Molecular photoswitches are attracting much attention lately mostly because of their possible applications in nano technology, and their role in biology. One of the widely studied representatives of photochromic molecules is azobenzene (AB). With light, by a static electric field, or with tunneling electrons this specie can be "switched" from the flat and energetically more stable trans form, into the compact cis form. The back reaction can be induced optically or thermally. Quantum chemical calculations, mostly based on density functional theory, on the AB molecule, AB derivatives and related systems are presented. All the calculations were done for isolated species, however, with implications for latest experimental results aiming at the switching of surface mounted ABs. In some of these experiments, it is assumed that the switching process is substrate mediated, by attaching an electron or a hole to the adsorbate forming short-lived anion or cation resonances. Therefore, we calculated also cationic and anionic ABs in this work. An influence of external electric fields on the potential energy surfaces, was also studied. Further, by the type, number and positioning of various substituent groups, systematic changes on activation energies and rates for the thermal cis-to-trans isomerization can be enforced. The nature of the transition state for ground state isomerization was investigated. Applying Eyring's transition state theory, trends in activation energies and rates were predicted and are, where a comparison was possible, in good agreement with experimental data. Further, thermal isomerization was studied in solution, for which a polarizable continuum model was employed. The influence of substitution and an environment leaves its traces on structural properties of molecules and quantitative appearance of calculated UV/Vis spectra, as well. Finally, an explicit treatment of a solid substrate was demonstrated for the conformational switching, by scanning tunneling microscope, of a 1,5-cyclooctadiene (COD) molecule at a Si(001) surface, treated by a cluster model. At first, we studied energetics and potential energy surfaces along relevant switching coordinates by quantum chemical calculations, followed by the switching dynamics using wave packet methods. We show that, in spite the simplicity of the model, our calculations support the switching of adsorbed COD, by inelastic electron tunneling at low temperatures.
Nowadays, the need to protect the environment becomes more urgent than ever. In the field of chemistry, this translates to practices such as waste prevention, use of renewable feedstocks, and catalysis; concepts based on the principles of green chemistry. Polymers are an important product in the chemical industry and are also in the focus of these changes. In this thesis, more sustainable approaches to make two classes of polymers, polypeptoids and polyesters, are described.
Polypeptoids or poly(alkyl-N-glycines) are isomers of polypeptides and are biocompatible, as well as degradable under biologically relevant conditions. In addition to that, they can have interesting properties such as lower critical solution temperature (LCST) behavior. They are usually synthesized by the ring opening polymerization (ROP) of N-carboxy anhydrides (NCAs), which are produced with the use of toxic compounds (e.g. phosgene) and which are highly sensitive to humidity. In order to avoid the direct synthesis and isolation of the NCAs, N-phenoxycarbonyl-protected N-substituted glycines are prepared, which can yield the NCAs in situ. The conditions for the NCA synthesis and its direct polymerization are investigated and optimized for the simplest N-substituted glycine, sarcosine. The use of a tertiary amine in less than stoichiometric amounts compared to the N-phenoxycarbonyl--sarcosine seems to accelerate drastically the NCA formation and does not affect the efficiency of the polymerization. In fact, well defined polysarcosines that comply to the monomer to initiator ratio can be produced by this method. This approach was also applied to other N-substituted glycines.
Dihydroxyacetone is a sustainable diol produced from glycerol, and has already been used for the synthesis of polycarbonates. Here, it was used as a comonomer for the synthesis of polyesters. However, the polymerization of dihydroxyacetone presented difficulties, probably due to the insolubility of the macromolecular chains. To circumvent the problem, the dimethyl acetal protected dihydroxyacetone was polymerized with terephthaloyl chloride to yield a soluble polymer. When the carbonyl was recovered after deprotection, the product was insoluble in all solvents, showing that the carbonyl in the main chain hinders the dissolution of the polymers. The solubility issue can be avoided, when a 1:1 mixture of dihydroxyacetone/ ethylene glycol is used to yield a soluble copolyester.
Ionizing radiation is used in cancer radiation therapy to effectively damage the DNA of tumors leading to cell death and reduction of the tumor tissue. The main damage is due to generation of highly reactive secondary species such as low-energy electrons (LEE) with the most probable energy around 10 eV through ionization of water molecules in the cells. A simulation of the dose distribution in the patient is required to optimize the irradiation modality in cancer radiation therapy, which must be based on the fundamental physical processes of high-energy radiation with the tissue. In the present work the accurate quantification of DNA radiation damage in the form of absolute cross sections for LEE-induced DNA strand breaks (SBs) between 5 and 20 eV is done by using the DNA origami technique. This method is based on the analysis of well-defined DNA target sequences attached to DNA origami triangles with atomic force microscopy (AFM) on the single molecule level. The present work focuses on poly-adenine sequences (5'-d(A4), 5'-d(A8), 5'-d(A12), 5'-d(A16), and 5'- d(A20)) irradiated with 5.0, 7.0, 8.4, and 10 eV electrons. Independent of the DNA length, the strand break cross section shows a maximum around 7.0 eV electron energy for all investigated oligonucleotides confirming that strand breakage occurs through the initial formation of negative ion resonances. Additionally, DNA double strand breaks from a DNA hairpin 5'-d(CAC)4T(Bt-dT)T2(GTG)4 are examined for the first time and are compared with those of DNA single strands 5'-d(CAC)4 and 5'- d(GTG)4. The irradiation is made in the most likely energy range of 5 to 20 eV with an anionic resonance maximum around 10 eV independently of the DNA sequence. There is a clear difference between σSSB and σDSB of DNA single and double strands, where the strand break for ssDNA are always higher in all electron energies compared to dsDNA by the factor 3. A further part of this work deals with the characterization and analysis of new types of radiosensitizers used in chemoradiotherapy, which selectively increases the DNA damage upon radiation. Fluorinated DNA sequences with 2'-fluoro-2'-deoxycytidine (dFC) show an increased sensitivity at 7 and 10 eV compared to the unmodified DNA sequences by an enhancement factor between 2.1 and 2.5. In addition, light-induced oxidative damage of 5'-d(GTG)4 and 5'-d((CAC)4T(Bt-dT)T2(GTG)4) modified DNA origami triangles by singlet oxygen 1O2 generated from three photoexcited DNA groove binders [ANT994], [ANT1083] and [Cr(ddpd)2][BF4]3 illuminated in different experiments with UV-Vis light at 430, 435 and 530 nm wavelength is demonstrated. The singlet oxygen induced generation of DNA damage could be detected in both aqueous and dry environments for [ANT1083] and [Cr(ddpd)2][BF4]3.
Die vorgelegte Dissertation präsentiert wissenschaftliche Ergebnisse, die in der Zeit vom Dezember 2012 bis August 2016, erarbeitet wurden. Der zentrale Inhalt der Arbeit ist die Simulation von Röntgenabsorptionsprozessen von verschiedenen Systemen in kondensierter Phase. Genauer gesagt, werden Nahkantenabsorptions- (NEXAFS) sowie Röntgenphotoelektronenspektren (XPS) berechnet. In beiden Fällen wird ein Röntgenphoton von einem molekularen System absorbiert. Aufgrund der hohen Photonenenergie wird ein stark gebundenes kernnahes Elektron angeregt. Bei der XPS gelangt dieses mit einer zu messenden kinetischen Energie in Kontinuumszustände. In Abhängigkeit der eingestrahlten Photonenenergie und der kinetischen Energie des austreten Elektrons, kann die Bindungsenergie berechnet werden, welche die zentrale Größe der XPS ist. Im Falle der NEXAFS-Spektroskopie wird das kernnahe Elektron in unbesetzte gebundene Zustände angeregt. Die zentrale Größe ist die Absorption als Funktion der eingestrahlten Photonenenergie. Das erste Kapitel meiner Arbeit erörtert detailliert die experimentellen Methoden sowie die daraus gewonnenen charakteristischen Größen.
Die experimentellen Spektren zeigen oft viele Resonanzen, deren Interpretation aufgrund fehlender Referenzmaterialien schwierig ist. In solchen Fällen bietet es sich an, die Spektren mittels quantenchemischer Methoden zu simulieren. Der dafür erforderliche mathematisch-physikalische Methodenkatalog wird im zweiten Kapitel der Arbeit erörtert.
Das erste von mir untersuchte System ist Graphen. In experimentellen Arbeiten wurde die Oberfläche mittels Bromplasma modifiziert. Die im Anschluss gemessenen NEXAFS-Spektren unterscheiden sich maßgeblich von den Spektren der unbehandelten Oberfläche. Mithilfe periodischer DFT-Rechnungen wurden verschiedene Gitterdefekte sowie bromierte Systeme untersucht und die NEXAFS-Spektren simuliert. Mittels der Simulationen können die Beiträge verschiedener Anregungszentren analysiert werden. Die Berechnungen erlauben den Schluss, dass Gitterdefekte maßgeblich für die entstandenen Veränderungen verantwortlich sind.
Polyvinylalkohol (PVA) wurde als zweites System behandelt. Hierbei sollte untersucht werden, wie groß der Einfluss der Molekularbewegung auf die Verbreiterung der Peaks im XP-Spektrum ist. Des Weiteren wurde untersucht, wie groß der Einfluss von intermolekularen Wechselwirkungen auf die Peakpositionen und Peakverbreiterung ist. Für die Berechnung dieses Systems wurde eine Kombination aus molekulardynamischen und quantenchemischen Methoden verwendet. Als Strukturen dienten Oligomermodelle, die unter dem Einfluss eines (ab initio) Potentials propagiert wurden. Entlang der erstellten Trajektorie wurden Schnappschüsse der Geometrien extrahiert und für die Berechnung der XP-Spektren verwendet. Die Spektren werden bereits mithilfe klassischer Molekulardynamik sehr gut reproduziert. Die erhaltenen Peakbreiten sind verglichen mit dem Experiment allerdings zu klein. Die Hauptursache der Peakverbreiterung ist die Molekularbewegung. Intermolekulare Wechselwirkungen verschieben die Peakpositionen um 0.6 eV zu kleineren Anregungsenergien.
Im dritten Teil der Arbeit stehen die NEXAFS-Spektren von ionischen Flüssigkeiten (ILs) im Fokus. Die experimentell gefundenen Spektren zeigen eine komplexe Struktur mit vielen Resonanzen. In der Arbeit wurden zwei ILs untersucht. Als Geometrien verwenden wir Clustermodelle, die aus experimentellen Kristallstrukturen extrahiert wurden. Die berechneten Spektren erlauben es, die Resonanzen den Anregungszentren zuzuordnen. Außerdem kann eine erstmals gemessene Doppelresonanz simuliert und erklärt werden. Insgesamt kann die Interpretation der Spektren mithilfe der Simulation signifikant erweitert werden.
In allen Systemen wurde zur Berechnung des NEXAFS-Spektrums eine auf Dichtefunktionaltheorie basierende Methode verwendet (die sogenannte Transition-Potential Methode). Gängige wellenfunktionsbasierte Methoden, wie die Konfigurationswechselwirkung mit Einfachanregungen (CIS), zeigen eine starke Blauverschiebung, wenn als Referenz eine Hartree-Fock Slaterdeterminante verwendet wird. Wir zeigen, dass die Verwendung von kernnah-angeregten Determinanten sowohl das resultierende Spektrum als auch die Anregungsenergien deutlich verbessert. Des Weiteren werden auch Referenzen aus Dichtefunktionalrechnungen getestet. Zusätzlich werden auch Referenzen mit gebrochenen Besetzungszahlen für kernnahe Elektronen verwendet. In der Arbeit werden die Resultate der verschiedenen Referenzen miteinander verglichen. Es zeigt sich, dass Referenzen mit gebrochenen Besetzungszahlen das Spektrum nicht weiter verbessern. Der Einfluss der verwendeten Elektronenstrukturmethode ist eher gering.
In dieser Arbeit werden drei Themen im Zusammenhang mit den spektroskopischen Eigenschaften von Cumarin- (Cou) und DBD-Farbstoffen ([1,3]Dioxolo[4,5-f][1,3]benzodioxol) behandelt. Der erste Teil zeigt die grundlegende spektroskopische Charakterisierung von 7-Aminocumarinen und ihre potentielle Anwendung als Fluoreszenzsonde für Fluoreszenzimmunassays. Im zweiten Teil werden mit die photophysikalischen Eigenschaften der Cumarine genutzt um Cou- und DBD-funktionalisierte Oligo-Spiro-Ketal-Stäbe (OSTK) und ihre Eigenschaften als Membransonden zu untersuchen. Der letzte Teil beschäftigt sich mit der Synthese und der Charakterisierung von Cou- und DBD-funktionalisierten Polyprolinen als Referenzsysteme für schwefelfunktionalisierte OSTK-Stäbe und ihrer Kopplung an Goldnanopartikel.
Immunochemische Analysemethoden sind in der klinischen Diagnostik sehr erfolgreich und werden heute auch für die Nahrungsmittelkontrolle und Überwachung von Umweltfragen mit einbezogen. Dadurch sind sie von großem Interesse für weitere Forschungen. Unter den verschiedenen Immunassays zeichnen sich lumineszenzbasierte Formate durch ihre herausragende Sensitivität aus, die dieses Format für zukünftige Anwendungen besonders attraktiv macht. Die Notwendigkeit von Multiparameterdetektionsmöglichkeiten erfordert einen Werkzeugkasten mit Farbstoffen, um die biochemische Reaktion in ein optisch detektierbares Signal umzuwandeln. Hier wird bei einem Multiparameteransatz jeder Analyt durch einen anderen Farbstoff mit einer einzigartigen Emissionsfarbe, die den blauen bis roten Spektralbereich abdecken, oder eine einzigartige Abklingzeit detektiert. Im Falle eines kompetitiven Immunassayformats wäre für jeden der verschiedenen Farbstoffe ein einzelner Antikörper erforderlich. In der vorliegenden Arbeit wird ein leicht modifizierter Ansatz unter Verwendung einer Cumarineinheit, gegen die hochspezifische monoklonale Antikörper (mAb) erzeugt wurden, als grundlegendes Antigen präsentiert. Durch eine Modifikation der Stammcumarineinheit an einer Position des Moleküls, die für die Erkennung durch den Antikörper nicht relevant ist, kann auf den vollen Spektralbereich von blau bis tiefrot zugegriffen werden. In dieser Arbeit wird die photophysikalische Charakterisierung der verschiedenen Cumarinderivate und ihrer entsprechenden Immunkomplexe mit zwei verschiedenen, aber dennoch hochspezifischen, Antikörpern präsentiert. Die Cumarinfarbstoffe und ihre Immunkomplexe wurden durch stationäre und zeitaufgelöste Absorptions- sowie Fluoreszenzemissionsspektroskopie charakterisiert. Darüber hinaus wurden Fluoreszenzdepolarisationsmessungen durchgeführt, um die Daten zu vervollständigen, die die verschiedenen Bindungsmodi der beiden Antikörper betonten. Im Gegensatz zu häufig eingesetzten Nachweissystemen wurde eine massive Fluoreszenzverstärkung bei der Bildung des Antikörper-Farbstoffkomplexes bis zu einem Faktor von 50 gefunden. Wegen der leichten Emissionsfarbenänderung durch das Anpassen der Cumarinsubstitution in der für die Antigenbindung nicht relevanten Position des Elternmoleküls, ist eine Farbstoff-Toolbox vorhanden, die bei der Konstruktion von kompetitiven Multiparameterfluoreszenzverstärkungsimmunassays verwendet werden kann.
Oligo-Spiro-Thio-Ketal-Stäbe werden aufgrund ihres hydrophoben Rückgrats leicht in Doppellipidschichten eingebaut und deshalb als optische Membransonde verwendet. Wegen ihres geringen Durchmessers wird nur eine minimale Störung der Doppellipidschicht verursacht. Durch die Markierung mit Fluoreszenzfarbstoffen sind neuartige Förster-Resonanz-Energietransfersonden mit hoch definierten relativen Orientierungen der Übergangsdipolmomente der Donor- und Akzeptorfarbstoffe zugänglich und macht die Klasse der OSTK-Sonden zu einem leistungsstarken, flexiblen Werkzeugkasten für optische Biosensoranwendungen. Mit Hilfe von stationären und zeitaufgelösten Fluoreszenzexperimenten wurde der Einbau von Cumarin- und DBD markierten OSTK-Stäben in großen unilamellaren Vesikeln untersucht und die Ergebnisse durch Fluoreszenzdepolarisationsmessungen untermauert.
Der letzte Teil dieser Arbeit beschäftigt sich mit der Synthese und Charakterisierung von Cou- und DBD-funktionalisierten Polyprolinen und ihrer Kopplung an Goldnanopartikel. Die farbstoffmarkierten Polyproline konnten erfolgreich hergestellt werden. Es zeigten sich deutlich Einflüsse auf die spektroskopischen Eigenschaften der Farbstoffe durch die Bindung an die Polyprolinhelix. Die Kopplung an die 5 nm großen AuNP konnte erfolgreich durchgeführt werden. Die Erfahrungen, die durch die Kopplung der Polyproline an die AuNP, gewonnen wurde, ist die Basis für eine Einzelmolekül-AFM-FRET-Nanoskopie mit OSTK-Stäben.
