We present novel experimental evidence on the availability and the status of exhaustivity inferences with focus partitioning in German, English, and Hungarian. Results suggest that German and English focus-background clefts and Hungarian focus share important properties, (É. Kiss 1998, 1999; Szabolcsi 1994; Percus 1997; Onea & Beaver 2009). Those constructions are anaphoric devices triggering an existence presupposition. EXH-inferences are not obligatory in such constructions in English, German, or Hungarian, against some previous literature (Percus 1997; Büring & Križ 2013; É. Kiss 1998), but in line with pragmatic analyses of EXH-inferences in clefts (Horn 1981, 2016; Pollard & Yasavul 2016). The cross-linguistic differences in the distribution of EXH-inferences are attributed to properties of the Hungarian number marking system.
Monolithic perovskite silicon tandem solar cells can overcome the theoretical efficiency limit of silicon solar cells. This requires an optimum bandgap, high quantum efficiency, and high stability of the perovskite. Herein, a silicon heterojunction bottom cell is combined with a perovskite top cell, with an optimum bandgap of 1.68 eV in planar p-i-n tandem configuration. A methylammonium-free FA(0.75)Cs(0.25)Pb(I0.8Br0.2)(3) perovskite with high Cs content is investigated for improved stability. A 10% molarity increase to 1.1 m of the perovskite precursor solution results in approximate to 75 nm thicker absorber layers and 0.7 mA cm(-2) higher short-circuit current density. With the optimized absorber, tandem devices reach a high fill factor of 80% and up to 25.1% certified efficiency. The unencapsulated tandem device shows an efficiency improvement of 2.3% (absolute) over 5 months, showing the robustness of the absorber against degradation. Moreover, a photoluminescence quantum yield analysis reveals that with adapted charge transport materials and surface passivation, along with improved antireflection measures, the high bandgap perovskite absorber has the potential for 30% tandem efficiency in the near future.
Nach einer Einführung in die Geschichte der Strategiespiele und im Speziellen von 4X-Spielen wird das Phänomen der „Hands-off-Games“ erläutert. Im Anschluss wird ein Vorschlag unterbreitet, wie 4X-Geschichtsspiele im Unterricht eingesetzt werden können. Dabei soll ein 4X-Strategiespiel zu einem historischen Thema entworfen werden. Die Modellierung erfolgt in drei Arbeitsschritten: Themenfindung, Modellfindung, Parametrierung. In den Entwurf des Modells fließen viele Überlegungen ein, die zentrale Fragen der Gemeinschaftskunde betreffen.
Abiotic stresses cause oxidative damage in plants. Here, we demonstrate that foliar application of an extract from the seaweed Ascophyllum nodosum, SuperFifty (SF), largely prevents paraquat (PQ)-induced oxidative stress in Arabidopsis thaliana. While PQ-stressed plants develop necrotic lesions, plants pre-treated with SF (i.e., primed plants) were unaffected by PQ. Transcriptome analysis revealed induction of reactive oxygen species (ROS) marker genes, genes involved in ROS-induced programmed cell death, and autophagy-related genes after PQ treatment. These changes did not occur in PQ-stressed plants primed with SF. In contrast, upregulation of several carbohydrate metabolism genes, growth, and hormone signaling as well as antioxidant-related genes were specific to SF-primed plants. Metabolomic analyses revealed accumulation of the stress-protective metabolite maltose and the tricarboxylic acid cycle intermediates fumarate and malate in SF-primed plants. Lipidome analysis indicated that those lipids associated with oxidative stress-induced cell death and chloroplast degradation, such as triacylglycerols (TAGs), declined upon SF priming. Our study demonstrated that SF confers tolerance to PQ-induced oxidative stress in A. thaliana, an effect achieved by modulating a range of processes at the transcriptomic, metabolic, and lipid levels.
Abdominal and general adiposity are independently associated with mortality, but there is no consensus on how best to assess abdominal adiposity. We compared the ability of alternative waist indices to complement body mass index (BMI) when assessing all-cause mortality. We used data from 352,985 participants in the European Prospective Investigation into Cancer and Nutrition (EPIC) and Cox proportional hazards models adjusted for other risk factors. During a mean follow-up of 16.1 years, 38,178 participants died. Combining in one model BMI and a strongly correlated waist index altered the association patterns with mortality, to a predominantly negative association for BMI and a stronger positive association for the waist index, while combining BMI with the uncorrelated A Body Shape Index (ABSI) preserved the association patterns. Sex-specific cohort-wide quartiles of waist indices correlated with BMI could not separate high-risk from low-risk individuals within underweight (BMI<18.5 kg/m(2)) or obese (BMI30 kg/m(2)) categories, while the highest quartile of ABSI separated 18-39% of the individuals within each BMI category, which had 22-55% higher risk of death. In conclusion, only a waist index independent of BMI by design, such as ABSI, complements BMI and enables efficient risk stratification, which could facilitate personalisation of screening, treatment and monitoring.
This paper addresses semantic/pragmatic variability of tag questions in German and makes three main contributions. First, we document the prevalence and variety of question tags in German across three different types of conversational corpora. Second, by annotating question tags according to their syntactic and semantic context, discourse function, and pragmatic effect, we demonstrate the existing overlap and differences between the individual tag variants. Finally, we distinguish several groups of question tags by identifying the factors that influence the speakers’ choices of tags in the conversational context, such as clause type, function, speaker/hearer knowledge, as well as conversation type and medium. These factors provide the limits of variability by constraining certain question tags in German against occurring in specific contexts or with individual functions.
Although the use of stable transformation technology has led to great insight into gene function, its application in high-throughput studies remains arduous. Agro-infiltration have been widely used in species such as Nicotiana benthamiana for the rapid detection of gene expression and protein interaction analysis, but this technique does not work efficiently in other plant species, including Arabidopsis thaliana. As an efficient high-throughput transient expression system is currently lacking in the model plant species A. thaliana, we developed a method that is characterized by high efficiency, reproducibility, and suitability for transient expression of a variety of functional proteins in A. thaliana and 7 other plant species, including Brassica oleracea, Capsella rubella, Thellungiella salsuginea, Thellungiella halophila, Solanum tuberosum, Capsicum annuum, and N. benthamiana. Efficiency of this method was independently verified in three independent research facilities, pointing to the robustness of this technique. Furthermore, in addition to demonstrating the utility of this technique in a range of species, we also present a case study employing this method to assess protein-protein interactions in the sucrose biosynthesis pathway in Arabidopsis.
Investigation of processes that contribute to the maintenance of genomic stability is one crucial factor in the attempt to understand mechanisms that facilitate ageing. The DNA damage response (DDR) and DNA repair mechanisms are crucial to safeguard the integrity of DNA and to prevent accumulation of persistent DNA damage. Among them, base excision repair (BER) plays a decisive role. BER is the major repair pathway for small oxidative base modifications and apurinic/apyrimidinic (AP) sites. We established a highly sensitive non-radioactive assay to measure BER incision activity in murine liver samples. Incision activity can be assessed towards the three DNA lesions 8-oxo-2’-deoxyguanosine (8-oxodG), 5-hydroxy-2’-deoxyuracil (5-OHdU), and an AP site analogue. We applied the established assay to murine livers of adult and old mice of both sexes. Furthermore, poly(ADP-ribosyl)ation (PARylation) was assessed, which is an important determinant in DDR and BER. Additionally, DNA damage levels were measured to examine the overall damage levels. No impact of ageing on the investigated endpoints in liver tissue were found. However, animal sex seems to be a significant impact factor, as evident by sex-dependent alterations in all endpoints investigated. Moreover, our results revealed interrelationships between the investigated endpoints indicative for the synergetic mode of action of the cellular DNA integrity maintaining machinery.
Anthropogenic changes in climate, land use, and disturbance regimes, as well as introductions of non-native species can lead to the transformation of many ecosystems. The resulting novel ecosystems are usually characterized by species assemblages that have not occurred previously in a given area. Quantifying the ecological novelty of communities (i.e., biotic novelty) would enhance the understanding of environmental change. However, quantification remains challenging since current novelty metrics, such as the number and/or proportion of non-native species in a community, fall short of considering both functional and evolutionary aspects of biotic novelty. Here, we propose the Biotic Novelty Index (BNI), an intuitive and flexible multidimensional measure that combines (a) functional differences between native and non-native introduced species with (b) temporal dynamics of species introductions. We show that the BNI is an additive partition of Rao's quadratic entropy, capturing the novel interaction component of the community's functional diversity. Simulations show that the index varies predictably with the relative amount of functional novelty added by recently arrived species, and they illustrate the need to provide an additional standardized version of the index. We present a detailed R code and two applications of the BNI by (a) measuring changes of biotic novelty of dry grassland plant communities along an urbanization gradient in a metropolitan region and (b) determining the biotic novelty of plant species assemblages at a national scale. The results illustrate the applicability of the index across scales and its flexibility in the use of data of different quality. Both case studies revealed strong connections between biotic novelty and increasing urbanization, a measure of abiotic novelty. We conclude that the BNI framework may help building a basis for better understanding the ecological and evolutionary consequences of global change.
Metagenomic sequencing has revolutionised our knowledge of virus diversity, with new virus sequences being reported faster than ever before. However, virus discovery from metagenomic sequencing usually depends on detectable homology: without a sufficiently close relative, so-called ‘dark’ virus sequences remain unrecognisable. An alternative approach is to use virus-identification methods that do not depend on detecting homology, such as virus recognition by host antiviral immunity. For example, virus-derived small RNAs have previously been used to propose ‘dark’ virus sequences associated with the Drosophilidae (Diptera). Here, we combine published Drosophila data with a comprehensive search of transcriptomic sequences and selected meta-transcriptomic datasets to identify a completely new lineage of segmented positive-sense single-stranded RNA viruses that we provisionally refer to as the Quenyaviruses. Each of the five segments contains a single open reading frame, with most encoding proteins showing no detectable similarity to characterised viruses, and one sharing a small number of residues with the RNA-dependent RNA polymerases of single- and double-stranded RNA viruses. Using these sequences, we identify close relatives in approximately 20 arthropods, including insects, crustaceans, spiders, and a myriapod. Using a more conserved sequence from the putative polymerase, we further identify relatives in meta-transcriptomic datasets from gut, gill, and lung tissues of vertebrates, reflecting infections of vertebrates or of their associated parasites. Our data illustrate the utility of small RNAs to detect viruses with limited sequence conservation, and provide robust evidence for a new deeply divergent and phylogenetically distinct RNA virus lineage.
The use of monoclonal antibodies is ubiquitous in science and biomedicine but the generation and validation process of antibodies is nevertheless complicated and time-consuming. To address these issues we developed a novel selective technology based on an artificial cell surface construct by which secreted antibodies were connected to the corresponding hybridoma cell when they possess the desired antigen-specificity. Further the system enables the selection of desired isotypes and the screening for potential cross-reactivities in the same context. For the design of the construct we combined the transmembrane domain of the EGF-receptor with a hemagglutinin epitope and a biotin acceptor peptide and performed a transposon-mediated transfection of myeloma cell lines. The stably transfected myeloma cell line was used for the generation of hybridoma cells and an antigen- and isotype-specific screening method was established. The system has been validated for globular protein antigens as well as for haptens and enables a fast and early stage selection and validation of monoclonal antibodies in one step.
Pollen records from Siberia are mostly absent in global or Northern Hemisphere synthesis works. Here we present a taxonomically harmonized and temporally standardized pollen dataset that was synthesized using 173 palynological records from Siberia and adjacent areas (northeastern Asia, 42-75 degrees N, 50-180 degrees E). Pollen data were taxonomically harmonized, i.e. the original 437 taxa were assigned to 106 combined pollen taxa. Age-depth models for all records were revised by applying a constant Bayesian age-depth modelling routine. The pollen dataset is available as count data and percentage data in a table format (taxa vs. samples), with age information for each sample. The dataset has relatively few sites covering the last glacial period between 40 and 11.5 ka (calibrated thousands of years before 1950 CE) particularly from the central and western part of the study area. In the Holocene period, the dataset has many sites from most of the area, with the exception of the central part of Siberia. Of the 173 pollen records, 81 % of pollen counts were downloaded from open databases (GPD, EPD, PANGAEA) and 10 % were contributions by the original data gatherers, while a few were digitized from publications. Most of the pollen records originate from peatlands (48 %) and lake sediments (33 %). Most of the records (83 %) have >= 3 dates, allowing the establishment of reliable chronologies. The dataset can be used for various purposes, including pollen data mapping (example maps for Larix at selected time slices are shown) as well as quantitative climate and vegetation reconstructions. The datasets for pollen counts and pollen percentages are available at https://doi.org/10.1594/PANGAEA.898616 (Cao et al., 2019a), also including the site information, data source, original publication, dating data, and the plant functional type for each pollen taxa.
A treemap is a visualization that has been specifically designed to facilitate the exploration of tree-structured data and, more general, hierarchically structured data. The family of visualization techniques that use a visual metaphor for parent-child relationships based “on the property of containment” (Johnson, 1993) is commonly referred to as treemaps. However, as the number of variations of treemaps grows, it becomes increasingly important to distinguish clearly between techniques and their specific characteristics. This paper proposes to discern between Space-filling Treemap TS, Containment Treemap TC, Implicit Edge Representation Tree TIE, and Mapped Tree TMT for classification of hierarchy visualization techniques and highlights their respective properties. This taxonomy is created as a hyponymy, i.e., its classes have an is-a relationship to one another: TS TC TIE TMT. With this proposal, we intend to stimulate a discussion on a more unambiguous classification of treemaps and, furthermore, broaden what is understood by the concept of treemap itself.
In this thesis, I examine different A-bar movement dependencies in Igbo, a Benue-Congo language spoken in southern Nigeria. Movement dependencies are found in constructions where an element is moved to the left edge of the clause to express information-structural categories such as in questions, relativization and focus. I show that these constructions in Igbo are very uniform from a syntactic point of view. The constructions are built on two basic fronting operations: relativization and focus movement, and are biclausal. I further investigate several morphophonological effects that are found in these A-bar constructions. I propose that these effects are reflexes of movement that are triggered when an element is moved overtly in relativization or focus. This proposal helps to explain the tone patterns that have previously been assumed to be a property of relative clauses. The thesis adds to the growing body of tonal reflexes of A-bar movement reported for a few African languages. The thesis also provides an insight into the complementizer domain (C-domain) of Igbo.
ABCB1/4 gallbladder cancer risk variants identified in India also show strong effects in Chileans

