We suggest several ideas which when combined could lead to a new mechanism for long-term pulsations of very hot and luminous stars. These involve the interplay between convection, radiation, atmospheric clumping and winds, which collectively feed back to stellar expansion and contraction. We discuss these ideas and point out the future work required in order to fill in the blanks.
Methanhydrate sind besonders in Verbindung mit den steigenden Weltmarktpreisen für Öl und Gas in den vergangenen Jahren mehr und mehr in den Fokus der Energiewirtschaft geraten, was zu einer starken Zunahme der angewandten Forschungsprojekte auf diesem Gebiet führte. Da Methanhydrat nur unter hohem Druck und niedrigen Temperaturen stabil ist, ist die Gewinnung natürlicher Proben für Laboruntersuchungen technisch sehr aufwendig und vor allem teuer. Zur Charakterisierung der Eigenschaften hydratführender Reservoire ist man häufig auf die Herstellung synthetischer Proben angewiesen. Die Eigenschaften der synthetisierten Proben sind dabei abhängig von der Herstellungsmethode und man ist noch immer auf der Suche nach Verfahren, mit denen sich möglichst „naturnahe“ Proben mit vertretbarem Aufwand erzeugen lassen.
In der vorliegenden Arbeit wurde eine neue, relativ schnell durchführbare Methode getestet, die im Porenraum von Sedimenten schwimmende bzw. gefügestützende Hydrate bildet, wie sie in der Natur vorkommen. Gleichzeitig erzeugt sie eine gleichmäßige Verteilung des Hydrats über die Probe und bietet gute Kontrolle über den Hydratgehalt. Sie funktioniert wie folgt: Eine mit einer KCl-Lösung gesättigte Sedimentprobe wird zu einem bestimmten Teil ausgefroren und das übrige Wasser mit Methan verdrängt. Durch Anlegen eines Methandrucks im Stabilitätsbereich wird das Eis zu Methanhydrat umgesetzt. Im Anschluss wird die Probe erneut mit einer KCl-Lösung gesättigt. Anhand seismischer Messungen konnte bestätigt werden, dass Hydrat mit dem gewünschten Hydrathabitus erzeugt wurde. Des Weiteren wurde gezeigt, dass die eishaltigen Proben aufgrund ähnlicher physikalischer Eigenschaften bereits vor der Umsetzung des Eises zu Methanhydrat als Näherung für Proben mit Porenraumhydrat verwendet werden können.
It is well established in language acquisition research that monolingual children and adult second language learners misinterpret sentences with the universal quantifier every and make quantifier-spreading errors that are attributed to a preference for a match in number between two sets of objects. The present Visual World eye-tracking study tested bilingual heritage Russian–English adults and investigated how they interpret of sentences like Every alligator lies in a bathtub in both languages. Participants performed a sentence–picture verification task while their eye movements were recorded. Pictures showed three pairs of alligators in bathtubs and two extra objects: elephants (Control condition), bathtubs (Overexhaustive condition), or alligators (Underexhaustive condition). Monolingual adults performed at ceiling in all conditions. Heritage language (HL) adults made 20% q-spreading errors, but only in the Overexhaustive condition, and when they made an error they spent more time looking at the two extra bathtubs during the Verb region. We attribute q-spreading in HL speakers to cognitive overload caused by the necessity to integrate conflicting sources of information, i.e. the spoken sentences in their weaker, heritage, language and attention-demanding visual context, that differed with respect to referential salience.

Nonlinear optical response of photochromic azobenzene-functionalized self-assembled monolayers

The combination of photochromic and nonlinear optical (NLO) properties of azobenzene-functionalized self-assembled monolayers (SAMs) constitutes an intriguing step towards novel photonic and optoelectronic devices. By utilizing the second-order NLO process of second harmonic generation (SHG), supported by density-functional theory and correlated wave function method calculations, we demonstrate that the photochromic interface provides the necessary prerequisites en route towards possible future technical applications: we find a high NLO contrast on the order of 16% between the switching states. These are furthermore accessible reversibly and with high efficiencies in terms of cross sections on the order of 10−18 cm2 for both photoisomerization reactions, i.e., drivable by means of low-power LED light sources. Finally, both photostationary states (PSSs) are thermally stable at ambient conditions.
In dieser Bachelorarbeit werden die Bedeutung und die Gebrauchsweisen des Vergangenheitstempus Perfekt in der deutschen und spanischen Sprache kontrastiv untersucht. Aufgrund des Zusammenwirkens der funktional-semantischen Kategorien Temporalität, Aspektualität und Modalität wird der Perfektgebrauch hinsichtlich dieser Kategorien analysiert. Es wird angenommen, dass sich das Perfekt im Spanischen und Deutschen nicht nur in Bezug auf seine temporalen und aspektuellen Werte, sondern auch hinsichtlich seiner modal-evidentiellen Werte und Ausdrucksweisen unterscheidet. Ziel dieses Beitrags ist es, die spezifischen temporalen und aspektuellen Bedeutungen dieses Vergangenheitstempus in der jeweiligen Sprache herauszustellen, kontrastiv zu betrachten und zu zeigen, inwiefern Perfektformen als Evidentialitätsmarker fungieren können. Der modal-evidentiellen Bedeutung des Perfekts kommt in dieser Arbeit eine zentrale Rolle zu, da insbesondere hinsichtlich der deutschen Perfekt-Konstruktionen noch wenige Forschungsansätze vorliegen. Die modalen und evidentiellen Bedeutungen des Perfekts werden in beiden Sprachen in der Schriftsprache unter Einbezug der Erkenntnisse der RAE (2009), Wachtmeister-Bermúdez (2005) und Diewald (2011) diskutiert und anhand von Beispielen aus spanischen und deutschen Pressetexten des CREA sowie der Zeit Online und Süddeutsche Zeitung belegt. Als Ergebnis dieser Untersuchung lässt sich feststellen, dass das Perfekt in beiden Sprachen als Evidentialitätsmarker dienen und verschiedene Arten von Evidenz markieren kann. Die Relevanz der kontrastiven Betrachtung kommt in zwei signifikanten Forschungsbereichen zum Tragen: zum Einen bei der Übersetzung, da die temporalen, aspektuellen und modal-evidentiellen Werte des Perfekts in der jeweiligen Sprache beachtet werden müssen und zum Anderen in der Fremdsprachendidaktik, da das Perfekt verschiedene Stellenwerte in den einzelnen Verbalsystemen einnimmt.
Der klassische Fernsehrundfunk genießt verfassungsrechtlich und einfachgesetzliche eine privilegierte Sonderstellung. Diese setzt sich bei der Kabelbelegung fort, wo er - insbesondere der öffentliche Rundfunk - gegenüber teemedialen Anrufangeboten bevorrechtigt wird. Hintergrund dieser Sonderstellung ist nicht zuletzt die auf den Rundfunk zentrierte Medienordnung. Die Arbeit untersucht, ob diese Medienordnung und damit die Sonderstellung des Rundfunks noch zeitgemäß ist und schlägt Anpassungen vor.
This study presents the development of 1D and 2D Surface Evolution Codes (SECs) and their coupling to any lithospheric-scale (thermo-)mechanical code with a quadrilateral structured surface mesh.
Both SECs involve diffusion as approach for hillslope processes and the stream power law to reflect riverbed incision. The 1D SEC settles sediment that was produced by fluvial incision in the appropriate minimum, while the supply-limited 2D SEC DANSER uses a fast filling algorithm to model sedimantation. It is based on a cellular automaton. A slope-dependent factor in the sediment flux extends the diffusion equation to nonlinear diffusion. The discharge accumulation is achieved with the D8-algorithm and an improved drainage accumulation routine. Lateral incision enhances the incision's modelling. Following empirical laws, it incises channels of several cells width.
The coupling method enables different temporal and spatial resolutions of the SEC and the thermo-mechanical code. It transfers vertical as well as horizontal displacements to the surface model. A weighted smoothing of the 3D surface displacements is implemented. The smoothed displacement vectors transmit the deformation by bilinear interpolation to the surface model. These interpolation methods ensure mass conservation in both directions and prevent the two surfaces from drifting apart.
The presented applications refer to the evolution of the Pamir orogen. A calibration of DANSER's parameters with geomorphological data and a DEM as initial topography highlights the advantage of lateral incision. Preserving the channel width and reflecting incision peaks in narrow channels, this closes the huge gap between current orogen-scale incision models and observed topographies.
River capturing models in a system of fault-bounded block rotations reaffirm the importance of the lateral incision routine for capturing events with channel initiation. The models show a low probability of river capturings with large deflection angles. While the probability of river capturing is directly depending on the uplift rate, the erodibility inside of a dip-slip fault speeds up headward erosion along the fault: The model's capturing speed increases within a fault.
Coupling DANSER with the thermo-mechanical code SLIM 3D emphasizes the versatility of the SEC. While DANSER has minor influence on the lithospheric evolution of an indenter model, the brittle surface deformation is strongly affected by its sedimentation, widening a basin in between two forming orogens and also the southern part of the southern orogen to south, east and west.
Aufgrund ihrer potenziell gesundheitsfördernden Wirkung sind die polyphenolischen Isoflavone für die menschliche Ernährung von großem Interesse. Eine Vielzahl an experimentellen und epidemiologischen Studien zeigen für die in Soja enthaltenen Isoflavone Daidzein und Genistein eine präventive Wirkung bezüglich hormon-abhängiger und altersbedingter Erkrankungen, wie Brust- und Prostatakrebs, Osteoporose, Herz-Kreislauf-Erkrankungen sowie des menopausalen Syndroms. Die Metabolisierung und Bioaktivierung dieser sekundären Pflanzenstoffe durch die humane intestinale Darmmikrobiota ist individuell unterschiedlich. Nur in einem geringen Teil der westlichen Bevölkerung wird der Daidzein-Metabolit Equol durch spezifische Darmbakterien gebildet. Ein isoliertes Equol-produzierendes Bakterium des menschlichen Darmtrakts ist Slackia isoflavoniconvertens. Anhand dieser Spezies sollten die bislang unbekannten, an der Umsetzung von Daidzein und Genistein beteiligten Enzyme identifiziert und charakterisiert werden.
Fermentationsexperimente mit S. isoflavoniconvertens zeigten, dass die Gene der Daidzein und Genistein-umsetzenden Enzyme nicht konstitutiv exprimiert werden, sondern induziert werden müssen. Mit Hilfe der zweidimensionalen differentiellen Gelelektrophorese wurden sechs Proteine detektiert, welche in einer S. isoflavoniconvertens-Kultur in Anwesenheit von Daidzein induziert wurden. Auf Grundlage einzelner Peptidsequenzen erfolgte die Sequenzierung eines Genkomplexes mit den in gleicher Orientierung angeordneten Genen der durch Daidzein induzierten Proteine. Sequenzvergleiche identifizierten zudem äquivalente Genprodukte zu den Proteinen von S. isoflavoniconvertens in anderen Equolproduzierenden Bakterien. Nach der heterologen Expression in Escherichia coli wurden drei dieser Gene durch enzymatische Aktivitätstests als Daidzein-Reduktase (DZNR), Dihydrodaidzein-Reduktase (DHDR) und Tetrahydrodaidzein-Reduktase (THDR) identifiziert. Die Kombination der E. coli-Zellextrakte führte zur vollständigen Umsetzung von Daidzein über Dihydrodaidzein zu Equol. Neben Daidzein setzte die DZNR auch Genistein zu Dihydrogenistein um. Dies erfolgte mit einer größeren Umsatzgeschwindigkeit im Vergleich zur Reduktion von Daidzein zu Dihydrodaidzein. Enzymatische Aktivitätstests mit dem Zellextrakt von S. isoflavoniconvertens zeigten ebenfalls eine schnellere Umsetzung von Genistein. Die Kombination der rekombinanten DHDR und THDR führte zur Umsetzung von Dihydrodaidzein zu Equol. Der korrespondierende Metabolit 5-Hydroxyequol konnte als Endprodukt des Genistein-Metabolismus nicht detektiert werden. Zur Reinigung der drei identifizierten Reduktasen wurden diese genetisch an ein Strep-tag fusioniert und mittels Affinitätschromatographie gereinigt. Die übrigen durch Daidzein induzierten Proteine IfcA, IfcBC und IfcE wurden ebenfalls in E. coli exprimiert und als Strep-Fusionsproteine gereinigt. Vergleichende Aktivitätstests identifizierten das induzierte Protein IfcA als Dihydrodaidzein-Racemase. Diese katalysierte die Umsetzung des (R)- und (S)-Enantiomers von Dihydrodaidzein und Dihydrogenistein zum korrespondierenden Racemat. Neben dem Elektronentransfer-Flavoprotein IfcBC wurden auch die THDR, DZNR und IfcE als FAD-haltige Flavoproteine identifiziert. Zudem handelte es sich bei IfcE um ein Eisen-Schwefel-Protein. Nach Induktion der für die Daidzein-Umsetzung kodierenden Gene wurden mehrere verschieden lange mRNA-Transkripte gebildet. Dies zeigte, dass die Transkription des durch Daidzein induzierten Genkomplexes in S. isoflavoniconvertens nicht in Form eines einzelnen Operonsystems erfolgte.
Auf Grundlage der identifizierten Daidzein-umsetzenden Enzyme kann der Mechanismus der bakteriellen Umsetzung von Isoflavonen durch S. isoflavoniconvertens eingehend erforscht werden. Die ermittelten Gensequenzen der durch Daidzein induzierten Proteine sowie die korrespondierenden Gene weiterer Equol-produzierender Bakterien bieten zudem die Möglichkeit der mikrobiellen Metagenomanalyse im humanen Darmtrakt.
MULTILIT

