In dieser Arbeit wurde gesteinsmagnetische und paleomagnetische Analysen an hemipelagischen Sequenzen von vier Lokationen analysiert. Die Kerne erreichen ein Alter von maximal 300 ky. In Kombination mit TEM, XRD, XRF und geochemischen Analysen zeigt die gesteinsmagnetische Studie, dass detritischer Magnetit das magnetische Signal der glazialen Sedimente dominiert. Die magnetischen Signale der interglazialen Sedimente wurden durch diagenetische Prozesse verändert. Mittels HIRM können Hämatit und Goethit quantifiziert werden. Diese Methode eignet sich, den detritischen Eintrag in den Baikalsee abzuschätzen. Relative Paleointensitäten des Erdmagnetfeldes ergaben reproduizerbare Muster, welche in Korrelation mit gutdatierten Referenzproben die Ableitung eines alternativen Altersmodells für die Datierung der Baikalsedimente ermöglichten. Bei Anwendung des paleomagnetischen Altersmodells beobachtet man, dass die Abkühlung im Baikalgebiet und im Oberflächenwasser des Nordatlantiks wie sie aus den δ18 O-Werten planktonischer Foraminiferen abgeleitet werden kann, zeitgleich ist. Wird das aus benthischen δ18 O-Werten abgeleitete Altermdodell auf den Baikalsee angewandt, ergibt sich eine deutliche Zeitverschiebung. Das benthische Altersmodell repräsentiert die globale Veränderung des Eisvolumens, welche später als die Vänderung der Oberflächenwassertemperatur auftritt. Die Kompilation paleomagnetischer Kurven ergab eine neue relative Paleointensitätskurve “Baikal 200”. Mittels Korngrössenanalyse des Detritus konnten drei Faziestypen mit unterschiedlicher Sedimentationsdynamik unterschieden werden: 1) Glaziale Peroiden werden durch hohe Tongehalte infolge von Windeintrag und durch grobe Sandfraktion mittels Transport durch lokale Winde über das Eis charakterisiert. Dieser Faziestyp deutet auf arides Klima. 2) Während der Glazial/Interglazial-Übergänge steigt die Siltfraktion an. Dies deutet auf erhöhte Feuchtigkeit und damit verbunden erhöhte Sedimentdynamik. Windtransport und in den Schnee der Eisdecke eingetragener Staub sind die vorherrschenden Prozesse, welche den Silt in hemipelagischer Position zur Ablagerung bringen. 3) Während des klimatischen Optimum des Eeemian werden Grösse und Quantität des Silts minimal, was auf eine geschlossene Vegetationsdecke im Hinterland deutet. N2 - Lake Baikal provides an excellent climatic archive for Central Eurasia as global climatic variations are continuously depicted in its sediments. We performed continuous rock magnetic and paleomagnetic analyses on hemipelagic sequences retrieved from 4 underwater highs reaching back 300 ka. The rock magnetic study combined with TEM, XRD, XRF and geochemical analyses evidenced that a magnetite of detrital origin dominates the magnetic signal in glacial sediments whereas interglacial sediments are affected by early diagenesis. HIRM roughly quantifies the hematite and goethite contributions and remains the best proxy for estimating the detrital input in Lake Baikal. Relative paleointensity records of the earth′s magnetic field show a reproducible pattern, which allows for correlation with well-dated reference curves and thus provides an alternative age model for Lake Baikal sediments. Using the paleomagnetic age model we observed that cooling in the Lake Baikal region and cooling of the sea surface water in the North Atlantic, as recorded in planktonic foraminifera δ18 O, are coeval. On the other hand, benthic δ18 O curves record mainly the global ice volume change, which occurs later than the sea surface temperature change. This proves that a dating bias results from an age model based on the correlation of Lake Baikal sedimentary records with benthic δ18 O curves. The compilation of paleomagnetic curves provides a new relative paleointensity curve, “Baikal 200”. With a laser-assisted grain size analysis of the detrital input, three facies types, reflecting different sedimentary dynamics can be distinguished. (1) Glacial periods are characterised by a high clay content mostly due to wind activity and by occurrence of a coarse fraction (sand) transported over the ice by local winds. This fraction gives evidence for aridity in the hinterland. (2) At glacial/interglacial transitions, the quantity of silt increases as the moisture increases, reflecting increased sedimentary dynamics. Wind transport and snow trapping are the dominant process bringing silt to a hemipelagic site (3) During the climatic optimum of the Eemian, the silt size and quantity are minimal due to blanketing of the detrital sources by the vegetal cover. Specifically, we consider the approach of belief change scenarios, a general framework that has been introduced for expressing different forms of belief change. The essential idea is that for a belief change scenario (K, R, C), the set of formulas K, representing the knowledge base, is modified so that the sets of formulas R and C are respectively true in, and consistent with the result. By restricting the form of a belief change scenario, one obtains specific belief change operators including belief revision, contraction, update, and merging. For both the general approach and for specific operators, we give a quantified Boolean formula such that satisfying truth assignments to the free variables correspond to belief change extensions