Background: The first large-scale genome-wide association study of gallbladder cancer (GBC) recently identified and validated three susceptibility variants in the ABCB1 and ABCB4 genes for individuals of Indian descent. We investigated whether these variants were also associated with GBC risk in Chileans, who show the highest incidence of GBC worldwide, and in Europeans with a low GBC incidence.
Methods: This population-based study analysed genotype data from retrospective Chilean case-control (255 cases, 2042 controls) and prospective European cohort (108 cases, 181 controls) samples consistently with the original publication.
Results: Our results confirmed the reported associations for Chileans with similar risk effects. Particularly strong associations (per-allele odds ratios close to 2) were observed for Chileans with high Native American (=Mapuche) ancestry. No associations were noticed for Europeans, but the statistical power was low.
Conclusion: Taking full advantage of genetic and ethnic differences in GBC risk may improve the efficiency of current prevention programs.
Macrophages have important protective functions during infection with herpes simplex virus type 1 (HSV-1). However, molecular mechanisms that restrict viral propagation and protect from severe disease are unclear. Here we show that macrophages take up HSV-1 via endocytosis and transport the virions into multivesicular bodies (MVBs). In MVBs, acid ceramidase (aCDase) converts ceramide into sphingosine and increases the formation of sphingosine-rich intraluminal vesicles (ILVs). Once HSV-1 particles reach MVBs, sphingosine-rich ILVs bind to HSV-1 particles, which restricts fusion with the limiting endosomal membrane and prevents cellular infection. Lack of aCDase in macrophage cultures or in Asah1(-/-) mice results in replication of HSV-1 and Asah1(-/-) mice die soon after systemic or intravaginal inoculation. The treatment of macrophages with sphingosine enhancing compounds blocks HSV-1 propagation, suggesting a therapeutic potential of this pathway. In conclusion, aCDase loads ILVs with sphingosine, which prevents HSV-1 capsids from penetrating into the cytosol.
Acquiring Syntactic Variability: The Production of Wh-Questions in Children and Adults Speaking Akan

This paper investigates the predictions of the Derivational Complexity Hypothesis by studying the acquisition of wh-questions in 4- and 5-year-old Akan-speaking children in an experimental approach using an elicited production and an elicited imitation task. Akan has two types of wh-question structures (wh-in-situ and wh-ex-situ questions), which allows an investigation of children’s acquisition of these two question structures and their preferences for one or the other. Our results show that adults prefer to use wh-ex-situ questions over wh-in-situ questions. The results from the children show that both age groups have the two question structures in their linguistic repertoire. However, they differ in their preferences in usage in the elicited production task: while the 5-year-olds preferred the wh-in-situ structure over the wh-ex-situ structure, the 4-year-olds showed a selective preference for the wh-in-situ structure in who-questions. These findings suggest a developmental change in wh-question preferences in Akan-learning children between 4 and 5 years of age with a so far unobserved u-shaped developmental pattern. In the elicited imitation task, all groups showed a strong tendency to maintain the structure of in-situ and ex-situ questions in repeating grammatical questions. When repairing ungrammatical ex-situ questions, structural changes to grammatical in-situ questions were hardly observed but the insertion of missing morphemes while keeping the ex-situ structure. Together, our findings provide only partial support for the Derivational Complexity Hypothesis.
One of the major risk factors for global death and disability is alcohol, tobacco, and illicit drug use. While there is increasing knowledge with respect to individual factors promoting the initiation and maintenance of substance use disorders (SUDs), disease trajectories involved in losing and regaining control over drug intake (ReCoDe) are still not well described. Our newly formed German Collaborative Research Centre (CRC) on ReCoDe has an interdisciplinary approach funded by the German Research Foundation (DFG) with a 12-year perspective. The main goals of our research consortium are (i) to identify triggers and modifying factors that longitudinally modulate the trajectories of losing and regaining control over drug consumption in real life, (ii) to study underlying behavioral, cognitive, and neurobiological mechanisms, and (iii) to implicate mechanism-based interventions. These goals will be achieved by: (i) using mobile health (m-health) tools to longitudinally monitor the effects of triggers (drug cues, stressors, and priming doses) and modify factors (eg, age, gender, physical activity, and cognitive control) on drug consumption patterns in real-life conditions and in animal models of addiction; (ii) the identification and computational modeling of key mechanisms mediating the effects of such triggers and modifying factors on goal-directed, habitual, and compulsive aspects of behavior from human studies and animal models; and (iii) developing and testing interventions that specifically target the underlying mechanisms for regaining control over drug intake.
Radical reactions have found many applications in carbohydrate chemistry, especially in the construction of carbon–carbon bonds. The formation of carbon–heteroatom bonds has been less intensively studied. This mini-review will summarize the efforts to add heteroatom radicals to unsaturated carbohydrates like endo-glycals. Starting from early examples, developed more than 50 years ago, the importance of such reactions for carbohydrate chemistry and recent applications will be discussed. After a short introduction, the mini-review is divided in sub-chapters according to the heteroatoms halogen, nitrogen, phosphorus, and sulfur. The mechanisms of radical generation by chemical or photochemical processes and the subsequent reactions of the radicals at the 1-position will be discussed. This mini-review cannot cover all aspects of heteroatom-centered radicals in carbohydrate chemistry, but should provide an overview of the various strategies and future perspectives
The Cluster mission has produced a large data set of electron flux measurements in the Earth's magnetosphere since its launch in late 2000. Electron fluxes are measured using Research with Adaptive Particle Imaging Detector (RAPID)/Imaging Electron Spectrometer (IES) detector as a function of energy, pitch angle, spacecraft position, and time. However, no adiabatic invariants have been calculated for Cluster so far. In this paper we present a step-by-step guide to calculations of adiabatic invariants and conversion of the electron flux to phase space density (PSD) in these coordinates. The electron flux is measured in two RAPID/IES energy channels providing pitch angle distribution at energies 39.2-50.5 and 68.1-94.5 keV in nominal mode since 2004. A fitting method allows to expand the conversion of the differential fluxes to the range from 40 to 150 keV. Best data coverage for phase space density in adiabatic invariant coordinates can be obtained for values of second adiabatic invariant, K, similar to 10(2), and values of the first adiabatic invariant mu in the range approximate to 5-20 MeV/G. Furthermore, we describe the production of a new data product "LSTAR," equivalent to the third adiabatic invariant, available through the Cluster Science Archive for years 2001-2018 with 1-min resolution. The produced data set adds to the availability of observations in Earth's radiation belts region and can be used for long-term statistical purposes.
The purpose of the present study was to investigate the role of gender and gender stereotype traits (masculinity, femininity) in cyber victimization behaviors (cyber relational victimization, cyber verbal victimization, hacking) through different technologies (mobile phones, gaming consoles, social networking sites). There were 456 8th graders (226 females; M age = 13.66, SD = 0.41) from two midwestern middle schools in the United States included in this study. They completed questionnaires on their endorsement of masculine and feminine traits, and self-reported cyber victimization through different technologies. The findings revealed main effects of types of cyber victimization for boys and of technology for girls. In particular, boys with feminine traits experienced the most victimization by cyber verbal aggression, cyber relational aggression, and hacking when compared to the other groups of boys. Girls with feminine traits experienced the most cyber victimization through social networking sites, gaming consoles, and mobile phones in comparison to the other groups of girls. For girls with feminine traits, they reported more cyber relational victimization and cyber verbal victimization through mobile phones and social networking sites, as well as more hacking via social networking sites. Such findings underscore the importance of considering gender stereotype traits, types of victimization, and technologies when examining cyber victimization.
Recent research indicates that affective responses during exercise are an important determinant of future exercise and physical activity. Thus far these responses have been measured with standardized self-report scales, but this study used biometric software for automated facial action analysis to analyze the changes that occur during physical exercise. A sample of 132 young, healthy individuals performed an incremental test on a cycle ergometer. During that test the participants’ faces were video-recorded and the changes were algorithmically analyzed at frame rate (30 fps). Perceived exertion and affective valence were measured every two minutes with established psychometric scales. Taking into account anticipated inter-individual variability, multilevel regression analysis was used to model how affective valence and ratings of perceived exertion (RPE) covaried with movement in 20 facial action areas. We found the expected quadratic decline in self-reported affective valence (more negative) as exercise intensity increased. Repeated measures correlation showed that the facial action mouth open was linked to changes in (highly intercorrelated) affective valence and RPE. Multilevel trend analyses were calculated to investigate whether facial actions were typically linked to either affective valence or RPE. These analyses showed that mouth open and jaw drop predicted RPE, whereas (additional) nose wrinkle was indicative for the decline in affective valence. Our results contribute to the view that negative affect, escalating with increasing exercise intensity, may be the body’s essential warning signal that physiological overload is imminent. We conclude that automated facial action analysis provides new options for researchers investigating feelings during exercise. In addition, our findings offer physical educators and coaches a new way of monitoring the affective state of exercisers, without interrupting and asking them.
Alexander von Humboldt betätigte sich in unterschiedlichsten Wissensbereichen und vielen Disziplinen. Obwohl er natürlich kein Jurist war, hat er sich vielfach mit Problemen befasst, die zu seinen Lebzeiten und danach zu grundlegenden Fragen des in der Entstehung begriffenen Völkerrechts avancierten. So beschäftigte er sich mit dem Verbot der Sklaverei und der Abschaffung der Rassendiskriminierung, mit der Fixierung von territorialen Grenzen von Staaten, mit Seegrenzen, der Förderung des Welthandels und der Hoheitsgewalt im Luft- und im Weltraum. Viele dieser Fragen wurden später in internationalen Verträgen kodifiziert, weshalb man Humboldt als Vordenker des Völkerrechts bezeichnen kann. Diese von der internationalen Humboldt-Forschung bislang nicht als Forschungsfeld erkannte Dimension seines Wirkens wird nachfolgend erstmals vertiefend beleuchtet.
En su breve visita a Canarias a finales del siglo XVIII, Alexander von Humboldt realizó interesantes observaciones sobre la geología de estas Islas. En relación con los fósiles inorgánicos, es decir, rocas y minerales, sus apreciaciones se enmarcan en sus estudios realizados en la Bergakademie de Freiberg, bajo la dirección de A. G. Werner, dentro del Neptunismo, y en sus experiencias en América e Italia. Aplicó en Canarias la universalidad de las formaciones geológicas wernerianas, algunas de las cuales creyó, erróneamente, haber observado en Tenerife, como la presencia de granitos o gneises. Humboldt estaba convencido de que el basalto
era de origen volcánico, y no se había formado por precipitación acuosa. Sería así uno de los más influyentes desertores del Neptunismo, y suscitó la visita a Canarias de numerosos geólogos a lo largo del siglo XIX.
Im Jahre 1789 hatte Alexander von Humboldt als Student in Göttingen mit großem Enthusiasmus ein Manuskript zur „Weberei der Alten“ verfasst. Es war das Ziel seiner Studie, die Technologie und die Terminologie der antiken Weberei zu erfassen und beide Gebiete vereint darzustellen. Dieses Manuskript ist nicht veröffentlicht worden und gilt als verschollen. Alexander von Humboldt hatte offenbar neue Erkenntnisse gewonnen, denen er so große Bedeutung beimaß, dass er den Verlust seines Manuskriptes noch über 50 Jahre nach dessen Abfassung bedauerte. Humboldt benennt glücklicherweise in überlieferten Briefen verschiedene Grundgedanken seiner damaligen Arbeit. In der vorliegenden Studie wird unter Einbeziehung aktueller technikhistorischer Erkenntnisse nachgewiesen, dass der junge Alexander von Humboldt das für alle Webtechniken gültige Prinzip in seinen Grundzügen erkannt hatte.
Though Humboldt’s travels to the Americas have been analyzed from a wide range of viewpoints, there are specific aspects that still await further investigation. Little is written about Humboldt in the field, specifically how he moved between different locations and simultaneously measured and mapped places and phenomena. The aim of this article is to discuss the triad movement-measure-ment-map that led to the development of specific practices of knowledge building on the move. Humboldt’s search for the connections between the watersheds of the Orinoco and the Amazon rivers and the resulting maps and drawings are used as an example to point out his cartographic impulse in his quest to understand and explain the physical world.

