The energy of such naturally occurring single-domain magnetic nanoparticles can reach up to 10-20 room k(B)T in the magnetic field of the Earth, which naturally led to supposition that they can serve as sensory elements in various animals. The plethora of S1P-mediated effects is due to the fact that the sphingolipid not only modulates intracellular functions but also acts as a ligand of G protein-coupled receptors after secretion into the extracellular environment. In the plasma, S1P is found in high concentrations, modulating immune cell trafficking and vascular endothelial integrity. The liver is engaged in modulating the plasma S1P content, as it produces apolipoprotein M, which is a chaperone for the S1P transport. Moreover, the liver plays a substantial role in glucose and lipid homeostasis. A dysfunction of glucose and lipid metabolism is connected with the development of liver diseases such as hepatic insulin resistance, non-alcoholic fatty liver disease, or liver fibrosis. Recent studies indicate that S1P is involved in liver pathophysiology and contributes to the development of liver diseases. In this review, the current state of knowledge about S1P and its signaling in the liver is summarized with a specific focus on the dysregulation of S1P signaling in obesity-mediated liver diseases. We investigate the C and N pools in two thermokarst-affected yedoma landscapes - on Sobo-Sise Island and on Bykovsky Peninsula in the north of eastern Siberia. Soil cores up to 3m depth were collected along geomorphic gradients and analysed for organic C and N contents. A high vertical sampling density in the profiles allowed the calculation of C and N stocks for short soil column intervals and enhanced understanding of within-core parameter variability. Profile-level C and N stocks were scaled to the landscape level based on landform classifications from 5 m resolution, multispectral RapidEye satellite imagery. Mean landscape C and N storage in the first metre of soil for Sobo-Sise Island is estimated to be 20.2 kg C m(-2) and 1.8 kg N m(-2) and for Bykovsky Peninsula 25.9 kg C m(-2) and 2.2 kg N m(-2). Radiocarbon dating demonstrates the Holocene age of thermokarst basin deposits but also suggests the presence of thick Holoceneage cover layers which can reach up to 2 m on top of intact yedoma landforms. Reconstructed sedimentation rates of 0.10-0.57 mm yr(-1) suggest sustained mineral soil accumulation across all investigated landforms. Both yedoma and thermokarst landforms are characterized by limited accumulation of organic soil layers (peat). We further estimate that an active layer deepening of about 100 cm will increase organic C availability in a seasonally thawed state in the two study areas by similar to 5.8 Tg (13.2 kg C m(-2)). Our study demonstrates the importance of increasing the number of C and N storage inventories in ice-rich yedoma and thermokarst environments in order to account for high variability of permafrost and thermokarst environments in pan-permafrost soil C and N pool estimates. The first corpus analysis revealed that definiteness, givenness and type of referring expression influenced word order variation in child language and child-directed speech when the type of referring expression distinguished between pronouns and lexical noun phrases. These results correspond to previous child language studies in English (e.g., de Marneffe et al. 2012). Extending the scope of previous studies, our second corpus analysis examined the role of different pronoun types on word order. It revealed that word order in child language and child-directed speech was predictable from the types of pronouns used. Different types of pronouns were associated with different sentence positions but also showed a strong correlation to givenness and definiteness. Yet, the distinction between pronoun types diminished the effects of givenness so that givenness had an independent impact on word order only in child-directed speech but not in child language. Our results support a multi-factorial approach to word order in German. Moreover, they underline the strong impact of the type of referring expression on word order and suggest that it plays a crucial role in the acquisition of the factors influencing word order variation. In Brazil, a few modeling studies assess freshwater nutrients, limiting policies on hydrologic ecosystem services. This paper aims to compare freshwater quality scenarios under different land-use and land-cover (LULC) change, one of them related to ecosystem-based adaptation (EbA), in Brazilian headwaters. Using the spatially semi-distributed Soil and Water Assessment Tool (SWAT) model, nitrate, total phosphorous (TP) and sediment were modeled in catchments ranging from 7.2 to 1037 km(2). These head-waters were eligible areas of the Brazilian payment for ecosystem services (PES) projects in the Cantareira water supply system, which had supplied water to 9 million people in the Sao Paulo metropolitan region (SPMR). We considered SWAT modeling of three LULC scenarios: (i) recent past scenario (S1), with historical LULC in 1990; (ii) current land-use scenario (S2), with LULC for the period 2010-2015 with field validation; and (iii) future land-use scenario with PES (S2 + EbA). This latter scenario proposed forest cover restoration through EbA following the river basin plan by 2035. These three LULC scenarios were tested with a selected record of rainfall and evapotranspiration observed in 2006-2014, with the occurrence of extreme droughts. To assess hydrologic services, we proposed the hydrologic service index (HSI), as a new composite metric comparing water pollution levels (WPL) for reference catchments, related to the grey water footprint (greyWF) and water yield. On the one hand, water quality simulations allowed for the regionalization of greyWF at spatial scales under LULC scenarios. According to the critical threshold, HSI identified areas as less or more sustainable catchments. On the other hand, conservation practices simulated through the S2 + EbA scenario envisaged not only additional and viable best management practices (BMP), but also preventive decision-making at the headwaters of water supply systems. We aimed to identify predictors of a successful tracheostomy decannulation. Methods: From 09/2014 to 03/2016, 831 tracheotomized and weaned patients (65.4 +/- 12.9 years, 68% male) were included consecutively in a prospective multicentric observation study. At admission, sociodemographic and clinical data (e.g. relevant neurological and internistic diseases, duration of mechanical ventilation, tracheotomy technique, and nutrition) as well as functional assessments (Coma Recovery Scale-Revised (CRS-R), Early Rehabilitation Barthel Index, Bogenhausener Dysphagia Score) were collected. Complications and the success of the decannulation procedure were documented at discharge. Results: Four hundred seventy patients (57%) were decannulated. The probability of decannulation was significantly negatively associated with increasing age (OR 0.68 per SD = 12.9 years, p < 0.001), prolonged duration of mechanical ventilation (OR 0.57 per 33.2 days, p < 0.001) and complications. An oral diet (OR 3.80; p < 0.001) and a higher alertness at admission (OR 3.07 per 7.18 CRS-R points; p < 0.001) were positively associated. Conclusions: This study identified practically measurable predictors of decannulation, which in the future can be used for a decannulation prognosis and supply optimization at admission in the neurological early rehabilitation clinic. Here, we describe for the first time an ancient and diverging lineage of the Italian wall lizard Podarcis siculus from the western Pontine Islands. We used nuclear and mitochondrial DNA sequences of 156 individuals with the aim of unraveling their phylogenetic position, while microsatellite loci were used to test several a priori insular biogeographic models of migration with empirical data. Our results suggest that the western Pontine populations colonized the islands early during their Pliocene volcanic formation, while populations from the eastern Pontine Islands seem to have been introduced recently. The inter-island genetic makeup indicates an important role of historical migration, probably due to glacial land bridges connecting islands followed by a recent vicariant mechanism of isolation. Moreover, the most supported migration model predicted higher gene flow among islands which are geographically arranged in parallel. Considering the threatened status of small insular endemic populations, we suggest this new evolutionarily independent unit be given priority in conservation efforts. Prior empirical research, however, has yielded inconclusive findings and has differed with respect to the specific aspects of narcissism and socioemotional cognition that have been examined. Here, we propose a differentiated facet approach that can be applied across research traditions and that distinguishes between facets of narcissism (agentic vs. antagonistic) on the one hand, and facets of socioemotional cognition ability (SECA; self-perceived vs. actual) on the other. Using five nonclinical samples in two studies (total N = 602), we investigated the effect of facets of grandiose narcissism on aspects of socioemotional cognition across measures of affective and cognitive empathy, Theory of Mind, and emotional intelligence, while also controlling for general reasoning ability. Across both studies, agentic facets of narcissism were found to be positively related to perceived SECA, whereas antagonistic facets of narcissism were found to be negatively related to perceived SECA. However, both narcissism facets were negatively related to actual SECA. Exploratory condition-based regression analyses further showed that agentic narcissists had a higher directed discrepancy between perceived and actual SECA: They self-enhanced their socio-emotional capacities. Implications of these results for the multifaceted theoretical understanding of the narcissism-SECA link are discussed. Articulated structures are a critical component of overall phenotypic diversity, but data gathered from these structures are difficult to incorporate into modern analyses because of the complexities associated with jointly quantifying 3D shape in multiple structures. While there are existing methods for analyzing shape variation in articulated structures in two-dimensional (2D) space, these methods do not work in 3D, a rapidly growing area of capability and research. Here, we describe a simple geometric rigid rotation approach that removes the effect of random translation and rotation, enabling the morphological analysis of 3D articulated structures. Our method is based on Cartesian coordinates in 3D space, so it can be applied to any morphometric problem that also uses 3D coordinates (e.g., spherical harmonics). We demonstrate the method by applying it to a landmark-based dataset for analyzing shape variation using geometric morphometrics. We have developed an R tool (ShapeRotator) so that the method can be easily implemented in the commonly used R package geomorph and MorphoJ software. This method will be a valuable tool for 3D morphological analyses in articulated structures by allowing an exhaustive examination of shape and size diversity. In this study, we analyze the influence of the thermal and density structure of the upper mantle on the lithospheric stress field and topography. We use a 3-D lithosphere–asthenosphere numerical model with power-law rheology, coupled to a spectral mantle flow code at 300 km depth. Our results are validated against the World Stress Map 2016 (WSM2016) and the observation-based residual topography. We derive the upper mantle thermal structure from either a heat flow model combined with a seafloor age model (TM1) or a global S-wave velocity model (TM2). We show that lateral density heterogeneities in the upper 300 km have a limited influence on the modeled horizontal stress field as opposed to the resulting dynamic topography that appears more sensitive to such heterogeneities. The modeled stress field directions, using only the mantle heterogeneities below 300 km, are not