The global drylands cover nearly half of the terrestrial surface and are home to more than two billion people. In many drylands, ongoing land-use change transforms near-natural savanna vegetation to agricultural land to increase food production. In Southern Africa, these heterogenous savanna ecosystems are also recognized as habitats of many protected animal species, such as elephant, lion and large herds of diverse herbivores, which are of great value for the tourism industry. Here, subsistence farmers and livestock herder communities often live in close proximity to nature conservation areas. Although these land-use transformations are different regarding the future they aspire to, both processes, nature conservation with large herbivores and agricultural intensification, have in common, that they change the vegetation structure of savanna ecosystems, usually leading to destruction of trees, shrubs and the woody biomass they consist of.
Such changes in woody vegetation cover and biomass are often regarded as forms of land degradation and forest loss. Global forest conservation approaches and international programs aim to stop degradation processes, also to conserve the carbon bound within wood from volatilization into earth’s atmosphere. In search for mitigation options against global climate change savannas are increasingly discussed as potential carbon sinks. Savannas, however, are not forests, in that they are naturally shaped by and adapted to disturbances, such as wildfires and herbivory. Unlike in forests, disturbances are necessary for stable, functioning savanna ecosystems and prevent these ecosystems from forming closed forest stands. Their consequently lower levels of carbon storage in woody vegetation have long been the reason for savannas to be overlooked as a potential carbon sink but recently the question was raised if carbon sequestration programs (such as REDD+) could also be applied to savanna ecosystems. However, heterogenous vegetation structure and chronic disturbances hamper the quantification of carbon stocks in savannas, and current procedures of carbon storage estimation entail high uncertainties due to methodological obstacles. It is therefore challenging to assess how future land-use changes such as agricultural intensification or increasing wildlife densities will impact the carbon storage balance of African drylands.
In this thesis, I address the research gap of accurately quantifying carbon storage in vegetation and soils of disturbance-prone savanna ecosystems. I further analyse relevant drivers for both ecosystem compartments and their implications for future carbon storage under land-use change. Moreover, I show that in savannas different carbon storage pools vary in their persistence to disturbance, causing carbon bound in shrub vegetation to be most likely to experience severe losses under land-use change while soil organic carbon stored in subsoils is least likely to be impacted by land-use change in the future.
I start with summarizing conventional approaches to carbon storage assessment and where and for which reasons they fail to accurately estimated savanna ecosystem carbon storage. Furthermore, I outline which future-making processes drive land-use change in Southern Africa along two pathways of land-use transformation and how these are likely to influence carbon storage. In the following chapters, I propose a new method of carbon storage estimation which is adapted to the specific conditions of disturbance-prone ecosystems and demonstrate the advantages of this approach in relation to existing forestry methods. Specifically, I highlight sources for previous over- and underestimation of savanna carbon stocks which the proposed methodology resolves. In the following chapters, I apply the new method to analyse impacts of land-use change on carbon storage in woody vegetation in conjunction with the soil compartment. With this interdisciplinary approach, I can demonstrate that indeed both, agricultural intensification and nature conservation with large herbivores, reduce woody carbon storage above- and belowground, but partly sequesters this carbon into the soil organic carbon stock. I then quantify whole-ecosystem carbon storage in different ecosystem compartments (above- and belowground woody carbon in shrubs and trees, respectively, as well as topsoil and subsoil organic carbon) of two savanna vegetation types (scrub savanna and savanna woodland). Moreover, in a space-for-time substitution I analyse how land-use changes impact carbon storage in each compartment and in the whole ecosystem. Carbon storage compartments are found to differ in their persistence to land-use change with carbon bound in shrub biomass being least persistent to future changes and subsoil organic carbon being most stable under changing land-use. I then explore which individual land-use change effects act as drivers of carbon storage through Generalized Additive Models (GAMs) and uncover non-linear effects, especially of elephant browsing, with implications for future carbon storage. In the last chapter, I discuss my findings in the larger context of this thesis and discuss relevant implications for land-use change and future-making decisions in rural Africa.