The laboratory mouse is the most common used mammalian research model in biomedical research. Usually these animals are maintained in germ-free, gnotobiotic, or specific-pathogen-free facilities. In these facilities, skilled staff takes care of the animals and scientists usually don’t pay much attention about the formulation and quality of diets the animals receive during normal breeding and keeping. However, mice have specific nutritional requirements that must be met to guarantee their potential to grow, reproduce and to respond to pathogens or diverse environmental stress situations evoked by handling and experimental interventions. Nowadays, mouse diets for research purposes are commercially manufactured in an industrial process, in which the safety of food products is addressed through the analysis and control of all biological and chemical materials used for the different diet formulations. Similar to human food, mouse diets must be prepared under good sanitary conditions and truthfully labeled to provide information of all ingredients. This is mandatory to guarantee reproducibility of animal studies. In this review, we summarize some information on mice research diets and general aspects of mouse nutrition including nutrient requirements of mice, leading manufacturers of diets, origin of nutrient compounds, and processing of feedstuffs for mice including dietary coloring, autoclaving and irradiation. Furthermore, we provide some critical views on the potential pitfalls that might result from faulty comparisons of grain-based diets with purified diets in the research data production resulting from confounding nutritional factors.
Alles auf (Studien-)Anfang? Faktoren für den Studienerfolg in der Eingangsphase und zur Studienmitte