perturbed much when the effects of lithosphere and crust above 300 km are added. In contrast, modeled stress magnitudes and dynamic topography are to a greater extent controlled by the upper mantle density structure. After correction for the chemical depletion of continents, the TM2 model leads to a much better fit with the observed residual topography giving a good correlation of 0.51 in continents, but this correction leads to no significant improvement of the fit between the WSM2016 and the resulting lithosphere stresses. In continental regions with abundant heat flow data, TM1 results in relatively small angular misfits. For example, in western Europe the misfit between the modeled and observation-based stress is 18.3°. Our findings emphasize that the relative contributions coming from shallow and deep mantle dynamic forces are quite different for the lithospheric stress field and dynamic topography. One of them is the conserved CDK5RAP2 protein. Here we focus on the role of Dictyostelium CDK5RAP2 for maintenance of centrosome integrity, its interaction partners and its dynamic behavior during interphase and mitosis. GFP-CDK5RAP2 is present at the centrosome during the entire cell cycle except from a short period during prophase, correlating with the normal dissociation of the corona at this stage. RNAi depletion of CDK5RAP2 results in complete disorganization of centrosomes and microtubules suggesting that CDK5RAP2 is required for organization of the corona and its association to the core structure. This is in line with the observation that overexpressed GFP-CDK5RAP2 elicited supernumerary cytosolic MTOCs. The phenotype of CDK5RAP2 depletion was very reminiscent of that observed upon depletion of CP148, another scaffolding protein of the corona. BioID interaction assays revealed an interaction of CDK5RAP2 not only with the corona markers CP148, gamma-tubulin, and CP248, but also with the core components Cep192, CP75, and CP91. Furthermore, protein localization studies in both depletion strains revealed that CP148 and CDK5RAP2 cooperate in corona organization. Here, I discuss their evolution and their key-functions in microtubule organization, mitosis, and cytokinesis. This is typically characterized by improvements in infants' abilities in discriminating among stimuli of familiar types, such as native speech tones and same-race faces. Simultaneously, infants begin to decline in their ability to discriminate among stimuli of types with which they have little experience, such as nonnative tones and other-race faces. The similarity in time-frames during which perceptual narrowing seems to occur in the domains of speech and face perception has led some researchers to hypothesize that the perceptual narrowing in these domains could be driven by shared domain-general processes. To explore this hypothesis, we tested 53 Caucasian 9-month-old infants from monolingual German households on their ability to discriminate among non-native Cantonese speech tones, as well among same-race German faces and other-race Chinese faces. We tested the infants using an infant-controlled habituation-dishabituation paradigm, with infants' preferences for looking at novel stimuli versus the habituated stimuli (dishabituation scores) acting as indicators of discrimination ability. As expected for their age, infants were able to discriminate between same-race faces, but not between other-race faces or non-native speech tones. Most interestingly, we found that infants' dishabituation scores for the non-native speech tones and other-race faces showed significant positive correlations, while the dishabituation scores for non-native speech tones and same-race faces did not. These results therefore support the hypothesis that shared domain-general mechanisms may drive perceptual narrowing in the domains of speech and face perception. As a striking example, defense-offense coevolution can reverse predator-prey cycles, so predator peaks precede prey peaks rather than vice versa. However, this has only rarely been shown in either model studies or empirical systems. Here, we investigate whether this rarity is a fundamental feature of reversed cycles by exploring under which conditions they should be found. For this, we first identify potential conditions and parameter ranges most likely to result in reversed cycles by developing a new measure, the effective prey biomass, which combines prey biomass with prey and predator traits, and represents the prey biomass as perceived by the predator. We show that predator dynamics always follow the dynamics of the effective prey biomass with a classic 1/4-phase lag. From this key insight, it follows that in reversed cycles (i.e., -lag), the dynamics of the actual and the effective prey biomass must be in antiphase with each other, that is, the effective prey biomass must be highest when actual prey biomass is lowest, and vice versa. Based on this, we predict that reversed cycles should be found mainly when oscillations in actual prey biomass are small and thus have limited impact on the dynamics of the effective prey biomass, which are mainly driven by trait changes. We then confirm this prediction using numerical simulations of a coevolutionary predator-prey system, varying the amplitude of the oscillations in prey biomass: Reversed cycles are consistently associated with regions of parameter space leading to small-amplitude prey oscillations, offering a specific and highly testable prediction for conditions under which reversed cycles should occur in natural systems. However, long-term exposure studies on such polymers are virtually missing. In this context, we synthesized a set of nine polymers that are typically used for low-fouling coatings, comprising the well-established poly(oligoethylene glycol methylether methacrylate), poly(3-(N-2-methacryloylethyl-N,N-dimethyl) ammoniopropanesulfonate) (sulfobetaine methacrylate), and poly(3-(N-3-methacryamidopropyl-N,N-dimethyl)ammoniopropanesulfonate) (sulfobetaine methacrylamide) as well as a series of hitherto rarely studied polysulfabetaines, which had been suggested to be particularly hydrolysis-stable. Hydrolysis resistance upon extended storage in aqueous solution is followed by H-1 NMR at ambient temperature in various pH regimes. Whereas the monomers suffered slow (in PBS) to very fast hydrolysis (in 1 M NaOH), the polymers, including the polymethacrylates, proved to be highly stable. No degradation of the carboxyl ester or amide was observed after one year in PBS, 1 M HCl, or in sodium carbonate buffer of pH 10. This demonstrates their basic suitability for anti-fouling applications. Poly(sulfobetaine methacrylamide) proved even to be stable for one year in 1 M NaOH without any signs of degradation. The stability is ascribed to a steric shielding effect. The hemisulfate group in the polysulfabetaines, however, was found to be partially labile. However, we lack an understanding of how the large variety of different defense traits with different competition costs affects coexistence and population dynamics. Our study focusses on two general defense mechanisms, that is, pre-attack (e.g., camouflage) and post-attack defenses (e.g., weaponry) that act at different phases of the predator—prey interaction. We consider a food web model with one predator, two prey types and one resource. One prey type is undefended, while the other one is pre- or post-attack defended paying costs either by a higher half-saturation constant for resource uptake or a lower maximum growth rate. We show that post-attack defenses promote prey coexistence and stabilize the population dynamics more strongly than pre-attack defenses by interfering with the predator's functional response: Because the predator spends time handling “noncrackable” prey, the undefended prey is indirectly facilitated. A high half-saturation constant as defense costs promotes coexistence more and stabilizes the dynamics less than a low maximum growth rate. The former imposes high costs at low resource concentrations but allows for temporally high growth rates at predator-induced resource peaks preventing the extinction of the defended prey. We evaluate the effects of the different defense mechanisms and costs on coexistence under different enrichment levels in order to vary the importance of bottom-up and top-down control of the prey community. Here, it is shown that such remodeling can also be induced by photoresponsive molecules. The morphological control of giant vesicles in the presence of a water-soluble ortho-tetrafluoroazobenzene photoswitch (F-azo) is demonstrated and it is shown that the shape transformations are based on an increase in membrane area and generation of spontaneous curvature. The vesicles exhibit budding and the buds can be retracted by using light of a different wavelength. In the presence of F-azo, the membrane area can increase by more than 5% as assessed from vesicle electrodeformation. To elucidate the underlying molecular mechanism and the partitioning of F-azo in the membrane, molecular dynamics simulations are employed. Comparison with theoretically calculated shapes reveals that the budded shapes are governed by curvature elasticity, that the spontaneous curvature can be decomposed into a local and a nonlocal contribution, and that the local spontaneous curvature is about 1/(2.5 mu m). The results show that exo- and endocytotic events can be controlled by light and that these photoinduced processes provide an attractive method to change membrane area and morphology. In the current study, we examined the nature of this control mechanism by testing the formation of comparative adjectives in English during language production. Event-related brain potentials (ERPs) were recorded during cued silent production, the first study of this kind for comparative adjective formation. We specifically examined the ERP correlates of producing synthetic relative to analytic comparatives, e.g. angriervs. more angry. A frontal, bilaterally distributed, enhanced negative-going waveform for analytic comparatives (vis-a-vis synthetic ones) emerged approximately 300ms after the (silent) production cue. We argue that this ERP effect reflects a control mechanism that constrains grammatically-based computational processes (viz. more comparative formation). We also address the possibility that this particular ERP effect may belong to a family of previously observed negativities reflecting cognitive control monitoring, rather than morphological encoding processes per se. In the arable hummocky soil landscape, erosion processes caused glacial till appearance closer to the soil surface at upslope positions. The objective of this work was to quantify the potential for subsurface lateral flow depending on the erosion-affected spatial hydropedological complexity. The eroded Haplic Luvisol profile was studied due to the presence of a relatively dense C horizon that varied in depth, thickness, and sloping angle. A two-dimensional numerical modeling and sensitivity analysis for the saturated hydraulic conductivity (K-s) of the C horizon and the depth to C horizon (i.e., soil solum thickness) was performed for rainstorms in 2011 and 2012 using HYDRUS-2D. A K-s value of <2.5 cm d(-1) for the C horizon was required for lateral flow initiation. Lateral flow was (i) increasing with decreasing solum thickness, indicating an erosion-induced feedback on subsurface lateral flow, and (ii) dependent on the soil moisture regime prior to rainstorms. The effect of lateral flow on the movement of a conservative tracer was simulated in the form of a "virtual experiment". Simulation scenarios revealed only a relatively small lateral shift of the tracer plume caused by a local decoupling of water (already lateral) from subsequent tracer movement (still vertical). Longer term simulations suggested that, for the present conditions, lateral flow was limited mostly to occasional summer storm events. Even without considering preferential flow contribution to lateral flow, highly complex hydropedologic interactions are present in erosion-affected heterogeneous soil profiles. Here, we propose a scheme to induce or inhibit these instabilities