Das vorliegende Buch vergleicht Strategien biologischer Systeme mit militärischen Strategien. Die zentrale Fragestellung ist dabei darauf gerichtet, ob es neben systemischen Gemeinsamkeiten auch gemeinsame oder ähnliche Strukturmuster und ähnliche Prozessabläufe beispielsweise sowohl im biologischen Abwehrmechanismus des Immunsystems und bei Insektenstaaten als auch bei Prozessen im Militär gibt. Vor diesem Hintergrund klaffen in der Theorie der Strategie, speziell in den Militärwissenschaften Lücken, denn der Systemansatz wird nicht konsequent beachtet, wie in diesem Buch mehrfach nachgewiesen ist. Von einem allgemeinen Verständnis der Strategie als bewusstem planerischem Vorgehen ist Abstand zu nehmen. Ausgehend von der Methode der Analogie und des Vergleichs wird im theoretischen Teil dieses Buches die Allgemeine Systemtheorie erläutert. Dabei werden der Begriff der Strategie ebenso wie die Begriffe Struktur und Prozess und Ansätze aus der Kriegsphilosophie von Clausewitz untersucht. Den Ausgangspunkt und schließlich auch wieder den Endpunkt der Überlegungen bilden neben dem notwendigen weiten Verständnis von Strategie, vor allem der Begriff der Organisation, ihrer Umwelt und der in diesem Zusammenhang bestehenden Wechselwirkung. Sowohl die Wechselwirkung von Umwelt und System als auch ihre Abhängigkeit durch strukturelle Kopplung werden beschrieben. Das Zusammenspiel und die daraus entstehende Komplexität der fünf Komponenten der Wahrnehmung, der Information und der Führung im Zusammenhang der Komponenten von Raum und Zeit in einem sozialen System lassen die klassische Ziel-Mittel-Zweck-Beziehung Clausewitz´scher Strategiedefinition verkürzt erscheinen. Anhand eines kurzen Rekurses der Methoden der Sozialen Netzwerkanalyse (SNA) wird der breite und tiefgehende Analyserahmen der Messung und Transparenzerreichung in Organisationen vorgestellt. Die SNA wird als Ausprägung der Netzwerk- und Graphentheorie, in die Allgemeine Systemtheorie integriert. Sie bildet eine zukunftsweisende Methode der Untersuchung von Netzwerken wie etwa dem Internet (Facebook, Xing etc.). Der aufgezeigte Theorierahmen bildet dabei zugleich eine Methode für den Systemvergleich und kann als Vorgehensmodell künftiger Strategieentwicklung genutzt werden. Der anschließende Systemvergleich wird mit mehreren Beispielen durchgeführt. Ausgehend von der Zelle als Grundeinheit werden Strukturen und Prozesse des Immunsystems mit solchen in militärischen Strukturen, weil sie im Lauf der Evolution enorme Leistungen in Reaktion, Anpassung und Optimierung vollbracht haben. Der Vergleich geht der Frage nach, ob in diesen Bereichen der Strategie und Organisation systemische Grundregeln existieren. Das Beispiel der Wechselwirkung zwischen Parasit und Wirt zeigt, dass jeder Fortschritt und Sieg angesichts der Systemeinbettung von Strategie nur relativ wirken kann. Die Analogie zwischen Viren und Bakterien sowie die Entwicklung des Begriffs der sozialen Mimikry führen zu einem erweiterten Verständnis der Strategie von Terroristen in sozialen Systemen. Verdeutlicht wird das Grundschema des Täuschens und Eindringens in Systeme sowie die Beeinflussung und Umsteuerung von Prozessen und Strukturen in einem System durch Kommunikation und Implementation von Codes. Am Beispiel des Immunsystems und der Bildung verschiedener Kommunikations- und