in the original approach. Hence, we reduce the problem of determining the results of a belief change operation to that of satisfiability. This approach has several benefits. First, it furnishes an axiomatic specification of belief change with respect to belief change scenarios. This then leads to further insight into the belief change framework. Second, this axiomatization allows us to identify strict complexity bounds for the considered reasoning tasks. Third, we have implemented these different forms of belief change by means of existing solvers for quantified Boolean formulas. T1 - Integral field spectroscopy of extended Ly alpha emission from the DLA galaxy in Q2233+131 N2 - This paper presents observations of an extended Lyman-alpha emission nebula surrounding the galaxy responsible for the Damped Lyman-alpha Absorption (DLA) line in the spectrum of the quasar Q2233+131. With the Potsdam Multi Aperture Spectrophotometer (PMAS) we measure the properties of the extended Lyalpha emission in an area of 3" x 5" having a total line flux of (2.8 +/- 0.3) x 10(-16) erg cm(-2) s(-1), which at redshift z = 3.15 corresponds to a luminosity of (2.4(-0.2)(+0.3)) x 10(43) erg s(-1) and a size of 23 x 38 kpc. The location of the emission is spatially coincident with the previously detected DLA galaxy, but extends significantly beyond its limb. We argue that the Lya emission is likely to be caused by an outflow from the DLA galaxy, presumably powered by star formation. In the case of negligible dust extinction, the Lya luminosity indicates a star-formation rate of 19 +/- 10 M-. yr(-1) consistent with that derived from the UV continuum flux from the parent galaxy. The wind velocity indicated by the integral field spectra is of the order of several hundred km s(-1). T1 - Mitochondrial DNA and microsatellite variation in the eider duck (Somateria mollissima) indicate stepwise postglacial colonization of Europe and limited current long-distance dispersal N2 - To unravel the postglacial colonization history and the current intercolony dispersal in the common eider, Somateria mollissima, we analysed genetic variation at a part of the mitochondrial control region and five unlinked autosomal microsatellite loci in 175 eiders from 11 breeding colonies, covering the entire European distribution range of this species. As a result of extreme female philopatry, mitochondrial DNA differentiation is substantial both among local colonies and among distant geographical regions. Our study further corroborates the previous hypothesis of a single Pleistocene refugium for European eiders. A nested clade analysis on mitochondrial haplotypes suggests that (i) the Baltic Sea eider population is genetically closest to a presumably ancestral population and that (ii) the postglacial recolonization progressed in a stepwise fashion via the North Sea region and the Faroe Islands to Iceland. Current long-distance dispersal is limited. Differentiation among colonies is much less pronounced at microsatellite loci. The geographical pattern of this nuclear genetic variation is to a large extent explained by isolation by distance. As female dispersal is very limited, the geographical pattern of nuclear variation is probably explained by male-mediated gene flow among breeding colonies. Our study provides genetic evidence for the assumed prominent postglacial colonization route shaping the present terrestrial fauna of the North Atlantic islands Iceland and the Faroes. T1 - Molecular and cytogenetic evidence for cryptic speciation within a rare endemic Malagasy lemur, the Northern Sportive Lemur (Lepilemur septentrionalis) N2 - Evolutionary relationships of different populations of the threatened malagasy lemur Lepilemur septentrimialis were assessed by sequence analysis of mitochondrial DNA (D-loop region and partial Cyt b gene). One hundred and fifty nine samples were collected from five main different localities in the northern part of Madagascar. We applied the phylogenetic species concept based on fixed diagnostic differences to determine the status of different geographical populations. No nucleotide site diagnoses Ankarana from Andrafiamena or Analamera. However, numerous fixed differences separate Sahafary from all other populations. These results were corroborated by phylogenetic trees. As previous cytogenetic studies, our molecular data suggest that two cryptic species of Lepilemur occur in the extreme north of Madagascar. This speciation is probably caused by chromosomal rearrangements in at least one of the evolutionary lineages. Our study comprises another striking example of how molecular genetic assay can detect phylogenetic discontinuities that are not reflected in traditional morphologically based taxonomies. T1 - Genetic analysis of a successful repatriation programme : giant Galapagos tortoises N2 - As natural populations of endangered species dwindle to precarious levels, remaining members are sometimes brought into captivity, allowed to breed and their offspring returned to the natural habitat. One goal of such repatriation programmes is to retain as much of the genetic variation of the species as possible. A taxon of giant GalApagos tortoises on the island of Espahola has been the subject of a captive breeding-repatriation programme for 33 