in vitro and in vivo. Methods: By Western blot and immunohistochemistry, we found that in GEP-NEN biopsy material overexpression of MDM2 was present in intestinal NEN. Therefore, we analyzed the effect of a small-molecule inhibitor, nutlin-3a, in p53 wild-type and mutant GEP-NEN cell lines by proliferation assay, flow cytometry, immunofluorescence, Western blot, and by multiplex gene expression analysis. Finally, we analyzed the antitumor effect of nutlin-3a in a xenograft mouse model in vivo. During the study, the tumor volume was determined. Results: The midgut wild-type cell line KRJ-I responded to the treatment with cell cycle arrest and apoptosis. By gene expression analysis, we could demonstrate that nutlins reactivated an antiproliferative p53 response. KRJ-I-derived xenograft tumors showed a significantly decreased tumor growth upon treatment with nutlin-3a in vivo. Furthermore, our data suggest that MDM2 also influences the expression of the oncogene FOXM1 in a p53-independent manner. Subsequently, a combined treatment of nutlin-3a and cisplatin (as chemoresistance model) resulted in synergistically enhanced antiproliferative effects. Conclusion: In summary, MDM2 overexpression is a frequent event in p53 wild-type intestinal neuroendocrine neoplasms and therefore recovery of a p53 response might be a novel personalized treatment approach in these tumors. PSI-Potsdam

An der Universität Potsdam wird seit 2015 im Rahmen der „Qualitätsoffensive Lehrerbildung“ das Projekt „Professionalisierung – Schulpraktische Studien – Inklusion“ (PSI-Potsdam) durchgeführt und am Zentrum für Lehrerbildung und Bildungsforschung (ZeLB) koordiniert. Zur ersten Projektförderphase (2015-2018) erschien der Band „PSI-Potsdam – Ergebnisbericht zu den Aktivitäten im Rahmen der Qualitätsoffensive Lehrerbildung (2015-2018)“ zum Auftakt der Reihe „Potsdamer Beiträge zur Lehrerbildung und Bildungsforschung“.
Der vorliegende Band aus der gleichen Reihe gibt in den Kapiteln „Erhebungen“, „Lehrkonzepte“ und „Vernetzungen“ einen Überblick über alle Teilprojekte der zweiten Projektförderphase (2019-2023). Wissenschaftler:innen aus verschiedenen Fachdidaktiken, Fachwissenschaften sowie aus den Bildungswissenschaften und der Inklusionspädagogik haben im Rahmen des Projektes kooperiert. Sowohl praxisnahe Forschung als auch die Entwicklung neuer Lehrkonzepte sowie Strategien zur Vernetzung innerhalb der Lehrkräftebildung stehen im Fokus dieses Bandes. Die Praxisphasen, die im Rahmen des „Potsdamer Modells der Lehrerbildung“ eine zentrale Rolle spielen, wurden in einer großen Studie über alle Praxisphasen untersucht.
Der Band gibt interessante Einblicke in die Ergebnisse der Teilprojekte und Anregungen sowohl für die eigene Forschung als auch für Entwicklungsarbeit wie zum Beispiel die Entwicklung neuer Lehrkonzepte. Herausgegeben wird dieser Band von PD Dr. Jolanda Hermanns (Gesamtkoordinatorin PSI-Potsdam und Chemiedidaktikerin).
Genisa-Blätter III

Ursprünglich sollten die „Genisa-Blätter III“ ein europäisches Projekt werden. Veröffentlicht werden sollten Quellen aus Genisot, die im vormals deutschsprachigen Raum gehoben wurden. Das Vorhaben scheiterte. Nunmehr sind im Band sieben Beiträge versammelt, in denen kenntnisreich Funde aus den Ablagen in Alsenz, Altenschönbach, Memmelsdorf, Reckendorf und Veitshöchheim besprochen werden. Der achte Aufsatz knüpft am internationalen Vorhaben an: Im Mittelpunkt des Artikels stehen drei Genisafunde aus Tschechien, genauer gesagt aus den beiden ostböhmischen Ortschaften Luže und Rychnov nad Kněžnou. Alle acht Beiträge gemeinsam verdeutlichen die inhaltliche Spannbreite von Genisot, die sich die Forschung zunutze machen kann: Sie reicht von Aspekten der Wirtschaft und des Rechts, über die Religion bis hin zum alltäglichen Leben und bietet jeweils eigene Geschichten im Spannungsfeld des möglichen Scheiterns.