Die hohen Abbruchquoten, insbesondere in der Studieneingangsphase, haben die Hochschulen in Deutschland veranlasst, eine Vielzahl von Maßnahmen zu ergreifen, über deren Wirkungen bisher allerdings wenig bekannt ist. Im vorliegenden Beitrag werden Befunde eines Forschungsprojekts speziell zur Studieneingangsphase sowie ergänzend zur Studienmitte vorgestellt, dessen Ziel es war, Bedingungen eines erfolgreichen Studieneinstiegs zu identifizieren und Empfehlungen für eine Optimierung des Studieneingangs abzuleiten. Das Forschungsdesign umfasste neben qualitativen Studien vor allem eine quantitative Längsschnittbefragung an fünf Universitäten (Potsdam, Mainz, Magdeburg, Kiel und Greifswald). Im Ergebnis der Analysen konnte die forschungsleitende Hypothese, dass Maßnahmen zum Studieneingang vor allem dann zur Erhöhung des Studienerfolgs einen Beitrag leisten, wenn sie zur akademischen und sozialen Integration in die Hochschule beitragen, bestätigt werden. Bedeutsam für den Studienerfolg sind demnach insbesondere solche Faktoren wie die Identifikation mit dem Studienfach, die Selbstwirksamkeit, die berufliche bzw. erfolgsorientierte Lernmotivation und die akademische Integration. Daneben konnte ein positiver Einfluss des sozialen Klimas sowie des Forschungs- und Praxisbezugs auf die Studienzufriedenheit nachgewiesen werden. Weiterführende Analysen zur Studienmitte verdeutlichen zudem, dass für die beiden Studienphasen (Eingang und Studienmitte) gleiche Faktoren bei zum Teil unterschiedlicher Gewichtung eine Rolle spielen. So ist die soziale Integration ein wesentlicher Prädiktor in beiden Phasen – in der Eingangsphase eher in die Studierendenschaft und im weiteren Studienverlauf (Studienmitte) eher in die akademische Gemeinschaft (in Form von Lehrenden). Insofern muss die Eingangsfrage wie folgt beantwortet werden: Ja, alles auf Anfang, aber dann mit den Bemühungen, soziale und akademische Integration aller Studierenden voll und ganz zu gewährleisten. Zudem machen die Befunde auf die bisher offenbar unterschätzte Rolle von Verwertungsmotiven aufmerksam.
Vitamin A, vitamin E and retinol-binding protein 4 (RBP4) are a focus of current obesity research in humans. The impact of body weight (BW) gain on fat-soluble vitamins and its associated parameters in equines has not been previously reported. Ten Shetland ponies and 9 Warmblood horses, all adult geldings, non-obese and healthy, were fed an excessive energy diet for 20 months to induce BW gain. Serum alpha-tocopherol (vitamin E), retinol (vitamin A), retinol-binding protein 4 (RBP4) and retinol/RBP4 ratio were analysed before BW gain induction and at six timepoints during the BW gaining period. The mean (+/- SD) % BW gain achieved during two years of excess energy intake was 29.9 +/- 19.4% for ponies and 17 +/- 6.74% for horses. Serum alpha-tocopherol increased significantly in ponies and horses during excess energy intake and circulating alpha-tocopherol levels correlated positively with alpha-tocopherol intake (r = .6; p < .001). Serum retinol concentrations showed variations during the study but without relation to intake. Serum RBP4 decreased at the end of the study. The retinol/RBP4 ratio increased with BW gain without differences between ponies and horses. In comparison with human research, the increase in the retinol/RBP4 ratio was unexpected and needs further elucidation.
Droughts in tropical South America have an imminent and severe impact on the Amazon rainforest and affect the livelihoods of millions of people. Extremely dry conditions in Amazonia have been previously linked to sea surface temperature (SST) anomalies in the adjacent tropical oceans. Although the sources and impacts of such droughts have been widely studied, establishing reliable multi-year lead statistical forecasts of their occurrence is still an ongoing challenge. Here, we further investigate the relationship between SST and rainfall anomalies using a complex network approach. We identify four ocean regions which exhibit the strongest overall SST correlations with central Amazon rainfall, including two particularly prominent regions in the northern and southern tropical Atlantic. Based on the time-dependent correlation between SST anomalies in these two regions alone, we establish a new early-warning method for droughts in the central Amazon basin and demonstrate its robustness in hindcasting past major drought events with lead-times up to 18 months.
Engineering biotechnological microorganisms to use methanol as a feedstock for bioproduction is a major goal for the synthetic metabolism community. Here, we aim to redesign the natural serine cycle for implementation in E. coli. We propose the homoserine cycle, relying on two promiscuous formaldehyde aldolase reactions, as a superior pathway design. The homoserine cycle is expected to outperform the serine cycle and its variants with respect to biomass yield, thermodynamic favorability, and integration with host endogenous metabolism. Even as compared to the RuMP cycle, the most efficient naturally occurring methanol assimilation route, the homoserine cycle is expected to support higher yields of a wide array of products. We test the in vivo feasibility of the homoserine cycle by constructing several E. coli gene deletion strains whose growth is coupled to the activity of different pathway segments. Using this approach, we demonstrate that all required promiscuous enzymes are active enough to enable growth of the auxotrophic strains. Our findings thus identify a novel metabolic solution that opens the way to an optimized methylotrophic platform.
The article introduces the efforts exerted to initiate multiplication of the DIES training courses to a wider audience of higher education managers and academics in the target regions. The DIES ‘National Multiplication Trainings’ Programme has supported three cohorts of alumni teams so far to implement national training courses in the area of higher education leadership and management. The article sets the context of this publication and reflects on the main evaluation results of the programme implementation itself.
Salt pans are highly dynamic environments that are difficult to study by in situ methods because of their harsh climatic conditions and large spatial areas. Remote sensing can help to elucidate their environmental dynamics and provide important constraints regarding their sedimentological, mineralogical, and hydrological evolution. This study utilizes spaceborne multitemporal multispectral optical data combined with spectral endmembers to document spatial distribution of surface crust types over time on the Omongwa pan located in the Namibian Kalahari. For this purpose, 49 surface samples were collected for spectral and mineralogical characterization during three field campaigns (2014–2016) reflecting different seasons and surface conditions of the salt pan. An approach was developed to allow the spatiotemporal analysis of the salt pan crust dynamics in a dense time-series consisting of 77 Landsat 8 cloud-free scenes between 2014 and 2017, covering at least three major wet–dry cycles. The established spectral analysis technique Sequential Maximum Angle Convex Cone (SMACC) extraction method was used to derive image endmembers from the Landsat time-series stack. Evaluation of the extracted endmember set revealed that the multispectral data allowed the differentiation of four endmembers associated with mineralogical mixtures of the crust’s composition in dry conditions and three endmembers associated with flooded or muddy pan conditions. The dry crust endmember spectra have been identified in relation to visible, near infrared, and short-wave infrared (VNIR–SWIR) spectroscopy and X-ray diffraction (XRD) analyses of the collected surface samples. According these results, the spectral endmembers are interpreted as efflorescent halite crust, mixed halite–gypsum crust, mixed calcite quartz sepiolite crust, and gypsum crust. For each Landsat scene the spatial distribution of these crust types was mapped with the Spectral Angle Mapper (SAM) method and significant spatiotemporal dynamics of the major surface crust types were observed. Further, the surface crust dynamics were analyzed in comparison with the pan’s moisture regime and other climatic parameters. The results show that the crust dynamics are mainly driven by flooding events in the wet season, but are also influenced by temperature and aeolian activity in the dry season. The approach utilized in this study combines the advantages of multitemporal satellite data for temporal event characterization with advantages from hyperspectral methods for the image and ground data analyses that allow improved mineralogical differentiation and characterization.
Concurrent observation technologies have made high-precision real-time data available in large quantities. Data assimilation (DA) is concerned with how to combine this data with physical models to produce accurate predictions. For spatial-temporal models, the ensemble Kalman filter with proper localisation techniques is considered to be a state-of-the-art DA methodology. This article proposes and investigates a localised ensemble Kalman Bucy filter for nonlinear models with short-range interactions. We derive dimension-independent and component-wise error bounds and show the long time path-wise error only has logarithmic dependence on the time range. The theoretical results are verified through some simple numerical tests.
Botulinum neurotoxins (BoNTs) are potent neurotoxins produced by bacteria, which inhibit neurotransmitter release, specifically in their physiological target known as motor neurons (MNs). For the potency assessment of BoNTs produced for treatment in traditional and aesthetic medicine, the mouse lethality assay is still used by the majority of manufacturers, which is ethically questionable in terms of the 3Rs principle. In this study, MNs were differentiated from human induced pluripotent stem cells based on three published protocols. The resulting cell populations were analyzed for their MN yield and their suitability for the potency assessment of BoNTs. MNs produce specific gangliosides and synaptic proteins, which are bound by BoNTs in order to be taken up by receptor-mediated endocytosis, which is followed by cleavage of specific soluble N-ethylmaleimide-sensitive-factor attachment receptor (SNARE) proteins required for neurotransmitter release. The presence of receptors and substrates for all BoNT serotypes was demonstrated in MNs generated in vitro. In particular, the MN differentiation protocol based on Du et al. yielded high numbers of MNs in a short amount of time with high expression of BoNT receptors and targets. The resulting cells are more sensitive to BoNT/A1 than the commonly used neuroblastoma cell line SiMa. MNs are, therefore, an ideal tool for being combined with already established detection methods.
The most severe flood events in Turkey were determined for the period 1960–2014 by considering the number of fatalities, the number of affected people, and the total economic losses as indicators. The potential triggering mechanisms (i.e., atmospheric circulations and precipitation amounts) and aggravating pathways (i.e., topographic features, catchment size, land use types, and soil properties) of these 25 events were analyzed. On this basis, a new approach was developed to identify the main influencing factor per event and to provide additional information for determining the dominant flood occurrence pathways for severe floods. The events were then classified through hierarchical cluster analysis. As a result, six different clusters were found and characterized. Cluster 1 comprised flood events that were mainly influenced by drainage characteristics (e.g., catchment size and shape); Cluster 2 comprised events aggravated predominantly by urbanization; steep topography was identified to be the dominant factor for Cluster 3; extreme rainfall was determined as the main triggering factor for Cluster 4; saturated soil conditions were found to be the dominant factor for Cluster 5; and orographic effects of mountain ranges characterized Cluster 6. This study determined pathway patterns of the severe floods in Turkey with regard to their main causal or aggravating mechanisms. Accordingly, geomorphological properties are of major importance in large catchments in eastern and northeastern Anatolia. In addition, in small catchments, the share of urbanized area seems to be an important factor for the extent of flood impacts. This paper presents an outcome that could be used for future urban planning and flood risk prevention studies to understand the flood mechanisms in different regions of Turkey.
We study populations of globally coupled noisy rotators (oscillators with inertia) allowing a nonequilibrium transition from a desynchronized state to a synchronous one (with the nonvanishing order parameter). The newly developed analytical approaches resulted in solutions describing the synchronous state with constant order parameter for weakly inertial rotators, including the case of zero inertia, when the model is reduced to the Kuramoto model of coupled noise oscillators. These approaches provide also analytical criteria distinguishing supercritical and subcritical transitions to the desynchronized state and indicate the universality of such transitions in rotator ensembles. All the obtained analytical results are confirmed by the numerical ones, both by direct simulations of the large ensembles and by solution of the associated Fokker-Planck equation. We also propose generalizations of the developed approaches for setups where different rotators parameters (natural frequencies, masses, noise intensities, strengths and phase shifts in coupling) are dispersed.
Utilising a reconstructed ancestral mitochondrial genome of a clade to design hybridisation capture baits can provide the opportunity for recovering mitochondrial sequences from all its descendent and even sister lineages. This approach is useful for taxa with no extant close relatives, as is often the case for rare or extinct species, and is a viable approach for the analysis of historical museum specimens. Asiatic linsangs (genus Prionodon) exemplify this situation, being rare Southeast Asian carnivores for which little molecular data is available. Using ancestral capture we recover partial mitochondrial genome sequences for seven banded linsangs (P. linsang) from historical specimens, representing the first intraspecific genetic dataset for this species. We additionally assemble a high quality mitogenome for the banded linsang using shotgun sequencing for time-calibrated phylogenetic analysis. This reveals a deep divergence between the two Asiatic linsang species (P. linsang, P. pardicolor), with an estimated divergence of ~12 million years (Ma). Although our sample size precludes any robust interpretation of the population structure of the banded linsang, we recover two distinct matrilines with an estimated tMRCA of ~1 Ma. Our results can be used as a basis for further investigation of the Asiatic linsangs, and further demonstrate the utility of ancestral capture for studying divergent taxa without close relatives.
Dieser Bericht nimmt die Institutionalisierung von Forschungsdatenmanagement (FDM) an den staatlichen Hochschulen Brandenburgs in den Blick. Er soll zur Beantwortung der folgenden Fragen beitragen: Warum wird FDM dort institutionalisiert? Wie wird FDM dort institutionalisiert? Welche Herausforderungen für eine weitere Institutionalisierung bestehen und wie kann ihnen begegnet werden? Zur Erhebung des Status Quo und zur Identifikation von Anforderungen und Weiterentwicklungspotenzialen wurde deshalb eine Organisationsuntersuchung durchgeführt. Untersuchung ist neben der Einhaltung wissenschaftlicher und professioneller Standards auch dem Anwendungsbezug der Ergebnisse, deren Anwendbarkeit im Kontext und der Partizipation der Beteiligten verpflichtet. Da es sich um ein für die betrachteten Einrichtungen noch vergleichsweise neues Themengebiet handelt, wurde ein exploratives Forschungsdesign unter Einsatz qualitativer Methoden gewählt. Grundlage der Ergebnisse sind ein Auftaktworkshop unter Beteiligung der Hochschulen, eine Dokumentenanalyse und 16 leitfadengestützte Expert*innen-Interviews auf Arbeits- und Leitungsebene der brandenburgischen Hochschulen. Die Ergebnisse werden anhand von vier Handlungsbereichen strukturiert vorgestellt: Relevanz von FDM und Institutionalisierung an den Hochschulen; Technologie und Infrastruktur; Personal und Qualifizierung; Organisation, Kooperation und Finanzierung. Dabei stellt der Bericht sowohl den Status Quo als auch die Ziele der Hochschulen sowie die Wünsche an eine Landesinitiative vor.
Objective
Plant carnivory is distributed across the tree of life and has evolved at least six times independently, but sequenced and annotated nuclear genomes of carnivorous plants are currently lacking. We have sequenced and structurally annotated the nuclear genome of the carnivorous Roridula gorgonias and that of a non-carnivorous relative, Madeira’s lily-of-the-valley-tree, Clethra arborea, both within the Ericales. This data adds an important resource to study the evolutionary genetics of plant carnivory across angiosperm lineages and also for functional and systematic aspects of plants within the Ericales.
Results
Our assemblies have total lengths of 284 Mbp (R. gorgonias) and 511 Mbp (C. arborea) and show high BUSCO scores of 84.2% and 89.5%, respectively. We used their predicted genes together with publicly available data from other Ericales’ genomes and transcriptomes to assemble a phylogenomic data set for the inference of a species tree. However, groups of orthologs showed a marked absence of species represented by a transcriptome. We discuss possible reasons and caution against combining predicted genes from genome- and transriptome-based assemblies.
Cardiac valves are essential for the continuous and unidirectional flow of blood throughout the body. During embryonic development, their formation is strictly connected to the mechanical forces exerted by blood flow. The endocardium that lines the interior of the heart is a specialized endothelial tissue and is highly sensitive to fluid shear stress. Endocardial cells harbor a signal transduction machinery required for the translation of these forces into biochemical signaling, which strongly impacts cardiac morphogenesis and physiology. To date, we lack a solid understanding on the mechanisms by which endocardial cells sense the dynamic mechanical stimuli and how they trigger different cellular responses. In the zebrafish embryo, endocardial cells at the atrioventricular canal respond to blood flow by rearranging from a monolayer to a double-layer, composed of a luminal cell population subjected to blood flow and an abluminal one that is not exposed to it. These early morphological changes lead to the formation of an immature valve leaflet. While previous studies mainly focused on genes that are positively regulated by shear stress, the mechanisms regulating cell behaviors and fates in cells that lack the stimulus of blood flow are largely unknown. One key discovery of my work is that the flow-sensitive Notch receptor and Krüppel-like factor (Klf) 2, one of the best characterized flow-regulated transcriptional factors, are activated by shear stress but that they function in two parallel signal transduction pathways. Each of these two pathways is essential for the rearrangement of atrioventricular cells into an immature double-layered valve leaflets. A second key discovery of my study is the finding that both Notch and Klf2 signaling negatively regulate the expression of the angiogenesis receptor Vegfr3/Flt4, which becomes restricted to abluminal endocardial cells of the valve leaflet. Within these cells, Flt4 downregulates the expressions of the cell adhesion proteins Alcam and VE-cadherin. A loss of Flt4 causes abluminal endocardial cells to ectopically express Notch, which is normally restricted to luminal cells, and impairs valve morphology. My study suggests that abluminal endocardial cells that do not experience mechanical stimuli loose Notch expression and this triggers expression of Flt4. In turn, Flt4 negatively regulates Notch on the abluminal side of the valve leaflet. These antagonistic signaling activities and fine-tuned gene regulatory mechanisms ultimately shape cardiac valve leaflets by inducing unique differences in the fates of endocardial cells.
In 1924, the Berlin ophthalmologist Arthur Czellitzer (1871–1943) and like-minded members of the local Jewish community founded the Society for Jewish Family Research. A year later, the Society launched the journal Jüdische Familienforschung (Jewish Family Research), edited by Czellitzer. The Society was an outstanding platform of professional academic and amateur researchers and promoted a type of Jewish genealogy and family history that was shaped by the historical-medical discourse of the time. The concepts and methods of both the biological sciences and Wissenschaft des Judentums shaped and defined the academic approach to family research and history in Czellitzer’s and the Society’s work. The Society soon became the leading international association for the academic Jewish genealogical research. Despite of its brutal end in 1938, Arthur Czellitzer’s and the Society’s works, the issues raised, and the methods they created shape Jewish family research and genealogy until today.
The East Asian monsoons characterize the modern-day Asian climate, yet their geological history and driving mechanisms remain controversial. The southeasterly summer monsoon provides moisture, whereas the northwesterly winter monsoon sweeps up dust from the arid Asian interior to form the Chinese Loess Plateau. The onset of this loess accumulation, and therefore of the monsoons, was thought to be 8 million years ago (Ma). However, in recent years these loess records have been extended further back in time to the Eocene (56-34 Ma), a period characterized by significant changes in both the regional geography and global climate. Yet the extent to which these reconfigurations drive atmospheric circulation and whether the loess-like deposits are monsoonal remains debated. In this thesis, I study the terrestrial deposits of the Xining Basin previously identified as Eocene loess, to derive the paleoenvironmental evolution of the region and identify the geological processes that have shaped the Asian climate.
I review dust deposits in the geological record and conclude that these are commonly represented by a mix of both windblown and water-laid sediments, in contrast to the pure windblown material known as loess. Yet by using a combination of quartz surface morphologies, provenance characteristics and distinguishing grain-size distributions, windblown dust can be identified and quantified in a variety of settings. This has important implications for tracking aridification and dust-fluxes throughout the geological record.
Past reversals of Earth’s magnetic field are recorded in the deposits of the Xining Basin and I use these together with a dated volcanic ash layer to accurately constrain the age to the Eocene period. A combination of pollen assemblages, low dust abundances and other geochemical data indicates that the early Eocene was relatively humid suggesting an intensified summer monsoon due to the warmer greenhouse climate at this time. A subsequent shift from predominantly freshwater to salt lakes reflects a long-term aridification trend possibly driven by global cooling and the continuous uplift of the Tibetan Plateau. Superimposed on this aridification are wetter intervals reflected in more abundant lake deposits which correlate with highstands of the inland proto-Paratethys Sea. This sea covered the Eurasian continent and thereby provided additional moisture to the winter-time westerlies during the middle to late Eocene.
The long-term aridification culminated in an abrupt shift at 40 Ma reflected by the onset of windblown dust, an increase in steppe-desert pollen, the occurrence of high-latitude orbital cycles and northwesterly winds identified in deflated salt deposits. Together, these indicate the onset of a Siberian high atmospheric pressure system driving the East Asian winter monsoon as well as dust storms and was triggered by a major sea retreat from the Asian interior. These results therefore show that the proto-Paratethys Sea, though less well recognized than the Tibetan Plateau and global climate, has been a major driver in setting up the modern-day climate in Asia.
Largescale patterns of global land use change are very frequently accompanied by natural habitat loss. To assess the consequences of habitat loss for the remaining natural and semi-natural biotopes, inclusion of cumulative effects at the landscape level is required. The interdisciplinary concept of vulnerability constitutes an appropriate assessment framework at the landscape level, though with few examples of its application for ecological assessments. A comprehensive biotope vulnerability analysis allows identification of areas most affected by landscape change and at the same time with the lowest chances of regeneration.
To this end, a series of ecological indicators were reviewed and developed. They measured spatial attributes of individual biotopes as well as some ecological and conservation characteristics of the respective resident species community. The final vulnerability index combined seven largely independent indicators, which covered exposure, sensitivity and adaptive capacity of biotopes to landscape changes. Results for biotope vulnerability were provided at the regional level. This seems to be an appropriate extent with relevance for spatial planning and designing the distribution of nature reserves.
Using the vulnerability scores calculated for the German federal state of Brandenburg, hot spots and clusters within and across the distinguished types of biotopes were analysed. Biotope types with high dependence on water availability, as well as biotopes of the open landscape containing woody plants (e.g., orchard meadows) are particularly vulnerable to landscape changes. In contrast, the majority of forest biotopes appear to be less vulnerable. Despite the appeal of such generalised statements for some biotope types, the distribution of values suggests that conservation measures for the majority of biotopes should be designed specifically for individual sites. Taken together, size, shape and spatial context of individual biotopes often had a dominant influence on the vulnerability score.
The implementation of biotope vulnerability analysis at the regional level indicated that large biotope datasets can be evaluated with high level of detail using geoinformatics. Drawing on previous work in landscape spatial analysis, the reproducible approach relies on transparent calculations of quantitative and qualitative indicators. At the same time, it provides a synoptic overview and information on the individual biotopes. It is expected to be most useful for nature conservation in combination with an understanding of population, species, and community attributes known for specific sites. The biotope vulnerability analysis facilitates a foresighted assessment of different land uses, aiding in identifying options to slow habitat loss to sustainable levels. It can also be incorporated into planning of restoration measures, guiding efforts to remedy ecological damage. Restoration of any specific site could yield synergies with the conservation objectives of other sites, through enhancing the habitat network or buffering against future landscape change.
Biotope vulnerability analysis could be developed in line with other important ecological concepts, such as resilience and adaptability, further extending the broad thematic scope of the vulnerability concept. Vulnerability can increasingly serve as a common framework for the interdisciplinary research necessary to solve major societal challenges.
High Mountain Asia (HMA) is dependent upon both the amount and timing of snow and glacier meltwater. Previous model studies and coarse resolution (0.25° × 0.25°, ∼25 km × 25 km) passive microwave assessments of trends in the volume and timing of snowfall, snowmelt, and glacier melt in HMA have identified key spatial and seasonal heterogeneities in the response of snow to changes in regional climate. Here we use recently developed, continuous, internally consistent, and high-resolution passive microwave data (3.125 km × 3.125 km, 1987–2016) from the special sensor microwave imager instrument family to refine and extend previous estimates of changes in the snow regime of HMA. We find an overall decline in snow volume across HMA; however, there exist spatially contiguous regions of increasing snow volume—particularly during the winter season in the Pamir, Karakoram, Hindu Kush, and Kunlun Shan. Detailed analysis of changes in snow-volume trends through time reveal a large step change from negative trends during the period 1987–1997, to much more positive trends across large regions of HMA during the periods 1997–2007 and 2007–2016. We also find that changes in high percentile monthly snow-water volume exhibit steeper trends than changes in low percentile snow-water volume, which suggests a reduction in the frequency of high snow-water volumes in much of HMA. Regions with positive snow-water storage trends generally correspond to regions of positive glacier mass balances.
Groundwater is the biggest single source of high-quality freshwater worldwide, which is also continuously threatened by the changing climate. In this paper, we investigate the response of the regional groundwater system to climate change under three global warming levels (1.5, 2, and 3 ∘C) in a central German basin (Nägelstedt). This investigation is conducted by deploying an integrated modeling workflow that consists of a mesoscale hydrologic model (mHM) and a fully distributed groundwater model, OpenGeoSys (OGS). mHM is forced with climate simulations of five general circulation models under three representative concentration pathways. The diffuse recharges estimated by mHM are used as boundary forcings to the OGS groundwater model to compute changes in groundwater levels and travel time distributions. Simulation results indicate that groundwater recharges and levels are expected to increase slightly under future climate scenarios. Meanwhile, the mean travel time is expected to decrease compared to the historical average. However, the ensemble simulations do not all agree on the sign of relative change. Changes in mean travel time exhibit a larger variability than those in groundwater levels. The ensemble simulations do not show a systematic relationship between the projected change (in both groundwater levels and travel times) and the warming level, but they indicate an increased variability in projected changes with adjusting the enhanced warming level from 1.5 to 3 ∘C. Correspondingly, it is highly recommended to restrain the trend of global warming.
To better understand the role of individual and lifestyle factors in human disease, an exposome-wide association study was performed to investigate within a single-study anthropometry measures and lifestyle factors previously associated with B-cell lymphoma (BCL). Within the European Prospective Investigation into Cancer and nutrition study, 2402 incident BCL cases were diagnosed from 475 426 participants that were followed-up on average 14 years. Standard and penalized Cox regression models as well as principal component analysis (PCA) were used to evaluate 84 exposures in relation to BCL risk. Standard and penalized Cox regression models showed a positive association between anthropometric measures and BCL and multiple myeloma/plasma cell neoplasm (MM). The penalized Cox models additionally showed the association between several exposures from categories of physical activity, smoking status, medical history, socioeconomic position, diet and BCL and/or the subtypes. PCAs confirmed the individual associations but also showed additional observations. The PC5 including anthropometry, was positively associated with BCL, diffuse large B-cell lymphoma (DLBCL) and MM. There was a significant positive association between consumption of sugar and confectionary (PC11) and follicular lymphoma risk, and an inverse association between fish and shellfish and Vitamin D (PC15) and DLBCL risk. The PC1 including features of the Mediterranean diet and diet with lower inflammatory score showed an inverse association with BCL risk, while the PC7, including dairy, was positively associated with BCL and DLBCL risk. Physical activity (PC10) was positively associated with DLBCL risk among women. This study provided informative insights on the etiology of BCL.
This study aimed to investigate the relationship between the acute to chronic workload ratio (ACWR), based upon participant session rating of perceived exertion (sRPE), using two models [(1) rolling averages (ACWRRA); and (2) exponentially weighted moving averages (ACWREWMA)] and the injury rate in young male team soccer players aged 17.1 ± 0.7 years during a competitive mesocycle. Twenty-two players were enrolled in this study and performed four training sessions per week with 2 days of recovery and 1 match day per week. During each training session and each weekly match, training time and sRPE were recorded. In addition, training impulse (TRIMP), monotony, and strain were subsequently calculated. The rate of injury was recorded for each soccer player over a period of 4 weeks (i.e., 28 days) using a daily questionnaire. The results showed that over the course of the study, the number of non-contact injuries was significantly higher than that for contact injuries (2.5 vs. 0.5, p = 0.01). There were also significant positive correlations between sRPE and training time (r = 0.411, p = 0.039), ACWRRA (r = 0.47, p = 0.049), and ACWREWMA (r = 0.51, p = 0.038). In addition, small-to-medium correlations were detected between ACWR and non-contact injury occurrence (ACWRRA, r = 0.31, p = 0.05; ACWREWMA, r = 0.53, p = 0.03). Explained variance (r 2) for non-contact injury was significantly greater using the ACWREWMA model (ranging between 21 and 52%) compared with ACWRRA (ranging between 17 and 39%). In conclusion, the results of this study showed that the ACWREWMA model is more sensitive than ACWRRA to identify non-contact injury occurrence in male team soccer players during a short period in the competitive season.

