Aus den Vorbemerkungen: In Deutschland werden kommunale Aufgaben traditionell auf zwei unterschiedlichen kommunalen Ebenen erbracht: der Ebene der Kreisverwaltung und der Ebene der einzelnen Gemeinden (in Brandenburg: Städte und Ämter). Angesichts der in Brandenburg teilweise recht dünnen Besiedlung des ländlichen Raumes und der daraus resultierenden kleinteiligen Gemeindestrukturen werden eine größere Zahl von Aufgaben, die für Bürger in deren Alltag durchaus relevant sind, durch die Kreisverwaltungen erbracht (Beispiele sind: Sozial- und Jugendhilfe, Kfz-Zulassung, Baugenehmigungsverfahren). Die Nutzung dieser Leistungen ist für die Bürger teilweise mit erheblichen Wegen und hohem Zeitaufwand verbunden. Daher wird in den Kommunalwissenschaften seit einiger Zeit der Gedanke einer zumindest teilweisen Integration der Leistungserbringung von Kreis- und Gemeindeverwaltungen diskutiert. Grundidee dabei: die Bürger sollen wohnortnah – z.B. in ihrem Amt – „unter einem Dach“ Dienste des Kreises ebenso wie die des Amtes beantragen und entgegennehmen können. Dies kann durch Übertragung von Aufgaben vom Kreis auf die Amts- oder Stadtverwaltung oder durch gemeinsame Leistungserbringung durch Kreis und Amt erreicht werden. Hier können mithilfe der Möglichkeiten von eGovernment klassische Grenzen überwunden werden. Das setzt voraus, dass eGovernment nicht nur auf die Schnittstelle zwischen der Verwaltung und ihrer Adressaten reduziert wird. Vielmehrbeinhaltet eGovernment auch ein Gestaltungspotenzial zur Reorganisation der internen Verwaltungsprozesse und Leistungsorganisation. Wichtiges Ziel des Landkreises Potsdam-Mittelmark ist es, unter Ausnutzung des IT-Gestaltungspotenzials Leistungen des Kreises vor Ort in den Gemeinden anzubieten. Leitbild dieser Neustrukturierung stellt das Konzept der integrierten Kommunalverwaltung dar, das der Kreistag im Jahr 2000 mit einem umfassenden Modernisierungsleitbild verabschiedet hat. Durch eine integrierte Kommunalverwaltung soll die oben erwähnte kommunale Zweigliederung (Kreis- und Gemeindeverwaltung) aus der Sicht des Bürgers reduziert werden. Der Landkreis Potsdam-Mittelmark will das technische Gestaltungspotenzial von eGovernment nutzen, um häufig nachgefragte Leistungen des Kreises auf die kreisangehörigen Gemeinden zu übertragen. Dazu arbeitet der Kreis mit den vier kreisangehörigen Pilotgemeinden Beelitz, Rehbrücke, Stahnsdorf und Wiesenburg in gemeinsamen Arbeitsgruppen zusammen.
Die Organisation der öffentlichen Aufgabenerfüllung ist in Deutschland in den letzten Jahren unter Einfluss von zunehmendem internationalen Wettbewerb der Wirtschaftsregionen, Europäisierung, allgemein sinkenden öffentlichen Einnahmen und vor allem der negativen demographischen Entwicklung wieder verstärkt unter Modernisierungsdruck geraten. Angesichts lahmender Binnenkonjuktur, Verlagerung von Produktionsstätten ins Ausland, steigender Arbeitslosigkeit sowie Bevölkerungstrukturkrise ist wohl in fast allen Teilen der Gesellschaft die Einsicht in die Notwendigkeit einer grundlegenden Modifizierung der Organisation von öffentlicher Aufgabenwahrnehmung vorhanden. Auch die Ausgestaltung der kommunalen Selbstverwaltung als Fundament der staatlichen Ordnung und konkret örtlicher Wirkungsbereich politisch-administrativen Handelns ist wieder verstärkt ins Zentrum von grundlegenden Reformüberlegungen geraten. Die Neujustierung des öffentlichen Zuständigkeitsgefüges in den Ländern sowie die äußere und innere Architektur der kommunalen Gebietskörperschaften werden erneut diskutiert. Immer mehr Bundesländer planen umfangreiche Modernisierungsmaßnahmen ihres Verwaltungsgefüges. Von großem Interesse ist deshalb die Frage, inwieweit durch Optimierung der Zusammenarbeit von Kreisen die überörtliche Aufgabenwahrnehmung gestärkt werden kann und welche Rahmenbedingungen dabei beachtet werden müssen. Die bestehenden öffentlich-rechtlichen Handlungsformen der interkommunalen Kooperation stammen überwiegend noch aus dem 19. Jahrhundert. Als sich das Kommunalwissenschaftliche Institut diesem Untersuchungsgegenstand im Jahre 2004 zuwandte, gab es nur wenige empirische Studien und Publikationen zu diesem Thema. Zwischenzeitlich hat die Debatte zu diesem Untersuchungsgegenstand in