While structural transitions of LEA proteins in response to changes in water availability are commonly observed and several molecular functions have been suggested, a systematic, comprehensive and comparative study of possible underlying sequence-structure-function relationships is still lacking. We performed molecular dynamics (MD) simulations as well as spectroscopic and light scattering experiments to characterize six members of two distinct, lowly homologous clades of LEA_4 family proteins from Arabidopsis thaliana. We compared structural and functional characteristics to elucidate to what degree structure and function are encoded in LEA protein sequences and complemented these findings with physicochemical properties identified in a systematic bioinformatics study of the entire Arabidopsis thaliana LEA_4 family. Our results demonstrate that although the six experimentally characterized LEA_4 proteins have similar structural and functional characteristics, differences concerning their folding propensity and membrane stabilization capacity during a freeze/thaw cycle are obvious. These differences cannot be easily attributed to sequence conservation, simple physicochemical characteristics or the abundance of sequence motifs. Moreover, the folding propensity does not appear to be correlated with membrane stabilization capacity. Therefore, the refinement of LEA_4 structural and functional properties is likely encoded in specific patterns of their physicochemical characteristics. Design Prospective observational study in two inpatient cardiac rehabilitation centres between September 2014 and August 2015 with a follow-up six months after cardiac rehabilitation. Method At admission to and discharge from cardiac rehabilitation, the cognitive performance of 401 patients (54.5 ± 6.3 years, 80% men) following an acute coronary syndrome and/or coronary artery bypass graft was tested using the Montreal Cognitive Assessment. Patients’ disease-related knowledge was determined using a quiz (22 items for medical knowledge and 12 items for healthy lifestyle and behaviour) at both times and at follow-up. The change in knowledge after cardiac rehabilitation was analysed in multivariable regression models. Potentially influencing parameters (e.g. level of education, medication, cardiovascular risk factors, coronary artery bypass graft, comorbidities, exercise capacity) were considered. Results During cardiac rehabilitation, disease-related knowledge was significantly enhanced in both scales. At follow-up, the average level of medical knowledge was significantly reduced, while lifestyle knowledge remained at a stable level. The maintenance of knowledge after cardiac rehabilitation was predominantly predicted by prior knowledge, cognitive performance at discharge from cardiac rehabilitation and, in the case of medical knowledge, by coronary artery bypass graft. Conclusion Patient education in cardiac rehabilitation led to enhanced disease-related knowledge, but the maintenance of this essentially depended on patients’ cognitive performance, especially after coronary artery bypass graft. Therefore, patient education concepts in cardiac rehabilitation should be reconsidered and adjusted as needed. This study shows that machine learning, equation-free statistical modeling of artificial intelligence, is a promising synthesis tool for discovering novel patterns and the source of controversy in a general hypothesis. We apply a decision tree algorithm, assuming that evidence from various contexts can be adequately integrated in a hierarchically nested structure. As a case study, we analyzed 163 articles that studied a prominent hypothesis in invasion biology, the enemy release hypothesis. We explored if any of the nine attributes that classify each study can differentiate conclusions as classification problem. Results corroborated that machine learning can be useful for research synthesis, as the algorithm could detect patterns that had been already focused in previous narrative reviews. Compared with the previous synthesis study that assessed the same evidence collection based on experts' judgement, the algorithm has newly proposed that the studies focusing on Asian regions mostly supported the hypothesis, suggesting that more detailed investigations in these regions can enhance our understanding of the hypothesis. We suggest that machine learning algorithms can be a promising synthesis tool especially where studies (a) reformulate a general hypothesis from different perspectives, (b) use different methods or variables, or (c) report insufficient information for conducting meta-analyses. High-resolution mapping of buried horizons using conventional soil surveys is destructive and time consuming. Geoelectrical sensors can offer a fast and non-destructive alternative for determining horizon positions and properties. In this paper, we compare the suitability of several geoelectrical methods for measuring the depth to buried horizons (Apb, Ahb and Hab) in the hummocky ground moraine landscape of northeastern Germany. Soil profile descriptions were developed for 269 locations within a 6-ha experimental field "CarboZALF-D". A stepwise linear discriminant analysis (LDA) estimated the lateral position of the buried horizons using electromagnetic induction data and terrain attributes. To predict the depth of a buried horizon, multiple linear regression (MLR) was used for both a 120-m transect and a 0.2-ha pseudo-three-dimensional (3D) area. At these scales, apparent electrical conductivity (ECa), electrical resistivity (ER) and terrain attributes were used as independent variables. The LDA accurately predicted Apb- and Ahb-horizons (a correct classification of 93%). The LDA of the Hab-horizon had a misclassification of 24%, which was probably related to the smaller test set and the higher depth of this horizon. The MLR predicted the depth of the Apb-, Ahb- and Hab-horizons with relative root mean square errors (RMSEs) of 7, 3 and 13%, respectively, in the pseudo-3D area. MLR had a lower accuracy for the 2D transect compared to the pseudo-3D area. Overall, the use of LDA and MLR has been an efficient methodological approach for predicting buried horizon positions. Highlights The suitability of geoelectrical measurements for digital modelling of diagnostic buried soil horizons was determined. LDA and MLR were used to detect multiple horizons with geoelectrical devices and terrain attributes. Geoelectrical variables were significant predictors of the position of the target soil horizons. The use of these tested digital technologies gives an opportunity to develop high-resolution soil mapping procedures. However, traditional observational methods exhibit limited positional accuracy. High-resolution GPS telemetry represents a promising method to overcome this drawback. In this study, we used three-dimensional GPS tracking data including high-accuracy tracks recorded at 3-s intervals to investigate the collision risk of breeding male Montagu's Harriers Circus pygargus in the Dutch–German border region. Avoidance of wind turbines was quantified by a novel approach comparing observed flights to a null model of random flight behaviour. On average, Montagu's Harriers spent as much as 8.2 h per day in flight. Most flights were at low altitude, with only 7.1% within the average rotor height range (RHR; 45–125 m). Montagu's Harriers showed significant avoidance behaviour, approaching turbines less often than expected, particularly when flying within the RHR (avoidance rate of 93.5%). For the present state, with wind farms situated on the fringes of the regional nesting range, collision risk models based on our new insights on flight behaviour indicated 0.6–2.0 yearly collisions of adult males (as compared with a population size of c. 40 pairs). However, the erection of a new wind farm inside the core breeding area could markedly increase mortality (up to 9.7 yearly collisions). If repowering of the wind farms was carried out using low-reaching modern turbines (RHR 36–150 m), mortality would more than double, whereas it would stay approximately constant if higher turbines (RHR 86–200 m) were used. Our study demonstrates the great potential of high-resolution GPS tracking