via a spatio-temporal feedback loop. In a piecewise-linear version of the FitzHugh-Nagumo model, transversal instabilities and spiral turbulence in the uncontrolled system are shown to be suppressed in the presence of control, thereby stabilizing plane wave propagation. The objective of this study was to elucidate the relationship between RBP4, TTR, triglyceride (TG) and type 2 diabetes risk in rural Thailand. Results: RBP4 and TTR levels, as well as homeostatic model assessment of insulin resistance (HOMA-IR) values, were significantly elevated among subjects with high triglyceride levels (p < 0.01, p < 0.05, p < 0.05, respectively). Triglyceride levels correlated with RBP4 (r = 0.34, p < 0.001) and TTR (r= 0.26, p < 0.01) levels, as well as HOMA-IR values (r= 0.16, p < 0.05). After adjustment for age and gender, the risk of hypertriglyceridemia was 3.7 times greater (95% Cl =1.42 -9.73, p = 0.008) in the highest RBP4 tertile as compared to the lowest tertile. Similarly, the highest TTR and HOMA-IR tertiles had greater risk of hypertriglyceridemia at 3.5 (95% Cl = 1.30-9.20, p = 0.01) and 3.6 (95% CI = 1.33- 9.58, p = 0.01) times higher than the respective lowest tertiles. The correlation between TTR and blood glucose was statistically significant (r 0.18, p < 0.05), but not found this relationship in RBP4. Conclusions: The associations of RBP4 and TTR with hypertriglyceridemia and insulin resistance may have important implications for the risk of heart disease and stroke. Water isotopes are key proxies to reconstructing past climatic conditions over the ice sheet and at the evaporation source. The accuracy of climate reconstructions depends on knowledge of all processes affecting water vapour, precipitation and snow isotopic compositions. Fractionation processes are well understood and can be integrated in trajectory-based Rayleigh distillation and isotope-enabled climate models. However, a quantitative understanding of processes potentially altering snow isotopic composition after deposition is still missing. In low-accumulation sites, such as those found in East Antarctica, these poorly constrained processes are likely to play a significant role and limit the interpretability of an ice core's isotopic composition. By combining observations of isotopic composition in vapour, precipitation, surface snow and buried snow from Dome C, a deep ice core site on the East Antarctic Plateau, we found indications of a seasonal impact of metamorphism on the surface snow isotopic signal when compared to the initial precipitation. Particularly in summer, exchanges of water molecules between vapour and snow are driven by the diurnal sublimation-condensation cycles. Overall, we observe in between precipitation events modification of the surface snow isotopic composition. Using high-resolution water isotopic composition profiles from snow pits at five Antarctic sites with different accumulation rates, we identified common patterns which cannot be attributed to the seasonal variability of precipitation. These differences in the precipitation, surface snow and buried snow isotopic composition provide evidence of post-deposition processes affecting ice core records in low-accumulation areas. Given that most peat accumulates in boreal regions, where low temperatures and water saturation preserve organic matter, the existence of peat in (sub)tropical regions remains enigmatic. Here we examined peat and plant chemistry across a latitudinal transect from the Arctic to the tropics. Near-surface low-latitude peat has lower carbohydrate and greater aromatic content than near-surface high-latitude peat, creating a reduced oxidation state and resulting recalcitrance. This recalcitrance allows peat to persist in the (sub)tropics despite warm temperatures. Because we observed similar declines in carbohydrate content with depth in high-latitude peat, our data explain recent field-scale deep peat warming experiments in which catotelm (deeper) peat remained stable despite temperature increases up to 9 degrees C. We suggest that high-latitude deep peat reservoirs may be stabilized in the face of climate change by their ultimately lower carbohydrate and higher aromatic composition, similar to tropical peats. Material and methods: A systematic search was performed in six databases (EBSCO, ScienceDirect, PubMed Central, ProQuest, Scopus, and Web of Science) with empirical studies published between 2006 and 2015 in English with regard to the Common Sense Model of Self-Regulation (CSMR) and familial/hereditary cancer. The key words used were: illness representations, common sense model, self regulatory model, familial/hereditary/genetic cancer, genetic cancer counseling. The selection of studies followed the PRISMA-P guidelines (Moher et al., 2009; Shamseer et al., 2015), which suggest a three-stage procedure. Results: Individuals create their own cognitive and emotional representation of the disease when their health is threatened, being influenced by the presence of a family history of cancer, causing them to adopt or not a salutogenetic behavior. Disease representations, particularly the cognitive ones, can be predictors of responses to health threats that determine different health behaviors. Age, family history of cancer, and worrying about the disease are factors associated with undergoing screening. No consensus has been reached as to which factors act as predictors of compliance with cancer screening programs. Conclusions: This model can generate interventions that are conceptually clear as well as useful in regulating the individuals’ behaviors by reducing the risk of developing the disease and by managing as favorably as possible health and/or disease. This complicates the differentiation between anthropogenic influence and natural variability as well as the detection of changes in water quality which indicate changes in single drivers. We suggest the new term "dominant changes" for changes in multivariate water quality data which concern (1) multiple variables, (2) multiple sites and (3) long-term patterns and present an exploratory framework for the detection of such dominant changes in data sets with irregular sampling in space and time. Firstly, a non-linear dimension-reduction technique was used to summarize the dominant spatiotemporal dynamics in the multivariate water quality data set in a few components. Those were used to derive hypotheses on the dominant drivers influencing water quality. Secondly, different sampling sites were compared with respect to median component values. Thirdly, time series of the components at single sites were analysed for long-term patterns. We tested the approach with a joint stream water and groundwater data set quality consisting of 1572 samples, each comprising sixteen variables, sampled with a spatially and temporally irregular sampling scheme at 29 sites in northeast Germany from 1998 to 2009. The first four components were interpreted as (1) an agriculturally induced enhancement of the natural background level of solute concentration, (2) a redox sequence from reducing conditions in deep groundwater to post-oxic conditions in shallow groundwater and oxic conditions in stream water, (3) a mixing ratio of deep and shallow groundwater to the streamflow and (4) sporadic events of slurry application in the agricultural practice. Dominant changes were observed for the first two components. The changing intensity of the first component was interpreted as response to the temporal variability of the thickness of the unsaturated zone. A steady increase in the second component at most stream water sites pointed towards progressing depletion of the denitrification capacity of the deep aquifer. However, it is not completely clear how inflammation arises in the IVD and which cellular compartments are involved in this process. Recently, the endoplasmic reticulum (ER) has emerged as a possible modulator of inflammation in age-related disorders. In addition, ER stress has been associated with the microenvironment of degenerated IVDs. Therefore, the aim of this study was to analyze the effects of ER stress on inflammatory responses in degenerated human IVDs and associated molecular mechanisms. Gene expression of ER stress marker GRP78 and pro-inflammatory cytokines IL-6, IL-8, IL-1 beta, and TNF-alpha was analyzed in human surgical IVD samples (n = 51, Pfirrmann grade 2-5). The expression of GRP78 positively correlated with the degeneration grade in lumbar IVDs and IL-6, but not with IL-1 beta and TNF-alpha. Another set of human surgical IVD samples (n = 25) was used to prepare primary cell cultures. ER stress inducer thapsigargin (Tg, 100 and 500 nM) activated gene and protein expression of IL-6 and induced phosphorylation of p38 MAPK. Both inhibition of p38 MAPK by SB203580 (10 mu M) and knockdown of ER stress effector CCAAT-enhancer-binding protein homologous protein (CHOP) reduced gene and protein expression of IL-6 in Tg-treated cells. Furthermore, the effects of an inflammatory microenvironment on ER stress were tested. TNF-alpha (5 and 10 ng/mL) did not activate ER stress, while IL-1 beta (5 and 10 ng/mL) activated gene and protein expression of GRP78, but did not influence [Ca2+](i) flux and expression of CHOP, indicating that pro-inflammatory cytokines alone may not induce ER stress in vivo. This study showed that IL-6 release in the IVD can be initiated following ER stress and that ER stress mediates IL-6 release through p38 MAPK and CHOP. Therapeutic targeting of ER stress response may reduce the consequences of the harsh microenvironment in degenerated IVD. However, individual isotope records are affected by a multitude of processes that may mask the true temperature variability. The relative magnitude of climate and non-climate contributions is expected to vary as a function of timescale, and thus it is crucial to determine those temporal scales on which the actual signal dominates the noise. At present, there are no reliable estimates of this timescale dependence of the signal-to-noise ratio (SNR). Here, we present a simple method that applies spectral analyses to stable-isotope data from multiple cores to estimate the SNR, and the signal and noise variability, as a function of timescale. The method builds on separating the contributions from a common signal and from local variations and includes a correction for the effects of diffusion and time uncertainty. We apply our approach to firn-core arrays from Dronning Maud Land (DML) in East Antarctica and from the West Antarctic Ice Sheet (WAIS). For DML and decadal to multi-centennial timescales, we find an increase in the SNR by nearly 1 order of magnitude (similar to 0.2 at decadal and similar to 1.0 at multi-centennial scales). The estimated spectrum of climate variability also shows increasing variability towards longer timescales, contrary to what is traditionally inferred from single records in this region. In contrast, the inferred variability spectrum for WAIS stays close to constant over decadal to centennial timescales, and the results even suggest a decrease in SNR over this range of timescales. We speculate that these differences between DML and WAIS are related to differences in the spatial and temporal scales of the isotope signal, highlighting the potentially more homogeneous atmospheric conditions on the Antarctic Plateau in contrast to the marine-influenced conditions on WAIS. In general, our approach provides a methodological basis for separating local proxy variability from coherent climate variations, which is applicable to a large set of palaeoclimate records. DTM-based studies, however, can contain remarkable biases if limitations and inaccuracies in these models are disregarded. In this work, four freely available datasets, including Shuttle Radar Topography Mission C-Band Synthetic Aperture Radar (SRTM C-SAR V3 DEM), Advanced Spaceborne Thermal Emission and Reflection Radiometer Global Digital Elevation Map (ASTER GDEM V2), and two nationwide airborne light detection and ranging (LiDAR)-derived DTMs (at 5-m and 1-m spatial resolution, respectively) were analysed in three geomorphologically contrasting, small (3–5 