Ihre Verwaltungen befinden sich dabei in einem herausfordernden Spannungsfeld: Einerseits haben die SDGs den holistischen Anspruch, vollständig in das Handeln der Kommunen integriert zu werden. Andererseits ist für eine effektive Umsetzung eine starke Anpassung der SDGs an den lokalen Kontext notwendig. Die vorliegende Arbeit betrachtet anhand einer Fallstudie die Frage, wie Kommunen die Nachhaltigkeitsziele der Vereinten Nationen in ihre Handlungsprogramme und Nachhaltigkeitsstrategien übersetzen, und welche Faktoren Einfluss auf diesen Prozess haben. Dabei wird ein translationstheoretischer Ansatz verwendet, der die Übertragung einer Idee in einen lokalen Kontext als aktiven Transfer versteht, bei dem das Handeln der beteiligten Akteure und deren Konstruktion der aufzunehmenden Idee im Fokus steht. Die Translation wird mit Hilfe von qualitativen Interviews nachvollzogen und analysiert. Die Ergebnisse zeigen, dass die SDGs zwar anhand ihrer Relevanz für die Kommune gefiltert werden, der normative Anspruch der SDGs aber erhalten bleibt und angesichts des als gering beurteilten Fortschritts der Kommune besonderes Gewicht erhält. Zentrale Einflussfaktoren für die Translation sind die verfügbaren personellen und finanziellen Ressourcen, die Akzeptanz für die SDGs in Verwaltung, Politik und Gesellschaft und nicht zuletzt das persönliche Engagement einzelner Verwaltungsmitarbeiter*innen. Cities are the Agenda 2030’s focal point. Their local governments however find themselves in a challenging dilemma: On the one hand, the SDG’s holistic approach warrants a wholesale integration into local policy. On the other hand, a substantial adaptation is necessary to integrate the Goals into the local context. This paper uses a case study to examine how municipalities translate the Sustainable Development Goals into their sustainability action plans and strategies. Moreover, it examines which factors are influential to this process. This study uses a translation theory perspective, which characterizes the transfer of an idea into the local context as an active process. It focusses on the actors and how they perceive the transferred idea. For this, qualitative interviews are conducted and analyzed. Thereby, this study shows that while SDGs are being filtered according to their relevancy for the municipality, their normative dimension remains intact. The municipal actors consider this dimension crucial vis-à-vis the lack of progress that they perceive in their municipality. This study finds that core influencing factors are the financial and personnel resources available, the acceptance of SDGs within the administration, politics and society as well as the activism of singular municipal actors. Also within the antibody field the cell-free technology has gained considerable attention with respect to the clinical research pipeline including antibody discovery and production. Besides the classical full-length monoclonal antibodies (mAbs), so-called "nanobodies" (Nbs) have come into focus. A Nb is the smallest naturally-derived functional antibody fragment known and represents the variable domain (VHH, similar to 15 kDa) of a camelid heavy-chain-only antibody (HCAb). Based on their nanoscale and their special structure, Nbs display striking advantages concerning their production, but also their characteristics as binders, such as high stability, diversity, improved tissue penetration and reaching of cavity-like epitopes. The classical way to produce Nbs depends on the use of living cells as production host. Though cell-based production is well-established, it is still time-consuming, laborious and hardly amenable for high-throughput applications. Here, we present for the first time to our knowledge the synthesis of functional Nbs in a standardized mammalian cell-free system based on Chinese hamster ovary (CHO) cell lysates. Cell-free reactions were shown to be time-efficient and easy-to-handle allowing for the "on demand" synthesis of Nbs. In this article, we argue that the translations of Lomonosov's Kratkij rossijskij letopisec should not be studied as an isolated act of cultural transfer but as an episode in a longer history of circulation of knowledge. We demonstrate the complexity of this circulation by reassessing the 'quality' of these translations and positioning them in that longer history of circulation of knowledge by analysing the distribution of historical concepts (Begriffe) in Lomonosov's original and its translations. We show weight gains of 20 healthy children weighed longitudinally from birth to age 1 year, published by Camerer in 1882. The data illustrate the historically prevalent concepts of infant nutrition practiced by German civil servants, lawyers, merchants, university professors, physicians, foresters and