for collision risk assessments. The resulting information on collision-related flight behaviour allows for performing detailed scenario analyses on wind farm siting and turbine design, in contrast to current environmental assessment practices. With regard to Montagu's Harriers, we conclude that although the deployment of higher wind turbines represents an opportunity to reduce collision risk for this species, precluding wind energy developments in core breeding areas remains the most important mitigation measure. However, there is a need for long-term data (over centennial or millennial timescales) to better understand natural community dynamics and the processes that govern the observed trends. Chironomids (Insecta: Diptera: Chironomidae) are often the most abundant insects in lake ecosystems, sensitive to environmental change, and, because their larval exoskeleton head capsules preserve well in lake sediments, they provide a unique record of insect community dynamics through time. Here, we provide the results of a metadata analysis of chironomid diversity across a range of spatial and temporal scales. First, we analyse spatial trends in chironomid diversity using Northern Hemispheric data sets overall consisting of 837 lakes. Our results indicate that in most of our data sets, summer temperature (T-jul) is strongly associated with spatial trends in modern-day chironomid diversity. We observe a strong increase in chironomid alpha diversity with increasing T-jul in regions with present-day T-jul between 2.5 and 14 degrees C. In some areas with T-jul > 14 degrees C, chironomid diversity stabilizes or declines. Second, we demonstrate that the direction and amplitude of change in alpha diversity in a compilation of subfossil chironomid records spanning the last glacial-interglacial transition (similar to 15,000-11,000 years ago) are similar to those observed in our modern data. A compilation of Holocene records shows that during phases when the amplitude of temperature change was small, site-specific factors had a greater influence on the chironomid fauna obscuring the chironomid diversity-temperature relationship. Our results imply expected overall chironomid diversity increases in colder regions such as the Arctic under sustained global warming, but with complex and not necessarily predictable responses for individual sites. It holds that a necessary condition of successful societies is that decision-makers base their decisions on a high level of attentiveness (concern and comprehension) towards subjectively valued and morally legitimate forms of life. Late modern societies consist of a plurality of forms of life, each providing grounds for what Alasdair MacIntyre has called internal goods—valued and morally valuable practices. The status of such goods is examined, and distinctions are drawn between their manifest and latent, and transposable and situationally specific, characteristics. We integrate this refined idea of internal goods into a developed conception of habitus that is both morally informed and situationally embedded. The sociological approach of strong structuration theory (SST) is employed to demonstrate how this conception of habitus can guide the critique of decision-making that damages internal goods. We identify the most pervasive and invidious forms of damaging decision-making in contemporary societies as those involving excessive forms of instrumental reasoning. We argue that our developed conception of habitus, anchored in the collectively valued practices of specific worlds, can be a powerful focus for resistance. Accounts of scholarship in higher education and of the white working class in America illustrate the specificities of singular, particular, social worlds and illuminate critical challenges raised by the perspective we advocate. How nocturnal animals move in an urban landscape, particularly in response to ALAN is largely unknown. Objectives We studied the movement responses, foraging and commuting, of common noctules (Nyctalus noctula) to urban landscape features in general and ALAN in particular. Methods We equipped 20 bats with miniaturized GPS loggers in the Berlin metropolitan area and related spatial positions of bats to anthropogenic and natural landscape features and levels of ALAN. Results Common noctules foraged close to ALAN only next to bodies of water or well vegetated areas, probably to exploit swarms of insects lured by street lights. In contrast, they avoided illuminated roads, irrespective of vegetation cover nearby. Predictive maps identified most of the metropolitan area as non-favoured by this species because of high levels of impervious surfaces and ALAN. Dark corridors were used by common noctules for commuting and thus likely improved the permeability of the city landscape. Conclusions We conclude that the spatial use of common noctules, previously considered to be more tolerant to light than other bats, is largely constrained by ALAN. Our study is the first individual-based GPS tracking study to show sensitive responses of nocturnal wildlife to light pollution. Approaches to protect urban biodiversity need to include ALAN to safeguard the larger network of dark habitats for bats and other nocturnal species in cities. The affective–reflective theory of physical inactivity and exercise (ART) proposes a theoretical framework for explaining how the automatic affective process (type‑1 process) will influence exercise behavior, i.e., through the automatic activation of exercise-related associations and a subsequent affective valuation of exercise. This study aimed to empirically test this assumption of the ART with data from 69 study participants. A single-measurement study, including within-subject experimental variation, was conducted. Automatic associations with exercise were first measured with a single-target implicit association test. The somato-affective core of the participants’ automatic valuation of exercise-related pictures was then assessed via heart rate variability (HRV) analysis, and the affective valence of the valuation was tested with a facial expression (FE; smile and frown) task. Exercise behavior was assessed via self-report. Multiple regression (path) analysis revealed that automatic associations predicted HRV reactivity (β = −0.24, p = .044); the signs of the correlation between automatic associations and the smile FE score was in the expected direction but remained nonsignificant (β = −0.21, p = .078). HRV reactivity predicted self-reported exercise behavior (β = −0.28, p = .013) (the same pattern of results was achieved for the frown FE score). The HRV-related results illustrate the potential role of automatic negative affective reactions to the thought of exercise as a restraining force in exercise motivation. For better empirical distinction between the two ART type‑1 process components, automatic associations and the affective valuation should perhaps be measured separately in the future. The results support the notion that automatic and affective processes should be regarded as essential aspects of the motivation to exercise. We propose to remedy this failure by redefining the mean squared displacements such that they reflect unambiguously the statistical properties of any stochastic process. In particular we study the effect of the initial condition in the Ornstein–Uhlenbeck process and its fractional extension. For the fractional Ornstein–Uhlenbeck process representing typical experimental situations in crowded environments such as living biological cells, we show that the stationarity of the process delicately depends on the initial condition. In recent years, metabolic engineering of microbes has gained strong momentum as a strategy for the production of valuable chemicals. As common microbial feedstocks like glucose directly compete with human consumption, the one carbon (C1) compound formate was suggested as an alternative feedstock. Formate can be easily produced by various means including electrochemical reduction of CO2 and could serve as a feedstock for microbial production, hence presenting a novel entry point for CO2 to the biosphere and a storage option for excess electricity. Compared to the gaseous molecule CO2, formate is a highly soluble