A new approach to achieve sub-pixel spatial resolution in a pnCCD detector with 75 x 75 mu m(2) pixel size is proposed for X-ray applications in single photon counting mode. The approach considers the energy dependence of the charge cloud created by a single photon and its split probabilities between neighboring pixels of the detector based on a rectangular model for the charge cloud density. For cases where the charge of this cloud becomes distributed over three or four pixels the center position of photon impact can be reconstructed with a precision better than 2 mu m. The predicted charge cloud sizes are tested at selected X-ray fluorescence lines emitting energies between 6.4 keV and 17.4 keV and forming charge clouds with size (rms) varying between 8 mu m and 10 mu m respectively. The 2 mu m enhanced spatial resolution of the pnCCD is verified by means of an x-ray transmission experiment throughout an optical grating.
Lamprophyre sind porphyrische, aus Mantelschmelzen gebildete Gesteine, die meist in Form von Gängen auftreten. Sie zeichnen sich durch auffällige und charakteristische texturelle, chemische und mineralogische Eigenschaften aus. Als ehemalige Mantelschmelzen liefern sie Information sowohl über Bedingungen der Schmelzbildung im Mantel als auch über geodynamische Prozesse, die zu metasomatischer Veränderung des Mantels geführt haben. Im Saxothuringikum Mitteleuropas, am Nordrand des Böhmischen Massivs, gibt es zahlreiche Lamprophyrvorkommen, die hier zur Charakterisierung der Mantelentwicklung während der variszischen Orogenese dienen. Die vorliegende Arbeit befaßt sich mit den mineralogischen, geochemischen und isotopischen (Sr-Nd-Pb) Signaturen von spätvariszischen kalkalkalischen Lamprophyren, von postvariszischen ultramafischen Lamprophyren, von Alkalibasalten der Lausitz und, zum Vergleich, von prävariszischen Gabbros. Darüberhinaus nutzt die Arbeit Lithium-Isotopensignaturen kombiniert mit Sr-Nd-Pb–Isotopendaten spätvariszischer kalkalkalischer Lamprophyre aus drei variszischen Domänen (Erzgebirge, Lausitz, Sudeten) zur Erkundung der lokalen Mantelüberprägungen während der variszischen Orogenese.
A new phenylanthrone, named knipholone cyclooxanthrone and a dimeric anthraquinone, 10-methoxy-10,7'-(chrysophanol anthrone)-chrysophanol were isolated from the roots of Kniphofia foliosa together with the rare naphthalene glycoside, dianellin. The structures were determined by NMR and mass spectroscopic techniques. The compounds showed antiplasmodial activities against the chloroquine-resistant (W2) and chloroquine-sensitive (D6) strains of Plasmodium falciparum with 10-methoxy-10,7'-(chrysophanol anthrone)-chrysophanol being the most active with IC50 values of 1.17 +/- 0.12 and 4.07 +/- 1.54 mu g/ml, respectively.
A deep observation campaign carried out by the High Energy Stereoscopic System (HESS) on Centaurus A enabled the discovery of gamma-rays from the blazar 1ES 1312-423, 2 degrees away from the radio galaxy. With a differential flux at 1 TeV of phi(1 TeV) = (1.9 +/- 0.6(stat) +/- 0.4(sys)) x 10(-13) cm(-2) s(-1) TeV-1 corresponding to 0.5 per cent of the Crab nebula differential flux and a spectral index Gamma = 2.9 +/- 0.5(stat) +/- 0.2(sys), 1ES 1312-423 is one of the faintest sources ever detected in the very high energy (E > 100 GeV) extragalactic sky. A careful analysis using three and a half years of Fermi Large Area Telescope (Fermi-LAT) data allows the discovery at high energies (E > 100 MeV) of a hard spectrum (Gamma = 1.4 +/- 0.4(stat) +/- 0.2(sys)) source coincident with 1ES 1312-423. Radio, optical, UV and X-ray observations complete the spectral energy distribution of this blazar, now covering 16 decades in energy. The emission is successfully fitted with a synchrotron self-Compton model for the non-thermal component, combined with a blackbody spectrum for the optical emission from the host galaxy.
Discovery of high and very high-energy emission from the BL Lacertae object SHBL J001355.9-185406

The detection of the high-frequency peaked BL Lac object (HBL) SHBL J001355.9-185406 (z = 0.095) at high (HE; 100 MeV < E < 300 GeV) and very high-energy (VHE; E > 100 GeV) with the Fermi Large Area Telescope (LAT) and the High Energy Stereoscopic System (H.E.S.S.) is reported. Dedicated observations were performed with the H. E. S. S. telescopes, leading to a detection at the 5.5 sigma significance level. The measured flux above 310 GeV is (8.3 +/- 1.7(stat) +/- 1.7(sys)) x 10(-13) photons cm(-2) s(-1) (about 0.6% of that of the Crab Nebula), and the power-law spectrum has a photon index of Gamma = 3.4 +/- 0.5(stat) +/- 0.2(sys). Using 3.5 years of publicly available Fermi-LAT data, a faint counterpart has been detected in the LAT data at the 5.5 sigma significance level, with an integrated flux above 300 MeV of (9.3 +/- 3.4(stat) +/- 0.8(sys)) x 10(-10) photons cm(-2) s(-1) and a photon index of Gamma = 1.96 +/- 0.20(stat) +/- 0.08(sys). X-ray observations with Swift-XRT allow the synchrotron peak energy in vF(v) representation to be located at similar to 1.0 keV. The broadband spectral energy distribution is modelled with a one-zone synchrotron self-Compton (SSC) model and the optical data by a black-body emission describing the thermal emission of the host galaxy. The derived parameters are typical of HBLs detected at VHE, with a particle-dominated jet.
The quasar PKS 1510-089 (z = 0.361) was observed with the H.E.S.S. array of imaging atmospheric Cherenkov telescopes during high states in the optical and GeV bands, to search for very high energy (VHE, defined as E >= 0.1 TeV) emission. VHE gamma-rays were detected with a statistical significance of 9.2 standard deviations in 15.8 h of H. E. S. S. data taken during March and April 2009. A VHE integral flux of I(0.15 TeV < E < 1.0TeV) = (1.0 +/- 0.2(stat) +/- 0.2(sys)) x 10(-11) cm(-2) s(-1) is measured. The best-fit power law to the VHE data has a photon index of G = 5.4 +/- 0.7(stat) +/- 0.3(sys). The GeV and optical light curves show pronounced variability during the period of H.E.S.S. observations. However, there is insufficient evidence to claim statistically significant variability in the VHE data. Because of its relatively high redshift, the VHE flux from PKS 1510-089 should suffer considerable attenuation in the intergalactic space due to the extragalactic background light (EBL). Hence, the measured gamma-ray spectrum is used to derive upper limits on the opacity due to EBL, which are found to be comparable with the previously derived limits from relatively-nearby BL Lac objects. Unlike typical VHE-detected blazars where the broadband spectrum is dominated by nonthermal radiation at all wavelengths, the quasar PKS 1510-089 has a bright thermal component in the optical to UV frequency band. Among all VHE detected blazars, PKS 1510-089 has the most luminous broad line region. The detection of VHE emission from this quasar indicates a low level of gamma - gamma absorption on the internal optical to UV photon field.
Context. Globular clusters (GCs) are established emitters of high-energy (HE, 100 MeV < E < 100 GeV) gamma-ray radiation which could originate from the cumulative emission of the numerous millisecond pulsars (msPSRs) in the clusters’ cores or from inverse Compton (IC) scattering of relativistic leptons accelerated in the GC environment. These stellar clusters could also constitute a new class of sources in the very-high-energy (VHE, E > 100 GeV) gamma-ray regime, judging from the recent detection of a signal from the direction of Terzan 5 with the H.E.S.S. telescope array. Aims. To search for VHE gamma-ray sources associated with other GCs, and to put constraints on leptonic emission models, we systematically analyzed the observations towards 15 GCs taken with the H. E. S. S. array of imaging atmospheric Cherenkov telescopes. Methods. We searched for point-like and extended VHE gamma-ray emission from each GC in our sample and also performed a stacking analysis combining the data from all GCs to investigate the hypothesis of a population of faint emitters. Assuming IC emission as the origin of the VHE gamma-ray signal from the direction of Terzan 5, we calculated the expected gamma-ray flux from each of the 15 GCs, based on their number of millisecond pulsars, their optical brightness and the energy density of background photon fields. Results. We did not detect significant VHE gamma-ray emission from any of the 15 GCs in either of the two analyses. Given the uncertainties related to the parameter determinations, the obtained flux upper limits allow to rule out the simple IC/msPSR scaling model for NGC6388 and NGC7078. The upper limits derived from the stacking analyses are factors between 2 and 50 below the flux predicted by the simple leptonic scaling model, depending on the assumed source extent and the dominant target photon fields. Therefore, Terzan 5 still remains exceptional among all GCs, as the VHE gamma-ray emission either arises from extra-ordinarily efficient leptonic processes, or from a recent catastrophic event, or is even unrelated to the GC itself.
Gamma-ray line signatures can be expected in the very-high-energy (E-gamma > 100 GeV) domain due to self-annihilation or decay of dark matter (DM) particles in space. Such a signal would be readily distinguishable from astrophysical gamma-ray sources that in most cases produce continuous spectra that span over several orders of magnitude in energy. Using data collected with the H. E. S. S. gamma-ray instrument, upper limits on linelike emission are obtained in the energy range between similar to 500 GeV and similar to 25 TeV for the central part of the Milky Way halo and for extragalactic observations, complementing recent limits obtained with the Fermi-LAT instrument at lower energies. No statistically significant signal could be found. For monochromatic gamma-ray line emission, flux limits of (2 x 10(-7)-2 x 10(-5)) m(-2)s(-1)sr(-1) and (1 x 10(-8)- 2 x 10(-6)) m(-2)s(-1)sr(-1) are obtained for the central part of the Milky Way halo and extragalactic observations, respectively. For a DM particle mass of 1 TeV, limits on the velocity- averaged DM annihilation cross section <sigma upsilon >(chi chi ->gamma gamma) reach similar to 10(-27)cm(3)s(-1), based on the Einasto parametrization of the Galactic DM halo density profile. HESS observations of the binary system PSR B1259-63/LS 2883 around the 2010/2011 periastron passage

Aims. We present very high energy (VHE; E > 100 GeV) data from the gamma-ray binary system PSR B1259-63/LS 2883 taken around its periastron passage on 15th of December 2010 with the High Energy Stereoscopic System (H. E. S. S.) of Cherenkov Telescopes. We aim to search for a possible TeV counterpart of the GeV flare detected by the Fermi LAT. In addition, we aim to study the current periastron passage in the context of previous observations taken at similar orbital phases, testing the repetitive behaviour of the source.
Methods. Observations at VHEs were conducted with H.E.S.S. from 9th to 16th of January 2011. The total dataset amounts to similar to 6 h of observing time. The data taken around the 2004 periastron passage were also re-analysed with the current analysis techniques in order to extend the energy spectrum above 3 TeV to fully compare observation results from 2004 and 2011.
Results. The source is detected in the 2011 data at a significance level of 11.5 sigma revealing an averaged integral flux above 1 TeV of (1.01 +/- 0.18(stat) +/- 0.20(sys)) x 10(-12) cm(-2) s(-1). The differential energy spectrum follows a power-law shape with a spectral index Gamma = 2.92 +/- 0.30(stat) +/- 0.20(sys) and a flux normalisation at 1 TeV of N-0 = (1.95 +/- 0.32(stat) +/- 0.39(sys)) x 10(-12) TeV-1 cm(-2) s(-1). The measured light curve does not show any evidence for variability of the source on the daily scale. The re-analysis of the 2004 data yields results compatible with the published ones. The differential energy spectrum measured up to similar to 10 TeV is consistent with a power law with a spectral index Gamma = 2.81 +/- 0.10(stat) +/- 0.20(sys) and a flux normalisation at 1 TeV of N-0 = (1.29 +/- 0.08(stat) +/- 0.26(sys)) x 10(-12) TeV-1 cm(-2) s(-1).
Conclusions. The measured integral flux and the spectral shape of the 2011 data are compatible with the results obtained around previous periastron passages. The absence of variability in the H.E.S.S. data indicates that the GeV flare observed by Fermi LAT in the time period covered also by H.E.S.S. observations originates in a different physical scenario than the TeV emission. Moreover, the comparison of the new results to the results from the 2004 observations made at a similar orbital phase provides a stronger evidence of the repetitive behaviour of the source.
Discovery of very high energy gamma-ray emission from the BL Lacertae
object PKS0301-243 with HESS