der Fachöffentlichkeit vor allem aus Gründen des europäischem Wettbewerbsrechtes, aber auch wegen des zunehmenden Handlungsdruckes zugenommen. Unstrittig ist insoweit, dass auf diesem Feld Modernisierungsbedarf besteht (bspw. Einführung gemeinsamer Kommunalunternehmen als Anstalten des öffentlichen Rechtes). Nach einer langen Rücklauf-, Auswertungs- und Studienerstellungsphase liegen erste Ergebnisse des Projektes vor. Ohne die wertvolle Unterstützung und den fachkundigen Rat von Vertretern vom Deutschen und Brandenburgischen Landkreistag sowie der Wissenschaft wäre diese Studie nicht zustande gekommen. Besonderer Dank gilt insofern dem Engagement von Prof. Nierhaus, Prof. em. Reichard, Dr. Schliesky, Dr. Rüge, Dr. Humpert und den Beteiligten aus den befragten Landkreisen bzw. kreisfreien Städten für das Forschungsprojekt. Das Autorenteam hofft, dass diese Studie mit der Förderung des Wissensstandes über die Kreiskooperation einen Beitrag für die Diskussion in der Fachöffentlichkeit und in der Verwaltungspraxis leistet.

Das zurückliegende Jahr stand an der Universität Potsdam auch im Zeichen des zwanzigjährigen Jubiläums der Hochschule. Am 15. Juli 1991, wurde sie gegründet und während einer Festwoche feierten Professorinnen und Professoren, Mitarbeiterinnen, Mitarbeiter und Studierende dieses Jubiläum gebührend. Seit der Gründung der größten brandenburgischen Hochschule sind wissenschaftliches Renommee, Ansehen und Attraktivität stetig gewachsen. Gerade in den letzten Jahren hat sie ihr Profil geschärft. Vor allem die Kognitions-, die Geo- und Biowissenschaften sind hier zu nennen. Aber auch die Lehrerbildung besitzt einen hohen Stellenwert. International anerkannte Forschungsbereiche, Wissenschaftspreise, eine erfolgreiche Drittmittelbilanz und nicht zuletzt die bauliche Entwicklung an allen drei Standorten sind sichtbare Indikatoren für die erfolgreiche Entwicklung, die die Universität Potsdam in den letzten zwei Jahrzehnten durchlaufen hat. Die drei ehemaligen Präsidenten sowie verschiedene andere Protagonisten werfen in dieser Ausgabe der Portal Alumni einen Blick auf unterschiedliche Aspekte der zurückliegenden Entwicklung der Universität. Vom Erfolg der Universität zeugt auch die wachsende Zahl der Absolventinnen und Absolventen, die die Universität verlassen. Portal Alumni stellt in der vorliegenden Ausgabe deshalb Absolventen und deren universitäre und berufliche Lebenswege genauer vor und lässt damit zugleich kaleidoskopartig 20 Jahre Studium an der Universität Potsdam Revue passieren.
Solid organ transplant (SOT) recipients receive therapeutic immunosuppression that compromises their immune response to infections and vaccines. For this reason, SOT patients have a high risk of developing severe coronavirus disease 2019 (COVID-19) and an increased risk of death from severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) infection. Moreover, the efficiency of immunotherapies and vaccines is reduced due to the constant immunosuppression in this patient group. Here, we propose adoptive transfer of SARS-CoV-2-specific T cells made resistant to a common immunosuppressant, tacrolimus, for optimized performance in the immunosuppressed patient. Using a ribonucleoprotein approach of CRISPR-Cas9 technology, we have generated tacrolimus-resistant SARS-CoV-2-specific T cell products from convalescent donors and demonstrate their specificity and function through characterizations at the single-cell level, including flow cytometry, single-cell RNA (scRNA) Cellular Indexing of Transcriptomes and Epitopes (CITE), and T cell receptor (TCR) sequencing analyses. Based on the promising results, we aim for clinical validation of this approach in transplant recipients. Additionally, we propose a combinatory approach with tacrolimus, to prevent an overshooting immune response manifested as bystander T cell activation in the setting of severe COVID-19 immunopathology, and tacrolimus-resistant SARS-CoV-2-specific T cell products, allowing for efficient clearance of viral infection. Our strategy has the potential to prevent severe COVID-19 courses in SOT or autoimmunity settings and to prevent immunopathology while providing viral clearance in severe non-transplant COVID-19 cases.