compound that can be easily handled and stored. It can serve as a carbon and energy source for natural formatotrophs, but these microbes are difficult to cultivate and engineer. In this work, I present the results of several projects that aim to establish efficient formatotrophic growth of E. coli – which cannot naturally grow on formate – via synthetic formate assimilation pathways. In the first study, I establish a workflow for growth-coupled metabolic engineering of E. coli. I demonstrate this approach by presenting an engineering scheme for the PFL-threonine cycle, a synthetic pathway for anaerobic formate assimilation in E. coli. The described methods are intended to create a standardized toolbox for engineers that aim to establish novel metabolic routes in E. coli and related organisms. The second chapter presents a study on the catalytic efficiency of C1-oxidizing enzymes in vivo. As formatotrophic growth requires generation of both energy and biomass from formate, the engineered E. coli strains need to be equipped with a highly efficient formate dehydrogenase, which provides reduction equivalents and ATP for formate assimilation. I engineered a strain that cannot generate reducing power and energy for cellular growth, when fed on acetate. Under this condition, the strain depends on the introduction of an enzymatic system for NADH regeneration, which could further produce ATP via oxidative phosphorylation. I show that the strain presents a valuable testing platform for C1-oxidizing enzymes by testing different NAD-dependent formate and methanol dehydrogenases in the energy auxotroph strain. Using this platform, several candidate enzymes with high in vivo activity, were identified and characterized as potential energy-generating systems for synthetic formatotrophic or methylotrophic growth in E. coli.   In the third chapter, I present the establishment of the serine threonine cycle (STC) – a synthetic formate assimilation pathway – in E. coli. In this pathway, formate is assimilated via formate tetrahydrofolate ligase (FtfL) from Methylobacterium extorquens (M. extorquens). The carbon from formate is attached to glycine to produce serine, which is converted into pyruvate entering central metabolism. Via the natural threonine synthesis and cleavage route, glycine is regenerated and acetyl-CoA is produced as the pathway product. I engineered several selection strains that depend on different STC modules for growth and determined key enzymes that enable high flux through threonine synthesis and cleavage. I could show that expression of an auxiliary formate dehydrogenase was required to achieve growth via threonine synthesis and cleavage on pyruvate. By overexpressing most of the pathway enzymes from the genome, and applying adaptive laboratory evolution, growth on glycine and formate was achieved, indicating the activity of the complete cycle. The fourth chapter shows the establishment of the reductive glycine pathway (rGP) – a short, linear formate assimilation route – in E. coli. As in the STC, formate is assimilated via M. extorquens FtfL. The C1 from formate is condensed with CO2 via the reverse reaction of the glycine cleavage system to produce glycine. Another carbon from formate is attached to glycine to form serine, which is assimilated into central metabolism via pyruvate. The engineered E. coli strain, expressing most of the pathway genes from the genome, can grow via the rGP with formate or methanol as a sole carbon and energy source. N2 - Um den steigenden Bedarf einer wachsenden Weltbevölkerung zu decken und gleichzeitig die CO2-Emissionen zu reduzieren, ist es notwendig, CO2 aufzufangen und zu recyceln. Durch gentechnische Veränderungen von Mikroorganismen ist es möglich diese zur Produktion wertvoller organischer Verbindungen zu nutzen. Da mikrobielle Kulturen primär mit Glucose gefüttert werden und somit mit menschlicher Nahrungsversorgung konkurrieren, wurde die C1-Verbindung Formiat als alternativer bakterieller Nährstoff vorgeschlagen.. Formiat kann durch unterschiedliche Verfahren hergestellt werden, unter anderem durch elektrochemische Reduktion von CO2. Dieses Verfahren ermöglicht das Recycling von CO2 und weiterhin eine Speichermöglichkeit für überschüssige Elektrizität. Formiat ist im Vergleich zum gasförmigen CO2 gut wasserlöslich, was den Transport und die Verwendung als mikrobiellen Nährstoff erleichtert. Natürlich vorkommende formatotrophe Mikroorganismen nutzen Formiat als Kohlenstoff- und Energiequelle. Diese lassen sich allerdings meist schwierig kultivieren und genetisch verändern. In dieser Arbeit stelle ich die Ergebnisse verschiedener Projekte vor, die gemeinsam darauf abzielen, effizientes formatotrophes Wachstum von E. coli – welches natürlicherweise nicht auf Formiat wachsen kann – mittels synthetischer Formiat-Assimilierungswege zu ermöglichen. In der ersten Studie stelle ich eine Strategie für wachstumsgekoppeltes Stoffwechsel-Engineering in E. coli vor. Ich erläutere diese Strategie anhand eines Beispiels, der schrittweisen Etablierung eines synthetischen Formiat- Assimilierungswegs, des PFL-Threonin-Zyklus. Die in diesem Kapitel beschriebenen Methoden sollen einen Leitfaden für Ingenieure bereitstellen, die neue Stoffwechselwege in E. coli und verwandten Organismen etablieren wollen. Im zweiten Kapitel stelle ich eine Studie über die katalytische Effizienz von C1-oxidierenden Enzymen vor. Da formatotrophes Wachstum sowohl die Erzeugung von Energie als auch von Biomasse aus Formiat erfordert, müssen synthetisch formatotrophe E. coli Stämme mit einer hocheffizienten Formiat-Dehydrogenase ausgestattet werden, welche Reduktionsäquivalente und ATP für die Assimilierung von Formiat liefert. Um die Effizienz verschiedener Enzyme testen und vergleichen zu können, entwickelte ich einen E. coli Stamm, der aus Acetat weder Reduktionsäquivalente noch Energie für das Zellwachstum erzeugen kann. Dieser „energie-auxotrophe“ Stamm benötigt eines zusätzlichen enzymatischen Systems zur NADH-Regenerierung, um auf Acetat wachsen zu können. Ich testete verschiedene NAD-abhängige Formiat- und Methanol-Dehydrogenasen in diesem Stamm und konnte zeigen, dass Wachstum auf Acetat durch Zugabe von Formiat oder Methanol ermöglicht wurde. Dies zeigt, dass der Stamm eine zuverlässige Testplattform für C1-oxidierende Enzyme darstellt. Unter Verwendung dieser Plattform wurden mehrere Kandidatenenzyme mit hoher in vivo-Aktivität identifiziert und als Kandidatenenzyme für synthetisches formatotrophes oder methylotrophes Wachstum in E. coli charakterisiert.   Im dritten Kapitel stelle ich die Etablierung des Serin-Threonin-Zyklus (STZ) – eines synthetischen Formiat-Assimilationswegs – in E. coli vor. In diesem Stoffwechselweg wird Formiat über die Formiat-Tetrahydrofolat-Ligase (FtfL) aus Methylobacterium extorquens (M. extorquens) assimiliert. Der Kohlenstoff aus Formiat wird an Glycin gebunden, um Serin zu produzieren, welches im nächsten Schritt in Pyruvat umgewandelt wird und so in den zentralen Kohlenstoffmetabolismus gelangt. Über den natürlichen Threonin-Synthese- und Spaltweg wird Glycin regeneriert und Acetyl-CoA als Produkt des Stoffwechselwegs generiert. Ich entwickelte mehrere E. coli Selektionsstämme, deren Wachstum von verschiedenen STZ-Modulen abhängt, und konnte Schlüsselenzyme bestimmen, die einen hohen Reaktionsfluss durch die Threonin-Synthese und -Spaltung ermöglichen. Ich konnte zeigen, dass die Expression einer Formiat-Dehydrogenase erforderlich ist, um Wachstum auf Pyruvat über Threonin zu erreichen. Durch Integration und Überexpression der meisten Enzyme des STZ auf Genomebene und Anwendung adaptiver Laborevolution wurde Wachstum auf Glycin und Formiat erreicht, was bedeutet, dass der gesamte Serin-Threonin-Zyklus in E. coli aktiv ist. Das vierte Kapitel zeigt die Etablierung des reduktiven Glycinwegs (rGW) – eines kurzen, linearen Formiat-Assimilierungswegs – in E. coli. Wie im STZ wird Formiat über M. extorquens FtfL assimiliert. Dabei wird das Kohlenstoffatom aus Formiat mit CO2 über die umgekehrte Reaktion des Glycin-Spaltungssystems zu Glycin kondensiert. Das Nachhaltigkeitsbewusstsein wurde dabei basierend auf dem von Balderjahn et al. (2013) entwickelten Consciousness-for-sustainable-consumption-Modell