Effekte einer reduzierten Dosis von Pflanzenschutzmitteln auf tritrophische Systeme im Ackerbau

Chemische Pflanzenschutzmittel (PSM) bekämpfen nicht nur Schadorganismen, sondern haben aufgrund ihrer hohen Toxizität auch negative Auswirkungen auf Nicht-Ziel-Organismen. Die Fragestellung der Arbeit war es, ob mit reduzierten Anwendungen von PSM ihr Gefährdungspotenzial für Prädatoren von Schädlingen verringert und dadurch das Potenzial der natürlichen Schädlingsregulation erhöht wird. In dreijährigen Freilanduntersuchungen wurden die Effekte einer dauerhaft reduzierten Dosis von chemischen PSM auf die ökologische Situation im Ackerbau anhand von drei Fallbeispielen in einem konventionell bewirtschafteten Betrieb in der Magdeburger Börde untersucht. Drei über 15 ha große Felder wurden dauerhaft in zwei Teilflächen geteilt, wobei eine Teilfläche mit der vom Landwirt gewünschten Dosis (100 %-Variante) und die andere mit jeweils genau der halben Dosis (50 %-Variante) behandelt wurde. Mittels dieser Halbfelder-Vergleiche wurden die ökologischen Situationen bezüglich des Auftretens von Blattläusen und ihren Prädatoren sowie Unkräutern vor und nach der jeweiligen PSM-Behandlung aufgenommen und ökonomische Parameter ermittelt. Ergänzend wurden im Labor Modellgefäßversuche mit abgestuften Dosierungen von Insektiziden und Herbiziden durchgeführt. Die Insektizidbehandlung übte einen großen Einfluss auf die Blattläuse und ihre Prädatoren aus, während alle vorherigen Herbizid- und Fungizidbehandlungen zu keinen Unterschieden in der Abundanz der Blattläuse und ihrer Prädatoren zwischen beiden Varianten hervorriefen. Die reduzierte Insektiziddosis führte zu keiner guten Blattlauskontrolle, während die Abundanz der blattlausspezifischen Prädatoren positiv beeinflusst wurde. Die Araneae reagierten auf die reduzierte Dosis mit einer teilweise erhöhten Aktivitätsdichte und Artendiversität. Dagegen waren diesbezüglich keine eindeutigen Effekte auf die Carabidae festzustellen. Es traten keine strukturellen Veränderungen in Form einer erhöhten Unkrautdichte durch die reduzierte Herbiziddosis auf. Erste Hinweise auf mögliche langfristige Auswirkungen einer dauerhaft reduzierten PSM-Anwendung konnten nur bei der Verunkrautung und der Aktivitätsdichte der Araneae beobachtet werden. Blattläuse profitierten demnach mehr von der reduzierten Anwendung der PSM als ihre Prädatoren, so dass zwar das Potenzial der natürlichen Blattlausregulation erhöht, die Selbstregulation aber nicht verbessert wurde. Die geschonten Prädatoren schafften es nicht, die vorhandene Restpopulation der Blattläuse zu reduzieren. Dagegen konnte in den Laborversuchen gezeigt werden, das schon bei deutlich reduzierten Insektiziddosen eine ausreichende Blattlausbekämpfung möglich ist und eine weitere Einsparung durch Ausnutzung der natürlichen Regulation durch Prädatoren erreicht werden kann. Allerdings ist eine Übertragung der Ergebnisse von Laboruntersuchungen auf Freilandbedingungen schwierig. Es kann zu einer Überschätzung der Prädatorleistung führen.
Biogene Amine sind eine Substanzklasse, die bei Vertebraten und Invertebraten eine wichtige Komponente des endokrinen Systems darstellen. Sie binden an spezifische Rezeptoren der Gruppe der G-Protein gekoppelten Rezeptoren. In dieser Arbeit wurden zwei neue Rezeptoren aus der Schabe Periplaneta americana kloniert. Durch verschiedene Ansätze konnten zwei vollständige cDNA-Sequenzen isoliert werden. Die Aminosäuresequenzen weisen die größte Ähnlichkeit zu bereits bekannten Tyraminrezeptoren aus Locusta/Bombyx bzw. zu Dopaminrezeptoren aus Apis/Drosophila auf. Entsprechend wurden diese Rezeptoren Pea (P. americana) TYR1 und PeaDOP2 genannt. Deutliche Hinweise auf ihre Funktion lassen sich an den abgeleiteten Aminosäuresequenzen ablesen. Aminosäuren, die wichtig bei der Bildung der Bindungstasche, der Rezeptoraktivierung und der Kopplung eines G-Proteins sind, treten bei beiden Rezeptoren auf. Sequenzalignments stellen die Rezeptoren in die Gruppe anderer Tyraminrezeptoren bzw. der Invertebraten-Typ Dopaminrezeptoren. Das Transkript der beiden Rezeptoren konnte durch RT-PCR in verschiedenen Geweben nachgewiesen werden. Ein Ziel der Arbeit war die Gewinnung eines polyklonalen Antiserums gegen PeaTYR1. Dieses Serum detektiert im Homogenat von Gehirnen mehrere Banden, darunter auch eine mit der kalkulierten Masse von PeaTYR1. Präabsorption des Serums mit dem Peptid, welches zur Reinigung verwendet wurde, zeigt dessen Spezifität. An Gehirnschnitten markiert das Serum große Teile des Protocerebrums aber auch feinere Strukturen der Antennalloben, der optischen Loben und des Zentralkomplexes. Ein weiteres Serum gegen Tyramin führte zu einer Markierung von mehreren Neuronengruppen, welche sich in die optischen Loben und den Zentralkomplex verzweigen. Der αPeaTYR1-CPL3 Antikörper markierte die Plasmamembran von transfizierten HEK293-Zellen. Die Lokalisierung von Rezeptor und Ligand deuten darauf hin, dass Tyramin die optische und olfaktorische Wahrnehmung beeinflussen könnte.
Being living systems unable to adjust their location to changing environmental conditions, plants display homeostatic networks that have evolved to maintain transition metal levels in a very narrow concentration range in order to avoid either deficiency or toxicity. Hence, plants possess a broad repertoire of mechanisms for the cellular uptake, compartmentation and efflux, as well as for the chelation of transition metal ions. A small number of plants are hypertolerant to one or a few specific transition metals. Some metal tolerant plants are also able to hyperaccumulate metal ions. The Brassicaceae family member Arabidopis halleri ssp. halleri (L.) O´KANE and AL´SHEHBAZ is a hyperaccumulator of zinc (Zn), and it is closely related to the non-hypertolerant and non-hyperaccumulating model plant Arabidopsis thaliana (L.) HEYNHOLD. The close relationship renders A. halleri a promising emerging model plant for the comparative investigation of the molecular mechanisms behind hypertolerance and hyperaccumulation. Among several potential candidate genes that are probably involved in mediating the zinc-hypertolerant and zinc-hyperaccumulating trait is AhHMA3. The AhHMA3 gene is highly similar to AtHMA3 (AGI number: At4g30120) in A. thaliana, and its encoded protein belongs to the P-type IB ATPase family of integral membrane transporter proteins that transport transition metals. In contrast to the low AtHMA3 transcript levels in A. thaliana, the gene was found to be constitutively highly expressed across different Zn treatments in A. halleri, especially in shoots. In this study, the cloning and characterisation of the HMA3 gene and its promoter from Arabidopsis halleri (L.) O´KANE and AL´SHEHBAZ and Arabidopsis thaliana (L.) HEYNHOLD is described. Heterologously expressed AhHMA3 mediated enhanced tolerance to Zn and to a much lesser degree to cadmium (Cd) but not to cobalt (Co) in metal-sensitive mutant strains of budding yeast. It is demonstrated that the genome of A. halleri contains at least four copies of AhHMA3, AhHMA3-1 to AhHMA3-4. A copy-specific real-time RT-PCR indicated that an AhHMA3-1 related gene copy is the source of the constitutively high transcript level in A. halleri and not a gene copy similar to AhHMA3-2 or AhHMA3-4. In accordance with the enhanced AtHMA3mRNA transcript level in A. thaliana roots, an AtHMA3 promoter-GUS gene construct mediated GUS activity predominantly in the vascular tissues of roots and not in shoots. However, the observed AhHMA3-1 and AhHMA3-2 promoter-mediated GUS activity in A. thaliana or A. halleri plants did not reflect the constitutively high expression of AhHMA3 in shoots of A. halleri. It is suggested that other factors e. g. characteristic sequence inserts within the first intron of AhHMA3-1 might enable a constitutively high expression. Moreover, the unknown promoter of the AhHMA3-3 gene copy could be the source of the constitutively high AhHMA3 transcript levels in A. halleri. In that case, the AhHMA3-3 sequence is predicted to be highly homologous to AhHMA3-1. The lack of solid localisation data for the AhHMA3 protein prevents a clear functional assignment. The provided data suggest several possible functions of the AhHMA3 protein: Like AtHMA2 and AtHMA4 it might be localised to the plasma membrane and could contribute to the efficient translocation of Zn from root to shoot and/or to the cell-to-cell distribution of Zn in the shoot. If localised to the vacuolar membrane, then a role in maintaining a low cytoplasmic zinc concentration by vacuolar zinc sequestration is possible. In addition, AhHMA3 might be involved in the delivery of zinc ions to trichomes and mesophyll leaf cells that are major zinc storage sites in A. halleri.
Im Rahmen der Untersuchung zur Entwicklung der pelagischen Gemeinschaft des ehemals sauren Tagebauseenkomplexes Goitsche (pH~3) während dessen Flutung und Neutralisierung wurden Wechselwirkungen zwischen der Zusammensetzung von Organismengemeinschaften und der Variabilität des abiotischen Umfeldes untersucht.Im Mittelpunkt standen zwei von ihrer Kausalität her unterschiedliche Aspekte: • Der erste Aspekt betraf die Reifung von Ökosystemen: War der Reifungsprozess von pelagischen Gemeinschaften anhand der von Odum (1969) formulierten Kriterien zur Energetik der Gemeinschaft, zu den Nährstoffkreisläufen sowie zu strukturellen Merkmalen auf Ökosystem- und Individuenebene zu erfassen? Führten der physiologische Stress durch den niedrigen pH und physikalische Störungen der Schichtung durch das einströmende Flutungswasser zu einer Umkehr des Reifungsprozesses? Auf welchen Organisationsebenen der Lebensgemeinschaften waren die Auswirkungen dieser Stressoren erkennbar? • Der zweite Aspekt behandelte die Entwicklung der Artenzahl, die Gleichverteilung der Dominanz von Arten (Eveness) und die Diversität von Planktongemeinschaften entlang des Produktivitätsgradienten. Speziell wurde untersucht, ob die Artenzahl und die Diversität eine monoton positive oder eine unimodale Funktion der Produktivität waren und ob die Eveness eine monoton abnehmende Funktion der Produktivität war. Zur besseren Vorhersagbarkeit der Entwicklung dieser Indizes wurden in einem nächsten Schritt zusätzliche biotische und abiotische Faktoren (z.B. Konsumenteneffekte, physikalische Störung, Immigration) berücksichtigt. Zuletzt wurde die Hypothese getestet, dass unter dem Einfluss von extremem physiologischem Stress keine Abhängigkeit zwischen den betrachteten Indizes und der Produktivität von Ökosystemen besteht. Die Untersuchungen der vorliegenden Arbeit führten zu folgenden Ergebnissen und Schlussfolgerungen: 1) Der Reifungsprozess der Planktongemeinschaft war unter neutralen Bedingungen nicht eindeutig an einzelnen Kriterien festzumachen. Vielmehr schienen idiosynkratische Effekte einzelner Arten auf die Zusammensetzung und Funktion der Organismengemeinschaft von Bedeutung. Coenobiumbildende Kieselalgen sowie größere Cladoceren und Copepoden dominierten sehr rasch die Planktongemeinschaft und vermittelten den Eindruck eines reifen Ökosystems fast unmittelbar nach der Neutralisierung des Tagebausees. 2) Der Einfluss von physiologischem Säurestress und physikalischer Störung der Schichtung durch das eintretende Flutungswasser war gegenüber einem neutralen, ungestörten Teilbecken des Tagebausees (Referenzzustand) eindeutig zu erkennen. Die isolierte Betrachtung der Wirkung der Stressoren lieferte hinsichtlich fast aller Kriterien Anzeichen einer Verjüngung des Systems sensu Odum (1969, 1985). 3) Im betrachteten Ökosystem existierte eine Hierarchie innerhalb der Stressoren. Der Einfluss des Säurestresses dominierte gegenüber dem Einfluss der physikalischen Störung, wahrscheinlich indem er die Reaktionsmöglichkeiten der Planktongemeinschaft einschränkte. 4) Für Primärproduzenten war die Artenzahl eine monoton positive Funktion der realisierten Biomasse (einem Surrogatparameter für die Produktivität des Systems). Die Eveness war eine monoton negative Funktion der Produktivität. Die beobachtete unimodale Beziehung zwischen der Diversität und der Produktivität der Primärproduzenten muss als eine Folge der mathematischen Formulierung dieser Indizes betrachtet werden. 5) Die Ergebnisse multivariater Modelle zur Vorhersage der Artenzahl und der Eveness der Primärproduzenten in Abhängigkeit zusätzlicher erfassbarer biotischer und abiotischer Faktoren ermöglichten eine differenziertere Betrachtung der Ergebnisse: • Im Tagebausee Goitsche war die Eveness hauptsächlich von dichteabhängigen Prozessen gesteuert (negative Abhängigkeit zum Quadrat der Biomassen und zu den Lichtverhältnissen). • Die Entwicklung der Artenzahl war neben dem primären Einfluss der zunehmenden Biomasse auch durch qualitative und quantitative Aspekte der Konsumentengemeinschaft (Diversität und Biomasse des Zooplanktons) beeinflusst. Der Einfluss einer erhöhten Immigration auf die Artenzahl wurde nur zu Beginn der Flutung des Tagebausees beobachtet. 6) Auf Ebene der Konsumenten war die einzige eindeutig feststellbare Abhängigkeit ein Anstieg der Artenzahl mit steigender Biomasse. Das Fehlen von weiteren Beziehungen zwischen Diversitätsindizes und dem Produktivitätsgradienten wird darauf zurückgeführt, dass auf den unteren trophischen Ebenen der Primärproduzenten Ressourceneffekte (Bottom-Up) stärker ausgeprägt sind, wohingegen auf höheren trophischen Ebenen Konsumenteneffekte (Top-Down) dominieren. 7) In durch physiologischen Stress beeinflussten Systemen bestand keine Abhängigkeit zwischen den Diversitätsindizes (Artenzahl, Eveness und Diversität) und der Produktivität.
The need to develop sustainable resource management strategies for semi-arid and arid rangelands is acute as non-adapted grazing strategies lead to irreversible environmental problems such as desertification and associated loss of economic support to society. In such vulnerable ecosystems, successful implementation of sustainable management strategies depends on well-founded under-standing of processes at different scales that underlay the complex system dynamic. There is ample evidence that, in contrast to traditional sectoral approaches, only interdisciplinary research does work for resolving problems in conservation and natural resource management. In this thesis I combined a range of modeling approaches that integrate different disciplines and spatial scales in order to contribute to basic guidelines for sustainable management of semi-arid and arid range-lands. Since water availability and livestock management are seen as most potent determinants for the dynamics of semi-arid and arid ecosystems I focused on (i) the interaction of ecological and hydro-logical processes and (ii) the effect of farming strategies. First, I developed a grid-based and small-scaled model simulating vegetation dynamics and inter-linked hydrological processes. The simulation results suggest that ecohydrological interactions gain importance in rangelands with ascending slope where vegetation cover serves to obstruct run-off and decreases evaporation from the soil. Disturbances like overgrazing influence these positive feedback mechanisms by affecting vegetation cover and composition. In the second part, I present a modeling approach that has the power to transfer and integrate ecological information from the small scale vegetation model to the landscape scale, most relevant for the conservation of biodiversity and sustainable management of natural resources. I combined techniques of stochastic modeling with remotely sensed data and GIS to investigate to which ex-tent spatial interactions, like the movement of surface water by run-off in water limited environments, affect ecosystem functioning at the landscape scale. My simulation experiments show that overgrazing decreases the number of vegetation patches that act as hydrological sinks and run-off increases. The results of both simulation models implicate that different vegetation types should not only be regarded as provider of forage production but also as regulator of ecosystem functioning. Vegetation patches with good cover of perennial vegetation are capable to catch and conserve surface run-off from degraded surrounding areas. Therefore, downstream out of the simulated system is prevented and efficient use of water resources is guaranteed at all times. This consequence also applies to commercial rotational grazing strategies for semi-arid and arid rangelands with ascending slope where non-degraded paddocks act as hydrological sinks. Finally, by the help of an integrated ecological-economic modeling approach, I analyzed the relevance of farmers’ ecological knowledge for longterm functioning of semi-arid and arid grazing systems under current and future climatic conditions. The modeling approach consists of an ecological and an economic module and combines relevant processes on either level. Again, vegetation dynamics and forage productivity is derived by the small-scaled vegetation model. I showed that sustainable management of semi-arid and arid rangelands relies strongly on the farmers’ knowledge on how the ecosystem works. Furthermore, my simulation results indicate that the projected lower annual rainfall due to climate change in combination with non-adapted grazing strategies adds an additional layer of risk to these ecosystems that are already prone to land degradation. All simulation models focus on the most essential factors and ignore specific details. Therefore, even though all simulation models are parameterized for a specific dwarf shrub savanna in arid southern Namibia, the conclusions drawn are applicable for semi-arid and arid rangelands in general.
