Investigating culture from a linguistic perspective : an exemplification with Hong Kong English

Ribulose 1,5-bisphosphate carboxylase/oxygenase (RuBisCO) is the key enzyme of the Calvin cycle, catalyzing the fixation of inorganic carbon dioxide to organic sugars. Unlike most enzymes, RuBisCO is extremely slow, substrate unspecific, and catalyzes undesired side-reactions, which are considered to be responsible for the slow deactivation observed in vitro, a phenomenon known as fallover. Despite the fact that amino acid sequences and the 3D structures of RuBisCO from a variety of species are known, the precise molecular mechanisms for the various side reactions are still unclear. In the present study, we investigate the kinetic properties of RuBisCO using mathematical models. Initially, we formulate a minimal model that quantitatively reflects the kinetic behavior of RuBisCOs from different organisms. By relating rate parameters for single molecular steps to experimentally determined K-m and V-max values, we can examine mechanistic differences among species. The minimal model further demonstrates that two inhibitor producing side reactions are sufficient to describe experimentally determined fallover kinetics. To explain the observed kinetics of the limited capacity of RuBisCO to accept xylulose 1,5-bisphosphate as substrate, the inclusion of other side reactions is necessary. Our model results suggest a yet undescribed alternative enolization mechanism that is supported by the molecular structure. Taken together, the presented models serve as a theoretical framework to explain a wide range of observed kinetic properties of RuBisCOs derived from a variety of species. Thus, we can support hypotheses about molecular mechanisms and can systematically compare enzymes from different origins.
STG decomposition is a promising approach to tackle the complexity problems arising in logic synthesis of speed independent circuits, a robust asynchronous (i.e. clockless) circuit type. Unfortunately, STG decomposition can result in components that in isolation have irreducible CSC conflicts. Generalising earlier work, it is shown how to resolve such conflicts by introducing internal communication between the components via structural techniques only.
Smart bioactive surfaces