durch die ökologische, soziale und die ökonomische Nachhaltigkeit sowie ergänzend durch das Tierschutzbewusstsein und Bewusstsein für lokale Produktion repräsentiert. Als Moderatoren dienten das Streben nach sozialer Anerkennung und Prestige, Materialismus, Hedonismus und Traditionsbewusstsein. Für die Aufdeckung möglicher Zusammenhänge zwischen den verschiedenen Dimensionen der Nachhaltigkeit und Luxuskonsum wurde eine Prädiktorenanalyse durchgeführt. Moderatorenanalysen offenbarten zusätzlich, ob ein Einfluss der verschiedenen Moderatoren auf die einzelnen Zusammenhänge vorlag. Die Untersuchung zeigte, dass jeweils das Umweltbewusstsein, das Bewusstsein für genügsamen Konsum sowie das Bewusstsein für schuldenfreien Konsum als Teil der ökonomischen Nachhaltigkeit und das Tierschutzbewusstsein einen Einfluss auf den Luxuskonsum ausüben. Darüber hinaus konnten insgesamt sieben Einflüsse durch die verschiedenen Moderatorvariablen auf die unterschiedlichen Zusammenhänge zwischen den Nachhaltigkeitsdimensionen und Luxuskonsum aufgedeckt werden. Increasing carbon fixation rate in plants is a promising approach to boost crop yields. The carbon-fixing enzyme Rubisco catalyzes, beside the carboxylation reaction, also an oxygenation reaction that generates glycolate-2P, which needs to be recycled via a metabolic route termed photorespiration. Photorespiration dissipates energy and most importantly releases previously fixed CO2, thus significantly lowering carbon fixation rate and yield. Engineering plants to omit photorespiratory CO2 release is the goal of the FutureAgriculture consortium and this thesis is part of this collaboration. The consortium aims to establish alternative glycolate-2P recycling routes that do not release CO2. Ultimately, they are expected to increase carbon fixation rates and crop yields. Natural and novel reactions, which require enzyme engineering, were considered in the pathway design process. Here I describe the engineering of two pathways, the arabinose-5P and the erythrulose shunt. They were designed to recycle glycolate-2P via glycolaldehyde into a sugar phosphate and thereby reassimilate glycolate-2P to the Calvin cycle. I used Escherichia coli gene deletion strains to validate and characterize the activity of both synthetic shunts. The strains’ auxotrophies can be alleviated by the activity of the synthetic route, thus providing a direct way to select for pathway activity. I introduced all pathway components to these dedicated selection strains and discovered inhibitions, limitations and metabolic cross talk interfering with pathway activity. After resolving these issues, I was able to show the in vivo activity of all pathway components and combine them into functional modules.. Specifically, I demonstrate the activity of a new-to-nature module of glycolate reduction to glycolaldehyde. Also, I successfully show a new glycolaldehyde assimilation route via arabinose-5P to ribulose-5P. In addition, all necessary enzymes for glycolaldehyde assimilation via L-erythrulose were shown to be active and an L-threitol assimilation route via L-erythrulose was established in E. coli. On their own, these findings demonstrate the power of using an easily engineerable microbe to test novel pathways; combined, they will form the basis for implementing photorespiration bypasses in plants. Therefore, different approaches are used to verify the flat platelet morphology of the AuNTs by oriented overgrowth with metal nanoparticles. The most important part for the morphological transformation of the AuNTs is the coating layer, containing surfactants or polymers. By using well established AuNTs stabilized by a dioctyl sodium sulfosuccinate (AOT) bilayer, different strategies of surface modification with noble metal nanoparticles are possible. On the one hand undulated superstructures were synthesized by in situ growth of hemispherical gold nanoparticles in the polyethyleneimine (PEI)-coated AOT bilayer of the AuNTs. On the other hand spiked AuNTs were obtained by a direct reduction of Au³⁺ ions in the AOT double layer in presence of silver ions and ascorbic acid as reducing agent. Additionally, crumble topping of the smooth AuNTs can be realized after an exchange of the AOT bilayer by hyaluronic acid, followed by a silver-ion mediated reduction with ascorbic acid. Furthermore, a decoration with silver nanoparticles after coating the AOT bilayer with the cationic surfactant benzylhexadecyldimethylammonium chloride (BDAC) can be realized. In that case the ultraviolet (UV)-absorption of the undulated Au@Ag nanoplatelets can be tuned depending on the degree of decoration with silver nanoparticles. Comparing the Raman scattering data for the plasmon driven dimerization of 4-nitrothiophenol (4-NTP) to 4,4′-dimercaptoazobenzene (DMAB) one can conclude that the most important effect of surface modification with a 75 times higher enhancement factor in SERS experiments becomes available by decoration with gold spikes. Therefore, we investigated the effects of two wind intensity levels by manipulating surrounding vegetation height in a grassland PSF field experiment. We grew four common plant species (two grasses and two non-leguminous forbs) with soil biota either previously conditioned by these or other species and tested the effect of wind on root:shoot ratio, fine root morphological traits as well as the outcome for PSFs. Wind intensity did not affect biomass allocation (i.e. root:shoot ratio) in any species. However, fine-root morphology of all species changed under high wind intensity. High wind intensity increased specific root length and surface area and decreased root tissue density, especially in the two grasses. Similarly, the direction of PSFs changed under high wind intensity in all four species, but differences in biomass production on the different soils between high and low wind intensity were marginal and most pronounced when comparing grasses with forbs. Because soils did not differ in plant-available nor total nutrient content, the results suggest that wind-induced changes in root morphology have the potential to influence plant–soil interactions. Linking wind-induced changes in fine-root morphology to effects on PSF improves our understanding of plant–soil interactions under changing environmental conditions. Monitoring and tracking of permafrost disturbances is important as they impact surrounding landscapes, ecosystems and infrastructure. Remote sensing provides the means to detect, map, and quantify these changes homogeneously across large regions and time scales. Existing Landsat-based algorithms assess different types of disturbances with similar spatiotemporal requirements. However, Landsat-based analyses are restricted in northern high latitudes due to the long repeat interval and frequent clouds, in particular at Arctic coastal sites. We therefore propose to combine Landsat and Sentinel-2 data for enhanced data coverage and present a combined annual mosaic workflow, expanding currently available algorithms, such as LandTrendr, to achieve more reliable time series analysis. We exemplary test the workflow for twelve sites across the northern high latitudes in Siberia. We assessed the number of images and cloud-free pixels, the spatial mosaic coverage and the mosaic quality with spectral comparisons. The number of available images increased steadily from 1999 to 2019 but especially from 2016 onward with the addition of Sentinel-2 images. Consequently, we have an increased number of cloud-free pixels even under challenging environmental conditions, which then serve as the input to the mosaicking process. In a comparison of annual mosaics, the Landsat+Sentinel-2 mosaics always fully covered the study areas (99.9–100 %), while Landsat-only mosaics contained data-gaps in the same years, only reaching coverage percentages of 27.2 %, 58.1 %, and 69.7 % for Sobo Sise, East Taymyr, and Kurungnakh in 2017, respectively. The spectral comparison of Landsat image, Sentinel-2 image, and Landsat+Sentinel-2 mosaic showed high correlation between the input images and mosaic bands (e.g., for Kurungnakh 0.91–0.97 between Landsat and Landsat+Sentinel-2 mosaic and 0.92–0.98 between Sentinel-2 and Landsat+Sentinel-2 