The active galactic nucleus PKS 0301-243 (z = 0.266) is a high-synchrotron-peaked BL Lac object that is detected at high energies (HE, 100 MeV < E < 100 GeV) by Fermi/LAT. This paper reports on the discovery of PKS 0301-243 at very high energies (E > 100 GeV) by the High Energy Stereoscopic System (H.E.S.S.) from observations between September 2009 and December 2011 for a total live time of 34.9 h. Gamma rays above 200 GeV are detected at a significance of 9.4 sigma. A hint of variability at the 2.5 sigma level is found. An integral flux I(E > 200GeV) = (3.3 +/- 1.1(stat) +/- 0.7(syst)) x 10(-12) ph cm(-2) s(-1) and a photon index Gamma = 4.6 +/- 0.7(stat) +/- 0.2(syst) are measured. Multi-wavelength light curves in HE, X-ray and optical bands show strong variability, and a minimal variability timescale of eight days is estimated from the optical light curve. A single-zone leptonic synchrotron self-Compton scenario satisfactorily reproduces the multi-wavelength data. In this model, the emitting region is out of equipartition and the jet is particle dominated. Because of its high redshift compared to other sources observed at TeV energies, the very high energy emission from PKS 0301-243 is attenuated by the extragalactic background light (EBL) and the measured spectrum is used to derive an upper limit on the opacity of the EBL.
Axionlike particles (ALPs) are hypothetical light (sub-eV) bosons predicted in some extensions of the Standard Model of particle physics. In astrophysical environments comprising high-energy gamma rays and turbulent magnetic fields, the existence of ALPs can modify the energy spectrum of the gamma rays for a sufficiently large coupling between ALPs and photons. This modification would take the form of an irregular behavior of the energy spectrum in a limited energy range. Data from the H. E. S. S. observations of the distant BL Lac object PKS 2155 - 304 (z = 0.116) are used to derive upper limits at the 95% C. L. on the strength of the ALP coupling to photons, g(gamma a) < 2.1 x 10(-11) GeV-1 for an ALP mass between 15 and 60 neV. The results depend on assumptions on the magnetic field around the source, which are chosen conservatively. The derived constraints apply to both light pseudoscalar and scalar bosons that couple to the electromagnetic field.
Very high-energy gamma-ray emission from PKS 0447-439 was detected with the H. E. S. S. Cherenkov telescope array in December 2009. This blazar is one of the brightest extragalactic objects in the Fermi bright source list and has a hard spectrum in the MeV to GeV range. In the TeV range, a photon index of 3.89 +/- 0.37 (stat) +/- 0.22 (sys) and a flux normalisation at 1 TeV, phi(1) (TeV) = (3.5 +/- 1.1(stat) +/- 0.9(sys)) x 10(-13) cm(-2) s(-1) TeV-1 were found. The detection with H. E. S. S. triggered observations in the X-ray band with the Swift and RXTE telescopes. Simultaneous UV and optical data from Swift UVOT and data from the optical telescopes ATOM and ROTSE are also available. The spectrum and light curve measured with H. E. S. S. are presented and compared to the multi-wavelength data at lower energies. A rapid flare is seen in the Swift XRT and RXTE data, together with a flux variation in the UV band, at a time scale of the order of one day. A firm upper limit of z < 0.59 on the redshift of PKS 0447-439 is derived from the combined Fermi-LAT and H. E. S. S. data, given the assumptions that there is no upturn in the intrinsic spectrum above the Fermi-LAT energy range and that absorption on the extragalactic background light (EBL) is not weaker than the lower limit provided by current models. The spectral energy distribution is well described by a simple one-zone synchrotron self-Compton scenario, if the redshift of the source is less than z less than or similar to 0.4.
Introducing the CTA concept

The Cherenkov Telescope Array (CTA) is a new observatory for very high-energy (VHE) gamma rays. CTA has ambitions science goals, for which it is necessary to achieve full-sky coverage, to improve the sensitivity by about an order of magnitude, to span about four decades of energy, from a few tens of GeV to above 100 TeV with enhanced angular and energy resolutions over existing VHE gamma-ray observatories. An international collaboration has formed with more than 1000 members from 27 countries in Europe, Asia, Africa and North and South America. In 2010 the CTA Consortium completed a Design Study and started a three-year Preparatory Phase which leads to production readiness of CTA in 2014. In this paper we introduce the science goals and the concept of CTA, and provide an overview of the project.
In aqueous solution, symmetric triblock copolymers with a thermoresponsive middle block and hydrophobic end blocks form flower-like core-shell micelles which collapse and aggregate upon heating through the cloud point (CP). The collapse of the micellar shell and the intermicellar aggregation are followed in situ and in real-time using time-resolved small-angle neutron scattering (SANS), while heating micellar solutions of a poly((styrene-d(8))-b-(N-isopropyl acrylamide)-b-(styrene-d(8))) triblock copolymer in D2O rapidly through their CP. The influence of polymer concentration as well as of the start and target temperatures is addressed. In all cases, the micellar collapse is very fast. The collapsed micelles immediately form small clusters which contain voids. They densify which slows down or even stops their growth. For low concentrations and target temperatures just above the CP, i.e. shallow temperature jumps, the subsequent growth of the clusters is described by diffusion-limited aggregation. In contrast, for higher concentrations and/or higher target temperatures, i.e. deep temperature jumps, intermicellar bridges dominate the growth. Eventually, in all cases, the clusters coagulate which results in macroscopic phase separation. For shallow temperature jumps, the cluster surfaces stay rough; whereas for deep temperature jumps, a concentration gradient develops at late stages. These results are important for the development of conditions for thermal switching in applications, e.g. for the use of thermoresponsive micellar systems for transport and delivery purposes.
Metabolically active microbial communities are present in a wide range of subsurface environments. Techniques like enumeration of microbial cells, activity measurements with radiotracer assays and the analysis of porewater constituents are currently being used to explore the subsurface biosphere, alongside with molecular biological analyses. However, many of these techniques reach their detection limits due to low microbial activity and abundance. Direct measurements of microbial turnover not just face issues of insufficient sensitivity, they only provide information about a single specific process but in sediments many different process can occur simultaneously. Therefore, the development of a new technique to measure total microbial activity would be a major improvement. A new tritium-based hydrogenase-enzyme assay appeared to be a promising tool to quantify total living biomass, even in low activity subsurface environments. In this PhD project total microbial biomass and microbial activity was quantified in different subsurface sediments using established techniques (cell enumeration and pore water geochemistry) as well as a new tritium-based hydrogenase enzyme assay. By using a large database of our own cell enumeration data from equatorial Pacific and north Pacific sediments and published data it was shown that the global geographic distribution of subseafloor sedimentary microbes varies between sites by 5 to 6 orders of magnitude and correlates with the sedimentation rate and distance from land. Based on these correlations, global subseafloor biomass was estimated to be 4.1 petagram-C and ~0.6 % of Earth's total living biomass, which is significantly lower than previous estimates. Despite the massive reduction in biomass the subseafloor biosphere is still an important player in global biogeochemical cycles. To understand the relationship between microbial activity, abundance and organic matter flux into the sediment an expedition to the equatorial Pacific upwelling area and the north Pacific Gyre was carried out. Oxygen respiration rates in subseafloor sediments from the north Pacific Gyre, which are deposited at sedimentation rates of 1 mm per 1000 years, showed that microbial communities could survive for millions of years without fresh supply of organic carbon. Contrary to the north Pacific Gyre oxygen was completely depleted within the upper few millimeters to centimeters in sediments of the equatorial upwelling region due to a higher supply of organic matter and higher metabolic activity. So occurrence and variability of electron acceptors over depth and sites make the subsurface a complex environment for the quantification of total microbial activity. Recent studies showed that electron acceptor processes, which were previously thought to thermodynamically exclude each other can occur simultaneously. So in many cases a simple measure of the total microbial activity would be a better and more robust solution than assays for several specific processes, for example sulfate reduction rates or methanogenesis. Enzyme or molecular assays provide a more general approach as they target key metabolic compounds. Since hydrogenase enzymes are ubiquitous in microbes, the recently developed tritium-based hydrogenase radiotracer assay is applied to quantify hydrogenase enzyme activity as a parameter of total living cell activity. Hydrogenase enzyme activity was measured in sediments from different locations (Lake Van, Barents Sea, Equatorial Pacific and Gulf of Mexico). In sediment samples that contained nitrate, we found the lowest cell specific enzyme activity around 10^(-5) nmol H_(2) cell^(-1) d^(-1). With decreasing energy yield of the electron acceptor used, cell-specific hydrogenase activity increased and maximum values of up to 1 nmol H_(2) cell^(-1) d^(-1) were found in samples with methane concentrations of >10 ppm. Although hydrogenase activity cannot be converted directly into a turnover rate of a specific process, cell-specific activity factors can be used to identify specific metabolism and to quantify the metabolically active microbial population. In another study on sediments from the Nankai Trough microbial abundance and hydrogenase activity data show that both the habitat and the activity of subseafloor sedimentary microbial communities have been impacted by seismic activities. An increase in hydrogenase activity near the fault zone revealed that the microbial community was supplied with hydrogen as an energy source and that the microbes were specialized to hydrogen metabolism.
The levels of environmental light experienced by organisms during the behavioral activity phase deeply influence the performance of important ecological tasks. As a result, their shape and coloring may experience a light-driven selection process via the day-night rhythmic behavior. In this study, we tested the phenotypic and genetic variability of the western Mediterranean squat lobster (Munida tenuimana). We sampled at depths with different photic conditions and potentially, different burrow emergence rhythms. We performed day-night hauling at different depths, above and below the twilight zone end (i.e., 700 m, 1200 m, 1350 m, and 1500 m), to portray the occurrence of any burrow emergence rhythmicity. Collected animals were screened for shape and size (by geometric morphometry), spectrum and color variation (by photometric analysis), as well as for sequence variation at the mitochondria] DNA gene encoding for the NADH dehydrogenase subunit I. We found that a weak genetic structuring and shape homogeneity occurred together with significant variations in size, with the smaller individuals living at the twilight zone inferior limit and the larger individuals above and below. The infra-red wavelengths of spectral reflectance varied significantly with depth while the blue-green ones were size-dependent and expressed in smaller animals, which has a very small spectral reflectance. The effects of solar and bioluminescence lighting are discussed as depth-dependent evolutionary forces likely influencing the behavioral rhythms and coloring of M. tenuimana.
Where girls the role of boys in CS - attitudes of CS students in a female-dominated environment