km2) catchments located in Mediterranean landscapes under intensive human influence (Mallorca Island, Spain). Vertical accuracy as well as the influence of each dataset’s characteristics on hydrological and geomorphological modelling applicability were assessed by using ground-truth data, classic geometric and morphometric parameters, and a recently proposed index of sediment connectivity. Overall vertical accuracy—expressed as the root mean squared error (RMSE) and normalised median deviation (NMAD)—revealed the highest accuracy for the 1-m (RMSE = 1.55 m; NMAD = 0.44 m) and 5-m LiDAR DTMs (RMSE = 1.73 m; NMAD = 0.84 m). Vertical accuracy of the SRTM data was lower (RMSE = 6.98 m; NMAD = 5.27 m), but considerably higher than for the ASTER data (RMSE = 16.10 m; NMAD = 11.23 m). All datasets were affected by systematic distortions. Propagation of these errors and coarse horizontal resolution caused negative impacts on flow routing, stream network, and catchment delineation, and to a lower extent, on the distribution of slope values. These limitations should be carefully considered when applying DTMs for catchment hydrogeomorphological modelling. This is in striking contrast with traditional conceptual models of rifting and rifted margin formation, which often assume 2-D deformation where the rift velocity is oriented perpendicular to the plate boundary. Here we quantify the validity of this assumption by analysing the kinematics of major continent-scale rift systems in a global plate tectonic reconstruction from the onset of Pangea breakup until the present day. We evaluate rift obliquity by joint examination of relative extension velocity and local rift trend using the script-based plate reconstruction software pyGPlates. Our results show that the global mean rift obliquity since 230 Ma amounts to 34 degrees with a standard deviation of 24 degrees, using the convention that the angle of obliquity is spanned by extension direction and rift trend normal. We find that more than similar to 70 % of all rift segments exceeded an obliquity of 20 degrees demonstrating that oblique rifting should be considered the rule, not the exception. In many cases, rift obliquity and extension velocity increase during rift evolution (e.g. Australia-Antarctica, Gulf of California, South Atlantic, India-Antarctica), which suggests an underlying geodynamic correlation via obliquity-dependent rift strength. Oblique rifting produces 3-D stress and strain fields that cannot be accounted for in simplified 2-D plane strain analysis. We therefore highlight the importance of 3-D approaches in modelling, surveying, and interpretation of most rift segments on Earth where oblique rifting is the dominant mode of deformation. This lack of data limits the use of objective function based evaluation prolific in other modelling fields, and restricts the application of sensitivity analyses in the models and the consequent assessment of model uncertainties. To overcome this deficiency, a novel model function approach has been developed, with each model function representing an aspect of model behaviour, which allows for the application of sensitivity analyses. The model function approach is used to assess the relative sensitivity of the CAESAR-Lisflood LEM to a set of model parameters by applying the Morris method sensitivity analysis for two contrasting catchments. The test revealed that the model was most sensitive to the choice of the sediment transport formula for both catchments, and that each parameter influenced model behaviours differently, with model functions relating to internal geomorphic changes responding in a different way to those relating to the sediment yields from the catchment outlet. The model functions proved useful for providing a way of evaluating the sensitivity of LEMs in the absence of data and methods for an objective function approach. All satellites observe differences in pixel values during repeat observations. In the case of night light data, these changes can be due to both environmental effects and changes in light emission. Here we examine the stability of individual locations of particular large scale light sources (e.g., airports and prisons) in the monthly composites of DNB data from April 2012 to September 2017. The radiances for individual pixels of most large light emitters are approximately normally distributed, with a standard deviation of typically 15-20% of the mean. Greenhouses and flares, however, are not stable sources. We observe geospatial autocorrelation in the monthly variations for nearby sites, while the correlation for sites separated by large distances is small. This suggests that local factors contribute most to the variation in the pixel radiances and furthermore that averaging radiances over large areas will reduce the total variation. A better understanding of the causes of temporal variation would improve the sensitivity of DNB to lighting changes. For example, adding repeated numbers (so-called tie problems, e.g., 2 + 2) not only has a speed and accuracy advantage over adding different numbers (e.g., 1 + 3) but may also lead to under-representation of the result relative to a standard value (Charras et al., 2012, 2014). Does the tie advantage merely reflect easier encoding or retrieval compared to non-ties, or also a distorted result representation? To answer this question, 47 healthy adults performed two tasks, both of which indicated under-representation of tie results: In a result-to-position pointing task (Experiment 1) we measured the spatial mapping of numbers and found a left-bias for tie compared to non-tie problems. In a result-to-line-length production task (Experiment 2) we measured the underlying magnitude representation directly and obtained shorter lines for tie-compared to non-tie problems. These observations suggest that the processing benefit of tie problems comes at the cost of representational reduction of result meaning. This conclusion is discussed in the context of a recent model of arithmetic heuristics and biases. The origin and genesis of the knickpoints can be manifold, considering that the present morphology is the result of the interactions of different factors such as tectonic movements, quaternary glaciations, river captures, variable lithology, and base-level changes. We analyzed the longitudinal profiles of the river channels in the Stura di Demonte Valley (Maritime Alps) to identify the knickpoints of such an alpine setting and to characterize their origins. The distribution and the geometry of stream profiles were used to identify the possible causes of the changes in stream gradients and to define zones with genetically linked knickpoints. Knickpoints are key geomorphological features for reconstructing the evolution of fluvial dissected basins, when the different perturbing factors affecting the ideally graded fluvial system have been detected. This study shows that even in a regionally small area, perturbations of river profiles are caused by multiple factors. Thus, attributing (automatically)-extracted knickpoints solely to one factor, can potentially lead to incomplete interpretations of catchment evolution. Calibration was performed manually. Subsequently, the modellers were asked for their experience, their calibration strategy, and whether they enjoyed the exercise. The exercise revealed that there is considerable modellers’ uncertainty even among the experienced modellers. It seemed to be equally important whether the modellers followed a good calibration strategy, and whether they enjoyed modelling. The exercise confirmed previous studies about the benefit of model ensembles: Different combinations of the simulation results (median, mean) outperformed the individual model simulations, while filtering the simulations even improved the quality of the model ensembles. Modellers’ experience, decisions, and attitude, therefore, have an impact on the hydrological model application and should be considered as part of hydrological modelling uncertainty. Until now, there is a paucity of studies that investigated both, the maternal and the fetal serum metabolome in the setting of GDM. Mounting evidence suggests that the fetus is not just passively affected by gestational disease but might play an active role in it. Metabolomic studies performed in maternal blood and fetal cord blood could help to better discern distinct fetal from maternal disease interactions. Methods: At the time of birth, serum samples from mothers and newborns (cord blood samples) were collected and screened for 163 metabolites utilizing tandem mass spectrometry. The cohort consisted of 412 mother/child pairs, including 31 cases of maternal GDM. Results: An initial non-adjusted analysis showed that eight metabolites in the maternal blood and 54 metabolites in the cord blood were associated with GDM. After Benjamini-Hochberg (BH) procedure and adjustment for confounding factors for GDM, fetal phosphatidylcholine acyl-alkyl C 32:1 and proline still showed an independent association with GDM. Conclusions: This study found metabolites in cord blood which were associated with GDM, even after adjustment for established risk factors of GDM. To the best of our knowledge, this is the first study demonstrating an independent association between fetal serum metabolites and maternal GDM. Despite several modular assembly methods developed in recent years, the plant biotechnology community has not widely adopted them yet, probably due to the lack of appropriate vectors and software tools. Here we present Plant X-tender, an extension of the highly efficient, scar-free and sequence-independent multigene assembly strategy AssemblX, based on overlap-depended cloning methods and rare-cutting restriction enzymes. Plant X-tender consists of a set of plant expression vectors and the protocols for most efficient cloning into the novel vector set needed for plant expression and thus introduces advantages of AssemblX into plant synthetic biology. The novel vector set covers different backbones and selection markers to allow full design flexibility. We have included ccdB counterselection, thereby allowing the transfer of multigene constructs into the novel vector set in a straightforward and highly efficient way. Vectors are available as empty backbones and are fully flexible regarding the orientation of expression cassettes and addition of linkers between them, if required. We optimised the assembly and subcloning protocol by testing different scar-less assembly approaches: the noncommercial SLiCE and TAR methods and the commercial Gibson assembly and NEBuilder HiFi DNA assembly kits. Plant X-tender was applicable even in combination with low efficient homemade chemically competent or electrocompetent Escherichia coli. We have further validated the developed procedure for plant protein expression by cloning two cassettes into the newly developed vectors and subsequently transferred them to Nicotiana benthamiana in a transient expression setup. Thereby we show that multigene constructs can be delivered into plant cells in a streamlined and highly efficient way. Our results will support faster introduction of synthetic biology into plant science. Topical application of ETR loaded tNGs to inflammatory skin equivalents or tape striped human skin resulted in efficient ETR delivery throughout the SC and into the viable epidermis that correlated with clear anti-inflammatory effects. Genetic incompatibilities were observed between lacertid populations through crossing experiments, which led to the delineation of two separate species within the L. viridis complex. The population history of these sister species and processes driving divergence are unknown. We constructed the first high-quality de novo genome assemblies for both L. viridis and L. bilineata through Illumina and PacBio sequencing, with annotation support provided from transcriptome sequencing of several tissues. To estimate gene flow between the two species and identify factors involved in reproductive isolation, we studied their evolutionary history, identified genomic rearrangements, detected signatures of selection on non-coding RNA, and on protein-coding