Sulphur, a macronutrient essential for plant growth, is among the most versatile elements in living organisms. Unfortunately, little is known about regulation of sulphate uptake and assimilation by plants. Identification of sulphate signalling processes will allow to control sulphate acquisition and assimilation and may prove useful in the future to improve sulphur-use efficiency in agriculture. Many of genes involved in sulphate metabolism are regulated on transcriptional level by products of other genes called transcription factors (TF). Several published experiments revealed TF genes that respond to sulphate deprivation, but none of these have been so far been characterized functionally. Thus, we aimed at identifying and characterising transcription factors that control sulphate metabolism in the model plant Arabidopsis thaliana. To achieve that goal we postulated that factors regulating Arabidopsis responses to inorganic sulphate deficiency change their transcriptional levels under sulphur-limited conditions. By comparing TF transcript profiles from plants grown on different sulphate regimes, we identified TF genes that may specifically induce or repress changes in expression of genes that allow plants to adapt to changes in sulphate availability. Candidate genes obtained from this screening were tested by reverse genetics approaches. Transgenic plants constitutively overproducing selected TF genes and mutant plants, lacking functional selected TF genes (knock out), were used. By comparing metabolite and transcript profiles from transgenic and wild type plants we aimed at confirming the role of selected AP2 TF candidate genes in plant adaptation to sulphur unavailability. After preliminary characterisation of WRKY24 and MYB93 TF genes, we postulate that these factors are involved in a complex multifactorial regulatory network, in which WRKY24 and MYB93 would act as superior factors regulating other transcription factors directly involved in the regulation of S-metabolism genes. Results obtained for plants overproducing TOE1 and TOE2 TF genes suggests that these factors may be involved in a mechanism, which is promoting synthesis of an essential amino acid, methionine, over synthesis of another amino acid, cysteine. Thus, TOE1 and TOE2 genes might be a part of transcriptional regulation of methionine synthesis. Approaches creating genetically manipulated plants may produce plant phenotypes of immediate biotechnological interest, such as plants with increased sulphate or sulphate-containing amino acid content, or better adapted to the sulphate unavailability.
Biomakromoleküle sind in der Natur für viele Abläufe in lebenden Organismen verantwortlich. Dies reicht vom Aufbau der extrazellulären Matrix und dem Cytoskelett über die Erkennung von Botenstoffen durch Rezeptoren bis hin zur Katalyse der verschiedensten Reaktionen in den Zellen selbst. Diese Aufgaben werden zum größten Teil von Proteinen übernommen, und besonders das spezifische Erkennen der Interaktionspartner ist für alle diese Moleküle äußerst wichtig, um eine fehlerfreie Funktion zu gewährleisten. Als Alternative zur evolutiven Erzeugung von optimalen Bindern und Katalysatoren auf der Basis von Aminosäuren und Nukleotiden wurden von Wulff, Shea und Mosbach synthetische molekular geprägte Polymere (molecularly imprinted polymers, MIPs) konzipiert. Das Prinzip dieser künstlichen Erkennungselemente beruht auf der Tatsache, dass sich funktionelle Monomere spezifisch um eine Schablone (Templat) anordnen. Werden diese Monomere dann vernetzend polymerisiert, entsteht ein Polymer mit molekularen Kavitäten, in denen die Funktionalitäten komplementär zum Templat fixiert sind. Dadurch ist die selektive Bindung des Templats in diese Kavitäten möglich. Aufgrund ihrer hohen chemischen und thermischen Stabilität und ihrer geringen Kosten haben “bio-inspirierte” molekular geprägte Polymere das Potential, biologische Erkennungselemente in der Affinitätschromatographie sowie in Biosensoren und Biochips zu ersetzen. Trotz einiger publizierter Sensorkonfigurationen steht der große Durchbruch noch aus. Ein Hindernis für Routineanwendungen ist die Signalgenerierung bei Bindung des Analyten an das Polymer. Eine Möglichkeit für die markerfreie Detektion ist die Benutzung von Kalorimetern, die Bindungs- oder Reaktionswärmen direkt messen können. In der Enzymtechnologie wird der Enzym-Thermistor für diesen Zweck eingesetzt, da enzymatische Reaktionen eine Enthalpie in einer Größenordnung von 5 – 100 kJ/mol besitzen. In dieser Arbeit wird die Herstellung von katalytisch geprägten Polymeren nach dem Verfahren des Oberflächenprägens erstmalig beschrieben. Die Methode zur Immobilisierung des Templats auf der Oberfläche von porösem Kieselgel sowie die Polymerzusammensetzung wurden optimiert. Weiter wird die Evaluation der katalytischen Eigenschaften über einen optischen Test, sowie das erste Mal die Kombination eines kalorimetrischen Transduktors – des Thermistors – mit der Analyterkennung durch ein katalytisch aktives MIP gezeigt. Bei diesen Messungen konnte zum ersten Mal gleichzeitig die Bindung/Desorption, sowie die katalytische Umwandlung des Substrats durch konzentrationsabhängige Wärmesignale nachgewiesen werden.
Global Circulation Models of climate predict not only a change of annual precipitation amounts but also a shift in the daily distribution. To improve the understanding of the importance of daily rain pattern for annual plant communities, which represent a large portion of semi-natural vegetation in the Middle East, I used a detailed, spatially explicit model. The model explicitly considers water storage in the soil and has been parameterized and validated with data collected in field experiments in Israel and data from the literature. I manipulated daily rainfall variability by increasing the mean daily rain intensity on rainy days (MDI, rain volume/day) and decreasing intervals between rainy days while keeping the mean annual amount constant. In factorial combination, I also increased mean annual precipitation (MAP). I considered five climatic regions characterized by 100, 300, 450, 600, and 800 mm MAP. Increasing MDI decreased establishment when MAP was >250 mm but increased establishment at more arid sites. The negative effect of increasing MDI was compensated by increasing mortality with increasing MDI in dry and typical Mediterranean regions (c. 360–720 mm MAP). These effects were strongly tied to water availability in upper and lower soil layers and modified by competition among seedlings and adults. Increasing MAP generally increased water availability, establishment, and density. The order of magnitudes of MDI and MAP effects overlapped partially so that their combined effect is important for projections of climate change effects on annual vegetation. The effect size of MAP and MDI followed a sigmoid curve along the MAP gradient indicating that the semi-arid region (≈300 mm MAP) is the most sensitive to precipitation change with regard to annual communitie
There is already strong evidence that temperate lakes have been highly vulnerable to human induced climate warming during the last century. Hitherto climate impact studies have mainly focussed on the impacts of the recent long-term warming in winter and spring and little is known on the influence of climate warming on temperate lakes in summer. In the present thesis, I studied some aspects, which may have been strongly involved in determining the response of a lake to climate warming in summer. Thereby I have focussed on climate induced impacts on the thermal characteristics and the phenology and abundance of summer plankton in a shallow polymictic lake (Müggelsee, Germany). First, the influence of climate warming on the phenology and abundance of the lake plankton was investigated across seasons. Fast-growing spring phytoplankton and zooplankton (Daphnia) advanced largely synchronously, whereas long-term changes in the phenology of slow-growing summer zooplankton were clearly species-specific and not synchronised. The phenology and/or abundance of several summer copepod species changed according to their individual thermal requirements at decisive developmental stages such as emergence from diapause in spring. The study emphasises that not only the degree of warming, but also its timing within the annual cycle is of great ecological importance. To analyse the impact of climate change on the thermal characteristics of the lake, I examined the long-term development of the daily epilimnetic temperature extrema during summer. The study demonstrated for the first time for lakes that the daily epilimnetic minima (during nighttime) have increased more rapidly than the daily epilimnetic maxima (during daytime), resulting in a distinct decrease in the daily epilimnetic temperature range. This day-night asymmetry in epilimnetic temperature was likely caused by an increased nighttime emission of long-wave radiation from the atmosphere. This underlines that not only increases in air temperature, but also changes in other meteorological variables such as wind speed, relative humidity and cloud cover may play an important role in determining the lake temperature with respect to further climate change. Furthermore, a short-term analysis on the mixing regime of the polymictic lake was conducted to examine the frequency and duration of stratification events and their impacts on dissolved oxygen, dissolved nutrients and summer phytoplankton. Even during the longest stratification events (heatwaves in 2003 and 2006) the thermal characteristics of the lake differed from those typically found in shallow dimictic lakes, which exhibit a continuous stratification during summer. Particularly, hypolimnetic temperatures were higher, favouring the depletion of oxygen and the accumulation of dissolved nutrient in the hypolimnion. Thermal stratification will be very likely amplified in the future, thus, I conclude that polymictic lakes will be very vulnerable to alterations in the thermal regime with respect to projections of further climate change during summer. Finally, a long-term case study on the long and short-term changes in the development of the planktonic larvae of the freshwater mussel Dreissena polymorpha was performed to analyse the impacts of simultaneous changes in the thermal and in the trophic regime of the lake. Both the climate warming and the decrease in external nutrient load were important in determining the abundance of the pelagic larvae by affecting different features of the life-history of this species throughout the warm season. The long-term increase in the abundance and length of larvae was related to the decrease in external nutrient loading and the change in phytoplankton composition. However, the recent heatwaves in 2003 and 2006 have offset this positive effect on larval abundance, due to unfavourable low oxygen concentrations that had resulted from extremely long stratification events, mimicking the effects of nutrient enrichment. Climate warming may thus induce counteracting effects in productive shallow lakes that underwent lake restoration through a decrease in external nutrient loading. I conclude that not only the nature of climate change and thus the timing of climate warming throughout the seasons and the occurrence of climatic extremes as heatwaves, but also site-specific lake conditions as the thermal mixing regime and the trophic state are crucial factors governing the impacts of climate warming on internal lake processes during summer. Consequently, further climate impact research on lake functioning should focus on how the different lake types respond to the complex environmental forcing in summer, to allow for a comprehensive understanding of human induced environmental changes in lakes.
Development and application of novel genetic transformation technologies in maize (Zea mays L.)

Plant genetic engineering approaches are of pivotal importance to both basic and applied research. However, rapid commercialization of genetically engineered crops, especially maize, raises several ecological and environmental concerns largely related to transgene flow via pollination. In most crops, the plastid genome is inherited uniparentally in a maternal manner. Consequently, a trait introduced into the plastid genome would not be transferred to the sexually compatible relatives of the crops via pollination. Thus, beside its several other advantages, plastid transformation provides transgene containment, and therefore, is an environmentally friendly approach for genetic engineering of crop plants. Reliable in vitro regeneration systems allowing repeated rounds of regeneration are of utmost importance to development of plastid transformation technologies in higher plants. While being the world’s major food crops, cereals are among the most difficult-to-handle plants in tissue culture which severely limits genetic engineering approaches. In maize, immature zygotic embryos provide the predominantly used material for establishing regeneration-competent cell or callus cultures for genetic transformation experiments. The procedures involved are demanding, laborious and time consuming and depend on greenhouse facilities. In one part of this work, a novel tissue culture and plant regeneration system was developed that uses maize leaf tissue and thus is independent of zygotic embryos and greenhouse facilities. Also, protocols were established for (i) the efficient induction of regeneration-competent callus from maize leaves in the dark, (ii) inducing highly regenerable callus in the light, and (iii) the use of leaf-derived callus for the generation of stably transformed maize plants. Furthermore, several selection methods were tested for developing a plastid transformation system in maize. However, stable plastid transformed maize plants could not be yet recovered. Possible explanations as well as suggestions for future attempts towards developing plastid transformation in maize are discussed. Nevertheless, these results represent a first essential step towards developing chloroplast transformation technology for maize, a method that requires multiple rounds of plant regeneration and selection to obtain genetically stable transgenic plants. In order to apply the newly developed transformation system towards metabolic engineering of carotenoid biosynthesis, the daffodil phytoene synthase (PSY) gene was integrated into the maize genome. The results illustrate that expression of a recombinant PSY significantly increases carotenoid levels in leaves. The beta-carotene (pro-vitamin A) amounts in leaves of transgenic plants were increased by ~21% in comparison to the wild-type. These results represent evidence for maize to have significant potential to accumulate higher amounts of carotenoids, especially beta-carotene, through transgenic expression of phytoene synthases. Finally, progresses were made towards developing transformation technologies in Peperomia (Piperaceae) by establishing an efficient leaf-based regeneration system. Also, factors determining plastid size and number in Peperomia, whose species display great interspecific variation in chloroplast size and number per cell, were investigated. The results suggest that organelle size and number are regulated in a tissue-specific manner rather than in dependency on the plastid type. Investigating plastid morphology in Peperomia species with giant chloroplasts, plasmatic connections between chloroplasts (stromules) were observed under the light microscope and in the absence of tissue fixation or GFP overexpression demonstrating the relevance of these structures in vivo. Furthermore, bacteria-like microorganisms were discovered within Peperomia cells, suggesting that this genus provides an interesting model not only for studying plastid biology but also for investigating plant-microbe interactions.
Biochemical and physiological studies of Arabidopsis thaliana Diacylglycerol Kinase 7 (AtDGK7)

A family of diacylglycerol kinases (DGK) phosphorylates the substrate diacylglycerol (DAG) to generate phosphatidic acid (PA) . Both molecules, DAG and PA, are involved in signal transduction pathways. In the model plant Arabidopsis thaliana, seven candidate genes (named AtDGK1 to AtDGK7) code for putative DGK isoforms. Here I report the molecular cloning and characterization of AtDGK7. Biochemical, molecular and physiological experiments of AtDGK7 and their corresponding enzyme are analyzed. Information from Genevestigator says that AtDGK7 gene is expressed in seedlings and adult Arabidopsis plants, especially in flowers. The AtDGK7 gene encodes the smallest functional DGK predicted in higher plants; but also, has an alternative coding sequence containing an extended AtDGK7 open reading frame, confirmed by PCR and submitted to the GenBank database (under the accession number DQ350135). The new cDNA has an extension of 439 nucleotides coding for 118 additional amino acids The former AtDGK7 enzyme has a predicted molecular mass of ~41 kDa and its activity is affected by pH and detergents. The DGK inhibitor R59022 also affects AtDGK7 activity, although at higher concentrations (i.e. IC50 ~380 µM). The AtDGK7 enzyme also shows a Michaelis-Menten type saturation curve for 1,2-DOG. Calculated Km and Vmax were 36 µM 1,2-DOG and 0.18 pmol PA min-1 mg of protein-1, respectively, under the assay conditions. Former protein AtDGK7 are able to phosphorylate different DAG analogs that are typically found in plants. The new deduced AtDGK7 protein harbors the catalytic DGKc and accessory domains DGKa, instead the truncated one as the former AtDGK7 protein (Gomez-Merino et al., 2005).
Different habitat models were created for the White Stork (Ciconia ciconia) in the region of the former German province of East Prussia (equals app. the current Russian oblast Kaliningrad and the Polish voivodship Warmia-Masuria). Different historical data sets describing the occurrence of the White Stork in the 1930s, as well as selected variables for the description of landscape and habitat, were employed. The processing and modeling of the applied data sets was done with a geographical information system (ArcGIS) and a statistical modeling approach that comes from the disciplines of machine-learning and data mining (TreeNet by Salford Systems Ltd.). Applying historical habitat descriptors, as well as data on the occurrence of the White Stork, models on two different scales were created: (i) a point scale model applying a raster with a cell size of 1 km2 and (ii) an administrative district scale model based on the organization of the former province of East Prussia. The evaluation of the created models show that the occurrence of White Stork nesting grounds in the former East Prussia for most parts is defined by the variables ‘forest’, ‘settlement area’, ‘pasture land’ and ‘proximity to coastline’. From this set of variables it can be assumed that a good food supply and nesting opportunities are provided to the White Stork in pasture and meadows as well as in the proximity to human settlements. These could be seen as crucial factors for the choice of nesting White Stork in East Prussia. Dense forest areas appear to be unsuited as nesting grounds of White Storks. The high influence of the variable ‘coastline’ is most likely explained by the specific landscape composition of East Prussia parallel to the coastline and is to be seen as a proximal factor for explaining the distribution of breeding White Storks. In a second step, predictions for the period of 1981 to 1993 could be made applying both scales of the models created in this study. In doing so, a decline of potential nesting habitat was predicted on the point scale. In contrast, the predicted White Stork occurrence increases when applying the model of the administrative district scale. The difference between both predictions is to be seen in the application of different scales (density versus suitability as breeding ground) and partly dissimilar explanatory variables. More studies are needed to investigate this phenomenon. The model predictions for the period 1981 to 1993 could be compared to the available inventories of that period. It shows that the figures predicted here were higher than the figures established by the census. This means that the models created here show rather a capacity of the habitat (potential niche). Other factors affecting the population size e.g. breeding success or mortality have to be investigated further. A feasible approach on how to generate possible habitat models was shown employing the methods presented here and applying historical data as well as assessing the effects of changes in land use on the White Stork. The models present the first of their kind, and could be improved by means of further data regarding the structure of the habitat and more exact spatially explicit information on the location of the nesting sites of the White Stork. In a further step, a habitat model of the present times should be created. This would allow for a more precise comparison regarding the findings from the changes of land use and relevant conditions of the environment on the White Stork in the region of former East Prussia, e.g. in the light of coming landscape changes brought by the European Union (EU).