A suitable vehicle for integration of bioactive plant constituents is proposed. It involves modification of proteins using phenolics and applying these for protection of labile constituents. It dissects the noncovalent and covalent interactions of beta-lactoglobulin with coffee-specific phenolics. Alkaline and polyphenol oxidase modulated covalent reactions were compared. Tryptic digestion combined with MALDI-TOF-MS provided tentative allocation of the modification type and site in the protein, and an in silico modeling of modified beta-lactoglobulin is proposed. The modification delivers proteins with enhanced antioxidative properties. Changed structural properties and differences in solubility, surface hydrophobicity, and emulsification were observed. The polyphenol oxidase modulated reaction provides a modified beta-lactoglobulin with a high antioxidative power, is thermally more stable, requires less energy to unfold, and, when emulsified with lutein esters, exhibits their higher stability against UV light. Thus, adaptation of this modification provides an innovative approach for functionalizing proteins and their uses in the food industry.
We report on the VERITAS observations of the high-frequency peaked BL Lac object 1ES 1959+650 in the period 2007-2011. This source is detected at TeV energies by VERITAS at 16.4 standard deviation (sigma) significance in 7.6 hr of observation in a low flux state. A multiwavelength spectral energy distribution (SED) is constructed from contemporaneous data from VERITAS, Fermi-LAT, RXTE PCA, and Swift UVOT. Swift XRT data is not included in the SED due to a lack of simultaneous observations with VERITAS. In contrast to the orphan gamma-ray flare exhibited by this source in 2002, the X-ray flux of the source is found to vary by an order of magnitude, while other energy regimes exhibit less variable emission. A quasi-equilibrium synchrotron self-Compton model with an additional external radiation field is used to describe three SEDs corresponding to the lowest, highest, and average X-ray states. The variation in the X-ray spectrum is modeled by changing the electron injection spectral index, with minor adjustments of the kinetic luminosity in electrons. This scenario produces small-scale flux variability of the order of less than or similar to 2 in the high energy (E > 1MeV) and very high energy (E > 100 GeV) gamma-ray regimes, which is corroborated by the Fermi-LAT, VERITAS, and Whipple 10 m telescope light curves.
We report on VERITAS observations of the BL Lac object B2 1215+30 between 2008 and 2012. During this period, the source was detected at very high energies (VHEs; E > 100 GeV) by VERITAS with a significance of 8.9s and showed clear variability on timescales larger than months. In 2011, the source was found to be in a relatively bright state and a power-law fit to the differential photon spectrum yields a spectral index of 3.6 +/- 0.4(stat) +/- 0.3(syst) with an integral flux above 200 GeV of (8.0 +/- 0.9(stat) +/- 3.2(syst)) x 10(-12) cm(-2) s(-1). No short term variability could be detected during the bright state in 2011. Multi-wavelength data were obtained contemporaneously with the VERITAS observations in 2011 and cover optical (Super-LOTIS, MDM, Swift/UVOT), X-ray (Swift/XRT), and gamma-ray (Fermi-LAT) frequencies. These were used to construct the spectral energy distribution (SED) of B2 1215+30. A one-zone leptonic model is used to model the blazar emission and the results are compared to those of MAGIC from early 2011 and other VERITAS-detected blazars. The SED can be reproduced well with model parameters typical for VHE-detected BL Lac objects.
We report the discovery of TeV gamma-ray emission coincident with the shell-type radio supernova remnant (SNR) CTA 1 using the VERITAS gamma-ray observatory. The source, VER J0006+729, was detected as a 6.5 standard deviation excess over background and shows an extended morphology, approximated by a two-dimensional Gaussian of semimajor (semiminor) axis 0.degrees 30 (0.degrees 24) and a centroid 5’ from the Fermi gamma-ray pulsar PSR J0007+7303 and its X-ray pulsar wind nebula (PWN). The photon spectrum is well described by a power-law dN/dE = N-0(E/3 TeV)(-Gamma), with a differential spectral index of Gamma = 2.2 +/- 0.2(stat) +/- 0.3(sys), and normalization N-0 = (9.1 +/- 1.3(stat) +/- 1.7(sys)) x 10(-14) cm(-2) s(-1) TeV-1. The integral flux, F-gamma = 4.0 x 10(-12) erg cm(-2) s(-1) above 1 TeV, corresponds to 0.2% of the pulsar spin-down power at 1.4 kpc. The energetics, colocation with the SNR, and the relatively small extent of the TeV emission strongly argue for the PWN origin of the TeV photons. We consider the origin of the TeV emission in CTA 1.
We present the results of a multiwavelength observational campaign on the TeV binary system LS I +61 degrees 303 with the VERITAS telescope array (>200 GeV), Fermi-LAT (0.3-300 GeV), and Swift/XRT (2-10 keV). The data were taken from 2011 December through 2012 January and show a strong detection in all three wavebands. During this period VERITAS obtained 24.9 hr of quality selected livetime data in which LS I +61 degrees 303 was detected at a statistical significance of 11.9 sigma. These TeV observations show evidence for nightly variability in the TeV regime at a post-trial significance of 3.6 sigma. The combination of the simultaneously obtained TeV and X-ray fluxes do not demonstrate any evidence for a correlation between emission in the two bands. For the first time since the launch of the Fermi satellite in 2008, this TeV detection allows the construction of a detailed MeV-TeV spectral energy distribution from LS I +61 degrees 303. This spectrum shows a distinct cutoff in emission near 4 GeV, with emission seen by the VERITAS observations following a simple power-law above 200 GeV. This feature in the spectrum of LS I +61 degrees 303, obtained from overlapping observations with Fermi-LAT and VERITAS, may indicate that there are two distinct populations of accelerated particles producing the GeV and TeV emission.
In order to predict which ecosystem functions are most at risk from biodiversity loss, meta-analyses have generalised results from biodiversity experiments over different sites and ecosystem types. In contrast, comparing the strength of biodiversity effects across a large number of ecosystem processes measured in a single experiment permits more direct comparisons. Here, we present an analysis of 418 separate measures of 38 ecosystem processes. Overall, 45 % of processes were significantly affected by plant species richness, suggesting that, while diversity affects a large number of processes not all respond to biodiversity. We therefore compared the strength of plant diversity effects between different categories of ecosystem processes, grouping processes according to the year of measurement, their biogeochemical cycle, trophic level and compartment (above- or belowground) and according to whether they were measures of biodiversity or other ecosystem processes, biotic or abiotic and static or dynamic. Overall, and for several individual processes, we found that biodiversity effects became stronger over time. Measures of the carbon cycle were also affected more strongly by plant species richness than were the measures associated with the nitrogen cycle. Further, we found greater plant species richness effects on measures of biodiversity than on other processes. The differential effects of plant diversity on the various types of ecosystem processes indicate that future research and political effort should shift from a general debate about whether biodiversity loss impairs ecosystem functions to focussing on the specific functions of interest and ways to preserve them individually or in combination.
Pseudotachylyte veins frequently associated with mylonites and ultramylonites occur within migmatitic paragneisses, metamonzodiorites, as well as felsic and mafic granulites at the base of the section of the Hercynian lower crust exposed in Calabria (Southern Italy). The crustal section is tectonically superposed on lower grade units. Ultramylonites and pseudotachylytes are particularly well developed in migmatitic paragneisses, whereas sparse fault-related pseudotachylytes and thin mylonite/ultramylonite bands occur in granulite-facies rocks. The presence of sillimanite and clinopyroxene in ultramylonites and mylonites indicates that relatively high-temperature conditions preceded the formation of pseudotachylytes. We have analysed pseudotachylytes from different rock types to ascertain their deep crustal origin and to better understand the relationships between brittle and ductile processes during deformation of the deeper crust. Different protoliths were selected to test how lithology controls pseudotachylyte composition and textures. In migmatites and felsic granulites, euhedral or cauliflower-shaped garnets directly crystallized from pseudotachylyte melts of near andesitic composition. This indicates that pseudotachylytes originated at deep crustal conditions (> 0.75 GPa). In mafic protoliths, quenched needle-to-feather-shaped high-alumina orthopyroxene occurs in contact with newly crystallized plagioclase. The pyroxene crystallizes in garnet-free and garnet-bearing veins. The simultaneous growth of orthopyroxene and plagioclase as well as almandine, suggests lower crustal origin, with pressures in excess of 0.85 GPa. The existence of melts of different composition in the same vein indicates the stepwise, non-equilibrium conditions of frictional melting. Melt formed and intruded into pre-existing anisotropies. In mafic granulites, brittle faulting is localized in a previously formed thin high-temperature mylonite bands. migmatitic gneisses are deformed into ultramylonite domains characterized by s-c fabric. Small grain size and fluids lowered the effective stress on the c planes favouring a seismic event and the consequent melt generation. Microstructures and ductile deformation of pseudotachylytes suggest continuous ductile flow punctuated by episodes of high-strain rate, leading to seismic events and melting.
Considerable effort has been devoted to the development of simulation algorithms for facies modeling, whereas a discussion of how to combine those techniques has not existed. The integration of multiple geologic data into a three-dimensional model, which requires the combination of simulation techniques, is yet a current challenge for reservoir modeling. This article presents a thought process that guides the acquisition and modeling of geologic data at various scales. Our work is based on outcrop data collected from a Jurassic carbonate ramp located in the High Atlas mountain range of Morocco. The study window is 1 km (0.6 mi) wide and 100 m (328.1 ft) thick. We describe and model the spatial and hierarchical arrangement of carbonate bodies spanning from largest to smallest: (1) stacking pattern of high-frequency depositional sequences, (2) facies association, and (3) lithofacies. Five sequence boundaries were modeled using differential global position system mapping and light detection and ranging data. The surface-based model shows a low-angle profile with modest paleotopographic relief at the inner-to-middle ramp transition. Facies associations were populated using truncated Gaussian simulation to preserve ordered trends between the inner, middle, and outer ramps. At the lithofacies scale, field observations and statistical analysis show a mosaiclike distribution that was simulated using a fully stochastic approach with sequential indicator simulation.
This study observes that the use of one single simulation technique is unlikely to correctly model the natural patterns and variability of carbonate rocks. The selection and implementation of different techniques customized for each level of the stratigraphic hierarchy will provide the essential computing flexibility to model carbonate settings. This study demonstrates that a scale-dependent modeling approach should be a common procedure when building subsurface and outcrop models.
The study of outcrop modeling is located at the interface between two fields of expertise, Sedimentology and Computing Geoscience, which respectively investigates and simulates geological heterogeneity observed in the sedimentary record. During the last past years, modeling tools and techniques were constantly improved. In parallel, the study of Phanerozoic carbonate deposits emphasized the common occurrence of a random facies distribution along single depositional domain. Although both fields of expertise are intrinsically linked during outcrop simulation, their respective advances have not been combined in literature to enhance carbonate modeling studies. The present study re-examines the modeling strategy adapted to the simulation of shallow-water carbonate systems, based on a close relationship between field sedimentology and modeling capabilities. In the present study, the evaluation of three commonly used algorithms Truncated Gaussian Simulation (TGSim), Sequential Indicator Simulation (SISim), and Indicator Kriging (IK), were performed for the first time using visual and quantitative comparisons on an ideally suited carbonate outcrop. The results show that the heterogeneity of carbonate rocks cannot be fully simulated using one single algorithm. The operating mode of each algorithm involves capabilities as well as drawbacks that are not capable to match all field observations carried out across the modeling area. Two end members in the spectrum of carbonate depositional settings, a low-angle Jurassic ramp (High Atlas, Morocco) and a Triassic isolated platform (Dolomites, Italy), were investigated to obtain a complete overview of the geological heterogeneity in shallow-water carbonate systems. Field sedimentology and statistical analysis performed on the type, morphology, distribution, and association of carbonate bodies and combined with palaeodepositional reconstructions, emphasize similar results. At the basin scale (x 1 km), facies association, composed of facies recording similar depositional conditions, displays linear and ordered transitions between depositional domains. Contrarily, at the bedding scale (x 0.1 km), individual lithofacies type shows a mosaic-like distribution consisting of an arrangement of spatially independent lithofacies bodies along the depositional profile. The increase of spatial disorder from the basin to bedding scale results from the influence of autocyclic factors on the transport and deposition of carbonate sediments. Scale-dependent types of carbonate heterogeneity are linked with the evaluation of algorithms in order to establish a modeling strategy that considers both the sedimentary characteristics of the outcrop and the modeling capabilities. A surface-based modeling approach was used to model depositional sequences. Facies associations were populated using TGSim to preserve ordered trends between depositional domains. At the lithofacies scale, a fully stochastic approach with SISim was applied to simulate a mosaic-like lithofacies distribution. This new workflow is designed to improve the simulation of carbonate rocks, based on the modeling of each scale of heterogeneity individually. Contrarily to simulation methods applied in literature, the present study considers that the use of one single simulation technique is unlikely to correctly model the natural patterns and variability of carbonate rocks. The implementation of different techniques customized for each level of the stratigraphic hierarchy provides the essential computing flexibility to model carbonate systems. Closer feedback between advances carried out in the field of Sedimentology and Computing Geoscience should be promoted during future outcrop simulations for the enhancement of 3-D geological models.
In this note we analyze the incentives to merge in a mixed duopoly if firms compete in prices or quantities. Our model framework mainly follows Barcena-Ruiz and Garzon (J Econ 80:27-42, 2003) who set up the model with quantity competition. We extend their analysis by analyzing the case of competition in prices. Further we compare the incentives to merge with Bertrand and Cournot competition. Comparing quantity with price competition we can show that a merger is more likely with Cournot competition than with Bertrand competition.
Bacteriophage P22 recognizes O-antigen polysaccharides of Salmonella enterica subsp. enterica (S.) with its tailspike protein (TSP). In the serovars S. Typhimurium, S. Enteritidis, and S. Paratyphi A, the tetrasaccharide repeat units of the respective O-antigens consist of an identical main chain trisaccharide but different 3,6-dideoxyhexose substituents. Here, the epimers abequose, tyvelose and paratose determine the specific serotype. P22 TSP recognizes O-antigen octasaccharides in an extended binding site with a single 3,6-dideoxyhexose binding pocket. We have isolated S. Paratyphi A octasaccharides which were not available previously and determined the crystal structure of their complex with P22 TSP. We discuss our data together with crystal structures of complexes with S. Typhimurium and S. Enteritidis octasaccharides determined earlier. Isothermal titration calorimetry showed that S. Paratyphi A octasaccharide binds P22 TSP less tightly, with a difference in binding free energy of similar to 7 kJ mol(-1) at 20 degrees C compared with S. Typhimurium and S. Enteritidis octasaccharides. Individual protein-carbohydrate contacts were probed by amino acid replacements showing that the dideoxyhexose pocket contributes to binding of all three serotypes. However, S. Paratyphi A octasaccharides bind in a conformation with an energetically unfavorable phi/epsilon glycosidic bond angle combination. In contrast, octasaccharides from the other serotypes bind as solution-like conformers. Two water molecules are conserved in all P22 TSP complexes with octasaccharides of different serotypes. They line the dideoxyhexose binding pocket and force the S. Paratyphi A octasaccharides to bind as nonsolution conformers. This emphasizes the role of solvent as part of carbohydrate binding sites.