mosaic) across all twelve study sites, testifying good quality mosaic results. Our results show that especially the results for northern, coastal areas was substantially improved with the Landsat+Sentinel-2 mosaics. By combining Landsat and Sentinel-2 data we accomplished to create reliably high spatial resolution input mosaics for time series analyses. Here, we aim to redesign the natural serine cycle for implementation in E. coli. We propose the homoserine cycle, relying on two promiscuous formaldehyde aldolase reactions, as a superior pathway design. The homoserine cycle is expected to outperform the serine cycle and its variants with respect to biomass yield, thermodynamic favorability, and integration with host endogenous metabolism. Even as compared to the RuMP cycle, the most efficient naturally occurring methanol assimilation route, the homoserine cycle is expected to support higher yields of a wide array of products. We test the in vivo feasibility of the homoserine cycle by constructing several E. coli gene deletion strains whose growth is coupled to the activity of different pathway segments. Using this approach, we demonstrate that all required promiscuous enzymes are active enough to enable growth of the auxotrophic strains. Our findings thus identify a novel metabolic solution that opens the way to an optimized methylotrophic platform. In contrast, the spontaneous condensation of formaldehyde with tetrahydrofolate (THF) has been proposed to contribute to the assimilation of this intermediate during growth on C1 carbon sources such as methanol. However, the in vivo rate of this condensation reaction is unknown and its possible contribution to growth remains elusive. Here, we used microbial platforms to assess the rate of this condensation in the cellular environment. We constructed Escherichia coli strains lacking the enzymes that naturally produce 5,10-methylene-THF. These strains were able to grow on minimal medium only when equipped with a sarcosine (N-methyl-glycine) oxidation pathway that sustained a high cellular concentration of formaldehyde, which spontaneously reacts with THF to produce 5,10-methylene-THF. We used flux balance analysis to derive the rate of the spontaneous condensation from the observed growth rate. According to this, we calculated that a microorganism obtaining its entire biomass via the spontaneous condensation of formaldehyde with THF would have a doubling time of more than three weeks. Hence, this spontaneous reaction is unlikely to serve as an effective route for formaldehyde assimilation. In this work, the wet Arabica coffee processing in batch and continuous modus were investigated. Coffee beans samples as well as by-products and wastewaters collected at different production steps were analyzed in terms of their content in total phenols, antioxidant capacity, caffeine content, organic acids, reducing sugars, free amino group and protein content. The results showed that 40% of caffeine was removed with pulp. Green coffee beans showed highest concentration of organic acids and sucrose (4.96 ± 0.25 and 5.07 ± 0.39 g/100 g DW for the batch and continuous processing). Batch green coffee beans contained higher amount of phenols. 5-caffeoylquinic Acid (5-CQA) was the main constituent (67.1 and 66.0% for the batch and continuous processing, respectively). Protein content was 15 and 13% in the green coffee bean in batch and continuous processing, respectively. A decrease of 50 to 64% for free amino groups during processing was observed resulting in final amounts of 0.8 to 1.4% in the processed beans. Finally, the batch processing still revealed by-products and wastewater with high nutrient content encouraging a better concept for valorization. The most prevalent causal account for developmental dyslexia is the phonological deficit hypothesis. As rhythm is a subpart of phonology, we hypothesized that reading deficits in dyslexia are associated with rhythm processing in speech and in music. In a rhythmic grouping task, adults with diagnosed dyslexia and age-matched controls listened to speech streams with syllables alternating in intensity, duration, or neither, and indicated whether they perceived a strong-weak or weak-strong rhythm pattern. Additionally, their reading and musical rhythm abilities were measured. Results showed that adults with dyslexia had lower musical rhythm abilities than adults without dyslexia. Moreover, lower musical rhythm ability was associated with lower reading ability in dyslexia. However, speech grouping by adults with dyslexia was not impaired when musical rhythm perception ability was controlled: like adults without dyslexia, they showed consistent preferences. However, rhythmic grouping was predicted by musical rhythm perception ability, irrespective of dyslexia. The results suggest associations among musical rhythm perception ability, speech rhythm perception, and reading ability. This highlights the importance of considering individual variability to better understand dyslexia and raises the possibility that musical rhythm perception ability is a key to phonological and reading acquisition. Out of a multitude of variables gathered at CR admission and discharge, we aimed to identify predictors of returning to work (RTW) and HRQL 6 months after CR. Design Prospective observational multi-centre study, enrolment in CR between 05/2017 and 05/2018. Method Besides general data (e.g. age, sex, diagnoses), parameters of risk factor management (e.g. smoking, hypertension), physical performance (e.g. maximum exercise capacity, endurance training load, 6-min walking distance) and patient-reported outcome measures (e.g. depression, anxiety, HRQL, subjective well-being, somatic and mental health, pain, lifestyle change motivation, general self-efficacy, pension desire and self-assessment of the occupational prognosis using several questionnaires) were documented at CR admission and discharge. These variables (at both measurement times and as changes during CR) were analysed using multiple linear regression models regarding their predictive value for RTW status and HRQL (SF-12) six months after CR. Results Out of 1262 patients (54±7 years, 77% men), 864 patients (69%) returned to work. Predictors of failed RTW were primarily the desire to receive pension (OR = 0.33, 95% CI: 0.22–0.50) and negative self-assessed occupational prognosis (OR = 0.34, 95% CI: 0.24–0.48) at CR discharge, acute coronary syndrome (OR = 0.64, 95% CI: 0.47–0.88) and comorbid heart failure (OR = 0.51, 95% CI: 0.30–0.87). High educational level, stress at work and physical and mental HRQL were associated with successful RTW. HRQL was determined predominantly by patient-reported outcome measures (e.g. pension desire, self-assessed health prognosis, anxiety, physical/mental HRQL/health, stress, well-being and self-efficacy) rather than by clinical parameters or physical performance. Conclusion Patient-reported outcome measures predominantly influenced return to work and HRQL in patients with heart disease. Therefore, the multi-component CR approach focussing on psychosocial support is crucial for subjective health prognosis and occupational resumption. The author, Joseph de Maimieux (1753–1820), is said to have coined the term pasigraphie. With the invention of pasigraphy, Maimieux pursued the goal of creating a conceptually based writing system that could be applied to all languages. In this way, a text written in this script could be read in all languages. The pasigraphic system is based on an a priori system of conceptual classification to which written characters are assigned. It is therefore initially an exclusively written language, on which Maimieux two years later also built a spoken language with the Pasilalie. The first arguments against the endeavour represented by the Pasigraphie were brought forward by Maimieux' immediate contemporaries, the ideologues. These objections were re-affirmed in particular by Johann Severin Vater. The decisive difference between Maimieux and his critics lies in the determination of the role of signs in the formation of concepts. While for Maimieux the signs only name the finished concepts, his critics argued that signs have a function in the constitution of