Alle Organismen sind für ihr Überleben auf Metalle angewiesen. Hierbei gibt es für jedes Metall einen Konzentrationsbereich, der das Optimum zwischen Metallmangel, -bedarf und -toxizität darstellt. Es gilt mittlerweile als erwiesen, dass alle Organismen zur Aufrechterhaltung des Metallgleichgewichts ein komplexes Netzwerk von Proteinen und niedermolekularen Verbindungen entwickelt haben. Die molekularen Komponenten dieses Netzwerks sind nur zu einem Teil bekannt und charakterisiert: In den letzten Jahren wurden einige Proteinfamilien identifiziert, deren Mitglieder Metalle durch Lipidmembranen transportieren. Eine dieser Metalltransporterfamilien ist die Cation Diffusion Facilitator (CDF)-Familie: Alle charakterisierten Mitglieder exportieren Metalle aus dem Zytoplasma – entweder in zelluläre Kompartimente oder aus der Zelle heraus. Von den zwölf Mitgliedern dieser Familie in Arabidopsis thaliana (A. thaliana) – Metall Toleranz Protein (MTP)-1 bis -12 – wurden bisher AtMTP1 und AtMTP3 charakterisiert. In dieser Arbeit wird die Charakterisierung von AtMTP2 beschrieben. Wie die homologen Proteine AtMTP1 und AtMTP3 führt AtMTP2 zu Zn-Toleranz, wenn es heterolog in Zn-sensitiven Hefemutanten exprimiert wird. Mit AtMTP2 transformierte Hefemutanten zeigten darüber hinaus erhöhte Co-Toleranz. Expression von chimären AtMTP2/GFP Fusionsproteinen in Hefe, A.thaliana protoplasten und in stabil transformierten A.thalinana Planzenlinien deutet auf Lokalisation of AtMTP2 in Membranen des Endoplasmatischen Retikulums (ER) hin, wenn GFP an den C-Terminus von MTP2 fusioniert wird. Fusion of GFP an den N-Terminus von AtMTP2 führte zu Lokalisation in der vakuolären Membran, was wahrscheinlichsten auf Fehllokalisierung durch Maskierung eines ER-Retentionsmotivs (XXRR) am N-Terminus von AtMTP2 zurückgeht. Dies legt nahe, dass AtMTP2 die erwähnten Metalle in das Endomembransystem der Zelle transportieren kann. Eine gewebespezifische Lokalisierung wurde mit Pflanzen durchgeführt, die das β-Glucuronidase (GUS)-Reporterprotein bzw. chimäre Fusionsproteine aus EGFP und AtMTP2 unter Kontrolle des nativen pMTP2-Promotors exprimierten. Diese Experimente bestätigten zum einen, dass der pMTP2-Promotor nur unter Zn-Defizienz aktiv ist. GUS-Aktivität wurde unter diesen Bedingungen in zwei Zonen der Wurzelspitze beobachtet: in den isodiametrischen Zellen der meristematischen Zone und in der beginnenden Wurzelhaarzone. Darüber hinaus konnte gezeigt werden, dass die EGFP-Fusionsproteine unter Kontrolle des nativen pMTP2-Promotors nur in epidermalen Zellen exprimiert werden. Für eine homozygote Knockout- Linie, mtp2-S3, konnte bisher kein eindeutiger Phänotyp identifiziert werden. Auf Grundlage der bisher durchgeführten Charakterisierung von AtMTP2 erscheinen zwei Modelle der Funktion von AtMTP2 in der Pflanze möglich: AtMTP2 könnte essentiell für die Versorgung des ER mit Zn unter Zn-Mangelbedingungen sein. Hierfür spricht, dass AtMTP2 in jungen, teilungsaktiven und damit Zn-benötigenden Wurzelzonen exprimiert wird. Die auf die Epidermis beschränkte Lokalisation könnte bei diesem Modell auf die Möglichkeit der zwischenzellulären Zn-Verteilung innerhalb des ER über Desmotubules hindeuten. Alternativ könnte AtMTP2 eine Funktion bei der Detoxifizierung von Zn unter Zn-Schock Bedingungen haben: Es ist bekannt, dass unter Zn- Mangelbedingungen die Expression der zellulären Zn-Aufnahmesysteme hochreguliert wird. Wenn nun die Zn-Verfügbarkeit im Boden z. B durch eine pH-Änderung innerhalb kurzer Zeit stark ansteigt, besteht die Notwendigkeit der Entgiftung von Zn innerhalb der Zelle, bis der starke Einstrom von Zn ins Zytoplasma durch die Deaktivierung der Zn-Aufnahmesysteme und einer geringeren Expression in der Pflanze gedrosselt ist. Ein ähnlicher Mechanismus wurde in der Bäckerhefe S. cerevisae beschrieben, in der darüber hinaus ein Zn-Transporter verstärkt exprimiert wird, der Zn durch Transport in die Vakuole entgiften kann. Es ist durchaus möglich, dass in Arabidopsis AtMTP2 die Zn-Detoxifizierung unter diesen speziellen Bedingungen durch Zn-Transport in das ER oder die Vakuole vermittelt. Zur Identifikation weiterer Komponenten des Metallhomöostasenetzwerks sind verschiedene Ansätze denkbar. In dieser Arbeit wurde in Hefe ein heterologer Screen durchgeführt, um Interaktoren für vier Mitglieder der Arabidopsis-CDF-Familie zu identifizieren. Unter den 11 im Hefesystem bestätigten Kandidaten befindet sich mit AtSPL1 ein AtMTP1-Interaktionskandidat, der möglicherweise eine Rolle bei der Cu-,Zn-Homöostase spielt. Als wahrscheinliche AtMTP3-Interaktionskandidaten wurde die c”-Untereinheit der vakuolären H+-ATPase AtVHA identifiziert sowie mit AtNPSN13 ein Protein, das vermutlich eine Rolle bei Fusionen von Vesikeln mit Zielmembranen spielt. Ein anderer Ansatz zur Identifikation neuer Metallhomöostasegene ist die vergleichende Elementanalyse von natürlichen oder mutagenisierten Pflanzenpopulationen. Voraussetzung für diesen Ansatz ist die schnelle und genaue Analyse des Elementgehalts von Pflanzen. Eine etablierte Methode zur simultanen Bestimmung von bis zu 65 Elementen in einer Probe ist die Inductively Coupled Plasma Optical Emission Spectrometry (ICP OES). Der limitierende Faktor für einen hohen Probendurchsatz ist die Notwendigkeit, Proben für die Analyse zu verflüssigen. Eine alternative Methode der Probenzuführung zum Analysegerät ist die elektrothermale Verdampfung (ETV) der Probe. Zur weitgehend automatisierten Analyse von Pflanzenmaterial mit minimiertem Arbeitsaufwand wurde eine Methode entwickelt, die auf der Kopplung der ETV mit der ICP OES basiert.
Es ist bekannt, dass Änderungen im Kohlenstoff- bzw. Stickstoffstaus der Pflanzen zu einer parallelen statt reziproken Änderung der kohlenstoff- und stickstoffhaltigen Primärmetabolite führen. Unter diesem Gesichtspunkt wurden in der vorliegenden Arbeit der Aminosäurestoffwechsel und der Sekundärstoffwechsel unter reduzierten Stickstoffbedingungen untersucht. Zur Beeinflussung des Stickstoffstoffwechsels wurden nitratmangelernährte Tabakwildtyppflanzen und Genotypen mit unterschiedlich stark reduzierter Nitratreduktase-Aktivität verwendet. Dieses experimentelle System erlaubt zusätzlich durch den Vergleich Nitrat defizienter Wildtyppflanzen mit Nitrat akkumulierenden NIA-Transformanten Prozesse zu identifizieren, die durch Nitrat gesteuert werden. Die Analysen der Primär- und Sekundärmetabolite wurde in allen Genotypen diurnal durchgeführt, um auch tageszeitlich abhängige Prozesse zu identifizieren. Die Analyse der absoluten Gehalte aller individuellen Aminosäuren enthüllte bei den meisten erstaunlich stabile diurnale Muster mit einem Anstieg während des Tages und einem Abfall in der Nacht in Wildtyppflanzen gewachsen mit ausreichend Nitrat. Dieses Ergebnis legt die Schlussfolgerung nahe, dass die Biosynthese der Aminosäuren koordiniert abläuft. In Pflanzen mit reduziertem Stickstoffstatus haben diese diurnalen Muster jedoch keinen Bestand. Die Kombination des erzeugten stickstoffbasierten Aminosäuredatensatz in Kombination mit einem bereits erzeugten Aminosäuredatensatz unter kohlenstofflimitierten Bedingungen von Matt et al. (2002) führte durch Hauptkomponentenanalyse (PCA) und Korrelationsanalyse zu dem Ergebnis, dass die Hypothese nach einer koordinierten Aminosäurebiosynthese nicht allgemeine Gültigkeit hat. Die PCA identifizierte Glutamin, Glutamat, Aspartat, Glycin, Pheny-lalanin und Threonin als Faktoren, die den Datensätzen ihre charakteristische Eigenschaft und deren Varianz verleihen. Die Korrelationsanalyse zeigte, dass die sehr guten Korrelationen der individuellen Aminosäuren untereinander in reduzierten Stickstoff- und Kohlenstoffbedingungen sich verschlechtern. Das Verhältnis einer einzelnen Aminosäure relativ zu den anderen führte zur Identifizierung einiger Aminosäuren, die individuelle Antworten auf Stickstoff- und/oder Kohlenstoffstatus zeigen, und/oder speziell auf Nitrat, Licht und/oder den E-nergiestatus der Thylakoidmembran. Glutamat beispielsweise verhält sich in den meisten Situationen stabil, Phenylalanin dagegen zeigt in jeder physiologischen Situation eine individuelle Antwort. Die Ergebnisse dieser Arbeit führen zu einer Erweiterung der Hypothese einer koordinierten Synthese der Aminosäuren dahingehend, dass diese nicht generell für alle Aminosäuren angenommen werden kann. Es gibt einige Aminosäuren deren, Anteile sich situationsbedingt anpassen. Die Reduktion des Stickstoffstatus in nitratmangelernährten Tabakwildtyppflanzen führte zu der, nach der „Carbon-Nutrient-Balance“ Hypothese erwarteten Verlagerung der kohlenstoffreichen Phenylpropanoide und des stickstoffreichen Nikotins. Die Erhöhung der Phenylpropanoidgehalte war nicht in der Nitrat akkumulierenden NIA-Transformante zu beobachten und somit konnte Nitrat als regulatorisches Element identifiziert werden. Ein Einfluss der Vorläufermetabolite konnte ausgeschlossen werden, da sowohl nitratmangelernährter Wildtyp als auch die Nitrat akkumulierende NIA-Transformante ähnliche Gehalte dieser aufwiesen. Genexpressionsanalysen über Mikroarray-Hybridisierung und quantitative RT-PCR zeigten, dass Nitrat durch noch nicht geklärte Mechanismen Einfluss auf die Expression einiger Gene nimmt, die dem Phenylpropanoidstoffwechsels zugeordnet sind. During the last decades, the global change of the environment has caused a dramatic loss of habitats and species. In Central Europe, open habitats are particularly affected. The main objective of this thesis was to experimentally test the suitability of wild megaherbivore grazing as a conservation tool to manage open habitats. We studied the effect of wild ungulates in a 160 ha game preserve in NE Germany in three successional stages (i) Corynephorus canescens-dominated grassland, (ii) ruderal tall forb vegetation dominated by Tanacetum vulgare and (iii) Pinus sylvestris-pioneer forest over three years. Our results demonstrate that wild megaherbivores considerably affected species composition and delayed successional pathways in open habitats. Grazing effects differed considerably between successional stages: species richness was higher in grazed ruderal and pioneer forest plots, but not in the Corynephorus sites. Species composition changed significantly in the Corynephorus and ruderal sites. Grazed ruderal sites had turned into sites with very short vegetation dominated by Agrostis spp. and the moss Brachythecium albicans, most species did not flower. Woody plant cover was significantly affected only in the pioneer forest sites. Young pine trees were severely damaged and tree height was considerably reduced, leading to a “Pinus-macchie”-appearance. Ecological patterns and processes are known to vary with spatial scale. Since grazing by megaherbivores has a strong spatial component, the scale of monitoring success of grazing may largely differ among and within different systems. Thus, the second aim of this thesis was to test whether grazing effects are consistent over different spatial scales, and to give recommendations for appropriate monitoring scales. For this purpose, we studied grazing effects on plant community structure using multi-scale plots that included three nested spatial scales (0.25 m2, 4 m2, and 40 m2). Over all vegetation types, the scale of observation directly affected grazing effects on woody plant cover and on floristic similarity, but not on the proportion of open soil and species richness. Grazing effects manifested at small scales regarding floristic similarity in pioneer forest and ruderal sites and regarding species richness in ruderal sites. The direction of scale-effects on similarity differed between vegetation types: Grazing effects on floristic similarity in the Corynephorus sites were significantly higher at the medium and large scale, while in the pioneer forest sites they were significantly higher at the smallest scale. Disturbances initiate vegetation changes by creating gaps and affecting colonization and extinction rates. The third intention of the thesis was to investigate the effect of small-scale disturbances on the species-level. In a sowing experiment, we studied early establishment probabilities of Corynephorus canescens, a key species of open sandy habitats. Applying two different regimes of mechanical ground disturbance (disturbed and undisturbed) in the three successional stages mentioned above, we focused on the interactive effects of small-scale disturbances, successional stage and year-to-year variation. Disturbance led to higher emergence in a humid and to lower emergence in a very dry year. Apparently, when soil moisture was sufficient, the main factor limiting C. canescens establishment was competition, while in the dry year water became the limiting factor. Survival rates were not affected by disturbance. In humid years, C. canescens emerged in higher numbers in open successional stages while in the dry year, emergence rates were higher in late stages, suggesting an important role of late successional stages for the persistence of C. canescens. We conclude that wild ungulate grazing is a useful tool to slow down succession and to preserve a species-rich, open landscape, because it does not only create disturbances, thereby supporting early successional stages, but at the same time efficiently controls woody plant cover. However, wild ungulate grazing considerably changed the overall appearance of the landscape. Additional measures like shifting exclosures might be necessary to allow vulnerable species to flower and reproduce. We further conclude that studying grazing impacts on a range of scales is crucial, since different parameters are affected at different spatial scales. Larger scales are suitable for assessing grazing impact on structural parameters like the proportion of open soil or woody plant cover, whereas species richness and floristic similarity are affected at smaller scales. Our results further indicate that the optimal strategy for promoting C. canescens is to apply disturbances just before seed dispersal and not during dry years. Further, at the landscape scale, facilitation by late successional species may be an important mechanism for the persistence of protected pioneer species.
This contribution describes a generator of stochastic time series of daily precipitation for the interior of Israel from c. 90 to 900 mm mean annual precipitation (MAP) as a tool for studies of daily rain variability. The probability of rainfall on a given day of the year is described by a regular Gaussian peak curve function. The amount of rain is drawn randomly from an exponential distribution whose mean is the daily mean rain amount (averaged across years for each day of the year) described by a flattened Gaussian peak curve. Parameters for the curves have been calculated from monthly aggregated, long-term rain records from seven meteorological stations. Parameters for arbitrary points on the MAP gradient are calculated from a regression equation with MAP as the only independent variable. The simple structure of the generator allows it to produce time series with daily rain patterns that are projected under climate change scenarios and simultaneously control MAP. Increasing within-year variability of daily precipitation amounts also increases among-year variability of MAP as predicted by global circulation models. Thus, the time series incorporate important characteristics for climate change research and represent a flexible tool for simulations of daily vegetation or surface hydrology dynamics.
Sucrose synthase (Susy) is a key enzyme of sucrose metabolism, catalysing the reversible conversion of sucrose and UDP to UDP-glucose and fructose. Therefore, its activity, localization and function have been studied in various plant species. It has been shown that Susy can play a role in supplying energy in companion cells for phloem loading (Fu and Park, 1995), provides substrates for starch synthesis (Zrenner et al., 1995), and supplies UDP-glucose for cell wall synthesis (Haigler et al., 2001). Analysis of the Arabidopsis genome identifies six Susy isoforms. The expression of these isoforms was investigated using promoter-reporter gene constructs (GUS) and real time RT-PCR. Although these isoforms are closely related at the protein level they have radically different spatial and temporal patterns of expression in the plant with no two isoforms showing the same distribution. More than one isoform is expressed in all organs examined. Some of them have high but specific expression in particular organs or developmental stages whilst others are constantly expressed throughout the whole plant and across various stages of development. The in planta function of the six Susy isoforms were explored through analysis of T-DNA insertion mutants and RNAi lines. Plants without the expression of individual isoforms show no differences in growth and development, and are not significantly different from wild type plants in soluble sugars, starch and cellulose contents under all growth conditions investigated. Analysis of T-DNA insertion mutant lacking Sus3 isoform that was exclusively expressed in stomata cells only had a minor influence on guard cell osmoregulation and/or bioenergetics. Although none of the sucrose synthases appear to be essential for normal growth under our standard growth conditions, they may be necessary for growth under stress conditions. Different isoforms of sucrose synthase respond differently to various abiotic stresses. It has been shown that oxygen deprivation up regulates Sus1 and Sus4 and increases total Susy activity. However, the analysis of the plants with reduced expression of both Sus1 and Sus4 revealed no obvious effects on plant performance under oxygen deprivation. Low temperature up regulates Sus1 expression but the loss of this isoform has no effect on the freezing tolerance of non acclimated and cold acclimated plants. These data provide a comprehensive overview of the expression of this gene family which supports some of the previously reported roles for Susy and indicates the involvement of specific isoforms in metabolism and/or signalling.