Ziel dieser Arbeit war die Synthese und Charakterisierung von neuartigen fluoreszierenden Copolymeren zur Analytdetektion in wässrigen Systemen. Das Detektionssystem sollte ein einfaches Schalten der Fluoreszenz bei Analytbindung „Aus“ bzw. Verdrängung „An“ ermöglichen. Dafür wurde die Synthese eines funktionalisierten Monomers so geplant, dass sich Fluorophor und Analyt innerhalb derselben Monomereinheit in direkter Nachbarschaft zueinander befinden. So sollten bei Erkennung des Analyten durch eine mit einem Fluoreszenzlöscher funktionalisierte Erkennungsstruktur Fluorophor und Löscher in einen vorgegebenen Abstand zueinander gezwungen und die Fluoreszenz des Fluorophors effizient gelöscht werden. Bei anschließender Verdrängung der Erkennungseinheit durch einen stärker bindenden Analyten sollte die Fluoreszenz wieder „angeschaltet“ werden. Eine weitere Zielstellung für das Detektionssystem war eine hohe Löslichkeit und Fluoreszenzintensität in Wasser. Da die Anwendung solcher Sensoren besonders in der Medizin und Biologie, z.B. für Schnellerkennungstest von Pathogenen, von Interesse ist, ist die Kompatibilität mit wässrigen Medien essentiell. Die funktionalisierten Monomere wurden frei radikalisch mit N Vinyl-pyrrolidon bzw. N Vinyl¬caprolactam zu wasserlöslichen, fluoreszierenden Copolymeren umgesetzt. In den N-Vinyl¬pyrrolidon-Polymeren (PNVP) wurde RhodaminB, in den thermoresponsiven N Vinyl¬caprolactam-Polymeren (PNVCL) ein Naphthalsäureimid als Fluorophor verwendet. Während Rhodamine eine hohe Fluoreszenzintensität, gute Quantenausbeuten und hohen Extinktionskoeffizienten in Wasser zeigen, sind Naphthalsäure¬imide umgebungssensitive Chromophore, die bei Änderung ihrer Lösungsmittelumgebung, wie z.B. beim Kollaps eines thermoresponsiven Polymers in Wasser, ihre Fluoreszenzintensität und Quantenausbeute drastisch ändern können. Der Vorteil der hier verwendeten Strategie der Monomersynthese liegt darin, dass bei jeder spezifischen Analytdetektion durch eine Erkennungseinheit die Fluoreszenz effizient gelöscht bzw. bei Verdrängung durch einen stärker bindenden Analyten wieder „angeschaltet“ wird. Dieses Prinzip wird bereits vielfach in der Biologie in sogenannten „Molecular Beacons“ ausgenutzt, wobei ein Fluorophor und ein Löscher durch spezifische DNA Basenpaarung in einen vorgegebenen Abstand zueinander gezwungen werden und so ein „Schalten“ der Fluoreszenz ermöglichen. Aufgrund der vorgegebenen Struktur der DNA Basensequenzen ist es jedoch nicht direkt auf andere Erkennungsreaktionen übertragbar. Daher wurde ein Modellsystem entwickelt, welches die Möglichkeit bietet Analyt, Erkennungseinheit und Signalgeber variabel, je nach Anforderungen des Systems, auszutauschen. So soll es möglich sein, den Sensor a priori für jede Erkennungs¬reaktion zu verwenden. Als Modell Bindungs¬paare wurden ß Cyclodextrin/Adamantan und Con¬cana¬valinA/Mannose ausgewählt. Adamantan bzw. Mannose wurde als Analyt zusammen mit dem Fluorophor in das Polymer eingebunden. ß Cyclo¬dextrin (ß CD) bzw. ConcanavalinA (ConA) wurde als Erkennungsstruktur an einem Fluoreszenzlöscher immobilisiert. Polymer-basierte Fluoreszenzsensoren sind in der Fachliteratur gut dokumentiert. In der Regel sind Signalgeber und Analyt jedoch statistisch im Polymer verteilt, da sie sich entweder in unterschiedlichen Monomereinheiten befinden oder die Funktionalisierung durch eine polymeranaloge Umsetzung erfolgt. Der gewählte Ansatz Fluorophor und Analyt innerhalb derselben Monomereinheit einzubinden, soll bei jeder Erkennungsreaktion des Analyten zu einer Änderung der Signalintensität des Fluorophors führen. Eine hohe Signalintensität bei Analytdetektion ist wünschenswert, insbesondere für Erkennungsreaktionen, die mit möglichst geringem apparativem Aufwand, am besten mit dem bloßen Auge zu verfolgen sein sollen. Des Weiteren ist es möglich den Fluorophorgehalt im Polymer genau einzustellen und so Selbstlöschung zu vermeiden. Die synthetisierten Polymere haben einen Fluorophorgehalt von 0,01 mol% bis 0,5 mol%. Für die RhodaminB haltigen Polymere zeigte sich, dass ein Fluorophorgehalt unterhalb 0,1 mol% im Polymer die höchsten Ausbeuten, Molmassen und Quantenausbeuten liefert. Für die Naphthalsäureimid haltigen Polymere hingegen wurden auch für einen Fluorophorgehalt von bis zu 1 mol% hohe Ausbeuten und Molmassen erreicht. Die Naphthalsäureimid haltigen Polymere haben jedoch in wässriger Lösungsmittelumgebung nur geringe Quantenausbeuten. Als Fluoreszenzlöscher wurden Goldnanopartikel synthetisiert, die mit den entsprechenden Erkennungsstrukturen (ß-CD oder ConA) für den verwendeten Analyten funktionalisiert wurden. Goldnanopartikel als Löscher bieten den Vorteil, dass ihre Dispergierbarkeit in einem Lösemittel durch Funktionalisierung ihrer Hülle gezielt gesteuert werden kann. Durch die hohe Affinität von Goldnanopartikeln zu Thiolen und Aminen konnten sie mit Hilfe einfacher Syntheseschritte mit Thio ß CD Derivaten bzw. ConA funktionalisiert werden. In der hier vorgelegten Arbeit sollte ein Modellsystem für einen solches fluoreszenz-basiertes Detektionssystem in Wasser entwickelt werden. Nachfolgend werden die zu erfüllenden strukturellen Voraussetzungen für die Synthese eines solchen Sensors nochmals zusammengefasst:
1. Verwendung eines Fluorophors, der eine hohe Signalintensität zeigt.
2. Analyt bzw. Erkennungseinheit soll sich im Abstand von wenigen Nanometern zum Signalgeber befinden, um bei jeder Detektionsreaktion die Signalintensität des Signalgebers beeinflussen zu können.
3. Die Detektionseinheit benötigt eine funktionelle Gruppe zur Immobilisierung. Immobilisierung kann z.B. durch Einbindung in ein Polymer erfolgen.
4. Der Fluorophor soll bei Änderung seiner lokalen Umgebung, durch Binden eines Löschers oder Änderung seiner Lösemittelumgebung seine Fluoreszenzeigenschaften drastisch ändern.
5. Die Reaktion sollte schnell und mit möglichst geringem apparativem Aufwand, am besten mit bloßem Auge zu verfolgen sein.
Für das ß-CD/Adamantan Modellsystem wurde ein Fluoreszenz Aus/An Sensor entwickelt, der bei Binden ß CD funktionalisierter Goldnanopartikel an das polymergebundene Adamantan die Fluoreszenz des RhodaminB Fluorophors effizient löscht und bei Verdrängung der Goldnanopartikel wieder zurück gewinnt. Dies konnte auch mit bloßem Auge verfolgt werden.
Für die Naphthalsäureimid Monomere, die mit NVCL copolymerisiert wurden, wurde abhängig von der lokalen Umgebung des Fluorophors eine unterschiedliche Verstärkung der Fluoreszenzintensität bei Überschreiten des Trübungspunktes des Polymers gefunden. Dabei zeigte sich, dass die Einführung eines Abstandshalters zwischen Polymerrückgrat und Fluorophor zu einer großen Fluoreszenz¬verstärkung führt, während sich ohne Abstandshalter die Fluoreszenzintensität bei Über¬schreiten des Trübungspunktes kaum ändert.
Ellipsometrische Lichtstreuung als neue Methode zur Charakterisierung der Grenzfläche von Kolloiden

Die ellipsometrische Lichtstreuung wird als eine neue, leistungsfähige Methode zur Charakterisierung von Schichten um kolloidale Partikel vorgestellt. Theoretische Grundlage der Methode ist die Mie-Theorie der Lichtstreuung. Experimentell wurde die Polarisationsoptik eines Null-Ellipsometers in den Strahlengang eines Lichtstreuaufbaus eingebaut. Wie in der Reflexionsellipsometrie um den Brewsterwinkel herum erhält man in der ellipsometrischen Streuung einen Winkelbereich, in dem die Methode empfindlich auf Schichten an der Oberfläche der Partikel ist. An verschiedenen Systemen wurde die Tauglichkeit der ellipsometrischen Streuung zur Charakterisierung von Schichten auf Partikeln demonstriert. So wurden Dicke und Brechungsindex einer thermosensitiven Schicht von Poly(N-isopropylacrylamid) auf einem Poly(methylmethacrylat)-Kern bestimmt. Damit ist es möglich, experimentell den Schichtbrechungsindex und damit den Quellungsgrad zu bestimmen. Des Weiteren wurde der Einfluss der NaCl-Konzentration auf die Polyelektrolythülle von Poly(methylmethacrylat)-Poly(styrolsulfonat)-Blockcopolymer-Partikeln untersucht. Die Polyelektrolytketten liegen im hier untersuchten Beispiel nicht gestreckt vor. Als drittes wurde die Verteilung von niedermolekularen Ionen um elektrostatisch stabilisierte Poly(styrol)-Latexpartikel in Wasser untersucht. Hier wurde gezeigt, dass die beobachteten Schichtdicken und Schichtbrechungsindizes viel größer sind, als nach der klassischen Poisson-Boltzmann-Theorie zu erwarten ist. Des Weiteren wurde die Doppelbrechung von unilamellaren Lipidvesikeln bestimmt. Außerdem wurden Messungen der dynamische Lichtstreuung im Intensitätsminimum der Ellipsometrie durchgeführt. Dabei wird ein Prozess mit einer Korrelationszeit, die unabhängig vom Streuvektor, aber abhängig von der verwendeten Wellenlänge ist, sichtbar. Die Natur dieses Prozesses konnte hier nicht vollständig geklärt werden.
The reliance on fossil fuels has resulted in an abnormal increase in the concentration of greenhouse gases, contributing to the global climate crisis. In response, a rapid transition to renewable energy sources has begun, particularly lithium-ion batteries, playing a crucial role in the green energy transformation. However, concerns regarding the availability and geopolitical implications of lithium have prompted the exploration of alternative rechargeable battery systems, such as sodium-ion batteries. Sodium is significantly abundant and more homogeneously distributed in the crust and seawater, making it easier and less expensive to extract than lithium. However, because of the mysterious nature of its components, sodium-ion batteries are not yet sufficiently advanced to take the place of lithium-ion batteries. Specifically, sodium exhibits a more metallic character and a larger ionic radius, resulting in a different ion storage mechanism utilized in lithium-ion batteries. Innovations in synthetic methods, post-treatments, and interface engineering clearly demonstrate the significance of developing high-performance carbonaceous anode materials for sodium-ion batteries. The objective of this dissertation is to present a systematic approach for fabricating efficient, high-performance, and sustainable carbonaceous anode materials for sodium-ion batteries. This will involve a comprehensive investigation of different chemical environments and post-modification techniques as well.
This dissertation focuses on three main objectives. Firstly, it explores the significance of post-synthetic methods in designing interfaces. A conformal carbon nitride coating is deposited through chemical vapor deposition on a carbon electrode as an artificial solid-electrolyte interface layer, resulting in improved electrochemical performance. The interaction between the carbon nitride artificial interface and the carbon electrode enhances initial Coulombic efficiency, rate performance, and total capacity. Secondly, a novel process for preparing sulfur-rich carbon as a high-performing anode material for sodium-ion batteries is presented. The method involves using an oligo-3,4-ethylenedioxythiophene precursor for high sulfur content hard carbon anode to investigate the sulfur heteroatom effect on the electrochemical sodium storage mechanism. By optimizing the condensation temperature, a significant transformation in the materials’ nanostructure is achieved, leading to improved electrochemical performance. The use of in-operando small-angle X-ray scattering provides valuable insights into the interaction between micropores and sodium ions during the electrochemical processes. Lastly, the development of high-capacity hard carbon, derived from 5-hydroxymethyl furfural, is examined. This carbon material exhibits exceptional performance at both low and high current densities. Extensive electrochemical and physicochemical characterizations shed light on the sodium storage mechanism concerning the chemical environment, establishing the material’s stability and potential applications in sodium-ion batteries.
Die Entwicklung nachhaltiger Bewirtschaftungs- und Produktionsmethoden ist eine der zentralen Fragestellungen der modernen Agrarwirtschaft. Die vorliegende Arbeit beschäftigt sich mit zwei Forschungsthemen, die das Konzept Nachhaltigkeit beinhalten. In beiden Fällen werden analytische Grundlagen für die Entwicklung entsprechender landwirtschaftlicher Arbeitsmethoden gelegt.
Das erste Thema ist an den sogenannten Präzisionsackerbau angelehnt. Bei diesem wird die Bearbeitung von Agrarflächen ortsabhängig ausgeführt. Das heißt, die Ausbringung von Saatgut, Dünger, Bewässerung usw. richtet sich nach den Eigenschaften des jeweiligen Standortes und wird nicht pauschal gleichmäßig über ein ganzes Feld verteilt. Voraussetzung hierfür ist eine genaue Kenntnis der Bodeneigenschaften. In der vorliegenden Arbeit sollten diese Parameter mittels der analytischen Technik der Laser-induzierten Breakdown Spektroskopie (LIBS), die eine Form der Elementaranalyse darstellt, bestimmt werden. Bei den hier gesuchten Bodeneigenschaften handelte es sich um die Gehalte von Nährstoffen sowie einige sekundäre Parameter wie den Humusanteil, den pH-Wert und den pflanzenverfügbaren Anteil einzelner Nährstoffe. Diese Eigenschaften wurden durch etablierte Referenzanalysen bestimmt. Darauf aufbauend wurden die Messergebnissen der LIBS-Untersuchungen durch verschiedene Methoden der sogenannten multivariaten Datenanalyse (MVA) ausgewertet. Daraus sollten Modelle zur Vorhersage der Bodenparameter in zukünftigen LIBS-Messungen erarbeitet werden. Die Ergebnisse dieser Arbeit zeigten, dass mit der Kombination von LIBS und MVA sämtliche Bodenparameter erfolgreich vorhergesagt werden konnten. Dies beinhaltete sowohl die tatsächlich messbaren Elemente als auch die sekundären Eigenschaften, welche durch die MVA mit den Elementgehalten in Zusammenhang gebracht wurden.
Das zweite Thema beschäftigt sich mit der Vermeidung von Verlusten durch Schädlingsbefall bei der Getreidelagerung. Hier sollten mittels der Ionenmobilitätsspektrometrie (IMS) Schimmelpilzkontaminationen detektiert werden. Dabei wurde nach den flüchtigen Stoffwechselprodukten der Pilze gesucht. Die durch Referenzmessungen mit Massenspektrometern identifizierten Substanzen konnten durch IMS im Gasvolumen über den Proben, dem sogenannten Headspace, nachgewiesen werden. Dabei wurde nicht nur die Anwesenheit einer Kontamination festgestellt, sondern diese auch charakterisiert. Die freigesetzten Substanzen bildeten spezifische Muster, anhand derer die Pilze identifiziert werden konnten. Hier wurden sowohl verschiedene Gattungen als auch einzelne Arten unterschieden. Die Messungen fanden auf verschiedenen Nährböden statt um den Einfluss dieser auf die Stoffwechselprodukte zu beobachten. Auch die sekundären Stoffwechselprodukte der Schimmelpilze, die Mykotoxine, konnten durch IMS detektiert werden.
Beide in dieser Arbeit vorgestellten Forschungsthemen konnten erfolgreich abgeschlossen werden. Sowohl LIBS als auch IMS erwiesen sich für den Nachweis der jeweiligen Analyten als geeignet, und der Einsatz moderner computergestützter Auswertemethoden ermöglichte die genaue Charakterisierung der gesuchten Parameter. Beide Techniken können in Form von mobilen Geräten verwendet werden und zeichnen sich durch eine schnelle und sichere Analyse aus. In Kombination mit entsprechenden Modellen der MVA sind damit alle Voraussetzungen für Vor-Ort-Untersuchungen und damit für den Einsatz in der Landwirtschaft erfüllt.
Wood is used for many applications because of its excellent mechanical properties, relative abundance and as it is a renewable resource. However, its wider utilization as an engineering material is limited because it swells and shrinks upon moisture changes and is susceptible to degradation by microorganisms and/or insects. Chemical modifications of wood have been shown to improve dimensional stability, water repellence and/or durability, thus increasing potential service-life of wood materials. However current treatments are limited because it is difficult to introduce and fix such modifications deep inside the tissue and cell wall. Within the scope of this thesis, novel chemical modification methods of wood cell walls were developed to improve both dimensional stability and water repellence of wood material. These methods were partly inspired by the heartwood formation in living trees, a process, that for some species results in an insertion of hydrophobic chemical substances into the cell walls of already dead wood cells, In the first part of this thesis a chemistry to modify wood cell walls was used, which was inspired by the natural process of heartwood formation. Commercially available hydrophobic flavonoid molecules were effectively inserted in the cell walls of spruce, a softwood species with low natural durability, after a tosylation treatment to obtain “artificial heartwood”. Flavonoid inserted cell walls show a reduced moisture absorption, resulting in better dimensional stability, water repellency and increased hardness. This approach was quite different compared to established modifications which mainly address hydroxyl groups of cell wall polymers with hydrophilic substances. In the second part of the work in-situ styrene polymerization inside the tosylated cell walls was studied. It is known that there is a weak adhesion between hydrophobic polymers and hydrophilic cell wall components. The hydrophobic styrene monomers were inserted into the tosylated wood cell walls for further polymerization to form polystyrene in the cell walls, which increased the dimensional stability of the bulk wood material and reduced water uptake of the cell walls considerably when compared to controls. In the third part of the work, grafting of another hydrophobic and also biodegradable polymer, poly(ɛ-caprolactone) in the wood cell walls by ring opening polymerization of ɛ-caprolactone was studied at mild temperatures. Results indicated that polycaprolactone attached into the cell walls, caused permanent swelling of the cell walls up to 5%. Dimensional stability of the bulk wood material increased 40% and water absorption reduced more than 35%. A fully biodegradable and hydrophobized wood material was obtained with this method which reduces disposal problem of the modified wood materials and has improved properties to extend the material’s service-life. Starting from a bio-inspired approach which showed great promise as an alternative to standard cell wall modifications we showed the possibility of inserting hydrophobic molecules in the cell walls and supported this fact with in-situ styrene and ɛ-caprolactone polymerization into the cell walls. It was shown in this thesis that despite the extensive knowledge and long history of using wood as a material there is still room for novel chemical modifications which could have a high impact on improving wood properties.
The urge of light utilization in fabrication of materials is as encouraging as challenging. Steadily increasing energy consumption in accordance with rapid population growth, is requiring a corresponding solution within the same rate of occurrence speed. Therefore, creating, designing and manufacturing materials that can interact with light and in further be applicable as well as disposable in photo-based applications are very much under attention of researchers. In the era of sustainability for renewable energy systems, semiconductor-based photoactive materials have received great attention not only based on solar and/or hydrocarbon fuels generation from solar energy, but also successful stimulation of photocatalytic reactions such as water splitting, pollutant degradation and organic molecule synthesisThe turning point had been reached for water splitting with an electrochemical cell consisting of TiO2-Pt electrode illuminated by UV light as energy source rather than an external voltage, that successfully pursued water photolysis by Fujishima and Honda in 1972. Ever since, there has been a great deal of interest in research of semiconductors (e.g. metal oxide, metal-free organic, noble-metal complex) exhibiting effective band gap for photochemical reactions. In the case of environmental friendliness, toxicity of metal-based semiconductors brings some restrictions in possible applications. Regarding this, very robust and ‘earth-abundant’ organic semiconductor, graphitic carbon nitride has been synthesized and successfully applied in photoinduced applications as novel photocatalyst. Properties such as suitable band gap, low charge carrier recombination and feasibility for scaling up, pave the way of advance combination with other catalysts to gather higher photoactivity based on compatible heterojunction.
This dissertation aims to demonstrate a series of combinations between organic semiconductor g-CN and polymer materials that are forged through photochemistry, either in synthesis or in application. Fabrication and design processes as well as applications performed in accordance to the scope of thesis will be elucidated in detail. In addition to UV light, more attention is placed on visible light as energy source with a vision of more sustainability and better scalability in creation of novel materials and solar energy based applications.
The need to reduce humankind reliance on fossil fuels by exploiting sustainably the planet renewable resources is a major driving force determining the focus of modern material research. For this reason great interest is nowadays focused on finding alternatives to fossil fuels derived products/materials. For the short term the most promising substitute is undoubtedly biomass, since it is the only renewable and sustainable alternative to fossil fuels as carbon source. As a consequence efforts, aimed at finding new synthetic approaches to convert biomass and its derivatives into carbon-based materials, are constantly increasing. In this regard, hydrothermal carbonisation (HTC) has shown to be an effective means of conversion of biomass-derived precursors into functional carbon materials. However the attempts to convert raw biomass, in particular lignocellulosic one, directly into such products have certainly been rarer. Unlocking the direct use of these raw materials as carbon precursors would definitely be beneficial in terms of HTC sustainability. For this reason, in this thesis the HTC of carbohydrate and protein-rich biomass was systematically investigated, in order to obtain more insights on the potentials of this thermochemical processing technique in relation to the production of functional carbon materials from crude biomass. First a detailed investigation on the HTC conversion mechanism of lignocellulosic biomass and its single components (i.e. cellulose, lignin) was developed based on a comparison with glucose HTC, which was adopted as a reference model. In the glucose case it was demonstrated that varying the HTC temperature allowed tuning the chemical structure of the synthesised carbon materials from a highly cross-linked furan-based structure (T = 180oC) to a carbon framework composed of polyaromatic arene-like domains. When cellulose or lignocellulosic biomass was used as carbon precursor, the furan rich structure could not be isolated at any of the investigated processing conditions. These evidences were indicative of a different HTC conversion mechanism for cellulose, involving reactions that are commonly observed during pyrolytic processes. The evolution of glucose-derived HTC carbon chemical structure upon pyrolysis was also investigated. These studies revealed that upon heat treatment (Investigated temperatures 350 – 900 oC) the furan-based structure was progressively converted into highly curved aromatic pre-graphenic domains. This thermal degradation process was observed to produce an increasingly more hydrophobic surface and considerable microporosity within the HTC carbon structure. In order to introduce porosity in the HTC carbons derived from lignocellulosic biomass, KOH chemical activation was investigated as an HTC post-synthesis functionalisation step. These studies demonstrated that HTC carbons are excellent precursors for the production of highly microporous activated carbons (ACs) and that the porosity development upon KOH chemical activation is dependent on the chemical structure of the HTC carbon, tuned by employing different HTC temperatures. Preliminary testing of the ACs for CO2 capture or high pressure CH4 storage yielded very promising results, since the measured uptakes of both adsorbates (i.e. CO2 and CH4) were comparable to top-performing and commercially available adsorbents, usually employed for these end-applications. The combined use of HTC and KOH chemical activation was also employed to produce highly microporous N-doped ACs from microalgae. The hydrothermal treatment of the microalgae substrate was observed to cause the depletion of the protein and carbohydrate fractions and the near complete loss (i.e. 90%) of the microalgae N-content, as liquid hydrolysis/degradation products. The obtained carbonaceous product showed a predominantly aliphatic character indicating the presence of alkyl chains presumably derived from the lipid fractions. Addition of glucose to the initial reaction mixture was found out to be extremely beneficial, because it allowed the fixation of a higher N amount, in the algae derived HTC carbons (i.e. 60%), and the attainment of higher product yields (50%). Both positive effects were attributed to Maillard type cascade reactions taking place between the monosaccharides and the microalgae derived liquid hydrolysis/degradation products, which were in this way recovered from the liquid phase. KOH chemical activation of the microalgae/glucose mixture derived HTC carbons produced highly microporous N-doped carbons. Although the activation process led to a major reduction of the N-content, the retained N-amount in the ACs was still considerable. These features render these materials ideal candidates for supercapacitors electrodes, since they provide extremely high surface areas, for the formation of electric double-layer, coupled to abundant heteroatom doping (i.e. N and O) necessary to obtain a pseudocapacitance contribution.