This paper presents an overview of the linguistic analyses developed in the MULTILIT project and the processing of the oral and written texts collected. The project investigates the language abilities of multilingual children and adolescents, in particular, those who have Turkish and/or Kurdish as a mother tongue. A further aim of the project is to examine from a psycholinguistic and sociolinguistic perspective the extent to which competence in academic registers is achieved on the basis of the languages spoken by the children, including the language(s) spoken at the home, the language of the country of residence and the first foreign language. To be able to examine these questions using corpus linguistic parameters, we created categories of analysis in MULTILIT.
The data collection comprises texts from bilingual and monolingual children and adolescents in Germany in their first language Turkish, their second language German und their foreign language English. Pupils aged between nine and twenty years of age produced monologue oral and written texts in the two genres of narrative and discursive. On the basis of these samples, we examine linguistic features such as lexical expression (lexical density, lexical diversity), syntactic complexity (syntactic and discursive packaging) as well as phonology in the oral texts and orthography in the written texts, with the aim of investigating the pupils’ growing mastery of these features in academic and informal registers.
To this end the raw data have been transcribed by the use of transcription conventions developed especially for the needs of the MULTILIT data. They are based on the commonly used HIAT and GAT transcription conventions and supplemented with conventions that provide additional information such as features at the graphic level.
The categories of analysis comprise a large number of linguistic categories such as word classes, syntax, noun phrase complexity, complex verbal morphology, direct speech and text structures. We also annotate errors and norm deviations at a wide range of levels (orthographic, morphological, lexical, syntactic and textual). In view of the different language systems, these criteria are considered separately for all languages investigated in the project.
Die Arbeit geht der Frage nach, wie Innovationen in einer Organisation des öffentlichen Sektors aufgenommen wurden und zu welchen Veränderungen dies führte. Im Vordergrund steht hier nicht die Innovation selbst, sondern vielmehr die Anpassungsmechanismen in der Organisation. Folgende Forschungsfragen wurden dazu gewählt:
1. Wie wurde das Instrument Zielsteuerung bzw. Zielvereinbarung im öffentlichen Sektor eingeführt und in die Managementroutinen integriert?
2. Welche Faktoren führen zu einer Integration der Zielsteuerung in die Managementroutinen?
3. Welche Empfehlungen für die Praxis lassen sich daraus ableiten?
Dazu wurde ein Landesbetrieb in Brandenburg detailliert untersucht und 31 Interviews mit Führungskräften der zweiten und dritten Managementebene geführt. In dieser Organisation wurde im Rahmen der deutschlandweiten Reformbewegung in der öffentlichen Verwaltung das Instrument Zielsteuerung bzw. Zielvereinbarung eingeführt und mit ganz konkreten Erwartungen verbunden. Als Untersuchungseinheit der möglichen Anpassungen und Veränderungen wurde das Konstrukt der Managementroutinen herangezogen, welche als kollektive Handlungsmuster ganz bewusst individuelle Verhaltensweisen ausklammerten.
Die Arbeit konnte eine Reihe von früheren Erkenntnissen bestätigen und zudem nachweisen, dass, entgegen des häufigen Vorurteils, Innovationen aus dem privatwirtschaftlichem Raum doch auch zu positiven Veränderungen in Organisationen der öffentlichen Hand führen können. Es kam hier jedoch nicht zur Entwicklung neuer, sondern zu einer Anpassung der bestehenden Routinen. Auf dieser Basis konnte festgestellt werden, dass ein stufenweiser Einführungsvorgang zunächst auf der Ebene der veränderten Zielvorstellungen der Führungskräfte zum Erfolg führte. Erst nach der Anpassung auf dieser „ostentativen“ Ebene kam es mit etwas Verzögerung zu einer Veränderung auf der Ebene der konkreten Handlungen. Im Hinblick auf die Einflussfaktoren der Innovation konnte festgestellt werden, dass viele Aspekte der Zielsetzungstheorie nach wie vor relevant sind und instabile politische Rahmenbedingungen zu wesentlichen Einschränkungen der Entfaltungsmöglichkeiten der Innovation führen können. Für viele Einflussfaktoren konnten allerdings sowohl positive als auch negative Wirkungen identifiziert werden.
Die vorliegende Arbeit beschäftigt sich mit der Fragestellung, inwieweit die Integrität der phonologischen Sprachverarbeitung für den erfolgreichen Schriftspracherwerb bei deutschsprachigen Kindern relevant ist. Hierbei bilden Fähigkeiten zur phonologischen Bewusstheit (PhB) den Schwerpunkt. Der erfolgreiche Schriftspracherwerb ist nicht nur für den Bildungserfolg mit den damit verbundenen beruflichen und sozio‑ökonomischen Perspektiven wichtig, sondern auch für die aktive Teilhabe am sozialen und kulturellen Leben in unserer Gesellschaft.
Die Bestandteile dieser publikationsbasierten Dissertation sind eine Monographie (Schnitzler, 2008), ein Beitrag in einem Sammelwerk (Schnitzler, 2013) sowie zwei Zeitschriftenartikel (Schnitzler, 2014, 2015). Die ersten beiden Publikationen beschäftigen sich mit der Entwicklung der PhB sowie Zusammenhängen zwischen PhB und Schriftsprachfertigkeiten. Die beiden Zeitschriftenartikel beschäftigen sich mit dem LRS‑Risiko deutschsprachiger Kinder, die im Vorschulalter aufgrund phonologischer Aussprachestörungen (PhAS) logopädisch behandelt werden. Hierzu wurden in Schnitzler (2015) die Ergebnisse einer selbst durchgeführten Studie dargestellt. In dieser Studie wurden mögliche Einflüsse zusätzlicher nicht‑phonologischer Symptome und der Art der phonologischen Aussprachestörung kontrolliert.
Die Ergebnisse weisen darauf hin, dass zum Schulbeginn und während der Schuleingangsphase genau beobachtet werden sollte, ob Kinder über altersentsprechende Fähigkeiten zur PhB verfügen und ob sie diese segmental‑phonologischen Wissensbestände bewusst aktivieren und beim Lesen und Schreiben effizient nutzen. Dies gilt insbesondere für Kinder, für die ein erhöhtes LRS‑Risiko besteht. Verfügen Kinder zu dieser Zeit über unzureichend spezifizierte phonologische Repräsentationen, ist eine frühzeitige Intervention im Sinne einer Prävention von LRS angezeigt.
Parts without a whole?

This explorative study gives a descriptive overview of what organizations do and experience when they say they practice design thinking. It looks at how the concept has been appropriated in organizations and also describes patterns of design thinking adoption. The authors use a mixed-method research design fed by two sources: questionnaire data and semi-structured personal expert interviews. The study proceeds in six parts: (1) design thinking¹s entry points into organizations; (2) understandings of the descriptor; (3) its fields of application and organizational localization; (4) its perceived impact; (5) reasons for its discontinuation or failure; and (6) attempts to measure its success. In conclusion the report challenges managers to be more conscious of their current design thinking practice. The authors suggest a co-evolution of the concept¹s introduction with innovation capability building and the respective changes in leadership approaches. It is argued that this might help in unfolding design thinking¹s hidden potentials as well as preventing unintended side-effects such as discontented teams or the dwindling authority of managers.
For the calculation of the work in an irreversible pressure-volume change, we propose approxima-tions, which in contrast to the usual representation in the literature reflect the work performed during expansion and compression symmetrically. The calculations are based on the Reversible-Share-Theorem: Is used the force to overcome for calculating the work, so it captures only the configurational reversible work share.
Die Interaktionen von komplexen Kohlenhydraten und Proteinen sind ubiquitär. Sie spielen wichtige Rollen in vielen physiologischen Prozessen wie Zelladhäsion, Signaltransduktion sowie bei viralen Infektionen. Die molekularen Grundlagen der Interaktion sind noch nicht komplett verstanden. Ein Modellsystem für Kohlenhydrat-Protein-Interaktionen besteht aus Adhäsionsproteinen (Tailspikes) von Bakteriophagen, die komplexe Kohlenhydrate auf bakteriellen Oberflächen (O-Antigen) erkennen. Das Tailspike-Protein (TSP), das in dieser Arbeit betrachtet wurde, stammt aus dem Bakteriophagen 9NA (9NATSP). 9NATSP weist eine hohe strukturelle Homologie zum gut charakterisierten TSP des Phagen P22 (P22TSP) auf, bei einer niedriger sequenzieller Ähnlichkeit. Die Substratspezifitäten beider Tailspikes sind ähnlich mit Ausnahme der Toleranz gegenüber den glucosylierten Formen des O-Antigens. Die Struktur der beiden Tailspikes ist bekannt, sodass sie ein geeignetes System für vergleichende Bindungsstudien darstellen, um die strukturellen Grundlagen für die Unterschiede der Spezifität zu untersuchen.
Im Rahmen dieser Arbeit wurde der ELISA-like tailspike adsorption assay (ELITA) etabliert, um Binderpaare aus TSPs und O-Antigen zu identifizieren. Dabei wurden 9NATSP und P22TSP als Sonden eingesetzt, deren Bindung an die intakten, an die Mikrotiterplatte adsorbierten Bakterien getestet wurde. Beim Test einer Sammlung aus 44 Salmonella-Stämmen wurden Stämme identifiziert, die bindendes O-Antigen exprimieren. Gleichzeitig wurden Unterschiede in der Bindung der beiden TSPs an Salmonella-Stämme mit gleichem O-Serotyp beobachtet. Die Ergebnisse der ELITA-Messung wurden qualitativ durch eine FACS-basierte Bindungsmessung bestätigt. Zusätzlich ermöglichte die FACS-Messung bei Stämmen, die teilweise modifizierte O-Antigene herstellen, den Anteil an Zellen mit und ohne Modifikation zu erfassen.
Die Oberflächenplasmonresonanz (SPR)-basierten Interaktionsmessungen wurden eingesetzt, um Bindungsaffinitäten für eine TSP-O-Antigen Kombination zu quantifizieren. Dafür wurden zwei Methoden getestet, um die Oligosaccharide auf einem SPR-Chip zu immobilisieren. Zum einen wurden die enzymatisch hergestellten O-Antigenfragmente mit einem bifunktionalen Oxaminadapter derivatisiert, der eine primäre Aminogruppe für die Immobilisierung bereitstellt. Ein Versuch, diese Oligosaccharidfragmente zu immobilisieren, war jedoch nicht erfolgreich. Dagegen wurde das nicht derivatisierte Polysaccharid, bestehend aus repetitivem O-Antigen und einem konservierten Kernsaccharid, erfolgreich auf einem SPR-Chip immobilisiert. Die Immobilisierung wurde durch Interaktionsmessungen mit P22TSP bestätigt. Durch die Immobilisierung des Polysaccharids sind somit quantitative SPR-Bindungsmessungen mit einem polydispersen Interaktionspartner möglich.
Eine Auswahl von Salmonella-Stämmen mit einer ausgeprägt unterschiedlichen Bindung von 9NATSP und P22TSP im ELITA-Testsystem wurde hinsichtlich der Zusammensetzung des O-Antigens mittels HPLC, Kapillargelelektrophorese und MALDI-MS analysiert. Dabei wurden nicht-stöchiometrische Modifikationen der O-Antigene wie Acetylierung und Glucosylierung detektiert. Das Ausmaß der Glucosylierung korrelierte negativ mit der Effizienz der Bindung und des Verdaus durch die beiden TSPs, wobei der negative Effekt bei 9NATSP weniger stark ausgeprägt war als bei P22TSP. Dies stimmt mit den Literaturdaten zu Infektivitätsstudien mit 9NA und P22 überein, die mit Stämmen mit vergleichbaren O-Antigenvarianten durchgeführt wurden. Die Korrelation zwischen der Glucosylierung und Bindungseffizienz konnte strukturell interpretiert werden.
Auf Grundlage der O-Antigenanalysen sowie der Ergebnisse der ELITA- und FACS-Bindungstests wurden die Salmonella-Stämme Brancaster und Kalamu identifiziert, die annähernd quantitativ glucosyliertes O-Antigen exprimieren. Damit eignen sich diese Stämme für weiterführende Studien, um die Zusammenhänge zwischen der Spezifität und der Organisation der Bindestellen der beiden TSPs zu untersuchen.
Die Wahl des richtigen Studienfaches und die daran anschließende
Studieneingangsphase sind oft entscheidend für den erfolgreichen Verlauf eines Studiums. Eine große Herausforderung besteht dabei darin, bereits in den ersten Wochen des Studiums bestehende Defizite in vermeintlich einfachen Schlüsselkompetenzen zu erkennen und diese so bald wie möglich zu beheben. Eine zweite, nicht minder wichtige Herausforderung ist es, möglichst frühzeitig für jeden einzelnen Studierenden zu erkennen, ob er bzw. sie das individuell richtige Studienfach gewählt hat, das den jeweiligen persönlichen Neigungen, Interessen und Fähigkeiten entspricht und zur Verwirklichung der eigenen Lebensziele beiträgt. Denn nur dann sind Studierende ausreichend stark und dauerhaft intrinsisch motiviert, um ein anspruchsvolles, komplexes Studium erfolgreich durchzuziehen. In diesem Beitrag fokussieren wir eine Maßnahme, die die Studierenden an einen Prozess zur systematischen Reflexion des eigenen Lernprozesses und der eigenen Ziele heranführt und beides in Relation setzt.
Assumed comparable environmental conditions of early Mars and early Earth in 3.7 Ga ago – at a time when first fossil records of life on Earth could be found – suggest the possibility of life emerging on both planets in parallel. As conditions changed, the hypothetical life on Mars either became extinct or was able to adapt and might still exist in biological niches. The controversial discussed detection of methane on Mars led to the assumption, that it must have a recent origin – either abiotic through active volcanism or chemical processes, or through biogenic production. Spatial and seasonal variations in the detected methane concentrations and correlations between the presence of water vapor and geological features such as subsurface hydrogen, which are occurring together with locally increased detected concentrations of methane, gave fuel to the hypothesis of a possible biological source of the methane on Mars.
Therefore the phylogenetically old methanogenic archaea, which have evolved under early Earth conditions, are often used as model-organisms in astrobiological studies to investigate the potential of life to exist in possible extraterrestrial habitats on our neighboring planet. In this thesis methanogenic archaea originating from two extreme environments on Earth were investigated to test their ability to be active under simulated Mars analog conditions. These extreme environments – the Siberian permafrost-affected soil and the chemoautotrophically based terrestrial ecosystem of Movile cave, Romania – are regarded as analogs for possible Martian (subsurface) habitats. Two novel species of methanogenic archaea isolated from these environments were described within the frame of this thesis.
It could be shown that concentrations up to 1 wt% of Mars regolith analogs added to the growth media had a positive influence on the methane production rates of the tested methanogenic archaea, whereas higher concentrations resulted in decreasing rates. Nevertheless it was possible for the organisms to metabolize when incubated on water-saturated soil matrixes made of Mars regolith analogs without any additional nutrients. Long-term desiccation resistance of more than 400 days was proven with reincubation and indirect counting of viable cells through a combined treatment with propidium monoazide (to inactivate DNA of destroyed cells) and quantitative PCR. Phyllosilicate rich regolith analogs seem to be the best soil mixtures for the tested methanogenic archaea to be active under Mars analog conditions. Furthermore, in a simulation chamber experiment the activity of the permafrost methanogen strain Methanosarcina soligelidi SMA-21 under Mars subsurface analog conditions could be proven. Through real-time wavelength modulation spectroscopy measurements the increase in the methane concentration at temperatures down to -5 °C could be detected.
The results presented in this thesis contribute to the understanding of the activity potential of methanogenic archaea under Mars analog conditions and therefore provide insights to the possible habitability of present-day Mars (near) subsurface environments. Thus, it contributes also to the data interpretation of future life detection missions on that planet. For example the ExoMars mission of the European Space Agency (ESA) and Roscosmos which is planned to be launched in 2018 and is aiming to drill in the Martian subsurface.
Direct assessment of attitudes toward socially sensitive topics can be affected by deception attempts. Reaction-time based indirect measures, such as the Implicit Association Test (IAT), are less susceptible to such biases. Neuroscientific evidence shows that deception can evoke characteristic ERP differences. However, the cerebral processes involved in faking an IAT are still unknown. We randomly assigned 20 university students (15 females, 24.65 +/- 3.50 years of age) to a counterbalanced repeated-measurements design, requesting them to complete a Brief-IAT (BIAT) on attitudes toward doping without deception instruction, and with the instruction to fake positive and negative doping attitudes. Cerebral activity during BIAT completion was assessed using high-density EEG. Event-related potentials during faking revealed enhanced frontal and reduced occipital negativity, starting around 150 ms after stimulus presentation. Further, a decrease in the P300 and LPP components was observed. Source analyses showed enhanced activity in the right inferior frontal gyrus between 150 and 200 ms during faking, thought to reflect the suppression of automatic responses. Further, more activity was found for faking in the bilateral middle occipital gyri and the bilateral temporoparietal junction. Results indicate that faking reaction-time based tests alter brain processes from early stages of processing and reveal the cortical sources of the effects. Analyzing the EEG helps to uncover response patterns in indirect attitude tests and broadens our understanding of the neural processes involved in such faking. This knowledge might be useful for uncovering faking in socially sensitive contexts, where attitudes are likely to be concealed.