years. Core breeders, consisting of 12 females and three males, have produced more than 1200 offspring that have been released on Espanola where in situ reproduction has recently been observed. Using microsatellite DNA markers, we have determined the maternity and paternity of 132 repatriated offspring. Contributions of the breeders are highly skewed. This has led to a further loss of genetic variation that is detrimental to the long-term survival of the population. Constance) N2 - During 1987-1998, the ciliates and their prey and predator communities in large, deep, mesotrophic Lake Constance were intensively studied as it underwent re-oligotrophication. Ciliate biomass exhibited the bimodal seasonal distribution typical for meso-eutrophic lakes, with high biomass in spring and summer and low biomass in winter and during the clear-water phase. Cluster analysis produced nine groups of temporally co-occurring ciliate morphotypes with potentially similar ecological characteristics. The clusters exhibited a larger seasonality than found in the size distribution, showing that similarly-sized ciliates had seasonally compensatory dynamics. Ciliate biomass declined by approx. 30 % during the 12 years of study, i.e. considerably less than daphnids (and total phosphorus). This yielded a significant increase in the ratio between summer ciliate and daphnid biomass as re-oligotrophication progressed, in contrast to previous studies. Few indications for a mechanistic link between phosphorus concentrations (which declined threefold during the study period) and ciliate biomass or community composition via group-specific food concentrations were found. The relative contribution of three of the nine clusters changed as re-oligotrophication progressed. Ciliate size distribution was related to reoligotrophication and daphnid biomass in summer. The smallest and largest ciliates gained in importance when daphnids decreased whereas large ciliates declined. Overall, summer daphnid biomas had a greater predictive power for attributes of the ciliate community than the other factors studied (phosphorus, prey biomass, copepod biomass). The extent of bottom-up and top-down control of ciliates appeared to be time and group specific. Overall, the ciliate community exhibited remarkably recurrent seasonal patterns despite major alternations in abiotic and biotic conditions. To examine how differences in plankton communities from highly eutrophic and more oligotrophic lakes are reflected in size spectra, a three-year time series of biomass size spectra was established for polymictic, eutrophic Lake Müggelsee, based on approximately weekly sampling and microscopic enumeration. The continuous but often bumpy size spectra reflected appropriately the seasonal and trophy-related variations in the plankton composition and growth conditions and the potential impact of daphnids on smaller plankton. We tested the hypothesis that more diverse plankton communities have smoother size spectra than impoverished ones. The spectra of L. Müggelsee and other more less eutrophic lakes covaried roughly with the functional diversity in total plankton composition but were unrelated to taxonomical diversity within the phyto- or mesozooplankton. The slopes of the normalised size spectra of Lake Müggelsee were generally more negative than -1, exhibited a recurrent seasonal pattern, and were strongly correlated with crustacean biomass. In contrast to less eutrophic systems, slopes could not be used to quantify energy fluxes within the foodweb due to highly variable algal P/B ratios and frequently bumpy size distributions. The latter indicated stronger deviations from the ideal concept of a steady energy flow along the size gradient than found in e. g. large, mesotrophic Lake Constance. Studying B cell activities in vivo or using them for tolerance induction requires that the cells remain unaltered in their biological behavior except for expression of the transgene. As we show here, murine B cells can efficiently be transduced by lentiviral, VSV-G-pseudotyped vectors without the necessity of prior activation. Culture with LPS gave enhanced transduction efficiencies but led to the upregulation of CD86 and proliferation of the cells. Transduction of naive B cells by lentiviral vectors was dependent on multiplicity of infection and did not lead to a concomitant activation. Furthermore, the transduced cells could be used for studies in the NOD mouse system without altering the onset of diabetes. We conclude that lentiviral gene transfer into naive B cells is a powerful tool for manipulation of B cells for therapeutic applications. T1 - Spatial pattern formation in semi-arid shrubland : a priori predicted versus observed pattern characteristics N2 - Ecologists increasingly use spatial statistics to study vegetation patterns. Mostly, however, these techniques are applied in a purely descriptive fashion without a priori statements on the pattern characteristics expected. We formulated such a priori predictions in a study of spatial pattern in a semi-arid Karoo shrubland, South Africa. Both seed dispersal and root competition have been discussed as processes shaping the spatial structure of this community. If either of the two processes dominates pattern formation, patterns within and