Land-use intensification is a major driver of biodiversity loss(1,2). Alongside reductions in local species diversity, biotic homogenization at larger spatial scales is of great concern for conservation. Biotic homogenization means a decrease in beta-diversity (the compositional dissimilarity between sites). Most studies have investigated losses in local (alpha)-diversity(1,3) and neglected biodiversity loss at larger spatial scales. Studies addressing beta-diversity have focused on single or a few organism groups (for example, ref. 4), and it is thus unknown whether land-use intensification homogenizes communities at different trophic levels, above-and belowground. Here we show that even moderate increases in local land-use intensity (LUI) cause biotic homogenization across microbial, plant and animal groups, both above- and belowground, and that this is largely independent of changes in alpha-diversity. We analysed a unique grassland biodiversity dataset, with abundances of more than 4,000 species belonging to 12 trophic groups. LUI, and, in particular, high mowing intensity, had consistent effects on beta-diversity across groups, causing a homogenization of soil microbial, fungal pathogen, plant and arthropod communities. These effects were nonlinear and the strongest declines in beta-diversity occurred in the transition from extensively managed to intermediate intensity grassland. LUI tended to reduce local alpha-diversity in aboveground groups, whereas the alpha-diversity increased in belowground groups. Correlations between the alpha-diversity of different groups, particularly between plants and their consumers, became weaker at high LUI. This suggests a loss of specialist species and is further evidence for biotic homogenization. The consistently negative effects of LUI on landscape-scale biodiversity underscore the high value of extensively managed grasslands for conserving multitrophic biodiversity and ecosystem service provision. Indeed, biotic homogenization rather than local diversity loss could prove to be the most substantial consequence of land-use intensification.
Although temporal heterogeneity is a well-accepted driver of biodiversity, effects of interannual variation in land-use intensity (LUI) have not been addressed yet. Additionally, responses to land use can differ greatly among different organisms; therefore, overall effects of land-use on total local biodiversity are hardly known. To test for effects of LUI (quantified as the combined intensity of fertilization, grazing, and mowing) and interannual variation in LUI (SD in LUI across time), we introduce a unique measure of whole-ecosystem biodiversity, multidiversity. This synthesizes individual diversity measures across up to 49 taxonomic groups of plants, animals, fungi, and bacteria from 150 grasslands. Multidiversity declined with increasing LUI among grasslands, particularly for rarer species and aboveground organisms, whereas common species and belowground groups were less sensitive. However, a high level of interannual variation in LUI increased overall multidiversity at low LUI and was even more beneficial for rarer species because it slowed the rate at which the multidiversity of rare species declined with increasing LUI. In more intensively managed grasslands, the diversity of rarer species was, on average, 18% of the maximum diversity across all grasslands when LUI was static over time but increased to 31% of the maximum when LUI changed maximally over time. In addition to decreasing overall LUI, we suggest varying LUI across years as a complementary strategy to promote biodiversity conservation.
Land-use intensification is a key driver of biodiversity change. However, little is known about how it alters relationships between the diversities of different taxonomic groups, which are often correlated due to shared environmental drivers and trophic interactions. Using data from 150 grassland sites, we examined how land-use intensification (increased fertilization, higher livestock densities, and increased mowing frequency) altered correlations between the species richness of 15 plant, invertebrate, and vertebrate taxa. We found that 54% of pairwise correlations between taxonomic groups were significant and positive among all grasslands, while only one was negative. Higher land-use intensity substantially weakened these correlations(35% decrease in rand 43% fewer significant pairwise correlations at high intensity), a pattern which may emerge as a result of biodiversity declines and the breakdown of specialized relationships in these conditions. Nevertheless, some groups (Coleoptera, Heteroptera, Hymenoptera and Orthoptera) were consistently correlated with multidiversity, an aggregate measure of total biodiversity comprised of the standardized diversities of multiple taxa, at both high and lowland-use intensity. The form of intensification was also important; increased fertilization and mowing frequency typically weakened plant-plant and plant-primary consumer correlations, whereas grazing intensification did not. This may reflect decreased habitat heterogeneity under mowing and fertilization and increased habitat heterogeneity under grazing. While these results urge caution in using certain taxonomic groups to monitor impacts of agricultural management on biodiversity, they also suggest that the diversities of some groups are reasonably robust indicators of total biodiversity across a range of conditions.