Untersuchungen zur räumlichen Analyse und Visualisierung von Mietpreisdaten für Immobilienportale

Die vorliegende Arbeit verfolgt das Ziel, aus geoinformatischer Sicht eine konzeptionelle Grundlage zur räumlichen Optimierung von Immobilienportalen zu schaffen. Die Arbeit geht dabei von zwei Hypothesen aus:
1. Verfahren der räumlichen Statistik und des Maschinellen Lernens zur Mietpreisschätzung sind den bisher eingesetzten Verfahren der hedonischen Regression überlegen und eignen sich zur räumlichen Optimierung von Immobilienportalen.
2. Die von Immobilienportalen publizierten webbasierten Mietpreiskarten geben nicht die tatsächlichen räumlichen Verhältnisse auf Immobilienmärkten wieder. Alternative webbasierte Darstellungsformen, wie z.B. Gridmaps, sind dem Status Quo der Immobilienpreiskarten von Immobilienportalen überlegen und visualisieren die tatsächlichen räumlichen Verhältnisse von Immobilienpreisen zweckmäßiger.
Beide Thesen können bewiesen werden.
Es erfolgt zunächst eine umfangreiche Erhebung des Forschungsbedarfs mittels Literaturstudien und technologischer Recherche. Zur Beantwortung der Forschungsfragen wird als quantitative Datenbasis ein 74.098 Mietangebote umfassender Datensatz (von Januar 2007 bis September 2013) eines Immobilienportals akquiriert. Dieser reicht jedoch nicht in vollem Umfang zur Beantwortung der Fragestellungen aus. Deshalb führt der Autor Experteninterviews zur Erhebung einer qualitativen Datenbasis. Deren Analyse ergibt in Kombination mit der Literaturstudie und der technologischen Recherche ein umfassendes, bisher so nicht verfügbares Bild. Es stellt den Status Quo der räumlichen Sicht sowie der raumanalytischen und geovisuellen Defizite von Immobilienportalen dar.
Zur Optimierung der raumanalytischen und geovisuellen Defizite werden forschungsbasierte Lösungsansätze herausgearbeitet und teilimplementiert. Methoden des Maschinellen Lernens und räumliche Schätzverfahren werden als Alternativen zu den von Immobilienportalen bisher genutzten „nicht räumlichen“ Analyseverfahren zur Preismodellierung untersucht. Auf Grundlage eines hierfür konzipierten Validierungsrahmens werden diese Methoden für die Nutzung im Kontext von Immobilienportalen adaptiert. Die prototypische Teilimplementierung zeigt die programmiertechnische Umsetzung des Konzeptes auf. Eine umfassende Analyse geeigneter Sekundärvariablensets zur Mietpreisschätzung liefert als methodisches Resultat, dass Interpolatoren, die Sekundärvariablen benötigen (Kriging with external drift, Ordinary Cokriging), kaum zu valideren Mietpreisschätzergebnissen gelangen als die Methode des Ordinary Kriging, die keine Sekundärvariablen benötigt. Die Methoden Random Forest aus dem Maschinellen Lernen und die Geographisch Gewichtete Regression hingegen bergen großes Potential zur Nutzung der räumlichen Mietpreisschätzung im Kontext von Immobilien-portalen. Die Forschungsergebnisse der räumlichen Preismodellierung werden in die räumliche Visualisierung von Mietpreisen transferiert.
Für die webbasierte Mietpreisdarstellung wird ein Set alternativer Darstellungsmethoden entwickelt, um Mietpreiskarten-Prototypen abzuleiten. Ein methodisches Ergebnis der Entwicklung der Mietpreiskarten-Prototypen ist die Entwicklung eines geeigneten Ansatzes der Loslösung des Preisbezugs von fachfremd verwendeten Bezugsgeometrien. Hierfür wird vom Autor der Begriff der zonenlosen Preiskarte geprägt. Diese werden mit Methoden des Gridmapping erstellt. Es werden optimale Rasterauflösungen zur Darstellung interpolierter Rastergrößen ermittelt. Zonenlose Preiskarten mit Methoden des Gridmapping, gepaart mit einer optionalen gebäudescharfen Darstellung in größeren Maßstäben, sind als Resultate der Forschung die bestmögliche, sich an realen Verhältnissen orientierende, räumliche Mietpreisdarstellung. Die entstandenen Prototypen sind eine Annäherung der wahren Verteilung des Mietpreises im Raum und um einiges schärfer, als die auf der hedonischen Regression basierenden Darstellungen. Somit kann die wahre „Topographie“ der Mietpreislandschaft abgebildet werden. Ein Einsatz der Karten für Nutzergruppen wie Makler, Investoren oder Kommunen zur Analyse städtischer Mietmärkte ist denkbar. Alle entstandenen Prototypen sind unter der Nutzung von Map APIs umgesetzt. Ein Ergebnis dessen ist, dass Map APIs noch an diversen „Kinderkrankheiten“ leiden und derart umgesetzte Mietpreiskarten noch einen weiten Weg vor sich haben, bis sie das Niveau thematischer Karten von Immhof oder Arnberger erreichen.
Die konzeptionellen Überlegungen und Teilimplementierungen münden in drei Prozessketten, die Umsetzungsoptionen für eine räumliche Optimierung von Immobilienportalen darstellen. Dabei werden zwei Szenarien für eine räumlich optimierte Mietpreisschätzung und ein Szenario für eine räumlich optimierte Mietpreisdarstellung herausgearbeitet.
Die vorliegende Publikation umfasst einen Teil der Dissertation „Wortstellungsvariation aus informationsstruktureller Perspektive. Eine Untersuchung der linken Satzperipherie im gesprochenen Deutsch“ von Sören Schalowski. In diesen Kapiteln wird aus synchroner Perspektive auf die Wortstellungsvariation in der linken Satzperipherie, also die Besetzungs- und Linearisierungsvarianten der syntaktischen Domäne vor dem finiten Verb, genauer eingegangen. Dabei werden die für das Standarddeutsche bekannten und in der Literatur diskutierten (oberflächlichen)
Abweichungen von der V2-Stellung in Aussagesätzen des Deutschen genauer diskutiert.

This article investigates collective team identification and team member alignment (i.e., the existence of short- and long-term team goals and team-based reward structures) as moderators of the association between task and relationship conflicts. Being indicators of cooperative goal interdependence in teams, both moderators are hypothesized to mitigate the positive association between the two conflict types. Findings from 88 development teams confirm the moderating effect for collective team identification, but not for team member alignment. Moreover, the moderating role of collective team identification is found to be dependent on the level of task conflict: It is more effective in decoupling task and relationship conflicts at medium as compared with high or low levels of task conflict.
The Norway lobster, Nephrops norvegicus, is a burrowing decapod with a rhythmic burrow emergence (24 h) governed by the circadian system. It is an important resource for European fisheries and its behavior deeply affects its availability. The current knowledge of Nephrops circadian biology is phenomenological as it is currently the case for almost all crustaceans. In attempt to elucidate the putative molecular mechanisms underlying circadian gene regulation in Nephrops, we used a transcriptomics approach on cDNA extracted from the eyestalk, a structure playing a crucial role in controlling behavior of decapods. We studied 14 male lobsters under 12–12 light-darkness blue light cycle. We used the Hiseq 2000 Illumina platform to sequence two eyestalk libraries (under light and darkness conditions) obtaining about 90 millions 100-bp paired-end reads. Trinity was used for the de novo reconstruction of transcriptomes; the size at which half of all assembled bases reside in contigs (N50) was equal to 1796 (light) and 2055 (darkness). We found a list of candidate clock genes and focused our attention on canonical ones: timeless, period, clock and bmal1. The cloning of assembled fragments validated Trinity outputs. The putative Nephrops clock genes showed high levels of identity (blastx on NCBI) with known crustacean clock gene homologs such as Eurydice pulchra (period: 47%, timeless: 59%, bmal1: 79%) and Macrobrachium rosenbergii (clock: 100%). We also found a vertebrate-like cryptochrome 2. RT-qPCR showed that only timeless had a robust diel pattern of expression. Our data are in accordance with the current knowledge of the crustacean circadian clock, reinforcing the idea that the molecular clockwork of this group shows some differences with the established model in Drosophila melanogaster.
The subject of the present thesis is the one-dimensional Bose gas. Since long-rang order is destroyed by infra-red fluctuations in one dimension, only the formation of a quasi-condensate is possible, which exhibits suppressed density fluctuations, but whose phase fluctuates strongly. It is shown that modified mean-field theories based on a symmetry-breaking approach can even characterise phase coherence properties of such a quasi-condensate properly. A correct description of the transition from the degenerate ideal Bose gas to the quasi-condensate, which is a smooth cross-over rather than a phase transition, is not possible though. Basic conditions for the applicability of the theories are not fulfilled in this regime, such that the existence of a critical point is predicted.
The theories are compared on the basis of their excitation sprectum, equation of state, density fluctuations and related correlation functions. High-temperature expansions of the corresponding integrals are derived analytically for the numerical evaluation of the self-consistent integral equations. Apart from that, the Stochastic Gross-Pitaevskii equation (SGPE), a non-linear Langevin equation, is analysed numerically by means of Monte-Carlo simulations and the results are compared to those of the mean-field theories. In this context, a lot of attention is payed to the appropriate choice of the parameters. The simulations prove that the SGPE is capable of describing the cross-over properly, but highlight the limitations of the widely used local density approximation as well.
The relationship between nutrition and the development of chronic diseases including metabolic syndrome, diabetes mellitus, cancer and cardiovascular disease has been well studied. On the other hand, changes in the GH-IGF-1 axis in association with nutrition-related diseases have been reported. The interplay between GH, total IGF-1 and different inhibitory and stimulatory kinds of IGF-1 binding proteins (IGFBPs) results in IGF-1 bioactivity, the ability of IGF-1 to induce phosphorylation of its receptor and consequently its signaling. Moreover, IGF-1 bioactivity is sufficient to reflect any change in the GH-IGF-1 system. Accumulating evidence suggests that both of high protein diet, characterized by increased glucagon secretion, and insulin-induced hypoglycemia increase mortality rate and the mechanisms are unclear. However both of glucagon and insulin-induced hypoglycemia are potent stimuli of GH secretion. The aim of the current study was to identify the impact of glucagon and insulin-induced hypoglycemia on IGF-1 bioactivity as possible mechanisms. In a double-blind placebo-controlled study, glucagon was intramuscularly administrated in 13 type 1 diabetic patients (6 males /7 females; [BMI]: 24.8 ± 0.95 kg/m2), 11 obese subjects (OP; 5/ 6; 34.4 ± 1.7 kg/m2), and 13 healthy lean participants (LP; 6/ 7; 21.7 ± 0.6 kg/m2), whereas 12 obese subjects (OP; 6/ 6; 34.4 ± 1.7 kg/m2), and 13 healthy lean participants (LP; 6/ 7; 21.7 ± 0.6 kg/m2) performed insulin tolerance test in another double-blind placebo-controlled study and changes in GH, total IGF-1, IGF binding proteins (IGFBPs) and IGF-1 bioactivity, measured by the cell-based KIRA method, were investigated. In addition, the interaction between the metabolic hormones (glucagon and insulin) and the GH-IGF-1 system on the transcriptional level was studied using mouse primary hepatocytes. In this thesis, glucagon decreased IGF-1 bioactivity in humans independently of endogenous insulin levels, most likely through modulation of IGFBP-1 and-2 levels. The glucagon-induced reduction in IGF-1 bioactivity may represent a novel mechanism underlying the impact of glucagon on GH secretion and may explain the negative effect of high protein diet related to increased cardiovascular risk and mortality rate. In addition, insulin-induced hypoglycemia was correlated with a decrease in IGF-1 bioactivity through up-regulation of IGFBP-2. These results may refer to a possible and poorly explored mechanism explaining the strong association between hypoglycemia and increased cardiovascular mortality among diabetic patients.
Massive, luminous stars reaching the Eddington limit in their interiors develop very dilute, extended envelopes. This effect is called envelope inflation. If the progenitors of Type Ib/c supernovae, which are believed to be Wolf-Rayet (WR) stars, have inflated envelopes then the shock breakout signals diffuse in them and can extend their rise times significantly. We show that our inflated, hydrogen-free, WR stellar models with a radius of ∼R⊙ can have shock breakout signals longer than ∼ 60 s. The puzzlingly long shock breakout signal observed in the Type Ib SN 2008D can be explained by an inflated progenitor envelope, and more such events might argue in favour of existence of inflated envelopes in general.
In the last decades, stellar atmosphere codes have become a key tool in understanding massive stars, including precise calculations of stellar and wind parameters, such as temperature, massloss rate, and terminal wind velocity. Nevertheless, for these models the hydrodynamic equation is not solved in the wind. Motivated by the results of the CAK theory, the models typically use a beta velocity law, which however turns out not to be adequate for stars with very strong winds, and treat the mass-loss rate as a free parameter. In a new branch of the Potsdam Wolf-Rayet model atmosphere (PoWR) code, we solve the hydrodynamic equation consistently throughout the stellar atmosphere. The PoWR code performs the calculation of the radiative force without approximations (e.g. Sobolev). We show the impact of hydrodynamically consistent modelling on OB and WR stars in comparison to conventional models and discuss the obtained velocity fields and their impact on the observed spectral lines.
This article shows a discussion about the key competencies
in informatics and ICT viewed from a philosophical foundation presented
by Martha Nussbaum, which is known as ‘ten central capabilities’.
Firstly, the outline of ‘The Capability Approach’, which has been presented
by Amartya Sen and Nussbaum as a theoretical framework of
assessing the state of social welfare, will be explained. Secondly, the
body of Nussbaum’s ten central capabilities and the reason for being
applied as the basis of discussion will be shown. Thirdly, the relationship
between the concept of ‘capability’ and ‘competency’ is to be
discussed. After that, the author’s assumption of the key competencies
in informatics and ICT led from the examination of Nussbaum’s ten
capabilities will be presented.
Colliding Wolf-Rayet (WR) winds produce thermal X-ray emission widely observed by X-ray telescopes. In wide WR+O binaries, such as WR 140, the X-ray flux is tied to the orbital phase, and is a direct probe of the winds’ properties. In the Galactic center, ~30 WRs orbit the super massive black hole (SMBH) within ~10”, leading to a smorgasbord of wind-wind collisions. To model the X-ray emission of WR 140 and the Galactic center, we perform 3D hydrodynamic simulations to trace the complex gaseous flows, and then carry out 3D radiative transfer calculations to compute the variable X-ray spectra. The model WR 140 RXTE light curve matches the data well for all phases except the X-ray minimum associated with periastron, while the model spectra agree with the RXTE hardness ratio and the shape of the Suzaku observations throughout the orbit. The Galactic center model of the Chandra flux and spectral shape match well in the region r ≤ 3”, but the model flux falls off too rapidly beyond this radius.