genes. Findings Here we show that gene flow was primarily unidirectional from L. bilineata to L. viridis after their split at least 1.15 million years ago. We detected positive selection of the non-coding repertoire; mutations in transcription factors; accumulation of divergence through inversions; selection on genes involved in neural development, reproduction, and behavior, as well as in ultraviolet-response, possibly driven by sexual selection, whose contribution to reproductive isolation between these lacertid species needs to be further evaluated. Conclusion The combination of short and long sequence reads resulted in one of the most complete lizard genome assemblies. The characterization of a diverse array of genomic features provided valuable insights into the demographic history of divergence among European green lizards, as well as key species differences, some of which are candidates that could have played a role in speciation. In montane and subalpine forests, environmental influences show a high spatial heterogeneity, which is reflected by a high spatial variability of humus forms. This study aims at examining spatial patterns of humus forms and their dependence on the spatial scale in a high mountain forest environment (Val di Sole/Val di Rabbi, Trentino, Italian Alps). On the basis of the distributions of environmental covariates across the study area, we described humus forms at the local scale (six sampling sites), slope scale (60 sampling sites) and landscape scale (30 additional sampling sites). The local variability of humus forms was analyzed with regard to the ground cover type. At the slope and landscape scale, spatial patterns of humus forms were modeled applying random forests and ordinary kriging of the model residuals. The results indicate that the occurrence of the humus form classes Mull, Mullmoder, Moder, Amphi and Eroded Moder generally depends on the topographical position. Local-scale patterns are mostly related to micro-topography (local accumulation and erosion sites) and ground cover, whereas slope-scale patterns are mainly connected with slope exposure and elevation. Patterns at the landscape scale show a rather irregular distribution, as spatial models at this scale do not account for local to slope-scale variations of humus forms. Moreover, models at the slope scale perform distinctly better than at the landscape scale. In conclusion, the results of this study highlight that landscape-scale predictions of humus forms should be accompanied by local- and slope-scale studies in order to enhance the general understanding of humus form patterns. We estimated within-lake variation in biomass using high-frequency chlorophyll fluorescence data from 18 globally distributed lakes. We tested how variation in fluorescence at monthly, daily, and hourly scales was related to high-frequency variability of wind, water temperature, and radiation within lakes as well as productivity and physical attributes among lakes. Within lakes, monthly variation dominated, but combined daily and hourly variation were equivalent to that expressed monthly. Among lakes, biomass variability increased with trophic status while, within-lake biomass variation increased with increasing variability in wind speed. Our results highlight the benefits of high-frequency chlorophyll monitoring and suggest that predicted changes associated with climate, as well as ongoing cultural eutrophication, are likely to substantially increase the temporal variability of algal biomass and thus the predictability of the services it provides. Aim: We aim to develop reference centiles for external skeletal robusticity of European girls and boys aged 0 to 18 years using the Frame Index as an indicator and identify population specific age-related patterns. Methods: We analysed cross-sectional & longitudinal data on body height and elbow breadth of boys and girls from Europe (0-18 years, n = 41.679), India (7-18 years, n = 3.297) and South Africa (3-18 years, n = 4.346). As an indicator of external skeletal robusticity Frame Index after Frisancho (1990) was used. We developed centiles for boys and girls using the LMS-method and its extension. Results: Boys have greater external skeletal robusticity than girls. Whereas in girls Frame Index decreases continuously during growth, an increase of Frame Index from 12 to 16 years in European boys can be observed. Indian and South African boys are almost similar in Frame Index to European boys. In girls, the pattern is slightly different. Whereas South African girls are similar to European girls, Indian girls show a lesser external skeletal robusticity. Conclusion: Accurate references for external skeletal robusticity are needed to evaluate if skeletal development is adequate per age. They should be used to monitor effects of changes in way of life and physical activity levels in children and adolescents to avoid negative health outcomes like osteoporosis and arthrosis. In the present study, the effective elastic moduli of two digital rock samples, the Fontainebleau and Bentheim sandstones, are numerically determined based on micro-CT images. Reduction in rock stiffness due to the dissolution of 10% calcite cement by volume out of the pore network is quantified for three synthetic spatial calcite distributions (coating, partial filling and random) using representative sub-cubes derived from the digital rock samples. Due to the reduced calcite content, bulk and shear moduli decrease by 34% and 38% in maximum, respectively. Total porosity is clearly the dominant parameter, while spatial calcite distribution has a minor impact, except for a randomly chosen cement distribution within the pore network. Moreover, applying an initial stiffness reduced by 47% for the calcite cement results only in a slightly weaker mechanical behaviour. Using the quantitative approach introduced here substantially improves the accuracy of predictions in elastic rock properties compared to general analytical methods, and further enables quantification of uncertainties related to spatial variations in porosity and mineral distribution. The present study aims (1) to evaluate the factorial validity of the TICS within the context of competitive sports and (2) to adapt a short version (TICS-36). The total sample consisted of 564 athletes (age in years: M = 19.1, SD = 3.70). The factor structure of the original TICS did not adequately fit the present data, whereas the short version presented a satisfactory fit. The results indicate that the TICS-36 is an economical instrument for gathering interpretable information about chronic stress. Cerebral cavernous malformation (CCM) is an inherited vascular disease that occurs when a second somatic mutation causes a loss of CCM1/KRIT1, CCM2, or CCM3 proteins. Here, we demonstrate that zebrafish Krit1 regulates the formation of cardiac valves. Expression of heg1, which encodes a binding partner of Krit1, is positively regulated by blood-flow. In turn, Heg1 stabilizes levels of Krit1 protein, and both Heg1 and Krit1 dampen expression levels of klf2a, a major mechanosensitive gene. Conversely, loss of Krit1 results in increased expression of klf2a and notch1b throughout the endocardium and prevents cardiac valve leaflet formation. Hence, the correct balance of blood-flow-dependent induction and Krit1 protein mediated repression of klf2a and notch1b ultimately shapes cardiac valve leaflet morphology. ECs derived from human induced pluripotent stem cells (hiPSCs) represent an alternative and potentially superior cell source, but traditional culture approaches and 2D differentiation protocols hardly allow for production of large cell numbers. Aiming at the production of ECs, we have developed a robust approach for efficient endothelial differentiation of hiPSCs in scalable suspension culture. The most commonly used approach involves mapping sequence reads to a reference genome. However, this process becomes increasingly challenging with an elevated genetic distance between target and reference or with the presence of contaminant sequences with high sequence similarity to the target species. The evaluation and testing of mapping efficiency and stringency are thus paramount for the reliable identification and analysis of ancient sequences. In this paper, we present ‘TAPAS’, (Testing of Alignment Parameters for Ancient Samples), a computational tool that enables the systematic testing of mapping tools for ancient data by simulating sequence data reflecting the properties of an ancient dataset and performing test runs using the mapping software and parameter settings of interest. We showcase TAPAS by using it to assess and improve mapping strategy for a degraded sample from a banded linsang (Prionodon linsang), for which no closely related reference is currently available. This enables a 1.8-fold increase of the number of mapped reads without sacrificing mapping specificity. The increase of mapped reads effectively reduces the need for additional sequencing, thus making more economical use of time, resources, and sample material. In the freshwater model species Daphnia magna, essential lipid requirements have been extensively studied. Dietary deficiencies in sterols and polyunsaturated fatty acids (PUFA) have been shown to constrain somatic growth and parthenogenetic reproduction of female Daphnia. In contrast, nutrient requirements of male Daphnia have not been studied yet. Supplementation experiments were conducted to investigate differences in sterol (cholesterol) and PUFA (eicosapentaenoic acid, EPA) requirements between female and male D. magna. Thresholds for sterol-limited juvenile growth were higher in females than in males, suggesting that females are more susceptible to dietary sterol deficiencies than males. Sex-specific differences in maximum somatic growth rates were evident primarily in the presence of dietary EPA; females could not exploit their generally higher growth potential in the absence of dietary PUFA. However, the thresholds for EPA-limited growth did not differ between sexes, suggesting that both sexes have similar dietary EPA requirements during juvenile growth. During a life history experiment, the gain in body dry mass was higher in females than in males, irrespective of food treatment. In both sexes, the gain in body dry mass increased significantly upon EPA supplementation, indicating that both sexes benefited from dietary EPA supply also later in life. However, the positive effects of EPA supplementation were most pronounced for female reproduction-related traits (i.e., clutch sizes, egg dry masses, and total dry mass investment in reproduction). The high maternal investment in reproduction resulted in a depletion of nutrients in female somata. In contrast, the comparatively low paternal investment in reproduction allowed for the accumulation of nutrients in male somata. We conclude that males are generally less susceptible to dietary nutrient deficiencies than females, because they can rely more on internal body stores. Our data suggest that the performances of the sexes are differentially influenced by lipid-mediated food quality, which may have consequences for sexual reproduction and thus the production of resting eggs and the maintenance of Daphnia populations. Yet, limited sample sizes and variation in space and habitat use across regions and life stages may compromise robustness of behavioral analyses and subsequent conservation plans. Here, we assessed variation in (i) home range sizes, (ii) home range selection, and (iii) fine-scale resource selection of white storks across breeding status and regions and test model transferability. Three study areas were chosen within the Central German breeding grounds ranging from agricultural to fluvial and marshland. We monitored GPS-locations of 62 adult white storks equipped with solar-charged GPS/3D-acceleration (ACC) transmitters in 2013-2014. Home range sizes were estimated using minimum convex polygons. Generalized linear mixed models were used to assess home range selection and fine-scale resource selection by relating the home ranges and foraging sites to Corine habitat variables and normalized difference vegetation