Germination rates and germination fractions of seeds can be predicted well by the hydrothermal time (HTT) model. Its four parameters hydrothermal time, minimum soil temperature, minimum soil moisture, and variation of minimum soil moisture, however, must be determined by lengthy germination experiments at combinations of several levels of soil temperature and moisture. For some applications of the HTT model it is more important to have approximate estimates for many species rather than exact values for only a few species. We suggest that minimum temperature and variation of minimum moisture can be estimated from literature data and expert knowledge. This allows to derive hydrothermal time and minimum moisture from existing data from germination experiments with one level of temperature and moisture. We applied our approach to a germination experiment comparing germination fractions of wild annual species along an aridity gradient in Israel. Using this simplified approach we estimated hydrothermal time and minimum moisture of 36 species. Comparison with exact data for three species shows that our method is a simple but effective method for obtaining parameters for the HTT model. Hydrothermal time and minimum moisture supposedly indicate climate related germination strategies. We tested whether these two parameters varied with the climate at the site where the seeds had been collected. We found no consistent variation with climate across species, suggesting that variation is more strongly controlled by site-specific factors.

In der vorliegenden Arbeit wurden cDNAs, kodierend für bisher unbekannte stärkeabbauende Enzyme, aus Kartoffel isoliert und funktionell analysiert. Die Isolation der cDNAs erfolgte mit Hilfe eines Systems, welches sich der funktionellen Expression von cDNA-Bibliotheken in E. coli bediente. Die mit diesem System zur Expression gebrachten cDNA-Bibliotheken wurden im Rahmen dieser Arbeit hergestellt. Zum einen handelte es sich um eine blattspezifische Phagen-cDNA-Bibliothek (Proben wurden während des Tag/Nacht Übergangs genommen), zum anderen um eine knollenspezifische cDNA-Bibliothek aus kaltgelagerten Knollen. Nach der Überführung der Phagen-Bibliotheken in Plasmid-Bibliotheken wurden diese funktionell in dem E. coli Stamm KV832 exprimiert. Der Stamm KV832 wurde aufgrund seiner Fähigkeit, lineare Glucane zu akkumulieren, ausgewählt. Werden Glucan akkumulierende KV832 Kolonien mit Jod bedampft, so zeigen diese eine typische Blaufärbung. Nach der Expression der Plasmid-Bibliotheken in KV832 wurden solche Kolonien weiter untersucht, welche in ihrer Färbung von den blauen Kolonien abwichen. Mittels eines zweiten E. coli Stamms, PGM −, welcher ebenfalls in der Lage ist, lineare Glucane zu akkumulieren, wurden die Ergebnisse für KV832 bestätigt. Die funktionelle Expression der Bibliotheken führte zur Isolation einer Reihe von unbekannten cDNAs. Zwei dieser cDNAs wurden im Rahmen dieser Arbeit weiterführend untersucht. Zum einen handelte es sich um eine cDNA, die für eine bis dahin unbekannte β-Amylase aus Kartoffel kodierte und deren Homolog aus Arabidopsis (CT-BMY) im Laufe dieser Arbeit von Lao et al. (1999) veröffentlicht wurde, zum anderen um eine cDNA, die für ein unbekanntes Enzym kodierte (DSD10). Das Arabidopsis Homolog zu DSD10 wurde im Zuge der Arabidopsis Genominitiative Ende 2000 publiziert. Im Rahmen dieser Arbeit konnte gezeigt werden, dass die isolierte β-Amylase cDNA für eine funktionelle β-Amylase kodiert und dieses Enzym in der Lage ist, neben löslicher auch rohe Stärke anzugreifen. Lokalisationsexperimente zeigten, dass das Enzym in isolierte Erbsenchloroplasten importiert wurde und dass die 100 N-terminalen Aminosäuren für den Import in die Plastiden ausreichten. Die β-Amylase wurde als PCT-BMYI bezeichnet. Die »antisense«-Inhibierung von PCT-BMYI führte zu einem Hochstärke-Phänotyp der Blätter, sowie zu einem Anstieg der Trockenmasse. Der Hochstärke-Phänotyp ist auf eine Reduktion der Stärkemobilisierung und die daraus folgende Akkumulation der Stärke während der Vegetationsperiode zurückzuführen. Damit konnte erstmals die physiologische Bedeutung einer β-Amylase für den Abbau der transitorischen Stärke gezeigt werden. Kein Einfluss zeigte die »antisense« Inhibierung von PCT-BMYI auf den kälteinduzierten Abbau der Speicherstärke in Knollen. Es konnte auch kein Unterschied im Keimverhalten oder der Entwicklung der neuen Pflanze beobachtet werden. Ein Teil der Ergebnisse zu PCT-BMYI wurde bereits publiziert (Scheidig et al., 2002). Die isolierten cDNAs dsd10, sgeI (die Volllängen cDNA zu dsd10) und das Arabidopsis Homolog asgeI kodieren für Enzyme, welche α-Amylase-Aktivität besitzen, aber keine Homologie zu bekannten α-Amylasen aufweisen. Ein mögliches Glucoamylase Motiv erwies sich für die Aktivität des Proteins als essentiell. Lokalisationsexperimente deuteten auf den Import des SGEI Proteins in isolierte Erbsenchloroplasten hin. Die »antisense«-Inhibierung von sgeI führte in den entsprechenden Linien zu einem Hochstärke-Phänotyp in Blättern, einem Anstieg der Trockenmasse in Blättern, sowie zu größeren Stärkekörnern in einer der untersuchten Linien. Ein nicht erwarteter Effekt zeigte sich in Blättern der entsprechenden Linien, welche für längere Zeit dunkel gehalten wurden. Die Blätter der untransformierten Kontrolle waren abgestorben, wohingegen die Blätter der SGEI »antisense« Linien grün und vital erschienen. Die α- und β-Amylase-Aktivität war in Blättern der SGEI »antisense« Linien reduziert, weshalb eine genaue Zuordnung der Funktion von SGEI nicht möglich war. Die vorliegenden Ergebnisse zu den SGEI »antisense« Linien deuten aber darauf hin, dass der beobachtete Hochstärke-Phänotyp nicht alleine auf die Reduktion der β-Amylase-Aktivität zurückzuführen ist. Ein Einfluss von SGEI auf den kälteinduzierten Abbau der Speicherstärke konnte nicht beobachtet werden. Es konnte auch hier kein Unterschied im Keimverhalten oder der Entwicklung der neuen Pflanze beobachtet werden.
In a recent contribution in Nature (vol. 442, pp. 555-558) Austin & Vivanco showed that sunlight is the dominant factor for decomposition of grass litter in a semi-arid grassland in Argentine. The quantification of this effect was portrayed as a novel finding. I put this result in the context of three other publications from as early as 1980 that quantified photodegradation. My synopsis shows that photodegradation is an important process in semi-arid grasslands in South America, North America and eastern Europe.
Apoptose, der programmierte Zelltod, spielt eine wichtige Rolle für das Gleichgewicht zwischen Proliferation und Sterben von Zellen und ist außerdem an der Beseitigung von infizierten und geschädigten Zellen beteiligt. Apoptose kann durch Stimulation von Rezeptoren aus der Familie der TNF-Rezeptoren wie CD95, ausgelöst werden. Nach Liganden-induzierter Trimerisierung der Rezeptoren bindet FADD an den zytoplasmatischen Teil des Rezeptors und rekrutiert Caspase-8 und/oder -10. Die räumliche Nähe der Caspasen in diesem als DISC bezeichneten Komplex führt zu ihrer auto- und transkatalytischen Spaltung und damit Aktivierung. Dadurch wird das apoptotische Programm gestartet, welches zum Tod der Zelle führt. Kontrolliert wird dieser Vorgang von einer Vielzahl anti-apoptotischer Proteine. Störungen in diesem System sind an der Entstehung einer Reihe von Krankheiten beteiligt. Die Blockade der Apoptoseinduktion kann zur Entstehung von Tumoren beitragen. Das klassische Hodgkin Lymphom ist eine maligne Erkrankung des lymphatischen Systems. Die Tumorzellen sind große, einkernige Hodgkin- oder mehrkernige Reed/Sternbergzellen (HRS-Zellen). Sie leiten sich von Keimzentrum-B-Zellen ab. In HRS-Zellen fehlt die Expression einer Vielzahl von typischen B-Zellmarkern, darunter die des B-Zellrezeptors. Solche B-Zellen sterben normalerweise während der Keimzentrumsreaktion durch Apoptose. An diesem Prozess ist CD95 beteiligt. In einer Reihe von malignen Erkrankungen wurden eine Herunterregulation der CD95-Expression oder Mutationen im CD95-Gen beobachtet. Es wird daher vermutet, dass CD95-induzierte Apoptose zur Entfernung von Tumorzellen beiträgt. Im Gegensatz dazu exprimieren sowohl primäre HRS-Zellen als auch etablierte HRS-Zelllinien in der Regel Wildtyp-CD95, sind aber trotzdem CD95-resistent. In dieser Arbeit konnte gezeigt werden, dass Komponenten des CD95-Systems, im Gegensatz zu anderen malignen Erkrankungen, in den HRS-Zellen hochreguliert sind, darunter CD95 selbst. In immunpräzipitierten DISCs von CD95-stimulierten HRS-Zellen wurde neben FADD und Caspase-8/-10 auch c-FLIP nachgewiesen. c-FLIP ist ein Caspase-8/-10-Homolog, das ebenfalls an FADD bindet, aber aufgrund fehlender katalytischer Aktivität die Aktivierung der Caspasen im DISC und damit die Apoptoseinduktion verhindert. Eine starke c-FLIP-Expression konnte in allen HRS-Zelllinien und in den HRS-Zellen nahezu aller untersuchter primärer Hodgkinfälle (55/59) gezeigt werden. Durch siRNA-vermittelte Herunterregulation von c-FLIP war es möglich, HRS-Zelllinien gegenüber CD95-induzierter Apoptose zu sensitivieren. Dies zeigt, dass die CD95-Rezeptor-induzierte Apoptose in den HRS-Zellen nicht strukturell, sondern funktionell inhibiert ist und c-FLIP stark zu dieser Inhibition beiträgt. Darüber hinaus konnte gezeigt werden, dass die c-FLIP-Expression in den HRS-Zellen von der konstitutiven Aktivität des Transkriptionsfaktors NF-κB abhängt, die charakteristisch für diese Zellen ist. Normalerweise wird NF-κB von Inhibitorproteinen, den IκBs, im Zytoplasma zurückgehalten. Diverse Stimuli können den IKK-Komplex aktivieren, der die IκBs an bestimmten Serinresten phosphoryliert. Dies hat die Ubiquitinylierung und den Abbau der IκBs zur Folge, wodurch NF-κB frei wird, in den Kern wandert und dort seine Zielgene aktiviert. Es wird angenommen, dass in HRS-Zellen ein konstitutiv aktiver IKK-Komplex und teilweise Mutationen der IκB-Proteine zur konstitutiven NF-κB-Aktivität beitragen. Zu den NF-κB-abhängigen Genen in den HRS-Zellen gehören solche mit anti-apoptotischer und Zellzyklus-treibender Wirkung. Die Inhibition der NF-κB-Aktivität in den HRS-Zellen führt zu Apoptose und eingeschränkter Proliferation. Von dreiwertigem Arsen ist bekannt, dass es die Induzierbarkeit des IKK-Komplexes inhibieren kann und damit letztendlich die Aktivierung von NF-κB. In dieser Arbeit konnte gezeigt werden, dass Arsen in HRS-Zellen den konstitutiv aktiven IKK-Komplex inhibiert. In Zelllinien mit intakten IκB-Proteinen führte dies zur NF-κB-Inhibition und Apoptoseinduktion. Die Reduktion der NF-κB-Aktivität ging mit der Herunterregulation von anti-apoptotischen und Proliferations-fördernden Zielgenen einher. Die ektope Überexpression von NF-κB hob die Apoptose-induzierende Wirkung von Arsen teilweise auf. Durch Arsen-Behandlung von Mäusen konnte das Tumorwachstum xenotransplantierter HRS-Zellen stark verlangsamt werden. In explantierten Tumorzellen konnte ebenfalls eine NF-κB-Inhibition nachgewiesen werden. Die NF-κB-Inhibition durch Arsen trägt also stark zur Apoptoseinduktion in den HRS-Zellen bei. Zusammengefasst zeigen die Ergebnisse dieser Arbeit, dass die Modulation der Apoptoseresistenz neue therapeutische Ansätze für die Behandlung des Hodgkin Lymphoms bieten könnte. Der Einsatz von Arsen ist dabei besonders interessant, da Arsen schon für die Behandlung anderer maligner Erkrankungen eingesetzt wird.
The aim of this study was to provide deeper insights in passerine phylogenetic relationships using new molecular markers. The monophyly of the largest avian order Passeriformes (~59% of all living birds) and the division into its suborders suboscines and oscines are well established. Phylogenetic relationships within the group have been extremely puzzling, as most of the evolutionary lineages originated through rapid radiation. Numerous studies have hypothesised conflicting passerine phylogenies and have repeatedly stimulated further research with new markers. In the present study, I used three different approaches to contribute to the ongoing phylogenetic debate in Passeriformes. I investigated the recently introduced gene ZENK for its phylogenetic utility for passerine systematics in combination and comparison to three already established nuclear markers. My phylogenetic analyses of a comprehensive data set yielded highly resolved, consistent and strongly supported trees. I was able to show the high utility of ZENK for elucidating phylogenetic relationships within Passeriformes. For the second and third approach, I used chicken repeat 1 (CR1) retrotransposons as phylogenetic markers. I presented two specific CR1 insertions as apomorphic characters, whose presence/absence pattern significantly contributed to the resolution of a particular phylogenetic uncertainty, namely the position of the rockfowl species Picathartes spp. in the passerine tree. Based on my results, I suggest a closer relationship of these birds to crows, ravens, jays, and allies. For the third approach, I showed that CR1 sequences contain phylogenetic signal and investigated their applicability in more detail. In this context, I screened for CR1 elements in different passerine birds, used sequences of several loci to construct phylogenetic trees, and evaluated their reliability. I was able to corroborate existing hypotheses and provide strong evidence for some new hypotheses, e.g. I suggest a revision of the taxa Corvidae and Corvinae as vireos are closer related to crows, ravens, and allies. The subdivision of the Passerida into three superfamilies, Sylvioidea, Passeroidea, and Muscicapoidea was strongly supported. I found evidence for a split within Sylvioidea into two clades, one consisting of tits and the other comprising warblers, bulbuls, laughingthrushes, whitethroats, and allies. Whereas Passeridae appear to be paraphyletic, monophyly of weavers and estrild finches as a separate clade was strongly supported. The sister taxon relationships of dippers and the thrushes/flycatcher/chat assemblage was corroborated and I suggest a closer relationship of waxwings and kinglets to wrens, tree-creepers, and nuthatches.
This study introduces a method for multiparallel analysis of small organic compounds in the unicellular green alga Chlamydomonas reinhardtii, one of the premier model organisms in cell biology. The comprehensive study of the changes of metabolite composition, or metabolomics, in response to environmental, genetic or developmental signals is an important complement of other functional genomic techniques in the effort to develop an understanding of how genes, proteins and metabolites are all integrated into a seamless and dynamic network to sustain cellular functions. The sample preparation protocol was optimized to quickly inactivate enzymatic activity, achieve maximum extraction capacity and process large sample quantities. As a result of the rapid sampling, extraction and analysis by gas chromatography coupled to time-of-flight mass spectrometry (GC-TOF) more than 800 analytes from a single sample can be measured, of which over a 100 could be positively identified. As part of the analysis of GC-TOF raw data, aliquot ratio analysis to systematically remove artifact signals and tools for the use of principal component analysis (PCA) on metabolomic datasets are proposed. Cells subjected to nitrogen (N), phosphorus (P), sulfur (S) or iron (Fe) depleted growth conditions develop highly distinctive metabolite profiles with metabolites implicated in many different processes being affected in their concentration during adaptation to nutrient deprivation. Metabolite profiling allowed characterization of both specific and general responses to nutrient deprivation at the metabolite level. Modulation of the substrates for N-assimilation and the oxidative pentose phosphate pathway indicated a priority for maintaining the capability for immediate activation of N assimilation even under conditions of decreased metabolic activity and arrested growth, while the rise in 4-hydroxyproline in S deprived cells could be related to enhanced degradation of proteins of the cell wall. The adaptation to sulfur deficiency was analyzed with greater temporal resolution and responses of wild-type cells were compared with mutant cells deficient in SAC1, an important regulator of the sulfur deficiency response. Whereas concurrent metabolite depletion and accumulation occurs during adaptation to S deprivation in wild-type cells, the sac1 mutant strain is characterized by a massive incapability to sustain many processes that normally lead to transient or permanent accumulation of the levels of certain metabolites or recovery of metabolite levels after initial down-regulation. For most of the steps in arginine biosynthesis in Chlamydomonas mutants have been isolated that are deficient in the respective enzyme activities. Three strains deficient in the activities of N-acetylglutamate-5-phosphate reductase (arg1), N2 acetylornithine-aminotransferase (arg9), and argininosuccinate lyase (arg2), respectively, were analyzed with regard to activation of endogenous arginine biosynthesis after withdrawal of externally supplied arginine. Enzymatic blocks in the arginine biosynthetic pathway could be characterized by precursor accumulation, like the amassment of argininosuccinate in arg2 cells, and depletion of intermediates occurring downstream of the enzymatic block, e.g. N2-acetylornithine, ornithine, and argininosuccinate depletion in arg9 cells. The unexpected finding of substantial levels of the arginine pathway intermediates N-acetylornithine, citrulline, and argininosuccinate downstream the enzymatic block in arg1 cells provided an explanation for the residual growth capacity of these cells in the absence of external arginine sources. The presence of these compounds, together with the unusual accumulation of N-Acetylglutamate, the first intermediate that commits the glutamate backbone to ornithine and arginine biosynthesis, in arg1 cells suggests that alternative pathways, possibly involving the activity of ornithine aminotransferase, may be active when the default reaction sequence to produce ornithine via acetylation of glutamate is disabled.