Die Zahl der deutschen Jugendlichen, die an IFD-Programmen teilnehmen, ist in den vergangenen Jahren erheblich gestiegen und die Volunteer-Branche hat sich stark diversifiziert. Was die verschiedenen Formen staatlicher und privatwirtschaftlicher IFD eint, ist die explizite Abgrenzung vom Tourismus. Bei dieser Abgrenzung handelt es sich jedoch um eine Exklusion, die das Ausgeschlossene gerade nicht als irrelevant, sondern für den Kommunikationszusammenhang als überaus bedeutsam markiert.
Ziel dieses Buches ist es, aufzuzeigen wie Tourismus als ANDERES konstruiert und dies zur Selbstidentifikation der Freiwilligen genutzt wird. Unter Zuhilfenahme des Laclau/Mouffe’schen Theoriegebäudes werden die Selbst- und Fremdbilder von deutschsprachigen Teilnehmer(innen) verschiedener IFD-Programme in Ghana hinsichtlich ihrer Deutungsansprüche und Identifikationsmechanismen betrachtet. Der Fokus richtet sich dabei auf Prozesse der Grenzziehung, die das Sprechen der Freiwilligen in Bereiche des EIGNENEN und des ANDEREN unterteilen. Im Sinne einer dem cultural turn verpflichteten Neuen Kulturgeographie wird der Verwendung räumlicher Identitätsanker dabei besondere Aufmerksamkeit gewidmet.
The Westerlund 1 (Wd1) cluster hosts a rich and varied collection of massive stars. Its dynamical youth and the absence of ongoing star formation indicate a coeval population. As such, the simultaneous presence of both late-type supergiants and Wolf-Rayet stars has defied explanation in the context of single-star evolution. Observational evidence points to a high binary fraction, hence this stellar population offers a robust test for stellar models accounting for both single-star and binary evolution. We present an optical to near-IR (VLT & NTT) spectroscopic analysis of 22 WR stars in Wd 1, delivering physical properties for the WR stars.
We discuss how these differ from the Galactic field population, and how they may be reconciled with the predictions of single and binary evolutionary models.
When Jesus Spoke Yiddish

In this paper, I wish to bring some evidences from a Yiddish manuscript of the “Toledot Yeshu” which has not yet been the object of research: MS. Günzburg, 1730 kept in the Russian State Library in Moscow and dated 17th century. The manuscript is part of the so-called ‘Herode-tradition’ of the “Toledot Yeshu”. This means that the Yiddish manuscript is connected to the version printed in Hebrew and accompanied by a Latin translation by the Swiss pastor and theologian Johann Jacob Uldrich (Huldricus, 1683–1731) in Leiden in 1705, bearing the title “Historia Jeschuae Nazareni”. Given the uncertainty about the exact dating of the Yiddish manuscript, a comparison between the Hebrew and the Yiddish can still allow some remarks concerning the characteristics of the Yiddish version and posit some questions about the transmission and the reception of this challenging and intriguing text.
In this contribution we present some preliminary results obtained from a SOAR-Goodman optical spectroscopic survey aimed to confirm the OIf* - OIf*/WN nature of a sample of Galactic candidates that were previously confirmed as massive stars based on near-infrared spectra taken with OSIRIS at SOAR. With only a few of such stars known in the Galaxy to date, our study significantly contributes to improve the number of known Galactic O2If* stars, as well as almost doubling the number of known members of the galactic sample of the rare type OIf*/WN.
The overarching goal of this dissertation is to provide a better understanding of the role of wind and water in shaping Earth’s Cenozoic orogenic plateaus - prominent high-elevation, low relief sectors in the interior of Cenozoic mountain belts. In particular, the feedbacks between surface uplift, the build-up of topography and ensuing changes in precipitation, erosion, and vegetation patterns are addressed in light of past and future climate change. Regionally, the study focuses on the two world’s largest plateaus, the Altiplano-Puna Plateau of the Andes and Tibetan Plateau, both characterized by average elevations of >4 km. Both plateaus feature high, deeply incised flanks with pronounced gradients in rainfall, vegetation, hydrology, and surface processes. These characteristics are rooted in the role of plateaus to act as efficient orographic barriers to rainfall and to force changes in atmospheric flow.
The thesis examines the complex topics of tectonic and climatic forcing of the surface-process regime on three different spatial and temporal scales: (1) bedrock wind-erosion rates are quantified in the arid Qaidam Basin of NW Tibet over millennial timescales using cosmogenic radionuclide dating; (2) present-day stable isotope composition in rainfall is examined across the south-central Andes in three transects between 22° S and 28° S; these data are modeled and assessed with remotely sensed rainfall data of the Tropical Rainfall Measuring Mission and the Moderate Resolution Imaging Spectroradiometer; (3) finally, a 2.5-km-long Mio-Pliocene sedimentary record of the intermontane Angastaco Basin (25°45’ S, 66°00’ W) is presented in the context of hydrogen and carbon compositions of molecular lipid biomarker, and oxygen and carbon isotopes obtained from pedogenic carbonates; these records are compared to other environmental proxies, including hydrated volcanic glass shards from volcanic ashes intercalated in the sedimentary strata.
There are few quantitative estimates of eolian bedrock-removal rates from arid, low relief landscapes. Wind-erosion rates from the western Qaidam Basin based on cosmogenic 10Be measurements document erosion rates between 0.05 to 0.4 mm/yr. This finding indicates that in arid environments with strong winds, hyperaridity, exposure of friable strata, and ongoing rock deformation and uplift, wind erosion can outpace fluvial erosion. Large eroded sediment volumes within the Qaidam Basin and coeval dust deposition on the Chinese Loess plateau, exemplify the importance of dust production within arid plateau environments for marine and terrestrial depositional processes, but also health issues and fertilization of soils.
In the south-central Andes, the analysis of 234 stream-water samples for oxygen and hydrogen reveals that areas experiencing deep convective storms do not show the commonly observed patterns of isotopic fractionation and the expected co-varying relationships between oxygen and hydrogen with increasing elevation. These convective storms are formed over semi-arid intermontane basins in the transition between the broken foreland of the Sierras Pampeanas, the Eastern Cordillera, and the Puna Plateau in the interior of the orogen. Here, convective rainfall dominates the precipitation budget and no systematic stable isotope-elevation relationship exists. Regions to the north, in the transition between the broken foreland and the Subandean foreland fold-and-thrust belt, the impact of convection is subdued, with lower degrees of storminess and a stronger expected isotope-elevation relationship. This finding of present-day fractionation trends of meteoric water is of great importance for paleoenvironmental studies in attempts to use stable isotope relationships in the reconstruction of paleoelevations.
The third part of the thesis focuses on the paleohydrological characteristics of the Mio-Pliocene (10-2 Ma) Angastaco Basin sedimentary record, which reveals far-reaching environmental changes during Andean uplift and orographic barrier formation. A precipitation- evapotranspiration record identifies the onset of a precipitation regime related to the South American Low Level Jet at this latitude after 9 Ma. Humid foreland conditions existed until 7 Ma, followed by orographic barrier uplift to the east of the present-day Angastaco Basin. This was superseded by rapid (~0.5 Myr) aridification in an intermontane basin, highlighting the effects of eastward-directed deformation. A transition in vegetation cover from a humid C3 forest ecosystem to semi-arid C4-dominated vegetation was coeval with continued basin uplift to modern elevations.
An observational measure of anger regulation in middle childhood was developed that facilitated the in situ assessment of five maladaptive regulation strategies in response to an anger-eliciting task. 599 children aged 6-10 years (M = 8.12, SD = 0.92) participated in the study. Construct validity of the measure was examined through correlations with parent- and self-reports of anger regulation and anger reactivity. Criterion validity was established through links with teacher-rated aggression and social rejection measured by parent-, teacher-, and self-reports. The observational measure correlated significantly with parent- and self-reports of anger reactivity, whereas it was unrelated to parent- and self-reports of anger regulation. It also made a unique contribution to predicting aggression and social rejection.
A flexible approach to assess fluorescence decay functions in complex energy transfer systems

Background: Time-correlated Forster resonance energy transfer (FRET) probes molecular distances with greater accuracy than intensity-based calculation of FRET efficiency and provides a powerful tool to study biomolecular structure and dynamics. Moreover, time-correlated photon count measurements bear additional information on the variety of donor surroundings allowing more detailed differentiation between distinct structural geometries which are typically inaccessible to general fitting solutions.
Results: Here we develop a new approach based on Monte Carlo simulations of time-correlated FRET events to estimate the time-correlated single photon counts (TCSPC) histograms in complex systems. This simulation solution assesses the full statistics of time-correlated photon counts and distance distributions of fluorescently labeled biomolecules. The simulations are consistent with the theoretical predictions of the dye behavior in FRET systems with defined dye distances and measurements of randomly distributed dye solutions. We validate the simulation results using a highly heterogeneous aggregation system and explore the conditions to use this tool in complex systems.
Conclusion: This approach is powerful in distinguishing distance distributions in a wide variety of experimental setups, thus providing a versatile tool to accurately distinguish between different structural assemblies in highly complex systems.
Vegetated dunes are recognized as important natural barriers that shelter inland ecosystems and coastlines suffering daily erosive impacts of the sea and extreme events, such as tsunamis. However, societal responses to erosion and shoreline retreat often result in man-made coastal defence structures that cover part of the intertidal and upper shore zones causing coastal squeeze and habitat loss, especially for upper shore biota, such as dune plants. Coseismic uplift of up to 2.0 m on the Peninsula de Arauco (South central Chile, ca. 37.5 degrees S) caused by the 2010 Maule earthquake drastically modified the coastal landscape, including major increases in the width of uplifted beaches and the immediate conversion of mid to low sandy intertidal habitat to supralittoral sandy habitat above the reach of average tides and waves. To investigate the early stage responses in species richness, cover and across-shore distribution of the hitherto absent dune plants, we surveyed two formerly intertidal armoured sites and a nearby intertidal unarmoured site on a sandy beach located on the uplifted coast of Llico (Peninsula de Arauco) over two years. Almost 2 years after the 2010 earthquake, dune plants began to recruit, then rapidly grew and produced dune hummocks in the new upper beach habitats created by uplift at the three sites. Initial vegetation responses were very similar among sites. However, over the course of the study, the emerging vegetated dunes of the armoured sites suffered a slowdown in the development of the spatial distribution process, and remained impoverished in species richness and cover compared to the unarmoured site. Our results suggest that when released from the effects of coastal squeeze, vegetated dunes can recover without restoration actions. However, subsequent human activities and management of newly created beach and dune habitats can significantly alter the trajectory of vegetated dune development. Management that integrates the effects of natural and human induced disturbances, and promotes the development of dune vegetation as natural barriers can provide societal and conservation benefits in coastal ecosystems.
Peer Assessment ist eine Methode, bei der die Teilnehmer eine gestellte Aufgabe nicht nur bearbeiten und einreichen, sondern – in einer zweiten Phase – diese auch gegenseitig überprüfen, kommentieren und bewerten. Durch diese Methode wird, auch in sehr großen Veranstaltungen, das Üben mit individuellen Bewertungen und individuellem Feedback möglich.
Im Wintersemester 2013/14 wurde dieser Ansatz in der Erstsemesterveranstaltung Programmieren an der Technischen Hochschule Nürnberg mit 340 Studierenden als semesterbegleitendes Online-Pflichtpraktikum erprobt. Bei gleichen Leistungsanforderungen wurde bei Studierenden, die erfolgreich am Praktikum teilnahmen, eine Reduzierung der Durchfallquote um durchschnittlich 60 % und eine Verbesserung der Durchschnittsnote um 0,6 – 0,9 Notenstufen erzielt. Zudem lernten die teilnehmenden Studierenden kontinuierlicher, bereiteten Lerninhalte besser nach und gelangten zu einer überwiegend positiven Einschätzung des Praktikums und der Methode. Im E-Learning System Moodle kann Peer Assessment, mit moderatem Umsetzungs- und Betreuungsaufwand, mit der Workshop-Aktivität realisiert werden. Im Beitrag wird auf die Schlüsselelemente des erfolgreichen Einsatzes von Peer Assessment eingegangen.
Fluid force microscopy combines the positional accuracy and force sensitivity of an atomic
force microscope (AFM) with nanofluidics via a microchanneled cantilever. However, adequate loading and cleaning procedures for such AFM micropipettes are required for various application situations. Here, a new frontloading procedure is described for an AFM micropipette functioning as a force- and pressure-controlled microscale liquid dispenser. This frontloading
procedure seems especially attractive when using target substances featuring high
costs or low available amounts. Here, the AFM micropipette could be filled from the tip side with liquid from a previously applied droplet with a volume of only a few μL using a short low-pressure pulse. The liquid-loaded AFM micropipettes could be then applied for experiments in air or liquid environments. AFM micropipette frontloading was evaluated with the well-known organic fluorescent dye rhodamine 6G and the AlexaFluor647-labeled antibody goat anti-rat IgG as an example of a larger biological compound. After micropipette usage, specific cleaning procedures were tested. Furthermore, a storage method is described, at which the AFM micropipettes could be stored for a few hours up to several days without drying out or clogging of the microchannel. In summary, the rapid, versatile and cost-efficient
frontloading and cleaning procedure for the repeated usage of a single AFM micropipette is beneficial for various application situations from specific surface modifications through to local manipulation of living cells, and provides a simplified and faster handling for already known experiments with fluid force microscopy.
We an optically-thick, transonic, steady wind model for a H-free Wolf-Rayet star. A bifurcation is found across a critical mass loss rate Mb. Slower winds M < Mb extend by several hydrostatic stellar radii, reproduce features of envelope in ation from Petrovic et al. (2006) and Gräfener et al. (2012), and are energetically unbound. This work is of particular interest for extended envelopes and winds, radiative hydrodynamic instabilities (eg. wind stagnation, clumping, etc.), and NLTE atmospheric models.
Voltage-gated potassium (K+) channels are present in all living systems. Despite high structural similarities in the transmembrane domains (TMD), this K+ channel type segregates into at least two main functional categories-hyperpolarization-activated, inward-rectifying (Kin) and depolarization-activated, outward-rectifying (Kout) channels. Voltage-gated K+ channels sense the membrane voltage via a voltage-sensing domain that is connected to the conduction pathway of the channel. It has been shown that the voltage-sensing mechanism is the same in Kin and Kout channels, but its performance results in opposite pore conformations. It is not known how the different coupling of voltage-sensor and pore is implemented. Here, we studied sequence and structural data of voltage-gated K+ channels from animals and plants with emphasis on the property of opposite rectification. We identified structural hotspots that alone allow already the distinction between Kin and Kout channels. Among them is a loop between TMD S5 and the pore that is very short in animal Kout, longer in plant and animal Kin and the longest in plant Kout channels. In combination with further structural and phylogenetic analyses this finding suggests that outward-rectification evolved twice and independently in the animal and plant kingdom.
Many studies on bacterial community composition (BCC) do not distinguish between particle associated (PA) and free-living (FL) bacteria or neglect the PA fraction by pre-filtration removing most particles. Although temporal and spatial gradients in environmental variables are known to shape BCC, it remains unclear how and to what extent PA and FL bacterial diversity responds to such environmental changes. To elucidate the BCC of both bacterial fractions related to different environmental settings, we studied surface samples of three Baltic Sea stations (marine, mesohaline, and oligohaline) in two different seasons (summer and fall/winter). Amplicon sequencing of the 16S rRNA gene revealed significant differences in BCC of both bacterial fractions among stations and seasons, with a particularly high number of PA operational taxonomic units (OTUs at genus-level) at the marine station in both seasons. "Shannon and Simpson indices" showed a higher diversity of PA than FL bacteria at the marine station in both seasons and at the oligohaline station in fall/winter. In general, a high fraction of bacterial OTUs was found exclusively in the PA fraction (52% of total OTUs). These findings indicate that PA bacteria significantly contribute to overall bacterial richness and that they differ from FL bacteria. Therefore, to gain a deeper understanding on diversity and dynamics of aquatic bacteria, PA and FL bacteria should be generally studied independently.

A project involving the composition of a number of pieces
of music by public participants revealed levels of engagement with and
mastery of complex music technologies by a number of secondary student
volunteers. This paper reports briefly on some initial findings of
that project and seeks to illuminate an understanding of computational
thinking across the curriculum.
We analyse whether a stellar atmosphere model computed with the code CMFGEN provides an optimal description of the stellar observations of WR 136 and simultaneously reproduces the nebular observations of NGC 6888, such as the ionization degree, which is modelled with the pyCloudy code. All the observational material available (far and near UV and optical spectra) were used to constrain such models. We found that the stellar temperature T∗, at τ = 20, can be in a range between 70 000 and 110 000 K, but when using the nebula as an additional restriction, we found that the stellar models with T∗ ∼ 70 000 K represent the best solution for both, the star and the nebula.
We present 3D numerical simulations of the NGC6888 nebula considering the proper motion and the evolution of the star, from the red supergiant (RSG) to the Wolf-Rayet (WR) phase. Our simulations reproduce the limb-brightened morphology observed in [OIII] and X-ray emission maps. The synthetic maps computed by the numerical simulations show filamentary and clumpy structures produced by instabilities triggered in the interaction between the WR wind and the RSG shell.