index in a presence/pseudo-absence design. We found strong variation in home range sizes across breeding stages with significantly larger home ranges in non-breeding compared to breeding white storks, but no variation between regions. Home range selection models had high explanatory power and well predicted overall density of Central German white stork breeding pairs. Also, they showed good transferability across regions and breeding status although variable importance varied considerably. Fine-scale resource selection models showed low explanatory power. Resource preferences differed both across breeding status and across regions, and model transferability was poor. Our results indicate that habitat selection of wild animals may vary considerably within and between populations, and is highly scale dependent. Thereby, home range scale analyses show higher robustness whereas fine-scale resource selection is not easily predictable and not transferable across life stages and regions. Such variation may compromise management decisions when based on data of limited sample size or limited regional coverage. We thus recommend home range scale analyses and sampling designs that cover diverse regional landscapes and ensure robust estimates of habitat suitability to conserve wild animal populations. However, their contribution to the organic carbon (OC) budget is unknown. We provide the first estimate of the contribution of RTSs to the nearshore OC budget of the Yukon Coast, Canada, and describe the evolution of coastal RTSs between 1952 and 2011 in this area. We (1) describe the evolution of RTSs between 1952 and 2011; (2) calculate the volume of eroded material and stocks of OC mobilized through slumping, including soil organic carbon (SOC) and dissolved organic carbon (DOC); and (3) estimate the OC fluxes mobilized through slumping between 1972 and 2011. We identified RTSs using high- resolution satellite imagery from 2011 and geocoded aerial photographs from 1952 and 1972. To estimate the volume of eroded material, we applied spline interpolation on an airborne lidar dataset acquired in July 2013. We inferred the stocks of mobilized SOC and DOC from existing related literature. Our results show a 73% increase in the number of RTSs and 14% areal expansion between 1952 and 2011. In the study area, RTSs displaced at least 16.6 x 10(6) m(3) of material, 53% of which was ice, and mobilized 145.9 x 10(6) kg of OC. Between 1972 and 2011, 49 RTSs displaced 8.6 x 10(3) m(3) yr(-1) of material, adding 0.6% to the OC flux released by coastal retreat along the Yukon Coast. Our results show that the contribution of RTSs to the nearshore OC budget is non-negligible and should be included when estimating the quantity of OC released from the Arctic coast to the ocean. Numerous scholars have problematized this approach. They have shown that increasing the levels of flood protection can attract more settlements and high-value assets in the areas protected by the new measures. Other studies have explored how structural measures can generate a sense of complacency, which can act to reduce preparedness. These paradoxical risk changes have been described as "levee effect", "safe development paradox" or "safety dilemma". In this commentary, we briefly review this phenomenon by critically analysing the intended benefits and unintended effects of structural flood protection, and then we propose an interdisciplinary research agenda to uncover these paradoxical dynamics of risk. Especially in intentionally planned change processes (e.g., change management), forgetting is an important precondition to impede the recall of obsolete routines and adapt to new strategic objectives accompanied by new organizational routines. We first comprehensively review the literature on the need for organizational forgetting and particularly on accidental vs. intentional forgetting. We discuss the current state of the art of theory and empirical evidence on forgetting from cognitive psychology in order to infer mechanisms applicable to the organizational context. In this respect, we emphasize retrieval theories and the relevance of retrieval cues important for forgetting. Subsequently, we transfer the empirical evidence that the elimination of retrieval cues leads to faster forgetting to the forgetting of organizational routines, as routines are part of organizational memory. We then propose a classification of cues (context, sensory, business process-related cues) that are relevant in the forgetting of routines, and discuss a meta-cue called the "situational strength" cue, which is relevant if cues of an old and a new routine are present simultaneously. Based on the classification as business process-related cues (information, team, task, object cues), we propose mechanisms to accelerate forgetting by eliminating specific cues based on the empirical and theoretical state of the art. We conclude that in intentional organizational change processes, the elimination of cues to accelerate forgetting should be used in change management practices. Weak vortex states, often associated with sudden stratospheric warmings (SSW), have been shown to increase the risk of cold-spells especially over Eurasia, but its role for North American winters is less clear. Using cluster analysis, we show that there are two dominant patterns of increased polar cap heights in the lower stratosphere. Both patterns represent a weak polar vortex but they are associated with different wave mechanisms and different regional tropospheric impacts. The first pattern is zonally symmetric and associated with absorbed upward-propagating wave activity, leading to a negative phase of the North Atlantic Oscillation (NAO) and cold-air outbreaks over northern Eurasia. This coupling mechanism is well-documented in the literature and is consistent with the downward migration of the northern annular mode (NAM). The second pattern is zonally asymmetric and linked to downward reflected planetary waves over Canada followed by a negative phase of the Western Pacific Oscillation (WPO) and cold-spells in Central Canada and the Great Lakes region. Causal effect network (CEN) analyses confirm the atmospheric pathways associated with this asymmetric pattern. Moreover, our findings suggest the reflective mechanism to be sensitive to the exact region of upward wave-activity fluxes and to be state-dependent on the strength of the vortex. Identifying the causal pathways that operate on weekly to monthly timescales can pave the way for improved sub-seasonal to seasonal forecasting of cold spells in the mid-latitudes. In this study, we focus on the functional characterization of the Arabidopsis thaliana TF GROWTH-REGULATING FACTOR9 (GRF9) and demonstrate that it exerts its negative effect on leaf growth by activating expression of the bZIP TF OBP3-RESPONSIVE GENE 3 (ORG3). While grf9 knockout mutants produce bigger incipient leaf primordia at the shoot apex, rosette leaves and petals than the wild type, the sizes of those organs are reduced in plants overexpressing GRF9 (GRF9ox). Cell measurements demonstrate that changes in leaf size result from alterations in cell numbers rather than cell sizes. Kinematic analysis and 5-ethynyl-2'-deoxyuridine (EdU) incorporation assay revealed that GRF9 restricts cell proliferation in the early developing leaf. Performing in vitro binding site selection, we identified the 6-base motif 5'-CTGACA-3' as the core binding site of GRF9. By global transcriptome profiling, electrophoretic mobility shift assay (EMSA) and chromatin immunoprecipitation (ChIP) we identified ORG3 as a direct downstream, and positively regulated target of GRF9. To gain a mechanistic understanding of the catalytic reactions, knowledge of the oxidation state of the active metals, ideally in operando, is therefore critical. L-edge X-ray absorption spectroscopy (XAS) is a powerful technique that is frequently used to infer the oxidation state via a distinct blue shift of L-edge absorption energies with increasing oxidation state. A unified description accounting for quantum-chemical notions whereupon oxidation does not occur locally on the metal but on the whole molecule and the basic understanding that L-edge XAS probes the electronic structure locally at the metal has been missing to date. Here we quantify how charge and spin densities change at the metal and throughout the molecule for both redox and core-excitation processes. We explain the origin of the L-edge XAS shift between the high-spin complexes Mn-II(acac)(2) and Mn-III(acac)(3) as representative model systems and use ab initio theory to uncouple effects of oxidation-state changes from geometric effects. The shift reflects an increased electron affinity of Mn-III in the core-excited states compared to the ground state due to a contraction of the Mn 3d shell upon core-excitation with accompanied changes in the classical Coulomb interactions. This new picture quantifies how the metal-centered core hole probes changes in formal oxidation state and encloses and substantiates earlier explanations. The approach is broadly applicable to mechanistic studies of redox-catalytic reactions in molecular systems where charge and spin localization/delocalization determine reaction pathways. The amine end-functionalized polystyrene surfactants with varying molecular weight were prepared via atom-transfer radical polymerization technique. By changing the concentration of these polymeric surfactants, Co NPs with different size, size distribution, and magnetic properties were obtained. Transmission electron microscopy characterization showed that the size of Co NPs stabilized with lower molecular weight polystyrene surfactants (M-n = 2300 g/mol) varied from 12-22 nm, while the size of Co NPs coated with polystyrene of middle (M-n = 4500 g/mol) and higher molecular weight (M-n = 10,500 g/mol) showed little change around 20 nm. Magnetic measurements revealed that the small cobalt particles were superparamagnetic, while larger particles were ferromagnetic and self-assembled into 1-D chain structures. Thermogravimetric analysis revealed that the grafting density of polystyrene with lower molecular weight is high. To the best of our knowledge, this is the first study to obtain both superparamagnetic and ferromagnetic Co NPs by changing the molecular weight and concentration of polystyrene through the dual-stage decomposition method. Here, we present the results of several unbiased small-molecule suppression screens in which we applied a total of 5,268 unique substances to CCM mutant worm, zebrafish, mouse, or human endothelial cells. We used a systems biology-based target prediction tool to integrate the results with the whole-transcriptome profile of zebrafish CCM2 mutants, revealing signaling pathways relevant to the disease and potential targets for small-molecule-based therapies. We found indirubin-3-monoxime to alleviate the lesion burden in murine preclinical models of CCM2 and CCM3 and suppress the loss-of-CCM phenotypes in human endothelial cells. Our multi-organism-based approach reveals new components of the CCM regulatory network and foreshadows novel small-molecule-based therapeutic applications for suppressing this devastating disease in patients. The extent of recombination depends on induction time and concentration of the chromophore phycocyanobilin (PCB), which can be easily adjusted. The tool presented here provides improved recombination control over the previously reported estradiol-dependent SCRaMbLE induction system, mediating a larger variety of possible recombination events in SCRaMbLE-ing a reporter plasmid. The diameter of the TiO2 nanoparticles (NPs) - mainly anatase with a minor fraction of brookite - and the Au NPs are on the order of 5 and 7-18 nm, respectively. Addition of poly(ethylene oxide) to the reaction mixture enables pore size tuning, thus providing access to different materials with different photocatalytic activities. Water splitting experiments using a sunlight simulator and a Xe lamp show that the new hybrid materials are effective water splitting catalysts and produce up to 30 mmol of hydrogen per 24 h. Overall the article demonstrates