the concepts. A universal language is therefore ultimately not possible, since there are no universal concepts and the individual languages lead to different conceptual structures. Im aktuellen Kontext des Klimawandels zeigt sich jedoch, wie stark sich Stoffflüsse auch kurzfristig ändern und dabei das Gesicht der Landschaft verwandeln können. Die Gobi-Wüste in der Inneren Mongolei (China), als Teil der gleichnamigen Trockenregionen Nordwestchinas, ist aufgrund der Ausgestaltung ihrer landschaftsprägenden Elemente sowie ihrer Landschaftsdynamik, im Zusammenhang mit der Lage zum Tibet-Plateau, in den Fokus der klimageschichtlichen Grundlagenforschung gerückt. Als großes Langzeitarchiv unterschiedlichster fluvialer, lakustriner und äolischer Sedimente stellt sie eine bedeutende Lokalität zur Rekonstruktion von lokalen und regionalen Stoffflüssen dar.. Andererseits ist die Gobi-Wüste zugleich auch eine bedeutende Quelle für den überregionalen Staubtransport, da sie aufgrund der klimatischen Bedingungen insbesondere der Erosion durch Ausblasung preisgegeben wird. Vor diesem Hintergrund erfolgten zwischen 2011 und 2014, im Rahmen des BMBF-Verbundprogramms WTZ Zentralasien – Monsundynamik & Geoökosysteme (Förderkennzeichen 03G0814), mehrere deutsch-chinesische Expeditionen in das Ejina-Becken (Innere Mongolei) und das Qilian Shan-Vorland. Im Zuge dieser Expeditionen wurden für eine Bestimmung potenzieller Sedimentquellen erstmals zahlreiche Oberflächenproben aus dem gesamten Einzugsgebiet des Heihe (schwarzer Fluss) gesammelt. Zudem wurden mit zwei Bohrungen im inneren des Ejina-Beckens, ergänzende Sedimentbohrkerne zum bestehenden Bohrkern D100 (siehe Wünnemann (2005)) abgeteuft, um weit reichende, ergänzende Informationen zur Landschaftsgeschichte und zum überregionalen Sedimenttransfer zu erhalten. Gegenstand und Ziel der vorliegenden Doktorarbeit ist die sedimentologisch-mineralogische Charakterisierung des Untersuchungsgebietes in Bezug auf potenzielle Sedimentquellen und Stoffflüsse des Ejina-Beckens sowie die Rekonstruktion der Ablagerungsgeschichte eines dort erbohrten, 19m langen Sedimentbohrkerns (GN100). Schwerpunkt ist hierbei die Klärung der Sedimentherkunft innerhalb des Bohrkerns sowie die Ausweisung von Herkunftssignalen und möglichen Sedimentquellen bzw. Sedimenttransportpfaden. Die methodische Herangehensweise basiert auf einem Multi-Proxy-Ansatz zur Charakterisierung der klastischen Sedimentfazies anhand von Geländebeobachtungen, lithologisch-granulometrischen und mineralogisch-geochemischen Analysen sowie statistischen Verfahren. Für die mineralogischen Untersuchungen der Sedimente wurde eine neue, rasterelektronenmikroskopische Methode zur automatisierten Partikelanalyse genutzt und den traditionellen Methoden gegenübergestellt. Die synoptische Betrachtung der granulometrischen, geochemischen und mineralogischen Befunde der Oberflächensedimente ergibt für das Untersuchungsgebiet ein logisches Kaskadenmodell mit immer wiederkehrenden Prozessbereichen und ähnlichen Prozesssignalen. Die umfangreichen granulometrischen Analysen deuten dabei auf abnehmende Korngrößen mit zunehmender Entfernung vom Qilian Shan hin und ermöglichen die Identifizierung von vier texturellen Signalen: den fluvialen Sanden, den Dünensanden, den Stillwassersedimenten und Stäuben. Diese Ergebnisse können als Interpretationsgrundlage für die Korngrößenanalysen des Bohrkerns genutzt werden. Somit ist es möglich, die Ablagerungsgeschichte der Bohrkernsedimente zu rekonstruieren und in Verbindung mit eigenen und literaturbasierten Datierungen in einen Gesamtkontext einzuhängen. Für das Untersuchungsgebiet werden somit vier Ablagerungsphasen ausgewiesen, die bis in die Zeit des letzten glazialen Maximums (LGM) zurückreichen. Während dieser Ablagerungsphasen kam es im Zuge unterschiedlicher Aktivitäts- und Stabilitätsphasen zu einer kontinuierlichen Progradation und Überprägung des Schwemmfächers. Eine besonders aktive Phase kann zwischen 8 ka und 4 ka BP festgestellt werden, während der es aufgrund zunehmender fluvialer Aktivitäten zu einer deutlich verstärkten Schwemmfächerdynamik gekommen zu sein scheint. In den Abschnitten davor und danach waren es vor allem äolische Prozesse, die zu einer Überprägung des Schwemmfächers geführt haben. Hinsichtlich der mineralogischen Herkunftssignale gibt es eine große Variabilität. Dies spiegelt die enorme Heterogenität der Geologie des Untersuchungsgebietes wider, wodurch die räumlichen Signale nicht sehr stark ausgeprägt sind. Dennoch, können für das Einzugsgebiet drei größere Bereiche deklariert werden, die als Herkunftsgebiet in Frage kommen. Das östliche Qilian Shan Vorland zeichnet sich dabei durch deutlich höhere Chloritgehalte als primäre Quelle für die Sedimente im Ejina-Becken aus. Sie unterscheiden sich insbesondere durch stark divergierende Chloritgehalte in der Tonmineral- und Gesamtmineralfraktion, was das östliche Qilian Shan Vorland als primäre Quelle für die Sedimente im Ejina-Becken auszeichnet. Dies steht in Zusammenhang mit den Grünschiefern, Ophioliten und Serpentiniten in diesem Bereich. Geochemisch deutet vor allem das Cr/Rb-Verhältnis eine große Variabilität innerhalb des Einzugsgebietes an. Auch hier ist es das östliche Vorland, welches aufgrund seines hohen Anteils an mafischen Gesteinen reich an Chromiten und Spinellen ist und sich somit vom restlichen Untersuchungsgebiet abhebt. Die zeitliche aber auch die generelle Variabilität der Sedimentherkunft lässt sich in den Bohrkernsedimenten nicht so deutlich nachzeichnen. Die mineralogisch-sedimentologischen Eigenschaften der erbohrten klastischen Sedimente zeugen zwar von zwischenzeitlichen Änderungen bei der Sedimentherkunft, diese sind jedoch nicht so deutlich ausgeprägt, wie es die Quellsignale in den Oberflächensedimenten vermuten lassen. Ein Grund dafür scheint die starke Vermischung unterschiedlichster Sedimente während des Transportes zu sein. Die Kombination der Korngrößenergebnisse mit den Befunden der Gesamt- und Schwermineralogie deuten darauf hin, dass es zwischenzeitlich eine Phase mit überwiegend äolischen Prozessen gegeben hat, die mit einem Sedimenteintrag aus dem westlichen Bei Shan in Verbindung stehen. Neben der Zunahme ultrastabiler Schwerminerale wie Zirkon und Granat und der Abnahme opaker Schwerminerale, weisen vor allem die heutigen Verhältnisse darauf hin. Der Vergleich der traditionellen Schwermineralanalyse mit der Computer-Controlled-Scanning-Electron-Microscopy (kurz: CCSEM), die eine automatisierte Partikelauswertung der Proben ermöglicht, zeigt den deutlichen Vorteil der modernen Analysemethode. Neben einem zeitlichen Vorteil, den man durch die automatisierte Abarbeitung der vorbereiteten Proben erlangen kann, steht vor allem die deutlich größere statistische Signifikanz des Ergebnisses im Vordergrund. Zudem können mit dieser Methode auch chemische Varietäten einiger Schwerminerale bestimmt werden, die eine noch feinere Klassifizierung und sicherere Aussagen zu einer möglichen Sedimentherkunft ermöglichen. Damit ergeben sich außerdem verbesserte Aussagen zu Zusammensetzungen und Entstehungsprozessen der abgelagerten Sedimente. Die Studie verdeutlicht, dass die Sedimentherkunft innerhalb des Untersuchungsgebietes sowie die ablaufenden Prozesse zum Teil stark von lokalen Gegebenheiten abhängen. Die Heterogenität der Geologie und die Größe des Einzugsgebietes sowie die daraus resultierende Komplexität der Sedimentgenese, machen exakte Zuordnungen zu klar definierten Sedimentquellen sehr schwer. Dennoch zeigen die Ergebnisse, dass die Sedimentzufuhr in das Ejina-Becken in erster Linie durch fluviale klastische Sedimente des Heihe aus dem Qilian Shan erfolgt sein muss. Die Untersuchungsergebnisse zeigen jedoch ebenso die Notwendigkeit einer ergänzenden Bearbeitung angrenzender Untersuchungsgebiete, wie beispielsweise den Gobi-Altai im Norden oder den Beishan im Westen, sowie die Verdichtung der Oberflächenbeprobung zur feineren Auflösung von lokalen Sedimentquellen. N2 - The Central Asian natural space, as it presents itself today, is the result of a combination of many different factors over millions of years. However, in the current context of climate change it becoming obvious how strongly material fluxes can