Diese Arbeit befasst sich mit der Herstellung und Charakterisierung von thermoresponsiven Filmen auf Goldelektroden durch Fixierung eines bereits synthetisierten thermoresponsiven Polymers. Als Basis für die Entwicklung der responsiven Grenzfläche dienten drei unterschiedliche Copolymere (Polymere I, II und III) aus der Gruppe der thermisch schaltbaren Poly(oligo(ethylenglykol)methacrylate).
Die turbidimetrischen Messungen der Copolymere in Lösungen haben gezeigt, dass der Trübungspunkt vom pH-Wert, der Gegenwart von Salzen sowie von der Ionenstärke der Lösung abhängig ist. Nach der Charakterisierung der Polymere in Lösung wurden Experimente der kovalenten Kopplung der Polymere I bis III an die Oberfläche der Gold-Elektroden durchgeführt. Während bei Polymeren I und II die Ankopplung auf einer Amidverbrückung basierte, wurde bei Polymer III als alternative Methode zur Immobilisierung eine photoinduzierte Anbindung unter gleichzeitiger Vernetzung gewählt. Der Nachweis der erfolgreichen Ankopplung erfolgte bei allen Polymeren elektrochemisch mittels Cyclovoltammetrie und Impedanzspektroskopie in K3/4[Fe(CN)6]-Lösungen. Wie die Ellipsometrie-Messungen zeigten, waren die erhaltenen Polymer-Filme unterschiedlich dick. Die Ankopplung über Amidverbrückung lieferte dünne Filme (10 – 15 nm), während der photovernetzte Film deutlich dicker war (70-80 nm) und die darunter liegende Oberfläche relativ gut isolierte.
Elektrochemische Temperaturexperimente an Polymer-modifizierten Oberflächen in Lösungen in Gegenwart von K3/4[Fe(CN)6] zeigten, dass auch die immobilisierten Polymere I bis III responsives Temperaturverhalten zeigen. Bei Elektroden mit den immobilisierten Polymeren I und II ist der Temperaturverlauf der Parameterwerte diskontinuierlich – ab einem kritischen Punkt (37 °C für Polymer I und 45 °C für Polymer II) wird zunächst langsame Zunahme der Peakströme wird deutlich schneller. Das Temperaturverhalten von Polymer III ist dagegen bis 50 °C kontinuierlich, der Peakstrom sinkt hier durchgehend.
Weiterhin wurde mit den auf Polymeren II und III basierten Elektroden deren Anwendung als responsive Matrix für Bioerkennungsreaktionen untersucht. Es wurde die Ankopplung von kleinen Biorezeptoren, TAG-Peptiden, an Polymer II- und Polymer III-modifizierten Elektroden durchgeführt. Das hydrophile FLAG-TAG-Peptid verändert das Temperaturverhalten des Polymer II-Films unwesentlich, da es die Hydrophilie des Netzwerkes nicht beeinflusst. Weiterhin wurde der Effekt der Ankopplung der ANTI-FLAG-TAG-Antikörper an FLAG-TAG-modifizierte Polymer II-Filme untersucht. Es konnte gezeigt werden, dass die Antikörper spezifisch an FLAG-TAG-modifiziertes Polymer II binden. Es wurde keine unspezifische Anbindung von ANTI-FLAG-TAG an Polymer II beobachtet. Die Temperaturexperimente haben gezeigt, dass die thermische Restrukturierung des Polymer II-FLAG-TAG-Filmes auch nach der Antikörper-Ankopplung noch stattfindet. Der Einfluss der ANTI-FLAG-TAG-Ankopplung ist gering, da der Unterschied in der Hydrophilie zwischen Polymer II und FLAG-TAG bzw. ANTI-FLAG-TAG zu gering ist.
Für die Untersuchungen mit Polymer III-Elektroden wurde neben dem hydrophilen FLAG-TAG-Peptid das deutlich hydrophobere HA-TAG-Peptid ausgewählt. Wie im Falle der Polymer II Elektrode beeinflusst das gekoppelte FLAG-TAG-Peptid das Temperaturverhalten des Polymer III-Netzwerkes nur geringfügig. Die gemessenen Stromwerte sind geringer als bei der Polymer III-Elektrode. Das Temperaturverhalten der FLAG-TAG-Elektrode ähnelt dem der reinen Polymer III-Elektrode – die Stromwerte sinken kontinuierlich bis die Temperatur von ca. 40 °C erreicht ist, bei der ein Plateau beobachtet wird. Offensichtlich verändert FLAG-TAG auch in diesem Fall nicht wesentlich die Hydrophilie des Polymer III-Netzwerkes. Das an Polymer III-Elektroden gekoppelte hydrophobe HA-TAG-Peptid beeinflusst dagegen im starken Maße den Quellzustand des Netzwerkes. Die Ströme für die HA-TAG-Elektroden sind deutlich geringer als die für die FLAG-TAG-Polymer III-Elektroden, was auf geringeren Wassergehalt und dickeren Film zurückzuführen ist. Bereits ab 30 °C erfolgt der Anstieg von Stromwerten, der bei Polymer III- bzw. bei Polymer III-FLAG-TAG-Elektroden nicht beobachtet werden kann. Das gekoppelte hydrophobe HA-TAG-Peptid verdrängt Wasser aus dem Polymer III-Netzwerk, was in der Stauchung des Films bereits bei Raumtemperatur resultiert. Dies führt dazu, dass der Film im Laufe des Temperaturanstieges kaum noch komprimiert. Die Stromwerte steigen in diesem Fall entsprechend des Anstiegs der temperaturabhängigen Diffusion des Redoxpaares. Diese Untersuchungen zeigen, dass das HA-TAG-Peptid als Ankermolekül deutlich besser für eine potentielle Verwendung der Polymer III-Filme für sensorische Zwecke geeignet ist, da es sich deutlich in der Hydrophilie von Polymer III unterscheidet.
The present thesis is to be brought into line with the current need for alternative and sustainable approaches toward energy management and materials design. In this context, carbon in particular has become the material of choice in many fields such as energy conversion and storage. Herein, three main topics are covered: 1)An alternative synthesis strategy toward highly porous functional carbons with tunable porosity using ordinary salts as porogen (denoted as “salt templating”) 2)The one-pot synthesis of porous metal nitride containing functional carbon composites 3)The combination of both approaches, enabling the generation of highly porous composites with finely tunable properties All approaches have in common that they are based on the utilization of ionic liquids, salts which are liquid below 100 °C, as precursors. Just recently, ionic liquids were shown to be versatile precursors for the generation of heteroatom-doped carbons since the liquid state and a negligible vapor pressure are highly advantageous properties. However, in most cases the products do not possess any porosity which is essential for many applications. In the first part, “salt templating”, the utilization of salts as diverse and sustainable porogens, is introduced. Exemplarily shown for ionic liquid derived nitrogen- and nitrogen-boron-co-doped carbons, the control of the porosity and morphology on the nanometer scale by salt templating is presented. The studies within this thesis were conducted with the ionic liquids 1-Butyl-3-methyl-pyridinium dicyanamide (Bmp-dca), 1-Ethyl-3-methyl-imidazolium dicyanamide (Emim-dca) and 1 Ethyl 3-methyl-imidazolium tetracyanoborate (Emim-tcb). The materials are generated through thermal treatment of precursor mixtures containing one of the ionic liquids and a porogen salt. By simple removal of the non-carbonizable template salt with water, functional graphitic carbons with pore sizes ranging from micro- to mesoporous and surface areas up to 2000 m2g-1 are obtained. The carbon morphologies, which presumably originate from different onsets of demixing, mainly depend on the nature of the porogen salt whereas the nature of the ionic liquid plays a minor role. Thus, a structural effect of the porogen salt rather than activation can be assumed. This offers an alternative to conventional activation and templating methods, enabling to avoid multiple-step and energy-consuming synthesis pathways as well as employment of hazardous chemicals for the template removal. The composition of the carbons can be altered via the heat-treatment procedure, thus at lower synthesis temperatures rather polymeric carbonaceous materials with a high degree of functional groups and high surface areas are accessible. First results suggest the suitability of the materials for CO2 utilization. In order to further illustrate the potential of ionic liquids as carbon precursors and to expand the class of carbons which can be obtained, the ionic liquid 1-Ethyl-3-methyl-imidazolium thiocyanate (Emim-scn) is introduced for the generation of nitrogen-sulfur-co-doped carbons in combination with the already studied ionic liquids Bmp-dca and Emim-dca. Here, the salt templating approach should also be applicable eventually further illustrating the potential of salt templating, too. In the second part, a one-pot and template-free synthesis approach toward inherently porous metal nitride nanoparticle containing nitrogen-doped carbon composites is presented. Since ionic liquids also offer outstanding solubility properties, the materials can be generated through the carbonization of homogeneous solutions of an ionic liquid acting as nitrogen as well as carbon source and the respective metal precursor. The metal content and surface area are easily tunable via the initial metal precursor amount. Furthermore, it is also possible to synthesize composites with ternary nitride nanoparticles whose composition is adjustable by the metal ratio in the precursor solution. Finally, both approaches are combined into salt templating of the one-pot composites. This opens the way to the one-step synthesis of composites with tunable composition, particle size as well as precisely controllable porosity and morphology. Thereby, common synthesis strategies where the product composition is often negatively affected by the template removal procedure can be avoided. The composites are further shown to be suitable as electrodes for supercapacitors. Here, different properties such as porosity, metal content and particle size are investigated and discussed with respect to their influence on the energy storage performance. Because a variety of ionic liquids, metal precursors and salts can be combined and a simple closed-loop process including salt recycling is imaginable, the approaches present a promising platform toward sustainable materials design.
In this work, the development of a new molecular building block, based on synthetic peptides derived from decorin, is presented. These peptides represent a promising basis for the design of polymer-based biomaterials that mimic the ECM on a molecular level and exploit specific biological recognition for technical applications. Multiple sequence alignments of the internal repeats of decorin that formed the inner and outer surface of the arch-shaped protein were used to develop consensus sequences. These sequences contained conserved sequence motifs that are likely to be related to structural and functional features of the protein. Peptides representative for the consensus sequences were synthesized by microwave-assisted solid phase peptide synthesis and purified by RP-HPLC, with purities higher than 95 mol%. After confirming the desired masses by MALDI-TOF-MS, the primary structure of each peptide was investigated by 1H and 2D NMR, from which a full assignment of the chemical shifts was obtained. The characterization of the peptides conformation in solution was performed by CD spectroscopy, which demonstrated that using TFE, the peptides from the outer surface of decorin show a high propensity to fold into helical structures as observed in the original protein. To the contrary, the peptides from the inner surface did not show propensity to form stable secondary structure. The investigation of the binding capability of the peptides to Collagen I was performed by surface plasmon resonance analyses, from which all but one of the peptides representing the inner surface of decorin showed binding affinity to collagen with values of dissociation constant between 2•10-7 M and 2.3•10-4 M. On the other hand, the peptides representative for the outer surface of decorin did not show any significant interaction to collagen. This information was then used to develop experimental demonstration for the binding capabilities of the peptides from the inner surface of decorin to collagen even when used in more complicated situations close to possible appications. With this purpose, the peptide (LRELHLNNN) which showed the highest binding affinity to collagen (2•10-7 M) was functionalized with an N-terminal triple bond in order to obtain a peptide dimer via copper(I)-catalyzed cycloaddition reaction with 4,4'-diazidostilbene-2,2'-disulfonic acid. Rheological measurements showed that the presence of the peptide dimer was able to enhance the elastic modulus (G') of a collagen gel from ~ 600 Pa (collagen alone) to ~ 2700 Pa (collagen and peptide dimer). Moreover, it was shown that the mechanical properties of a collagen gel can be tailored by using different molar ratios of peptide dimer respect to collagen. The same peptide, functionalized with the triple bond, was used to obtain a peptide-dye conjugate by coupling it with N-(5'-azidopentanoyl)-5-aminofluorescein. An aqueous solution (5 vol% methanol) of the peptide dye conjugate was injected into a collagen and a hyaluronic acid (HA) gel and images of fluorescence detection showed that the diffusion of the peptide was slower in the collagen gel compared to the HA gel. The third experimental demonstration was gained using the peptide (LSELRLHNN) which showed the lower binding affinity (2.3•10-4 M) to collagen. This peptide was grafted to hyaluronic acid via EDC-chemistry, with a degree of functionalization of 7 ± 2 mol% as calculated by 1H-NMR. The grafting was further confirmed by FTIR and TGA measurements, which showed that the onset of decomposition for the HA-g-peptide decreased by 10 °C compared to the native HA. Rheological measurements showed that the elastic modulus of a system based on collagen and HA-g-peptide increased by almost two order of magnitude (G' = 200 Pa) compared to a system based on collagen and HA (G' = 0.9 Pa). Overall, this study showed that the synthetic peptides, which were identified from decorin, can be applied as potential building blocks for biomimetic materials that function via biological recognition.
Diese Arbeit befasst sich mit der ganzheitlichen Betrachtung der Fluideigenschaften eines unterpermischen Reservoirs am Geothermie Forschungsstandort Groß Schönebeck (GrSk) bei Reservoirbedingungen und im Betrieb der Geothermieanlage. Die Untersuchungen zur Fluidherkunft ergeben, dass es sich um ein konnates Wasser meteorischen Ursprungs ohne den Einfluss der darüberliegenden Zechsteinwässer handelt. Die Ionen und Isotopenverhältnisse im Formationswasser gelöster Komponenten in GrSk belegen einen gemeinsamen Genesepfad mit Wässern anderer Rotliegend-Reservoire des Nordostdeutschen Beckens (NEGB). Die Isotopenverhältnisse von ⁸⁷Sr/⁸⁶Sr ≈ 0,7158 und von δ³⁴SV CDT ≈ 4,1 ‰ des Sulfats weisen auf die Anreicherung des Fluids mit schweren Isotopen durch die Fluid Gestein-Wechselwirkung mit Vulkaniten und Rotliegend Sandsteinen des Unteren Perms hin.
Das im Formationswasser bei Reservoirbedingungen gelöste Gas (Gas/Wasser ≤ 2 bei STP) enthält Stickstoff (δ¹⁵NAir ≈ 0,6 ‰) und thermogenes Methan (δ¹³CV-PDB ≈ - 18 ‰) aus organischen Karbonablagerungen (Kerogen Typ - III Kohlen) hoher Reife. Die Isotopenverhältnisse der Edelgase belegen eine krustale Herkunft des Gasgemisches. Die berechnete Verweilzeit τ (⁴He) der Gase im Reservoir liegt zwischen 275 und 317 Ma und überschreitet damit bei gegebener Konzentration von Mutternukliden im Reservoirgestein das allgemein angenommene Zeitalter der Sedimentgruppe. Das lässt sich durch eine Zuwanderung von Gasen aus älteren Sedimentfolgen erklären.
Die Veränderungen der physikochemischen Fluidparameter während des Anlagenbetriebs sind hauptsächlich temperaturbedingt. Bei stabilen Produktionsbedingungen und einer Temperatur von ca. 100 °C stabilisieren sich auch die Fluideigenschaften. Bei In situ Bedingungen übertage beträgt die Dichte ρ = 1,1325 ± 0,0002 g ∙ mL⁻¹, das Redoxpotential Eh = -105,5 ± 1,3 mV und der pH = 6,61 ± 0,002. Die relative Zusammensetzung der Gasphase bei stabilen Produktionsbedingungen zeigt dagegen eine geringe Erhöhung des Stickstoffanteils sowie des Anteils der Kohlenwasserstoffe (Ethan, Propan, usw.) und Abnahme des relativen Methananteils im Laufe des Betriebs.
Die quantitative Untersuchung der sekundären mineralischen Ausfällungen im Fluid mittels sequentieller Extraktion zeigte, dass Schwermetalle als eine Hauptkomponente der Fluidfestphase größtenteils in Verbindung mit organischen Molekülen vorliegen. Experimente zum Einfluss organischer Verbindungen unterschiedlicher Substanzklassen auf eine Mobilisierung der Schwermetalle aus dem Reservoirgestein ergaben, dass die Verbindungen wie Fettsäuren und PAK (polyzyklische aromatische Kohlenwasserstoffe) die Freisetzung von Kupfer, Nickel, Chrom und Blei verhindern bzw. zu derer Immobilisierung beitragen. Im Gegensatz dazu wird die Mobilität von Zink in Anwesenheit von diesen Verbindungen erhöht. Niedermolekulare Monocarbonsäuren und stickstoffhaltige Heteroaromaten tragen, mit Ausnahme von Blei, zur Freisetzung bzw. Mobilisierung von Schwermetallen aus dem Reservoirgestein bei.
Die gewonnenen Erkenntnisse dieser Arbeit bestätigen das Risiko massiver Ausfällungen auf der kalten Seite der Geothermieanlage bei Inbetriebnahme des Kraftwerks, wenn keine an den Fluidchemismus angepassten Präventionsmethoden eingesetzt werden. Die Isotopenzusammensetzung der Fluidkomponenten sowie geringfügige Schwankungen der Gaszusammensetzung im kontinuierlichen Anlagenbetrieb lässt eine Kommunikation des unterpermischen Reservoirs mit dem darunter liegenden Oberkarbon vermuten, was eine nachträgliche Veränderung der Fluidzusammensetzung beim Dauerbetrieb der Anlage bedeuten kann.
The needs for sustainable energy generation, but also a sustainable chemistry display the basic motivation of the current thesis. By different single investigated cases, which are all related to the element carbon, the work can be devided into two major topics. At first, the sustainable synthesis of “useful” carbon materials employing the process of hydrothermal carbonisation (HC) is described. In the second part, the synthesis of heteroatom - containing carbon materials for electrochemical and fuel cell applications employing ionic liquid precursors is presented. On base of a thorough review of the literature on hydrothermolysis and hydrothermal carbonisation of sugars in addition to the chemistry of hydroxymethylfurfural, mechanistic considerations of the formation of hydrothermal carbon are proposed. On the base of these reaction schemes, the mineral borax, is introduced as an additive for the hydrothermal carbonisation of glucose. It was found to be a highly active catalyst, resulting in decreased reaction times and increased carbon yields. The chemical impact of borax, in the following is exploited for the modification of the micro- and nanostructure of hydrothermal carbon. From the borax - mediated aggregation of those primary species, widely applicable, low density, pure hydrothermal carbon aerogels with high porosities and specific surface areas are produced. To conclude the first section of the thesis, a short series of experiments is carried out, for the purpose of demonstrating the applicability of the HC model to “real” biowaste i.e. watermelon waste as feedstock for the production of useful materials. In part two cyano - containing ionic liquids are employed as precursors for the synthesis of high - performance, heteroatom - containing carbon materials. By varying the ionic liquid precursor and the carbonisation conditions, it was possible to design highly active non - metal electrocatalyst for the reduction of oxygen. In the direct reduction of oxygen to water (like used in polymer electrolyte fuel cells), compared to commercial platinum catalysts, astonishing activities are observed. In another example the selective and very cost efficient electrochemical synthesis of hydrogen peroxide is presented. In a last example the synthesis of graphitic boron carbon nitrides from the ionic liquid 1 - Ethyl - 3 - methylimidazolium - tetracyanoborate is investigated in detail. Due to the employment of unreactive salts as a new tool to generate high surface area these materials were first time shown to be another class of non - precious metal oxygen reduction electrocatalyst.