Palaeoenvironmental implications of evaporative gaylussite crystals from Lonar Lake, central India

We have undertaken petrographic, mineralogical, geochemical and isotopic investigations on carbonate minerals found within a 10-m-long core from Lonar Lake, central India, with the aim of evaluating their potential as palaeoenvironmental proxies. The core encompasses the entire Holocene and is the first well-dated high-resolution record from central India. While calcite and/or aragonite were found throughout the core, the mineral gaylussite was found only in two specific intervals (46303890 and 2040560 cal a BP). Hydrochemical and isotope data from inflowing streams and lake waters indicate that evaporitic processes play a dominant role in the precipitation of carbonates within this lake. Isotopic (18O and 13C) studies on the evaporative gaylussite crystals and residual bulk carbonates (calcite) from the long core show that evaporation is the major control on 18O enrichment in both the minerals. However, in case of 13C additional mechanisms, for example methanogenesis (gaylussite) and phytoplankton productivity (calcium carbonate), play an additional important role in some intervals. We also discuss the relevance of our investigation for palaeoclimate reconstruction and late Holocene monsoon variability.
We have undertaken a high resolution palaeoclimate reconstruction on radiocarbon dated palaeolake sediments from the Spiti valley, NW Himalaya. This site lies in the climatically sensitive winter westerlies and Indian Summer Monsoon (ISM) transitional regime and provides an opportunity to reconstruct the precipitation seasonality, and extreme precipitation events that are characterised by intensified erosion. The lake sediments reveal distinct lithofacies that provide evidence of changes in depositional environment and climate during early to mid Holocene (8.7-6.1 cal ka BP). We have identified three stages during the period of lake's existence: the Stage I (8.7-7.6 cal ka BP) is marked by lake establishment; Stage II (similar to 7.6-6.8 cal ka BP) by sustained cooler periods and weakened summer monsoon, and Stage III (similar to 6.8-6.1 cal ka BP) by a shift from colder to warmer climate with stronger ISM. We have identified several short term cooler periods at ca. 8.7, 8.5, 8.3 and 7.2-6.9 cal ka BP. Based on an overview of regional climate records we show that there is an abrupt switch in precipitation seasonality ca. 6.8 cal ka BP that is followed by the onset of the intensified monsoon in the NW Himalaya. Im Rahmen der aktuellen Debatte über Veränderungen im Schulsystem rückt auch die Lehramtsausbildung stärker in den Fokus universitärer und öffentlicher Diskussionen. Um Unterricht und Schule weiterzuentwickeln, bedarf es, nach Ansicht des Autors, kompetenter Lehrkräfte, welche reflexiv und forschungsgeleitet ihre Arbeit als einen permanenten Entwicklungsprozess betrachten. Das „Praxisforschen“ ist eine Form des Lehrens und Lernens, mittels dessen die Hochschullehre verbessert werden kann und Lehramtsstudierende gut auf die zukünftigen Anforderungen als Lehrende vorbereitet werden. „Praxisforschen“ bedeutet, anhand konkreter Forschungsfragen Schule und/oder die eigene Unterrichtstätigkeit weiterzuentwickeln. Studierende durchlaufen hierzu einen typischen Forschungsprozess, vom Entwickeln einer Fragestellung bis zur Auswertung und Darstellung der Ergebnisse. Der Artikel beschreibt die Umsetzung des Praxisforschens, wie es momentan in den Begleitseminaren zum Praxissemester an der Universität Potsdam durchgeführt wird und gibt praktische Hinweise für das forschende Lernen mit Studierenden.
We report the detection of a new TeV gamma-ray source, VER J0521+211, based on observations made with the VERITAS imaging atmospheric Cherenkov Telescope Array. These observations were motivated by the discovery of a cluster of >30 GeV photons in the first year of Fermi Large Area Telescope observations. VER J0521+211 is relatively bright at TeV energies, with a mean photon flux of (1.93 +/- 0.13(stat) +/- 0.78(sys)) x 10(-11) cm(-2) s(-1) above 0.2 TeV during the period of the VERITAS observations. The source is strongly variable on a daily timescale across all wavebands, from optical to TeV, with a peak flux corresponding to similar to 0.3 times the steady Crab Nebula flux at TeV energies. Follow-up observations in the optical and X-ray bands classify the newly discovered TeV source as a BL Lac-type blazar with uncertain redshift, although recent measurements suggest z = 0.108. VER J0521+211 exhibits all the defining properties of blazars in radio, optical, X-ray, and gamma-ray wavelengths.
We report results from TeV gamma-ray observations of the microquasar Cygnus X-3. The observations were made with the Very Energetic Radiation Imaging Telescope Array System (VERITAS) over a time period from 2007 June 11 to 2011 November 28. VERITAS is most sensitive to gamma rays at energies between 85 GeV and 30 TeV. The effective exposure time amounts to a total of about 44 hr, with the observations covering six distinct radio/X-ray states of the object. No significant TeV gamma-ray emission was detected in any of the states, nor with all observations combined. The lack of a positive signal, especially in the states where GeV gamma rays were detected, places constraints on TeV gamma-ray production in Cygnus X-3. We discuss the implications of the results.
We report on the detection of a very rapid TeV gamma-ray flare from BL Lacertae on 2011 June 28 with the Very Energetic Radiation Imaging Telescope Array System (VERITAS). The flaring activity was observed during a 34.6 minute exposure, when the integral flux above 200 GeV reached (3.4 +/- 0.6) x 10(-6) photons m(-2) s(-1), roughly 125% of the Crab Nebula flux measured by VERITAS. The light curve indicates that the observations missed the rising phase of the flare but covered a significant portion of the decaying phase. The exponential decay time was determined to be 13 +/- 4 minutes, making it one of the most rapid gamma-ray flares seen from a TeV blazar. The gamma-ray spectrum of BL Lacertae during the flare was soft, with a photon index of 3.6 +/- 0.4, which is in agreement with the measurement made previously by MAGIC in a lower flaring state. Contemporaneous radio observations of the source with the Very Long Baseline Array revealed the emergence of a new, superluminal component from the core around the time of the TeV gamma-ray flare, accompanied by changes in the optical polarization angle. Changes in flux also appear to have occurred at optical, UV, and GeV gamma-ray wavelengths at the time of the flare, although they are difficult to quantify precisely due to sparse coverage. A strong flare was seen at radio wavelengths roughly four months later, which might be related to the gamma-ray flaring activities. We discuss the implications of these multiwavelength results.
This review presents recommended nomenclature for the biosynthesis of ribosomally synthesized and post-translationally modified peptides (RiPPs), a rapidly growing class of natural products. The current knowledge regarding the biosynthesis of the >20 distinct compound classes is also reviewed, and commonalities are discussed.