Die Etablierung der Transkription von kompletten Genen auf planaren Oberflächen soll eine Verbindung zwischen der Mikroarraytechnologie und der Transkriptomforschung herstellen. Darüber hinaus kann mit diesem Verfahren ein Brückenschlag zwischen der Synthese der Gene und ihrer kodierenden Proteine auf einer Oberfläche erfolgen. Alle transkribierten RNAs wurden mittels RT-PCR in cDNA umgeschrieben und in einer genspezifischen PCR amplifiziert. Die PCR-Produkte wurden hierfür entweder per Hand oder maschinell auf die Oberfläche transferiert. Über eine Oberflächen-PCR war es möglich, die Gensequenz des Reportergens EGFP direkt auf der Oberfläche zu synthetisieren und anschließend zu transkribieren. Somit war eine Transkription mit weniger als 1 ng an Matrize möglich. Der Vorteil einer Oberflächen-Transkription gegenüber der in Lösung liegt in der mehrfachen Verwendung der immobilisierten Matrize, wie sie in dieser Arbeit dreimal erfolgreich absolviert wurde. Die Oberflächen-Translation des EGFP-Gens konnte ebenfalls zweimal an einer immobilisierten Matrize gezeigt werden, wobei Zweifel über eine echte Festphasen-Translation nicht ausgeräumt werden konnten. Zusammenfassend kann festgestellt werden, dass die Transkription und Translation von immobilisierten Gensequenzen auf planaren Oberflächen möglich ist, wofür die linearen Matrizen direkt auf der Oberfläche synthetisiert werden können.
Daylength is one of several parameters controlling flowering time in many plant species. The day length is perceived in leaves, but how the floral signal is transduced to the shoot apex via the phloem to induce flowering remains to be elucidated. This study aimed at the identification of new candidates involved in the induction of flowering by employing three plant species, Arabidopsis thaliana, Sinapis alba and Brassica napus in combination with transcript profiling by Affymetrix chip hybridization, metabolite profiling by gas chromatography – mass spectrometry and targeted protein analysis using antibodies. All analyses were performed on tissue-specific samples and focused on phloem sap or phloem exudates. To find common transcript and metabolite candidates potentially associated with the floral transition, two independent induction systems in Arabidopsis were used: a photoextension system, whereby plants received fourteen additional hours of light, and a parallel dexamethasone-inducible system, which was centered on the induction of the known flowering gene CONSTANS (CO). Identification of signals preceding the CO cascade was possible using the light extension regime, while downstream events dependent on CO activation were compared in both systems. Altogether, a number of interesting transcript and metabolite candidates were identified in both systems with some degree of overlap. Sinapis alba was used to investigate the universality of the floral signals between species. Comparisons of metabolite data revealed a few common candidates that may prove interesting for further studies. In addition, a targeted approach was carried out to investigate the presence of the Flowering Locus T (FT) protein during different stages of flower development using an antibody. Interesting changes in the sizes of antigens from rape phloem were seen and appeared consistent in Arabidopsis and to a lesser extent in Sinapis. Overall, the broad surveying approaches for transcripts and metabolites used in this study revealed several new potential candidates involved in the induction and/or regulation of flowering. As far as the protein work, additional experiments will reveal the link between FT and floral induction as well as its role in maintaining the floral state using the abovementioned plant species.
The ultimate aim of this study is to better understand the relevance of weak electricity in the adaptive radiation of the African mormyrid fish. The chosen model taxon, the genus Campylomormyrus, exhibits a wide diversity of electric organ discharge (EOD) waveform types. Their EOD is age, sex, and species specific and is an important character for discriminating among species that are otherwise cryptic. After having established a complementary set of molecular markers, I examined the radiation of Campylomormyrus by a combined approach of molecular data (sequence data from the mitochondrial cytochrome b and the nuclear S7 ribosomal protein gene, as well as 18 microsatellite loci, especially developed for the genus Campylomormyrus), observation of ontogeny and diversification of EOD waveform, and morphometric analysis of relevant morphological traits. I built up the first convincing phylogenetic hypothesis for the genus Campylomormyrus. Taking advantage of microsatellite data, the identified phylogenetic clades proved to be reproductively isolated biological species. This way I detected at least six species occurring in sympatry near Brazzaville/Kinshasa (Congo Basin). By combining molecular data and EOD analyses, I could show that there are three cryptic species, characterised by their own adult EOD types, hidden under a common juvenile EOD form. In addition, I confirmed that adult male EOD is species-specific and is more different among closely related species than among more distantly related ones. This result and the observation that the EOD changes with maturity suggest its function as a reproductive isolation mechanism. As a result of my morphometric shape analysis, I could assign species types to the identified reproductively isolated groups to produce a sound taxonomy of the group. Besides this, I could also identify morphological traits relevant for the divergences between the identified species. Among them, the variations I found in the shape of the trunk-like snout, suggest the presence of different trophic specializations; therefore, this trait might have been involved in the ecological radiation of the group. In conclusion, I provided a convincing scenario envisioning an adaptive radiation of weakly electric fish triggered by sexual selection via assortative mating due to differences in EOD characteristics, but caused by a divergent selection of morphological traits correlated with the feeding ecology.
Charakterisierung von Transportmechanismen in der Speicheldrüse der Schabe Periplaneta americana

Die Aktivierung der Speichelsekretion erfolgt in der innervierten Speicheldrüse der Schabe Periplaneta americana durch die biogenen Amine Dopamin (DA) und Serotonin (5-HT). Die Acini der Speicheldrüse sezernieren einen Primärspeichel, der in den Ausführgängen modifiziert wird. Die durch DA und 5-HT aktivierten Signalwege sowie die an der Elektrolyt- und Flüssigkeitssekretion bzw. Speichel-modifikation beteiligten Transportmechanismen sind weitgehend unbekannt. Mikrofluorometrische Ca<sup>2+-, Na<sup>+- und pH-Messungen in Kombination mit pharmakologischen Experimenten, biochemische Messungen der Aktivitäten von Ionentransport-ATPasen sowie videomikroskopische Analysen zu transepithelialen Wasserbewegungen wurden in dieser Arbeit durchgeführt. Sie sollten Informationen über die an der Speichelbildung und -modifikation beteiligten Transportmechanismen und die Signalwege liefern, welche durch DA und/oder 5-HT aktiviert werden. Wesentliche Ergebnisse dieser Arbeit waren: <ul> <li>Messungen des intrazellulären pH (pHi) in Gangzellen zeigten, dass isolierte Ausführgänge mit Acini bei Stimulierung mit DA und 5-HT stark ansäuerten. In isolierten Ausführgängen ohne Acini verursachte nur DA eine schwache Ansäuerung. Da nur die Ausführgänge dopaminerg innerviert sind, die Acini jedoch dopaminerg und serotonerg, zeigt dieses Ergebnis, dass die DA- und/oder 5-HT-induzierte Primärspeichelbildung die Ursache für die pHi-Änderungen in den Gangzellen ist. pHi-Messungen in den Gangzellen geben also auch Hinweise auf Transportvorgänge in den Acini.</li> <li> Der Na<sup>+-K<sup>+-2Cl<sup>--Symporter und der Cl<sup>--HCO3<sup>--Antiporter, gekoppelt mit dem Na<sup>+ H<sup>+-Antiporter (NHE) waren an der NaCl-Aufnahme in die peripheren Zellen der Acini zur Bildung des NaCl-reichen Primärspeichels beteiligt. Die Aktivität dieser Transporter hing von der CO2/HCO3<sup>--Verfügbarkeit ab und war Ca<sup>2+-abhängig.</li> <li>Die starke Ansäuerung in den Gangzellen hing nicht von der Aktivität der apikalen vakuolären Protonen-ATPase (V-H<sup>+-ATPase), aber von der Aktivität der basolateralen Na<sup>+-K<sup>+-ATPase ab, die anscheinend in den Ausführgängen die Speichelmodifikation energetisiert.</li> <li>In isolierten Ausführgängen mit Acini waren die V-H<sup>+-ATPase und Na<sup>+-abhängige Transporter (u. a. NHE) an der Erholung von einer DA-induzierten oder einer NH4Cl-Vorpuls-induzierten Ansäuerung in den Gangzellen beteiligt. Bei der Regulation des pHi in unstimulierten Gangzellen spielten diese Transporter keine Rolle.</li> <li>In isolierten Ausführgängen mit Acini induzierte DA in den Gangzellen einen Anstieg der [Na<sup>+]i und, zeitlich verzögert, auch der [Ca<sup>2+]i. Der [Na<sup>+]i-Anstieg war von der Aktivität der Acini abhängig und erfolgte möglicherweise über apikale Na+-Kanäle. Der [Ca<sup>2+]i-Anstieg war graduiert und tonisch. Der DA-induzierte [Na<sup>+]i-Anstieg in den Gangzellen und deren Depolarisation führten dazu, dass der basolaterale Na<sup>+-Ca<sup>2+-Antiporter in den Ca<sup>2+-Influx-Modus umkehrte. Die daraus resultierende tonische [Ca<sup>2+]i-Erhöhung könnte an der Regulation der Na<sup>+-Rückresorption beteiligt sein.</li> <li>Zum Nachweis transepithelialer Flüssigkeitsbewegungen in isolierten Ausführgängen wurde eine videomikroskopische Methode entwickelt. Isolierte Ausführgänge ohne Acini resorbierten im unstimulierten Zustand Flüssigkeit aus dem Ausführganglumen. Möglicherweise sezernieren die Acini auch im unstimulierten Zustand mit geringerer Rate einen Primärspeichel, der in den Ausführgängen resorbiert wird. Die Resorption war ATP-abhängig. Der ATP-verbrauchende Transportmechanismus konnte nicht identifiziert werden. Weder die Na<sup>+-K<sup>+-ATPase noch die V-H<sup>+-ATPase waren an der Resorption beteiligt.</li> </ul> Diese Arbeit trug zur Kenntnis der komplexen Funktionsweise von Speicheldrüsen in Insekten bei und erweiterte das lückenhafte Wissen über die zellulären Wirkungen biogener Amine in Insekten. Zudem wurden in dieser Arbeit viele Parallelen zu Funktionsweisen der Speicheldrüsen in Vertebraten deutlich.
Die Enzymsuperfamilie der löslichen Sulfotransferasen (SULT) spielt eine wichtige Rolle in der Phase II des Fremdstoffmetabolismus. Sie katalysieren den Transfer einer Sulfonylgruppe auf nucleophile Gruppen endogener und exogener Substrate. Die Sulfokonjugation von Fremdstoffen erhöht deren Wasserlöslichkeit und behindert die passive Permeation von Zellmembranen. Dadurch wird die Ausscheidung dieser konjugierten Substanzen erleichtert. In Abhängigkeit von der Struktur des Zielmoleküls kann die Sulfokonjugation aber auch zur metabolischen Aktivierung von Fremdstoffen durch die Bildung instabiler Metabolite führen. Die SULT-vermittelte Aktivierung promutagener Substanzen ist somit von toxikologischem Interesse. Für die Detektion SULT-vermittelter Mutagenität mittels bakterieller in-vitro Testsysteme ist die heterologe Expression der fremdstoffmetabolisierenden Enzyme direkt in den Indikatorzellen notwendig. S. typhimurium exprimieren selbst keine SULT, und externe Metabolisierungssysteme sind problematisch, weil die negativ geladenen, kurzlebigen Metabolite nur schlecht die Zellmembran penetrieren können. Die Expression humaner Enyme in Bakterien ist jedoch zum Teil sehr kritisch. So zeigen z.B. sehr ähnliche Enzyme (SULT1A2*1 und *2) deutliche Unterschiede im Expressionsniveau bei exakt gleichen äußeren Bedingungen. Dies erschwert den Vergleich der enzymatischen Aktivitäten dieser Enzyme im in-vitro Testsystem. Andere Enzyme (z.B. SULT2B1b) werden unter Verwendung ihrer Wildtyp-cDNA zum Teil nicht detektierbar exprimiert. Deshalb sollte in dieser Arbeit eine Methode zur Optimierung der heterologen Expression fremdstoffmetabolisierender Enzyme für Genotoxizitätsuntersuchungen etabliert werden. Es wurde bereits gezeigt dass synonyme Codonaustausche am 5’-Ende der humanen SULT1A2-cDNA zu einer Erhöhung der Expression des entsprechenden Enzyms in S. typhimurium führten. Dementsprechend wurden in dieser Arbeit Codonaustausche am 5’-Ende der cDNA verschiedener SULT (1A1*1, 1A2*1, 2B1b) sowie der Ratten Glutathion-S-Transferase Theta 2 (rGSTT2) und dem Reportergen Luciferase durchgeführt. Die Expression der so generierten Konstrukte wurde in verschiedenen S. typhimurium und E. coli Stämmen quantifiziert und die Aktivität der überexprimierten Enzyme im Ames-Test bzw. im Enzym-Aktivitätsassay überprüft. Durch das Einführen seltener Codons in die cDNA konnte die Proteinexpression von SULT1A1*1, SULT1A2*1 und SULT2B1b maximal 7-fach, 18-fach und 100-fach im Vergleich zur Wildtyp-cDNA gesteigert werden. Die Expression der rGSTT2 wurde ebenfalls durch das Einführen seltener Codons erhöht (maximal 5-fach). Bei dem Reportergen Luciferase jedoch führte das Austauschen häufiger Codons gegen seltene Codons zu einer Reduktion der Proteinexpression um 80 %. Die Expression von Fusionsproteinen aus 2B1b (5’-Ende) und Luciferase (3’-Ende) wurde durch das Einführen seltener Codons ebenfalls um 50 % reduziert. Die S. typhimurium Stämme mit optimierter SULT 1A1*1- bzw. 1A2*1-Expression wurden im Ames-Test eingesetzt und zeigten im Vergleich zu den geringer exprimierenden Stämmen eine höhere Sensitivität. Für SULT2B1b konnte keine Mutagenaktivierung im Ames-Test nachgewiesen werden. Allerdings zeigte ein Enzym-Aktivitätsassay mit Dehydroepiandosteron, dass das bakteriell exprimierte Enzym funktionell war. Da in der Literatur der Effekt seltener Codons auf die Expression in Bakterien bisher fast ausschließlich als inhibitorisch beschrieben wurde, sollte die Wirkungsweise der hier beobachteten Expressionserhöhung durch seltene Codons genauer untersucht werden. Dazu wurden verschiedene Konstrukte der SULT1A2*1 und der SULT2B1b, die unterschiedlich viele seltene Codons in verschiedenen Kombinationen besaßen, hergestellt. Es konnten jedoch keine einzelnen Codons, die für die Expressionssteigerung allein verantwortlich waren, identifiziert werden. Die Plasmidkopienzahl in den verschiedenen SULT2B1b-Klonen war konstant und die SULT2B1b-mRNA-Konzentration zeigte nur moderate Schwankungen, die nicht als Ursache für die dramatische Erhöhung der SULT2B1b-Expression in Frage kommen. Die berechnete Stabilität der potentiellen mRNA-Sekundärstrukturen wurde durch die seltenen Codons häufig stark gesenkt und ist als eine mögliche Ursache für die Expressionssteigerung anzusehen. Zusätzlich erhöhten die seltenen Codons den Consensus der Downstream Box zur 16S rRNA, was ebenfalls eine Ursache für die Expressionssteigerung sein kann. In dieser Arbeit konnte somit die Expression der humanen SULT1A1*1, 1A2*1 und der 2B1b sowie der rGSTT2 erfolgreich mittels synonymer Codonaustausche erhöht werden. Die so optimierten S. typhimurium Stämme zeigten im Ames-Test eine erhöhte Sensitivität gegenüber SULT aktivierten Promutagenen bzw. erhöhte Aktivität in spezifischen Enymaktivitätsassays.