Die Doktorarbeit behandelt die Synthese Kohlenstoffnitrid basierter Materialien in eutektischen Mischungen bivalenter Metallchloride. Der Fokus liegt dabei auf ZnCl2-, SnCl2- und CoCl2-haltigen eutektischen Mischungen, in denen die Kondensation gebräuchlicher organischer Precursoren durchgeführt wird. Im Rahmen dessen wird untersucht wie durch die Reaktionsführung in Salzschmelzen unterschiedlicher Lewis-Acidität, neben der Molekülstruktur andere charakteristische Eigenschaften, wie Morphologie, Kristallinität und spezifische Oberflächen, der Materialien kontrolliert werden können. Darüber hinaus werden die optischen Eigenschaften der Materialien erörtert und in diesem Zusammenhang die Eignung als Photokatalysatoren für den oxidativen Abbau organischer Farbstoffe und für die photokatalytische Wasserreduktion bzw. –oxidation untersucht. Zusätzlich wird gezeigt, wie im System LiCl/KCl in einem einstufigen Prozess edelmetallfreie Kohlenstoffnitrid Komposite zur photokatalytischen Wasserreduktion hergestellt werden können.
Nanostructured inorganic materials are routinely synthesized by the use of templates. Depending on the synthesis conditions of the product material, either “soft” or “hard” templates can be applied. For sol-gel processes, usually “soft” templating techniques are employed, while “hard” templates are used for high temperature synthesis pathways. In classical templating approaches, the template has the unique role of structure directing agent, in the sense that it is not participating to the chemical formation of the resulting material. This work investigates a new templating pathway to nanostructured materials, where the template is also a reagent in the formation of the final material. This concept is described as “reactive templating” and opens a synthetic path toward materials which cannot be synthesised on a nanometre scale by classical templating approaches. Metal nitrides are such kind of materials. They are usually produced by the conversion of metals or metal oxides in ammonia flow at high temperature (T > 1000°C), which make the application of classical templating techniques difficult. Graphitic carbon nitride, g-C3N4, despite its fundamental and theoretical importance, is probably one of the most promising materials to complement carbon in material science and many efforts are put in the synthesis of this material. A simple polyaddition/elimination reaction path at high temperature (T = 550°C) allows the polymerisation of cyanamide toward graphitic carbon nitride solids. By hard templating, using nanostructured silica or aluminium oxide as nanotemplates, a variety of nanostructured graphitic carbon nitrides such as nanorods, nanotubes, meso- and macroporous powders could be obtained by nanocasting or nanocoating. Due to the special semi-conducting properties of the graphitic carbon nitride matrix, the nanostructured graphitic carbon nitrides show unexpected catalytic activity for the activation of benzene in Friedel-Crafts type reactions, making this material an interesting metal free catalyst. Furthermore, due to the chemical composition of g-C3N4 and the fact that it is totally decomposed at temperatures between 600°C and 800°C even under inert atmosphere, g-C3N4 was shown to be a good nitrogen donor for the synthesis of early transition metal nitrides at high temperatures. Thus using the nanostructured carbon nitrides as “reactive templates” or “nanoreactors”, various metal nitride nanostructures, such as nanoparticles and porous frameworks could be obtained at high temperature. In this approach the carbon nitride nanostructure played both the role of the nitrogen source and of the exotemplate, imprinting its size and shape to the resulting metal nitride nanostructure.
Molecules are often naturally embedded in a complex environment. As a consequence, characteristic properties of a molecular subsystem can be substantially altered or new properties emerge due to interactions between molecular and environmental degrees of freedom. The present thesis is concerned with the numerical study of quantum dynamical and stationary properties of molecular vibrational systems embedded in selected complex environments.
In the first part, we discuss "strong-coupling" model scenarios for molecular vibrations interacting with few quantized electromagnetic field modes of an optical Fabry-Pérot cavity. We thoroughly elaborate on properties of emerging "vibrational polariton" light-matter hybrid states and examine the relevance of the dipole self-energy. Further, we identify cavity-induced quantum effects and an emergent dynamical resonance in a cavity-altered thermal isomerization model, which lead to significant suppression of thermal reaction rates. Moreover, for a single rovibrating diatomic molecule in an optical cavity, we observe non-adiabatic signatures in dynamics due to "vibro-polaritonic conical intersections" and discuss spectroscopically accessible "rovibro-polaritonic" light-matter hybrid states.
In the second part, we study a weakly coupled but numerically challenging quantum mechanical adsorbate-surface model system comprising a few thousand surface modes. We introduce an efficient construction scheme for a "hierarchical effective mode" approach to reduce the number of surface modes in a controlled manner. In combination with the multilayer multiconfigurational time-dependent Hartree (ML-MCTDH) method, we examine the vibrational adsorbate relaxation dynamics from different excited adsorbate states by solving the full non-Markovian system-bath dynamics for the characteristic relaxation time scale. We examine half-lifetime scaling laws from vibrational populations and identify prominent non-Markovian signatures as deviations from Markovian reduced system density matrix theory in vibrational coherences, system-bath entanglement and energy transfer dynamics.
In the final part of this thesis, we approach the dynamics and spectroscopy of vibronic model systems at finite temperature by formulating the ML-MCTDH method in the non-stochastic framework of thermofield dynamics. We apply our method to thermally-altered ultrafast internal conversion in the well-known vibronic coupling model of pyrazine. Numerically beneficial representations of multilayer wave functions ("ML-trees") are identified for different temperature regimes, which allow us to access thermal effects on both electronic and vibrational dynamics as well as spectroscopic properties for several pyrazine models.
Die Beeinflussung optischer Eigenschaften durch Bestrahlung stellt eine Grundlage für die Herstellung anisotroper optischer Komponenten dar. In dünnen Schichten von Azobenzen-Polymeren kann optische Anisotropie durch linear polarisierte Bestrahlung induziert oder modifiziert werden. Ziel der Arbeit war es, wesentliche Struktur-Eigenschafts-Beziehungen zum Prozess der Photoorientierung zu erarbeiten, um so eine Optimierung der Materialien für verschiedene Anwendungen ermöglichen. In isotropen Schichten flüssigkristalliner und amorpher Azobenzen-Polymeren wird das Ausmaß der induzierten optischen Anisotropie günstig durch eine Donor/Akzeptor-Substitution in 4,4'-Position beeinflusst. Die Induktionsgeschwindigkeit ist in Schichten flüssigkristalliner Polymeren deutlich geringer, jedoch lassen sich höhere Werte der Doppelbrechung und des Dichroismus erreichen. In Copolymeren bewirkt die Photoorientierung der Azobenzen-Seitengruppen eine kooperative Orientierung von formanisotropen Seitengruppen. Die Mesogenität der nicht-photochromen Seitengruppen erhöht das Ausmaß der induzierten optischen Anisotropie. Die Stabilität der induzierten Doppelbrechung und des Dichroismus wird durch diese Gruppen gesteigert. In Schichten flüssigkristalliner Polymeren wird die induzierte optische Anisotropie beim Tempern im Bereich der Mesophasen erheblich verstärkt. Dabei reicht ein geringes Maß an induzierter Anisotropie aus, um Doppelbrechungs- und Dichroismuswerte zu erzielen, wie sie für LC-Domänen typisch sind. In orientierten Schichten von Azobenzen-Polymeren wird das Resultat der linear polarisierten Bestrahlung durch die Stärke der anisotropen Wechselwirkungen in den flüssigkristallinen Domänen oder den LB-Multilayern bestimmt. Eine lichtinduzierte Reorientierung kann nur erreicht werden, wenn diese Wechselwirkungen überwunden werden können. Erfolgt eine Photoreorientierung in den orientierten Polymerschichten, werden in Copolymeren formanisotrope Seitengruppen ebenfalls kooperativ reorientiert. Eine vorgelagerte UV-Bestrahlung kann durch Erzeugung eines hohen Anteils an nicht-mesogenen Z-Isomeren die anisotropen Wechselwirkungen stark schwächen und so die Seitengruppen entkoppeln. Aus diesem Zustand erfolgt die Photoreorientierung mit einer der Photoorientierung in isotropen Schichten vergleichbaren Effizienz. Die erarbeiteten Struktur-Eigenschafts-Beziehungen liefern einen Beitrag zur Optimierung derartiger Materialien für Anwendungen in den Bereichen optischer Funktionsschichten, holographischer Datenspeicherung oder der Generierung von Oberflächenreliefgittern.
As climate change worsens, there is a growing urgency to promote renewable energies and improve their accessibility to society. Here, solar energy harvesting is of particular importance. Currently, metal halide perovskite (MHP) solar cells are indispensable in future solar energy generation research. MHPs are crystalline semiconductors increasingly relevant as low-cost, high-performance materials for optoelectronics. Their processing from solution at low temperature enables easy fabrication of thin film elements, encompassing solar cells and light-emitting diodes or photodetectors. Understanding the coordination chemistry of MHPs in their precursor solution would allow control over the thin film crystallization, the material properties and the final device performance.
In this work, we elaborate on the key parameters to manipulate the precursor solution with the long-term objective of enabling systematic process control. We focus on the nanostructural characterization of the initial arrangements of MHPs in the precursor solutions. Small-angle scattering is particularly well suited for measuring nanoparticles in solution. This technique proved to be valuable for the direct analyzes of perovskite precursor solutions in standard processing concentrations without causing radiation damage. We gain insights into the chemical nature of widely used precursor structures such as methylammonium lead iodide (MAPbI3), presenting first insights into the complex arrangements and interaction within this precursor state. Furthermore, we transfer the preceding results to other more complex perovskite precursors. The influence of compositional engineering is investigated using the addition of alkali cations as an example. As a result, we propose a detailed working mechanism on how the alkali cations suppress the formation of intermediate phases and improve the quality of the crystalline thin film. In addition, we investigate the crystallization process of a tin-based perovskite composition (FASnI3) under the influence of fluoride chemistry. We prove that the frequently used additive, tin fluoride (SnF2), selectively binds undesired oxidized tin (Sn(IV)) in the precursor solution. This prevents its incorporation into the actual crystal structure and thus reduces the defect density of the material. Furthermore, SnF2 leads to a more homogeneous crystal growth process, which results in improved crystal quality of the thin film material.
In total, this study provides a detailed characterization of the complex system of perovskite precursor chemistry. We thereby cover relevant parameters for future MHP solar cell process control, such as (I) the environmental impact based on concentration and temperature (II) the addition of counter ions to reduce the diffuse layer surrounding the precursor nanostructures and (III) the targeted use of additives to eliminate unwanted components selectively and to ensure a more homogeneous crystal growth.
Hybrid nanomaterials offer the combination of individual properties of different types of nanoparticles. Some strategies for the development of new nanostructures in larger scale rely on the self-assembly of nanoparticles as a bottom-up approach. The use of templates provides ordered assemblies in defined patterns. In a typical soft-template, nanoparticles and other surface-active agents are incorporated into non-miscible liquids. The resulting self-organized dispersions will mediate nanoparticle interactions to control the subsequent self-assembly. Especially interactions between nanoparticles of very different dispersibility and functionality can be directed at a liquid-liquid interface.
In this project, water-in-oil microemulsions were formulated from quasi-ternary mixtures with Aerosol-OT as surfactant. Oleyl-capped superparamagnetic iron oxide and/or silver nanoparticles were incorporated in the continuous organic phase, while polyethyleneimine-stabilized gold nanoparticles were confined in the dispersed water droplets. Each type of nanoparticle can modulate the surfactant film and the inter-droplet interactions in diverse ways, and their combination causes synergistic effects. Interfacial assemblies of nanoparticles resulted after phase-separation. On one hand, from a biphasic Winsor type II system at low surfactant concentration, drop-casting of the upper phase afforded thin films of ordered nanoparticles in filament-like networks. Detailed characterization proved that this templated assembly over a surface is based on the controlled clustering of nanoparticles and the elongation of the microemulsion droplets. This process offers versatility to use different nanoparticle compositions by keeping the surface functionalization, in different solvents and over different surfaces. On the other hand, a magnetic heterocoagulate was formed at higher surfactant concentration, whose phase-transfer from oleic acid to water was possible with another auxiliary surfactant in ethanol-water mixture. When the original components were initially mixed under heating, defined oil-in-water, magnetic-responsive nanostructures were obtained, consisting on water-dispersible nanoparticle domains embedded by a matrix-shell of oil-dispersible nanoparticles.
Herein, two different approaches were demonstrated to form diverse hybrid nanostructures from reverse microemulsions as self-organized dispersions of the same components. This shows that microemulsions are versatile soft-templates not only for the synthesis of nanoparticles, but also for their self-assembly, which suggest new approaches towards the production of new sophisticated nanomaterials in larger scale.
In recent years, the aim of supramolecular syntheses is not only the creation of particular structures but also the introduction of specific functions in these supramolecules. The present work describes the use of the ionic self-assembly (ISA) route to generate nanostructured materials with integrated functionality. Since the ISA strategy has proved to be a facile method for the production of liquid-crystalline materials, we investigated the phase behaviour, physical properties and function of a variety of ISA materials comprising a perylene derivative as the employed oligoelectrolyte. Functionality was introduced into the materials through the use of functional surfactants. In order to meet the requirements to produce functional ISA materials through the use of functional surfactants, we designed and synthesized pyrrole-derived monomers as surfactant building blocks. Owing to the presence of the pyrrole moiety, these surfactants are not only polymerizable but are also potentially conductive when polymerized. We adopted single-tailed and double-tailed N-substituted pyrrole monomers as target molecules. Since routine characterization analysis of the double-tailed pyrrole-containing surfactant indicated very interesting, complex phase behaviour, a comprehensive investigation of its interfacial properties and mesophase behavior was conducted. The synthesized pyrrole-derived surfactants were then employed in the synthesis of ISA complexes. The self-assembled materials were characterized and subsequently polymerized by both chemical and electrochemical methods. The changes in the structure and properties of the materials caused by the in-situ polymerization were addressed. In the second part of this work, the motif investigated was a property rather than a function. Since chiral superstructures have obtained much attention during the last few years, we investigated the possibility of chiral ISA materials through the use of chiral surfactants. Thus, the work involved synthesis of novel chiral surfactants and their incorporation in ISA materials with the aim of obtaining ionically self-assembled chiral superstructures. The results and insights presented here suggest that the presented synthesis strategy can be easily extended to incorporate any kind of charged tectonic unit with desired optical, electrical, or magnetic properties into supramolecular assemblies for practical applications.
Phototropic microalgae have a large potential for producing valuable substances for the feed, food, cosmetics, pigment, bioremediation, and pharmacy industries as well as for biotechnological processes. Today it is estimated that the microalgal aquaculture worldwide production is 5000 tons of dry matter per year (not taking into account processed products) making it an approximately $1.25 billion U.S. per year industry. In this work, several spectroscopic techniques were utilized for the investigation of microalgae cells. Specifically, photondensity wave spectroscopy was applied as a technique for the on-line observation of the culture. For effective evaluation of the photosynthetic growth processes, fast and non-invasive sensor systems that analyze the relevant biological and technical process parameters are preferred. Traditionally, the biomass in a photobioreactor is quantified with the help of turbidimetry measurements, which require extensive calibration. Another problem frequently encountered when using spectral analysis for investigating solutions is that samples of interest are often undiluted and highly scattering and do not adhere to Beer-Lambert's law. Due to the fluorescence properties of chlorophyll, fluorescence spectroscopy techniques including fluorescence lifetime imaging and single photon counting could be applied to provide images of the cells as well as determine the effects of excitation intensity on the fluorescence lifetime, which is an indicator of the condition of the cell. A photon density wave is a sinusoidally intensity-modulated optical wave stemming from a point-source of light, which propagates through diffuse medium and exhibits amplitude and phase variations. Light propagation though strongly scattering media can be described by the P1 approximation to the Boltzmann transport equation. Photon density wave spectroscopy enables the ability to differentiate between scattered and absorbed light, which is desired so that an independent determination of the reduced scattering and absorption coefficients can be made. The absorption coefficient is related to the pigment content in the cells, and the reduced scattering coefficient can be used to characterize physical and morphological properties of the medium and was here applied for the determination of the average cell size.
Im Rahmen dieser Dissertation wurde der Sauerstoff im Grundgerüst der [1,3]-Dioxolo[4.5-f]benzodioxol-Fluoreszenzfarbstoffe (DBD-Fluoreszenzfarbstoffe) vollständig mit Schwefel ausgetauscht und daraus eine neue Klasse von Fluoreszenzfarbstoffen entwickelt, die Benzo[1,2-d:4,5-d']bis([1,3]dithiol)-Fluorophore (S4-DBD-Fluorophore). Insgesamt neun der besonders interessanten, difunktionalisierten Vertreter konnten synthetisiert werden, die sich in ihren elektronenziehenden Gruppen und in ihrer Anordnung unterschieden.
Durch den Austausch von Sauerstoff mit Schwefel kam es zu teilweise auffälligen Veränderungen in den Fluoreszenzparametern, wie eine Abnahme der Fluoreszenzquantenausbeuten und -lebenszeiten aber auch eine deutliche Rotverschiebung in den Absorptions- und Emissionswellenlängen mit großen STOKES-Verschiebungen. Damit sind die S4-DBD-Fluorophore eine wertvolle Ergänzung für die DBD-Farbstoffe.
Die Ursachen für die Abnahme der Lebenszeiten und Quantenausbeuten konnte auf eine hohe Besetzung des Triplett-Zustandes zurückgeführt werden, welcher durch die verstärkten Spin-Bahn-Kopplungen des Schwefels hervorgerufen wird. Zusammen mit dem Arbeitskreis physikalische Chemie der Universität Potsdam konnten auch die photophysikalischen Prozesse über die Transienten-Absorptionsspektroskopie (TAS) aufgeklärt werden.
Eine Strategie zur Funktionalisierung der S4-DBD-Farbstoffe am Thioacetalgerüst konnte entwickelt werden. So gelang es Alkohol-, Propargyl-, Azid-, NHS-Ester-, Carbonsäure-, Maleimid- und Tosyl-Gruppen an S4-DBD-Dialdehyden anzubringen.
Erweiternd wurden molekulare Stäbe auf Basis von Schwefel-Oligo-Spiro-Ketalen (SOSKs) untersucht, bei denen Sauerstoff durch Schwefel ersetzt wurde. Hier konnten die Synthesen der löslichkeitsvermittelnden TER-Muffe und auch des Tetrathiapentaerythritols als Grundbaustein deutlich verbessert werden. Aus diesen konnte ein einfaches SOSK-Polymer hergestellt werden. Weitere Versuche zum Aufbau eines Stabes müssen aber noch untersucht werden. Um einen S-OSK-Stab aufzubauen hat sich dabei die Dithiocarbonat-Gruppe in ersten Versuchen als potenzielle geeignete Schutzgruppe für das Tetrathiapentaerythritol herausgestellt.
In den letzten Jahrzehnten fand auch in der Beschichtungsindustrie ein Umdenken hin zu umweltfreundlicheren Farben und Lacken statt. Allerdings basieren auch neue Lösungen meist nicht auf Biopolymeren und in einem noch geringeren Anteil auf wasserbasierten Beschichtungssystemen aus nachwachsenden Rohstoffen. Dies stellt den Anknüpfungspunkt dieser Arbeit dar, in der untersucht wurde, ob das Biopolymer Stärke das Potenzial zum wasserbasierten Filmbildner für Farben und Lacke besitzt. Dabei müssen angelehnt an etablierte synthetische Marktprodukte die folgenden Kriterien erfüllt werden: Die wässrige Dispersion muss mindestens einen 30%igen Feststoffgehalt haben, bei Raumtemperatur verarbeitet werden können und Viskositäten zwischen 10^2-10^3 mPa·s aufweisen. Die finale Beschichtung muss einen geschlossenen Film bilden und sehr gute Haftfestigkeiten zu einer spezifischen Oberfläche, in dieser Arbeit Glas, besitzen. Als Grundlage für die Modifizierung der Stärke wurde eine Kombination von molekularem Abbau und chemischer Funktionalisierung ausgewählt. Da nicht bekannt war, welchen Einfluss die Stärkeart, die gewählte Abbaureaktion als auch verschiedene Substituenten auf die Dispersionsherstellung und deren Eigenschaften sowie die Beschichtungseigenschaften ausüben könnten, wurden die strukturellen Parameter getrennt voneinander untersucht.