Daneben dient die Zeitschrift als Forum zur Positionierung der Fächer Jüdische Studien und Judaistik innerhalb des wissenschaftlichen Diskurses sowie zur Diskussion ihrer historischen und gesellschaftlichen Verantwortung.
The main goal of our target article was to provide concrete recommendations for improving the replicability of research findings. Most of the comments focus on this point. In addition, a few comments were concerned with the distinction between replicability and generalizability and the role of theory in replication. We address all comments within the conceptual structure of the target article and hope to convince readers that replication in psychological science amounts to much more than hitting the lottery twice.
Replicability of findings is at the heart of any empirical science. The aim of this article is to move the current replicability debate in psychology towards concrete recommendations for improvement. We focus on research practices but also offer guidelines for reviewers, editors, journal management, teachers, granting institutions, and university promotion committees, highlighting some of the emerging and existing practical solutions that can facilitate implementation of these recommendations. The challenges for improving replicability in psychological science are systemic. Improvement can occur only if changes are made at many levels of practice, evaluation, and reward.
Für die vorliegende Studie »Qualitative Untersuchung zur Akzeptanz des neuen Personalausweises und Erarbeitung von Vorschlägen zur Verbesserung der Usability der Software AusweisApp« arbeitete ein Innovationsteam mit Hilfe der Design Thinking Methode an der Aufgabenstellung »Wie können wir die AusweisApp für Nutzer intuitiv und verständlich gestalten?« Zunächst wurde die Akzeptanz des neuen Personalausweises getestet. Bürger wurden zu ihrem Wissensstand und ihren Erwartungen hinsichtlich des neuen Personalausweises befragt, darüber hinaus zur generellen Nutzung des neuen Personalausweises, der Nutzung der Online-Ausweisfunktion sowie der Usability der AusweisApp. Weiterhin wurden Nutzer bei der Verwendung der aktuellen AusweisApp beobachtet und anschließend befragt. Dies erlaubte einen tiefen Einblick in ihre Bedürfnisse. Die Ergebnisse aus der qualitativen Untersuchung wurden verwendet, um Verbesserungsvorschläge für die AusweisApp zu entwickeln, die den Bedürfnissen der Bürger entsprechen. Die Vorschläge zur Optimierung der AusweisApp wurden prototypisch umgesetzt und mit potentiellen Nutzern getestet. Die Tests haben gezeigt, dass die entwickelten Neuerungen den Bürgern den Zugang zur Nutzung der Online-Ausweisfunktion deutlich vereinfachen. Im Ergebnis konnte festgestellt werden, dass der Akzeptanzgrad des neuen Personalausweises stark divergiert. Die Einstellung der Befragten reichte von Skepsis bis hin zu Befürwortung. Der neue Personalausweis ist ein Thema, das den Bürger polarisiert. Im Rahmen der Nutzertests konnten zahlreiche Verbesserungspotenziale des bestehenden Service Designs sowohl rund um den neuen Personalausweis, als auch im Zusammenhang mit der verwendeten Software aufgedeckt werden. Während der Nutzertests, die sich an die Ideen- und Prototypenphase anschlossen, konnte das Innovtionsteam seine Vorschläge iterieren und auch verifizieren. Die ausgearbeiteten Vorschläge beziehen sich auf die AusweisApp. Die neuen Funktionen umfassen im Wesentlichen: · den direkten Zugang zu den Diensteanbietern, · umfangreiche Hilfestellungen (Tooltips, FAQ, Wizard, Video), · eine Verlaufsfunktion, · einen Beispieldienst, der die Online-Ausweisfunktion erfahrbar macht. Insbesondere gilt es, den Nutzern mit der neuen Version der AusweisApp Anwendungsfelder für ihren neuen Personalausweis und einen Mehrwert zu bieten. Die Ausarbeitung von weiteren Funktionen der AusweisApp kann dazu beitragen, dass der neue Personalausweis sein volles Potenzial entfalten kann.

In a systematic approach we synthesized a new series of fluorescent probes incorporating donoracceptor (D-A) substituted 1,2,3-triazoles as conjugative -linkers between the alkali metal ion receptor N-phenylaza-[18]crown-6 and different fluorophoric groups with different electron-acceptor properties (4-naphthalimide, meso-phenyl-BODIPY and 9-anthracene) and investigated their performance in organic and aqueous environments (physiological conditions). In the charge-transfer (CT) type probes 1, 2 and 7, the fluorescence is almost completely quenched by intramolecular CT (ICT) processes involving charge-separated states. In the presence of Na+ and K+ ICT is interrupted, which resulted in a lighting-up of the fluorescence in acetonitrile. Among the investigated fluoroionophores, compound 7, which contains a 9-anthracenyl moiety as the electron-accepting fluorophore, is the only probe which retains light-up features in water and works as a highly K+/Na+-selective probe under simulated physiological conditions. Virtually decoupled BODIPY-based 6 and photoinduced electron transfer (PET) type probes 35, where the 10-substituted anthracen-9-yl fluorophores are connected to the 1,2,3-triazole through a methylene spacer, show strong ion-induced fluorescence enhancement in acetonitrile, but not under physiological conditions. Electrochemical studies and theoretical calculations were used to assess and support the underlying mechanisms for the new ICT and PET 1,2,3-triazole fluoroionophores.
Herein, we report the synthesis of two phenylaza-[18]crown-6 lariat ethers with a coumarin fluorophore (1 and 2) and we reveal that compound 1 is an excellent probe for K+ ions under simulated physiological conditions. The presence of a 2-methoxyethoxy lariat group at the ortho position of the anilino moiety is crucial to the substantially increased stability of compounds 1 and 2 over their lariat-free phenylaza-[18] crown-6 ether analogues. Probe 1 shows a high K+/Na+ selectivity and a 2.5-fold fluorescence enhancement was observed in the presence of 100 mm K+ ions. A fluorescent membrane sensor, which was prepared by incorporating probe 1 into a hydrogel, showed a fully reversible response, a response time of 150 s, and a signal change of 7.8% per 1 mm K+ within the range 1-10 mm K+. The membrane was easily fabricated (only a single sensing layer on a solid polyester support), yet no leaching was observed. Moreover, compound 1 rapidly permeated into cells, was cytocompatible, and was suitable for the fluorescent imaging of K+ ions on both the extracellular and intracellular levels.
In aquatic systems, terrestrial dissolved organic matter (t-DOM) is known to stimulate bacterial activities in the water column, but simultaneous effects of autumnal leaf input on water column and sediment microbial dynamics in littoral zones of lakes remain largely unknown. The study's objective was to determine the effects of leaf litter on bacterial metabolism in the littoral water and sediment, and subsequently, the consequences for carbon cycling and food web dynamics. Therefore, in late fall, we simultaneously measured water and sediment bacterial metabolism in the littoral zone of a temperate shallow lake after adding terrestrial particulate organic matter (t-POM), namely, maize leaves. To better evaluate bacterial production (BP) and community respiration (CR) in sediments, we incubated sediment cores with maize leaves of different quality (nonleached and leached) under controlled laboratory conditions. Additionally, to quantify the incorporated leaf carbon into microbial biomass, we determined carbon isotopic ratios of fatty acids from sediment and leaf-associated microbes from a laboratory experiment using C-13-enriched beech leaves. The concentrations of dissolved organic carbon (DOC) increased significantly in the lake after the addition of maize leaves, accompanied by a significant increase in water BP. In contrast, sediment BP declined after an initial peak, showing no positive response to t-POM addition. Sediment BP and CR were also not stimulated by t-POM in the laboratory experiment, either in short-term or in long-term incubations, except for a short increase in CR after 18 hours. However, this increase might have reflected the metabolism of leaf-associated microorganisms. We conclude that the leached t-DOM is actively incorporated into microbial biomass in the water column but that the settling leached t-POM (t-POML) does not enter the food web via sediment bacteria. Consequently, t-POML is either buried in the sediment or introduced into the aquatic food web via microorganisms (bacteria and fungi) directly associated with t-POML and via benthic macroinvertebrates by shredding of t-POML. The latter pathway represents a benthic shortcut which efficiently transfers t-POML to higher trophic levels.
Standard quantum chemical methods are used for accurate calculation of thermochemical properties such as enthalpies of formation, entropies and Gibbs energies of formation. Equilibrium reactions are widely investigated and experimental measurements often lead to a range of reaction Gibbs energies and equilibrium constants. It is useful to calculate these equilibrium properties from quantum chemical methods in order to address the experimental differences. Furthermore, most standard calculation methods differ in accuracy and feasibility of the system size. Hence, asystematic comparison of equilibrium properties calculated with different numerical algorithms would provide a useful reference. We select two well-known gas phase equilibrium reactions with small molecules: covalent dimer formation of NO2 (2NO(2) reversible arrow N2O4) and the synthesis of NH3 (N-2 + 3 H-2 reversible arrow 2NH(3)). We test four quantum chemical methods denoted by G3B3, CBS-APNO, W1 and CCSD(T) with aug-cc-pVXZ basis sets (X = 2, 3, and 4), to obtain thermochemical data for NO2, N2O4, and NH3. The calculated standard formation Gibbs energies Delta(f)G degrees are used to calculate standard reaction Gibbs energies Delta(r)G degrees and standard equilibrium constants K-eq for the two reactions. Standard formation enthalpies Delta H-f degrees are calculated in a more reliable way using high-level methods such as W1 and CCSD(T). Standard entropies S degrees for the molecules are calculated well within the range of experiments for all methods, however, the values of standard formation Gibbs energies Delta(f)G degrees show some dependence on the choice of the method. High-level methods perform better for the calculation of molecular energies, however, simpler methods such as G3B3 and CBS-APNO perform quite well in the calculation of total reaction energies and equilibrium constants, provided that the chemical species involved do not exhibit molecular geometries that are difficult to handle by the applied method. The temperature dependence of standard reaction Gibbs energy Delta(r)G degrees for the NH3 reaction is discussed by using the calculated standard formation Gibbs energies Delta(f)G degrees of the reaction species at 298.15 K. The corresponding equilibrium constant K-eq as a function of temperature is found to be close to experimental values.

Several mechanisms are proposed to be part of the earthquake triggering process, including static stress interactions and dynamic stress transfer. Significant differences of these mechanisms are particularly expected in the spatial distribution of aftershocks. However, testing the different hypotheses is challenging because it requires the consideration of the large uncertainties involved in stress calculations as well as the appropriate consideration of secondary aftershock triggering which is related to stress changes induced by smaller pre- and aftershocks. In order to evaluate the forecast capability of different mechanisms, I take the effect of smaller--magnitude earthquakes into account by using the epidemic type aftershock sequence (ETAS) model where the spatial probability distribution of direct aftershocks, if available, is correlated to alternative source information and mechanisms. Surface shaking, rupture geometry, and slip distributions are tested. As an approximation of the shaking level, ShakeMaps are used which are available in near real-time after a mainshock and thus could be used for first-order forecasts of the spatial aftershock distribution. Alternatively, the use of empirical decay laws related to minimum fault distance is tested and Coulomb stress change calculations based on published and random slip models. For comparison, the likelihood values of the different model combinations are analyzed in the case of several well-known aftershock sequences (1992 Landers, 1999 Hector Mine, 2004 Parkfield). The tests show that the fault geometry is the most valuable information for improving aftershock forecasts. Furthermore, they reveal that static stress maps can additionally improve the forecasts of off--fault aftershock locations, while the integration of ground shaking data could not upgrade the results significantly. In the second part of this work, I focused on a procedure to test the information content of inverted slip models. This allows to quantify the information gain if this kind of data is included in aftershock forecasts. For this purpose, the ETAS model based on static stress changes, which is introduced in part one, is applied. The forecast ability of the models is systematically tested for several earthquake sequences and compared to models using random slip distributions. The influence of subfault resolution and segment strike and dip is tested. Some of the tested slip models perform very good, in that cases almost no random slip models are found to perform better. Contrastingly, for some of the published slip models, almost all random slip models perform better than the published slip model. Choosing a different subfault resolution hardly influences the result, as long the general slip pattern is still reproducible. Whereas different strike and dip values strongly influence the results depending on the standard deviation chosen, which is applied in the process of randomly selecting the strike and dip values.
A Biosensor for aromatic aldehydes comprising the mediator dependent PaoABC-Aldehyde oxidoreductase

