The paired salivary glands in the cockroach are composed of acini with ion-transporting peripheral P-cells and protein-secreting central C-cells, and a duct system for the modification of the primary saliva. Secretory activity is controlled by serotonergic and dopaminergic neurons, whose axons form a dense plexus on the glands. The spatial relationship of release sites for serotonin and dopamine to the various cell types was determined by anti-synapsin immunofluorescence confocal microscopy and electron microscopy. Every C-cell apparently has only serotonergic synapses on its surface. Serotonergic and dopaminergic fibres on the acini have their release zones at a distance of similar to0.5 mum from the P-cells. Nerves between acinar lobules may serve as neurohaemal organs and contain abundant dopaminergic and few serotonergic release sites. Some dopaminergic and serotonergic release sites reside in the duct epithelium, the former throughout the duct system, the latter only in segments next to acini. These findings are consistent with the view that C-cells respond exclusively to serotonin, P-cells to serotonin and dopamine, and most duct cells only to dopamine. Moreover, the data suggest that C-cells are stimulated by serotonin released close to their surface, whereas P-cells and most duct cells are exposed to serotonin/dopamine liberated at some distance
Die Entwicklung von Dickdarmkrebs wird durch eine Reihe von Lebens- und Essgewohnheiten sowie Umweltfaktoren begünstigt. Den letzteren beiden sind Substanzen zuzurechnen, die bei der Zubereitung der Nahrung entstehen und mit ihr aufgenommen werden. Zu diesen Verbindungen gehört das 2-Amino-1-methyl-6-phenylimidazo[4,5-b]pyridin (PhIP) aus der Substanzklasse der heterozyklischen aromatischen Amine. Es entsteht bei der Erhitzung zahlreicher proteinhaltiger Nahrungsmittel und die Zielorgane in Nagerstudien stimmen mit der Häufung von Krebsinzidenzen in westlichen Industrienationen überein. Dieser Zusammenhang konnte jedoch bis heute nicht endgültig bewiesen werden. Fütterungsversuche mit Ratten wurden mit Konzentrationen der Substanz durchgeführt, die weit über der menschlichen Exposition liegen. Durch das Verfüttern einer humanrelevanten Dosis PhIP sollte geklärt werden, ob auch geringe Konzentrationen dickdarmkrebstypische Mutationen, präneoplastische Läsionen oder Tumore induzierten. Die mit humanrelevanten Dosen gefütterten Tiere wiesen weniger Läsionen als die Hoch-Dosis-PhIP-Gruppe auf, in der allerdings keinerlei maligne Tumoren des Dickdarms auftraten. Hinweise auf dickdarmkrebstypische Mutationen fanden sich ebenfalls in beiden Gruppen, wobei hier keine Dosisabhängigkeit beobachtet werden konnte. Die Sequenzierung ergab ein deutlich von Literaturdaten abweichendes Spektrum. In Bezug auf das verwendete Tiermodell wurden erhebliche Abweichungen in der Empfindlichkeit der Tiere gegenüber der Substanz im Vergleich zu ähnlichen Studien festgestellt. Beide Fütterungsgruppen zeigten deutlich weniger Läsionen; als mögliche Gründe wurden Unterschiede in der Futterzusammensetzung und –zubereitung sowie in der Tierhaltung und –herkunft ausgemacht. Es konnte erstmalig ein Zusammenhang zwischen PhIP in niedrigen Dosen in der Nahrung und der Induktion von Entzündungen gezeigt werden. Diese waren sowohl makroskopisch als auch histologisch sichtbar, der genaue Mechanismus ihrer Entstehung ist jedoch unbekannt. Die zusammenfassende Betrachtung aller Ergebnisse lässt vermuten, dass PhIP allein über lange Zeiträume aber in geringen Dosen verabreicht nicht für die hohe Zahl an Krebserkrankungen in westlichen Industrienationen ursächlich ist.
Heterocyclic aromatic amines (HCAs) are formed in meat cooked at high temperatures for a long time or over an open flame. In this context 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP), the most abundant HCA in cooked meat, has been suggested to be involved in colon and prostate carcinogenesis. In the latter case it has been reported that: (1) roughly 50% of Fischer F344 male rats treated with PhIP develop carcinomas in the ventral prostate lobe at 1 year of age; (2) inflammation precedes prostatic intraepithelial neoplasia in PhIP-fed rats; (3) inflammation specifically occurs in the ventral prostate lobe of PhIP-fed rats. To test whether PhIP by itself leads to inflammation in the colon and whether a human-relevant concentration of PhIP is able to induce preneoplastic lesions in the colon, male F344 rats were fed 0.1 or 100 ppm PhIP for up to 10 months and thereafter the colon tissue was analyzed histochemically. In none of the experimental groups signs of acute or chronic colonic inflammation were observed. 0.1 ppm PhIP leads to the development of hyperplastic and dysplastic lesions in the colon of single animals, but the incidence of these lesions does not reach a statistical significance. In contrast, in rats fed 100 ppm PhIP for 10 months hyperplastic and dysplastic colonic lesions were induced in a statistically significant number of animals. It is concluded that: (1) the induction of preneoplastic lesions in rat colon by PhIP is not preceded or accompanied by an inflammatory process; (2) a human-relevant concentration of PhIP alone is not sufficient to initiate colon carcinogenesis in rats.