between shrub functional groups are expected to show distinct deviations from null models. We predicted the type and scale of these deviations and compared predicted to observed pattern characteristics. As predicted by the seed dispersal hypothesis, small-scale co-occurrence within and between groups of colonisers and successors was increased as compared to complete spatially random arrangement of shrubs. The root competition predictions, however, were not met as shrubs of similar rooting depth co- occurred more frequently than expected under random shrub arrangement. Since the distribution of rooting groups to the given shrub locations also failed to match the root competition predictions, there was little evidence for dominance of root competition in pattern formation. Although other processes may contribute to small-scale plant co-occurrence, the sufficient and most parsimonious explanation for the observed pattern is that its formation was dominated by seed dispersal. To characterise point patterns we applied both cumulative (uni- and bivariate K-function) and local (pair- and mark-correlation function) techniques. Based on our results we recommend that future studies of vegetation patterns include local characteristics as they independently describe a pattern at different scales and can be easily related to processes changing with interplant distance in a predictable fashion. In semi-arid Kalahari savannas, heterogeneity is created by savanna structure, i.e. by the spatial arrangement and temporal dynamics of woody plant and open grassland microsites. We formulate a conceptual model describing the effects of savanna dynamics on the population dynamics of the animal-dispersed shrub Grewia flava. From empirical results we derive model rules describing effects of savanna structure on several processes in Grewia's life cycle. By formulating the model, we summarise existing information on Grewia demography and identify gaps in this knowledge. Despite a number of such gaps, the model can be used to make certain quantitative predictions. As an example, we apply the model to investigate the role of seed dispersal in Grewia encroachment on rangelands. Model results show that cattle promote encroachment by depositing substantial numbers of seeds in open areas, where Grewia is otherwise dispersal-limited. Finally, we draw some general conclusions about Grewia's life history and population dynamics. Under natural conditions, concentrated seed deposition under woody plants appears to be a key process causing the observed association between Grewia and other woody plants. Furthermore, low rates of recruitment and high adult survival result in slow-motion dynamics of Grewia populations. As a consequence, Grewia populations interact with savanna dynamics on long temporal and short to intermediate spatial scales. In contrast, a shallower but more expanded freshwater lake existed at 110 kyr BP. The best analog for the most extreme highstand at 135 kyr BP is the highstand during the Early Holocene humid period from 10 to 6 kyr BP. The environmental conditions as reconstructed from diatom assemblages suggest long-lasting episodes of increased humidity during the high lake periods. This contrasts to the modern situation with a relatively shallow Lake Naivasha characterized by rapid water level fluctuations within a few decades. The most likely cause for the variable hydrological conditions since 175 kyr BP is orbitally driven insolation changes on the equator and increased lateral moisture transport from the ocean. These modular tool systems are interfaces between spindle and workpiece and consist of several complicatedly formed parts. Their thermal behavior is dominated by nonlinearities, delay and hysteresis effects even in tools with simpler geometry and it is difficult to describe it theoretically. Due to the dominant nonlinear nature of this behavior the so far used linear regression between the temperatures and the displacements is insufficient. Therefore, in this study we test the hypothesis whether we can reliably predict such thermal displacements via nonlinear temperature-displacement regression functions. These functions are estimated firstly from learning measurements using the alternating conditional expectation (ACE) algorithm and then tested on independent data sets. First, we analyze data that were generated by a finite element spindle model. We find that our approach is a powerful tool to describe the relation between temperatures and displacements for simulated data. Next, we analyze the temperature-displacement relationship in a silent real experimental setup, where the tool system is thermally forced. Again, the ACE-algorithm is powerful to estimate the deformation with high precision. The corresponding errors obtained by using the nonlinear regression approach are 10-fold lower in comparison to multiple linear regression analysis. Finally, we investigate the thermal behavior of a modular tool system in a working milling machine and get again promising results. This model describes a phytoplankton population in a chemostat under periodic supply of nutrients. Previously it has been proven under very general assumptions that such systems are not able to exhibit chaotic dynamics. Here we show that the simple introduction