Die vorliegende Arbeit wurde im Zeitraum von Oktober 2002 bis November 2005 an dem Institut für Biochemie und Biologie der Universität Potsdam in Kooperation mit dem Institut für Chemie des GKSS Forschungszentrums in Teltow unter der Leitung von Herrn Prof. Dr. B. Micheel und Herrn Prof. Dr. Th. Groth angefertigt. Im Rahmen dieser Arbeit wurden die Wechselwirkungen von Immunzellen mit verschiedenen Kultursubstraten untersucht. Dafür wurden drei verschiedene Hybridomzelllinien eingesetzt. Eine Hybridomzelllinie (K2) ist im Laufe dieser Arbeit hergestellt und etabliert worden. Der Einsatz von synthetischen und proteinbeschichteten Kulturoberflächen führte bei Hybridomzellen zu einer deutlich gesteigerten Antikörpersynthese im Vergleich zu herkömmlichen Zellkulturmaterialien. Obwohl diese Zellen in der Regel als Suspensionszellen kultiviert werden, führten die eingesetzten Polymermembranen (PAN, NVP) zu einer verbesserten Antikörpersynthese (um 30%) gegenüber Polystyrol als Referenz. Es konnte gezeigt werden, dass es einen Zusammenhang zwischen der Produktivität und dem Adh asionsverhalten der Hybridomzellen gibt. Um den Einfluss von Proteinen der extrazellulären Matrix auf Zellwachstum und Antikörpersynthese von Hybridomzellen zu untersuchen, wurden proteinbeschichtete Polystyrol-Oberflächen eingesetzt. Für die Modifikationen wurden Fibronektin, Kollagen I, Laminin und BSA ausgewählt. Die Modifikation der Polystyrol-Oberfläche mit geringen Mengen Fibronektin (0,2-0,4 µg/ml) führte zu einer beträchtlichen Steigerung der Antikörpersynthese um 70-120%. Für Kollagen I- und BSA-Beschichtungen konnten Steigerungen von 40% beobachtet werden. Modifikationen der Polystyrol-Oberfläche mit Laminin zeigten nur marginale Effekte. Durch weitere Versuche wurde bestätigt, dass die Adhäsion der Zellen an Kollagen I- und Laminin-beschichteten Oberflächen verringert ist. Die alpha2-Kette des alpha2beta1-Integrins konnte auf der Zelloberfläche nicht nachgewiesen werden. Durch ihr Fehlen wird wahrscheinlich die Bindungsfähigkeit der Zellen an Kollagen I und Laminin beeinflusst. Durch die Ergebnisse konnte gezeigt werden, dass Hybridomzellen nicht nur Suspensionszellen sind und das Kultursubstrate das Zellwachstum und die Produktivität dieser Zellen stark beeinflussen können. Der Einsatz von synthetischen und proteinbeschichteten Kultursubstraten zur Steigerung der Antikörpersynthese kann damit für die industrielle Anwendung von großer Relevanz sein. Für die Modellierung einer Lymphknotenmatrix wurden Fibronektin, Kollagen I, Heparansulfat und N-Acetylglucosamin-mannose in verschiedenen Kombinationen an Glasoberflächen adsorbiert und für Versuche zur In-vitro-Immunisierung eingesetzt. Es konnte gezeigt werden, dass die Modifikation der Oberflächen die Aktivierung und Interaktion von dendritischen Zellen, T- und B-Lymphozyten begünstigt, was durch den Nachweis spezifischer Interleukine (IL12, IL6) und durch die Synthese spezifischer Antikörper bestätigt wurde. Eine spezifische Immunreaktion gegen das Antigen Ovalbumin konnte mit den eingesetzten Zellpopulationen aus Ovalbumin-T-Zell-Rezeptor-transgenen Mäusen nachgewiesen werden. Die In-vitro-Immunantwort wurde dabei am stärksten durch eine Kombination von Kollagen I, Heparansulfat und N-Acetylglucosamin-mannose auf einer Glasoberfläche gefördert. Die Etablierung einer künstlichen Immunreaktion kann eine gesteuerte Aktivierung bzw. Inaktivierung von körpereigenen dendritischen Zellen gegen bestehende Krankheitsmerkmale in vitro ermöglichen. Durch die Versuche wurden Grundlagen für spezifische Immunantworten erarbeitet, die u.a. für die Herstellung von humanen Antikörpern eingesetzt werden können.
Flüssigkeitssekretion und Proteinsekretion werden in Speicheldrüsen von Insekten über Hormone und Neurotransmitter gesteuert. Diese entfalten ihre physiologische Wirkung in den sekretorischen Drüsenzellen hauptsächlich über den zyklischen Adenosinmonophosphat (cAMP)-Signalweg und den Inositoltrisphosphat (IP<SUB>3</SUB>) / Ca<sup>2+-Signalweg. Die Mechanismen möglicher Wechselwirkungen zwischen diesen Signalwegen und ihre physiologischen Auswirkungen sind unzureichend bekannt. Im Mittelpunkt dieser Arbeit stand die Frage, ob und wie sich der Ca<sup>2+-Signalweg und der cAMP-Signalweg in der Speicheldrüse der Diptere Calliphora vicina beeinflussen. Substanzen wie 5-Fluoro-α-Methyltryptamin und Histamin wurden in früheren Arbei-ten als Agonisten genutzt, um in den Speicheldrüsen von C. vicina selektiv den cAMP-Signalweg (getrennt vom IP<SUB>3</SUB>/Ca<sup>2+-Signalweg) zu aktivieren. Es zeigte sich in transepithelialen Potentialmessungen und mikrofluorometrischen Ca<sup>2+-Untersuchungen, dass beide Substanzen sowohl den cAMP-Weg als auch den Ca<sup>2+-Signalweg aktivierten. Die physiologischen Ursachen der Histamin-induzierten Ca<sup>2+-Erhöhung wurden genauer untersucht. Zusammengefasst zeigten diese Untersuchungen, dass Histamin wie 5-HT den cAMP-Weg und die Phosphoinositidkaskade aktivierte. Im Gegensatz zu den 5-HT-induzierten Ca<sup>2+-Oszillationen, welche durch interzelluläre Ca<sup>2+-Wellen synchronisiert werden, verursachte Histamin bei niedrigen Konzentrationen lokale Ca<sup>2+-Oszillationen in einzelnen Zellen (keine Wellen). Bei höheren Histamin-Konzentrationen war eine anhaltende Ca<sup>2+-Erhöhung oder ein synchrones <quote>Ca<sup>2+-beating</quote> in der gesamten Drüse zu beobachten. Des Weiteren wurde die Frage untersucht, ob eine Erhöhung der intrazellulären cAMP-Konzentration den IP<SUB>3</SUB> Ca<sup>2+-Signalweg in den Epithelzellen der Speicheldrüse beeinflussen kann. Es zeigte sich, dass cAMP den durch schwellennahe 5-HT-Konzentrationen induzierten Ca<sup>2+-Anstieg verstärkte. Diese Verstärkung wurde durch eine PKA-vermittelte Sensitivierung des IP<SUB>3</SUB>-Rezeptor/Ca<sup>2+-Kanals für IP<SUB>3</SUB> verursacht. Immunzytochemische Untersuchungen deuten dar-auf hin, dass die Proteinkinase A eng mit dem IP<SUB>3</SUB>-Rezeptor/Ca<sup>2+-Kanal assoziiert ist. Diese Messungen zeigen erstmals, dass auch bei Invertebraten der Botenstoff cAMP, PKA-vermittelt, den IP<SUB>3</SUB>-Rezeptor/Ca<sup>2+-Kanal des ER für IP<SUB>3</SUB> sensitiviert.
Kaliumionen (K<sup>+) sind die am häufigsten vorkommenden anorganischen Kationen in Pflanzen. Gemessen am Trockengewicht kann ihr Anteil bis zu 10% ausmachen. Kaliumionen übernehmen wichtige Funktionen in verschiedenen Prozessen in der Pflanze. So sind sie z.B. essentiell für das Wachstum und für den Stoffwechsel. Viele wichtige Enzyme arbeiten optimal bei einer K<sup>+ Konzentration im Bereich von 100 mM. Aus diesem Grund halten Pflanzenzellen in ihren Kompartimenten, die am Stoffwechsel beteiligt sind, eine kontrollierte Kaliumkonzentration von etwa 100 mM aufrecht. Die Aufnahme von Kaliumionen aus dem Erdreich und deren Transport innerhalb der Pflanze und innerhalb einer Pflanzenzelle wird durch verschiedene Kaliumtransportproteine ermöglicht. Die Aufrechterhaltung einer stabilen K<sup>+ Konzentration ist jedoch nur möglich, wenn die Aktivität dieser Transportproteine einer strikten Kontrolle unterliegt. Die Prozesse, die die Transportproteine regulieren, sind bis heute nur ansatzweise verstanden. Detailliertere Kenntnisse auf diesem Gebiet sind aber von zentraler Bedeutung für das Verständnis der Integration der Transportproteine in das komplexe System des pflanzlichen Organismus. In dieser Habilitationsschrift werden eigene Publikationen zusammenfassend dargestellt, in denen die Untersuchungen verschiedener Regulationsmechanismen pflanzlicher Kaliumkanäle beschrieben werden. Diese Untersuchungen umfassen ein Spektrum aus verschiedenen proteinbiochemischen, biophysikalischen und pflanzenphysiologischen Analysen. Um die Regulationsmechanismen grundlegend zu verstehen, werden zum einen ihre strukturellen und molekularen Besonderheiten untersucht. Zum anderen werden die biophysikalischen und reaktionskinetischen Zusammenhänge der Regulationsmechanismen analysiert. Die gewonnenen Erkenntnisse erlauben eine neue, detailliertere Interpretation der physiologischen Rolle der Kaliumtransportproteine in der Pflanze.
In silico identification of genes regulated by abscisic acid in Arabidopsis thaliana (L.) Heynh.

Abscisic acid (ABA) is a major plant hormone that plays an important role during plant growth and development. During vegetative growth ABA mediates (in part) responses to various environmental stresses such as cold, drought and high salinity. The response triggered by ABA includes changes in the transcript level of genes involved in stress tolerance. The aim of this project was the In silico identification of genes putatively regulated by ABA in A. thaliana. In silico predictions were combined with experimental data in order to evaluate the reliability of computational predictions. Taking advantage of the genome sequence of A. thaliana publicly available since 2000, 1 kb upstream sequences were screened for combinations of cis-elements known to be involved in the regulation of ABA-responsive genes. It was found that around 10 to 20 percent of the genes of A. thaliana might be regulated by ABA. Further analyses of the predictions revealed that certain combinations of cis-elements that confer ABA-responsiveness were significantly over-represented compared with results in random sequences and with random expectations. In addition, it was observed that other combinations that confer ABA-responsiveness in monocotyledonous species might not be functional in A. thaliana. It is proposed that ABA-responsive genes in A. thaliana show pairs of ABRE (abscisic acid responsive element) with MYB binding sites, DRE (dehydration responsive element) or with itself. The analysis of the distances between pairs of cis-elements suggested that pairs of ABREs are bound by homodimers of ABRE binding proteins. In contrast, pairs between MYB binding sites and ABRE, or DRE and ABRE showed a distance between cis-elements that suggested that the binding proteins interact through protein complexes and not directly. The comparison of computational predictions with experimental data confirmed that the regulatory mechanisms leading to the induction or repression of genes by ABA is very incompletely understood. It became evident that besides the cis-elements proposed in this study to be present in ABA-responsive genes, other known and unknown cis-elements might play an important role in the transcriptional regulation of ABA-responsive genes. For example, auxin-related cis elements, or the cis-elements recognized by the NAM-family of transcription factors (Non-Apical meristem). This work documents the use of computational and experimental approaches to analyse possible interactions between cis-elements involved in the regulation of ABA-responsive genes. The computational predictions allowed the distinction between putatively relevant combinations of cis-elements from irrelevant combinations of cis-elements in ABA-responsive genes. The comparison with experimental data allowed to identify certain cis-elements that have not been previously associated to the ABA-mediated transcriptional regulation, but that might be present in ABA-responsive genes (e.g. auxin responsive elements). Moreover, the efforts to unravel the gene regulatory network associated with the ABA-signalling pathway revealed that NAM-transcription factors and their corresponding binding sequences are important components of this network.
The advent of large-scale and high-throughput technologies has recently caused a shift in focus in contemporary biology from decades of reductionism towards a more systemic view. Alongside the availability of genome sequences the exploration of organisms utilizing such approach should give rise to a more comprehensive understanding of complex systems. Domestication and intensive breeding of crop plants has led to a parallel narrowing of their genetic basis. The potential to improve crops by conventional breeding using elite cultivars is therefore rather limited and molecular technologies, such as marker assisted selection (MAS) are currently being exploited to re-introduce allelic variance from wild species. Molecular breeding strategies have mostly focused on the introduction of yield or resistance related traits to date. However given that medical research has highlighted the importance of crop compositional quality in the human diet this research field is rapidly becoming more important. Chemical composition of biological tissues can be efficiently assessed by metabolite profiling techniques, which allow the multivariate detection of metabolites of a given biological sample. Here, a GC/MS metabolite profiling approach has been applied to investigate natural variation of tomatoes with respect to the chemical composition of their fruits. The establishment of a mass spectral and retention index (MSRI) library was a prerequisite for this work in order to establish a framework for the identification of metabolites from a complex mixture. As mass spectral and retention index information is highly important for the metabolomics community this library was made publicly available. Metabolite profiling of tomato wild species revealed large differences in the chemical composition, especially of amino and organic acids, as well as on the sugar composition and secondary metabolites. Intriguingly, the analysis of a set of S. pennellii introgression lines (IL) identified 889 quantitative trait loci of compositional quality and 326 yield-associated traits. These traits are characterized by increases/decreases not only of single metabolites but also of entire metabolic pathways, thus highlighting the potential of this approach in uncovering novel aspects of metabolic regulation. Finally the biosynthetic pathway of the phenylalanine-derived fruit volatiles phenylethanol and phenylacetaldehyde was elucidated via a combination of metabolic profiling of natural variation, stable isotope tracer experiments and reverse genetic experimentation.
Weltweit versuchen Wissenschaftler, künstliche Viren für den Gentransfer zu konstruieren, die nicht reproduktionsfähig sind. Diese sollen die Vorteile der natürlichen Viren besitzen (effizienter Transport von genetischem Material), jedoch keine Antigene auf ihrer Oberfläche tragen, die Immunreaktionen auslösen. Ziel dieses Projektes ist es, einen künstlichen Viruspartikel herzustellen, dessen Basis eine Polyelektrolytenhohlkugel bildet, die mit einer Lipiddoppelschicht bedeckt ist. Um intakte Doppelschichten zu erzeugen, muss die Wechselwirkung zwischen Lipid und Polyelektrolyt (z.B. DNA) verstanden und optimiert werden. Dazu ist es notwendig, die strukturelle Grundlage der Interaktion aufzuklären. Positiv geladene Lipide gehen zwar starke Wechselwirkungen mit der negativ geladenen DNA ein, sie wirken jedoch toxisch auf biologische Zellen. In der vorliegenden Arbeit wurde daher die durch zweiwertige Kationen vermittelte Kopplung von genomischer oder Plasmid-DNA an zwitterionische oder negativ geladene Phospholipide an zwei Modellsystemen untersucht. 1. Modellsystem: Lipidmonoschicht an der Wasser/Luft-Grenzfläche Methoden: Filmwaagentechnik in Kombination mit IR-Spektroskopie (IRRAS), Röntgenreflexion (XR), Röntgendiffraktion (GIXD), Brewsterwinkel-Mikroskopie (BAM), Röntgenfluoreszenz (XRF) und Oberflächenpotentialmessungen Resultate: A) Die Anwesenheit der zweiwertigen Kationen Ba2+, Mg2+, Ca2+ oder Mn2+ in der Subphase hat keinen nachweisbaren Einfluss auf die Struktur der zwitterionischen DMPE- (1,2-Dimyristoyl-phosphatidyl-ethanolamin) Monoschicht. B) In der Subphase gelöste DNA adsorbiert nur in Gegenwart dieser Kationen an der DMPE-Monoschicht. C) Sowohl die Adsorption genomischer Kalbsthymus-DNA als auch der Plasmid-DNA pGL3 bewirkt eine Reduktion des Neigungswinkels der Alkylketten, die auf einen veränderten Platzbedarf der Kopfgruppe zurückzuführen ist. Durch die Umorientierung der Kopfgruppe wird die elektrostatische Wechselwirkung zwischen den positiv geladenen Stickstoffatomen der Lipidkopfgruppen und den negativ geladenen DNA-Phosphaten erhöht. D) Die adsorbierte DNA weist eine geordnete Struktur auf, wenn sie durch Barium-, Magnesium-, Calcium- oder Manganionen komplexiert ist. Der Abstand zwischen parallelen DNA-Strängen hängt dabei von der Größe der DNA-Fragmente sowie von der Art des Kations ab. Die größten Abstände ergeben sich mit Bariumionen, gefolgt von Magnesium- und Calciumionen. Die kleinsten DNA-Abstände werden durch Komplexierung mit Manganionen erhalten. Diese Ionenreihenfolge stellt sich sowohl für genomische DNA als auch für Plasmid-DNA ein. E) Die DNA-Abstände werden durch die Kompression des Lipidfilms nicht beeinflusst. Zwischen der Lipidmonoschicht und der adsorbierten DNA besteht demnach nur eine schwache Wechselwirkung. Offensichtlich befindet sich die durch zweiwertige Kationen komplexierte DNA als weitgehend eigenständige Schicht unter dem Lipidfilm. 2. Modellsystem: Lipiddoppelschicht an der fest/flüssig-Grenzfläche Methoden: Neutronenreflexion (NR) und Quarzmikrowaage (QCM-D) Resultate: A) Das zwitterionische Phospholipid DMPC (1,2-Dimyristoyl-phosphatidylcholin) bildet keine Lipiddoppelschicht auf planaren Polyelektrolytmultischichten aus, deren letzte Lage das positiv geladene PAH (Polyallylamin) ist. B) Hingegen bildet DMPC auf dem negativ geladenen PSS (Polystyrolsulfonat) eine Doppelschicht aus, die jedoch Defekte aufweist. C) Eine Adsorption von genomischer Kalbsthymus-DNA auf dieser Lipidschicht findet nur in Gegenwart von Calciumionen statt. Andere zweiwertige Kationen wurden nicht untersucht. D) Das negativ geladene Phospholipid DLPA (1,2-Dilauryl-phosphatidsäure) bildet auf dem positiv geladenen PAH eine Lipiddoppelschicht aus, die Defekte aufweist. E) DNA adsorbiert ebenfalls erst in Anwesenheit von Calciumionen in der Lösung an die DLPA-Schicht. F) Durch die Zugabe von EDTA (Ethylendiamintetraessigsäure) werden die Calciumionen dem DLPA/DNA-Komplex entzogen, wodurch dieser dissoziiert. Demnach ist die calciuminduzierte Bildung dieser Komplexe reversibel.