A novel aldehyde oxidoreductase (PaoABC) from Escherichia coli was utilized for the development of an oxygen insensitive biosensor for benzaldehyde. The enzyme was immobilized in polyvinyl alcohol and currents were measured for aldehyde oxidation with different one and two electron mediators with the highest sensitivity for benzaldehyde in the presence of hexacyanoferrate(III). The benzaldehyde biosensor was optimized with respect to mediator concentration, enzyme loading and pH using potassium hexacyanoferrate(III). The linear measuring range is between 0.5200 mu M benzaldehyde. In correspondence with the substrate selectivity of the enzyme in solution the biosensor revealed a preference for aromatic aldehydes and less effective conversion of aliphatic aldehydes. The biosensor is oxygen independent, which is a particularly attractive feature for application. The biosensor can be applied to detect contaminations with benzaldehyde in solvents such as benzyl alcohol, where traces of benzaldehyde in benzyl alcohol down to 0.0042?% can be detected.
An unusual behavior of the periplasmic aldehyde oxidoreductase (PaoABC) from Escherichia coil has been observed from electrochemical investigations of the enzyme catalyzed oxidation of aromatic aldehydes with different mediators under different conditions of ionic strength. The enzyme has similarity to other molybdoenzymes of the xanthine oxidase family, but the catalytic behavior turned out to be very different. Under steady state conditions the turnover of PaoABC is maximal at pH 4 for the negatively charged ferricyanide and at pH 9 for a positively charged osmium complex. Stopped-flow kinetic measurements of the catalytic half reaction showed that oxidation of benzaldehyde proceeds also above pH 7. Thus, benzaldehyde oxidation can proceed under acidic and basic conditions using this enzyme, a property which has not been described before for molybdenum hydroxylases. It is also suggested that the electron transfer with artificial electron acceptors and PaoABC can proceed at different protein sites and depends on the nature of the electron acceptor in addition to the ionic strength. Die vorliegende Arbeit untersucht das sozio-kulturelle und institutionelle Umfeld der Organisationen des öffentlichen Sektors in der Mongolei, das signifikante Einflüsse auf die aktuellen Reformbemühungen in der öffentlichen Verwaltung hat. Die Studie stützt sich auf die Kultur- und Werttheorie. Die regelkonforme Verhaltensweise, Gemeinschaftsfavorisierende strenge Hierarchie, die fatalistische Annahme einer Autorität als unvermeidlich und unkontrollierbar sowie ein auf möglichst eigenständige Entscheidung und Meinungsbildung angestrebter Individualismus sind die weitverbreiteten kulturellen Verhaltensformen bei den Organisationen des öffentlichen Sektors der Mongolei. Dementsprechend streben die Beschäftigten des öffentlichen Dienstes uneigennützig das Wohlergehen der Bevölkerung, die Einhaltung der öffentlichen Regeln, die einvernehmlichen Beziehungen der Menschen zueinander sowie die Sicherheit und Nachhaltigkeit des Lebens an. Bestimmte Wertvorstellungen zur Selbstbestimmung, wie persönliche Geisteshaltung, eigenständiges Handeln sowie Kreativität sind für sie sehr wichtig. Dieser sozio- kulturelle Kontext hat große Auswirkungen auf das Arbeitsverhalten und auf die Aktivitäten der Beschäftigten des öffentlichen Dienstes zur Umsetzung von Reformen in der öffentlichen Verwaltung. Daher ist eine institutionelle Führung als Förderer und Beschützer von Wertesystemen bei der Umsetzung von Reformen in den hiesigen Institutionen unerlässlich.
Ionogel fiber mats were made by electrospinning poly(methylmethacrylate) (PMMA) and the ionic liquid (IL) bis(1-butyl-3-methyl-imidazolium) tetrachloridocupraten, [Bmim](2)[CuCl4], from acetone. The morphology of the electrospun ionogels strongly depends on the spinning parameters. Dense and uniform fiber mats were only obtained at concentrations of 60 to 70 g of polymer and IL mass combined. Lower concentrations led to a low number of poorly defined fibers. High voltages of 20 to 25 kV led to well-defined and uniform fibers; voltages between 15 and 20 kV again led to less uniform and less dense fibers. At 10 kV and lower, no spinning could be induced. Finally, PMMA fibers electrospun without IL show a less well-defined morphology combining fibers and oblong droplets indicating that the IL has a beneficial effect on the electrospinning process. The resulting materials are prototypes for new functional materials, for example in sterile filtration.
We consider systems of Euler-Lagrange equations with two degrees of freedom and with Lagrangian being quadratic in velocities. For this class of equations the generic case of the equivalence problem is solved with respect to point transformations. Using Lie's infinitesimal method we construct a basis of differential invariants and invariant differentiation operators for such systems. We describe certain types of Lagrangian systems in terms of their invariants. The results are illustrated by several examples.
The new N-heterocyclic carbene (NHC) precursors 4,5-dicyano-1, -dimesityl- (9) and 4, 5-dicyano-1, 3-dineopentyl-2-(pentafluorophenyl)imidazoline (14) were synthesized. 9 could be determined by X-ray crystallography. With the 2-pentafluorophenyl-substituted imidazolines 9 and 14, the [AgCl(NHC)], [RhCl(COD)(NHC)], and [RhCl(CO)(2)(NHC)] complexes [NHC = 4, 5-dicyano-1, 3-dimesitylimidazol-2-ylidene (3) and 4, 5-dicyano-1, 3-dineopentylimidazol-2-ylidene (4)] were obtained. Crystal structures of [AgCl(3)] (15), [RhCl(COD)(3)] (17), [RhCl(COD)(4)] (18), and [RhCl(CO)(2)(3)] (19) were solved and with the crystal data of 19, the percent buried volume (%V-bur) of 31.8(+/- 0.1)% was determined for NHC 3. Infrared spectra of the imidazolines 9 and 14 and of the complexes 15-20 were recorded and the CO stretching frequencies of complexes 19 and 20 were used to determine the 3 ( (-1)) and 4 ( (-1)), thus proving that 1, 3-substitution of maleonitrile-NHCs does not have a significant effect for the high -acceptor strength of these carbenes.
The distribution of poikilotherms is determined by the thermal structure of the marine environment that they are exposed to. Recent research has indicated that changes in migration phenology of beluga whales in the Arctic are triggered by changes in the thermal structure of the marine environment in their summering area. If sea temperatures reflect the spatial distribution of food resources, then changes in the thermal regime will affect how homogeneous or clumped food is distributed. We explore, by individual-based modelling, the hypothesis that changes in migration phenology are not necessarily or exclusively triggered by changes in food abundance, but also by changes in the spatial aggregation of food. We found that the level of food aggregation can significantly affect the relationship between the timing of the start of migration to the winter grounds and the total prey capture of individuals. Our approach strongly indicates that changes in the spatial distribution of food resources should be considered for understanding and quantitatively predicting changes in the phenology of animal migration.

Antarctic glacier forfields are extreme environments and pioneer sites for ecological succession. The Antarctic continent shows microbial community development as a natural laboratory because of its special environment, geographic isolation and little anthropogenic influence. Increasing temperatures due to global warming lead to enhanced deglaciation processes in cold-affected habitats and new terrain is becoming exposed to soil formation and accessible for microbial colonisation. This study aims to understand the structure and development of glacier forefield bacterial communities, especially how soil parameters impact the microorganisms and how those are adapted to the extreme conditions of the habitat. To this effect, a combination of cultivation experiments, molecular, geophysical and geochemical analysis was applied to examine two glacier forfields of the Larsemann Hills, East Antarctica. Culture-independent molecular tools such as terminal restriction length polymorphism (T-RFLP), clone libraries and quantitative real-time PCR (qPCR) were used to determine bacterial diversity and distribution. Cultivation of yet unknown species was carried out to get insights in the physiology and adaptation of the microorganisms. Adaptation strategies of the microorganisms were studied by determining changes of the cell membrane phospholipid fatty acid (PLFA) inventory of an isolated bacterium in response to temperature and pH fluctuations and by measuring enzyme activity at low temperature in environmental soil samples. The two studied glacier forefields are extreme habitats characterised by low temperatures, low water availability and small oligotrophic nutrient pools and represent sites of different bacterial succession in relation to soil parameters. The investigated sites showed microbial succession at an early step of soil formation near the ice tongue in comparison to closely located but rather older and more developed soil from the forefield. At the early step the succession is influenced by a deglaciation-dependent areal shift of soil parameters followed by a variable and prevalently depth-related distribution of the soil parameters that is driven by the extreme Antarctic conditions. The dominant taxa in the glacier forefields are Actinobacteria, Acidobacteria, Proteobacteria, Bacteroidetes, Cyanobacteria and Chloroflexi. The connection of soil characteristics with bacterial community structure showed that soil parameter and soil formation along the glacier forefield influence the distribution of certain phyla. In the early step of succession the relative undifferentiated bacterial diversity reflects the undifferentiated soil development and has a high potential to shift according to past and present environmental conditions. With progressing development environmental constraints such as water or carbon limitation have a greater influence. Adapting the culturing conditions to the cold and oligotrophic environment, the number of culturable heterotrophic bacteria reached up to 108 colony forming units per gram soil and 148 isolates were obtained. Two new psychrotolerant bacteria, Herbaspirillum psychrotolerans PB1T and Chryseobacterium frigidisoli PB4T, were characterised in detail and described as novel species in the family of Oxalobacteraceae and Flavobacteriaceae, respectively. The isolates are able to grow at low temperatures tolerating temperature fluctuations and they are not specialised to a certain substrate, therefore they are well-adapted to the cold and oligotrophic environment. The adaptation strategies of the microorganisms were analysed in environmental samples and cultures focussing on extracellular enzyme activity at low temperature and PLFA analyses. Extracellular phosphatases (pH 11 and pH 6.5), β-glucosidase, invertase and urease activity were detected in the glacier forefield soils at low temperature (14°C) catalysing the conversion of various compounds providing necessary substrates and may further play a role in the soil formation and total carbon turnover of the habitat. The PLFA analysis of the newly isolated species C. frigidisoli showed that the cold-adapted strain develops different strategies to maintain the cell membrane function under changing environmental conditions by altering the PLFA inventory at different temperatures and pH values. A newly discovered fatty acid, which was not found in any other microorganism so far, significantly increased at decreasing temperature and low pH and thus plays an important role in the adaption of C. frigidisoli. This work gives insights into the diversity, distribution and adaptation mechanisms of microbial communities in oligotrophic cold-affected soils and shows that Antarctic glacier forefields are suitable model systems to study bacterial colonisation in connection to soil formation.
The “triple bottom line” concept (planet, people, and profit) represents an important guideline for the sustainable, hence future-oriented, development of societies and for the behaviors of all societal members. For institutions promoting societal change, as well as for companies being confronted with growing expectations regarding compelling contributions to sustainable changes, it is of great importance to know if, and to what extent, consumers have already internalized the idea of sustainability. Against the background of existing research gaps regarding a comprehensive measurement of the consciousness for sustainable consumption (CSC), the authors present the result of a scale development. Consciousness was operationalized by weighting personal beliefs with the importance attached by consumers to sustainability dimensions. Four separate tests of the CSC scale indicated an appropriate psychometric quality of the scale and provided support for this new measurement approach that incorporates the environmental, social and economic dimensions of sustainability.

Compared with the vast amount of studies concerning environmentally conscious consumer behaviour, relatively little is known about socially conscious consumption. The present paper focuses on fair consumption as an important aspect of social consumption. In our study, consciousness for fair consumption (CFC) is defined as a latent disposition of consumers to prefer products that are produced and traded in compliance with fair labour and business practices. A scale to measure CFC was conceptualized and tested in three independent empirical studies. Two studies were conducted at European universities (2010 and 2012) and used 352 and 362 undergraduate business students respectively. The third study, conducted in 2011, used 141 employees at a European university. The results confirmed the reliability and validity of the new CFC scale across samples. While being moderately related to other aspects of sustainable consumption such as ecological concern and moral reasoning, CFC was significantly distinct from those concepts. Most importantly, it was established that the CFC, as measured by the new CFC scale, is a strong determinant of consumption of fair trade products that has been neglected in existing research.