We applied the Social Cognitive Theory to investigate whether parent–child relationships, bullying victimization, and teacher–student relationships are directly as well as indirectly via self-efficacy in social conflicts associated with adolescents’ willingness to intervene in a bullying incident. There were 2071 (51.3% male) adolescents between the ages of 12 and 17 from 24 schools in Germany who participated in this study. A mediation test using structural equation modeling revealed that parent–child relationships, bullying victimization, and teacher–student relationships were directly related to adolescents’ self-efficacy in social conflicts. Further, teacher–student relationships and bullying victimization were directly associated with adolescents’ willingness to intervene in bullying. Finally, relationships with parents, peers and teachers were indirectly related to higher levels of students’ willingness to intervene in bullying situations due to self-efficacy in social conflicts. Thus, our analysis confirms the general assumptions of Social Cognitive Theory and the usefulness of applying its approach to social conflicts such as bullying situations.
This study sought to analyze the relationship between in-season training workload with changes in aerobic power (VO2max), maximum and resting heart rate (HRmax and HRrest), linear sprint medium (LSM), and short test (LSS), in soccer players younger than 16 years (under-16 soccer players). We additionally aimed to explain changes in fitness levels during the in-season through regression models, considering accumulated load, baseline levels, and peak height velocity (PHV) as predictors. Twenty-three male sub-elite soccer players aged 15.5 ± 0.2 years (PHV: 13.6 ± 0.4 years; body height: 172.7 ± 4.2 cm; body mass: 61.3 ± 5.6 kg; body fat: 13.7% ± 3.9%; VO2max: 48.4 ± 2.6 mL⋅kg–1⋅min–1), were tested three times across the season (i.e., early-season (EaS), mid-season (MiS), and end-season (EnS) for VO2max, HRmax, LSM, and LSS. Aerobic and speed variables gradually improved over the season and had a strong association with PHV. Moreover, the HRmax demonstrated improvements from EaS to EnS; however, this was more evident in the intermediate period (from EaS to MiS) and had a strong association with VO2max. Regression analysis showed significant predictions for VO2max [F(2, 20) = 8.18, p ≤ 0.001] with an R2 of 0.45. In conclusion, the meaningful variation of youth players’ fitness levels can be observed across the season, and such changes can be partially explained by the load imposed.

Even though the majority of individuals know that exercising is healthy, a high percentage struggle to achieve the recommended amount of exercise. The (social-cognitive) theories that are commonly applied to explain exercise motivation refer to the assumption that people base their decisions mainly on rational reasoning. However, behavior is not only bound to reflection. In recent years, the role of automaticity and affect for exercise motivation has been increasingly discussed. In this dissertation, central assumptions of the affective–reflective theory of physical inactivity and exercise (ART; Brand & Ekkekakis, 2018), an exercise-specific dual-process theory that emphasizes the role of a momentary automatic affective reaction for exercise-decisions, were examined. The central aim of this dissertation was to investigate exercisers and non-exercisers automatic affective reactions to exercise-related stimuli (i.e., type-1 process). In particular, the two components of the ART’s type-1 process, that are, automatic associations with exercise and the automatic affective valuation to exercise, were under study.
In the first publication (Schinkoeth & Antoniewicz, 2017), research on automatic (evaluative) associations with exercise was summarized and evaluated in a systematic review. The results indicated that automatic associations with exercise appeared to be relevant predictors for exercise behavior and other exercise-related variables, providing evidence for a central assumption of the ART’s type-1 process. Furthermore, indirect methods seem to be suitable to assess automatic associations. The aim of the second publication (Schinkoeth, Weymar, & Brand, 2019) was to approach the somato-affective core of the automatic valuation of exercise using analysis of reactivity in vagal HRV while viewing exercise-related pictures. Results revealed that differences in exercise volume could be regressed on HRV reactivity. In light of the ART, these findings were interpreted as evidence of an inter-individual affective reaction elicited at the thought of exercise and triggered by exercise-stimuli. In the third publication (Schinkoeth & Brand, 2019, subm.), it was sought to disentangle and relate to each other the ART’s type-1 process components—automatic associations and the affective valuation of exercise. Automatic associations to exercise were assessed with a recoding-free variant of an implicit association test (IAT). Analysis of HRV reactivity was applied to approach a somatic component of the affective valuation, and facial reactions in a facial expression (FE) task served as indicators of the automatic affective reaction’s valence. Exercise behavior was assessed via self-report. The measurement of the affective valuation’s valence with the FE task did not work well in this study. HRV reactivity was predicted by the IAT score and did also statistically predict exercise behavior. These results thus confirm and expand upon the results of publication two and provide empirical evidence for the type-1 process, as defined in the ART. This dissertation advances the field of exercise psychology concerning the influence of automaticity and affect on exercise motivation. Moreover, both methodical implications and theoretical extensions for the ART can be derived from the results.
The decision to exercise is not only bound to rational considerations but also automatic affective processes. The affective–reflective theory of physical inactivity and exercise (ART) proposes a theoretical framework for explaining how the automatic affective process (type‑1 process) will influence exercise behavior, i.e., through the automatic activation of exercise-related associations and a subsequent affective valuation of exercise. This study aimed to empirically test this assumption of the ART with data from 69 study participants. A single-measurement study, including within-subject experimental variation, was conducted. Automatic associations with exercise were first measured with a single-target implicit association test. The somato-affective core of the participants’ automatic valuation of exercise-related pictures was then assessed via heart rate variability (HRV) analysis, and the affective valence of the valuation was tested with a facial expression (FE; smile and frown) task. Exercise behavior was assessed via self-report. Multiple regression (path) analysis revealed that automatic associations predicted HRV reactivity (β = −0.24, p = .044); the signs of the correlation between automatic associations and the smile FE score was in the expected direction but remained nonsignificant (β = −0.21, p = .078). HRV reactivity predicted self-reported exercise behavior (β = −0.28, p = .013) (the same pattern of results was achieved for the frown FE score). The HRV-related results illustrate the potential role of automatic negative affective reactions to the thought of exercise as a restraining force in exercise motivation. For better empirical distinction between the two ART type‑1 process components, automatic associations and the affective valuation should perhaps be measured separately in the future. The results support the notion that automatic and affective processes should be regarded as essential aspects of the motivation to exercise.
Bayesian Data Assimilation to Support Informed Decision Making in Individualized Chemotherapy

An essential component of therapeutic drug/biomarker monitoring (TDM) is to combine patient data with prior knowledge for model-based predictions of therapy outcomes. Current Bayesian forecasting tools typically rely only on the most probable model parameters (maximum a posteriori (MAP) estimate). This MAP-based approach, however, does neither necessarily predict the most probable outcome nor does it quantify the risks of treatment inefficacy or toxicity. Bayesian data assimilation (DA) methods overcome these limitations by providing a comprehensive uncertainty quantification. We compare DA methods with MAP-based approaches and show how probabilistic statements about key markers related to chemotherapy-induced neutropenia can be leveraged for more informative decision support in individualized chemotherapy. Sequential Bayesian DA proved to be most computationally efficient for handling interoccasion variability and integrating TDM data. For new digital monitoring devices enabling more frequent data collection, these features will be of critical importance to improve patient care decisions in various therapeutic areas.

The rapidly growing amount and diversity of data are confronting us more than ever with the need to make informed predictions under uncertainty. The adverse impacts of climate change and natural hazards also motivate our search for reliable predictions. The range of statistical techniques that geomorphologists use to tackle this challenge has been growing, but rarely involves Bayesian methods. Instead, many geomorphic models rely on estimated averages that largely miss out on the variability of form and process. Yet seemingly fixed estimates of channel heads, sediment rating curves or glacier equilibrium lines, for example, are all prone to uncertainties. Neighbouring scientific disciplines such as physics, hydrology or ecology have readily embraced Bayesian methods to fully capture and better explain such uncertainties, as the necessary computational tools have advanced greatly. The aim of this article is to introduce the Bayesian toolkit to scientists concerned with Earth surface processes and landforms, and to show how geomorphic models might benefit from probabilistic concepts. I briefly review the use of Bayesian reasoning in geomorphology, and outline the corresponding variants of regression and classification in several worked examples.
The objective of the study is to develop a better understanding of the capillary circulation in contracting muscles. Ten subjects were measured during a submaximal fatiguing isometric muscle action by use of the O2C spectrophotometer. In all measurements the capillary-venous oxygen saturation of hemoglobin (SvO2) decreases immediately after the start of loading and levels off into a steady state. However, two different patterns (type I and type II) emerged. They differ in the extent of deoxygenation (–10.37 ±2.59 percent points (pp) vs. –33.86 ±17.35 pp, P = .008) and the behavior of the relative hemoglobin amount (rHb). Type I reveals a positive rank correlation of SvO2 and rHb (? = 0.735, P <.001), whereas a negative rank correlation (? = –0.522, P <.001) occurred in type II, since rHb decreases until a reversal point, then increases averagely 13% above the baseline value and levels off into a steady state. The results reveal that a homeostasis of oxygen delivery and consumption during isometric muscle actions is possible. A rough distinction in two types of regulation is suggested.
Being born large for gestational age is associated with increased global placental DNA methylation