farmers. Breastfeeding by the mother was not truly appreciated in those days; children were often breastfed by wet nurses or received bottled milk. Bottle feeding mainly used diluted cow’s milk with some added carbohydrates, without evidence that appropriate amounts of oil, butter or other fatty components were added. French children from 1914 showed similar weight gain patterns suggesting similar feeding practices. The historical data suggest that energy deficient infant formula was fed regularly in the late 19th and early 20th century Europe, regardless of wealth and social class. The data question current concerns that temporarily feeding energy deficient infant formula may warrant serious anxieties regarding long-term cognitive, social and emotional behavioral development. Due to their prokaryotic origin, modern-day chloroplast genomes harbor 100 to 200 genes. These genes encode for core components of the photosynthetic complexes and the chloroplast gene expression machinery, making most of them essential for the viability of the organism. The regulation of those genes is predominated by translational adjustments. The powerful technique of ribosome profiling was successfully used to generate highly resolved pictures of the translational landscape of Arabidopsis thaliana cytosol, identifying translation of upstream open reading frames and long non-coding transcripts. In addition, differences in plastidial translation and ribosomal pausing sites were addressed with this method. However, a highly resolved picture of the chloroplast translatome is missing. Here, with the use of chloroplast isolation and targeted ribosome affinity purification, I generated highly enriched ribosome profiling datasets of the chloroplasts translatome for Nicotiana tabacum in the dark and light. Chloroplast isolation was found unsuitable for the unbiased analysis of translation in the chloroplast but adequate to identify potential co-translational import. Affinity purification was performed for the small and large ribosomal subunit independently. The enriched datasets mirrored the results obtained from whole-cell ribosome profiling. Enhanced translational activity was detected for psbA in the light. An alternative translation initiation mechanism was not identified by selective enrichment of small ribosomal subunit footprints. In sum, this is the first study that used enrichment strategies to obtain high-depth ribosome profiling datasets of chloroplasts to study ribosome subunit distribution and chloroplast associated translation. Ever-changing light intensities are challenging the photosynthetic capacity of photosynthetic organism. Increased light intensities may lead to over-excitation of photosynthetic reaction centers resulting in damage of the photosystem core subunits. Additional to an expensive repair mechanism for the photosystem II core protein D1, photosynthetic organisms developed various features to reduce or prevent photodamage. In the long-term, photosynthetic complex contents are adjusted for the efficient use of experienced irradiation. However, the contribution of chloroplastic gene expression in the acclimation process remained largely unknown. Here, comparative transcriptome and ribosome profiling was performed for the early time points of high-light acclimation in Nicotiana tabacum chloroplasts in a genome-wide scale. The time- course data revealed stable transcript level and only minor changes in translational activity of specific chloroplast genes during high-light acclimation. Yet, psbA translation was increased by two-fold in the high light from shortly after the shift until the end of the experiment. A stress-inducing shift from low- to high light exhibited increased translation only of psbA. This study indicate that acclimation fails to start in the observed time frame and only short-term responses to reduce photoinhibition were observed. Diese kodieren für zentrale Komponenten der Photosynthesekomplexe und des Genexpressionsapparates, was sie für die Lebensfähigkeit des gesamten Organismus essenziell macht. Die leistungsstarke Methode Ribosome Profiling wurde bereits erfolgreich eingesetzt, um hochaufgelöste Bilder der zytosolischen Translationslandschaft von Arabidopsis thaliana zu erstellen, wobei Translation von der Hauptsequenz vorgelagerten, kodierenden Sequenzen und langen, nicht-kodierenden Transkripten identifiziert wurde. Ferner wurden mit dieser Technik Regulationen der Plastidentranslation und spezifische Regionen mit unterschiedlicher Elongationsgeschwindigkeit aufgedeckt. Es fehlen jedoch hochaufgelöste Datensätze des Chloroplasten-Translatoms. Chloroplastenisolation und Affinitätsaufreinigung chloroplastidiärer Ribosomen wurde verwendet, um hochangereicherte Ribosome Profiling-Datensätze des Chloroplastentranslatoms