Das erste Themengebiet beinhaltete den oxidativen Abbau von Kartoffel- und Palerbsenstärke mittels des Hypochlorit-Abbaus (OCl-) und des ManOx-Abbaus (H2O2, KMnO4). Mit beiden Abbaureaktionen konnten vergleichbare gewichtsmittlere Molmassen (Mw) von 2·10^5-10^6 g/mol (GPC-MALS) hergestellt werden. Allerdings führten die gewählten Reaktionsbedingungen beim ManOx-Abbau zur Bildung von Gelpartikeln. Diese lagen im µm-Bereich (DLS und Kryo-REM-Messungen) und hatten zur Folge, dass die ManOx-Proben deutlich erhöhte Viskositäten (c: 7,5 %; 9-260 mPa·s) im Vergleich zu den OCl--Proben (4-10 mPa·s) bei scherverdünnendem Verhalten besaßen und die Eigenschaften von viskoelastischen Gelen (G‘ > G‘‘) zeigten. Des Weiteren wiesen sie reduzierte Heißwasserlöslichkeiten (95 °C, vorrangig: 70-99 %) auf. Der OCl--Abbau führte zu hydrophileren (Carboxylgruppengehalt bis zu 6,1 %; ManOx: bis zu 3,1 %), nach 95 °C-Behandlung vollständig wasserlöslichen abgebauten Stärken, die ein Newtonsches Fließverhalten mit Eigenschaften einer viskoelastischen Flüssigkeit (G‘‘ > G‘) hatten. Die OCl--Proben konnten im Vergleich zu den ManOx-Produkten (10-20 %) zu konzentrierteren Dispersionen (20-40 %) verarbeitet werden, die gleichzeitig die Einschränkung von anwendungsrelevanten Mw auf < 7·10^5 g/mol zuließen (Konzentration sollte > 30 % sein). Außerdem führten nur die OCl--Proben der Kartoffelstärke zu transparenten (alle anderen waren opak) geschlossenen Beschichtungsfilmen. Somit hebt sich die Kombination von OCl--Abbau und Kartoffelstärke mit Hinblick auf die Endanwendung ab.
Das zweite Themengebiet umfasste Untersuchungen zum Einfluss von Ester- und Hydroxyalkylether-Substituenten auf Basis einer industriell abgebauten Kartoffelstärke (Mw: 1,2·10^5 g/mol) vor allem auf die Dispersionsherstellung, die rheologischen Eigenschaften der Dispersionen und die Beschichtungseigenschaften in Kombination mit Glassubstraten. Dazu wurden Ester und Ether mit DS/MS-Werten von 0,07-0,91 synthetisiert. Die Derivate konnten zu wasserbasierten Dispersionen mit Konzentrationen von 30-45 % verarbeitet werden, wobei bei hydrophoberen Modifikaten ein Co-Lösemittel, Diethylenglycolmonobutylether (DEGBE), eingesetzt werden musste. Die Feststoffgehalte sanken dabei für beide Derivatklassen vor allem mit zunehmender Alkylkettenlänge. Die anwendungsrelevanten Viskositäten (323-1240 mPa·s) stiegen auf Grund von Wechselwirkungen tendenziell mit DS/MS und Alkylkettenlänge an. Hinsichtlich der Beschichtungseigenschaften erwiesen sich die Ester vergleichend zu den Ethern als die bevorzugte Substituentenklasse, da nur die Ester geschlossene, defektfreie und mehrheitlich transparente Beschichtungsfilme bildeten, die exzellente bis sehr gute Haftfestigkeiten (ISO Klasse: 0 und 1) auf Glas besaßen. Die Ether bildeten mehrheitlich brüchige Filme. Basierend auf der Kombination der Ergebnisse aus Lösemittelaustausch, den rheologischen Untersuchungen und zusätzlichen Oberflächenspannungsmessungen (30-61 mN/m) konnte geschlossen werden, dass wahrscheinlich fehlende oder schlechte Haftfestigkeiten vorrangig akkumuliertem Wasser in den Beschichtungsfilmen (visuell: trüb oder weiß) geschuldet sind, während die Brüchigkeit vermutlich auf Wechselwirkungen (H-Brücken Wechselwirkungen, hydrophobe Wechselwirkungen) zwischen den Polymeren zurückgeführt werden kann.
Insgesamt scheint die Kombination aus Kartoffelstärke basierend auf dem OCl--Abbau mit Mw < 7·10^5 g/mol und einem Estersubstituenten eine gute Wahl für wasserbasierte Dispersionen mit hohen Feststoffkonzentrationen (> 30 %), guter Filmbildung und exzellenten Haftungen auf Glas zu sein.
Solar photocatalysis is the one of leading concepts of research in the current paradigm of sustainable chemical industry. For actual practical implementation of sunlight-driven catalytic processes in organic synthesis, a cheap, efficient, versatile and robust heterogeneous catalyst is necessary. Carbon nitrides are a class of organic semiconductors who are known to fulfill these requirements.
First, current state of solar photocatalysis in economy, industry and lab research is overviewed, outlining EU project funding, prospective synthetic and reforming bulk processes, small scale solar organic chemistry, and existing reactor designs and prototypes, concluding feasibility of the approach.
Then, the photocatalytic aerobic cleavage of oximes to corresponding aldehydes and ketones by anionic poly(heptazine imide) carbon nitride is discussed. The reaction provides a feasible method of deprotection and formation of carbonyl compounds from nitrosation products and serves as a convenient model to study chromoselectivity and photophysics of energy transfer in heterogeneous photocatalysis.
Afterwards, the ability of mesoporous graphitic carbon nitride to conduct proton-coupled electron transfer was utilized for the direct oxygenation of 1,3-oxazolidin-2-ones to corresponding 1,3-oxazlidine-2,4-diones. This reaction provides an easier access to a key scaffold of diverse types of drugs and agrochemicals.
Finally, a series of novel carbon nitrides based on poly(triazine imide) and poly(heptazine imide) structure was synthesized from cyanamide and potassium rhodizonate. These catalysts demonstrated a good performance in a set of photocatalytic benchmark reactions, including aerobic oxidation, dual nickel photoredox catalysis, hydrogen peroxide evolution and chromoselective transformation of organosulfur precursors.
Concluding, the scope of carbon nitride utilization for net-oxidative and net-neutral photocatalytic processes was expanded, and a new tunable platform for catalyst synthesis was discovered.
The Reversible Addition Fragmentation Chain Transfer (RAFT) process using the new RAFT agent benzyldithiophenyl acetate is shown to be a powerful polymerization tool to synthesize novel well-defined amphiphilic diblock copolymers composed of the constant hydrophobic block poly(butyl acrylate) and of 6 different hydrophilic blocks with various polarities, namely a series of non-ionic, non-ionic comb-like, anionic and cationic hydrophilic blocks. The controlled character of the polymerizations was supported by the linear increase of the molar masses with conversion, monomodal molar mass distributions with low polydispersities and high degrees of end-group functionalization. The new macro-surfactants form micelles in water, whose size and geometry strongly depend on their composition, according to dynamic and static light scattering measurements. The micellization is shown to be thermodynamically favored, due to the high incompatibility of the blocks as indicated by thermal analysis of the block copolymers in bulk. The thermodynamic state in solution is found to be in the strong or super strong segregation limit. Nevertheless, due to the low glass transition temperature of the core-forming block, unimer exchange occurs between the micelles. Despite the dynamic character of the polymeric micellar systems, the aggregation behavior is strongly dependent on the history of the sample, i.e., on the preparation conditions. The aqueous micelles exhibit high stability upon temperature cycles, except for an irreversibly precipitating block copolymer containing a hydrophilic block exhibiting a lower critical solution temperature (LCST). Their exceptional stability upon dilution indicates very low critical micelle concentrations (CMC) (below 4∙10<sup>-4 g∙L<sup>-1). All non-ionic copolymers with sufficiently long solvophobic blocks aggregated into direct micelles in DMSO, too. Additionally, a new low-toxic highly hydrophilic sulfoxide block enables the formation of inverse micelles in organic solvents. The high potential of the new polymeric surfactants for many applications is demonstrated, in comparison to reference surfactants. The diblock copolymers are weakly surface-active, as indicated by the graduate decrease of the surface tension of their aqueous solutions with increasing concentration. No CMC could be detected. Their surface properties at the air/water interface confer anti-foaming properties. The macro-surfactants synthesized are surface-active at the interface between two liquid phases, too, since they are able to stabilize emulsions. The polymeric micelles are shown to exhibit a high ability to solubilize hydrophobic substances in water.
In this work, the nonaqueous synthesis of binary and ternary metal oxide nanoparticles is investigated for a number of technologically important materials. A strong focus was put on studying the reaction mechanisms leading to particle formation upon solvothermal treatment of the precursors, as an understanding of the formation processes is expected to be crucial for a better control of the systems, offering the potential to tailor particle size and morphology. The synthesis of BaTiO3 was achieved by solvothermal reaction of metallic barium and titanium isopropoxide in organic solvents. Phase-pure, highly crystalline particles about 6 nm in size resulted in benzyl alcohol, whereas larger particles could be obtained in ketones such as acetone or acetophenone. In benzyl alcohol, a novel mechanism was found to lead to BaTiO3, involving a C–C coupling step between the isopropoxide ligand and the benzylic carbon of the solvent. The resulting coupling product, 4-phenyl-2-butanol, is found in almost stoichiometric yield. The particle formation in ketones proceeds via a Ti-mediated aldol condensation of the solvent, involving formal elimination of water which induces formation of the oxide. These processes also occurred when reacting solely the titanium alkoxide with ketones or aldehydes, leading to highly crystalline anatase nanoparticles for all tested solvents. In ketones, also the synthesis of nanopowders of lead zirconate titanate (PZT) was achieved, which were initially amorphous but could be crystallized by calcination at moderate temperatures. Additionally, PZT films were prepared by simply casting a suspension of the powder onto Si substrates followed by calcination.Solvothermal synthesis however is not restricted to alkoxides as precursors but is also achieved from metal acetylacetonates. The use of benzylamine as solvent proved particularly versatile, making possible the synthesis of nanocrystalline In2O3, Ga2O3, ZnO and iron oxide from the respective acetylacetonates. During the synthesis, the acetylacetonate ligand undergoes a solvolysis under C–C cleavage, resulting in metal-bound enolate ligands which, in analogy to the synthesis in ketones, induce ketimine and aldol condensation reactions. In the last section of this work, surface functionalization of anatase nanoparticles is explored. The particles were first capped with various organic ligands via a facile in situ route, which resulted in altered properties such as enhanced dispersibility in various solvents. In a second step, short functional oligopeptide segments were attached to the particles by means of a catechol linker to achieve advanced self-assembly properties.
This thesis provides a novel view on the early stage of crystallization utilizing calcium carbonate as a model system. Calcium carbonate is of great economical, scientific and ecological importance, because it is a major part of water hardness, the most abundant Biomineral and forms huge amounts of geological sediments thus binding large amounts of carbon dioxide. The primary experiments base on the evolution of supersaturation via slow addition of dilute calcium chloride solution into dilute carbonate buffer. The time-dependent measurement of the Ca2+ potential and concurrent pH = constant titration facilitate the calculation of the amount of calcium and carbonate ions bound in pre-nucleation stage clusters, which have never been detected experimentally so far, and in the new phase after nucleation, respectively. Analytical Ultracentrifugation independently proves the existence of pre-nucleation stage clusters, and shows that the clusters forming at pH = 9.00 have a proximately time-averaged size of altogether 70 calcium and carbonate ions. Both experiments show that pre-nucleation stage cluster formation can be described by means of equilibrium thermodynamics. Effectively, the cluster formation equilibrium is physico-chemically characterized by means of a multiple-binding equilibrium of calcium ions to a ‘lattice’ of carbonate ions. The evaluation gives GIBBS standard energy for the formation of calcium/carbonate ion pairs in clusters, which exhibits a maximal value of approximately 17.2 kJ mol^-1 at pH = 9.75 and relates to a minimal binding strength in clusters at this pH-value. Nucleated calcium carbonate particles are amorphous at first and subsequently become crystalline. At high binding strength in clusters, only calcite (the thermodynamically stable polymorph) is finally obtained, while with decreasing binding strength in clusters, vaterite (the thermodynamically least stable polymorph) and presumably aragonite (the thermodynamically intermediate stable polymorph) are obtained additionally. Concurrently, two different solubility products of nucleated amorphous calcium carbonate (ACC) are detected at low binding strength and high binding strength in clusters (ACC I 3.1EE-8 M^2, ACC II 3.8EE-8 M^2), respectively, indicating the precipitation of at least two different ACC species, while the clusters provide the precursor species of ACC. It is proximate that ACC I may relate to calcitic ACC –i.e. ACC exhibiting short range order similar to the long range order of calcite and that ACC II may relate to vateritic ACC, which will subsequently transform into the particular crystalline polymorph as discussed in the literature, respectively. Detailed analysis of nucleated particles forming at minimal binding strength in clusters (pH = 9.75) by means of SEM, TEM, WAXS and light microscopy shows that predominantly vaterite with traces of calcite forms. The crystalline particles of early stages are composed of nano-crystallites of approximately 5 to 10 nm size, respectively, which are aligned in high mutual order as in mesocrystals. The analyses of precipitation at pH = 9.75 in presence of additives –polyacrylic acid (pAA) as a model compound for scale inhibitors and peptides exhibiting calcium carbonate binding affinity as model compounds for crystal modifiers- shows that ACC I and ACC II are precipitated in parallel: pAA stabilizes ACC II particles against crystallization leading to their dissolution for the benefit of crystals that form from ACC I and exclusively calcite is finally obtained. Concurrently, the peptide additives analogously inhibit the formation of calcite and exclusively vaterite is finally obtained in case of one of the peptide additives. These findings show that classical nucleation theory is hardly applicable for the nucleation of calcium carbonate. The metastable system is stabilized remarkably due to cluster formation, while clusters forming by means of equilibrium thermodynamics are the nucleation relevant species and not ions. Most likely, the concept of cluster formation is a common phenomenon occurring during the precipitation of hardly soluble compounds as qualitatively shown for calcium oxalate and calcium phosphate. This finding is important for the fundamental understanding of crystallization and nucleation-inhibition and modification by additives with impact on materials of huge scientific and industrial importance as well as for better understanding of the mass transport in crystallization. It can provide a novel basis for simulation and modelling approaches. New mechanisms of scale formation in Bio- and Geomineralization and also in scale inhibition on the basis of the newly reported reaction channel need to be considered.

In dieser Arbeit ist es gelungen, die Bedeutung physikalisch-chemischer Mechanismen in der Biomineralisation gegenüber der bisher angenommenen Dominanz spezifischer biomolekularer Erkennungsmechanismen aufzuzeigen. Dazu wurden drei Ansätze verfolgt: Zum einen wurden Studien zur Calciumcarbonatkristallisation durchgeführt. Zum anderen wurde das Biomineral Perlmutt intensiv untersucht. Als drittes wurde ein Modellsystem entwickelt, mit dem künstliches Perlmutt synthetisiert und ein Mechanismus für die Perlmuttmineralisation vorgeschlagen werden konnte. Im ersten Schritt wurden in einem simplen Kristallisationsansatz komplexe Calciumcarbonatüberstrukturen ohne die Verwendung von Additiven synthetisiert. Es wurde gezeigt, daß diese durch orientierte Anlagerung von Nanopartikeln gebildet werden, bei der dipolare Felder eine wichtige Rolle zu spielen scheinen. Dieser Mechansimus war bislang für Calciumcarbonat unbekannt und ermöglicht die Synthese komplexer Kristallmorphologien, wodurch die Frage aufgeworfen wird, ob er bei der Biomineralbildung von Bedeutung sein kann. Durch Einsatz minimaler Mengen eines einfachen, synthetischen Additivs bei der Kristallisation wurden zu Überstrukturen angeordnete Aragonitplättchen synthetisiert, die von einer wenige nm dicken Schicht aus amorphen Calciumcarbonat umgeben sind. Eine solche Schicht wurde auch bei den Aragonitplättchen Perlmutts entdeckt (s.u.) und bietet möglicherweise in verschiedenen Systemen eine Erklärung für die Stabilisierung der sonst metastabilen Aragonitphase. Im zweiten Schritt wurden bei der Untersuchung von natürlichem Perlmutt zwei bislang unbekannte Strukturmerkmale entdeckt: Es gibt Bereiche, die nicht aus den charakteristischen Plättchen bestehen, sondern wesentlich weniger stark mineralisert sind. Die Mineralphase besteht in diesen Bereichen aus Nanopartikeln. Es wurde weiterhin gezeigt, daß die Aragonitplättchen von einer wenige nm dicken Schicht aus amorphem Calciumcarbonat umgeben ist. Die gängigen Modelle der Perlmuttbildung sind mit diesen Beobachtungen nicht zu vereinbaren und somit zu hinterfragen. Dagegen deuten diese Ergebnisse ein Wachstum von Perlmutt über ACC-Nanopartikel an. Unter der Annahme der Bedeutung physikalisch-chemischer Mechanismen in der Biomineralisation wurde schließlich als dritter Schritt ein Ansatz zur in vitro-Retrosynthese von Biomineralien ausgehend von ihrer unlöslichen Matrix entwickelt. Mit diesem Ansatz ist es erstmals gelungen, künstliches Perlmutt auf synthetischem Wege herzustellen, das morphologisch nicht vom Original zu unterscheiden ist. Die existierenden Unterschiede konnten zeigen, daß der Mineralisationsprozeß nicht auf ein spezifisches Mikroumgebungssystem beschränkt, sondern "allgemeiner gültig"' sein muß. Bei der Retrosynthese gibt es zwei Schlüsselfaktoren: Zum einen die demineralisierte unlösliche Perlmuttmatrix als dreidimensionales Gerüst für das künstliche Perlmutt, zum anderen amorphe Precursorpartikel, die die Mineralphase bilden. Es werden keinerlei Proteine oder andere Biomoleküle verwendet. Dieser Ansatz bietet die Möglichkeit, den Mineralisationsprozeß an einem in vitro-Modellsystem zu verfolgen, was für das in vivo-System, wenn überhaupt, nur unter starken Einschränkungen möglich ist. Es wurde gezeigt, daß das künstliche Perlmutt über die Mesoskalentransformation von ACC-Precursorn innerhalb der Matrix gebildet wird und als möglicher Mechanismus bei der Biomineralisation von natürlichem Perlmutt diskutiert. Es konnte in der vorliegenden Arbeit konsequent gezeigt werden, daß die Imitation von Biomineralisationsprozessen in in vitro-Ansätzen möglich ist, wobei chemisch-physikalische Parameter dominieren. In zukünftigen Studien sollten einerseits die mechanischen Eigenschaften des künstlichen Perlmutts untersucht werden, wofür sich in Vorversuchen im Rahmen dieser Arbeit die Nanoindentierung als geeignet herausgestellt hat. Es sollte geprüft werden, ob das hier ermittelte Prinzip zur Mineralisierung in der Materialentwicklung angewendet werden kann. Andererseits sollte die Retrosynthese auf andere Systeme ausgeweitet und in vivo-Studien durchgeführt werden, um die Gültigkeit der vorgeschlagenen Mechanismen zu überprüfen.