The purpose of this highlight is to define the emerging field of bioactive surfaces. In recent years, various types of synthetic materials capable of "communicating'' with biological objects such as nucleic acids, proteins, polysaccharides, viruses, bacteria or living cells have been described in the literature. This novel area of research certainly goes beyond the traditional field of smart materials and includes different types of sophisticated interactions with biological entities, such as reversible adhesion, conformational control, biologically-triggered release and selective permeation. These novel materials may be 2D planar surfaces as well as colloidal objects or 3D scaffolds. Overall, they show great promise for numerous applications in biosciences and biotechnology. For instance, practical applications of bioactive surfaces in the fields of bioseparation, cell engineering, biochips and stem-cell differentiation are briefly discussed herein.
This work generates, through a sample of numerical simulations of the restricted three-body problem, diagrams of semimajor axis and eccentricity which defines stable and unstable zones for particles in S-type orbits around Pluto and Charon. Since we consider initial conditions with 0 <= e <= 0.99, we found several new stable regions. We also identified the nature of each one of these newly found stable regions. They are all associated to families of periodic orbits derived from the planar circular restricted three-body problem. We have shown that a possible eccentricity of the Pluto-Charon system slightly reduces, but does not destroy, any of the stable regions.
In terms of the constructivist-orientated sociology of knowledge the social change of Christian religion turns out to be a reconstruction of central forms of knowledge. Mission and Ecumenism are two cornerstones of development This will be demonstrated using, Protestantism as an example. While the recourse on the notion of mission is enabling Christian religion to objectify itself as an integrative world religion against its societal environment, the Ecumenical leitmotif focuses on the denominational fragmented inner relations. Thereby Mission and Ecumenism are not only acting as semantics of,self-description. As distinct knowledge systems they are also standing for a programmatic change of the theological reservoir of knowledge.
A confocal set-up is presented that improves micro-XRF and XAFS experiment with high-pressure e diamond-anvil cells (DACs) In this experiment a probing volume is defined by the focus of the incoming synchrotron radiation beam and that of a polycapillary X-ray half-lens with a very long working distance, which is placed in front of the fluorescence detector This set-up enhances the quality of the fluorescence and XAFS spectra, and thus the sensitivity for detecting elements at low concentrations. It efficiently suppresses signal from outside the sample chamber, which stems from elastic and inelastic scattering of the incoming beam by the diamond anvils as well as from excitation of fluorescence from the body of the DAC
Amphibole and mica Ar-40/Ar-39 ages as well as zircon, rutile and titanite U-Pb geochronology of eclogites and associated host rocks from the Higher Himalayan Crystalline Nappes (Indian Plate) in the Upper Kaghan Valley, Pakistan allow distinction of a multistage exhumation history. An Eocene age for peak-pressure metamorphism has been obtained by phengite Ar-40/Ar-39 (47.3 +/- 0.3 Ma) and zircon U-Pb (47.3 +/- 0.4 and 47.4 +/- 0.3 Ma) ages from cover and basement gneisses. A very short-lived metamorphic peak and rapid cooling is documented by an amphibole Ar-40/Ar-39 age of 46.6 +/- 0.5 Ma and a rutile U-Pb age of 44.1 +/- 1.3 Ma from eclogites. Phengite and biotite ages from cover and basement sequences metamorphosed during the Himalayan orogeny are 34.5 +/- 0.2 to 28.1 +/- 0.2 Ma whereas youngest biotites, yielding 23.6 +/- 0.1 and 21.7 +/- 0.2 Ma, probably reflect argon partial resetting. The amphibole age, together with those derived from phengite and zircon demonstrate a rate of initial exhumation of 86-143 mm/a i.e. an extremely rapid transport of the Indian Plate continental crust from ultra-high pressure (UHP) conditions back to crustal levels (47-46 Ma for transport from 140 to 40 km depth). Subsequent exhumation (46-41 Ma, 40-35 km) slowed to about 1 mm/a at the base of the continental crust but increased again later towards slightly higher exhumation rates of ca. 2 mm/a (41-34 Ma, 35- 20 km). This indicates a change from buoyancy-driven exhumation at mantle depths to compression forces related to continent-continent collision and accompanied crustal folding, thrusting and stacking that finally exposed the former deeply-buried rocks.
Metabasites were sampled from rock series of the subducted margin of the Indian Plate, the so-called Higher Himalayan Crystalline, in the Upper Kaghan Valley, Pakistan. These vary from corona dolerites, cropping out around Saif- ul-Muluk in the south, to coesite-eclogite close to the suture zone against rocks of the Kohistan arc in the north. Bulk rock major- and trace-element chemistry reveals essentially a single protolith as the source for five different eclogite types, which differ in fabric, modal mineralogy as well as in mineral chemistry. The study of newly-collected samples reveals coesite (confirmed by in situ Raman spectroscopy) in both garnet and omphacite. All eclogites show growth of amphiboles during exhumation. Within some coesite-bearing eclogites the presence of glaucophane cores to barroisite is noted whereas in most samples porphyroblastic sodic-calcic amphiboles are rimmed by more aluminous calcic amphibole (pargasite, tschermakite, and edenite). Eclogite facies rutile is replaced by ilmenite which itself is commonly surrounded by titanite. In addition, some eclogite bodies show leucocratic segregations containing phengite, quartz, zoisite and/or kyanite. The important implication is that the complex exhumation path shows stages of initial cooling during decompression (formation of glaucophane) followed by reheating: a very similar situation to that reported for the coesite-bearing eclogite series of the Tso Morari massif, India, 450 km to the south-east.
In 1915, Alfred Wegener published his hypotheses of plate tectonics that revolutionised the world for geologists. Since then, many scientists have studied the evolution of continents and especially the geologic structure of orogens: the most visible consequence of tectonic processes. Although the morphology and landscape evolution of mountain belts can be observed due to surface processes, the driving force and dynamics at lithosphere scale are less well understood despite the fact that rocks from deeper levels of orogenic belts are in places exposed at the surface. In this thesis, such formerly deeply-buried (ultra-) high-pressure rocks, in particular eclogite facies series, have been studied in order to reveal details about the formation and exhumation conditions and rates and thus provide insights into the geodynamics of the most spectacular orogenic belt in the world: the Himalaya. The specific area investigated was the Kaghan Valley in Pakistan (NW Himalaya). Following closure of the Tethyan Ocean by ca. 55-50 Ma, the northward subduction of the leading edge of India beneath the Eurasian Plate and subsequent collision initiated a long-lived process of intracrustal thrusting that continues today. The continental crust of India – granitic basement, Paleozoic and Mesozoic cover series and Permo-Triassic dykes, sills and lavas – has been buried partly to mantle depths. Today, these rocks crop out as