Steuerungsmechanismen von Zellsystemen sowie Beispielen von Schwarmbildung und der Organisation sozialer Insekten werden eine Vielzahl heuristischer Hinweise für neue Ansätze für die Organisation von Streitkräften und ihrer Steuerung gefunden. Neben der Erarbeitung eines grundlegenden Strategiebegriffs anhand von Wahrnehmung und Selektion als Grundprozess der Erzeugung von Strategie wird eine differenzierte Betrachtung von Begriffen wie Redundanz und Robustheit sowie eine relativierende Sichtweise von Risiko, Gefahr und Schaden gewonnen. Der Vergleich mit dem Immunsystems zeigt einfache Beispiele der Informationsspeicherung und -übertragung, die zudem Bypassfähigkeiten sowie dezentrale Eskalations- und Deeskalationsprinzipien veranschaulichen. Dies eröffnet in Analogie dieser Prinzipien einen weiten Raum Sicherheitsarchitekturen zu überdenken und neu zu strukturieren. Zudem kann die räumliche Ausbreitung von Information und Kräften als ein gemeinsames Grundproblem der Entwicklung und Wirksamkeit von Strategien sowohl in der Natur, als auch im Militär identifiziert werden. Die Betrachtung zeigt zudem wie Zellen mit fehlgeleiteten Prozessen und Strukturen umgehen. Die Analogie deutet auf das Erfordernis einer Veränderung im Umgang mit Fehlern und ihrer Rückführ- und Umkehrbarkeit im weitesten Sinne. Das Buch eröffnet überdies ein neues Verständnis von Staat, Gewaltenteilung und Institutionen in einem sozialen System. Die Ergebnisse sind auch auf andere Forschungsbereiche, Organisationen und unterschiedlichste soziale Systeme übertragbar. Es eröffnet sich ein breites Anwendungsspektrum für künftige strategische Untersuchungen.
Charakterisierung von ausgewählten Protein-Phosphatasen und MATE-Proteinen aus Arabidopsis thaliana

Im ersten Teil der vorliegenden Arbeit wurde die Genexpression der Protein Phosphatase-gene TOPP1, TOPP2, TOPP5, STH1 und STH2 analysiert. Alle fünf ausgewählten Gene kodieren für PP des PP1/PP2A-Typs. Es wurde untersucht, ob homologen PP-Isoformen individuelle Expressionsmuster zugewiesen werden konnten. Besonderes Augenmerk richtete sich dabei auf die Expression von PP-Genen in den Schließzellen von A. thaliana. In mehreren Inhibitorstudien wurde beschrieben, dass PP1/PP2A-Proteine eine wichtige Rolle in der Signaltransduktion pflanzlicher Schließzellen spielen. Bisher konnte allerdings noch keine der entsprechenden katalytischen Untereinheiten auf molekularer Ebene identifiziert werden. Im Rahmen dieser Arbeit wurde zum ersten Mal nachgewiesen, dass mit TOPP1 ein Protein des PP1-Typs präferenziell in den Schließzellen von A. thaliana exprimiert wird. Ein Vergleich der Genexpression von TOPP1, TOPP2 und TOPP5 zeigte für die drei homologen Gene sehr Isoform-spezifische Expressionsmuster. Dies war ein deutlicher Hinweis, dass diese eng verwandten PP trotz großer Übereinstimmung auf Aminosäureebene vermutlich unterschiedliche Funktionen in planta haben. Die Untersuchung der Genexpression von STH1 und STH2 zeigte, dass die fast identischen Proteine zum Teil in unterschiedlichen Geweben vorkommen. Die Transkripte der beiden Gene, welche eine eigene Untergruppe von PP2A-verwandten Sequenzen bilden, konnten aus EF isoliert werden. Der in dieser Arbeit entwickelte Screeningansatz ermöglichte es, die sehr ähnlichen cDNA-Fragmente eindeutig voneinander zu