The dissertation proposes that the spread of photography and popular cinema in 19th- and 20th-century-India have shaped an aesthetic and affective code integral to the reading and interpretation of Indian English novels, particularly when they address photography and/or cinema film, as in the case of the four corpus texts. In analyzing the nexus between ‘real’ and ‘reel’, the dissertation shows how the texts address the reader as media consumer and virtual image projector. Furthermore, the study discusses the Indian English novel against the backdrop of the cultural and medial transformations of the 20th century to elaborate how these influenced the novel’s aesthetics. Drawing upon reception aesthetics, the author devises the concept of the ‘implied spectator’ to analyze the aesthetic impact of the novels’ images as visual textures.
No God in Sight (2005) by Altaf Tyrewala comprises of a string of 41 interior monologues, loosely connected through their narrators’ random encounters in Mumbai in the year 2000. Although marked by continuous perspective shifts, the text creates a sensation of acute immediacy. Here, the reader is addressed as implied spectator and is sutured into the narrated world like a film spectator ― an effect created through the use of continuity editing as a narrative technique.
Similarly, Ruchir Joshi’s The Last Jet Engine Laugh (2002) coll(oc)ates disparate narrative perspectives and explores photography as an artistic practice, historiographic recorder and epistemological tool. The narrative appears guided by the random viewing of old photographs by the protagonist and primary narrator, the photographer Paresh Bhatt. However, it is the photographic negative and the practice of superimposition that render this string of episodes and different perspectives narratively consequential and cosmologically meaningful. Photography thus marks the perfect symbiosis of autobiography and historiography.
Tabish Khair’s Filming. A Love Story (2007) immerses readers in the cine-aesthetic of 1930s and 40s Bombay film, the era in which the embedded plot is set. Plotline, central scenes and characters evoke the key films of Indian cinema history such as Satyajit Ray’s “Pather Panchali” or Raj Kapoor’s “Awara”. Ultimately, the text written as film dissolves the boundary between fiction and (narrated) reality, reel and real, thereby showing that the images of individual memory are inextricably intertwined with and shaped by collective memory. Ultimately, the reconstruction of the past as and through film(s) conquers trauma and endows the Partition of India as a historic experience of brutal contingency with meaning.
The Bioscope Man (Indrajit Hazra, 2008) is a picaresque narrative set in Calcutta - India’s cultural capital and birthplace of Indian cinema at the beginning of the 20th century. The autodiegetic narrator Abani Chatterjee relates his rise and fall as silent film star, alternating between the modes of tell and show. He is both autodiegetic narrator and spectator or perceiving consciousness, seeing himself in his manifold screen roles. Beyond his film roles however, the narrator remains a void. The marked psychoanalytical symbolism of the text is accentuated by repeated invocations of dark caves and the laterna magica. Here too, ‘reel life’ mirrors and foreshadows real life as Indian and Bengali history again interlace with private history. Abani Chatterjee thus emerges as a quintessentially modern man of no qualities who assumes definitive shape only in the lost reels of the films he starred in.
The final chapter argues that the static images and visual frames forwarded in the texts observe an integral psychological function: Premised upon linear perspective they imply a singular, static subjectivity appealing to the postmodern subject. In the corpus texts, the rise of digital technology in the 1990s thus appears not so much to have displaced older image repertories, practices and media techniques, than it has lent them greater visibility and appeal. Moreover, bricolage and pastiche emerge as cultural techniques which marked modernity from its inception. What the novels thus perpetuate is a media archeology not entirely servant to the poetics of the real. The permeable subject and the notion of the gaze as an active exchange as encapsulated in the concept of darshan - ideas informing all four texts - bespeak the resilience of a mythical universe continually re-instantiated in new technologies and uses. Eventually, the novels convey a sense of subalternity to a substantially Hindu nationalist history and historiography, the centrifugal force of which developed in the twentieth century and continues into the present.
Die Honigbiene Apis mellifera zeigt innerhalb einer Kolonie eine an das Alter gekoppelte Arbeitsteilung. Junge Honigbienen versorgen die Brut (Ammenbienen), während ältere Honigbienen (Sammlerinnen) außerhalb des Stocks Pollen und Nektar eintragen. Die biogenen Amine Octopamin und Tyramin sind an der Steuerung der Arbeitsteilung maßgeblich beteiligt. Sie interagieren mit Zielzellen über die Bindung an G Protein gekoppelte Rezeptoren. A. mellifera besitzt fünf charakterisierte Octopaminrezeptoren (AmOctαR1, AmOctβR1-4), einen charakterisierten Tyraminrezeptor (AmTyr1) sowie einen weiteren putativen Tyraminrezeptor.
In der vorliegenden Arbeit wurde dieser putative Aminrezeptor als zweiter Tyraminrezeptor (AmTyr2) identifiziert, lokalisiert und pharmakologisch charakterisiert.
Die von der cDNA abgeleitete Aminosäuresequenz weist strukturelle Eigenschaften und konservierte Motive von G Protein gekoppelten Rezeptoren auf. Phylogenetisch ordnet sich der AmTyr2 Rezeptor bei den Tyramin 2 Rezeptoren anderer Insekten ein. Die funktionelle und pharmakologische Charakterisierung des putativen Tyraminrezeptors erfolgte in modifizierten HEK293 Zellen, die mit der Rezeptor cDNA transfiziert wurden. Die Applikation von Tyramin aktiviert Adenylylcyclasen in diesen Zellen und resultiert in einem Anstieg des intrazellulären cAMP Gehalts. Der AmTyr2 Rezeptor kann durch Tyramin in nanomolaren Konzentrationen halbmaximal aktiviert werden. Während es sich bei Octopamin um einen wirkungsvollen Agonisten des Rezeptors handelt, sind Mianserin und Yohimbin effektive Antagonisten. Für die Lokalisierung des Rezeptorproteins wurde ein polyklonaler Antikörper generiert. Eine AmTyr2-ähnliche Immunreaktivität zeigt sich im Gehirn in den optischen Loben, den Antennalloben, dem Zentralkomplex und in den Kenyon Zellen der Pilzkörper.
Des Weiteren wurde die Rolle der Octopamin- und Tyraminrezeptoren bei der Steuerung der altersabhängigen Arbeitsteilung analysiert.
Die Genexpression des AmOctαR1 in verschiedenen Gehirnteilen korreliert unabhängig vom Alter mit der sozialen Rolle, während sich die Genexpression von AmOctβR3/4 und den Tyraminrezeptoren AmTyr1 und AmTyr2 maximal mit dem Alter aber nicht der sozialen Rolle ändert. Sammlerinnen weisen einen höheren Octopamingehalt im Gesamtgehirn auf als Ammenbienen; bei Tyramin zeigen sich keine Unterschiede. Während Tyramin offensichtlich keine direkte Rolle spielt, werden durch Octopamin gesteuerte Prozesse der altersabhängigen Arbeitsteilung bei der Honigbiene vermutlich über den AmOctαR1 vermittelt.
Die Ergebnisse der vorliegenden Arbeit zeigen die wichtige Rolle von biogenen Aminen, insbesondere Octopamin bei der sozialen Organisation von Insektenstaaten.
The transmission of wildlife zoonoses to humans depends, amongst others, on complex interactions of host population ecology and pathogen dynamics within host populations. In Europe, the Puumala virus (PUUV) causes nephropathia epidemica in humans. In this study we investigated complex interrelations within the epidemic system of PUUV and its rodent host, the bank vole (Myodes glareolus). We suggest that beech fructification and bank vole abundance are both decisive factors affecting human PUUV infections. While rodent host dynamics are expected to be directly linked to human PUUV infections, beech fructification is a rather indirect predictor by serving as food source for PUUV rodent hosts. Furthermore, we examined the dependence of bank vole abundance on beech fructification. We analysed a 12-year (2001-2012) time series of the parameters: beech fructification (as food resource for the PUUV host), bank vole abundance and human incidences from 7 Federal States of Germany. For the first time, we could show the direct interrelation between these three parameters involved in human PUUV epidemics and we were able to demonstrate on a large scale that human PUUV infections are highly correlated with bank vole abundance in the present year, as well as beech fructification in the previous year. By using beech fructification and bank vole abundance as predictors in one model we significantly improved the degree of explanation of human PUUV incidence. Federal State was included as random factor because human PUUV incidence varies considerably among states. Surprisingly, the effect of rodent abundance on human PUUV infections is less strong compared to the indirect effect of beech fructification. Our findings are useful to facilitate the development of predictive models for host population dynamics and the related PUUV infection risk for humans and can be used for plant protection and human health protection purposes.
Hintergrund: In Deutschland stellt der akute Myokardinfarkt (MI) eine der häufigsten Todesursachen dar. Als Ursache für regionale Unterschiede bei den Mortalitätsraten werden divergente Versorgungsstrukturen vermutet. Ziel der Untersuchung war, diese Fragestellung anhand anonymisierter krankenkassenbasierter Abrechnungsdaten zu evaluieren.
Methodik: Standardisierte Hospitalisierungs- sowie Krankenhaus- und Ein-Jahres-Mortalitätsraten nach MI wurden anhand anonymisierter Versichertendaten einer gesetzlichen Krankenkasse für das Jahr 2012 und die Bundesländer Berlin, Brandenburg und Mecklenburg-Vorpommern ermittelt (n=1.387.084, 46.3% male, 60.9 ± 18,2 years). Weiterhin wurden prädiktive Einflussfaktoren auf die Ein-Jahres-Mortalität, auf die Durchführung invasiver Prozeduren und auf eine leitliniengerechte pharmakotherapeutische Sekundärprävention analysiert.
Ergebnisse: 6.733 Patienten (73,7 ±13,0 Jahre, 56,7% männlich) wurden identifiziert. Obwohl für das Bundesland Berlin eine höhere Hospitalisierungsrate als in Mecklenburg-Vorpommern ermittelt werden konnte, ließen sich bei der Krankenhaus- und 1-Jahres-Mortalität keine signifikant abweichenden Raten zwischen den Bundesländern beobachten. Die Durchführung einer Koronarangiographie (OR: 0,42 [0,35-0,51]) und eine leitliniengerechte Pharmakotherapie (OR: 0,14 [0,12-0,17] waren mit einer geringeren 1-Jahres-Mortalität assoziiert. Die Durchführung einer Koronarangiographie und eine leitliniengerechte Pharmakotherapie von Patienten nach Myokardinfarkt wurde hingegen primär durch Alter und Geschlecht, nicht aber durch das Bundesland determiniert.
Folgerung: Eine regional divergierende stationäre und postinfarzielle Versorgung auf Bundesland-Ebene kann anhand der vorliegenden Daten nicht nachgewiesen werden.
Gangsta-Rap ist besonders in Deutschland stark männlich dominiert und als Musikgenre kommerziell sehr erfolgreich. Er gilt als eine der wenigen Zonen bislang weitestgehend unbedrohter Männlichkeitsentwürfe.
Die Ende des Jahres 2011 in Erscheinung getretene Gangsta-Rapperin Schwesta Ewa könnte gerade aufgrund ihres biologischen Geschlechts die Chance nutzen, Männlichkeitskonzeptionen und Vorstellungen von Weiblichkeit im Gangsta-Rap infrage zu stellen. Doch welche Konstruktionen von Männlichkeit und Weiblichkeit stellen Männer und Frauen in der Praxis genau auf? Zur Beantwortung dieser Forschungsfrage werden primär die soziologischen Konzepte zu hegemonialer Männlichkeit (Connell), Habitus (Bourdieu) und männlichem Geschlechtshabitus (Meuser) herangezogen. Die theoriegeleitete, fallspezifische Zuordnung und Analyse von Songtexten erfolgt durch die empirische Methode der inhaltlich strukturierenden Inhaltsanalyse. Ein Ergebnis ist, dass Schwesta Ewa eine komplexe Eigen-Weiblichkeitskonstruktion aufstellt. Über den Vergleich mit dem untersuchten Gangsta-Rapper Kollegah wird u. a. die These von Connell und Messerschmidt empirisch nachgewiesen, wonach auch Frauen Träger von Aspekten hegemonialer Männlichkeit sein können.
Die erste Forschungsfrage untersucht vor allem die homosoziale Dimension sowie das für Gangsta-Rap konstitutive Moment von Eigenaufwertung und Fremdabwertung. Welche Muster von Anerkennung existieren jedoch zwischen Männern und Frauen? Für diese zweite Forschungsfrage wird eine Diskursanalyse durchgeführt, mit der viele Künstler/-innen und ihre Songtexte untersucht werden. Es wird ermittelt, dass Gangsta-Rap auch als Repräsentation konservativer Geschlechterbilder zu verstehen ist. Ebenso sind Schilderungen zu alltäglich erfahrbaren sozialen Phänomenen wie Liebe und Freundschaft als auch zu den damit einhergehenden sozialen Rollen vorzufinden. Die Perspektive gegengeschlechtlicher Wertschätzung kann aber gleichzeitig ablaufende Abwertung beibehalten.
We present the first physical characterization of the young open cluster VVVCL041. We spectroscopically observed the cluster main-sequence stellar population and a very-massive star candidate: WR62-2. CMFGEN modelling to our near-infrared spectra indicates that WR62-2 is a very luminous (10^6.4±0.2 L⊙)and massive (∼ 80M⊙) star.
Diese Arbeit befasst sich mit der Rheinischen Verlaufsform (RV) im rheinfränkischen Dialekt. Nach dem DUDEN handelt es sich bei der RV um eine Konstruktion, die aus dem Kopulaverb sein und einer PP mit am und nominalisiertem Infinitiv besteht und dem Ausdruck von progressivem Aspekt dient.
Die vorliegenden Arbeiten zur RV beschäftigen sich im Wesentlichen entweder mit der Ausprägung der Konstruktion im Standarddeutschen (z.B. Reimann (1999), Krause (2002), Rödel (2003), Rödel (2004a), Rödel (2004b), van Pottelberge (2004)) oder im Ripuarischen (z.B. Andersson (1989), Bhatt & Schmidt (1993)) und kommen zu unterschiedlichen Ergebnissen bezüglich der Verwendungsmöglichkeiten und des Aufbaus der Konstruktion, insbesondere des Status des Infinitivs in der Verlaufsform.
Hauptziel dieser Arbeit ist es, zu zeigen, dass sich die Grammatikalisierung der Verlaufsform von der im DUDEN beschriebenen Konstruktion zu einer analytischen Verbform entlang eines festen Grammatikalisierungspfades vollzieht und die entsprechenden Teilschritte bei der Entwicklung zu einer analytischen Verbform herauszuarbeiten. Auf dieser Grundlage wird in der Arbeit dargestellt, wie sich mittels eines geeigneten Sets an Indikatoren der Grammatikalisierungsgrad der Verlaufsform in einem Dialektraum oder einem diatopischen Register konkret feststellen lässt.
In 1945, Zinovii Shenderovich Tolkatchev (1903–1977), a Soviet artist of Jewish origin, created a striking series of five images entitled “Jesus in Majdanek”. The series was the culmination of Tolkatchev‘s intensive preoccupation with the experience he, as a Red Army soldier, endured upon taking part in liberation of the concentration camps Majdanek and Auschwitz. Shocked by the actual sights he witnessed, he depicted Jesus as an actual camp inmate, wearing a striped uniform marked by every possible defamation sign – the Jewish yellow star, the red triangle of political prisoners, and the individual prison number, the numerical tattoo on his lower arm can also be seen. The different stages of camp life are portrayed as the traditional Passion of Christ. While showing the actual situations the artist based himself upon the well known European Renaissance paintings canonically depicting Jesus‘ suffering. The article places Tolkatchev‘s series in a broader cultural and visual context by exploring the development of the ‘historical Jesus’ in the 19th century European thought and Russian realist art, and by examining the impact of the German avant-garde. By doing so, a deeper understanding of the universal message Tolkatchev’s works entail is offered.
Plant cell walls are complex structures that underpin plant growth and are widely exploited in diverse human activities thus placing them with a central importance in biology. Cell walls have been a prominent area of research for a long time, but the chemical complexity and diversity of cell walls not just between species, but also within plants, between cell-types, and between cell wall micro-domains pose several challenges. Progress accelerated several-fold in cell wall biology owing to advances in sequencing technology, aided soon thereafter by advances in omics and imaging technologies. This development provides additional perspectives of cell walls across a rapidly growing number of species, highlighting a myriad of architectures, compositions, and functions.
Furthermore, rather than the component centric view, integrative analysis of the different cell wall components across system-levels help to gain a more in-depth understanding of the structure and biosynthesis of the cell envelope and its interactions with the environment.
To this end, in this work three case studies are detailed, all pertaining to the integrative analysis of heterogeneous cell wall related data arising from different system-levels and analytical techniques. A detailed account of multiblock methods is provided and in particular canonical correlation and regression methods of data integration are discussed. In the first integrative analysis, by employing canonical correlation analysis - a multivariate statistical technique to study the association between two datasets - novel insight to the relationship between glycans and phenotypic traits is gained. In addition, sparse partial least squares regression approach that adapts Lasso penalization and allows for the selection of a subset of variables was employed. The second case study focuses on an integrative analysis of images obtained from different spectroscopic techniques. By employing yet another multiblock approach - multiple co-inertia analysis, insitu biochemical composition of cell walls from different cell-types is studied thereby highlighting the common and complementary parts of the two hyperspectral imaging techniques. Finally, the third integrative analysis facilitates gene expression analysis of the Arabidopsis root transcriptome and translatome for the identification of cell wall related genes and compare expression patterns of cell wall synthesis genes. The computational analysis considered correlation and variation of expression across cell-types at both system-levels, and also provides insight into the degree of co-regulatory relationships that are preserved between the two processes.
The integrative analysis of glycan data and phenotypic traits in cotton fibers using canonical methods led to the identification of specific polysaccharides which may play a major role during fiber development for the final fiber characteristics. Furthermore, this analysis provides a base for future studies on glycan arrays in case of developing cotton fibers. The integrative analysis of images from infrared and Raman spectroscopic approaches allowed the coupling of different analytical techniques to characterize complex biological material, thereby, representing various facets of their chemical properties. Moreover, the results from the co-inertia analysis demonstrated that the study was well adapted as it is relevant for coupling data tables in a symmetric way. Several indicators are proposed to investigate how the global and block scores are related. In addition, studying the root cells of \textit{Arabidopsis thaliana} allowed positing a novel pipeline to systematically investigate and integrate the different levels of information available at the global and single-cell level. The conducted analysis also confirms that previously identified key transcriptional activators of secondary cell wall development display highly conserved patterns of transcription and translation across the investigated cell-types. Moreover, the biological processes that display conserved and divergent patterns based on the cell-type-specific expression and translation levels are identified.
Welfare states and policies have changed greatly over the past decades, mostly characterized by retrenchments in terms of government spending or in terms of restricted access to certain benefits. In the area of family policies, however, a lot of countries have simultaneously expanded provisions and transfers for families. Bringing together the macro analysis of policy variation and household income changes on the micro-level, the main research question of the dissertation is to what extent economic consequences following separation and divorce in families with children have changed between the 1980s and the 2000s in Germany and the United States. The second research question of the dissertation regards the differences in dissolution outcomes between married and cohabiting parents in Germany.
The dissertation thus aims to link institutional regulations of welfare states with the actual income situation of families. To achieve this, a research design was developed that has never been used for the analysis of the economic consequences of family dissolution. For this, the two longest running panel datasets, German Socio-economic Panel (GSOEP) and the US American Panel Study of Income Dynamics (PSID), have been used. The analytic strategy applied to estimate the effects of family dissolution on household income is a difference-in-difference design combined with coarsened exact matching (CEM).
To begin with, the dissertation confirmed many findings of previous research, for example regarding the gender differences in family dissolution outcomes. Mothers experience clearly higher relative income losses and consequently higher risks of poverty than fathers. This finding is universal, that is it holds for both countries, for all time periods observed, and for all measures of economic outcome that were employed. Another confirmed finding is the higher level of welfare state intervention in Germany compared to the United States.
The dissertation also revealed a number of novel findings. The results show that the expansion of family policies in Germany over time has not been accompanied by substantially decreasing income losses for mothers. Though income losses have slightly decreased over time, they have become more persistent during the years following family dissolution. The impact of the German welfare state has meanwhile been quite stable.
American mothers’ income losses took place on a slightly lower level than those of German mothers. Only during the 1980s their relative losses were clearly lower than those of German mothers. And also American mothers did not recover as much from their income losses during the 2000s than they used to during the 1980s. For them, the 1996 welfare reform brought a considerable decrease in welfare state support. Accordingly, the results for American mothers can certainly be described as a shift from public to private provision.
The general finding of previous studies that fathers do not have to suffer income losses, or if at all rather moderate ones compared to mothers, can be confirmed. Nevertheless, both German and US American fathers face a deterioration of the economic consequences of family dissolution over time. German fathers’ relative income changes are still positive though they have decreased over time. One reason for this decrease is the increasing loss of partner earnings following union dissolution. Also among American fathers, income gains still prevail in the year of family dissolution. Two years later, however, they have been facing income losses already since the 1980s which have furthermore increased considerably over time.
Zooming in on Germany, family dissolution outcomes by marital status show negligible differences between cohabiting and married mothers in disposable income, but considerable differences in losses of income before taxes and transfers. It is the impact of the welfare state that equalizes the differences in income losses between these two groups of mothers. For married mothers, losses are not as high in the year of event but they have difficulties to recover from these losses. Without the income buffering of the welfare state, married mothers would, three years after family dissolution, remain with relative income losses double as high as for cohabiting mothers.
Compared to mothers, differences between married and cohabiting fathers are visible in changes of income before as well as after taxes and transfers. The welfare state does not alter the difference between the two groups of fathers. With regard to both income concepts, cohabiting fathers fare worse than married fathers. Cohabiting fathers suffer moderate income losses of disposable income while married fathers experience moderate income gains. Accounting for support payments is decisive for fathers’ income changes. If these payments are not deducted from disposable income, both married and cohabiting fathers experience gains in disposable income following family dissolution.
Several authors highlighted that the time course of an experiment itself could have a substantial influence on the interpretability of experimental effects. Since mixed effects modeling had enabled researchers to investigate more complex problems with more precision than before, two naming experiments were conducted with college students, with and without non-words intermixed, and analyzed with regard to frequency, quality, interactive and trial-history effects. The present analyses build on and extend the Bates, Kliegl, Vasishth, and Baayen (2015) approach in order to converge on a parsimonious model that accounts for autocorrelated errors caused by trial history. For three of four cases, a history-sensitive model improved the model fit over a history-naïve model and explained more deviance. In one of these cases, the herein presented approach helped reveal an interaction between stimulus frequency and quality that was not significant without a trial history account. Main and joint effects, limitations, as well as directions for further research, are briefly discussed.
Many chemical reactions in biological cells occur at very low concentrations of constituent molecules. Thus, transcriptional gene-regulation is often controlled by poorly expressed transcription-factors, such as E.coli lac repressor with few tens of copies. Here we study the effects of inherent concentration fluctuations of substrate-molecules on the seminal Michaelis-Menten scheme of biochemical reactions. We present a universal correction to the Michaelis-Menten equation for the reaction-rates. The relevance and validity of this correction for enzymatic reactions and intracellular gene-regulation is demonstrated. Our analytical theory and simulation results confirm that the proposed variance-corrected Michaelis-Menten equation predicts the rate of reactions with remarkable accuracy even in the presence of large non-equilibrium concentration fluctuations. The major advantage of our approach is that it involves only the mean and variance of the substrate-molecule concentration. Our theory is therefore accessible to experiments and not specific to the exact source of the concentration fluctuations.
Physical computing covers the design and realization of interactive
objects and installations and allows students to develop concrete,
tangible products of the real world that arise from the learners’
imagination. This way, constructionist learning is raised to a level that
enables students to gain haptic experience and thereby concretizes the
virtual. In this paper the defining characteristics of physical computing
are described. Key competences to be gained with physical computing
will be identified.
The high-latitudinal thermospheric processes driven by the solar wind and Interplanetary Magnetic Field (IMF) interaction with the Earth magnetosphere are highly variable parts of the complex dynamic plasma environment, which represent the coupled Magnetosphere – Ionosphere – Thermosphere (MIT) system. The solar wind and IMF interactions transfer energy to the MIT system via reconnection processes at the magnetopause. The Field Aligned Currents (FACs) constitute the energetic links between the magnetosphere and the Earth ionosphere. The MIT system depends on the highly variable solar wind conditions, in particular on changes of the strength and orientation of the IMF.
In my thesis, I perform an investigation on the physical background of the complex MIT system using the global physical - numerical, three-dimensional, time-dependent and self-consistent Upper Atmosphere Model (UAM). This model describes the thermosphere, ionosphere, plasmasphere and inner magnetosphere as well as the electrodynamics of the coupled MIT system for the altitudinal range from 80 (60) km up to the 15 Earth radii.
In the present study, I developed and investigated several variants of the high-latitudinal electrodynamic coupling by including the IMF dependence of FACs into the UAM model. For testing, the various variants were applied to simulations of the coupled MIT system for different seasons, geomagnetic activities, various solar wind and IMF conditions. Additionally, these variants of the theoretical model with the IMF dependence were compared with global empirical models. The modelling results for the most important thermospheric parameters like neutral wind and mass density were compared with satellite measurements. The variants of the UAM model with IMF dependence show a good agreement with the satellite observations. In comparison with the empirical models, the improved variants of the UAM model reproduce a more realistic meso-scale structures and dynamics of the coupled MIT system than the empirical models, in particular at high latitudes. The new configurations of the UAM model with IMF dependence contribute to the improvement of space weather prediction.
During the last two decades, instability training devices have become a popular means in athletic training and rehabilitation of mimicking unstable surfaces during movements like vertical jumps. Of note, under unstable conditions, trunk muscles seem to have a stabilizing function during exercise to facilitate the transfer of torques and angular momentum between the lower and upper extremities. The present thesis addresses the acute effects of surface instability on performance during jump-landing tasks. Additionally, the long-term effects (i.e., training) of surface instability were examined with a focus on the role of the trunk in athletic performance/physical fitness.
Healthy adolescent, and young adult subjects participated in three cross-sectional and one longitudinal study, respectively. Performance in jump-landing tasks on stable and unstable surfaces was assessed by means of a ground reaction force plate. Trunk muscle strength (TMS) was determined using an isokinetic device or the Bourban TMS test. Physical fitness was quantified by standing long jump, sprint, stand-and-reach, jumping sideways, Emery balance, and Y balance test on stable surfaces. In addition, activity of selected trunk and leg muscles and lower limb kinematics were recorded during jump-landing tasks.
When performing jump-landing tasks on unstable compared to stable surfaces, jump performance and leg muscle activity were significantly lower. Moreover, significantly smaller knee flexion angles and higher knee valgus angles were observed when jumping and landing on unstable compared to stable conditions and in women compared to men. Significant but small associations were found between behavioral and neuromuscular data, irrespective of surface condition. Core strength training on stable as well as on unstable surfaces significantly improved TMS, balance and coordination.
The findings of the present thesis imply that stable rather than unstable surfaces provide sufficient training stimuli during jump exercises (i.e., plyometrics). Additionally, knee motion strategy during plyometrics appears to be modified by surface instability and sex. Of note, irrespective of surface condition, trunk muscles only play a minor role for leg muscle performance/activity during jump exercises. Moreover, when implemented in strength training programs (i.e., core strength training), there is no advantage in using instability training devices compared to stable surfaces in terms of enhancement of athletic performance.
Development of geophysical methods to characterize methane hydrate reservoirs on a laboratory scale