that the combination of a renewable and robust scaffold such as B. mori silk with a photoactive material provides a promising approach to new monolithic photocatalysts that can easily be recycled and show great potential for application in lightweight devices for green fuel production. The present study examined the semantic relatedness and semantic similarity of literal and non-literal (metonymic and metaphorical) senses of three word classes: nouns, verbs, and adjectives. Two methods were used: a psycholinguistic experiment and a distributional analysis of corpus data. In the experiment, participants were presented with 6-12 short phrases containing a polysemous word in literal, metonymic, or metaphorical senses and were asked to classify them so that phrases with the same perceived sense were grouped together. To investigate the impact of professional background on their decisions, participants were controlled for linguistic vs. non-linguistic education. For nouns and verbs, all participants preferred to group together phrases with literal and metonymic senses, but not any other pairs of senses. For adjectives, two pairs of senses were often grouped together: literal with metonymic, and metonymic with metaphorical. Participants with a linguistic background were more accurate than participants with non-linguistic backgrounds, although both groups shared principal patterns of sense classification. For the distributional analysis of corpus data, we used a semantic vector approach to quantify the similarity of phrases with literal, metonymic, and metaphorical senses in the corpora. We found that phrases with literal and metonymic senses had the highest degree of similarity for the three word classes, and that metonymic and metaphorical senses of adjectives had the highest degree of similarity among all word classes. These findings are in line with the experimental results. Overall, the results suggest that the mental representation of a polysemous word depends on its word class. In nouns and verbs, literal and metonymic senses are stored together, while metaphorical senses are stored separately; in adjectives, metonymic senses significantly overlap with both literal and metaphorical senses. Anti-genotoxicity against the human liver carcinogen aflatoxin B1 (AFB(1)) as determined by the comet assay and radical oxygen species (ROS) scavenging capacity of ethanolic and aqueous extracts were investigated in human derived liver (HepG2) cells. ROS scavenging activity was assessed using electron paramagnetic spin resonance and quantification of ARE/Nrf2 mediated gene expression. The cultivation was done under different environmental conditions. The processing included fermentation and cooking; postharvest ultraviolet irradiation (UV-C) treatment was also investigated. Overall, S. scabrum extracts showed strong health promoting potential, the highest potential was observed with the fermented extract, which showed a 60% reduction of AFB(1) induced DNA damage and a 38% reduction in FeSO4 induced oxidative stress. The content of total polyphenols, carotenoids and chlorophylls was indeed affected by cultivation and processing. Based on the present in vitro findings consumption of S. scabrum leaves could be further encouraged, preferentially after cooking or fermentation of the plant. A recently developed data-assimilation-based framework that allows model parameters to vary through time in response to signals of change in observations is considered for a medium-sized catchment (2880 km(2)) in northern Vietnam experiencing substantial but gradual land cover change. We investigate the efficacy of the method as well as the importance of the chosen model structure in ensuring the success of a time-varying parameter method. The method was used with two lumped daily conceptual models (HBV and HyMOD) that gave good-quality streamflow predictions during pre-change conditions. Although both time-varying parameter models gave improved streamflow predictions under changed conditions compared to the time-invariant parameter model, persistent biases for low flows were apparent in the HyMOD case. It was found that HyMOD was not suited to representing the modified baseflow conditions, resulting in extreme and unrealistic time-varying parameter estimates. This work shows that the chosen model can be critical for ensuring the time-varying parameter framework successfully models streamflow under changing land cover conditions. It can also be used to determine whether land cover changes (and not just meteorological factors) contribute to the observed hydrologic changes in retrospective studies where the lack of a paired control catchment precludes such an assessment. Further, the impact of breadmaking on these metabolites using pea and lupin material of two ontogenetic stages as an added ingredient in wheat-based bread was assessed. In "pea microgreen bread" no decrease of quercetin was found with regard to the non-processed plant material. However kaempferol glycosides showed slight decreases induced by the breadmaking process in "pea microgreen bread" and "pea leaf bread." In "lupin microgreen bread" no decrease of genistein compared to the non-processed plant material was found. Chrysoeriol glycosides showed slight decreases induced by the breadmaking process in "lupin microgreen bread" and "lupin leaf bread." In all breads, carotenoids and chlorophylls were depleted however pheophytin formation was caused. In this study, we address the spatial deformation pattern in the data-scarce western Himalaya. We calculated catchment averaged, normalized river-steepness indices of non-glaciated drainage basins with tributary catchment areas between 5 and 200 km(2) (n = 2138). We analyzed the spatial distribution of the relative change of river steepness both along and across strike to gain information about the regional distribution of differential uplift pattern and relate this to the activity of distinctive fault segments. For our study area, we observe a positive correlation of averaged k(sn) values with long-term exhumation rates derived from previously published thermochronologic datasets combined with thermal modeling as well as with millennial timescale denudation rates based on cosmogenic nuclide dating. Our results indicate three tectono-geomorphic segments with distinctive landscape morphology, structural architecture, and fault geometry along the western Himalaya: Garhwal-Sutlej, Chamba, and Kashmir Himalaya (from east to west). Moreover, our data recognize distinctive fault segments showing varying thrust activity along strike of the Main Frontal Thrust, the Main Boundary Thrust, and in the vicinity of the steep topographic transition between the Lesser and Greater Himalaya. In this region, we relate out-of-sequence deformation along major basement thrust ramps, such as the Munsiari Thrust with deformation along a mid-crustal ramp along the basal decollement. We suggest that during the Quaternary, all major fault zones in the Western Himalaya experienced out-of-sequence faulting and have accommodated some portion of crustal shortening. In the actively deforming western Tarim Basin in NW China, Pliocene-Pleistocene conglomerates were dated at eight sites, integrating Al-26/Be-10 burial dating with previously published magnetostratigraphic sections. These samples were collected from growth strata on the flanks of growing folds and from sedimentary units beneath active faults to place timing constraints on the initiation of deformation of structures within the basin and on shortening rates on active faults. However, stopover sites are crucial for migratory birds. They are often used by a multitude of species, which could lead to increased competition. In this work, we investigated niche use of 8 migratory and closely related Emberiza bunting species at a stopover site in Far East Russia, situated on the poorly studied East Asian fly-way. We used bird ringing data to evaluate morphological similarity as well as niche overlap on the trophic, spatial, and temporal dimension. Bill morphology was used as a proxy for their trophic niche. We were able to prove that a majority of the species occupies well-defined stopover niches on at least one of the dimensions. Niche breadth and niche overlap differ between spring and autumn season with higher overlap found during spring. Morphological differences are mostly related to overall size and wing pointedness. The temporal dimension is most important for segregation among the studied species. Furthermore, all species seem to exhibit a rather strict and consistent phenological pattern. Their occurrence at the study site is highly correlated with their geographic origin and the length of their migration route. We assume that buntings are able to use available resources opportunistically during stopover, while trying to follow a precise schedule in order to avoid competition and to ensure individual fitness. The exact timing of events and the significance of geochronological information are debated due to the interplay of tectonic, metamorphic, and metasomatic processes. Here we present new geochronological data using Rb-Sr internal mineral isochrons and in situ Ar-40/Ar-39 laser ablation data to provide constraints on the relative importance of fluid-mediated mineral replacement reactions and diffusion for the interpretation of radiogenic isotope signatures, and on the use of these isotopic systems for dating metamorphic and variably deformed rocks. Our study focuses on the shear zone at the contact between two major lithological units of the Sesia zone, the eclogitic micaschists and the gneiss minuti. Metasedimentary rocks of the eclogitic micaschists unit contain phengite with step-like zoning in major element chemistry as evidence for petrologic disequilibrium. Distinct Ar-40/Ar-39 spot ages of relict phengite cores and over-printed rims demonstrate the preservation of individual age domains in the crystals. The eclogitic micaschists also show systematic Sr isotope disequilibria among different phengite populations, so that minimum ages of relict assemblage crystallization can be differentiated from the timing of late increments of deformation. The preservation of these disequilibrium features shows the lack of diffusive re-equilibration and underpins that fluid-assisted dissolution and recrystallization reactions are the main factors controlling the isotope record in these subduction-related metamorphic rocks. Blueschist-facies mylonites record deformation along the major shear zone that separates the eclogitic micaschists from the gneiss minuti. Two Rb-Sr isochrones that comprise several white mica fractions and glaucophane constrain the timing of this deformation and accompanying near-complete blueschist-facies re-equilibration of the Rb-Sr system to 60.1 +/- 0.9 Ma and 60.9 +/- 2.1 Ma, respectively. Overlapping ages in eclogitic micaschists of 60.1 +/- 1.1 (Rb-Sr isochron of sheared matrix assemblage), 58.6 +/- 0.8, and 60.9 +/- 0.4 Ma (white mica Ar-40/Ar-39 inverse isochron ages) support the significance of this age and show that fluid-rock interaction and partial re-equilibration occurred as much as several kilometers away from the shear zone. An earlier equilibration during high-pressure conditions in the eclogitic mica schists is recorded in minimum Rb-Sr ages for relict assemblages (77.2 +/- 0.8 and 72.4 +/- 1.1 Ma) and an Ar-40/Ar-39 inverse isochron age of 75.4 +/- 0.8 Ma for white mica cores, again demonstrating that the two isotope systems provide mutually supporting geochronological information. Local reactivation and recrystallization along the shear zone lasted >15 m.y., as late increments of deformation are recorded in a greenschist-facies mylonite by a Rb-Sr isochron age of 46.5 +/- 0.7 Ma. The integrated SENSKIN technology will be implemented in the case of steel and concrete bridges, and tested, field-evaluated and benchmarked on actual bridge environment against a conventional health monitoring solution developed by Mistras Group Hellas. The main objective of the present work is to implement the autonomous, fully functional strain monitoring system based on commercially available