Being born small (SGA) or large for gestational age (LGA) is associated with adverse birth outcomes and metabolic diseases in later life of the offspring. It is known that aberrations in growth during gestation are related to altered placental function. Placental function is regulated by epigenetic mechanisms such as DNA methylation. Several studies in recent years have demonstrated associations between altered patterns of DNA methylation and adverse birth outcomes. However, larger studies that reliably investigated global DNA methylation are lacking. The aim of this study was to characterize global placental DNA methylation in relationship to size for gestational age. Global DNA methylation was assessed in 1023 placental samples by LC-MS/MS. LGA offspring displayed significantly higher global placental DNA methylation compared to appropriate for gestational age (AGA; p<0.001). ANCOVA analyses adjusted for known factors impacting on DNA methylation demonstrated an independent association between placental global DNA methylation and LGA births (p<0.001). Tertile stratification according to global placental DNA methylation levels revealed a significantly higher frequency of LGA births in the third tertile. Furthermore, a multiple logistic regression analysis corrected for known factors influencing birth weight highlighted an independent positive association between global placental DNA methylation and the frequency of LGA births (p=0.001).
Depending on the biochemical and biotechnical approach, the aim of this work was to understand the mechanism of protein-glucan interactions in regulation and control of starch degradation. Although starch degradation starts with the phosphorylation process, the mechanisms by which this process is controlling and adjusting starch degradation are not yet fully understood. Phosphorylation is a major process performed by the two dikinases enzymes α-glucan, water dikinase (GWD) and phosphoglucan water dikinase (PWD). GWD and PWD enzymes phosphorylate the starch granule surface; thereby stimulate starch degradation by hydrolytic enzymes. Despite these important roles for GWD and PWD, so far the biochemical processes by which these enzymes are able to regulate and adjust the rate of phosphate incorporation into starch during the degradation process haven‘t been understood. Recently, some proteins were found associated with the starch granule. Two of these proteins are named Early Starvation Protein 1 (ESV1) and its homologue Like-Early Starvation Protein 1 (LESV). It was supposed that both are involved in the control of starch degradation, but their function has not been clearly known until now. To understand how ESV1 and LESV-glucan interactions are regulated and affect the starch breakdown, it was analyzed the influence of ESV1 and LESV proteins on the phosphorylating enzyme GWD and PWD and hydrolysing enzymes ISA, BAM, and AMY. However, the analysis determined the location of LESV and ESV1 in the chloroplast stroma of Arabidopsis. Mass spectrometry data predicted ESV1and LESV proteins as a product of the At1g42430 and At3g55760 genes with a predicted mass of ~50 kDa and ~66 kDa, respectively. The ChloroP program predicted that ESV1 lacks the chloroplast transit peptide, but it predicted the first 56 amino acids N-terminal region as a chloroplast transit peptide for LESV. Usually, the transit peptide is processed during transport of the proteins into plastids. Given that this processing is critical, two forms of each ESV1 and LESV were generated and purified, a full-length form and a truncated form that lacks the transit peptide, namely, (ESV1and tESV1) and (LESV and tLESV), respectively. Both protein forms were included in the analysis assays, but only slight differences in glucan binding and protein action between ESV1 and tESV1 were observed, while no differences in the glucan binding and effect on the GWD and PWD action were observed between LESV and tLESV. The results revealed that the presence of the N-terminal is not massively altering the action of ESV1 or LESV. Therefore, it was only used the ESV1 and tLESV forms data to explain the function of both proteins.
However, the analysis of the results revealed that LESV and ESV1 proteins bind strongly at the starch granule surface. Furthermore, not all of both proteins were released after their incubation with starches after washing the granules with 2% [w/v] SDS indicates to their binding to the deeper layers of the granule surface. Supporting of this finding comes after the binding of both proteins to starches after removing the free glucans chains from the surface by the action of ISA and BAM. Although both proteins are capable of binding to the starch structure, only LESV showed binding to amylose, while in ESV1, binding was not observed. The alteration of glucan structures at the starch granule surface is essential for the incorporation of phosphate into starch granule while the phosphorylation of starch by GWD and PWD increased after removing the free glucan chains by ISA. Furthermore, PWD showed the possibility of starch phosphorylation without prephosphorylation by GWD.
Biochemical studies on protein-glucan interactions between LESV or ESV1 with different types of starch showed a potentially important mechanism of regulating and adjusting the phosphorylation process while the binding of LESV and ESV1 leads to altering the glucan structures of starches, hence, render the effect of the action of dikinases enzymes (GWD and PWD) more able to control the rate of starch degradation. Despite the presence of ESV1 which revealed an antagonistic effect on the PWD action as the PWD action was decreased without prephosphorylation by GWD and increased after prephosphorylation by GWD (Chapter 4), PWD showed a significant reduction in its action with or without prephosphorylation by GWD in the presence of ESV1 whether separately or together with LESV (Chapter 5). However, the presence of LESV and ESV1 together revealed the same effect compared to the effect of each one alone on the phosphorylation process, therefore it is difficult to distinguish the specific function between them. However, non-interactions were detected between LESV and ESV1 or between each of them with GWD and PWD or between GWD and PWD indicating the independent work for these proteins. It was also observed that the alteration of the starch structure by LESV and ESV1 plays a role in adjusting starch degradation rates not only by affecting the dikinases but also by affecting some of the hydrolysing enzymes since it was found that the presence of LESV and ESV1leads to the reduction of the action of BAM, but does not abolish it.
Ferruginous conditions were a prominent feature of the oceans throughout the Precambrian Eons and thus throughout much of Earth’s history. Organic matter mineralization and diagenesis within the ferruginous sediments that deposited from Earth’s early oceans likely played a key role in global biogeochemical cycling. Knowledge of organic matter mineralization in ferruginous sediments, however, remains almost entirely conceptual, as modern analogue environments are extremely rare and largely unstudied, to date. Lake Towuti on the island of Sulawesi, Indonesia is such an analogue environment and the purpose of this PhD project was to investigate the rates and pathways of organic matter mineralization in its ferruginous sediments.
Lake Towuti is the largest tectonic lake in Southeast Asia and is hosted in the mafic and ultramafic rocks of the East Sulawesi Ophiolite. It has a maximum water depth of 203 m and is weakly thermally stratified. A well-oygenated surface layer extends to 70 m depth, while waters below 130 m are persistently anoxic. Intensive weathering of the ultramafic catchment feeds the lake with large amounts of iron(oxy)hydroxides while the runoff contains only little sulfate, leading to sulfate-poor (< 20 µM) lake water and anoxic ferruginous conditions below 130 m. Such conditions are analogous to the ferruginous water columns that persisted throughout much of the Archean and Proterozoic eons. Short (< 35 cm) sediment cores were collected from different water depths corresponding to different bottom water redox conditions. Also, a drilling campaign of the International Continental Scientific Drilling Program (ICDP) retrieved a 114 m long sediment core dedicated for geomicrobiological investigations from a water depth of 153 m, well below the depth of oxygen penetration at the time of sampling. Samples collected from these sediment cores form the fundament of this thesis and were used to perform a suite of biogeochemical and microbiological analyses.
Geomirobiological investigations depend on uncontaminated samples. However, exploration of subsurface environments relies on drilling, which requires the use of a drilling fluid. Drilling fluid infiltration during drilling can not be avoided. Thus, in order to trace contamination of the sediment core and to identify uncontaminated samples for further analyses a simple and inexpensive technique for assessing contamination during drilling operations was developed and applied during the ICDP drilling campaign. This approach uses an aqeous fluorescent pigment dispersion commonly used in the paint industry as a particulate tracer. It has the same physical properties as conventionally used particulate tracers. However, the price is nearly four orders of magnitude lower solving the main problem of particulate tracer approaches. The approach requires only a minimum of equipment and allows for a rapid contamination assessment potentially even directly on site, while the senstitivity is in the range of already established approaches. Contaminated samples in the drill core were identified and not included for further geomicrobiological investigations.
Biogeochemical analyses of short sediment cores showed that Lake Towutis sediments are strongly depleted in electron acceptors commonly used in microbial organic matter mineralization (i.e. oxygen, nitrate, sulfate). Still, the sediments harbor high microbial cell densities, which are a function of redox conditions of Lake Towuti’s bottom water. In shallow water depths bottom water oxygenation leads to a higher input of labile organic matter and electron acceptors like sulfate and iron, which promotes a higher microbial abundance. Microbial analyses showed that a versatile microbial community with a potential to perform metabolisms related to iron and sulfate reduction, fermentation as well as methanogenesis inhabits Lake Towuti’s surface sediments.
Biogeochemical investigations of the upper 12 m of the 114 m sediment core showed that Lake Towuti’s sediment is extremely rich in iron with total concentrations up to 2500 µmol cm-3 (20 wt. %), which makes it the natural sedimentary environment with the highest total iron concentrations studied to date. In the complete or near absence of oxygen, nitrate and sulfate, organic matter mineralization in ferruginous sediments would be expected to proceed anaerobically via the energetically most favorable terminal electron acceptors available - in this case ferric iron. Astonishingly, however, methanogenesis is the dominant (>85 %) organic matter mineralization process in Lake Towuti’s sediment. Reactive ferric iron known to be available for microbial iron reduction is highly abundant throughout the upper 12 m and thus remained stable for at least 60.000 years. The produced methane is not oxidized anaerobically and diffuses out of the sediment into the water column. The proclivity towards methanogenesis, in these very iron-rich modern sediments, implies that methanogenesis may have played a more important role in organic matter mineralization thoughout the Precambrian than previously thought and thus could have been a key contributor to Earth’s early climate dynamics.
Over the whole sequence of the 114 m long sediment core siderites were identified and characterized using high-resolution microscopic and spectroscopic imaging together with microchemical and geochemical analyses. The data show early diagenetic growth of siderite crystals as a response to sedimentary organic matter mineralization. Microchemical zoning was identified in all siderite crystals. Siderite thus likely forms during diagenesis through growth on primary existing phases and the mineralogical and chemical features of these siderites are a function of changes in redox conditions of the pore water and sediment over time. Identification of microchemical zoning in ancient siderites deposited in the Precambrian may thus also be used to infer siderite growth histories in ancient sedimentary rocks including sedimentary iron formations.