Biogene Amine sind kleine organische Verbindungen, die sowohl bei Wirbeltieren als auch bei Wirbellosen als Neurotransmitter, Neuromodulatoren und/oder Neurohormone wirken können. Sie bilden eine bedeutende Gruppe von Botenstoffen und entfalten ihre Wirkungen über die Bindung an eine bestimmte Klasse von Rezeptorproteinen, die als G-Protein-gekoppelte Rezeptoren bezeichnet werden. Bei Insekten gehören zur Substanzklasse der biogenen Amine die Botenstoffe Dopamin, Tyramin, Octopamin, Serotonin und Histamin. Neben vielen anderen Wirkung ist z.B. gezeigt worden, daß einige dieser biogenen Amine bei der Honigbiene (Apis mellifera) die Geschmacksempfindlichkeit für Zuckerwasser-Reize modulieren können. Ich habe verschiedene Aspekte der aminergen Signaltransduktion an den „Modellorganismen“ Honigbiene und Amerikanische Großschabe (Periplaneta americana) untersucht. Aus der Honigbiene, einem „Modellorganismus“ für das Studium von Lern- und Gedächtnisvorgängen, wurden zwei Dopamin-Rezeptoren, ein Tyramin-Rezeptor, ein Octopamin-Rezeptor und ein Serotonin-Rezeptor charakterisiert. Die Rezeptoren wurden in kultivierten Säugerzellen exprimiert, um ihre pharmakologischen und funktionellen Eigenschaften (Kopplung an intrazelluläre Botenstoffwege) zu analysieren. Weiterhin wurde mit Hilfe verschiedener Techniken (RT-PCR, Northern-Blotting, in situ-Hybridisierung) untersucht, wo und wann während der Entwicklung die entsprechenden Rezeptor-mRNAs im Gehirn der Honigbiene exprimiert werden. Als Modellobjekt zur Untersuchung der zellulären Wirkungen biogener Amine wurden die Speicheldrüsen der Amerikanischen Großschabe genutzt. An isolierten Speicheldrüsen läßt sich sowohl mit Dopamin als auch mit Serotonin Speichelproduktion auslösen, wobei Speichelarten unterschiedlicher Zusammensetzung gebildet werden. Dopamin induziert die Bildung eines völlig proteinfreien, wäßrigen Speichels. Serotonin bewirkt die Sekretion eines proteinhaltigen Speichels. Die Serotonin-induzierte Proteinsekretion wird durch eine Erhöhung der Konzentration des intrazellulären Botenstoffs cAMP vermittelt. Es wurden die pharmakologischen Eigenschaften der Dopamin-Rezeptoren der Schaben-Speicheldrüsen untersucht sowie mit der molekularen Charakterisierung putativer aminerger Rezeptoren der Schabe begonnen. Weiterhin habe ich das ebony-Gen der Schabe charakterisiert. Dieses Gen kodiert für ein Enzym, das wahrscheinlich bei der Schabe (wie bei anderen Insekten) an der Inaktivierung biogener Amine beteiligt ist und im Gehirn und in den Speicheldrüsen der Schabe exprimiert wird.
Die Honigbiene Apis mellifera gilt seit langem als Modell-Organismus zur Untersuchung von Lern- und Gedächtnisvorgängen sowie zum Studium des Sozialverhaltens und der Arbeitsteilung. Bei der Steuerung und Regulation dieser Verhaltensweisen spielt das Indolalkylamin Serotonin eine wesentliche Rolle. Serotonin entfaltet seine Wirkung durch die Bindung an G-Protein-gekoppelte Rezeptoren (GPCRs). In der vorliegenden Arbeit wird der erste Serotonin-Rezeptor aus der Honigbiene molekular charakterisiert. Durch die Anwendung zwei verschiedener Klonierungsstrategien konnten drei cDNA-Sequenzen isoliert werden, die für potentielle Serotonin-Rezeptoren kodieren. Die Sequenzen weisen die größte Ähnlichkeit zu dem 5-HT7- und 5-HT2-Rezeptor von Drosophila melanogaster bzw. dem 5-HT1-Rezeptor von Panulirus interruptus auf. Die isolierten Serotonin-Rezeptoren der Honigbiene wurden dementsprechend Am(Apis mellifera)5-HT1, Am5-HT2 und Am5-HT7 benannt. Das Hydropathieprofil des Am5-HT1-, Am5-HT2- und Am5-HT7-Rezeptors deutet auf das Vorhandensein des charakteristischen heptahelikalen Aufbaus G-Protein-gekoppelter Rezeptoren hin. Die abgeleiteten Aminosäuresequenzen zeigen typische Merkmale biogener Amin-Rezeptoren. Aminosäuren, die eine Bedeutung bei der Bildung der Liganden-Bindungstasche, der Rezeptor-Aktivierung und der Kopplung eines G-Proteins an den Rezeptor haben, sind in allen drei Rezeptoren konserviert. Interessanterweise ist jedoch das in den meisten biogenen Amin-Rezeptoren vorhandene DRY-Motiv in dem Am5-HT2- und Am5-HT7-Rezeptor nicht konserviert. Das Vorhandensein einer PDZ-Domäne in dem Am5-HT1- und Am5-HT7-Rezeptor lässt vermuten, dass diese Rezeptoren als Adapterproteine fungieren, die Signalmoleküle zu einem Signaltransduktionskomplex vereinigen. RT-PCR-Experimente zeigen die Expression der Rezeptoren in verschiedenen Geweben der Honigbiene. Auffallend ist die hohe Expression im Zentralgehirn. Des Weiteren konnte die Expression der Serotonin-Rezeptoren in den optischen Loben, Antennalloben sowie in der Peripherie, d.h. in der Flugmuskulatur und den Malpighischen Gefäßen nachgewiesen werden. Durch in situ Hybridisierungen wurde die Expression in Gefrierschnitten von Gehirnen adulter Sammlerinnen im Detail untersucht. Transkripte der Rezeptoren sind in den Somata von intrinsischen Pilzkörperzellen, Neuronen der optischen Loben und Neuronen der Antennalloben vorhanden. In einem heterologen Expressionssystem wurde der intrazelluläre Signalweg des Am5-HT7-Rezeptors untersucht. Die Aktivierung des stabil exprimierten Rezeptors durch Serotonin führt zur Bildung von cAMP. Der 5-HT7-Rezeptor spezifische Agonist 5-CT zeigt eine mit Serotonin vergleichbare Fähigkeit, die intrazelluläre cAMP-Konzentration zu erhöhen. Am5-HT7 gehört daher funktionell zu der Gruppe der 5-HT7-Rezeptoren. Der EC50-Wert von 1,06~nM (5-HT), ist im Vergleich zu anderen 5-HT7-Rezeptoren äußert niedrig. Des Weiteren wurde gezeigt, dass das basale cAMP-Niveau in den transfizierten Zellen im Vergleich zu nicht transfizierten Zellen deutlich erhöht ist. Das heißt, dass der Rezeptor auch in der Abwesenheit eines Liganden aktiv ist. Diese konstitutive Aktivität ist auch von anderen biogenen Amin-Rezeptoren bekannt. Methiothepin wurde als wirksamer inverser Agonist des Am5-HT7-Rezeptors identifiziert, da es in der Lage ist, der konstitutiven Aktivität entgegenzuwirken. Die Ergebnisse der vorliegenden Arbeit deuten darauf hin, dass die Serotonin-Rezeptoren in verschiedenen Regionen des ZNS der Honigbiene an der Informationsverarbeitung beteiligt sind. Es kann eine Beeinflussung von Lern- und Gedächtnisprozessen sowie des olfaktorischen und visuellen Systems durch diese Rezeptoren vermutet werden. Mit der Klonierung und funktionellen Charakterisierung des ersten Serotonin-Rezeptors der Honigbiene ist eine Grundlage für die Untersuchung der molekularen Mechanismen der serotonergen Signaltransduktion geschaffen worden.
In semi-arid savannas, unsustainable land use can lead to degradation of entire landscapes, e.g. in the form of shrub encroachment. This leads to habitat loss and is assumed to reduce species diversity. In BIOTA phase 1, we investigated the effects of land use on population dynamics on farm scale. In phase 2 we scale up to consider the whole regional landscape consisting of a diverse mosaic of farms with different historic and present land use intensities. This mosaic creates a heterogeneous, dynamic pattern of structural diversity at a large spatial scale. Understanding how the region-wide dynamic land use pattern affects the abundance of animal and plant species requires the integration of processes on large as well as on small spatial scales. In our multidisciplinary approach, we integrate information from remote sensing, genetic and ecological field studies as well as small scale process models in a dynamic region-wide simulation tool. <hr> Interdisziplinäres Zentrum für Musterdynamik und Angewandte Fernerkundung Workshop vom 9. - 10. Decisions for the conservation of biodiversity and sustainable management of natural resources are typically related to large scales, i.e. the landscape level. However, understanding and predicting the effects of land use and climate change on scales relevant for decision-making requires to include both, large scale vegetation dynamics and small scale processes, such as soil-plant interactions. Integrating the results of multiple BIOTA subprojects enabled us to include necessary data of soil science, botany, socio-economics and remote sensing into a high resolution, process-based and spatially-explicit model. Using an example from a sustainably-used research farm and a communally used and degraded farming area in semiarid southern Namibia we show the power of simulation models as a tool to integrate processes across disciplines and scales.
Mit der politischen Wende in den Staaten des ehemaligen Ostblockes wurde für viele militärisch genutzte Flächen ein tiefgreifender Nutzungswandel eingeleitet. Truppenübungsplätze als stark gestörte Bestandteile unserer Kulturlandschaft weisen auf großen Flächen naturschutzfachlich wertvolle Habitatmosaike mit speziellen Lebensgemeinschaften auf. Der Nutzungswandel ist mit einer Veränderung der Vegetationsstrukturen (Sukzession) und weiteren landschaftsökologischen Prozessen verbunden. Der ehemalige Truppenübungsplatz Döberitz im Norden der Landeshauptstadt Potsdam kann auf eine lange militärische Nutzungsgeschichte verweisen (erste Manöver des Soldatenkönigs im Jahr 1713). Nach 1992 wurden das NSG Döberitzer Heide (3.415 ha) und das NSG Ferbitzer Bruch (1.155 ha) ausgewiesen. Als Schutzgebiete nach der Vogelschutzrichtlinie sind sie Bestandteile des kohärenten Schutzgebietssystems Natura 2000 der europäischen Gemeinschaft. Trotz des Schutzstatus und der militärischen Altlasten unterliegt das Gebiet als größte zusammenhängende Naturfläche im engeren Verflechtungsraum des Landes Brandenburg einem hohen Nutzungsdruck. <hr> Interdisziplinäres Zentrum für Musterdynamik und Angewandte Fernerkundung Workshop vom 9. - 10. From its first use in the field of biochemistry, instrumental analysis offered a variety of invaluable tools for the comprehensive description of biological systems. Multi-selective methods that aim to cover as many endogenous compounds as possible in biological samples use different analytical platforms and include methods like gene expression profile and metabolite profile analysis. The enormous amount of data generated in application of profiling methods needs to be evaluated in a manner appropriate to the question under investigation. The new field of system biology rises to the challenge to develop strategies for collecting, processing, interpreting, and archiving this vast amount of data; to make those data available in form of databases, tools, models, and networks to the scientific community. On the background of this development a multi-selective method for the determination of phytohormones was developed and optimised, complementing the profile analyses which are already in use (Chapter I). The general feasibility of a simultaneous analysis of plant metabolites and phytohormones in one sample set-up was tested by studies on the analytical robustness of the metabolite profiling protocol. The recovery of plant metabolites proved to be satisfactory robust against variations in the extraction protocol by using common extraction procedures for phytohormones; a joint extraction of metabolites and hormones from plant tissue seems practicable (Chapter II). Quantification of compounds within the context of profiling methods requires particular scrutiny (Chapter II). In Chapter III, the potential of stable-isotope in vivo labelling as normalisation strategy for profiling data acquired with mass spectrometry is discussed. First promising results were obtained for a reproducible quantification by stable-isotope in vivo labelling, which was applied in metabolomic studies. In-parallel application of metabolite and phytohormone analysis to seedlings of the model plant Arabidopsis thaliana exposed to sulfate limitation was used to investigate the relationship between the endogenous concentration of signal elements and the ‘metabolic phenotype’ of a plant. An automated evaluation strategy was developed to process data of compounds with diverse physiological nature, such as signal elements, genes and metabolites – all which act in vivo in a conditional, time-resolved manner (Chapter IV). Final data analysis focussed on conditionality of signal-metabolome interactions.
The major aim of this work was the identification of new phloem sap proteins and a metabolic characterisation of this transport fluid. The experiments were performed on the three plant species C. sativus, C. maxima and B. napus. To characterise the phloem samples from B. napus, a new model plant for phloem analysis, western blot tests together with metabolite profiling were performed. GC-MS metabolite profiling and enzyme assays were used for measuring metabolites in the phloem of B. napus. Results from the phloem sap measurements showed, as expected, a typical sugar distribution for apoplasmic phloem loaders with sucrose being the predominant sugar. In stem extracts, the most abundant sugar was glucose with much lower fructose and sucrose levels. With the GC-MS approach it was possible to identify a number of metabolites which showed a differential distribution when phloem and stem tissue extracts were compared. For protein identification, two different approaches were employed (i) screening expression libraries with total phloem protein specific antisera and (ii) protein separation on 2 DE gels followed by ESI-MS/MS sequence analyses. For the first approach, three different phloem protein-specific antisera were produced and expression libraries were constructed. Phloem protein antisera were tested for specificity and some attempts to estimate specific epitopes were undertaken. Screening of the libraries resulted in the identification of 14 different proteins from all investigated species. Analyses of B. napus phloem sap proteins from 2 DE with ESI-MS/MS resulted in the identification of 5 different proteins. The phloem localisation of the identified proteins was additionally confirmed by western blot tests using specific antibodies. In order to functionally characterise some selected phloem proteins from B. napus, the group of potential calcium-binding polypeptides was analysed for functional Ca<sup>+2 binding properties and several Ca<sup>+2–binding proteins could be isolated. However, their sequences could as yet not be determined. Another approach used for functional protein characterisation was the analysis of Arabidopsis T-DNA insertion mutants. Four available mutants with insertions in phloem protein-specific genes were chosen from the SALK and GABI-Kat collections and selected homozygous lines were tested for the presence of the investigated proteins. In order to verify if the product of one of the mutated gene (GRP 7) is transported through the phloem, grafting experiments were performed followed by western blot analyses. Although the employed antiserum against GRP 7 protein did not allow distinguishing between the mutant and the wild type plants, successful Arabidopsis grafting could be established as a promising method for further studies on protein translocation through the phloem.
Das Borna Disease Virus (BDV, Bornavirus) besitzt ein einzelsträngiges RNA-Genom negativer Polarität und ist innerhalb der Ordnung Mononegavirales der Prototyp einer eigenen Virusfamilie, die der Bornaviridae. Eine außergewöhnliche Eigenschaft des Virus ist seine nukleäre Transkription und Replikation, eine weitere besteht in seiner Fähigkeit, als neurotropes Virus sowohl in vivo als auch in vitro persistente Infektionen zu etablieren. Die zugrunde liegenden Mechanismen sowohl der Replikation als auch der Persistenz sind derzeit noch unzureichend verstanden, auch deshalb, weil das Virus noch relativ „jung“ ist: Erste komplette Sequenzen des RNA-Genoms wurden 1994 publiziert und erst vor einigen Monaten gelang die Generierung rekombinanter Viren auf der Basis klonierter cDNA. Im Mittelpunkt dieser Arbeit standen das p10 Protein und das Phosphoprotein (P), die von der gemeinsamen Transkriptionseinheit II in überlappenden Leserahmen kodiert werden. Als im Kern der Wirtszelle replizierendes Virus ist das Bornavirus auf zelluläre Importmechanismen angewiesen, um den Kernimport aller an der Replikation beteiligten viralen Proteine zu gewährleisten. Das p10 Protein ist ein negativer Regulator der viralen RNA-abhängigen RNA-Polymerase (L). In vitro Importexperimente zeigten, dass p10 über den klassischen Importin alpha/beta abhängigen Kernimportweg in den Nukleus transportiert wird. Dies war unerwartet, da p10 kein vorhersagbares klassisches Kernlokalisierungssignal (NLS) besitzt und weist darauf hin, dass der zelluläre Importapparat offensichtlich flexibler ist als allgemein angenommen. Die ersten 20 N-terminalen AS vermitteln sowohl Kernimport als auch die Bindung an den Importrezeptor Importin alpha. Durch Di-Alanin-Austauschmutagenese wurden die für diesen Transportprozess essentiellen AS identifiziert und die Bedeutung hydrophober und polarer AS-Reste demonstriert. Die Fähigkeit des Bornavirus, persistente Infektionen zu etablieren, wirft die Frage auf, wie das Virus die zellulären antiviralen Abwehrmechanismen, insbesondere das Typ I Interferon (IFN)-System, unterwandert. Das virale P Protein wurde in dieser Arbeit als potenter Antagonist der IFN-Induktion charakterisiert. Es verhindert die Phosphorylierung des zentralen Transkriptionsfaktors IRF3 durch die zelluläre Kinase TBK1 und somit dessen Aktivierung. Der Befund, dass P mit TBK1 Komplexe bildet und zudem auch als Substrat für die zelluläre Kinase fungiert, erlaubt es, erstmalig einen Mechanismus zu postulieren, in dem ein virales Protein (BDV-P) als putatives TBK1-Pseudosubstrat die IRF3-Aktivierung kompetitiv hemmt.
Die Tailspike Proteine (TSP) der Bakteriophagen P22, Sf6 und HK620 dienen der Erkennung von Kohlenhydratstrukturen auf ihren gram-negativen Wirtsbakterien und zeigen, von den ersten 110 Aminosäuren des N-Terminus abgesehen, keine Sequenzübereinstimmung. Mit Röntgenkristallstrukturanalyse konnte gezeigt werden, dass HK620TSP und Sf6TSP ebenfalls zu einer parallelen, rechtsgängigen beta-Helix falten, wie dies schon für P22TSP bekannt war. Die Kohlenhydratbindestelle ist bei Sf6TSP im Vergleich zu P22TSP zwischen die Untereinheiten verschoben.