The Alborz range of N Iran provides key information on the spatiotemporal evolution and characteristics of the Arabia-Eurasia continental collision zone. The southwestern Alborz range constitutes a transpressional duplex, which accommodates oblique shortening between Central Iran and the South Caspian Basin. The duplex comprises NW-striking frontal ramps that are kinematically linked to inherited E-W-striking, right-stepping lateral to obliquely oriented ramps. New zircon and apatite (U-Th)/He data provide a high-resolution framework to unravel the evolution of collisional tectonics in this region. Our data record two pulses of fast cooling associated with SW-directed thrusting across the frontal ramps at similar to 18-14 and 9.5-7.5 Ma, resulting in the tectonic repetition of a fossil zircon partial retention zone and a cooling pattern with a half U-shaped geometry. Uniform cooling ages of similar to 7-6 Ma along the southernmost E-W striking oblique ramp and across its associated NW-striking frontal ramps suggests that the ramp was reactivated as a master throughgoing, N-dipping thrust. We interpret this major change in fault kinematics and deformation style to be related to a change in the shortening direction from NE to N/NNE. The reduction in the obliquity of thrusting may indicate the termination of strike-slip faulting (and possibly thrusting) across the Iranian Plateau, which could have been triggered by an increase in elevation. Furthermore, we suggest that similar to 7-6-m.y.-old S-directed thrusting predated inception of the westward motion of the South Caspian Basin. Citation: Ballato, P., D. F. Stockli, M. R. With this paper, we assess the present-day conductive thermal field of the Glueckstadt Graben in NW Germany that is characterized by large salt walls and diapirs structuring the graben fill. We use a finite element method to calculate the 3D steady-state conductive thermal field based on a lithosphere-scale 3D structural model that resolves the first-order structural characteristics of the graben and its underlying lithosphere. Model predictions are validated against measured temperatures in six deep wells. Our investigations show that the interaction of thickness distributions and thermal rock properties of the different geological layers is of major importance for the distribution of temperatures in the deep subsurface of the Glueckstadt Graben. However, the local temperatures may result from the superposed effects of different controlling factors. Especially, the upper sedimentary part of the model exhibits huge lateral temperature variations, which correlate spatially with the shape of the thermally highly conductive Permian salt layer. Variations in thickness and geometry of the salt cause two major effects, which provoke considerable lateral temperature variations for a given depth. (1) The "chimney effect" causes more efficient heat transport within salt diapirs. As a consequence positive thermal anomalies develop in the upper part and above salt structures, where the latter are covered by much less conductive sediments. In contrast, negative thermal anomalies are noticeable underneath salt structures. (2) The "thermal blanketing effect" is caused by thermally low conductive sediments that provoke the local storage of heat where these insulating sediments are present. The latter effect leads to both local and regional thermal anomalies. Locally, this translates to higher temperatures where salt margin synclines are filled with thick insulating clastic sediments. For the regional anomalies the cumulative insulating effects of the entire sediment fill results in a long-wavelength variation of temperatures in response to heat refraction effects caused by the contrast between insulating sediments and highly conductive crystalline crust. Finally, the longest wavelength of temperature variations is caused by the depth position of the isothermal lithosphere-asthenosphere boundary defining the regional variations of the overall geothermal gradient. We find that a conductive thermal model predicts observed temperatures reasonably well for five of the six available wells, whereas the steady-state conductive approach appears not to be valid for the sixth well.
The course timetabling problem can be generally defined as the task of assigning a number of lectures to a limited set of timeslots and rooms, subject to a given set of hard and soft constraints. The modeling language for course timetabling is required to be expressive enough to specify a wide variety of soft constraints and objective functions. Furthermore, the resulting encoding is required to be extensible for capturing new constraints and for switching them between hard and soft, and to be flexible enough to deal with different formulations. In this paper, we propose to make effective use of ASP as a modeling language for course timetabling. We show that our ASP-based approach can naturally satisfy the above requirements, through an ASP encoding of the curriculum-based course timetabling problem proposed in the third track of the second international timetabling competition (ITC-2007). Our encoding is compact and human-readable, since each constraint is individually expressed by either one or two rules. Each hard constraint is expressed by using integrity constraints and aggregates of ASP. Each soft constraint S is expressed by rules in which the head is the form of penalty (S, V, C), and a violation V and its penalty cost C are detected and calculated respectively in the body. We carried out experiments on four different benchmark sets with five different formulations. We succeeded either in improving the bounds or producing the same bounds for many combinations of problem instances and formulations, compared with the previous best known bounds.

Furthermore, the resulting encoding is required to be extensible for capturing new constraints and for switching them between hard and soft, and to be flexible enough to deal with different formulations. In this paper, we propose to make effective use of ASP as a modeling language for course timetabling. We show that our ASP-based approach can naturally satisfy the above requirements, through an ASP encoding of the curriculum-based course timetabling problem proposed in the third track of the second international timetabling competition (ITC-2007). Our encoding is compact and human-readable, since each constraint is individually expressed by either one or two rules. Each hard constraint is expressed by using integrity constraints and aggregates of ASP. Each soft constraint S is expressed by rules in which the head is the form of penalty (S, V, C), and a violation V and its penalty cost C are detected and calculated respectively in the body. We carried out experiments on four different benchmark sets with five different formulations. We succeeded either in improving the bounds or producing the same bounds for many combinations of problem instances and formulations, compared with the previous best known bounds.
The time-dependent approach to electronic spectroscopy, as popularized by Heller and coworkers in the 1980's, is applied here in conjunction with linear-response, time-dependent density functional theory to study vibronic absorption, emission and resonance Raman spectra of several diamondoids. Two-state models, the harmonic and the Condon approximations, are used for the calculations, making them easily applicable to larger molecules. The method is applied to nine pristine lower and higher diamondoids: adamantane, diamantane, triamantane, and three isomers each of tetramantane and pentamantane. We also consider a hybrid species “Dia = Dia” – a shorthand notation for a recently synthesized molecule comprising two diamantane units connected by a C[double bond, length as m-dash]C double bond. We resolve and interpret trends in optical and vibrational properties of these molecules as a function of their size, shape, and symmetry, as well as effects of “blending” with sp2-hybridized C-atoms. Time-dependent correlation functions facilitate the computations and shed light on the vibrational dynamics following electronic transitions.

Soil moisture dynamics are affected by complex interactions among several factors. Understanding the relative importance of these factors is still an important challenge in the study of water fluxes and solute transport in unsaturated media. In this study, the spatio-temporal variability of surface soil moisture was investigated in a 10 ha flat cropped field located in northern Italy. Soil moisture was measured on a regular 50 x 50 m grid on seven dates during the growing season. For each measurement campaign, the spatial variability of the soil moisture was compared with the spatial variability of the soil texture and crop properties. In particular, to better understand the role of the vegetation, the spatio-temporal variability of two different parameters - leaf area index and crop height - was monitored on eight dates at different crop development stages. Statistical and geostatistical analysis was then applied to explore the interactions between these variables. In agreement with other studies, the results show that the soil moisture variability changes according to the average value within the field, with the standard deviation reaching a maximum value under intermediate mean soil moisture conditions and the coefficient of variation decreasing exponentially with increasing mean soil moisture. The controls of soil moisture variability change according to the average soil moisture within the field. Under wet conditions, the spatial distribution of the soil moisture reflects the variability of the soil texture. Under dry conditions, the spatial distribution of the soil moisture is affected mostly by the spatial variability of the vegetation. The interaction between these two factors is more important under intermediate soil moisture conditions. These results confirm the importance of considering the average soil moisture conditions within a field when investigating the controls affecting the spatial variability of soil moisture. This study highlights the importance of considering the spatio-temporal variability of the vegetation in investigating soil moisture dynamics, especially under intermediate and dry soil moisture conditions. The results of this study have important implications in different hydrological applications, such as for sampling design, ranking stability application, indirect measurements of soil properties and model parameterisation.
Whenever eye movements are measured, a central part of the analysis has to do with where subjects fixate and why they fixated where they fixated. To a first approximation, a set of fixations can be viewed as a set of points in space; this implies that fixations are spatial data and that the analysis of fixation locations can be beneficially thought of as a spatial statistics problem. We argue that thinking of fixation locations as arising from point processes is a very fruitful framework for eye-movement data, helping turn qualitative questions into quantitative ones. We provide a tutorial introduction to some of the main ideas of the field of spatial statistics, focusing especially on spatial Poisson processes. We show how point processes help relate image properties to fixation locations. In particular we show how point processes naturally express the idea that image features' predictability for fixations may vary from one image to another. We review other methods of analysis used in the literature, show how they relate to point process theory, and argue that thinking in terms of point processes substantially extends the range of analyses that can be performed and clarify their interpretation.
The spatial distribution of soil types is controlled by a set of environmental factors such as climate, organisms, parent material and topography as well as time and space. A change of these factors will lead to a change in the spatial distribution of soil types. In this study, we use a digital soil mapping approach to improve our knowledge about major soil type distributing factors in the steppe regions of Inner Mongolia (China) which currently undergo tremendous environmental change, e.g. climate and land use change. We use Random Forests in an effort to map Reference Soil Groups according to the World Reference Base for Soil Resources (WRB) in the Xilin River catchment. We benefit from the superior prediction capabilities of RF and additional interpretive results in order to identify the major environmental factors that control spatial patterns of soil types. The nine WRB soil groups that were identified and spatially predicted for the study area are Arenosol, Calcisol, Cambisol, Chernozem, Cryosol, Gleysol, Kastanozem, Phaeozem and Regosol.
Model and prediction performances of the RF model are high with an Out-of-Bag error of 51.6% for the model and a misclassification error for the predicted map of 28.9%. The main controlling factors of soil type distribution are land use, a set of topographic variables, geology and climate. However, land use and climate are of major importance and topography and geology are of minor importance. The visualizations of the predictions, the variable importance measures as result of RF and the comparisons of these with the spatial distribution of the environmental factors delivered additional, quantitative information of these controlling factors and revealed that intensively grazed areas are subjected to soil degradation. However, most of the area is still governed by natural soil forming processes which are driven by climate, topography and geology. Most importantly though, our study revealed that a shift towards warmer temperatures and lower precipitation regimes will lead to a change of the spatial distribution of RSGs towards steppe soils that store less carbon, i.e. a decrease of spatial extent of Phaeozems and an increase of spatial extent of Chernozems and Kastanozems.
A qualitative exploratory study was conducted to reveal students' argumentation skills in the context of the topic of evolution. Transcripts from problem-centred interviews on secondary students' beliefs about evolutionary processes of adaptation were analysed using a content analysis approach. For this purpose two categorical systems were deductively developed: one addressing the complexity of students' arguments, the other focusing on students' use of argumentation schemes. Subsequently, the categorical systems were inductively elaborated upon the basis of the analysed material showing a satisfactory inter-rater reliability. Regarding the arguments' complexity, students produced mainly single claims or claims with a single justification consisting of either data or warrants. With regard to argumentation schemes students drew their arguments mainly using causal schemes, analogies, or illustrative examples. Results are discussed in light of possible implications for teaching evolutionary theory using classroom argumentation.
The standard charging process for polymer ferroelectrets, e. g., from polypropylene foams or layered film systems involves the application of high DC fields either to metal electrodes or via a corona discharge. In this often-used process, the DC field triggers the internal breakdown and limits the final charge densities inside the ferroelectret cavities and, thus, the final polarization. Here, an AC + DC charging procedure is proposed and demonstrated in which a high-voltage high-frequency (HV-HF) wave train is applied together with a DC poling voltage. Thus, the internal dielectric-barrier discharges in the ferroelectret cavities are induced by the HV-HF wave train, while the final charge and polarization level is controlled separately through the applied DC voltage. In the new process, the frequency and the amplitude of the HV-HF wave train must be kept within critical boundaries that are closely related to the characteristics of the respective ferroelectrets. The charging method has been tested and investigated on a fluoropolymer-film system with a single well-defined cylindrical cavity. It is found that the internal electrical polarization of the cavity can be easily controlled and increases linearly with the applied DC voltage up to the breakdown voltage of the cavity. In the standard charging method, however, the DC voltage would have to be chosen above the respective breakdown voltage. With the new method, control of the HV-HF wave-train duration prevents a plasma-induced deterioration of the polymer surfaces inside the cavities. It is observed that the frequency of the HV-HF wave train during ferroelectret charging and the temperature applied during poling of ferroelectrics serve an analogous purpose. The analogy and the similarities between the proposed ferroelectret charging method and the poling of ferroelectric materials or dipole electrets at elevated temperatures with subsequent cooling under field are discussed.
Temporal evolution of the re-breakdown voltage in small gaps from nanoseconds to milliseconds