of algal mortality may lead to chaotic oscillations of algal density in the forced chemostat. Our numerical simulations show that the existence of chaos is intimately related to plankton overshooting in the unforced model. We provide a simple measure, based on stability analysis, for estimating the amount of overshooting. These findings are not restricted to the Droop model but hold also for other chemostat models with mortality. It shows several jets, knots and filaments in opposite directions from the central star. A couple of twisted long filaments could be interpreted as due to point-symmetric type ejection. If such is the case, the progenitor would be a binary precessing system. High resolution spectroscopy shows that most of the material is approaching or receding from the star. However the line profiles are very complex, showing several components at different velocities. Our high resolution spectroscopic data show that the different structures (knots, filaments, ...) present different radial velocities spreading from 240 to more than 400 km/s. The system velocity is 300 km/s. There are high velocity knots located to the north of the central star, moving at more than 100 km/s relative to the system velocity. The antioxidant activity of the protein-phenol derivatives was investigated using a modified TEAC assay. The results show that the covalent attachment of quercetin to BSA decreases the total antioxidant activity in comparison to an equivalent amount of free quercetin depending on the degree of derivatization. The derivative with the highest amount of covalently bound quercetin (2:1 derivative) showed an antioxidant activity of only 79% compared to an equivalent amount of free quercetin. After the enzymatic proteolysis of the BSA quercetin derivatives with trypsin, the total antioxidant activity of the degradation products increases in comparison to the respective undigested derivatives, but does not reach the activity of an equivalent amount of free quercetin. Even after 240 minutes of tryptic degradation there is still a lack in antioxidant activity (for the 7:1 derivative nearly 33%) as compared to free quercetin. The reaction products were analyzed in terms of covalent binding at the nucleophilic side chains of proteins. Changes in molecular properties of the proteins derivatives were documented by SDS-PAGE, IEF and SELDI-TOF-MS. The structural changes induced were studied using circular dichroism (CD). The in vitro digestibility was assessed with trypsin. The results show that the occurrence of the catechol moiety, i.e. the two adjacent (ortho) aromatic hydroxyl groups on ring B of the flavonoid structural skeleton appears to be perquisite condition for covalent binding to proteins. The catechol moiety on ring A was less reactive. Its absence lead to a slight or no significant reaction, although non-covalent interactions may still be possible even when lacking this structural element. A comparison of the data is also made with quercetin representing the flavonols. A1 - Feudel, Ulrike T1 - An approach to a process model of laser beam melt ablation using methods of linear and non-linear data analysis N2 - As a non-contact process laser beam melt ablation offers several advantages compared to conventional processing mechanisms. During ablation the surface of the workpiece is molten by the energy of a CO2-laser beam, this melt is then driven out by the impulse of an additional process gas. Although the idea behind laser beam melt ablation is rather simple, the process itself has a major limitation in practical applications: with increasing ablation rate surface quality of the workpiece processed declines rapidly. With different ablation rates different surface structures can be distinguished, which can be characterised by suitable surface parameters. The corresponding regimes of pattern formation are found in linear and non-linear statistical properties of the recorded process emissions as well. While the ablation rate can be represented in terms of the line-energy, this parameter does not provide sufficient information about the full behaviour of the system. The dynamics of the system is dominated by oscillations due to the laser cycle but includes some periodically driven non-linear processes as well. Upon the basis of the measured time series, a corresponding model is developed. The deeper understanding of the process can be used to develop strategies for a process control. N2 - Despite the stated primary goal of this article to investigate sluicing, any discussion of the general conditions on ellipsis must begin with the best investigated case, VP ellipsis in Czech and English. I therefore start with these cases, describing the general results and theoretical findings in this area, and then move to a theory of sluicing (IP-ellipsis) in order to discover how these results apply to a more sophisticated theory of focus- and isomorphism-requirements (e.g. Merchand 2001; Tancredi 1992; Rooth (1992). Finally we turn to the most important question of this study: we ask, how these theoretical findings can be applied to corpus data. We assume that both VP-e. and IP-e. are only licensed under the condition of focus feature marking, but not purely as a requirement on isomorphism of two adjunct structures.