This thesis aimed to investigate several fundamental and perplexing questions relating to the phloem loading and transport mechanisms of Cucurbita maxima, by combining metabolomic analysis with cell biological techniques. This putative symplastic loading species has long been used for experiments on phloem anatomy, phloem biochemistry, phloem transport physiology and phloem signalling. Symplastic loading species have been proposed to use a polymer trapping mechanism to accumulate RFO (raffinose family oligosaccharides) sugars to build up high osmotic pressure in minor veins which sustains a concentration gradient that drives mass flow. However, extensive evidence indicating a low sugar concentration in their phloem exudates is a long-known problem that conflicts with this hypothesis. Previous metabolomic analysis shows the concentration of many small molecules in phloem exudates is higher than that of leaf tissues, which indicates an active apoplastic loading step. Therefore, in the view of the phloem metabolome, a symplastic loading mechanism cannot explain how small molecules other than RFO sugars are loaded into phloem. Most studies of phloem physiology using cucurbits have neglected the possible functions of vascular architecture in phloem transport. It is well known that there are two phloem systems in cucurbits with distinctly different anatomical features: central phloem and extrafascicular phloem. However, mistaken conclusions on sources of cucurbit phloem exudation from previous reports have hindered consideration of the idea that there may be important differences between these two phloem systems. The major results are summarized as below: 1) O-linked glycans in C.maxima were structurally identified as beta-1,3 linked glucose polymers, and the composition of glycans in cucurbits was found to be species-specific. Inter-species grafting experiments proved that these glycans are phloem mobile and transported uni-directionally from scion to stock. 2) As indicated by stable isotopic labelling experiments, a considerable amount of carbon is incorporated into small metabolites in phloem exudates. However, the incorporation of carbon into RFO sugars is much faster than for other metabolites. 3) Both CO2 labelling experiments and comparative metabolomic analysis of phloem exudates and leaf tissues indicated that metabolic processes other than RFO sugar metabolism play an important role in cucurbit phloem physiology. 4) The underlying assumption that the central phloem of cucurbits continuously releases exudates after physical incision was proved wrong by rigorous experiments including direct observation by normal microscopy and combined multiple-microscopic methods. Errors in previous experimental confirmation of phloem exudation in cucurbits are critically discussed. 5) Extrafascicular phloem was proved to be functional, as indicated by phloem-mobile carboxyfluorescein tracer studies. Commissural sieve tubes interconnect phloem bundles into a complete super-symplastic network. 6) Extrafascicular phloem represents the main source of exudates following physical incision. The major transported metabolites by these extrafacicular phloem are non-sugar compounds including amino acids, O-glycans, amines. 7) Central phloem contains almost exclusively RFO sugars, the estimated amount of which is up to 1 to 2 molar. The major RFO sugar present in central phloem is stachyose. 8) Cucurbits utilize two structurally different phloem systems for transporting different group of metabolites (RFO sugars and non-RFO sugar compounds). This implies that cucurbits may use spatially separated loading mechanisms (apoplastic loading for extrafascicular phloem and symplastic loading for central phloem) for supply of nutrients to sinks. 9) Along the transport systems, RFO sugars were mainly distributed within central phloem tissues. There were only small amounts of RFO sugars present in xylem tissues (millimolar range) and trace amounts of RFO sugars in cortex and pith. The composition of small molecules in external central phloem is very different from that in internal central phloem. 10) Aggregated P-proteins were manually dissected from central phloem and analysed by both SDS-PAGE and mass spectrometry. Partial sequences of peptides were obtained by QTOF de novo sequencing from trypsin digests of three SDS-PAGE bands. None of these partial sequences shows significant homology to known cucurbit phloem proteins or other plant proteins. This proves that these central phloem proteins are a completely new group of proteins different from those in extrafascicular phloem. The extensively analysed P-proteins reported in literature to date are therefore now shown to arise from extrafascicular phloem and not central phloem, and therefore do not appear to be involved in the occlusion processes in central phloem.
Post-translational redox-regulation is a well-known mechanism to regulate enzymes of the Calvin cycle, oxidative pentose phosphate cycle, NADPH export and ATP synthesis in response to light. The aim of the present thesis was to investigate whether a similar mechanism is also regulating carbon storage in leaves. Previous studies have shown that the key-regulatory enzyme of starch synthesis, ADPglucose pyrophosphorylase (AGPase) is inactivated by formation of an intermolecular disulfide bridge between the two catalytic subunits (AGPB) of the heterotetrameric holoenzyme in potato tubers, but the relevance of this mechanism to regulate starch synthesis in leaves was not investigated. The work presented in this thesis shows that AGPase is subject to post-translational redox-regulation in leaves of pea, potato and Arabidopsis in response to day night changes. Light was shown to trigger posttranslational redox-regulation of AGPase. AGPB was rapidly converted from a dimer to a monomer when isolated pea chloroplasts were illuminated and from a monomer to a dimer when preilluminated leaves were darkened. Conversion of AGPB from dimer to monomer was accompanied by an increase in activity due to changes in the kinetik properties of the enzyme. Studies with pea chloroplast extracts showed that AGPase redox-activation is mediated by thioredoxins f and m from spinach in-vitro. In a further set of experiments it was shown that sugars provide a second input leading to AGPase redox activation and increased starch synthesis and that they can act as a signal which is independent from light. External feeding of sugars such as sucrose or trehalose to Arabidopsis leaves in the dark led to conversion of AGPB from dimer to monomer and to an increase in the rate of starch synthesis, while there were no significant changes in the level of 3PGA, an allosteric activator of the enyzme, and in the NADPH/NADP+ ratio. Experiments with transgenic Arabidopsis plants with altered levels of trehalose 6-phosphate (T6P), the precursor of trehalose synthesis, provided genetic evidence that T6P rather than trehalose is leading to AGPase redox-activation. Compared to Wt, leaves expressing E.coli trehalose-phosphate synthase (TPS) in the cytosol showed increased activation of AGPase and higher starch level during the day, while trehalose-phosphate phosphatase (TPP) overexpressing leaves showed the opposite. These changes occurred independently of changes in sugar and sugar-phosphate levels and NADPH/NADP+ ratio. External supply of sucrose to Wt and TPS-overexpressing leaves led to monomerisation of AGPB, while this response was attenuated in TPP expressing leaves, indicating that T6P is involved in the sucrose-dependent redox-activation of AGPase. To provide biochemical evidence that T6P promotes redox-activation of AGPase independently of cytosolic elements, T6P was fed to intact isolated chloroplasts for 15 min. incubation with concentrations down to 100 µM of T6P, but not with sucrose 6-phosphate, sucrose, trehalose or Pi as controls, significantly and specifically increased AGPB monomerisation and AGPase activity within 15 minutes, implying T6P as a signal reporting the cytosolic sugar status to the chloroplast. The response to T6P did not involve changes in the NADPH/NADP+ ratio consistent with T6P modulating redox-transfer to AGPase independently of changes in plastidial redox-state. Acetyl-CoA carboxylase (ACCase) is known as key-regulatory enzyme of fatty acid and lipid synthesis in plants. At the start of the present thesis there was mainly in vitro evidence in the literature showing redox-regulation of ACCase by DTT, and thioredoxins f and m. In the present thesis the in-vivo relevance of this mechanism to regulate lipid synthesis in leaves was investigated. ACCase activity measurement in leaf tissue collected at the end of the day and night in Arabidopsis leaves revealed a 3-fold higher activation state of the enzyme in the light than in the dark. Redox-activation was accompanied by change in kinetic properties of ACCase, leading to an increase affinity to its substrate acetyl-CoA . In further experiments, DTT as well as sucrose were fed to leaves, and both treatments led to a stimulation in the rate of lipid synthesis accompanied by redox-activation of ACCase and decrease in acetyl-CoA content. In a final approach, comparison of metabolic and transcript profiling after DTT feeding and after sucrose feeding to leaves provided evidence that redox-modification is an important regulatory mechanism in central metabolic pathways such as TCA cycle and amino acid synthesis, which acts independently of transcript levels.
In den letzten 20 Jahren hat sich der Maiszünsler (Ostrinia nubilalis HÜBNER), aus der Schmetterlingsfamilie der Pyralidae oder Zünsler, zum bedeutendsten tierischen Schädling des Maises (Zea mays) entwickelt. Eine Möglichkeit den Befall des Maiszünslers abzuwenden, bietet der Anbau von Bacillus thuringiensis-Mais (Bt-Mais). Mit Hilfe der Gentechnik wurden Gene des Bakteriums Bacillus thuringiensis übertragen, die einen für Fraßinsekten giftigen Wirkstoff bilden, wodurch die Pflanzen während der kompletten Vegetation vor den Larven des Maiszünslers geschützt sind. Ziel des vorliegenden Projektes war es, in einer 3-jährigen Studie die Auswirkungen des großflächigen Anbaus von Bt-Mais auf die ökologische Situation und den Handlungsrahmen des integrierten Pflanzenschutzes komplex zu untersuchen. Dazu wurden in Betrieben im Oderbruch, das als permanentes Befallsgebiet des Maiszünslers gilt, in den Jahren 2002 bis 2004 jährlich zwei Felder mit jeweils einer Bt-Sorte und einer konventionellen Sorte angelegt. Zusätzlich wurden biologische und chemische Maiszünsler-Bekämpfungsvarianten geprüft. Durch verschiedene Methoden wie Bonituren, Ganzpflanzenernten, Bodenfallenfänge und Beobachtungen des Wahlverhaltens von (Flug-)insekten konnten Aussagen zum Vorkommen von Insekten und Spinnentieren getroffen werden, wobei hierfür Daten aus Untersuchungen der Jahre 2000 und 2001 im Oderbruch ergänzend herangezogen werden konnten. Durch Ertragsmessungen, Energie- und Qualitätsermittlungen, sowie Fusarium- und Mykotoxinanalysen konnte der Anbau von Bt-Mais als neue Alternative zur Bekämpfung des Maiszünslers bewertet werden. Bezüglich des Auftretens von Insekten und Spinnentieren wurden im Mittel der fünfjährigen Datenerhebung beim Vergleich der Bt-Sorte zur konventionellen Sorte, mit Ausnahme der fast 100 %igen Bekämpfung des Maiszünslers, keine signifikanten Unterschiede festgestellt. Hierfür wurde ein besonderes Augenmerk auf Thripse, Wanzen, Blattläuse und deren Fraßfeinde, sowie mittels Bodenfallenfängen auf Laufkäfer und Spinnen gerichtet. Die erwarteten ökonomischen Vorteile wie etwa Ertragsplus oder bessere Nährstoff- und Energiegehalte durch geringeren Schaden beim Anbau von Bt-Mais als Silomais blieben in den Untersuchungsjahren aus. Allerdings zeigten Fusarium- und Mykotoxinanalysen eine geringere Belastung des Bt-Maises, was möglicherweise auf den geringeren Schaden zurückzuführen ist, da beschädigte Pflanzen für Fusarium und Mykotoxine anfälliger sind. Desweiteren konnten erste methodische Ansätze für ein auf EU-Ebene gefordertes, den Anbau von Bt-Mais begleitendes Monitoring, erarbeitet werden. So konnten Vorschläge für geeignete Methoden, deren Umfang sowie des Zeitpunktes der Durchführungen gemacht werden.
Arabidopsis thaliana trichomes exhibit strong birefringence under polarized light, a characteristic of cell walls containing large amounts of highly ordered cellulose microfibrils. The tbr1 mutant of Arabidopsis lacks trichome birefringence and is deficient in secondary cell wall cellulose synthesis (Potikha and Delmer, 1995). The TBR gene was identified by recombinational mapping, candidate gene sequencing and molecular complementation using genomic cosmid clones, as well as a p35S:TBR genomic DNA construct, fully rescuing the mutant phenotype in both cases. The only mutant allele available (tbr-1) carries a substitution (G to E) in a conserved aminoacid domain of the protein. TBR gene structure was proved to have a longer size than the one found to be annotated at the time of identification in the data-base. A full cDNA clone containing the full transcript was available and also complementation experiments using different gene fragments (annotated and suggested) leaded to the result that TBR gene is indeed, longer. TBR encodes a novel plant-specific protein with predicted plasma membrane localization, therefore being consistent with idea that is required for-, or is a novel component of a functional cellulose synthase complex. TBR is part of an Arabidopsis gene/protein family, (TBL-trichome birefringence like) which, depending on homology, comprises up to 20 members, none of which has a biological or biochemical function attributed. T-DNA insertion lines in TBR gene and two close homologues have been screened by PCR, but no homozygous were found and no trichomes phenotype was identified. Promoter-GUS lines were produced for TBR, as well as for its two closest homologues (one being a segmentally duplicated gene on chromosome III), using 1.6-2 kb of promoter sequence upstream of the annotated start codons. The TBR promoter was the only one of the three that yielded trichome expression, this probably explaining the phenotype of the TBR mutant. Moreover, TBR is expressed in leaves, in growing lateral roots, and in vascular tissues of young Arabidopsis seedlings and plantlets. Later on, the expression appears in inflorescens, stems, flowers and green siliques. This expression pattern is largely overlapping with those of the two analyzed homologues and it corresponds with data of RT-PCR expression profiling performed for TBR and the two analyzed homologues in different tissues, at different developmental stages. Biochemical analysis of cell wall (leaves and trichomes), as GC and MALDI-TOF, were performed, but revealed no major differences between tbr1 and wild type plants. Scanning electron microscopy analysis and cell wall polysaccharides antibody labeling showed a clear difference in the trichomes cell wall structure between mutant plant and wild type.
Nitrogen is often a limiting factor for plant growth due to its heterogenous distribution in the soil and to seasonal and diurnal changes in growth rates. In most soils, NH4+ and NO3 – are the predominant sources of inorganic nitrogen that are available for plant nutrition. In this context, plants have evolved mechanisms that enable them to optimize nitrogen acquisition, which include transporters specialized in the uptake of nitrogen and susceptible to a regulation that responds to nitrogen limiting or excess conditions. Although the average NH4+ concentrations of soils are generally 100 to 1000 times lower than those of NO3 – (Marschner, 1995), most plants preferentially take up NH4+ when both forms are present because unlike NO3– , NH4+ has not to be reduced prior to assimilation and thus requires less energy for assimilation (Bloom et al., 1992). Apart from high uptake rates in roots, high intracellular ammonium concentrations also result from quantitatively important internal breakdown of amino acids (Feng et al., 1998), and originates in high quantities during photorespiration (Mattson et al., 1997, Pearson et al., 1998). Thus, NH4+ is a key component of nitrogen metabolism for all plants and can accumulate to varying concentrations in all compartments of the cell, including the cytosol, the vacuole and in the apoplast (Wells and Miller, 2000; Nielsen and Schjoerring, 1998). Two related families of ammonium transporters (AMT1 and AMT2), containing six genes which encode transporter proteins that are specific for ammonium had been identified prior to this thesis and some genes had partially been characterised in Arabidopsis (Gazzarrini et al., 1999; Sohlenkamp et al. 2002; Kaiser et al., 2002). However, these studies were not sufficient to assign physiological functions to the individual transporters and AMT1.4 and AMT1.5 had not been studied prior to this thesis. Given this background, it was considered desirable to acquire a deeper knowledge of the physiological functions of the six Arabidopsis ammonium transporters. To this end, tissue specific expression profiles of the individual wildtype AtAMT genes were performed by quantitative real time PCR (qRT-PCR) and promoter-GUS expression. Modern approaches such as the use of T-DNA insertional mutants and RNAi hairpin constructs were employed to reduce the expression levels of AMT genes. Transcript levels were determined, and physiological, biochemical and developmental analysis such as growth tests on different media and 14C-MA and NH4+ uptake studies with the isolated insertional mutants and RNAi lines were performed to deepen the knowledge of the individual functions of the six AMTs in Arabidopsis. In addition, double mutants of the insertional mutants were created to investigate the extent in which homologous genes could compensate for lost transporter functions. The results described in this thesis show that the six AtAMT genes display a high degree of specifity in their tissue specific expression and are likely to play complementary roles in ammonium uptake into roots, in shoots, and in flowers. AtAMT1.1 is likely to be a ‘work horse’ for cellular ammonium transport and reassimilation. A major role is probably the recapture of photorespiratory NH3/NH4+ escaping from the cytosol. In roots, it is likely to transport NH4+ from the apoplast into cortical cells. AtAMT1.3 and AtAMT1.5 appear to be specialised in the acquisition of external NH4+ from the soil. Furthermore, AtAMT1.5 plays an additional role in the reassimilation of NH3/NH4+ released during the breakdown of storage proteins in the cotyledons of germinating seedlings. It was difficult to distinguish a specialisation between the transporters AtAMt1.2 and AtAMt1.1, however the root and flower specific expression patterns are different and indicate alternative functions of both. AtAMT1.4 has a very distinct expression which is restricted to the vascular bundels of leaves and to pollen only, where it is likely to be involved in the loading of NH4+ into the cells.The AtAMT2.1 expression pattern is confined to vascular bundels and meristematic active tissues in leaves where ammonium concentrations can reach very high levels. Additionally, the Vmax of AtAMT2 increases with increasing external pH, contrasting to AtAMT1.1. Thus, AtAMT2.1 it might be specialised in ammonium transport in ammonium rich environments, where the functions of other transporters are limited, enabling cells to take up NH4+ over a wide range of concentrations. The root hair expression ascribes an additional role in NH3/NH4+ acquisition where it possibly serves as a transporter that is able to acquire ammonium from basic soils where other transporters become less effective.RNAi lines showing a reduction in AtAMT gene mRNA levels and NH4+ transport kinetics, grew slower and flowering time was delayed. This indicates that NH4+ is a crucial and limiting factor for plant growth.