A detailed understanding of electric breakdown in dielectrics is of scientific and technological interest. In gaseous dielectrics, a so-called re-breakdown is sometimes observed after extinction of the previous discharge. Although time-dependent re-breakdown voltage is essentially known, its behavior immediately after the previous discharge is not precisely understood. We present an electronic circuit for accurate measurements of the time-dependent re-breakdown voltage in small gaps from tens of nanoseconds to several milliseconds after the previous spark. Results from such experiments are compared with earlier findings, and relevant physical mechanisms such as heating of the gas, decay of the plasma, and ionization of excited atoms and molecules are discussed. It is confirmed that the thermal model is not valid at times below several microseconds.
Although ring faults are present at many ancient, deeply eroded volcanoes, they have been detected at only very few modern volcanic centers. At the so far little studied Tendurek volcano in eastern Turkey, we generated an ascending and a descending InSAR time series of its surface displacement field for the period from 2003 to 2010. We detected a large (similar to 105km(2)) region that underwent subsidence at the rate of similar to 1cm/yr during this period. Source modeling results show that the observed signal fits best to simulations of a near-horizontal contracting sill located at around 4.5km below the volcano summit. Intriguingly, the residual displacement velocity field contains a steep gradient that systematically follows a system of arcuate fractures visible on the volcano’s midflanks. RapidEye satellite optical images show that this fracture system has deflected Holocene lava flows, thus indicating its presence for at least several millennia. We interpret the arcuate fracture system as the surface expression of an inherited ring fault that has been slowly reactivated during the detected recent subsidence. These results show that volcano ring faults may not only slip rapidly during eruptive or intrusive phases, but also slowly during dormant phases.
Data integration aims to combine data of different sources and to provide users with a unified view on these data. This task is as challenging as valuable. In this thesis we propose algorithms for dependency discovery to provide necessary information for data integration. We focus on inclusion dependencies (INDs) in general and a special form named conditional inclusion dependencies (CINDs): (i) INDs enable the discovery of structure in a given schema. (ii) INDs and CINDs support the discovery of cross-references or links between schemas. An IND “A in B” simply states that all values of attribute A are included in the set of values of attribute B. We propose an algorithm that discovers all inclusion dependencies in a relational data source. The challenge of this task is the complexity of testing all attribute pairs and further of comparing all of each attribute pair's values. The complexity of existing approaches depends on the number of attribute pairs, while ours depends only on the number of attributes. Thus, our algorithm enables to profile entirely unknown data sources with large schemas by discovering all INDs. Further, we provide an approach to extract foreign keys from the identified INDs. We extend our IND discovery algorithm to also find three special types of INDs: (i) Composite INDs, such as “AB in CD”, (ii) approximate INDs that allow a certain amount of values of A to be not included in B, and (iii) prefix and suffix INDs that represent special cross-references between schemas. Conditional inclusion dependencies are inclusion dependencies with a limited scope defined by conditions over several attributes. Only the matching part of the instance must adhere the dependency. We generalize the definition of CINDs distinguishing covering and completeness conditions and define quality measures for conditions. We propose efficient algorithms that identify covering and completeness conditions conforming to given quality thresholds. The challenge for this task is twofold: (i) Which (and how many) attributes should be used for the conditions? (ii) Which attribute values should be chosen for the conditions? Previous approaches rely on pre-selected condition attributes or can only discover conditions applying to quality thresholds of 100%. Our approaches were motivated by two application domains: data integration in the life sciences and link discovery for linked open data. We show the efficiency and the benefits of our approaches for use cases in these domains.
1. Models aim to predict phytoplankton dynamics based on observed initial conditions and a set of equations and parameters. However, our knowledge about initial conditions in nature is never perfect. Thus, if phytoplankton dynamics are sensitive to small variations in initial conditions, they are difficult to predict. 2. We used time-series data from indoor mesocosm experiments with natural phyto- and zooplankton communities to quantify the extent to which small initial differences in the species, functional group and community biomass in parallel treatments were amplified or buffered over time. We compared the differences in dynamics between replicates and among all mesocosms of 1year. 3. Temperature-sensitive grazing during the exponential growth phase of phytoplankton caused divergence. In contrast, negative density dependence caused convergence. 4. Mean differences in biomass between replicates were similar for all hierarchical levels. This indicates that differences in their initial conditions were amplified to the same extent. Even though large differences in biomass occasionally occurred between replicates for a short time, dynamics returned to the same path at all hierarchical levels. This suggests that internal feedback mechanisms make the spring development of phytoplankton highly predictable.
Under dilute in vitro conditions transcription factors rapidly locate their target sequence on DNA by using the facilitated diffusion mechanism. However, whether this strategy of alternating between three-dimensional bulk diffusion and one-dimensional sliding along the DNA contour is still beneficial in the crowded interior of cells is highly disputed. Here we use a simple model for the bacterial genome inside the cell and present a semi-analytical model for the in vivo target search of transcription factors within the facilitated diffusion framework. Without having to resort to extensive simulations we determine the mean search time of a lac repressor in a living E. coli cell by including parameters deduced from experimental measurements. The results agree very well with experimental findings, and thus the facilitated diffusion picture emerges as a quantitative approach to gene regulation in living bacteria cells. Furthermore we see that the search time is not very sensitive to the parameters characterizing the DNA configuration and that the cell seems to operate very close to optimal conditions for target localization. Local searches as implied by the colocalization mechanism are only found to mildly accelerate the mean search time within our model.
Development of apical membrane organization and V-ATPase regulation in blowfly salivary glands

Secretory cells in blowfly salivary gland are specialized via morphological and physiological attributes in order to serve their main function, i.e. the transport of solutes at a high rate in response to a hormonal stimulus, namely serotonin (5-HT). This study examines the way that 5-HT-insensitive precursor cells differentiate into morphologically complex 5-HT-responsive secretory cells. By means of immunofluorescence microscopy, immunoblotting and measurements of the transepithelial potential changes, we show the following. (1) The apical membrane of the secretory cells becomes organized into an elaborate system of canaliculi and is folded into pleats during the last pupal day and the first day of adulthood. (2) The structural reorganization of the apical membrane is accompanied by an enrichment of actin filaments and phosphorylated ERM protein (phospho-moesin) at this membrane domain and by the deployment of the membrane-integral part of vacuolar-type H+-ATPase (V-ATPase). These findings suggest a role for phospho-moesin, a linker between actin filaments and membrane components, in apical membrane morphogenesis. (3) The assembly and activation of V-ATPase can be induced immediately after eclosion by way of 8-CPT-cAMP, a membrane-permeant cAMP analogue. (4) 5-HT, however, produces the assembly and activation of V-ATPase only in flies aged for at least 2 h after eclosion, indicating that, at eclosion, the 5-HT receptor/adenylyl cyclase/cAMP signalling pathway is inoperative upstream of cAMP. (5) 5-HT activates both the Ca2+ signalling pathway and the cAMP signalling cascade in fully differentiated secretory cells. However, the functionality of these signalling cascades does not seem to be established in a tightly coordinated manner during cell differentation.
Background: DNA fragments carrying internal recognition sites for the restriction endonucleases intended for cloning into a target plasmid pose a challenge for conventional cloning.
Results: A method for directional insertion of DNA fragments into plasmid vectors has been developed. The target sequence is amplified from a template DNA sample by PCR using two oligonucleotides each containing a single deoxyinosine base at the third position from the 5' end. Treatment of such PCR products with endonuclease V generates 3' protruding ends suitable for ligation with vector fragments created by conventional restriction endonuclease reactions.
Conclusions: The developed approach generates terminal cohesive ends without the use of Type II restriction endonucleases, and is thus independent from the DNA sequence. Due to PCR amplification, minimal amounts of template DNA are required. Using the robust Taq enzyme or a proofreading Pfu DNA polymerase mutant, the method is applicable to a broad range of insert sequences. Appropriate primer design enables direct incorporation of terminal DNA sequence modifications such as tag addition, insertions, deletions and mutations into the cloning strategy. Further, the restriction sites of the target plasmid can be either retained or removed.

Black shales are sedimentary rocks with a high content of organic carbon, which leads to a dark grayish to black color. Due to their potential to contain oil or gas, black shales are of great interest for the support of the worldwide energy supply. An integrated seismic investigation of the Lower Palaeozoic black shales was carried out at the Danish island Bornholm to locate the shallow-lying Alum Shale layer and its surrounding formations and to characterize its potential as a source rock. Therefore, two seismic experiments at a total of three crossing profiles were carried out in October 2010 and in June 2012 in the southern part of the island. Two different active measurements were conducted with either a weight drop source or a minivibrator. Additionally, the ambient noise field was recorded at the study location over a time interval of about one day, and also a laboratory analysis of borehole samples was carried out. The seismic profiles were positioned as close as possible to two scientific boreholes which were used for comparative purposes. The seismic field data was analyzed with traveltime tomography, surface wave inversion and seismic interferometry to obtain the P-wave and S-wave velocity models of the subsurface. The P-wave velocity models which were determined for all three profiles clearly locate the Alum Shale layer between the Komstad Limestone layer on top and the Læså Sandstone Formation at the base of the models. The black shale layer has P-wave velocities around 3 km/s which are lower compared to the adjacent formations. Due to a very good agreement of the sonic log and the vertical velocity profiles of the two seismic lines, which are directly crossing the borehole where the sonic log was conducted, the reliability of the traveltime tomography is proven. A correlation of the seismic velocities with the content of organic carbon is an important task for the characterization of the reservoir properties of a black shale formation. It is not possible without calibration but in combination with a full 2D tomographic image of the subsurface it gives the subsurface distribution of the organic material. The S-wave model obtained with surface wave inversion of the vibroseis data of one of the profiles images the Alum Shale layer also very well with S-wave velocities around 2 km/s. Although individual 1D velocity models for each of the source positions were determined, the subsurface S-wave velocity distribution is very uniform with a good match between the single models. A really new approach described here is the application of seismic interferometry to a really small study area and a quite short time interval. Also new is the selective procedure of only using time windows with the best crosscorrelation signals to achieve the final interferograms. Due to the small scale of the interferometry even P-wave signals can be observed in the final crosscorrelations. In the laboratory measurements the seismic body waves were recorded for different pressure and temperature stages. Therefore, samples of different depths of the Alum Shale were available from one of the scientific boreholes at the study location. The measured velocities have a high variance with changing pressure or temperature. Recordings with wave propagation both parallel and perpendicular to the bedding of the samples reveal a great amount of anisotropy for the P-wave velocity, whereas the S-wave velocity is almost independent of the wave direction. The calculated velocity ratio is also highly anisotropic with very low values for the perpendicular samples and very high values for the parallel ones. Interestingly, the laboratory velocities of the perpendicular samples are comparable to the velocities of the field experiments indicating that the field measurements are sensitive to wave propagation in vertical direction. The velocity ratio is also calculated with the P-wave and S-wave velocity models of the field experiments. Again, the Alum Shale can be clearly separated from the adjacent formations because it shows overall very low vP/vS ratios around 1.4. The very low velocity ratio indicates the content of gas in the black shale formation. With the combination of all the different methods described here, a comprehensive interpretation of the seismic response of the black shale layer can be made and the hydrocarbon source rock potential can be estimated.