eclogites, amphibolites and gneisses within the Higher Himalayan Crystalline between low-grade metamorphosed rocks (600-640°C/ ca. 5 kbar) of the Lesser Himalaya and Tethyan sediments. Beside tectonically driven exhumation mechanisms the channel flow model, that describes a denudation focused ductile extrusion of low viscosity material developed in the middle to lower crust beneath the Tibetan Plateau, has been postulated. To get insights into the lithospheric and crustal processes that have initiated and driven the exhumation of this (ultra-) high-pressure rocks, mineralogical, petrological and isotope-geochemical investigations have been performed. They provide insights into 1) the depths and temperatures to which these rocks were buried, 2) the pressures and temperatures the rocks have experienced during their exhumation, 3) the timing of these processes 4) and the velocity with which these rocks have been brought back to the surface. In detail, through microscopical studies, the identification of key minerals, microprobe analyses, standard geothermobarometry and modelling using an effective bulk rock composition it has been shown that published exhumation paths are incomplete. In particular, the eclogites of the northern Kaghan Valley were buried to depths of 140-100 km (36-30 kbar) at 790-640°C. Subsequently, cooling during decompression (exhumation) towards 40-35 km (17-10 kbar) and 630-580°C has been superseded by a phase of reheating to about 720-650°C at roughly the same depth before final exhumation has taken place. In the southern-most part of the study area, amphibolite facies assemblages with formation conditions similar to the deduced reheating phase indicate a juxtaposition of both areas after the eclogite facies stage and thus a stacking of Indian Plate units. Radiometric dating of zircon, titanite and rutile by U-Pb and amphibole and micas by Ar-Ar reveal peak pressure conditions at 47-48 Ma. With a maximum exhumation rate of 14 cm/a these rocks reached the crust-mantle boundary at 40-35 km within 1 Ma. Subsequent exhumation (46-41 Ma, 40-35 km) decelerated to ca. 1 mm/a at the base of the continental crust but rose again to about 2 mm/a in the period of 41-31 Ma, equivalent to 35-20 km. Apatite fission track (AFT) and (U-Th)/He ages from eclogites, amphibolites, micaschists and gneisses yielded moderate Oligocene to Miocene cooling rates of about 10°C/Ma in the high altitude northern parts of the Kaghan Valley using the mineral-pair method. AFT ages are of 24.5±3.8 to 15.6±2.1 Ma whereas apatite (U-Th)/He analyses yielded ages between 21.0±0.6 and 5.3±0.2 Ma. The southern-most part of the Valley is dominated by younger late Miocene to Pliocene apatite fission track ages of 7.6±2.1 and 4.0±0.5 Ma that support earlier tectonically and petrologically findings of a juxtaposition and stack of Indian Plate units. As this nappe is tectonically lowermost, a later distinct exhumation and uplift driven by thrusting along the Main Boundary Thrust is inferred. A multi-stage exhumation path is evident from petrological, isotope-geochemical and low temperature thermochronology investigations. Buoyancy driven exhumation caused an initial rapid exhumation: exhumation as fast as recent normal plate movements (ca. 10 cm/a). As the exhuming units reached the crust-mantle boundary the process slowed down due to changes in buoyancy. Most likely, this exhumation pause has initiated the reheating event that is petrologically evident (e.g. glaucophane rimmed by hornblende, ilmenite overgrowth of rutile). Late stage processes involved widespread thrusting and folding with accompanied regional greenschist facies metamorphism, whereby contemporaneous thrusting on the Batal Thrust (seen by some authors equivalent to the MCT) and back sliding of the Kohistan Arc along the inverse reactivated Main Mantle Thrust caused final exposure of these rocks. Similar circumstances have been seen at Tso Morari, Ladakh, India, 200 km further east where comparable rock assemblages occur. In conclusion, as exhumation was already done well before the initiation of the monsoonal system, climate dependent effects (erosion) appear negligible in comparison to far-field tectonic effects.
Atrial natriuretic peptide (ANP) stimulates lipid mobilization and lipid oxidation in humans. The mechanism appears to promote lipid mobilization during exercise. We tested the hypothesis that water immersion augments exercise- induced ANP release and that the change in ANP availability is associated with increased lipid mobilization and lipid oxidation. In an open randomized and cross-over fashion we studied 17 men (age 31 +/- 3.6 years; body mass index 24 +/- 1.7 kg/m(2); body fat 17 +/- 6.7%) on no medication. Subjects underwent two incremental exercise tests on a bicycle ergometer. One test was conducted on land and the other test during immersion in water up to the xiphoid process. In a subset (n = 7), we obtained electromyography recordings in the left leg. We monitored gas exchange, blood pressure, and heart rate. In addition, we obtained blood samples towards the end of each exercise step to determine ANP, norepinephrine, epinephrine, lactate, free fatty acids, insulin, and glucose concentrations. Heart rate, systolic blood pressure, and oxygen consumption at the anaerobic threshold and during peak exercise were similar on land and with exercise in water. The respiratory quotient was mildly reduced when subjects exercised in water. Glucose and lactate measurements were decreased whereas free fatty acid concentrations were increased with exercise in water. Water immersion attenuated epinephrine and norepinephrine and augmented ANP release during exercise. Even though water immersion blunts exercise-induced sympathoadrenal activation, lipid mobilization and lipid oxidation rate are maintained or even improved. The response may be explained by augmented ANP release.
The population status of the harbour porpoise (Phocoena phocoena) in the Baltic area has been a continuous matter of debate. Here we present the by far most comprehensive genetic population structure assessment to date for this region, both with regard to geographic coverage and sample size: 497 porpoise samples from North Sea, Skagerrak, Kattegat, Belt Sea, and Inner Baltic Sea were sequenced at the mitochondrial Control Region and 305 of these specimens were typed at 15 polymorphic microsatellite loci. Samples were stratified according to sample type (stranding vs. by- caught), sex, and season (breeding vs. non-breeding season). Our data provide ample evidence for a population split between the Skagerrak and the Belt Sea, with a transition zone in the Kattegat area. Among other measures, this was particularly visible in significant frequency shifts of the most abundant mitochondrial haplotypes. A particular haplotype almost absent in the North Sea was the most abundant in Belt Sea and Inner Baltic Sea. Microsatellites yielded a similar pattern (i.e., turnover in occurrence of clusters identified by STRUCTURE). Moreover, a highly significant association between microsatellite assignment and unlinked mitochondrial haplotypes further indicates a split between North Sea and Baltic porpoises. For the Inner Baltic Sea, we consistently recovered a small, but significant separation from the Belt Sea population. Despite recent arguments that separation should exceed a predefined threshold before populations shall be managed separately, we argue in favour of precautionary acknowledging the Inner Baltic porpoises as a separate management unit, which should receive particular attention, as it is threatened by various factors, in particular local fishery measures.