Carbonates play a key role in the chemistry and dynamics of our planet. They are directly connected to the CO2 budget of our atmosphere and have a great impact on the deep carbon cycle. Moreover, recent studies have shown that carbonates are stable along the geothermal gradient down to Earth's lower mantle conditions, changing their crystal structure and related properties. Subducted carbonates may also react with silicates to form new phases. These reactions will redistribute elements, such as calcium (Ca), magnesium (Mg), iron (Fe) and carbon in the form of carbon dioxide (CO2), but also trace elements, that are carried by the carbonates. The trace elements of most interest are strontium (Sr) and rare earth elements (REE) which have been found to be important constituents in the composition of the primitive lower mantle and in mineral inclusions found in super-deep diamonds. However, the stability of carbonates in presence of mantle silicates at relevant temperatures is far from being well understood. Related to this, very little is known about distribution processes of trace elements between carbonates and mantle silicates. To shed light on these processes, we studied reactions between Sr- and REE-containing CaCO3 and Mg/Fe-bearing silicates of the system (Mg,Fe)2SiO4 - (Mg,Fe)SiO3 at high pressure and high temperature using synchrotron radiation based μ-X-ray diffraction (μ-XRD) and μ-X-ray fluorescence (μ-XRF) with μm-resolution in a laser-heated diamond anvil cell. X-ray diffraction is used to derive the structural changes of the phase reactions whereas X-ray fluorescence gives information on the chemical changes in the sample. In-situ experiments at high pressure and high temperature were performed at beamline P02.2 at PETRA III (Hamburg, Germany) and at beamline ID27 at ESRF (Grenoble, France). In addition to μ-XRD and μ-XRF, ex-situ measurements were made on the recovered sample material using transmission electron microscopy (TEM) and provided further insights into the reaction kinetics of carbonate-silicate reactions.
Our investigations show that CaCO3 is unstable in presence of mantle silicates above 1700 K and a reaction takes place in which magnesite plus CaSiO3-perovskite are formed. In addition, we observed that a high content of iron in the carbonate-silicate system favours dolomite formation during the reaction. The subduction of natural carbonates with significant amounts of Sr leads to a comprehensive investigation of the stability not only of CaCO3 phases in contact with mantle silicates but also of SrCO3 (and of Sr-bearing CaCO3). We found that SrCO3 reacts with (Mg,Fe)SiO3-perovskite to form magnesite and gained evidence for the formation of SrSiO3-perovskite.
To complement our study on the stability of SrCO3 at conditions of the Earth's lower mantle, we performed powder X-ray diffraction and single crystal X-ray diffraction experiments at ambient temperature and up to 49 GPa. We observed a transformation from SrCO3-I into a new high-pressure phase SrCO3-II at around 26 GPa with Pmmn crystal structure and a bulk modulus of 103(10) GPa. This information is essential to fully understand the phase behaviour and stability of carbonates in the Earth's lower mantle and to elucidate the possibility of introducing Sr into mantle silicates by carbonate-silicate reactions.
Simultaneous recording of μ-XRD and μ-XRF in the μm-range over the heated areas provides spatial information not only about phase reactions but also on the elemental redistribution during the reactions. A comparison of the spatial intensity distribution of the XRF signal before and after heating indicates a change in the elemental distribution of Sr and an increase in Sr-concentration was found around the newly formed SrSiO3-perovskite. With the help of additional TEM analyses on the quenched sample material the elemental redistribution was studied at a sub-micrometer scale. Contrary to expectations from combined μ-XRD and μ-XRF measurements, we found that La and Eu were not incorporated into the silicate phases, instead they tend to form either isolated oxide phases (e.g. Eu2O3, La2O3) or hydroxyl-bastnäsite (La(CO3)(OH)). In addition, we observed the transformation from (Mg,Fe)SiO3-perovskite to low-pressure clinoenstatite during pressure release. The monoclinic structure (P21/c) of this phase allows the incorporation of Ca as shown by additional EDX analyses and, to a minor extent, Sr too.
Based on our experiments, we can conclude that a detection of the trace elements in-situ at high pressure and high temperature remains challenging. However, our first findings imply that silicates may incorporate the trace elements provided by the carbonates and indicate that carbonates may have a major effect on the trace element contents of mantle phases.
Carrion plays an essential role in shaping the structure and functioning of ecosystems and has far‐reaching implications for biodiversity conservation. The change in availability and type of carcasses throughout ecosystems can involve negative effects for scavenging communities. To address this issue, there have been recent conservation management measures of carrion provision in natural systems. However, the optimal conditions under which exposing carcasses to optimize conservation outcomes are still limited. Here, we used camera traps throughout elevational and vegetational gradients to monitor the consumption of 48 deer carcasses over a study period of six years by evaluating 270,279 photographs resulting out of 15,373 trap nights. We detected 17 species visiting carcass deployments, including five endangered species. Our results show that large carcasses, the winter season, and a heterogeneous surrounding habitat enhanced the frequency of carcass visits and the species richness of scavenger assemblages. Contrary to our expectations, carcass species, condition (fresh/frozen), and provision schedule (continuous vs single exposure) did not influence scavenging frequency or diversity. The carcass visitation frequency increased with carcass mass and lower temperatures. The effect of large carcasses was especially pronounced for mesopredators and the Eurasian lynx (Lynx lynx ). Lynx were not too influenced in its carrion acquisition by the season, but exclusively preferred remote habitats containing higher forest cover. Birds of prey, mesopredators, and top predators were also positively influenced by the visiting rate of ravens (Corvus corax ), whereas no biotic or abiotic preferences were found for wild boars (Sus scrofa ). This study provides evidence that any ungulate species of carrion, either in a fresh or in previously frozen condition, attracts a high diversity of scavengers especially during winter, thereby supporting earlier work that carcass provisions may support scavenger communities and endangered species.
Objective:
Stunting (height-for-age < −2 SD) is one of the forms of undernutrition and is frequent among children of low- and middle-income countries. But stunting perSe is not a synonym of undernutrition. We investigated association between body height and indicators of energetic undernutrition at three critical thresholds for thinness used in public health: (1) BMI SDS < −2; (2) mid-upper arm circumference divided by height (MUAC (mm) × 10/height (cm) < 1·36) and (3) mean skinfold thickness (SF) < 7 mm and to question the reliability of thresholds as indicators of undernutrition.
Design:
Cross-sectional study; breakpoint analysis.
Setting:
Rural and urban regions of Indonesia and Guatemala – different socio-economic status (SES).
Participants:
1716 Indonesian children (6·0–13·2 years) and 3838 Guatemalan children (4·0–18·9 years) with up to 50 % stunted children.
Results:
When separating the regression of BMI, MUAC or SF, on height into distinguishable segments (breakpoint analysis), we failed to detect relevant associations between height, and BMI, MUAC or SF, even in the thinnest and shortest children. For BMI and SF, the breakpoint analysis either failed to reach statistical significance or distinguished at breakpoints above critical thresholds. For MUAC, the breakpoint analysis yielded negative associations between MUAC/h and height in thin individuals. Only in high SES Guatemalan children, SF and height appeared mildly associated with R2 = 0·017.
Conclusions:
Currently used lower thresholds of height-for-age (stunting) do not show relevant associations with anthropometric indicators of energetic undernutrition. We recommend using the catch-up growth spurt during early re-feeding instead as immediate and sensitive indicator of past undernourishment. We discuss the primacy of education and social-economic-political-emotional circumstances as responsible factors for stunting.
Machine learning (ML) algorithms are being increasingly used in Earth and Environmental modeling studies owing to the ever-increasing availability of diverse data sets and computational resources as well as advancement in ML algorithms. Despite advances in their predictive accuracy, the usefulness of ML algorithms for inference remains elusive. In this study, we employ two popular ML algorithms, artificial neural networks and random forest, to analyze a large data set of flood events across Germany with the goals to analyze their predictive accuracy and their usability to provide insights to hydrologic system functioning. The results of the ML algorithms are contrasted against a parametric approach based on multiple linear regression. For analysis, we employ a model-agnostic framework named Permuted Feature Importance to derive the influence of models' predictors. This allows us to compare the results of different algorithms for the first time in the context of hydrology. Our main findings are that (1) the ML models achieve higher prediction accuracy than linear regression, (2) the results reflect basic hydrological principles, but (3) further inference is hindered by the heterogeneity of results across algorithms. Thus, we conclude that the problem of equifinality as known from classical hydrological modeling also exists for ML and severely hampers its potential for inference. To account for the observed problems, we propose that when employing ML for inference, this should be made by using multiple algorithms and multiple methods, of which the latter should be embedded in a cross-validation routine.
Increased N400 amplitudes on indefinite articles (a/an) incompatible with expected nouns have been initially taken as strong evidence for probabilistic pre-activation of phonological word forms, and recently been intensely debated because they have been difficult to replicate. Here, these effects are simulated using a neural network model of sentence comprehension that we previously used to simulate a broad range of empirical N400 effects. The model produces the effects when the cue validity of the articles concerning upcoming noun meaning in the learning environment is high, but fails to produce the effects when the cue validity of the articles is low due to adjectives presented between articles and nouns during training. These simulations provide insight into one of the factors potentially contributing to the small size of the effects in empirical studies and generate predictions for cross-linguistic differences in article induced N400 effects based on articles’ cue validity. The model accounts for article induced N400 effects without assuming pre-activation of word forms, and instead simulates these effects as the stimulus-induced change in a probabilistic representation of meaning corresponding to an implicit semantic prediction error.
The Fram Strait is an area with a relatively low and irregular distribution of diatom microfossils in surface sediments, and thus microfossil records are scarce, rarely exceed the Holocene, and contain sparse information about past richness and taxonomic composition. These attributes make the Fram Strait an ideal study site to test the utility of sedimentary ancient DNA (sedaDNA) metabarcoding. Amplifying a short, partial rbcL marker from samples of sediment core MSM05/5-712-2 resulted in 95.7 % of our sequences being assigned to diatoms across 18 different families, with 38.6 % of them being resolved to species and 25.8 % to genus level. Independent replicates show a high similarity of PCR products, especially in the oldest samples. Diatom sedaDNA richness is highest in the Late Weichselian and lowest in Mid- and Late Holocene samples. Taxonomic composition is dominated by cold-water and sea-ice-associated diatoms and suggests several reorganisations – after the Last Glacial Maximum, after the Younger Dryas, and after the Early and after the Mid-Holocene. Different sequences assigned to, amongst others, Chaetoceros socialis indicate the detectability of intra-specific diversity using sedaDNA. We detect no clear pattern between our diatom sedaDNA record and the previously published IP25 record of this core, although proportions of pennate diatoms increase with higher IP25 concentrations and proportions of Nitzschia cf. frigida exceeding 2 % of the assemblage point towards past sea-ice presence.
The significant environmental and socioeconomic consequences of hydrometeorological extreme events, such as extreme rainfall, are constituted as a most important motivation for analyzing these events in the south-central Andes of NW Argentina. The steep topographic and climatic gradients and their interactions frequently lead to the formation of deep convective storms and consequently trigger extreme rainfall generation.
In this dissertation, I focus on identifying the dominant climatic variables and atmospheric conditions and their spatiotemporal variability leading to deep convection and extreme rainfall in the south-central Andes.
This dissertation first examines the significant contribution of temperature on atmospheric humidity (dew-point temperature, Td) and on convection (convective available potential energy, CAPE) for deep convective storms and hence, extreme rainfall along the topographic and climatic gradients. It was found that both climatic variables play an important role in extreme rainfall generation. However, their contributions differ depending on topographic and climatic sub-regions, as well as rainfall percentiles.
Second, this dissertation explores if (near real-time) the measurements conducted by the Global Navigation Satellite System (GNSS) on integrated water vapor (IWV) provide reliable data for explaining atmospheric humidity. I argue that GNSS-IWV, in conjunction with other atmospheric stability parameters such as CAPE, is able to decipher the extreme rainfall in the eastern central Andes. In my work, I rely on a multivariable regression analysis described by a theoretical relationship and fitting function analysis.
Third, this dissertation identifies the local impact of convection on extreme rainfall in the eastern Andes. Relying on a Principal Component Analysis (PCA) it was found that during the existence of moist and warm air, extreme rainfall is observed more often during local night hours. The analysis includes the mechanisms for this observation.
Exploring the atmospheric conditions and climatic variables leading to extreme rainfall is one of the main findings of this dissertation. The conditions and variables are a prerequisite for understanding the dynamics of extreme rainfall and predicting these events in the eastern Andes.
The performance of the recently commissioned spectrometer PEAXIS for resonant inelastic soft X-ray scattering (RIXS) and X-ray photoelectron spectroscopy and its hosting beamline U41-PEAXIS at the BESSY II synchrotron are characterized. The beamline provides linearly polarized light from 180 eV to 1600 eV allowing for RIXS measurements in the range 200-1200 eV. The monochromator optics can be operated in different configurations to provide either high flux with up to 10(12) photons s(-1) within the focal spot at the sample or high energy resolution with a full width at half maximum of <40 meV at an incident photon energy of similar to 400 eV. The measured total energy resolution of the RIXS spectrometer is in very good agreement with theoretically predicted values obtained by ray-tracing simulations. PEAXIS features a 5 m-long RIXS spectrometer arm that can be continuously rotated about the sample position by 106 degrees within the horizontal photon scattering plane, thus enabling the study of momentum-transfer-dependent excitations. Selected scientific examples are presented to demonstrate the instrument capabilities, including measurements of excitations in single-crystalline NiO and in liquid acetone employing a fluid cell sample manipulator. Planned upgrades of the beamline and the RIXS spectrometer to further increase the energy resolution to similar to 100 meV at 1000 eV incident photon energy are discussed.
Understanding the hydrologic connectivity between kettle holes and shallow groundwater, particularly in reaction to the highly variable local meteorological conditions, is of paramount importance for tracing water in a hydro(geo)logically complex landscape and thus for integrated water resource management. This article is aimed at identifying the dominant hydrological processes affecting the kettle holes' water balance and their interactions with the shallow groundwater domain in the Uckermark region, located in the north-east of Germany. For this reason, based on the stable isotopes of oxygen (delta O-18) and hydrogen (delta H-2), an isotopic mass balance model was employed to compute the evaporative loss of water from the kettle holes from February to August 2017. Results demonstrated that shallow groundwater inflow may play the pivotal role in the processes taking part in the hydrology of the kettle holes in the Uckermark region. Based on the calculated evaporation/inflow (E/I) ratios, most of the kettle holes (86.7%) were ascertained to have a partially open, flow-through-dominated system. Moreover, we identified an inverse correlation between E/I ratios and the altitudes of the kettle holes. The same holds for electrical conductivity (EC) and the altitudes of the kettle holes. In accordance with the findings obtained from this study, a conceptual model explaining the interaction between the shallow groundwater and the kettle holes of Uckermark was developed. The model exhibited that across the highest altitudes, the recharge kettle holes are dominant, where a lower ratio of E/I and a lower EC was detected. By contrast, the lowest topographical depressions represent the discharge kettle holes, where a higher ratio of E/I and EC could be identified. The kettle holes existing in between were categorized as flow-through kettle holes through which the recharge takes place from one side and discharge from the other side.
Bacterial chemotaxis-a fundamental example of directional navigation in the living world-is key to many biological processes, including the spreading of bacterial infections. Many bacterial species were recently reported to exhibit several distinct swimming modes-the flagella may, for example, push the cell body or wrap around it. How do the different run modes shape the chemotaxis strategy of a multimode swimmer? Here, we investigate chemotactic motion of the soil bacterium Pseudomonas putida as a model organism. By simultaneously tracking the position of the cell body and the configuration of its flagella, we demonstrate that individual run modes show different chemotactic responses in nutrition gradients and, thus, constitute distinct behavioral states. On the basis of an active particle model, we demonstrate that switching between multiple run states that differ in their speed and responsiveness provides the basis for robust and efficient chemotaxis in complex natural habitats.
Model-derived relationships between chlorophyll a (Chl-a) and nutrients and temperature have fundamental implications for understanding complex interactions among water quality measures used for lake classification, yet accuracy comparisons of different approaches are scarce. Here, we (1) compared Chl-a model performances across linear and nonlinear statistical approaches; (2) evaluated single and combined effects of nutrients, depth, and temperature as lake surface water temperature (LSWT) or altitude on Chl-a; and (3) investigated the reliability of the best water quality model across 13 lakes from perialpine and central Balkan mountain regions. Chl-a was modelled using in situ water quality data from 157 European lakes; elevation data and LSWT in situ data were complemented by remote sensing measurements. Nonlinear approaches performed better, implying complex relationships between Chl-a and the explanatory variables. Boosted regression trees, as the best performing approach, accommodated interactions among predictor variables. Chl-a-nutrient relationships were characterized by sigmoidal curves, with total phosphorus having the largest explanatory power for our study region. In comparison with LSWT, utilization of altitude, the often-used temperature surrogate, led to different influence directions but similar predictive performances. These results support utilizing altitude in models for Chl-a predictions. Compared to Chl-a observations, Chl-a predictions of the best performing approach for mountain lakes (oligotrophic-eutrophic) led to minor differences in trophic state categorizations. Our findings suggest that both models with LSWT and altitude are appropriate for water quality predictions of lakes in mountain regions and emphasize the importance of incorporating interactions among variables when facing lake management challenges.
Numerous studies investigate which sanctioning institutions prevent cartel formation but little is known as to how these sanctions work. We contribute to understanding the inner workings of cartels by studying experimentally the effect of sanctioning institutions on firms’ communication. Using machine learning to organize the chat communication into topics, we find that firms are significantly less likely to communicate explicitly about price fixing when sanctioning institutions are present. At the same time, average prices are lower when communication is less explicit. A mediation analysis suggests that sanctions are effective in hindering cartel formation not only because they introduce a risk of being fined but also by reducing the prevalence of explicit price communication.