für Nicotiana tabacum im Dunkeln und unter Licht zu erzeugen. Wenngleich sich die Chloroplastenisolation als ungeeignet für eine unverfälschte Analyse der Translation im Chloroplast erwies, ermöglichte sie die Identifizierung von potentiellem co-translationalen Proteinimport. Die entsprechenden Datensätze spiegelten die Ergebnisse des zellulären Ribosome Profilings wider. Für psbA wurde im Licht erhöhte Translationsaktivität festgestellt. Alternative Initiationsmechanismen konnten durch spezifische Anreicherung der kleinen ribosomalen Untereinheit nicht verifiziert werden. Hier wurde ein vergleichendes Transkript- und Ribosomen Profiling für die frühen Zeitpunkte der Akklimatisierung unter Starklicht in Tabakchloroplasten in einem genomweiten Maßstab durchgeführt. Die Zeitverlaufsdaten zeigten ein unverändertes Transkriptniveau und nur geringe Änderungen der translationalen Aktivität von chloroplastidiären Genen im Hochlicht im Vergleich zu Kontrollproben. Die psbA-Translation war jedoch unter Hochlicht schon kurz nach Beginn bis zum Ende des Experiments um etwa das Zweifache erhöht. Der stressinduzierende Wechsel von Schwach- zu Hochlicht bewirkte ebenfalls eine auf psbA-beschränkt, erhöhte Translation. One of the puzzles in evolutionary genetics is therefore to uncover evolutionary driving forces that result in specific codon choice. In many bacteria, the first 5-10 codons of protein-coding genes are often codons that are less frequently used in the rest of the genome, an effect that has been argued to arise from selection for slowed early elongation to reduce ribosome traffic jams. However, genome analysis across many species has demonstrated that the region shows reduced mRNA folding consistent with pressure for efficient translation initiation. This raises the possibility that unusual codon usage is a side effect of selection for reduced mRNA structure. Here we discriminate between these two competing hypotheses, and show that in bacteria selection favours codons that reduce mRNA folding around the translation start, regardless of whether these codons are frequent or rare. Among them, the largest proportion of cellular resources is dedicated to protein translation. Synthetic biology often exploits these resources in executing orthogonal genetic circuits, yet the burden this places on the cell is rarely considered. Here, we develop a minimal model of ribosome allocation dynamics capturing the demands on translation when expressing a synthetic construct together with endogenous genes required for the maintenance of cell physiology. Critically, it contains three key variables related to design parameters of the synthetic construct covering transcript abundance, translation initiation rate, and elongation time. We show that model-predicted changes in ribosome allocation closely match experimental shifts in synthetic protein expression rate and cellular growth. Intriguingly, the model is also able to accurately infer transcript levels and translation times after further exposure to additional ambient stress. Our results demonstrate that a simple model of resource allocation faithfully captures the redistribution of protein synthesis resources when faced with the burden of synthetic gene expression and environmental stress. The tractable nature of the model makes it a versatile tool for exploring the guiding principles of efficient heterologous expression and the indirect interactions that can arise between synthetic circuits and their host chassis because of competition for shared translational resources. However, since many steps of ribosome biogenesis are not thermodynamically favorable at biological conditions, it requires many assembly factors. One group of assembly factors, circularly permuted GTPases, was implicated in 30S subunit maturation in E. coli, by a protein RsgA. RsgA orthologues are present in bacteria and plastid-containing species and in silico analysis revealed presence of a RsgA-like protein in Arabidopsis thaliana. To functionally characterize the Arabidopsis orthologue, two AtRsgA T-DNA insertion lines were analyzed in this study. The exon line (rsgA-e) led to embryo lethality, while the intron line (rsgA-i) caused severe dwarf, pale green phenotype. Further investigation of rsgA-i mutant line revealed defects in chloroplast biogenesis which led to increased number of chloroplasts, decreased chloroplast size, decreased air space between mesophyll cells and smaller shoot apical meristems, which showed unusual proplastid accumulation. Moreover, rsgA-i plants showed reduction in chlorophyll A and B content, decreased electron transport rate and photosynthetic efficiency. Further analyses revealed that the protein is involved in chloroplast 30S subunit maturation. Interestingly, we observed that