Nanofibrous mats are interesting scaffold materials for biomedical applications like tissue engineering due to their interconnectivity and their size dimension which mimics the native cell environment. Electrospinning provides a simple route to access such fiber meshes. This thesis addresses the structural and functional control of electrospun fiber mats. In the first section, it is shown that fiber meshes with bimodal size distribution could be obtained in a single-step process by electrospinning. A standard single syringe set-up was used to spin concentrated poly(ε-caprolactone) (PCL) and poly(lactic-co-glycolic acid) (PLGA) solutions in chloroform and meshes with bimodal-sized fiber distribution could be directly obtained by reducing the spinning rate at elevated humidity. Scanning electron microscopy (SEM) and mercury porosity of the meshes suggested a suitable pore size distribution for effective cell infiltration. The bimodal fiber meshes together with unimodal fiber meshes were evaluated for cellular infiltration. While the micrometer fibers in the mixed meshes generate an open pore structure, the submicrometer fibers support cell adhesion and facilitate cell bridging on the large pores. This was revealed by initial cell penetration studies, showing superior ingrowth of epithelial cells into the bimodal meshes compared to a mesh composed of unimodal 1.5 μm fibers. The bimodal fiber meshes together with electrospun nano- and microfiber meshes were further used for the inorganic/organic hybrid fabrication of PCL with calcium carbonate or calcium phosphate, two biorelevant minerals. Such composite structures are attractive for the potential improvement of properties such as stiffness or bioactivity. It was possible to encapsulate nano and mixed sized plasma-treated PCL meshes to areas > 1 mm2 with calcium carbonate using three different mineralization methods including the use of poly(acrylic acid). The additive seemed to be useful in stabilizing amorphous calcium carbonate to effectively fill the space between the electrospun fibers resulting in composite structures. Micro-, nano- and mixed sized fiber meshes were successfully coated within hours by fiber directed crystallization of calcium phosphate using a ten-times concentrated simulated body fluid. It was shown that nanofibers accelerated the calcium phosphate crystallization, as compared to microfibers. In addition, crystallizations performed at static conditions led to hydroxyapatite formations whereas in dynamic conditions brushite coexisted. In the second section, nanofiber functionalization strategies are investigated. First, a one-step process was introduced where a peptide-polymer-conjugate (PLLA-b-CGGRGDS) was co-spun with PLGA in such a way that the peptide is enriched on the surface. It was shown that by adding methanol to the chloroform/blend solution, a dramatic increase of the peptide concentration at the fiber surface could be achieved as determined by X-ray photoelectron spectroscopy (XPS). Peptide accessibility was demonstrated via a contact angle comparison of pure PLGA and RGD-functionalized fiber meshes. In addition, the electrostatic attraction between a RGD-functionalized fiber and a silica bead at pH ~ 4 confirmed the accessibility of the peptide. The bioactivity of these RGD-functionalized fiber meshes was demonstrated using blends containing 18 wt% bioconjugate. These meshes promoted adhesion behavior of fibroblast compared to pure PLGA meshes. In a second functionalization approach, a modular strategy was investigated. In a single step, reactive fiber meshes were fabricated and then functionalized with bioactive molecules. While the electrospinning of the pure reactive polymer poly(pentafluorophenyl methacrylate) (PPFPMA) was feasible, the inherent brittleness of PPFPMA required to spin a PCL blend. Blends and pure PPFPMA showed a two-step functionalization kinetics. An initial fast reaction of the pentafluorophenyl esters with aminoethanol as a model substance was followed by a slow conversion upon further hydrophilization. This was analysed by UV/Vis-spectroscopy of the pentaflurorophenol release upon nucleophilic substitution with the amines. The conversion was confirmed by increased hydrophilicity of the resulting meshes. The PCL/PPFPMA fiber meshes were then used for functionalization with more complex molecules such as saccharides. Aminofunctionalized D-Mannose or D-Galactose was reacted with the active pentafluorophenyl esters as followed by UV/Vis spectroscopy and XPS. The functionality was shown to be bioactive using macrophage cell culture. The meshes functionalized with D-Mannose specifically stimulated the cytokine production of macrophages when lipopolysaccharides were added. This was in contrast to D-Galactose- or aminoethanol-functionalized and unfunctionalized PCL/PPFPMA fiber mats.
Poröse Sol-Gel-Materialien finden in vielen Bereichen Anwendung bzw. sind Gegenstand der aktuellen Forschung. Zu diesen Bereichen zählen sowohl klassische Anwendungen, wie z. B. die Verwendung als Katalysator, Molekularsieb oder Trockenmittel, als auch nichtklassische Anwendungen, wie z. B. der Einsatz als Kontrastmittel in der Magnet-Resonanz-Tomographie oder in Form von dünnen Zeolithfilmen als Isolatoren in Mikrochips. Auch für den Einsatz in der Photonik werden poröse Materialien in Betracht gezogen, wie die Entwicklung des Zeolith-Farbstoff-Lasers zeigt. Mikroporöse Zeolithe können generell über einfache Ionenaustauschreaktionen mit Lanthanoidionen in lumineszente Materialien umgewandelt werden. Neben der Erzeugung eines lumineszenten Materials, dessen Lumineszenzeigenschaften charakterisiert werden müssen, bietet die Nutzung von Lanthanoidionen die Möglichkeit diese Ionen als Sonde zur Charakterisierung der Ion-Wirt-Wechselwirkungen zu funktionalisieren, was z. B. in Bezug auf die Anwendung als Katalysator von großer Bedeutung ist. Dabei werden die einzigartigen Lumineszenzeigenschaften der Lanthanoidionen, in diesem Fall von Europium(III) und Terbium(III), genutzt. In dieser Arbeit wurden Lanthanoid-dotierte mikroporöse Zeolithe, mikroporös-mesoporöse Hybridmaterialien und mesoporöse Silikate hinsichtlich ihrer Lumineszenzeigenschaften und ihrer Wechselwirkung des Wirtsmaterials mit den Lanthanoidionen mittels zeitaufgelöster Lumineszenzspektroskopie untersucht. Zeitaufgelöste Emissionsspektren (TRES) liefern dabei sowohl Informationen in der Wellenlängen- als auch in der Zeitdomäne. Erstmalig wurden die TRES mittels einer umfangreichen Auswertemethodik behandelt. Neben der Anpassung des Abklingverhaltens mit einer diskreten Zahl von Exponentialfunktionen, wurden unterstützend auch Abklingzeitverteilungsanalysen durchgeführt. Zeitaufgelöste flächennormierte Emissionsspektren (TRANES), eine Erweiterung der normalen TRES, konnten erstmals zur Bestimmung der Zahl der emittierenden Lanthanoidspezies in porösen Materialien genutzt werden. Durch die Berechnung der Decayassoziierten Spektren (DAS) konnten den Lanthanoidspezies die entsprechenden Lumineszenzspektren zugeordnet werden. Zusätzlich konnte, speziell im Fall der Europium-Lumineszenz, durch Kombination von zeitlicher und spektraler Information das zeitabhängige Asymmetrieverhältnis R und die spektrale Evolution des 5D0-7F0-Übergangs mit der Zeit t untersucht und somit wesentliche Informationen über die Verteilung der Europiumionen im Wirtsmaterial erhalten werden. Über die Abklingzeit und das Asymmetrieverhältnis R konnten Rückschlüsse auf die Zahl der OH-Oszillatoren in der ersten Koordinationssphäre und die Symmetrie der Koordinationsumgebung gezogen werden. Für die mikroporösen und mikroporös-mesoporösen Materialien wurden verschiedene Lanthanoidspezies, im Regelfall zwei, gefunden, welche entsprechend der beschriebenen Methoden charakterisiert wurden. Diese Lanthanoidspezies konnten Positionen in den Materialien zugeordnet werden, die sich im tief Inneren des Porensystems oder auf bzw. nahe der äußeren Oberfläche oder in den Mesoporen befinden. Erstere Spezies ist aufgrund ihrer Position im Material gut vor Feuchtigkeitseinflüssen geschützt, was sich deutlich in entsprechend langen Lumineszenzabklingzeiten äußert. Zusätzlich ist diese Europiumspezies durch unsymmetrische Koordinationsumgebung charakterisiert, was auf einen signifikanten Anteil an Koordination der Lanthanoidionen durch die Sauerstoffatome im Wirtsgitter zurückzuführen ist. Ionen, die sich nahe oder auf der äußeren Oberfläche befinden, sind dagegen für Feuchtigkeit zugänglicher, was in kürzeren Lumineszenzabklingzeiten und einer symmetrischeren Koordinationsumgebung resultiert. Der Anteil von Wassermolekülen in der ersten Koordinationssphäre ist hier deutlich größer, als bei den Ionen, die sich tiefer im Porensystem befinden und entspricht in vielen Fällen der Koordinationszahl eines vollständig hydratisierten Lanthanoidions. Auch der Einfluss von Oberflächenmodifikationen auf die Speziesverteilung und das Verhalten der Materialien gegenüber Feuchtigkeit wurde untersucht. Dabei gelang es den Einfluss der Feuchtigkeit auf die Lumineszenzeigenschaften und die Speziesverteilung durch die Oberflächenmodifikation zu verringern und die Lumineszenzeigenschaften teilweise zu konservieren. Im Fall der mesoporösen Silikamonolithe wurde auch eine heterogene Verteilung der Lanthanoidionen im Porensystem gefunden. Hier wechselwirkt ein Teil der Ionen mit der Porenwand, während sich die restlichen Ionen in der wäßrigen Phase innerhalb des Porensystems aufhalten. Das Aufbringen von Oberflächenmodifikationen führte zu einer Wechselwirkung der Ionen mit diesen Oberflächenmodifikationen, was sich in Abhängigkeit von der Oberflächenbeladung in den enstprechenden Lumineszenzeigenschaften niederschlug.
Cellulose is the most abundant biopolymer on earth. In this work it has been used, in various forms ranging from wood to fully processed laboratory grade microcrystalline cellulose, to synthesise a variety of metal and metal carbide nanoparticles and to establish structuring and patterning methodologies that produce highly functional nano-hybrids. To achieve this, the mechanisms governing the catalytic processes that bring about graphitised carbons in the presence of iron have been investigated. It was found that, when infusing cellulose with an aqueous iron salt solution and heating this mixture under inert atmosphere to 640 °C and above, a liquid eutectic mixture of iron and carbon with an atom ratio of approximately 1:1 forms. The eutectic droplets were monitored with in-situ TEM at the reaction temperature where they could be seen dissolving amorphous carbon and leaving behind a trail of graphitised carbon sheets and subsequently iron carbide nanoparticles. These transformations turned ordinary cellulose into a conductive and porous matrix that is well suited for catalytic applications. Despite these significant changes on the nanometre scale the shape of the matrix as a whole was retained with remarkable precision. This was exemplified by folding a sheet of cellulose paper into origami cranes and converting them via the temperature treatment in to magnetic facsimiles of those cranes. The study showed that the catalytic mechanisms derived from controlled systems and described in the literature can be transferred to synthetic concepts beyond the lab without loss of generality. Once the processes determining the transformation of cellulose into functional materials were understood, the concept could be extended to other metals and metal-combinations. Firstly, the procedure was utilised to produce different ternary iron carbides in the form of MxFeyC (M = W, Mn). None of those ternary carbides have thus far been produced in a nanoparticle form. The next part of this work encompassed combinations of iron with cobalt, nickel, palladium and copper. All of those metals were also probed alone in combination with cellulose. This produced elemental metal and metal alloy particles of low polydispersity and high stability. Both features are something that is typically not associated with high temperature syntheses and enables to connect the good size control with a scalable process. Each of the probed reactions resulted in phase pure, single crystalline, stable materials. After showing that cellulose is a good stabilising and separating agent for all the investigated types of nanoparticles, the focus of the work at hand is shifted towards probing the limits of the structuring and pattering capabilities of cellulose. Moreover possible post-processing techniques to further broaden the applicability of the materials are evaluated. This showed that, by choosing an appropriate paper, products ranging from stiff, self-sustaining monoliths to ultra-thin and very flexible cloths can be obtained after high temperature treatment. Furthermore cellulose has been demonstrated to be a very good substrate for many structuring and patterning techniques from origami folding to ink-jet printing. The thereby resulting products have been employed as electrodes, which was exemplified by electrodepositing copper onto them. Via ink-jet printing they have additionally been patterned and the resulting electrodes have also been post functionalised by electro-deposition of copper onto the graphitised (printed) parts of the samples. Lastly in a preliminary test the possibility of printing several metals simultaneously and thereby producing finely tuneable gradients from one metal to another have successfully been made. Starting from these concepts future experiments were outlined. The last chapter of this thesis concerned itself with alternative synthesis methods of the iron-carbon composite, thereby testing the robustness of the devolved reactions. By performing the synthesis with partly dissolved scrap metal and pieces of raw, dry wood, some progress for further use of the general synthesis technique were made. For example by using wood instead of processed cellulose all the established shaping techniques available for wooden objects, such as CNC milling or 3D prototyping, become accessible for the synthesis path. Also by using wood its intrinsic well defined porosity and the fact that large monoliths are obtained help expanding the prospect of using the composite. It was also demonstrated in this chapter that the resulting material can be applied for the environmentally important issue of waste water cleansing. Additionally to being made from renewable resources and by a cheap and easy one-pot synthesis, the material is recyclable, since the pollutants can be recovered by washing with ethanol. Most importantly this chapter covered experiments where the reaction was performed in a crude, home-built glass vessel, fuelled – with the help of a Fresnel lens – only by direct concentrated sunlight irradiation. This concept carries the thus far presented synthetic procedures from being common laboratory syntheses to a real world application. Based on cellulose, transition metals and simple equipment, this work enabled the easy one-pot synthesis of nano-ceramic and metal nanoparticle composites otherwise not readily accessible. Furthermore were structuring and patterning techniques and synthesis routes involving only renewable resources and environmentally benign procedures established here. Thereby it has laid the foundation for a multitude of applications and pointed towards several future projects reaching from fundamental research, to application focussed research and even and industry relevant engineering project was envisioned.
Lignin valorization

The topic of this project is the use of lignin as alternative source of aromatic building blocks and oligomers to fossil feedstocks. Lignin is known as the most abundant aromatic polymer in nature and is isolated from the lignocellulosic component of plants by different possible extraction treatments. Both the biomass source and the extraction method affect the structure of the isolated lignin, therefore influencing its further application. Lignin was extracted from beech wood by two different hydrothermal alkaline treatments, which use NaOH and Ba(OH)2 as base and by an acid-catalyzed organosolv process. Moreover, lignin was isolated from bamboo, beech wood and coconut by soda treatment of the biomasses. A comparison of the structural features of such isolated lignins was performed through the use of a wide range of analytical methods. Alkaline lignins resulted in a better candidate as carbon precursor and macromonomers for the synthesis of polymer than organosolv lignin. In fact, alkaline lignins showed higher residual mass after carbonization and higher content of the reactive hydroxy functionalities. In contrast, the lignin source turned out to slightly affect the lignin hydroxyl content.
One of the most common lignin modifications is its deconstruction to obtain aromatic molecules, which can be used as starting materials for the synthesis of fine chemicals. Lignin deconstruction leads to a complex mixture of aromatic molecules. A gas chromatographic analytical method was developed to characterize the mixture of products obtained by lignin deconstruction via heterogeneous catalytic hydrogenolysis. The analytical protocol allowed the quantification of three main groups of molecules by means of calibration curves, internal standard and a preliminary silylation step of the sample. The analytical method was used to study the influence of the hydrogenolysis catalyst, temperature and system (flow and batch reactor) on the yield and selectivity of the aromatic compounds.
Lignin extracted from beech wood by a hydrothermal process using Ba(OH)2 as base, was functionalized by aromatic nitration in order to add nitrogen functionalities. The final goal was the synthesis of a nitrogen doped carbon. Nitrated lignin was reduced to the amino form in order to compare the influence of different nitrogen functionalities on the porosity of the final carbon. The carbons were obtained by ionothermal treatment of the precursors in the presence of the eutectic salt mixture KCl/ZnCl2 Such synthesized carbons showed micro-, macro- and mesoporosity and were tested for their electrocatalytic activity towards the oxygen reduction reaction. Mesoporous carbon derived from nitro lignin displayed the highest electrocatalytic activity.
Lignins isolated from coconut, beech wood and bamboo were used as macromonomers for the synthesis of biobased polyesters. A condensation reaction was performed between lignin and a hyper branched poly(ester-amine), previously obtained by condensation of triethanolamine and adipic acid. The influence of the lignin source and content on the thermochemical and mechanical properties of the final material was investigated. The prepolymer showed adhesive properties towards aluminum and its shear strength was therefore measured. The gluing properties of such synthesized glues turned out to be independent from the lignin source but affected by the amount of lignin in the final material.
This work shows that, although still at a laboratory scale, the valorization of lignin can overcome the critical issues of lignin´s structure variability and complexity.
Funktionalisierte Poly(2-oxazoline) als neue Materialien stellen sowohl unter strukturellen Gesichtspunkten als auch im Hinblick auf potentielle Anwendungen eine interessante Polymerklasse dar. Die Ausbildung von hierarchischen Strukturen mit Poly(2-oxazolinen) über intermolekulare Wasserstoffbrückenbindungen ist hierbei ein bisher nicht beachteter Aspekt. Über einen bioinspirierten Ansatz sollten gezielt funktionelle Gruppen, die für einen hierarchischen Aufbau, z.B. in Proteinen, verantwortlich sind, in vereinfachter Weise auf die synthetische Substanzklasse der Poly(2-oxazoline) übertragen werden. Die vorliegende Arbeit beschäftigt sich mit der modularen Synthese neuer, funktionalisierter Poly(2-oxazolin) Homo- und Copolymere. Ausgehend von der Synthese von 2-(3-Butenyl)-2-oxazolin wurden definierte Präpolymere in einer kationischen Isomerisierungspolymerisation unter kontrolliert/„lebenden“ Bedingungen hergestellt. In einer anschließenden „Thio-Click“ (Thiol-En-Reaktion) Modifizierungsreaktion wurden die gewünschten funktionellen Gruppen quantitativ eingeführt. Hydroxylierte Poly(2-oxazoline) wurden hinsichtlich ihres Aggregationsverhaltens in Wasser untersucht. Bereits die jeweiligen Homopolymere bildeten aufgrund von intermolekularen Wasserstoffbrückenbindungen supramolekulare tubuläre Nanofasern aus. Durch Einsatz verschiedener analytischer Methoden konnte die innere Struktur der Nanoröhren beschrieben und ein entsprechendes Modell aufgestellt werden. Die dargestellten funktionellen Poly(2-oxazoline) wurden hinsichtlich ihrer Anwendung als potentielle, synthetische „antifreeze additives“ untersucht. Alle Polymere besitzen eine ausgeprägte Tendenz zur Nukleierung von Wasser und führen daher zu signifikanten Änderungen der Eismorphologie. Des weiteren wurde ein carboxyliertes Derivat zur biomimetischen Mineralisation von Kalziumcarbonat eingesetzt und nach phänomenologischen Gesichtspunkten untersucht.