Gas hydrates are crystalline solids composed of water and gas molecules. They are stable at elevated pressure and low temperatures. Therefore, natural gas hydrate deposits occur at continental margins, permafrost areas, deep lakes, and deep inland seas. During hydrate formation, the water molecules rearrange to form cavities which host gas molecules. Due to the high pressure during hydrate formation, significant amounts of gas can be stored in hydrate structures. The water-gas ratio hereby can reach up to 1:172 at 0°C and atmospheric pressure. Natural gas hydrates predominantly contain methane. Because methane constitutes both a fuel and a greenhouse gas, gas hydrates are a potential energy resource as well as a potential source for greenhouse gas.
This study investigates the physical properties of methane hydrate bearing sediments on a laboratory scale. To do so, an electrical resistivity tomography (ERT) array was developed and mounted in a large reservoir simulator (LARS). For the first time, the ERT array was applied to hydrate saturated sediment samples under controlled temperature, pressure, and hydrate saturation conditions on a laboratory scale. Typically, the pore space of (marine) sediments is filled with electrically well conductive brine. Because hydrates constitute an electrical isolator, significant contrasts regarding the electrical properties of the pore space emerge during hydrate formation and dissociation. Frequent measurements during hydrate formation experiments permit the recordings of the spatial resistivity distribution inside LARS. Those data sets are used as input for a new data processing routine which transfers the spatial resistivity distribution into the spatial distribution of hydrate saturation. Thus, the changes of local hydrate saturation can be monitored with respect to space and time.
This study shows that the developed tomography yielded good data quality and resolved even small amounts of hydrate saturation inside the sediment sample. The conversion algorithm transforming the spatial resistivity distribution into local hydrate saturation values yielded the best results using the Archie-var-phi relation. This approach considers the increasing hydrate phase as part of the sediment frame, metaphorically reducing the sample’s porosity. In addition, the tomographical measurements showed that fast lab based hydrate formation processes cause small crystallites to form which tend to recrystallize.
Furthermore, hydrate dissociation experiments via depressurization were conducted in order to mimic the 2007/2008 Mallik field trial. It was observed that some patterns in gas and water flow could be reproduced, even though some setup related limitations arose.
In two additional long-term experiments the feasibility and performance of CO2-CH4 hydrate exchange reactions were studied in LARS. The tomographical system was used to monitor the spatial hydrate distribution during the hydrate formation stage. During the subsequent CO2 injection, the tomographical array allowed to follow the CO2 migration front inside the sediment sample and helped to identify the CO2 breakthrough.
Mentoring in a Digital World

This paper focuses on the results of the evaluation of the first
pilot of an e-mentoring unit designed by the Hands-On ICT consortium,
funded by the EU LLL programme. The overall aim of this two-year
activity is to investigate the value for professional learning of Massive
Online Open Courses (MOOCs) and Community Online Open Courses
(COOCs) in the context of a ‘community of practice’. Three units in the
first pilot covered aspects of using digital technologies to develop creative
thinking skills. The findings in this paper relate to the fourth unit
about e-mentoring, a skill that was important to delivering the course
content in the other three units. Findings about the e-mentoring unit
included: the students’ request for detailed profiles so that participants
can get to know each other; and, the need to reconcile the different
interpretations of e-mentoring held by the participants when the course
begins. The evaluators concluded that the major issues were that: not all
professional learners would self-organise and network; and few would
wish to mentor their colleagues voluntarily. Therefore, the e-mentoring
issues will need careful consideration in pilots two and three to identify
how e-mentoring will be organised.
Spatio-temporal control of cellular uptake achieved by photoswitchable cell-penetrating peptides

The selective uptake of compounds into specific cells of interest is a major objective in cell biology and drug delivery. By incorporation of a novel, thermostable azobenzene moiety we generated peptides that can be switched optically between an inactive state and an active, cell-penetrating state with excellent spatio-temporal control.
Winter storms are the most costly natural hazard for European residential property. We compare four distinct storm damage functions with respect to their forecast accuracy and variability, with particular regard to the most severe winter storms. The analysis focuses on daily loss estimates under differing spatial aggregation, ranging from district to country level. We discuss the broad and heavily skewed distribution of insured losses posing difficulties for both the calibration and the evaluation of damage functions. From theoretical considerations, we provide a synthesis between the frequently discussed cubic wind–damage relationship and recent studies that report much steeper damage functions for European winter storms. The performance of the storm loss models is evaluated for two sources of wind gust data, direct observations by the German Weather Service and ERA-Interim reanalysis data. While the choice of gust data has little impact on the evaluation of German storm loss, spatially resolved coefficients of variation reveal dependence between model and data choice. The comparison shows that the probabilistic models by Heneka et al. (2006) and Prahl et al. (2012) both provide accurate loss predictions for moderate to extreme losses, with generally small coefficients of variation. We favour the latter model in terms of model applicability. Application of the versatile deterministic model by Klawa and Ulbrich (2003) should be restricted to extreme loss, for which it shows the least bias and errors comparable to the probabilistic model by Prahl et al. (2012).