In nature, plants are often subjected to periods of recurrent environmental stress that can strongly affect their development and productivity. To cope with these conditions, plants can remember a previous stress, which allows them to respond more efficiently to a subsequent stress, a phenomenon known as priming. This ability can be maintained at the somatic level for a few days or weeks after the stress is perceived, suggesting that plants can store information of a past stress during this recovery phase. While the immediate responses to a single stress event have been extensively studied, knowledge on priming effects and how stress memory is stored is still scarce. At the molecular level, memory of a past condition often involves changes in chromatin structure and organization, which may be maintained independently from transcription. In this review, we will summarize the most recent developments in the field and discuss how different levels of chromatin regulation contribute to priming and plant abiotic stress memory.

In this dissertation a series of experimental studies are presented which demonstrate that the exhaustive inference of focus-background it-clefts in English and their cross-linguistic counterparts in Akan, French, and German is neither robust nor systematic. The inter-speaker and cross-linguistic variability is accounted for with a discourse-pragmatic approach to cleft exhaustivity, in which -- following Pollard & Yasavul 2016 -- the exhaustive inference is derived from an interaction with another layer of meaning, namely, the existence presupposition encoded in clefts.
Coarse-grained molecular model for the Glycosylphosphatidylinositol anchor with and without protein

Glycosylphosphatidylinositol (GPI) anchors are a unique class of complex glycolipids that anchor a great variety of proteins to the extracellular leaflet of plasma membranes of eukaryotic cells. These anchors can exist either with or without an attached protein called GPI-anchored protein (GPI-AP) both in vitro and in vivo. Although GPIs are known to participate in a broad range of cellular functions, it is to a large extent unknown how these are related to GPI structure and composition. Their conformational flexibility and microheterogeneity make it difficult to study them experimentally. Simplified atomistic models are amenable to all-atom computer simulations in small lipid bilayer patches but not suitable for studying their partitioning and trafficking in complex and heterogeneous membranes. Here, we present a coarse-grained model of the GPI anchor constructed with a modified version of the MARTINI force field that is suited for modeling carbohydrates, proteins, and lipids in an aqueous environment using MARTINI's polarizable water. The nonbonded interactions for sugars were reparametrized by calculating their partitioning free energies between polar and apolar phases. In addition, sugar-sugar interactions were optimized by adjusting the second virial coefficients of osmotic pressures for solutions of glucose, sucrose, and trehalose to match with experimental data. With respect to the conformational dynamics of GPI-anchored green fluorescent protein, the accessible time scales are now at least an order of magnitude larger than for the all-atom system. This is particularly important for fine-tuning the mutual interactions of lipids, carbohydrates, and amino acids when comparing to experimental results. We discuss the prospective use of the coarse-grained GPI model for studying protein-sorting and trafficking in membrane models.

Similarly, it is acknowledged that in order to prove the Collatz conjecture, one must demonstrate that this tree covers all (odd) natural numbers. A structured reachability analysis is hitherto not available. This paper investigates the problem from a graph theory perspective. We define a tree that consists of nodes labeled with Collatz sequence numbers. This tree will be transformed into a sub-tree that only contains odd labeled nodes. The analysis of this tree will provide new insights into the structure of Collatz sequences. The findings are of special interest to possible cycles within a sequence. Next, we describe the conditions which must be fulfilled by a cycle. Finally, we demonstrate how these conditions could be used to prove that the only possible cycle within a Collatz sequence is the trivial cycle, starting with the number 1, as conjectured by Lothar Collatz.

Finally, we transform the Collatz graph into a binary tree, following the approach of Kleinnijenhuis, which could form the basis for a comprehensive proof of the conjecture.
Polynucleobacter asymbioticus strain QLW-P1DMWA-1T represents a group of highly successful heterotrophic ultramicrobacteria that is frequently very abundant (up to 70% of total bacterioplankton) in freshwater habitats across all seven continents. This strain was originally isolated from a shallow Alpine pond characterized by rapid changes in water temperature and elevated UV radiation due to its location at an altitude of 1300 m. To elucidate the strain’s adjustment to fluctuating environmental conditions, we recorded changes occurring in its transcriptomic and proteomic profiles under contrasting experimental conditions by simulating thermal conditions in winter and summer as well as high UV irradiation. To analyze the potential connection between gene expression and regulation via methyl group modification of the genome, we also analyzed its methylome. The methylation pattern differed between the three treatments, pointing to its potential role in differential gene expression. An adaptive process due to evolutionary pressure in the genus was deduced by calculating the ratios of non-synonymous to synonymous substitution rates for 20 Polynucleobacter spp. genomes obtained from geographically diverse isolates. The results indicate purifying selection.

One of the most important social cognitive skills in humans is the ability to “put oneself in someone else’s shoes,” that is, to take another person’s perspective. In socially situated communication, perspective taking enables the listener to arrive at a meaningful interpretation of what is said (sentence meaning) and what is meant (speaker’s meaning) by the speaker. To successfully decode the speaker’s meaning, the listener has to take into account which information he/she and the speaker share in their common ground (CG). We here further investigated competing accounts about when and how CG information affects language comprehension by means of reaction time (RT) measures, accuracy data, event-related potentials (ERPs), and eye-tracking. Early integration accounts would predict that CG information is considered immediately and would hence not expect to find costs of CG integration. Late integration accounts would predict a rather late and effortful integration of CG information during the parsing process that might be reflected in integration or updating costs. Other accounts predict the simultaneous integration of privileged ground (PG) and CG perspectives. We used a computerized version of the referential communication game with object triplets of different sizes presented visually in CG or PG. In critical trials (i.e., conflict trials), CG information had to be integrated while privileged information had to be suppressed. Listeners mastered the integration of CG (response accuracy 99.8%). Yet, slower RTs, and enhanced late positivities in the ERPs showed that CG integration had its costs. Moreover, eye-tracking data indicated an early anticipation of referents in CG but an inability to suppress looks to the privileged competitor, resulting in later and longer looks to targets in those trials, in which CG information had to be considered. Our data therefore support accounts that foresee an early anticipation of referents to be in CG but a rather late and effortful integration if conflicting information has to be processed. We show that both perspectives, PG and CG, contribute to socially situated language processing and discuss the data with reference to theoretical accounts and recent findings on the use of CG information for reference resolution.
Many technical challenges still need to be overcome to improve the quality of the green coffee beans. In this work, the wet Arabica coffee processing in batch and continuous modus were investigated. Coffee beans samples as well as by-products and wastewaters collected at different production steps were analyzed in terms of their content in total phenols, antioxidant capacity, caffeine content, organic acids, reducing sugars, free amino group and protein content. The results showed that 40% of caffeine was removed with pulp. Green coffee beans showed highest concentration of organic acids and sucrose (4.96 ± 0.25 and 5.07 ± 0.39 g/100 g DW for the batch and continuous processing). Batch green coffee beans contained higher amount of phenols. 5-caffeoylquinic Acid (5-CQA) was the main constituent (67.1 and 66.0% for the batch and continuous processing, respectively). Protein content was 15 and 13% in the green coffee bean in batch and continuous processing, respectively. A decrease of 50 to 64% for free amino groups during processing was observed resulting in final amounts of 0.8 to 1.4% in the processed beans. Finally, the batch processing still revealed by-products and wastewater with high nutrient content encouraging a better concept for valorization.
Completely water-based systems are of interest for the development of novel material for various reasons: On one hand, they provide benign environment for biological systems and on the other hand they facilitate effective molecular transport in a membrane-free environment. In order to investigate the general potential of aqueous two-phase systems (ATPSs) for biomaterials and compartmentalized systems, various solid particles were applied to stabilize all-aqueous emulsion droplets. The target ATPS to be investigated should be prepared via mixing of two aqueous solutions of water-soluble polymers, which turn biphasic when exceeding a critical polymer concentration. Hydrophilic polymers with a wide range of molar mass such as dextran/poly(ethylene glycol) (PEG) can therefore be applied. Solid particles adsorbed at the interfaces can be exceptionally efficient stabilizers forming so-called Pickering emulsions, and nanoparticles can bridge the correlation length of polymer solutions and are thereby the best option for water-in-water emulsions.
The first approach towards the investigation of ATPS was conducted with all aqueous dextran-PEG emulsions in the presence of poly(dopamine) particles (PDP) in Chapter 4. The water-in-water emulsions were formed with a PEG/dextran system via utilizing PDP as stabilizers. Studies of the formed emulsions were performed via laser scanning confocal microscope (CLSM), optical microscope (OM), cryo-scanning electron microscope (SEM) and tensiometry. The stable emulsions (at least 16 weeks) were demulsified easily via dilution or surfactant addition. Furthermore, the solid PDP at the water-water interface were crosslinked in order to inhibit demulsification of the Pickering emulsion. Transmission electron microscope (TEM) and scanning electron microscope (SEM) were used to visualize the morphology of PDP before and after crosslinking. PDP stabilized water-in-water emulsions were utilized in the following Chapter 5 to form supramolecular compartmentalized hydrogels. Here, hydrogels were prepared in pre-formed water-in-water emulsions and gelled via α-cyclodextrin-PEG (α-CD-PEG) inclusion complex formation. Studies of the formed complexes were performed via X-ray powder diffraction (XRD) and the mechanical properties of the hydrogels were measured with oscillatory shear rheology. In order to verify the compartmentalized state and its triggered decomposition, hydrogels and emulsions were assessed via OM, SEM and CLSM. The last chapter broadens the investigations from the previous two systems by utilizing various carbon nitrides (CN) as different stabilizers in ATPS. CN introduces another way to trigger demulsification, namely irradiation with visible light. Therefore, emulsification and demulsification with various triggers were probed. The investigated all aqueous multi-phase systems will act as model for future fabrication of biocompatible materials, cell micropatterning as well as separation of compartmentalized systems.
The analysis of behavioral models is of high importance for cyber-physical systems, as the systems often encompass complex behavior based on e.g. concurrent components with mutual exclusion or probabilistic failures on demand. The rule-based formalism of probabilistic timed graph transformation systems is a suitable choice when the models representing states of the system can be understood as graphs and timed and probabilistic behavior is important. However, model checking PTGTSs is limited to systems with rather small state spaces.
We present an approach for the analysis of large scale systems modeled as probabilistic timed graph transformation systems by systematically decomposing their state spaces into manageable fragments. To obtain qualitative and quantitative analysis results for a large scale system, we verify that results obtained for its fragments serve as overapproximations for the corresponding results of the large scale system. Hence, our approach allows for the detection of violations of qualitative and quantitative safety properties for the large scale system under analysis. We consider a running example in which we model shuttles driving on tracks of a large scale topology and for which we verify that shuttles never collide and are unlikely to execute emergency brakes. In our evaluation, we apply an implementation of our approach to the running example.
Gender-inclusive language has evolved into a much-debated topic during the past years, discussed interdisciplinarily from theoretical to psycholinguistics, sociology, and economy – and by anyone who uses language.
Studies on German that primarily relied on questionnaires (reviewed in Braun et al. 2005), cloze tests (Klein 1988), and categorisation tasks with picture matching (Irmen & Köhncke 1996) disqualify the generically used masculine forms as pseudo-generic – failing their grammatically prescribed function to include referents of any Gender. Gender-balanced expressions (pair and split forms like Lehrer und Lehrerinnen) make explicit reference to female presence and participation, and thus elevate a more equitable interpretation.
Online methods to investigate the processing of Gender-sensitive language are surprisingly rare among research on the phenomenon, except for reaction time measures (Irmen & Köhncke 1996, Irmen & Kaczmarek 2000) and eye-tracking in reading (Irmen & Schumann 2011).
In addition, Gender-neutral language (GNL) has not been focused on in the majority of experiments, and when it was among the stimuli, results were inconclusive (De Backer & De Cuypere 2012) or found such alternatives to be ineffective (resembling masculine generics, Braun et al. 2005), despite the fact that guidelines on non-discriminatory language use commonly recommend these.
Gender-neutral (GN) expressions for personal reference in German include
• nominalised participles; nominalisations in general: Interessierte, Lehrende
• collective singulars: Publikum, Kollegium
• compounds (e.g., with a notion of “-person”): Ansprechpersonen, Lehrkräfte
• paraphrases that background a (gendered) subject: e.g., passives, relatives
In a visual world eye-tracking study, the comprehension of plural generics using masculine nouns and GN forms was tested for roles and occupations.
In complex stimulus scenarios, reference had to be established to referent images presented on a screen. At the end of each item, a question was asked in order to (re)identify the image that matched the referents of the respective setting best. Images depicted 1) a single person (protagonist), 2) an all-female group, 3) an all-male group, 4) a mixed Gender group of female and male members. The group referents were introduced with either a) masculine nouns (die Lehrer), b) female-specific feminine nouns (die Lehrerinnen), or c) one of the upper three nominal GN variants (die Lehrkräfte).
Results confirm the frequent male bias in masculine forms that are used as generics, that is, their male-specific interpretation. Furthermore, stereotypicality of nouns had an impact on responses. The GN alternatives, which are generally known to aim for indefinite reference (“marked” for Gender-fair language) were found to be most qualified to elicit mixed Gender group interpretations. When reference was established with GN terms, an inclusive response was consistently elicited. This was both indicated by eye movements and response proportions, but to a different extent depending on the particular GN noun type. Concepts that abstract from Gender in their linguistic forms (“neutralising” it) appear to be more inclusive, and thus better candidates for generic reference than masculines.