In dieser Arbeit wird ein chemisches Abgussverfahren für selbstorganisierte Strukturen in Lösung verwendet, das es ermöglicht definierte poröse Materialien mit Strukturierung auf der Nanometerskala herzustellen. Ähnlich wie beim Gussverfahren von Werkstücken wird die Vorlage durch ein geeignetes Material abgebildet. Durch Entfernen dieser Vorlage erhält man ein poröses (mit Hohlräumen durchsetztes) Negativ derselben. Die auf diese Weise erhaltenen Materialien sind in mehrerer Hinsicht interessant: So lassen sich aus ihrer Morphologie Rückschlüsse über die Natur der selbstorganisierten Strukturen erhalten, da der hier verwendete Abbildungsprozess selbst kleinste strukturelle Details erfasst. Die Hohlräume der synthetisierten porösen Stoffe hingegen können als winzige Reaktionsgefäße, sogenannte "Nano-Reaktoren" verwendet werden. Dies ermöglicht sowohl die Synthese von Nano-Partikeln, die auf anderem Wege nicht zugänglich sind, als auch die Möglichkeit Einflüsse der räumlichen Restriktion auf die Reaktion zu untersuchen. Besonders räumlich ausgedehnte Strukturen sollten hierbei Auffälligkeiten zeigen. Somit ist die Gliederung der Arbeit vorgegeben: - Die Herstellung und Charakterisierung von porösen Stoffen und selbstorganisierten Strukturen - Ihre Verwendung als "Nano-Reaktor"
Für den Einsatz in Autobatterien gibt es besondere Anforderungen an den Elektrolyten im Bereich der Energie- und Leistungsdichten, um beispielsweise thermische Verluste gering zu halten. Hochleitfähige Elektrolyte mit Leitfähigkeiten im Millisiemensbereich sind hier ebenso notwendig wie auch sichere, d.h. möglichst nicht brennbare und einen niedrigen Dampfdruck besitzende Materialien. Um diese Vorgaben zu erreichen, ist es notwendig, einen polymeren Separator zu entwickeln, welcher auf brennbare organische Lösungsmittel verzichtet und damit eine drastische Steigerung der Sicherheit gewährleistet. Gleichzeitig müssen hierbei die Leistungsvorgaben bezüglich der Leitfähigkeit erfüllt werden. Zu diesem Zweck wurde ein Konzept basierend auf der Kombination von einer polymeren sauerstoffreichen Matrix und einer ionischen Flüssigkeit entwickelt und verifiziert. Dabei wurden folgende Erkenntnisse gewonnen: 1. Es wurden neuartige diacrylierte sauerstoffreiche Matrixkomponenten mit vielen Carbonylfunktionen, für eine gute Lithiumleitfähigkeit, synthetisiert. 2. Es wurden mehrere neue ionische Flüssigkeiten sowohl auf Imidazolbasis als auch auf Ammoniumbasis synthetisiert und charakterisiert. 3. Die Einflüsse der Kationenstruktur und der Einfluss der Gegenionen im Bezug auf Schmelzpunkte und Leitfähigkeiten wurden untersucht. 4. Aus den entwickelten Materialien wurden Blendsysteme hergestellt und mittels Impedanzspektrometrie untersucht: Leitfähigkeiten von 10-4S/cm bei Raumtemperatur sind realisierbar. 5. Die Blendsysteme wurden auf ihre thermische Stabilität hin untersucht: Stabilitäten bis 250°C sind erreichbar. Dabei wird keine kristalline Struktur beobachtet.
Die intrazelluläre Markierung mit geeigneten Reagenzien ermöglicht ihre bildgebende Darstellung in lebenden Organismen. Dieses Verfahren (auch „Zell-Tracking“ genannt) wird in der Grundlagenforschung zur Entwicklung zellulärer Therapien, für die Erforschung pathologischer Prozesse, wie der Metastasierung, sowie für Therapiekontrollen eingesetzt. Besondere Bedeutung haben in den letzten Jahren zelluläre Therapien mit Stammzellen erlangt, da sie großes Potential bei der Regeneration von Geweben bei Krankheiten wie Morbus Parkinson oder Typ-1-Diabetes versprechen. Für die Entwicklung einer zellulären Therapie sind Informationen über den Verbleib der applizierten Zellen in vivo (Homing-Potential), über ihre Zellphysiologie sowie über die Entstehung möglicher Entzündungen notwendig. Das Ziel der vorliegenden Arbeit war daher die Synthese von Markierungsreagenzien, die nicht nur eine effiziente Zellmarkierung ermöglichen, sondern einen synergistischen Effekt hinsichtlich des modalitätsübergreifenden Einsatzes in den bildgebenden Verfahren MRT und Laser-Ablation(LA)-ICP-MS erlauben. Die MRT-Bildgebung ermöglicht die nicht invasive Nachverfolgung markierter Zellen in vivo und die LA-ICP-MS die anschließende ex vivo Analytik zur Darstellung der Elementverteilung (Bioimaging) in einer Biopsieprobe oder in einem Gewebeschnitt. Für diese Zwecke wurden zwei verschiedene Markierungsreagenzien mit dem kontrastgebenden Element Gadolinium synthetisiert. Gadolinium eignet sich aufgrund seines hohen magnetischen Moments hervorragend für die MRT-Bildgebung und da es in Biomolekülen nicht natürlich vorkommt, konnten die Reagenzien gleichermaßen für die Zellmarkierung und das Bioimaging mit der LA-ICP-MS untersucht werden. Für die Synthese eines makromolekularen Reagenzes wurde das kommerziell verfügbare Dendrimer G5-PAMAM über bifunktionelle Linker mit dem Chelator DOTA funktionalisiert, um anschließend Gadolinium zu komplexieren. Ein zweites, nanopartikuläres Reagenz wurde über eine Solvothermal-Synthese erhalten, bei der Ln:GdVO4-Nanokristalle mit einer funktionellen Polyacrylsäure(PAA)-Hülle dargestellt wurden. Die Dotierung der Ln:GdVO4-PAA Nanokristalle mit verschiedenen Lanthanoiden (Ln=Eu, Tb) zeigte ihre prinzipielle Multiplexfähigkeit in der LA-ICP-MS. Beide Markierungsreagenzien zeichneten sich durch gute Bioverträglichkeiten und r1-Relaxivitäten aus, was zudem ihr Potential für Anwendungen als präklinische „blood-pool“ MRT-Kontrastmittel belegte. Die Untersuchung der Zellmarkierung erfolgte anhand einer Tumorzelllinie und einer Stammzelllinie, wobei beide Zellarten erfolgreich intrazellulär mit beiden Reagenzien markiert wurden. Nach der Zellmarkierung veranschaulichte die in vitro MRT-Bildgebung von Zell-Phantomen eine deutlichere Kontrastverstärkung der Zellen nach der Markierung mit den Nanokristallen im Vergleich zum kommerziellen Kontrastmittel Magnevist®. Die hohe Effizienz der Zellmarkierung mit den Nanokristallen und die damit verbundenen hohen Signalintensitäten in einer einzelnen Zelle erlaubten beim Bioimaging mit der LA-ICP-MS, Messungen bis zu einer Auflösung von 4 µm Laser Spot Size. Nach der Zellmarkierung mit den DOTA(Gd3+)-funktionalisierten G5-PAMAM Dendrimeren waren hingegen Aufnahmen mit der LA-ICP-MS nur bis zu einer Auflösung von 12 µm Laser Spot Size möglich. Insgesamt waren die Ln:GdVO4-PAA Nanokristalle mit größerer Ausbeute und kostengünstiger herstellbar als die DOTA(Gd3+)-funktionalisierten G5-PAMAM Dendrimere und zeigten zudem eine effizientere Zellmarkierung. Die Ln:GdVO4-PAA Nanokristalle erscheinen somit für das Zell-Tracking als besonders vielversprechend. Darauf aufbauend wurden die Nanokristalle zur Etablierung der Antikörper-Konjugation ausgewählt, was sie für die molekulare in vivo Bildgebung sowie für die Immuno-Bildgebung von Gewebeschnitten oder Biopsie-Proben mit der LA-ICP-MS anwendbar macht.
The main focus of the present thesis was to investigate the stabilization ability of poly(ionic liquid)s (PILs) in several examples as well as develop novel chemical structures and synthetic routes of PILs. The performed research can be specifically divided into three parts that include synthesis and application of hybrid material composed of PIL and cellulose nanofibers (CNFs), thiazolium-containing PILs, and main-chain imidazolium-type PILs.
In the first chapter, a vinylimidazolium-type IL was polymerized in water in the presence of CNFs resulting in the in situ electrostatic grafting of polymeric chains onto the surface of CNFs. The synthesized hybrid material merged advantages of its two components, that is, superior mechanical strength of CNFs and anion dependent solution properties of PILs. In contrast to unmodified CNFs, the hybrid could be stabilized and processed in organic solvents enabling its application as reinforcing agent for porous polyelectrolyte membranes.
In the second part, PILs and ionic polymers containing two types of thiazolium repeating units were synthesized. Such polymers displayed counterion dependent thermal stability and solubility in organic solvents of various dielectric constants. This new class of PILs was tested as stabilizers and phase transfer agents for carbon nanotubes in aqueous and organic media, and as binder materials to disperse electroactive powders and carbon additives in solid electrode in lithium-ion batteries. The incorporation of S and N atoms into the polymeric structures make such PILs also potential precursors for S, N - co-doped carbons.
In the last chapter, reactants originating from biomass were successfully harnessed to synthesize main-chain imidazolium-type PILs. An imidazolium-type diester IL obtained via a modified Debus-Radziszewski reaction underwent transesterification with diol in a polycondensation reaction. This yielded a polyester-type PIL which CO2 sorption properties were investigated. In the next step, the modified Debus-Radziszewski reaction was further applied to synthesize main-chain PILs according to a convenient, one-step protocol, using water as a green solvent and simple organic molecules as reagents. Depending on the structure of the employed diamine, the synthesized PILs after anion exchange showed superior thermal stability with unusually high carbonization yields.
Overall, the outcome of these studies will actively contribute to the current research on PILs by introducing novel PIL chemical structures, improved synthetic routes, and new examples of stabilized materials. The synthesis of main-chain imidazolium-type PILs by a modified Debus-Radziszewski reaction is of a special interest for the future work on porous ionic liquid networks as well as colloidal PIL nanoparticles.
The development of novel programmable materials aiming to control friction in real-time holds potential to facilitate innovative lubrication solutions for reducing wear and energy losses. This work describes the integration of light-responsiveness into two lubricating materials, silicon oils and polymer brush surfaces.
The first part focusses on the assessment on 9-anthracene ester-terminated polydimethylsiloxanes (PDMS-A) and, in particular, on the variability of rheological properties and the implications that arise with UV-light as external trigger. The applied rheometer setup contains an UV-transparent quartz-plate, which enables radiation and simultaneous measurement of the dynamic moduli. UV-A radiation (354 nm) triggers the cycloaddition reaction between the terminal functionalities of linear PDMS, resulting in chain extension. The newly-formed anthracene dimers cleave by UV-C radiation (254 nm) or at elevated temperatures (T > 130 °C). The sequential UV-A radiation and thermal reprogramming over three cycles demonstrate high conversions and reproducible programming of rheological properties. In contrast, the photochemical back reaction by UV-C is incomplete and can only partially restore the initial rheological properties. The dynamic moduli increase with each cycle in photochemical programming, presumably resulting from a chain segment re-arrangement as a result of the repeated partial photocleavage and subsequent chain length-dependent dimerization. In addition, long periods of radiation cause photooxidative degradation, which damages photo-responsive functions and consequently reduces the programming range. The absence of oxygen, however, reduces undesired side reactions. Anthracene-functionalized PDMS and native PDMS mix depending on the anthracene ester content and chain length, respectively, and allow fine-tuning of programmable rheological properties. The work shows the influence of mixing conditions during the photoprogramming step on the rheological properties, indicating that material property gradients induced by light attenuation along the beam have to be considered. Accordingly, thin lubricant films are suggested as potential application for light-programmable silicon fluids.
The second part compares strategies for the grafting of spiropyran (SP) containing copolymer brushes from Si wafers and evaluates the light-responsiveness of the surfaces. Pre-experiments on the kinetics of the thermally initiated RAFT copolymerization of 2-hydroxyethyl acrylate (HEA) and spiropyran acrylate (SPA) in solution show, first, a strong retardation by SP and, second, the dependence of SPA polymerization on light. Surprisingly, the copolymerization of SPA is inhibited in the dark. These findings contribute to improve the synthesis of polar, spiropyran-containing copolymers. The comparison between initiator systems for the grafting-from approach indicates PET-RAFT superior to thermally initiated RAFT, suggesting a more efficient initiation of surface-bound CTA by light. Surface-initiated polymerization via PET-RAFT with an initiator system of EosinY (EoY) and ascorbic acid (AscA) facilitates copolymer synthesis from HEA and 5-25 mol% SPA. The resulting polymer film with a thickness of a few nanometers was detected by atomic force microscopy (AFM) and ellipsometry. Water contact angle (CA) measurements demonstrate photo-switchable surface polarity, which is attributed to the photoisomerization between non-polar spiropyran and zwitterionic merocyanine isomer. Furthermore, the obtained spiropyran brushes show potential for further studies on light-programmable properties. In this context, it would be interesting to investigate whether swollen spiropyran-containing polymers change their configuration and thus their film thickness under the influence of light. In addition, further experiments using an AFM or microtribometer should evaluate whether light-programmable solvation enables a change in frictional properties between polymer brush surfaces.
Die vorliegende Arbeit beschäftigt sich mit der Synthese und Charakterisierung mesoporöser monolithischer Silica und deren Hybridmaterialien mit Ionischen Flüssigkeiten (ILs, ionic liquids). Zur Synthese der Silicaproben wurde ein Sol-Gel-Verfahren, ausgehend von einer Präkursorverbindung wie Tetramethylorthosilicat angewendet. Der Katalysator mit der geringsten Basizität führte zum Material mit der kleinsten Porengröße und der größten spezifischen Oberfläche. Eine Kombination von porösen Silica mit ILs führt zur Materialklasse der Silica-Ionogele. Diese Hybridmaterialien verbinden die Eigenschaften eines porösen Festkörpers mit denen einer IL (Leitfähigkeit, weites elektrochemisches Fenster, gute thermische Stabilität) und bieten vielfältige Einsatzmöglichkeiten z.B. in der Katalyse- Solar- und Sensortechnik. Um diese Materialien für ihren Verwendungszweck zu optimieren, bedarf es deren umfassenden Charakterisierung. Daher wurde in der vorliegenden Arbeit das thermische Verhalten von Silica-Ionogelen unter Verwendung verschiedener 1-Ethyl-3-methylimidazolium [Emim]-basierter ILs untersucht. Interessanterweise zeigen die untersuchten ILs deutliche Änderungen in ihrem thermischen Verhalten, wenn diese in porösen Materialien eingeschlossen werden (Confinement). Während sich die untersuchten reinen ILs durch klar unterscheidbare Phasenübergänge auszeichnen, konnten für die entsprechenden Hybridmaterialien deutlich schwächer ausgeprägte Übergänge beobachtet werden. Einzelne Phasenübergänge wurden unterdrückt (Glas- und Kristallisationsübergänge), während z.B. Schmelzübergänge in verbreiterten Temperaturbereichen, zum Teil als einzeln getrennte Schmelzpeaks beobachtet wurden. Diese Untersuchungen belegen deutliche Eigenschaftsänderungen der ILs in eingeschränkten Geometrien. Über Festkörper-NMR-Spektroskopie konnte außerdem gezeigt werden, daß die ILs in den mesoporösen Silicamaterialien eine unerwartet hohe Mobilität aufweisen. Die ILs können als quasi-flüssig bezeichnet werden und zeigen die nach bestem Wissen höchste Mobilität, die bisher für vergleichbare Hybridmaterialien beobachtet wurde. Durch Verwendung von funktionalisierten Präkursoren, sowie der Wahl der Reaktionsbedingungen, kann die Oberfläche der Silicamaterialien chemisch funktionalisiert werden und damit die Materialeigenschaften in der gewünschten Weise beeinflußt werden. In der vorliegenden Arbeit wurde der Einfluß der Oberflächenfunktionalität auf das thermische Verhalten hin untersucht. Dazu wurden zwei verschiedene Möglichkeiten der Funktionalisierung angewendet und miteinander verglichen. Bei der in-situ-Funktionalisierung wird die chemische Funktionalität während der Sol-Gel-Synthese über ein entsprechend funktionalisiertes Silan mit in das Silicamaterial einkondensiert. Eine postsynthetische Funktionalisierung erfolgt durch Reaktion der Endgruppen eines Silicamaterials mit geeigneten Reaktionspartnern. Um den Einfluß der physikalischen Eigenschaften der Probe auf die Reaktion zu untersuchen, wurden pulverisierte und monolithische Silicamaterialien miteinander verglichen. Im letzten Teil der Arbeit wurde die Vielfältigkeit, mit der Silicamaterialien postsynthetisch funktionalisiert werden können demonstriert. Durch die Kenntnis von Struktur-Eigenschaftsbeziehungen können die Eigenschaften von Silica-Ionogelen durch die geeignete Kombination von fester und mobiler Phase in der gewünschten Weise verändert werden. Die vorliegende Arbeit soll einen Beitrag zur Untersuchung dieser Beziehungen leisten, um das Potential dieser interessanten Materialien für Anwendungen nutzen zu können.
Within this work, three physicochemical methods for the hydrophobization of initially hydrophilic solid particles are investigated. The modified particles are then used for the stabilization of oil-in-water (o/w) emulsions. For all introduced methods electrostatic interactions between strongly or weakly charged groups in the system are es-sential. (i) Short chain alkylammonium bromides (C4 – C12) adsorb on oppositely charged solid particles. Macroscopic contact angle measurements of water droplets under air and hexane on flat silica surfaces in dependency of the surface charge density and alkylchain-length allow the calculation of the surface energy and give insights into the emulsification properties of solid particles modified with alkyltrimethylammonium bromides. The measure-ments show an increase of the contact angle with increasing surface charge density, due to the enhanced adsorp-tion of the oppositely charged alkylammonium bromides. Contact angles are higher for longer alkylchain lengths. The surface energy calculations show that in particular the surface-hexane or surface-air interfacial en-ergy is being lowered upon alkylammonium adsorption, while a significant increase of the surface-water interfa-cial energy occurs only at long alkyl chain lengths and high surface charge densities. (ii) The thickness and the charge density of an adsorbed weak polyelectrolyte layer (e.g. PMAA, PAH) influence the wettability of nanoparticles (e.g. alumina, silica, see Scheme 1(b)). Furthermore, the isoelectric point and the pH range of colloidal stability of particle-polyelectrolyte composites depend on the thickness of the weak polye-lectrolyte layer. Silica nanoparticles with adsorbed PAH and alumina nanoparticles with adsorbed PMAA be-come interfacially active and thus able to stabilize o/w emulsions when the degree of dissociation of the polye-lectrolyte layer is below 80 %. The average droplet size after emulsification of dodecane in water depends on the thickness and the degree of dissociation of the adsorbed PE-layer. The visualization of the particle-stabilized o/w emulsions by cryogenic SEM shows that for colloidally stable alumina-PMAA composites the oil-water interface is covered with a closely packed monolayer of particles, while for the colloidally unstable case closely packed aggregated particles deposit on the interface. (iii) By emulsifying a mixture of the corrosion inhibitor 8-hydroxyquinoline (8-HQ) and styrene with silica nanoparticles a highly stable o/w emulsion can be obtained in a narrow pH window. The amphoteric character of 8-HQ enables a pH dependent electrostatic interaction with silica nanoparticles, which can render them interfa-cially active. Depending on the concentration and the degree of dissociation of 8-HQ the adsorption onto silica results from electrostatic or aromatic interactions between 8-HQ and the particle-surface. At intermediate amounts of adsorbed 8-HQ the oil wettability of the particles becomes sufficient for stabilizing o/w emulsions. Cryogenic SEM visualization shows that the particles arrange then in a closely packed shell consisting of partly of aggregated domains on the droplet interface. For further increasing amounts of adsorbed 8-HQ the oil wet-tability is reduced again and the particles ability to stabilize emulsions decreases. By the addition of hexadecane to the oil phase the size of the droplets can be reduced down to 200 nm by in-creasing the silica mass fraction. Subsequent polymerization produces corrosion inhibitor filled (20 wt-%) poly-styrene-silica composite particles. The measurement of the release of 8-hydroxyquinoline shows a rapid increase of 8-hydroxyquinoline in a stirred aqueous solution indicating the release of the total content in less than 5 min-utes. The method is extended for the encapsulation of other organic corrosion inhibitors. The silica-polymer-inhibitor composite particles are then dispersed in a water based alkyd emulsion, and the dispersion is used to coat flat aluminium substrates. After drying and cross-linking the polmer-film Confocal Laser Scanning Micros-copy is employed revealing a homogeneous distribution of the particles in the film. Electrochemical Impedance Spectroscopy in aqueous electrolyte solutions shows that films with aggregated particle domains degrade with time and don’t provide long-term corrosion protection of the substrate. However, films with highly dispersed particles have high barrier properties for corrosive species. The comparison of films containing silica-polystyrene composite particles with and without 8-hydroxyquinoline shows higher electrochemical impedances when the inhibitor is present in the film. By applying the Scanning Vibrating Electrode Technique the localized corrosion rate in the fractured area of scratched polymer films containing the silica-polymer-inhibitor composite particles is studied. Electrochemical corrosion cannot be suppressed but the rate is lowered when inhibitor filled composite particles are present in the film. By depositing six polyelectrolyte layers on particle stabilized emulsion droplets their surface morphology changes significantly as shown by SEM visualization. When the oil wettability of the outer polyelectrolyte layer increases, the polyelectrolyte coated droplets can act as emulsion stabilizers themselves by attaching onto bigger oil droplets in a closely packed arrangement. In the presence of 3 mM LaCl3 8-HQ hydrophobized silica particles aggregate strongly on the oil-water inter-face. The application of an ultrasonic field can remove two dimensional shell-compartments from the droplet surface, which are then found in the aqueous bulk phase. Their size ranges up to 1/4th of the spherical particle shell.