Global food production needs to be increased by 60-110% between 2005 and 2050 to meet growing food and feed demand. Intensification and/or expansion of agriculture are the two main options available to meet the growing crop demands. Land conversion to expand cultivated land increases GHG emissions and impacts biodiversity and ecosystem services. Closing yield gaps to attain potential yields may be a viable option to increase the global crop production. Traditional methods of agricultural intensification often have negative externalities. Therefore, there is a need to explore location-specific methods of sustainable agricultural intensification. We identified regions where the achievement of potential crop calorie production on currently cultivated land will meet the present and future food demand based on scenario analyses considering population growth and changes in dietary habits. By closing yield gaps in the current irrigated and rain-fed cultivated land, about 24% and 80% more crop calories can respectively be produced compared to 2000. Most countries will reach food self-sufficiency or improve their current food self-sufficiency levels if potential crop production levels are achieved. As a novel approach, we defined specific input and agricultural management strategies required to achieve the potential production by overcoming biophysical and socioeconomic constraints causing yield gaps. The management strategies include: fertilizers, pesticides, advanced soil management, land improvement, management strategies coping with weather induced yield variability, and improving market accessibility. Finally, we estimated the required fertilizers (N, P2O5, and K2O) to attain the potential yields. Globally, N-fertilizer application needs to increase by 45-73%, P2O5-fertilizer by 22-46%, and K2O-fertilizer by 2-3 times compared to the year 2010 to attain potential crop production. The sustainability of such agricultural intensification largely depends on the way management strategies for closing yield gaps are chosen and implemented.
Anthropogenic activities have transformed the Earth's environment, not only on local level, but on the planetary-scale causing global change. Besides industrialization, agriculture is a major driver of global change. This change in turn impairs the agriculture sector, reducing crop yields namely due to soil degradation, water scarcity, and climate change. However, this is a more complex issue than it appears. Crop yields can be increased by use of agrochemicals and fertilizers which are mainly produced by fossil energy. This is important to meet the increasing food demand driven by global demographic change, which is further accelerated by changes in regional lifestyles. In this dissertation, we attempt to address this complex problem exploring agricultural potential globally but on a local scale. For this, we considered the influence of lifestyle changes (dietary patterns) as well as technological progress and their effects on climate change, mainly greenhouse gas (GHG) emissions. Furthermore, we examined options for optimizing crop yields in the current cultivated land with the current cropping patterns by closing yield gaps. Using this, we investigated in a five-minute resolution the extent to which food demand can be met locally, and/or by regional and/or global trade. Globally, food consumption habits are shifting towards calorie rich diets. Due to dietary shifts combined with population growth, the global food demand is expected to increase by 60-110% between 2005 and 2050. Hence, one of the challenges to global sustainability is to meet the growing food demand, while at the same time, reducing agricultural inputs and environmental consequences. In order to address the above problem, we used several freely available datasets and applied multiple interconnected analytical approaches that include artificial neural network, scenario analysis, data aggregation and harmonization, downscaling algorithm, and cross-scale analysis.
Globally, we identified sixteen dietary patterns between 1961 and 2007 with food intakes ranging from 1,870 to 3,400 kcal/cap/day. These dietary patterns also reflected changing dietary habits to meat rich diets worldwide. Due to the large share of animal products, very high calorie diets that are common in the developed world, exhibit high total per capita emissions of 3.7-6.1 kg CO2eq./day. This is higher than total per capita emissions of 1.4-4.5 kg CO2eq./day associated with low and moderate calorie diets that are common in developing countries. Currently, 40% of the global crop calories are fed to livestock and the feed calorie use is four times the produced animal calories. However, these values vary from less than 1 kcal to greater 10 kcal around the world. On the local and national scale, we found that the local and national food production could meet demand of 1.9 and 4.4 billion people in 2000, respectively. However, 1 billion people from Asia and Africa require intercontinental agricultural trade to meet their food demand. Nevertheless, these regions can become food self-sufficient by closing yield gaps that require location specific inputs and agricultural management strategies. Such strategies include: fertilizers, pesticides, soil and land improvement, management targeted on mitigating climate induced yield variability, and improving market accessibility. However, closing yield gaps in particular requires global N-fertilizer application to increase by 45-73%, P2O5 by 22-46%, and K2O by 2-3 times compare to 2010. Considering population growth, we found that the global agricultural GHG emissions will approach 7 Gt CO2eq./yr by 2050, while the global livestock feed demand will remain similar to 2000. This changes tremendously when diet shifts are also taken into account, resulting in GHG emissions of 20 Gt CO2eq./yr and an increase of 1.3 times in the crop-based feed demand between 2000 and 2050. However, when population growth, diet shifts, and technological progress by 2050 were considered, GHG emissions can be reduced to 14 Gt CO2eq./yr and the feed demand to nearly 1.8 times compare to that in 2000. Additionally, our findings shows that based on the progress made in closing yield gaps, the number of people depending on international trade can vary between 1.5 and 6 billion by 2050. In medium term, this requires additional fossil energy. Furthermore, climate change, affecting crop yields, will increase the need for international agricultural trade by 4% to 16%.
In summary, three general conclusions are drawn from this dissertation. First, changing dietary patterns will significantly increase crop demand, agricultural GHG emissions, and international food trade in the future when compared to population growth only. Second, such increments can be reduced by technology transfer and technological progress that will enhance crop yields, decrease agricultural emission intensities, and increase livestock feed conversion efficiencies. Moreover, international trade dependency can be lowered by consuming local and regional food products, by producing diverse types of food, and by closing yield gaps. Third, location specific inputs and management options are required to close yield gaps. Sustainability of such inputs and management largely depends on which options are chosen and how they are implemented. However, while every cultivated land may not need to attain its potential yields to enable food security, closing yield gaps only may not be enough to achieve food self-sufficiency in some regions. Hence, a combination of sustainable implementations of agricultural intensification, expansion, and trade as well as shifting dietary habits towards a lower share of animal products is required to feed the growing population.
Das im vorliegenden Beitrag beschriebene hochschuldidaktische Format des Kolloquiums dient dem kollegialen Austausch Lehrender über typische Probleme ihres Lehralltags und über mögliche Lösungswege. Dafür wird das Erfahrungswissen der Lehrenden als Praktikerinnen und Praktiker aktiviert. Das Format wurde 2012 an der Universität Potsdam eingeführt und wurde mittlerweile auf die Universität Magdeburg übertragen. Der Artikel beschreibt die Kennzeichen des Formats und diskutiert seinen theoretischen Hintergrund.
Dynamic C and N stocks