unterscheiden. Die gefundenen Isoform-spezifischen Expressionsmuster waren ein deutlicher Hinweis auf unterschiedliche Funktionen in planta. Zur weiteren Untersuchung der PP-Funktionen in planta wurden Pflanzen mit veränderter Genaktivität von TOPP2 oder STH1 untersucht. In Pflanzen mit RNAi-vermittelter Reduktion des TOPP2-Transkriptgehalts ließ sich ein deutlich verändertes Blattwachstum beobachten. Die eingerollten oder asymmetrisch entwickelten Blätter waren vermutlich ein Hinweis, dass diese PP1-Isoform auch in A. thaliana eine Rolle bei der Zellteilung spielt. Für TOPP2-Expression in Hefen wurde diese Funktion schon nachgewiesen. Die Analyse von Insertions-mutanten mit T-DNA Insertionen in beiden STH1-Allelen waren neben den Expressions-studien ein weiterer Hinweis, dass sich STH1 nicht funktionell durch STH2 ersetzen lässt. Die Experimente in dieser Arbeit zeigten, dass das Fehlen der STH1-Genaktivität zu einem deutlichen Blattphänotyp mit gezahnten Blatträndern führte. Für STH2-Insertionsmutanten wurde dieses veränderte Wachstum nicht beschrieben. Im zweiten Teil dieser Arbeit wurde das Gen NIC1, welches für ein MATE-Membranprotein kodiert, identifiziert und charakterisiert. Die Sequenzierung des Genoms von A. thaliana hatte gezeigt, dass mindestens 56 MATE-Gene in dieser Pflanze vorhanden sind. Zum Zeitpunkt der Identifikation von NIC1 war keines dieser Gene charakterisiert. Außer für das MATE-Protein ERC1 aus S. cerevisiae gab es keine Studien zu eukaryotischen Mitgliedern dieser großen Familie von Membranproteinen. Anhand NIC1 wurden heterologe Expressionssysteme zur funktionellen Charakterisierung von MATE-Proteinen aus Pflanzen etabliert. Die cDNA von NIC1 wurde nach ihrer Klonierung in X. laevis Oozyten und S. cerevisiae exprimiert. In S. cerevisiae erhöhte die NIC1-Expression die Lithumtoleranz der Hefen und führte zu einer Verminderung der Natriumtoleranz. Parallele Versuche mit NIC2 und NIC4 (in den Diplomarbeiten von Blazej Dolniak und Mandy Kursawe) zeigten, dass auch diese beiden Proteine die Salztoleranz von S. cerevisiae beeinflussten. Während NIC2 die Lithium- und Natriumtoleranz erhöhte, führte NIC4-Expresion zu einer höheren Sensibilität gegenüber diesen beiden Kationen. Die unterschiedlichen Eigenschaften der drei homologen Proteine zeigten sich auch bei ihrer Expression in X. laevis Oozyten. NIC1 induzierte in den Oozyten auswärts gerichtete Chloridströme, die spannungsabhängig waren und durch mikromolare Konzentrationen der trivalenten Kationen Lanthan oder Gadolinium inhibiert werden konnten. NIC4 induzierte Barium-inhibierbare Kaliumströme, die spannungsunabhängig waren. Für NIC2 ließ sich in diesem Expressionssystem keine Aktivität detektieren. Zur Untersuchung der NIC1-Funktion in planta wurde die Genaktivität in transgenen Pflanzen lokalisiert und reduziert. Die NIC1-Genexpression war hauptsächlich in den vaskulären Geweben der Pflanze detektierbar und einer Verminderung des NIC1-Transkriptgehalts beeinflusste die Entwicklungsgeschwindigkeit der Pflanzen. Sie entwickelten sich deutlich langsamer als der parallel kultivierte Wildtyp. Der deutliche Phänotyp bei Veränderung der Genaktivität von nur einem der mindestens 56 vorhandenen MATE-Gene in A. thaliana zeigte, dass vermutlich keine weitere MATE-Isoform in der Lage ist, die Funktion von NIC1 zu übernehmen.