Bisweilen wird Walser unterstellt, er suche über die Ausschließung des Jüdischen das Nationale zu stärken und behaupte die Unversöhnlichkeit von Tätergemeinschaft und Opfergemeinschaft nach 1945. Das bislang in der Forschung wenig beachtete Hörspiel "Das Gespenst von Gattnau" lässt die Annahme einer anderen Tendenz zu: die Tendenz zur Versöhnung.
Cenozoic uplift of the East African Plateau has been associated with fundamental climatic and environmental changes in East Africa and adjacent regions. While this influence is widely accepted, the timing and the magnitude of plateau uplift have remained unclear. This uncertainty stems from the lack of datable, geomorphically meaningful reference horizons that could record surface uplift. Here, we document the existence of significant relief along the East African Plateau prior to rifting, as inferred from modeling the emplacement history of one of the longest terrestrial lava flows, the similar to 300-km-long Yatta phonolite flow in Kenya. This 13.5 Ma lava flow originated on the present-day eastern Kenya Rift flank, and utilized a riverbed that once routed runoff from the eastern rim of the plateau. Combining an empirical viscosity model with subsequent cooling and using the Yatta lava flow geometry and underlying paleotopography (slope angle), we found that the prerift slope was at least 0.2 degrees, suggesting that the lava flow originated at a minimum elevation of 1400 m. Hence, high paleotopography in the Kenya Rift region must have existed by at least 13.5 Ma. We infer from this that middle Miocene uplift occurred, which coincides with the two-step expansion of grasslands, as well as important radiation and speciation events in tropical Africa.
Live cell flattening