The drainage and cultivation of fen peatlands create complex small-scale mosaics of soils with extremely variable soil organic carbon (SOC) stocks and groundwater levels (GWLs). To date, the significance of such sites as sources or sinks for greenhouse gases such as CO2 and CH4 is still unclear, especially if the sites are used for cropland. As individual control factors such as GWL fail to account for this complexity, holistic approaches combining gas fluxes with the underlying processes are required to understand the carbon (C) gas exchange of drained fens. It can be assumed that the stocks of SOC and N located above the variable GWL - defined as dynamic C and N stocks - play a key role in the regulation of the plant- and microbially mediated CO2 fluxes in these soils and, inversely, for CH4. To test this assumption, the present study analysed the C gas exchange (gross primary production - GPP; ecosystem respiration - R-eco; net ecosystem exchange - NEE; CH4) of maize using manual chambers for 4 years. The study sites were located near Paulinenaue, Germany, where we selected three soil types representing the full gradient of GWL and SOC stocks (0-1 m) of the landscape: (a) Haplic Arenosol (AR; 8 kg C m(-2)); (b) Mollic Gleysol (GL; 38 kg C m(-2)); and (c) Hemic Histosol (HS; 87 kg C m(-2)). Daily GWL data were used to calculate dynamic SOC (SOCdyn) and N (N-dyn) stocks.
Average annual NEE differed considerably among sites, ranging from 47 +/- 30 g C m(-2) yr(-1) in AR to -305 +/- 123 g C m(-2) yr(-1) in GL and -127 +/- 212 g C m(-2) yr(-1) in HS. While static SOC and N stocks showed no significant effect on C fluxes, SOCdyn and N-dyn and their interaction with GWL strongly influenced the C gas exchange, particularly NEE and the GPP : R-eco ratio. Moreover, based on nonlinear regression analysis, 86% of NEE variability was explained by GWL and SOCdyn. The observed high relevance of dynamic SOC and N stocks in the aerobic zone for plant and soil gas exchange likely originates from the effects of GWL-dependent N availability on C formation and transformation processes in the plant-soil system, which promote CO2 input via GPP more than CO2 emission via R-eco.
The process-oriented approach of dynamic C and N stocks is a promising, potentially generalisable method for system-oriented investigations of the C gas exchange of groundwater-influenced soils and could be expanded to other nutrients and soil characteristics. However, in order to assess the climate impact of arable sites on drained peatlands, it is always necessary to consider the entire range of groundwater-influenced mineral and organic soils and their respective areal extent within the soil landscape.
The invention and development of next or second generation sequencing methods has resulted in a dramatic transformation of ancient DNA research and allowed shotgun sequencing of entire genomes from fossil specimens. However, although there are exceptions, most fossil specimens contain only low (similar to 1% or less) percentages of endogenous DNA. The only skeletal element for which a systematically higher endogenous DNA content compared to other skeletal elements has been shown is the petrous part of the temporal bone. In this study we investigate whether (a) different parts of the petrous bone of archaeological human specimens give different percentages of endogenous DNA yields, (b) there are significant differences in average DNA read lengths, damage patterns and total DNA concentration, and (c) it is possible to obtain endogenous ancient DNA from petrous bones from hot environments. We carried out intra-petrous comparisons for ten petrous bones from specimens from Holocene archaeological contexts across Eurasia dated between 10,0001,800 calibrated years before present (cal. BP). We obtained shotgun DNA sequences from three distinct areas within the petrous: a spongy part of trabecular bone (part A), the dense part of cortical bone encircling the osseous inner ear, or otic capsule (part B), and the dense part within the otic capsule (part C). Our results confirm that dense bone parts of the petrous bone can provide high endogenous aDNA yields and indicate that endogenous DNA fractions for part C can exceed those obtained for part B by up to 65-fold and those from part A by up to 177-fold, while total endogenous DNA concentrations are up to 126-fold and 109-fold higher for these comparisons. Our results also show that while endogenous yields from part C were lower than 1% for samples from hot (both arid and humid) parts, the DNA damage patterns indicate that at least some of the reads originate from ancient DNA molecules, potentially enabling ancient DNA analyses of samples from hot regions that are otherwise not amenable to ancient DNA analyses.
Two of the most controversial issues concerning the late Cenozoic evolution of the Andean orogen are the timing of uplift of the intraorogenic Puna plateau and its eastern border, the Eastern Cordillera, and ensuing changes in climatic and surface-process conditions in the intermontane basins of the NW-Argentine Andes. The Eastern Cordillera separates the internally drained, arid Puna from semi-arid intermontane basins and the humid sectors of the Andean broken foreland and the Subandean fold-and-thrust belt to the east. With elevations between 4,000 and 6,000 m the eastern flanks of the Andes form an efficient orographic barrier with westward-increasing elevation and asymmetric rainfall distribution and amount with respect to easterly moisture-bearing winds. This is mirrored by pronounced gradients in the efficiency of surface processes that erode and re-distribute sediment from the uplifting ranges. Although the overall pattern of deformation and uplift in this sector of the southern central Andes shows an eastward migration of deformation, a well-developed deformation front does not exist and uplift and associated erosion and sedimentary processes are highly disparate in space and time. In addition, periodic deformation within intermontane basins, and continued diachronous foreland uplifts associated with the reactivation of inherited basement structures furthermore make a rigorous assessment of the spatiotemporal uplift patterns difficult.
This thesis focuses on the tectonic evolution of the Eastern Cordillera of NW Argentina, the depositional history of its intermontane sedimentary basins, and the regional topographic evolution of the eastern flank of the Puna Plateau. The intermontane basins of the Eastern Cordillera and the adjacent morphotectonic provinces of the Sierras Pampeanas and the Santa Bárbara System are akin to reverse fault bounded, filled, and partly coalesced sedimentary basins of the Puna Plateau. In contrast to the Puna basins, however, which still form intact morphologic entities, repeated deformation, erosion, and re-filling have impacted the basins in the Eastern Cordillera. This has resulted in a rich stratigraphy of repeated basin fills, but many of these basins have retained vestiges of their early depositional history that may reach back in time when these areas were still part of a contiguous and undeformed foreland basin. Fortunately, these strata also contain abundant volcanic ashes that are not only important horizons to decipher tectono-sedimentary events through U-Pb geochronology and geochemical correlation, but they also represent terrestrial recorders of the hydrogen-isotope composition of ancient meteoric waters that can be compared to the isotopic composition of modern meteoric water. The ash horizons are thus unique recorders of past environmental conditions and lend themselves to tracking the development of rainfall barriers and tectonically forced climate and environmental change through time.
U-Pb zircon geochronology and paleocurrent reconstructions of conglomerate sequences in the Humahuaca Basin of the Eastern Cordillera at 23.5° S suggest that the basin was an integral part of a largely unrestricted depositional system until 4.2 Ma, which subsequently became progressively decoupled from the foreland by range uplifts to the east that forced easterly moisture-bearing winds to precipitate in increasingly eastward locations. Multiple cycles of severed hydrological conditions and drainage re-capture are identified together with these processes that were associated with basin filling and sediment evacuation, respectively. Moreover, systematic relationships among faults, regional unconformities and deformed landforms reveal a general pattern of intra-basin deformation that appears to be linked with basin-internal deformation during or subsequent to episodes of large-scale sediment removal. Some of these observations are supported by variations in the hydrogen stable isotope composition of volcanic glass from the Neogene to Quaternary sedimentary record, which can be related to spatiotemporal changes in topography and associated orographic effects. δDg values in the basin strata reveal two main trends associated with surface uplift in the catchment area between 6.0 and 3.5 Ma and the onset of semiarid conditions in the basin following the attainment of threshold elevations for effective orographic barriers to the east after 3.5 Ma. The disruption of sediment supply from western sources after 4.2 Ma and subsequent hinterland aridification, moreover, emphasize the possibility that these processes were related to lateral orogenic growth of the adjacent Puna Plateau. As a result of the hinterland aridification the regions in the orogen interior have been characterized by an inefficient fluvial system, which in turn has helped maintaining internal drainage conditions, sediment storage, and relief reduction within high-elevation basins.
The diachronous nature of basin formation and impacts on the fluvial system in the adjacent broken foreland is underscored by the results of detailed sediment provenance and paleocurrent analyses, as well as U-Pb zircon geochronology in the Lerma and Metán basins at ca. 25° S. This is particularly demonstrated by the isolated uplift of the Metán range at ~10 Ma, which is more than 50 km away from the presently active orogenic front along the eastern Puna margin and the Eastern Cordillera to the west. At about 5 Ma, Puna-sourced sediments disappear from the foreland record, documenting further range uplifts in the Eastern Cordillera and hydrological isolation of the neighboring Angastaco Basin from the foreland. Finally, during the late Pliocene and Quaternary, deformation has been accommodated across the entire foreland and is still active. To elucidate the interactions between tectonically controlled changes in elevation and their impact on atmospheric circulation processes in this region, this thesis provides additional, temporally well-constrained hydrogen stable isotope results of volcanic glass samples from the broken foreland, including the Angastaco Basin, and other intermontane basins farther south. The results suggest similar elevations of intermontane basins and the foreland sectors prior to ca. 7 Ma. In case of the Angastaco Basin the region was affected by km-scale surface uplift of the basin. A comparison with coeval isotope data collected from sedimentary sequences in the Puna plateau explains rapid shifts in the intermontane δDg record and supports the notion of recurring phases of enhanced deep convection during the Pliocene, and thus climatic conditions during the middle to late Pliocene similar to the present day.
Combined, field-based and isotope geochemical methods used in this study of the NW-Argentine Andes have thus helped to gain insight into the systematics, rate changes, interactions, and temporal characteristics among tectonically controlled deformation patterns, the build-up of topography impacting atmospheric processes, the distribution of rainfall, and resulting surface processes in a tectonically active mountain belt. Ultimately, this information is essential for a better understanding of the style and the rates at which non-collisional mountain belts evolve, including the development orogenic plateaus and their bordering flanks. The results presented in this study emphasize the importance of stable isotope records for paleoaltimetric and paleoenvironmental studies in mountain belts and furnishes important data for a rigorous interpretation of such records.
It has been observed for many African languages that focussed subjects
have to appear outside of their syntactic base position, as opposed to
focussed objects, which can remain in-situ. This is known as subjectobject
asymmetry of focus marking, which Fiedler et al. (2010) claim
to hold also for Akan. Genzel (2013), on the other hand, argues that
Akan does not exhibit a subject-object focus asymmetry. A questionnaire
study and a production experiment were carried out to investigate
whether focussed subjects may indeed be realized in-situ in Akan. The
results suggest that (i) focussed subjects do not have to be obligatorily
realized ex-situ, and that (ii) the syntactic preference for the realization
of a focussed subject highly depends on exhaustivity.
During the course of their ontogenesis plants are continuously exposed to a large variety of abiotic stress factors which can damage tissues and jeopardize the survival of the organism unless properly countered. While animals can simply escape and thus evade stressors, plants as sessile organisms have developed complex strategies to withstand them. When the intensity of a detrimental factor is high, one of the defense programs employed by plants is the induction of programmed cell death (PCD). This is an active, genetically controlled process which is initiated to isolate and remove damaged tissues thereby ensuring the survival of the organism. The mechanism of PCD induction usually includes an increase in the levels of reactive oxygen species (ROS) which are utilized as mediators of the stress signal. Abiotic stress-induced PCD is not only a process of fundamental biological importance, but also of considerable interest to agricultural practice as it has the potential to significantly influence crop yield. Therefore, numerous scientific enterprises have focused on elucidating the mechanisms leading to and controlling PCD in response to adverse conditions in plants. This knowledge may help develop novel strategies to obtain more resilient crop varieties with improved tolerance and enhanced productivity. The aim of the present review is to summarize the recent advances in research on ROS-induced PCD related to abiotic stress and the role of the organelles in the process.
The polymorphism of immunogenes of the major histocompatibility complex (MHC) is thought to influence the functional plasticity of immune responses and, consequently, the fitness of populations facing heterogeneous pathogenic pressures. Here, we evaluated MHC variation (allelic richness and divergence) and patterns of selection acting on the two highly polymorphic MHC class II loci (DRB and DQB) in the endangered primate Madame Berthe's mouse lemur (Microcebus berthae). Using 454 pyrosequencing, we examined MHC variation in a total of 100 individuals sampled over 9 years in Kirindy Forest, Western Madagascar, and compared our findings with data obtained previously for its sympatric congener, the grey mouse lemur (Microcebus murinus). These species exhibit a contrasting ecology and demography that were expected to affect MHC variation and molecular signatures of selection. We found a lower allelic richness concordant with its low population density, but a similar level of allelic divergence and signals of historical selection in the rare feeding specialist M. berthae compared to the widespread generalist M. murinus. These findings suggest that demographic factors may exert a stronger influence than pathogen-driven selection on current levels of allelic richness in M. berthae. Despite a high sequence similarity between the two congeners, contrasting selection patterns detected at DQB suggest its potential functional divergence. This study represents a first step toward unravelling factors influencing the adaptive divergence of MHC genes between closely related but ecologically differentiated sympatric lemurs and opens new questions regarding potential functional discrepancy that would explain contrasting selection patterns detected at DQB.
The objectives of this study were to examine (a) the effect
of dynamic assessment (DA) in a 3D Immersive Virtual Reality
(IVR) environment as compared with computerized 2D and noncomputerized
(NC) situations on cognitive modifiability, and (b) the
transfer effects of these conditions on more difficult problem solving
administered two weeks later in a non-computerized environment. A
sample of 117 children aged 6:6-9:0 years were randomly assigned
into three experimental groups of DA conditions: 3D, 2D, and NC, and
one control group (C). All groups received the pre- and post-teaching
Analogies subtest of the Cognitive Modifiability Battery (CMB-AN).
The experimental groups received a teaching phase in conditions similar
to the pre-and post-teaching phases. The findings showed that cognitive
modifiability, in a 3D IVR, was distinctively higher than in the two
other experimental groups (2D computer group and NC group). It was
also found that the 3D group showed significantly higher performance
in transfer problems than the 2D and NC groups.
Im Rahmen der EU-weiten REACH-Verordnung haben Alternativmethoden zum Tierversuch in der Toxikologie an Bedeutung gewonnen. Die Alternativmethoden gliedern sich auf in In-vitro- und In-silico-Methoden. In dieser Dissertation wurden verschiedene Konzepte der In-silico-Toxikologie behandelt.
Die bearbeiteten Themen reichen von quantitativen Strukturaktivitätsbeziehungen (QSAR) über eine neue Herangehensweise an das gängige Konzept zur Festlegung von Grenzwerten bis hin zu computerbasierten Modellierungen zum Alkohol- und Bisphenol-A-Stoffwechsel.
Das Kapitel über QSAR befasst sich im Wesentlichen mit der Erstellung und Analyse einer Datenbank mit 878 Substanzen, die sich aus Tierversuchsstudien aus dem Archiv des Bundesinstituts für Risikobewertung zusammensetzt. Das Design wurde dabei an eine bereits bestehende Datenbank angepasst, um so einen möglichst großen Datenpool zu generieren. In der Analyse konnte u.a. gezeigt werden, dass Stoffe mit niedrigerem Molekulargewicht ein erhöhtes Potential für toxikologische Schäden aufwiesen als größere Moleküle.
Mit Hilfe des sogenannten TTC-Konzepts können Grenzwerte für Stoffe geringer Exposition festgelegt werden, zu denen keine toxikologischen Daten zur Verfügung stehen. In dieser Arbeit wurden für die Stoffe dreier Datenbanken entsprechende Grenzwerte festgelegt. Es erfolgte zunächst eine gängige strukturbasierte Aufteilung der Substanzen in die Kategorien "nicht toxisch", "möglicherweise toxisch" und "eindeutig toxisch". Substanzen, die aufgrund ihrer Struktur in eine der drei Klassen eingeordnet werden, erhalten den entsprechenden Grenzwert. Da in die dritte Klasse auch Stoffe eingeordnet werden, deren Toxizität nicht bestimmbar ist, ist sie sehr groß. Daher wurden in dieser Arbeit die ersten beiden Klassen zusammengelgt, um einen größeren Datenpool zu ermöglichen. Eine weitere Neuerung umfasst die Erstellung eines internen Grenzwerts. Diese Vorgehensweise hat den Vorteil, dass der Expositionsweg herausgerechnet wird und somit beispielsweise Studien mit oraler Verabreichung mit Studien dermaler Verabreichung verglichen werden können.
Mittels physiologisch basiertem kinetischem Modelling ist es möglich, Vorgänge im menschlichen Körper mit Hilfe spezieller Software nachzuvollziehen. Durch diese Vorgehensweise können Expositionen von Chemikalien simuliert werden. In einem Teil der Arbeit wurden Alkoholexpositionen von gestillten Neugeborenen simuliert, deren Mütter unmittelbar zuvor alkoholische Getränke konsumiert hatten. Mit dem Modell konnte gezeigt werden, dass die Expositionen des Kindes durchweg gering waren. Nach einem Glas Wein wurden Spitzenkonzentrationen im Blut von Neugeborenen von 0,0034 Promille ermittelt. Zum Vergleich wurde die Exposition durch ein für Säuglinge zugelassenes alkoholhaltiges pflanzliches Arzneimittel simuliert. Hier wurden Spitzenkonzentrationen von 0,0141 Promille erreicht. Daher scheinen Empfehlungen wie gelegentlicher Konsum ohne schädigende Wirkung auf das Kind wissenschaftlich fundiert zu sein.
Ein weiteres Kinetik-Modell befasste sich mit dem Stoffwechsel von Bisphenol A. Teils widersprüchliche Daten zur Belastung mit BPA in der wissenschaftlichen Literatur führen wiederholt zu Anregungen, den Grenzwert der Chemikalie anzupassen. Die Funktionalität der am Metabolismus beteiligten Enzyme kann je nach Individuum unterschiedlich ausgeprägt sein. Mittels Modellings konnte hier gezeigt werden, dass dies maßgeblich dazu führt, dass sich berechnete Plasmaspiegel von Individuen bis zu 4,7-fach unterscheiden.
Die Arbeit konnte somit einen Beitrag zur Nutzung und Weiterentwicklung von In-silico-Modellen für diverse toxikologische Fragestellungen leisten.
Plasma convection in the high-latitude ionosphere provides important information about magnetosphere-ionosphere-thermosphere coupling. In this study we estimate the along-track component of plasma convection within and around the polar cap, using electron density profiles measured by the three Swarm satellites. The velocity values estimated from the two different satellite pairs agree with each other. In both hemispheres the estimated velocity is generally anti-sunward, especially for higher speeds. The obtained velocity is in qualitative agreement with Super Dual Auroral Radar Network data. Our method can supplement currently available instruments for ionospheric plasma velocity measurements, especially in cases where these traditional instruments suffer from their inherent limitations. Also, the method can be generalized to other satellite constellations carrying electron density probes.
Peak oil is forcing our society to shift from fossil to renewable resources. However, such renewable resources are also scarce, and they too must be used in the most efficient and sustainable way possible. Biorefining is a concept that represents both resource efficiency and sustainability. This approach initiates a cascade use, which means food and feed production before material use, and an energy-related use at the end of the value-added chain. However, sustainability should already start in the fields, on the agricultural side, where the industrially-used biomass is produced. Therefore, the aim of my doctoral thesis is to analyse the sustainable feedstock supply for biorefineries. In contrast to most studies on biorefineries, I focus on the sustainable provision of feedstock and not on the bioengineering processing of whatever feedstock is available.
Grasslands provide a high biomass potential. They are often inefficiently used, so a new utilisation concept based on the biorefining approach can increase the added value from grasslands. Fodder legumes from temporary and permanent grasslands were chosen for this study. Previous research shows that they are a promising feedstock for industrial uses, and their positive environmental impact is an important byproduct to promote sustainable agricultural production systems.
Green Biorefineries are a class of biorefineries that use fresh green biomass, such as grasses or fodder legumes, as feedstock. After fractionation, an organic solution (press juice) forms; this is used for the production of organic acids, chemicals and extracts, as well as fertilisers. A fibre component (press cake) is also created to produce feed, biomaterials and biogas. This thesis examines a specific value chain, using alfalfa and clover/grass as feedstock and generating lactic acid and one type of cattle feed from it. The research question is if biomass production needs to be adapted for the utilisation of fodder legumes in the Green Biorefinery approach. I have attempted to give a holistic analysis of cultivation, processing and utilisation of two specific grassland crops. Field trials with alfalfa and clover/grass at different study sites were carried out to obtain information on biomass quality and quantity depending on the crop, study site and harvest time. The fresh biomass was fractionated with a screw press and the composition of press juices and cakes was analysed. Fermentation experiments took place to determine the usability of press juices for lactic acid production. The harvest time is not of high importance for the quality of press juices as a fermentation medium. For permanent grasslands, late cuts, often needed for reasons of nature conservation, are possible without a major influence on feedstock quality. The press cakes were silaged for feed-value determination.
Following evidence that both intermediate products are suitable feedstocks in the Green Biorefinery approach, I developed a cost-benefit analysis, comparing different production scenarios on a farm. Two standard crop rotations for Brandenburg, producing either only market crops or market crops and fodder legumes for ruminant feed production, were compared to a system that uses the cultivated fodder legumes for the Green Biorefinery value chain instead of only feed production. Timely processing of the raw material is important to maintain quality for industrial uses, so on-site processing at the farm is assumed in Green Biorefinery scenario. As a result, more added value stays in the rural area. Two farm sizes, common for many European regions, were chosen to examine the influence of scale. The cost site of farmers has also been analysed in detail to assess which farm characteristics make production of press juices for biochemical industries viable. Results show that for large farm sizes in particular, the potential profits are high. Additionally, the wider spectrum of marketable products generates new sources of income for farmers.
The holistic analysis of the supply chain provides evidence that the cultivation processes for fodder legumes do not need to be adapted for use in Green Biorefineries. In fact, the new utilisation approach even widens the cultivation and processing spectrum and can increase economic viability of fodder legume production in conventional farming.
We report the results from two experiments investigating how referential context information affects native and non-native readers’ interpretation of ambiguous relative clauses in sentences such as The journalist interviewed the assistant of the inspector who was looking very serious. The preceding discourse context was manipulated such that it provided two potential referents for either the first (the assistant) or the second (the inspector) of the two noun phrases that could potentially host the relative clause, thus biasing towards either an NP1 or an NP2 modification reading. The results from an offline comprehension task indicate that both native English speakers’ and German and Chinese-speaking ESL learners’ ultimate interpretation preferences were reliably influenced by the type of referential context. In contrast, in a corresponding self-paced-reading task we found that referential context information modulated only the non-native participants’ disambiguation preferences but not the native speakers’. Our results corroborate and extend previous findings suggesting that non-native comprehenders’ initial analysis of structurally ambiguous input is strongly influenced by biasing discourse information.