change in the short-term and thereby transform the face of the landscape. As a large long-term archive of different fluvial, lacustrine and aeolian sediments, the Gobi Desert in Inner Mongolia (China) represents a significant locality for the reconstruction of local and regional material flows as well as an important source for supra-regional dust transport due to erosion by aeolian processes. This, in connection with its location to the Tibetan Plateau, has moved the Gobi Desert into the focus of climate-historical basic research. Against this background, several German-Chinese expeditions to Ejina Basin (Inner Mongolia) and the Qilian Shan foreland were accomplished between 2011 and 2014. They were part of the BMBF joint program WTZ Central Asia - Monsoon Dynamics & Geo-Ecosystems (Grant No. 03G0814). In the course of these expeditions numerous surface samples from the entire catchment area of the Heihe (Black River) were collected for the determination of potential sediment sources. Additionally, two drillings in the interior of the Ejina Basin were conducted as a continuation of the existing drill-core D100 (see Wünnemann (2005)) and for gathering complementary information on landscape history and supra-regional sediment transfer. The goals of this doctoral thesis are a sedimentological-mineralogical characterization of the study area with regard to potential sediment sources and material fluxes of the Ejina Basin as well as the reconstruction of the sedimentation history of a 19 m sediment drill-core (GN100) drilled in the northern part of the basin. The main focus is on the clarification of the sediment provenance within the drill core as well as the designation of provenance signals and possible sediment sources or sediment transport paths. The methodological approach is based on a multi-proxy approach to characterize the clastic sedimentary facies using field observations, lithological-granulometric and mineralogical-geochemical analyses as well as statistical methods. For the mineralogical investigations of the sediments, a new scanning electron microscopic method for automated particle analysis was used and compared with traditional methods. The synoptic analysis of the granulometric, geochemical and mineralogical findings of the surface sediments result in a logical cascade model with recurring process areas and similar process signals. The results of the extensive granulometric analyses indicate decreasing grain sizes as the distance from the Qilian Shan increases and allow the identification of four textural signals: fluvial sands, dune sands, still water sediments and dusts. They can be used as the basis of interpretation for the grain size analyses of the drill core. This makes it possible to reconstruct the depositional history of the core sediments and to put them in an overall context in conjunction with own and literature-based datings. For the study area four depositional phases can be identified which date back until the time of the Last Glacial Maximum (LGM). During these depositional phases, various phases of activity and stability led to a continuous progradation and overprinting of the alluvial fan. A particularly active phase can be observed between 8 ka and 4 ka BP. During this period there seems to have been a significant increase in alluvial fan dynamics due to increasing fluvial activity. In the periods before and after mainly aeolian processes have led to an overprinting of the alluvial fan. Regarding the mineralogical provenance signals there is a great variability. This reflects the enormous geological heterogeneity of the study area, by which the spatial signals are not very pronounced. Nonetheless, within the catchment three larger areas can be declared which can be considered as possible sediment sources. These are the eastern and western foreland of the Qilian Shan mountains as wells as the Bei Shan in the west of the Ejina basin. They differ, in particular, from strongly diverging chlorite contents in the clay mineral and total mineral fraction. The eastern Qilian Shan foreland is characterized by significantly higher chlorite contents as the primary source for the sediments in the Ejina Basin. This is related to the greenschists, ophiolites and serpentinites in this area. Geochemically, the Cr/Rb-ratio in particular indicates a large variability within the catchment area. In this case its again the eastern Qilian Shan foreland which stands out from the rest of the study area with high values due to its high proportion of mafic rocks which are rich in chromites and spinels. Within the drill core sediments of GN100 the temporal but also the general variability of the sediment provenance according to the surface sediment results cannot be clearly traced. The mineralogical-sedimentological characteristics of the drilled clastic sediments exhibit meantime changes in sediment provenance. However, they are not as pronounced as the source signals within the surface sediments suggest. One reason seems to be the strong mixing of different sediments during transport. The combination of the particle size results with the results of the total and heavy mineralogy indicate that there must have been a phase in the meantime with predominantly aeolian processes.. These processes were connected with a sediment input from the Bei Shan mountains in the west. In addition to the increase in ultrastable heavy minerals such as zirconium and garnet and the decline of opaque heavy minerals, mainly present conditions point to this. The comparison of the traditional heavy mineral analysis (polarized light microscopy) with computer-controlled scanning electron microscopy (short: CCSEM) shows the clear advantage of the modern analysis method. Above all is the significant greater statistical significance of the counting results. In addition, this method can also be used to determine chemical varieties of some heavy minerals, which allows an even finer classification and more reliable conclusions about a possible sediment provenance. Furthermore, better statements can be made about the composition and the development process of the deposited sediments. This thesis shows that the sediment provenance within the study area as well as the ongoing processes partly depend strongly on local conditions. Therefore, the heterogeneity of the geology and the size of the catchment as well as the resulting complexity of the sediment genesis makes it very difficult to make exact assignments to clearly defined sediment sources. Nevertheless, the results indicate a primary input of fluvial clastic sediments of the Heihe coming from the Qilian Shan mountains. However, the findings also indicate the need to complement the study with adjacent study areas such as the Gobi Altai in the north or the Beishan in the west as well as a densification of the surface sampling for a better resolution of possible local sediment sources. The test was assessed in patients receiving phenprocoumon (NCT04074980). Methods: Venous plasma international normalized ratio (INR) was measured using the LumiraDx INR Test. LumiraDx INR Test-ascertained capillary whole blood INR was compared with venous plasma INR measured using the IL ACL Elite Pro and Sysmex CS-5100 reference instruments. Results: A total of 102 patients receiving phenprocoumon were recruited. The INR results from venous plasma and capillary whole blood that were analyzed on the LumiraDx INR Test correlated well with those measured using the IL ACL Elite Pro (plasma: n = 25, r = 0.981; capillary blood: n = 74, r = 0.949) and the Sysmex CS-5100 (n = 73, r = 0.950). Conclusions: The LumiraDx INR Test showed high accuracy in analyzing venous plasma and capillary whole blood from patients receiving phenprocoumon. The magnetite nanoparticles dispersed in olive oil are processed into the gel and remain in the macroporous aerogel after removing the oil components. The coexistence of macropores from the Janus droplets and mesopores from freeze-drying of the hydrogels in combination with the magnetic properties offer a special hierarchical pore structure, which is of relevance for smart supercapacitors, biosensors, and spilled oil sorption and separation. The morphology of the final structure was investigated