Die Kombination von Polymeren mit Peptiden vereint die Eigenschaften beider Stoffklassen miteinander. Dabei können die strukturbildenden Eigenschaften der Peptide genutzt werden, um Polymere zu organisieren. In der vorliegenden Arbeit wurde ein Polymer-Peptid-Konjugat verwendet, das sich in Wasser zu Bändern anordnet. Die treibende Kraft für diesen Prozess ist die Anordnung des Peptidteils zu β-Faltblattstrukturen. Das Polymer-Peptid-Aggregat besitzt einen Peptidkern mit funktionalen Oberflächen, der lateral von einer Polyethylenoxidschale umgeben ist. Durch Änderung der Peptidsequenz war es bisher möglich, die Eigenschaften dieser Fasern zu variieren. In der Arbeit wird ein modularer Ansatz zur vielfältigen Modifizierung einer Polymer-Peptid-Faser entwickelt. So ist es möglich, die Eigenschaften der Fasern einzustellen, ohne die strukturbildende β-Faltblattsequenz verändern zu müssen. Um weitere Funktionen an den Fasern anzubringen, wurde die 1,3-dipolaren Addition verwendet. Diese Reaktion beschreibt die konzertierte Umlagerung eines Azides mit einem Alkin. Sie ist in den meisten Lösungsmitteln unter hohen Ausbeuten durchführbar. Im Rahmen der Arbeit wird die Erzeugung von Aziden untersucht und auf die Polymer-Peptid-Fasern übertragen. Der Diazotransfer stellte dabei die Methode der Wahl dar, so können Azidgruppen aus Aminen gewonnen werden. Unter Verwendung der 1,3-dipolaren Addition konnten verschiedene alkinfunktionale Moleküle kovalent an die azidfunktionalisierten Polymer-Peptid-Fasern gebunden werden. So wurde ein Fluoreszenzfarbstoff an die Fasern gebunden, der eine Abbildung der Fasern mittels konfokaler Mikroskopie erlaubte. Weiterhin wurden die Eigenschaften der Fasern durch Addition dreier carboxylfunktionaler Moleküle modifiziert. Diese Fasern konnten weiter genutzt werden, um Kalzium zu binden. Dabei variierte die Anzahl der gebundenen Kalziumionen in Abhängigkeit der jeweiligen Fasermodifikation erheblich. Weitere Untersuchungen, die Morphologie von Kalziumcarbonatkristallen betreffend, werden aktuell durchgeführt. Die kovalente Anbringung eines reduzierenden Zuckers an die Polymer-Peptid-Fasern erlaubt die Abscheidung von Silber aus Tollens Reagenz. Durch eine Entwicklung analog zur Schwarz-Weiss-Photographie können in nachfolgenden Arbeiten so Silberdrähte in Nanogröße erzeugt werden. An die azidfunktionalen Fasern können weitere funktionale Moleküle angebracht werden, um die Eigenschaften und das Anwendungsspektrum der Polymer-Peptid-Fasern zu erweitern.
Im Rahmen einer explorativen Entwicklung wurde in der vorliegenden Studie ein Konzept zur Wissenschaftskommunikation für ein Graduiertenkolleg, in dem an photochemischen Prozessen geforscht wird, erstellt und anschließend evaluiert. Der Grund dafür ist die immer stärker wachsende Forderung nach Wissenschaftskommunikation seitens der Politik. Es wird darüber hinaus gefordert, dass die Kommunikation der eigenen Forschung in Zukunft integrativer Bestandteil des wissenschaftlichen Arbeitens wird. Um junge Wissenschaftler bereits frühzeitig auf diese Aufgabe vorzubereiten, wird Wissenschaftskommunikation auch in Forschungsverbünden realisiert.
Aus diesem Grund wurde in einer Vorstudie untersucht, welche Anforderungen an ein Konzept zur Wissenschaftskommunikation im Rahmen eines Forschungsverbundes gestellt werden, indem die Einstellung der Doktoranden zur Wissenschaftskommunikation sowie ihre Kommunikationsfähigkeiten anhand eines geschlossenen Fragebogens evaluiert wurden. Darüber hinaus wurden aus den Daten Wissenschaftskommunikationstypen abgeleitet. Auf Grundlage der Ergebnisse wurden unterschiedliche Wissenschaftskommunikationsmaßnahmen entwickelt, die sich in der Konzeption, den Rezipienten, sowie der Form der Kommunikation und den Inhalten unterscheiden.
Im Rahmen dieser Entwicklung wurde eine Lerneinheit mit Bezug auf die Inhalte des Graduiertenkollegs, bestehend aus einem Lehr-Lern-Experiment und den dazugehörigen Begleitmaterialien, konzipiert. Anschließend wurde die Lerneinheit in eine der Wissenschaftskommunikationsmaßnahmen integriert. Je nach Anforderung an die Doktoranden, wurden die Maßnahmen durch vorbereitende Workshops ergänzt.
Durch einen halboffenen Pre-Post-Fragebogen wurde der Einfluss der Wissenschaftskommunikationsmaßnahmen und der dazugehörigen Workshops auf die Selbstwirksamkeit der Doktoranden evaluiert, um Rückschlüsse darauf zu ziehen, wie sich die Wahrnehmung der eigenen Kommunikationsfähigkeiten durch die Interventionen verändert. Die Ergebnisse deuten darauf hin, dass die einzelnen Wissenschaftskommunikationsmaßnahmen die verschiedenen Typen in unterschiedlicher Weise beeinflussen. Es ist anzunehmen, dass es abhängig von der eigenen Einschätzung der Kommunikationsfähigkeit unterschiedliche Bedürfnisse der Förderung gibt, die durch dedizierte Wissenschaftskommunikationsmaßnahmen berücksichtigt werden können.
Auf dieser Grundlage werden erste Ansätze für eine allgemeingültige Strategie vorgeschlagen, die die individuellen Fähigkeiten zur Wissenschaftskommunikation in einem naturwissenschaftlichen Forschungsverbund fördert.
Ammonia is a chemical of fundamental importance for nature`s vital nitrogen cycle. It is crucial for the growth of living organisms as well as food and energy source. Traditionally, industrial ammonia production is predominated by Haber- Bosch process (HBP) which is based on direct conversion of N2 and H2 gas under high temperature and high pressure (~500oC, 150-300 bar). However, it is not the favorite route because of its thermodynamic and kinetic limitations, and the need for the energy intense production of hydrogen gas by reforming processes. All these disfavors of HBP open a target to search for an alternative technique to perform efficient ammonia synthesis via electrochemical catalytic processes, in particular via water electrolysis, using water as the hydrogen source to save the process from gas reforming.
In this study, the investigation of the interface effects between imidazolium-based ionic liquids and the surface of porous carbon materials with a special interest in the nitrogen absorption capability. As the further step, the possibility to establish this interface as the catalytically active area for the electrochemical N2 reduction to NH3 has been evaluated. This particular combination has been chosen because the porous carbon materials and ionic liquids (IL) have a significant importance in many scientific fields including catalysis and electrocatalysis due to their special structural and physicochemical properties. Primarily, the effects of the confinement of ionic liquid (EmimOAc, 1-Ethyl-3-methylimidazolium acetate) into carbon pores have been investigated. The salt-templated porous carbons, which have different porosity (microporous and mesoporous) and nitrogen species, were used as model structures for the comparison of the IL confinement at different loadings. The nitrogen uptake of EmimOAc can be increased by about 10 times by the confinement in the pores of carbon materials compared to the bulk form. In addition, the most improved nitrogen absorption was observed by IL confinement in micropores and in nitrogen-doped carbon materials as a consequence of the maximized structural changes of IL. Furthermore, the possible use of such interfaces between EmimOAc and porous carbon for the catalytic activation of dinitrogen during the kinetically challenging NRR due to the limited gas absorption in the electrolyte, was examined. An electrocatalytic NRR system based on the conversion of water and nitrogen gas to ammonia at ambient operation conditions (1 bar, 25 °C) was performed in a setup under an applied electric potential with a single chamber electrochemical cell, which consists of the combination of EmimOAc electrolyte with the porous carbon-working electrode and without a traditional electrocatalyst. Under a potential of -3 V vs. SCE for 45 minutes, a NH3 production rate of 498.37 μg h-1 cm-2 and FE of 12.14% were achieved. The experimental observations show that an electric double-layer, which serves the catalytically active area, occurs between a microporous carbon material and ions of the EmimOAc electrolyte in the presence of sufficiently high provided electric potential. Comparing with the typical NRR systems which have been reported in the literature, the presented electrochemical ammonia synthesis approach provides a significantly higher ammonia production rate with a chance to avoid the possible kinetic limitations of NRR. In terms of operating conditions, ammonia production rate and the faradic efficiency without the need for any synthetic electrocatalyst can be resulted of electrocatalytic activation of nitrogen in the double-layer formed between carbon and IL ions.
Die vorliegende Arbeit beschäftigt sich mit der Synthese monodisperser, multifunktionaler Poly(amidoamine) (PAAs). Die Klasse der PAAs ist besonders interessant für eine Anwendung im Bereich der Biomedizin, da sie meist nicht toxisch ist, eine sehr geringe Immunogenizität zeigt und eine erhöhte Zellmembranpermeabilität besitzt. Allerdings ist der Einsatz linearer PAAs bisher limitiert, da ihre Synthese nur den Zugang von hoch-polydispersen Systemen mit einer streng alternierenden oder statistischen Verteilung von Funktionalitäten erlaubt. Es ist daher von großem Interesse diese Polymerklasse durch die Möglichkeit eines sequenzdefinierten Aufbaus und der Integration von neuen Funktionalitäten zu verbessern. Um dies zu ermöglichen, wurden, vergleichbar mit der etablierten Festphasensynthese von Peptiden, schrittweise funktionale Disäure- und Diamin-Bausteine an ein polymeres Träger-Harz addiert. Der sequenzielle Aufbau ermöglicht die Synthese monodisperser PAAs und die Kontrolle über die Monomersequenz. Die Wahl der Monomer-Bausteine und ihrer Funktionalitäten kann dabei für jede Addition neu getroffen werden und entscheidet so über die Sequenz der Funktionalitäten im Polymerrückgrat. Die verwendete Chemie entspricht dabei der Standardpeptidchemie, so dass mit Hilfe eines Peptidsynthese-Automaten die Synthese vollständig automatisiert werden konnte. Die Verwendung spezieller Trägerharze, die bereits mit einem synthetischen Polymerblock wie PEO oder auch mit einem Peptid vorbeladen waren, erlaubt die direkte Synthese von PEO- und Peptid-PAA Blockcopolymeren. Da die hier dargestellten PAAs später auf ihre Eignung als multivalente Polykationen in der Gentherapie getestet werden sollten, wurden zunächst Bausteine gewählt, die den Einbau verschiedener Aminfunktionalitäten ermöglichen. Die Bausteine müssen dabei so gewählt sein, dass sie kompatibel sind mit der Chemie des Peptidsynthesizers und eine quantitative Addition ohne Neben- oder Abbruchreaktionen garantieren. Darüber hinaus ist der Einbau von Peptidsequenzen und Disulfid-Einheiten in die PAA-Kette möglich, die z. B. für einen selektiven Abbau des Polymers im Organismus genutzt werden können. Zusammenfassend lässt sich feststellen, dass die in dieser Arbeit vorgestellten PAA-Systeme großes Potenzial als nicht-virale Vektoren für die Gentransfektion bieten. Sie sind nicht toxisch und zeigen Zellaufnahme-Effizienzen von bis zu 77%. Die Gentransfereffizienz ist im Vergleich zu etablierten Polymer-Vektoren zwar noch sehr gering, aber die bisherigen Versuche zeigen bereits eine mögliche Ursache, nämlich die schlechte Freisetzung des Genmaterials innerhalb der Zelle. Eine Lösung dieses Problems bietet jedoch die weitere Modifizierung der PAA-Systeme durch den Einbau von Sollbruchstellen. Diese Sollbruchstellen ermöglichen einen programmierten Abbau des Polymers innerhalb der Zelle und damit sollte die Freisetzung des Genmaterials vom Träger deutlich erleichtert werden. Mögliche Bruchstellen sind z. B. enzymatisch gezielt spaltbare Peptideinheiten oder Disulfid-Einheiten, wie sie bereits als Bausteine für die PAA-Synthese vorgestellt wurden (vergl. Kapitel 4.4). Da nur innerhalb der Zelle ein reduzierendes elektrochemisches Potential besteht, werden z. B. Disulfid-Einheiten auch nur dort gespalten und bieten außerhalb der Zelle ausreichende Stabilität zum Erhalt der Polyplexstruktur. Neben einer Anwendung in der Gentherapie bieten die hier vorgestellten PAA-Systeme den Vorteil einer systematischen Untersuchung von Struktur-Eigenschafts-Beziehungen der Polyplexe. Es wurden verschiedene Zusammenhänge zwischen der chemischen Struktur der PAA-Segmente und der Art und Stärke der DNS-Komprimierung aufgezeigt. Die Komprimierungsstärke wiederum zeigte deutlichen Einfluss auf die Internalisierungsrate und damit auch Transfektionseffizienz. Darüber hinaus zeigte sich ein drastischer Einfluss des PEO-Blocks auf die Stabilisierung der Polyplexe sowie deren intrazelluläre Freisetzung bei Zusatz von Chloroquin. Dennoch bleiben aufgrund der Komplexität der Zusammenhänge noch viele Mechanismen der Transfektion unverstanden, und es muss Aufgabe folgender Arbeiten sein, das Potential der hier eingeführten monodispersen PAA-Systeme weiter auszuloten. So wäre z. B. eine Korrelation der Kettenlänge mit den Parametern der Polyplexbildung, der Zellaufnahme und Transfektionseffizienz von großem Interesse. Darüber hinaus bietet der Einbau von Sollbruchstellen wie kurzen Peptidsequenzen oder den hier bereits eingeführten Disulfid-Einheiten neue Möglichkeiten der gezielten Freisetzung und des programmierten Abbaus, die näher untersucht werden müssen. Neben der Anwendung im Bereich der Gentransfektion sind außerdem andere Gebiete für den Einsatz von monodispersen multifunktionalen PAAs denkbar, da diese kontrollierbare und einstellbare Wechselwirkungen ermöglichen.
The optical properties of chromophores, especially organic dyes and optically active inorganic molecules, are determined by their chemical structures, surrounding media, and excited state behaviors. The classical optical go-to techniques for spectroscopic investigations are absorption and luminescence spectroscopy. While both techniques are powerful and easy to apply spectroscopic methods, the limited time resolution of luminescence spectroscopy and its reliance on luminescent properties can make its application, in certain cases, complex, or even impossible. This can be the case when the investigated molecules do not luminesce anymore due to quenching effects, or when they were never luminescent in the first place. In those cases, transient absorption spectroscopy is an excellent and much more sophisticated technique to investigate such systems. This pump-probe laser-spectroscopic method is excellent for mechanistic investigations of luminescence quenching phenomena and photoreactions. This is due to its extremely high time resolution in the femto- and picosecond ranges, where many intermediate or transient species of a reaction can be identified and their kinetic evolution can be observed. Furthermore, it does not rely on the samples being luminescent, due to the active sample probing after excitation. In this work it is shown, that with transient absorption spectroscopy it was possible to identify the luminescence quenching mechanisms and thus luminescence quantum yield losses of the organic dye classes O4-DBD, S4-DBD, and pyridylanthracenes. Hence, the population of their triplet states could be identified as the competitive mechanism to their luminescence. While the good luminophores O4-DBD showed minor losses, the S4-DBD dye luminescence was almost entirely quenched by this process. However, for pyridylanthracenes, this phenomenon is present in both the protonated and unprotonated forms and moderately effects the luminescence quantum yield. Also, the majority of the quenching losses in the protonated forms are caused by additional non-radiative processes introduced by the protonation of the pyridyl rings. Furthermore, transient absorption spectroscopy can be applied to investigate the quenching mechanisms of uranyl(VI) luminescence by chloride and bromide. The reduction of the halides by excited uranyl(VI) leads to the formation of dihalide radicals X^(·−2). This excited state redox process is thus identified as the quenching mechanism for both halides, and this process, being diffusion-limited, can be suppressed by cryogenically freezing the samples or by observing these interactions in media with a lower dielectric constant, such as ACN and acetone.
The self-assembly of amphiphilic polymers in aqueous systems is important for a plethora of applications, in particular in the field of cosmetics and detergents. When introducing thermoresponsive blocks, the aggregation behavior of these polymers can be controlled by changing the temperature. While confined to simple diblock copolymer systems for long, the complexity - and thus the versatility - of such smart systems can be strongly enlarged, once designed monomers, specific block sizes, different architectures, or additional functional groups such as hydrophobic stickers are implemented. In this work, the structure-property relationship of such thermoresponsive amphiphilic block copolymers was investigated by varying their structure systematically. The block copolymers were generally composed of a permanently hydrophobic sticker group, a permanently hydrophilic block, and a thermoresponsive block exhibiting a Lower Critical Solution Temperature (LCST) behavior. While the hydrophilic block consisted of N,N dimethylacrylamide (DMAm), different monomers were used for the thermoresponsive block, such as N n propylacrylamide (NPAm), N iso propylacrylamide (NiPAm), N,N diethylacrylamide (DEAm), N,N bis(2 methoxyethyl)acrylamide (bMOEAm), or N acryloylpyrrolidine (NAP) with different reported LCSTs of 25, 32, 33, 42 and 56 °C, respectively. The block copolymers were synthesized by successive reversible addition fragmentation chain transfer (RAFT) polymerization. For the polymers with the basic linear, the twinned hydrophobic and the symmetrical quasi miktoarm architectures, the results were well defined block sizes and end groups as well as narrow molar mass distributions (Ɖ ≤ 1.3). More complex architectures, such as the twinned thermoresponsive and the non-symmetrical quasi miktoarm one, were achieved by combining RAFT polymerization with a second technique, namely atom transfer radical polymerization (ATRP) or single unit monomer insertion (SUMI), respectively. The obtained block copolymers showed well defined block sizes, but due to the complexity of these reaction paths, the dispersities were generally higher (Ɖ ≤ 1.8) and some end groups were lost.
The thermoresponsive behavior of the block copolymers was investigated by turbidimetry and dynamic light scattering (DLS). Below the phase transition temperature, the polymers were soluble in water and small micellar structures were visible. However, above the phase transition temperature, the aggregation behavior was strongly dependent on the architecture and the chemical structure of the thermoresponsive block. Thermoresponsive blocks comprising PNAP and PbMOEAm with DPn = 40 showed no cloud point (CP), since their already high LCSTs were further increased by the attached hydrophilic block. Depending on the architecture as well as on the block size, block copolymers with PNiPAm, PDEAm and PNPAm showed different CP’s. Large aggregates were visible for block copolymers with PNiPAm and PDEAm above their CP. For PNPAm containing block copolymers, the phase transition was very sensitive towards the architecture resulting in either small or large aggregates.
In addition, fluorescence studies were performed using PDMAm and PNiPAm homo and block copolymers with linear architecture, functionalized with complementary fluorescence dyes introduced at the opposite chain ends. The thermoresponsive behavior was studied in pure aqueous solution as well as in an oil in water (o/w) microemulsion. The findings indicate that the block copolymer behaves as polymeric surfactant at low temperatures, with one relatively small hydrophobic end group and an extended hydrophilic chain forming ‘hairy micelles’ similar as the other synthesized architectures. Above the phase transition temperature of the PNiPAm block, however, the copolymer behaves as associative telechelic polymer with two non-symmetrical hydrophobic end groups, which do not mix. Thus, instead of a network of bridged ‘flower micelles’, large dynamic aggregates are formed. These are connected alternatingly by the original micellar cores as well as by clusters of the collapsed PNiPAm blocks. This type of bridged micelles is even more favored in the o/w microemulsion than in pure aqueous solution.