while chloroplast-targeted and chloroplast-encoded proteins are generally downregulated in the mutant, a contrasting upregulation of the corresponding transcripts is observed, indicating an elaborate compensatory mechanism. To conclude, the study presented here reveals a ribosome assembly factor and a compensatory mechanism activated during impaired chloroplast function. The formation of a functional protein through successful mRNA translation is central to a coordinated cellular response. To what extent the response towards reactive oxygen species (ROS) is regulated at the translational level is poorly understood. Here we analysed leaf- and tissue-specific translatomes using a set of transgenic Arabidopsis thaliana lines expressing a FLAG-tagged ribosomal protein to immunopurify polysome-bound mRNAs before and after oxidative stress. We determined transcript levels of 171 ROS-responsive genes upon paraquat treatment, which causes formation of superoxide radicals, at the whole-organ level. Furthermore, the translation of mRNAs was determined for five cell types: mesophyll, bundle sheath, phloem companion, epidermal and guard cells. Mesophyll and bundle sheath cells showed the strongest response to paraquat treatment. Interestingly, several ROS-responsive transcription factors displayed cell type-specific translation patterns, while others were translated in all cell types. In part, cell type-specific translation could be explained by the length of the 5-untranslated region (5-UTR) and the presence of upstream open reading frames (uORFs). Our analysis reveals insights into the translational regulation of ROS-responsive genes, which is important to understanding cell-specific responses and functions during oxidative stress. The study illustrates the response of different Arabidopsis thaliana leaf cells and tissues to oxidative stress at the translational level, an aspect of reactive oxygen species (ROS) biology that has been little studied in the past. Our data reveal insights into how translational regulation of ROS-responsive genes is fine-tuned at the cellular level, a phenomenon contributing to the integrated physiological response of leaves to stresses involving changes in ROS levels. One of the puzzles in evolutionary genetics is therefore to uncover evolutionary driving forces that result in specific codon choice. In many bacteria, the first 5-10 codons of protein-coding genes are often codons that are less frequently used in the rest of the genome, an effect that has been argued to arise from selection for slowed early elongation to reduce ribosome traffic jams. However, genome analysis across many species has demonstrated that the region shows reduced mRNA folding consistent with pressure for efficient translation initiation. This raises the possibility that unusual codon usage is a side effect of selection for reduced mRNA structure. Here we discriminate between these two competing hypotheses, and show that in bacteria selection favours codons that reduce mRNA folding around the translation start, regardless of whether these codons are frequent or rare. Experiments confirm that primarily mRNA structure, and not codon usage, at the beginning of genes determines the translation rate. Sie basiert auf einem Korpus von 3.200 französischen Originaltiteln und ihren deutschen Neutiteln und schließt eine Forschungslücke der Filmtitelübersetzung für das Sprachenpaar deutsch-französisch. Im theoretischen Teil werden die text- und übersetzungswissenschaftlichen Grundlagen dargelegt. Filmtitel bilden eine eigene Textsorte, die unter Zuhilfenahme der Textualitätskriterien von de Beaugrande/Dressler spezifiziert wird. Anhand ausgewählter Beispiele aus dem Korpus werden maßgebliche Funktionen von Filmtiteln, wie Werbung, Information, Identifikation, Kontakt und Interpretation erörtert. Auf E. Prunčs Translationstypologie basieren jene fünf Strategien, die bei der Übertragung von französischen Filmtiteln in den deutschen Sprach- und Kulturraum zum Einsatz kommen: Identität, Analogie, Variation, Innovation sowie hybride Formen. Ausführlich werden Übersetzungen von Umtitelungen abgegrenzt. Die Auswertung des Korpus ergibt, dass Titelinnovation die am häufigsten angewandte Strategie beim Titeltransfer im gesamten Untersuchungszeitraum darstellt, während Titelidentitäten am seltensten zum Einsatz kommen. Die Betrachtung kürzerer Zeitspannen zeigt gewisse Tendenzen auf, beispielsweise die deutliche Zunahme von Hybridtiteln in jüngster Zeit. Erstmals wird in dieser Arbeit das Phänomen der Mehrfachbetitelungen in verschiedenen deutschsprachigen Ländern aufgegriffen, indem nach Motiven für unterschiedliche Neutitel in Deutschland, der ehemaligen DDR und Österreich gesucht wird. Den Abschluss bildet eine Betrachtung der Filmtitel aus