in dependence on initial compositions. More hydrophobic aerogels with magnetic responsiveness were synthesized by bisacrylamide-crosslinking of the hydrogel. The crosslinked aerogels can be successfully used in magnetically responsive clean up experiments of the cationic dye methylene blue. In recent years, there are a variety of endeavors to incorporate platform magnetometers, which are initially designed for navigation purposes, to study ionospheric currents. Because of the suboptimal resolution and significant noise of the platform magnetometers, however, most of the studies were confined to high-latitude auroral regions, where magnetic field deflections from ionospheric currents easily exceed 100 nT. This study aims to demonstrate the possibility of diagnosing weak low-/mid-latitude ionospheric currents based on platform magnetometers. We use navigation magnetometer data from two satellites, CryoSat-2 and the Gravity Recovery and Climate Experiment Follow-On (GRACE-FO), both of which have been intensively calibrated based on housekeeping data and a high-precision geomagnetic field model. Analyses based on 8 years of CryoSat-2 data as well as similar to 1.5 years of GRACE-FO data reproduce well-known climatology of inter-hemispheric field-aligned currents (IHFACs), as reported by previous satellite missions dedicated to precise magnetic observations. Also, our results show that C-shaped structures appearing in noontime IHFAC distributions conform to the shape of the South Atlantic Anomaly. The F-region dynamo currents are only partially identified in the platform magnetometer data, possibly because the currents are weaker than IHFACs in general and depend significantly on altitude and solar activity. Still, this study evidences noontime F-region dynamo currents at the highest altitude (717 km) ever reported. We expect that further data accumulation from continuously operating missions may reveal the dynamo currents more clearly during the next solar maximum. Wir richten dabei den Blick auf die vom Wissenschaftsrat attestierten Schwachstellen im Bereich empirisch-analytischer Methoden und erläutern ihre Auswirkungen auf Interdisziplinarität, Internationalität und Politikberatung der deutschen Friedens- und Konfliktforschung. Wir argumentieren, unter Verweis auf den Bericht des Wissenschaftsrats, dass eine breitere Methodenausbildung und -kenntnis von großer Bedeutung für interdisziplinäre und internationale Zusammenarbeit, aber auch für die Politikberatung ist. Zukünftige Initiativen innerhalb der Friedens- und Konfliktforschung sollten die Methodenvielfalt des Forschungsbereichs angemessen berücksichtigen und einen besonderen Fokus auf die Ausbildung im Bereich empirisch-analytischer Methoden legen, um das Forschungsfeld in diesem Bereich zu stärken. N2 - This article reflects on and adds to the ongoing discussion of the German Science Council’s recommendations for the further development of peace and conflict research. We focus on the gaps in empirical-analytical research methodology identified by the German Science Council and elaborate how they affect interdisciplinary cooperation, international visibility and policy-oriented research within the German peace and conflict research community. We follow the analysis of the Science Council’s report and argue that a diversified training in as well as knowledge of empirical research methodology is of central importance for interdisciplinary and international cooperation as well as comprehensive policy-oriented research. Future initiatives within the peace and conflict research community should strive to reflect the methodological diversity of our research community and put a special emphasis on training in empirical-analytical research methodology, in order to strengthen the methodological expertise in this realm. This paper examines different UN simulations in political science courses to demonstrate their pedagogical value and provide empirical evidence for their effectiveness regarding three levels of knowledge (factual, procedural and soft skills). Despite comprehensive theoretical claims about the positive effects of active learning environments on learning outcomes, substantial empirical evidence is limited. Here, we focus on simulations to systematically test previous claims and demonstrate their pedagogical value. Model United Nations (MUNs) have been a popular teaching device in political science. To gain comprehensive data about the active learning effects of MUNs, we collect data and evaluate three simulations covering the whole range of simulation characteristics: a short in-class simulation of the UN Security Council, a regional MUN with different committees being simulated, and two delegations to the National Model United Nations, for which the students prepare for 1 year. Comparative results prove that simulations need to address certain characteristics in order to produce extensive learning outcomes. Only comprehensive simulations are able to achieve all envisioned learning outcomes regarding factual and procedural knowledge about the UN and soft skills. Small microplastics enter the water either directly or accumulate through disintegration of larger plastic particles. These particles might then be ingested by filter-feeding zooplankton, such as rotifers. Particles released into the water may also interact with the biota through the formation of aggregates, which might alter the uptake by zooplankton. In this study, we tested for size-specific aggregation of polystyrene microspheres and their ingestion by a common freshwater rotifer Brachionus calyciflorus. The ingestion of three sizes of polystyrene microspheres (MS) 1-, 3-, and 6-mu m was investigated. Each MS size was tested in combination with three different treatments: MS as the sole food intake, MS in association with food algae and MS aggregated with biogenic matter. After 72 h incubation in pre-filtered natural river water, the majority of the 1-mu m spheres occurred as aggregates. The larger the particles, the higher the relative number of single particles and the larger the aggregates. All particles were ingested by the rotifer following a Type-II functional response. The presence of algae did not influence the ingestion of the MS for all three sizes. The biogenic aggregation of microspheres led to a significant size-dependent alteration in their ingestion. Rotifers ingested more microspheres (MS) when exposed to aggregated 1- and 3-mu m MS as compared to single spheres, whereas fewer aggregated 6-mu m spheres were ingested. This indicates that the small particles when aggregated were in an effective size range for Brachionus, while the aggregated larger spheres became too large to be efficiently ingested. These observations provide the first evidence of a size- and aggregation-dependent feeding interaction between microplastics and rotifers. Microplastics when aggregated with biogenic particles in a natural environment can rapidly change their size-dependent availability. The aggregation properties of microplastics should be taken into account when performing experiments mimicking the natural environment. These include the liver itself (nonalcoholic steatohepatitis or NASH; fibrosis and cirrhosis, and hepatocellular carcinoma) or other organs such as the vessels and the heart (cardiovascular disease) or the islets of Langerhans (type 2 diabetes). In addition to elevated caloric intake and a sedentary lifestyle, genetic and epigenetic predisposition contribute to the development of NAFLD and the secondary diseases. Scope of review: We present data from genome-wide association studies (GWAS) and functional studies in rodents which describe polymorphisms identified in genes relevant for the disease as well as changes caused by altered DNA methylation and gene regulation via specific miRNAs. The review also provides information on the current status of the use of genetic and epigenetic factors as risk markers. Major conclusion: With our overview we provide an insight into the genetic and epigenetic landscape of NAFLD and argue about the applicability of currently defined risk scores for risk stratification and conclude that further efforts are needed to make the scores more usable and meaningful.