off-the-shelf components, that will be used to accomplish direct comparison between the performance of the innovative SENSKIN sensors and the conventional strain sensors commonly used for structural monitoring of bridges. For this purpose, the mini Structural Monitoring System (mini SMS) of Physical Acoustics Corporation, a comprehensive data acquisition unit designed specifically for long-term unattended operation in outdoor environments, was selected. For the completion of the conventional system, appropriate foil-type strain sensors were selected, driven by special conditioners manufactured by Mistras Group. A comprehensive description of the strain monitoring system and its peripheral components is provided in this paper. For the evaluation of the integrated system’s performance and the effect of various parameters on the long-term behavior of sensors, several test steel pieces instrumented with different strain sensors configurations were prepared and tested in both laboratory and field ambient conditions. Furthermore, loading tests were performed aiming to validate the response of the system in monitoring the strains developed in steel beam elements subject to bending regimes. Since the last century, deforestation and expanding agricultural activities have been changing the ecosystem and its provision of moisture to the atmosphere. However, it remains uncertain how the ongoing land use change will influence rainfall, runoff, and water availability as findings from previous studies differ. Using moisture tracking experiments based on observational data, we provide a spatially detailed analysis recognizing potential teleconnection between source and sink regions of atmospheric moisture. We apply land use scenarios in upwind moisture sources and quantify the corresponding rainfall and runoff changes in downwind moisture sinks. We find spatially varying responses of water regimes to land use changes, which may explain the diverse results from previous studies. Parts of the Peruvian Amazon and western Bolivia are identified as the sink areas most sensitive to land use change in the Amazon and we highlight the current water stress by Amazonian land use change on these areas in terms of the water availability. Furthermore, we also identify the influential source areas where land use change may considerably reduce a given target sink's water reception (from our example of the Ucayali River basin outlet, rainfall by 5–12 % and runoff by 19–50 % according to scenarios). Sensitive sinks and influential sources are therefore suggested as hotspots for achieving sustainable land–water management. This effect describes that comparable talented students in high-achieving school settings have a lower ASC compared to equally talented students attending low-achieving settings. Past research has focused on students’ domain-specific ASC, while little is known about the relation of achievement-related classroom compositions and the various facets of ASC. Additionally, BFLPE-research has been critiqued to build its theoretical frame on social comparison theory, without providing sufficient empirical support. To address this gap, we analyzed how the single student’s social, criterial, absolute, and individual ASC relate to class-level achievement of 8th graders. Applying Multilevel Structural Equation Modeling (MLSEM) we found that all facets of ASC were significantly related to average-class achievement, while student’s social ASC revealed the strongest associated. The results reveal explicitly that average-class achievement is strongly related to social comparison processes. The latter are very attractive materials for signal amplification because liposomes can be loaded with a large amount of signaling molecules, leading to a high sensitivity. In addition, liposomes can be used as a cell-like "bioscaffold" to directly test recognition schemes aiming at cell-related processes. This study demonstrates an easy and fast approach to link the novel hydrophobic optical probe based on [1,3]dioxolo[4,5-f]-[1,3]benzodioxole (DBD dye mm239) with tunable optical properties to hydrophilic recognition elements (e.g., antibodies) using liposomes for signal amplification and as carrier of the hydrophobic dye. The fluorescence properties of mm239 (e.g., long fluorescence lifetime, large Stokes shift, high photostability, and high quantum yield), its high hydrophobicity for efficient anchoring in liposomes, and a maleimide bioreactive group were applied in a unique combination to build a concept for the coupling of antibodies or other protein markers to liposomes (coupling to membranes can be envisaged). The concept further allowed us to avoid multiple dye labeling of the antibody. Here, anti-TAMRA-antibody (DC7-Ab) was attached to the liposomes. In proof-of-concept, steady-state as well as time-resolved fluorescence measurements (e.g., fluorescence depolarization) in combination with single molecule detection (fluorescence correlation spectroscopy, FCS) were used to analyze the binding interaction between DC7-Ab and liposomes as well as the binding of the antigen rhodamine 6G (R6G) to the antibody. The ratio of surfactant/DNA concentration and solution ionic strength determines the result of DNA-surfactant interaction: Complexes with a micelle-like surfactant associates on DNA, which induces DNA shrinkage, DNA precipitation or DNA condensation with the emergence of nanosized particles. UV and fluorescence spectroscopy, low gradient viscometry and flow birefringence methods were employed to investigate DNA-surfactant and surfactant-surfactant interaction at different NaCl concentrations, [NaCl]. It was observed that [NaCl] (or the Debye screening radius) determines the surfactant-surfactant interaction in solutions without DNA. Monomers, micelles and non-micellar associates of azobenzene-containing surfactants with head-to-tail orientation of molecules were distinguished due to the features of their absorption spectra. The novel data enabled us to conclude that exactly the type of associates (together with the concentration of components) determines the result of DNA-surfactant interaction. Predomination of head-to-tail associates at 0.01 M < [NaCl] < 0.5 M induces DNA aggregation and in some cases DNA precipitation. High NaCl concentration (higher than 0.8 M) prevents electrostatic attraction of surfactants to DNA phosphates for complex formation. DAPI dye luminescence in solutions with DNA-surfactant complexes shows that surfactant tails overlap the DNA minor groove. The addition of di- and trivalent metal ions before and after the surfactant binding to DNA indicate that the bound surfactant molecules are located on DNA in islets. In recent studies, we found an association of MDD with an increase of acid sphingomyelinase (ASM) activity. Thus, we asked whether chronic psychosocial stress as a detrimental factor contributing to the emergence of MDD would also affect ASM activity and sphingolipid (SL) metabolism. To induce chronic psychosocial stress in male mice we employed the chronic subordinate colony housing (CSC) paradigm and compared them to non-stressed single housed control (SHC) mice. We determined Asm activity in liver and serum, hepatic SL concentrations as well as hepatic mRNA expression of genes involved in SL metabolism. We found that hepatic Asm activity was increased by 28% (P = 0.006) and secretory Asm activity by 47% (P = 0.002) in stressed mice. C16:0-Cer was increased by 40% (P = 0.008). Gene expression analysis further revealed an increased expression of tumor necrosis factor (TNF)-alpha (P = 0.009) and of several genes involved in SL metabolism (Cers5, P = 0.028; Cers6, P = 0.045; Gba, P = 0.049; Gba2, P = 0.030; Ormdl2, P = 0.034; Smpdl3B; P = 0.013). Our data thus provides first evidence that chronic psychosocial stress, at least in mice, induces alterations in SL metabolism, which in turn might be involved in mediating the adverse health effects of chronic psychosocial stress and peripheral changes occurring in mood disorders. Here, crosslinked poly(epsilon-caprolactone) thermosets (cPCLs) were prepared from linear starting material, whereby the amount of extractable polymer was varied. Fractions of 5-60 wt % of non-crosslinked polymer chains, which freely interpenetrate the crosslinked network, were achieved leading to differences in the resulting phase of the bulk material. This can be described as "sponge-like" with open or closed compartments depending on the amount of interpenetrating polymer. The crosslinking density and the average network chain length remained in a similar range for all network structures, while the theoretical accessible volume for reptation of the free polymer content is affected. This feature could influence or introduce new functions into the material created by thermomechanical treatment. The effect of interpenetrating PCL in cPCLs on the reversible actuation was analyzed by cyclic, uniaxial tensile tests. Here, high reversible strains of up to Delta epsilon = 24% showed the enhanced actuation performance of networks with a non-crosslinked PCL content of 30 wt % resulting from the crystal formation in the phase of the non-crosslinked PCL and co-crystallization with network structures. Additional functionalities are reprogrammability and self-healing capabilities for networks with high contents of extractable polymer enabling reusability and providing durable actuator materials. Most of the United States, Central Europe, and Northeast and West Africa, as well as large parts of India and Indonesia, require the strongest adaptation effort. More than half of the United States needs to at least double their protection within the next two decades. Radiocontrast agents elicit the third most common cause of nephropathy among hospitalized patients, accounting for 11-12% of cases. CIN is connected with clinically significant consequences, including increased morbidity, prolonged hospitalization, increased risk of complications, potential need for dialysis, and increased mortality rate. The number of in hospital examinations using iodine-based contrast media has been significantly increasing over the last decade. In order to protect patients from possible complications of such examinations, new biomarkers are needed that are able to predict a risk of contrast-induced nephropathy. Urinary and plasma cyclic guanosine monophosphate (cGMP) concentrations are influenced by renal function. Urinary cGMP is primarily of renal cellular origin. Therefore, we assessed if urinary cGMP concentration may predict major adverse renal events (MARE) after contrast media exposure during coronary angiography. Methods Urine samples were prospectively collected from non-randomized consecutive patients with either diabetes or preexisting impaired kidney function receiving intra-arterial contrast medium (CM) for emergent or elective coronary angiography at the Charite Campus Mitte, University Hospital Berlin. Urinary cGMP concentration in spot urine was analyzed 24 hours after CM exposure. Patients were followed up over 90 days for occurrence of death, initiation of dialysis, doubling of plasma creatinine concentration or MARE. Results In total, 289 consecutive patients were included into the study. Urine cGMP/creatinine ratio 24 hours before CM exposure expressed as mean +/- SD was predictive for the need of dialysis (no dialysis: 89.77 +/- 92.85 mu M/mM, n = 277; need for dialysis: 140.3 +/- 82.90 mu M/mM, n = 12, p = 0.008), death (no death during follow-up: 90.60 +/- 92.50 mu M/mM, n = 280; death during follow-up: 169.88 +/- 81.52 mu M/mM, n = 9; p = 0.002), and the composite endpoint MARE (no MARE: 86.02 +/- 93.17 mu M/mM, n = 271; MARE: 146.64 +/- 74.68 mu M/mM, n = 18, p<0.001) during the follow-up of 90 days after contrast media application. cGMP/creatinine ratio stayed significantly increased at values exceeding 120 pM/mM in patients who developed MARE, required dialysis or died. Conclusions Urinary cGMP/creatinine ratio >= 120 mu M/mM before CM exposure is a promising biomarker for the need of dialysis and all-cause mortality 90 days after CM exposure in patients with preexisting renal impairment or diabetes.