rechtlicher und ökonomischer Perspektive, denn zusammen mit ihren Filmen stellen Titel von hoher Kommerzialität geprägte Texte dar, und wie jedes Wirtschaftsgut erfahren auch sie eine präzise juristische Regulierung. N2 - The empirical study is an analyse of the interlingual transfer of french and german movie titles of the past century. It is based on a corpus of 3,200 french original titles and their new german titles, closing a gap in the research of german and french movie title translation. The theoretical part introduces the basics of the linguistics of texts and translation. Movie titles are a specific kind of text, which is distinguished by the textual criteria of de Beaugrande/Dressler. By using choosen examples of the corpus the mainly functions of movie titles are displayed, as there are promotion, information, identification, contact and interpretation. There are five strategies, basing on the translation typology of Prunč, which are used whenever french movie titles were transferred into german language and culture: identity, analogy, variation, innovation and hybrid forms. The difference between translation and transtiteling is explained in detail. Results of the analytical part are that title innovations are the strategy most often applied when transferring titles, while title identities are used only rarely. The outcome of observing shorter periods is that there are different tendencies, like the significant increase of hybrid titles in the last decades. For the first time this study analyses multiple titles in different german speaking countries, searching for the motivation of alternative new titles in Germany, the former GDR and Austria. The final part is the juristical and economic consideration of movie titles, since motion pictures and their titles are a highly commercialised kind of text and therefore subjects of their corresponding laws. Darüber hinaus kann mit diesem Verfahren ein Brückenschlag zwischen der Synthese der Gene und ihrer kodierenden Proteine auf einer Oberfläche erfolgen. Alle transkribierten RNAs wurden mittels RT-PCR in cDNA umgeschrieben und in einer genspezifischen PCR amplifiziert. Die PCR-Produkte wurden hierfür entweder per Hand oder maschinell auf die Oberfläche transferiert. Über eine Oberflächen-PCR war es möglich, die Gensequenz des Reportergens EGFP direkt auf der Oberfläche zu synthetisieren und anschließend zu transkribieren. Somit war eine Transkription mit weniger als 1 ng an Matrize möglich. Der Vorteil einer Oberflächen-Transkription gegenüber der in Lösung liegt in der mehrfachen Verwendung der immobilisierten Matrize, wie sie in dieser Arbeit dreimal erfolgreich absolviert wurde. Die Oberflächen-Translation des EGFP-Gens konnte ebenfalls zweimal an einer immobilisierten Matrize gezeigt werden, wobei Zweifel über eine echte Festphasen-Translation nicht ausgeräumt werden konnten. Zusammenfassend kann festgestellt werden, dass die Transkription und Translation von immobilisierten Gensequenzen auf planaren Oberflächen möglich ist, wofür die linearen Matrizen direkt auf der Oberfläche synthetisiert werden können. N2 - In vitro mRNA synthesis and in vitro translation are of great interest for biochemical and molecular biological basic research, and also for biotechnology and other applications. Solid phase coupled synthesis is very useful for the development of high throughput procedures to elucidate and manipulate gene products. An artificial gene was constructed combining the T7 promoter and terminator with the EGFP-gene from the plasmid pEGFP. The functionality of the construct was shown by in vitro translation. The gene-construct was immobilised on a planar glass surface. The transcription was performed on the immobilised gene and mRNA was determined by RT-PCR. These results demonstrate that the complete gene is transcribed from the covalently coupled PCR product. Thus, it is possible to transfer a standard transcription technique onto an On-chip reaction. The direct PCR amplification of transcriptionable sequences of EGFP bound on surfaces was successfully used for solid phase transcription. Successful transcriptions were also performed at least to 1 ng of used template. The RNA synthesis was also successful in the second and third reaction on the same slide as observed by signals after RT-PCR. It seems to be possible to transfer the translation of reportergenes in a solid phase coupled synthesis, too. For further integration of cellular procedures on a chip, the cell-free RNA synthesis on immobilised templates is an crucial technical hurdle to conquer. Major advantages of using immobilised templates for transcription are, low risk of contamination occuring in solution, and no necessity of further purification steps for downstream applications of the RNA product.