Eukaryotic cell flattening is valuable for improving microscopic observations, ranging from bright field (BF) to total internal reflection fluorescence (TIRF) microscopy. Fundamental processes, such as mitosis and in vivo actin polymerization, have been investigated using these techniques. Here, we review the well known agar overlayer protocol and the oil overlay method. In addition, we present more elaborate microfluidics-based techniques that provide us with a greater level of control. We demonstrate these techniques on the social amoebae Dictyostelium discoideum, comparing the advantages and disadvantages of each method.
An efficient route from myo- to neo-inositol is described. The key steps of the sequence are oxidation of the hydroxy group at C-5 to the corresponding ketone, followed by a highly (dr = 7.8:1) stereoselective reduction. The route includes nine steps with an overall yield of 51% and is therefore superior to all hitherto reported methods for the preparation of neo-inositol.
Calibration of the global hydrological model WGHM with water mass variations from GRACE gravity data

Since the start-up of the GRACE (Gravity Recovery And Climate Experiment) mission in 2002 time dependent global maps of the Earth's gravity field are available to study geophysical and climatologically-driven mass redistributions on the Earth's surface. In particular, GRACE observations of total water storage changes (TWSV) provide a comprehensive data set for analysing the water cycle on large scales. Therefore they are invaluable for validation and calibration of large-scale hydrological models as the WaterGAP Global Hydrology Model (WGHM) which simulates the continental water cycle including its most important components, such as soil, snow, canopy, surface- and groundwater. Hitherto, WGHM exhibits significant differences to GRACE, especially for the seasonal amplitude of TWSV. The need for a validation of hydrological models is further highlighted by large differences between several global models, e.g. WGHM, the Global Land Data Assimilation System (GLDAS) and the Land Dynamics model (LaD). For this purpose, GRACE links geodetic and hydrological research aspects. This link demands the development of adequate data integration methods on both sides, forming the main objectives of this work. They include the derivation of accurate GRACE-based water storage changes, the development of strategies to integrate GRACE data into a global hydrological model as well as a calibration method, followed by the re-calibration of WGHM in order to analyse process and model responses. To achieve these aims, GRACE filter tools for the derivation of regionally averaged TWSV were evaluated for specific river basins. Here, a decorrelation filter using GRACE orbits for its design is most efficient among the tested methods. Consistency in data and equal spatial resolution between observed and simulated TWSV were realised by the inclusion of all most important hydrological processes and an equal filtering of both data sets. Appropriate calibration parameters were derived by a WGHM sensitivity analysis against TWSV. Finally, a multi-objective calibration framework was developed to constrain model predictions by both river discharge and GRACE TWSV, realised with a respective evolutionary method, the ε-Non-dominated-Sorting-Genetic-Algorithm-II (ε-NSGAII). Model calibration was done for the 28 largest river basins worldwide and for most of them improved simulation results were achieved with regard to both objectives. From the multi-objective approach more reliable and consistent simulations of TWSV within the continental water cycle were gained and possible model structure errors or mis-modelled processes for specific river basins detected. For tropical regions as such, the seasonal amplitude of water mass variations has increased. The findings lead to an improved understanding of hydrological processes and their representation in the global model. Finally, the robustness of the results is analysed with respect to GRACE and runoff measurement errors. As a main conclusion obtained from the results, not only soil water and snow storage but also groundwater and surface water storage have to be included in the comparison of the modelled and GRACE-derived total water budged data. Regarding model calibration, the regional varying distribution of parameter sensitivity suggests to tune only parameter of important processes within each region. Furthermore, observations of single storage components beside runoff are necessary to improve signal amplitudes and timing of simulated TWSV as well as to evaluate them with higher accuracy. The results of this work highlight the valuable nature of GRACE data when merged into large-scale hydrological modelling and depict methods to improve large-scale hydrological models.

Die These des Aufsatzes lautet, dass es sich bei Sebastian Brants „Narrenschiff“ um einen Text handelt, der entsprechend der Prinzipien verstanden werden kann, die Rudolf Agricola für das Sammeln von Argumenten formuliert hat. Das Gründungsdokument dieser humanistischen Methode, die unter dem Begriff der „loci communes“ Geschich¬te gemacht hat, ist ein Brief Agricolas aus dem Jahr 1484. Ein erster Teil des Aufsatzes stellt Agricolas Argumen¬tationstheorie dar, ein zweiter Teil zeigt, dass Brants „Narrenschiff“ als Ergebnis dieser Argumentationstheorie verstanden werden kann.

Das Gründungsdokument dieser humanistischen Methode, die unter dem Begriff der „loci communes“ Geschichte gemacht hat, ist ein Brief Agricolas aus dem Jahr 1484. Ein erster Teil des Aufsatzes stellt Agricolas Argumentationstheorie dar, ein zweiter Teil zeigt, dass Brants „Narrenschiff“ als Ergebnis dieser Argumentationstheorie verstanden werden kann.

Quantifying uncertainty, variability and likelihood for ordinary differential equation models

Background
In many applications, ordinary differential equation (ODE) models are subject to uncertainty or variability in initial conditions and parameters. Both, uncertainty and variability can be quantified in terms of a probability density function on the state and parameter space.
Results
The partial differential equation that describes the evolution of this probability density function has a form that is particularly amenable to application of the well-known method of characteristics. The value of the density at some point in time is directly accessible by the solution of the original ODE extended by a single extra dimension (for the value of the density). This leads to simple methods for studying uncertainty, variability and likelihood, with significant advantages over more traditional Monte Carlo and related approaches especially when studying regions with low probability.
Conclusions
While such approaches based on the method of characteristics are common practice in other disciplines, their advantages for the study of biological systems have so far remained unrecognized. Several examples illustrate performance and accuracy of the approach and its limitations.
